The experience of pain is mediated by a specialized sensory system, the nociceptive system. There is considerable evidence that the cGMP/cGMP kinase I (cGKI) signaling pathway modulates the nociceptive processing within the spinal cord. However, downstream targets of cGKI in this context have not been identified to date. In this study we investigated whether cysteine-rich protein 2 (CRP2) is a downstream effector of cGKI in the spinal cord and is involved in nociceptive processing. Immunohistochemistry of the mouse spinal cord revealed that CRP2 is expressed in superficial laminae of the dorsal horn. CRP2 is colocalized with cGKI and with markers of primary afferent C fibers. Importantly, the majority of CRP2 mRNA-positive dorsal root ganglion (DRG) neurons express cGKI and CRP2 is phosphorylated in a cGMP-dependent manner. To elucidate the functional role of CRP2 in nociception, we investigated the nociceptive behavior of CRP2-deficient (CRP2-/-) mice. Touch perception and acute thermal nociception were unaltered in CRP2-/- mice. However, CRP2-/- mice showed an increased nociceptive behavior in models of persistent pain as compared to wild type mice. Intrathecal administration of cGKI activating cGMP analogs increased the nociceptive behavior in wild type but not in CRP2-/- mice, indicating that the presence of CRP2 was essential for cGMP/cGKI-mediated nociception. These data indicate that CRP2 is a new downstream effector of cGKI-mediated spinal nociceptive processing and point to an inhibitory role of CRP2 in the generation of inflammatory pain.
Art, Culture and Society Vol 1 is the first in a series of books to be published by Twaweza Communications on the relationship between art and society, with special reference to Kenya. It is part of a cultural leadership initiative being undertaken by the organization through a reexamination of the arts as they are produced and studied. This volume brings together important reflections on the arts and is a major step in encouraging dialogue on the relationship between creativity and the human condition in the region. Significantly, it creates a space for university-based academics to engage in dialogue with artists and writers based outside institutions of higher learning. The conversations will bridge the gap between the two domains for knowledge production and enrich creative enterprise in Kenya, in theory and practice. As the essays in this collection show, the present global situation demands a way to conceptualise and theorise an ever growing cultural interconnectedness, sometimes manifested in art; and interconnectedness that draws from a myriad of cultures and experiences. Through the bridges of contact and cultural exchange distant images are mediated and brought closer to us. They are reinterpreted and modified. In the final analysis, culture is shown to be an important aspect of human creativity but separateness and boundedness is contested. Instead, culture is shown to be malleable and fluid. The essays bring in a new freshness to our reading of the creative arts coming out of Kenya.
Background Cryptic species are two or more distinct but morphologically similar species that were classified as a single species. During the past two decades we observed an exponential growth of publications on cryptic species. Recently published reviews have demonstrated cryptic species have profound consequences on many biological disciplines. It has been proposed that their distribution is non-random across taxa and biomes. Results We analysed a literature database for the taxonomic and biogeographical distribution of cryptic animal species reports. Results from regression analysis indicate that cryptic species are almost evenly distributed among major metazoan taxa and biogeographical regions when corrected for species richness and study intensity. Conclusion This indicates that morphological stasis represents an evolutionary constant and that cryptic metazoan diversity does predictably affect estimates of earth´s animal diversity. Our findings have direct theoretical and practical consequences for a number of prevailing biological questions with regard to global biodiversity estimates, conservation efforts and global taxonomic initiatives.
This issue of L1 – Educational Studies in Language and Literature is the largest single issue we have produced since our introduction in 2000. Containing seven articles, it covers a range of L1 issues: reform movements, the role of literature, culture and multiculturalism in L1, literacy, technology, reading comprehension and the role of oral and written language in L1 Teacher Education. Authors represent a similar diverse national scope: New Zealand, North Cyprus, France, USA, Hong Kong and Israel. The issue reflects the sentiments expressed in our IAIMTE Conference 2007 theme in Exeter, UK: "Crossing Cultural Boundaries." It represents both the diversity in the field and the simultaneous opportunity to speak to a wide range of critical L1 issues between the covers of a single copy of the journal.
This paper analyses cross-border contagion in a sample of European banks from January 1994 to January 2003. We use a multinomial logit model to estimate the number of banks in a given country that experience a large shock on the same day (“coexceedances”) as a function of variables measuring common shocks and coexceedances in other countries. Large shocks are measured by the bottom 95th percentile of the distribution of the first difference in the daily distance to default of the bank. We find evidence in favour of significant cross-border contagion. We also find some evidence that since the introduction of the euro cross-border contagion may have increased. The results seem to be very robust to changes in the specification.

Scholars of institutional design attribute large importance to the choice of new institutions. The comparative analysis of how Rwanda and Zambia crafted their new electoral systems and the systems of government regards procedural, structural and rational choice variables which may influence the option for particular solutions. External influences and the type of transition are determinants that can decide which actors make their interests prevail. The degree of innovation or conservatism of new institutions is mainly a result of the speed of the process and the kind of actors involved. However, rational reflections on how to produce legitimacy and minimize personal risks which take into consideration the state of conflict in the country decide on the speed and on innovative outcomes. The structured analysis of only two cases uncovers already that it is rather difficult to realise the transfer of design recommendations into reality.
Background Bacterial DNA containing motifs of unmethylated CpG dinucleotides (CpG-ODN) initiate an innate immune response mediated by the pattern recognition receptor Toll-like receptor 9 (TLR9). This leads in particular to the expression of proinflammatory mediators such as tumor necrosis factor (TNF-alpha) and interleukin-1beta (IL-1beta). TLR9 is expressed in human and murine pulmonary tissue and induction of proinflammatory mediators has been linked to the development of acute lung injury. Therefore, the hypothesis was tested whether CpG-ODN administration induces an inflammatory response in the lung via TLR9 in vivo. Methods Wild-type (WT) and TLR9-deficient (TLR9-D) mice received CpG-ODN intraperitoneally (1668-Thioat, 1 nmol/g BW) and were observed for up to 6 hrs. Lung tissue and plasma samples were taken and various inflammatory markers were measured. Results In WT mice, CpG-ODN induced a strong activation of pulmonary NFKB as well as a significant increase in pulmonary TNF-alpha and IL-1beta mRNA/protein. In addition, cytokine serum levels were significantly elevated in WT mice. Increased pulmonary content of lung myeloperoxidase (MPO) was documented in WT mice following application of CpG-ODN. Bronchoalveolar lavage (BAL) revealed that CpG-ODN stimulation significantly increased total cell number as well as neutrophil count in WT animals. In contrast, the CpG-ODN-induced inflammatory response was abolished in TLR9-D mice. Conclusion This study suggests that bacterial CpG-ODN causes lung inflammation via TLR9.
This paper extends the internal frank report 28 as follows: It is shown that for a call-by-need lambda calculus LRCCP-Lambda extending the calculus LRCC-Lambda by por, i.e in a lambda-calculus with letrec, case, constructors, seq and por, copying can be done without restrictions, and also that call-by-need and call-by-name strategies are equivalent w.r.t. contextual equivalence.
Call-by-need lambda calculi with letrec provide a rewritingbased operational semantics for (lazy) call-by-name functional languages. These calculi model the sharing behavior during evaluation more closely than let-based calculi that use a fixpoint combinator. In a previous paper we showed that the copy-transformation is correct for the small calculus LR-Lambda. In this paper we demonstrate that the proof method based on a calculus on infinite trees for showing correctness of instantiation operations can be extended to the calculus LRCC-Lambda with case and constructors, and show that copying at compile-time can be done without restrictions. We also show that the call-by-need and call-by-name strategies are equivalent w.r.t. contextual equivalence. A consequence is correctness of all the transformations like instantiation, inlining, specialization and common subexpression elimination in LRCC-Lambda. We are confident that the method scales up for proving correctness of copy-related transformations in non-deterministic lambda calculi if restricted to "deterministic" subterms.
This paper presents psycholinguistic evidence on the factors governing the resolution of German personal pronouns. To determine the relative influence of linear order versus grammatical function of potential antecedents, two interpretation-preference tasks were designed. Their specific aim was to disentangle salience factors conflated in previous research on pronoun interpretation, such as linear or-der, first mention and topicalization. Experiment 1 tested pronoun resolution to non-sentence-initial position (scrambling) and Experiment 2 tested pronoun resolution to sentence-initial position (topicalization). The results across different verb types and across different syntactic contexts in Experiments 1 and 2 show that grammatical function, yet neither linear order, first mention nor topicalization predicts pronoun resolution in German.
The adult morphology of the recently established species Somatochlora borisi Marinov, 2001 is outlined. The species has a unique combination of features, especially when compared to representatives of the two closest European genera, Cordulia Leach, 1815 and Somatochlora Selys, 1871 but also compared to other Holarctic genera and species within the Corduliinae (sensu Garrison et al. 2006). The extent of these morphological differences suggests that the species can not be assigned to any of the extant genera, and therefore the new genus Corduliochlora is being established.
Information supply is the genuine task of academic institutions as well as of publishers. Publishers profit from copyright provisions which give them exclusive rights in their products. The same copyright provisions are often the limiting factor when academic institutions try to improve their service to the academic community. This is the case in particular when it comes to digital access to information. In a so-called "Second Basket", the German copyright act has just been revised, introducing explicit legal exemptions for document deliveries and on the spot consultation of works contained in public libraries' collections. At the same time, unresolved issues remain with respect to existing legal exemptions as well as the new ones. What will the legal parameters look like for academic institutions once the "Second basket" has been put into force? How can libraries work with these provisions in practice?
A systematic revision of the genus Dichaeu (Orchidaceae) in Costa Rica is presented. The taxonomic history of the genus and its phylogenetic position are discussed, with emphasis on infragenenc grouping. Characters of vegetative and floral morphology are treated, and their taxonoiilic significance is discussed. Twenty-nine Dichnea taxa are recognized for the flora of Costa Rica, and a key to species is provided. Each taxon is described on the basis of Costa Rican material, illustrated in a composite plate, and its distribution in the country is assessed. Distribution maps for all the taxa are given. Overall distribution, derivation of name, notes on species ecology, and diagnostic features are presented for each taxon. Lectotypes are selectcd for D. acostae Schltr., D. acroblephara Schltr., D. amparoana Schltr., D. costaricensis Schltr., D. dammeriana Kraenzl., D. lycopodioides Rchb. f. ex Kraenzl., D. poicillantha Schltr., D. selaginella Schltr., D. tuercklheimii Schltr., Epidendrum echinocarpon Sw., and E. trichocarpon Sw. A new species, D. gomez-lauritoi, is described and illustrated from the wet Caribbean plains of central Costa Rica.
Bypassing of DNA lesions by damage-tolerant DNA polymerases depends on the interaction of these enzymes with the monoubiquitylated form of the replicative clamp protein, PCNA. We have analyzed the contributions of ubiquitin and PCNA binding to damage bypass and damage-induced mutagenesis in Polymerase {eta} (encoded by RAD30) from the budding yeast Saccharomyces cerevisiae. We report here that a ubiquitin-binding domain provides enhanced affinity for the ubiquitylated form of PCNA and is essential for in vivo function of the polymerase, but only in conjunction with a basal affinity for the unmodified clamp, mediated by a conserved PCNA interaction motif. We show that enhancement of the interaction and function in damage tolerance does not depend on the ubiquitin attachment site within PCNA. Like its mammalian homolog, budding yeast Polymerase {eta} itself is ubiquitylated in a manner dependent on its ubiquitin-binding domain.
The following new species are described from the Maghreb: Tapinocyba algirica n. sp. and Walckenaeria heimbergi n. sp. The unknown male of Minicia elegans and the unknown females of Alioranus pauper, Cherserigone graciipes and Entelecara truncatifrons are described. Tmeticus hipponense is transfered to the genus Gongylidiellum and HybocoptliS ericicola is removed from synonymy with H. corrugis and revalidated. The Maghrebian species of the genera Alioranus, Brachycerasphora, Cherserigone, Didectoprocnemis, Entelecara, Eperigone, Erigone, Gnathonarium, Gonatium, Gongylidiellum, Hybocoptus, Lessertia, Maso, Mierargus, Microetenonyx, Minicia, Monocephalus, Nematogmus, Ostearius, Prinerigone, Styloetetor, Tapinocyba, Triehoncoides and Trichoncus are all revised. As a final paper in a series on the Linyphiidae of the Maghreb, all the remaining genera are reviewed. A total of 169 species of Linyphiidae has currently been recorded in the Maghreb.
During the second part of the TROCCINOX campaign that took place in Brazil in early 2005, chemical species were measured on-board the high-altitude research aircraft Geophysica (ozone, water vapor, NO, NOy, CH4 and CO) in the altitude range up to 20 km (or up to 450 K potential temperature), i.e. spanning the entire TTL region roughly extending between 350 and 420 K. Here, analysis of transport across the TTL is performed using a new version of the Chemical Lagrangian Model of the Stratosphere (CLaMS). In this new version, the stratospheric model has been extended to the earth surface. Above the tropopause, the isentropic and cross-isentropic advection in CLaMS is driven by meteorological analysis winds and heating/cooling rates derived from a radiation calculation. Below the tropopause, the model smoothly transforms from the isentropic to the hybrid-pressure coordinate and, in this way, takes into account the effect of large-scale convective transport as implemented in the vertical wind of the meteorological analysis. As in previous CLaMS simulations, the irreversible transport, i.e. mixing, is controlled by the local horizontal strain and vertical shear rates. Stratospheric and tropospheric signatures in the TTL can be seen both in the observations and in the model. The composition of air above ≈350 K is mainly controlled by mixing on a time scale of weeks or even months. Based on CLaMS transport studies where mixing can be completely switched off, we deduce that vertical mixing, mainly driven by the vertical shear in the tropical flanks of the subtropical jets and, to some extent, in the the outflow regions of the large-scale convection, offers an explanation for the upward transport of trace species from the main convective outflow at around 350 K up to the tropical tropopause around 380 K.
