Non-matrix-matched calibration of laser ablation ICPMS (trace/major) element data is a common quantification strategy. However, LA sampling is associated with downhole elemental fractionation, potentially causing inaccuracies if the magnitude of fractionation between the sample and reference material (RM) differs. Here, we estimate fractionation factors (FFs) for different elements (El) in a range of RMs relative to NIST SRM610/612 (FFEl/Ca-NIST) and evaluate element-specific corrections for downhole fractionation using these measured FFEl/Ca-NIST. Significantly different mean El/Ca values were observed before and after correction, particularly for the alkali elements (all RMs), and B, Fe, and Zn (some RMs), notably improving accuracy, especially for the alkali elements. In cases where this methodology does not result in an accuracy improvement, this may help identify underlying issues in reported/reference values for RMs, given that this phenomenon should be accounted for. Overall, we recommend considering routine assessment of FFs and applying a FF correction to enhance data quality.
Der Natrium-abhängige Kaliumkanal Slack (KNa1.1, Slo2.2, KCNT1) nimmt eine Schlüsselrolle in der Regulation neuronaler Erregbarkeit ein, indem er die Ausbildung und Feuerungsfrequenz von Aktionspotentialen kontrolliert. Sowohl in Mäusen als auch in Menschen wird Slack besonders hoch in nicht-peptidergen C-Faser-Neuronen exprimiert. Wissenschaftliche Erkenntnisse der letzten Jahre konnten die Beteiligung von Slack-Kanälen in der Signalverarbeitung neuropathischer Schmerzen, aber auch in verschiedenen Arten von Pruritus, feststellen. Dabei zeigen Slack-defiziente Mäuse ein verstärktes mechanisches Schmerzverhalten nach einer peripheren Nervenverletzung und ein erhöhtes Kratzverhalten in akuten Juckreiz-Modellen. Das als Slack-Aktivator identifizierte trizyklische Neuroleptikum Loxapin zeigt sowohl analgetische als auch antipruritische Effekte in Mäusen, jedoch ist sein klinischer Einsatz auf Grund schwerwiegender antipsychotischer Nebenwirkungen limitiert. Basierend auf Loxapins Leitstruktur wurden daher in dieser Arbeit neue Slack-Aktivatoren mit einem verbesserten pharmakologischen Profil designed und ihr Potential für die Therapie von Schmerzen sowie akutem und chronischem Pruritus in vivo untersucht.
The hepatitis B virus is one of the most common causes of virus-related chronic liver disease and remains a major global health problem affecting 296 million people worldwide. Despite an available and highly effective vaccination, hepatitis B infections lead to an annual mortality rate of approximately 0.8 million people. The global prevalence is heterogeneously distributed and reflects a high infections and chronicity, particularly in low-income countries, due to a lack of vaccination strategies, underdiagnosis and low treatment rates. A complete cure remains undiscovered to this day. Based on their genetic makeup, the virus is categorized into nine genotypes with a genetic difference of more than 8% within the sequence. In addition to their geographical distribution, hepatitis B virus genotypes also differ in terms of their clinical outcome, pathogenesis and treatment response.
The viral protein HBx is known to interact with several cellular signaling pathways and is thereby accounted as the driving force in the development of hepatitis B virus-associated pathogenesis and progression of hepatocellular carcinoma. In particular, HBx interacts with mitochondria and induces profound alterations in the mitochondrial morphology and function with a severe impact on the liver’s physiology and with an emerging role in liver-related disease progression.
This study aims to investigate the genotype-related impact of HBx with regard to their interaction with cellular signaling pathways. A particular focus was placed on mitochondria-dependent interactions and signaling pathways in order to broaden the understanding of the genetic diversity of the genotypes.
Differences between genotypes of HBx were examined and compared through in vitro experiments based on a cell culture-based system. Plasmid DNA encoding the HBx protein of the different genotypes was transiently transfected into Huh7 or HepG2 cells and examined for molecular and protein-biochemical effects on the host cell, usually 72 hours after transfection. This study focused on the most common genotypes A, B, C, D, E and G worldwide.
Based on initial kinome profiling analyses, it was found that HBx differs greatly within their genetic variants and suggests different effects on overall cell function and in particular on mitochondrial kinases. Furthermore, confocal laser scanning microscopy reveals profound HBx-mediated changes in the mitochondrial network structure, however with major differences among the different genotypes. In particular, HBx of genotypes A and G causes enormous fragmentation of mitochondrial structures, accompanied by emergent changes in mitochondrial function. Due to an increased interaction with the voltage-dependent anion channel 3, a significant loss of mitochondrial membrane potential was also observed, together with an increased radial oxygen stress level and an induction of central mitochondria-dependent inflammatory mediators. In contrast, the contribution of HBx-genotype B and E reveals only moderate effects in these regards. Using a pH-sensitive reporter system, HBx genotypes which previously indicated a strong distribution in the mitochondrial morphology and function, also showed an elevated mitophagy through the PINK1/Parkin-mediated pathway. This study provides direct evidence that HBx-mediated changes in host cell signaling pathways, especially in mitochondrial-associated pathways, fundamentally dependent on the different genotypes. In addition, the results also indicate an important role of HBx in the process of genotype-dependent liver pathogenesis and provide insight into the underlying cellular mechanisms and signaling pathways.

In vier Studien werden jüdische Erfahrungen der Moderne zwischen Zeigen und Beobachten, sozialer Schau und antisemitischer Bedrängnis als Modi des Theatralen zum Thema. Dabei rückt Theater als Praxis ins Zentrum, mit der Zugehörigkeit in der Moderne verhandelt, selbstbewusst behauptet oder befragt wurde. Während sich Juden und Jüdinnen "wie auf einer Bühne" permanenter Beobachtung ausgesetzt sahen, kommentierten sie dies wiederum theatral, erprobten eigene Strategien sozialer Teilhabe und beteiligten sich an der Ausgestaltung von bürgerlichen Sozialrollen in der Großstadt. Theater zeigt sich so als vielschichtige kulturelle Praxis wie als "Spiel- und Spiegelform des Lebens" (Stefan Zweig).
