Chronisch-entzündliche Dermatosen sind in Deutschland weit verbreitet und haben einen enormen Einfluss auf die Lebensqualität der Erkrankten. Das umfassende Verständnis der molekularen Prozesse und Signalwege bildet die Basis, um mögliche Beziehungen zwischen den Hauterkrankungen aufzudecken. Der Nachweis von Ähnlichkeit und Übereinstimmung in den Signalwegen bietet die Aussicht, dass etablierte Therapien auch bei anderen Erkrankungen helfen können.
Der Zweck dieser Arbeit ist der Nachweis der Expression von IL-1β, IL-17A, IL-22, IL-23 und TNF-α in drei chronisch-entzündlichen Dermatosen: Acne inversa (AI), Sinus pilondalis (SP) und Perifolliculitis capitis abscedens et suffodiens (PCAS). Bei allen untersuchten Diagnosen handelt es sich um potentiell verwandte Erkrankungen der Haarfollikel, die durch Verlegung der Ausführungsgänge zu ausgedehnten Entzündungsreaktionen mit Bildung von Knoten, Abszessen und Fisteln führen. Bereits nachgewiesen ist, dass IL-1β, IL-17A, IL-22, IL-23 und TNF-α eine wichtige Rolle in der Pathogenese von Acne inversa spielen und Antikörpertherapien an einigen dieser Zielproteine spezifisch angreifen, um den Verlauf der Erkrankung zu verbessern.
Die Expression der Proteine wurde in Läsionen von Patienten mit den drei Indikationen immunhistochemisch an paraffiniertem Gewebe untersucht. Wie erwartet, zeigten sich in der überwiegenden Anzahl der Proben große entzündliche Infiltrate und hier wurde die Expression aller untersuchten Zytokine in unterschiedlicher Intensität nachgewiesen. Vielversprechend war insbesondere die Expression von IL-17A und IL-23 in SP und PCAS. Die Expression von IL-1β war insgesamt eher gering ausgeprägt; bei AI noch etwas höher als in SP und PCAS. Die Färbungen auf IL-22 zeigten sich kräftig in allen untersuchten Dermatosen. Allerdings gibt es bisher keine zugelassene Therapie zur Modulation dieses speziellen Zytokins. IL-22 scheint eine zentrale Rolle in
der Pathogenese der AI zu spielen. Mit TNF-α-Blockern wurden schon gute therapeutische Ergebnisse bei AI und PCAS erzielt. Deshalb ist der Nachweis von TNF-α in den entzündlichen Läsionen zu erwarten gewesen. Auch bei den SP-Proben fanden sich deutlich erhöhte Protein-Level, sodass auch hier eine gezielte Therapie von Vorteil sein könnte. Wegen des geringen Probenumfangs und der Methodik sind weitere gezielte Untersuchungen notwendig. Dennoch konnten viele Gemeinsamkeiten der Zytokinexpression ausgemacht werden, was vielversprechende Hinweise auf mögliche Behandlungsansätze bei AI, SP und PCAS zulässt. Diese Arbeit bietet einen ersten Blick auf den immunologischen Phänotyp der verwandten Dermatosen.
Im Rahmen dieser Arbeit wurde die schnelle Energietransfer- (EET) und Elektronentransfer (ET)-Dynamik unterschiedlichster Quantenpunkte (QD) spektroskopisch untersucht. Die untersuchten Systeme bestanden in den meisten Fällen aus Donor-Akzeptor-Paaren, bei denen die Halbleiternanokristalle als Donor fungierten. Der Fokus lag dabei auf der gezielten Anpassung des Donors, um die optimale Funktionalität zu erreichen. Die Untersuchung der Nanokristalle erstreckte sich daher von einfachen Kernen über verschiedene Kern-Schale-Partikel bis hin zu völlig anderen Strukturen wie Nanoplatelets (NPL). Als Akzeptor wurden eine Vielzahl von Molekülen verwendet, die sich als Elektronen- und/oder Energieakzeptoren für die verschiedenen QDs eignen.
Der Begriff der Kompetenz ist allgegenwärtig und wird fast schon inflationär eingesetzt, um beispielsweise Qualität und Wertigkeit einer Dienstleistung oder eines Produktes hervorzuheben, sowie häufig verwendet, um eine besondere Bedeutung und hohe Leistungsfähigkeit zu vermitteln.
Innerhalb des formalisierten schulischen, aber auch des beruflichen und des universitären Bildungssystems nimmt der Kompetenzbegriff, nachdem er aufgrund seiner Popularität den Begriff der Schlüsselqualifikation ablöste, eine zentrale Stellung ein. Kompetenzorientierter Unterricht soll nicht nur dafür sorgen, dass Deutschland im internationalen Vergleich schulischer Bildungsergebnisse besteht, sondern auch ermöglichen, dass Bildung und Ausbildung an gesellschaftlichen Erfordernissen und individuellen Voraussetzungen gleichermaßen ausgerichtet werden können.
In der Kunstpädagogik ist die Kompetenzorientierung nicht unstrittig. Zwischen dem Begrüßen als Möglichkeit des Anschlusses an internationale Bildungsstandards und der vehementen Ablehnung als Versuch der Ökonomisierung und Technokratisierung von Bildungsprozessen sind nur wenige Alternativen auszumachen, die eine fachspezifische Aneignung und Reformulierung des Begriffs beabsichtigen.
Die hier vorliegende Forschungsarbeit untersucht die Anwendung, Schulung und Erweiterung von Kompetenzen in einem bildnerischen Projekt in der Jugendhilfe, einem Teilbereich der Sozialen Arbeit. In außerschulischen Bildungskontexten wird der Begriff der Kompetenz bislang noch wenig rezipiert. Was nicht zuletzt daran liegt, dass Handlungsfelder außerhalb formalisierter Lernsettings lange nicht als einflussreiche Bildungsorte in den Blick genommen wurden. Erst langsam beginnt sich eine Forschung zu etablieren, die Bildung in Bezug auf die gesamte Lebenswelt versteht. Dabei werden Kompetenzen ähnlich wie im schulischen Bildungssystem oftmals auf vermittelbare Wissensbestände, Fertigkeiten oder methodische Vorgehensweisen bezogen und somit vor allem kognitiv und domänenspezifisch bestimmt. Ein weiter gefasstes Verständnis begreift Kompetenz jedoch als Fähigkeit, subjektiv erfolgreich mit der Umwelt zu interagieren, wobei diese Fähigkeit in der Interaktion ausgebildet und modifiziert wird. Mit diesem Verständnis lassen sich auch motivationale und volitionale Aspekte integrieren.
Kompetenz wird damit als eine sowohl sachliche als auch soziale und personale Bestandteile beinhaltende Befähigung zur Lebensbewältigung betrachtet, die auf eine weitestgehende Selbstbestimmung abzielt und ein auf die Zukunft gerichtetes Handeln ermöglicht. Kompetenz ist damit nicht nur als Folge oder Ergebnis von Bildungsprozessen anzusehen, die in der erwünschten Form und Ausprägung entwickelt werden sollen. Kompetenz kann vielmehr als eine dem Individuum eigene Befähigung angesehen werden, die in Bildungsprozessen subjektiv sinnhaft aktiviert, weiter ausgebildet und verändert wird. Damit geraten sowohl die subjektiven Sinnstrukturen als auch die soziokulturellen Kontexte, in denen Kompetenz als Bewältigungskönnen ausgebildet werden muss, stärker in den Blick.
Structure-function relationships in substrate binding protein dependent secondary transporters

This work provides new insights into the relevance of SBP dependent secondary transport systems, especially in the thus far under-researched subgroup of TAXI transporters. Importantly, we identified and characterized the TAXI transport system TAXIPm-PQM from Proteus mirabilis. We demonstrated that, in contrast to previously characterized SBP dependent secondary transport systems, TAXIPm-PQM is a proton coupled system and transports the C5-dicarboxylate α- ketoglutarate. Since initially the transport of α-ketoglutarate could only be demonstrated in vivo but not in vitro using established protocols (Mulligan et al. 2009), we investigated in detail the differences between the in vivo and in vitro assay. This resulted in a bioinformatic analysis of TRAP and TAXI signal peptides, which strongly implied that TAXIPm-P requires a transmembrane anchor to allow for transport. We then provided TAXIPm-P surface tethered to the membrane in in vitro transport assays and confirmed the prediction of our bioinformatic analysis that TAXIPm-PQM deploys a membrane-anchored instead of a soluble SBP. Furthermore, the TAXI transport system TAXIMh-PQM from Marinobacter hydrocarbonoclasticus transports fumarate only if both membrane domains Q and M are present. For further characterization, Michaelis-Menten kinetics and affinities were determined for both TAXI transport systems TAXIPm-PQM from Proteus mirabilis and TAXIMh-PQM from Marinobacter hydrocarbonoclasticus. In addition, nanobodies were selected for the membrane domain TAXIPm-QM from Proteus mirabilis to stabilize different conformations which can serve in subsequent structural elucidation studies. Furthermore, the TRAP SBP TRAPHi-SiaP from Haemophilus influenzae was shown to interact not only with its corresponding membrane domain TRAPHi-SiaQM but with at least one additional transporter. It was thereby excluded that TRAPHi- SiaP transfers N-acetylneuraminic acid to the only native E. coli TRAP transporter TRAPEc-YiaMNO and suggested to rather interact with a SBP dependent ABC transport system as this protein family represents the largest SBP dependent protein group in E. coli (Moussatova et al. 2008).
Baleen whales (Mysticeti) are a clade of highly adapted carnivorous marine mammals that can reach extremely large body sizes and feature characteristic keratinaceous baleen plates used for obligate filter feeding. From a conservation perspective, nearly all baleen whale species were hunted extensively over a roughly 100 years lasting time period that depleted many of the respective whale stocks with so far unknown consequences for e.g. their molecular viability. From an evolutionary perspective, the lack of fossil records together with conflicting molecular patterns resulted in a still unclear and debated phylogeny of modern baleen whales, particularly in rorquals (Balaenopteridae). In this dissertation, I will demonstrate the application of baleen whale genomes to tackle these open questions by using modern approaches of conservation and evolutionary genomics.
Conservation genomic aspects of baleen whales were addressed in two projects, both using whole genome data of either an Icelandic fin whale (Balaenoptera physalus) population or multiple blue whale (Balaenoptera musculus) populations to evaluate the impact of the industrial whaling era on their molecular viability. The results suggest a substantial drop in effective population size of both species but also a lack of manifestation in genotypes of the fin whale population when compared to the blue whale populations. Especially the rare and short runs of homozygosity (ROH), usually indicative for inbreeding, suggest frequent outcrossing in fin whales while all analyzed blue whale populations featured long and frequent ROH. In addition to these analyses, genome data of blue whale populations was further used to evaluate if northern hemisphere blue whales diverged into different subspecies. Population genetic and gene flow analyses showed clearly separated and well isolated populations in accordance with their assumed geographical distance. In contrast, the genome-wide divergence between all blue whale populations was low compared to other cetacean populations and to the next closely related sei whale species. Because this includes the morphologically different and well recognized pygmy blue whale subspecies, a proposal was made to equally categorize the two northern-hemisphere blue whale populations as subspecies.
Evolutionary aspects were addressed in a third project, by constructing the genome of the pygmy right whale (Caperea marginata) and testing its potential in phylogenetics and cancer research. Phylogenomic analyses using fragments of a whole-genome alignment featuring nearly all extant baleen whales, allowed the revision of the complex evolutionary relationships of rorquals by quantifying and characterizing the amounts of conflicts in early diverging branches. These relationships were further used to identify phylogenetically independent pairs of baleen whales with a maximum of diverging body size differences to compare rates of positive selection between their genomes. The results suggest nearly evenly distributed frequencies of alternative topologies which supports the representation of the early divergence of rorquals as a hard polytomy with high amounts of introgression and incomplete lineage sorting. Within the set of available genomic data, three independent pairs of baleen whales with diverging body sizes were found and comparisons of positive selection rates resulted in many potentially body size and cancer related genes. The lack of conserved selection patterns, however, suggest a more convergent evolution of size and cancer resistance like previously discussed in paleontology.
In conclusion, the application of whole genome data using methods of conservation genetics allowed for a comprehensive estimation about the molecular viability of blue and fin whales as well as an assessment of the taxonomic status of northern-hemisphere blue whale populations. The rather different results between blue and fin whales underlines the importance of genomic monitoring of baleen whales because different species show rather different molecular consequences of their potentially varying depletions. Furthermore, as showcased for the northern-hemisphere blue whale, many important isolated populations of baleen whales may still be unknown to conservation management and genome-wide comparisons will most likely contribute to overcome this under-classification problem. The application of whole genome data in evolutionary research allowed the characterization of the complex patterns of molecular conflicts within baleen whales and especially rorquals that will contribute to the still rather unclear understanding of their evolution. The here found molecular support for the idea of convergent evolution of gigantism in whales will further guide the search for molecular patterns responsible for Peto’s paradox.
Precise intensity monitoring at CRYRING@ESR: on designing a Cryogenic Current Comparator for FAIR

In the field of today’s beam intensity diagnostic there is a significant gap in the non-interceptive, calibrated measurement of the absolute intensity of continuous (unbunched) dc beams with current amplitudes below 1 μA. At the Facility for Antiproton and Ion Research (FAIR) low-intensity DC beams will occur during slow extraction from the synchrotrons as well as for coasting beams of highly-charged or exotic nuclei in the storage rings. The lack of adequate beam instrumentation limits the experimental program as well as the accuracy of experimental results.
The Cryogenic Current Comparator (CCC) can close the diagnostic gap with a high-precision dc current reading independent of ion-species and of beam parameters. However, the established detector design based on a core with high magnetic permeability and on a radial shield geometry has well-known weaknesses concerning magnetic shielding efficiency and intrinsic current noise. To eliminate these weaknesses, a novel coreless CCC with a co-axial shield was constructed and combined with a high-performance SQUID contributed by the Leibniz-Institute of Photonic Technology (Leibniz-IPHT Jena). The new axial CCC model was compared to a radial CCC with the established design provided by the Friedrich-Schiller-University Jena. According to numerical simulations prepared at TU Darmstadt and test measurements of the detectors in the laboratory, the new design offered a significant improvement of the shielding factor – from 75dB to 207dB at the required dimensions – and eliminated all noise contributions from the core material, promising an improved current resolution. Although the lower inductance of the pickup coil reduced the coupling to the beam significantly, the noise properties of the new CCC type were comparable to the classical version with a high-permeability core. However, the expected decrease of the low-frequency noise and thus an increase of the current resolution could not be observed at this stage of development.
Consequently, the classical CCC based on the radial shielding and high-permeability core had to be installed in CRYRING@ESR to provide best possible intensity measurements for the upcoming experimental campaign. In CRYRING the CCC was operated with beam currents between 1nA and 20μA and with different ion species (H, Ne, O, Pb, U). It was shown that the CCC provides a noise-limited current resolution of better than 3.2 nArms at a bandwidth of 200 kHz as well as a noise level below 40 pA/√Hz above 1 kHz. During the operation, the main noise sources of the accelerator environment had to be identified and suitable mitigation strategies were developed. Temperature and pressure fluctuations were suppressed with a newly-designed cryogenic support system based on a 70 l helium bath cryostat, developed and built in collaboration with the Institut für Luft- und Kältetechnik Dresden, in combination with a helium re-liquefier. The cryogenic operating time was restricted to around 7 days, which must be expanded significantly in the future. Digital filters were developed to remove the perturbations of the helium liquefier and of the neighboring dipole magnets. Given the promising results the CCC system can be considered as a prototype for future CCCs at FAIR.
Compaction and spheroid formation modulates stemness and differentiation of human pancreas organoids

The incidence of diabetes type 1 (T1D) in children and young adults is increasing worldwide. T1D is well treated by insulin administration. However, there is currently no long-lasting cure for this ailment. The success rate of pancreatic islet transplantation to treat T1D is limited by the availability of patient-matched islets and the necessity of using life-long immunosuppressive medication. The difficulties caused by transplantation can be overcome by generating bio-engineered pancreatic islets from patient-derived progenitor cells. Aim of this thesis is to establish new strategies for the generation and analysis of pancreatic lineages derived from human progenitor cells. It reports on the optimization of a technique to form human pancreatic spheroids from hollow monolayered human pancreas organoids (hPOs) to investigate how cell-cell and cell-matrix interaction can be leveraged to induce endocrine differentiation of the pancreas progenitor cell organoids. We introduce cell aggregation protocols to generate endocrine pancreas cell lineages from ductal pancreatic cells. Next, we study the effect of co-culture with stromal and endothelial cells to promote cell differentiation toward a pancreatic fate enhancing β cells productivity.
This thesis has focused on identifying the differences in gene expression along with phenotypical transformation during differentiation of human pancreatic organoids (hPOs) towards human β cells to be used in the future of cellular therapeutics in treating T1D patients.
Gastroschisis und Omphalozele zählen zu den häufigsten angeborenen Bauchwanddefekten. Dabei wird zwischen einfacher (ohne sekundäre Darmveränderungen) und komplexer (mit sekundären Darmveränderungen) Gastroschisis unterschieden. Bei der Omphalozele ist es wichtig, ob eine Protrusion der Leber vorliegt. Die Therapie beider Fehlbildungen besteht aus der Reposition der Bauchorgane in den Bauchraum und einem operativen Bauchdeckenverschluss in den ersten Lebenstagen.
Daten zu Langzeitverläufen nach dem operativen Bauchdeckenverschluss, insbesondere in Hinblick auf gastrointestinale Komplikationen mit erneutem chirurgischen Interventionsbedarf, sind in der Literatur rar.
