In a recent article dealing with the Tangale Peak or Kilang, as it is called in the local Tangale language, Herrmann JUNGRAITHMAYR presents an account narrated by a Tangale elder about the attempted ascent of that characteristic mountain by a British colonial officer and his subsequent death.1 Kilang mountain is a basaltic cone approximately 1300 m high, about 8 km southwest of Kaltungo, one of the principal settlements of the Tangale people, in southern Bauchi State, northeastern Nigeria. During a research stay at the National Archives in Kaduna in November 1993 I was able to consult a file containing various documents relating to this incident in detail.2 In the following note I present an outline of the events based on the evidence in the colonial records. By doing this I not only intend to shed more light on a tragic event from the very early years of the colonial era. The picture of the circumstances emerging from the investigations of the colonial authorities may serve as a background to the narrative by the Tangale elder presented in JUNGRAITHMAYR's publication.
The craggy and hilly Muri Mountains, which are situated to the north of the Benue Lowlands, are an area with a complex pattern of settlement. This roughly 80 km long and 20 km wide mountainous area is inhabited by about 20 ethnic groups belonging to different language families. The present ethnic and linguistic situation is understood as the result of a complex series of migrations and adaptations to the natural environment. This paper will describe actual movements of settlements and consider certain conditions which may have been relevant in the decision to leave a settlement or choose a new one. The most important conditions will be the accessibility of arable land and/or pasture, accessibility of water, and conditions dependent on the historical and political context such as affording of security and possibility of defence. Therefore an interdisciplinary approach seems to be appropriate to evaluate the natural conditions for settlement and cultivation of the various places from a geographer´s point of view, to interrogate into the historical aspects and motifs of the settlement patterns and migrations with a thorough ethnological background, as well as to gain additional information from a linguistic analysis of toponymes and contact phenomena of the languages spoken in the area.
Population structure was estimated in a continuous population of a small land snail (Trochoidea geyeri). Mark-recapture experiments and randomly amplified polymorphic DNA analyses indicate that the population structure can be described by the isolation by distance model of Wright (1946). Estimates of density and dispersal suggest a neighbourhood size of 70-208 individuals on an area of 13-21 m². A principal component analysis of the randomly amplified polymorphic DNA data reveals clinal variation of genetic composition across the population, as predicted by the neighbourhood concept. An analysis of molecular variance indicates substantial genetic structuring. Comparisons of the genetic distances, expressed as euclidean distances among individuals, versus the geographic distance between sampling sites yield a highly significant positive correlation (Mantel test: r = 0.567, p<0.0001). The revealed pattern of populational subdivision on a microgeographic scale seems to be one of the principal processes generating and maintaining genetic diversity within populations of small land gastropods.

In the present study the population genetic structure of the terrestrial snail Pomatias elegans was related to habitat structure on a microspatial scale. The genetic variability of 1607 individuals from 51 sampling sites in five different populations in Provence, France, was studied with an allozyme marker using population genetic methods, Mantel tests and spatial autocorrelation techniques were applied to different connectivity networks accounting for the structural features of the landscape. It is suggested that the population structure is, to a large extent, a function of the habitat quality, quantified as population density, and of the spatial arrangement of the habitat in the landscape and not of the geographical distance per se. In fragmented habitats, random genetic drift was the prevailing force for sampling sites separated by a few hundred meters.
Background: Growth rate is central to the development of cells in all organisms. However, little is known about the impact of changing growth rates. We used continuous cultures to control growth rate and studied the transcriptional program of the model eukaryote Saccharomyces cerevisiae, with generation times varying between 2 and 35 hours.
Results: A total of 5930 transcripts were identified at the different growth rates studied. Consensus clustering of these revealed that half of all yeast genes are affected by the specific growth rate, and that the changes are similar to those found when cells are exposed to different types of stress (>80% overlap). Genes with decreased transcript levels in response to faster growth are largely of unknown function (>50%) whereas genes with increased transcript levels are involved in macromolecular biosynthesis such as those that encode ribosomal proteins. This group also covers most targets of the transcriptional activator RAP1, which is also known to be involved in replication. A positive correlation between the location of replication origins and the location of growth-regulated genes suggests a role for replication in growth rate regulation.
Conclusion: Our data show that the cellular growth rate has great influence on transcriptional regulation. This, in turn, implies that one should be cautious when comparing mutants with different growth rates. Our findings also indicate that much of the regulation is coordinated via the chromosomal location of the affected genes, which may be valuable information for the control of heterologous gene expression in metabolic engineering.
The main purpose of the paper is to explain the divergent paths of development of ethno-territorial protest movements in modern democratic political systems. By focusing on the interaction between these movements and the state, the different systems of accommodation between the relevant regional and central elites will be analyzed. The study concentrates on the case studies of Québec (Canada) and Corsica (France). The paper is divided into three parts. The first part describes the traditional systems of accommodation in France and Canada. The second part is focused on the process of socio-economic modernization in the 1950s and 1960s in those countries that threatened the established patterns of elite accommodation. The third part deals with the consequences for the established patterns of elite-accommodation and new concepts of territorial management that the central states tried to establish. By looking at the different degrees of centralization and decentralization in the mentioned political systems, the question of access to the political system by new social and political actors will be discussed in detail.
Length variations of repetitive sequences in different AT-rich loop-coding regions of mitochondrial 16S rDNA in two gastropod species were discovered during intraspecific haplotype surveys. Examination of the discrete length variation of the basic repeat unit in a phylogenetic framework led to the conclusion of a microsatellite-like mutational dynamic. The observations suggest that the presence of a repetitive sequence structure alone is sufficient to trigger this dynamic.
I analysed the importance of shell size, shell shape, habitat preferences and availability, experienced climate, active dispersal and influence of Pleistocene glaciations for the range sizes of 37 Western Palaearctic Helicidae s.l. species for which a phylogeny was available. In both cross-species and phylogenetically controlled analyses, the range sizes were positively correlated to climatic tolerance, shell size, active dispersal and influence of Pleistocene glaciations. In addition, range sizes increased significantly with latitude. Multiple regression suggested that, predominantly, the influence of Pleistocene glaciations, tolerance to large annual temperature ranges and shell size influenced the distributional range sizes. Habitat preference, range and availability, active dispersal and shell shape explained no additional variance. The results suggest that the processes influencing species range size of the Helicidae s.l. are mainly related to the climatic shifts after the Pleistocene.
Alzheimer's disease-related mutations in the presenilin-1 gene (PS1) are leading to an elevated production of neurotoxic beta-amyloid 1-42 and may additionally enhance oxidative stress. Here, we provide in vivo evidence indicating that brains of transgenic mice expressing different human Alzheimer-linked PS1 mutations exhibit a reduced activity of two antioxidant enzymes. For this purpose, mice transgenic for human PS1 and for single and multiple PS1 mutations were generated. Mice with multiple PS1 mutations showed a significantly decreased activity of the antioxidant enzymes Cu/Zn superoxide dismutase and glutathione reductase already at an age of 3-4 months. As expected, this effect was less pronounced for the mice with a single PS1 mutation. By contrast, animals bearing normal human PS1 showed significantly elevated enzyme activities relative to non-transgenic littermate controls.
Background Reliable taxonomic identification at the species level is the basis for many biological disciplines. In order to distinguish species, it is necessary that taxonomic characters allow for the separation of individuals into recognisable, homogeneous groups that differ from other such groups in a consistent way. We compared here the suitability and efficacy of traditionally used shell morphology and DNA-based methods to distinguish among species of the freshwater snail genus Radix (Basommatophora, Pulmonata). Results Morphometric analysis showed that shell shape was unsuitable to define homogeneous, recognisable entities, because the variation was continuous. On the other hand, the Molecularly defined Operational Taxonomic Units (MOTU), inferred from mitochondrial COI sequence variation, proved to be congruent with biological species, inferred from geographic distribution patterns, congruence with nuclear markers and crossing experiments. Moreover, it could be shown that the phenotypically plastic shell variation is mostly determined by the environmental conditions experienced. Conclusion Contrary to DNA-taxonomy, shell morphology was not suitable for delimiting and recognising species in Radix. As the situation encountered here seems to be widespread in invertebrates, we propose DNA-taxonomy as a reliable, comparable, and objective means for species identification in biological research.
Enhanced apoptosis and elevated levels of reactive oxygen species (ROS) play a major role in aging. In addition, several neurodegenerative diseases are associated with increased oxidative stress and apoptosis in neuronal tissue. Antioxidative treatment has neuro-protective effects. The aim of the present study was to evaluate changes of susceptibility to apoptotic cell death by oxidative stress in aging and its inhibition by the antioxidant Ginkgo biloba extract EGb761. We investigated basal and ROS-induced levels of apoptotic lymphocytes derived from the spleen in young (3 months) and old (24 months) mice. ROS were induced by 2-deoxy-D-ribose (dRib) that depletes the intracellular pool of reduced glutathione. Lymphocytes from aged mice accumulate apoptotic cells to a significantly higher extent under basal conditions compared to cells from young mice. Treatment with dRib enhanced this difference, implicating a higher sensitivity to ROS in aging. Apoptosis can be reduced in vitro by treatment with EGb761. In addition, mice were treated daily with 100mg/kg EGb761 per os over a period of two weeks. ROS-induced apoptosis was significantly reduced in the EGb761 group. Interestingly, this effect seemed to be more pronounced in old mice.
Apoptosis seems to be involved in immunosenescence associated with aging. Moreover, in lymphocytes (PBL) of patients with Alzheimer's disease, an increased susceptibility to the apoptotic pathway has been described possibly due to impaired protection of oxidative stress. Accordingly, it seemed to be of particular interest to investigate the contribution of normal aging to the susceptibility from human lymphocytes to programmed cell death. We could show that PBL from elderly individuals (>60 years) accumulate apoptosing cells to a significant higher extent in spontaneous and activation-induced cell death compared to younger controls (<35 years). Treatment with the oxidative stressor 2-deoxy-D-ribose or with agonistic-CD95-antibody pronounced this effect even more implicating a higher sensitivity to reactive oxygen species and a higher functional CD95 expression, respectively. In addition, expression of the activation markers HLA-DR and CD95 was significantly increased in CD3+-cells of aged subjects, while expression of CD25 did not seem to be affected by age. Expression of Bcl-2 was increased in aging and correlated with the number of apoptotic cells.