Stimulated emission depletion (STED) microscopy is a super-resolution technique that surpasses the diffraction limit and has contributed to the study of dynamic processes in living cells. However, high laser intensities induce fluorophore photobleaching and sample phototoxicity, limiting the number of fluorescence images obtainable from a living cell. Here, we address these challenges by using ultra-low irradiation intensities and a neural network for image restoration, enabling extensive imaging of single living cells. The endoplasmic reticulum (ER) was chosen as the target structure due to its dynamic nature over short and long timescales. The reduced irradiation intensity combined with denoising permitted continuous ER dynamics observation in living cells for up to 7 hours with a temporal resolution of seconds. This allowed for quantitative analysis of ER structural features over short (seconds) and long (hours) timescales within the same cell, and enabled fast 3D live-cell STED microscopy. Overall, the combination of ultra-low irradiation with image restoration enables comprehensive analysis of organelle dynamics over extended periods in living cells.
Correlative dynamic imaging of cellular landmarks, such as nuclei and nucleoli, cell membranes, nuclear envelope and lipid droplets is critical for systems cell biology and drug discovery, but challenging to achieve with molecular labels. Virtual staining of label-free images with deep neural networks is an emerging solution for correlative dynamic imaging. Multiplexed imaging of cellular landmarks from scattered light and subsequent demultiplexing with virtual staining leaves the light spectrum for imaging additional molecular reporters, photomanipulation, or other tasks. Current approaches for virtual staining of landmark organelles are fragile in the presence of nuisance variations in imaging, culture conditions, and cell types. We report training protocols for virtual staining of nuclei and membranes robust to variations in imaging parameters, cell states, and cell types. We describe a flexible and scalable convolutional architecture, UNeXt2, for supervised training and self-supervised pre-training. The strategies we report here enable robust virtual staining of nuclei and cell membranes in multiple cell types, including human cell lines, neuromasts of zebrafish and stem cell (iPSC)-derived neurons, across a range of imaging conditions. We assess the models by comparing the intensity, segmentations, and application-specific measurements obtained from virtually stained and experimentally stained nuclei and cell membranes. The models rescue missing labels, non-uniform expression of labels, and photobleaching. We share three pre-trained models (VSCyto3D, VSNeuromast, and VSCyto2D) and a PyTorch-based pipeline (VisCy) for training, inference, and deployment that leverages current community standards for image data and metadata.
Snapshots of acetyl-CoA synthesis, the final step of CO₂ fixation in the Wood-Ljungdahl pathway

In the ancient microbial Wood-Ljungdahl pathway, CO2 is fixed in a multi-step process with acetyl-CoA synthesis at the bifunctional carbon monoxide dehydrogenase/acetyl-CoA synthase complex (CODH/ACS). Here, we present catalytic snapshots of the CODH/ACS from the gas-converting acetogen Clostridium autoethanogenum, characterizing the molecular choreography of the overall reaction including electron transfer to the CODH for CO2 reduction, methyl transfer from the corrinoid iron-sulfur protein (CoFeSP) partner to the ACS active site and acetyl-CoA production. Unlike CODH, the multidomain ACS undergoes large conformational changes to form an internal connection to the CODH active site, accommodate the CoFeSP for methyl transfer and protect the reaction intermediates. Altogether, the structures allow us to draw a detailed reaction mechanism of this enzyme crucial for CO2 fixation in anaerobic organisms.
By analyzing (27.12±0.14)×108 ψ(3686) events accumulated with the BESIII detector, the decay ηc(2S)→K+K−η is observed for the first time with a significance of 6.2σ after considering systematic uncertainties. The product of the branching fractions of ψ(3686)→γηc(2S) and ηc(2S)→K+K−η is measured to be B(ψ(3686)→γηc(2S))×B(ηc(2S)→K+K−η)=(2.39±0.32±0.34)×10−6, where the first uncertainty is statistical, and the second one is systematic. The branching fraction of ηc(2S)→K+K−η is determined to be B(ηc(2S)→K+K−η)=(3.42±0.46±0.48±2.44)×10−3, where the third uncertainty is due to the branching fraction of ψ(3686)→γηc(2S). Using a recent BESIII measurement of B(ηc(2S)→K+K−π0), we also determine the ratio between the branching fractions of ηc(2S)→K+K−η and ηc(2S)→K+K−π0 to be 1.49±0.22±0.25, which is consistent with the previous result of BaBar at a comparable precision level.
Search for X(3872)→π⁰π⁰χc₁,₂

Using 10.1 fb−1 of e+e− collision data collected by the BESIII detector with center-of-mass energies between 4.15 GeV and 4.30 GeV, we search for the decays X(3872)→π0π0χc1,2, where the X(3872) is produced in e+e−→γX(3872). No evidence above 3σ is found for either decay. Upper limits at the 90% C.L. on the branching fractions of X(3872)→π0π0χc1,2 normalized to the branching fraction of X(3872)→π+π−J/ψ are set to be B(X(3872)→π0π0χc1)/B(X(3872)→π+π−J/ψ)<1.1 and B(X(3872)→π0π0χc2)/B(X(3872)→π+π−J/ψ)<0.5, taking into account both statistical and systematic uncertainties.

Here, we report the synthesis of a new heavy fermion compound, Ce3Bi4Ni3. It is an isoelectronic analog of the prototypical Kondo insulator Ce3Bi4Pt3 and of the recently discovered Weyl-Kondo semimetal Ce3Bi4Pd3. In contrast to the volume-preserving Pt-Pd substitution, structural and chemical analyses reveal a positive chemical pressure effect in Ce3Bi4Ni3 relative to its heavier counterparts. Based on the results of electrical resistivity, Hall effect, magnetic susceptibility, and specific heat measurements, we identify an energy gap of 65–70 meV, about eight times larger than that in Ce3Bi4Pt3 and about 45 times larger than that of the Kondo-insulating background hosting the Weyl nodes in Ce3Bi4Pd3. We show that this gap as well as other physical properties do not evolve monotonically with increasing atomic number, i.e., in the sequence Ce3Bi4Ni3−Ce3Bi4Pd3−Ce3Bi4Pt3, but instead with increasing partial electronic density of states of the 𝑑 orbitals at the Fermi energy. This work opens the possibility to investigate the conditions under which topological states develop in this series of strongly correlated 3-4-3 materials.

