Arbeiten zur experimentellen Erhärtung der Vorstellungen über die Biogenese von Terpenen aus den Hemiterpenalkoholen Dimethylvinylcarbinol oder Prenol führten zur Auffindung der Dichloressigsäure als eines aliphatischen Polymerisationsbeschleunigers sowohl dieser beiden Alkohole als auch des ihnen zugrunde liegenden Dienkohlenwasserstoffs, des Isoprens. Die Ergebnisse mit dieser einfachen Carbonsäure regen zur Prüfung natürlicher organischer Säuren als Fermentmodelle an.
Bei langdauernder Injektion von Benzylthioharnstoff, der schwer resorbiert wird, entsteht beim Kaninchen keine Struma baśedowificata, sondern nur ein Präbasedow-Zustand. Eine Aktivierung der Schilddrüsen(SD)-Follikel tritt bei kurzfristiger Verfütterung von Benzylthioharnstoff auf.
Der symm. Dibenzylthioharnstoff erzeugt bei täglicher Injektion ein Vorstadium der Struma diffusa parenchymatosa. Seine Resorption ist schlecht.
Die Isothioharnstoffe sind für den Tierversuch wahrscheinlich zu giftig.
Thiouracil, subcutan gespritzt, ruft SD-Vergrößerung hervor und ist bei weitem nicht so toxisch wie Allylthioharnstoff. Die Größen der entarteten Drüsen sind den zugeführten Thiouracilmengen nicht proportional und meines Erachtens von der Konstitution der Versuchstiere weitgehend abhängig.
2-Thio-barbitursäure wirkt bei einer Injektion über drei Monate schwach schilddrüsenaktiv.
Die Natriumsalze des S-Kupfer-N-Allyl-N'-[m-carboxyphenyl]-iso-thioharnstoffes (Cuprion) und des S-Gold-N-Allyl-N'-[m-carboxyphenyl]-iso-thioharnstoffes (Lopion), parenteral gegeben, üben einen nachteiligen Einfluß auf die Schilddrüse aus. Lopion wird besser als Cuprion vertragen. Der N-Allyl-N'-[m-carboxyphenyl]-iso-thioharnstoff wird offenbar schnell vom Körper ausgeschieden, weshalb große Mengen des Grundkörpers erforderlich sind, um eine stärkere SD-Aktivität hervorzurufen.
Sulfanilylthioharnstoff verändert ein wenig die Kaninchen-SD, wenn er über zwei Monate verabfolgt wird.
Mengt man Sulfanilylguanidin dem Futter bei, dann wird die SD in geringem Grade aktiviert. Eine leichte Hypertrophie der Drüse wurde bei subcutaner Behandlung eines Tieres festgestellt.
Möglicherweise führen unter gewissen Bedingungen Benzylthioharnstoff, Sulfanilylguanidin, Cupro-N-Allyl-N'- [m-carboxyphenyl]-iso-thioharnstoff und Thiouracil, in dieser Reihe zunehmend wirksam, zu einer SD-Hypertrophie.
Die Art und Weise, wie die Kaninchen auf die schilddrüsenwirksamen Substanzen reagierten, weist auf die Entwicklung eines anormalen Zustandes der Thyreoidea hin. Unsere Befunde stehen mit der Erklärung der thyreostatischen Wirksamkeit chemischer Verbindungen durch amerikanische Autoren in Einklang.
Die im Trockenweißkraut vorkommende Kropfnoxe wird durch haushaltsübliches Kochen zerstört. Durch Wasserdampfdestillation verliert das Kraut rund 50% seines Gesamtschwefelgehaltes sowie seine strumigene Aktivität. Eine Isolierung der wahrscheinlich S-haltigen Noxe ist bisher nicht gelungen.
Die Noxe des getrockneten Weißkrautes ist im Gegensatz zu der des weißen Senfsamens in heißem Alkohol unlöslich.
Das Senfölglykosid Sinalbin scheint bei Anwesenheit von Myrosinase die Schilddrüse im Sinne einer Struma diffusa parenchymatosa zu beeinflussen.
Die Reizschwelle des in kürzester Zeit basedowifizierend wirkenden Allylthioharnstoffs liegt bei gewöhnlichen Kaninchen zwischen 30 und 40 mg pro 1 kg Körpergewicht, für Angorakaninchen darunter. Allylthioharnstoff verändert die Schilddrüse zunächst im Sinne einer Struma diffusa parenchymatosa, die aber sehr schnell in eine Struma basedowificata übergeht. Tyronorman beeinflußt die Allylthioharnstoffwirkung, wenn es gleichzeitig mit der chemischen Noxe verabfolgt wird; die Drüse verharrt dabei in einem Präbasedowzustand.
Dijodtyrosin ruft unter den gleichen Bedingungen eine ähnliche, aber stärkere Wirkung hervor. Bei kurzfristiger Behandlung einer bereits längere Zeit durch Allylthioharnstoff geschädigten SD kommt es zur Ausbildung einer SD, die histologisch der Jodbasedow-Struma gleicht. Benzylthioharnstoff verändert die SD vorwiegend im Sinne einer Struma diffusa parenchymatosa und steht somit in seiner Wirkung der im Weißkraut vorhandenen Kropfnoxe nahe. Auch für Benzylthioharnstoff scheint die Reizschwelle im gleichen Größenbereich wie beim Allylthioharnstoff zu liegen.
Zur Biochemie der Schilddrüsenfunktion VII : Anzeichen der tierexperimentellen E-Hypervitaminose

Vitamin-E-reiche Fütterung von männlichen jungen Kaninchen führt nach mehreren Wochen zu krankhaften Erscheinungen an der Schilddrüse (SD). Die drei Symptome der entstandenen E-Hypervitaminose in dem innersekretorischen Organ sind: 1. Abnorme Volumen- und Gewichtszunahme, 2. Jodfreiheit bzw. -mangel und 3. histologische Veränderung (gesteigertes Epithelwachstum). Vitamin-E-Gaben neutralisieren nicht die schilddrüsenschädigende Wirkung von Kohlkropfnahrung. Ein täglicher α-Toko-pherol-Zusatz zum Normalfutter verhindert nicht den kropferzeugenden Thiouracil-Effekt an Kaninchen, sondern wirkt im gleichen Sinne.