New evidence is provided for a grammatical principle that singles out contrastive focus (Rooth 1996; Truckenbrodt 1995) and distinguishes it from discourse-new “informational” focus. Since the prosody of discourse-given constituents may also be distinguished from discourse-new, a three-way distinction in representation is motivated. It is assumed that an F-feature marks just contrastive focus (Jackendoff 1972, Rooth 1992), and that a G-feature marks discoursegiven constituents (Féry and Samek-Lodovici 2006), while discoursenew is unmarked. A crucial argument for G-marking comes from second occurrence focus (SOF) prosody, which arguably derives from a syntactic representation where SOF is both F-marked and G-marked. This analysis relies on a new G-Marking Condition specifying that a contrastive focus may be G-marked only if the focus semantic value of its scope is discourse-given, i.e. only if the contrast itself is given.

The article puts forward a discourse-pragmatic approach to the notoriously evasive phenomena of contrastivity and emphasis. It is argued that occurrences of focus that are treated in terms of "contrastive focus", "kontrast" (Vallduví & Vilkuna 1998) or "identificational focus" (É. Kiss 1998) in the literature should not be analyzed in familiar semantic terms like introduction of alternatives or exhaustivity. Rather, an adequate analysis must take into account discourse-pragmatic notions like hearer expectation or discourse expectability of the focused content in a given discourse situation. The less expected a given content is judged to be for the hearer, relative to the Common Ground, the more likely a speaker is to mark this content by means of special grammatical devices, giving rise to emphasis.

Ohne Lebensgeschichtliches ungebührlich hineinzuziehen oder gar für die Dichtung selbst zu überfordern, verdient berücksichtigt zu werden, dass Hofmannsthal wenige Wochen vor Niederschrift dieser Terzinen menschliches Vergehen in überaus persönlicher und angreifender Nähe wahrnehmen musste, den Tod der hochverehrten Josephine von Wertheimstein; diese Erfahrung bestätigte sein beklommenes Ahnen, ließ ihn zutiefst erleiden, "wie viel unendliche Schönheit da für immer weggegangen […] Es war schon […] früher so grauenhaft, sie zu sehen; ihre edle, großartige Schönheit war in etwas Schattenhaftes, Verblichenes, Hilfloses verwandelt […]". Diese Erscheinung, schon zuvor "Symbol für unzählige Dinge", blickt auch aus den Terzinen hervor.
Abeillé and Godard (2007) describe a variety of Spanish whose complex predicates differ structurally from the more familiar flat VP type of complex predicate common to other varieties of Spanish and Romance. I present a verb cluster analysis of this variety which both captures these structural differences, and at the same time preserves those features that are common across both construction types. Coupled with a simple morphological treatment of affixation, this analysis predicts the range of 'clitic climbing' facts. The parsimony of the affixation analysis is afforded by an alternative approach to the constraints on reflexive affix distribution in Spanish complex predicates. I depart radically from previous morpho-lexical approaches to the phenomenon, instead showing how the constraints follow from independently motivated binding principles. This approach not only handles more of the Spanish data, but also has the potential to provide a unified account of the phenomenon across Romance.
In 1991, the newly elected National Government of New Zealand set in train a major reform of the New Zealand national curriculum and, a little later, a major reform of the New Zealand qualifications system. These reforms have had a major impact on the construction of English as a subject in New Zealand secondary schools, and the work and professional identity of teachers. This article uses as a basis for analysis a framework which posits four paradigms for subject English and proceeds to examine the current national English curriculum in New Zealand for its underlying discourses. In specific terms, it explores questions of partition and progression, and terminology. In respect of progression, it argues that the current curriculum has imposed a flawed model on teachers and students, in part because of its commitment to the assignment of decontextualised outcomes statements ('achievement objects') to staged levels of student development (levels). It also argues that much of the terminology used by the document has had a negative impact on metalinguistic classroom practice. Finally, while it views the national English curriculum as a discursively mixed bag, it notes an absence of critical discourses and a tendency, in recent qualifications reforms, to construct English teachers as technicians and the subject as skills-based.
The rate of species extinctions due to anthropogenic activities has dramatically increased within the past few centuries (Dirzo & Raven, 2003; Novacek & Cleland, 2001). Although the mechanisms and ultimate causes leading to the extinction of species remain largely unclear (Frankham et al., 2002), five threats to global biodiversity have frequently been referred to as the most important: habitat destruction and fragmentation, global climate change, hunting and overuse of food resources, biological invasions and environmental pollution (Dudgeon et al., 2006; Lewis, 2006; Novacek & Cleland, 2001). Different research fields, as conservation biology, ecology and ecotoxicology, investigate the effects of these factors on organisms and found strong evidence for their negative impact on regional and global biodiversity.
In most cases, natural populations will be impacted not only by one threat, but rather a combination of them (Buckley & Roughgarden, 2004; Kappelle et al., 1999). Multiple environmental stress factors can have cumulative negative effects on the survival of populations (Sih et al., 2004). To understand, how natural populations respond to combinations of different stress factors is thus of crucial importance in order to understand our present and future impact on all scales of biodiversity (Warren et al., 2001).
The effects of anthropogenically introduced chemicals on organisms and ecosystems are investigated in the field of ecotoxicology. Research in this area has led to a large body of information concerning the impact of chemical stress on the fitness of model species in the laboratory. In contrast to this, there is an obvious lack of knowledge on the effects of contaminants on natural populations and communities (Bickham et al., 2000; Bourdeau et al., 1990). For instance, ecotoxicologists have just started to investigate the impact of environmental pollution on the genetic variability of natural populations (Bickham et al., 2000; Whitehead et al., 2003). Genetic variation provides the raw material for populations in order to adapt to changing environmental conditions and is thus the substrate for evolution and long-term survival of populations and species (Frankham, 2005). The amount of genetic variation in populations is positively correlated with the effective population size (Frankham, 1996). Habitat destruction and fragmentation has divided the ranges of many species into small and isolated refuges. Without migration from adjacent habitats, isolated populations will decrease in their level of genetic diversity through random loss of alleles (Hedrick, 2000). Frankham (1995) for instance, showed that 32 of the 37 endangered species (which occur in small populations per definition) of different animals and plant taxa display reduced levels of heterozygosity compared to closely related and more frequent species.
In strongly human impacted landscapes, both factors, environmental pollution and habitat destruction, can be expected to occur frequently together. It is thus of crucial importance to investigate the impact of reduced genetic diversity and inbreeding on the response to chemical stress. In addition, chemical exposure has frequently been discussed to have an impact on the extent of genetic variability in exposed populations (Guttman, 1994; Staton et al., 2001; van Straalen & Timmermans, 2002). However, evidence for this 'genetic erosion hypothesis' remained scarce to date, most likely because of the difficulty to single out the impact of pollution stress from a background of multiple factors which influence patterns of genetic variability in natural populations (Belfiore, 2001; Staton et al., 2001; van Straalen & Timmermans, 2002).
Consequences of altered eicosanoid patterns for nociceptive processing in mPGES-1-deficient mice

Cyclooxygenase-2 (COX-2)-dependent prostaglandin (PG) E2 synthesis in the spinal cord plays a major role in the development of inflammatory hyperalgesia and allodynia. Microsomal PGE2 synthase-1 (mPGES-1) isomerizes COX-2-derived PGH2 to PGE2. Here, we evaluated the effect of mPGES-1-deficiency on the noci-ceptive behavior in various models of nociception that depend on PGE2 synthesis. Surprisingly, in the COX-2-dependent zymosan-evoked hyperalgesia model, the nociceptive behavior was not reduced in mPGES-1-deficient mice despite a marked decrease of the spinal PGE2 synthesis. Similarly, the nociceptive behavior was unaltered in mPGES-1-deficient mice in the formalin test. Importantly, spinal cords and primary spinal cord cells derived from mPGES-1-deficient mice showed a redirection of the PGE2 synthesis to PGD2, PGF2α and 6-keto-PGF1α (stable metabolite of PGI2). Since the latter prostaglandins serve also as mediators of noci-ception they may compensate the loss of PGE2 synthesis in mPGES-1-deficient mice.

The translation eukaryotic elongation factor 1alpha (eEF1A) is a monomeric GTPase involved in protein synthesis. In addition, this protein is thought to participate in other cellular functions such as actin bundling, cell cycle regulation, and apoptosis. Here we show that eEF1A is associated with the alpha2 subunit of the inhibitory glycine receptor in pulldown experiments with rat brain extracts. Moreover, additional proteins involved in translation like ribosomal S6 protein and p70 ribosomal S6 protein kinase as well as ERK1/2 and calcineurin were identified in the same pulldown approaches. Glycine receptor activation in spinal cord neurons cultured for 1 week resulted in an increased phosphorylation of ribosomal S6 protein. Immunocytochemistry showed that eEF1A and ribosomal S6 protein are localized in the soma, dendrites, and at synapses of cultured hippocampal and spinal cord neurons. Consistent with our biochemical data, immunoreactivities of both proteins were partially overlapping with glycine receptor immunoreactivity in cultured spinal cord and hippocampal neurons. After 5 weeks in culture, eEF1A immunoreactivity was redistributed to the cytoskeleton in about 45% of neurons. Interestingly, the degree of redistribution could be increased at earlier stages of in vitro differentiation by inhibition of either the ERK1/2 pathway or glycine receptors and simultaneous N-methyl-D-aspartate receptor activation. Our findings suggest a functional coupling of eEF1A with both inhibitory and excitatory receptors, possibly involving the ERK-signaling pathway.
Based on Krifka (1992) and de Kuthy (2000), this paper develops an architecture for complex topic-comment structures in HPSG and applies it to predicate fronting in English with the goal of capturing the insights of Ward (1988) on this construction. We argue that predicate fronting is a distributed constructional form consisting of an auxiliary occurring in a predicate preposing phrase. The use of predicate preposing is a function of a combination of simultaneous constraints on its theme structure, its background-focus distribution, and its presuppositional structure. It is shown that these constraints can be made explicit within the HPSG architecture developed here.
Complement control is a well-known phenomenon in Turkish linguistics, and different proposals for analysing it are available. The majority of these treat control as a structural phenomenon, cf. Kerslake (1987), Özsoy (1987; 2001) and Kural (1998). In sum, control is predicted only in sentences with complement clauses formed with the suffixes -mEk and -mE, which can be case-marked, but the appearance of a possessive marker definitely precludes control. As far as the control relations are concerned, the research so far has attested the classical cases of subject and object control. In addition to that, variable control is discussed by Taylan (1996). The status of the controlled element is discussed by Bozşahin (in press), which concludes that the syntactic subject is appointed by this function in Turkish.
In this paper I will argue that the currently established approach to control is insufficient. The shortcomings of a strictly configurational approach become clear if a broader perspective on control is adopted. I follow the approach to control outlined by Stiebels (this volume), and show that two types of control must be distinguished. Inherent control is encoded in the lexical entry of the verb. Verbs which show inherent control either select only control-inducing structures or trigger control in environments not requiring control. Structural control, on the other hand, arises through the use of a control-inducing structure with a verb which does not inherently require control. Structural control verbs show control only with control-inducing structures. No control occurs with such verbs in other configurations. The data discussed in this paper will show that control is a ‘mixed’ phenomenon, since it may arise structurally or semantically. Its explanation must therefore consider the semantics of the relevant matrix verbs and the syntactic properties of complement clauses on an equal basis.
Exchanges in Europe are in a process of consolidation. After the failure of the proposed merger between Deutsche Börse and Euronext, these two groups are likely to become the nuclei for further mergers and co-operation with currently independent exchanges. A decision for one of the groups entails a decision for the respective trading platform. Against that background we evaluate the attractiveness of the two dominant continental European trading systems. Though both are anonymous electronic limit order books, there are important differences in the trading protocols. We use a matched-sample approach to compare execution costs in Euronext Paris and Xetra. We find that both quoted and effective spreads are lower in Xetra. When decomposing the spread we find no systematic differences in the adverse selection component. Realized spreads, on the other hand, are significantly higher in Euronext. Neither differences in the number of liquidity provision agreements nor differences in the minimum tick size or in the degree of domestic competition for order flow explain the different spread levels. We thus conclude that Xetra is the more efficient trading system. JEL Classification: G10, G15
Why is it hard to divide attention between dissimilar activities, such as reading and listening to a conversation? We used functional magnetic resonance imaging (fMRI) to study interference between simple auditory and visual decisions, independently of motor competition. Overlapping activity for auditory and visual tasks performed in isolation was found in lateral prefrontal regions, middle temporal cortex and parietal cortex. When the visual stimulus occurred during the processing of the tone, its activation in prefrontal and middle temporal cortex was suppressed. Additionally, reduced activity was seen in modality-specific visual cortex. These results paralleled impaired awareness of the visual event. Even without competing motor responses, a simple auditory decision interferes with visual processing on different neural levels, including prefrontal cortex, middle temporal cortex and visual regions.