Ziel dieser Studie ist es daher, die Inzidenz und die Art von operativen Eingriffen nach dem Bauchdeckenverschluss bei Patienten mit einer Gastroschisis und einer Omphalozele nach einem Bauchdeckenverschluss am eigenen Patientenkollektiv zu untersuchen. Hierzu wurden die Akten aller Patienten mit Gastroschisis und Omphalozele, die in der Klinik für Kinderchirurgie und Kinderurologie des Universitätsklinikums Frankfurt von 2010 bis 2019 behandelt wurden, retrospektiv ausgewertet. Die Inzidenz von Operationen nach Bauchdeckenverschluss wurde anhand der mittleren kumulativen Ein-Jahres-Anzahl an Operationen pro Patienten und der kumulativen Ein-Jahres-Inzidenz der Operationen ermittelt.
Insgesamt wurden 61 Patienten identifiziert. Nach Ausschluss von vier Patienten (Krankenhausverlegung (ein Patient) und Versterben (drei Patienten)) konnten die Verläufe von 33 Patienten mit Gastroschisis (18 mit einfacher und 15 mit komplexer Gastroschisis) und 24 Patienten mit Omphalozele (je zwölf mit und ohne Leberprotrusion) ausgewertet werden.
Bei 23 Patienten mit Gastroschisis und bei 20 Patienten mit Omphalozele kam es innerhalb des ersten Jahres nach Bauchdeckenverschluss zu erneuten Operationen. Das Risiko sich einer erneuten Operation unterziehen zu müssen, war bei Patienten mit komplexer Gastroschisis signifikant höher als bei Patienten mit einfacher Gastroschisis (kumulative Ein-Jahres-Inzidenz: 64,3% vs. 24,4%; p= 0,05). Zwischen den beiden 6 Formen der Omphalozele bestand kein Unterschied in der Inzidenz chirurgischer Eingriffe. Im Median kam es bei Patienten mit Gastroschisis nach 84 Tagen und bei Patienten mit Omphalozele nach 114,5 Tagen zu einer erneuten Operation.
74% der Patienten mit Gastroschisis und 30% der Patienten mit Omphalozele wurden aufgrund einer gastrointestinalen Komplikation operiert. Die Patienten mit komplexer Gastroschisis hatten im Vergleich zu Patienten mit einfacher Gastroschisis ein signifikant erhöhtes Risiko für eine Operation zur Behandlung einer gastrointestinalen Komplikation (kumulative Ein-Jahres-Inzidenz 64,3% vs. 11,1%; p= 0,015). In Bezug auf die Anzahl der Operationen pro Patienten und Jahr hatten die Patienten mit komplexer Gastroschisis und Patienten mit Omphalozele und Leberprotrusion die meisten operativen Eingriffe (kumulative Ein-Jahres-Anzahl 1,664 und 1,417 vs. einfache Gastroschisis (0,326) und Omphalozele ohne Leberprotrusion (0,333)). Dieses Verhältnis spiegelt sich auch in der kumulativen Ein-Jahres-Anzahl an Operationen mit gastrointestinalen Indikationen wider (komplexe Gastroschisis: 1,462 Operationen; Omphalozele mit Leberprotrusion: 0,500 Operationen; einfache Gastroschisis: 0,111 Operationen; Omphalozele ohne Leberprotrusion: keine Operation)
Nach dem Bauchdeckenverschluss kam es zu zwei Todesfällen bei Patienten mit komplexer Gastroschisis und zu drei Todesfällen bei den Patienten mit Omphalozele und Leberprotrusion. Bei den beiden Patienten mit komplexer Gastroschisis lag eine gastrointestinale Ursache vor (Leberversagen, Dünndarmvolvulus). Die Patienten mit der Omphalozele verstarben aufgrund assoziierter kardialer und pulmonaler Erkrankungen. Die Daten zeigen, dass Patienten mit Gastroschisis und Omphalozele im ersten Lebensjahr nach dem Bauchdeckenverschluss ein hohes Risiko für weitere Operationen haben. Patienten mit komplexer Gastroschisis und mit einer Omphalozele und Leberprotrusion haben das größte Risiko für eine erneute Operation nach Bauchdeckenverschluss. Bei Patienten mit einfacher Gastroschisis und Omphalozele ohne Leberprotrusion ist dieses Risiko gering. Die Mehrzahl der Operationen erfolgt aufgrund von gastrointestinalen Ursachen. Nur bei Patienten mit komplexer Gastroschisis tragen gastrointestinale Komplikationen zu einer erhöhten Mortalitätsrate bei. Die Ergebnisse dieser Arbeit können für Aufklärungs- und Beratungsgespräche von Eltern von Kindern mit Gastroschisis und Omphalozele herangezogen werden.
N6-methyladenosine (m6A) is the most abundant and well understood modification in eukaryotic mRNA and was first identified in polyadenylated parts of the mRNA.The distinct distribution of m6A in the transcriptome with special enrichment in long internal exons, 39UTRs and around stop codons was uncovered by early biochemical work and later on antibody based sequencing techniques. The so called m6A writer, reader and eraser machinery is responsible for the dynamic and with that regulatory nature of the m6A modification. As m6A writer, the human N6-methyltransferase complex (MTC) cotranscriptionally methylates the central adenine within a RRACH (preferably GGACU) sequence context to form m6A in the nascent RNA chain.9–15 The catalytic core of the complex is formed by the two proteins METTL3 and METTL14, with the active site located in the methyltransferase domain (MTD) of METTL3.16–18 The DPPW motif near the methyl donor S-adenosylmethionine (SAM) binding site in this MTD was postulated to bind the target adenine during catalysis. Moreover, a positively charged groove in the METTL3-METTL14 interface, the C-terminal RGG domain in METTL14 and the zinc finger motifs in METTL3 were identified as important domains for RNA binding. However, to date there are no full-length or substrate-RNA-bound structures of the catalytic METTL3-METTL14 complex.
In addition, a set of accessory proteins assembles to the METTL3-METTL14 heterodimer to form the full MTC, mediated by WTAP that firmly binds to the N-terminal leader helix in METTL3.20 WTAP was shown to locate the whole complex to the nuclear speckles and can modulate m6A deposition to specific sites in the RNA. Moreover, WTAP acts as binding platform for other accessory proteins including VIRMA, RBM15, ZC3H13 and HAKAI that are mostly identified to mediate position specific methylation. For example, RBM15 was shown to mediates region-selective methylation in a WTAP dependent manner, directing specificity towards U-rich sequences.
The observed specificity of the methyltransferase complex to methylate only site specific DRACH sequenced is still poorly understood. Some possible modulators like the role of the accessory proteins are under investigation, however, the structural context of the RNA methylation sites or a structural preference of the complex have been mainly neglected so far. Moreover, the structural dynamics of this methylation process still remain elusive. This thesis contributes to the afore-mentioned aspects by analysis of the methylation process regarding RNA structure sensitivity with enzymatic activity assays and its dynamic nature by implementing a smFRET approach.
We hypothesized the target RNA secondary structure to be an additional important modulator of methylation efficiency, based on the RNA binding elements of the complex (positively charged binding groove, zinc finger domain, RGG domain) and the supposed target adenine binding in the active site. Here, we postulated the possibility for a flipped-out adenine to be of special relevance, which is closely related to the local stability of the target adenine containing structure. Moreover, efficient binding of the protein complex to the RNA should require the ability to anchor the RNA on both sides of the target sequence.
Neuroendokrine Tumoren (NET) sind eine seltene Krankheit mit einem breitgefächerten heterogenen Erscheinungsbild, wodurch sich die Diagnose der Tumoren aus einer Vielzahl aus Gründen häufig um Jahre verzögert (1). In dieser Arbeit analysierten wir einen großen Datensatz in einem tertiären Referenzzentrum (UKF) von 1984-2019, um die Symptomatik vor der Diagnose des Tumors sowie den Zeitraum von der Tumormanifestation bis zur Diagnose weiter zu klären. Für die deskriptiven Analysen kamen SPSS, Cox-Regression und Log-Rank-Test zur Anwendung.
Insgesamt schloss die retrospektive Studie 488 gastroenteropankreastische (GEP)-NET mit 486 Patienten ≥ 18 Jahren ein, wovon knapp mehr als die Hälfte männlich (52,9%) waren. Das mittlere Alter bei Erstdiagnose (ED) betrug 58 Jahre (477/486, 9 unbekannt). Die häufigsten Primärtumorlokalisationen stellten Pankreas (143/488 Patienten) und Dünndarm (145/488 Patienten) dar. Die Mehrheit der NET waren langsam wachsende G1-Tumoren mit einem Ki67 < 3% (155/330). Die Hälfte der Patienten entwickelten im Verlauf Fernmetastasen, wobei die meisten bereits bei der ED vorlagen und insbesondere die Leber als Metastasierungsorgan dominierte. Bei mehr als 60% der Patienten konnten Angaben zur klinischen Symptomatik vor der ED detektiert werden, wovon wiederum mehr als die Hälfte symptomatisch waren. 42% der symptomatischen Patienten zeigten NET-spezifische Symptome (Bauchschmerzen 77/128; 60,2%, Durchfall 51/128; 39,8%, Flush 19/128; 14,8%, Karzinoidsyndrom 8/128; 6,3% Tachykardie 6/128; 4,7%). In der primären bildgebenden Diagnostik dominierten konventionelle Bildgebungen wie Sonographie und Computertomographie (CT), wobei nuklearmedizinische Diagnostik eine Seltenheit darstellte. Mehr als 30% der Tumoren wurden als Zufallsbefunde im Rahmen einer bildgebenden Diagnostik oder Operation diagnostiziert. Die Mehrheit der Patienten stellte sich initial außerhalb unserer Klinik vor, nur etwa 15% wurden innerhalb unserer Klinik insbesondere in der Gastroenterologie vorstellig, wo der NET diagnostiziert wurde.
Die Phase von der Tumormanifestation bis zur ED aller NET betrug im Median 17 Tage. Das Vorhandensein von Fernmetastasen sowie Symptomen führte zu keiner signifikanten Kürzung der Phase und einer schnelleren ED des NET (Median 65,5 vs. 90 Tage, p = 0,4).
Large international airports were identified as sources of ultrafine particles (UFPs) (Hu et al., 2009; Yu et al., 2012; Hsu et al., 2013; Keuken et al., 2015; Hudda and Fruin, 2016). Since September 2017 UFP emissions originating from the Frankfurt International Airport, Germany are monitored by the Hessian Agency for Nature Conservation, Environment and Geology (HLNUG) showing elevated UFP concentrations during airport operating hours (05:00–23:00 CET) (Ditas et al., 2022). Referring to that, the organic chemical composition of aviation-related UFPs emerging from the Frankfurt Airport was analysed by performing a comprehensive non-target screening of UFP filter samples.
Aluminium-filter samples were collected at an air quality monitoring station 4 km north of the Frankfurt Airport, using a 13-stage impactor system (Nano-MOUDI). The chemical
characterization of UFPs in the size range of 10-18 nm, 18-32 nm and 32-56 nm was accomplished by ultra-high-performance liquid chromatography, heated electrospray ionisation and mass analysis using an Orbitrap high-resolution mass spectrometer. Non-target screening revealed that the majority of detected compounds belong to homologous series of two different types of organic esters, which are base stocks of aircraft lubrication oils.
In reference to five different jet engine lubrication oils of various manufacturers, the corresponding lubricant base stocks and their additives, two amines and one organophosphate, were identified in the UFPs by the use of matching retention time, exact mass and MS/MS fragmentation pattern of single organic molecules. The quantitative analysis of the jet engine oil constituents in the aviation-related UFPs with diameters < 56 nm was accomplished by standard addition. By characterizing the Nano-MOUDI, loss factors for each size stage were determined and used for correction accordingly. Particle-number size distribution measurements, conducted parallel to the filter sampling, enabled the determination of the jet engine oil contribution to the UFP mass.
Furthermore, the nucleation and particle formation potential of a commonly used synthetic jet engine lubrication oil was investigated in the laboratory. Thermodenuder experiments at 20 °C and 300 °C were carried out to monitor the gas-to-particle partitioning behaviour of jet engine oils. At 300 °C a significantly higher number of particles with a mean diameter of ~10 nm are formed, leading to a more than fivefold increase in total particle numbers compared to 20 °C. Particle diameters of the newly formed oil particles in the laboratory experiment appeared in the same size region as UFPs emerging from Frankfurt Airport. Particles originating from the Frankfurt city centre direction showed larger diameters.
Results indicate that aircraft emissions strongly influence the total mass of 10-18 nm particles. The jet oil fraction decreases for bigger particles (e.g., 18-56 nm), implying that these oils form new particles in the cooling exhaust gases of aircraft engines. In addition, non-target screening and in vitro bioassays on aviation-related PM2.5 filter samples were combined to provide indications for potential toxicologically relevant compounds in dependence of different wind directions and airport operations. Most recently, the applied non-target screening method was also used to identify seasonal variations in the organic aerosol composition in Beijing.
The production of ribosomes is a complicated multistep, that is susceptible to changes occurring within the cell and its environment. The process itself requires many proteins, known as ribosome biogenesis factors (RBFs) and many non-coding RNAs like the small nucleolar RNAs (snoRNAs). While RBFs are required for the accurate processing of the pre-rRNA into mature rRNAs, the snoRNAs act to coordinate and guide enzymes for post-transcriptional modifications, chiefly 2´-O-ribose methylation and pseudouridylation. While ribosome biogenesis is mostly described in human and yeast model eucaryotes, similar detailed studies in the model plant Arabidopsis thaliana are far less explored and understood. Furthermore, for many experimentally confirmed modification sites the according snoRNAs and for many pre-rRNA processing steps the responsible RBFs are missing. Therefore, it is expected that a high number of snoRNAs and RBFs are not identified till yet. For this reason, RNA-deep sequencing was performed in order to identify novel snoRNAs and MS analysis data of nucleoli and nuclei of A. thaliana from a former PhD student were used in order to find new proteins involved in pre-rRNA processing.
In here, it is shown that with RNA deep-sequencing still new snoRNAs and snRNAs can be identified and that detection of predicted snoRNAs can be fulfilled with a) antisense oligonucleotides tagged with fluorescence dyes and b) with radioactive labeled antisense probes. Furthermore, a secondary structure map of the 60S and 40S subunit highlighting the predicted and moreover verified modification sites in 5.8S, 25S and 18S rRNA was created. Especially, the correlation between the modification sites and the guiding snoRNA is highlighted further shedding light on overview about current pre-rRNA modification sites and corresponding guiding snoRNAs. The next chapter reveals the complex and multi-layered existence of the 5.8S rRNA and its numerous precursors. The mutant prp24 (also known as seap1) encoding AtPRP24, is recognized as factor being important for splicing as it is promoting the recruitment of the U4 and U6 snRNAs to the spliceosome. In here, it was found that AtPRP24 is involved in processing of 5.8S rRNA precursors, recognizable by precursors that are over accumulating in the mutant. Moreover, it could be shown for the first time that the plant-specific precursor 5´-5.8S is exported to the cytoplasm, where final cleavage steps of 5.8S rRNA takes place. In the prp24.2 mutant, this precursor is exported at an increased rate to the cytoplasm, where it can be detected in the actively translating ribosomes (polysomes). A lower sensitivity of the mutant seeds to cycloheximide (CHX) suggests that due to the extension at the 5´-end of 5.8S, the structure of the 60S subunit has altered CHX binding. In conclusion, this work highlights the importance and complexity of 5.8S rRNA and its precursors for ribosome biogenesis and displays new insights into pre-rRNA processing in A. thaliana.
The formation of aliphatic α-amino acids by X-ray induced carboxylation of simple amines or amination of simple carboxylic acids is not favored over the formation of other amino acids. The new carboxylic and amino groups are more or less distributed statistically over the carbon atoms of the starting material. On radiationchemical formation of aliphatic hydrocarbons over C3, therefore, an increasing amount of unusual amino acids is produced. The results are influenced by various parameters such as temperature, pH, concentration, linear energy transfer and total dosis of radiation applied. Also peptides can be formed radiationchemically. However, the formation of greater molecules by radiationchemical processes under the conditions of a primitive earth seems to have only a low probability. The reaction mechanisms of radiationchemical carboxylation and amination are discussed.
Xylose, an abundant sugar fraction of lignocellulosic biomass, is a five-carbon skeleton molecule. Since decades, utilization of this sugar has gained much attention and has been in particular focus as a substrate for production of biofuels like ethanol by microbial hosts, including Saccharomyces cerevisiae. In this yeast, xylose is naturally not used as a carbon source, but its utilization could be achieved by metabolic engineering either via the oxidoreductive route or through the isomerase pathway. Both pathways share xylulose as a common intermediate that must be phosphorylated before entering the endogenous metabolism via the non-oxidative pentose phosphate pathway (noxPPP). Besides this, in some bacteria a non-phosphorylating oxidative pathway for xylose degradation exists, known as Weimberg pathway, where a molecule of xylose is converted by a series of enzymes - xylose dehydrogenase (XylB), xylonate dehydratase (XylD), 3-keto-2-deoxy-xylonate dehydratase (XylX) and α-ketoglutarate semialdehyde dehydrogenase (KsaD) - to form α-ketoglutarate (AKG). Besides having several useful properties as a product, AKG could also be used for cell growth as an intermediate of the tricarboxylic acid (TCA) cycle. One target of the present study is to establish a functional Weimberg pathway in S. cerevisiae. Previous studies have shown that this task is not trivial, for instance due to the toxicity of xylonate (the first metabolite of the pathway) and the involvement of an iron-sulfur cluster dependent enzyme, the D-xylonate dehydratase. The assembly of iron-sulfur clusters on a heterologous protein in yeast is known to be challenging.