Heterostructures of graphene in proximity to magnetic insulators open the possibility to investigate exotic states emerging from the interplay of magnetism, strain and charge transfer between the layers. Recent reports on the growth of self-integrated atomic wires of β-RuCl3 on graphite suggest these materials as versatile candidates to investigate these effects. Here we present detailed first principles calculations on the charge transfer and electronic structure of β-RuCl3/heterostructures and provide a comparison with the work function analysis of the related honeycomb family members α-RuX3 (X = Cl,Br,I). We find that proximity of the two layers leads to a hole-doped graphene and electron-doped RuX3 in all cases, which is sensitively dependent on the distance between the two layers. Furthermore, strain effects due to lattice mismatch control the magnetization which itself has a strong effect on the charge transfer. Charge accumulation in β-RuCl3 strongly drops away from the chain making such heterostructures suitable candidates for sharp interfacial junctions in graphene-based devices.
Antimatter is a research topic of fundamental interest. Sufficient matter-antimatter asymmetry in the early Universe created the matter-dominated world today. The origin of this asymmetry is not completely understood to date. High-energy nuclear collisions create conditions similar to the Universe microseconds after the Big Bang, with comparable amounts of matter and antimatter. Much of the antimatter created escapes the rapidly expanding fireball without annihilation, making such collisions an effective experimental tool to create heavy antimatter nuclear objects and study their properties. In this paper, we report the first observation of the antimatter hypernucleus 4Λ¯H¯¯¯¯, composed of an Λ¯, an antiproton and two antineutrons. The discovery was made through its two-body decay after production in ultrarelativistic heavy-ion collisions by the STAR experiment at the Relativistic Heavy Ion Collider. In total, 15.6 candidate 4Λ¯H¯¯¯¯ antimatter hypernuclei are obtained with an estimated background count of 6.4. Lifetimes of the antihypernuclei 3Λ¯H¯¯¯¯ and 4Λ¯H¯¯¯¯ are measured and compared with the lifetimes of their corresponding hypernuclei, testing the symmetry between matter and antimatter. Various production yield ratios among (anti)hypernuclei and (anti)nuclei are also measured and compared with theoretical model predictions, shedding light on their production mechanism.
We report results on an elastic cross section measurement in proton-proton collisions at a center-of-mass energy s√=510 GeV, obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section is measured in the four-momentum transfer squared range 0.23≤−t≤0.67 GeV2. We find that a constant slope B does not fit the data in the aforementioned t range, and we obtain a much better fit using a second-order polynomial for B(t). The t dependence of B is determined using six subintervals of t in the STAR measured t range, and is in good agreement with the phenomenological models. The measured elastic differential cross section dσ/dt agrees well with the results obtained at s√=546 GeV for proton--antiproton collisions by the UA4 experiment. We also determine that the integrated elastic cross section within the STAR t-range is σfidel=462.1±0.9(stat.)±1.1(syst.)±11.6(scale) μb.

canning tunneling microscopy (STM) is perhaps the most promising way to detect the superconducting gap size and structure in the canonical unconventional superconductor Sr2RuO4 directly. However, in many cases, researchers have reported being unable to detect the gap at all in simple STM conductance measurements. Recently, an investigation of this issue on various local topographic structures on a Sr-terminated surface found that superconducting spectra appeared only in the region of small nanoscale canyons, corresponding to the removal of one RuO surface layer. Here, we analyze the electronic structure of various possible surface structures using first principles methods, and argue that bulk conditions favorable for superconductivity can be achieved when removal of the RuO layer suppresses the RuO4 octahedral rotation locally. We further propose alternative terminations to the most frequently reported Sr termination where superconductivity surfaces should be observed.
Motivated by recently reported magnetic-field induced topological phases in ultracold atoms and correlated Moiré materials, we investigate topological phase transitions in a minimal model consisting of interacting spinless fermions described by the Hofstadter model on a square lattice. For interacting lattice Hamiltonians in the presence of a commensurate magnetic flux it has been demonstrated that the quantized Hall conductivity is constrained by a Lieb-Schultz-Mattis (LSM)-type theorem due to magnetic translation symmetry. In this work, we revisit the validity of the theorem for such models and establish that a topological phase transition from a topological to a trivial insulating phase can be realized but must be accompanied by spontaneous magnetic translation symmetry breaking caused by charge ordering of the spinless fermions. To support our findings, the topological phase diagram for varying interaction strength is mapped out numerically with exact diagonalization for different flux quantum ratios and band fillings using symmetry indicators. We discuss our results in the context of the LSM-type theorem.
On the potential for GWAS with phenotypic population means and allele-frequency data (popGWAS)

This study explores the potential of a novel genome-wide association study (GWAS) approach for identifying loci underlying quantitative polygenic traits in natural populations. Extensive population genetic forward simulations demonstrate that the approach is generally effective for oligogenic and moderately polygenic traits and relatively insensitive to low heritability, but applicability is limited for highly polygenic architectures and pronounced population structure. The required sample size is moderate with very good results being obtained already for a few dozen populations scored. The method performs well in predicting population means even with a moderate false positive rate. When combined with machine learning for feature selection, this rate can be further reduced. The data efficiency of the method, particularly when using pooled sequencing, makes GWAS studies more accessible for research in biodiversity genomics. Overall, this study highlights the promise of this popGWAS approach for dissecting the genetic basis of complex traits in natural populations.