Es wird ein quantenmechanisches Näherungsverfahren entwickelt, das die Wechselwirkungsenergie der π -Elektronen im Grundzustand aromatischer Kohlenwasserstoffe praktisch ohne Rechenarbeit aus der Strukturformel abzulesen gestattet. Die Resultate des Verfahrens sind im großen und ganzen ebensogut mit der Erfahrung in Übereinstimmung, wie die Resultate des Slater-Paulingschen Verfahrens. Im Falle des Stoffpaares Anthracen-Phenanthren sind sie den Resultaten des Slater-Paulingschen Verfahrens überlegen. Mit Hilfe des Verfahrens lassen sich die Wechselwirkungsenergien der π- Elektronen für ganze Stoffklassen überblicken. Einige Beispiele hierzu werden mitgeteilt.
CH4 und NH4+werden quantenmechanisch als Pseudo-Neon-Atome nach einer Methode behandelt, die der Slaterschen Methode für Atome entspricht. Die Eigenfunktionen nullter Näherung werden aus Eigenfunktionen eines Zentralproblems aufgebaut. Es er-gibt sich sehr gute Übereinstimmung mit den empirischen Daten über -Atomabstände, Suszeptibilitäten und das C-H-Bindungsmoment.
Als Biradikale bezeichnet man Moleküle, bei denen der tiefste Singulettzustand und der tiefste Triplett-zustand praktisch miteinander entartet sind. Bisher liegt nur ein Versuch von Hückel 1 vor. für den Schlenkschen Kohlenwasserstoff die Lage der fraglichen Tenne zueinander theoretisch zu bestimmen. Die Hückelsche Rechnung wurde mit Hilfe des "zweiten" Näherungsverfahrens ausgeführt. Das dem genannten Problem wesentlich besser angepaßte "erste" Näherungsverfahren (nach Slater-Hückel-Pauling) ist bisher nicht angewandt worden. Wir haben für zwei Modellmoleküle, und zwar das klassisch formulierbare Butadien (I) und das klassisch nicht formulierbare, also in gewissem Sinne "metachinoide" Trimethylenmethyl (II) CH"=CH-CH=CH2
Die Resultate des vorstehend 1 veröffentlichten Näherungsverfahrens zur quanten-mechanischen Berechnung der Energie des π-Elektronensystems aromatischer Kohlen-wasserstoffe werden mit denen des Hückel sehen Näherungsverfahrens verglichen. Bei den 18 Molekülen, die zum Vergleich herangezogen werden konnten, ergab sich sehr gute Übereinstimmung.
Resultate der quantenmechanischen Theorie der aromatischen Kohlenwasserstoffe werden mit experimentellen Daten über Anlagerungs-und Umlagerungsreaktionen von aromatischen Systemen verglichen. Zum Vergleich mit der Theorie werden heran-gezogen-, 1. Die Hydrierung aromatischer Kohlenwasserstoffe, 2. die Anlagerung von Maleinsäureanhydrid an aromatische Kohlenwasserstoffe, 3. die Redoxpotentiale der Chinone, 4. die Anthron-Anthranol-Umlagerung. Es zeigt sich, daß die Theorie geeignet ist, den qualitativen Zusammenhang eines großen empirischen Materials befriedigend darzustellen.
Über das dielektrische Verhalten des Diphenyläthers in Mischungen mit unpolaren Flüssigkeiten

Es werden Schwingkreismodelle angegeben, deren Säkulargleichungen mit denen formal identisch sind, die sich bei der Anwendung der Methode der Moleküleigenfunktionen auf das Problem der π -Elektronenzustände in Molekülen ungesättigter und aromatischer Kohlenwasserstoffe ergeben. Damit ergibt sich die Möglichkeit, die quantenmechanischen Säkularprobleme durch Messung der Eigenfrequenzen der Modelle zu bestimmen.
Mit Hilfe der quantenmechanischen Theorie der aromatischen Verbindungen wird gezeigt, daß aus den neuen experimentellen Befunden von Gillet über die Addition von Maleinsäureanhydrid an 9.10-Diphenyl-anthracen und 9-Phenylanthracen auf eine Verdrehung der Phenylringe gegen das Anthracenskelett geschlossen werden kann.
Es wird gezeigt, daß man die langwelligen Absorptionsspektren der magnetisch normalen oktaedrischen Komplexionen mit den Übergangsmetallionen Ti3+, V3+, Cr3+, Mn3+ und Fe3+ verstehen kann, wenn man annimmt, daß die schwachen langwelligen Banden durch Übergänge zwischen den Aufspaltungsprodukten der jeweiligen Grundterme entstehen, wobei die Auf-spaltungen durch das elektrostatische Feld der Liganden zustande kommen.
Die Lichtabsorptionseigenschaften einer Reihe von Lösungen der Komplexionen des dreiwertigen Vanadins vom Typus [V A6]3+, wo A H2O, CH3OH, C2H5OH sowie iso-C4H9OH ist, wurden gemessen. Ferner wurden die Absorptionsspektren von kristallisiertem Ammonium-sowie Caesiumvanadin(III)-alaun aufgenommen.
Intensitätsverhältnisse und spektrale Lage der in allen Spektren auftretenden langwelligen Banden stehen in guter Übereinstimmung mit den Aussagen der Theorie für den Fall eines Zentralions mit zwei d-Elektronen bei Oh-Symmetrie des Komplexfeldes.
Die Ergebnisse quantentheoretischer Modellrechnungen zur Deutung der von WALDEN sowie von STRAUSS und DÜTZMANN beobachteten elektrolytischen Dissoziation organischer Chlorverbindungen in flüssigem SO2 werden mitgeteilt. Anschließend werden die Ergebnisse von Leitfähigkeitsmessungen an Tritylchlorid in SO2-Lösung dargestellt.
Untersuchungen an uv-bestrahlten Schwimmhäuten von Fröschen und Rückenhäuten von Mäusen ergaben, daß mit Hilfe der Nadi - Reaktion (GRÄFF) Oxydationspotentiale an der Bestrahlungsstelle nachgewiesen werden können. Kurzwelliges UV unter λ = 300 mμ hat hierbei die beste Wirkung. Die Stellen stärkster Oxydationskraft liegen bei Schwimmhäuten an den Melanophoren des Corium, bei Mäuserücken-Häuten verteilt im Corium. Bei den Froschhäuten kann zusätzlich bei unphysiologischem pH 4,2 Oxydationswirkung in der Epidermis nachgewiesen werden. Strukturelle Veränderungen an der Schwimmhaut werden polarisationsoptisch deutlich.
