Chromalveolates are a diverse group of protists that include many ecologically and medically relevant organisms such as diatoms and apicomplexan parasites. They possess plastids generally surrounded by four membranes, which evolved by engulfment of a red alga. Today, most plastid proteins must be imported, but many aspects of protein import into complex plastids are still cryptic. In particular, how proteins cross the third outermost membrane has remained unexplained. We identified a protein in the third outermost membrane of the diatom Phaeodactylum tricornutum with properties comparable to those of the Omp85 family. We demonstrate that the targeting route of P. tricornutum Omp85 parallels that of the translocation channel of the outer envelope membrane of chloroplasts, Toc75. In addition, the electrophysiological properties are similar to those of the Omp85 proteins involved in protein translocation. This supports the hypothesis that P. tricornutum Omp85 is involved in precursor protein translocation, which would close a gap in the fundamental understanding of the evolutionary origin and function of protein import in secondary plastids.
The donor-free silanimines tBu2Si=N-SiRtBu2 (R = tBu, Ph), which are prepared from tBu2ClSiN3 and NaSiRtBu2 at −78 ◦C inBu2O, decompose in benzene at room temperature with the formation of isobutene. Products of ene reactions of isobutene and tBu2Si=N-SiRtBu2 (R = tBu, Ph) are formed. X-Ray quality crystals of H2C=C(CH2SitBu2-NH-SiPhtBu2)2 (monoclinic, space group C2/c, Z = 4) were grown from a benzene solution at ambient temperature, whereas single crystals of H2C=C(CH2SitBu2-NH-SitBu3)2 (monoclinic, space group P21, Z = 2) were obtained by recrystallization from THF.

The title molecule, C34H28I4·4C6H6, has crystallographic 4 symmetry and crystallizes with four symmetry-related benzene solvent molecules. The phenyl group is eclipsed with one of the adamantane C—C bonds. The tetraphenyladamantane units and the benzene solvent molecules are connected by weak intermolecular phenyl–benzene C—H⋯π and benzene–benzene C—H⋯π interactions. In the crystal, molecules are linked along the c-axis direction via the iodophenyl groups by a combination of weak intermolecular I⋯I [3.944 (1) Å] and I⋯π(phenyl) [3.608 (6) and 3.692 (5) Å] interactions.
Die vorliegende Arbeit hat die Charakterisierung und Untersuchung des Stabilitätsverhaltens von
Parvulustat (PL), einem Homologen des α-Amylase-Inhibitors Tendamistat, zum Inhalt. Zur
weitreichenden Charakterisierung wurden verschiedene Proteinregionen des Parvulustats der CTerminus,
das hydrophobe Cluster, die Disulfidbrücken sowie die Proline auf ihren jeweiligen
Einfluss auf die Struktur und die thermodynamische Stabilität untersucht. In der vorliegenden
Zusammenfassung werden die Ergebnisse dieser Studien komprimiert präsentiert:
· Charakterisierung des Parvulustat-Wildtyps
Es galt vorweg herauszufinden, wie sich der Inhibitor unter nativen und denaturierten
Bedingungen verhält, um Rückschlüsse auf seine Merkmale und Strukturen zu ziehen. Die
erzielten Ausbeuten und die hohe Reinheit des isolierten Parvulustats erlaubten eine umfassende
Charakterisierung, einschließlich zahlreicher Kristallisationsexperimente, die vermuten lassen,
dass eine Kristallisation möglich sein sollte. Aufgrund der Tatsache, dass die Struktur des
Parvulustats bis 2009 unbekannt war, wurde die sekundäre Struktur mittels Circulardichroismus
und Fluoreszenzspektroskopie untersucht. Die Analyse des fernen UV-CD-Spektrums bei pH 7,0
und 25°C offenbarte eine „all-b-sheet“ Protein-Struktur. Mittels Fluoreszenzspektroskopie wurde
deutlich, dass die aromatischen Aminosäuren exponiert vorliegen. Um zunächst einen Einblick in
die Strukturveränderungen und die thermodynamische Stabilität zu erhalten, wurde der
temperaturinduzierte Entfaltungsübergang mittels CD-Spektroskopie verfolgt. Das Angleichen der
bei 230 nm gemessenen CD-Daten nach der linearen Extrapolations-Methode für eine
Zweizustands-Faltung ergab einen Tm-Wert von 82°C und D H(Tm) von 201,6 kJ/mol. Die
beträchtlichen Werte veranschaulichen die hohe Stabilität des Parvulustats. Eine aus den CDMessungen
bei 50° ergebende Übergangskurve zeigte, dass sich die Sekundärstruktur mit einem
Übergangsmittelpunkt bei 5,62 M GdnHCl kooperativ und reversibel entfaltet. Das Protein
entfaltet sogar infolge einer pH-Wert-Senkung bis auf pH 1 nicht vollständig, sondern es wechselt
direkt in einen Säure-Zustand („acid-state“). Dieser Zustand zeigt spektroskopisch die gleichen
Eigenschaften wie das native Protein, wobei die volle Inhibierungs-Aktivität nicht erhalten bleibt.
In sehr basischem Milieu bei pH 14 nimmt Parvulustat einen alkalisch denaturierten
Zwischenzustand IB an, der sich erheblich von dem GdnHCl-denaturierten, dem säurebehandelten
oder vom „molten globule“ Zustand unterscheidet. Allgemein behielt Parvulustat
über ein breites pH-Wert Spektrum (1,0-10,0) die native Struktur, bzw. eine „native like“ Struktur,
was erneut auf die enorme Stabilität des Proteins hindeutet. Die von Rehm et al. (Rehm et al.,

2009) aufgeworfene Hypothese des „induced fit“ Inhibierungsmechanismus des Parvulustats
konnte durch die in dieser Arbeit durchgeführten Experimenten bekräftigt werden. Mittels
Sekundärstrukturbestimmungen des Parvulustats unter Komplexbildung mit der a-Amylase
konnte eindeutig gezeigt werden, dass strukturelle Veränderungen am Inhibitor im Komplex
vorliegen. Durch zahlreiche Tests konnte festgestellt werden, dass die WRY-Region des
Parvulustats sich der Struktur der a-Amylase anpasst. Die Komplexierung des Parvulustats
bewirkte aber eine thermodynamische Destabilisierung der Inhibitor-Struktur.
· Einfluss des C-Terminus auf die Stabilität des Parvulustats
Um die Ursachen für die hohe Stabilität des Parvulustats auch im Vergleich zu Tendamistat (Tm:
79°C) zu finden, wurde der Einfluss des hoch flexiblen C-Terminus untersucht. Die Derivate mit
um zwei (PL-2AA), vier (PL-4AA) und sieben (PL-7AA) Aminosäuren verkürztem C-Terminus
wurden isoliert und analysiert. Die Entfaltungstemperaturen der verkürzten Derivate des
Parvulustats sinken mit abnehmender Zahl der Aminosäuren. Die Ergebnisse suggerieren, dass
der C-Terminus des Parvulustats eine entscheidende Rolle in der strukturellen Vollständigkeit des
Proteins während der thermischen Entfaltung spielt und damit auch in der Faltung (Tab.: 2.1). Der
Vergleich der Inhibitoraktivitäten der verkürzten Proteine mit dem nativen Parvulustat ergibt für
die Varianten PL-2AA und PL-4AA eine dem Wildtyp ähnliche Aktivität. Die Variante PL-7AA
weist eine leicht verringerte Aktivität auf.

Einfluss hydrophober Oberflächenclustern auf Stabilität und Faltung
des Parvulustats
Parvulustat besitzt in der Mitte des ersten b-Faltblatts, um die Position 22 liegend, einen
hydrophoben Oberflächencluster. Wie mit Hilfe von Substitutionsexperimenten in dieser Region
gezeigt werden konnte, ist die thermodynamische Stabilität des Parvulustats in hohem Maße von
der Bildung dieses kleinen aber wichtigen hydrophoben Kerns bestimmt. Demnach muss das b-
Faltblatt I und das b-Hairpin I eine entscheidende Rolle in der Faltung von Parvulustat spielen.
Position 22 ist in zweierlei Hinsicht für die Stabilität des Parvulustats wichtig: einerseits durch die
energetisch wichtigen Wasserstoffbrückenbindungen und zum zweiten steuert die Stelle zur
Formation des hydrophoben Kerns bei. Die Daten suggerieren, dass es auch eine
thermodynamische Kopplung zwischen dem hydrophoben Effekt und der Präsenz von
Wasserstoffbrückenbindungen geben könnte. Zumindest aus der Sicht der Stabilität ist es
eindeutig, dass interatomare Interaktionen wie Wasserstoffbrückenbindungen, van-der-Waalsund
Dipol-Dipol-Wechselwirkungen notwendig sind, um eine stabile Bildung von b-Faltblättern
in Parvulustat und seinen Derivaten zu verwirklichen.
· Der Effekt der Proline auf die Stabilität des Parvulustats
Der Effekt der Substitution des Prolins durch Alanin an verschiedenen Positionen des Parvulustats
wurde ebenfalls untersucht. Im Ganzen betrachtet führt auch die Mehrfachsubstitution von Prolin
zu keiner nennenswerten strukturellen Veränderung im Parvulustat. Die durchgeführten
thermischen Entfaltungsexperimente bestätigen diese Beobachtungen. Alle Einzelmutanten (P5A,
P42A, P48A, P72A) zeigten eine höhere thermische Stabilität als der Wildtyp (Abb. 2.1).

Die fast gleich bleibenden Tm-Werte bzw. die Erhöhung der Stabilität des Parvulustats sind durch
die strukturelle Fluktuationen zu erklären, denn die rigide XAS-Pro-Bindung wurde durch die
flexible XAS-Ala-Bindung ersetzt.
· Der Einfluss der Disulfidbindung auf die Stabilität des Parvulustats
Der Einfluss der zwei Disulfidbrücken des Parvulustats wurde bezüglich Aktivität, spektraler
Eigenschaften sowie Stabilität untersucht. Hierfür wurden 25 Inhibitorvarianten mittels gezielter
Mutagenese gewonnen, in denen jeweils zwei Cysteinreste, die im natürlich vorkommenden
Parvulustat Disulfidbrücken bilden, durch andere Aminosäuren ersetzt wurden. Die Ergebnisse
zeigten, dass die Faltung Parvulustats ein zwei Zustand Verhalten besitzt, das außer in
Tendamistat in keinem anderen disulfid-verknüpftem Protein gefunden wurde. Dieses Verhalten
wurde durch die Entfernung von Disulfidbrücken nicht beeinflusst. Wie durch die CD- und
fluoreszenzspektroskopischen Experimente belegt werden konnte, ist die native Struktur des
Parvulustats durch die Entfernung der C43-C70-Disulfidbindung tiefgreifend verändert worden.
Passend zu den Veränderungen der Struktur haben die Mutationen schwerwiegende
Destabilisierungseffekte auf das Protein verursacht, was auch an der Erniedrigung der Freien
Gibbs Energie der Denaturierung und der Tm-Werte zu erkennen ist. Die Untersuchung des
Einflusses der Aminosäure-Substitution in den Positionen 43 und 70 auf die thermodynamische
Stabilität des Parvulustats führt zum Ergebnis, dass die Hydrophobizität und Polarität des Restes
70 einen bedeutenden Effekt auf die Stabilität des Proteins besitzt. Die Betrachtung der
thermodynamischen Daten macht deutlich, dass der Beitrag der freien Energie zur Stabilisierung
nicht nur abhängig von der eingeführten Aminosäure ist, sondern zum Teil auch von dem
strukturellen Kontext abhängt. Die Daten zeigen, dass die Substitution des Alanins an der Stelle
70 durch Leucin oder Threonin die Struktur um 0,3 bzw. 1,7 kJ/mol stabilisieren. Zusätzliche
Beiträge zum Unterschied bezüglich des ΔG°-Werts zwischen den Varianten C43AC70L/T und
C43L/TC70A können sich auch durch die unterschiedliche lokale Umgebung, wie z.B. die
Seitenketten von benachbarten Aminosäuren um die zwei Mutationsstellen ergeben. Der Tm-Wert
des Wildtyp-Proteins beträgt 82°C. Die Vergleiche dieser Größe mit der von der stabilsten
Cystein-defizitären Doppelmutante C43AC70T (47,3°C) ergibt eine Differenz von 34,7°C. Dieses
Ergebnis untermauert, dass die Disulfidbindung 2 eine extrem wichtige strukturelle Komponente
in der ungewöhnlich hohen Stabilität des Parvulustats darstellt.