Abstract
Seed harvesting from wild plant populations is key for ecological restoration, but may threaten the persistence of source populations. Consequently, several countries have set guidelines limiting the proportions of harvestable seeds. However, these guidelines are so far inconsistent, and they lack a solid empirical basis. Here, we use high-resolution data from 298 plant species to model the demographic consequences of seed harvesting. We find that the current guidelines do not protect populations of annuals and short-lived perennials, while they are overly restrictive for long-lived plants. We show that the maximum possible fraction of seed production – what can be harvested without compromising the long-term persistence of populations – is strongly related to the generation time of the target species. When harvesting every year, this safe seed fraction ranges from 80% in long-lived species to 2% in most annuals. Less frequent seed harvesting substantially increases the safe seed fraction: In the most vulnerable annual species, it is safe to harvest 5%, 10% or 30% of population seed production when harvesting every two, five or ten years, respectively. Our results provide a quantitative basis for seed harvesting legislations worldwide, based on species’ generation time and harvesting regime.
Significance The UN Decade on Ecosystem Restoration, 2021-2030, foresees upscaling restoration, and the demand for native seed is skyrocketing. Seeds for restoring native vegetation are often harvested in wild, but too intensive harvest can threaten the donor populations. Existing guidelines that set limits to wild seed harvest are mostly based on expert opinions, yet they commonly lack empirical basis and vary among regions in one order of magnitude. We show that the current guidelines urgently need to be reformulated, because they are overly restrictive in long-lived species, while they do not protect annual plants from extinction. Using matrix population models of nearly 300 plant species, we provide a quantitative basis for a new seed harvesting legislation world-wide.
Evidence for the singly Cabibbo suppressed decay Λ+c→pπ0 is reported for the first time with a statistical significance of 3.7σ based on 6.0 fb−1 of e+e− collision data collected at center-of-mass energies between 4.600 and 4.843 GeV with the BESIII detector at the BEPCII collider. The absolute branching fraction of Λ+c→pπ0 is measured to be (1.56+0.72−0.58±0.20)×10−4. Combining with the branching fraction of Λ+c→nπ+, (6.6±1.3)×10−4, the ratio of the branching fractions of Λ+c→nπ+ and Λ+c→pπ0 is calculated to be 3.2+2.2−1.2. As an important input for the theoretical models describing the decay mechanisms of charmed baryons, our result indicates that the non-factorizable contributions play an essential role and their interference with the factorizable contributions should not be significant. In addition, the absolute branching fraction of Λ+c→pη is measured to be (1.63±0.31stat±0.11syst)×10−3.
We measure triangular flow relative to the reaction plane at 3 GeV center-of-mass energy in Au+Au collisions at the BNL Relativistic Heavy Ion Collider. A significant v3 signal for protons is observed, which increases for higher rapidity, higher transverse momentum, and more peripheral collisions. The triangular flow is essentially rapidity-odd with a slope at mid-rapidity, dv3/dy|(y=0), opposite in sign compared to the slope for directed flow. No significant v3 signal is observed for charged pions and kaons. Comparisons with models suggest that a mean field potential is required to describe these results, and that the triangular shape of the participant nucleons is the result of stopping and nuclear geometry.
The Born cross sections for the process e+e−→ωη′ are measured at 22 center-of-mass energies from 2.000 to 3.080 GeV using data collected with the BESIII detector at the BEPCII collider. A resonant structure is observed with a statistical significance of 9.6σ. A Breit-Wigner fit determines its mass to be MR=(2153±30±31) MeV/c2 and its width to be ΓR=(167±77±7) MeV, where the first uncertainties are statistical and the second are systematic.
Using a sample of (10087±44)×106 J/ψ events, which is about 45 times larger than that was previously analyzed, a further investigation on the J/ψ→γ3(π+π−) decay is performed. A significant distortion at 1.84 GeV/c2 in the line-shape of the 3(π+π−) invariant mass spectrum is observed for the first time, which could be resolved by two overlapping resonant structures, X(1840) and X(1880). The new state X(1880) is observed with a statistical significance larger than 10σ. The mass and width of X(1880) are determined to be 1882.1±1.7±0.7 MeV/c2 and 30.7±5.5±2.4 MeV, respectively, which indicates the existence of a pp¯ bound state.
Using 9.0 fb−1 of e+e− collision data collected at center-of-mass energies from 4.178 to 4.278 GeV with the BESIII detector at the BEPCII collider, we perform the first search for the radiative transition χc1(3872)→γψ2(3823). No χc1(3872)→γψ2(3823) signal is observed. The upper limit on the ratio of branching fractions B(χc1(3872)→γψ2(3823),ψ2(3823)→γχc1)/B(χc1(3872)→π+π−J/ψ) is set as 0.075 at the 90\% confidence level. Our result contradicts theoretical predictions under the assumption that the χc1(3872) is the pure charmonium state χc1(2P).
Using data samples with an integrated luminosity of 22.42 fb−1 collected by the BESIII detector operating at the BEPCII storage ring, we measure the cross sections of the 𝑒+𝑒−→𝜂𝐽/𝜓 process at center-of-mass energies from 3.808 to 4.951 GeV. Three structures are observed in the line shape of the measured cross sections. A maximum-likelihood fit with 𝜓(4040), two additional resonances, and a nonresonant component are performed. The mass and width of the first additional state are (4219.7±2.5±4.5) MeV/𝑐2 and (80.7±4.4±1.4) MeV, respectively, consistent with the 𝜓(4230). For the second state, the mass and width are (4386±13±17) MeV/𝑐2 and (177±32±13) MeV, respectively, consistent with the 𝜓(4360). The first uncertainties are statistical, and the second ones are systematic. The statistical significance of 𝜓(4040) is 8.0𝜎 and those for 𝜓(4230) and 𝜓(4360) are more than 10.0𝜎.