UV-mikrospektrophotometrische Messungen der Nucleinsäuren- und Eiweißkörper-Konzentration sowie der Kern- und Nukleolengröße nach Virusinfektion und unspezifischer Reizung der Chorion-Allantoismembran zeigen, daß es in beiden Fällen zu einer gleich starken Stimulierung des nucleinsäuren- und eiweißkörperbildenden Systems der Zelle kommt. Bei der Infektion mit Vaccinevirus auf das Ektoderm setzt die Reaktion der Membranzellen in der Eklipse ein, nach Infektion mit Newcastle-Disease-Virus fällt der Titeranstieg mit der Zellreaktion zeitlich zusammen.
A nucleoprotein of a vitrous consistency was extracted from the gonads of the coalfish (Gadus virens).
The preparation of deoxyribonucleic acid (DNA) from this nucleoprotein and from staphylococci is described. Both of these different kinds of DNA have been mixed with bovine serum albumin or cytochrom c respectively to produce solutions which subsequently were spread onto the Langmuir trough under defined conditions.
After transfer of aliquots from the surface monolayers to carbon support films the preparations were examined with the electron microscope. The micrographs show threads of various lengths, partly stretched, partly folded in loops, consisting of DNA molecules embedded in a protein envelope.
Measurements and calculations of 5900 particles of the complex of Gadus virens-DNA-Albumin, with relatively short threads show a distribution of discontinuous character. If length is plotted against number then it occurs that there are maxima of different lengths of threads. The abscissae of these maxima obey the ratio 1 : 2 : 4 : 8. This holds for longer threads too the maxima of which, however, have smaller ordinate values.
Kristallisierte Lactat-Dehydrogenasen aus Schweineherz, Kaninchenskelettmuskel, Rattenherz und Rattenskelettmuskel wurden mit Trypsin abgebaut und die durch Hochspannungs-Elektrophorese der Verdauungsansätze erhaltenen Peptidmuster nach Anfärbung mit Ninhydrin und diazotierter Sulfanilsäure miteinander verglichen. Es ergaben sich die in den Abb. 1-4 dargestellten Unterschiede.
Durch fluoreszenz-spektrographische Intensitätsmessungen mit einem aus Laboratoriumsmitteln gebauten, einfachen Fluoreszenzspektrometer wurde die Löslichkeit des carcinogenen Kohlenwasserstoffs 3.4-Benzpyren in verdünnten, wäßrigen Lösungen von β-Lactoglobulin und Milchsäure-Dehydro-genase bestimmt. Die molare Lösungsvermittlung der Proteine für 3.4-Benzpyren ist erheblich größer als die des Koffeins. Die Fluoreszenz des 3.4-Benzpyrens wird in den Proteinlösungen nicht durch molekularen Sauerstoff gelöscht. Das beobachtete Spektrum gleicht demjenigen alkoholischer Lösungen von 3.4-Benzpyren ohne Konzentrationslöschung.
Die grundlegenden Untersuchungen von KALLMANN führten in den letzten Jahren meist auf empirischem Wege zur Entdeckung zahlreicher organischer Flüssigkeits-Szintillatoren, die besonders in der Meßtechnik hochenergetischer Teilchen und solcher mit sehr geringer Strahlungsenergie, Eingang gefunden haben. Es soll in dieser Arbeit versucht werden, einen Zusammenhang zu geben zwischen der chemischen Konstitution und den Szintillator-Eigenschaften einiger organischer Verbindungen.
Das Hückelsche zweite Näherungsverfahren wird durch Mitberücksichtigung der höheren Atomzustände erweitert. Dabei ergibt sich die Erklärung für das Scheibesche Phänomen. Der Begriff „theoretische Sonderenergie (Resonanzenergie)“ wird richtiggestellt. Die bekannten Schwierigkeiten, die sich beim Vergleich spektroskopischer und kalorischer Energiewerte im Rahmen der Einelektronentheorie der π-Elektronensysteme bisher immer ergeben haben, verschwinden.
Es wird versucht, Beweise für die indirekte Strahlenwirkung zu erbringen und die einzelnen Reaktionsschritte, die bei Röntgenstrahlen-Wirkung auf wäßrige Lysinlösungen ablaufen, zu erkennen. Zur Untersuchung der durch HO2 oder OH-Radikale ausgelösten Wirkungen wurden 2 Versuchsreihen durchgeführt:
1. Bestrahlung bei Anwesenheit von Sauerstoff,
2. Bestrahlung bei Sauerstoffmangel.
Außerdem wurde der Einfluß der Dosis, Dosisleistung, Temperatur und verschiedener Substrat-Konzentrationen untersucht.
Schließlich wurde versucht, die experimentell gewonnenen Ergebnisse mit Hilfe von Radikalwirkungen, die sich auf
a) Decarboxylierungen,
b) Desaminierungen,
c) und Veränderungen des Kohlenstoff-Gerüstes erstrecken, zu deuten.
Es wurden mehrere unkonjugierte 2.4-Diaminopteridine erstmals synthetisiert. Die Wachstumshemmung verschiedener Mikroorganismen durch 2.4-Diamino-6-[1.2-dihydroxypropyl-(ʟ-erythro)] pteridin (Aminobiopterin) und anderer unkonjugierter 2.4-Diamino-pteridine läßt sich nur mit Folsäure oder Thymin, nicht dagegen mit Biopterin aufheben.
The enzyme 20-β-Hydroxy-steroid-dehydrogenase obtained from the culture of Streptomyces hydrogenans and dissolved in 0.05 M Tris puffer, pH 7.3, has been investigated by means of a ultracentrifuge at 20 °C. The sedimentation- as well as the diffusion-coefficients obtained from various solutions at different concentrations were extrapolated to the concentration c = 0. The resulting zero-value for the sedimentation coefficient is s0 = 6.64 s and for the diffusion coefficient is D0 = 5.51 × 10-7 cm2/sec. Supposing the partial specific volume of the enzyme under consideration analogously to other similar proteins is V+=0.749 ml/g, the molecular weight has been estimated as M = 118 400.
Wir ermittelten die Geschwindigkeitskonstanten sowie die Arrhenius sehen Aktivierungsenergien und Aktionskonstanten der alkalischen Hydrolyse einer Reihe von Di- und Tripeptiden. Die zur Gewinnung der kinetischen Daten führende quantitative Analyse der Hydrolysate gelang, indem die Peptide vor ihrer Hydrolyse geeignet mit 14C markiert, die Hydrolysate mit Hilfe der Papierchromatographie bzw. der Zonenelektrophorese getrennt und die isolierten Hydrolyseprodukte auf Grund ihrer Radioaktivität quantitativ bestimmt wurden.