The relevance of physiological immune aging is of great interest with respect to determining disorders with pathologic immune function in aging individuals. In recent years, the relevance of changes in peripheral lymphocytes in age-associated neurologic diseases has become more evident. Due to the lack of immunological studies, covering more than one event after mitogenic activation, we envisaged a new concept in the present study, aiming to investigate several events, starting from T cell receptor (TCR) ligation up to T cell proliferation. In addition, we addressed the question whether changes are present in the subsets (CD4, CD8) with aging. Phosphorylation of tyrosine residues declines with increasing age in CD4+ cells. Fewer levels of CD69 positive cells after 4 h mitogenic activation, altered expression of cytokines (IL2, IFN-gamma and TNF-alpha; 22 h) and lower proliferation (72 h) were determined in aging. Moreover, it could be shown that CD8+ lymphocytes react more effectively to mitogenic stimulation with reference to CD69 expression and proliferation in both age groups (<35 and >60 years old). These data indicate that T cell activation, mediated by TCR engagement, is significantly impaired in aging and both subsets are affected. However, bypassing the TCR does not fully restore T cell function, indicating that there are more mechanisms involved than impaired signal transduction through TCR only. The results will be discussed in relation to their relevance in neurodegenerative and psychiatric disorders.
In vivo manipulation of interleukin-2 expression by a retroviral tetracycline (tet)-regulated system

We have used the tetracycline (tet)-regulated system as described previously to evaluate the applicability of controlled gene expression in cancer gene therapy. As a model gene, we used the human interleukin-2 (IL-2) gene, which has been placed under the transcriptional control of the tetO/promoter. Human melanoma cells were transduced by two modified retroviral tet vectors containing the transactivator regulatory unit and the IL-2 gene driven by the tetO/promoter, respectively. In the absence of tet, IL-2 expression in the target cells was stable over several months. IL-2 production was in the range of 40 U/106 cells/24 hours. A fine tuning of IL-2 expression could be achieved by culturing the transduced cells with increasing doses of tet, whereby a concentration of 500 ng/mL tet in the culture medium abrogated IL-2 expression. Most importantly for clinical application, IL-2 expression by the transduced melanoma cells could also be regulated in vivo. When nu/nu mice were inoculated with the transduced tumor cells, they failed to develop tumors. Instead, the inhibition of IL-2 expression in the transduced tumor cells by oral administration of tet led to subcutaneous tumor growth; this growth rate was comparable with the growth rate of subcutaneously inoculated untransduced parental cells. The finding demonstrates the applicability of the tet-regulated system in cancer gene therapy.
Many cases of early-onset inherited Alzheimer's disease (AD) are caused by mutations in the presenilin-1 (PS1) gene. Expression of PS1 mutations in cell culture systems and in primary neurons from transgenic mice increases their vulnerability to cell death. Interestingly, enhanced vulnerability to cell death has also been demonstrated for peripheral lymphocytes from AD patients. We now report that lymphocytes from PS1 mutant transgenic mice show a similar hypersensitivity to cell death as do peripheral cells from AD patients and several cell culture systems expressing PS1 mutations. The cell death-enhancing action of mutant PS1 was associated with increased production of reactive oxygen species and altered calcium regulation, but not with changes of mitochondrial cytochrome c. Our study further emphasizes the pathogenic role of mutant PS1 and may provide the fundamental basis for new efforts to close the gap between studies using neuronal cell lines transfected with mutant PS1, neurons from transgenic animals, and peripheral cells from AD patients. In large models of neuronal cell death, there is a tight correlation between Cdk5 deregulation and cell-cycle dysfunction. However, pathways that link Cdk5 to the cell cycle during neuronal death are still unclear. We have investigated the molecular events that precede p25/Cdk5-triggered neuronal death using a neuronal cell line that allows inducible p25 expression. In this system, no sign of apoptosis was seen before 24 hours of p25 induction. Thus, at that time, cell-cycle-regulatory proteins were analysed by immunoblotting and some of them showed a significant deregulation. Interestingly, after time-course experiments, the earliest feature correlated with p25 expression was the phosphorylation of the retinoblastoma protein (Rb). Indeed, this phosphorylation was observed 6 hours after p25 induction and was abolished in the presence of a Cdk5 inhibitor, roscovitine, which does not inhibit the usual Rb cyclin-D kinases Cdk4 and Cdk6. Furthermore, analyses of levels and subcellular localization of Cdk-related cyclins did not reveal any change following Cdk5 activation, arguing for a direct effect of Cdk5 activity on Rb protein. This latter result was clearly demonstrated by in vitro kinase assays showing that the p25-Cdk5 complex in our cell system phosphorylates Rb directly without the need for any intermediary kinase activity. Hence, Rb might be an appropriate candidate that connects Cdk5 to cell-cycle deregulation during neuronal cell death.
The identification of specific genetic (presenilin-1 [PS1] and amyloid precursor protein [APP] mutations) and environmental factors responsible for Alzheimer's disease (AD) has revealed evidence for a shared pathway of neuronal death. Moreover, AD-specific cell defects may be observed in many other nonneuronal cells (e.g., lymphocytes). Thus, lymphocytes may serve as a cellular system in which to study risk factors of sporadic, as well as genetic AD in vivo. The aim of our present study was to clarify whether lymphocytes bearing genetic or sporadic risk factors of AD share an increased susceptibility to cell death. Additionally we examined whether a cell typespecific vulnerability pattern was present and how normal aging, the main risk factor of sporadic AD, contributes to changes in susceptibility to cell death. Here, we report that lymphocytes affected by sporadic or genetic APP and PS1 AD risk factors share an increased vulnerability to cell death and exhibit a similar cell type-specific pattern, given that enhanced vulnerability was most strongly developed in the CD4+ T-cell subtype. In this paradigm, sporadic risk factors revealed the highest impact on cell type-specific sensitivity of CD4+ T cells to apoptosis. In contrast, normal aging results in an increased susceptibility to apoptosis of both, CD4+ and CD8+ T cells.
Introduction: Reactive oxygen species (ROS) have been implicated in neurodegeneration and seem to be involved in the physiology and pathophysiology of several diseases, including normal aging and Alzheimer’s disease (AD). Enhanced ROS production in aging or AD is not restricted to the brain, but can also been seen in several peripheral tissues. The objective of the present study was to evaluate whether the mechanisms involved in the generation of oxidative stress in normal senescence and Alzheimer’s disease are identical or not. Methods: We analysed intracellular basal levels of ROS in lymphocytes from AD patients and healthy young and aged not-demented subjects as well as ROS levels following stimulation with d-ribose and staurosporine in all three groups. ROS levels were measured by flow cytometry using the intracellular fluorescence dye dihydrorhodamine123 (DHR123). Results: Our study shows that AD lymphocytes have increased basal levels of ROS, low susceptibility to ROS stimulation by 2-deoxy-D-ribose (dRib) and an increased response to staurosporine when compared with age-matched controls. Discussion: The data suggest that the defect(s) responsible for enhanced ROS production in AD may involve different or additional biological pathways than those involved in enhanced ROS generation during aging.
As major sources of reactive oxygen species (ROS), mitochondrial structures are exposed to high concentrations of ROS and may therefore be particularly susceptible to oxidative damage. Mitochondrial damage could play a pivotal role in the cell death decision. A decrease in mitochondrial energy charge and redox state, loss of transmembrane potential (depolarization), mitochondrial respiratory chain impairment, and release of substances such as calcium and cytochrome c all contribute to apoptosis. These mitochondrial abnormalities may constitute a part of the spectrum of chronic oxidative stress in Alzheimer's disease. Accumulation of amyloid beta (Abeta) in form of senile plaques is also thought to play a central role in the pathogenesis of Alzheimer's disease mediated by oxidative stress. In addition, increasing evidence shows that Abeta generates free radicals in vitro, which mediate the toxicity of this peptide. In our study, PC12 cells were used to examine the protective features of EGb 761(definition see editorial) on mitochondria stressed with hydrogen peroxide and antimycin, an inhibitor of complex III. In addition, we investigated the efficacy of EGb 761 in Abeta-induced MTT reduction in PC12 cells. Moreover, we examined the effects of EGb 761 on ROS levels and ROS-induced apoptosis in lymphocytes from aged mice after in vivo administration. Here, we will report that EGb 761 was able to protect mitochondria from the attack of hydrogen peroxide, antimycin and Abeta. Furthermore, EGb 761 reduced ROS levels and ROS-induced apoptosis in lymphocytes from aged mice treated orally with EGb 761 for 2 weeks. Our data further emphasize neuroprotective properties of EGb 761, such as protection against Abeta-toxicity, and antiapoptotic properties, which are probably due to its preventive effects on mitochondria.
FGF-2, a potent multifunctional and neurotrophic growth factor, is widely expressed in the brain and upregulated in cerebral ischemia. Previous studies have shown that intraventricularly or systemically administered FGF-2 reduces the size of cerebral infarcts. Whether endogenous FGF-2 is beneficial for the outcome of cerebral ischemia has not been investigated. We have used mice with a null mutation of the fgf2 gene to explore the relevance of endogenous FGF-2 in brain ischemia. Focal cerebral ischemia was produced by occlusion of the middle cerebral artery (MCAO). We found a 75% increase in infarct volume in fgf2 knock-out mice versus wild type littermates (P < 0.05). This difference in the extent of ischemic damage was observed after 24 h, and correlated with decreased viability in fgf2 mutant mice following MCA occlusion. Increased infarct volume in fgf2 null mice was associated with a loss of induction in hippocampal BDNF and trkB mRNA expression. These findings indicate that signaling through trkB may contribute to ameliorating brain damage following ischemia and that bdnf and trkB may be target genes of FGF-2. Together, our data provide the first evidence that endogenous FGF-2 is important in coping with ischemic brain damage suggesting fgf2 as one crucial target gene for new therapeutic strategies in brain ischemia.