Poster presentation: The transcription factor NF-kappaB plays a pivotal role in the development and maintenance of the central nervous system and its constitutive activation in neurons has been previously reported. NF-kappaB is post-translationally activated upon phosphorylation of the IkappaBalpha inhibitory protein by the activated IkappaB kinase (IKKalpha/beta) and the subsequent degradation of IkappaBalpha by the proteasome. Recently, we had demonstrated an unexpected accumulation of three components of the NF-kappaB cascade in the axon initial segment (AIS): Activated IKK, phosphorylated IkappaBalpha and phosphorylated-p65(Ser536). These are all associated with detergent-insoluble cytoskeletal components of the AIS. We observed further compartimentalization as pIKKalpha/beta primarily associated with the membrane cytoskeleton, whereas pIkappaBalpha was sequestered to fasciculated microtubules. Colchicine-induced depolymerization of microtubules was associated with reduced sequestration of pIkappaBalpha in the AIS, which could be blocked by use of proteasome inhibitors like Mg-132 or Lactacystin. Concurrently, enhanced nuclear immunoreactivity for the NF-kappaB subunit p65 was noted. Using NF-kappaB-dependent reporter gene assays, a significant increase in NF-kappaB activity was observed after depolymerization of microtubules and this was inhibited by the microtubule-stabilizing drug paclitaxel. The use of transiently transfected, photoactivatable-GFP p65 fusion proteins will allow us to specifically analyse the compartimentalized signal transduction pathways in unique spatial and temporal resolution. Taken together, these observations provide strong evidence for compartmentalized activation of NF-kappaB in the AIS and modulation of neuronal NF-kappaB activity by microtubule dynamics.
Prepositional phrase (PP) attachment is one of the major sources for errors in traditional statistical parsers. The reason for that lies in the type of information necessary for resolving structural ambiguities. For parsing, it is assumed that distributional information of parts-of-speech and phrases is sufficient for disambiguation. For PP attachment, in contrast, lexical information is needed. The problem of PP attachment has sparked much interest ever since Hindle and Rooth (1993) formulated the problem in a way that can be easily handled by machine learning approaches: In their approach, PP attachment is reduced to the decision between noun and verb attachment; and the relevant information is reduced to the two possible attachment sites (the noun and the verb) and the preposition of the PP. Brill and Resnik (1994) extended the feature set to the now standard 4-tupel also containing the noun inside the PP. Among many publications on the problem of PP attachment, Volk (2001; 2002) describes the only system for German. He uses a combination of supervised and unsupervised methods. The supervised method is based on the back-off model by Collins and Brooks (1995), the unsupervised part consists of heuristics such as ”If there is a support verb construction present, choose verb attachment”. Volk trains his back-off model on the Negra treebank (Skut et al., 1998) and extracts frequencies for the heuristics from the ”Computerzeitung”. The latter also serves as test data set. Consequently, it is difficult to compare Volk’s results to other results for German, including the results presented here, since not only he uses a combination of supervised and unsupervised learning, but he also performs domain adaptation. Most of the researchers working on PP attachment seem to be satisfied with a PP attachment system; we have found hardly any work on integrating the results of such approaches into actual parsers. The only exceptions are Mehl et al. (1998) and Foth and Menzel (2006), both working with German data. Mehl et al. report a slight improvement of PP attachment from 475 correct PPs out of 681 PPs for the original parser to 481 PPs. Foth and Menzel report an improvement of overall accuracy from 90.7% to 92.2%. Both integrate statistical attachment preferences into a parser. First, we will investigate whether dependency parsing, which generally uses lexical information, shows the same performance on PP attachment as an independent PP attachment classifier does. Then we will investigate an approach that allows the integration of PP attachment information into the output of a parser without having to modify the parser: The results of an independent PP attachment classifier are integrated into the parse of a dependency parser for German in a postprocessing step.

We discuss the present collective flow signals for the phase transition to the quark-gluon plasma (QGP) and the collective flow as a barometer for the equation of state (EoS). We emphasize the importance of the flow excitation function from 1 to 50A GeV: here the hydrodynamicmodel has predicted the collapse of the v1-flow at ~ 10A GeV and of the v2-flow at ~ 40A GeV. In the latter case, this has recently been observed by the NA49 collaboration. Since hadronic rescattering models predict much larger flow than observed at this energy, we interpret this observation as potential evidence for a first order phase transition at high baryon density pB.
Working closely with teaching and research staff is critical to the success of libraries and information services. Indeed, the degree of integration with a University's academic work is one of the factors that distinguish a successful service from a poor one. This paper will consider the relationship between information services and how universities operate. Using the challenges facing institutions as a starting point - including the move towards a single European higher education market - the impact of information provision on institutional strategies will be explored. Information resources underpin all learning, teaching and research activities and the presentation will consider the professional practice which ensures that libraries and computing services are fully exploited. The focus on the experience of students is leading some institutions to integrate information services with a wide range of other activities and the paper will consider the opportunities and challenges which this brings, including the need to build working relationships with a broader range of professional groups.
La présente étude revisite la liste des langues bantoues du Gabon contenues dans la classification des langues bantoues de Guthrie (1967 - 1971), en y ajoutant une nouvelle langue, des nouveaux dialectes, et pour la première fois des sous dialectes. Prenant en compte les acquis des classifications antérieures des langues bantoues du Gabon, elle revient sur les propositions de codification de Maho (2003,2006 et 2007) visant à ajouter des nouvelles langues et de nouveaux dialectes dans la liste de Guthrie sans remettre en question le système de codification établi par ce dernier. Des nouvelles propositions sont formulées ici qui réanalysent ou réaménagent les modèles de codification avancés par cet auteur, tout en se portant en faux contre une sacralisation inavouée de la classification de Guthrie qui viserait à mettre ses propositions d’indexation des langues bantoues à l’abris de toute modification ou réaménagement. Enfin, la présente classification revient sur un certain nombre de dénominations et de problèmes orthographiques des noms de langues relevées dans les anciennes classifications, pour proposer des corrections qui tiennent comptent soit du bon sens, soit du point de vue des locuteurs ou des conventions de notation des langues bantoues arrêtées par les linguistes de l’Ecole de Tervuren.
This paper argues that new digital genres clash with notions of a 'traditional' version of English, as represented in post-16 Advanced Level Literature exam courses in England. This argument is set within the context of an ongoing political imperative to integrate ICT into the school curriculum together with general optimism amongst many English teachers regarding the potential of particular uses of ICT to enhance teaching and learning in aspects of English (Andrews, 2001; Stevens & McGuinn, 2004). The paper focuses on hypertext which has been the subject of some exciting theoretical claims about its value for literary study, ranging from access to searchable databases, texts and research, to democratising the publishing process and changing the relationship between reader, writer and text (Delany & Landow, 1991; Landow, 1994; Joyce, 1996). The paper draws on a case study of an Advanced Level Literature classroom 'design' within the ESRC InterActive Education Project. The class experimented with the use of hypertext as a tool for researching and writing about literature. This revealed the dissonance between the subject culture of English Literature and the subject culture of ICT. Students attempted to negotiate altered reading and writing practices which were not readily compatible with the assessment demands and classroom practices of Advanced Level English Literature. This negotiation involved different levels of student resistance and compliance with the project of integrating technology into English literature study. The paper ends with some speculations about which aspects of 'traditional' English should be retained and valued in an age of information saturation and multimedia hype.

Die vorliegende Arbeit untersuchte die CIN- Inzidenz und die HPV- Prävalenz bei Frauen mit einer HIV- Infektion, die aus dem Patientinnenkollektiv der Universitäts-frauenklinik Frankfurt am Main, rekrutiert wurden. Die Daten von 305 Frauen wurden in die Studie eingebracht. Dieses Kollektiv wur-de einer HIV- negativen Kontrollgruppe gegenübergestellt. Die wichtigsten Ergebnisse der Untersuchung waren: - die CIN- Inzidenz in dem HIV-positiven Kollektiv lag bei 27% (n=83 von 305) - die CIN- Inzidenz in der HIV- negativen Kontrollgruppe lag bei 31% (n=14 von 45) - HIV- positive Frauen mit CIN 1-3 hatten eine signifikant höhere HPV- Prävalenz (78%) als HIV- positive Frauen mit CIN 0 (27%) - die CIN- Inzidenz bei HIV- und HPV- positiven Frauen war mit 52 % (n=65 von 126) signifikant höher als die CIN- Inzidenz bei HIV- positi-ven aber HPV- negativen Frauen, die bei 10% (n=18 von179) lag - die CIN- Inzidenz bei HIV- negativen und HPV- positiven Frauen mit 88% (n=14 von 18) signifikant erhöht war, während HIV- und HPV- negative Frauen eine CIN- Inzidenz von 0 % hatten (n=0 von 29) - die CIN- Inzidenz bei HIV- und HPV- positiven Frauen unabhängig von der CD4 Zellzahl war - die CIN- Inzidenz bei HIV- positiven und HPV- negativen Frauen ab-hängig von der CD4 Zellzahl ist und bei Zellzahlen unter 200/ul signifkant erhöht ist (39% versus 7%, p< 0,01) - eine antiretrovirale Therapie beeinflusst die CIN- Inzidenz anscheinend nicht - der Infektionsweg der betroffenen Frauen (Drogenabusus, heterose-xuelle Kontakte, Abstammung aus Hochprävalenzländern) keinen signifikanten Unterschied bezüglich der CIN- Inzidenz ausmacht - Frauen mit Nikotinkonsum in der Anamnese haben eine signifikant erhöhte CIN- Inzidenz (38%,n=63 von 163) als Frauen ohne Nikotin-konsum (14%, n=20 von 143) Insgesamt reihen sich die Untersuchungergebnisse in die umfangreiche Datenlage der aktuellen Literatur weitgehend ein. Zudem ist das hier untersuchte Kollektiv zahlenmäßig, sicherlich eines der größten im deutschen und internationalen Ver-gleich. Schlussfolgern lässt sich eine erhöhtes CIN- Risiko bei HIV- positiven Frauen. Die hier erhobenen Daten erhärten einmal mehr, die Forderung eines intensivierten Screeningprogramms für die betroffenen Frauen.
Chthonius heterodactylus Tömösváry, 1882 is recorded for the first time from the Czech Republic (Hranická chasm). The occurence of this Carpathian species in Central Europe is discussed and the positions of the type localities are corrected.
Koalas are popular zoo animals, but difficult in husbandry. In addition to their specialised diet of eucalyptus leaves, they are prone to “stress” and disease. Particularly in European zoos, themonitoring of theirwell-being has high priority and they are protected from possible stressors. However, stress signs in koalas are vague and monitoring techniques like weighing might result in discomfort itself. Additionally, husbandry routines are planned according to keeper’s schedule, not to the endogenous rhythms of the koalas. Therefore it is necessary to investigate activity pattern in captive koalas and the signals influencing them. These signals have to be assessed on the strength and quality of their impact. A total of 17 koalas have been observed in three zoological gardens in Australia and Europe. Koalas kept in outdoor enclosures with little human contact (Koala Walkabout, Taronga Zoo, Sydney) showed a uniform activity pattern, which was clearly entrained by light. Activity levels were higher during the night, and there was a pronounced resting period in the morning which corresponds with low body temperature measured by Degabriele and Dawson (1979). Activity peaks were related to twilight and changed during the year related to day lengths. However, there was a clear influence from the introduction of fresh browse which resulted in a distinct feeding peak in the afternoon. With short day lengths, this stimulus competed with dusk. Activity patterns from koalas in indoor enclosures (Zoo Duisburg, Vienna Zoo) varied between individuals and in some cases lacked a detectable rhythm. Though activity peaks were related to light, entrainment to sunlight was weak. In winter, koalas reacted primarily to the artificial light, but some also showed activity peaks related to sunlight. Activity patterns in these koalas were less structured and differed severely from patterns expected according to literature. Activity was often related to the keeper’s presence and food introduction. Frequency of feeding bouts was considerably higher at Vienna Zoo compared to the other zoos and the bouts were shorter in duration. Time budgets of the koalas were within the range given in free-range studies. Feeding showed seasonal changes and was increased in lactating females. Koalas at Vinna Zoo had a high level of locomotor activity compared to the size of the enclosure. Koalas at Koala Walkabout were not used to handling, so they resisted the keeper. The koalas at the two European zoos were handled regularly and settled down quickly. However, handling took place in the morning; in most koalas, there was no activity prior to it. In Vienna, resting periods were interrupted daily due to weighing. Food introduction at KoalaWalkabout took place in the afternoon. It was preceded by locomotor activity and triggered a long feeding bout in the koalas. It is not clear, whether food had true Zeitgeber properties or masked the endogenous rhythm. In the two European zoos, food was introduced in the morning. The peaks related to this were smaller than those at Koala Walkabout. Activity was rarely observed prior to food introduction. The koalas at Koala Encounter, Taronga Zoo (Sydney),were regularly confronted with visitors, though no contact was allowed. Direct observation by the keepers did rarely show any stress signs. Activity patterns at night were strikingly similar to Koala Walkabout, but differed dramatically during the day. Food was introduced three times a day, which usually resulted in activity that interrupted a resting period. Generally, the koalas at Koala Encounter were more active than those at KoalaWalkabout. They also displayed a high level of locomotor activity, especially on the ground, which is an accepted sign of discomfort in koalas (Wood 1978; Zoological Society of San Diego 2001; Yusuf& Rosenthal unpublished data). In summary, this chronoethological study of the captive koalas showed that there are several problems with koala husbandry. Artificial light regimes for koalas are not sufficient for entrainment and result in unstructured activity pattern. This is especially the case in winter, when the day in Europe is artificially extended. Due to the mainly nocturnal behaviour of koalas, such an extension might not be necessary and therefore should be avoided. Handling in Europe took place during the physiological resting time of the koalas. Interruptions of resting times are considered as stressors (Wood 1978) and should be avoided. Handling in the afternoon would be more suitable for the koalas and triggered activity in the two koalas at Vienna Zoo. It is also arguable if daily weighing is necessary to monitor health in captive koalas or if the frequent interruption of resting countervail the advantages of constant monitoring. Frequent contact with visitors, evenwithout the so-called cuddling, has a considerable impact on activity patterns and time budget of koalas, even if no immediate stress signs are displayed. Such contact should therefore be reduced to a minimum and chronoethological observations of the koalas should be used. A study on koalas with direct visitor contact is also advisable to revise the current legislation on “koala cuddling”. Koalas frequently rested in living trees if they had access to it. Since no food-poisoning has been reported from koalas using living non-food trees, the provision of living trees with an appropriate canopy should be included in the husbandry guidelines. Increased locomotor activity has been shown to be related to conditions of discomfort or stress and possibly to oestrus. This is in accordance with literature (Wood 1978; Zoological Society of San Diego 2001). Further observation, combined with hormone analysis, are advisable to establish this parameter for evaluation of well-being. Chronoethology has proven to be useful for the evaluation of husbandry conditions and group dynamics. Different to other, traditional ethologicalmethods, it indicated problems and enabled me to advise more appropriate times for handling and food introduction. It is desirable that zoos already using 24-hour video observation include chronoethological aspects into their analysis.