Der zeitliche Verlauf der alkalischen Hydrolyse von Di- und Tripeptiden wird durch ein Zeitgesetz 1. Ordnung bezüglich des Peptids und bezüglich der Hydroxylionen dargestellt. Die Abstufungen der Aktivierungsenergien, die den einzelnen Peptidbindungen in Di- und Tripeptiden zugeordnet sind, lassen sich durch die Annahme deuten, daß die Hydrolyse nach dem BAC 2-Mechanismus verläuft. Die Aktionskonstanten wachsen, wenn auch nicht regelmäßig, mit den Aktivierungsenergien.
Mit Alkalimetallen reduzierte Lösungen von 2,2′-Dipyridyl liefern ein ESR-Signal, dessen Hyperfeinstruktur vom Reduktionsmittel abhängt. Bei Verwendung von Lithium beobachtet man das Spektrum des Ionenpaares (Li+, dip-), mit Kalium dagegen erhält man im wesentlichen das ESR-Spektrum des Anionradikals dip-. Seine Hyperfeinstruktur wird quantitativ interpretiert. Abweichungen zwischen den daraus entnommenen experimentellen Spindichten und theoretischen Spindichten, die mittels der HÜCKELschen Theorie erhalten wurden, deuten auf Grenzen in der Anwendbarkeit dieser Theorie.

Beiträge zur Erweiterung der Hückel'schen Theorie der Pi-Elektronensysteme Hückel entwickelte 1931 ein Verfahren zur quantenmechanischen Berechnung zahlreicher Eigenschaften von zr-Elektronensystemen. Obwohl die Theorie halbtheoretisch ist, hat sie einen wesentlichen Beitrag zum Verständnis solcher Eigenschaften von ungesättigten und aromatischen Verbindungen geliefert, die dem Grundzustand der Moleküle zugeschrieben werden können, so z. B. der Resonanzstabilität und der Reaktivität. Doch ist sie nur in der Lage, die Eigenschaften dieser Verbindungen richtig zu beschreiben, die durch angeregte Zustände der Moleküle mitbestimmt werden, wie z. B. die Spektren, wenn man für das Resononanzintegral einen entsprechenden Wert verwendet, der von dem für den Grundzustand benutzten bedeutend abweichen kan. Im ersten Abschnitt der vorliegenden Arbeit wird die Hückel'schen Theorie der Pi-Elektronensysteme diskutiert. Einen nur annähernd vollständigen Überblick über die Beiträge zu dieser Theorie zu geben, würde den Rahmen der Arbeit überschreiten. Daher wurde sich im Wesentlichen darauf beschänkt, die Grundgedanken und die Näherungen des Verfahrens klar darzustellen und kritisch zu betrachten, sowie die Anwendungsmöglicheiten zu beschreiben. Hartmann hat 1960 eine Erweiterung der Hückel 'schen Theorie vorgeschlagen, die auch solche Eigenschaften ungesättigter und aromatischer Verbindunsen richtig erfaßt, die aur der Beteiligung angeregter Zustände beruhen, wie am Beispiel der Spektren gezeigt wurde. Im zweiten Teil dieser Arbeit wird die Hückel-Hartmann'sche Theorie der Pi-Elektronensysteme dargestellt und die Ergebnisse mit denen der Hückel'schen Theorie verglichen. Hartmann selbst hat die Erweiterung nur für gleichatomige Pi-Elektronensysteme und unter zahlreichen Vernachlässigunen durchgeführt. Dieser Umstand und die wenigen zu der Theorie erst vorliegenden Arbeiten ließen es wünschensert erscheinen, die Hückel-Hartmannsche Theorie der Pi-Elektronensysteme systematisch und in möglichst allgemeiner Form zu diskutieren. Im dritten Abschnitt der Arbeit wird daher eine Generalisierung der Hückel-Hartmannt'schen Theorie durchgeführt. Mit Hilfe des entwickelten Verfahrens wird anschließend der Einfluß der Überlappungen auf die Ergebnisse untersucht und eine Erweiterung der Hückel-Hartmann'schen Theorie auf heteroatomare Pi-Elektronensysteme angegeben.
In order to determine the influence of OH and O2H-radicals on proteins, bovine serum albumin (BSA) in aqueous solution was treated with Fenton’s reagent [Fe(II)SO4+EDTA+H2O2] and with ultraviolet light (λ > 2800 Å) in the presence of H2O2. The action of free radicals produced in this way did not change the properties of the native protein with respect to the sedimentation in the ultracentrifuge or optical rotatory dispersion and electrophoresis under normal conditions. Ampèrometric titration indicated partial oxidation of SH-groups and of 3—5 SS-groups which are not reducible by NaBH4.
Heat aggregation investigated by means of light-scattering was suppressed at pH 7.5 and strongly accelerated at pH 4.6 (range of coagulation), the latter being a result of increased entropy of activation of coagulation velocity.
The difference spectrum against native BSA had positive values of Δε and two maxima at 2480 and 2950 Å.
Ultracentrifugation at room temperature in phosphate buffer (pH 7.3, μ=0.18) furnishes a molecular weight of 63 300. In a solution of 8 M urea and borate buffer (pH 9, μ=0.05) fragments with molecular weights between 25 000 and 37 000 were observed while in phosphate buffer (pH 7.3, without urea) at temperatures higher than 46 °C an anomalous behaviour of the concentration gradient indicated an effect which possibly depends on a dissociation equilibrium.
As a consequence oxygen radicals seem to attack not only SH- and SS-groups but at least one covalent bond of the peptide chain. Some experiments of heat aggregation with BSA treated with γ-rays (60Co) gave the same results as BSA treated with Fenton’s reagent or UV-light+H2O2.
Für eine Gruppe von Di- und Tripeptiden wurden die Geschwindigkeitskonstanten sowie die Arrhenius schen Aktivierungsenergien und Aktionskonstanten der sauren Hydrolyse der einzelnen Peptidbindungen gemessen. Zeitgesetz und Abstufungen der Aktivierungsenergien lassen sich nur unter bestimmten Annahmen über die Solvatation der Reaktionsteilnehmer auf der Grundlage des Ingold- Benderschen Mechanismus interpretieren. Aktionskonstante und Aktivierungsenergien weisen einen symbaten Gang auf.