Das Ziel der Arbeit war es dennoch die Daten der Stabilitäten einzelner Disulfidmutanten zu
sammeln und zu erfassen, um dadurch allgemeine Grundregeln für eine rationale Gestaltung der
Elimination beider Disulfidbrücken im Sinne einer vorhersagbaren Auswirkung auf die
Proteinstabilität zu bekommen.

Der Versuch ein disulfidfreies Protein in großen Mengen aus S. lividans zu isolieren, stellte sich
aber als extrem schwierig dar. Nach der Änderung des Stamms des Wirtsorganismus
(Streptomyces lividans TK23), Erhöhung der Qualität der Protoplasten und der Expressions-
Bedingungen (19°C, 150 rpm) konnten auf dem SDS-Gel stärkere Banden des Derivats
C9AC25TC43AC70T-4AA beobachtet werden. Die Expression der Mutante
C9AC25TC43AC70L-4AA konnte hingegen nicht nachgewiesen werden. Die anschließende
Reinigung des Derivats C9AC25TC43AC70T-4AA des Parvulustats mittels Gelfiltration und RPHPLC
bzw. Isolierung des Proteins aus dem SDS-Gel brachte das erwünschte Produkt. Dieses
konnte mit der MALDI-Massenspektroskopie eindeutig nachgewiesen werden. Das aktive
Cystein-freie Derivat C9AC25TC43AC70T-4AA konnte auch auf dem a-Amylase Plattentest
nachgewiesen werden. Diese Ergebnisse zeigen, dass eine Expression und der Nachweis einer
cystein-freien Variante möglich sind, dennoch haben die Ausbeuten dieses Proteins für
weiterreichende Analytik nicht ausgereicht. Demnach sind die Disulfidbrücken für die Stabilität
und Struktur des Parvulustats von enormer Bedeutung dennoch sind sie nicht zwingend
erforderlich für die Aktivität und die Expression in S. lividans.
The display of foreign polypeptides and proteins on the surface of viruses or cells provides an important tool for the engineering of biomolecules and the analysis of their interactions with binding partners. The most extensively used display platform is the coat protein of the filamentous bacteriophage (Smith, 1985). Phage display libraries have often been selected for polypeptides, e.g. single chain (sc) antibodies that bind to a protein of interest, but in vivo selection could only be demonstrated for peptides so far. An alternative display platform is the retrovirus murine leukemia virus (MLV). Here, polypeptides are displayed at the N-terminus of the viral envelope glycoprotein. Proof of principle for this platform was demonstrated for protease substrate libraries, which can be selected through coupling proteolytic activation with viral infectivity (Buchholz et al., 1998). Selection of the library CX4A on living cells resulted in viruses with more than three orders of magnitude improved spreading efficiency through tumor cells (Hartl et al., 2005). Also scAb libraries have recently been displayed and selected using retroviruses (Urban et al., 2005). The library scFvlibxMo displays the repertoire of phage display preselected sc antibodies for laminin-1 binding. The retrovirus based selection process resulted in laminin-specific sc antibodies with improved expression levels in mammalian cells.
This thesis describes the in vivo (i.e. in mouse tumor models) selection of the C-X4-A and scFvlibxMo for tumor homing upon systemic delivery.
For selection of the protease substrate library C-X4-A a subcutaneous tumor was induced in SCID mice followed by three systemic injections of the library. The selection process was monitored over a period of 34 days. After the incubation period mice were sacrificed and virus load in organs and tumor determined. PCR analysis after 34 days showed that virus from the library had preferentially infected the tumor. Sequence analysis showed the selection of protease substrates with the most prominent one with a frequency of over 65%. The four most prominent protease substrate variants where reconstituted into the original viral backbone for further investigation (C-SK-A, C-HI-A, C-HM-A and C-HS-A). Interestingly, these viruses exhibited a reduced spreading capacity in vitro on HT1080 cells as compared to the C-AK-A virus, which had previously been selected on HT1080 cells. When assayed for tumor homing, however, viruses C-HI-A and C-HS-A had clearly improved in comparison to C-AK-A. Tumor tissue had been infected at rates of over 55% while virus load of extratumoral organs was very low (infection rates <0.7 for C-HS-A and <0.02 for C-HI-A). Tumor targeting capacity had thus been improved over 10-fold by the in vivo selection of the C-X4-A library.
The experimental set up for the in vivo selection of the scFvlibxMo library was performed according to that of the C-X4-A library. Fingerprint analysis of the selected viruses that infected tumor tissue resulted in the identification of seven antibody variants showing unique CDR3 sequences. Two prominent clones (M49T-A and M49T-B) were cloned back into the MoMLV genome for further analysis of the reconstituted viruses. While variant B bound laminin-1 efficiently, variant A was unable to do so, although it was selected at highest frequency (76%). Both reconstituted viruses were equally well infectious and spread through HT1080rec1 cells at a similar efficiency as MoMLV. In an in vivo competition experiment the selected viruses clearly out-competed a laminin-1 binding reference virus L36xMo for tumor homing. To understand the molecular driving forces behind the in vivo selection process the epitope of the selected scFv M49T-A was identified using a phage peptide library approach. In silico analysis led to the identification of a small group of possible antigens, including tenascin, fibronectin and collagen.
The data described in this thesis demonstrate that the retrovirus display platform is capable of allowing the in vivo selection of protease substrates and scFvs. Notably, the replication competence of the system introduced an additional level of complexity to the library. The performed in vivo selections significantly enhanced tumor tropism. Selective infection of tumor cells combined with transfer of anti-tumoral genes is an attractive strategy for cancer therapy being in focus of current research. The viruses selected in this thesis build prime candidates for targeted retrovirus based tumor therapy.
Sucrose- and H+-dependent charge movements associated with the gating of sucrose transporter ZmSUT1

Background: In contrast to man the majority of higher plants use sucrose as mobile carbohydrate. Accordingly proton-driven sucrose transporters are crucial for cell-to-cell and long-distance distribution within the plant body. Generally very negative plant membrane potentials and the ability to accumulate sucrose quantities of more than 1 M document that plants must have evolved transporters with unique structural and functional features.
Methodology/Principal Findings: To unravel the functional properties of one specific high capacity plasma membrane sucrose transporter in detail, we expressed the sucrose/H+ co-transporter from maize ZmSUT1 in Xenopus oocytes. Application of sucrose in an acidic pH environment elicited inward proton currents. Interestingly the sucrose-dependent H+ transport was associated with a decrease in membrane capacitance (Cm). In addition to sucrose Cm was modulated by the membrane potential and external protons. In order to explore the molecular mechanism underlying these Cm changes, presteady-state currents (Ipre) of ZmSUT1 transport were analyzed. Decay of Ipre could be best fitted by double exponentials. When plotted against the voltage the charge Q, associated to Ipre, was dependent on sucrose and protons. The mathematical derivative of the charge Q versus voltage was well in line with the observed Cm changes. Based on these parameters a turnover rate of 500 molecules sucrose/s was calculated. In contrast to gating currents of voltage dependent-potassium channels the analysis of ZmSUT1-derived presteady-state currents in the absence of sucrose (I = Q/τ) was sufficient to predict ZmSUT1 transport-associated currents.
Conclusions: Taken together our results indicate that in the absence of sucrose, ‘trapped’ protons move back and forth between an outer and an inner site within the transmembrane domains of ZmSUT1. This movement of protons in the electric field of the membrane gives rise to the presteady-state currents and in turn to Cm changes. Upon application of external sucrose, protons can pass the membrane turning presteady-state into transport currents.
Das extrem thermophile Eubakterium Thermus thermophilus ist in den letzten Jahren zu einem Modell für thermophile Organismen geworden und verdankt seinen Statuszum Teil seiner hohe Wachstumsrate, den guten Zellerträgen und der konstitutivenExpression eines natürlichen Kompetenzapparates, der seine genetische Manipulation ermöglicht. Die Verfügbarkeit von kompatiblen Plasmiden und bis zu vier thermostabilen Antibiotikaresistenzmarkern konnten den Wert des Organismus in Hinblick auf biotechnologische Anwendungen noch weiter steigern und tatsächlich besteht nach wie vor ein ungebrochenes Interesse an der Struktur- und Funktionsaufklärung thermophiler Proteine. Der Focus der hier vorliegenden Arbeit richtete sich auf eine der insgesamt zwei terminalen Oxidasen der Atmungskette von T. thermophilus, die Cytochrom ba3 Oxidase. Es wurden verschiedene rekombinante Varianten des Proteins, die sich hinsichtlich der Position und Länge des verwendeten Histidin-Tags unterschieden, kloniert, exprimiert und aufgereinigt. Das Einfügen eines internen His12-Tags in einen periplasmatischen Loop zwischen den Transmembranhelices IV und V führte zu einer rekombinanten Version der Oxidase, die in ihren Eigenschaften dem nativen Wildtyp entsprach und sich durch die in dieser Arbeit etablierte Aufreinigungsstrategie relativ schnell, in guten Ausbeuten und hoher Reinheit aufreinigen ließ. Weiterhin konnten verschiedene Punktmutationen von möglicherweise am Elektronentransfer beteiligten Aminosäureresten generiert und die resultierenden Proteine aufgereinigt und über ihre enzymatische Aktivität charakterisiert werden. Eine weiterführende Charakterisierung der Mutanten erfolgte im Rahmen einer Kooperation und ist bisher noch nicht abgeschlossen. Das Herzstück dieser Arbeit machte jedoch die Definition der Transkriptionseinheit der Cytochrom ba3 Oxidase und die sich daraus ergebenden Fragestellungen aus. So konnte gezeigt werden, dass das ba3 Operon zusätzlich zu den Strukturgenen der dort kodierten Untereinheiten noch mindestens ein, höchst wahrscheinlich jedoch zwei zusätzliche Gene enthält: cbaX und cbaY. Bioinformatische Charakterisierungen ordneten CbaY der diversen Gruppe von sekundären Transportern zu, und es konnte experimentell gezeigt werden, dass seine Anwesenheit für die Expression der Cytochrom ba3 Oxidase förderlich ist. CbaX hingegen konnte über die durchgeführte Homologiesuche keine Funktion zugeordnet werden; uncharakterisierte Homologe waren einzig in der Thermaceae Gruppe zu finden. Durch Deletions- und Komplementationsstudien konnte dem Protein eine entscheidende Rolle in der Assemblierung der ba3 Oxidase bescheinigt werden. CbaX scheint eine zentrale Aufgabe bei der Häm a Insertion in Untereinheit I zu spielen und könnte, ohne Sequenzhomologie aufzuweisen, die Rolle des Surf1-Proteins übernehmen, welches in den sequenzierten Organismen der Thermaceae Gruppe nicht konserviert ist. Die homologe Expression und Aufreinigung von CbaX führte nicht zur erwarteten Ausbeute und Reinheit des Proteins, konnte aber durch immunologische Experimente eine potentielle Interaktion von CbaX und der Cytochrom ba3 Oxidase nachweisen.
