Gefängnisfilm

Der Gefängnisfilm bildete sich als Subgenre des klassischen Gangsterfilms parallel zu dessen Entstehung zu Beginn der 1930er Jahre heraus. Die Haftanstalt nimmt in diesem Genre eine ebenso mythische wie notwendige Position ein, da der Gefängnisaufenthalt entweder das Ende einer Gangsterkarriere bedeutet oder eine Fortsetzung dieser Funktion unter veränderten Voraussetzungen. „Gefängnis und Gangster-Existenz bedingen einander so sehr, daß das eine ohne das andere kaum vorstellbar erscheint“, wie es in Kellners Gangsterfilm (1977) heißt. Der Gefängnisfilm ist letztlich ein Gangsterfilm, der seinen Haupthandlungsschauplatz in die Haftanstalt verlegt hat und dort entweder von der Läuterung des Gangsters, von dem Schicksal eines zu Unrecht Verurteilten oder einem Gefängnisaufstand erzählt.
Over the past few decades, changes in market conditions such as globalisation and deregulation of financial markets as well as product innovation and technical advancements have induced financial institutions to expand their business activities beyond their traditional boundaries and to engage in cross-sectoral operations. As combining different sectoral businesses offers opportunities for operational synergies and diversification benefits, financial groups comprising banks, insurance undertakings and/or investment firms, usually referred to as financial conglomerates, have rapidly emerged, providing a wide range of services and products in distinct financial sectors and oftentimes in different geographic locations. In the European Union (EU), financial conglomerates have become part of the biggest and most active financial market participants in recent years. Financial conglomerates generally pose new problems for financial authorities as they can raise new risks and exacerbate existing ones. In particular, their cross-sectoral business activities can involve prudentially substantial risks such as the risk of regulatory arbitrage and contagion risk arising from intra-group transactions. Moreover, the generally large size of financial conglomerates as well as the high complexity and interconnectedness of their corporate structures and risk exposures can entail substantial systemic risk and can therefore threaten the stability of the financial system as a whole. Until a few years ago, there was no supervisory framework in place which addressed a financial conglomerate in its entirety as a group. Instead, each group entity within a financial conglomerate was subject to the supervisory rules of its pertinent sector only. Such silo supervisory approach had the drawback of not taking account of risks which arise or aggravate at the group level. It also failed to consider how the risks from different business lines within the group interrelate with each other and affect the group as a whole. In order to address this lack of group-wide prudential supervision of financial conglomerates, the European legislator adopted the Financial Conglomerates Directive 2002/87/EC8 (‘FCD’) on 16 December 2002. The FCD was transposed into national law in the member states of the EU (‘Member States’) by 11 August 2004 for application to financial years beginning on 1 January 2005 and after. The FCD primarily aims at supplementing the existing sectoral directives to address the additional risks of concentration, contagion and complexity presented by financial conglomerates. It therefore provides for a supervisory framework which is applicable in addition to the sectoral supervision. Most importantly, the FCD has introduced additional capital requirements at the conglomerate level so as to prevent the multiple use of the same capital by different group entities. This paper seeks to examine to what extent the FCD provides for an adequate capital regulation of financial conglomerates in the EU while taking into account the underlying sectoral capital requirements and the inherent risks associated with financial conglomerates. In Part 1, the definition and the basic corporate models of financial conglomerates will be presented (I), followed by an illustration of the core motives behind the phenomenon of financial conglomeration (II) and an overview of the development of the supervision over financial conglomerates in the EU (III). Part 2 begins with a brief elaboration on the role of regulatory capital (I) and gives a general overview of the EU capital requirements applicable to banks and insurance undertakings respectively. A delineation of the commonalities and differences of the banking and the insurance capital requirements will be provided (II). It continues to further examine the need for a group-wide capital regulation of financial conglomerates and analyses the adequacy of the FCD capital requirements. In this context, the technical advice rendered by the Joint Committee on Financial Conglomerates (JCFC) as well as the currently ongoing legislative reforms at the EU level will be discussed (III). The paper finally closes with a conclusion and an outlook on remaining open issues (IV).
This study examines the legal environment of netting agreements covering financial contracts. It concludes that an international instrument should be developed capable of improving the effectiveness of netting agreements in mitigating systemic risk. To this end, two different aspects of the enforceability of netting agreements are considered: (i) the general enforceability of netting, and (ii) the possibility of precluding the operation of netting a mechanism by way of a regulatory moratorium for considerations of systemic stability. The first part of the study presents the use of netting and the various forms it may take before going on to explain the benefits and drawbacks of enforceable netting agreements. Benefits for individual firms consist in lower counterparty risk and more favourable capital requirements. Benefits for the financial market as a whole flow from greater financial market stability since the contagion of systemically relevant institutions by the default or insolvency of another institution is limited, thus helping to avoid systemic effects. Additionally, the use of netting arrangements can improve overall market liquidity. A potential drawback of enforceability of netting, in certain situations, is that the operation of a netting mechanism could actually work against the purpose of systemic stability where the transfer of parts of the business of an insolvent financial institution to a solvent bridge entity would enhance or maintain value to a greater extent than the operation of a netting agreement would. Regulatory authorities are considering under which conditions a moratorium to halt the netting mechanism until the situation is solved could avoid this threat to systemic stability. The second part of the study examines whether there is the potential to support the purpose of enhanced systemic stability by way of international harmonisation of private and insolvency law. As regards the issue of general enforceability, the global picture of netting legislation is heterogeneous. Given the great practical relevance of the matter, an international instrument could be very useful. As to the issue of private law consequences of regulatory moratoria, the absence of a harmonised framework appears to lead to actual cross-border inconsistency and legal uncertainty as regards financial contracts that are governed by a foreign law. Taking these to aspects into account, this paper recommends that work on developing an international instrument be undertaken. The final part of the study suggests a set of preliminary guidelines for the development of suchan instrument. In the light of the findings of the previous sections, a mixed, two-step approach is recommended. First, a non-binding instrument could be developed, serving as a benchmark and reservoir of legal solutions in respect of the relevant issues. Secondly, isolated aspects relating to both the general enforceability of netting and the accommodation of a regulatory moratorium in foreign private and insolvency law could be dealt with in an international Convention, in particular where cross-border situations involving netting require uniformity of applicable legal rules.
Das OLG Düsseldorf hat über Managerfehler im Vorfeld der Finanzmarktkrise bei der IKB Deutsche Industriebank AG zu entscheiden. Der Beschluss wird Anfangs- und Referenzpunkt der organhaftungsrechtlichen Krisenaufarbeitung sein, und führende Berater attestieren ihm ein Wirkungspotential, wie es zuletzt dem ARAG-Urteil des BGH zukam. Der Beitrag stimmt dem Beschlussergebnis zu, zeigt aber auf, wie Kernaussagen schnell in eine prinzipienlose Ausdehnung des Organhaftungsrechts münden können.
Die Möglichkeiten zum Recherchieren von botanischer Literatur über Hessen in sieben Online-Datenbanken werden vergleichend betrachtet. Unter anderem werden Testrecherchen zu Beiträgen in den Zeitschriften Hessische Floristische Briefe und Botanik und Naturschutz in Hessen ausgewertet. Es stellt sich heraus, dass im Feld der getesteten Datenbanken die Hessische Bibliographie sowie Biological Abstracts jeweils eine besondere Stellung einnehmen; mehrere weitere, überwiegend lizenzfrei nutzbare Datenbanken liefern ebenfalls substanzielle Beiträge. Die von der Universitätsbibliothek Johann Christian Senckenberg in Frankfurt am Main aufgebaute Virtuelle Fachbibliothek Biologie (vifabio) erleichtert den Zugang zu einigen wesentlichen Ressourcen; unter anderem können Hessische Bibliographie und Biological Abstracts sowie weitere Datenbanken in vifabio mit einer einzigen Suchanfrage parallel durchsucht werden.
Am 18.März 2010 stellte die Initiative „Wir wollen lernen“ den Antrag auf die Durchführung eines Volksentscheids. Verhindert werden soll die am 7.Oktober 2009 ins Schulgesetz aufgenommene schulpolitische Vereinbarung von Schwarz-Grün. Die Voraussetzung für den Antrag war ein erfolgreiches Volksbegehren, bei dem die Initiative 184000 Unterschriften sammelte, dreimal mehr als erforderlich. Zur Abstimmung, die am 18.Juli 2010 stattfinden wird, stehen die Forderungen an Bürgerschaft und Senat der Stadt * „eine Ausgliederung der Klassen 5 und 6 aus dem Gymnasium und anderen weiterführenden Schulen und deren Anbindung an die Grundschule als ‚Primarschule’ zu unterlassen“, * „Gymnasien und weiterführende Schulen in der bisherigen Form“ zu erhalten, * den Eltern wie bislang das Recht einzuräumen, „die Schulform für ihre Kinder nach Klasse 4 zu wählen.“ Sollte bei der Abstimmung eine Mehrheit der rund 245000 erforderlichen Wähler/innen zustimmen, wäre die Schulreform von Schwarz-Grün in wesentlichen Punkten gescheitert. ...