To establish the Weimberg pathway in yeast, the genes xylB, xylD, and xylX were obtained from Caulobacter cresentus and ksaD was from Corynebacterium glutamicum. In a variant, the dehydratase xylD was replaced with orf41 from Arthrobacter nicotinovorans, which is believed to be independent of iron-sulfur clusters. Growth of yeast cells on xylose as a sole carbon source was expected as an indicator of a functional Weimberg pathway. However, the heterologous expression of the codon optimized genes was not sufficient to reach this goal. Due to the complexity of the interactions of the heterologous pathway with the endogenous cellular processes, it was assumed that potential limitations could be overcome by adaptive laboratory evolution, using xylose as a sole source of carbon. Increasing selection pressure was applied on a strain with Weimberg pathway genes integrated into the genome over several generations. As a variant of the evolutionary engineering approach, mutator strains were generated. For this, RAD27 and MSH2 genes were deleted, which are involved in nucleotide excision and mismatch repair mechanisms, respectively. Some of the resulting strains PRY24, PRY25, PRY27 and PRY28 were able grow in xylose as a sole carbon source after evolutionary engineering. As a control, a non-mutator strain PRY19 was also included. Strikingly, only the mutator strains were able to consume xylose as a sole carbon source, which shows the feasibility of the approach.
In addition to the mutator strain strategy, a further approach employed in the present study was the simultaneous expression of the Weimberg pathway in the cytosol and mitochondria. This was based on the reasoning that the iron-sulfur cluster biogenesis on XylD may be improved in the organelle and that the AKG is an intermediate of the TCA cycle. In the strain AHY02, all enzymes of the pathway were tagged with mitochondrial targeting signals in addition to a full cytosolically localized pathway. The localization of the mitochondrial variants was confirmed by fluorescence microscopy. Together with AHY02, CEN.PK2-1C wild type strain was also included as a control for evolution. When a selection pressure on xylose was applied, both strains - AHY02 and CEN.PK2-1C - were able to grow in the course of evolution. Deletion of the xylulokinase (XKS1) gene was found to be detrimental for both evolved strains in xylose-containing media. This suggests that the evolution of the endogenous oxidoreductive and noxPPP genes is responsible for growth of the evolved cells. For the evolved strain AHY02, it could also be possible that the Weimberg pathway genes supported to growth in addition to the oxidoreductive route. To elucidate the underlying molecular mechanisms, genome sequencing and reverse engineering approaches would be necessary in future.
In addition to screening for growth on xylose as a sole carbon source, a less stringent screening system was created to examine even a minor flux of xylose towards AKG. For this, all genes necessary for conversion of isocitrate to AKG where deleted, yielding a glutamate auxotrophic strain. In this system, the cells can grow on other carbon sources, whereas xylose is only provided as a source of AKG for the synthesis of glutamate...
This thesis deals with several aspects of non-perturbative calculations in low-dimensional quantum field theories. It is split into two main parts:
The first part focuses on method development and testing. Using exactly integrable QFTs in zero spacetime dimensions as toy models, the need for non-perturbative methods in QFT is demonstrated. In particular, we focus on the functional renormalization group (FRG) as a non-perturbative exact method and present a novel fluid-dynamic reformulation of certain FRG flow equations. This framework and the application of numerical schemes from the field of computational fluid dynamics (CFD) to the FRG is tested and benchmarked against exact results for correlation functions. We also draw several conclusions for the qualitative understanding and interpretation of renormalization group (RG) flows from this fluid-dynamic reformulation and discuss the generalization of our findings to realistic higher-dimensional QFTs.
The topics discussed in the second part are also manifold. In general, the second part of this thesis deals with the Gross-Neveu (GN) model, which is a prototype of a relativistic QFT. Even though being a model in two spacetime dimensions, it shares many features of realistic models and theories for high-energy particle physics, but also emerges as a limiting case from systems in solid state physics. Especially, it is interesting to study the model at non-vanishing temperatures and densities, thus, its thermodynamic properties and phase structure.
First, we use this model to test and apply our findings of the first part of this thesis in a realistic environment. We analyze how the fluid-dynamic aspects of the FRG realize themselves in the RG flow of a full-fledged QFT and how we profit from this numeric framework in actual calculations. Thereby, however, we also aim at answering a long-standing question: Is there still symmetry breaking and condensation at non-zero temperatures in the GN model, if one relaxes the commonly used approximation of an infinite number of fermion species and works with a finite number of fermions? In short: Is matter (in the GN model) in a single spatial dimension at non-zero temperature always gas-like?
In general, we also use the GN model to learn about the correct description of QFTs at non-zero temperatures and densities. This is of utmost relevance for model calculations in low-energy quan- tum chromodynamics (QCD) or other QFTs in medium and we draw several conclusions for the requirements for stable calculations at non-zero chemical potential.
Identification of new natural products from nematode-associated bacteria using mass spectrometry

This work aims to find unknown natural products produced by bacteria, that live in close association with nematodes and to elucidate their structure by using mass spectrometry.
The first chapter of this work is dedicated to the detection of hitherto unknown natural products by using a metabolomics approach and subsequent structure elucidation of said compounds. This chapter includes metabolomics analysis of Xenorhabdus szentirmaii wild type and knockout mutants, overproduction of the target compound, identification of derivatives from other strains and MS based structure elucidation.
The second and third chapters are about natural products that protect C. elegans from B. thuringiensis infections.
The second chapter deals with natural products that protect the nematode host without killing the pathogen. I deployed molecular biology methods to generate deletion and overproduction strains of a target compound, identified it via LC-MS/MS analysis and used LC-MS/MS and lipidomics to analyse the chemical properties of the active compound.
The third chapter aims at finding natural products, which are produced by Pseudomonas strains MYb11 and MYb12, respectively. These natural products display the ability to protect C. elegans by killing B. thuringiensis. I identified said compounds via fractionation and subsequent bioactivity testing. After identification, I generated production strains of the target compounds and elucidated the structure of the bioactive derivative.
The last chapter deals with the structure elucidation of peptides produced by an unusual GameXPeptide synthetase in Xenorhabdus miraniensis. I analysed producer strains of GameXPeptides using LC-MS and elucidated the structural differences between the known GameXPeptides, produced by P. luminescens TT01, and the unusual ones produced by X. miraniensis.
Investigation of the kinematics involved in compton scattering and hard X-ray photoabsorption

The present work investigates the kinematics of Compton scattering at gaseous, internally-cool helium and molecular nitrogen targets in the high- and the low-energy regime. Additionally, photoionization at molecular nitrogen with high-energy photons is investigated. These exeprimental regimes were previously inaccessible due to the extremely small cross sections involved. Nowadays, the third- and fourth-generation synchrotron machines produce sufficient photon flux, enabling the investiagtion of the above processes. The utilized cold-target recoil-ion momentum spectroscopy (COLTRIMS) technique further increases the detection efficiency of the observed processes, since it enables full-solid-angle detection by exploiting momentum conservation.
Compton scattering is investigated at both high (helium and N2) and low (helium) photon energies. In the high-energy regime, the impulse approximation is mostly valid, which is not the case for the low-energy regime. The impulse approximation assumes that the Compton-scattering process takes place at a free electron with a momentum distribution as if it was bound, thus ignoring the binding energy of the system. In the low-energy regime, the impulse approximation is not valid.
Photoionization is investigated at high photon energies, where the linear momentum of the photon cannot be neglected, as is the fashion of the commonly used dipole approximation.
Die vorliegende Dissertation mit dem Titel “Structural dynamics of eukaryotic H/ACA RNPs from Saccharomyces cerevisiae & Structural dynamics of the Guanidine-II riboswitch from Escherichia coli” besteht aus zwei Projekten. Das erste Projekt befasst sich mit den eukaryotischen H/ACA Ribonukleoproteinen (RNP) aus der Hefe. Diese können sequenzspezifisch in der RNA ein Uridin Nukleotid in das Rotationsisomer Pseudouridin (Ψ) umwandeln. Die H/ACA RNPs bestehen aus einer Leit-RNA und vier Proteinen, der katalytisch aktiven Pseudouridylase Cbf5, Nhp2, Gar1 und Nop10. Die Leit-RNA besteht in Eukaryoten konserviert aus zwei Haarnadelstrukturen, die von einem H-Box oder ACA-Box Sequenzmotiv gefolgt sind. In jeder dieser Haarnadeln befindet sich ein ungepaarter Bereich, die sogenannte Pseudouridylierungstasche, wo durch komplementäre Basenpaarung die Ziel-RNA gebunden wird. Fehlerhafte H/ACA RNPs können beim Menschen zu schweren Krankheiten wie verschiedenen Krebsarten oder dem Knochenmarksversagen Dyskeratosis congenita führen, aber sie bieten auch Möglichkeiten zum Einsatz als Therapiemethode. In dieser Arbeit wurde hauptsächlich der zweiteilige Aufbau der H/ACA RNPs untersucht.
Dafür wurden zunächst die einzelnen Komponenten hergestellt werden. Cbf5, Nop10 und Gar1 wurden zusammen heterolog in E. coli exprimiert und gereinigt. Außerdem wurden mehrere Deletionsvarianten von Gar1 hergestellt. Zusätzlich wurde die Leit-RNA unmarkiert über T7 Transkription synthetisiert, sowie sechs verschiedene FRET-Konstrukte mit verschiedenen Markierungschemas der Fluorophore Cy3 und Cy5 über DNA-geschiente Ligation. Anschließend wurde über Größenausschlusschromatographie und radioaktiven Aktivitätsassays geprüft, dass sich die aktiven H/ACA RNPs in vitro aus den einzelnen Komponenten rekonstituieren lassen.
In smFRET Experimenten wurden einzelne Haarnadelstrukturen mit dem zweiteiligen Komplexen verglichen. Dabei konnte gezeigt werden, dass die H3 Haarnadel durch die Anwesenheit von H5 dynamischer und heterogener wurde, während H5 überwiegend unbeeinflusst war. Außerdem konnte die dreidimensionale Orientierung der Haarnadelstrukturen in verschiedenen Assemblierungsschritten mittels smFRET untersucht werden. Hier deutete sich an, dass in Abwesenheit von Proteinen beide Haarnadeln eher entgegengesetzt stehen als in einer parallelen Konformation. Cbf5 scheint den Linker zwischen den Beiden auszustrecken bzw. zu orientieren und die Haarnadelstrukturen etwas gegeneinander zu neigen. Ein Zusammenspiel von Nhp2 und Gar1 war nötig um die oberen Bereiche der Haarnadeln zusammenzuziehen. Es konnte auch ein Modell für den vollen H/ACA RNP vorgeschlagen werden. Im kompletten Komplex könnte das Zusammenziehen der Haarnadelstrukturen durch Nhp2 und Gar1 mit dem Effekt von Cbf5 konkurrieren und könnte hauptsächlich den oberen Bereich von H3 betreffen. Zum Schluss wurde das Zusammenspiel von Gar1 und Nhp2 auf eine Abhängigkeit von den RGG Domänen von Gar1 hin untersucht. Hier besteht möglicherweise eine Hierarchie, die eine Kooperativität von den N- und C-terminalen Domänen benötigt.
Das zweite Projekt befasst sich mit dem Guanidin-II Riboschalter aus E. coli. Der Riboschalter kann das toxische Molekül Guanidinium (Gdm+) spezifisch in seiner Aptamerdomäne binden und dadurch die Genexpression von Proteinen zur Detoxifizierung von Gdm+ aktivieren. Der Riboschalter besteht aus zwei Haarnadelstrukturen, mit einer Schleife, die aus der Sequenz ACGR besteht, wobei R ein Purin ist. In einem vorgeschlagenen Modell soll die Ribosomenbindestelle (Shine-Dalgarno Sequenz) in Abwesenheit von Ligand mit dem Linker komplementär Basenpaaren und so die Translation verhindern. Mit Ligand würde sich dann eine Schleifen-Schleifen Interaktion mit den beiden CG Basen ausbilden, wodurch die Anti-Shine-Dalgarno Sequenz nicht mehr zugänglich wäre. Bisherige Studien arbeiteten zumeist nur mit der Aptamerdomäne, den einzelnen Haarnadeln oder noch kleineren Elementen. In dieser Arbeit wurden die Strukturdynamiken von verschiedenen Längen, auch mit der Expressionsplatform, untersucht. Außerdem wurden verschiedene Mutationen analysiert und die Effekte auf den Riboschalter in seiner natürlichen Umgebung in E. coli.
Zunächst mussten insgesamt 24 FRET-Konstrukte hergestellt werden, die sich in Länge, Markierungsschema und Mutationen unterschieden. Hierfür wurde DNA-geschiente Ligation verwendet. Dank der verschiedenen Fluorophorpositionen konnte ein konformationelles Modell für die Aptamerdomäne vorgeschlagen werden. In diesem Modell könnte in Abwesenheit von Ionen das Aptamer offen vorliegen. Durch Mg2+ würde sich bereits eine lockere Schleifen-Schleifen Interaktion ausbilden. Zusätzlich deuten die Ergebnisse auf eine neue Konformation hin, der stabilisierten Schleifen-Schleifen Interaktion, bei der der Linker zusätzlich mit den Haarnadelstrukturen interagiert, beispielswese mit den Purinen an der vierten Schleifenposition...
Adhesion to host cells is the first and most crucial step in infections with pathogenic Gram negative bacteria and is often mediated by trimeric autotransporter adhesins (TAAs). TAA-producing bacteria are the causative agent of many human diseases and TAA targeted anti-adhesive compounds might counteract such bacterial infections. The modularly structured Bartonella adhesin A (BadA) is one of the best characterised TAAs and serves as an attractive adhesin to study the domain-function relationship of TAAs during infection. BadA is a major virulence factor of B. henselae and is essential for the initial attachment to host cells via adhesion to extracellular matrix proteins. B. henselae is the causative agent of cat scratch disease and adheres to fibronectin using its long BadA fibres. The life cycle of this pathogen, with alternating host conditions, drives evolutionary and host-specific adaptations.
Human, feline, and laboratory adapted B. henselae isolates display genomic and phenotypic differences. By analysing the genomes of eight B. henselae strains using long-read sequencing, a variable genomic badA island with a diversified and highly repetitive badA gene flanked by badA pseudogenes was identified. Moreover, numerous conserved flanking genes were characterised, however, their influence on the regulation of badA expression and modification remains to be explored. It seems that B. henselae G 5436 is the evolutionary ancestor of the other B. henselae strains analysed in this work. The diversity of the badA island among the B. henselae strains indicates that the downstream badA-like domain region might be used as a ‘toolbox’ for rearrangements in the badA gene. Overall, it is suggested that badA-domain duplications, insertions, and/or deletions are the result of active phase variation via site-specific recombination and contribute to rapid host adaptation in the scope of pathogenicity, immune evasion, and/or enhanced long-term colonisation.
The model strain B. henselae Marseille expresses a badA gene that includes 30 repetitive neck/stalk domains, each consisting of several predicted structural motifs. To further elucidate the motif sequences that mediate fibronectin binding, various modified badA constructs were generated. Their ability to bind fibronectin was assessed via whole-cell ELISA and fluorescence microscopy. In conclusion, it is suggested that BadA adheres to fibronectin in a cumulative fashion with quick saturation via unpaired β-strands appearing in structural motifs present in BadA neck/stalk domains 19, 27, and other homologous domains. Furthermore, antibodies targeting a 15-mer amino acid sequence in the DALL motif of BadA neck/stalk domain 27 were able to reduce fibronectin binding of the B. henselae mutant strain S27. Moreover, this DALL motif sequence is conserved in the genome of all analysed B. henselae strains. The identification of common binding motifs between BadA and fibronectin supports the development of new anti-adhesive compounds that might inhibit the initial adherence of B. henselae and other TAA-producing pathogens during infection.
This thesis is focusing on the impact of Paratethys and Mediterranean water bodies over the Eurasian climate and the interplay between climate, tectonics and biosphere during the late Miocene. This target was the interval between 12.7 and 7.65 Ma for Paratethys, following the Eastern Paratethys restriction and isolation, and 7.2−6.5 Ma (the early Messinian) in Mediterranean, zooming on the effects of gateway restrictions over the eastern Mediterranean and the new born Aegean domain. In both cases restriction is overlapping with large scale climatic changes and tectonic reconfiguration, leading a sort of symbiotic relationship.
Paratethys was a giant epicontinental sea that covered a large part of Eurasia since Paleogene. Due to the Eurasia-Afro-Arabia collision and formation of the Alpine-Himalayan belt (Rögl, 1999; Popov et al., 2006), the Paratethys was divided during the late Miocene in smaller basins that in time were isolated of each other. The protracted isolation and intense continentalisation of paratethyan realm led to changes in humidity distribution, basin connectivity, sediment sources and salinity. These changes had in turn major consequences over water circulation, water availability, vegetation cover and biota. These changes are more intense after 11.6 Ma, when the Eastern Paratethys lost any sustained marine connection, evolving into an enclosed system with endemic fauna (Harzhauser and Piller, 2007).
Mediterranean Sea is a Mezozoic oceanic relic squeezed between Africa, Europe, Anatolia and Arabia, as Africa continued to subduct beneath the European plate. As opposed to Paratethys, it maintained the open connection with the ocean until Messinian, when the two Atlantic gateways (Betic and Rifian corridors) closed for a short time, isolating the basin. The cut off resulted in a dramatic drop down and onset of evaporitic precipitation in marginal basins, the event receiving the name of Messinian Salinity Crisis (5.97−5.55). The restriction affected all marine ecosystems, due to changes in salinity and stratification of water column.
The main objectives of this thesis were:
(1) build valid paleo-temperatures records for both basins based on biomarkers;
(2) reconstruct the hydrology for the late Miocene time interval;
(3) identify vegetation composition and changes;
(4) identify paleo-fires in the late Miocene sediment records;
(5) identify the biotic response to the overall climate and tectonic changes.
All the above objectives were attained with results published in specific journals (Chapters 5−7).
Based on Panagia section (Taman Peninsula, Russia) the longest Paratethys temperature record was completed (~5 Myr), covering the interval between 12.7 and 7.65 Ma. A comprehensive SST and MAT records was obtained, as well as soil pH and carbon (δ13C) and hydrogen (δ2H) stable isotopic compositions on n-alkanes and alkenones. The main findings are concentrated around three prolonged periods with severe droughts affecting the late Miocene circum-Paratethys region peaking at 9.65, 9.4 and 7.9 Ma, associated with a transition towards open land vegetation, intensification of fire activity and enhanced evaporation and aridity.