Background: ClC-7 is a ubiquitous transporter which is broadly expressed in mammalian tissues. It is implied in the pathogenesis of lysosomal storage disease and osteopetrosis. Because of its endosomal/lysosomal localization it is still poorly characterized. Methodology/Principal Findings: An electrophysiological characterization of rat ClC-7 using solid-supported membrane-based electrophysiology is presented. The measured currents show the characteristics of ClC-7 and confirm its function as a Cl−/H+-antiporter. We have used rat ClC-7 in CHO cells as a model system to investigate the functionality and cellular localization of the wt transporter and its variant G213R ClC-7 which is the analogue of human G215R ClC-7 responsible for autosomal dominant osteopetrosis type II. Our study shows that rat G213R ClC-7 is functional but has a localization defect in CHO cells which prevents it from being correctly targeted to the lysosomal membrane. The electrophysiological assay is tested as a tool for drug discovery. The assay is validated with a number of drug candidates. It is shown that ClC-7 is inhibited by DIDS, NPPB and NS5818 at micromolar concentrations. Conclusions/Significance: It is suggested that the scenario found in the CHO model system also applies to the human transporter and that mislocalization rather than impaired functionality of G215R ClC-7 is the primary cause of the related autosomal dominant osteopetrosis type II. Furthermore, the robust solid-supported membrane-based electrophysiological assay is proposed for rapid screening for potential ClC-7 inhibitors which are discussed for treatment of osteoporosis.
The continuous progress in the structural and functional characterization of aquaporins increasingly attracts attention to study their roles in certain mammalian diseases. Although several structures of aquaporins have already been solved by crystallization, the challenge of producing sufficient amounts of functional proteins still remains. CF (cell free) expression has emerged in recent times as a promising alternative option in order to synthesize large quantities of membrane proteins, and the focus of this report was to evaluate the potential of this technique for the production of eukaryotic aquaporins. We have selected the mouse aquaporin 4 as a representative of mammalian aquaporins. The protein was synthesized in an E. coli extract based cell-free system with two different expression modes, and the efficiencies of two modes were compared. In both, the P-CF (cell-free membrane protein expression as precipitate) mode generating initial aquaporin precipitates as well as in the D-CF (cell-free membrane protein expression in presence of detergent) mode, generating directly detergent solubilized samples, we were able to obtain mg amounts of protein per ml of cell-free reaction. Purified aquaporin samples solubilized in different detergents were reconstituted into liposomes, and analyzed for the water channel activity. The calculated Pf value of proteoliposome samples isolated from the D-CF mode was 133 µm/s at 10°C, which was 5 times higher as that of the control. A reversible inhibitory effect of mercury chloride was observed, which is consistent with previous observations of in vitro reconstituted aquaporin 4. In this study, a fast and convenient protocol was established for functional expression of aquaporins, which could serve as basis for further applications such as water filtration.
Structured RNA regions are important gene control elements in prokaryotes and eukaryotes. Here, we show that the mRNA of a cyanobacterial heat shock gene contains a built-in thermosensor critical for photosynthetic activity under stress conditions. The exceptionally short 5´-untranslated region is comprised of a single hairpin with an internal asymmetric loop. It inhibits translation of the Synechocystis hsp17 transcript at normal growth conditions, permits translation initiation under stress conditions and shuts down Hsp17 production in the recovery phase. Point mutations that stabilized or destabilized the RNA structure deregulated reporter gene expression in vivo and ribosome binding in vitro. Introduction of such point mutations into the Synechocystis genome produced severe phenotypic defects. Reversible formation of the open and closed structure was beneficial for viability, integrity of the photosystem and oxygen evolution. Continuous production of Hsp17 was detrimental when the stress declined indicating that shutting-off heat shock protein production is an important, previously unrecognized function of RNA thermometers. We discovered a simple biosensor that strictly adjusts the cellular level of a molecular chaperone to the physiological need.
Employing NMR spectroscopy, it is not only possible to calculate the three dimensional structures of single proteins, but also to study dynamics and conformational changes of protein-complexes. In fact that is an important aspect, since the protein function depends on dynamics and interactions with other molecules. Therefore the study of protein-protein interactions is of highest importance for a better understanding of biological processes. Based on NMR methods, in this thesis we were able to determine protein-protein interactions within the enterobacterial Rcs signalling complex which is regulated via a phosphorelay. Originally identified as regulator of capsule synthesis, the Rcs phosphorelay is now considered to be implicated in stress response caused by disturbances in the peptidoglycan layer. Beyond that the Rcs system is involved in multiplex transcriptional networks including cell division, motility, biofilm formation and virulence. Because of such global nature and its extraordinary structural organisation involving membrane integrated sensor proteins (RcsC, RcsD), coactivators (RcsF, RcsA) and a transcription factor (RcsB), the Rcs system is one of the most remarkable phosphorelays in the family of enterobacteriacaea. During the complex phosphotransfer the histidine phosphotransferase (HPt) domain of the intermediary RcsD protein mediates the phosphotransfer between RcsC and RcsB, and probably modulates the phosphorylation state of the response regulator RcsB. Therefore the present work has been focused on the interface between RcsD and RcsB in more detail. In the first part of the thesis a new domain within the RcsD protein has been identified and structurally analysed by liquid NMR spectroscopy. RcsD is an inner membrane bound hybrid sensor like-kinase composed of a periplasmic sensor domain and a cytoplasmic portion. The cytoplasmic part contains the histidine like-kinase (HK) domain and the histidine phosphotransferase (HPt) domain. By analysis of the secondary structure in more detail, it was shown here that the two domains are intermitted by an additional 13.3 kDa domain. Corresponding to the position of the ABL (α−β−loop) domain of RcsC, located C-terminal to the RcsC-HK domain, the new identified domain was named RcsD-ABL. The central structural element of RcsD-ABL is a β-sheet composed of six strands with a β1−β2−β3−β4−β6−β5 topology and surrounded by two α-helices α1 and α2. In the second part of the thesis, RcsD-ABL is identified as a binding domain for the response regulator RcsB by NMR titration experiments. Such a binding domain for a response regulator has so far only been described for the histidine kinase CheA. In reportergene assays with β-galactosidase and ONPG as substrate it was shown that overexpression of RcsD-ABL in high amounts inhibited binding of RcsB to its target promoter. The β-galactosidase activity was reduced by 80 % with respect to cells carrying no plasmid encoding RcsD-ABL. The mapping of the binding interface was successfully achieved by chemical shift perturbations, a fast mapping protocol and selective labelling. It was shown that the interaction between RcsD-ABL and RcsB takes place via a binding interface comprising mainly the two α-helices of RcsD-ABL and the α-helices α7, α8 and α10 in the effector domain of RcsB. In the third part of the thesis, the interaction of RcsB with RcsD-ABL was related to that with RcsD-HPt. Using NMR titration experiments and ITC measurements, a comparison of the binding constants (Kd) of RcsB interacting either with the isolated RcsD-ABL (2 PM) or the isolated RcsDHPt domain (40 PM) revealed a higher affinity of RcsD-ABL to RcsB. A conjugate of RcsD-ABL-HPt interacting with RcsB decreased the Kd in the one-site fitting mode to 10 PM. However, the two-site fitting mode applied for RcsD-ABL-HPt/RcsB interaction resulted in a Kd (RcsD-ABL) of 2 PM and a Kd (RcsD-HPt) of 8 PM, indicating that RcsD-ABL enhances the binding of RcsD-HPt to RcsB. In the last part of the thesis, it was partly possible together with the data obtained from NMR titration experiments, PRE measurements and a HADDOCK protocol to develop a geometrical model for the interaction of RcsD with RcsB. In this model the receiver domain of RcsB interacts with the RcsD-HPt domain and the RcsB effector domain interacts with the RcsD-ABL domain. These results lead to surprising insights on the regulation of phosphorelays, since normally the effector domain binds to DNA. Here the effector domain is recognized by the newly identified RcsD-ABL domain. Prospectively, further investigations of phosphorylation affects and mutational studies will be of great interest.
Neue Synthesestrategien als Thema des Chemieunterrichts - Experimente zur Kombinatorischen Chemie

In der vorliegenden Dissertation wurde die experimentelle Umsetzung der Thematik Kombinatorische Chemie für den Chemieunterricht in der Sekundarstufe II ausgearbeitet. Dazu wurden geeignete Experimente erarbeitet, die einen Einblick in die Kombinatorische Chemie und somit in die neuen Arbeitmethoden der heutigen Chemie schaffen. Mit deren Hilfe gängige Synthesemethoden wie die Split and Combine-Synthese, Teabag-Methode, Spot-Methode und Pin-Methode aufgezeigt werden können und von den Schülern selbst durchgeführt werden. Als ein vorteilhaftes Modell zum Aufzeigen der Arbeitsweise der Kombinatorischen Chemie haben sich Farbstoffe erwiesen, da sie schnell darstellbar sind und keiner aufwendigen Detektion der entstandenen Produkte bedürfen. Als erstes Farbstoffsystem wurden Azofarbstoffe ausgewählt, da sie auch im fakultativen Themenbereich „Farben“ im Chemieunterricht thematisiert werden. Die Übertragung der normalerweise in Lösung hergestellten Azofarbstoffe auf die Festphasenchemie musste zuerst eine geeignete und kostengünstige Festphase gefunden werden. Hierbei haben sich im Handel erhältliche Ionenaustauscher (Harze ohne Linker), die auch zur Wasseraufbereitung verwendet werden, als geeignet herausgestellt. Die erforderlichen Reaktionsbedingungen der Lösungsreaktionen wurden erprobt, optimiert und auf die Festphasenchemie übertragen. Eine Übertragung der Syntheseprinzipien auf drei weitere Farbstoffsysteme wie der Fluoreszenzfarbstoffe der Pyrydin- und Pyrylium-Reihe sowie der Oxidationsfarbstoffe schloß sich an. Nunmehr stehen für den schulischen Einsatz, aber auch für die Ausbildung, einfache Systeme zur Verfügung, die anhand schnell und problemlos durchzuführender Experimente einen Zugang zu unterschiedlichen Methoden der Kombinatorischen Chemie vermitteln können. Dabei bieten die Experimente Anregungen für weitere Arbeiten. Als problematisch stellte sich jedoch der Zugang zu mehrstufigen kombinatorischen Synthesen auf dem Gebiet der Farbstoffe dar. Hier stößt man mit den aus didaktischen Überlegungen sehr einfach gewählten Beispielen an eine Grenze. Ergänzend wurde die Thematik Kombinatorische Chemie mit hochbegabten Schülern der Internatsschule Schloß Hansenberg und interessierten Schülern aus dem Raum Frankfurt, die an dem Projekt Science Camp teilnahmen, erprobt und das Verständnis für diese Thematik mittels eines Fragebogens untersucht. Ein direkter Vergleich beider Schülergruppen konnte aufgrund unterschiedlicher schulischer Bildung nicht vorgenommen werden, weshalb die Auswertung lediglich erste Eindrücke vermittelt. Danach ist das bisher ausgearbeitete Konzept in Lerngruppen mit hochbegabten Schülern gut einsetzbar. Ein Vergleich mit durchschnittlich leistungsstarken Lerngruppen steht noch aus. Eine wünschenswerte Erweiterung und ein Ausbau der Thematik aus Sicht der Schulchemie sollte anorganische Systeme mit einbeziehen und Strategien der Dekonvolution.