This paper presents results of corpus analytic investigations of children's use of referring expressions and considers possible implications of this work for questions relating to development of theory of mind. The study confirms previous findings that children use the full range of referring forms (definite and indefinite articles, demonstrative determiners, and demonstrative and personal pronouns) appropriately by age 3 or earlier. It also provides support for two distinct stages in mind-reading ability. The first, which is implicit and non-propositional, includes the ability to assess cognitive statuses such as familiarity and focus of attention in relation to the intended referent; the second, which is propositional and more conscious, includes the ability to assess epistemic states such as knowledge and belief. Distinguishing these two stages supports attempts to reconcile seemingly inconsistent results concerning the age at which children develop theory of mind. It also makes it possible to explain why children learn to use forms correctly be-fore they exhibit the pragmatic ability to consider and calculate quantity implicatures.
In the context of AIDS and a declining economy, one strategy for children to ensure their own livelihood is to engage in domestic employment. Here, Michael Bourdillon presents the findings of research based on interviews and discussions with child domestic workers in Zimbabwe. It looks at the circumstances that pushed them into employment, the hardships and humiliations they face therein, as well as the benefits they derive, including, in some cases, education. Most children wanted improvements in their living and working conditions. They did not want to be stopped from working, perceiving that this would worsen their already harsh lives. While child domestic wok is problematic, and often lays children open to various types of abuse, it can also offer critical support and patronage to very disadvantaged children.
Chemical ozone loss in winter 1991–1992 is recalculated based on observations of the HALOE satellite instrument, ER-2 aircraft measurements and balloon data. HALOE satellite observations are shown to be reliable in the lower stratosphere below 400 K, at altitudes where profiles are most likely disturbed by the enhanced sulfate aerosols, as a result of the Mt. Pinatubo eruption in June 1991. Very large chemical ozone loss was observed below 400 K from Kiruna balloon observations between December and March 1992. Additionally, for the two winters after the Mt. Pinatubo eruption, HALOE satellite observations show a stronger extent of chemical ozone loss at lower altitudes compared to other Arctic winter between 1991 and 2003. In stipe of already occurring deactivation of chlorine in March 1992, Mipas-B and LPMA balloon observations indicate still chlorine activation at lower altitudes, consistent with observed chemical ozone loss occurring between February and March and April. Enhanced chemical ozone loss in the Arctic winter 1991–1992 as calculated in earlier studies is corroborated here.
This preliminary checklist of Aphodiini south of the United States is prepared to provide published data for a future web-based checklist of all New World Aphodiinae. All species names are used in combination with their currently accepted generic name, creating many new combinations. A few genus-species combinations are discussed. The interplay of charmonium production and suppression in In+In and Pb+Pb reactions at 158 AGeV and in Au+Au reactions at sqrt(s)=200 GeV is investigated with the HSD transport approach within the hadronic comover model' and the QGP melting scenario'. The results for the J/Psi suppression and the Psi' to J/Psi ratio are compared to the recent data of the NA50, NA60, and PHENIX Collaborations. We find that, at 158 AGeV, the comover absorption model performs better than the scenario of abrupt threshold melting. However, neither interaction with hadrons alone nor simple color screening satisfactory describes the data at sqrt(s)=200 GeV. A deconfined phase is clearly reached at RHIC, but a theory having the relevant degrees of freedom in this regime (strongly interacting quarks/gluons) is needed to study its transport properties.
Almost two thousand years ago the question was asked, "And who is my neighbor?", and Jesus answered it with the parable of the Good Samaritan (Luke 10. 29-37), and so the idea of charity was inseparably linked with Christianity. However, it is almost a common opinion that Western Christianity and Russian Orthodoxy have quite different approaches to this item. According to that opinion, the Catholic Church had more concern about one's bodily needs and explicit welfare, while the Russian Orthodoxy paid less attention to the ''explicit man" and took more care of "the inner man". The Russian Orthodox tradition - though it has its own method in ascetism, apotheosis, general and private prayer - pays too little attention to the needy and deprived members of society. To raise the question about mollification of the deprived people's torments and to see it as a way towards the Kingdom of God seems alien to the very essence of Russian Orthodoxy. Did the Russian Orthodox Church ever refuse to help beggars and cripples? Or do the facts still show another picture? Since when did Russian Orthodox Church start to neglect that task? Which reasons have proved that charity has been unable to flourish in the Orthodox Church and especially Russian Orthodox Church as flourished her spirituality and arts: icon painting, choral singing, and church architecture? Let us have a brief outlook on the history of the Russian Orthodox Church and find out if there are any reasons to explain the reason.
1. Die Deletionsderivate der Kompetenzproteine PilN und PilQ wurden als Fusionsproteine mit dem Maltosebindeprotein überproduziert, über eine Amylosesäule aufgereinigt und in nativer Form für die Generierung von polyklonalen Kaninchen-Antikörpern eingesetzt. 2. Unter Einsatz der spezifischen alpha-PilN- und alpha-PilQ-Antikörpern konnte das PilN-und PilQ-Kompetenzprotein im Rohextrakt von T. thermophilus HB27 detektiert werden. 3. Die Überprüfung bereits vorliegender Antikörper gegen die Kompetenzproteine PilM, PilW und PilA4 ergaben, dass es sich hier ebenfalls um spezifische Antikörper handelt. 4. Mittels Western-Blot-Analysen unterschiedlicher Mutanten konnte gezeigt werden, dass die Markerinsertion in den Genen pilM, pilN, pilO und pilW zu keinem polaren Effekt der jeweils stromabwärts gelegenen Gene führt. 5. Analysen der subzellulären Lokalisation der Kompetenzproteine ergaben, dass PilM und PilN ausschließlich in der inneren Membran lokalisiert sind, während PilW, PilQ und PilA4 sowohl in der inneren als auch äußeren Membran detektiert wurden. Die größte Menge an PilQ wurde allerdings in der äußeren Membran gefunden. 6. Mutantenstudien ergaben, dass eine pilQ-Mutantion zur Abwesenheit von PilW und PilA4 in der äußeren Membran führte. Ebenso führte eine pilW-Mutantion zu Abwesenheit von PilQ und PilA4 in der äußeren Membran. Diese Ergebnisse indizieren Interaktionen zwischen PilW, PilQ und PilA4. 7. In der pilD-Mutante wurde kein PilA4 mehr in der äußeren Membran detektiert. Dieser Befund untermauert den Schluss, dass es sich bei PilD um eine PilA4 prozessierende Präpilin-Peptidase handelt. 8. Western-Blot-Analysen der gereinigten Pili führen zu dem Schluss, dass das Kompetenzprotein PilA4 die strukturelle Untereinheit der Pilus-Struktur repräsentiert. Das Sekretin-ähnliche Kompetenzprotein PilQ wurde ebenfalls in der gereinigten Pilus-Fraktion detektiert und könnte Teil der globulären Struktur an den Pilusenden sein. 9. Elektronenmikroskopische Aufnahmen von Dünnschnitten durch T. thermophilus HB27 Zellen ließen den ungewöhnlichen Aufbau der Zellperipherie erkennen. Zwischen der inneren Membran (8 nm dick) und der äußeren Membran (8 nm dick) befand sich neben dem dünnen Peptidoglykan (8 nm dick) eine 40 nm dicke kontrastarme Schicht, die radial von Fäden durchzogen schien. 10. Die Dünnschnitte wurden für Immunogoldmarkierungen mit alpha-PilQ-Antikörpern eingesetzt, um PilQ zu detektieren. Allerdings konnte in den elektronenmikroskopischen Analysen keine spezifische Goldmarkierung nachgewiesen werden. Ursache hierfür könnte die Unzugänglichkeit des nativen PilQ in den Dünnschnitten sein. 11. Die Immunogoldmarkierung des Gefrierbruchs von ganzen Thermus-Zellen mit alpha-PilQ-Antikörpern führte zur Detektion von PilQ-haltigen Ring-ähnlichen Strukturen in der äußeren Membran. Der Durchmesser des PilQ-Komplexes mit 17 - 18 nm ist mit denen der aus Sekretinen bestehenden Ringsystemen anderer Organismen vergleichbar. 12. In den T. thermophilus HB27 Membransolubilisaten wurde ein >669 kDa PilW-PilQ-Komplex identifiziert. Dieser Komplex ließ sich optimal mit n-Dodecyl-beta-D-maltosid (1 mg Detergenz pro mg Membranprotein) aus der Membranfraktion solubilisieren. Stabilitätsuntersuchungen ergaben, dass dieser Komplex bei 4 °C einige Tage, bei Raumtemperatur und bei Anwesenheit von 1 M NaCl einige Stunden stabil ist. 13. Mittels einer DEAE-Austausch-Chromatographie konnte der PilW-PilQ-Komplex angereichert werden. Die Western-Blot-Analysen ergaben, dass das Kompetenzprotein PilM mit dem PilW-PilQ-Komplex koeluiert. 14. In der MALDI-TOF-Massenspektrometrie konnte das Kompetenzprotein PilQ und Untereinheiten der DNA-gerichteten RNA-Polymerase, ein Membran-Lipoprotein sowie Energie-Stoffwechsel-Proteine in der angereicherten PilW-PilQ-haltigen Fraktion nachgewiesen werden. Diese Proteine müssen auf die Beteiligung an der Transformation untersucht werden. 15. Immunelektronenmikroskopische Analysen des angereicherten PilW-PilQ-Komplexes führten zur Identifizierung von Ringstrukturen mit einem Durchmesser von 17 nm. Dieser Durchmesser entspricht den Sekretin-Komplexen von P. aeruginosa (18,3 ± 1,2 nm) bzw. N. gonorrhoeae (15,5 - 16,5 nm) und dem Durchmesser der über Immunogoldmarkierung detektierten PilQ-haltigen Strukturen in T. thermophilus HB27.