In der Doktorarbeit wurde ein Verfahren zur Ermittlung der Schwerpunkthöhe eines Fahrzeugs aus den Messwerten von Sensoren, die serienmäßig in vielen geländegängigen Fahrzeugen verbaut sind, entwickelt. Dieses Verfahren benötigt nur die Signale von Sensoren des elektronischen Stabilitätssystems (ESP) und eines Fahrwerks mit Luftfeder. Um die Höhe des Schwerpunkts zu bestimmen, wurde ein Modell entworfen, das die Drehbewegung des Fahrzeugs um seine Längsachse beschreibt. Eine der unbekannten Größen in diesem Modell ist das Produkt m_g\Deltah, wobei mit m_g die gefederte Masse des Fahrzeugs und mit Deltah der Abstand zwischen dem Schwerpunkt und der Wankachse des Fahrzeugs bezeichnet wird. Die Höhe des Schwerpunkts wird berechnet, indem zu diesem Abstand der als bekannt vorausgesetzte Abstand der Wankachse von der Straße addiert wird. Es wurden drei Varianten des Modells betrachtet. Die eine Modellvariante (stationäres Modell) beschreibt das Fahrzeugverhalten nur in solchen Fahrsituationen exakt, in denen die Wankgeschwindigkeit und die Wankbeschleunigung vernachlässigbar klein sind. In dieser Modellvariante wurden die Federkräfte mit einem detaillierten Modell der Luftfeder berechnet. Eine Eingangsgröße dieses Modells ist der Druck in den Gummibälgen der Luftfeder. Um diesen Druck zu ermitteln, wurde ein Algorithmus auf dem Steuergerät des Luftfedersystems implementiert. Um die Genauigkeit des Luftfedermodells zu testen und um die Abmessungen bestimmter Bauteile der Luftfeder zu ermitteln, wurden Messungen am Federungsprüfstand durchgeführt und eine Methode entwickelt, wie aus diesen Messungen die gesuchten Größen berechnet werden können. Bei den zwei übrigen Modellvarianten (dynamisches Modell) gelten die Einschränkung für die Fahrsituationen nicht. Die einzelnen Varianten des dynamischen Modells unterscheiden sich darin, dass das eine Mal die Feder- und Dämpferkonstanten als bekannt vorausgesetzt und das andere Mal aus den Sensorsignalen geschätzt werden. Passend zu jeder Modellvariante wurde ein Verfahren gewählt, mit dem Schätzwerte für das Produkt m_g\Deltah berechnet wurden. Des Weiteren wurde auch eine Methode entwickelt, mit der die Masse mg geschätzt wurde, ohne zuvor ein Wert für das Produkt m_g\Deltah zu ermitteln. Die Schätzwerte wurden unter Verwendung von Daten ermittelt, die bei einer Simulation und bei Messfahrten gewonnen worden sind. Das Ergebnis des Vergleiches der betrachteten Modellvarianten ist, dass die eine Variante des dynamischen Modells zum Teil falsche Werte für m_g\Deltah liefert, weil die Modellgleichungen ein nicht beobachtbares System bilden. Die andere Variante dieses Modells liefert nicht bei jeder Beladung exakte Werte, was vor allem daran liegt, dass in den Modellgleichungen dieses Modells ein konstanter Wert für die Federsteifigkeit angenommen wird. Bei Fahrzeugen mit Luftfeder ändert sich jedoch dieser Wert in Abhängigkeit von der Fahrzeugmasse. Die Werte von m_g\Deltah und mg können am genauesten mit dem stationären Modell ermittelt werden. Des Weiteren wurden Methoden entwickelt, die die Genauigkeit der durch den Schätzalgorithmus ermittelten Werte verbessern. So wurde zusätzlich zu dem Produkt m_g\Deltah und der Masse mg auch die Verteilung des Gewichtes auf die Vorder- und Hinterachse betrachtet. Es wurde ermittelt, welche Zusammenhänge zwischen dieser Verteilung und dem Produkt m_g\Deltah sowie zwischen dieser Verteilung und der Masse des Fahrzeugs bestehen. So konnte der Fehler in den Schätzwerten dieser Größen minimiert werden. Außerdem wurde auch der Zusammenhang zwischen dem Produkt m_g\Deltah und der Masse des Fahrzeugs ermittelt. Damit konnten die Schätzwerte dieser Größen genauer bestimmt werden. Aus den so gewonnenen Werten kann die Schwerpunkthöhe von einem Mercedes ML auf etwa 8cm genau berechnet werden. Diese Genauigkeit reicht aus, um das elektronische Stabilitätsprogramm auf die aktuelle Beladung des Fahrzeugs abzustimmen und damit einen Gewinn an Agilität für dieses Fahrzeug zu realisieren.
Background: Environmental noise is ubiquitous in population growth processes, with a well acknowledged potential to affect populations regardless of their sizes. It therefore deserves consideration in population dynamics modelling. The usual approach to incorporating noise into population dynamical models is to make some model parameter(s) (typically the growth rate, the carrying capacity, or both) stochastic and responsive to environment fluctuations. It is however still unclear whether including noise in one or/and another parameter makes a difference to the model performance. Here we investigated this issue with a focus on model fit and predictive accuracy. To do this, we developed three population dynamical models of the Ricker type with the noise included in the growth rate (Model 1), in the carrying capacity (Model 2), and in both (Model 3). We generated several population time series under each model, and used a Bayesian approach to fit the three models to the simulated data. We then compared the model performances in fitting to the data and in forecasting future observations. Results: When the mean intrinsic growth rate, r, in the data was low, the three models had roughly comparable performances, irrespective of the true model and the level of noise. As r increased, Models 1 performed best on data generated from it, and Model 3 tended to perform best on data generated from either Models 2 or Model 3. Model 2 was uniformly outcompeted by the other two models, regardless of the true model and the level of noise. The correlation between the deviance information criterion (DIC) and the mean square error (MSE) used respectively as measure of fit and predictive accuracy was broadly positive. Conclusion: Our results suggested that the way environmental noise is incorporated into a population dynamical model may profoundly affect its performance. Overall, we found that including noise in one or/and another parameter does not matter as long as the mean intrinsic growth rate, r, is low. As r increased, however, the three models performed differently. Models 1 and 3 broadly outperformed Model 2, the first having the advantage of being simple and more computationally tractable. A comforting result emerging from our analysis is the broad positive correlation between MSEs and DICs, suggesting that the latter may also be informative about the predictive performance of a model.

We show that average excess returns during the last two years of the presidential cycle are significantly higher than during the first two years: 9.8 percent over the period 1948 – 2008. This pattern in returns cannot be explained by business-cycle variables capturing time-varying risk premia, differences in risk levels, or by consumer and investor sentiment. In this paper, we formally test the presidential election cycle (PEC) hypothesis as the alternative explanation found in the literature for explaining the presidential cycle anomaly. PEC states that incumbent parties and presidents have an incentive to manipulate the economy (via budget expansions and taxes) to remain in power. We formulate eight empirically testable propositions relating to the fiscal, monetary, tax, unexpected inflation and political implications of the PEC hypothesis. We do not find statistically significant evidence confirming the PEC hypothesis as a plausible explanation for the presidential cycle effect. The existence of the presidential cycle effect in U.S. financial markets thus remains a puzzle that cannot be easily explained by politicians employing their economic influence to remain in power. The Anthonomus juniperinus group, with descriptions of two new species (Coleoptera: Curculionidae)

The Anthonomus juniperinus (Sanborn) species group is defined and two new species, Anthonomus sanborni, new species, and A. rileyi new species, from the United States are described, keyed and illustrated. The three species of the group are associated with the plant genus Juniperus and the larvae of A. juniperinus are known to develop in fungal galls of Gymnosporangium spp. as well as fruits of the Eastern redcedar, Juniperus virginiana L. The biology of the group and its taxonomic relationships to other species of Anthonomus Germar are also discussed.
The fauna of Phanaeini of the northeast of Brazil was investigated through fieldwork in the States of Ceará, Maranhão and Piauí, and through study of preserved material from other states. Seven species of Phanaeini are newly recorded from these three states. Of these, two species are also new records for the northeast region: Phanaeus melibaeus Blanchard and an unidentified Dendropaemon Perty species. A total of 13 new state records are given for eight of the 15 species of Phanaeini recorded from the northeast to date, including three new state genus records. A key is provided for identification of all species. Detailed distributional information is presented together with habitat and bait preferences and other ecological data for each species. The diversity and distribution of the tribe in the northeast is discussed in the context of regional biotopes and wider geographic ranges. The fauna is shown to be more diverse than previously believed, containing both endemic and widespread elements occurring in species assemblages that differ according to habitat type and elevation, leading to substantial complementarity of diversity amongst the main biogeographic provinces and biotopes of the region.