Qualität und Qualitätsmanagement in der universitären naturwissenschaftlichen Lehrerfortbildung

Vor dem Hintergrund der politischen Entwicklungen in Hessen und der allgemeinen Debatte über Qualität im Bildungsbereich sollte im Lehrerfortbildungszentrum Chemie (lfbz-Chemie) der Universität Frankfurt am Main eine systematische Qualitätsentwicklung etabliert und dieses Vorhaben wissenschatflich begleitet werden. Die wissenschaftliche Arbeit besteht aus drei Säulen: - Eine erste, qualitative Studie sollte Qualitätskriterien und -indikatoren liefern. - In einer Fallstudie am lfbz-Chemie Frankfurt sollte die Einführung von Instrumenten des Qualitätsmanagements aus dem gewerblichen Weiterbildungsbereich beobachtet und bewertet werden. - Eine bundesweite Umfrage unter universitären Fortbildungsanbietern im naturwissenschaftlichen Bereich sollte sowohl die Struktur der anbietenden Institutionen als auch die allgemeine Einstellung der Befragten zum professionellen Qualitätsmanagement (QM) nach einem im gewerblichen Bereich geläufigen Modell (LQW) beleuchten. Die Qualitätsindikatoren und -kriterien fielen sehr spezifisch für die naturwissenschaftliche Lehrerfortbildung aus. Allein die drei folgenden Bereiche erfassen zwei Drittel aller Nennungen: die „Qualität der Teamer bzw. Moderatoren“, die „Fortbildungsgestaltung“ und die „Zielgruppenorientierung (Schulbezug)“. Nimmt man die Anzahl der Indikatoren und Kriterien pro Bereich als Maßstab, fokussieren die Befragten sehr stark auf Anforderungen, die die Professionalität des Personals betreffen. Zur Orientierung für die Qualitätsarbeit am lfbz-Chemie (Fallstudie) wurde das QM-Modell LQW 2 gewählt; als kleinere Instrumente kamen die Tabellen nach dem Vorbild der Balanced Scorecard (BSC) nach Kaplan und Norton sowie die Stärken-Schwächen-Analyse zum Einsatz. Als erstes Ergebnis der Fallstudie kristallisierten sich drei Arbeitsbereiche heraus: Personalorganisation, Evaluation und Innovationen. Diese Arbeitsfelder ergänzten die identifizierten Anforderungen an Fortbildung somit um Bereiche, die eher der organisatorischen Ebene zuzuordnen sind. Es ließ sich in der Fallstudie feststellen, dass ein ganzes Qualitätsmanagementmodell wie LQW 2 nur als Anregung für den einen oder anderen, als wichtig erachteten Bereich dienen konnte, während sich kleinere Instrumente eher als handhabbar erwiesen. Außerdem konnte postuliert werden, dass - ein persönlicheres Eingehen auf die Ziele und Aufgaben der einzelnen Personen im Sinne eines Total Quality Managements vermutlich zu besseren Ergebnissen geführt hätte als die Konzentration auf allgemeine Themen und die Orientierung am eher abstrakten Modell, - Erfolge sehr nachdrücklich kommuniziert werden müssen und - möglichst eine Person aus dem Stammpersonal für den Bereich Qualität verantwortlich sein sollte. Um ergänzend zur Fallstudie zu allgemeineren Schlüssen gelangen zu können, wurde eine Charakterisierung der universitären Fortbildungsanbieter mittels Clusteranalyse mit Daten der bundesweiten Umfrage durchgeführt. Sie zeigte vier Typen von Anbietern auf. Insgesamt fanden sich neben der Gruppe der großen Anbieter, zu denen auch das lfbz-Chemie gehört, nur vergleichsweise kleine Anbieter. Die in der Fallstudie erhaltenen Ergebnisse können somit nur eingeschränkt auf die meisten Anbieter übertragen werden. Insgesamt konnten Stärken und Schwächen der universitären Anbieter identifiziert werden, mit denen sich die universitäre naturwissenschaftliche Lehrerfortbildung von der gewerblichen Weiterbildung abhebt. Stärken lagen vor allem in den besonderen Kompetenzen, die an der Hochschule anzutreffen sind. Problemfelder lagen ausgerechnet hauptsächlich im Personalbereich. Als spezifische Schwächen der universitären Anbieter im Personalbereich kann z. B. die Stellung der Fortbildung als Nebenaufgabe oder der Verlust von Know-how durch die Fluktuation des Personals gelten. Auch allgemein kann ein Modell wie LQW 2 als Anregung für die Qualitätsarbeit in der universitären Lehrerfortbildung dienen, nicht jedoch umfassend umgesetzt werden. Die „kleineren“ Instrumente können dagegen eher als allgemein geeignet betrachtet werden. Während die „großen“ Anbieter durch verbesserte personelle Ressourcen für Aufgaben wie die Qualitätsarbeit unterstützt werden könnten, bietet sich für die „kleinen“ Anbieter eher die Unterstützung durch eine zentrale Stelle der Universität an, um die Fortbildungsanbieter doch noch in ein professionelles Qualitätsmanagement einbinden zu können.
The title compound. C15H14N2O4, (I), has a gauche–gauche (O/C/C/C—O/C/C/C or GG) conformation and is a positional isomer of propane-1,3-diyl bis(pyridine-3-carboxylate), (II). The molecule of (I) lies on a twofold rotation axis, which passes through the central C atom of the aliphatic chain, giving one half-molecule per asymmetric unit. There is excellent agreement of the geometric parameters of (I) and (II). The most obvious differences between them are the O/C/C/C—O/C/C/C torsion angles [56.6 (2)° in (I) and 174.0 (3)/70.2 (3)° in (II) for GG and TG conformations, respectively] and the dihedral angle between the planes of the aromatic rings [80.3 (10)° in (I) and 76.5 (3)° in (II)]. The crystal structure is stabilized by weak C—H ... N and C—H ... O hydrogen bonding.

In the title compound, C17H11NO4, the dihedral angle between the two benzene rings is 8.66 (3)°. The nitro group is twisted by 4.51 (9)° out of the plane of the aromatic ring to which it is attached. The presence of intermolecular C—H ... O contacts in the crystal structure leads to the formation of chains along the c axis.
In the crystal of the title compound [systematic name: 2-(3,5-diamino-6-chloropyrazin-2-ylcarbonyl)guanidinium chloride methanol disolvate], C6H9ClN7O+·Cl-·2CH3OH , the components are connected by N—H ... N, N—H ... Cl, N—H ... O, O—H ... Cl and O—H ... O hydrogen bonds into a three-dimensional network. The dihedral angle between the aromatic ring and the guanidine residue is 6.0 (2)°.
In contrast to the previous structure determinations of the title structure, (NH4)2[MoS4], the present determination at 173 K localized the positions of the H atoms. The title structure belongs to the beta-K2SO4 family and all the ions are located on crystallographic mirror planes. The ions are held together by N—H ... S hydrogen bonds (some of which are bifurcated), forming a three-dimensional network. One of the N atoms has nine contacts to the S atoms shorter than 4 Å, and the other has ten.
The title compound (also know as azorellanone), C20H32O2, is built up from three fused carbocycles, one five-membered ring and two six-membered rings. The five membered-ring has an envelope conformation, whereas the six-membered rings have a distorted half-chair and a twist–boat conformation. In the crystal, molecules are linked by O—H ... O interactions into zigzag chains with graph-set notation C(8) along [010]. The absolute configuration was assigned on the basis of earlier chemical studies.
The dihydropyrimidine ring of the title compound, C13H15ClN2S, adopts an envelope conformation with five almost coplanar atoms (r.m.s. deviation = 0.054 Å) and the C atom bearing the two methyl substituents deviating from this plane by 0.441 (2) Å. The best plane through the five almost coplanar atoms forms a dihedral angle of 89.56 (5)° with the benzene ring. The crystal packing is characterized by centrosymmetric dimers connected by pairs of N—H ... S hydrogen bonds.

The title compound, C12H8BBr, crystallizes with three essentially planar molecules (r.m.s. deviations = 0.018, 0.020 and 0.021Å) in the asymmetric unit: since the title compound is rigid, there are no conformational differences between these three molecules. The crystal packing resembles a herringbone pattern.
The title compound, C22H28N2O6, crystallizes with four half-molecules in the asymmetric unit: each molecule is located about a crystallographic inversion centre. The central methylene groups of two molecules are disordered over two sets of equally occupied sites. The crystal packing is characterized by sheets of molecules parallel to (114).
In the title compound, C27H19N3O4, the phenol and pyrazole rings are almost coplanar [dihedral angle = 0.95 (12)°] due to an intramolecular O—H ... N hydrogen bond, whereas the phenyl ring is tilted by 40.81 (7)° with respect to the plane of the pyrazole ring. The aromatic ring with a nitrophenoxy substituent makes a dihedral angle of 54.10 (7)° with the pyrazole ring.
The title compound, C15H14N2O4, has a trans–gauche [O/C/C/C–O/C/C/C] (TG) conformation. The angle between the planes of aromatic rings is 76.4 (3)°. The crystal structure is stabilized by van der Waals interactions and C—H ... O hydrogen bonds. The crystal used was a non-merohedral twin with a fractional contribution of the minor component of 0.443 (5).

The biguanidium group is not planar [dihedral angle between the two CN3 groups = 52.0 (1)°] and is rotated with respect to the phenyl group [tau = 54.3 (3)°]. In the crystal, N—H ... N hydrogen-bonded centrosymmetric dimers are connected into ribbons, which are further stabilized by N—H ... Cl interactions, forming a three-dimensional hydrogen-bonded network.
The title compound, [Na(CF3O3S)(C12H24O6)], features a sodium cation that is coordinated by eight O atoms in an irregular hexagonal bipyramidal environment. The equatorial positions are occupied by the six O atoms of an 18-crown-6 ether ring. In the axial positions, there is one O atom of a trifluoromethanesulfonate anion and an ether O atom of a symmetry-equivalent crown ether ring. In this way, centrosymmetric dimers are formed.
The asymmetric unit of the title compound, [K(C5HF6N2)(H2O)2]n, is composed of two 3,5-bis(trifluoromethyl)pyrazolide anions, two potassium cations and four water molecules. The water molecules and 3,5-bis(trifluoromethyl)pyrazolide anions act as bridges between the potassium cations. Each potassium cation is surrounded by four O atoms [K—O = 2.705 (3)–2.767 (3) Å] and four F atoms [K—F = 2.870 (7)–3.215 (13) Å]. The water molecules and the 3,5-bis(trifluoromethyl)pyrazolide anions are connected by O—H ... N hydrogen bonds, forming layers in the ab plane. All –CF3 groups show rotational disorder between two orientations each.
The two rings in the title compound, C11H12N2O4S, are roughly coplanar [dihedral angle = 6.77 (8)°]. Whereas the two outer methyl groups of the three methoxy groups are almost coplanar with the aromatic ring to which they are attached [C—C—O—C torsion angles = 8.5 (3) and -8.3 (3)°], the methyl group of the central methoxy substituent is not [C—C—C—C = -78.4 (3)°]. The crystal packing is stabilized by N—H ... O hydrogen bonding.
In the title compound, C11H11N3O2, the dihedral angle between the central ethanone fragment and the 4-methoxyphenyl group is 2.9 (2)°, while that between the ethanone fragment and the triazole ring is 83.4 (2)°. The dihedral angle between the planes of the triazole and benzene rings is 81.7 (1)°. The 4-methoxyphenyl group is cis with respect to the ethanone fragment O atom across the exocyclic C—C bond. In the crystal, molecules are linked by C—H ... N interactions into C(9) chains along [001].