Die Arbeit überprüft die Zusammensetzung der F1FO-ATP-Synthase in Säugetiermitochondrien, dem Enzymkomplex, der das meiste ATP für den Energiebedarf einer Zelle liefert. Es sind zwei neue Proteine identifiziert und als ATP-Synthase assoziiert verifiziert worden, das sog. dapit protein (diabetes-associated protein in insulin-sensitive tissue; Datenbanknummer in NCBI für Rattus norvegicus, gi|19424210) bzw. 6.8 kDa mitochondrial proteolipid (Datenbanknummer in NCBI für Rattus norvegicus, gi|109478763). Bis jetzt sind beide Proteine nicht zusammen mit dem Komplex V detektiert worden, da es sich bei beiden Proteinen um sehr kleine Membranproteine (kleiner 7 kDa) handelt und sie sehr leicht in Gegenwart von Detergenzien verloren gehen. Die etablierte Strategie zur milden Aufreinigung von Komplex V, die eingesetzte gelelektrophoretische Trennung und die gewonnenen Erkenntnisse zur Identifizierung solch kleiner Proteine können sicherlich auch Lösungsansätze für andere ungelöste Problemfälle in der Proteinkomplexanalytik liefern. Da beide neuen Proteine in die Modulation des metabolischen Zellzustandes involviert sein könnten, sind die erarbeiteten Daten für weitere funktionelle und biochemische Untersuchungen der ATP-Synthase äußerst nützlich. Außerdem könnten die Ergebnisse für neurologische und klinische Studien hinsichtlich der Ursachenforschung von Funktionsstörungen in den Mitochondrien von Interesse sein, da eines der zwei neuen Proteine früher schon mit Diabetes in Zusammenhang gebracht worden ist (dapit, diabetesassociated protein in insulin-sensitive tissue). Für ein bakterielles Multihäm c-Typ Cytochrom konnte massenspektrometrisch gezeigt werden, dass es auf eine unkonventionelle Weise Häm bindet. Durch massenspektrometrische Charakterisierung des Proteins konnte erstmals nachgewiesen werden, dass es nicht nur die Häm c-Bindemotive CX2-4CH und CXXCK, sondern auch Häm c-Bindemotive der Form CXnCH in Bakterien gibt. Diese Erkenntnis führt in der Molekularbiologie zu neuen Fragen, z. B. welche speziellen Lyasen (cytochrome c haem lyases) letztendlich für das Einfügen der Häm-Gruppe an solche neuen Motive verantwortlich sind. Auch die computerbasierte Vorhersage von c-Typ Cytochromen wird dieses Wissen wohl zukünftig in Suchstrategien umsetzen, um die neuen Häm c-Bindemotive bei der Genomanalyse von Organismen nicht zu übersehen. In dem Feld der Identifizierung und Charakterisierung von Membranproteinen im Allgemeinen konnten grundlegende Erkenntnisse zum Umgang mit alternativen Enzymen und deren Potential für einen zukünftigen Einsatz erarbeitet werden. Schwerpunktmäßig wurden die Enzyme Chymotrypsin, Elastase und Pepsin untersucht. Es konnte für alle drei Kandidaten gezeigt werden, dass sie bevorzugt an einer begrenzten Anzahl von Aminosäuren spalten. Besonders für Elastase ist diese Erkenntnis neu, da sie in der Literatur bisher als unspezifisches Enzym wie Proteinase K geführt wurde. Auch wenn die Spezifität der drei Enzyme nicht zu 100% wie bei Trypsin festgelegt werden kann, sondern es sich nur um eine Bevorzugung gewisser Aminosäuren handelt, sind die enzymatischen Spaltungen reproduzierbar. Selbst eine Auswertung der MS-Spektren mittels Peptide Mass Fingerprint (PMF) ist deshalb auch bei diesen weniger spezifischen Enzymen möglich. Die Intensität der MS-Signale muss aber berücksichtigt werden, was bei bisherigen PMF-Suchen jedoch nicht in der Art und Weise geschieht, wie es für diese Enzyme nötig wäre. An einigen Membranproteinen konnte letztendlich bereits beispielhaft gezeigt werden, dass der Einsatz von weniger spezifischen Enzymen für die Identifizierung des Proteins und der nachfolgenden Charakterisierung (z. B. Identifizierung von posttranslationale Modifikationen) vorteilhaft ist. Für Elastase konnte in diesem Zusammenhang auch demonstriert werden, dass sie problemlos in Lösungsmittelsystemen mit einem hohen organischen Anteil (Acetonitril, Isopropanol, Methanol) einsetzbar ist. 100% Sequenzabdeckung lassen sich aber auch bei weniger spezifischen Enzymen trotz der größeren Anzahl an Schnittmöglichkeiten nur erahnen. Zwei Hauptursachen hierfür sind wahrscheinlich die schlechte Zugänglichkeit des Enzyms zum Membranprotein bzw. die Bevorzugung bestimmter enzymatischer Fragmente in MALDI und ESI. Polyacrylamidgele mit alternativen Quervernetzern, bei denen sich die Geldichte vor dem Verdau verringern lässt, könnten die Zugänglichkeit zum Membranprotein zukünftig vielleicht positiv beeinflussen. Der Einsatz von organischen Lösungsmitteln und bestimmter Detergenzien beim Verdau verbessert ebenfalls die Zugänglichkeit zum Membranprotein. Die Zahl der Tenside, die mit der Massenspektrometrie sehr gut kompatibel sind, ist aber sehr gering, wie Untersuchungen in dieser Arbeit ebenfalls ergeben haben. Außerdem beschränkt sich die Anwendung von diesen Detergenzien ausschließlich auf MALDI. Die zu erwartenden Fortschritte bei der Identifizierung und Charakterisierung von Membranproteinen umschreibt daher besonders gut ein Aphorismus von Christian Morgenstern (deutscher Schriftsteller; 1871 – 1914): „Es gibt nur ein Neues: Die Nuance.“ Einige Nuancen sind in dieser Arbeit enthalten. In der Zukunft werden aber viele weitere solcher Nuancen das Überwinden der Hürde „Membran Proteomics“ immer realistischer werden lassen.
In der vorliegenden Arbeit sollte das basolaterale Targeting des Transmembranproteins shrew-1 in polarisierten Epithelzellen analysiert werden. Es konnte gezeigt werden, dass die cytoplasmatische Domäne von shrew-1 mehrere spezifische basolaterale Sortingmotive enthält. Die Funktionalität dieser Motive wurde anhand Mutationsanalysen von Schlüsselaminosäuren untersucht. Substitution dieser Aminosäuren führt zu einer apikalen Lokalisation von shrew-1 in polarisierten MDCK Zellen. Durch Analyse der Proteinverteilung von shrew-1 Varianten in polarisierten LLC-PK1 Zellen wurde deutlich, dass das Sorting von shrew-1 in die basolaterale Plasmamembran ein AP-1B-abhängiger Prozess ist. Außerdem konnte mittels Coimmunopräzipitation eine Interaktion zwischen shrew-1 und der Untereinheit my1B aus dem Adapterproteinkomplex AP-1B nachgewiesen werden. Untersuchungen des Targetings von shrew-1 Varianten in polarisierten MDCK und LLCPK1 Zellen mit Hilfe der Transzytoseexperimente zeigten, dass die apikal lokalisierte Mutante shrew-1-NTD5 auf dem Weg zur apikalen Membranregion, trotz fehlender Sortinginformation, die basolaterale Plasmamembran durchquert. Durch Inhibition der Membranfusion mittels Tanninsäure konnte zusätzlich gezeigt werden, dass die Passage der basolateralen Plasmamembran für das Targeting von sowohl shrew-1 als auch von shrew-1-NTD5 essentiell ist. Die Beobachtungen des Turnovers von shrew-1 in der Plasmamembran von lebenden Zellen zeigten, dass shrew-1 aktiv endozytiert wird und dass nachfolgend ein Recycling des Proteins zur Plasmamembran stattfindet. Anhand der durchgeführten Untersuchungen lässt sich zusammenfassend ein Targetingmodell für shrew-1 in polarisierten Epithelzellen aufstellen, das ein postendozytotisches Sorting beschreibt: Dabei wird shrew-1 zunächst in Post-Golgi-Carriern auf unbekanntem Weg zur basolateralen Plasmamembran gebracht, wo seine unmittelbare Internalisierung und ein Weitertransport zum Recyclingendosom stattfinden. Der im Recyclingendosom lokalisierte und am Sorting beteiligte Adapterproteinkomplex AP-1B vermittelt dann den Rücktransport von shrew-1 zur basolateralen Plasmamembran.
Dem Nahrungsmittelallergiker bietet sich zum jetzigen Zeitpunkt lediglich eine Möglichkeit für ein symptomfreies Leben, er muss die allergieauslösenden Nahrungsmittel aus seinem Speiseplan eliminieren. Neben der Entwicklung von immuntherapeutischen Maßnahmen wird unabhängig davon auf pflanzentechnologischer Seite die Reduktion der allergenen Potenz pflanzlicher Lebensmittel diskutiert, um die Lebensqualität von Nahrungsmittelallergikern zu verbessern. Die Kooperation zwischen dem Institut für Biochemie der Friedrich-Alexander Universität Erlangen-Nürnberg und dem Paul-Ehrlich-Institut leistet einen Beitrag zu dieser Diskussion. Mit der Tomate als Modellpflanze wurde die allergene Potenz pflanzlicher Nahrungsmittel mit Hilfe der RNAi-Technologie reduziert. Diese Dissertation befasst sich mit der Identifikation und Charakterisierung der Zielproteine für das RNAi vermittelte „gene silencing“. Im Fokus der Untersuchungen steht das Lipid Transfer Protein der Tomate. Bei der Charakterisierung des Tomaten LTP wurden die IgE-bindenden Epitope sowie die Stabilität bei Pepsinverdau und bei der Verarbeitung zu tomatenhaltigen Produkten untersucht. Das Tomaten LTP besteht aus einer Multigenfamilie. Durch seine zweidimensionale Auftrennung konnten Informationen über das Vorkommen und Expressionsniveau möglicher Isoformen gewonnen werden. Die rekombinante Herstellung von zwei ausgewählten Isoformen ermöglichte einen Vergleich der IgE-Bindungskapazität und der biologischen Potenz. Die in Erlangen erzeugten gentechnisch veränderten Pflanzen wurden dem Paul-Ehrlich-Institut für eine Allergenitätsbewertung zur Verfügung gestellt. Die Gewinnung von Erkenntnissen über die Effizienz der Methode und die Stabilität des „gene silencing“ über mehrere Generationen hinweg standen hier im Mittelpunkt. Die Untersuchungen zur Prävalenz der IgE-Bindung an die drei bekannten Tomatenallergene Profilin (Lyc e 1), Invertase (Lyc e 2) und das LTP der Tomate, erfolgten bei einer Patientengruppe bestehend aus 70 Seren von Tomatenallergikern aus Deutschland (n=36) und Spanien (n=34). Als häufigstes Symptom nach dem Verzehr von Tomate wurde das OAS angegeben. Die Häufigkeit der IgE-Bindung an Lyc e 1 lag bei ca. 36%, für Lyc e 2 bei ca. 56% und für das LTP der Tomate bei ca. 24%. Bei der Unterscheidung der deutschen und spanischen Patientengruppe ergaben sich höhere Prävalenzen der IgE-Bindung an alle drei Allergene bei der spanischen Patientengruppe. Das Tomaten LTP wurde als Minorallergen Lyc e 3 in die IUIS Allergendatenbank aufgenommen. Die Aufreinigung von nLyc e 3 erfolgte durch eine zweistufige Chromatographie aus Tomatenschalenextrakt, die Identifizierung als LTP mittels N-terminaler Sequenzierung sowie die Verifikation der Sekundärstruktur mittels CD-Spektroskopie. Biochemische Untersuchungen zeigten eine hohe Stabilität von Lyc e 3 in Tomatenschalenextrakt nach Pepsinverdau. Auch im Schalenextrakt von gekochten und in getrockneten Tomaten wurde immunreaktives Lyc e 3 identifiziert. Die Konformation des Lyc e 3 ist an der IgE- und IgG-Bindung maßgeblich beteiligt. Natürliche Lyc e 3 Isoformen konnten nicht über die zweidimensionale Gelelektrophorese identifiziert werden. Eine Sequenzierung des Proteins mittels nano-HPLC-nano ESI-MS/MS bestätigte die Präsenz ausschließlich einer dominanten Isoform im Tomatenschalenextrakt. Bei der Klonierung und Expression zweier ausgewählter Lyc e 3 Isoformen (Lyc e 3.1 und Lyc e 3.2) wurden unterschiedliche Strategien verfolgt. Durch Überexpression eines Thioredoxinfusionsproteins im Vektor pET-32a in E.coli Origami und die Abspaltung des Thioredoxinanhangs durch Thrombin konnte ausreichend Protein für die nachfolgenden Untersuchungen produziert werden. Jedoch zeigten die rekombinanten Proteine, vermutlich durch eine an dem Zielprotein verbleibende Anhangssequenz, bei der Messung der CD-Spektren einen Unterschied zu der natürlichen Präparation. Die Herstellung von rekombinanten Isoformen ohne Anhangssequenz mit einer Sekundärstruktur analog der natürlichen Präparation blieb erfolglos. Ein Vergleich der IgE-Bindungskapazität der rekombinanten Proteine, gemessen in der dosisabhängigen ELISA-Inhibition, konnte auf Grund ihrer identischen Sekundärstruktur durchgeführt werden. Da rLyc e 3.2 eine höhere IgE-Bindungskapazität zu besitzen scheint, wurde mit dieser Isoform eine dosisabhängige ELISA-Inhibition mit nLyc e 3 trotz unterschiedlicher Sekundärstruktur durchgeführt. Die IgE-Bindungskapazitäten sind für die herangezogenen Seren sehr individuell. Zur Bestimmung der allergenen Potenz der rekombinanten und natürlichen Präparationen diente der basophile Histamin Freisetzungstest. Die natürliche Präparation besitzt tendenziell die stärkste allergene Potenz. Die allergene Potenz der rekombinanten Isoformen ist jedoch wie die IgE-Bindungskapazität für jeden Patienten sehr spezifisch. Im Rahmen der Analyse der gentechnisch veränderten Pflanzen wurden WT und RNAi-Pflanzen mittels Immunoblot, dosisabhängiger ELISA-Inhibition und im basophilen Histamin Freisetzungstest untersucht. Im Immunoblot konnte keine Bindung von allergenspezifischen Antikörpern bei den RNAi-Pflanzen im Vergleich zum WT festgestellt werden. Bei der dosisabhängigen ELISA-Inhibition mit Lyc e 1 oder Lyc e 3 auf der Festphase war die IgE-Bindungskapaziät der RNAi Pflanzen um ein zehnfaches (Lyc e 1-RNAi) bzw. ein zehn- bis hundertfaches (Lyc e 3-RNAi) reduziert. Eine Bestätigung der Ergebnisse mit den Lyc e 3-RNAi Pflanzen lieferten ein basophiler Histamin Freisetzungstest und ein in vivo Hauttest. Durch die zusätzliche Analyse von Tochtergenerationen wurde deutlich, dass die Reduktion der Allergenexpression stabil war. Im Rahmen dieser Studie konnte gezeigt werden, dass die RNAi-Technologie eine geeignete Methode ist, um die Expression von Allergenen gezielt und stabil zu unterdrücken. Sie könnte eine Möglichkeit darstellen, die allergene Potenz von Nahrungsmitteln zu verringern und die Lebensqualität von Nahrungsmittelallergikern zu verbessern.
