Extraction of network topology from multi-electrode recordings : is there a small-world effect?
(2011)
The simultaneous recording of the activity of many neurons poses challenges for multivariate data analysis. Here, we propose a general scheme of reconstruction of the functional network from spike train recordings. Effective, causal interactions are estimated by fitting generalized linear models on the neural responses, incorporating effects of the neurons’ self-history, of input from other neurons in the recorded network and of modulation by an external stimulus. The coupling terms arising from synaptic input can be transformed by thresholding into a binary connectivity matrix which is directed. Each link between two neurons represents a causal influence from one neuron to the other, given the observation of all other neurons from the population. The resulting graph is analyzed with respect to small-world and scale-free properties using quantitative measures for directed networks. Such graph-theoretic analyses have been performed on many complex dynamic networks, including the connectivity structure between different brain areas. Only few studies have attempted to look at the structure of cortical neural networks on the level of individual neurons. Here, using multi-electrode recordings from the visual system of the awake monkey, we find that cortical networks lack scale-free behavior, but show a small, but significant small-world structure. Assuming a simple distance-dependent probabilistic wiring between neurons, we find that this connectivity structure can account for all of the networks’ observed small-world-ness. Moreover, for multi-electrode recordings the sampling of neurons is not uniform across the population. We show that the small-world-ness obtained by such a localized sub-sampling overestimates the strength of the true small-world structure of the network. This bias is likely to be present in all previous experiments based on multi-electrode recordings.

Rare Earth Elements (REE) have become the new strategic economic weapon for the modern age. Used in the manufacturing of products ranging from mobile phones to jet fighter engines, REEs have become the new “oil” of today in terms of economic and strategic importance. Currently, 95% of REEs mined globally are mined in China, giving China a monopoly on the industry. Deng Xiaoping foresaw the importance of REEs in 1992 when he commented: “as there is oil in the Middle East, there is rare earth in China.” Recently, China temporarily stopped exports of REEs to Japan, the EU and the US as an unofficial response to varying political and economic issues. This stoppage raised concerns as to the dependability of China and REE exports. Using the theory of neo-mercantilism, this paper analyzes China’s actions in the REE market and its subsequent economic and political implications. It concludes with a look at how countries are trying to position themselves away from a dependency on China.
Japan's quest for energy security : risks and opportunities in a changing geopolitical landscape
(2011)
For much of the 20th century, economic growth was fueled by cheap oil-based energy supply. Due to increasing resource constraints, however, the political and strategic importance of oil has become a significant part of energy and foreign policy making in East and Southeast Asian countries. In Japan, the rise of China’s economic and military power is a source of considerable concern. To enhance energy security, the Japanese government has recently amended its energy regulatory framework, which reveals high political awareness of risks resulting from the looming key resources shortage and competition over access. An essential understanding that national energy security is a politically and economically sensitive area with a clear international dimension affecting everyday life is critical in shaping a nation’s energy future.
It has often been asked whether today´s Japan will be able to move into new and promising industries, or whether it is locked into an innovation system with an inherent inability to give birth to new industries. One argument reasons that the thick institutional complementarities among labour, innovation, and finance among its enterprises and the public sector favour industrial development in sectors of intermediate uncertainty, while it is difficult to move into areas of major uncertainty. In this paper, we present the case of the silver industry or, somewhat more prosaically, the 60+ or even 50+ industry, for which most would agree that Japan has indeed become a lead market and lead producer on the global market. For an institutional economist, the case of the silver industry is particularly interesting, because Japan´s success is based on the cooperation of existing actors, the enterprise and public sector in particular, which helped overcome the information uncertainties and asymmetries involved in the new market by relying on several established mechanisms developed well before. In that sense, Japan´s silver industry presents a case of of what we propose to call successful institutional path activation with the effect of an innovative market creation, instead of the problematic lockin effects that are usually associated with the term path dependence.
The emergence of Capitalism is said to always lead to extreme changes in the structure of a society. This view implies that Capitalism is a universal and unique concept that needs an explicit institutional framework and should not discriminate between a German or US Capitalism. In contrast, this work argues that the ‘ideal type’ of Capitalism in a Weberian sense does not exist. It will be demonstrated that Capitalism is not a concept that shapes a uniform institutional framework within every society, constructing a specific economic system. Rather, depending on the institutional environment - family structures in particular - different forms of Capitalism arise. To exemplify this, the networking (Guanxi) Capitalism of contemporary China will be presented, where social institutions known from the past were reinforced for successful development. It will be argued that especially the change, destruction and creation of family and kinship structures are key factors that determined the further development and success of the Chinese economy and the type of Capitalism arising there. In contrast to Weber, it will be argued that Capitalism not necessarily leads to a process of destruction of traditional structures and to large-scale enterprises under rational, bureaucratic management, without leaving space for socio-cultural structures like family businesses. The flexible global production increasingly favours small business production over larger corporations. Small Chinese family firms are able to respond to rapidly changing market conditions and motivate maximum efforts for modest pay. The structure of the Chinese family proved to be very persistent over time and to be able to accommodate diverse economic and political environments while maintaining its core identity. This implies that Chinese Capitalism may be an entirely new economic system, based on Guanxi and the family.