Im März 2000 hat der Europäische Rat von Lissabon das strategische Ziel der EU formuliert, "die Union zum wettbewerbsfähigsten und dynamischsten wissensbasierten Wirtschaftsraum der Welt zu machen – ... der fähig ist, ein dauerhaftes Wirtschaftswachstum mit mehr und besseren Arbeitsplätzen und einem größeren sozialen Zusammenhalt zu schaffen". Die Umsetzung soll u. a. durch die "Offene Methode der Koordinierung" (OMK) vorangetrieben werden. Damit wird eine Leitungs- und Koordinierungsfunktion des Rates auch auf solchen Politikfeldern angestrebt, die nach dem Vertragsrecht in die alleinige Gestaltungskompetenz der Mitgliedstaaten fallen. Die OMK lässt sich als eine Art Benchmarking-Verfahren ohne unmittelbar verpflichtende Konsequenzen für die Mitgliedstaaten beschreiben. Ein ähnliches Vorgehen wird seit 1997 in der Beschäftigungspolitik praktiziert. Mit der Lissabon-Strategie wird die OMK sukzessive auf weitere sozialpolitische Bereiche ausgedehnt. Im Bereich der Alterssicherung kommt sie ebenso zum Einsatz wie bei der Bekämpfung von Armut und sozialer Ausgrenzung. Künftig wird auch die Gesundheitspolitik der Mitgliedsländer auf dem Prüfstand der OMK stehen.
Die Rolle des Kulturbildes im interkulturell motivierten Deutschunterricht am Beispiel des Iran

Der Vortrag basiert auf meiner Dissertation über die "Interkulturelle Kommunikation an iranischen Sprachinstituten und Universitäten". Er setzt sich zunächst mit den Vorstellungen über die Kultur des deutschsprachigen Raums im Iran und dessen Reflexion in Unterrichtsprozessen auseinander. In diesem Zusammenhang wird die Rolle der Beziehung zwischen Kultur und Sprache im Fremdsprachenunterricht herausgearbeitet. Daher steht die Beschäftigung mit Kulturfragen im Mittelpunkt. Es handelt sich dabei vor allem um zwei typische Beispiele für "interkulturelle Missverständnisse", die bei iranischen Deutschlernenden in ihrem Verständnis der deutschen Kultur sehr häufig vorkommen. Zwei weitere Beispiele werden gezeigt, in denen die kontrastierten Kulturen miteinander übereinstimmen und somit eine feste und unproblematische Grundlage für den Umgang miteinander darstellen. Im Folgenden soll die Rolle der Deutschlehrer und der institutionellen Bedingungen im Unterricht analysiert werden. Gerade der Deutschunterricht in islamischen Ländern ist darauf angewiesen, dass der Lehrer Sprachsituationen bereitstellt, anhand derer er versucht, die von der Ausgangsgesellschaft stammenden interkulturellen Konflikte aufzugreifen und letztlich zu bewältigen. Meine Unterrichtsanalysen zeigen, dass diese Vorgehensweise der Lehrer ohne Steuerung der Institution nicht möglich ist. Da die interkulturell motivierte Deutschvermittlung institutionelle Voraussetzungen verlangt, stellt der Vortrag auch die Frage, ob eine regionale von den beiden Regierungen unabhängige Institution für die Beschäftigung mit den möglichen interkulturellen Konflikten notwendig ist und warum sie bis jetzt nicht zustande gekommen ist. Trotz der neusten politischen Bemühungen um die kulturellen Annäherungen zwischen beiden Ländern gelten immer noch beide Kulturen als "kontrastiert". Der Deutschunterricht in der iranischen Gesellschaft kann diese Kluft nur dann schmälern, wenn die Deutsch anbietenden Institutionen inter- und transkulturelle Aspekte stärker berücksichtigen.
Since the early 1960s ethno-territorial movements have increasingly challenged established patterns of political integration in western democracies. The purpose of this paper is to compare the formation and different paths of development of such nationalist movements in Quebec (Canada) and Corsica (France). The paper presents a brief outline of an argument more fully developed in a study on the two cases. After a short introduction I will first discuss theoretical aspects of minority nationalism. Secondly, I will try to give an overview over major findings and conclusions of my study.
Background The reciprocal (9;22) translocation fuses the bcr (breakpoint cluster region) gene on chromosome 22 to the abl (Abelson-leukemia-virus) gene on chromosome 9. Depending on the breakpoint on chromosome 22 (the Philadelphia chromosome – Ph+) the derivative 9+ encodes either the p40(ABL/BCR) fusion transcript, detectable in about 65% patients suffering from chronic myeloid leukemia, or the p96(ABL/BCR) fusion transcript, detectable in 100% of Ph+ acute lymphatic leukemia patients. The ABL/BCRs are N-terminally truncated BCR mutants. The fact that BCR contains Rho-GEF and Rac-GAP functions strongly suggest an important role in cytoskeleton modeling by regulating the activity of Rho-like GTPases, such as Rho, Rac and cdc42. We, therefore, compared the function of the ABL/BCR proteins with that of wild-type BCR. Methods We investigated the effects of BCR and ABL/BCRs i.) on the activation status of Rho, Rac and cdc42 in GTPase-activation assays; ii.) on the actin cytoskeleton by direct immunofluorescence; and iii) on cell motility by studying migration into a three-dimensional stroma spheroid model, adhesion on an endothelial cell layer under shear stress in a flow chamber model, and chemotaxis and endothelial transmigration in a transwell model with an SDF-1α gradient. Results Here we show that both ABL/BCRs lost fundamental functional features of BCR regarding the regulation of small Rho-like GTPases with negative consequences on cell motility, in particular on the capacity to adhere to endothelial cells. Conclusion Our data presented here describe for the first time an analysis of the biological function of the reciprocal t(9;22) ABL/BCR fusion proteins in comparison to their physiological counterpart BCR.
Kurzfassung eines Vortrags gehalten auf der GMA-Tagung vom 04.11.-06.11.2005 in Münster. Aufgrund wenig überzeugender Leistungen seiner Studierenden im vorklinischen Studienabschnitt hat der Fachbereich Medizin der Goethe-Universität zum Wintersemester 2001/02 eine völlig neue Studienordnung eingeführt, deren wesentliche Ziele das erfolgreiche Erlernen natur-, geistes- und sozialwissenschaftlicher Komponenten der normalen Biologie des Menschen mit optimaler Vorbereitung auf den klinischen Studienabschnitt binnen zweier Studienjahre und die Kompatibilität mit der ÄAppO und der ZÄAppO (jeweils alte und neue Version bzw. Vorentwurf) sind. Grundprinzipien des Curriculums sind: 1. Logisch aufeinander abgestimmte Themenblöcke, 2. Integration von makroskopischer und mikroskopischer Anatomie, 3. Interdisziplinärität der Themenblöcke, 4. Immanente Wiederholung von Lehrinhalten durch enge Verzahnung von Plenar- und Kleingruppenveranstaltungen, 5. Aufteilung der Studienanfänger in gemischte Gruppen nach den Kriterien Geschlecht, Herkunft und Studienfach, 6. Erlernen des Umgangs mit Patienten schon vom ersten Semester an durch "Anatomie am Lebenden"-Kurse und durch den Kurs Einführung in die klinische Medizin. Wichtigste Ergebnisse: Steigerung der Referenzgruppe beim Physikum von ca. 25% auf >60%, relativ guter Prüfungserfolg, hervorragende Sozialisation durch die frühe randomisierte Zuordnung in Kleingruppen und der sehr glatte Übergang in den klinischen Abschnitt des Medizin- bzw. Zahnmedizinstudiums.
Von Ahasver, dem ‚Ewigen Juden’ wird schon im Druck von 1602 erzählt, er habe, nachdem er von Christus zur ewigen Wanderschaft verdammt worden war, die Stadt Jerusalem verlassen und durch alle Teile der Welt wandern müssen. Als er nach Jahrhunderten wieder in das Heilige Land gekommen sei, habe er das Land verwüstet und Jerusalem so vollständig zerstört vorgefunden, dass er es nicht mehr erkannt habe. Mit dieser Bemerkung signalisiert der Autor des kleinen, aber überaus wirkungsmächtigen Traktats den Lesern, dass damit mehr gemeint ist als nur eine Episode auf der Wanderschaft Ahasvers. [...]
Gute naturwissenschaftliche Vorkenntnisse sind insbesondere für den vorklinischen Studienabschnitt wichtig. Wegen der heterogenen Auswahl von Leistungskursen und Abituranforderungen kann jedoch nicht unbedingt von einem einheitlichen Wissensstand ausgegangen werden. Daher wurde versucht, mit einem Testbogen aus insgesamt 40 Aufgaben zur Biologie, Chemie, Mathematik und Physik den Wissensstand der Studienanfänger in Humanmedizin in Deutschland zu quantifizieren. Der Fragebogen enthielt neben Faktenaufgaben auch Anwendungen vor allem mathematischer und chemischer Prinzipien. Alle Fragen mussten durch Freitextantworten oder Skizzen beantwortet werden. Teilgenommen haben insgesamt 2 935 Studienanfänger des Wintersemesters 2004/2005 von 14 deutschen Universitäten (etwa 40% des Jahrganges). Im Mittel wurden 14,34 der 40 Aufgaben richtig beantwortet; etwas bessere Kenntnisse wurden in den 15 Biologiefragen (6,89) und den 8 Mathematikfragen erreicht (3,23), während vor allem in Chemie (2,18 von 10 Fragen) und Physik (1,55 von 8 Fragen) große Wissenslücken bestehen. Die Ergebnisse bestätigen, dass die naturwissenschaftlichen Vorkenntnisse der Studienanfänger schlecht sind; sie erfordern einen größeren Zeitaufwand für die Vermittlung des Abiturwissens im ersten vorklinischen Semester. Sinnvoll erscheint alternativ die verpflichtende Teilnahme an Zusatzkursen in diesen Fächern vor Aufnahme des eigentlichen Fachstudiums.