The central structural element of the title compound, C24H29NO2, is a carbazole unit substituted with two acetyl residues and an octyl chain. The title compound, [systematic name: 5a-acetoxymethyl-3-isopropyl-8-methyl-1,2,3,3a,4,5,5a,6,7,10,10a,10b-dodecahydro-7,10-endo-epidioxycyclohepta[e]indene-3a-carboxylic acid], C22H32O6 (I), is closely related to methyl 5a-acetoxymethyl-3-isopropyl-8-methyl-1,2,3,3a,4,5,5a,6,7,10,10a,10b-dodecahydro-7,10-endo-epidioxycyclohepta[e]indene-3a-carboxylate, (II) [Brito et al., (2008 [triangle]). Acta Cryst. E64, o1209]. There are two molecules in the asymmetric unit, which are linked by two strong intramolecular O—H ... O hydrogen bonds with graph-set motif R 2 2(8). In both (I) and (II), the conformation of the three fused rings are almost identical. The five-membered ring has an envelope conformation, the six-membered ring has a chair conformation and the seven-membered ring has a boat conformation. The most obvious differences between the two compounds is the observed disorder of the acetoxymethyl fragments in both molecules of the asymmetric unit of (I). This disorder is not observed in (II). The crystal structure and the molecular conformation is stabilized by intermolecular C—H ... O hydrogen bonds. The ability to form hydrogen bonds is different in the two compounds. The crystal studied was a non-merohedral twin, the ratio of the twin components being 0.28 (1):0.72 (1)
In the title compound, C4H7N3O·C2H6OS, creatinine [2-amino-1-methyl-1H-imidazol-4(5H)one] exists in the amine form. The ring is planar (r.m.s. deviation for all non-H atoms = 0.017 Å). In the crystal, two creatinine molecules form centrosymmetric hydrogen-bonded dimers linked by pairs of N—H[cdots, three dots, centered]N hydrogen bonds. In addition, creatinine is linked to a dimethyl sulfoxide molecule by an N—H[cdots, three dots, centered]O interaction. The packing shows layers parallel to (120).
The title compound, [Li3(C4F9O)3(C3H6O)3], features an open Li/O cube with an Li ion missing at one corner. Three of the four bridging O atoms of the cube carry a fluorinated tert-butyl residue, whereas the fourth is part of an acetone molecule. Two of the Li atoms are further bonded to a non-bridging acetone molecule. Two of the lithium ion coordination geometries are very distorted LiO4 tetrahedra; the third could be described as a very distorted LiO3 T-shape with two distant F-atom neighbours. The Li[cdots, three dots, centered]Li contact distances for the three-coordinate Li+ ion [2.608 (14) and 2.631 (12) Å] are much shorter that the contact distance [2.940 (13) Å] between the tetrahedrally coordinated species.

The analysis of biomolecular macrocomplexes requires certain preconditions to be fulfilled. The preparation of biomolecular samples usually results in low yields. Due to this constraint of low availability any method should provide a sufficient sensitivity to cope with typical sample amounts. Biomolecules also often show a reduced stability, i.e. a propensity for fragmentation upon ionisation, which requires reasonable soft methods for the investigation. Furthermore macromolecular complexes usually are composed by means of non-covalent interactions presenting additional demands on the softness. This holds true for specific complexes like protein-ligand or DNA double strand binding. For the formation of non-covalent, specific complexes the biomolecules’ native structure and environment are a basic prerequisite and hence crucial. Therefore it is desirable during analysis to keep the biomolecules in a native environment to preserve their structure and weak interactions. One suitable method for analysing biomolecules is mass spectrometry. Mass spectrometry is capable of high throughput screening as well as determining masses with high accuracy and high sensitivity. Especially since the availability of MALDI-MS and ESI-MS mass spectrometry evolved to a versatile tool to investigate biomolecular complexes. Both, MALDI- and ESI-MS are sufficiently soft methods to observe fragile biomolecules. Yet both methods have their advantages and disadvantages. During the recent years an alternative mass spectrometric approach has been developed in our group, termed LILBID-MS (Laser Induced Liquid Bead Ionisation/Desorption). In LILBID microdroplets of aqueous solution containing buffer, salt and further additives among the analyte molecules are injected into vacuum and irradiated one-by-one by mid-IR laser pulses. The absorption of the energy by the water leads to a rapid ablation of the preformed analyte ions. LILBID is highly tolerant for the addition of salts and detergents allowing to study biomolecular complexes in a native environment. As LILBID-MS is soft enough to avoid fragmentation, specific non-covalent complexes can be analysed directly from their native environment by this method. In addition dissociation can be induced on demand by increasing the laser intensity which allows for the study of subunit compositions. A further prominent property of LILBID is the possibility to study hydrophobic membrane proteins due to the tolerated use of detergents. During the course of this work, several instrumental improvements mostly concerning ion focussing and beam steering were introduced. Together with refinements of different modes of measurement the result is a significantly improved signal-to-noise ratio as well as a further improvement in sensitivity. In addition the accessible m/z range for a given flight time has been vastly increased. The new possibilities that LILBID now offers for the study of biomolecular complexes were investigated. The ability to detect specific binding in LILBID-MS was investigated by means of nucleic acids and their interaction with proteins. It could be shown that the stability of a 16bp dsDNA corresponds to that in solution phase regarding the dependency on concentration and type of the salts used. In addition a competitive experiment with the well-known transcription factor p50 was used to demonstrate the detection of sequence-specific binding with LILBID. The improved sensitivity allowed to detect single stranded DNA at nanomolar concentrations and even the 2686bp plasmid pUC19 could be easily detected without fragmentation using a concentration of only 80nM. In case of the transcription factor p63 the mass spectrometric analysis could help to identify a new model of activation and inhibition. For the first time known quarternary structures of membrane proteins like the light-driven proton pump bacteriorhodopsin and the potassium channel KcsA could be detected with mass spectrometry. For the light-driven proton pump proteorhodopsin the type and the concentration of the used detergents significantly influenced the stability of this protein as well as the preferred quarternary structure.
Die Hämophile ist eine X-chromosomal vererbbare Blutungserkrankung, die durch einen funktionellen Mangel der Gerinnungsaktoren Faktor VIII oder Faktor IX im Fall der Hämo-philie A oder Hämophilie B hervorgerufen wird. Der niedrige therapeutische Schwellenwert in der Behandlung der Hämophilie (Faktorenspiegel >1%) macht diese Krankheit zu einem idealen Zielobjekt in der Gentherapie. Eine schwerwiegende Komplikation in der Behandlung der Hämophilen besteht in der Entwicklung von neutralisierenden Antikörpern gegen das the-rapeutisch substituierte Protein. Akute Blutungen in solchen Hemmkörper-Patienten werden derzeit durch FVIII- und FIX-unabhängigen Bypass-Agenzien kontrolliert. Im Rahmen dieser Arbeit wurde nach einer erfolgten Optimierung eines nicht-viralen Ansatzes zur Transgenex-pression von FIX in der Leber der Maus dieses Gentransfersystem dazu verwendet, um eine Entwicklung von neuen, auf FIX basierenden, FVIII-Bypass-Strategien in vivo zu ermöglichen. Im ersten Teil der vorliegenden Arbeit wurden Plasmidvektoren und Vektorrückgrat-freie Minicircle-Konstrukte auf ihre Effizienz zur Expression des FIX-Transgens nach hydrodyna-misch basiertem Gentransfer in die Leber der Maus untersucht. Der Gentransfer der Mini-circle führte hierbei zu einer höheren Transgenexpression und einem geringeren Abfall der FIX-Antigen-Spiegel über die Zeit. Darüber hinaus war das FIX-Protein selbst bei den höchs-ten Expressionsleveln (700% des physiologischen Levels) völlig funktional aktiv. Der Abfall der Transgenexpression über die Zeit konnte größtenteils auf den Verlust des Vektors zurück-geführt werden. Ein signifikanter Silencing-Effekt konnte nur in den Plasmidvektor- und nicht in den Minicircle-behandelten Gruppen beobachtet werden. Diesbezüglich wurde nach 100 Tagen Persistenz der nicht-viralen Vektoren in der Mausleber eine signifikante Akkumulation an CpG-Methylierungen in regulatorischen Sequenzen des hAAT-Promoters im Plasmidvek-tor gegenüber den Minicirclen ermittelt. Außerdem konnte ebenfalls eine Demethylierung von Cytosin-Nukleotiden in bakteriellen Dcm-Sequenzmotiven über die Zeit beobachtet werden, die eine signifikant höhere Rate für das Minicircle-Konstrukt im hAAT-Promoter aufwies. Die erprobten nicht-viralen Vektoren versprechen somit eine ausreichende Effizienz, um eine Translation in den Menschen, z. B. über eine spezifische Behandlung von Lebersegmenten, möglich erscheinen zu lassen. Die Methylierungsanalyse von Promoterelementen auf Sequenzebene erbrachte zudem konkrete Ansätze zum Design von nicht-viralen Vektoren mit einer noch stabileren FIX-Expression. Im zweiten Teil dieser Arbeit wurde mit Hilfe eines Screening-Systems zur Aktivitätstestung von rekombinant exprimierten FIX-Varianten die Mutation K265T als die entscheidende Sub-stitution im 99-loop identifiziert, welche die FVIII-unabhängige Aktivität im FIX-Protein unter physiologischen Bedingungen begünstigt. Das Wildtyp- IX-Protein braucht hingegen FVIII um eine 106-fache Steigerung seiner proteolytischen Aktivität zu erreichen. Durch Kombination der Mutation K265T mit V181I und I383V konnte diese spezifische FVIII-Bypass-Aktivität in der resultierenden FIX-Variante (ITV) auf 15,6% (verglichen zu FIX in Anwesenheit von FVIII) gesteigert werden. Durch Inkubation der FIX-Varianten mit Plasma-proben aus Hemmkörper-Patienten konnte die Funktionalität der FIX-Varianten selbst in der Anwesenheit von inhibitorischen FVIII-Antikörpern demonstriert werden. Durch Untersu-chungen des Aktivierungszustands von FIX konnte zudem gezeigt werden, dass die im Rah-men dieser Arbeit generierten FIX-Varianten nicht voraktiviert sind und die messbare FVIII-Bypass-Aktivität FIX spezifisch ist. Darüber hinaus konnte hier erstmals eine hämostatische Effizienz von FIX-Varianten mit einer FVIII-unabhängigen Aktivität in einem relevanten Krankheitsmodel demonstriert werden. Diesbezüglich resultierte der nicht-virale Gentransfer der FIX-Varianten in FVIII-K.O.-Mäusen in einer dosisabhängigen Verkürzung der aPTT-basierenden Gerinnungszeit und in einer Reduktion des Blutverlustes im tail-clip-Assay in An- oder Abwesenheit von inhibitorischen FVIII-Antikörpern. Desweiteren konnte zudem mittels Vakzinierung von FIX-WT-tolerierten Mäusen mit den FIX-Varianten demonstriert werden, dass die hier eingeführten Mutationen keine hoch-immunogenen Epitope des FIX-Proteins betreffen. Schließlich konnte die Expression der FIX-Varianten in hämostatisch normalen Mäusen nicht mit der Entstehung von thromboembolischen Ereignissen selbst in einem intakten Gerinnungssystem assoziiert werden. Die hier beschriebenen FIX-Varianten könnten somit einen sicheren und effizienten Therapieansatz zur Behandlung der Hämophilie A mit inhibitorischen Antikörpern darstellen und hinsichtlich der Halbwertszeit und des möglichen throboembolischen Risikos zu einer wesentlichen Verbesserung der bereits verfügbaren Bypass-Strategien beitragen.