Four new species of Cydistomyia Taylor from New Guinea, C. missimiensis, C. madangiensis, C. waigani, and C. moresbyensis, are described and figured. A revised key to the females of New Guinea Cydistomyia and New Guinea collection records for 57 additional species of Tabanidae are provided. A table with the approximate longitudes and latitudes of all but one locality listed is provided.
Hiltonius carpinus carpinus Chamberlin, 1943 (Spirobolida: Spirobolidae), is authoritatively recorded from the United States for the first time; it is known only from southern/southeastern Arizona but should be expected in adjoining counties of New Mexico. The northernmost locality is the Pinaleno Mountains, Graham County, and its distribution extends to southern Mexico; the other subspecies, H. c. vulcan (Chamberlin, 1953), occurs in Guatemala. The range of H. c. carpinus includes the type locality of the enigmatic H. fossulifer (Pocock, 1908), lending credence to prior suggestions that the names are synonymous. Three new Mexican states – Durango, Jalisco, and Nuevo León – are documented for H. c. carpinus.
New species are described in the tribe Piezocerini: Gorybia rondonia sp. nov. from Brazil (Rondônia) and G. bahiensis sp. nov. from Brazil (Bahia); in the tribe Hexoplonini: Calycibidion rubricolle sp. nov. from Brazil (Bahia); in the tribe Ibidionini, Tropidina: Tropidion argentina sp. nov. from Argentina (La Rioja) and T. boliviensis sp. nov. from Bolivia (Santa Cruz); in the tribe Rhinotragini: Ommata (Ommata) albitarsis sp. nov. from Brazil (Rondônia); and in the tribe Rhopalophorini: Cosmisoma viridescens sp. nov. from Brazil (Bahia). To validate the tribal names, Hexoplon Thomson, 1864 and Tropidion Thomson, 1867, are here designated type genera of Hexoplonini and Tropidiina, respectively.
The human immunodeficiency virus type 1 (HIV-1) coreceptor use and viral evolution were analyzed in blood samples from an HIV-1 infected patient undergoing allogeneic stem cell transplantation (SCT). Coreceptor use was predicted in silico from sequence data obtained from the third variable loop region of the viral envelope gene with two software tools. Viral diversity and evolution was evaluated on the same samples by Bayesian inference and maximum likelihood methods. In addition, phenotypic analysis was done by comparison of viral growth in peripheral blood mononuclear cells and in a CCR5 (R5)-deficient T-cell line which was controlled by a reporter assay confirming viral tropism. In silico coreceptor predictions did not match experimental determinations that showed a consistent R5 tropism. Anti-HIV directed antibodies could be detected before and after the SCT. These preexisting antibodies did not prevent viral rebound after the interruption of antiretroviral therapy during the SCT. Eventually, transplantation and readministration of anti-retroviral drugs lead to sustained increase in CD4 counts and decreased viral load to undetectable levels. Unexpectedly, viral diversity decreased after successful SCT. Our data evidence that only R5-tropic virus was found in the patient before and after transplantation. Therefore, blocking CCR5 receptor during stem cell transplantation might have had beneficial effects and this might apply to more patients undergoing allogeneic stem cell transplantation. Furthermore, we revealed a scenario of HIV-1 dynamic different from the commonly described ones. Analysis of viral evolution shows the decrease of viral diversity even during episodes with bursts in viral load.
Background: The importance of the Notch signaling in the development of glomerular diseases has been recently described. Therefore we analyzed in podocytes the expression and activity of ADAM10, one important component of the Notch signaling complex. Methods: By Western blot, immunofluorescence and immunohistochemistry analysis we characterized the expression of ADAM10 in human podocytes, human urine and human renal tissue. Results: We present evidence, that differentiated human podocytes possessed increased amounts of mature ADAM10 and released elevated levels of L1 adhesion molecule, one well known substrate of ADAM10. By using specific siRNA and metalloproteinase inhibitors we demonstrate that ADAM10 is involved in the cleavage of L1 in human podocytes. Injury of podocytes enhanced the ADAM10 mediated cleavage of L1. In addition, we detected ADAM10 in urinary podocytes from patients with kidney diseases and in tissue sections of normal human kidney. Finally, we found elevated levels of ADAM10 in urinary vesicles of patients with glomerular kidney diseases. Conclusions: The activity of ADAM10 in human podocytes may play an important role in the development of glomerular kidney diseases.
Background: B. burgdorferi sensu lato (sl) is the etiological agent of Lyme borreliosis in humans. Spirochetes have adapted themselves to the human immune system in many distinct ways. One important immune escape mechanism for evading complement activation is the binding of complement regulators Factor H (CFH) or Factor H-like protein1 (FHL-1) to Complement Regulator-Acquiring Surface Proteins (CRASPs). Results: We demonstrate that B. garinii OspA serotype (ST4) PBi resist complement-mediated killing by binding of FHL-1. To identify the primary ligands of FHL-1 four CspA orthologs from B. garinii ST4 PBi were cloned and tested for binding to human CFH and FHL-1. Orthologs BGA66 and BGA71 were found to be able to bind both complement regulators but with different intensities. In addition, all CspA orthologs were tested for binding to mammalian and avian CFH. Distinct orthologs were able to bind to CFH of different animal origins. Conclusions: B. garinii ST4 PBi is able to evade complement killing and can bind FHL-1 to membrane expressed proteins. Recombinant proteins BGA66 can bind FHL-1 and human CFH, while BGA71 can bind only FHL-1. All recombinant CspA orthologs from B. garinii ST4 PBi can bind CFH from different animal origins. This partly explains the wide variety of animals that can be infected by B. garinii.
Die Untersuchung versucht als Genderstudie eine Antwort auf die Frage nach den Verhaltensweisen, dem Leben und Erleben von Frauen in Krisensituationen des Zwanzigsten Jahrhunderts zu geben. Der politische und soziale Fokus liegt in der Zeit des Nationalsozialismus und des Zweiten Weltkrieges.
Es handelt sich dabei um eine große Spannbreite von unterschiedlichen Biografien. Frauen sind als Zeitzeuginnen besonders tauglich, weil sie nicht nur durch ihr Geschlecht, sondern auch durch viele andere gesellschaftliche Positionen, Situationen und Handlungsspielräume im Alltag, und vor allem im Kriegsalltag und an der Heimatfront, geprägt sind.
Die Arbeit knüpft an wichtige Erkenntnisse der Frauen- und Genderforschung, der historischen Sozialforschung, der Biographie- und gerontologischen Forschung sowie der Oral History an.
Das Sample besteht aus acht Fraueninterviews, vier Gruppen von jeweils zwei Frauen. Da sind zwei adlige Frauen, die beide fast hundert Jahre lang und darüber hinaus gelebt haben. Beide haben ihren besonderen Bezugspunkt durch ihre Brüder als Beteiligte des militärischen Widerstands am 20. Juli 1944. Die anderen sechs Frauenbiografien stehen exemplarisch für ebenfalls drei besondere Gruppen. Da sind zwei jüdische Frauen, die Auschwitz überlebt haben, zwei Russinnen, die in ihren "Großen Vaterländischen Krieg" involviert waren und zwei bei der Wehrmacht dienstverpflichtete deutsche Frauen.
Das Sichtbarmachen von Lebenszusammenhängen kann die Ghettoisierung von Alten verhindern und Verbindungen knüpfen, die einer alternden Gesellschaft die Zukunft erleichtert.