This volume highlights the proceedings of the two policy dialogue conferences held by the Working Group on Finance and Education (WGFE) in 2004. Part I of the document discusses the endemic crisis that higher educationhas been beset with since the outset of the post colonial period in Africa. It highlights the critical state of higher education systems in Burkina Faso, Mali, Nigeria and Senegal by scrutinizing the causes, manifestations and consequences of the crisis to posit useful recommendations and possible solutions. Part II is a comprehensive review of the challenges facing the financing and planning of all levels and types ofeducation - from kindergarten to graduate school - in selected African countries. The papers reveal the sources and mechanisms of funding education in Africa, drawing attention to the experiences of communities confronted with new funding sources. A new trend, which consists of designing decade long educational development plans, has emerged and is rapidly expanding in numerous African countries. This experience is examined and shared by the authors. This book has contributions in both French and English.
cGMP- and cAMP-dependent protein kinases (cGK and cAK) mediate the inhibitory effects of endothelium-derived messenger molecules nitric oxide and prostacyclin on platelets. To understand the mechanisms involved in platelet inhibition we searched for new substrates of cGK and cAK. We identified Rap1GAP2, the only GTPase-activating protein of Rap1 in platelets. Rap1 is a guanine-nucleotide binding protein that controls integrin activity, platelet adhesion and aggregation. Rap1GAP2 is required to turn over Rap1-GTP to Rap1-GDP resulting in the inactivation of integrins and reduced cellular adhesion. Using phospho-specific antibodies we demonstrate phosphorylation of endogenous Rap1GAP2 on serine 7 by cGK and cAK in intact platelets. Yeast-two-hybrid screening revealed an interaction of the phosphoserine/-threonine binding adapter protein 14-3-3 with Rap1GAP2, and we mapped the 14-3-3 binding site to the N-terminus of Rap1GAP2 close to the cGK/cAK phosphorylation site. We could show that 14-3-3 binding to Rap1GAP2 requires phosphorylation of serine 9. Platelet activation by ADP and thrombin treatment induces Rap1GAP2 serine 9 phosphorylation and enhances the attachment of 14-3-3 to Rap1GAP2. In contrast, phosphorylation of serine 7 by cGK/cAK leads to the detachment of 14-3-3. Furthermore, Rap1GAP2 serine 7 phosphorylation correlates with the inhibition of Rap1-GTP formation by cGMP and cAMP in platelets. Cell adhesion experiments provide additional evidence that Rap1GAP2 is activated by the detachment of 14-3-3. Point mutants of Rap1GAP2 deficient in 14-3-3 binding inhibit Rap1-mediated cell adhesion significantly stronger than a Rap1GAP2 mutant that binds 14-3-3 constitutively. Our findings define a novel regulatory mechanism that might contribute to both platelet activation and endothelial inhibition of platelet adhesion and aggregation.
Defects in podocyte signaling are the basis of many inherited glomerular diseases leading to glomerulosclerosis. CD2-associated protein (CD2AP) is highly expressed in podocytes and is considered to play an important role in the maintenance of the glomerular slit diaphragm. Mice deficient for CD2AP (CD2AP(-/-)) appear normal at birth but develop a rapid onset nephrotic syndrome at 3 weeks of age. We demonstrate that impaired intracellular signaling with subsequent podocyte damage is the reason for this delayed podocyte injury in CD2AP(-/-) mice. We document that CD2AP deficiency in podocytes leads to diminished signal initiation and termination of signaling pathways mediated by receptor tyrosine kinases (RTKs). In addition, we demonstrate that CIN85, a paralog of CD2AP, is involved in termination of RTK signaling in podocytes. CIN85 protein expression is increased in CD2AP(-/-) podocytes in vitro. Stimulation of CD2AP(-/-) podocytes with various growth factors, including insulin-like growth factor 1, vascular endothelial growth factor, and fibroblast growth factor, resulted in a significantly decreased phosphatidylinositol 3-kinase/AKT and ERK signaling response. Moreover, increased CIN85 protein is detectable in podocytes in diseased CD2AP(-/-) mice, leading to decreased base-line activation of ERK and decreased phosphorylation after growth factor stimulation in vivo. Because repression of CIN85 protein leads to a restored RTK signaling response, our results support an important role of CD2AP/CIN85 protein balance in the normal signaling response of podocytes.
Introduction: Metabolic acidosis during hemorrhagic shock is common and conventionally considered to be due to hyperlactatemia. There is increasing awareness, however, that other nonlactate, unmeasured anions contribute to this type of acidosis.
Methods: Eleven anesthetized dogs were hemorrhaged to a mean arterial pressure of 45 mm Hg and were kept at this level until a metabolic oxygen debt of 120 mLO2/kg body weight had evolved. Blood pH, partial pressure of carbon dioxide, and concentrations of sodium, potassium, magnesium, calcium, chloride, lactate, albumin, and phosphate were measured at baseline, in shock, and during 3 hours post-therapy. Strong ion difference and the amount of weak plasma acid were calculated. To detect the presence of unmeasured anions, anion gap and strong ion gap were determined. Capillary electrophoresis was used to identify potential contributors to unmeasured anions.
Results: During induction of shock, pH decreased significantly from 7.41 to 7.19. The transient increase in lactate concentration from 1.5 to 5.5 mEq/L during shock was not sufficient to explain the transient increases in anion gap (+11.0 mEq/L) and strong ion gap (+7.1 mEq/L), suggesting that substantial amounts of unmeasured anions must have been generated. Capillary electrophoresis revealed increases in serum concentration of acetate (2.2 mEq/L), citrate (2.2 mEq/L), alpha-ketoglutarate (35.3 microEq/L), fumarate (6.2 microEq/L), sulfate (0.1 mEq/L), and urate (55.9 microEq/L) after shock induction.
Conclusion: Large amounts of unmeasured anions were generated after hemorrhage in this highly standardized model of hemorrhagic shock. Capillary electrophoresis suggested that the hitherto unmeasured anions citrate and acetate, but not sulfate, contributed significantly to the changes in strong ion gap associated with induction of shock.
Aims: The purpose of this study was to evaluate the feasibility and short-term efficacy of transcatheter paravalvular leak closure using different occlusion devices. Methods and Results: Twenty one patients underwent transcatheter closure of either aortic or mitral paravalvular leak from June 2002 to February 2006 using the Amplatzer PDA, ASD or VSD occluder. All patients had symptoms and signs of haemolysis and/or cardiac decompensation with dyspnoea. Implantation of a device was technically successful in twenty patients (95 %). Immediate residual leak was found in seventeen patients (85 %). Significant shunting persisted in nine cases during follow up (45 %). Permanent leaflet obstruction was observed in one patient. Severe complications during follow up led to early death in one patient and surgical intervention in three. A successful second catheter treatment was performed in another three patients. The event-free survival from reoperation, death and stroke at the end of the observation period was 80 %. Conclusion: Transcatheter closure of paravalvular leaks is a technically feasible, but demanding procedure. Residual leaks are common and may worsen pre-existing haemolysis. Due to the significant ongoing morbidity in this group of patients and the complexity of follow up individual patient results differ considerably. Nevertheless, it is possible to achieve some symptomatic relief, thus an interventional approach should be discussed as a potential treatment option for those patients with a limited defect and who are not deemed suitable for another operation.

We introduce a multivariate multiplicative error model which is driven by componentspecific observation driven dynamics as well as a common latent autoregressive factor. The model is designed to explicitly account for (information driven) common factor dynamics as well as idiosyncratic effects in the processes of high-frequency return volatilities, trade sizes and trading intensities. The model is estimated by simulated maximum likelihood using efficient importance sampling. Analyzing five minutes data from four liquid stocks traded at the New York Stock Exchange, we find that volatilities, volumes and intensities are driven by idiosyncratic dynamics as well as a highly persistent common factor capturing most causal relations and cross-dependencies between the individual variables. This confirms economic theory and suggests more parsimonious specifications of high-dimensional trading processes. It turns out that common shocks affect the return volatility and the trading volume rather than the trading intensity. JEL Classification: C15, C32, C52

Im Rahmen der Untersuchungen an Trockenbiotopen im Allertal (Landkreis Börde, Sachsen- Anhalt) wurden im Jahr 2007 auf mehreren flächenhaften Naturdenkmalen vegetationskundliche Kartierarbeiten durchgeführt. Während dieser Kartierarbeiten wurde ein neuer Fundort von Bupleurum virgatum CAV. [Bupleurum gerardii auct.], dem Ruten-Hasenohr, nachgewiesen. Die Art gilt in Sachsen-Anhalt sowie in ganz Deutschland als vom Aussterben bedroht (FRANK et al. 2004, KORNECK et al. 1996). Das Ruten-Hasenohr ist sehr unauffällig und leicht zu übersehen. Bisher war es nur noch vom Alten Falkenstein im Selketal bekannt (BENKERT et al. 1996, HERRMANN & KISON 2004, HERRMANN 2005). Der Nachweis im Allertal macht Hoffnung, dass die Art auf ähnlichen Standorten noch nachweisbar sein könnte. Daher erfolgt hier eine Beschreibung der Standortverhältnisse am Generalsberg S Groß-Bartensleben (MTB Helmstedt, 3732/4).
Das Bumpmapping-Verfahren, eine Methode zur realistischen Darstellung rauer Oberflächen, existiert schon seit 30 Jahren, aber erst durch aktuelle Entwicklungen der Hardware lässt es sich in Echtzeitumgebungen einsetzen. Die aktuellen Verfahren ermöglichen viele darüber hinausgehende Effekte, jedoch haben sie auch mit Problemen zu kämpfen. Das Ziel dieser Diplomarbeit ist die Weiterentwicklung der Verfahren zu betrachten. In dieser Arbeit werden die Grenzen der aktuellen Bumpmapping-Algorithmen aufgezeigt und nach neuen Wegen geforscht. Das erste Verfahren erzeugt durch ein Multipassrendern fraktale Landschaften im Shader. Die darin verwendeten Methoden lassen sich für einen weiteren Algorithmus nutzen, mit dem feine Unebenheiten der Oberfläche an jedem Pixel ausgewertet werden. So können anisotrope Materialien wie gebürstete Metalle oder Mikropartikellacke simuliert werden. Den Abschluss bilden zwei neue Verfahren für prozedurale Shader. Die zu imitierende Oberfläche wird im Modell nachgebildet und per Raytracing für jeden Oberflächenpixel ausgewertet. Durch diese Methode werden viele Probleme texturbasierter Verfahren komplett umgangen.

Die Geschichte der floristischen Erforschung, Daten zum Naturschutz und Grundlagenhinweise runden die Einleitung ab. Im Hauptteil werden in systematischer Reihenfolge alle Arten vorgestellt. Durch die Rasterverbreitungskarten erkennt der Leser sofort die Verbreitung der jeweiligen Pflanzenarten im Gebiet. Es werden Angaben zum Standort, Häufigkeit und zur Gefährdung der Pflanzen gemacht. Von den selteneren Arten sind alle Fundortangaben aufgeführt. So erhält der Leser einen Einblick in die sehr abwechselungsreiche Sauerländer Flora. The bryophytes (mosses, liverworts and hornworts) that occur in the Blue Mountains region of New South Wales (latitude 33˚–34˚ S, longitude 151˚–151˚40’ E) are listed and information is provided on their distribution in the region. Species lists are based on herbarium specimens and field collections. 348 bryophyte taxa have been recorded from 70 families, including 225 moss taxa (in 108 genera from 45 families), 120 liverwort taxa (in 51 genera from 24 families) and 3 hornwort taxa (in 3 genera from one family). The moss families with most taxa are the Pottiaceae (with 23 taxa in 13 genera), Bryaceae (with 15 taxa in 3 genera) and Fissidentaceae (with 13 taxa). The largest genera are Fissidens (13 taxa), Campylopus (9) and Macromitrium (8). The liverwort family with the most taxa is Lepidoziaceae, with 29 taxa in 10 genera. The largest liverwort genera are Frullania (11 taxa) and Riccardia (8). The species lists include collections from both bushland and urban areas. Natural features of the Blue Mountains, including topography, altitude, climate and vegetation appear to be important factors influencing the number of bryophyte species recorded from each location. The number of collections from particular locations has been considerably influenced by ease of access, particularly proximity to roads, public transport and railway stations. The species lists include many records from areas that were not accessible to the early collectors of the late 19th and early 20th centuries such as Wollemi National Park, Gardens of Stone National Park, Newnes Plateau and Kanangra-Boyd National Park.