Mit der Neuregelung der Studienplatzverteilung von 2005 haben die Universitäten in Deutschland die Möglichkeit, bis zu 60% der Studienplätze nach universitätseigenen Kriterien zu vergeben. Implizit wird vom Gesetzgeber und der öffentlichen Meinung gefordert, nicht-leistungsbezogene Kriterien und Persönlichkeitsmerkmale verstärkt zur Bewerberauswahl einzusetzen (Motivation, Identifikation, Vermeidung von Fehlvorstellungen). Da in Anbetracht der Bewerberzahlen mündliche Auswahlgespräche als ungeeignet erscheinen, wurde vom Fachbereich Medizin der Johann Wolfgang Goethe Universität ein Fragebogen entworfen, um nichtschulische Leistungen zu erfassen. Dieser Fragebogen wurde am Beginn des Wintersemesters 2005/2006 von allen Studienanfängern der JWG-Universität Frankfurt und der Medizinischen Universität Innsbruck ausgefüllt. Entgegen der initialen Erwartungen der Verfasser gaben nur etwa 15% Prozent Medizin-spezifische berufliche Vorerfahrungen an (Rettungsdienst, Ausbildung als Krankenschwester/pfleger oder ähnliches); dagegen wurden von etwa 60% angegeben, mindestens ein Musikinstrument zu spielen oder länger sportlich aktiv gewesen zu sein. Die Zusammenstellung der Selbstangaben zeigt, dass Medizin-relevante Vorkenntnisse nur bei einem kleinen Anteil der Studienbewerber in größerem Umfang vorhanden sind. Aufgrund der großen Streuung in der Art und Dauer der angegebenen Vorleistungen sollte die Erhebung von Parametern zur Beurteilung von soft skills, z.B. durch Online-Fragebogen, als (Vor)Selektionsinstrument nur sehr vorsichtig eingesetzt werden.
Maßgeblich unter dem wachsenden externen Druck hat die didaktische Qualifizierung in den medizinischen Fakultäten an Bedeutung gewonnen. Im Rahmen der Professionalisierung der medizinischen Aus-, Fort- und Weiterbildung ist eine pädagogisch-didaktische Ausbildung der Lehrenden unumgänglich. Um Orientierung und Argumentationshilfe zu geben, werden in einer dreiteiligen Artikelfolge Stellenwert der Medizindidaktik, Anforderungsprofil der Angebote und Konzepte zur Implementierung und zur Erfolgsmessung für den deutschsprachigen Raum beleuchtet. In Teil II beleuchten wir den Ausbildungsbedarf und erstellen ein Anforderungsprofil für ein strukturiertes systematisches Qualifizierungsangebot. Der Hauptbedarf besteht in der Qualifizierung der Lehrenden, die den täglichen Unterricht durchführen. Insbesondere sie brauchen das Handwerkszeug, das ihnen erlaubt, ihre verschiedenen Lehraufgaben effizienter zu bewältigen. Seitens der Fakultät und der Studierenden bestehen Ansprüche an qualifizierte Lehrende; ebenso bestehen Forderungen seitens der Lehrenden an adäquat qualifizierende Kurse. Zur Umsetzung dieser berechtigten Ansprüche sind modular aufgebaute Programme nötig, die eine zumindest national qualitativ und quantitativ vergleichbare Ausbildung gewährleisten. Anforderungen an derartige Kurse sind bereits definiert und lokal zum Beispiel in Baden-Württemberg und zum Teil auch in Nordrhein-Westfalen umgesetzt. Es gilt nun, diese auf breiter Basis in die Praxis umzusetzen.
Alle Leistungsnachweise des klinischen Studienabschnittes nach neuer Ärztlicher Approbationsordnung müssen benotet werden; hierzu sind in der Regel schriftliche Prüfungen notwendig. Bisher erprobte Methoden beinhalten die Prüfung passiven Wissens (Einfachauswahlfragen, multiple choice-Fragen, progress test-Fragen) und aktiven Wissens (short essay questions, long essay questions). Vor- und Nachteile dieser Verfahren werden diskutiert, sowie die zur Erstellung, Durchführung und Auswertung schriftlicher Prüfungen notwendigen Ressourcen.

Im internationalen Vergleich fehlt in Deutschland ein allgemein verbindliches "Basis-Programm", das die täglich Lehrenden systematisch auf ihre Ausbildungsaufgaben vorbereitet. Dies ist bisher nur lokal umgesetzt wie zum Beispiel in Baden-Württemberg mit dem ministeriell zertifizierten Programm der Medizindidaktischen Qualifikation I und II. Vergleichbares ist in Nordrhein-Westfalen und Bayern im Aufbau.
The interaction of T cells and antigen-presenting cells is central to adaptive immunity and involves the formation of immunological synapses in many cases. The surface molecules of the cells form a characteristic spatial pattern whose formation mechanisms and function are largely unknown. We perform computer simulations of recent experiments on geometrically repatterned immunological synapses and explain the emerging structure as well as the formation dynamics. Only the combination of in vitro experiments and computer simulations has the potential to pinpoint the kind of interactions involved. The presented simulations make clear predictions for the structure of the immunological synapse and elucidate the role of a self-organizing attraction between complexes of T cell receptor and peptide–MHC molecule, versus a centrally directed motion of these complexes.
Enzymatic and antisense effects of a specific anti-Ki-ras ribozyme in vitro and in cell culture

Due to their mode of action, ribozymes show antisense effects in addition to their specific cleavage activity. In the present study we investigated whether a hammerhead ribozyme is capable of cleaving mutated Ki-ras mRNA in a pancreatic carcinoma cell line and whether antisense effects contribute to the activity of the ribozyme. A 2[prime]-O-allyl modified hammerhead ribozyme was designed to cleave specifically the mutated form of the Ki-ras mRNA (GUU motif in codon 12). The activity was monitored by RT-PCR on Ki-ras RNA expression by determination of the relative amount of wild type to mutant Ki-ras mRNA, by 5-bromo-2[prime]-deoxy-uridine incorporation on cell proliferation and by colony formation in soft agar on malignancy in the human pancreatic adenocarcinoma cell line CFPAC-1, which is heterozygous for the Ki-ras mutation. A catalytically inactive ribozyme was used as control to differentiate between antisense and cleavage activity and a ribozyme with random guide sequences as negative control. The catalytically active anti-Ki-ras ribozyme was at least 2-fold more potent in decreasing cellular Ki-ras mRNA levels, inhibiting cell proliferation and colony formation in soft agar than the catalytically inactive ribozyme. The catalytically active anti-Ki-ras ribozyme, but not the catalytically inactive or random ribozyme, increased the ratio of wild type to mutated Ki-ras mRNA in CFPAC-1 cells. In conclusion, both cleavage activity and antisense effects contribute to the activity of the catalytically active anti-Ki-ras hammerhead ribozyme. Specific ribozymes might be useful in the treatment of pancreatic carcinomas containing an oncogenic GTT mutation in codon 12 of the Ki-ras gene.

The conditionally-lethal pso4-1 mutant allele of the spliceosomal-associated PRP19 gene allowed us to study this gene’s influence on pre-mRNA processing, DNA repair and sporulation. Phenotypes related to intron-containing genes were correlated to temperature. Splicing reporter systems and RT–PCR showed splicing efficiency in pso4-1 to be inversely correlated to growth temperature. A single amino acid substitution, replacing leucine with serine, was identified within the N-terminal region of the pso4-1 allele and was shown to affect the interacting properties of Pso4-1p. Amongst 24 interacting clones isolated in a two-hybrid screening, seven could be identified as parts of the RAD2, RLF2 and DBR1 genes. RAD2 encodes an endonuclease indispensable for nucleotide excision repair (NER), RLF2 encodes the major subunit of the chromatin assembly factor I, whose deletion results in sensitivity to UVC radiation, while DBR1 encodes the lariat RNA splicing debranching enzyme, which degrades intron lariat structures during splicing. Characterization of mutagen-sensitive phenotypes of rad2{Delta}, rlf2{Delta} and pso4-1 single and double mutant strains showed enhanced sensitivity for the rad2{Delta} pso4-1 and rlf2{Delta} pso4-1 double mutants, suggesting a functional interference of these proteins in DNA repair processes in Saccharomyces cerevisiae.
Chemically modified bases are frequently used to stabilize nucleic acids, to study the driving forces for nucleic acid structure formation and to tune DNA and RNA hybridization conditions. In particular, fluorobenzene and fluorobenzimidazole base analogues can act as universal bases able to pair with any natural base and to stabilize RNA duplex formation. Although these base analogues are compatible with an A-form RNA geometry, little is known about the influence on the fine structure and conformational dynamics of RNA. In the present study, nano-second molecular dynamics (MD) simulations have been performed to characterize the dynamics of RNA duplexes containing a central 1'-deoxy-1'-(2,4-difluorophenyl)-ß-D-ribofuranose base pair or opposite to an adenine base. For comparison, RNA with a central uridine:adenine pair and a 1'-deoxy-1'-(phenyl)-ß-D-ribofuranose opposite to an adenine was also investigated. The MD simulations indicate a stable overall A-form geometry for the RNAs with base analogues. However, the presence of the base analogues caused a locally enhanced mobility of the central bases inducing mainly base pair shear and opening motions. No stable ‘base-paired’ geometry was found for the base analogue pair or the base analogue:adenine pairs, which explains in part the universal base character of these analogues. Instead, the conformational fluctuations of the base analogues lead to an enhanced accessibility of the bases in the major and minor grooves of the helix compared with a regular base pair.