The results of spectroscopic and paramagnetic resonance investigations show that the previously prepared substance with the empirical composition [VIdip3] J · ½ py is most presumably a mixed crystal of two similar compounds of bivalent and zero-valent vanadium. The ion [VIdip3] + exists only in solution and gives no paramagnetic resonance signal, which is understood on account of a low-lying singlet ground state.
Die Winkelabhängigkeit des ZEEMAN-Effektes der Kernquadrupolresonanzen (35Cl) eines Einkristalls von ortho-Dichlorbenzol wurde bei — 35°C vermessen. Das Kristallsystem ist monoklin. Die z-Achsen der Feldgradiententensoren im Molekül bilden einen Winkel von (64,7 ±0,5)°. Der Asymmetrieparameter des Feldgradienten hat einen Wert von 0,100 ± 0,01. Die Winkelmeßeinrichtung wird beschrieben.
Nach Kultivierung von Enterococcus Stei mit 14C-markiertem 5-Chlor-, 5-Brom- oder 5-Jod-Uracil wurde aus den Zellen die DNS isoliert und hoch gereinigt. Durch UV-Bestrahlung dieser DNS in wäßriger Lösung werden die eingebauten 5-Halogen-Uracile photochemisch verändert. Beim Abbau dieser bestrahlten DNS findet man neben geringen Mengen nicht-identifizierter Photoprodukte als überwiegendes Strahlenprodukt nach Hydrolyse mit Perchlorsäure Uracil und nach fermentativem Abbau Uracildesoxyribosid. Die Dehalogenierung von BU und JU in der DNS verläuft in Abhängigkeit von der Bestrahlungsstärke etwa gleich schnell, während CU sehr viel langsamer dehalogeniert wird.
Die photochemische Dehalogenierung des BU erfolgt in der nativen DNS am leichtesten, weniger gut in der Hitze-denaturierten DNS und nur in geringem Maße in der Apurinsäure.
1- (2̸.3′.4′-O-Triacetyl-1.β-D-glucopyranosyl) 3-carboxamido-pyridiniumchlorid wird aus α-1-Chlor-2.3.4-O-triacetyl-glucopyranose und Nikotinamid hergestellt. Die freie Hydroxylgruppe in 6-Stellung der Glucose wird mit Phosphoroxychlorid verestert. Durch Acylwanderung entsteht außerdem ein isomeres Produkt. Die Acetylgruppen lassen sich sauer verseifen. Durch Kondensation mit Adenosinmonophosphat erhält man ein Gemisch beider Nikotinamidglucosid-Adenin-Dinucleotid-Isomerer. Die Verbindungen sind trotz hoher Affinität zu nucleophilen Agentien auf Grund der sterischen Konfiguration enzymatisch inaktiv.
A study on the effect of UV-irradiated polyuridylic acid on the incorporation of phenylalanine into the polypeptide precipitable through trichloroacetic acid, in a cell-free system from E. coli was made. Attempts were made to reactivate the UV-inactivated polyuridylic acid through hydrogen peroxide, uranyl acetate and visible light. We could show that polyuridylic acid irradiated at a dose of 1.2 ×105 ergs/mm2 could be completely reactivated, while the one irradiated at a higher dose of 2.4 ×105 ergs/mm2 could not be completely reactivated under the conditions of our experiment. We have studied the effects of hydrogen peroxide and uranyl acetate on UV-irradiated polyuridylic acid chemically as well. Our results altogether show that the photoreactivating effect of uranyl acetate and hydrogen peroxide is due to their ability to split the uracil dimers formed during UV-irradiation.
The carcinogenic hydrocarbon 3.4-benzopyrene is soluble in aqueous solutions of different proteins. The solubilities are easily determined by the fluorimetric method. The fluorescence o. the hydrocarbon in the protein solutions is not quenched by molecular oxygen. Nevertheless only in presence of air (oxygen) an irreversible decrease of the fluorescence intensity occurs under irradiation with UV-light of wavelength 366 mμ, which is considerably faster than under nitrogen or in solutions of the hydrocarbon in ethanol or aqueous caffeine.
In the systems investigad, a correlation was found between the half-life period of the reaction and the SH-group activities. The participation of protein-SH-Groups in the 3.4-benzopyrene photoreaction is demonstrated by ampèrometric Ag⊕-titrations.
The influence of protein denaturation and inhibiting additives on the photoreaction are investigated by the fluorimetric method.
Irradiation- and oxygen-dependence of the reaction are analogous to the observations of photodynamic action and skin cancer induction by 3.4-benzopyrene.
By 366 mµ irradiation of β-lactoglobuline solutions containing 3.4-benzopyrene the heatdenaturation characteristics of the protein are changed. The same changes are produced without 3.4-benzopyrene by UV-light of the wavelength 280 mµ. Treatment of the β-lactoglobuline solutions with an amount of cigarette smoke, which certainly does not contain 3.4-benzopyrene in sufficient concentration, acts in the same direction.
Along with the changes in the protein properties the typical fluorescence of 3.4-benzopyrene vanishes. The hydrocarbon does not act as a catalyst in photodynamic action, but is chemically altered as well as the protein, at least in the system under investigation.
The kinetic data of the hydrolysis of some serine peptides in diluted hydrochloric acid and in pure water and of the rearrangement of O-glycyl-DL-serine to glycyl-DL-serine were determined.
The hydrolysis of glycyl-DL-serine and DL-alanyl-DL-serine proceeds surprisingly rapidly in pure water as compared with the hydrolysis of those peptides in 0.5 N hydrochloric acid as well as the hydrolysis of glycyl-DL-alanine in purely aqueous solution. The O → N migration of the glycyl residue in O-glycyl-DL-serine which probably is an intermediate in the cleavage of glycyl-DL-serine in purely aqueous solution represents a three center reaction in which the nucleophilic attack on the O-peptide and peptide bond, respectively, involves a free basic amino group. The analogy between the serine peptide interconversion and the hydrolysis catalyzed by certain proteolytic enzymes is referred to.
Under the conditions of freeze drying are formed in hydrochloric acid solutions of DL-alanyl-DL-serine O-peptide and depsipeptide.
Diluted aqueous solutions of some proteins (bovine serum albumin, β-Lactoglobubin, Peroxidase) show weak phosphorescence lasting over several minutes after they have been irradiated with light in the range 3500-4200 A. Addition of Eosin after the irradiation amplifies in some cases the intensity of luminescence to a value of about hundred. If Eosin is present at the irradiation process the excitation to phosphorescence is possible with light of the wavelength 5460 A.