The time intervals with dryer conditions recorded in Panagia coincide with periods of mammal turnover and dispersal in Eurasia indicating that major environmental changes occurred in the circum-Paratethys region and Paratethys fragmentation had a great impact on the terrestrial ecosystems, when periods of prolonged droughts generated biotic crises and animal displacements across the Eurasian continent. The δ13CC29n-alkane values and charcoal morphologies from Panagia indicate an increased contribution of C4 plants adapted to drier conditions at 9.66 Ma. Similarly high δ13CC29n-alkane values continue until 9.4 Ma, when in Western Europe increased seasonality accelerated the demise of the evergreen subtropical woodlands and expansion of grasslands from Anatolia and Middle East to Europe.
As a result of basin fragmentation and climatic stress, the Eastern Paratethys sub-basins progressively lost their marine properties and turned into brackish-fresh water bodies fed primarily by riverine input. The shallower areas became in time emerged, obstructing connections and isolating the biota, inducing rapid adjusting or extinctions. Thus, the Paratethys harbored a highly endemic fauna (Rögl, 1999), such as dwarf whales, dolphins, seals (among mammals), as well as fish and other taxa (mollusks, ostracods, diatoms, foraminifera, algae, etc.).
Collectively the data structured and analyzed in chapter five support a model in which the Eastern Paratethys evolved as a largely (en)closed system, registering paleoenvironmental signals that are governed by interbasinal connectivity (or lack of it) and regional climate changes in the basin catchment. Acting as an important source of humidity for Western and Central Asia, the size and areal extent of the Paratethys water body is likely to have had a major impact on hydroclimate patterns in the Eurasian interior, with the cumulative fluctuations in both hydrology and surface temperature enhancing the aridity and seasonality, with different partition of moisture over the year. Our combined data suggests a decoupling of Paratethys from the global system as isolation advanced, dominated by regional tectonics and ultimately the Paratethys volume and areal extent reduction.
Sowohl die Lebenserwartung als auch die Prävalenz HIV-infizierter Patient*innen ist stetig ansteigend,aufgrund der HAART und durch verbesserte diagnostische Methoden. Nicht-AIDS-definierenden Erkrankungen sind heutzutage die führenden Todesursachen. Durch verzögerte Diagnosestellung und zurückhaltenden Therapien gynäkologischer Malignome ist die Prognose im Vergleich zur Normalbevölkerung schlechter.
In dieser retrospektiven Fall-Kontroll-Studie des Universitätsklinikums Frankfurt am Mains wurden die Therapie und das Outcome gynäkologischer Malignome von 23 HIV-infizierten Patientinnen aus den Jahren 2009-2019 mit einer Kontrollgruppe aus dem gynäkologischen Krebszentrum der Klinik Essen Mitte verglichen, um herauszufinden, inwiefern eine HIV-Infektion das Outcome der Patientinnen beeinflusst.
Das gynäkologische Malignom, dominierend das Zervixkarzinom, trat durchschnittlich ein Jahrzehnt nach der HIV-Diagnose auf. Im Unterschied zu anderen Studien, ist unser Kollektiv überwiegend hellhäutig. Ein bekannter Drogenabusus ist häufig und zusammenhängend mit weiteren Koinfektionen.
Die HIV-Erkrankung ist bei mehr als der Hälfte der Patientinnen bereits fortgeschritten, jedoch ließ sich kein Zusammenhang zwischen dem Auftreten gynäkologischer Malignome und einer CD4-Zellzahl <500 CD4-Zellen/µl nachweisen. Die antiretrovirale Therapie entsprach größtenteils nicht den aktuellen Leitlinien.
Bis auf fünf Frauen wurden alle Frauen leitliniengerecht therapiert. Eine Korrelation zwischen der Therapie und der Tumorentität, der CD4-Zellzahl, dem Alter oder dem Stadium des Malignoms konnte nicht gezeigt werden.
Insgesamt liegt die 5-Jahresüberlebensrate der Kohorte bei 74.8%. Eine nicht leitliniengerechte Therapie ist nicht direkt mit einem schlechteren Outcome verbunden, jedoch mit einem weitaus kürzeren Follow-Up-Zeitraum von durchschnittlich 0.22 Jahren im Vergleich zu 4.85 Jahren bei leitlinienkonform therapierten Patientinnen. Es liegt ein statistisch signifikanter Unterschied zwischen der Kontrollgruppe und unserer Kohorte vor, sodass angenommen werden kann, dass bei Vorliegen einer HIV-Infektion die Therapie des gynäkologischen Malignoms häufiger nicht leitliniengerecht ist.
Bislang existieren nur wenige Studien, die die Therapie und das Outcome gynäkologischer Malignome bei HIV-infizierten Patientinnen untersuchen. Die Interaktion einer ART mit antineoplastischen Medikamenten und die Anwendung von Checkpointinhibitoren und einer „targeted therapy“ sollten Gegenstand weiterer Untersuchungen sein. Dafür sollten HIV-Patientinnen in Therapiestudien inkludiert werden, sodass geeignete Leitlinien erarbeitet werden können.
Das Verständnis von Tumorerkrankungen wurde durch neue technologische und ökonomische Verbesserungen für die Next-Generation-Sequencing Analyse (NGS) gefördert. Die Komplexität der Interpretation genomischer Daten erschwert jedoch die Anwendung von NGS-Analysen im klinischen Kontext. Die Herausforderung besteht darin bei stetig wachsendem medizinischem Wissen dieses im klinischen Kontext zu interpretieren und eine personalisierte Therapieempfehlung abzugeben. Einen ressourcensparenden Ansatz können KI-unterstützende Software-Programme bieten, welche die genomischen Varianten mit der aktuellen Literatur vergleichen, eine Bewertung der Therapieoptionen geben und klinische Studien empfehlen können. In dieser retrospektiven Arbeit wurden Patient:innen mit metastasiertem Brustkrebs (n=77) mittels gezielter NGS-Analyse anhand von sogenannten Genpanels mit 126 bzw. 540 krebsrelevanten Genen im Zeitraum von 01/2019-02/2022 untersucht. Mit Hilfe von bioinformatischen Methoden wurden patientenspezifische genomische Veränderungen mit Behandlungsoptionen abgeglichen. Diese Methoden stützen sich vollständig auf öffentliche Datenbanken über somatische Varianten mit prädiktiver Evidenz für das Ansprechen auf bestimmte Medikamente. Diese Versorgungsforschung einer repräsentativen Kohorte des Universitätsklinikums Frankfurt in Kooperation mit Regionalverbund OncoNet Rhein-Main wurden systematisch ausgewertet inklusive der Bedeutung genomischer Varianten. Das OncoNet Rhein-Main ist eine Kooperation aus führenden onkologischen Zentren und Praxen im Rhein-Main-Gebiet, welche sich als Netzwerk der Aufgabe angenommen haben Patient:innen optimal therapeutisch zu versorgen. Für 51% (39/77) der Patient:innen konnte mindestens eine gezielte Therapieoption mit einem effektivem Biomarker im gleichen Tumortyp gemäß Zulassung der Europäischen Arzneimittelbehörde (EMA) gefunden werden.
Bei 12/77 (16%) wurde mindestens eine Alteration mit einem effektivem Biomarker und einer OFF-Label Therapieoption gefunden. Bei 30% der Patient:innen wurden Veränderungen in optionalen Biomarkern gefunden, welche Resistenzmechanismen erklären. Die umfassende molekulare Analyse von Patient:innen mit fortgeschrittenem Brustkrebs erlaubt die Behandlungsoption zu verbessern und ermöglicht durch die Analyse von bekannten Resistenzmarkern auch den klinischen Verlauf besser zu verstehen. Die interdisziplinäre Besprechung der Befunde im molekularem Tumorboard ist im Hinblick auf kontinuierliches Lernen aller Beteiligten sowie zur Qualitätssicherung eine entscheidende obligate Maßnahme.
Prevenzione e repressione nel contrasto al riciclaggio : un’indagine comparata tra Italia e Germania

The doctoral thesis provides a comparative study of the Italian and the German crime of money laundering. Although the topic of the essay is double, its final purpose concerns solely the Italian criminal provision (article 648-bis of the Italian Criminal code, art. 648-bis c.p.) in the attempt to thoroughly investigate its offensiveness, underlying legal interest and legitimation as well as to propose a restrictive interpretation.
The first chapter defines the research topic and the comparative methodology which shall be used throughout the thesis. The comparison is hereby intended as a tool to not only describe, but also contextualize the German legislation and create a “dialogue” with the italian criminal provision. Also, the first chapter lays down the three fundamental questions upon whom the analysis will be built (“from where?”, “what?” and “who?”) and offers a conceptual framework of the crime of money laundering by enlightening the existing liaison between this criminal offence and the paradigm of crime-prevention; European and supranational law; and the activities of prosecution and intelligence.
The second chapter delves into the question “from where?”, that is, the question of the role and the meaning of the predicate offence in money laundering’s prevention as well as repression. Therefore, the topic of the predicate crime is examined both from the angle of the anti-money laundering obligations for private actors and from the strictly penal angle. The question of the contribution of the predicate offence to money laundering’s offensiveness is specifically investigated with regard to both supranational and national law (including the most recent reforms that took place in Italy and Germany).
The third chapter is dedicated to the question of “what” is money laundering. This topic is first of all addressed from a pre-normative point of view. To this end, the chapter begins with a survey of relevant criminological theories that have been developed in order to explain, on an empirical basis, the phenomenon of money laundering and its effects on economy and society. The chapter then moves on to the criminal law provisions by examining the shape and current interpretation of the national crimes of money laundering and interrogating their suitability in the light of the criminological suggestions.
With a view to drawing some conclusions on the Italian crime of money laundering, the fourth and last chapter explores the question of “who” launders, meaning the possibility for the author of the predicate offence to be punished as self-launderer. In this context, self-laundering is examined as a form of both rationalization and extremization of the crime of money laundering. Lastly, the doctoral thesis summarizes its findings and proposes a restrictive interpretation of art. 648-bis c.p.
Magnetische Quadrupole und Solenoide sind ein elementarer Bestandteil einer Beschleunigeranlage und begrenzen die transversale Ausdehnung eines Teilchenstrahls durch eine Reflexion der Teilchen in Richtung der Beschleunigerachse. Die konventionelle Bauweise als Elektromagnet besteht aus einem Eisenjoch welches mit Spulen umwickelt ist. In dieser Arbeit werden diese Magnetstrukturen auf Basis von Permanentmagneten designt und hinsichtlich ihrer Qualität zum Strahltransport optimiert, sowie Feldmessungen an permanentmagnetischen Quadrupolen durchgeführt. Diese wurden mit 3D-gedruckten Halterungen aus Kunststoff gefertigt, was eine Vielzahl von Formvariationen ermöglicht. Darauf aufbauend wurde ein im Vakuum befindlicher Aufbau entwickelt, mit welchem die Strahlenvelope im inneren eines permanentmagnetischen Quadrupol Tripletts diagnostiziert werden kann. Dies greift auf ein am Institut für angewandte Physik entwickeltes System zur nicht-invasiven Strahldiagnose mithilfe von Raspberry Pi Einplatinencomputern und Kameras in starken Magnetfeldern zurück.
Die in dieser Arbeit vorgestellte Konfiguration eines PMQ’s ist eine Weiterentwicklung des am CERN im Linac4, einem Alvarez-Driftröhrenbeschleuniger zur Beschleunigung von H– , verwendeten Designs. Bei diesem sind je acht quaderförmige Permanentmagnete aus Samarium Cobalt (SmCo) in die Driftröhren des Beschleunigers integriert.
Darauf aufbauend wurden die geometrischen Designparameter hinsichtlich ihres Einflusses auf die Qualität des Magnetfelds untersucht. In einem magnetischen Quadrupol zur Strahlfokussierung wird dies durch einen linearen Anstieg des Magnetfeldes von Quadrupolachse zu Polflächen charakterisiert. Das Design wurde im Zuge dessen zur Verwendung von industriellen Standardgeometrien von Quadermagneten und der Erhöhung der magnetischen Flussdichte erweitert. Dazu wurde untersucht wie sich das Hinzufügen von zusätzlichen Magneten auswirkt und ob eine bessere Feldqualität durch andere Magnetformen erreicht wird.
Die Kombination mehrerer PMQ in geringem Abstand (<10 mm) führt abhängig von der Geometrie der PMQ-Singlets zu einer erheblichen Verschlechterung der Feldlinearität, was eine Erhöhung des besetzten Phasenraumvolumens der Teilchen nach sich zieht.
Am Beispiel von PMQ-Tripletts werden die zu beachtenden Designparameter analysiert und Lösungsansätze vorgestellt. Die auftretenden Effekte werden anhand von Strahldynamiksimulation veranschaulicht. Für eine Anwendung der vorgestellten Designs wurde eine Magnethülle mit einer Wabenstruktur zur Aufnahme der Einzelmagnete entwickelt. Diese besteht aus zwei Halbschalen, welche jeweils den Kompletteinschluss aller Magnete garantiert und eine einfache Montage um ein Strahlrohr ermöglicht. Diese wurden in der Institutswerkstatt aus Kunststoff via 3D-Druck gefertigt. Aufgrund der höheren erreichbaren Magnetisierung wurden Neodym-Eisen-Bor-Magnete (Nd2F14B, Br =1,36 T) für den Bau der entwickelten Strukturen verwendet. Für eine Magnetfeldmessung zur Bestätigung der magnetostatischen Simulationen und einer Bewertung der Druckqualität wurde eine motorisierte xyz-Stage zur Bewegung einer Hallsonde aufgebaut. Die Messungen zeigen eine gute Zentrierung des Magnetfeldes, sodass PMQ mit einer Kunststoffhalterung eine schnelle und billige Möglichkeit sind, kurzfristig eine Quadrupol-Konfiguration aufzubauen. Die Kosten belaufen sich für einen einzelnen PMQ je nach Länge auf 50€ bis 100€.
Basierend auf der PMQ-Struktur wurde ein PMQ-Triplett in ein Vakuum versetzt und mit Raspberry Pi Kameras im Zwischenraum der Singlets ausgestattet. Dies ermöglichte die Aufnahme der Strahlenvelope innerhalb des Tripletts anhand der durch einen Heliumstrahl induzierten Fluoreszenz und erste Erkenntnisse für notwendige Weiterentwicklungen wurden gesammelt. Auf den genauen technischen Aufbau wird im abschließenden Kapitel der Arbeit detailliert eingegangen.
In der einfachsten Form wird ein PM-Solenoid anhand eines einzelnen axial magnetisierten Hohlzylinders realisiert und erzeugt näherungsweise die Feldverteilung einer Zylinderspule. Durch die radialen Magnetfeldkomponenten an den Rändern des Solenoiden erhalten Teilchen eine tangentiale Geschwindigkeitskomponente und führen eine Gyrationsbewegung entlang der Solenoidachse aus. Diese reduziert den Strahlradius und die Teilchen behalten eine Geschwindigkeitskomponente, welche zur Solenoidachse zeigt. Für eine Maximierung dieser Fokussierung muss das Magnetfeld auf die Zylinderachse konzentriert werden. Insbesondere bei einer Verlängerung des Hohlzylinders wird die Kopplung der Polflächen über das Innenvolumen abgeschwächt. Aufgrund dessen wurde ein Design bestehend aus drei Hohlzylindersegmenten entwickelt. Dieses setzt sich aus zwei radial und einem axial magnetisierten Hohlzylinder zusammen und erhöht die mittlere magnetische Flussdichte für ausgewählte Geometrien um einen Faktor zwei im Vergleich zu einem einzelnen Hohlzylinder gleicher Geometrie. Dies ist gleichzusetzen mit einer Vervierfachung der Fokussierstärke, welche quadratisch mit der mittleren magnetischen Flussdichte skaliert. Die Strahldynamischen Konsequenzen werden anhand von Simulationen mit generierten Magnetfeldverteilungen erläutert. Für eine kostengünstige Bauweise wurde eine Design basierend auf quaderförmigen Magneten entwickelt.
Die Digitalisierung weiter Teile der Lebenswelt ist vor allem mit der Verbreitung und extensiven Nutzung des Smartphones verknüpft. Eine Mehrzahl der älteren Menschen in Deutschland nutzt Smartphones selbstverständlich im Alltag. Die vorliegende Studie adressiert erstens wie stark sich das Smartphone bereits in den Alltag älterer Menschen als vergleichsweise neue Technologie eingeschrieben hat. Im Mittelpunkt steht dabei, welche Funktionen genutzt werden, wie oft und wann zum Smartphone gegriffen wird und welche Gemeinsamkeiten und Unterschiede in der Nutzung identifizierbar sind. Darüber hinaus wird zweitens der Zusammenhang zwischen alltäglicher Smartphonenutzung, Medienkompetenz und technikbezogenen Einstellungen in den Blick genommen. Drittens geht die Studie der Frage nach inwiefern die Smartphonenutzung mit zentralen Variablen der Lebensqualität im Alter, der sozialer Eingebundenheit sowie dem subjektiven Wohlbefinden assoziiert ist. Die vorliegende Arbeit ist eine quantitativ empirische Studie, die einem mikrolängsschnittlichem Design folgt. Zur Beantwortung der Forschungsfragen wurden objektive Nutzungsdaten, Daten aus einem ambulanten Assessment sowie Daten eines querschnittlichen Fragebogens kombiniert. Die Ergebnisse zeigen, dass die Befragten älteren Menschen im Hinblick auf Nutzungsdauer, -intensität als auch inhaltlicher Vielfalt hohe inter- als auch intraindividuelle Unterschiede aufwiesen. Gemeinsamkeiten der Nutzung waren die Kommunikation oder Alltagsorganisation. Das Smartphone wurde aber auch zum Spielen oder für gesundheitsbezogene Handlungen genutzt. Gleichzeitig ging eine zeitlich intensive auch mit einer inhaltlich breit gefächerten Nutzung einher. Die Unterschiede in der Nutzung konnten primär durch technikbezogene Einstellungen und deklarative Wissensbestände zu Computer und Smartphone erklärt werden. Intensivnutzer wiesen positivere Einstellungen gegenüber Technik auf und verfügten über höhere Wissensbestände. Trotz intensiver Nutzung des Smartphones zur Kommunikation zeigte sich kein klarer Zusammenhang mit sozialer Eingebundenheit und subjektivem Wohlbefinden. Vielmehr konnte in diesem Kontext die wahrgenommene Qualität der erlebten sozialen Beziehungen als zentraler Prädiktor identifiziert werden. Zusammengenommen zeigte sich eine tiefe Integration des Smartphones in den Alltag älterer Smartphonenutzer.