The radiation-sensitive mutant pso4-1 of Saccharomyces cerevisiae shows a pleiotropic phenotype, including sensitivity to DNA cross-linking agents, nearly blocked sporulation and reduced mutability. We have cloned the putative yeast DNA repair gene PSO4 from a genomic library by complementation of the blocked UV-induced mutagenesis and of sporulation in diploids homozygous for pso4-1. Sequence analysis revealed that gene PSO4 consists of 1512 bp located upstream of UBI4 on chromosome XII and encodes a putative protein of 56.7 kDa. PSO4 is allelic to PRP19, a gene encoding a spliceosome-associated protein, but shares no significant homology with other yeast genes. Gene disruption with a destroyed reading frame of our PSO4 clone resulted in death of haploid cells, confirming the finding that PSO4/PRP19 is an essential gene. Thus, PSO4 is the third essential DNA repair gene found in the yeast S.cerevisiae.
Mismatch repair is a highly conserved system that ensures replication fidelity by repairing mispairs after DNA synthesis. In humans, the two protein heterodimers hMutSα (hMSH2‐hMSH6) and hMutLα (hMLH1‐hPMS2) constitute the centre of the repair reaction. After recognising a DNA replication error, hMutSα recruits hMutLα, which then is thought to transduce the repair signal to the excision machinery. We have expressed an ATPase mutant of hMutLα as well as its individual subunits hMLH1 and hPMS2 and fragments of hMLH1, followed by examination of their interaction properties with hMutSα using a novel interaction assay. We show that, although the interaction requires ATP, hMutLα does not need to hydrolyse this nucleotide to join hMutSα on DNA, suggesting that ATP hydrolysis by hMutLα happens downstream of complex formation. The analysis of the individual subunits of hMutLα demonstrated that the hMutSα–hMutLα interaction is predominantly conferred by hMLH1. Further experiments revealed that only the N‐terminus of hMLH1 confers this interaction. In contrast, only the C‐terminus stabilised and co‐immunoprecipitated hPMS2 when both proteins were co‐expressed in 293T cells, indicating that dimerisation and stabilisation are mediated by the C‐terminal part of hMLH1. We also examined another human homologue of bacterial MutL, hMutLβ (hMLH1–hPMS1). We show that hMutLβ interacts as efficiently with hMutSα as hMutLα, and that it predominantly binds to hMutSα via hMLH1 as well.
Aim of the study: Investigation, whether water-filtered infrared-A (wIRA) irradiation during moderate bicycle ergometer endurance exercise has effects especially on local fat reduction and on weight reduction beyond the effects of ergometer exercise alone.
Methods: Randomised controlled study with 40 obese females (BMI 30-40 (median: 34.5), body weight 76-125 (median: 94.9) kg, age 20-40 (median: 35.5) years, isocaloric nutrition), 20 in the wIRA group and 20 in the control group. In both groups each participant performed 3 times per week over 4 weeks for 45 minutes bicycle ergometer endurance exercise with a constant load according to a lactate level of 2 mmol/l (aerobic endurance load, as determined before the intervention period). In the wIRA group in addition large parts of the body (including waist, hip, and thighs) were irradiated during all ergometries of the intervention period with visible light and a predominant part of water-filtered infrared-A (wIRA), using the irradiation unit “Hydrosun® 6000” with 10 wIRA radiators (Hydrosun® Medizintechnik, Müllheim, Germany, radiator type 500, 4 mm water cuvette, yellow filter, water-filtered spectrum 500-1400 nm) around a speed independent bicycle ergometer.
Main variable of interest: change of “the sum of circumferences of waist, hip, and both thighs of each patient” over the intervention period (4 weeks). Additional variables of interest: body weight, body mass index BMI, body fat percentage, fat mass, fat-free mass, water mass (analysis of body composition by tetrapolar bioimpedance analysis), assessment of an arteriosclerotic risk profile by blood investigation of variables of lipid metabolism (cholesterol, triglycerides, high density lipoproteins HDL, low density lipoproteins LDL, apolipoprotein A1, apolipoprotein B), clinical chemistry (fasting glucose, alanin-aminotransferase ALT (= glutamyl pyruvic transaminase GPT), gamma-glutamyl-transferase GGT, creatinine, albumin), endocrinology (leptin, adiponectin (= adipo Q), homocysteine, insulin). All variables were at least measured before and after the intervention period. Ergometry (ECG, blood pressure behaviour, lactate curve with power at 2, 3 and 4 mmol/l) before the intervention period. In addition: nutrition training ahead of and during the intervention period with a nutrition protocol over one week for assessment of the daily energy intake; calculation of basic metabolic rate and total energy requirement. Assessment of undesired effects.
Only methods of non-parametric statistics were used, both descriptive (median, percentiles of 25 and 75 (= interquartile range), minimum, maximum) and confirmatory (two-sided Mann-Whitney U test for unpaired samples for the only one main variable of interest). Total error probability: .05 (5%). An intention to treat analysis ITT with last observed carry forward method was used preferably (presented results) and in addition an on treatment analysis OT. Only 2 (treatment group) and 4 (control group) drop-outs occurred (mostly due to lack of time).
Results: The “sum of circumferences of waist, hip, and both thighs of each patient” decreased during the 4 weeks significantly more (p<.001) in the wIRA group than in the control group: medians and interquartile ranges: -8.0 cm (-10.5 cm/-4.1 cm) vs. -1.8 cm (-4.4 cm/0.0 cm).
As well “body weight of each patient” decreased during the 4 weeks markedly more in the wIRA group than in the control group: medians and interquartile ranges: -1.9 kg (-4.0 kg/0.0 kg) vs. 0.0 kg (-1.5 kg/+0.4 kg); median of body weight changed from 99.3 kg to 95.6 kg (wIRA) vs. 89.9 kg to 89.6 kg (control). A similar effect showed the body mass index BMI.
Blood variables of interest remained unchanged or showed some slight improvements during the treatment period, concerning most variables with no obvious differences between the two groups; insulin showed a slight trend to decrease in the wIRA group and to increase in the control group.
Undesired effects of the treatment were not seen.
Discussion: The results of the study suggest, that wIRA – during moderate bicycle ergometer endurance exercise as lipolytic stimulus – increases local lipolysis with a local fat reduction (thighs) in the otherwise bradytrophic fatty tissue. The presumably underlying mechanisms of wIRA have already been proven: wIRA acts both by thermal effects and by non-thermal effects. Thermal effects of wIRA are the generation of a therapeutic field of warmth with the increase of tissue temperature, tissue oxygen partial pressure, and tissue blood flow, and by this regional metabolism. As fatty tissue normally has a slow metabolism (bradytrophic and hypothermic tissue) with a low rate of lipolysis, wIRA can increase lipolysis in fatty tissue and the mobilized fats are burned in musculature during the ergometer exercise.
Conclusion: The results of the study indicate, that wIRA irradiation during moderate ergometer endurance exercise can be used – in combination with an appropriate nutrition – to improve body composition, especially local fat distribution, and the reduction of fat and body weight in obese persons.
RNA interference (RNAi) has emerged as a powerful tool to induce loss-of-function phenotypes by post-transcriptional silencing of gene expression. In this study we wondered whether inducible RNAi-cassettes integrated into cellular DNA possess the power to trigger neoplastic growth. For this purpose inducible RNAi vectors containing tetracycline (Tet)-responsive derivatives of the H1 promoter for the conditional expression of short hairpin RNA (shRNA) were used to target human polo-like kinase 1 (Plk1), which is overexpressed in a broad spectrum of human tumors. In the absence of doxycycline (Dox) HeLa clones expressing TetR, that carry the RNAi-cassette stably integrated, exhibited no significant alteration in Plk1 expression levels. In contrast, exposure to Dox led to marked downregulation of Plk1 mRNA to 3% and Plk1 protein to 14% in cell culture compared to mismatch shRNA/Plk1-expressing cells. As a result of Plk1 depletion cell proliferation decreased to 17%. Furthermore, for harnessing RNAi for silencing disease-related genes in vivo we transplanted inducible RNAi-HeLa cells onto nude mice. After administration of Dox knockdown of Plk1 expression was observed correlating to a significant inhibition of tumor growth. Taken together, our data revealed that genomically integrated RNAi-elements are suitable to hamper tumor growth by conditional expression of shRNA.
Natural objects provide partially redundant information to the brain through different sensory modalities. For example, voices and faces both give information about the speech content, age, and gender of a person. Thanks to this redundancy, multimodal recognition is fast, robust, and automatic. In unimodal perception, however, only part of the information about an object is available. Here, we addressed whether, even under conditions of unimodal sensory input, crossmodal neural circuits that have been shaped by previous associative learning become activated and underpin a performance benefit. We measured brain activity with functional magnetic resonance imaging before, while, and after participants learned to associate either sensory redundant stimuli, i.e. voices and faces, or arbitrary multimodal combinations, i.e. voices and written names, ring tones, and cell phones or brand names of these cell phones. After learning, participants were better at recognizing unimodal auditory voices that had been paired with faces than those paired with written names, and association of voices with faces resulted in an increased functional coupling between voice and face areas. No such effects were observed for ring tones that had been paired with cell phones or names. These findings demonstrate that brief exposure to ecologically valid and sensory redundant stimulus pairs, such as voices and faces, induces specific multisensory associations. Consistent with predictive coding theories, associative representations become thereafter available for unimodal perception and facilitate object recognition. These data suggest that for natural objects effective predictive signals can be generated across sensory systems and proceed by optimization of functional connectivity between specialized cortical sensory modules.
