RNA interference (RNAi) has emerged as a powerful tool to induce loss-of-function phenotypes by post-transcriptional silencing of gene expression. In this study we wondered whether inducible RNAi-cassettes integrated into cellular DNA possess the power to trigger neoplastic growth. For this purpose inducible RNAi vectors containing tetracycline (Tet)-responsive derivatives of the H1 promoter for the conditional expression of short hairpin RNA (shRNA) were used to target human polo-like kinase 1 (Plk1), which is overexpressed in a broad spectrum of human tumors. In the absence of doxycycline (Dox) HeLa clones expressing TetR, that carry the RNAi-cassette stably integrated, exhibited no significant alteration in Plk1 expression levels. In contrast, exposure to Dox led to marked downregulation of Plk1 mRNA to 3% and Plk1 protein to 14% in cell culture compared to mismatch shRNA/Plk1-expressing cells. As a result of Plk1 depletion cell proliferation decreased to 17%. Furthermore, for harnessing RNAi for silencing disease-related genes in vivo we transplanted inducible RNAi-HeLa cells onto nude mice. After administration of Dox knockdown of Plk1 expression was observed correlating to a significant inhibition of tumor growth. Taken together, our data revealed that genomically integrated RNAi-elements are suitable to hamper tumor growth by conditional expression of shRNA.
We present a report on the second annual symposium of the Dry Grassland Working Group, which took place 26.-28.08.05 in Münster and was entitled ‘Observation scales in dry grasslands’ The Dry Grassland Working Group is a discussion forum for all who are dealing with dry grasslands in central Europe. It organises annual symposia on various topics and aims to prepare the volumes on dry grasslands and closely related vegetation classes within the the ‘Synopsis of the plant communities of Germany’ The Synopsis will be based on a TURBOVEG database. As its structure and reference lists are now almost fixed, we will probably be able to start with data input at the beginning of 2006.
subkontinentale Art wertet, dargestellt.
Klees in Sachsen-Anhalt.
dargestellt.
We propose to use the hadron number fluctuations in the limited momentum regions to study the evolution of initial flows in high energy nuclear collisions. In this method by a proper preparation of a collision sample the projectile and target initial flows are marked in fluctuations in the number of colliding nucleons. We discuss three limiting cases of the evolution of flows, transparency, mixing and reflection, and present for them quantitative predictions obtained within several models. Finally, we apply the method to the NA49 results on fluctuations of the negatively charged hadron multiplicity in Pb+Pb interactions at 158A GeV and conclude that the data favor a hydrodynamical model with a significant degree of mixing of the initial flows at the early stage of collisions.
A new imaging method that combines high-efficiency fast-neutron detection with sub-ns time resolution is presented. This is achieved by exploiting the high neutron detection efficiency of a thick scintillator and the fast timing capability and flexibility of light-pulse detection with a dedicated image intensifier. The neutron converter is a plastic scintillator slab or, alternatively, a scintillating fibre screen. The scintillator is optically coupled to a pulse counting image intensifier which measures the 2-dimensional position coordinates and the Time-Of-Flight (TOF) of each detected neutron with an intrinsic time resolution of less than 1 ns. Large-area imaging devices with high count rate capability can be obtained by lateral segmentation of the optical readout channels.
In this contribution we present algorithms for model checking of analog circuits enabling the specification of time constraints. Furthermore, a methodology for defining time-based specifications is introduced. An already known method for model checking of integrated analog circuits has been extended to take into account time constraints. The method will be presented using three industrial circuits. The results of model checking will be compared to verification by simulation.
Ludwig Leichhardt had to abandon a large and important collection of botanical specimens during his Expedition from Moreton Bay to Port Essington. Here we attempt to assess the significance of the lost collection by identifying the botanical references in his detailed published journal from the journey. From Leichhardt’s description of the plants and their habitats, and with our accurate knowledge of current distribution, it has been possible, in most cases, to identity his botanical references to a single species. In other cases there is lower degree of certainty. Well over one hundred of the species recorded in Leichhardt’s journal would have been new to science at the time if specimens had survived. The record does identify some potential locations for species that would represent range extensions and suggests an indigenous status for a number of plant species that where previously considered exotic. Certainly Leichhardt was a talented botanist and his significant contribution to Australian natural science should be recognised.
