Objectives: Given the high need and the absence of specific antivirals for treatment of COVID-19 (the disease caused by severe acute respiratory syndrome-associated coronavirus-2 [SARS-CoV-2]), human immunodeficiency virus (HIV) protease inhibitors are being considered as therapeutic alternatives. Methods: Prezcobix/Rezolsta is a fixed-dose combination of 800 mg of the HIV protease inhibitor darunavir (DRV) and 150 mg cobicistat, a CYP3A4 inhibitor, which is indicated in combination with other antiretroviral agents for the treatment of HIV infection. There are currently no definitive data on the safety and efficacy of DRV/cobicistat for the treatment of COVID-19. The in vitro antiviral activity of darunavir against a clinical isolate from a patient infected with SARS-CoV-2 was assessed. Results: DRV showed no antiviral activity against SARS-CoV-2 at clinically relevant concentrations (EC50 > 100 μM). Remdesivir, used as a positive control, demonstrated potent antiviral activity (EC50 = 0.38 μM). Conclusions: Overall, the data do not support the use of DRV for the treatment of COVID-19.
Electrical stimulation (EStim) has been shown to promote bone healing and regeneration both in animal experiments and clinical treatments. Therefore, incorporating EStim into promising new bone tissue engineering (BTE) therapies is a logical next step. The goal of current BTE research is to develop combinations of cells, scaffolds, and chemical and physical stimuli that optimize treatment outcomes. Recent studies demonstrating EStim’s positive osteogenic effects at the cellular and molecular level provide intriguing clues to the underlying mechanisms by which it promotes bone healing. In this review, we discuss results of recent in vitro and in vivo research focused on using EStim to promote bone healing and regeneration and consider possible strategies for its application to improve outcomes in BTE treatments. Technical aspects of exposing cells and tissues to EStim in in vitro and in vivo model systems are also discussed.
The aim of this clinical study was to describe the quality of posterior composite restorations (n = 329) performed on a group of patients (n=219) during an observation period of three years at various intervals (6, 12, 18, 24 and 36 months) after application.
The parameters were assessed both In vivo and In vitro using clinical examinations, impressions and photography according to modified FDI criteria. For the statistical analysis of the results, the Wilcoxon test with a significance level of p = 0.05 was applied.
After three years, In vivo five from the seven parameters exhibited significant changes. Only "retention" and "approximal contact" remained unchanged. In vitro studied parameters "anatomical form", "occlusal contour/wear" and "approximal contact" did not result in any significant changes, however "marginal adaptation", "surface luster" and "overhangs" deteriorated significantly.
In summary, the results of this study show that composite posterior restorations were clinically acceptable in terms of specific parameters. However, unsatisfactory results have arisen in relation to the handling of composites, stated In vivo and In vitro especially in the reconstruction of the marginal adaptation, surface and overfilling.
Introduction: Over the last years, electronic cigarettes (ECs) have become more popular, particularly in individuals who want to give up smoking tobacco. The aim of the present study was to assess the influence of the different e-smoking liquids on the viability and proliferation of human periodontal ligament fibroblasts.
Method and materials: For this study six test solutions with components from ECs were selected: lime-, hazelnut- and menthol-flavored liquids, nicotine, propylene glycol, and PBS as control group. The fibroblasts were incubated up to 96 h with the different liquids, and cell viability was measured by using the PrestoBlue® reagent, the ATP detection and the migration assay. Fluorescence staining was carried out to visualize cell growth and morphology. Data were statistically analyzed by two-tailed one-way ANOVA.
Results: The cell viability assay showed that the proliferation rates of the cells incubated with nicotine or the various flavored liquids of the e-cigarettes were reduced in comparison to the controls, though not all reductions were statistically significant. After an incubation of 96 h with the menthol-flavored liquid the fibroblasts were statistically significant reduced (p < 0.001). Similar results were found for the detection of ATP in fibroblasts; the incubation with menthol-flavored liquids (p < 0.001) led to a statistically significant reduction. The cell visualization tests confirmed these findings.
Conclusion: Within its limits, the present in vitro study demonstrated that menthol additives of e-smoking have a harmful effect on human periodontal ligament fibroblasts. This might indicate that menthol additives should be avoided for e-cigarettes.