We present the first observation of the singly Cabibbo-suppressed decay Λ+c→ΛK+π0 with a significance of 5.7σ and the first evidence of Λ+c→ΛK+π+π− decay with a significance of 3.1σ, based on e+e− annihilation data recorded by the BESIII detector at the BEPCII collider. The data correspond to an integrated luminosity of 6.4 fb−1, in the center-of-mass energy range from 4.600 GeV to 4.950 GeV. We determine the branching fractions of Λ+c→ΛK+π0 and Λ+c→ΛK+π+π− relative to their Cabibbo-favored counterparts to be B(Λ+c→ΛK+π0)B(Λ+c→Λπ+π0)=(2.09±0.39stat.±0.07syst.)×10−2 and B(Λ+c→ΛK+π+π−)B(Λ+c→Λπ+π+π−)=(1.13±0.41stat.±0.06syst.)×10−2, respectively. Moreover, by combining our measured result with the world average of B(Λ+c→Λπ+π0), we obtain the branching fraction B(Λ+c→ΛK+π0)=(1.49±0.27stat.±0.05syst.±0.08ref.)×10−3. This result significantly departs from theoretical predictions based on quark SU(3) flavor symmetry, which is underpinned by the presumption of meson pair S-wave amplitude dominance.
The epitranscriptome embodies many new and largely unexplored functions of RNA. A major roadblock in the epitranscriptomics field is the lack of transcriptome-wide methods to detect more than a single RNA modification type at a time, identify RNA modifications in individual molecules, and estimate modification stoichiometry accurately. We address these issues with CHEUI (CH3 (methylation) Estimation Using Ionic current), a new method that concurrently detects N6-methyladenosine (m6A) and 5-methylcytidine (m5C) in individual RNA molecules from the same sample, as well as differential methylation between any two conditions. CHEUI processes observed and expected nanopore direct RNA sequencing signals with convolutional neural networks to achieve high single-molecule accuracy and outperforms other methods in detecting m6A and m5C sites and quantifying their stoichiometry. CHEUI’s unique capability to identify two modification types in the same sample reveals a non-random co-occurrence of m6A and m5C in mRNA transcripts in cell lines and tissues. CHEUI unlocks an unprecedented potential to study RNA modification configurations and discover new epitranscriptome functions.
Using e+e− annihilation data corresponding to an integrated luminosity of 2.93 fb−1 taken at the center-of-mass energy s√=3.773~GeV with the BESIII detector, a joint amplitude analysis is performed on the decays D0→π+π−π+π− and D0→π+π−π0π0(non-η). The fit fractions of individual components are obtained, and large interferences among the dominant components of D0→a1(1260)π, D0→π(1300)π, D0→ρ(770)ρ(770) and D0→2(ππ)S are found in both channels. With the obtained amplitude model, the CP-even fractions of D0→π+π−π+π− and D0→π+π−π0π0(non-η) are determined to be (75.2±1.1stat.±1.5syst.)% and (68.9±1.5stat.±2.4syst.)%, respectively. The branching fractions of D0→π+π−π+π− and D0→π+π−π0π0(non-η) are measured to be (0.688±0.010stat.±0.010syst.)% and (0.951±0.025stat.±0.021syst.)%, respectively. The amplitude analysis provides an important model for binning strategy in the measurements of the strong phase parameters of D0→4π when used to determine the CKM angle γ(ϕ3) via the B−→DK− decay.

Combining with the branching fraction of Λ+c→nπ+, (6.6±1.3)×10−4, the ratio of the branching fractions of Λ+c→nπ+ and Λ+c→pπ0 is calculated to be 3.2+2.2−1.2. As an important input for the theoretical models describing the decay mechanisms of charmed baryons, our result indicates that the non-factorizable contributions play an essential role and their interference with the factorizable contributions should not be significant. In addition, the absolute branching fraction of Λ+c→pη is measured to be (1.63±0.31stat±0.11syst)×10−3, which is consistent with previous measurements.

Using a sample of (10087±44)×106 𝐽/𝜓 events, which is about 45 times larger than that was previously analyzed, a further investigation on the 𝐽/𝜓→𝛾3(𝜋+𝜋−) decay is performed. Observation of η_(c)(1S, 2S) and χ_(cJ) decays to 2(π⁺π^(−))η via ψ(3686) radiative transitions

Based on 2.7×109 ψ(3686) decays collected with the BESIII detector, the radiative decay ψ(3686)→γ2(π+π−)η is investigated to measure properties of S- and P-wave charmonium states. The branching fraction of the decay ηc(1S)→2(π+π−)η, which is found to have a strong dependence on the interference pattern between ηc(1S) and non-ηc(1S) processes, is measured in both destructive and constructive interference scenarios for the first time. The mass and width of the ηc(1S) are measured to be M=(2984.14±0.13±0.38) MeV/c2 and Γ=(28.82±0.11±0.82) MeV, respectively. Clear signals for the decays of the χcJ(J=0,1,2) and the ηc(2S) to 2(π+π−)η are also observed for the first time, and the corresponding branching fractions are measured. The ratio of the branching fractions between the ηc(2S) and ηc(1S) decays is significantly lower than the theoretical prediction, which might suggest different dynamics in their decays.
A light scalar X0 or vector X1 particles have been introduced as a possible explanation for the (g−2)μ anomaly and dark matter phenomena.
Using (8.998±0.039)×109 $\jpsi$ events collected by the BESIII detector, we search for a light muon philic scalar X0 or vector X1 in the processes J/ψ→μ+μ−X0,1 with X0,1 invisible decays. No obvious signal is found, and the upper limits on the coupling g′0,1 between the muon and the X0,1 particles are set to be between 1.1×10−3 and 1.0×10−2 for the X0,1 mass in the range of 1<M(X0,1)<1000 MeV/c2 at 90% confidence level.
Using 2.93 fb−1 of e+e− collision data collected with the BESIII detector at the center-of-mass energy of 3.773 GeV, we investigate the semileptonic decays D+→π+π−ℓ+νℓ (ℓ=e and μ). The D+→f0(500)μ+νμ decay is observed for the first time. By analyzing simultaneously the differential decay rates of D+→f0(500)μ+νμ and D+→f0(500)e+νe in different ℓ+νℓ four-momentum transfer intervals, the product of the relevant hadronic form factor ff0+(0) and the magnitude of the c→d Cabibbo-Kobayashi-Maskawa matrix element |Vcd| is determined to be ff0+(0)|Vcd|=0.0787±0.0060stat±0.0033syst for the first time. With the input of |Vcd| from the global fit in the standard model, we determine ff0+(0)=0.350±0.027stat±0.015syst. The absolute branching fractions of D+→f0(500)(π+π−)μ+νμ and D+→ρ0(π+π−)μ+νμ are determined as (0.72±0.13stat±0.10syst)×10−3 and (1.64±0.13stat±0.11syst)×10−3. Combining these results with those of previous BESIII measurements on their semielectronic counterparts from the same data sample, we test lepton flavor universality by measuring the branching fraction ratios BD+→ρ0μ+νμ/BD+→ρ0e+νe = 0.88±0.10 and BD+→f0(500)μ+νμ/BD+→f0(500)e+νe = 1.14±0.28, which are compatible with the standard model expectation.