Der L-Carnitin/gamma-Butyrobetain Antiporter CaiT ist ein Mitglied der Betain/Carnitin/Cholin Transporter (BCCT) Familie. Sekundärtransporter der BCCT Familie transportieren Substrate, die eine positiv-geladene quartäre Ammoniumgruppe besitzen. CaiT besteht aus 504 Amiosäuren und besitzt ein moleculares Gewicht von etwa 56 kDa. In Enterobakterien wie Escherichia coli, Proteus mirabilis und Salmonella typhimurium wird die Expression des caiTABCDE Operons unter anaeroben Bedingungen induziert. Unter diesen Bedinungen ist CaiT der Haupttransporter des Betain-Derivates L-Carnitin. In Enterobakterien wird L-Carnitin unter anaeroben Bedingungen aufgenommen und dehydratisiert wobei Crotonobetain ensteht. Crotonobetain wird anschließend zum Endprodukt gamma-Butyrobetain reduziert. Gamma-Butyrobetain ist das Gegensubstrat, das aus der Zelle hinaustransportiert wird, wenn L-Carnitin in die Zelle aufgenommen wird. Der Austauschmechanismus von LCarnitin gegen gamma-Butyrobetain geschieht ohne das Vorhandensein eines elektrochemischen Gradients, d.h. CaiT ist sowohl H+- als auch Na+-unabhängig. Ein Ziel dieser Arbeit war es die drei-dimensionale (3D) Struktur von CaiT mittels Röntgenstrukturanalyse zu lösen. Weiterhin sollten mit Hilfe der 3D-Struktur und funktionellen Studien detailiertere Erkenntnisse über den kationenunabhängigen Antiportmechanismus von CaiT ermittelt werden. Im Rahmen dieser Arbeit wurden die 3D-Röntgenkristallstrukturen von drei CaiT-Homologen der Enterobakterien P. mirabilis (PmCaiT), E. coli (EcCaiT) und S. typhimurium (StCaiT) mittels molekularem Ersatz (engl.: molecular replacement, MR) mit einem Alanin-Model des CaiT verwandten Na+/Glycinbetain Symporters BetP gelöst. PmCaiT konnte mit einer Auflösung von 2.3 Å gelöst werden. Das Protein kristallisierte in der Kristallraumgruppe H3, mit drei Molekülen in der asymmetrischen Einheit (engl.: asymmetric unit, AU). Die drei PmCaiT-Moleküle ordneten sich innerhalb der AU um eine kristallographische dreifach Symmetrieachse an. EcCaiT wurde mittels MR mit einem Alanin-Model von PmCaiT bei einer Auflösung von 3.5 Å gelöst. EcCaiT kristallisierte in der Kristallraumgruppe P32, ebenfalls mit drei Molekülen in der AU, jedoch ohne kristallographische Symmetry. Während der Verfeinerung des EcCaiT-Models wurde eine strenge dreifache nichtkristallographische Symmetry (engl.: non-crystallographic symmetry, NCS) angewandt. StCaiT, das ebenfalls mittels MR mit einem Alanin-Model von PmCaiT, aber bei einer Auflösung von 4.0 Å gelöst wurde, kristallisierte in der Kristallraumgruppe P65, ebenfalls mit drei StCaiT-Molekülen in der AU, ohne kristallographische Symmetry. Bei der Verfeinerung des StCaiT-Modells wurde wie bei EcCaiT eine strenge NCS angewandt. Da die Auflösung von 4.0 Å bei StCaiT zu niedrig ist um detailierte moleculare Erkenntnisse zu gewinnen, wurden Protein- sowie Substratinteraktionen nur an den Strukturen von PmCaiT und EcCaiT analysiert. Alle drei CaiT-Homologe weisen jedoch einen ähnlichen strukturellen Aufbau auf. In der Röntgenkristallstruktur bildet CaiT ein symmetrisches Trimer, das über ionische und polare Wechselwirkungen zwischen den Protomeren stabilisiert wird. Der trimere Oligomerisierungszustand von CaiT in Detergenzlösung sowie in zweidimensionalen Lipidmembrankristallen wurde bereits in früheren Arbeiten gezeigt. Jedes der drei CaiT-Protomere besteht aus zwölf Transmembranhelices (TMH), die N- und C-terminalen Domänen des Proteins befinden sich auf der cytoplasmatischen Seite. Zehn der TMH bilden zwei invertierte Wiederholungseinheiten aus jeweils fünf TMH. Die erste Einheit besteht aus den TMH 3 – 7, die invertierte zweite Einheit besteht aus den TMH 8 – 12. Beide Wiederholungseinheiten sind strukturell nahezu identisch und lassen sich fast vollständig übereinanderlegen, jedoch weisen die Aminosäuren der beiden Einheiten keine signifikante Sequenzidentität auf. Die ersten beiden Helices der Wiederholungseinheiten, die TMH 3 – 4 und die TMH 8 – 9, bilden ein antiparalleles vier-Helix-Bündel, in dem in CaiT zwei Substratbindestellen lokalisiert sind. Eine derartige Transporterarchitektur wurde erstmals in der Struktur des Na+/Alanin Symporters LeuTAa des thermophilen Bakteriums Aquifex aeolicus gezeigt. Bislang wurden, inklusive CaiT, sieben Sekundärtransporterstrukturen gelöst, die diese LeuT-Transporterarchitektur aufweisen. Ungewöhnlich dabei ist, dass diese sieben Sekundärtransporter fünf verschiedenen Transporterfamilien angehören und eine Verwandschaft auf Basis der Aminosäuren nicht zu finden ist. Da jedoch die tertiäre Struktur dieser Tansporter konserviert ist, kann davon ausgegangen werden, dass sie alle von einem Urprotein entstanden sind, welches zunächst aus fünf TMH bestanden haben muss. Im Laufe der Evolution hat sich das Urgen des Urproteins zunächst dupliziert und die weitere Evolution hat zwar die Aminosäuresequenz verändert und den Umweltbedingungen angepasst, jedoch ist die tertiäre Struktur erhalten geblieben. Da sich die tertiäre Struktur der sieben Sekundärtransporter so stark ähnelt, ist zu vermuten, dass auch der Transportmechanismus ähnlich, jedoch nicht identisch ist. Nach dem strukturellen Aufbau der Transporter, der Lage der Substratbindestellen in den jeweiligen Transportern und der Tatsache, dass es sich bei diesen Proteinen um Membranproteine handelt, wurde ein Transportmechanismus aufgestellt, in dem die Bindestelle des zu transportierende Substrats alternierend zu beiden Seiten der Membran zugänglich ist, ohne jedoch jemals den Substratweg innerhalb des Proteins vollständig zu öffnen. Dieser Mechanismus wurde als “alternating access mechanism” beschrieben. Anhand der unterschiedlichen Zustände, in denen einige der Transporter kristallisierten, kann abgeleitet werden, welche Konformationsänderungen erforderlich sind um das Substrat von einer Seiter der Membran auf die andere zu transportieren. Bisher kristallisierten einzelne der sechs Transporter in der nach außen gerichteten offenen Form, der nach außen gerichteten Form, in der die Substratbindestelle jedoch nicht mehr zugänglich ist, in einer Form, die keine Öffnungspräferenz der Substratbindestelle zu einer Seite der Membran hat und in der nach innen gerichteten Form, in der die Substratbindestelle jedoch nicht geöffnet ist. CaiT kristallisierte in der noch fehlenden Konformation, der nach innen gerichteten Form, in der die Substratbindestelle zugänglich ist. Mit dieser noch fehlenend Konformation kann der Transportzyklus des “alternating access mechanism” vollständig beschrieben werden. Alle drei CaiT-Homologe kristallisierten in der nach innen gerichteten, offenen Konformation. Im Gegensatz zur EcCaiT-Struktur kristallisierte PmCaiT in der substratungebundenen Form. In der StCaiT-Struktur konnte aufgrund der niedrigen Auflösung kein Substrat nachgewiesen werden. In der EcCaiT-Struktur sind zwei gamma-Butyrobetain-Moleküle gebunden. Das erste Molekül wurde in der zentralen Substratbindestelle, der sogenannten Tryptophan-Box bestehend aus vier Tryptophanen, im Zentrum des Protein lokalisiert. Das zweite gamma-Butyrobetain-Molekül wurde in einer Vertiefung an der extrazellulären Proteinoberfläche gefunden. Beide Substrate werden hauptsächlich über Kation-Pi-Interaktionen zwischen der positiv geladenen quatären Ammoniumgruppe des Substrats und des Pi-Elektronensystems der Tryptophane in den jeweiligen Bindestellen gebunden. Eine besondere Eigenschaft von CaiT ist der H+- bzw. Na+-unabhängige Substrattransport. Die CaiT-Struktur erklärt warum kein zusätzliches Kation benötigt wird um Substrat zu binden oder zu transportieren. In der EcCaiT-Struktur ist eine wichtige polare nicht-bindende Interaktion zwischen der Carboxylgruppe des gamma-Butyrobetains und dem Schwefelatom eines Methionins in der zentrale Bindestelle zu erkennen. Dieses Methionin ist konserviert in den prokaryotischen CaiTs und in den Na+-unabhängigen eukaryotischen L-Carnitin Transportern (OCTN), jedoch ist es nicht konserviert im Na+-abhängigen verwandten Glycinbetain Transporter BetP. In BetP ist diese Position des Methionins durch ein Valin ersetzt. Die Mutation des Methionins in CaiT zu Valin ermöglicht zwar immernoch die H+- bzw. Na+-unabhängige Bindung des Substrates durch die Tryptophan-Box, jedoch ist der Substrattransport nahezu vollständig zerstört. Eine derart wichtige Substratkoordinierende Funktion des Schwefelatoms eines Methionins wurde bisher nicht beschrieben. Eine weitere Stelle, die in H+- bzw. Na+-abhängigen Transporter mit H+ bzw. Na+ besetzt ist, ist in CaiT von einem positiv geladenen Arginin eingenommen. Eine positive Ladung an dieser Stelle stabilisiert den Bereich im Protein in der Nähe der zentralen Substratbindestelle. Die Mutation des Arginins zu Glutamat in CaiT erzielt eine vollständige Inaktivierung des Substrattansports. Durch Zugabe von Na+ im Transportansatz kann die Substrattransportaktivität der Glutamat-Mutante jedoch teilweise zurückerlangt werden. Diese eben beschriebenen Aminosäurereste in den beiden Stellen des Proteins erklären die Kationenunabhängigkeit von CaiT. Die Aktivierung des Antiportmechanismus in CaiT wurde mit Hilfe von Bindungsstudien an rekonstituiertem Protein ermittelt. Diese Messungen ergaben für das Wildtypprotein ein sigmoidales Substratbindungsverhalten, was auf ein positiv-kooperatives Bindungsverhalten hindeutet. Die beiden Substratbindestellen im Protein sowie die beiden unterschiedlichen Substrate, L-Carnitin und gamma-Butyrobetain, lassen auf einen heterotropen positiv-kooperativen Bindungs- und einen allosterisch regulierten Transportmechanismus schließen. Bei diesem Mechanismus erhöht die Bindung eines Substrats in der regulatorischen Bindestelle durch induzierte Konformationsänderungen die Affinität eines anderen Substrats in einer weiteren Substratbindestelle. Die regulatorische Bindestelle in CaiT befindet sich an der extrazellulären Proteinoberfläche. Eine Schwächung der Substrataffinität in dieser Bindestelle durch Einführung einer Mutation, verstärkt das sigmoidale Substratbindungsverhalten und hat einen negativen Einfluss auf den Substrattransport. Durch die in dieser Arbeit gelösten 3D-Röntgenkristallstrukturen der zwei CaiT-Homologen, PmCaiT und EcCaiT, sowie den durchgeführten funktionellen Studien sowohl an Wildtypprotein wie auch an Mutanten konnte ein L-Carnitin/gamma-Butyrobetain Antiport-Mechanismus für CaiT vorzuschlagen werden.