Riboswitches reflect a novel concept in gene regulation that is particularly suited for technological adaptation. Therefore, we characterized thermodynamically the ligand binding properties of a synthetic, tetracycline (tc)-binding RNA aptamer, which regulates gene expression in a dose-dependent manner when inserted into the untranslated region of an mRNA. In vitro, one molecule of tc is bound by one molecule of partially pre-structured and conformationally homogeneous apo-RNA. The dissociation constant of 770 pM, as determined by fluorimetry, is the lowest reported so far for a small molecule-binding RNA aptamer. Additional calorimetric analysis of RNA point mutants and tc derivatives identifies functional groups crucial for the interaction and including their respective enthalpic and entropic contributions we can propose detailed structural and functional roles for certain groups. The conclusions are consistent with mutational analyses in vivo and support the hypothesis that tc-binding reinforces the structure of the RNA aptamer, preventing the scanning ribosome from melting it efficiently.
In November 2005, a survey was begun of the wells in and around Hagia Sophia Church in Istanbul. The long-term goal of the survey is the understanding of the function of the tunnels and the water systems used for Hagia Sophia and its surroundings during the Byzantine and the Ottoman periods. Alternate research methods, such as geophysical research, will be used in future surveys. The 2005 survey examined the channels that run from under the narthex and continue northwards and the southwards of the building as well as channels that run towards the atrium, hippodrome, and garden in the north. The survey resulted in the first photos of the well-bottoms in the history of Hagia Sophia.
Cadmium (Cd) has been in industrial use for a long period of time. Its serious toxicity moved into scientific focus during the middle of the last century. In this review, we discuss historic and recent developments of toxicological and epidemiological questions, including exposition sources, resorption pathways and organ damage processes.
In this article we examine and "exapt" Wurzel's concept of superstable markers in an innovative manner. We develop an extended view of superstability through a critical discussion of Wurzel's original definition and the status of marker-superstability versus allomorphy in Natural Morphology: As we understand it, superstability is - above and beyond a step towards uniformity - mainly a symptom for the weakening of the category affected (cf. 1.,2. and 4.). This view is exemplified in four short case studies on superstability in different grammatical categories of four Germanic languages: genitive case in Mainland Scandinavian and English (3.1), plural formation in Dutch (3.2), second person singular ending -st in German (3.3), and ablaut generalisation in Luxembourgish (3.4).
Proton pumping respiratory complex I (NADH:ubiquinone oxidoreductase) is a major component of the oxidative phosphorylation system in mitochondria and many bacteria. In mammalian cells it provides 40% of the proton motive force needed to make ATP. Defects in this giant and most complicated membrane-bound enzyme cause numerous human disorders. Yet the mechanism of complex I is still elusive. A group exhibiting redox-linked protonation that is associated with iron-sulfur cluster N2 of complex I has been proposed to act as a central component of the proton pumping machinery. Here we show that a histidine in the 49-kDa subunit that resides near iron-sulfur cluster N2 confers this redox-Bohr effect. Mutating this residue to methionine in complex I from Yarrowia lipolytica resulted in a marked shift of the redox midpoint potential of iron-sulfur cluster N2 to the negative and abolished the redox-Bohr effect. However, the mutation did not significantly affect the catalytic activity of complex I and protons were pumped with an unchanged stoichiometry of 4 H+/2e−. This finding has significant implications on the discussion about possible proton pumping mechanism for complex I.