Using 7.33~fb−1 of e+e− collision data collected by the BESIII detector at center-of-mass energies in the range of s√=4.128−4.226~GeV, we search for the rare decays D+s→h+(h0)e+e−, where h represents a kaon or pion. By requiring the e+e− invariant mass to be consistent with a ϕ(1020), 0.98<M(e+e−)<1.04 ~GeV/c2, the decay D+s→π+ϕ,ϕ→e+e− is observed with a statistical significance of 7.8σ, and evidence for the decay D+s→ρ+ϕ,ϕ→e+e− is found for the first time with a statistical significance of 4.4σ. The decay branching fractions are measured to be B(D+s→π+ϕ,ϕ→e+e−)=(1.17+0.23−0.21±0.03)×10−5, and B(D+s→ρ+ϕ,ϕ→e+e−)=(2.44+0.67−0.62±0.16)×10−5, where the first uncertainties are statistical and the second systematic. No significant signal for the three four-body decays of D+s→π+π0e+e−, D+s→K+π0e+e−, and D+s→K0Sπ+e+e− is observed. For D+s→π+π0e+e−, the ϕ mass region is vetoed to minimize the long-distance effects. The 90% confidence level upper limits set on the branching fractions of these decays are in the range of (7.0−8.1)×10−5.
Based on (2.712±0.014)×109 ψ(3686) events collected by the BESIII collaboration, evidence of the hadronic decay hc→K0SK+π−+c.c. is found with a significance of 4.3σ in the ψ(3686)→π0hc process. The branching fraction of hc→K0SK+π−+c.c. is measured to be (7.3±0.8±1.8)×10−4, where the first and second uncertainties are statistical and systematic, respectively. Combining with the exclusive decay width of ηc→KK¯π, our result indicates inconsistencies with both pQCD and NRQCD predictions.

While high-quality climate reconstructions of some past warm periods in the Cenozoic era now exist, the geological processes responsible for driving the observed longterm changes in atmospheric CO2 are not sufficiently well understood. The long-term change in atmospheric CO2 across the Cenozoic has been proposed to be driven by processes such as terrestrial weathering, organic carbon production and burial, reverse weathering, and volcanic degassing. One way of constraining the relative importance of the various driving forces proposed so far is to better understand the degree to which ocean chemistry has changed because the chemistry of seawater responds to geologic processes that drive atmospheric CO2. In addition, knowledge of the concentration of the major elements in seawater is crucial for accurately applying proxies such as those based on the boron isotopic composition and Mg/Ca of marine carbonates (a proxy for palaeo pH/CO2 and palaeotemperature, respectively). Previously reported records of seawater composition are primarily derived from fluid inclusions in marine evaporites; however, the results are sparse due to the limited availability of such deposits. In this thesis, changes in the Eocene seawater chemistry were reconstructed using trace element (elements/Ca) and isotopic (δ26Mg) proxies in a Larger Benthic Foraminifera (LBFs), i.e., Nummulites sp., to constrain the driving processes of long-term changes in seawater chemistry.
To achieve the objective of this thesis, first, a measurement protocol was established using LA-ICPMS to measure the K/Ca ratio simultaneously with other element/calcium ratios, which is challenging due to the interference of ArH+ on K+. Utilising this newly established measurement protocol, laboratory-cultured Operculina ammonoides grown at different seawater calcium concentrations ([Ca2+]), repeated at different temperatures, as well as modern O. ammonoides collected from different regions exhibiting a range of seawater parameters, were investigated. A significant correlation was observed between K/Casw and K/CaLBF, allowing K/CaLBF to potentially be used as a proxy for seawater major ion reconstructions. In addition, modern O. ammonoides demonstrated no significant influence of most seawater parameters (temperature, salinity, pH, or [CO32-]) on K/CaLBF. Modern O.
ammonoides were also assessed for their Mg isotopic composition (δ26Mg), revealing no significant effect of temperature or salinity on δ26MgLBF. Furthermore, the Mg isotopic fractionation in O. ammonoides was found to be close to that of inorganic calcite, indicating minimal vital effects in these large benthic foraminifera.
Operculina ammonoides is the nearest living relative of the abundant Eocene genus Nummulites, enabling the reconstruction of seawater chemistry using the calibration based on O. ammonoides. The trace elemental/calcium proxies, including Na/Ca, K/Ca, and Mg/Ca, as well as the δ26Mg proxy, were investigated in Eocene Nummulites. The result showed that during the Eocene, [Ca2+]sw was 1.6-2 times higher, while [K+]sw was ~2 times lower than the modern seawater composition. Furthermore, [Mg2+]sw decreased from the early Eocene (54.3− +9 7..69 mmol kg-1 at ~55 Ma) to Late Eocene (37.8− +4 4..3 4 mmol kg-1 at ~31 Ma), followed by
an increase toward modern seawater [Mg]. In contrast, the variability in δ26Mgsw values remained within a narrow range of ~0.3 ‰ throughout the Cenozoic. The reconstructed [Ca2+]sw agrees with the suggestion that Cenozoic seawater chemistry changes can be explained via a change in the seafloor spreading rate. When combined with existing records, the observed minimal change in δ26Mgsw with an increase in [Mg2+]sw suggests an additional possible role of a decrease in the formation of authigenic clay minerals coincident with the Cenozoic decline in deep ocean temperature, which is also supported by the increase in the [K+]sw reconstructed here for the first time. This finding highlights that the reduction in seafloor-spreading rate and decline in reverse weathering during the Cenozoic era has played a significant role in the evolution of seawater chemistry, emphasizing the importance of these processes in driving long-term changes in the carbon cycle.