In den siebziger Jahren überragte die Deutung der Werke Brechts die anderer Autoren an Umfang und Intensität um ein Vielfaches. Alles wurde diskutiert, seheinbar nichts ausgelassen. Inzwischen ist die Diskussion zum Erliegen gekommen, da die gesellschaftliche und politische Dimension der Literatur, deren Repräsentant Brecht wie kein anderer war, in den neuen kulturwissensehaftlichen Konzeptionen keine Rolle mehr spielt. Die Protagonisten der Brechtforschung und einige ihrer Anhänger publizieren zwar weiter, stoßen aber nicht auf Widerspruch, so daß sich Defizite fortsetzen. Dies ist auch bei einem 1952 erschienenen Band mit dem Titel "Theaterarbeit" der Fall, üher den nichts geschrieben wurde, vermutlich weil der Name Brechts hier nur als Teil einer "Redaktion" auftaucht und theoretische Äußerungen eher im Hintergrund stehen. In Editionen und Handbüchern wird er nicht oder nur am Rande berücksichtigt: In der "Großen kommentierten Berliner und Frankfurter Ausgabe" sind selbst die aus dem Band übernommenen Texte nur unzureichend ediert. In der fünfbändigen Ausgabe des Brecht-Handbuchs (2001-2003) wird der Band nicht als eigenständiges Werk behandelt, so daß die Mängel der Edition nicht korrigiert wurden. Dennoch ist die Theaterarbeit mit 400 großformatigen Seiten, aufwendiger Typographie und zahlreichen Abbildungen nicht nur repräsentativer als alle Publikationen Brechts, sie enthält auch die einzige umfassende Darstellung der epischen Dramaturgie zu seinen Lebzeiten. Durch die Vernachlässigung des Bandes hat eine unbedachte Brechtphilologie deshalb fortgesetzt, was die Kulturinstitutionen der DDR seit Beginn der fünfziger Jahre aus politischen Gründen betrieben haben: die Ausgrenzung eines Grundlagenwerks.
Hintergrund Die chronische Herzinsuffizienz erfordert als Systemerkrankung hausärztliche sowie spezialärztliche Versorgung. Die evidenzbasierte DEGAM-Leitlinie (LL) zur hausärztlichen Versorgung der Herzinsuffizienz wurde formal interdisziplinär konsentiert, nachdem der Entwurf ein mehrstufiges internes und externes Reviewverfahren durchlaufen hatte. Methode Wissenschaftliche Fachgesellschaften und Organisationen (FG/O) wurden zu einem Nominalen Gruppenprozeß (NGP) eingeladen und entsandten autorisierte Teilnehmer. Diese erhielten den LL-Entwurf inkl. Methodenreport sowie eine Liste zentraler LL-Empfehlungen für ein persönliches Ranking (44-Items; 6-stufige Likert-Skala). Beim Konsentierungstreffen wurden aus dem 1. Ranking Themen ohne deutliche Übereinstimmung (Likert =4) identifiziert, unter Hinzunahme weiterer Themenvorschläge in priorisierter Reihenfolge diskutiert und erneut abgestimmt. Der überarbeitete LL-Entwurf wurde in einem zweiten Ranking im Delphi-Verfahren konsentiert. Ergebnisse Im Abstimmungsprozess mit 10 Vertretern aus 11 FG/O wurden ~35 Themen diskutiert. Bei zwei Empfehlungen mit fehlender Evidenz wurde ein von internationalen LL abweichender Konsens getroffen (z.B. ß-Blocker bei asymptomatischen Patienten nur nach durchgemachtem Herzinfarkt). Vier Formulierungen bewertenden Charakters zur BNP-Bestimmung wurden zugunsten einer Negativempfehlung gestrichen, eine Empfehlung mit der STIKO harmonisiert (Pneumokokkenimpfung), bei weiteren wurden ergänzende Konditionen im Wortlaut eingefügt oder sprachliche Änderungen vorgenommen. Fünf Themen wurden neu erstellt (z.B. kontraindizierte Pharmaka). Bis auf drei (z.B. Flussdiagramme nicht vollständig konsensfähig: unangemessene Vereinfachung vs. fehlende Praktikabilität) wurden alle Empfehlungen der LL konsentiert. Schlussfolgerungen Der NGP ist für evidenzbasierte LL eine geeignete Vorgehensweise. Interdisziplinarität ist insbesondere bei Entscheidungsunsicherheit (fehlende oder inkonsistente Evidenz) und zur Schnittstellendefinition wertvoll.
Although the concept that dendritic cells (DCs) recognize pathogens through the engagement of Toll-like receptors is widely accepted, we recently suggested that immature DCs might sense kinin-releasing strains of Trypanosoma cruzi through the triggering of G-protein-coupled bradykinin B2 receptors (B2R). Here we report that C57BL/6.B2R-/- mice infected intraperitoneally with T. cruzi display higher parasitemia and mortality rates as compared to B2R+/+ mice. qRT-PCR revealed a 5-fold increase in T. cruzi DNA (14 d post-infection [p.i.]) in B2R-/- heart, while spleen parasitism was negligible in both mice strains. Analysis of recall responses (14 d p.i.) showed high and comparable frequencies of IFN-gamma-producing CD4+ and CD8+ T cells in the spleen of B2R-/- and wild-type mice. However, production of IFN-gamma by effector T cells isolated from B2R-/- heart was significantly reduced as compared with wild-type mice. As the infection continued, wild-type mice presented IFN-gamma-producing (CD4+CD44+ and CD8+CD44+) T cells both in the spleen and heart while B2R-/- mice showed negligible frequencies of such activated T cells. Furthermore, the collapse of type-1 immune responses in B2R-/- mice was linked to upregulated secretion of IL-17 and TNF-alpha by antigen-responsive CD4+ T cells. In vitro analysis of tissue culture trypomastigote interaction with splenic CD11c+ DCs indicated that DC maturation (IL-12, CD40, and CD86) is controlled by the kinin/B2R pathway. Further, systemic injection of trypomastigotes induced IL-12 production by CD11c+ DCs isolated from B2R+/+ spleen, but not by DCs from B2R-/- mice. Notably, adoptive transfer of B2R+/+ CD11c+ DCs (intravenously) into B2R-/- mice rendered them resistant to acute challenge, rescued development of type-1 immunity, and repressed TH17 responses. Collectively, our results demonstrate that activation of B2R, a DC sensor of endogenous maturation signals, is critically required for development of acquired resistance to T. cruzi infection. Author Summary: Antibodies and IFN-gamma-producing effector T cells are essential for the immune control of infection by Trypanosoma cruzi, the intracellular protozoa that causes human Chagas disease. Despite the potency of anti-parasite immunity, the parasites are not cleared from their intracellular niches. Instead, a low grade chronic infection prevails, provoking severe immunopathology in the myocardium. Although it is well established that innate sentinel cells sense T. cruzi through receptors for microbial structures, such as Toll-like receptors, it remained unclear whether endogenous inflammatory signals also contribute to the development of adaptive immunity. The present study was motivated by awareness that T. cruzi trypomastigotes (extracellular infective forms) are equipped with proteases that liberate the pro-inflammatory bradykinin peptide from an internal segment of kininogens. Here we demonstrate that splenic dendritic cells (DCs), the antigen-presenting cells that coordinate the adaptive branch of immunity in lymphoid tissues, are potently activated via G-protein-coupled bradykinin B2 receptors (B2R). Analysis of the outcome of infection in B2R-knockout mice revealed that the mutant mice developed a typical susceptible phenotype, owing to impaired development of IFN-gamma-producing effector T cells. Notably, the immune dysfunction of B2R-knockout mice was corrected upon cell transfer of wild-type DCs, thus linking development of protective T cells to DCs' sensing of endogenous danger signals (kinins) released by trypomastigotes.
The Botsotso literary journal started in 1996 as a monthly 4 page insert in the New Nation, an independent anti-apartheid South African weekly and reached over 80,000 people at a time ? largely politisized black workers and youth ? with a selection of poems, short stories and short essays that reflected the deep changes taking place in the country at that time. Since the closure of the New Nation in 1999, the journal has evolved into a stand-alone compilation featuring the same mix of genres, and with the addition of photo essays and reviews. The Botsotso editorial policy remains committed to creating a mix of voices which highlight the diverse spectrum of South African identities and languages, particularly those that are dedicated to radical expression and examinations of South Africa's complex society.
Eine ethnographische Studie zu bolivianischen Migrantinnen, die in Barcelona als undokumentierte "Domestic Workers" (private Kinder- und Altenbetreuung, sowie Haushaltshilfe) arbeiten, um mit ihrem Einkommen den Lebensunterhalt ihrer Familien im Herkunftsland zu sichern. Die Arbeit analysiert Motive und Entscheidungsprozesse des Migrationsprojekts aus einer gendersensiblen Perspektive und stellt die Strategien zur Grenzüberwindung in den Rahmen von verschärften EU-Grenzregimen. Weiter wird die prekäre Wohn- und Arbeitssituation der "illegalen" MigrantInnen, insbesondere der Arbeitsplatz Privathaushalt, thematisiert und die Rolle von Hilfsorganisationen kritisch beleuchtet.
Am Peischlachtörl (2484 m Meereshöhe, nördliche Schobergruppe, Nationalpark Hohe Tauern) wurden Bültenböden vegetations- und bodenkundlich untersucht und mit einem typischen alpinen Rasen (Caricetum curvulae) verglichen. Insgesamt wurden 380 Vegetationsaufnahmen à 10 x 10 cm entlang von 14 Kleintransekten durchgeführt, um das Vegetationsmosaik von Bülten und Senken darzustellen. Die Untersuchungen umfassten auch die Bestimmung der Bodentypen, pH-Messungen und Korngrößenanalysen. Bodentemperaturmessungen erfolgten in 10 und 50 cm Bodentiefe von 2003 bis 2005. Es zeigte sich eine deutliche Vegetationszonierung von den Bülten zu den Senken. Die Bultkuppen werden von einer Gesellschaft bestimmt, die dem Caricetum curvulae zugeordnet werden kann, mit einer Loiseleuria procumbens-Ausbildung an einer kleineren Anzahl von Kuppen, einer Oreochloa disticha-Ausbildung für den überwiegenden Anteil der Kuppen sowie einer Ligusticum mutellina-Ausbildung im Übergang zu den Senken. Der alpine Rasen (Caricetum curvulae) lässt sich gut mit der Vegetation der Bultkuppen vergleichen, wobei der überwiegende Teil der Rasen-Aufnahmen zur Oreochloa disticha-Ausbildung gezählt werden kann. Die Senken der Bültenböden werden von einer Luzula alpinopilosa-Ausbildung charakterisiert. Eine pflanzensoziologische Zuordnung dieser Gesellschaft ist sehr schwierig, da Schneebodenarten, Gräser der subalpinen Stufe und Caricetum curvulae-Arten vermischt vorkommen. Im Bereich der Bülten wurde eine pseudovergleyte Braunerde bzw. ein Semipodsol festgestellt, in der Senke ein Pseudogley und im alpinen Rasen ein Eisen-Humus-Podsol. Bültenböden und alpiner Rasen unterscheiden sich signifikant hinsichtlich der Korngrößen: der Boden des alpinen Rasens ist wesentlich skelett- und sandreicher. Kuppen und Senken unterscheiden sich nur unwesentlich in ihrer Körnung. - Der erste Winter zeichnete sich durch fehlenden Bodenfrost aus, während im zweiten Winter sowohl der Boden des alpinen Rasens als auch die Bülten tief durchfroren. We analyze the effect of committee formation on how corporate boards perform two main functions: setting CEO pay and overseeing the financial reporting process. The use of performance-based pay schemes induces the CEO to manipulate earnings, which leads to an increased need for board oversight. If the whole board is responsible for both functions, it is inclined to provide the CEO with a compensation scheme that is relatively insensitive to performance in order to reduce the burden of subsequent monitoring. When the functions are separated through the formation of committees, the compensation committee is willing to choose a higher pay-performance sensitivity as the increased cost of oversight is borne by the audit committee. Our model generates predictions relating the board committee structure to the pay-performance sensitivity of CEO compensation, the quality of board oversight, and the level of earnings management.