Die Verfügbarkeit synthetischer Oligonukleotide hat der Entwicklung einer Vielzahl molekularbiologischer, biochemischer und medizinischer Anwendungen den Weg geebnet. Und sind viele diese Anwendungen für sich genommen schon hochinteressant, so eröffnet die Kombination mehrerer Methoden oft noch ganz neue Möglichkeiten. In der vorliegenden Doktorarbeit ist es gelungen, die Technik der photolabilen Schützung auf die Anwendungen von siRNAs und molecular beacons zu übertragen und diesen damit die Option der orts- und zeitaufgelösten Aktivierung zu ermöglichen. Durch die Einführung eines Nukleotids mit 2-(2-nitrophenyl)propyl-geschützter Nukleobase in eine siRNA, konnte der katalytische Schritt der RNA-Interferenz, die mRNA-Spaltung, unterbunden werden. Hierzu wurde das photolabil modifizierte Nukleotid so in der siRNA positioniert, dass es gegenüber der mRNA-Schnittstelle bzw. in unmittelbarerer Nachbarschaft zu dieser lag. Dabei war das modifizierte Nukleotid selbst kein Ribonukleotid sondern ein Desoxynukleotid. Zuvor konnte gezeigt werden, dass die Einführung einzelner Desoxynukleotide in eine siRNA keinerlei Einfluss auf deren Aktivität hat. Als Modellsystem diente der RFP/eGFP-Reportergenassay, wobei die Plasmide mit der siRNA in die verwendeten HeLa-Zellen kotransfiziert wurden. Die verwendete siRNA regulierte dabei die eGFP-mRNA, die gemessene Fluoreszenz wurde auf die RFP-Fluoreszenz normiert. In der Studie gelang es, ein sauberes „An/Aus-Verhalten“ zu erzielen, das heißt, die modifizierte siRNA zeigte zunächst keinerlei Einfluss auf die eGFP-mRNA. Bestrahlte man diese siRNA jedoch für drei Minuten bei 366 nm, erzielte man eine Unterdrückung der eGFP-Expression, die der einer unmodifizierten siRNA entsprach. Dies funktionierte für vor der Transfektion bestrahlte siRNAs ebenso, wie für solche, die erst nach der Transfektion in der Zelle entschützt wurden. Ein bis jetzt ungeklärtes und noch näher zu untersuchendes Phänomen ist die Stabilität der Modifikationen in der Zelle. Aus bisher nicht eindeutig zu benennender Ursache fand nach einer definierten Zeit eine Aktivierung der ausgeschalteten siRNA statt, ohne dass diese bestrahlt wurde. Versuche mit photolabil modifizierten Nukleotiden an anderen Positionen innerhalb der siRNA, sowie eine Fluoreszenz-Korrelationsspektroskopie-Studie mit fluoreszenzmarkierter siRNA und fluoreszenzmarkiertem RISC erlaubten es Rückschlüsse auf den Schritt der RNAi zu ziehen, der durch die Einführung der Basenmodifikation blockiert wird. Offenbar handelt es sich tatsächlich um den katalytischen Schritt der mRNA-Spaltung, ein Einbau der modifizierten siRNA in den RISC findet statt. Zudem zeigte die erfolgreiche Inaktivierung der für die FCS-Studie genutzten anti-TK siRNA, dass der Ansatz, die Modifikation im Bereich der Schnittstelle einzubauen, von der anti-eGFP siRNA auf andere siRNAs übertragbar ist. Im zweiten erfolgreichen Projekt gelang es, molecular beacons durch Einführung zahlreicher photolabiler Basenmodifikationen lichtaktivierbar zu machen. Hierzu wurde ein bereits beschriebener GAPDH-molecular beacon verwendet. Modifiziert man die Schleife dieses molecular beacon mit sieben photolabilen Basenschutzgruppen (NPP und NPE) so gelingt es die Bindung desselben an seine komplementäre Ziel-RNA komplett zu unterbinden, während ein GAPDH-beacon mit drei oder fünf Modifikationen noch in verringertem Maße bindungsfähig ist. Dieses Verhalten wurde sowohl mittels einfachen Auslesens der Fluoreszenzintensität, als auch anhand eines PA-Geles belegt. Eine große Herausforderung bei diesem Projekt stellte die Aufreinigung des hochmodifizierten molecular beacon dar, der neben Fluorophor und Quencher auch zahlreiche Photoschutzgruppen trägt. Diese gelang schließlich durch den Einsatz einer extra densely bond-RP-HPLC-Säule und wiederholter HPL-Chromatographie. Ebenfalls konnte der große Vorteil eines lichtaktivierbaren molecular beacon, die Möglichkeit der präzisen Ortsauflösung der Aktivierung, dargestellt werden. Hierzu wurde modellhaft die Ziel-RNA auf einer Objektträgeroberfläche immobilisiert. Die dann aufgetragene molecular beacon-Lösung zeigte zunächst keine Fluoreszenz. Diese trat erst nach Bestrahlung und auch nur begrenzt auf den wenige Quadratmikrometer großen Bestrahlungsbereich auf.
In this thesis, the structure of the C-terminal domain of presenilin-1, the catalytic component of the y-secretase complex, is investigated by NMR spectroscopy. The ysecretase complex has a definitive role in the pathogenic development of Alzheimer's disease, in that it mediates the cleavage of aprecursor to create the amyloid ß peptide. Aggregates of amyloid ß which form amyloid plaques are the most overt clinieal feature observed in the post-mortem brains of Alzheimer's patient. In addition, many of the mutations found in the aggressive early onset familial Alzheimer's disease have been linked to presenilin-1, highlighting its importance in disease progression and deeming it an important target for investigation. One of the greatest challenges for the structural investigation of the y-secretase components is their low expression yields in cell-based systems. We therefore applied continuous-exchange cell-free expression to obtain sufficient amounts of protein for our structural studies. An added benefit of the cell-free expression system is the freedom to incorporate any desired combination of stable-isotope labels directly into sampies. We were therefore able to develop a labeling scheme which targets the amino acid composition of transmembrane a-helices, allowing us to simplify an assignment procedure whieh tends to be cumbersome and diffieult for most a-helical transmembrane proteins. The y-secretase complex is a member of the intramembrane cleaving proteases which, as their name implies, cleave their transmembrane substrates within the bilayer. Single particle analysis of the y-secretase (1) as weil as crystal structures of rhomboid (2) and S2P (3) have revealed the presence of hydrophilie po res within the membrane where catalysis occurs. In light of evidence that certain elements of CTF reside in close proximity or even contribute to the formation of the hydrophilic pore, we chose to study the structure of CTF in mieelles, whieh may be better suited to accommodate CTF in isolation as compared with solid membranes in the absence of the other y-secretase components. The structure of CTF was solved to 1.7 A (backbone r.m.s.d) and revealed the presence of unusual features, including a partially membrane-spanning helix which situates the catalytic asparte at its N-terminus in what would be the center of the membrane where catalysis is proposed to occur, as weil as a severely kinked helix which is partially embedded beneath the surface of the membrane (P6). Interestingly, similar features have been observed in the crystal structure of the GlpG rhomboid. In addition, a soluble helix was found in the long N-terminal loop of CTF which until now has been described as unstructured. The first part of the thesis is designed to provide an introduction to Alzheimer's disease, the role of y-secretase and its presenilin-l catalytic component in disease progression, as weil as cell-free expression and liquid-state NMR techniques involved in the structural investigation of membrane proteins. In chapter 2, the reader is familiarized with the history, the clinical manifestation, and biochemical features of Alzheimer's disease. The chapter goes further to describe the role of the y-secretase complex and its individual components in disease progression and substrate processing. Chapter 3 focuses more specifically on presenilin-l in the context of the newly emerging class of intramembrane proteases. In chapter 4, attention is shifted to the cell-free expression system with special focus on the expression of membrane proteins, and chapter 5 explores the various liquid-state NMR techniques that were required for the characterization of CTF. The second part of the thesis is cumulative and contains original research, method, and review articles that were produced during the course of study. Chapter 6 explores the various techniques and innovations used to study membrane proteins using continuous exchange cell-free expression coupled with NMR spectroscopy. In chapter 7, a new technique, transmembrane segment targeted labeling, is described as a tool that facilitates the backbone assignment of transmembrane proteins which display severe overlap in NMR spectra. Chapter 8 presents the novel NMR structure of the C-terminal fragment of presenilin-l solved in SOS micelles.
ABCB9 is a peptide transporter belonging to the ATP-binding cassette (ABC) transporter subfamily B. Due to its high sequence identity to the transporter associated with antigen processing (TAP) the protein was named TAP-like (TAPL). The primary aim of this PhD thesis was the functional characterization of the TAPL transport complex. Despite the lack of TAPL function in the classical MHC class I pathway an involvement of TAPL in antigen presentation was still suggested. Apart from the crucial role of TAP for peptide delivery into the ER, TAP-independent translocation pathways in professional antigen presenting cells (pAPC) have been proposed, but not identified so far. Remarkably, TAPL mRNA and protein expression is strongly induced during differentiation of monocytes to immature and mature dendritic cells. This result was confirmed in the promonocytic cell line THP-1, which was used as a model system for monocyte to macrophage differentiation. By using quantitative immunofluorescence microscopy and subcellular fractionation, TAPL was detected in the lysosomal compartment co-localizing with the lysosome associated membrane protein 2 (LAMP-2) thus excluding the ER-localization formerly reported. Furthermore, by in vitro assays, a TAPL-specific and ATPdependent translocation of peptides into isolated lysosomes was demonstrated. Hence, TAPL is a candidate mediating peptide transport in alternative antigen presentation pathways in pAPCs. The presence of an extra N-terminal transmembrane domain (TMD0) lacking sequence homology to any known protein distinguishes TAPL from most other ABC transporters of its subfamily. By dissecting the TAPL translocation complex into its four putative transmembrane helices containing TMD0 and the core complex, distinct functions to the core complex and TMD0 were assigned. The core-TAPL complex composed of six predicted transmembrane helices and the nucleotide-binding domain (NBD) was expressed transiently in HeLa or stably in Raji cells. Crude membranes containing core-TAPL showed the same peptide transport activity as wt-TAPL demonstrating that the six core helices and the NBD are sufficient for peptide transport. This result also shows that the core transport complex is correctly targeted to and assembled in the membrane. Strikingly, in contrast to the wt transporter, the core complex localizes only partially to lysosomes and is mistargeted to the plasma membrane as observed by immunofluorescence microscopy and confirmed biochemically by cell surface biotinylation. Thus, a crucial role for TMD0 in proper subcellular targeting can be postulated. The vast majority of biological processes are mediated by protein complexes, hence characterization of such protein-protein-interactions is essential for understanding protein function on the cellular level. To identify interaction partners of TAPL, the transporter was isolated by tandem affinity purification. By tandem mass spectrometry the membrane proteins LAMP-1 and LAMP-2 were deciphered as specific proteins interacting with wt-TAPL. Notably, core-TAPL lacks these interactions indicating a role for TMD0 in recruiting other proteins. These results were verified for endogenous TAPL by co-immunoprecipitation. Using cells deficient in LAMP-1 and/or in LAMP-2 an escort function for the LAMP proteins was excluded. Very importantly, the physiological function of the LAMP-1and LAMP-2 interaction with TAPL is an increase in stability, since in their absence half-life of TAPL is drastically reduced.