Wir ermittelten die Geschwindigkeitskonstanten sowie die Arrhenius sehen Aktivierungsenergien und Aktionskonstanten der alkalischen Hydrolyse einer Reihe von Di- und Tripeptiden. Die zur Gewinnung der kinetischen Daten führende quantitative Analyse der Hydrolysate gelang, indem die Peptide vor ihrer Hydrolyse geeignet mit 14C markiert, die Hydrolysate mit Hilfe der Papierchromatographie bzw. der Zonenelektrophorese getrennt und die isolierten Hydrolyseprodukte auf Grund ihrer Radioaktivität quantitativ bestimmt wurden.
Der zeitliche Verlauf der alkalischen Hydrolyse von Di- und Tripeptiden wird durch ein Zeitgesetz 1. Ordnung bezüglich des Peptids und bezüglich der Hydroxylionen dargestellt. Die Abstufungen der Aktivierungsenergien, die den einzelnen Peptidbindungen in Di- und Tripeptiden zugeordnet sind, lassen sich durch die Annahme deuten, daß die Hydrolyse nach dem BAC 2-Mechanismus verläuft. Die Aktionskonstanten wachsen, wenn auch nicht regelmäßig, mit den Aktivierungsenergien.
Attention deficit hyperactivity disorder (ADHD) is a neurodevelopmental disorder that typically begins in childhood and is associated with the cardinal symptoms of inattentiveness, hyperactivity, and impulsiveness. In a significant number of cases, ADHD persists into adulthood and leads to profound psychosocial impairment and costs to the population. The course of the disorder and the severity of psychosocial impairment are further influenced by the presence of comorbidities. The risk of developing psychiatric comorbidities such as affective disorders, personality disorders and substance use disorders is increased compared to the general population. Studies also indicate that ADHD is associated with a higher burden of somatic disorders such as obesity, diabetes mellitus, asthma and migraine. In the last decades, there has been a growing body of research that identified sex-related differences in ADHD, but there is still insufficient evidence on specific issues. In addition to the sex-ratio, which is more balanced in adulthood compared to childhood, there are also indications that differences exist at the symptom level and that the comorbid disorders that occur more frequently in ADHD also seem to differ in men and women, although the studies are not yet clear on this. Using resting-state analyses of functional magnetic resonance imaging (fMRI), we aimed to address the question of whether we can detect sex differences in ADHD and selected comorbidities (substance use disorder, depression, obesity) based on altered functional connectivity profiles. A central role for the pathogenesis of ADHD is the dysregulation of dopaminergic neurotransmission, specifically altered reward processing, as an expression of impaired impulse control. In the present study, we focused on a neuroanatomical hub, namely, the external part of the globus pallidus (GPe), which we defined as a "region of interest" for the analyses performed. There is growing evidence that the globus pallidus not only plays a role in the extrapyramidal motor system, but also integrates cognitive and reward-related information, functions that are impaired in ADHD. In a first step, we looked for sex differences in ADHD patients (n=137) and separately in healthy controls (HC) (n=45), then we compared a similar group of HC and ADHD patients to compare sex-differences in ADHD patients and HC. In a second step, we investigated whether the neural basis of comorbidity patterns differed between male and female patients. Analysis of the images of 182 participants was performed using the SPM-based CONN toolbox V 18.b. When comparing subjects with ADHD and HC, we observed an interaction between the GPe and the middle left temporal gyrus, with the effect being more pronounced in healthy subjects. When analyzing the large ADHD sample, an interaction between the GPe and the frontal pole/middle right frontal gyrus was observed. The connectivity between the GPe and the frontal and temporal brain areas appeared to be more pronounced in female ADHD patients than in males, with the sex-effect being reversed and more pronounced in healthy subjects. The results suggest that in patients with ADHD there is a loss of sex-specialization in GPe-connectivity. Males with ADHD and depression showed lower functional connectivity between the GPe and parts of the occipital cortex than females with ADHD and depression. To our knowledge, this is the first study to investigate sex-specific functional connectivity networks using a seed-based connectivity analysis of the external globus pallidus in adult ADHD patients with and without comorbidities. The study serves to improve our knowledge of GPe involvement in ADHD and sex-specific recruitment of this network. Taken as a whole, this study contributes to our understanding of the neurobiological correlates of ADHD and suggests possible differences between males and females with ADHD centered on altered connectivity with the GPe, helping to provide a different perspective on current research and new ideas for further studies.
Locomotion, the way animals independently move through space by active muscle contractions, is one of the most apparent animal behaviors. However, in many situations it is more beneficial for animals to actively prevent locomotion, for instance to briefly stop before reorienting with the aim of avoiding predators, or to save energy and recuperate from stress during sleep. The molecular and cellular mechanisms underlying such locomotion inhibition still remain elusive. So, the aim of this study was to utilize the practical genetic model organism Caenorhabditis elegans to efficiently tackle relevant questions on how animals are capable of suppressing locomotion.
Nerve cells, mostly called neurons, are known to control locomotion patterns by activating some and inhibiting other muscle groups in a spatiotemporal manner via local secretion of molecules known as neurotransmitters. This study particularly focuses on whether neuropeptides modulate such neurotransmission to prevent locomotion. Neuropeptides are small protein-like molecules that are secreted by specific neurons and that act in the brain by activating G protein-coupled receptors (GPCRs) expressed in other target neurons. They can act as hormones, neuromodulators or neurotransmitters. DNA sequences coding for neuropeptides and their cognate receptors are similar across diverse species and thus indicate evolutionary conservation of their molecular signaling pathways. This could potentially also imply that regulatory functions of specific neuropeptides are also similar across species and are thus meaningful to unravel more general mechanisms for instance underlying locomotion inhibition.
Specifically, we find that the modulatory interneuron RIS constitutes a dedicated stop neuron of which the activity is sufficient to initiate rapid locomotion arrest in C. elegans while maintaining its body posture. Similar to its known function in larval sleep, RIS requires RFamide neuropeptides encoded by the flp 11 gene for this activity, in addition to GABA. Furthermore, we find that spontaneous calcium activity transients in RIS are compartmentalized and correlated with locomotion stop. These findings illustrate that a single neuron can regulate both stopping and sleeping phenotypes.
Secondly, we show that C. elegans RPamide neuropeptides encoded by nlp-22 and nlp-2 regulate sleep and wakefulness, respectively. We unexpectedly find that these peptides activate gonadotropin-releasing hormone (GnRH)-like receptors dose dependently and we highlight their sequence resemblance to other bilaterian GnRH-like neuropeptides. In addition, we show that these receptors are expressed in distinct subsets of neurons that are associated with motor behavior. Finally, we show that nlp 22 encoded peptides signal through GNNR 6 receptors to regulate larval sleep and that nlp 2 encoded peptides require both GNRR 3 and GNRR 6 receptors to promote wakefulness.
In sum, we find that locomotion inhibition in C. elegans is regulated by multiple, but evolutionary conserved RFamide and GnRH-like RPamide neuropeptidergic signaling pathways.
Caspase-2 is the evolutionary most conserved member of the caspase family and was shown to be involved in genotoxic stress induced apoptosis, control of aneuploidy, and ageing related metabolic changes. However, its role in apoptosis seems redundant due to the observation, that knockout does not inhibit apoptotic signalling exclusively. Instead, knockout of caspase-2 leads to tumor susceptibility in vivo, which led to the assumption, that caspase-2 has non-apoptotic functions and can act as a tumor suppressor. The underlying mechanism of the tumor suppressor activity of caspase-2 has not been clarified so far. Furthermore, caspase-2, has a prominent, and as pro-enzyme exclusive localisation in the nucleus and other subcellular compartments, implicating a distinct and location specific role.
In this study, a novel caspase-2 specific substrate, termed p54nrb, was identified. P54nrb is harbouring a caspase-2 specific cleavage site at the aspartate residue D422, and cleavage of p54nrb leads apparently to disruption of its putative DNA binding domain at the C-terminus.
P54nrb is a nuclear multifunctional RNA and DNA binding protein, known for roles in transcriptional regulation, DNA unwinding and repair, RNA splicing, and retention of defective RNA. Overexpression of p54nrb has been observed in several human cancers, such as cervix carcinoma, melanoma, and colon carcinoma.
Data from this study revealed, that depletion of p54nrb in tumor cell lines results in a loss of resistance to drug induced cell death and to reduced capability of anchorage independent growth, which is functionally equivalent to a reduced tumorigenic potential. Meanwhile, p54nrb depletion alone is not cytotoxic.
The investigation of p54nrb dependent gene regulations by high resolution quantitative proteomics uncovered an altering expression of multiple tumorigenic genes. For two of these candidates, the tumorigenic protease cathepsin-Z and the anti-apoptotic gelsolin, p54nrb dependent expression was detected universally in all three investigated tumor cell lines, cervix carcinoma, melanoma, and colon carcinoma. Additionally, a direct interaction of p54nrb with the cathepsin Z and gelsolin encoding DNA, but not with their corresponding mRNA, could be demonstrated.
Conjointly, this study unveils a novel mechanistic feature of caspase-2 as a tumor suppressor. The caspase-2—p54nrb axis can orchestrate the levels of several tumorigenic proteins and thereby determine the cell death susceptibility and long-term tumor survival. These findings might be of great value for future therapeutic interventions and for overcoming drug resistance of tumors.
The peptide loading complex (PLC) is a central machinery in adaptive immunity ensuring antigen presentation by major histocompatibility complex class I (MHC I) molecules to immune cells. If nucleated cells present foreign antigenic peptides from various origins (e.g., viral infected or cancer cells) on their cell surface they are targeted and eliminated by effector cells of the immune system to protect the organism against the hazard. The antigen presentation process starts with proteasomal degradation. Peptide loading and quality control of most, if not all, MHC I is performed by the PLC. Despite the main components, architecture, and general functions of this labile and multi-subunit assembly have been described, knowledge about the inner mechanics of MHC I loading and quality control in the PLC is limited. Detailed structural insights into the interactions and functions of key elements are lacking. In this PhD thesis, structural and functional aspects of the PLC in peptide loading and quality control of MHC I are unraveled, and the PLC was analyzed from an evolutionary perspective.
First, composition and architecture of native PLC isolated from different mammalian species was analyzed. Comparison of detergent-solubilized PLC from cow and sheep spleens with PLC isolated from human source showed a compositional conservation in mammals, with the central components TAP, ERp57, tapasin, calreticulin, and the MHC I heterodimer were conserved in these species. Negative-stain electron microscopy (EM) analyses revealed an identical overall architecture of PLCs from human, sheep, and cow with two major densities at opposing sides of the plane of the detergent micelle corresponding to endoplasmic reticulum (ER) luminal and cytosolic domains. Interestingly, the glucose-regulated protein 78 (GRP78) was associated only with the PLC from sheep and cow as revealed by mass spectrometry. This ER chaperone is involved in initial folding steps of MHC I but was not co-purified with human PLC, rendering it an interesting target for future functional and in-depth structural studies.
The human PLC was stabilized by reconstitution in membrane mimicking systems that replace the detergent, which is necessary to solubilize the complex. This stabilization allowed detailed structural analysis by single-particle cryogenic electron microscopy (cryo-EM). The structure of the MHC I editing module in the PLC, composed of tapasin, ERp57, calreticulin, MHC I, and β-2-microglobulin (β2m), was solved at an overall resolution of 3.7 Å. Within the structure, two important features were visualized: (i) the editing loop of tapasin, which is directly involved in peptide proofreading of MHC I; (ii) the A-branch of the Asn86 tethered N-linked glycan on MHC I. Both features are crucial elements in the quality control and peptide editing process on MHC I. The editing loop interacts with the peptide binding groove in MHC I. It disturbs the interaction between a cargo peptide C terminus and the F-pocket in the binding groove by displacing Tyr84 and the helices α1 and α2. The helix displacement widens the F-pocket which allows a faster peptide exchange on MHC I. The glycan is bound in its monoglucosylated form (Glc1Man9GlcNAc2) by the lectin domain of calreticulin. The A-branch of this glycan is stretched between MHC I Asn86 and the lectin domain, leading to the hypothesis that the glycan will be released from calreticulin once MHC I is loaded with a favored peptide (pMHC I).
For investigation of the glycan status of MHC I, intact protein liquid chromatography coupled mass spectrometry (LC-MS) was performed under denaturating conditions. An allosteric coupling between peptide loading and removal of the terminal glucose by α-Glucosidase II (GluII) was discovered. In addition, the PLC remained fully intact after peptide loading, which demonstrated GluII action on the PLC once MHC I is loaded.
With establishing GluII as transient interaction partner, this work deepens the knowledge of the molecular sociology of the PLC and how the PLC is involved in the endoplasmic reticulum quality control (ERQC). Further investigation of the ER aminopeptidases ERAP1 and ERAP2 showed that these enzymes neither alone nor together stably interact with the PLC. In contrast, both work independent from the PLC on free peptides in the ER.
LC-MS analysis of the PLC components revealed a very unusual glycosylation pattern of tapasin. Tapasin was observed with N-linked glycans ranging from the full glycan (Man9GlcNAc2) to heavily trimmed glycans, where only a single GlcNAc remained attached to Asn233. In the PLC, tapasin is probably shielded from degradation by ERQC and can remain functional and intact without a full N-linked glycan.
Es wurde das Leitfähigkeitsverhalten von reinem, lufthaltigem Wasser bei kontinuierlicher und impulsgetasteter Röntgenbestrahlung (60 kV8) untersucht. Hierbei ergaben sich zwei einander überlagerte Effekte: 1. Ein der Röntgen-Dosisleistung proportionaler irreversibler Leitfähigkeitsanstieg, der vermutlich auf eine Strahlenreaktion des gelösten CO2 zurückzuführen ist, 2. eine reversible Leitfähigkeitserhöhung während der Bestrahlung, die sich mit der Entstehung einer Ionenart mit einer mittleren Lebensdauer von ca. 0,15 sec erklären läßt. Es wird angenommen, daß es sich dabei um Radikalionen O2⊖ handelt, welche durch die Reaktion der als Strahlungsprodukt entstehenden Η-Radikale mit dem gelösten Sauerstoff gebildet werden. Ein möglicher chemischer Reaktionsmechanismus wird angegeben, der zu befriedigender quantitativer Übereinstimmung der Versuchsergebnisse mit Ausbeutewerten und Reaktionskonstanten aus der Literatur führt.
Auf der Oberfläche von Erythrozyten, Thrombozyten und Neutrophilen befinden sich mehrere hundert verschiedene polymorphe, ungekoppelt vererbte Blutgruppenantigene. Dementsprechend birgt jede Bluttransfusion das Risiko einer Immunisierung gegen fremde Blutgruppenmerkmale. Auch während der Schwangerschaft können aufgrund väterlich vererbter Antigene Alloantikörper induziert werden. Deshalb muss das Blut vor jeder Transfusion oder während einer Schwangerschaft auf das Vorhandensein irregulärer erythrozytärer Antikörper untersucht werden. Dabei greifen die aktuellen diagnostischen Verfahren auf primäre, stabilisierte Testerythrozyten von Blutspendern zurück, deren relevante Blutgruppenantigene bekannt sind. Antikörperspezifitäten können anhand von Agglutinationsreaktionen der Testzellen mit dem zu untersuchenden Patientenplasma auf ein oder mehrere Antigene zurückgeführt werden. Ist jedoch ein Antikörper gegen ein häufiges, ein hochfrequentes oder ein nicht-polymorphes, ubiquitäres Antigen gerichtet, kann in Ermangelung Antigen-negativer Testzellen keine adäquate Diagnostik gewährleistet, die Verträglichkeit der Transfusion also nicht definitiv sichergestellt werden. Auch der medizinische Einsatz therapeutischer Antikörper, welche Antigene adressieren, die auch auf Erythrozyten exprimiert werden, führt zunehmend zu Problemen. Tests auf granulozytäre Antikörper sind mangelhaft bezüglich ihrer Robustheit, besitzen eine unzureichende Auflösung und sind zudem meist zeitaufwändig und daher teuer. Antikörper gegen humane Plättchenantigene spielen insbesondere in der Schwangerschaft eine Rolle; sie vermögen bei Neugeborenen thrombozytopenische Blutungen bis hin zu massiven Hirnblutungen zu verursachen, die zu schweren Entwicklungsstörungen führen können. Bisher erfolgt jedoch mangels geeigneter Reagenzien keine standardisierte pränatale Untersuchung auf thrombozytäre Antikörper. In dieser Arbeit wurde ein neuartiges Verfahren für die Identifikation und Differenzierung irregulärer Blutgruppenantikörper etabliert, welches auf gentechnisch hergestellten, xenogenen Testzellen basiert, die einzelne definierte humane Blutgruppenantigene auf ihrer Oberfläche präsentieren. Die nicht humanen Zellen co exprimieren Fluorochrome, anhand derer Antikörper-markierte Testzellen durchflusszytometrisch voneinander unterscheidbar sind. Weiterhin können die generierten Testzellen zur Depletion von Antikörpern aus polyagglutinierenden Plasmen unter Erhalt der anderen Antikörperspezifitäten verwendet werden. Diese Technologie könnte die konventionelle Diagnostik erheblich erleichtern und bietet zudem die Möglichkeit, therapeutische Antikörper (wie z. B. anti-CD38, anti CD47, etc.), die häufig zu Interferenzen mit der Routinediagnostik führen, spezifisch prädiagnostisch aus Patientenproben zu entfernen.