Protein-Protein Interaktionen der NADH:Ubichinon-Oxidoreduktase (Kolmplex I) aus Y. lipolytica In der inneren Mitochondrienmembran sind die Atmungskettenkomplexe I bis IV und die ATP-Synthase lokalisiert. Parallel zum Elektronentransport werden Protonen von der NADH:Ubichinon-Oxidoreduktase (Komplex I), der bihydrochinon:Cytochrom c-Oxidoreduktase (Komplex III) und der Cytochrom c-Oxidase (Komplex IV) über die Membran in den Intermembranraum gepumpt. Der resultierende chemiosmotische Gradient wird von der ATPase verwendet um ATP zu generieren. Die NADH:Ubichinon- xidoreduktase ist der Startpunkt des Elektronentransfers. Der Komplex I wurde in verschiedenen Organismen in Interaktion mit Komplex III und IV beobachtet. Diese funktionellen Einheiten werden als Respirasomen oder Superkomplexe bezeichnet (Pfeiffer und Schägger, 2000). Der Komplex I in prokaryotischen Zellen stellt die kleinstmögliche Variante dieses Enzyms dar. Da er in der Lage ist, alle bioenergetischen Reaktionen durchzuführen, werden die beteiligten 14 Untereinheiten als zentrale Untereinheiten bezeichnet (Fearnley et al. 1992). In den Eukaryoten findet sich ein Enzym, das je nach Organismus bis zu 31 weitere, sogenannte akzessorische Untereinheiten aufweist. Da diese Untereinheiten keine Homologie mit bakteriellen Proteinen aufweisen, wird davon ausgegangen, dass sie eher eine Funktion im Assemblierungsprozess ausüben als am Elektronentransfer oder der Translokation der Protonen beteiligt zu sein. Ziel dieser Arbeit war es daher mögliche Interaktionspartner des Komplex I oder weitere Untereinheiten zu finden. Für die Detektion möglicher Interakteure sollten neben einem Two-Hybrid-Screen noch weitere Methoden etabliert werden. Als Modellsystem wurde Yarrowia lipolytica verwendet. Unter Verwendung des BacterioMatchII® Systems konnte die mitochondriale Malat-Dehydrogenase, ein Enzym des Citrat-Zyklus, als ein Interaktionspartner des Komplex I gefunden werden. Bei der Reaktion entsteht NADH, das Energieäquivalent, welches dem Komplex I Elektronen liefert. Im Kontext des Substratchannelings deutet die Interaktion der mitochondrialen Malat- ehydrogenase auf hochgeordnete Stukturen im Matrixraum hin, die einen Transfermechanismus oder eine zielgerichtete Diffusion des NADH gewährleisten könnten. Durch die Kombination der Methoden Immunpräzipitation, Gelelektrophorese und MALDI-MS konnten erstmals Hinweise auf Superkomplexe in Y. lipolytica gefunden werden, was dann durch mehrdimensionale Gelelektrophorese bestätigt werden konnte. In dieser Arbeit konnten die Superkomplexe I1III2 ,I1III2IV1 und I1III2IV2 identifiziert werden. Ein vollständiges Respirasom mit einer Stöchiometrie von I1III2IV4 konnte nicht gezeigt werden. Desweiteren konnte die Anwesenheit eines Komplex I-Dimers nachgewiesen werden. In dem bisher postulierten Modell der respiratory strings werden die Superkomplexe durch tetramere Komplex IV-Module zu hochmolekularen Strukturen verknüpft (Wittig et al., 2006). Die detektierten Komplex I-Dimere lassen vermuten, dass sie zusammen mit dem tetrameren Komplex IV als Linker-Moleküle die Superkomplexe zu einem zweidimensionalen respiratory patch verbinden. Zudem konnte eine neue Untereinheit des Komplex I, genannt NEBM, durch das Zusammenspiel von LILBID- und MALDI-Massenspektrometrie und Gelelektophorese identifiziert werden. Durch MALDI-Massenspektrometrie wurde diese Untereinheit de novo sequenziert. Die Charakteristik des identifizierten Proteins entspricht den sogenannten single-transmembrane domain Untereinheiten (Brandt et al., 2005; Brandt 2006), die eventuell eine Funktion in der Assemblierung des Komplex I einnehmen könnten. Weder bei den Messungen der NADH:HAR- noch der dNADH:DBQ-Aktivität zeigte der Deletionsstamm enzymatisch aktiven Komplex I. Weder in blau-nativer Elektrophorese noch in silbergefärbten 2D SDS-Gelen war ein Komplex I noch ein Subkomplex sichtbar und auch In-Gel-Aktivitäts-Tests verliefen für die Detektion von Komplex I im Deletionsstamm negativ. Erst durch eine Western Blot Analyse konnte ein Subkomplex mit einer Größe von ca. 300kDa detektiert werden. Unter Berücksichtigung der für den Rinderkomplex vorgeschlagenen Einteilung des Komplex I in die Subkomplexe Iα und Iβ (Brandt et al. 2006) könnte der detektierte Subkomplex Iβ zugeordnet werden, da er zwei Untereinheiten beinhaltet, die diesem Komplex zuzuordnen sind. Die NEBM Untereinheit könnte also an der Assemblierung der NADH:Ubichinon-Oxidoreduktase beteiligt sein. Erstmals konnten Superkomplexe in Y. lipolytica Mitochondrien nachgewiesen werden. Die Identifizierung der neuen NEBM Untereinheit vervollständigt die Kenntnisse über die Untereinheitenzusammensetzung. Das Wechselspiel mit anderen Proteinen und die Zusammensetzung des Komplexes können für die Strukturaufklärung und zur Analyse des Reaktionsmechanismus wichtig sein. Somit leisten diese Ergebnisse einen wichtigen Beitrag zur Charakterisierung der NADH:Ubichinon-Oxidoreduktase aus Y. lipolytica.
Introduction: Hypothermia improves survival and neurological recovery after cardiac arrest. Pro-inflammatory cytokines have been implicated in focal cerebral ischemia/reperfusion in-jury. It is unknown whether cardiac arrest also triggers the release of cerebral inflammatory molecules, and whether therapeutic hypothermia alters this inflammatory response. This study sought to examine whether hypothermia or the combination of hypothermia with anes-thetic postconditioning with sevoflurane affect cerebral inflammatory response after cardio-pulmonary resuscitation. Methods: Thirty pigs (28 - 34kg) were subjected to cardiac arrest following temporary coro-nary artery occlusion. After 7 minutes of ventricular fibrillation and 2 minutes of basic life support, advanced cardiac life support was started according to the current AHA guidelines. Return of spontaneous circulation was achieved in 21 animals who were randomized to ei-ther normothermia at 38degreesC, hypothermia at 33degreesC or hypothermia at 33degreesC combined with se-voflurane (each group: n = 7) for 24 hours. The effects of hypothermia and the combination of hypothermia with sevoflurane on cerebral inflammatory response after cardiopulmonary resuscitation were studied using tissue samples from the cerebral cortex of pigs euthanized after 24 hours and employing quantitative RT-PCR and ELISA techniques. Results: Global cerebral ischemia following resuscitation resulted in significant upregulation of cerebral tissue inflammatory cytokine mRNA expression (mean +/- SD; interleukin (IL)-1beta 8.7 +/- 4.0, IL-6 4.3 +/- 2.6, IL-10 2.5 +/- 1.6, tumor necrosis factor (TNF)alpha 2.8 +/- 1.8, intercellular adhesion molecule-1 (ICAM-1) 4.0 +/- 1.9-fold compared with sham control) and IL-1beta protein concentration (1.9 +/- 0.6-fold compared with sham control). Hypothermia was associated with a significant (P <0.05 versus normothermia) reduction in cerebral inflammatory cytokine mRNA expression (IL-1beta 1.7 +/- 1.0, IL-6 2.2 +/- 1.1, IL-10 0.8 +/- 0.4, TNFalpha 1.1 +/- 0.6, ICAM-1 1.9 +/- 0.7-fold compared with sham control). These results were also confirmed for IL-1beta on protein level. Experimental settings employing hypothermia in combination with sevoflurane showed that the volatile anesthetic did not confer additional anti-inflammatory effects com-pared with hypothermia alone. Conclusions: Mild therapeutic hypothermia resulted in decreased expression of typical ce-rebral inflammatory mediators after cardiopulmonary resuscitation. This may confer, at least in part, neuroprotection following global cerebral ischemia and resuscitation.
Dynamical systems driven by Gaussian noises have been considered extensively in modeling, simulation, and theory. However, complex systems in engineering and science are often subject to non-Gaussian fluctuations or uncertainties. A coupled dynamical system under a class of Lévy noises is considered. After discussing cocycle property, stationary orbits, and random attractors, a synchronization phenomenon is shown to occur, when the drift terms of the coupled system satisfy certain dissipativity and integrability conditions. The synchronization result implies that coupled dynamical systems share a dynamical feature in some asymptotic sense.