After denaturation processes which destroy the configuration of proteins (Urea, Guanidine-HCI. detergents, heat at higher pH) the ability of phosphorescence disappears altogether; likewise after blocking the SH-groups by benzochinone or a total oxidation or reduction of the SS-groups which causes an complete unfolding of the peptide chain.
In solutions of bovine serum-albumin irradiated with 3650 Å at room temperature and afterwards frozen to -178°C no radicals could be observed by measurements of electron-spin-resonance but they were detectable if the irradiation took place in the presence of H2O2.
The reactions Xanthinoxidase-Xanthine-O2, Peroxidase-H2O2 and bovine serum-albumin-H2O2-Fe (II) EDTA are accompanied by chemiluminescence. By comparison with the behaviour of oxidised serum-albumin it could be shown that the chemical reaction produces an excited state of the native protein.
The observations lead to the conclusion that the weak phosphorescence of long duration originates from a triplet-state which is sufficiently populated only as the consequence of cooperative phenomena attending the undisturbed α-Helix-structure of the protein.
In order to determine the intermolecular forces in the process of the heat aggregation of globular proteins in solution, selected proteins with different amounts of disulfide- and thiolgroups were investigated by specific inhibition experiments and by degradation analysis, using lightscattering and ultracentrifugation methods.
In accordance with the mechanism of the heat aggregation, which in general (SH —SS-proteins) may be characterized as a coupled coagulation- and exchange-reaction, auxiliary valences and covalent bonds take part in the aggregation process.
Besides the pʜ-range of lanthionine-formation, the coagulation-mechanism by weak intermolecular forces exceeds the covalent type of aggregation.
If only one of the sulphur functional groups is present in the protein molecules the aggregation is merely the result of the coagulation-mechanism, i. e. the degradation by urea, guanidine·HCl, variation of pʜ etc. leads back to the monomer.
In the case of SH —SS-proteins the degradation rate depends on the temperature and duration of aggregation: In the range of predenaturation and under isoelectric conditions the native monomer is restored while increasing net charge leads more and more to covalently bound aggregates which are due to disulfide- and lanthionine-groups. High alkalinity promotes the formation of lanthionine.
Regarding the weak intermolecular bonds the application of specific criteria in degradation and inhibition experiments proves that Η-bonds and hydrophobic interactions participate in the aggregation process while ion pair bonds may be excluded. The hydrophobic interactions do not become apparent, until partial denaturation of the aggregating protein takes place.
The proportion of the total aggregation at extreme pʜ-values which is produced by the coagulation mechanism may be explained in a tentative way by assuming specific electrostatic short range interactions between the partially dehydrated molecules, leading to fibrillar associates.
Steroid initiated enzyme induction (Δ5-Ketosteroid-Isomerase, 3α-Hydroxysteroid-Dehydrogenase, and 3β.17β-Hydroxysteroid-Dehydrogenase) in Pseudomonas testosteroni was investigated with respect to the kinetics of induction, operon control of the induced enzymes, and the relative strengths of various inducers. The induction process was followed indirectly by selective inhibition of different stages in the protein synthetic pathway. Comparisons between bacterial and mammalian steroid induction are discussed.
Glycol causes a denaturation of the DNA double helix structure in solution. As could be shown earlier, heat denaturation of DNA leads to an increased dimerization of thymine following uvirradiation. In contrast to this, thymine dimer is not increased - but is even slightly decreased - when DNA is uv-irradiated in the presence of glycol. These results are discussed with regard to the distortion of the hydration layer and the hydrophobic stacking of bases, as influenced by glycol.
Investigations were made of energy-transfer in liquid scintillators under UV- and β-excitation. The influences of n-hexane and methanol as diluting solvents on the scintillation process in p- terphenyl-toluene were measured. Differences of energy transfer under β - and UV-irradiation are discussed. Radiationless energy-transfer occurs over distances from 15 to 32 Angström units. Quenching effects of phenol on p-terphenyl, 2,5-diphenyl-oxazole, and 2,4 (or 5) -diphenyl-imidazole were studied. Oxygen-quenching in liquid scintillators containing hydroxy-benzenes was investigated. The results are in correspondence with the KALLMANN-mechanism of radiationless energy-transfer. — Further investigations were made of the connection between the chemical structure of aryl-imidazoles and their scintillation properties. Scintillation properties depend on spectral data. The results aprove HELLERS postulates, concerning structural requirements for good scintillation properties of organic liquid scintillators.
In wäßrig gepufferten Mitochondrien-Suspensionen konnte ein selektives Lösungsvermögen für carcinogene Kohlenwasserstoffe mit Hilfe fluoreszenz-spektroskopischer Messungen festgestellt werden. Gleichzeitig wird eine starke Verminderung der Atmung der Mitochondrien durch die Einwirkung der gelösten, polycyclischen Aromaten beobachtet. Nicht carcinogene Kohlenwasserstoffe werden weder gelöst, noch zeigen sie einen Einfluß auf die Atmung. Sehr schwer lösliche Carcinogene und schwächer wirksame Kohlenwasserstoffe werden in den angewandten Versuchszeiten weder gelöst, noch vermindern sie die Geschwindigkeit des Sauerstoffverbrauchs.
Mitochondrien älterer, oder mit Vitamin B2-Mangel behafteter Tiere zeigen erhöhte Depression der Atmung bei Zugabe von DMBA bzw. 3.4-Benzpyren.
Es wird versucht, diese Effekte einer reinen Promotor-Aktivität der polycyclischen, aromatischen Kohlenwasserstoffe zuzuschreiben, die an den Strukturelementen der Zellatmung (Mitochondrien) einsetzt.
Interactions of eosin with three different substrates, β-lactoglobuline, bovine serum albumin and cysteine, in aqueous solutions of pH 7 under illumination with light of wavelengths 5200—5400 Å are investigated by changes in absorption spectrum characteristics, SH-group activities and phosphorescence intensities.
Only with bovine serum albumin the major part of protein conversion, as shown by spectral changes and diminution of SH-groups due to eosin-sensitized photo-oxidation. In β-lactoglobuline an oxidizing photoreaction occurs, by which eosin is vanishing to the same degree as the protein shows loss of SH-groups and spectral alterations indicating attack on aromatic amino acid residues. There is no red shift of the eosin absorption band at 5170 Å as is observed in solutions of bovine serum albumin, where the intensity of phosphorscence is about 100 fold compared with the intensity obtained by solutions of β-lactoglobulin.