Recent advances in artificial neural networks enabled the quick development of new learning algorithms, which, among other things, pave the way to novel robotic applications. Traditionally, robots are programmed by human experts so as to accomplish pre-defined tasks. Such robots must operate in a controlled environment to guarantee repeatability, are designed to solve one unique task and require costly hours of development. In developmental robotics, researchers try to artificially imitate the way living beings acquire their behavior by learning. Learning algorithms are key to conceive versatile and robust robots that can adapt to their environment and solve multiple tasks efficiently. In particular, Reinforcement Learning (RL) studies the acquisition of skills through teaching via rewards. In this thesis, we will introduce RL and present recent advances in RL applied to robotics. We will review Intrinsically Motivated (IM) learning, a special form of RL, and we will apply in particular the Active Efficient Coding (AEC) principle to the learning of active vision. We also propose an overview of Hierarchical Reinforcement Learning (HRL), an other special form of RL, and apply its principle to a robotic manipulation task.
Ziel: Ziel dieser Studie war es die Verweildauerraten, sowie die komplikationslosen Verweil-dauerraten von rein zahn, rein implantat und kombiniert zahn-implantatgetragenen Galvano-Konusprothesen auf keramischen Primärteilen im Oberkiefer zu untersuchen. Zusätzlich wurden durchgeführte Reparaturen, sowie die Patientenzufriedenheit ausgewertet.
Methode: 83 Patienten, welche im Zeitraum von 1999-2012 am ZZMK eine Galvano-Konusprothese im Oberkiefer erhalten haben wurden retrospektiv nachuntersucht. Die Patientenzufriedenheit wurde anhand eines Fragebogens erhoben, welcher mittels 22 Fragen die Bereiche „Ästhetik“, „Prothesenhalt-bzw. funktion“ und die „Reinigungsmöglichkeit der Prothese“ aus Patientensicht evaluiert. Als Zielereignis der Kaplan-Meier Verweildaueranalyse wurde die erste Reparatur, das Versagen der Prothese, sowie der Pfeiler- bzw. der Implantatverlust definiert. Reparatur- und Nachsorgemaßnahmen wurden deskriptiv erhoben.
Ergebnisse: Es konnten 83 Prothesen und 477 Pfeiler nachuntersucht werden. Der mittlere Beobachtungszeitraum betrug 3,9 Jahre. Die 3- ,5- und 10- Jahres Verweildauerraten der Prothesen lagen bei 98,2%, 95,5% bzw. 70,7%. Die konstruktionsbezogene Pfeilerüberlebensrate bis zum ersten Pfeilerzahnverlust betrug 98,2% nach 3 Jahren, 92,9% nach 5 Jahren und 29,2% nach 10 Jahren. Die häufigste Ursache eines Versagens war der Mehrfachverlust von Pfeilern (n=5). Eine Prothese versagte aufgrund eines Gerüstbruchs (n=1). Es konnte keine signifikanten Unterschiede in der Verweildauerrate bis zum Prothesenverlust, sowie in der komplikationslosen Verweildauerrate zwischen rein zahn, rein implantat und kombiniert zahn-implantagetragenen Prothesen festgestellt werden. Zu den häufigsten Reparaturen zählten Erweiterungen nach Pfeilerverlusten, Verblendreparaturen, Dezementierungen der Primärkronen und Frakturen von Prothesenzähnen. Die häufigste Nachsorgemaßnahme war die Druckstellenentfernung. Die Patientenzufriedenheit mit der Galvano-Konusprothese erwies sich als sehr hoch. Die Ästhetik, der Prothesenhalt und die Reinigungsmöglichkeit des Zahnersatzes wurden unabhängig der Verankerungsart durchgängig positiv bewertet.
Schlussfolgerung: Die Galvano-Konusprothese hat sich als hochwertiger herausnehmbarer Zahnersatz bewährt, welcher hohe Verweildauerraten aufweist und dessen Reparaturanfälligkeit vergleichbar mit anderen Doppelkronenversorgungen ist.
Die akute myeloische Leukämie (AML) ist eine aggressive Erkrankung des Knochenmarks, welche die Hämatopoese beeinträchtigt und zu Knochenmarksversagen führt. Trotz des Fortschritts in der AML-Therapie bleibt die Prognose für die meisten Patienten schlecht, sodass neue Therapieansätze für die Behandlung dringend benötigt werden. Autophagie, ein kataboler Abbauprozess von zellulären Komponenten, ist nachweislich an der Entstehung von AML beteiligt. Als zentraler Regulator von Zellüberleben, Homöostase und Stoffwechsel, dient die Autophagie als Nährstoffquelle durch die Wiederverwertung von Makromolekülen während begrenzter Energieversorgung. AML-Zellen benötigen ein konstantes Nährstoff- und Energieniveau, um ihre Vermehrung aufrechtzuerhalten. Dies wird durch eine Umstellung von Stoffwechselwegen, insbesondere des mitochondrialen Stoffwechsels einschließlich der oxidativen Phosphorylierung (OXPHOS) und des Tricarbonsäurezyklus (TCA), erreicht.
Mehrere Studien haben die Hemmung der Autophagie für die Behandlung von Krebs als vielversprechenden Ansatz vorgestellt. Doch eine Monotherapie mit Autophagie-Inhibitoren erzielte nur eine geringfügige Wirksamkeit. Eine mögliche Erklärung hierfür ist die Entstehung von Kompensationsmechanismen, die zum Ausgleich der Autophagie-Hemmung in Krebszellen entstehen. Bis heute sind diese Kompensationsmechanismen kaum untersucht. Ziel dieser Arbeit ist es, ein geeignetes Autophagie-Gen zu identifizieren, mit dem sich die Rolle der Autophagie-Hemmung für das Überleben von AML-Zellen untersuchen lässt. Zusätzlich sollen die kompensatorischen Mechanismen, die durch die Autophagie-Hemmung in AML-Zellen entstehen können, untersucht werden, um neue metabolische Angriffspunkte zu identifizieren, die für Kombinationstherapien genutzt werden können.
Zu Beginn der Arbeit wurde ein gezielter CRISPR/Cas9 Screen in zwei humanen AML-Zelllinien durchgeführt, um Autophagie-Gene zu identifizieren, deren Verlust eine Proliferationsstörung in AML-Zellen verursacht, welche überwunden werden kann. Validierungsexperimente zeigten, dass der Verlust von ATG3 das Zellwachstum signifikant verminderte. Außerdem zeigte die Messung des Autophagie-Fluxes, dass der Verlust von ATG3 die Autophagie stark beeinträchtigte. Dies wurde durch eine Western-Blot-Analyse, die eine beeinträchtigte LC3-Lipidierung zeigte, und durch eine Immunfluoreszenzanalyse der Autophagosomen-Bildung mittels konfokaler Mikroskopie, die eine geringere Anzahl von Autophagosomen in ATG3-defizienten Zellen ergab, bestätigt. Deshalb wurde der Knockdown von ATG3 in AML Zellen verwendet, um die Mechanismen, die zum Ausgleichen der Autophagie-Hemmung entstehen, zu untersuchen. Zuerst wurde die Zellproliferation in fünf verschiedenen AML Zelllinien über sieben Tage betrachtet. In allen Zellenlinien führte der Verlust von ATG3 mittels small hairpin RNA zu verminderter Zellproliferation. Diese Ergebnisse zeigen die wichtige Rolle von ATG3 in der Autophagie und dass Autophagie-Hemmung durch ATG3-Verlust das Wachstum von AML-Zellen beeinträchtigt.
Da der Verlust von ATG3 die Proliferation von AML-Zellen beeinträchtigte, wurde eine Zellzyklusanalyse durchgeführt. Eine reduzierte S-Phase bestätigte die verminderte Proliferation in ATG3-depletierten AML-Zellen, doch der Zellzyklus war grundsätzlich nicht gestoppt. Darüber hinaus ergab die Analyse der Apoptose, dass diese unter dem Verlust von ATG3 erhöht war, aber etwa 50% der Zellen blieben vital. Diese Beobachtungen deuten darauf hin, dass AML-Zellen trotz des Verlusts der ATG3-abhängigen Autophagie weiter proliferieren können.
Um die Mechanismen zur Kompensation der Autophagie-Hemmung zu untersuchen, wurden die Auswirkungen des ATG3-Verlusts auf die mitochondriale Homöostase untersucht. Die Mitophagie sowie das mitochondriale Membranpotenzial und die Masse unterschieden sich zwischen Kontroll- und ATG3-depletierten AML-Zellen nicht, was darauf hindeutet, dass die mitochondriale Homöostase durch den Verlust von ATG3 nicht beeinträchtigt ist. Als nächstes wurde die mitochondriale Funktion durch Messung des ATP-Spiegels und der OXPHOS untersucht. Die ATP-Level und die OXPHOS waren nach dem Verlust von ATG3 in AML-Zellen erhöht, was auf eine gesteigerte mitochondriale Aktivität bei Autophagie-Defizienz hinweist.
Hintergrund: Asthma ist die häufigste chronische Krankheit bei Kindern und Jugendlichen. Asthma-Exazerbationen sind ein häufiger Grund für Vorstellungen in der Notaufnahme und für Krankenhausaufenthalte. Die routinemäßige Gabe von Antibiotika bei einem akuten schweren Asthma wird von den Leitlinien nicht empfohlen, trotzdem erfolgt sie sowohl im ambulanten als auch im stationären Bereich. Das primäre Ziel dieser Promotion war es, die Häufigkeit der Gabe von Antibiotika während Asthma-Exazerbationen, die eingesetzten Antibiotikaklassen, den Einfluss des CRP Spiegels und die klinischen Kriterien, die zur Antibiotikagabe führten, zu analysieren.
Methoden: Es handelte sich um eine retrospektive Analyse von 660 Kindern und Jugendlichen im Alter von 6 bis 17 Jahren, die während eines Zeitraums von 10 Jahren (2008 - 2018) mit der ICD Diagnose Asthma Bronchiale (J.45) stationär am Universitätsklinik Frankfurt/Main stationär behandelt wurden. Akutes, schweres Asthma wurde definiert als Dyspnoe, Tachypnoe, Sauerstoffbedarf und/oder systemische Steroidtherapie. Antibiotikagabe ohne Indikation wurde definiert als Gabe von Antibiotika bei einem CRP <2 mg/dl, keiner radiologischen Evidenz einer Pneumonie, oder keiner Indikation zur Durchführung eines Röntgenbildes und keinem anderen Grund zur Antibiotikagabe. Analysiert wurden u.a. Alter, Geschlecht, Intensivstationsaufenthalt, Liegedauer, Erstdiagnose oder bekanntes Asthma, Sauerstoffbedarf, Laborparameter, Lungenfunktion, Röntgen, Phänotypen, Therapie und Anzahl der Re-Hospitalisierungen.
Ergebnisse: 350 Aufnahmen erfüllten die Einschlusskriterien. Es erfolgte bei jüngeren Kindern häufig eine Antibiotikagabe (6 - 11 Jahre 57,6% und 12 - 17 Jahre 39,7%). Bei 26% der Aufnahmen wurde ein Antibiotikum ohne Indikation verabreicht, dabei waren Makrolide die häufigste Antibiotikaklasse. Die Gruppe der PatientInnen, die ein Antibiotikum ohne Indikation erhielten, unterschieden sich von der Gruppe, die kein Antibiotikum erhielten in Alter (9 ± 6 Jahre vs. 11 ± 6 Jahre; p-Wert 0,024), Sauerstoffbedarf (46,2% vs. 26,3%; p<0.001) und CRP-Wert (0,79 ± 0,92 mg/dl vs. 0,23 ± 0,46 mg/dl; p<0.001), aber nicht im Vorhandensein von Fieber (14,3% vs. 10,6%; n.s.). 10,3 % der PatientInnen wurden in den 10 Jahren re-hospitalisiert. Es fanden sich klinisch keine Unterschiede zwischen den re-hospitalisierten PatientInnen und den anderen Patienten.
Schlussfolgerung: Antibiotikagabe ohne klare Indikation während einem akuten, schweren Asthma war in unserer Kohorte häufig. Klinische Parameter einer schwereren Erkrankung beeinflussten die Entscheidung zur Antibiotikagabe, obwohl keine Hinweise auf eine bakterielle Infektion vorlagen und nicht zu einem verbesserten Outcome führten. Gründe der Re-Hospitalisierung von PatientInnen sollten weiter untersucht werden, mit besonderem Fokus auf ausreichende Allergiediagnostik und Evaluierung der Compliance.
Bei über der Hälfte der Patienten mit einer operationsbedürftigen MI liegt gleichzeitig eine Insuffizienz der TK vor. In den meisten Fällen handelt es sich hierbei um eine funktionelle Insuffizienz welche aus einer RV-Pathologie, bedingt durch eine Volumen- oder Druckbelastung resultiert. In der Vergangenheit bestand die Überzeugung eine konservative Behandlung der TI sei ausreichend und nach Beheben der ursächlichen Grunderkrankung sei diese selbstlimitierend. Neuere Studien konnten jedoch belegen, dass eine bestehende TI auch nach operativer Versorgung einer ursächlichen LV-pathologie keine ausreichende Rückbildungstendenz aufweist, sondern im Verlauf sogar noch progredient ist. Die persistierende TI führt zu einer deutlich erhöhten Morbidität und Mortalität. Ist eine zweizeitige Operation an der TK im Verlauf erforderlich, so ist die Früh- und Spätmortalität deutlich erhöht und die Langzeitergebnisse sind schlecht. Diese Ergebnisse haben dazu geführt, dass die Indikation zur operativen Versorgung der TI ≥ 2 zu einem früheren Zeitpunkt und weniger restriktiv gestellt wird. Jedoch weisen die aktuellen ESC/EACTS-Guidelines insbesondere hinsichtlich der Versorgung einer TI < 2 nur einen niedrigen Evidenz-Grad auf, der richtige Operationszeitpunkt bleibt weiterhin umstritten. Der Grund für eine eher zurückhaltende Einstellung ist vor allem die Angst vor einer erhöhten Morbidität und Mortalität durch den additiven Eingriff. Zudem sind kaum Studien zu einer operativen Versorgung einer TI < 2 vorhanden. Ziel der vorliegenden Arbeit war daher der Gewinn weiterer Erkenntnisse, dieses bis dato kaum untersuchten Patientengutes, insbesondere hinsichtlich der peri- und postoperativen Mortalität, sowie der Auswirkungen der prophylaktischen TKR im Langzeitverlauf. Im Rahmen dieser monozentrischen, prospektiven Studie wurden 264 Patienten eingeschlossen, welche sich im Zeitraum von 2009 bis 2015 einer TKR im Rahmen einer MKR an der Klinik für Thorax- Herz- und thorakaler Gefäßchirurgie des Universitätsklinikums Frankfurt am Main unterzogen. Das Patientenkollektiv wurde nachfolgend in zwei Gruppen unterteilt, nach dem Schweregrad der präoperativ bestehenden TI in eine „prophylaktische” pTKR-Gruppe bei einer TI < 2 und eine „therapeutische” tTKR-Gruppe bei einer TI ≥ 2. Primärer Endpunkt war die Erfassung der Früh- und Spätmortalität. Sekundäres Endziel war die Untersuchung des postoperativen Verlaufes der TI, sowie der Verlauf der kardialen Funktion. Die 30-Tages-Mortalität betrug 12% in der pTKR-Gruppe und 17% in der tTKR-Gruppe. Der wichtigste Einflussfaktor war die EKZ-Dauer. Der Unterschied zwischen den beiden Gruppen war statistisch nicht signifikant. Im Vergleich zu anderen Studien zeigte sich eine höhere 30-Tages-Mortalität, jedoch handelte es sich bei diesen zumeist um einen isolierten Eingriff an der Mitralklappe, mit einer deutlich kürzeren EKZ. Hinsichtlich der Gesamtmortalität zeigte sich ein 1-, 5- und 7-Jahres-Überleben in der pTKR-Gruppe von 81%, 66% und 56%, sowie 65%, 52% und 41% in der tTKR-Gruppe. Die höhere Gesamtmortalität dieser Studie im Vergleich zu anderen Arbeiten ist durch das deutlich ältere und multimorbide Patientenkollektiv erklärt. Bei dem Vergleich von Patienten, welche im Rahmen dieser Studie eine prophylaktische TKR erhielten gegenüber den Patienten, bei welchen im Rahmen von Vergleichsarbeiten bewusst auf eine TKR verzichtet wurde (NTKR-Gruppe), zeigte sich ein verbessertes Langzeitüberleben der pTKR-Gruppe. Bei den Arbeiten, bei welchen kein Überlebensvorteil unserer pTKR-Gruppe, gegenüber deren NTKR-Gruppe gezeigt werden konnte, zeigten sich dennoch positive Effekte der begleitenden Klappenoperation. Die erhöhte Frühsterblichkeit dieser Arbeit ist dem Umstand geschuldet, dass durch die zur Indikationsstellung herangezogenen Risikofaktoren ein Hochrisikokollektiv selektioniert wurde, mit einer konsekutiv erhöhten Sterblichkeit auch abhängig vom Ausmass der TI. Bereits in der Vergangenheit konnte eine Verbesserung des Langzeitüberlebens durch die Durchführung einer TKR bei einer TI ≥ 2, gegenüber dem Verzicht auf diese nachgewiesen werden. Anhand des Vergleiches von anderen Arbeiten mit der vorliegenden Arbeit konnte dieser Überlebensvorteil auch für die Durchführung einer pTKR bei einer TI < 2 gezeigt werden. Zudem konnte dargelegt werden, dass es sich bei der begleitenden pTKR um eine effektive und dauerhafte Methode zur Vermeidung einer Klappeninsuffizienz handelt. Somit konnte gezeigt werden, dass auch bei Patienten mit einer TI < 2 eine additiven TKR die Entwicklung einer späten TI verhindert.