Background The proto-oncogene pituitary tumor-transforming gene (PTTG) has been shown to be abundantly overexpressed in a large variety of neoplasms likely promoting neo-vascularization and tumor invasiveness. In this study, we investigated a potential role for PTTG mRNA expression as a marker to evaluate the future clinical outcome of patients diagnosed with primary cancer of the head and neck. Methods Tumor samples derived from primary tumors of 89 patients suffering from a squamous cell carcinoma were analyzed for PTTG mRNA-expression and compared to corresponding unaffected tissue. Expression levels were correlated to standard clinico-pathological parameters based on a five year observation period. Results In almost all 89 tumor samples PTTG was found to be overexpressed (median fold increase: 2.1) when compared to the unaffected tissue specimens derived from the same patient. The nodal stage correlated with PTTG transcript levels with significant differences between pN0 (median expression: 1.32) and pN+ (median expression: 2.12; P = 0.016). In patients who developed a tumor recurrence we detected a significantly higher PTTG expression in primary tumors (median expression: 2.63) when compared to patients who did not develop a tumor recurrence (median expression: 1.29; P = 0.009). Since the median expression of PTTG in patients with tumor stage T1/2N0M0 that received surgery alone without tumor recurrence was 0.94 versus 3.82 in patients suffering from a tumor recurrence (P = 0.006), PTTG expression might provide a feasible mean of predicting tumor recurrence. Conclusion Elevated PTTG transcript levels might be used as a prognostic biomarker for future clinical outcome (i.e. recurrence) in primary squamous cell carcinomas of the head and neck, especially in early stages of tumor development.
Zu der grössern Zahl interessanter Urkunden, deren erste Kenntniss man dem Sammlerfleisse des Nikolaus Kindlinger verdankt, gehören einige, die das Bischofs- und Dienstmannenrecht von Köln im 12. und 13. Jahrhundert zum Gegenstand haben und dem Herausgeber bei seiner Bearbeitung des kurkölnischen Archivs bekannt geworden waren. An einem wenig zugängIichen Platze untergebracht, als urkundliche Beigabe zu einer Untersuchung, mit der sie nur lose zusammenhängen, haben sie längst verdient, der Forschung näher und bequemer gerückt zu werden. Ist dieser Vortheil nun auch schon den Hauptstücken zu Theil geworden, so hat es doch bis jetzt an einer kritischen Revision der Texte, an einer Vereinigung der Urkunden an einer Stelle und an einer zusammenhiingenden sachlichen Erläuterung derselben gefehlt. Diese Aufgaben zu lösen ist der nachstehende Aufsatz bestimmt, der in drei Abtheilungen: I. das lateinische Dienstrecht von Köln (Jura ministerialium beati Petri); II. das deutsche Dienstrecht von Köln; III. den Aufsatz : de servitio cotidiano Coloniensis archiepiscopi behandeln will.

Am Ende des zweiten Jahrtausends wie am Ende des ersten sind die Menschen von eschatologischen Vorstellungen beunruhigt und fasziniert zugleich. Der Tod wird wieder entindividualisiert und mir Vorliebe im traditionellen Bild des Totentanzes und des tanzenden Todes ausgedrückt. Dies hat auch Auswirkungen auf die wissenschaftliche Beschäftigung mit diesem Thema, die in den letzten Jahren intensiviert wurde. Einige der neueren Arbeiten zu den spätmittelalterlichen Totentänzen und zur Geschichte ihrer produktiven Rezeption werden hier vorgestellt und im Kontext der Traditionsforschung diskutiert.
Aus der Notwendigkeit heraus, "nachhaltig die Funktionen des Bodens zu sichern" (§1 BBodSchG), und damit auch Bodenschutz vorsorgend in Planungsprozesse zu integrieren, wurde ein neues Bodenschutzkonzept entwickelt. Es basiert auf einer differenzierten, aber gleichzeitig nachvollziehbaren Bodenbewertung. Das Problem bei der Bodenbewertung ist, dass etwas bewertet werden soll, für das - je nach Fragestellung - immer wieder neue Ziele definiert werden müssen. Deshalb liegt der Bodenbewertung ein Zielsystem zu Grunde, das Schutzziele klar festlegt und mit Hilfe dessen die Bodenbewertung nachvollziehbar wird. Für das Bodenschutzkonzept werden aus der Vielzahl möglicher Kriterien wichtige vorgestellt, aus denen die - bezogen auf dieses Zielsystem - wesentlichen ausgewählt werden können. Um aussagekräftige Daten für diese Kriterien zu erhalten stützt sich die Bodenbewertung auf bodenkundliche sowie landschaftsgenetisch-geomorphologische Zusammenhänge. Die eigentliche Bewertung erfolgt dann in drei Schritten: zuerst eine Einzelbewertung, dann zusammengefasst nach den Bodenfunktionen Lebensraumfunktion, Regelungsfunktion, Informationsfunktion, dem Eigenwert des Bodens (Schutzwürdigkeit) sowie der Empfindlichkeit und Gefährdung (Schutzbedürftigkeit). Im dritten Schritt werden diese Bewertungen dann zu einer gewichteten, verbal-argumentativen Gesamtbewertung der Schutzwürdigkeit und Schutzbedürftigkeit zusammengefasst. Mit Hilfe des Bewertungsverfahrens werden auch Zielkonflikte zwischen den unterschiedlichen Schutzgütern offengelegt. Schutzmaßnahmen ergeben sich dann stringent aus den vorher im Zielsystem gesetzten Prämissen, d.h., Ziele und Maßnahmen sind begründbar gewählt, stehen in einem ökologischen Gesamtzusammenhang und lassen sich sehr gut nachvollziehen. Das hier vorgestellte, neue Bodenschutzkonzept ist für verschiedene Planungsebenen geeignet. Es ist in unterschiedlichen Naturräumen anwendbar, kann verschiedene Schutzziele mit Hilfe des Zielsystems bestimmen und so z.B. die Naturraumvielfalt in einem Gebiet ebenso berücksichtigen wie die Meinungsvielfalt, was unter vorsorgendem Bodenschutz zu verstehen sei.
Multiple choice (MC)-Klausuren sind im deutschen Medizinstudium trotz weitgehend fehlender Daten zur Validität dieser Prüfungsform zur Regelprüfung geworden. Darüber hinaus ist unklar, in welchem Ausmaß die Studierenden - auch solche mit guten Prüfungsergebnissen - den geprüften Lernstoff tatsächlich beherrschen. Am Fachbereich Medizin der Johann-Wolfgang-Goethe-Universität Frankfurt wurde am Ende des SS 2003 im Fach Mikrobiologie für die Studierenden des 2. klinischen Semesters eine MC-basierte Abschlussprüfung geschrieben. Die Studierenden des 1. klinischen Semesters hatten - bedingt durch Umstellungen des Curriculums - eine identische Ausbildung. Diese wurde durch eine inhaltlich weitgehend identische, im Format aber andere Klausur abgeschlossen, in der sowohl offene Fragen enthalten waren als auch Fragen, bei denen die Studierenden jede Aussage einzeln auf Korrektheit bewerten mussten. Der Vergleich der Ergebnisse für inhaltlich gleiche Fragen zeigt, dass die Studierenden im MC-Format eine hohe Quote richtiger Antworten erzielen, diese jedoch durch ein geändertes Fragenformat stark reduziert wird. So erreichten nur 20 - 30% der Studierenden ein vollständig richtiges Ergebnis, wenn jede Aussage einzeln bewertet werden musste, während die inhaltlich gleiche Frage im MC-Format 80 - 90% richtige Ergebnisse erzielte. In freien Fragen konnten nur 30 - 40% der Studierenden die richtige Antwort aktiv niederschreiben, während 90 -99% der Studierenden die richtige Lösung passiv erkannten. Wir interpretieren diese Ergebnisse dahin, dass der Entscheidungszwang in MC-basierten Fragen einen starken Einfluss auf die Quote richtiger Antworten hat, und die Prüfungsergebnisse damit wesentlich durch das Format beeinflusst werden, das Wissen dagegen nicht beherrscht wird. Die Ergebnisse dieser Studie legen nahe, Sorgfalt bei der Auswahl des Prüfungsverfahrens walten zu lassen und der Steuerung des studentischen Lernverhaltens durch das Prüfungsformat wesentlich mehr Aufmerksamkeit zu widmen als bisher.
Die "Relatio de legatione Constantinopolitana" des Liudprand von Cremona und seine Gesandtschaft des Jahres 968 nach Konstantinopel zum Kaiser Nikephoros II. Phokas ist von byzantinistischer Seite bereits mehrfachi untersucht worden. Daß Liudprand und seine Schriften hauptsächlich Gegenstand der mediävistischen Forschung geblieben sind, ist nur natürlich. Und daß dabei die im engeren Sinne byzantinistischen Belange zweitrangig sind, ist ebenfalls normal. Gelegentlich jedoch können - so scheint mir - Informationen, die Liudprand über Byzanz mitteilt, auch in einem westlichen Kontext gesehen werden. Der Bischof von Cremona schrieb ja schließlich nicht für die Byzantiner; sondern sah als sein vorrangiges Publikum Otto I. und dessen Hof an. Ich werde deshalb an dieser Stelle versuchen, zunächst die Hintergründe für Liudprand von Cremonas Kapitel 39-41 der "Relatio" auf der Basis des eben angesprochenen byzantinischen Hintergrunds auszuleuchten und werde in einem zweiten Teil, der allerdings aufs engste mit dem Background für diese Kapitel der "Relatio" des Liudprand zusammenhängt, auf zwei Schriften eines bedeutenden byzantinischen Gelehrten des 10. Jhs. - Niketas Paphlagon - aufmerksam machen, die nicht nur einen sehr interessanten Hinweis auf Kontakte dieses byzantinischen Theologen mit westlichen Bischöfen enthalten, sondern durch ihren Inhalt endzeitliche Erwartungen in Byzanz wie im lateinisclien Westen belegen. Sie sind bisher völlig unbeachtet geblieben. Ich beabsichtige allerdings nicht, in die alte Debatte der Mediävistik um den "terreur de l'an mille" einzugreifen. Mir geht es allein darum, zu zeigen, daß auch in Byzanz ein Wissen um die Gefahren der Zeit um das Jahr 1000 a.D. existierte, auch wenn dieses sich teilweise aus ganz anderen Quellen als analoge Phänomene im Westen speiste. ...