Die Lyme-Borreliose, eine durch Zecken übertragene Multisystem-Erkrankung, wird durch die Spirochäte B. burgdorferi s. l. hervorgerufen. Die in vitro-Empfindlichkeit von Borrelien gegenüber einer ganzen Reihe von Substanzen ist bislang nicht oder nur unzureichend bekannt. In der vorliegenden Arbeit wurde daher die in vitro-Empfindlichkeit von B. burgdorferi gegenüber einer Reihe von neu entwickelten Antibiotika im Vergleich zu bekannten Substanzen unter standardisierten Bedingungen untersucht. Im 1. Teil der Arbeit wurde ein neues kolorimetrisches Mikrodilutionstestverfahren in Kombination mit konventioneller Subkultivierung in Flüssigmedium zur Bestimmung der in vitro-Empfindlichkeit von 17 repräsentativ ausgewählten Borrelienstämmen gegenüber dem neu entwickelten Ketolid Cethromycin eingesetzt. Das Verfahren ist standardisiert, gut reproduzierbar und erlaubt es, mehrere Antibiotika bzw. Isolate gleichzeitig effizient zu testen. Die MHK90 für Cethromycin lag für die getesteten Isolate einheitlich bei  0,002 µg/ml. Die MBK90 lag bei 0,75 µg/ml. Im 2. Teil der Arbeit wurden auf der Basis von konventionellen Zellzählungen mittels Dunkelfeldmikroskopie zusätzlich Untersuchungen zur Aktivität von Cethromycin, Sitafloxacin und Gemifloxacin im Vergleich zu Ciprofloxacin und den bekanntermaßen gut gegen Borrelien wirksamen Substanzen Doxycyclin und Ceftriaxon für je ein repräsentatives Isolat der humanpathogenen Genospezies B. burgdorferi s. s., B. garinii und B. afzelii durchgeführt. Von den untersuchten neuen Substanzen zeigten Cethromycin (MHK [Mittelwert]: ≤ 0,002 µg/ml; MBK [Mittelwert]: 0,3 µg/ml) und Gemifloxacin (MHK [Mittelwert]: 0,06 µg/ml; MBK [Mittelwert]: 8 µg/ml) im Vergleich zu den in der Therapie der Lyme-Borreliose bewährten Substanzen Ceftriaxon (MHK [Mittelwert]: 0,016 µg/ml; MBK [Mittelwert]: 1,2 µg/ml) und Doxycyclin (MHK [Mittelwert]: 0,25 µg/ml; MBK: von 24 bis >32 µg/ml) die vielversprechendste Aktivität gegen B. burgdorferi. Diese Ergebnisse konnten in Hinblick auf die Aktivität der getesteten Antibiotika gegenüber Borrelien durch die zusätzlich über 120 h durchgeführten Time-Kill-Experimenten abgesichert werden. Im 3. Teil der Arbeit wurden dann unter Zuhilfenahme eines theoretischen Modells mittels Einbeziehung weiterer pharmakologischer und pharmakokinetischer Daten für alle getesteten Substanzen sogenannte Bakterizidie-Indizes (BIs) auf der Basis der erzielten in vitro-Resultate berechnet. BIs von > 2,8 stehen dabei für eine gute bis sehr gute antimikrobielle Aktivität. Für Cethromycin zeigte sich bei den untersuchten Isolaten ein rascher Anstieg der BIs schon nach einer sehr kurzen Inkubationszeit von 48 h. Dabei lagen die Werte für Cethromycin (BIs 5,0-7,59) nach 120 h sogar über denen von Doxycyclin (BIs 2,81-3,15). Ebenso lagen die errechneten BIs für Gemifloxacin nach 120 h (BIs 3,33-8,05) höher als für Doxycyclin. Ceftriaxon war allerdings die bei weitem am besten wirksame Substanz und zeigte auch die höchsten BIs von 14,96 bis 18,48. Unter Berücksichtigung aller verfügbaren Daten ergab sich aus der vorliegenden Arbeit zusammenfassend folgende Rangordnung für die in vitro-Aktivität der getesteten antibiotischen Substanzen: Ceftriaxon > Cethromycin > Gemifloxacin > Doxycyclin > Sitafloxacin > Ciprofloxacin. Für neu entwickelte Antibiotika stellen in vitro-Untersuchungen einen ersten Schritt für eine Charakterisierung ihrer antimikrobiellen Wirksamkeit gegen Infektionserreger dar. Die Einbeziehung weiterer pharmakologischer Daten in die Interpretation solcher experimentell gewonnenen Ergebnisse ist innovativ, vermittelt zusätzliche Informationen und erlaubt eine vergleichende modellhafte Abschätzung der Aktivität solcher Substanzen unter Berücksichtigung wichtiger pharmakokinetischer und pharmakodynamischer Parameter. Zusammenfassend legen die in der vorliegenden Arbeit herausgearbeiteten Ergebnisse eine vielversprechende Aktivität von zwei neuen Substanzen (Cethromycin und Gemifloxacin) gegen B. burgdorferi nahe und lassen daher eine weitergehende klinische Prüfung dieser Antibiotika für eine mögliche therapeutische Anwendung bei Lyme-Borreliose sinnvoll erscheinen.
Pneumoperitoneum mit Kohlendioxid steht im Verdacht, die Proliferation intraabdomineller Tumorzellen zu beeinflussen. Vorangegangene Versuche haben bereits gezeigt, daß isobares Kohlendioxid die Proliferation von Kolonkarzinomzellen in vitro und in der Ratte stimulieren. In der vorliegenden Arbeit wurden Zellen des Colon-Karzinoms CX-2 und des Pancreas-Karzinoms DAN-G mit Kohlendioxid unter iso- und hyperbaren Bedingungen behandelt. Das Tumorwachstum wurde bis zum 15. Tag nach der Exposition durch DNA-Färbung mit Pico-Green gemessen. An den ersten vier Tagen nach Expositon ist das Tumorwachstum bei beiden Zellinien signifikant vermindert. Ab dem fünften Tag ist das Wachstum von DAN-G bei allen Drücken bis zum Ende des Untersuchungszeitraumes signifikant erhöht. Bei CX-2 hängen Ausmaß und Dauer der Proliferationssteigerung von der Höhe der jeweiligen Drücke ab. Schlußfolgerung: Kohlendioxid beeinflußt das Wachstum humaner Karzinomzellen. Das Ausmaß des Effektes hängt im Wesentlichen von der Höhe des Gasdruckes während der Exposition ab.