Tree-related microhabitats (TreMs) describe the microhabitats that a tree can provide for a multitude of other taxonomic groups and have been proposed as an important indicator for forest biodiversity (Asbeck et al., 2021). So far, the focus of TreM studies has been on temperate forests, although many trees in the tropics harbour exceptionally high numbers of TreMs. In this study, TreMs in the lowland tropical forests of the Choco (Ecuador) and in the mountain tropical forests of Mount Kilimanjaro (Tanzania) were surveyed. Our results extend the existing typology of TreMs of Larrieu et al. (2018) to include tropical forests and enabled a comparison of the relative recordings and diversity of TreMs between tropical and temperate forests. A new TreM form, Root formations, and three new TreM groups, concavities build by fruits or leaves, dendrotelms, and root formations, were established. In total, 15 new TreM types in five different TreM groups were specified. The relative recordings of most TreMs were similar between tropical and temperate forests. However, ivy and lianas, and ferns were more common in the lowland rainforest than in temperate forests, and bark microsoil, limb breakage, and foliose and fruticose lichens in tropical montane forest than in lowland rainforest. Mountain tropical forests hosted the highest diversity for common and dominant TreM types, and lowland tropical forest the highest diversity for rare TreMs. Our extended typology of tree-related microhabitats can support studies of forest-dwelling biodiversity in tropical forests. Specifically, given the ongoing threat to tropical forests, TreMs can serve as an additional tool allowing rapid assessments of biodiversity in these hyperdiverse ecosystems.
Die Epoche der Weimarer Republik war gekennzeichnet durch eine Vielzahl von Umbrüchen im politischen, ökonomischen und gesellschaftlichen Bereich. Diese durch den Ersten Weltkrieg verursachten radikalen Veränderungen der Lebensverhältnisse hatten auch einen weitreichenden Einfluss auf den Alltag und die öffentliche Wahrnehmung der Frauen dieser Zeit: So konnten diese ab 1918 an den Wahlen teilnehmen, konnten sich an allen deutschen Universitäten einschreiben und in bestimmtem Maße finanziell unabhängig werden. Das Bild der so genannten "Neuen Frau", das gleichzeitig in der Öffentlichkeit generiert wurde, war insbesondere gekennzeichnet durch innerliche und äußerliche Emanzipation und stellte damit einen radikalen Gegenpol zur traditionellen Weiblichkeit des 19. Jahrhunderts dar. Viele Facetten der "Neuen Frau" sind in der Forschung weitgehend erschlossen - die sportliche Frau, die automobile Frau, die Frau als flapper oder garçonne - nur einer Ausprägung des neuen Frauentypus wurde bisher wenig Beachtung geschenkt: der Frau als Verbrecherin. Ziel der vorliegenden interdisziplinär und medienkomparatistisch angelegten Dissertation ist es daher, übergreifende Semantiken und Ästhetiken der geschlechtsspezifischen Verbrechensdarstellung in der Weimarer Republik offenzulegen und die unterschiedlich erzeugten, vermittelten und klassifizierenden Vorstellungen von Verbrecherinnen zu analysieren und zu kontextualisieren. Das zugrundeliegende Material umfasst dabei fachwissenschaftliche, publizistische und literarische Darstellungen von Verbrecherinnen, wobei das Hauptaugenmerk auf der Analyse von Artikeln aus Illustrierten Magazinen liegt, da diese, neben Radio und Film, zu den wirkungsmächtigsten Medien der Weimarer Republik gehören.
The PhD addresses the feasibility of reconstructing open charm mesons with the Compressed Baryonic Matter experiment, which will be installed at the FAIR accelerator complex at Darmstadt/Germany. The measurements will be carried out by means of a dedicated Micro Vertex Detector (MVD), which will be equipped with CMOS Monolithic Active Pixel Sensors (MAPS). The feasibility of reconstructing the particles with a proposed detector setup was studied.
To obtain conclusive results, the properties of a MAPS prototype were measured in a beam test at the CERN-SPS accelerator. Based on the results achieved, a dedicated simulation software for the sensors was developed and implemented into the software framework of CBM (CBMRoot). Simulations on the reconstruction of D0-mesons were carried out. It is concluded that the reconstruction of those particles is possible.
The PhD introduces the physics motivation of doing open charm measurements, represents the results of the measurements of MAPS and introduces the innovative simulation model for those sensors as much as the concept and results of simulations of the D0 reconstruction.
Why do banks issue contingent convertible debt? To answer this question we study comprehensive data covering all issues by publicly traded banks in Europe of contingent convertible bonds (CoCos) that count as additional tier 1 capital (AT1). We find that banks with lower asset volatility are more likely to issue AT1 CoCos than their riskier counterparts, but that CDS spreads do not react following issue announcements. Our estimates therefore suggest that agency costs play a crucial role in banks' ability to successfully issue CoCos. The agency costs may be higher for CoCos than for equity explaining why we observe riskier or lowly capitalized banks to issue equity rather than CoCos.
Over the last few decades, considerable attention has focused on small-scale studies of invasive plants and invaded systems. Unfortunately, small scale studies rarely provide comprehensive insight into the complexities of biological invasions at macroscales. Systematic and repeated monitoring of biological invasions at broad scales are rare. In this report, we highlight a unique invasive plant database from the national Forest Inventory and Analysis (FIA) program of the United States Forest Service. We demonstrate the importance and capability of this subcontinental-wide database by showcasing several critical macroscale invasion patterns that have emerged from its initial analysis: (1) large portion of the forests systems (39%) in the United States are impacted by invasive plants, (2) forests in the eastern United States harbor more invasive species than the western regions, (3) human land-use legacies at regional to national scalesb may drive large-scale invasion patterns. This accumulated dataset, which continues to grow in temporal richness with repeated measurements, will allow the understanding of invasion patterns and processes at multi-spatial and temporal scales. Such insights are not possible from smaller-scale studies, illustrating the benefit that can be gained by investing in the development of regional to continental-wide invasion monitoring programs elsewhere.