Die Fibel, das zeigt die gewiß nicht affirmativ gemeinte Lehrlingslektion überdeutlich, muß an die sozialen Erfahrungen der Kinder anknüpfen, um einmal Motivationen schaffen für das Lesenlernen, wozu am Ende des ersten Schuljahres bereits das "weiterführende" Lesen gehört; davon jedoch nicht abzutrennen ist das erzieherische Ziel, dem das Lesen vom ersten Schultag an verpflichtet sein sollte: die sozialen Erfahrungen der Kinder in ihrer gesellschaftlichen Verursachung aufklären zu helfen. Vor dieser generellen Forderung, der viele Fibelherausgeber zustimmen werden, erweisen sich neuere Fibeln insgesamt als in hohem Maße reformbedürftig, auch wenn hier und da schon erfreuliche Reformtendenzen wahrnehmbar sind. Einige Beobachtungen, die direkt Gegenstand der Analyse waren oder nur am Rande notiert wurden, lassen sich am Ende dieser Arbeit - zumindest als Hypothesen für die weitere Diskussion - generalisieren. Fibeln, von der Lesebuchdiskussion nicht berücksichtigt, wurden von dieser dennoch mitbeeinflußt. Die kindliche Umwelt, breit gefächert in die verschiedenen Aktionsbereiche, ist längst zentraler Gegenstand neuerer Fibeln. Die zivilisatorisch veralteten Texte und Illustrationen mit der Mutter am Waschzuber und dem Sämann mit der Säschüssel über der Schulter sind verschwunden bzw. ersetzt durch modernisierte; d. h. aber auch, daß Waschtag und Landarbeit, um bei den genannten Beispielen zu bleiben, noch vielfach Gegenstand von Fibellektionen sind, auch wenn sie in ihrer Bedeutung für die Erfahrungswelt der meisten Kinder gewiß keine Priorität mehr besitzen. Daraus folgt für viele Fibeln nach der Phase der technologischen Modernisierung der alten Inhalte die Notwendigkeit, die Relevanz dieser Inhalte für Sechs/Siebenjährige zu überprüfen. Aber auch dieser Prozeß, der in vollem Gange ist, betrifft noch nicht den Punkt einer Fibelreform, da sich auch bei einer konsequenten Berücksichtigung von Themenbereichen aus der kindlichen Umwelt deren Konfliktzonen aussparen lassen. Genau dieses Verfahren aber provoziert den entscheidenden Vorbehalt auch noch gegen fast neueren Fibeln, in denen wichtige, weil erfahrungsträchtige kindliche Spiel- und Aktionsräume beschrieben werden, ohne daß die dabei auftretenden individuellen, altersbedingten oder durch gesellschaftliche Verhältnisse verursachte Konflikte Berücksichtigung fänden. So entsteht weiterhin das unwahre Bild einer heilen Welt, in der Kinder durchweg eine liebevolle, fleißige Mutter haben und ab und an auch einen netten Vater; in der heiter, ohne Zank und Verdruß, mit Puppen und Eisenbahnen gespielt, in der gesungen, gerodelt und gebadet wird. Verschwiegen werden all die üblen Erfahrungen die Kinder machen müssen: mit ihren Spielsachen, die, kaum benutzt, kaputt gehen; mit anderen Kindern, die aggressiv sind und nicht spielen können; mit ihren Eltern, die geschieden sind, sich längst hätten trennen sollen oder einfach nur normal sind, was schon schlimm genug sein kann; mit Eltern, die ihre Arbeit ruiniert, die nervös, mürrisch oder brutal sind; mit Wohnungen, die keinen Spielraum bieten; mit Hinterhöfen, Vorgärten, Grünanlagen und Straßen, die verboten sind; mit Spielplätzen, die weit weg, zu klein oder von pedantischen Rentnern bewacht werden; mit Schulen, die all das nicht kümmert und darum vielen Kindern verhaßt oder sterbenslangweilig sind usw. Fibelherausgeber, die bereit sind, die kindlichen Bedürfnisse, die sich in solchen negativen Erfahrungen anmelden, in Texten und Illustrationen zu vertreten, müssen auch berücksichtigen, daß die landläufigen Vorstellungen von kindlicher Umwelt bisher geprägt waren von der Umwelt der Mittelschicht Es wäre zwar ein Fortschritt und in gewissem Umfang auch im Interesse aller Kinder, wenn wenigstens, wie das in "Wunderbare Sachen" in einer Reihe von Lektionen konsequent geschieht, Konflikte von Mittelschichtkindern dargestellt würden, auf die Dauer aber ist nicht zu verantworten, all jene Konfliktzonen zu vermeiden, die für Kinder dadurch entstehen oder als besonders schlimm empfunden werden, weil die Eltern zu den gesellschaftlich Benachteiligten gehören. So können beispielsweise Kinder aus der Mittelschicht, wenn sie in der Großstadt außerhalb der Wohnung etwas unternehmen wollen, genau so übel dran sein wie Arbeiterkinder; aber zum einen leben sie für gewöhnlich in geräumigen Wohnungen und/oder ihre Eltern haben am Wochenende und in den Ferien mehr Möglichkeiten, den kindlichen Spiel- und Erfahrungsraum auszuweiten und wissen im allgemeinen mehr darüber, wie notwendig dies für die Entwicklung der Kinder ist. Oder ein anderes Beispiel: die Erfahrungen, die Kinder mit ihren Vätern am Feierabend machen, sind sehr unterschiedlich; auch wenn der leitende Angestellte von seiner Arbeit ähnlich frustriert sein kann wie ein Akkordarbeiter, so hat er im allgemeinen in seiner langen Schulzeit so viel lernen können, daß er dies nicht ausgerechnet seiner Frau und seinen Kindern heimzahlen sollte. Es kann nicht darum gehen, nach den harmlosen, weil konfliktfreien Fibeln für Mittelschichtkinder, mit Eigenheim, Garten und anderen Wohlstandsattributen, nun weniger harmlose für Arbeiterkinder zu produzieren, in denen, ohne Umschweife und unabhängig vom Bewußtseinsstand der Eltern, Schulanfängern die gesellschaftlichen Widersprüche vor Augen geführt werden. Wichtig für eine inhaltliche Revision der Fibel wird vielmehr sein, die sozialen Erfahrungen der Kinder insgesamt ernst zu nehmen, die negativen nicht einfach auszusparen und darüber hinaus auch Konflikte zu behandeln, die für Kinder materiell benachteiligter Eltern besonders schmerzlich sind. Basis solcher Reformen müssen in jedem Falle die sozialen Erfahrungen der Kinder sein; denn nur so kann die Gefahr von Indoktrination vermieden werden, die auch dort gegeben ist, wo über die Köpfe von Kindern hinweg Aufklärung betrieben wird. Für inhaltliche Reformen dieser Art gibt es in einigen Fibeln bescheidene Ansätze - mehr noch nicht.
Wenn auch heute, nachdem Staat und Kommunen die Führung übernommen haben, der konfessionellen Wohlfahrtspflege nicht mehr die Bedeutung zukommt, wie in früheren Zeiten, so weist sie doch auch gegenwärtig noch höchst beachtenswerte Leistungen auf; sie ergänzt die öffentliche Wohlfahrtspflege in einem Maße, daß sie in dem weitverzweigten Betriebe unserer öffentlichen Einrichtungen gar nicht wegzudenken ist. Das gilt sowohl von der christlichen, wie der jüdischen Charitas. Aber während die Leistungen der ersteren vor allen Augen offen daliegen, blüht die jüdische Wohlfahrtspflege vielfach im Verborgenen. Sie ist außerdem so weit verzweigt und so reich gegliedert, daß sie kaum von wenigen, und auch von den Fachmännern nicht übersehen werden kann; dies um so mehr, als auch die Literatur die Materie bisher sehr stiefmütterlich behandelt hat und, von dem Aufsatze Breslauers: "Die jüdische Wohltätigkeit und Wohlfahrtspflege in Deutschland" (Archiv für Volkswohlfahrt 1908 S. 97) abgesehen, keine zusammenfassende Bearbeitung dieses Gegenstandes vorhanden ist. ES dürfte daher nicht überflüssig sein, einmal im Zusammenhang darzustellen, wie sich die jüdische WohIfahrtspflege historisch entwickelt hat, welchen Umfang sie in der Gegenwart angenommen hat, ihre charakteristischen Merkmale herauszuschälen und darzulegen, welche Bedeutung ihr im Rahmen der allgemeinen Wohlfahrtspflege zukommt. Dies soll Aufgabe dieser Untersuchung sein, bei der in erster Linie die Verhältnisse in Deutschland berücksichtigt werden sollen.
Unsere Betrachtungen lassen erkennen, daß das gotische Maßwerk reichlich Gelegenheit bietet, den mathematischen Unterricht zu beleben. Insbesondere sind Aufgaben über das Apollonische Problem, die sich durch Rechnung und Zeichnung ohne Schwierigkeit lösen lassen, bemerkenswert. Es wurde eine ganz allgemeine Formel als Lösung des Apollonischen Problems aufgestellt, aus der sich die Lösungen für alle in Betracht kommenden Maßverkaufgaben ableiten lassen.
Background: PPARs exhibit anti-inflammatory capacities and are potential modulators of the inflammatory response. We hypothesized that their expression and/or function may be altered in cystic fibrosis (CF), a disorder characterized by an excessive host inflammatory response.
Methods: PPARα, β and γ mRNA levels were measured in peripheral blood cells of CF patients and healthy subjects via RT-PCR. PPARα protein expression and subcellular localization was determined via western blot and immunofluorescence, respectively. The activity of PPARα was analyzed by gel shift assay.