The aerobic eosin photoreaction in solutions of β-lactoglobulin is faster than aerobic photobleaching of the dye. Still faster is its bleaching photoreaction with cysteine, which is nearly independent of oxygen.
Wäßrige Sephadex-Gele nehmen Vielfache der in Wasser gelösten Anteile polycyclischer, aromatischer Kohlenwasserstoffe auf, wenn diese als Suspension vorliegen. Beobachtbar ist dieser Effekt an der Erhöhung der Fluoreszenzintensität des gelösten Anteils der Aromaten bei Zugabe von Sephadex-Körnern und anschließender kräftiger Rührung. Verwertbar ist dieses Phänomen zumindest für die Ermittlung der Lage der Emissionsbanden der Kohlenwasserstoffe in Wasser, denn es bestehen nur geringe Unterschiede zwischen den Fluoreszenzsprektren in Sephadex und Wasser. Weiterhin ist beachtlich, daß die carcinogenen Kohlenwasserstoffe nach diesen ersten orientierenden Versuchen, zu einer Gruppe gehören, die eine geringe Diffusionsgeschwindigkeit in das Sephadex-Gel, bei gleichzeitigem Erreichen relativ hoher Endkonzentrationen, besitzen. Der Quotient aus beiden Größen wird daher für die Carcinogene maximal. Nach Desoxycholsäure, Detergentien, Purinen, Desoxynucleinsäuren, Adenosintriphosphat 24 und Proteinen wurde damit gefunden, daß auch Polysaccharid-artige Systeme in der Lage sind, Lösungs-vermittelnd auf Kohlenwasserstoffe in wäßriger Phase zu wirken.
Es verspricht daher, interessant zu werden, an Sephadex, als primitivem Modell von Zellbestandteilen, in der Zukunft standartisierte Kohlenwasserstoff-Suspensionen in dieser Weise zu untersuchen und gleiche Experimente mit stärker hydrophobiertem Gel durchzuführen.
Die Synthese des Coenzymmodells Flavin-benzimidazol-dinucleotid * gelang durch Kondensation von Benzimidazolribotid-imidazolid 1 oder Benzimidazolribotid-guanidiniumamidat 2 mit Flavinmononucleotid. Das Coenzymmodell war enzymatisch nicht aktiv und bildete keinen Enzym-Coenzym-Komplex. Im Absorptionsspektrum konnte eine Extinktionszunahme nach der Spaltung der Pyrophosphatbrücke nur im Bereich von 260 mμ beobachtet werden. Das Molekül liegt daher vermutlich in einer gefalteten Form vor. Ein Komplex zwischen Flavin- und Benzimidazolteil konnte nicht nachgewiesen werden. Eine Fluoreszenzunterdrückung, die im FAD durch die Komplexbildung zwischen Flavin- und Adeninteil bedingt wird, wurde im FBD-Coenzymmodell nicht beobachtet.
Mitochondrien aus Rattenleber (RL) und Rinderherzmuskel (BH) erzeugen bei Behandlung mit O2 eine schwache Chemilumineszenz in dem Spektralbereich zwischen 400 und 650 mµ, deren Intensität bei RL-Mitochondrien durch vorheriges Einfrieren und Auftauen wie durch Ultraschallbehandlung größer wird. Bei beiden Arten verursacht Zusatz von Acridinorange eine wesentliche Verstärkung der Lumineszenz, gleichzeitig wird die O2-Aufnahme gehemmt. RL-Mitochondrien erzeugen unter diesen Bedingungen eine kurzzeitige, BH-Mitochondrien eine langsam ansteigende langandauernde Strahlung; das gleiche Verhalten zeigen aus BH-Mitochondrien gewonnene „electrontransfer-particles“ (ETP). Der zeitliche Ablauf und die Beeinflußbarkeit durch Effektoren der Atmungskette ist andersartig als bei der von VLADIMIROV gefundenen Chemilumineszenz von RL-Mitochondrien. Als Träger der Lumineszenz wird angeregter O2 diskutiert.
This paper reports about the fine structure in the O—K-spectra of the oxides BeO, MgO, CaO, SrO, BaO, Sc2O5, Y2O3, La2O3, Sm2O3, Yb2O3, NiO and ZnO. The spectra show the satellite lines α3, α4, α5, α6 on the short wavelength side of the main line α1,2 and a shoulder β′ on its long wavelength side. The wavelengths of all lines depend on the nature of the oxide. For the positions of the lines Kα1.2 in the spectra no systematic relation to other data of the oxides is observed. On the other hand the distance of the a4-satellite from the α1,2-line decreases with increasing electronegativity of the metal atom in the oxide. This distance can be used as a measure for the ionic character of the metal-oxygen bond in these compounds.
The rotational spectrum of several isotopic species of HSiCl3 and CH3SiCl3 was studied in the region from 8 to 40 Gc. From the derived rotational constants the following structural parameters were obtained using KRAITCHMAN'S equations: dSi-H= (1.4655±0.0002) A, dSi-Cl= (2.0118±0.0009) A, ∢ Cl—Si—Cl= (110,60±0.25)°. Furthermore the constants for centrifugal distortion DJ= (1.2 ± 0.4) kc for HSiCl3 and DJ= (0.19 ± 0.04) kc for CH3SiCl3, for the quadrupole coupling e Q Vzz= + 12.8 Mc and the dipole moment μ= (0.86 ± 0.01) D for HSiCl3 and μ= (1.91 ± 0.01) D for CH3SiCl3 were determined. The interaction of the overall-rotation with the internal rotation is discussed for CH3SiCl3, and the hindering barrier is estimated to be less than 200 cm-1.
Die „Selbstenergien“ Gn in 1 wurden durch näherungsweise Auswertung einer größeren Klasse von Selbstenergiediagrammen approximativ berechnet. Das Gleichungssystem (3.3) in 1 für die renormierten Semiinvarianten wurde umgeformt und durch zusätzliche Näherungsannahmen vereinfacht. Durch Näherungsansätze für die Semiinvarianten M2, M3,... konnten einfache Gleichungen für die Magnetisierung M1 hergeleitet werden. Diese Gleichungen wurden numerisch gelöst. Auf der Grundlage der Beziehungen (3.5) und (4.8) in 1 wurden ferner die innere Energie, die freie Energie und die Atomwärme des zweidimensionalen Ising-Ferromagneten sowie die Druck-Dichte-Isothermen des zweidimensionalen Gittergases numerisch ausgerechnet.