Die Rolle von Antiepileptika im Zusammenhang mit dem Auftreten letaler kardialer Arrhythmien wird kontrovers diskutiert. Zum aktuellen Zeitpunkt ist bei Patienten mit Epilepsie die Ursache für die im Vergleich zur Normalbevölkerung erhöhte Mortalität nicht hinreichend geklärt. Die Ätiologie und Pathophysiologie des Sudden unexpected death in epilepsy patients (SUDEP) ist nur teilweise bekannt, wodurch die Durchführung präventiver Maßnahmen nur bedingt möglich ist. Es konnten bestimmte Risikofaktoren für das Auftreten eines SUDEP identifiziert werden, wobei unter anderem die antikonvulsive Polytherapie einen unabhängigen Risikofaktor darstellt. Bezüglich der pathogenetischen Mechanismen spielen neben der Entstehung eines neurogenen pulmonalen Ödems und einer zentralen Apnoe auch kardiale Arrhythmien eine bedeutende Rolle. Valproinsäure als ein sehr häufig eingesetztes Antiepileptikum wird verdächtigt, eine Verlängerung der QT-Zeit im Elektrokardiogramm (EKG) zu bewirken. Eine Verlängerung der Plateauphase des Aktionspotentials führt zu einer instabilen Repolarisation, wodurch einfallende frühe Nachdepolarisationen bereits wieder ein Aktionspotential auslösen können. Ein erworbenes Long-QT-Syndrom (LQTS), zu dem auch die medikamentenbedingte QT-Verlängerung zählt, birgt das Risiko, durch die Auslösung von Herzrhythmusstörungen im Sinne von Torsade de Pointes an einem plötzlichen Herztod zu versterben. Im Rahmen dieser prospektiven Studie wurden 50 Kinder mit Epilepsie unter der Therapie mit Valproinsäure bezüglich des Auftretens kardialer Nebenwirkungen untersucht. Das Studienprotokoll beinhaltete ein standardisiertes EKG mit Bestimmung der Zeiten, Lagetypen, Hypertrophie- und Schädigungszeichen. Parallel wurde die Dosierung von Valproinsäure und deren Plasmaspiegel bestimmt. Ziel der vorliegenden Arbeit war es herauszufinden, ob die antikonvulsive Therapie mit Valproinsäure durch die Auslösung eines medikamentenbedingten LQTS zu einem erhöhten kardialen SUDEP-Risko bei pädiatrischen Epilepsiepatienten führen könnte. Die 25 weiblichen und 25 männlichen Patienten im mittleren Alter von 12,2 Jahren wurden wegen verschiedener Epilepsieformen behandelt. Sie nahmen Valproinsäure durchschnittlich über eine Zeitdauer von 5,8 Jahren ein (Range 3 Monate bis 32 Jahre), wobei 58% eine Monotherapie, 26% in Zweier- und 16% eine Dreier-56 Kombinationstherapie erhielten. Die durchschnittliche Dosierung lag bei 26,6 mg/ kgKG/ Tag (Range 10,5 bis 63,8 mg/ kgKG/ Tag), der im Plasma gemessene Valproinsäure-Spiegel zwischen 19,7 und 147,4 μg/ml (Mittelwert 88,8 ± 24,3 μg/ml; Median 91 μg/ml). Die gemessenen EKG-Zeiten lagen alle im altersentsprechenden Normbereich: --> PR-Intervall: Range 100-170 ms, Median: 124 ms, Mittelwert: 126,7 ± 18,5 ms --> QRS-Dauer: Range 56-108 ms, Median 79 ms, Mittelwert 79,94 ± 11,3 ms --> QT-Zeit: Range 264-400 ms, Median 348 ms, Mittelwert 343,34 ± 35,9 ms -->QTc-Zeit (Normwert < 440 ms bei Jungen und < 460 ms bei Mädchen): Range 377-433 ms, Median 403 ms, Mittelwert 402,9 ± 11,1 ms. Bezüglich der Lagetypen fanden sich bei 42% ein Indifferenz-, bei 46% ein Steil- und bei 8% ein Linkstyp. Bei einem Patienten mit Tuberöser Sklerose lag ein Rechtstyp vor. Dieser wies einen vollständigen Rechtsschenkelblock auf, welcher bei Zustand nach reseziertem Rhabdomyom des rechten Ventrikels durch die operative Durchtrennung des Reizleitungssystems verursacht wurde. Den Ergebnissen dieser Untersuchung zufolge lassen sich bei Kindern unter Therapie mit Valproinsäure keine Veränderungen im EKG, insbesondere keine QT-Verlängerungen, als möglicher Risikofaktor für eine kardiale Arrhythmie nachweisen. Aus diesen Daten lässt sich schlussfolgern, dass dieses Antiepileptikum nicht das Risiko erhöht, an einem QT-bedingten SUDEP zu versterben. Trotz eines erhöhten Risikos unter einer antiepileptischen Polytherapie kann bisher nicht von einer erhöhten Inzidenz durch die Therapie mit einem bestimmten Antikonvulsivum ausgegangen werden. Weitere Untersuchungen sind notwendig, um die Rolle der einzelnen Pharmaka in diesem Zusammenhang besser einschätzen zu können. Bisher ist davon auszugehen, dass eine gute Anfallskontrolle das SUDEP-Risiko entscheidend verringert. Bis der Pathomechanismus des SUDEP detaillierter geklärt ist, gelten eine Optimierung der antiepileptischen Therapie und eine Vermeidung von bisher bekannten Risikofaktoren als sinnvollste präventive Maßnahmen. Abschließend ist festzuhalten, dass sich im Rahmen dieser Untersuchung bei Kindern unter antikonvulsiver Therapie mit Valproinsäure kein Hinweis auf QT-Verlängerungen als möglicher Risikofaktor für einen SUDEP fand.
Die zellfreie Proteinsynthese hat sich in den letzten Jahren zu einem potenten Werkzeug – auch in der Produktion von Membranproteinen – entwickelt. Da keine lebenden Zellen genutzt werden, kann der Prozess der präparativen Membranproteinproduktion vereinfacht und individuell optimiert werden. Im Gegensatz zu konventionellen zellbasierten Expressionssystemen gewährleistet die zellfreie Proteinsynthese die direkte Zugänglichkeit zum Reaktionsort und damit die Möglichkeit der unmittelbaren Kontrolle. Dies ermöglicht eine genaue Anpassung der Reaktionsbedingungen auf das Zielprotein. Die Verbesserung und Entwicklung neuer Modi der zellfreien Membranproteinsynthese war ein Teil der vorliegenden Arbeit. Setzt man dem Zellfrei-System von Außen keine hydrophobe Umgebung zu, so präzipitiert das neu-synthetisierte Membranprotein im Reaktionsmix (P-CF). Interessanter Weise unterscheiden sich diese Präzipitate von den aus der E.coli zellbasierten Proteinproduktion bekannten Einschlußkörperchen, da sie sich teilweise leicht in mildem Detergenz resolubilisieren lassen. Zudem konnte für verschiedene Transportproteine, die aus Präzpitat resolubilisiert und danach in Liposomen rekonstituiert wurden, spezifische Transportaktivität gezeigt werden (z.B. eukaryotische Ionentransporter, Multi-Drug Resistenzproteine von E.coli). Alternativ können die Membranproteine direkt, durch die Zugabe von Detergenzien in den Reaktionsmix, solubilisiert werden (D-CF). Um die einzelnen Expressionsmodi zu optimieren wurden 24 gebräuchliche Detergenzien auf ihre Eigenschaft hin gestestet, strukturell sehr unterschiedliche Membranproteine zu solubilisieren. Die Familie der langkettigen Polyoxyethylen-alkyl Ether hat sich dabei als sehr geeignet erwiesen um das prokaryotische α-helikale Multi-Drug Resistenzprotein EmrE, den bakteriellen vornehmlich aus ß-sheets bestehenden Transporter Tsx und den eukaryotischen G-Protein gekoppelten Vasopressin Rezeptor V2R direkt im D-CF Modus zu solubilisieren. Zudem konnte eine Abhängigkeit der spezifischen Aktivität von Tsx vom verwendeten Expressionsmodus bzw. des verwendeten Detergenz mit Hilfe der Black Lipid Membrane´ Methode gezeigt werden. Die Expression eines repäsentativen Teils von 134 Zielproteinen des inneren Membranproteoms von E.coli wurde in drei verschiedenen Zellfrei-Expressionsmodi getestet. Ein an jedes Zielprotein des Membranroteoms C-terminal fusioniertes GFP diente der Konzentrationsbestimmung im D-CF Expressionsmodus. Die Faltung von GFP ist in Anwesenheit von Detergenz signifikant reduziert. Zunächst wurden alle Zielproteine in einem batch´ System im D-CF Modus im Mikrotiterplatten Maßstab mit Hilfe eines Roboters hergestellt. Die Etablierung einer robotergestützten Plattform, welche das Pipettieren, Inkubieren und Detektieren kombiniert, diente als Grundlage für den Herstellungsprozess des Membranroteoms von E.coli in einem Medium-Durchsatz Verfahren in batch´ Konfiguration. In dieser ersten Stufe des Screens im D-CF Modus konnten 84 Zielproteine (63%) aufgrund der detektierten GFP-Fluoreszens in Mengen von 1 bis 60μg pro mL Reaktion als erfolgreich produziert identifiziert werden. Zudem wurde das Membranproteom in dem effektiveren continous exchange (CE) Verfahren im P-CF, wie auch im D-CF Modus wiederholt exprimiert. Im Vergleich zur batch´ Konfiguration konnten im CE D-CF Modus deutlich mehr Zielproteine (75%) als positiv identifiziert werden. 16 Zielproteine wurden dabei bereits in Expressionsmengen von mehr als 100μg solubilisierte Membranproteinfusion pro mL Reaktionsmix gewonnen. 