Results: In lymphocytes, the expression of PPARα mRNA, but not of PPARβ, was reduced (-37%; p < 0.002) in CF patients compared with healthy persons and was therefore further analyzed. A similar reduction of PPARα was observed at protein level (-26%; p < 0.05). The transcription factor was mainly expressed in the cytosol of lymphocytes, with low expression in the nucleus. Moreover, DNA binding activity of the transcription factor was 36% less in lymphocytes of patients (p < 0.01). For PPARα and PPARβ mRNA expression in monocytes and neutrophils, no significant differences were observed between CF patients and healthy persons. In all cells, PPARγ mRNA levels were below the detection limit.
Conclusion: Lymphocytes are important regulators of the inflammatory response by releasing cytokines and antibodies. The diminished lymphocytic expression and activity of PPARα may therefore contribute to the inflammatory processes that are observed in CF.
In den Jahren 1992-1999, 2001 und 2002 wurden vom Landesmuseum Kärnten am Zollfeld (KG Maria Saal, pol. Bez. Klagenfurt Land) archäologische Untersuchungen im westlichen Stadtrandbereich des Municipium Claudium Virunum durchgeführt. Dabei wurde der Ostteil zweier Insulae erfasst, die durch den Ost-West verlaufenden Cardo Maximus voneinander getrennt werden. Im Zuge der Ausgrabungen konnte auch eine große Zahl an Wandmalereifragmenten geborgen werden. Der überwiegende Teil kam zwar nur noch in Form von klein- bis kleinstteiligen Fragmenten zu Tage, die Menge der Fundstücke und die Tatsache, dass es sich bei diesen um die ersten stratifizierbaren Wandmalereien der Provinzhauptstadt Noricums handelt, machte eine ausführliche Bearbeitung des Materials notwendig. Im Zuge der Bearbeitung konnten auch zwei Rekonstruktionsversuche erarbeitet werden, die hier im Anschluss vorgestellt werden sollen

Background: The flavin in its FMN and FAD forms is a versatile cofactor that is involved in catalysis of most disparate types of biological reactions. These include redox reactions such as dehydrogenations, activation of dioxygen, electron transfer, bioluminescence, blue light reception, photobiochemistry (as in photolyases), redox signaling etc. Recently, hitherto unrecognized types of biological reactions have been uncovered that do not involve redox shuffles, and might involve the reduced form of the flavin as a catalyst. The present work addresses properties of reduced flavin relevant in this context. Results: N(5)-H exchange reactions of the flavin reduced form and its pH dependence were studied using the 15N-NMR-signals of 15N-enriched, reduced flavin in the pH range from 5 to 12. The chemical shifts of the N(3) and N(5) resonances are not affected to a relevant extent in this pH range. This contrasts with the multiplicity of the N(5)-resonance, which strongly depends on pH. It is a doublet between pH 8.45 and 10.25 that coalesces into a singlet at lower and higher pH values. From the line width of the 15N(5) signal the pH-dependent rate of hydrogen exchange was deduced. The multiplicity of the 15N(5) signal and the proton exchange rates are little dependent on the buffer system used. Conclusion: The exchange rates allow an estimation of the pKa value of N(5)-H deprotonation in reduced flavin to be ≥ 20. This value imposes specific constraints for mechanisms of flavoprotein catalysis based on this process. On the other hand the pK ≈ 4 for N(5)-H protonation (to form N(5)+-H2) would be consistent with a role of N(5)-H as a base.
Background While there is enough convincing evidence in childhood acute lymphoblastic leukemia (ALL), the data on the pre-natal origin in childhood acute myeloid leukemia (AML) are less comprehensive. Our study aimed to screen Guthrie cards (neonatal blood spots) of non-infant childhood AML and ALL patients for the presence of their respective leukemic markers. Methods We analysed Guthrie cards of 12 ALL patients aged 2–6 years using immunoglobulin (Ig) and T-cell receptor (TCR) gene rearrangements (n = 15) and/or intronic breakpoints of TEL/AML1 fusion gene (n = 3). In AML patients (n = 13, age 1–14 years) PML/RARalpha (n = 4), CBFbeta/MYH11 (n = 3), AML1/ETO (n = 2), MLL/AF6 (n = 1), MLL/AF9 (n = 1) and MLL/AF10 (n = 1) fusion genes and/or internal tandem duplication of FLT3 gene (FLT3/ITD) (n = 2) were used as clonotypic markers. Assay sensitivity determined using serial dilutions of patient DNA into the DNA of a healthy donor allowed us to detect the pre-leukemic clone in Guthrie card providing 1–3 positive cells were present in the neonatal blood spot. Results In 3 patients with ALL (25%) we reproducibly detected their leukemic markers (Ig/TCR n = 2; TEL/AML1 n = 1) in the Guthrie card. We did not find patient-specific molecular markers in any patient with AML. Conclusion In the largest cohort examined so far we used identical approach for the backtracking of non-infant childhood ALL and AML. Our data suggest that either the prenatal origin of AML is less frequent or the load of pre-leukemic cells is significantly lower at birth in AML compared to ALL cases.
Background: Experimental animal data show that protection against severe acute respiratory syndrome coronavirus (SARS-CoV) infection with human monoclonal antibodies (mAbs) is feasible. For an effective immune prophylaxis in humans, broad coverage of different strains of SARS-CoV and control of potential neutralization escape variants will be required. Combinations of virus-neutralizing, noncompeting mAbs may have these properties. Methods and Findings: Human mAb CR3014 has been shown to completely prevent lung pathology and abolish pharyngeal shedding of SARS-CoV in infected ferrets. We generated in vitro SARS-CoV variants escaping neutralization by CR3014, which all had a single P462L mutation in the glycoprotein spike (S) of the escape virus. In vitro experiments confirmed that binding of CR3014 to a recombinant S fragment (amino acid residues 318–510) harboring this mutation was abolished. We therefore screened an antibody-phage library derived from blood of a convalescent SARS patient for antibodies complementary to CR3014. A novel mAb, CR3022, was identified that neutralized CR3014 escape viruses, did not compete with CR3014 for binding to recombinant S1 fragments, and bound to S1 fragments derived from the civet cat SARS-CoV-like strain SZ3. No escape variants could be generated with CR3022. The mixture of both mAbs showed neutralization of SARS-CoV in a synergistic fashion by recognizing different epitopes on the receptor-binding domain. Dose reduction indices of 4.5 and 20.5 were observed for CR3014 and CR3022, respectively, at 100% neutralization. Because enhancement of SARS-CoV infection by subneutralizing antibody concentrations is of concern, we show here that anti-SARS-CoV antibodies do not convert the abortive infection of primary human macrophages by SARS-CoV into a productive one. Conclusions: The combination of two noncompeting human mAbs CR3014 and CR3022 potentially controls immune escape and extends the breadth of protection. At the same time, synergy between CR3014 and CR3022 may allow for a lower total antibody dose to be administered for passive immune prophylaxis of SARS-CoV infection.

Die Gestade bedeckt noch jetzt bis zu einer Höhe von 1000 Fuss jenes Gehölz, welches dem ganzen Mittelmeergebiet eigen ist und vorzüglich durch Oelbäume, Myrthen, Judasbäume, Pistacien, Johannisbrod, Oleander, Asarolen, Erdbeerbäume nebst mehreren anderen vertreten wird. Ueber dieser Zone kommt das Reich der immergrünen Stecheichen, die in vielen Sorten allgemein verbreitet sind. Zwischen ihnen eingemengt Galleichen und syrischer Ahorn, mit einem Unterholz von 10—12 Straucharten. Von 3000—4000 Fuss finden sich die Reste der Föhre im Libanon (Pinus Brutia), die inselartig in grossen und mächtigen Stämmen in diesem weiten Gürtel zerstreut zu finden sind. Hier beginnt auch schon die Cypresse an steilen Thallehnen kleine Bestände zu bilden, und die vielgestaltige Zerreiche ist nicht selten. Der höchste Theil der Baumvegetation, meist aus Nadelholz bestehend, bildet den obern Saum von 4000—6000 Fuss über dem Meere. Hier sind vorherrschend die weiten Bestände von wilden Cypressen, Libanoncedern, cilicischen Tannen, Baumwachholdern und einer subalpinischen Eichenart. Ueber die Baumgrenze hinaus reichen noch krüppelnde Baumwachholder, Tragantsträucher und die hingestreckte Kirsche. Die krautartige Vegetation ist in den niederen Regionen zu wenig gekannt, jene in den höheren wird im Verlauf dieser Zeilen bekannt werden.

An alternative theoretical description of axial electron channeling in the multi-GeV region has been developed. We solve a kinetic equation to evaluate an electron distribution function in axially oriented single crystals. Based on the single-string model, the required matrix elements for radiation and scattering by lattice vibrations are calculated employing solutions of the Dirac equation in cylindrical coordinates. Results obtained for 150-GeV electrons propagating along the <110> axis of germanium are in good agreement with experimental observations.
The experimental cold-fission yields for the system 233U(nth, f) are analyzed as function of the effective total excitation energy (TXE). The nuclear level density effect is taken into account at higher TXE, in order to benefit by the lower experimental data uncertainty as well as to avoid the quantitative account of the level densities close to fragment ground states. In this way the odd-even staggering which appears in the yields extrapolated at zero excitation energy by using the level densities, vanishes. We conclude that the cold nuclear fragmentation theory including the dynamical model describes well the experimental data.
The components of the nuclear inertia tensor, functions of the separation distance R and of the radius of the light fragment R2, BRR(R,R2), BRR2(R,R2), and BR2R2(R,R2) are calculated within the Werner-Wheeler approximation, by using the parametrization of two intersected symmetric or asymmetric spheres. Analytical relationships are derived. When projected to a path R2=R2(R), the reduced mass is obtained at the touching point. The two one-dimensional parametrizations with R2=const, and the volume V2=const previously studied, are found to be particular cases of the present more general approach. Illustrations for the cold fission, cluster radioactivity, and α decay of 252Cf are given.