The radial artery as a conduit for coronary artery bypass grafting : review of current knowledge
(2006)
The effect of coronary artery bypass grafting (CABG) lasts as long as the grafts are patent. The internal mammary artery has been considered the "golden" graft due to the superb long-term patency, exceeding 90% at 10 years. The saphenous vein grafts, unfortunately, tend to occlude with a rate of 10-15% within a year after surgery, and eventually, at 10 years after the operation, as much as 60-70% of these vein grafts are either occluded or have angiographic evidence of atherosclerosis. The search for another "arterial conduit", the radial artery, has intensified through the last 15 years in hope to provide a better graft than the saphenous vein for CABG. This article reviews the current knowledge for the radial artery as a conduit in CABG. (Anadolu Kardiyol Derg 2006; 6: 153-62)
A survey covering almost all known sites and most areas of potential habitat of the rare plant Blandfordia cunninghamii (family Blandfordiaceae) in 2004 recorded over 4000 plants from 27 locations, with 80% of the plants in the upper Blue Mountains west of Sydney (lat 33° 40' S, long 150° 20' E), and the remainder as a disjunct occurrence on Mount Kembla in the Illawarra. Habitat requirements of Blandfordia cunninghamii were found to be southern aspect (SE to SW), a slope of > 30°, high rainfall (>1200 mm a year), good drainage, partial canopy cover (30-50%), and acid clayey sands with a pH of 4.5-5, at an altitude between 500 and 950 m. Using International Union for Conservation of Nature (IUCN) parameters, we consider that the number of plants (less than 10 000), their Extent of Occurrence (940 square km), Area of Occupancy (80 km2) are below the threshold for Vulnerable. There is observed decline in habitat and numbers and we conclude that there may be less than 10 locations (under IUCN definitions). This would mean that the species could be considered Vulnerable under IUCN Criteria.
Gene trapping is a method of generating murine embryonic stem (ES) cell lines containing insertional mutations in known and novel genes. A number of international groups have used this approach to create sizeable public cell line repositories available to the scientific community for the generation of mutant mouse strains. The major gene trapping groups worldwide have recently joined together to centralize access to all publicly available gene trap lines by developing a user-oriented Website for the International Gene Trap Consortium (IGTC). This collaboration provides an impressive public informatics resource comprising ~45 000 well-characterized ES cell lines which currently represent ~40% of known mouse genes, all freely available for the creation of knockout mice on a non-collaborative basis. To standardize annotation and provide high confidence data for gene trap lines, a rigorous identification and annotation pipeline has been developed combining genomic localization and transcript alignment of gene trap sequence tags to identify trapped loci. This information is stored in a new bioinformatics database accessible through the IGTC Website interface. The IGTC Website (www.genetrap.org) allows users to browse and search the database for trapped genes, BLAST sequences against gene trap sequence tags, and view trapped genes within biological pathways. In addition, IGTC data have been integrated into major genome browsers and bioinformatics sites to provide users with outside portals for viewing this data. The development of the IGTC Website marks a major advance by providing the research community with the data and tools necessary to effectively use public gene trap resources for the large-scale characterization of mammalian gene function.
Human impacts on the landscape have increased the penalties for Black-tailed Godwits laying their eggs too late, especially in the very intensive agricultural landscapes of The Netherlands. Thus, godwits have experienced a dramatic change of their fitness landscape, because the advance in mowing date made late clutches worthless destroying either eggs or chicks. To determine the driving forces of the recent population decline we study the individual variation in timing of breeding with respect to reproductive success in a population unaffected by mowing. Our results show that even in a low intensity agricultural area it is very important for godwits to breed early in the season.