99 Zielproteine (74%) konnten als positiv identifiziert werden, nachdem die unlösliche Fraktion der CE P-CF Reaktion elektrophoretisch getrennt und angefärbt wurde. Für 66 Kandidaten (49%) stellt das produzierte Protein nach Coomassie-Färbung eine dominante Bande, und damit (semi-)präparative Proteinmengen, dar. Der Erhalt von Detergenz-solubilisierten Membranproteinproben von hoher Qualität ist ein wichtiger Schritt zur Gewinnung struktureller sowie biochemischer Daten. Das E.coli α-helikale Multi- Drug Resistenz Protein SugE konnte im CE P-CF Verfahren in präparativen Mengen von mehr als 2mg Protein pro mL des Reaktionsansatzes gewonnen werden. Durchgeführte analytische Größenausschlusschromatographie zeigte, dass der Transporter unter optimierten Reaktionsbedingungen in einem homogenen, schlanken Peak eluiert. Mittels elektronenmikroskopischer Gefrierbruchanalysen konnte eine effiziente und homogene Rekonstitution von SugE in E.coli Liposomen gezeigt werden. Bindungsstudien unter der Verwendung fluoreszensbasierter Anisotropie-Messungen haben gezeigt dass Proflavin – im Gegenteil zu Ethidium – ein Substrat von SugE ist. YedZ ist ein 24kDa leucinreiches Membranprotein mit sechs putativen Transmembransegmenten und enthält zwei Kofaktoren, ein Häm b und ein Flavin-mononukleotid (FMN). Im P-CF Modus exprimiertes YedZ kann effizient in den Detergenzien LMPG, LPPG, SDS und DPC resolubilisiert werden. Analytische Größenausschlusschromatographie zeigte einen symmetrischen Elutionspeak der apo-Form. Mittels CD-Spektroskopie des gereinigten apo-YedZ in 0.02% DDM wurde ein α-helikaler Sekundärstrukturanteil von 55% ermittelt. Zur Gewinnung von holo-YedZ wurde anstatt Hämb das chemisch verwandte Hemin eingesetzt. Die aufgenommenen UV/Vis Spektren der zellfrei produzierten holo-YedZ Proteinprobe in ihrer oxydierten und reduzierten Form, zeigen zu einer in vivo exprimierten Vergleichsprobe identische Absorptionsmaxima. Für sechs G-Protein gekoppelten Rezeptoren konnte die zellfreie Expression in präparativen Mengen gezeigt werden. Das Steroid-Derivat Digitonin, sowie einzelne Mitglieder der Detergenzfamilie der langkettigen Polyoxyethylen-alkyl Ether, wurden als am geeignetsten für die lösliche Expression der GPCRs im CE D-CF Verfahren ermittelt. Löslich in Anwesenheit von Brij78 produzierter GPCR Proben, zeigten nach Negativfärbung in elektronenmikroskopischen Einzelpartikelanalysen eine homogene Probenpräparation und geben Hinweis auf eine strukturelle Dimerisierug der Rezeptoren. Detergenzsolubilisierte Rezeptoren konnten in Liposomen, basierend auf E.coli Lipid-Mischungen, rekonstituiert werden. Elektronen-mikroskopische Gefrierbruchanalysen zeigten eine homogene Rekonstitution, welche auf eine funktionelle Faltung der Rezeptoren schließen lässt.
Zahlreiche Studien haben gezeigt, dass sich trotz der Verwendung von Dentinadhäsiven in deninbegrenzten Klasse-II-Kavitäten keine randspaltfreien Füllungen erzielen lassen. Die Ergebnisse dieser in-vitro-Studie zeigen dies ebenfalls, dass mit Dentinadhäsiven der fünften und sechsten Generation keine komplett spaltfreien Kompositfüllungen im Dentin hergestellt werden können. Zu diesem Schluss kam auch Manhart, J. et al 2000. Selbst bei Anwendung moderner Dentinadhäsivsysteme unter Einhaltung aller Verarbeitungsvorschriften lässt sich das Zustandekommen einer Komposit-Dentinhaftung in Klasse-II-Kavitäten nicht mit Sicherheit vorhersagen. Bei allen getesteten Dentinadhäsiven wurde an den Kavitätenrändern Farbstoffpenetration nachgewiesen. Die Farbstoffpenetration zeigte an den Kavitäten die Tendenz von approximal nach pulpal geringer zu werden. Optibond Solo mit Füllstoffgehalt erreichte in der Farbstoffpenetration am zervikalenFüllungsrand mit (54,54 %) und ohne (31,89 %) Liquorsimulation die besten Resultate. Auch am oralen Füllungsrand hatte Optibond Solo mit (36,38 %) und ohne (37,09 %) Liquorfluss die besten Ergebnisse. An der vestibulären Kavitätengrenze waren Optibond Solo und Syntac Sprint an der Spitze: mit Liquorsimulation Syntac Sprint 32,68 %, Optibond Solo 38,42 %; ohne Liquorsimulation Syntac Sprint 35,97 %, Optibond Solo 35,3 %. Alle Ränder zusammen betrachtet, wies ebenfalls Optibond Solo die geringste Farbstoffpenetration auf: mit Liquor 43,2 %, ohne Liquor 34,72 %. Das wasserhaltige Syntac SC belegte den letzten Platz bei Auswertung aller Ränder zusammen mit Liquor 82,89 %, ohne Liquorsimulation war hingegen Etch & Prime 3.0 absolutes Schlusslicht (80,03 %). Das schlechteste Ergebnis erreichte Syntac SC ebenfalls am vestibulären Füllungsrand ohne (75,59 %) und mit Liquorsimulation (81,16 %), sowie zervikal mit Liquor (95,32 %). Syntac SC konnte genauso mit der angewandten Moist-bonding-Technik in der REM (20 % "perfekte Ränder" vor und 8 % nach Thermocycling) nur den letzten Platz einnehmen. Das acetonbasierende Adhäsiv Syntac Sprint erlangte in der REM sowohl vor (77 %) als auch nach (76 %) Temperaturwechselbadbelastung den höchsten Anteil an "perfekten Rändern", gefolgt von Optibond Solo mit 65 % vor und 50 % nach Thermocycling. Die Temperaturwechselbadbelastung hatte Einfluss auf die Qualität der Randadaptation. Bei allen untersuchten Dentinadhäsiven reduzierte sich der prozentuale Anteil an perfekten Rändern nach Thermocycling im Vergleich zuvor. Die Qualität des Zustandekommens eines akzeptablen Füllungsrandes ist von der chemischen Zusammensetzung, insbesondere dem enthaltenen Lösungsmittel, dem Füllstoffgehalt und der Dentinfeuchtigkeit abhängig. Die Durchführung der Simulation des Liquors führte am zervikalen Rand bei Syntac SC (ohne: 92,54 %; mit: 95,32 %), Optibond Solo (ohne: 31,89 %; mit: 54,55 %) und Scotchbond 1 (ohne: 63,44 %; mit: 83,24 %) zur erhöhten Farbstoffpenetration - im Falle von Etch & Prime 3.0 (ohne: 93,11 %; mit: 86,42 %), Syntac Sprint (ohne: 81,98 %; mit: 73,75 %) und Prime & Bond NT (ohne: 88,86 %; mit: 81,01 %) jedoch zu einer verringerten Penetration des Methylenblaus.
The purpose of this essay is to assess the automatic exchange of information as described in EU Directive 2003/48 of 3 June 2003 on taxation of savings income in the form of interest payments with regard to the fundamental right of the individual to a private life, to banking secrecy and the freedoms on which the European internal market is based. The assessment reveals the conflicts of interests and values involved in the holding by banks (particularly those offering private banking services) of increasingly extensive, detailed and intimate information about their clients and in the automatic processing of that information by ever more powerful and sophisticated systems. Banking secrecy plays an essential role in protecting clients against the dangers which the disclosure of such information without their permission might produce. Banking secrecy exists not only in Luxembourg but also in many other European countries, and in Germany and France in particular it is not very different from the system applying in Luxembourg. While the French and German tax authorities do have some investigative powers not enjoyed by their Luxembourg counterparts, those powers are strictly circumscribed and cannot rely on the electronic exchange of information set out in EU Directive 2003/48/EC. While banking secrecy is totally incompatible with the electronic exchange of information, the core question is whether the latter can be reconciled with the respect for private life. In a Europe that sets itself up as the cradle of human rights, the general and en-masse exchange of private information cannot provide adequate and sufficient guarantees that the information exchanged will not be misused. The amount of interference in private life is clearly out of proportion to the public interest involved and is contrary to sub-section 2, article 8 of the European Convention for the Protection of Human Rights and Fundamental Freedoms and to articles 7 and 8 of the Charter of Fundamental Rights of the European Union. Since the automatic exchange of information at least potentially risks restricting the free flow of capital among Member States and discouraging the use of transborder banking services, its compliance with the fundamental principles of the internal market also needs to be closely examined. The restrictions imposed by such exchange very probably go beyond the limits within which the free movement of capital and services is possible. The European Court of Justice has found that there is no proportionality if the measures supposedly undertaken in the general interest are actually based on a general presumption of tax evasion or tax fraud. However, it would be true to say that the ECJ does not always examine the tax restrictions placed on the free movement of capital particularly thoroughly to ensure that they are necessary or proportionate. The economic effectiveness of the automatic exchange of information is far from being proved and involves significant cost to the banks providing the information and to the tax authorities using it. To date the system does not appear to have produced any significant new tax revenue nor does it prevent the continuing outflow of capital from Europe. Yet withholding at source, which respects individual and economic freedoms, does generate tax revenue that is cost-free to the State. Exchange of information on request in justified cases using the OECD Tax Convention on Income and Capital model does also fight tax fraud while at the same time providing citizens with the guarantees required to ensure their private lives are respected. A combination of these two systems - withholding at source and exchange of information on request in justified cases - would create the proper balance between the public and private interest that the automatic exchange of information cannot provide.