An chemischen Synapsen diffundieren von der präsynaptischen Nervenendigung ausgeschüttete Neurotransmitter durch den synaptischen Spalt und aktivieren Rezeptormoleküle in der postsynaptischen Plasmamembran. Eine schnelle und zuverlässige Kommunikation an Synapsen bedingt, dass die Rezeptoren in Clustern direkt gegenüber den aktiven Zonen der Präsynapsen angereichert sind. Eine hohe Rezeptorendichte in der postsynaptischen Membran wird durch sog. Gerüstproteine, die mit den Rezeptormolekülen assoziieren, erreicht. Bisher wurden für verschiedene Synapsen unterschiedliche Gerüstproteine identifiziert. An glutamatergen Synapsen werden Rezeptoren durch u.a. das Postsynaptic Density 95 (PSD 95)-Protein, an cholinergen Synapsen durch Rapsyn, und an GABAergen und glyzinergen Synapsen durch Gephyrin verankert. An inhibitorischen Synapsen wurde bisher ausschliesslich Gephyrin als Ankerprotein für Glyzin- und GABAARezeptoren identifiziert. An exzitatorischen glutamatergen Synapsen dagegen regulieren weitere Gerüstproteine wie Shank, Homer und das Glutamatrezeptor interagierende Protein (GRIP) die Lokalisation, den Transport und die Stabilität verschiedener Glutamatrezeptor- Subtypen. Gephyrin wurde ursprünglich als peripheres Membranprotein zusammen mit dem Glyzin-rezeptorkomplex aufgereinigt. Es bindet an die große intrazelluläre Schleife der ß-Untereinheit des Glyzinrezeptors. Experimente mit Antisense-Oligonukleotiden und Gephyrin-defizienten Mäusen zeigten, dass Gephyrin essentiell für die synaptische Lokalisation von Glyzinrezeptoren und α2- und γ2-Untereinheiten enthaltenden GABAARezeptoren ist. Die Rezeptorlokalisation an der Synapse wird durch eine stabile Verankerung des Gephyrin-Gerüstes am Zytoskelett gewährleistet. Die Gerüstbildung erfolgt durch die Oligomerisierung zweier Gephyrin-Domänen, der aminoterminalen G-Domäne und der carboxyterminalen E-Domäne, wodurch ein hexagonales Gephyrinnetzwerk entsteht, welches für die Clusterbildung an der Synapse notwendig ist. Live-imaging Studien in Neuronenkulturen zeigten, daß an Rezeptor-Transportvesikel gebundenes Gephyrin kontinuierlich an und von aktiven Synapsen weg transportiert wird, was eine hochdynamische Modulation des Gephyrin-Gerüsts nahelegt. Der retro- und anterograde Transport von Gephyrin wird durch Dynein bzw. Kinesin bewerkstelligt. Einige zytosolische Proteine wie Profilin, das Mena/Vasodilator-stimulierte Phosphoprotein (VASP) und Collybistin (Cb) spielen in der Zytoskelett-Regulation eine Rolle und interagieren mit Gephyrin. Cb ist notwendig für die Gephyrin-Clusterbildung an bestimmten GABAergen Synapsen. Im Hippocampus Cb-defizienter Mäuse befindet sich kein Gephyrin an den Postsynapsen. Cb gehört zur Familie der Guanin-Nukleotid-Austauschfaktoren (GEF), die durch eine DH-PH (Dbl Homologie-/Pleckstrin-Homologie) Tandem-Domäne charakterisiert sind und existiert in zwei Spleißvarianten, CbI und CbII. Die DH-Domäne aktiviert spezifisch das kleine G-Protein Cdc42, und die PH-Domäne interagiert mit Phosphoinositol-3-Phosphat (PI3P). In vitro-Studien haben gezeigt, dass die Interaktion von Gephyrin mit Cb II eine Reduktion der GEF-Aktivität für Cdc42 bewirkt. Basierend auf diesen Resultaten wurde vorgeschlagen, dass Gephyrin die Cdc42-Aktivierung durch Cb beendet. Dieser Mechanismus könnte für die initiale Bildung inhibitorischer Synapsen notwendig sein. Analysen mit Domänen-deletierten Cb-Konstrukten zeigten, dass sowohl die DH- als auch die PH-Domäne für die Bildung von submembranären Gephyrin-Microclustern an der Plasmamembran notwendig sind. In dieser Arbeit wurde der Mechanismus der Gephyrin-Gerüstbildung durch Cb weiter untersucht. Insbesondere wurde geklärt, inwiefern Cdc42-Aktivierung bzw. PI3P-Bindung durch Cb an der Gephyrin-Gerüstbildung beteiligt ist. Zusätzlich wurde anhand einer eigens erzeugten GFP-Gephyrin transgenen Maus untersucht, ob sich die Stabilität des Gephyrin-Gerüsts während der Differenzierung ändert und in die Regulation der Stabilität und Plastizität inhibitorischer Synapsen involviert ist. Die Rolle von Cdc42 in der Gephyrin-Gerüstbildung wurde mittels einer konstitutiv aktiven Spleißvariante von Cb untersucht. Basierend auf Homologien zu anderen bereits charakterisierten GEFs und der publizierten Kristallstruktur des CbII-Cdc42 Komplexes wurden Aminosäurereste in der DH-Domäne von Cb mutiert, um die Cdc42-Aktivierung zu unterbrechen (CB T61A, K192A und NE232-233AA). In vitro GTPase-Aktivierungsassays und Filopodien-Induzierung in NIH-3T3-Zellen bestätigten, dass diese Mutationen die Cdc42-Aktivierung reduzierten bzw. aufhoben. Dennoch induzierten sie die Gephyrin-Gerüstbildung in heterologen Zellen und hippocampalen Neuronen ähnlich effektiv wie Wildtyp-Cb II. Die Rolle von Cdc42 in der synaptischen Gephyrin-Clusterbildung wurde außerdem in konditionell Cdc42-defizienten Mäusen, in denen das Cdc42-Gen selektiv im Vorderhin inaktiviert wurde, untersucht. Die Dichte von Gephyrin- und das GABAA-Rezeptor-Clustern war bei Verlust von Cdc42 im Hippocampus nicht verändert. Dies steht im Gegensatz zu einem fast 80%igen Verlust von Gephyrin- und GABAA-Rezeptor-Clustern im Hippocampus von Cb-defizienten Mäusen. Diese Ergebnisse zeigen, dass Cdc42 für die Gephyrin-Gerüstbildung an inhibitorischen Synapsen nicht notwendig ist. Die Rolle der PH-Domäne von Cb bei der Gephyrin-Clusterbildung wurde ebenfalls durch Mutagenese-Experimente analysiert. Viele PH-Domänen haben die Fähigkeit, Phosphoinositide zu binden und damit Membranbindung zu vermitteln, während andere PHDomänen, die nicht an Phosphoinositide binden, nur nach Bindung weiterer Liganden mit Membranen assozieren. In früheren Arbeiten war gezeigt worden, dass humanes Cb PI3P bindet. Hier wurde eine mögliche Beteiligung von Phospholipid-Bindung an der Gephyrin-Gerüstbildung durch Substitution zweier basischer Reste in der β3- und β4- Schleife, R303 und R304, mit Asparaginen untersucht. Ein Lipid-Dot-Blot-Assay mit der Cb II-RR303-304NN-Mutante zeigte einen völligen Verlust der PI3P-Bindung in vitro. Die Expression dieser Mutante in heterologen Zellen und hippocampalen Neuronen zeigte, dass die Gerüstbildung und die synaptische Lokalisation von Gephyrin Phosphoinositidbindung erfordern. Zusammenfassend zeigen diese Ergebnisse, dass die Aktivierung von Cdc42, nicht aber die PI3P- Bindung, für die Cb II-vermittelte Gephyrin-Gerüstbildung entbehrlich ist. Cb hat also zumindest zwei biologische Funktionen: Einerseits ist die DH-Domänen vermittelte Aktivierung von Cdc42 notwendig für die Regulation des Aktinzytoskeletts und die Bildung von Filopodien; andererseits wird die PH-Domänen-abhängige und Cdc42-unabhängige Phospholipidbindung für die Gephyrin-Gerüstbildung an inhibitorischen Postsynapsen benötigt. Um die Dynamik von Gephyrin an inhibitorischen Synapsen zu untersuchen, wurde eine transgene Maus entwickelt, welche GFP-Gephyrin unter der Kontrolle des Neuronspezifischen Thy1-Promotors exprimiert. In verschiedenen Hirnregionen der transgenen Mauslinie, wie Hippocampus, Stammhirn, Rückenmark und Kortex, wurde punktuelle synaptische GFP- Fluoreszenz beobachtet. Diese GFP-Gephyrin-Fluoreszenz kolokalisierte mit einem inhibitorischen Präsynapsenmarker, dem vesikulären inhibitorischen Aminosäuretransporter (VIAAT). Immunfärbungen von Hirnschnitten mit Gephyrin-spezifischen Antikörpern zeigten, daß die durchschnittliche Größe und Dichte der GFP-Gephyrin-Cluster mit denen endogener Gephyrin-Cluster identisch waren. Eine Western-Blot Analyse inhibitorischer synaptischer Proteine zeigte keinen Unterschied zwischen Thy1-GFP-Gephyrin- und Wildtyp-Mäusen auf, ebensowenig wie elektrophysiologische Untersuchungen von GFP-Gephyrin-positiven und -negativen Neuronen. Sowohl die durchschnittliche Amplitude der mIPSCs als auch deren Frequenz waren nicht signifikant verändert, was dafür spricht, dass die transgene Expression von GFP- Gephyrin keine funktionellen Veränderungen verursacht. Verhaltensversuche zeigten gleiche Ergebnisse für Thy1-GFP-Gephyrin und WTMäuse. Daher kann die GFP-Gephyrin-Maus als verlässliche Reporterlinie für Studien zur Gephyrin-Dynamik an inhibitorischen Synapsen im Hippocampus und in einigen anderen Hirnregionen eingesetzt werden. Die Dynamik des synaptischen Gephyrin-Gerüsts wurde an inhibitorischen Postsynapsen in organotypischen entorhinal-hippocampalen Schnittkulturen aus GFPGephyrin-Mäusen untersucht. Fluorescence Recovery after Photobleaching (FRAP)-Analysen individueller GFP-Gephyrin-Cluster in 1-Woche- und 4-Wochen-alten Kulturen zeigten eine entwicklungsabhängige Stabilisierung der GFP-Gephyrin-Cluster auf. Diese Stabilisierung ist eng verbunden mit einer Größenzunahme der Gephyrin-Cluster an GABAergen Synapsen. Mit elektrophysiologischen Ableitungen wurde eine Reifung der GABAergen synaptischen Übertragung ebenfalls während dieser Periode beobachtet. Die Stabilisierung und Grössenzunahme des Gephyrin-Gerüsts spiegelte sich in einer erhöhten miniature inhibitory postsynaptic current (mIPSC)- Amplitude wieder. Außerdem wies die Zunahme der mIPSCFrequenz auf eine effiziente Reifung der präsynaptischen Endigungen hin, die immunhistochemisch durch eine Zunahme der VIAAT-Immunfluoreszenz erhärtet werden konnte. Ein möglicher Einfluss der GABAergen, synaptischen Aktivität auf die Grösse und Stabilität der Gephyrin-Cluster wurde in reifen Neuronenkulturen durch pharmakologische Modulation der GABAA-Rezeptoren untersucht. G-protein coupled receptors (GPCRs) are the key players in signal perception and transduction and one of the currently most important class of drug targets. An example of high pharmacological relevance is the human endothelin (ET) system comprising two rhodopsin-like GPCRs, the endothelin A (ETA) and the endothelin B (ETB) receptor. Both receptors are major modulators in cardiovascular regulation and show striking diversities in biological responses affecting vasoconstriction and blood pressure regulation as well as many other physiological processes. Numerous disorders are associated with ET dysfunction and ET antagonism is considered an efficient treatment of diseases like heart failure, hypertension, diabetes, artherosclerosis and even cancer. This study exemplifies strategies and approaches for the preparative scale synthesis of GPCRs in individual cell-free (CF) systems based on E. coli, a newly emerging and promising technique for the production of even very difficult membrane proteins. The preparation of high quality samples in sufficient amounts is still a major bottleneck for the structural determination of the ET receptors. Heterologous overexpression has been a challenge now for decades but extensive studies with conventional cell-based systems had only limited success. A central milestone of this study was the development of efficient preparative scale expression protocols of the ETA receptor in qualities sufficient for structural analysis by using individual CF systems. Newly designed optimization strategies, the implementation of a variety of CF expression modes and the development of specific quality control assays finally resulted in the production of several milligrams of ETA receptor per one millilitre of reaction mixture. The versatility of CF expression was extensively used to modulate GPCR sample quality by modification of the solubilization environment with detergents and lipids in a variety of combinations at different stages of the production process. Downstream processing procedures of CF synthesized GPCRs were systematically optimized and sample properties were analysed with respect to homogeneity, protein stability and receptor ligand binding competence. Evaluation was accomplished by an array of complementary and specifically modified techniques. Depending on its hydrophobic environment, CF production of the ETA receptor resulted in non-aggregated, monodisperse forms with sufficient long-term stability and high degrees of secondary structure thermostability. The obtained results document the CF production of the ETA receptor in two different modes as an example of a class A GPCR in ligand-binding competent and non-aggregated form in quantities sufficient for structural approaches. The presented strategy could serve as basic guideline for the production of related receptors in similar systems.