Die FOURIER-Nullkomponente der Bindungsmatrix Gij G(0) in 1 Beziehung (5.16) wurde durch Summation spezieller Diagramme in der sogen. „Kettenapproximation“ und der „Wassermelonenapproximation“ näherungsweise berechnet. Auf der Basis von (5.16) in 1 wurde in der Kettenapproximation und der Wassermelonenapproximation die magnetische Suszeptibilität und die Magnetisierung des dreidimensionalen ISING-Modells bestimmt. In der Kettenapproximation wurden ferner die freie Energie, die innere Energie und die Atomwärme des dreidimensionalen ISING-Ferromagneten sowie die Druck-Dichte-Isothermen des dreidimensionalen Gittergases ausgerechnet.
Gegenstand der vorliegenden Untersuchung ist das hydrolytische Verhalten von Glycyl-seryl-alanin beim Erhitzen in einer rein wäßrigen Lösung bei einem pH-Wert in der Nähe des isoelektrischen Punktes. Die Reaktionsmöglichkeiten des Tripeptids und aller Folgeprodukte, die Alanin enthalten, wurden qualitativ aufgeklärt und die kinetischen Daten der einzelnen Reaktionen durch quantitative Hydrolyseversuche an den mit dem Kohlenstoffisotop 14C geeignet radioaktiv markierten Verbindungen Glycyl-seryl-alanin, Seryl-alanin, Alanyl-serin und Seryl-alanyl-diketopiperazin bei mehreren Temperaturen zwischen 55 und 95 °C bestimmt. Die Trennung der Hydrolysatproben erfolgte mit Hilfe der Papierchromatographie bzw. der Papierelektrophorese; für die Konzentrationsbestimmung der radioaktiven Substanzen wurde ein Verfahren ausgearbeitet, das eine Radioaktivitätsmessung mit hoher reproduzierbarer Ausbeute direkt auf den zur Trennung benutzten Filterpapieren nach der Flüssigkeitsszintillations-Methode ermöglicht.
Alle beobachteten Reaktionen verlaufen unter den Versuchsbedingungen nach einem Zeitgesetz 1. Ordnung. Die Konzentrations-Zeit-Funktionen der einzelnen Reaktionsteilnehmer wurden durch Auflösung der simultanen linearen Differentialgleichungen 1. Ordnung bestimmt und zur Berechnung der Reaktionsgeschwindigkeits-Konstanten nach einem iterativen Ausgleichsverfahren herangezogen.
In der Diskussion wird versucht, die beobachteten thermodynamischen Daten in ihrer Größenordnung aus anderen Eigenschaften und aus der Molekularstruktur der einzelnen Verbindungen abzuleiten und durch bestimmte Reaktionsmechanismen zu interpretieren.
Induction of the enzyme Δ5-3-ketosteroid isomerase in Pseudomonas testosteroni was found to be strongly inhibited by reserpine and by the alkaloids of Vinca and Ergot. Morphine, colchicine and papaverine caused weaker inhibition whilst a series of other alkaloids were almost ineffective. Ergot alkaloids were inhibitory towards all steroids tested, androgens, oestrogens and progestagens, and a similar effect was shown with the other inducible enzymes, 3α- and 3β.17β-hydroxysteroid-dehydrogenase.
Experiments with cell-free protein synthesis indicate that reserpine inhibits the induction of messenger RNA.
The non-specific inhibition of the poly U directed polymerisation of phenylalanine through polyanions was studied. This inhibition was found to be in order as follows: dextransulfate, polyethylensulfate, heparine, ribosomal RNA and alginate. It was found that poly A, poly AP and poly AG cause a specific inhibition of the poly U directed synthesis of polyphenylalanine. Poly AG and poly AP, but not poly A were found to inhibit the poly C directed polymerisation of proline as well. The mechanism of these two types of inhibition caused by polyanions has been discussed.
The effect of NNMG on the template activities of different polynucleotides (polyuridylic acid, polycytidylic acid, polyadenylic acid and copolymer of adenylic and guanylic acid 5,5:1) and t-RNS was studied. The maximum inhibition of the messenger activity was found for poly-C, followed by poly-Α and poly-U. The acceptor activity of t-RNA was found to be inhibited by NNMG: maximum for proline, followed by serine, leucine, phenylalanine and lysine. The mechanism of these inhibitions was studied using NNMG radioactively labelled on the methyl group. Different amounts of radioactivity were found in the various polynucleotides and t-RNS.
It is possible to determine the anomeric configuration of nucleosides by a simple, spectrophotometric assay, using the nucleoside phosphorylase activity of cell-free extracts from E. coli. β-nucleo-sides are split, a-anomers remain unchanged. For a single estimation 20-40 µg of nucleoside are required. 6-Azauracil- and 8-azaguanine-β-ᴅ-riboside and some nucleoside phosphates are resistant, a fact, which is of interest in view of the specificity of nucleoside phosphorylases.
The ribonucleic acid of reovirus was extracted with 2 M sodium perchlorate solution and spread by the protein monolayer technique. Areas of the monolayer were transferred to support films, rotary shadowed, and observed in the electron microscope. Filaments of RNA obtained by extraction prior to spreading were similar in appearance and in distribution of contour lengths (0.2 to 1.2 μ) to those obtained by phenol extraction of the virus. Most of the filaments resulting from extraction of the virus suspension during spreading on a sodium perchlorate solution, however, were longer than 1 μ. The lengths of the longest filaments exceeded the 5 μ length predicted from chemical data for one single piece of complementary-stranded RNA in the reovirus particle.
The short filaments, 1.2 μ and less in length, fell into a tri-modal pattern of length distribution with peaks at 0.35 μ, 0.60 μ and 1.10 μ. These shorter lengths probably resulted from breakage of the intact RNA during the extraction procedures. The consistently observed pattern of length distribution suggests that they represent relatively stable subunits of the molecule.
Sodium perchlorate extracted reovirus RNA was thermally denatured in formaldehyde prior to spreading by the protein monolayer technique. Length distributions and relative numbers of filaments in the peaks of the tri-modal distribution pattern were similar to those found for unheated material when extracted prior to spreading. This similarity indicates that heating subsequent to extraction produced no further filament breakage. The thin, kinky appearance of the heated filaments, and the appearance of congruent pairs, indicated that heating had separated the strands of the complementary-stranded RNA subunits.