Die MicroRNA-193b (miR-193b) fungiert in Akuter Myeloischer Leukämie (AML) als Tumorsuppressor und als Biomarker zur Prädiktion der Überlebenswahrscheinlichkeit bei erwachsenen sowie kindlichen AML-Patienten. Die Expression der miR-193b ist in allen AML-Entitäten reduziert, mit Ausnahme der Akuten Promyelozyten Leukämie (APL), bei der das Level an miR-193b erhöht ist. APL ist auch die einzige Form der AML, bei der All-Trans-Retinoinsäure (ATRA) leit-liniengerecht und effektiv zur Therapie angewendet wird.
In Vorarbeiten konnte gezeigt werden, dass der antileukämische irreversible Inhibitor der Lysin-spezifischen Histon Demethylase 1 (LSD1) GSK-LSD1 die Expression von miR-193b induziert. Dadurch wird in murinen Homebox protein A9 (Hoxa9)/Meis Homebox Protein 1 (Meis1) AML-Zellen zumindest ein Teil der tumorsuppressiven Wirkung von GSK-LSD1 über die Hochregulation von miR-193b vermittelt.
Das Ziel dieser Arbeit bestand darin, die Gültigkeit des potenziellen GSK-LSD1-Wirkmechanismus über die Induktion von miR-193b in humanen Zellen zu untersuchen. Weitere Ziele waren die Erforschung der Rolle der miR-193b bei dem Behandlungseffekt von ATRA in humaner AML und APL sowie die Untersuchung einer potenziell protoonkogenen Eigenschaft der miR-193b in APL. Der methodische Ansatz dieser Arbeit basierte auf einer Senkung bzw. Erhöhung des intrazellulären miR-193b-Levels mittels Locked nucleic acids (LNA) bzw. lentiviraler miR-193b-Überexpression und anschließender Behandlung der Zellen mit den Wirkstoffen. Es wurden Proliferationskurven erstellt und der Differenzierungsgrad durchflusszytometrisch bestimmt. Eine hohe Aufnahmeeffizienz und Effektivität der LNA und des lentiviralen Überexpressionsvektors wurde durchflusszytometrisch und per quantitativer Polymerase-Kettenreaktion (qPCR) bestätigt.
Zur Untersuchung der Rolle der miR-193b bei dem Behandlungseffekt von GSK-LSD1 wurden die AML-Linien ML-2 und MOLM-13 sowie die APL-Zelllinie NB-4 mit LNA transfiziert bzw. mit dem Überexpressionsvektor transduziert und mit GSK-LSD1 behandelt. Die Populationen mit reduziertem miR-193b-Level wiesen nach 96 Stunden eine unverändert reduzierte Proliferation und Differenzierung auf. Hingegen zeigten miR-193b-überexprimierende Zellen nach GSK-LSD1-Behandlung einen signifikant stärkeren antiproliferativen Effekt. ML-2 Zellen wiesen zudem eine signifikant gesteigerte Differenzierung auf, gemessen an der Expression des Differenzierungsmarkers CD11b.
Die erhöhten miR-193b-Level in APL-Patienten konnten in der APL-Zelllinie NB-4 mittels qPCR bestätigt werden. NB-4 weist eine über 60-fach höhere miR-193b Expression als ML-2 (AML) auf. Zur Überprüfung einer onkogenen Eigenschaft der miR-193b in APL wurde das zelluläre miR-193b-Level in NB-4-Zellen durch LNA reduziert und anschließend die Proliferationskurve über 12 Tage bestimmt und die Differenzierungsmarker CD11b und CD13 gemessen. Proliferation und Differenzierungsgrad blieben nach Reduktion der miR-193b in NB-4-Zellen unverändert.
Die gleichzeitige Behandlung von NB-4 mit ATRA und miR-193b-LNA zeigte nach 96 Stunden den gleichen antiproliferativen und differenzierungsinduzierenden Effekt wie bei ATRA allein. Auch bei den AML-Zelllinien ML-2 und MOLM-13 wirkte die ATRA-Behandlung bei reduziertem miR-193b-Level unverändert differenzierungsinduzierend und antiproliferativ. Hingegen führte eine ATRA-Behandlung der AML-Zelllinien bei erhöhtem miR-193b-Level zu einer Verstärkung des antiproliferativen Effekts. Bei beiden AML-Zelllinien wurde die Proliferation mehr als doppelt so stark reduziert als bei alleiniger ATRA-Behandlung.
Diese Arbeit konnte zeigen, dass miR-193b kein Protoonkogen in der APL-Zelllinie NB-4 ist. Außerdem weisen die Ergebnisse dieser Arbeit darauf hin, dass die Wirkungseffekte von sowohl GSK-LSD1 als auch von ATRA, weder in APL noch in AML, über die Induktion der miR-193b vermittelt werden. Hingegen zeigt sich bei beiden Wirkstoffen in AML-Zellen ein additiver antileukämischer Effekt bei Behandlung mit gleichzeitiger Erhöhung des miR-193b-Levels. Die molekulare Begründung hierfür ist möglicherweise eine gemeinsame Modulation der Mitogen-aktivierten Protein Kinase (MAPK)-Signaltransduktionskaskade und bedarf weiterer Experimente zur Überprüfung dieser Hypothese. Ausblickend stellen die Ergebnisse dieser Arbeit die miR-193b als einen interessanten Kombinationspartner für die Therapie mit GSK-LSD1 oder ATRA, bzw. generell für den MAPK Signalweg beeinflussende Medikamente, dar, sobald eine sichere und effiziente Einschleusung von MicroRNAs in Patienten möglich ist.
In der vorliegenden Arbeit wurden selbstanordnende Monolagen (SAMs) entwickelt, welche als gassensitive Schichten auf Goldoberflächen mit kanzerogenem Benzol reversibel wechselwirken. Der Fokus lag dabei auf elektronenarmen Systemen und insbesondere auf hochfluorierten Aromaten. Insgesamt wurden 18 neuartige SAM-Präkursoren mit Substituenten, die starke Dipolmomente induzieren, hergestellt, die mit Hilfe von Thiolat- bzw. Selenolat-Ankergruppen an Goldoberflächen angebunden wurden. Die Charakterisierung dieser Schichten erfolgte mit diversen oberflächenanalytischen Methoden, wie Ellipsometrie und Infrarot-Reflexions-Absorptions-Spektroskopie (IRRAS). Für die Erforschung der Selektivität und Sensitivität der präparierten Monolagen wurden Austrittsarbeitsänderungen mit Hilfe einer KELVIN-Sonde gemessen und aus diesen Erkenntnissen systematische Veränderungen an den sensoraktiven Molekülen vorgenommen. So wurden der Fluorierungsgrad des aromatischen Systems, die Kettenlänge des Spacers sowie weitere vielversprechende Strukturmotive untersucht. Insbesondere hochfluoriere Derivate mit CF3-Substituenten zeigten die höchsten Sensitivitäten gegenüber Benzol. Weiterhin wurde ein neues Infrarot-Messprinzip entwickelt und etabliert, welches in situ Messungen während eines Überleitens gasförmiger Analyten ermöglichte. Diese Ergebnisse belegen eine Interaktion zwischen Analyten und Monolage.
Ein weiteres Projekt befasste sich mit elektronenarmen aromatischen Monolagen, die über Cystamin-Einheiten an Goldoberflächen angebunden wurden, sich aber in den dipolaren Kopfgruppen unterschieden. Dieses Vorgehen ermöglicht die Herstellung von SAMs gleicher Packungsdichten, sodass lediglich die unterschiedlichen Substituenten die elektronischen Eigenschaften beeinflussen. Die resultierenden Monolagen wurden mittels mit Rastertunnelmikroskopie, Ellipsometrie, IRRAS sowie Wasserkontaktwinkel-Goniometrie untersucht. Durch eine relativ einfache Veränderung der Kopfgruppe, wobei F3C , F3CS, H3C- sowie NC-Reste eingeführt wurden, war es möglich, die Austrittsarbeit einer Goldoberfläche in einem Bereich von 4.9 bis 5.6 eV einzustellen. Zusätzlich konnte ein gemischtes Disulfid synthetisiert werden, welches eine echte gemischte SAM auf der Goldoberfläche bildete. Dieses Konzept könnte in Zukunft eine noch präzisere Modifizierung der Austrittsarbeit zulassen, was die Entwicklung von effizienteren elektronischen Bauelementen auf Basis von organischen n Typ-Halbleitern ermöglicht.
Abschließend wurden teilfluorierte Benzonitril-Derivate mit Thiolat- bzw. Selenolat-Ankergruppen erforscht, welche als SAMs Informationen über den dynamischen Ladungstransport lieferten. Die Präkursoren unterscheiden sich lediglich im Fluorierungsmuster, welches das Dipolmoment der Gesamtstruktur beeinflusst. Die hergestellten Monolagen wurden mit einer Vielzahl oberflächenanalytischer Methoden (IRRAS, Ellipsometrie, Röntgenphotoelektronen- sowie Röntgen-Nahkanten-Absorptions-Spektroskopie) charakterisiert, die alle die Bildung geordneter SAMs belegten und auf eine eher aufrechte Orientierung der Moleküle auf der Oberfläche hindeuteten. Die Bestimmung der Elektronentransfer-Zeiten mit Hilfe der sogenannten core-hole-clock-Methode zeigte überraschenderweise, dass durch eine Änderung des Substitutionsmusters der Fluoratome der Elektronenübergang nicht beeinflusst werden. Dementsprechend hat das Dipolmoment innerhalb des molekularen Gerüsts keinen signifikanten Einfluss auf die Elektronentransfer-Dynamik.
Interactional niche in the development of geometrical and spatial thinking in the familial context

In the analysis of mathematics education in early childhood it is necessary to consider the familial context, which has a significant influence on development in early childhood. Many reputable international research studies emphasize that the more children experience mathematical situations in their families, the more different emerging forms of participation occur for the children that enable them to learn mathematics in the early years. In this sense mathematical activities in the familial context are cornerstones of children’s mathematical development, which is also affected by the ethnic, cultural, educational and linguistic features of their families. Germany has a population of approximately 82 million, about 7.2 million of whom are immigrants (Statisches Bundesamt 2009, pp.28-32). Children in immigrant families grow up with multiculturalism and multilingualism, therefore these children are categorized as a risk group in Germany. “Early Steps in Mathematics Learning – Family Study” (erStMaL-FaSt) is the one of the first familial studies in Germany to deal with the impact of familial socialization on mathematics learning. The study enables us to observe children from different ethnic groups with their family members in different mathematical play situations. The family study (erStMaL-FaSt) is empirically performed within the framework of the erStMaL (Early Steps in Mathematics Learning) project, which relates to the investigation of longitudinal mathematical cognitive development in preschool and early primary-school ages from a socio-constructivist perspective. This study uses two selected mathematical domains, Geometry and Measurement, and four play situations within these two mathematical domains.
My PhD study is situated in erStMaL-FaSt. Therefore, in the beginning of this first chapter, I briefly touch upon IDeA Centre and the erStMaL project and then elaborate on erStMaL-FaSt. As parts of my research concepts, I specify two themes of erStMaL-FaSt: family and play. Thereafter I elaborate upon my research interest. The aim of my study is the research and development of theoretical insights in the functioning of familial interactions for the formation of geometrical (spatial) thinking and learning of children of Turkish ethnic background. Therefore, still in Chapter 1, I present some background on the Turkish people who live in Germany and the spatial development of the children.
This study is designed as a longitudinal study and constructed from interactionist and socio-constructivist perspectives. From a socio-constructivist perspective the cognitive development of an individual is constitutively bound to the participation of this individual in a variety of social interactions. In this regard the presence of each family member provides the child with some “learning opportunities” that are embedded in the interactive process of negotiation of meaning about mathematical play. During the interaction of such various mathematical learning situations, there occur different emerging forms of participation and support. For the purpose of analysing the spatial development of a child in interaction processes in play situations with family members, various statuses of participation are constructed and theoretically described in terms of the concept of the “interactional niche in the development of mathematical thinking in the familial context” (NMT-Family) (Acar & Krummheuer, 2011), which is adapted to the special needs of familial interaction processes. The concept of the “interactional niche in the development of mathematical thinking” (NMT) consists of the “learning offerings” provided by a group or society, which are specific to their culture and are categorized as aspects of “allocation”, and of the situationally emerging performance occurring in the process of meaning negotiation, both of which are subsumed under the aspect of the “situation”, and of the individual contribution of the particular child, which constitutes the aspect of “child’s contribution” (Krummheuer 2011a, 2011b, 2012, 2014; Krummheuer & Schütte 2014). Thereby NMT-Family is constructed as a subconcept of NMT, which offers the advantage of closer analyses and comparisons between familial mathematical learning occasions in early childhood and primary school ages.
Within the scope of NMT-Family, a “mathematics learning support system” (MLSS) is an interactional system which may emerge between the child and the family members in the course of the interaction process of concrete situations in play (Krummheuer & Acar Bayraktar, 2011). All these topics are addressed in Chapter 2 as theoretical approaches and in Chapter 3 as the research method of this study. Die CMV-Infektion ist die häufigste opportunistische Infektion nach Nierentransplantation und wird mit einem erhöhten Risiko für Mortalität und Transplantatverlust assoziiert. Das Risiko an einer CMV-Infektion zu erkranken, wird vom Serostatus des Spenders und Empfängers beeinflusst. Dabei sind besonders Patienten mit Hochrisikokonstellation (D+/R–) betroffen. Bei seropositiven Transplantatempfängern (D+/R+, D–/R+) wird ebenfalls ein erhöhtes Risiko durch Reaktivierung des latent vorkommenden Virus oder Sekundärinfektion mit einem exogenen Virusstamm vermutet. Mithilfe von präventiven Maßnahmen wie der präemptiven und prophylaktischen Therapie kann die Rate an Infektionen gesenkt werden.
In dieser Studie wurden 441 Patienten, die in dem Zeitraum von 2006-2013 am Universitätsklinikum Frankfurt eine Nierentransplantation erhielten, retrospektiv untersucht. Ziel der Studie war es, den Einfluss des CMV-Serostatus, der Infektionsprophylaxe sowie der CMV-Infektion auf die Mortalität, das Transplantatüberleben und die Rejektion zu untersuchen. Da CMV-Infektionen meist in den ersten drei Monaten nach Transplantation oder nach Absetzen einer antiviralen Medikation auftreten, wählten wir eine zweijährige Verlaufsbeobachtung. Unsere Hypothese war, dass der Serostatus trotz regelhafter präventiver Maßnahmen mit einer Verschlechterung der Nierenfunktion assoziiert ist. Aufgrund neuer Empfehlungen wurde ab 2010 bei der Mehrheit der Transplantatempfänger eine Prophylaxe mit (Val)Ganciclovir durchgeführt. Folglich wurden die Zeiträume von 2006-2009 sowie von 2010-2013 miteinander verglichen.
In den Jahren von 2010-2013 konnten wir die höchste Infektionsinzidenz für die Hochrisikogruppe, gefolgt von der intermediären Risikogruppe (D+/R+, D–/R+), nachweisen. Beide Gruppen dokumentierten vermehrt CMV-Erkrankungen und Viruslasten im oberen Bereich (> 10.000 Kopien/ml). Diese zeigten jedoch keinen statistisch signifikanten Unterschied zu den anderen Serostatus-Konstellationen auf. Die Infektionsinzidenz in diesem Zeitraum war wiederum mit einem erhöhten Transplantatverlust assoziiert. Durch den prophylaktischen Einsatz von (Val)Ganciclovir konnte das Transplantatüberleben signifikant verlängert werden.
Im Vergleich der Zeiträume vor und nach 2010 stellte sich die D+/R+ Gruppe mit einem verbesserten Transplantatüberleben, die D–/R+ Gruppe mit verbessertem Patientenüberleben dar. Diese Gruppen konnten vermutlich von einer Ausweitung der Prophylaxe profitieren. Bezüglich der Rejektionen empfehlen wir für eine aussagekräftige Auswertung die Beachtung potenzieller Einflussgrößen wie die Immunsuppression und HLA-Merkmale. Rein deskriptiv lag das Auftreten von Rejektionen zeitlich vor den Infektionsereignissen. Die Niedrigrisikogruppe zeigte das geringste Abstoßungsrisiko.
Der Serostatus alleinig erscheint in unserer Studie keinen ausreichenden Hinweis auf das Outcome nach Transplantation zu geben. Allerdings waren CMV-Infektionen in den verschiedenen Zeiträumen maßgeblich mit einem verringerten Transplantat- oder Patientenüberleben assoziiert und sollten deshalb vermieden werden. Die Prophylaxe mit (Val)Ganciclovir stellte sich im gesamten untersuchten Zeitraum als effektive Therapie für ein verbessertes Transplantatüberleben dar.
Ein limitierender Faktor war die Vergleichbarkeit der Gruppen durch die unterschiedliche Patientenanzahl. Daten zur Infektion lagen uns erst ab dem Jahr 2009 vor. Bezüglich der Mortalität schätzen wir die Nachbeobachtungszeit von zwei Jahren zu gering ein, um signifikante Ergebnisse zu erzielen. Hervorzuheben ist, dass durch die Umstellung der Therapiemaßnahmen zwei interessante Patientenkollektive geschaffen wurden, an welchen die Auswirkungen der Prophylaxe dargestellt werden konnten. Mithilfe unserer Daten aus insgesamt acht Jahren mit zweijähriger Verlaufsbeobachtung, schätzen wir das am Universitätsklinikum Frankfurt durchgeführte Therapieregime nach 2010 als wirksame CMV-Prophylaxe ein. Um die Infektionsrate künftig weiterhin zu reduzieren, sind weitere Studien nötig, mit deren Hilfe Risikopatienten identifiziert, die Diagnostik erweitert und Therapien verbessert werden. Ansätze hierfür gibt es beispielsweise beim Einsatz des mTOR-Inhibitors Everolimus, der mit einer niedrigen Rate an CMV-Infektionen assoziiert wird. Auch die Anwendung neuerer Testmethoden, wie z. B. die Messung CMV-spezifischer T-Zellen, sollen dazu beitragen, Patienten mit erhöhtem Infektionsrisiko frühzeitig zu erkennen.