This paper analyzes the impact of blockownership dispersion on firm value. Blockholdings by multiple blockholders is a widespread phenomenon in the U.S. market. It is not clear, however, whether dispersion among blockholder is preferable to having a more concentrated ownership structure. To test for the direction of the effect, we use a large dataset of U.S. firms that combines blockholder information, shareholder rights information, debt ratings, accounting information, and financial markets information. We find that a large fraction of aggregated block ownership negatively affects Tobin’s Q. The negative impact is larger if blockowners are more dispersed, suggesting that a concentrated ownership structure is to be preferred on average. Results are robust to controlling for blockholder type as well as proxies for shareholder rights. Our empirical findings are also confirmed if we study the impact of ownership dispersion on firm debt ratings rather than Tobin’s Q. JEL Classification: G3, G32
We estimate the risk and expected returns of private equity investments based on the market prices of exchange traded funds of funds that invest in unlisted private equity funds. Our results indicate that the market expects unlisted private equity funds to earn abnormal returns of about one to two percent. We also find that the market expects listed private equity funds to earn zero to marginally negative abnormal returns net of fees. Both listed and unlisted private equity funds have market betas close to one and positive factor loadings on the Fama-French SMB factor. Private equity fund returns are positively correlated with GDP growth and negatively correlated with the credit spread. Finally, we find that market returns of exchange traded funds of funds and listed private equity funds predict changes in self-reported book values of unlisted private equity funds.
A call on art investments

The art market has seen boom and bust during the last years and, despite the downturn, has received more attention from investors given the low interest environment following the financial crisis. However, participation has been reserved for a few investors and the hedging of exposures remains dificult. This paper proposes to overcome these problems by introducing a call option on an art index, derived from one of the most comprehensive data sets of art market transactions. The option allows investors to optimize their exposure to art. For pricing purposes, non-tradability of the art index is acknowledged and option prices are derived in an equilibrium setting as well as by replication arguments. In the former, option prices depend on the attractiveness of gaining exposure to a previously non-traded risk. This setting further overcomes the problem of art market exposures being dificult to hedge. Results in the replication case are primarily driven by the ability to reduce residual hedging risk. Even if this is not entirely possible, the replication approach serves as pricing benchmark for investors who are significantly exposed to art and try to hedge their art exposure by selling a derivative. The objective of this study is to determine whether specific industries across countries or within countries are more likely to reach a stage of profitability and make a successful exit. In particular, we assess whether firms in certain industries are more prone to exit via IPO, be acquired, or exit through a leveraged buy-out. We are also interested in analyzing whether substantial differences across industries and countries arise when looking separately at the success’ rate of firms which have received venture funding at the early seed and start-up stages, vis-à-vis firms that received funding at later stages. Our results suggest that, inasmuch as some of the differences in performance can be explained by country-specific factors, there are also important idiosyncratic differences across industries: In particular, firms in the biotech and the medical / health / life science sectors tend to be significantly more likely to have a successful exit via IPO, while firms in the computer industry and communications and media are more prone to exit via merger or acquisition. We study the impact of the arrival of macroeconomic news on the informational and noise-driven components in high-frequency quote processes and their conditional variances. Bid and ask returns are decomposed into a common ("efficient return") factor and two market-side-specific components capturing market microstructure effects. The corresponding variance components reflect information-driven and noise-induced volatilities. We find that all volatility components reveal distinct dynamics and are positively influenced by news. The proportion of noise-induced variances is highest before announcements and significantly declines thereafter. Moreover, news-affected responses in all volatility components are influenced by order flow imbalances. JEL Classification: C32, G14, E44

Intracarotid administration of human bone marrow mononuclear cells in rat photothrombotic ischemia

Background: Increasing evidence suggests that cell therapy improves functional recovery in experimental models of stroke and myocardial infarction. So far only small pilot trials tested the effects of cell therapy in stroke patients, whereas large clinical trials were conducted in patients with ischemic heart disease. To investigate the therapeutic benefit of cell therapy to improve the recovery after stroke, we determined the efficacy of bone marrow derived mononuclear cells, which were shown to improve the recovery in experimental and clinical acute myocardial infarction studies, in a rat stroke model. Methods: Adult male Wistar rats were randomly assigned to receive either five million human bone marrow mononuclear cells (hBMC) or placebo intraarterially 3 days after photothrombotic ischemia. For immunosuppression the animals received daily injections of cyclosporine throughout the experiment, commencing 24 hours before the cell transplantation. A battery of behavioural tests was performed before and up to 4 weeks after ischemia. Results: Body temperature and body weight revealed no difference between groups. Neurological deficits measured by the Rotarod test, the adhesive-removal test and the cylinder test were not improved by hBMC transplantation compared to placebo. Conclusions: This study demonstrates that hBMC do not improve functional recovery when transplanted intraaterially 3 days after the onset of focal cerebral ischemia. A possible reason for the failed neurological improvement after cell therapy might be the delayed treatment initiation compared to other experimental stroke studies that showed efficacy of bone marrow mononuclear cells.
Characteristics of the Christian movement in North Korea and the former GDR are fundamentally different and based on the facts presented before. While the Christians in the North can only operate in the underground, at risk of being sentenced to work in a labour camp, to torture or even to execution when identified as Christian, the Christians in the GDR were freer although they suffered from repression, too. Compared to non-members they had clearly many disadvantages. However, as a state-independent organization, they were officially recognised and therefore an institution that was capable of leading a dialogue with the government. Moreover, they were able to connect themselves to other political alternative groups representing together a quite large group that was dissatisfied with the present situation without having the prior goal to overthrow the state. Awareness, dialogue and organization - these factors can be considered as decisive advantages in this process. As for North Korea creating awareness and dialogue seems to be a target for support. Even from a humanitarian angle alone. Awareness comes through information and that should be increased, e.g. through mutual development projects, continuous human and food aid, or why not initiating town twining? Just to name a few. More contact will generate more awareness and people will start rethinking their attitudes. Even if the elite benefits it may have a positive impact. Dialogue is pivotal in order to exchange one’s ideas und rethink one’s standpoints. Thus, a dialogue with the outside world shall be maintained and a dialogue among the people within the country shall be supported. Time will show if the introduction of mobile phones and the internet in North Korea will increase dialogue and will finally lead to the third suggested step of grass roots community building - organization. An ecclesial movement alone did not cause the change in East Germany and it will not do in North Korea. But it provided an important ground to connect people and accelerate change in the long run.
Vascularization of the vertebrate brain takes place during embryonic development from a preformed perineural vascular plexus. As a consequence of the intimate contact with neuroectodermal cells the vessels, which are entering the brain exclusively via sprouting angiogenesis, acquire and maintain unique barrier properties known as the blood-brain barrier (BBB). The endothelial BBB depends upon the close association of endothelial cells with pericytes, astrocytes, neurons and microglia, which are summarized in the term neuro-vascular unit. Although it is known since decades that the CNS tissue provides the cues for BBB induction and differentiation in endothelial cells, the molecular mechanism remained obscure. Only recently, the canonical Wnt/beta-catenin pathway and the Wnt7a/7b growth factors have been implicated in brain angiogenesis on the one hand and in BBB induction on the other. This breakthrough in understanding the differentiation of the brain vasculature prompted us to review these findings embedded in the emerging concepts of Wnt signaling in the vasculature. In particular, interactions with other pathways that are crucial for vascular development such as VEGF, Notch, angiopoietins and Sonic hedgehog are discussed. Finally, we considered the potential role of the Wnt pathway in vascular brain pathologies in which BBB function is hampered, as for example in glioma, stroke and Alzheimer's disease.