Ziel: Nebennierenraumforderungen können sonographisch mit guter Sensitivität und Spezifität erkannt werden. Ziel der vorliegenden Studie war es, die Wertig-keit der Kontrastmittel-Sonographie (CEUS = contrast enhanced ultrasound) unter Verwendung von Zeit-Intensitäts-Kurven für die Charakterisierung von Nebennierenraumforderungen zu bewerten.
Material und Methoden: 108 Patienten mit 116 Raumforderungen der Neben-niere erhielten einen Ultraschall der Nebenniere, inklusive einer Kontrastmittel-Sonographie mit dem Kontrastmittel SonoVue®. Die Kontrastmitteldynamik wurde mittels Zeit-Intensitätskurven aufgezeichnet. Anhand der Kontrastmittel-anflutungszeit wurden folgende vier Kontrastmittelanreicherungsmuster ver-wendet, welche in einer Pilotstudie1 entwickelt wurden: Anreicherungsmuster I = früh arterielle Kontrastmittelanreicherung, Anreicherungsmuster II = arterielle Kontrastmittelanreicherung, Anreicherungsmuster III = späte Kontrastmittelan-reicherung, Anreicherungsmuster IV = keine Kontrastmittelanreicherung. Zu-sätzlich erhielten alle Patienten eine Beurteilung der Nebenniere durch ein zweite Bildgebendes Verfahren (Computertomographie (CT) oder Magnetresonanztomographie (MRT)) sowie eine laborchemische Hormondiagnostik. In Fällen mit malignitätsverdächtigem Befund erfolgte eine Biopsie oder Adrenalektomie.
Ergebnisse: CEUS- Anreicherungsmuster I und II konnten bei allen Patienten mit primärem oder sekundärem Malignom der Nebenniere (n = 16) nachgewie-sen werden. Die Sensitivität der CEUS für die Diagnose einer malignen Neben-nierenraumforderung betrug 100% (95% CI [75; 100]) und die Spezifität 67% (95% CI [56; 75]). Bei 40 Nebennierenraumforderungen war die Histologie die Referenzmethode. In 68% dieser Nebennierenraumforderungen waren MRT/CT und CEUS kongruent in der Beurteilung der Dignität. Schlussfolgerung: Die Kontrastmittel-Sonographie ist eine nützliche ergän-zende Methode in der Aufarbeitung von Nebennierenraumforderungen mit exzellenter Sensitivität für die Diagnose eines Malignoms.
Most elements heavier than iron are synthesized in stars during neutron capture reactions in the r- and s-process. The s-process nucleosynthesis is composed of the main and weak component. While the s-process is considered to be well understood, further investigations using nucleosynthesis simulations rely on measured neutron capture cross sections as crucial input parameters. Neutron capture cross sections
relevant for the s-process can be measured using various experimental methods. A prominent example is the activation method relying on the 7Li(p,n)7Be reaction as a neutron source, which has the advantage of high neutron intensities and is able to create a quasi-stellar neutron spectrum at kBT = 25 keV. Other neutron sources able to provide quasi-stellar spectra at different energies suffer from lower neutron intensities. Simulations using the PINO tool suggest the neutron activation of samples with different neutron spectra, provided by the 7Li(p,n)7Be reaction, and a subsequent linear combination of the obtained spectrum-averaged cross sections
to determine the Maxwellian-averaged cross section (MACS) at various energies of astrophysical relevance. To investigate the accuracy of the PINO tool at proton energies between the neutron emission threshold at Ep = 1880.4 keV and 2800 keV,
measurements of the 7Li(p,n)7Be neutron fields are presented, which were carried out at the PTB Ion Accelerator Facility at the Physikalisch-Technische Bundesanstalt in Braunschweig. The neutron fields of ten different proton energies were measured.
The presented neutron fields show a good agreement at proton energies Ep = 1887, 1897, 1907, 1912 and 2100 keV. For the other proton energies, E p = 2000, 2200, 2300, 2500, and 2800 keV, differences between measurement and simulation were found and discussed. The obtained results can be used to benchmark and adapt the PINO tool and provide crucial information for further improvement of the neutron activation method for astrophysics.
An application for the 7Li(p,n)7Be neutron fields is presented as an activation experiment campaign of gallium, an element that is mostly produced during the weak s-process in massive stars. The available cross section data for the 69,71Ga(n,γ)
reactions, mostly determined by activation measurements, show differences up toa factor of three. To improve the data situation, activation measurements were carried out using the 7Li(p,n)7Be reaction. The neutron capture cross sections for
a quasi-stellar neutron spectrum at kBT = 25 keV were determined for 69Ga and 71Ga.
In this dissertation the formal abstraction and verification of analog circuit is examined. An approach is introduced that automatically abstracts a transistor level circuit with full Spice accuracy into a hybrid automaton (HA) in various output languages. The generated behavioral model exhibits a significant simulation speed-up compared to the original netlist, while maintaining an acceptable accuracy, and can be therefore used in various verification and validation routines. On top of that, the generated models can be formally verified against their Spice netlists, making the obtained models correct by construction.
The generated abstract models can be extended to enclose modeling as well as technology dependent parameter variations with little over approximations. As these models enclose the various behaviors of the sampled netlists, the obtained models are of significant importance as they can replace several simulations with just a single reachability analysis or symbolic simulation. Moreover, these models can be as well be used in different verification routines as demonstrated in this dissertation.
As the obtained models are described by HAs with linear behaviors in the locations, the abstract models can be as well compositionally linked, allowing thereby the abstraction of complex analog circuits.
Depending on the specified modeling settings, including for example the number of locations of the HA and the description of the system behavior, the accuracy, speedup, and various additional properties of the HA can be influenced. This is examined in detail in this dissertation. The underlying abstraction process is first covered in detail. Several extensions are then handled including the modeling of the HAs with parameter variations. The obtained models are then verified using various verification methodologies. The accuracy and speed-up of the abstraction methodology is finally evaluated on several transistor level circuits ranging from simple operational amplifiers up to a complex circuits.