Epithelial cells enable essential physiological functions, including absorption, morphogenesis, secretion, and transport. To execute these functions, epithelial cells often form three-dimensional shapes that include curved sheets of cells surrounding a pressurized fluid-filled lumen. These three-dimensional tissues (called domes) are essential for organ function, but when they are not working properly, developmental defects, inflammation, and cancer can ensue. Recently, it has been shown that the cells that form domes show active superelasticity on micropatterned plates.
We show here that the immortalized renal proximal tubule epithelial cell line, LLC-PK1, stereotypically forms tubules in 10 days. Tubule formation takes place in 4 stages. When cells are plated on a culture dish, they form a monolayer on the 1st day; on the 3rd day, three-dimensional structures are formed, called domes; and after the 4.5th day, these domes start fusing to begin the transition stage and transit to the tubule stage. At the end of the 10th day, differentiated, elongated, and matured tubes form (Figure 3.1). Therefore, tubule formation is a self-organized, stereotypic morphogenetic program under long-term, unperturbed tissue culture conditions.
We propose that tubulogenesis is a two-step process in proximal tubules by doming and wrapping. The process begins with dome formation, and as the cell layers come together in the transition stage at the edge of the dome, this leads to the formation of the lumen of the eventual tubule. We also found that F-actin provides the mechanical strength during the formation of these three-dimensional structures during tubule formation. To better understand this 4-step process on a molecular level, we performed proteomics of tubule formation to identify the different proteins that play a significant role in proximal tubule development. Importantly, we identified proximal tubule markers like synaptopondin, angiotensin 1-10, collectrin, polycystin 1, and polycystin 2. These proteins play an important role in renal tube formation and differentiation.
Cell division is carried out by highly conserved cyclin-CDK complexes, which phosphorylate various cellular components. Cyclin-CDKs act differently depending on the cell cycle phase and work cooperatively to create DNA replication and cytokinesis. Therefore, we identified that cyclin-B1, marker of proliferation Ki-67, the RAD51 recombinase, and proliferating cell nuclear antigen (PNCA) are upregulated in the monolayer stage, and the expression decreases as tubule formation takes place. The proximal tubule reabsorbs 60-65% of the glomerulus filtrate. Therefore, it requires a lot of energy generated by using the fatty acid oxidation (FAO) pathway. In our model, we found FAO expression is higher than that of the other metabolic pathways.
We found expression of an intricate protein network in mitochondria, which we interpret as a sign of mitochondrial homeostasis being vital for the FAO pathway to work. Furthermore, we also identified different types of transporters at each stage of proximal tubule formation, and we could recognize different cytoskeletal components playing a significant role in each stage of proximal tubule formation, for instance, at the monolayer stage, vimentin expression is high, and its expression is reduced as tubules form. Hence, this 2D system, at this step of characterization, seems suitable to use to study differential transport protein expression and how this might relate to physiological functions and syndromes.
Next, we inhibited different transporters using specific inhibitors and analyzed the effect on dome and tubule formation. We identified that Na+/K+ ATPase and vacuolar H+ ATPase play a significant role in the process of epithelial dynamics. Digoxin (a Na+/K+ ATPase inhibitor) treatment inhibits dome and tubule formation. Bafilomycin (a v-ATPase inhibitor) treatment demonstrated a delay in dome and tube formation. Therefore, this study shows that this 2D proximal tubule novel system can be used for screening of pharmacological leads in the context of specific aspects of kidney physiology.
Despite the recent success in growing kidney organoids, they are not well suited to investigate various pathophysiological conditions in vitro for several reasons: They grow in 3D and form a tissue that later needs to be dissected/cleared and stained to investigate pathophysiological changes. Moreover, organoids require complex and expensive protocols for generation and are challenging to use in screening approaches. Therefore, we set out to demonstrate feasibility for our 2D system using normal renal epithelial cells, which are the origin of various pathological conditions, to study pathophysiological conditions.
This thesis investigates the structure of research articles in the field of Computational Linguistics with the goal of establishing that a set of distinctive linguistic features is associated with each section type. The empirical results of the study are derived from the quantitative and qualitative evaluation of research articles from the ACL Anthology Corpus. More than 20,000 articles were analyzed for the purpose of retrieving the target section types and extracting the predefined set of linguistic features from them. Approximately 1,100 articles were found to contain all of the following five section types: abstract, introduction, related work, discussion, and conclusion. These were chosen for the purpose of comparing the frequency of occurrence of the linguistic features across the section types. Making use of frameworks for Natural Language Processing, the Stanford CoreNLP Module, and the Python library SpaCy, as well as scripts created by the author, the frequency scores of the features were retrieved and analyzed with state-of-the-art statistical techniques.
The results show that each section type possesses an individual profile of linguistic features which are associated with it more or less strongly. These section-feature associations are shown to be derivable from the hypothesized purpose of each section type.
Overall, the findings reported in this thesis provide insights into the writing strategies that authors employ so that the overall goal of the research paper is achieved.
The results of the thesis can find implementation in new state-of-the-art applications that assist academic writing and its evaluation in a way that provides the user with a more sophisticated, empirically based feedback on the relationship between linguistic mechanisms and text type. In addition, the potential of the identification of text-type specific linguistic characteristics (a text-feature mapping) can contribute to the development of more robust language-based models for disinformation detection.
Due to their sessile nature, plants are constantly exposed to an everchanging environment. When these changes exceed certain limits, they can significantly impact plant growth and development, which, in case of crop plants, has consequences on food security. Exposure to high temperatures causes heat stress (HS), one of the most devastating stresses that plants can face. The survival and recovery from HS are dependent on the activation of the HS response (HSR), a collection of molecular mechanisms conferring HS tolerance by maintaining the cellular homeostasis. Stress responses follow a strictly orchestrated network of signal perception and -transduction, ultimately resulting in an adaptive cellular output. Thereby, the massive reshaping of the transcriptome plays a major part, in which heat stress transcription factors (HSFs) play the key role by inducing the expression of HS-responsive genes, including heat shock proteins and other transcription factors. Additionally, alternative splicing (AS), the selective usage of splice sites, contributes to the rapid adjustment of the transcriptome landscape by producing different mRNA variants from a single gene. Consequently, this results in the reduction of translatable transcripts by nonsense-mediated mRNA-decay or nuclear retention, but also enhances the proteome diversity by allowing the synthesis of protein isoforms with distinct functions. AS thereby modulates the activity of important regulatory factors like HSFA2 in Solanum lycopersicum (tomato). HSFA2 is the key factor of acquired thermotolerance (ATT), which enables the ability to survive a potentially lethal HS through pre-exposure to a preceding mild HS. Temperature-dependent AS leads to the synthesis of two HSFA2 protein variants, whereby inhibition of splicing ensures the synthesis of the stable isoform HSFA2-I that is required for ATT.
Transcriptome analysis of several plant species exposed to HS has highlighted the strong impact of high temperatures on the regulation of pre-mRNA splicing. Despite its importance, little is known about the molecular basis of the AS regulation in plants. Particularly for an economically important crop like tomato, understanding the regulation of HS-sensitive AS will contribute to the description of such an important regulatory mechanism but also might offer new insights for increasing HS resilience. Serine/arginine-rich proteins (SR proteins) are central regulators of constitutive and AS by modulating the splice site selection by the spliceosome. This study describes two members of the RS2Z subfamily of SR proteins in tomato, namely RS2Z35 and RS2Z36, which act as core regulators of AS under HS and consequently as central factors for thermotolerance. This study investigates the interaction of the two RS2Z proteins with the HSFA2 pre-mRNA and provides evidence for their function as splicing repressors in this particular AS event. Thereby, RS2Z proteins play an important role in the HSR by modulating the AS of the key factor of the ATT. Furthermore, based on global transcriptome analysis of knockout mutants of single or both RS2Z genes, it is demonstrated that RS2Z proteins are involved in the splicing of pre-mRNAs of almost 2000 genes. Moreover, RS2Z proteins act as splicing regulators and take part in a large portion of HS-induced AS events, thus playing a broader role in AS regulation. Furthermore, the HS-induced RS2Z36 is involved in basal thermotolerance (BTT), highlighting its importance for the basic HS resilience capacity of tomato. In addition, RNA sequencing demonstrates that RS2Z proteins–especially RS2Z36–regulate the expression of proteins involved in plant immunity. The study thereby provides experimental evidence for the important and essential role of SR proteins for plant thermotolerance and suggests the existence of RS2Z-mediated crossroads of different stress responses.
This thesis presents a first-of-its-kind phenomenological framework that formally describes the development of acquired epilepsy and the role of the neuro-immune axis in this development. Formulated as a system of nonlinear differential equations, the model describes the interaction of processes such as neuroinflammation, blood- brain barrier disruption, neuronal death, circuit remodeling, and epileptic seizures. The model allows for the simulation of epilepsy development courses caused by a variety of neurological injuries. The simulation results are in agreement with ex- perimental findings from three distinct animal models of epileptogenesis. Simula- tions capture injury-specific temporal patterns of seizure occurrence, neuroinflam- mation, blood-brain barrier leakage, and progression of neuronal death. In addition, the model provides insights into phenomena related to epileptogenesis such as the emergence of paradoxically long time scales of disease development after injury, the dose-dependence of epileptogenesis features on injury severity, and the variability of clinical outcomes in subjects exposed to identical injury. Moreover, the developed framework allows for the simulation of therapeutic interventions, which provides insights into the injury-specificity of prominent intervention strategies. Thus, the model can be used as an in silico tool for the generation of testable predictions, which may aid pre-clinical research for the development of epilepsy treatments.
Hintergrund: Der Rettungsdienst versorgt täglich viele Patient/-innen in unterschiedlichen Umgebungen und ist damit auch potentieller Überträger nosokomialer Infektionen. Zur Händehygiene, als entscheidende Säule der Infektionsprophylaxe, liegen bislang nur wenige Daten aus dem Rettungsdienst vor.
Methoden: Prospektive multizentrische Studie mit Fragebogen zur Selbst- und Fremdeinschätzung der Compliance und beeinflussender Faktoren (abgeleitet von der WHO Perception Survey for Health-Care Workers) sowie direkte Compliance-Beobachtung nach WHO-Standard bei Rettungsdienstpersonal zweier Berufsfeuerwehren in Deutschland.
Ergebnisse: Es wurden 207 Fragebögen eingereicht und während ca. 66h Beobachtungszeit wurden 674 Händedesinfektionsgelegenheiten protokolliert. Der präventive Effekt der HH wurde allgemein von den Mitarbeitenden anerkannt. Die Selbsteinschätzung (MW: 80%) und beobachtete Compliance-Rate (38%) zeigten eine deutliche Diskrepanz und die Compliance variierte zwischen den verschiedenen Indikationen. Besonders niedrig zeigte sich die Compliance rund um die Durchführung aseptischer Tätigkeiten. Hier zeichnete sich ein geringes Risikobewusstsein für nicht sichtbare Verunreinigungen ab. Hürden für die Umsetzung der Händehygiene stellten vor allem die Vorrangigkeit anderer Maßnahmen, Unterbrechung des Arbeitsablaufes und Zeitmangel dar.
Schlussfolgerungen: Die beobachtete Compliance-Rate im Rettungsdienst lag unterhalb der innerklinischen Durchschnittswerte. Insbesondere die Compliance im Rahmen aseptischer Tätigkeiten muss dringend gesteigert werden. Dies erfordert einen multimodalen Lösungsansatz, der die Optimierung der Ausbildung, Algorithmen, Materialverfügbarkeit und Praktikabilität der Händedesinfektion im Rettungsdienst beinhaltet.
In the recent past, we are making huge progress in the field of Artificial Intelligence. Since the rise of neural networks, astonishing new frontiers are continuously being discovered. The development is so fast that overall no major technical limits are in sight. Hence, digitization has expanded from the base of academia and industry to such an extent that it is prevalent in the politics, mass media and even popular arts. The DFG-funded project Specialized Information Service for Biodiversity Research and the BMBF-funded project Linked Open Tafsir can be placed exactly in that overall development. Both projects aim to build an intelligent, up-to-date, modern research infrastructure on biodiversity and theological studies for scholars researching in these respective fields of historical science. Starting from digitized German and Arabic historical literature containing so far unavailable valuable knowledge on biodiversity and theological studies, at its core, our dissertation targets to incorporate state-of-the-art Machine Learning methods for analyzing natural language texts of low-resource languages and enabling foundational Natural Language Processing tasks on them, such as Sentence Boundary Detection, Named Entity Recognition, and Topic Modeling. This ultimately leads to paving the way for new scientific discoveries in the historical disciplines of natural science and humanities. By enriching the landscape of historical low-resource languages with valuable annotation data, our work becomes part of the greater movement of digitizing the society, thus allowing people to focus on things which really matter in science and industry.
Hintergrund: Das Benigne Prostatasyndrom (BPS) stellt die klinische Ausprägung der Benignen Prostatahyperplasie (BPH), eine Volkskrankheit der männlichen Bevölkerung, dar. Die Prostataarterienembolisation (PAE) ist ein junges, minimal-invasives Therapieverfahren, bei dem die Gefäße, welche die Prostata versorgen, mittels Embolisaten blockiert werden. Die Blutversorgung des embolisierten Gewebes wird dadurch unterbunden und nekrotisiert in der Folge. Dieser Infarkt lässt sich über eine Diffusionsminderung in der Magnetresonanztomographie (MRT) nachweisen. Aufgrund des technisch anspruchsvollen Verfahrens und der Komplexität der prostatischen Gefäße findet die Magnetresonanzangiographie (MRA) immer mehr Beachtung zur strahlenfreien Visualisierung des Gefäßverlaufs. Das Ziel dieser Doktorarbeit bestand darin, mögliche prädiktive Faktoren für den Therapierfolg der PAE aufzudecken und dabei die Interventionsplanung, unter Anwendung und Nichtanwendung einer MRA, zu vergleichen.
Material und Methodik: In diese retrospektive Arbeit wurden 259 Patienten, die an dem Institut für Diagnostische und Interventionelle Radiologie der Universitätsklinik Frankfurt am Main eine PAE erhielten, eingeschlossen. Das Kollektiv wurde in zwei Gruppen, abhängig vom Vorhandensein einer präinterventionellen MRA, eingeteilt. Bei 137 Patienten wurde die PAE mit einer präinterventionellen MRA Rekonstruktion ausgeführt (= Gruppe A). Bei 122 Patienten erfolgte die PAE ohne vorangehende MRA Rekonstruktion (= Gruppe B). Mit Hilfe der Fragebögen International Prostate Symptom Score (IPSS), International Index of Erectile Function (IIEF) und Quality of Life (QoL) sowie MRT-Messungen (Diffusionskoeffizient, Prostatavolumen) wurden die Daten vor und nach der PAE evaluiert. Als statistische Methoden wurden die deskriptive Datenanalyse, der gepaarte Wilcoxon-Test, der Wilcoxon-Mann-Whitney-U-Test für nicht-parametrische Variablen sowie die Korrelationsmatrix nach Spearman verwendet.
Ergebnisse: In Gruppe A konnte eine signifikant höhere mediane Prostatavolumenreduktion (rho = 0,1827, p = 0,032448) und eine Reduktion der Strahlendosis (p = 0,000000) im Vergleich zu Gruppe B erreicht werden. In Gruppe A wurde im Verlauf eine signifikant höhere Abnahme des Diffusionskoeffizienten festgestellt als in Gruppe B (p = 0,036303). Ein infarziertes Areal nach der PAE wurde zu 59,3% (n = 67) von Patienten der Gruppe A erreicht, zu 40,7% (n = 46) erreichten dies Patienten der Gruppe B. Innerhalb der Gruppe A korrelierte der initial gemessene Diffusionskoeffizient mit dem präinterventionellen IPSS (rho = 0,3259, p = 0,0213). Weiterhin korrelierten bei Einsatz einer MRA der vor der Behandlung gemessene Diffusionskoeffizient sowie der IPSS mit der Abnahme der Beschwerden (rho = 0,3138, p = 0,0285; rho = 0,6723, p = 0,0000) und der Volumenreduktion (rho = 0,3690, p = 0,0024; rho = 0,4681, p = 0,0020). Die Reduktion des Diffusionskoeffizienten korrelierte mit der Höhe der initialen Symptomatik (rho = 0,3703, p = 0,0127) und mit der Verbesserung der Beschwerden (rho = 0,3830, p = 0,0107).
Schlussfolgerung: In Zusammenschau aller Ergebnisse sind die präinterventionelle Messung des Diffusionskoeffizienten, IPSS Erhebung sowie die Prostatavolumetrie in Verbindung mit der MRA prognostisch sinnvoll. Was die vorliegende Studie leistet, sind erste Erkenntnisse auf einem bisher kaum am Menschen untersuchten Gebiet. Dieser strahlensparende, prognostische Ansatz könnte zu einer besseren Patientenselektion beitragen und weitere Ansätze für den Einsatz von Künstlicher Intelligenz schaffen. Damit könnten Strahlendosis, Kosten und Risiken minimiert werden.
Die Paulinenpflege Winnenden e.V. wurde 1823 gegründet als Rettungshaus für verwahrloste Kinder. Bald wurde daraus eine Doppelanstalt einerseits für Kinder, die aus sozialen Gründen nicht zuhause leben konnten, andererseits für hör- und sprachbehinderte Kinder. Zum 200-jährigen Jubiläum im Jahr 2023 erscheint dieses wissenschaftliche Werk. Dieses Werk erscheint zusätzlich in weitgehend identischer Weise als Buch.