Die Grewioideae sind eine Unterfamilie der Malvaceae sensu Bayer et al. (1999). Sie umfassen mit 25 Gattungen und ca. 700 Arten einen Großteil der früheren Tiliaceae. Diese waren vor allem aufgrund der durchweg dithecischen Antheren und der oft zahlreichen, freien Stamina zu einer Familie zusammengefasst worden, auch wenn die enge Verwandtschaft mit den Sterculiaceae, Bombacaceae und Malvaceae bekannt war und die Abgrenzung dieser Familien untereinander oft als künstlich angesehen wurde. ... Insgesamt 45 Arten aus allen 25 Gattungen der Malvaceae-Grewioideae (sensu Bayer & Kubitzki 2003) wurden hinsichtlich ihrer Morphologie im Blüten-, Frucht- und vegetativen Bereich untersucht. Der Schwerpunkt lag dabei auf einer vergleichenden Bearbeitung der Blütenstruktur, für die neben morphologischen auch ontogenetische und anatomische Untersuchungen durchgeführt wurden. Die Ergebnisse waren Grundlage für eine Datenmatrix mit 55 Merkmalen, die ebenso wie DNA-Sequenzdaten (ndhF) für phylogenetische Analysen verwendet wurden. Die Analysen beider Datensätze ergeben eine Zweiteilung der Grewioideae. Auf der Basis der Ergebnisse wird vorgeschlagen, die Grewioideae in die zwei Triben Apeibeae und Grewieae zu unterteilen. Diese Gliederung widerspricht früheren Klassifikationen, ist aber durch strukturelle Merkmale gut gestützt. Die Apeibeae zeichnen sich durch hornförmige Verlängerungen der Sepalen-Spitze und stachelige Emergenzen auf der Fruchtoberfläche aus, Reduktionsformen sind durch Übergänge nachweisbar (Ausnahme: Glyphaea). Leitbündelstränge innerhalb der Fruchtwand verlaufen einzeln. Bestäubungsbiologisch relevante Nektarien, soweit vorhanden, befinden sich vor den Petalen auf einem Androgynophor. Das Androeceum entwickelt sich zentrifugal in einer unterschiedlichen Zahl einheitlicher Kreise. Den Grewieae fehlen Fortsätze der Sepalen-Spitzen sowie stachelige Emergenzen der Früchte. Sie sind durch Nektarien auf den Petalen charakterisiert (Ausnahme: Mollia). Ihre Fruchtwand weist einen geschlossenen Leitbündelmantel auf. Steinkerne oder dorsal geflügelte Fruchtfächer kommen vor. Das Androeceum entwickelt sich oft ungleichmäßig mit einer Förderung des antesepalen Sektors, bisweilen ausgehend von Komplexprimordien. Innerhalb der Triben lassen sich teils neue, teils früher schon bekannte Gruppen erkennen; andere sind anhand der vorliegenden Daten nicht aufzulösen. Dies gilt insbesondere für die Verwandtschaft von Grewia, bei der die Gattungsgrenzen einer kritischen Revision bedürfen. Die Kartierung der morphologischen Merkmale auf den Konsensusbaum der DNA-Sequenzdaten ergab, dass insbesondere der Androgynophor innerhalb der Grewioideae mehrfach parallel entstanden ist und im Gegensatz zu früheren Klassifikationen nicht als Tribus-Merkmal herangezogen werden kann. Auf der Suche nach einem gemeinsamen Bauplan, der den stark unterschiedlichen Blütenstrukturen der Schwestergruppen Grewioideae und Byttnerioideae zugrunde liegen könnte, wurde insbesondere das Androeceum vergleichend untersucht. Bei beiden Unterfamilien findet man polystemone Androeceen, die bei den Byttnerioideae in staminodiale antesepale und fertile antepetale Sektoren differenziert sind. Dies wird in der Literatur oft als obdiplostemones Arrangement zweier Kreise interpretiert, von denen der fertile antepetale Teil dédoubliert. Bei den Grewioideae findet man keine derartige Differenzierung; sie sind in ihrer androecealen Struktur außerordentlich variabel. Die Befunde der Morphologie, Ontogenie und Anatomie widersprechen sich dabei jeweils dergestalt, dass eine theoretische Gliederung des polystemonen Androeceums in zwei Kreise kaum zu rechtfertigen ist. In diesem Zusammenhang wird die in der Literatur verbreitete Vorstellung einer sekundären Polyandrie kritisch beleuchtet. Für die untersuchte Gruppe wird ein Zusammenhang zwischen Bestäubungsbiologie und Blütenstruktur als nahe liegender angesehen, der zu unterschiedlichen Gruppenbildungen innerhalb polystemoner Androeceen führen kann. Während Petalen und Stamina bei den oft fliegenblütigen Byttnerioideae eine spezielle Funktionseinheit bilden, findet man bei den größtenteils bienenblütigen Grewioideae in Zusammenhang mit den Petalen oft Nektar. Die fertilen Stamina können den gesamten antesepalen und antepetalen Raum einnehmen. So stand an der Basis der Grewioideae möglicherweise der Wechsel der Bestäubergruppe und die Auflösung eines blütenbiologisch fixierten Musters, ein Vorgang, wie er innerhalb der Malvaceae mehrfach auf unterschiedliche Weise erfolgt sein könnte. Auch andere Gruppen weisen Blütenstrukturen mit vorwiegend freien Stamina und verborgenem Nektar in Zusammenhang mit Bienenbestäubung auf. Es mag diese blütenbiologisch bedingte Ähnlichkeit gewesen sein, die zur Vereinigung nicht näher verwandter Gruppen zur früheren Familie Tiliaceae geführt hat.
Dichter stellen seit der Antike selbst einen beliebten Gegenstand von Dichtung vor. Einen Höhepunkt erreichte das literarische Interesse an der Künstler- und Dichterpersönlichkeit bekanntlich in der Literatur der Romantik, die das Faszinosum individueller künstlerischer Schaffenskraft selbstreflexiv ins Zentrum der künstlerischen Gestaltung rückte. Nimmt man die deutsche Literatur insgesamt in den Blick, so lassen sich die Dichtergedichte, Dichterdramen, Dichtererzählungen und -romane in ihrer Vielzahl kaum übersehen. Unter ihnen finden sich einige der bekanntesten Werke der deutschen Literatur wie Goethes Drama "Torquato Tasso", Novalis' Künstlerroman "Heinrich von Ofterdingen" und Büchners Erzählung "Lenz". Kein zweiter Schriftsteller aber hat andere Dichter so häufig zum Gegenstand seines Schreibens gemacht wie Robert Walser, dem die Auseinandersetzung mit fremden Autoren zur Grundlage des eigenen literarischen Schaffens wurde.
Als eine Möglichkeit, emotionale Wirkungen von Literatur zu beschreiben, wird eine evolutionspsychologische Heuristik vorgeschlagen. Während der rein textwissenschaftliche Ansatz nur die Darstellung von Emotionen in Texten untersuchen kann, ermöglicht der literaturpsychologische Ansatz ein Inbeziehungsetzen von Werkstruktur und Leserpsyche und damit hypothetische Aussagen über emotionale Wirkungsphänomene. Die Evolutionspsychologie eröffnet einen Zugang insbesondere zu einigen sehr basalen Wirkungsmechanismen, die der Introspektion in der Regel nicht zugänglich sind. So erlaubt die biologische Evolutionstheorie die Annahme einzelner emotionaler Programme, die sich über ihre jeweilige adaptive Funktion in der menschlichen Entwicklungsgeschichte plausibilisieren lassen. Für jedes dieser Emotionsprogramme lässt sich ein spezifisches auslösendes Reizschema festmachen, auf das hin der literarische Text analysiert werden kann. Wird ein Emotionsprogramm durch einen textuellen Reiz ausgelöst, kann man von einer Attrappenwirkung sprechen, da das auslösende Objekt in der Regel nicht identisch ist mit dem, an welchem sich das betreffende Emotionsprogramm evolutionsgeschichtlich herausgebildet hat, sondern lediglich dasselbe Reizschema erfüllt. Zuerst wird die evolutionspsychologische Emotionstheorie vorgestellt, die diesem Modell der literarischen Attrappe zugrunde liegt. Zentral ist dabei die Unterscheidung von Auslösemechanismus und Verlaufsprogramm. Während der Auslösemechanismus den Auslösereiz identifiziert, erste körperliche Reaktionen initiiert und eine spezifische kognitive Verarbeitungsstrategie in Gang setzt, nimmt das Verlaufsprogramm einer Emotion eine zweite Situationseinschätzung vor und passt das Verhalten im Rahmen der vom jeweiligen Emotionsprogramm vorgesehenen Optionen an. Diese Entkopplung von Stimulus und Verhaltensreaktion durch das dazwischengeschaltete Verlaufs-programm erklärt z.B., warum wir auf fiktionale Reize zwar ähnlich, aber nicht identisch reagieren wie auf wirkliche Reize: Durch Fiktionen werden dieselben Emotionen ausgelöst wie in wirklichen Situationen, aber sie nehmen wegen des anders gearteten Situationsfeedbacks einen anderen Verlauf. Im Standardfall werden wir zwar ähnlich 'fühlen', d.h. dieselbe physische und kognitive Aktivierung wahrnehmen, aber anders – nämlich in der Regel gar nicht – handeln. Im zweiten Abschnitt wird der adaptive Wert imaginärer Welten für die ontogenetische Entwicklung des Gehirns dargestellt. Um angeborene Mechanismen im Laufe der individuellen Entwicklung aufzubauen, in Funktionsbereitschaft zu halten und an die jeweils gegebene kulturelle Umwelt anzupassen, müssen Anlässe geschaffen werden, diese Mechanismen wiederholt (und möglichst in pragmatisch entlasteten Situationen) zu aktivieren. Literatur scheint besonders geeignet, eine Reihe von kognitiven und emotionalen Adaptationen zu aktivieren und dadurch ontogenetisch einzuüben. Dazu, solche Gelegenheiten wahrzunehmen, motiviert uns die Evolution durch Funktionslust, d.h. durch eine die Handlung begleitende Ausschüttung von Lusthormonen, die dafür sorgt, dass wir solche scheinbar nutzlosen ('spielerischen') Betätigungen freiwillig aufnehmen und mit der nötigen Ausdauer betreiben. Die evolutionspsychologische Konzeption der Funktionslust kann somit erklären, warum wir die emotionale Wirkung von Kunstwerken genießen und warum es zu solchen paradox scheinenden Phänomenen wie der 'Lust der Tränen' oder dem 'angenehmen Grauen' kommt. Im dritten Abschnitt werden am Beispiel der Landschaftsästhetik evolutionär begründete Präferenzsysteme ("aesthetics") vorgestellt und am literarischen locus amoenus bzw. locus terribilis exemplifiziert. Aus der Notwendigkeit für unsere Vorfahren, nahrungsreiche und sichere Habitate zu identifizieren, lassen sich eine Reihe von Präferenzen für landschaftliche Gegebenheiten, die diesem Bedarf entsprachen und die wir heute als grundlos schön oder angenehm empfinden, ableiten. Reize wie beispielsweise vegetative Üppigkeit, gute Sicht und Schutz vor Witterung steuern als "Annäherungs-" oder "appetitive Reize" unser Verhalten, ohne dass wir darüber nachdenken müssten. Ihre Verwendung in der Literatur wie z.B. im Topos vom 'Lustort' sollte folglich eine positive emotionale Reaktion bewirken. Umgekehrt lassen sich eine Reihe von "Abstoßungs-" oder "aversiven Reizen" (Jeffrey A. Gray) konstatieren, die ein furchtvoll vermeidendes oder abwartendes Verhalten hervorrufen. In der Literatur sind solche Reize vor allem aus dem Motivbereich des 'Erhabenen' bekannt. Am Leitfaden zweier filmwissenschaftlicher Studien werden im vierten Abschnitt zwei spezifische emotionale Reaktionen mit den ihnen korrespondierenden Textreizen vorgestellt. Die Reaktion des Weinens (die Emotion der Rührung) wird auf eine angeborene Kapitulationsreaktion (Helmuth Plessner) gegenüber Situationen der Überforderung und Ohnmacht zurückgeführt und mit resultativen Handlungs-ereignissen in narrativen Texten in Verbindung gebracht. Wann immer es in einer Handlung zu einer Entwicklung kommt, in der es nichts mehr 'zu hoffen und zu bangen' gibt, sondern der Leser einen Ist-Zustand (den Tod einer Figur, den Beweis der Tugend o.Ä.) als definitiv anerkennen muss, wird aufgrund einer strukturellen Isomorphie dieser Erlebniseinheit mit dem evolutionär verankerten Auslöseschemata der Kapitulationsreaktion diese Reaktion des sentimentalen Weinens ausgelöst. Ebenfalls als ein bereichsspezifisches psychisches Reaktionsprogramm wird anschließend die Reaktion des Lachens angeführt. Lachen als Signal der Kooperationsbereitschaft dürfte sich evolutionär besonders in Situationen von gelindem sozialem Stress bewährt haben, d.h. in Situationen, in denen die soziale Kohäsion in Gefahr, aber noch zu retten war. Entsprechend lassen sich Textstrukturen, die im Sinne eines 'unschwerwiegenden Fehlers' Verständnishürden aufbauen und dadurch Stress in der Kommunikationssituation des Lesers herstellen, als geeignete Reize, eine Lachreaktion auszulösen, identifizieren. Die solchermaßen 'humoristische' literarische Sprechinstanz wirkt zudem als sozialer Annäherungsreiz. Mit der Frage der sozialen Attraktivität literarischer Sprecher und fiktiver Figuren befasst sich der fünfte und letzte Abschnitt. Hier wird eine Typologie von vier gestaffelten Sympathiegraden vorgestellt, die es erlaubt, bekannte Phänomene des quasisozialen Leserverhaltens zu gruppieren und bestimmten psychischen Mechanismen zuzuordnen. Als der niedrigste Sympathiegrad kann das Erkennen psychisch verfasster Lebewesen gelten, das sich auf unsere Fähigkeit zur Unterscheidung belebter und unbelebter Objekte zurückführen lässt und darüber entscheidet, auf welche Instanzen im literarischen Text wir uns empathisch einlassen. Der zweite Sympathiegrad wird als 'Parteinahme' betitelt und meint das Erkennen eines 'Mitglieds der eigenen Gruppe' (der eigenen Wertgemeinschaft o.Ä.; in der evolutionären Vergangenheit: des eigenen Clans). Er entscheidet darüber, mit wem der Leser 'mitfiebert'. Als dritter Sympathiegrad kann die freundschaftkonstituierende Vertrautheit mit fiktiven Personen angeführt werden. Diese Familiarität erklärt die affektive Bindung des Lesers an Fiktionen und, als Effekt davon, den Trennungsschmerz beim Beenden eines Buches. Der vierte Sympathiegrad involviert weniger freundschaftliche Bindung als vielmehr 'Liebe' zu einem bestimmten Individuum. Liebestypische Äußerungsformen lassen sich vor allem im Fankult (z.B. im Wertherkult) beobachten.