The identity of Anthribus heros Fabricius (Coleoptera: Anthribidae), and lectotype designation

The structural analysis of the redox complex between the soluble cytochrome c552 and the membrane-integral cytochrome ba3 oxidase of Thermus thermophilus is complicated by the transient nature of this protein-protein interaction. Using NMR-based chemical shift perturbation mapping, however, we identified the contact regions between cytochrome c552 and the CuA domain, the fully functional water-soluble fragment of subunit II of the ba3 oxidase. First we determined the complete backbone resonance assignments of both proteins for each redox state. Subsequently, two-dimensional [15N,1H]TROSY spectra recorded for each redox partner both in free and complexed state indicated those surface residues affected by complex formation between the two proteins. This chemical shift analysis performed for both redox states provided a topological description of the contact surface on each partner molecule. Remarkably, very pronounced indirect effects, which were observed on the back side of the heme cleft only in the reduced state, suggested that alterations of the electron distribution in the porphyrin ring due to formation of the protein-protein complex are apparently sensed even beyond the heme propionate groups. The contact residues of each redox partner, as derived from the chemical shift perturbation mapping, were employed for a protein-protein docking calculation that provided a structure ensemble of 10 closely related conformers representing the complex between cytochrome c552 and the CuA domain. Based on these structures, the electron transfer pathway from the heme of cytochrome c552 to the CuA center of the ba3 oxidase has been predicted.
Distribution of plant size and reproductive success is investigated in the temperate epiphytic orchid Sarcochilus australis (Lindl.) Rchb. f. at Kinglake National Park, Victoria, in south-eastern Australia, and applied to estimating the effective population size. Plant size distribution (leaf number, length of longest leaf and number of flowers) was not normally distributed. Most individuals were vegetative and it is estimated that more than half of all individuals are too small to flower, however exceptionally large individuals even though rare are able to have more than one active inflorescence. Flowering probability is plant size dependent and follows a sigmoid curve. The minimum observed leaf size of a flowering individual was 26 mm, however these small individuals have a low probability of flowering (<30%), while large plants (> 80 mm) have a much higher probability of flowering (90%). The effective population size (Ne) of the Kinglake population of Sarcochilus australis was estimated from the distribution of flower production, and shown to be small (Ne = 10–19%) and comparatively similar to some of the other published estimates of effective populations size in orchids. From this basic survey of size distribution in Sarcochilus australis it is predicted that genetic diversity is low.
This is a splendid formulation of the problem. Hermann’s article should be compulsory reading for students of historical syntax. In search of a solution to this problem, I have applied the following procedure. First I have put together the minor catechisms with those parts of the Enchiridion which translate the same German text. Words which are missing in any of the three versions have been italicized. The result is shown below.
The moss flora of the Los Ruiles Nature Reserve, Maule region (VII), central Chile was investigated. Los Ruiles is a small forest reserve dominated by Nothofagus trees and surrounded by plantations of Pinus radiata. The moss species list contains 72 taxa, among which are 36 species newly reported for the Maule region. Several species reach their northernmost known limit in the reserve, including Achrophyllum magellanicum var. magellanicum, Ancistrodes genuflexa, Cryphaea consimilis, Dendrocryphaea lechleri, Lembophyllum orbiculatum, Leptostomum menziesii, Symblepharis krausei, and Zygodon papillatus. To ensure the survival of these rare or local bryophytes, an increase of the proportion of Nothofagus trees in the forests surrounding the reserve is desirable.
The beetle fauna of Dominica, Lesser Antilles (Insecta: Coleoptera) : diversity and distribution
(2006)
The beetle fauna of the island of Dominica is summarized. It is presently known to contain 269 genera, and 361 species (in 42 families), of which 347 are named at a species level. Of these, 62 species are endemic to the island. The other naturally occurring species number 262, and another 23 species are of such wide distribution that they have probably been accidentally introduced and distributed, at least in part, by human activities. Undoubtedly, the actual numbers of species on Dominica are many times higher than now reported. This highlights the poor level of knowledge of the beetles of Dominica and the Lesser Antilles in general. Of the species known to occur elsewhere, the largest numbers are shared with neighboring Guadeloupe (201), and then with
South America (126), Puerto Rico (113), Cuba (107), and Mexico-Central America (108). The Antillean island chain probably represents the main avenue of natural overwater dispersal via intermediate stepping-stone islands. The distributional patterns of the species shared with Dominica and elsewhere in the Caribbean suggest stages in a dynamic taxon cycle of species origin, range expansion, distribution contraction, and re-speciation.