Background: Studies on the development of the nervous system and the musculature of invertebrates have become more sophisticated and numerous within the last decade and have proven to provide new insights into the evolutionary history of organisms. In order to provide new morphogenetic data on opisthobranch gastropods we investigated the neuromuscular development in the nudibranch Aeolidiella stephanieae Valdez, 2005 using immunocytochemistry as well as F-actin labelling in conjunction with confocal laser scanning microscopy (cLSM). Results: The ontogenetic development of Aeolidiella stephanieae can be subdivided into 8 stages, each recognisable by characteristic morphological and behavioural features as well as specific characters of the nervous system and the muscular system, respectively. The larval nervous system of A. stephanieae includes an apical organ, developing central ganglia, and peripheral neurons associated with the velum, foot and posterior, visceral part of the larva. The first serotonergic and FMRFamidergic neural structures appear in the apical organ that exhibits an array of three sensory, flask-shaped and two non-sensory, round neurons, which altogether disappear prior to metamorphosis. The postmetamorphic central nervous system (CNS) becomes concentrated, and the rhinophoral ganglia develop together with the anlage of the future rhinophores whereas oral tentacle ganglia are not found. The myogenesis in A. stephanieae begins with the larval retractor muscle followed by the accessory larval retractor muscle, the velar or prototroch muscles and the pedal retractors that all together degenerate during metamorphosis, and the adult muscle complex forms de novo. Conclusions: Aeolidiella stephanieae comprises features of the larval and postmetamorphic nervous as well as muscular system that represent the ground plan of the Mollusca or even the Trochozoa (e. g. presence of the prototrochal or velar muscle ring). On the one hand, A. stephanieae shows some features shared by all nudibranchs like the postmetamorphic condensation of the CNS, the possession of rhinophoral ganglia and the lack of oral tentacle ganglia as well as the de novo formation of the adult muscle complex. On the other hand, the structure and arrangement of the serotonergic apical organ is similar to other caenogastropod and opisthobranch gastropods supporting their sister group relationship.
Die schwarz-gelbe Regierung in Nordrhein-Westfalen übernahm 2005 von einer rot-grünen Koalition ein viergliedriges Schulsystem mit Hauptschule, Realschule, Gymnasium und Gesamtschule. Dazu kommt noch als fünfte Schulform die Sonderschule. Die letzte SPD-geführte Landesregierung konnte sich im Wahlkampf 2004, nach PISA 2000 und 2003 ein stärker integratives Schulsystem zwar vorstellen, wollte sich aber für den Umbau Zeit lassen und damit nicht den Wahlkampf belasten, den sie dann doch verlor. Die neue Landesregierung versucht in der laufenden Legislaturperiode dieses überkommene System zu stabilisieren versucht, indem sie das Siechtum der Hauptschule stoppen will und die boomende Nachfrage nach Gesamtschulplätzen so massiv ignoriert, dass in den Schuljahren 2008/09 und 2009/10 jeweils rund 15000 an Gesamtschulen angemeldeten Kinder keinen Platz fanden und so deren Eltern um das Recht der freien Schulwahl gebracht wurden. (Merkelbach 2009, S.4-6) Diese von einer CDU-Ministerin betriebene Politik ging zuletzt auch dem Koalitionspartner FDP zu weit und sie hat zugleich die beiden Oppositionsparteien zu weitgehenden Reformkonzepten herausgefordert. Es wird darum in der heißen Phase des Wahlkampfes heftiger zugehen als vor der letzten Landtagswahl.

Seit ca. 20 Jahren wird das digitale Medium als Träger für literarische Texte verwendet. Als Pioniertat wird häufig der Hyperroman afternoon (1987) von Michael Joyce genannt. Am Anfang wurde ein für heutige Begriffe primitives Programm namens Storyspace verwendet, das nur auf Apple-Computern verwendbar war. Eastgate, ein kalifornischer Verlag, brachte dieses Programm und die ersten Hypertexte auf den Markt, und zwar auf den heute wegen ihrer geringen Speicherkapazität schon fast ausgestorbenen floppy disks, später auf CD-ROMs. Entwickelt wurde Storyspace von Mark Bernstein, dem Chef von Eastgate, zusammen mit J. David Bolter, einem Hypertexttheoretiker, und dem Autor Michael Joyce. Heute kann man einfache Hypertexte mit jeder Textverarbeitung oder mit einem Programm zur Erstellung von Webseiten erzeugen. Besonders nützlich sind allerdings die in Storyspace leicht zu generierenden maps, das sind Übersichten über die einzelnen Textblöcke und ihre Verlinkungen. Diese Baupläne sind auch für die Analyse von Hypertexten äußerst wertvoll. Ganz allgemein hat sich die Technik seit den Anfängen der digitalen Literatur, sowohl was Hardware als auch was Software betrifft, rapide weiter entwickelt und verschiedenste Formen der Nutzung des Mediums ermöglicht. Noch immer werden digitale Texte kommerziell auf CD-ROMs vertrieben, aber die überwiegende Mehrzahl digitaler Literatur kann man online lesen. Bei Eastgate sind momentan ca. 30 Werke lieferbar, was im Vergleich zur online verfügbaren Literatur nur eine verschwindend geringe Zahl ist. Man hat mit Recht die auf CD-ROMs vertriebenen Hypertexte als Laborprodukte bezeichnet, weil sie künstlich abgegrenzt sind. Der wahre Ort für Hypertexte - und selbst ein einziger gigantischer Hypertext - ist natürlich das World Wide Web.
Do you know what I think? asks Adrian Leverkuehn. "Musik ist die Zweideutigkeit als System." Music is Janus-faced by its very nature. It can move and paralyze. "What passion cannot music raise and quell," exclaims John Dryden in his Song for St. Cecilia's Day, 1687. Music is an expert in the use of opiates, asserts Settembrini in The Magic Mountain, and Nietzsche speaks of her dual, intoxicating and befogging, nature. Shakespeare's Desdemona "will sing the savageness out of a bear" (IV, i) and the merchants in Novalis' Heinrich von Ofterdingen tell the story of another Orpheus whose song so charms a sea "monster" that it saves the singer's life and returns his treasure to him. John Dryden's Thimotheus "to his breathing flute and sounding lyre, could swell the soul to rage, or kindle soft desire" (Alexander's Feast, 1697). "Musica Consolatrix" and "Musica Tremenda". She is the "Mysterium tremendum et fascinosum" in Kleist's novella about the power of music. While English late 17th and early 18th century literature offers a particularly rich harvest of poetry celebrating the contradictory qualities, or effects, of music, there is in fact testimony to this at all stages of our tradition.

The musical ending [of Goethe's Novelle] recalls the fascination with "music as metaphor", "the power of music", among recent and contemporary poets from Pope and Dryden and Collins to E.T.A. Hoffmann and Kleist and, of course to Goethe himself. Music saves Faust's life on Easter morning at the end of a dreadful night, and we'll encounter a similar role of music in his Trilogie der Leidenschaft which we'll read in this context.
A single mother and her grown children. A team now. The fathers have come and gone and are barely remembered. These are her children. By contrast, Matthew (27; 56) identifies an anonymous woman as "the mother of Zebedee's children." We'll talk about it, for what it may mean. More important is the fact that this group is headed by a dominant female. Let's see if it makes a difference. Demian, as you'll remember, was the product of matriarchy, as it were, and seemed to be none the worse for it. It wasn't even worth mentioning. Fifty years later, Edgar Wibeau of Plenzdorf's The New Sorrows of Young W. (1972), a modern version of Goethe's bestselling novel Werther written 200 years earlier, and one of the most brilliant pieces of theatre post-Brecht, does find it worth mentioning. He is "sick & tired" of being paraded as living proof that "a single mother can successfully raise a male."
At the beginning of The Judgment, we find Georg Bendemann, who has just finished a letter to his friend in Russia, reliving once more the agonizing decision to write the letter in the first place. The decision had not been easy. Like many of Kafka's characters, Georg Bendemann is obsessed with the idea of analysis, with the painstaking exploration of all sides of a given issue. "What could one write to such a man without hurting him?" had been the question. "On the other hand, by writing only casual gossip or not at all one would doubtless increase the friends isolation" had been the counter-argument. What follows now is an exercise in looking at alternatives that spawn new alternatives that leaves the reader dazzled. Each conclusion is in turn explored to its possible opposite implications, which are in turn qualified, which leads to more questions followed by more partial conclusions plus qualifications thereof. The process could continue ad infinitum, in fact, has gone on for years--we are merely presented with a condensed version of it.

In fact, in an annoyed moment, the diplomat Antonio sees him as a useless and pampered brat and undiplomatically says so, triggering a fateful sequence of events that generate a seemingly unstoppable momentum.

"The sun shines, and Hitler is master of this city. The sun shines, and dozens of my friends are in prison, possibly dead. Thousands of people like Frl. Schroeder are acclimatizing themselves, like an animal which changes its coat for the winter. After all, whatever government is in power, they are doomed to live in this town." These are among the final entries in Christopher Isherwood's Berlin Diaries. Hitler has legally assumed power and Isherwood, who "can't altogether believe that any of this has really happened," will leave the city he has come to love and return to England. The Nazi Movement that began a decade ago in seedy Bavarian beer halls has now conquered its very antithesis, Prussia. It seems unstoppable. The people, as always, will adapt or perish.
The tentative title of my presentation is GENDER AMBIVALENCE (AMBIGUITY?) and deals with Renaissance iconography in Thomas Mann's Death in Venice. It will show that homosexual attraction, although obvious in the story (and Mann's life) is a secondary issue and that the main concern lies with the problem provided by an aesthetic principle. (Parallel narratives, the plot is not the real/only story).