The mango flower beetle, Protaetia fusca (Herbst), an Asiatic pest of several commercial crops, was first found in Florida in 1985. All subsequent Florida specimens are recorded here, as well as the first documented records from the Bahamas and Barbados. Illustrations are provided and comparisons are made to the similar native Euphoria sepulcralis (Fabricius), which is also recorded for the first time from the Bahamas. Extensive host records and a bibliography (especially tracking P. fusca spread in Hawaii) are included.
Ursachen dafür lassen sich u.a. invorgängigen institutionellen Erfahrungen vermuten.
The first key is completed for the Palaearctic Pristiphora Latereille, 1810 species. Pristiphora araratensis sp. n. is descdbed. Pristiphora kamtchatica Malaise, 1931, Pristiphora mesatlantica Lacourt, 1976 and Pristiphora amelanchieris (Takeuchi, 1922) are new synonyms of Pristiphora insularis Rohwer, 1910.
Deanolis sublimbalis SNELLEN (Lepidoptera: Pyralidae), the red banded mango caterpillar (RMBC), is a Southeast Asian insect species. It is now widely distributed throughout this region (India, Burma, Thailand, China, Brunei, Philippines, Indonesia, and Papua New Guinea) and was recently detected for the first time on mainland Australia, but so far has not been recorded in Pakistan, Nepal, and Malaysia (WATERHOUSE 1998). In Papua New Guinea (PNG) it is nowadays widely distributed throughout the mainland and islands (WATERHOUSE 1998). Although infestation levels of 40 – 50 % were recorded in the Philippines (TIPON 1979), very little is known about the biology of this pest, and there are only few references in the literature, the results differing significantly (WATERHOUSE 1998). Due to the contradictory opinions on the biology published, the following study was undertaken to obtain the basic information required for the development of an appropriate management technique for this pest. In particular, the fundamental data to clarify in the course of this study were: 1. life history and behaviour of RMBC, especially when mango is out of season. 2. search for other host plants of RMBC. 3. search for natural enemies of RMBC.
To evade the host's immune response, herpes simplex virus employs the immediate early gene product ICP47 (IE12) to suppress antigen presentation to cytotoxic T-lymphocytes by inhibition of the ATP-binding cassette transporter associated with antigen processing (TAP). ICP47 is a membrane-associated protein adopting an alpha-helical conformation. Its active domain was mapped to residues 3-34 and shown to encode all functional properties of the full-length protein. The active domain of ICP47 was reconstituted into oriented phospholipid bilayers and studied by proton-decoupled 15N and 2H solid-state NMR spectroscopy. In phospholipid bilayers, the protein adopts a helix-loop-helix structure, where the average tilt angle of the helices relative to the membrane surface is approximately 15 degrees (+/- 7 degrees ). The alignment of both structured domains exhibits a mosaic spread of approximately 10 degrees . A flexible dynamic loop encompassing residues 17 and 18 separates the two helices. Refinement of the experimental data indicates that helix 1 inserts more deeply into the membrane. These novel insights into the structure of ICP47 represent an important step toward a molecular understanding of the immune evasion mechanism of herpes simplex virus and are instrumental for the design of new therapeutics.
In the Solling mountains (Southern Lower Saxony, Germany) the fauna was sampled for one year with stem eclectors in adjacent spruce, mixed (spruce/beech), and beech stands. The tree age was more than 90 years. Four sampling treatments were established: eclectors on spruce stems in the spruce stand (1), on beech stems in the beech stand (2), and on spruce (3) and beech stems in the mixed stand (4). The following harvestmen species, with 1601 individuals in total, were found: Mitopus morio, Oligolophus tridens, Platybunus bucephalus, Leiobunum blackwalli, and Leiobunum rotundum. The number of individuals was highest on spruce stems in the more open spruce stand, mainly due to Mitopus morio, whereas number of species was highest on beech stems in the mixed stand. Both the number of individuals and species were lowest on beech stems in the beech stand. Here, additional information about the phenology of the harvestmen species is given.