Saccade-related modulations of neuronal excitability support synchrony of visually elicited spikes
(2011)
During natural vision, primates perform frequent saccadic eye movements, allowing only a narrow time window for processing the visual information at each location. Individual neurons may contribute only with a few spikes to the visual processing during each fixation, suggesting precise spike timing as a relevant mechanism for information processing. We recently found in V1 of monkeys freely viewing natural images, that fixation-related spike synchronization occurs at the early phase of the rate response after fixation-onset, suggesting a specific role of the first response spikes in V1. Here, we show that there are strong local field potential (LFP) modulations locked to the onset of saccades, which continue into the successive fixation periods. Visually induced spikes, in particular the first spikes after the onset of a fixation, are locked to a specific epoch of the LFP modulation. We suggest that the modulation of neural excitability, which is reflected by the saccade-related LFP changes, serves as a corollary signal enabling precise timing of spikes in V1 and thereby providing a mechanism for spike synchronization.
In binocular rivalry, presentation of different images to the separate eyes leads to conscious perception alternating between the two possible interpretations every few seconds. During perceptual transitions, a stimulus emerging into dominance can spread in a wave-like manner across the visual field. These traveling waves of rivalry dominance have been successfully related to the cortical magnification properties and functional activity of early visual areas, including the primary visual cortex (V1). Curiously however, these traveling waves undergo a delay when passing from one hemifield to another. In the current study, we used diffusion tensor imaging (DTI) to investigate whether the strength of interhemispheric connections between the left and right visual cortex might be related to the delay of traveling waves across hemifields. We measured the delay in traveling wave times (ΔTWT) in 19 participants and repeated this test 6 weeks later to evaluate the reliability of our behavioral measures. We found large interindividual variability but also good test–retest reliability for individual measures of ΔTWT. Using DTI in connection with fiber tractography, we identified parts of the corpus callosum connecting functionally defined visual areas V1–V3. We found that individual differences in ΔTWT was reliably predicted by the diffusion properties of transcallosal fibers connecting left and right V1, but observed no such effect for neighboring transcallosal visual fibers connecting V2 and V3. Our results demonstrate that the anatomical characteristics of topographically specific transcallosal connections predict the individual delay of interhemispheric traveling waves, providing further evidence that V1 is an important site for neural processes underlying binocular rivalry.
Recently, new soil data maps were developed, which include vertical soil properties like soil type. Implementing those into a multilayer Soil-Vegetation-Atmosphere-Transfer (SVAT) scheme, discontinuities in the water content occur at the interface between dissimilar soils. Therefore, care must be taken in solving the Richards equation for calculating vertical soil water fluxes. We solve a modified form of the mixed (soil water and soil matric potential based) Richards equation by subtracting the equilibrium state of soil matrix potential ψE from the hydraulic potential ψh. The sensitivity of the modified equation is tested under idealized conditions. The paper will show that the modified equation can handle with discontinuities in soil water content at the interface of layered soils.
Autism spectrum disorders (ASD) have been associated with sensory hypersensitivity. A recent study reported visual acuity (VA) in ASD in the region reported for birds of prey. The validity of the results was subsequently doubted. This study examined VA in 34 individuals with ASD, 16 with schizophrenia (SCH), and 26 typically developing (TYP). Participants with ASD did not show higher VA than those with SCH and TYP. There were no substantial correlations of VA with clinical severity in ASD or SCH. This study could not confirm the eagle-eyed acuity hypothesis of ASD, or find evidence for a connection of VA and clinical phenotypes. Research needs to further address the origins and circumstances associated with altered sensory or perceptual processing in ASD.
A hygroscopicity tandem differential mobility analyzer (HTDMA) was used to measure the water uptake (hygroscopicity) of secondary organic aerosol (SOA) formed during the chemical and photochemical oxidation of several organic precursors in a smog chamber. Electron ionization mass spectra of the non-refractory submicron aerosol were simultaneously determined with an aerosol mass spectrometer (AMS), and correlations between the two different signals were investigated. SOA hygroscopicity was found to strongly correlate with the relative abundance of the ion signal m/z 44 expressed as a fraction of total organic signal (f44). m/z 44 is due mostly to the ion fragment CO2+ for all types of SOA systems studied, and has been previously shown to strongly correlate with organic O/C for ambient and chamber OA. The analysis was also performed on ambient OA from two field experiments at the remote site Jungfraujoch, and the megacity Mexico City, where similar results were found. A simple empirical linear relation between the hygroscopicity of OA at subsaturated RH, as given by the hygroscopic growth factor (GF) or "ϰorg" parameter, and f44 was determined and is given by ϰorg = 2.2 × f44 − 0.13. This approximation can be further verified and refined as the database for AMS and HTDMA measurements is constantly being expanded around the world. The use of this approximation could introduce an important simplification in the parameterization of hygroscopicity of OA in atmospheric models, since f44 is correlated with the photochemical age of an air mass.
During the measurement campaign FROST 2 (FReezing Of duST 2), the Leipzig Aerosol Cloud Interaction Simulator (LACIS) was used to investigate the influence of various surface modifications on the ice nucleating ability of Arizona Test Dust (ATD) particles in the immersion freezing mode. The dust particles were exposed to sulfuric acid vapor, to water vapor with and without the addition of ammonia gas, and heat using a thermodenuder operating at 250 °C. Size selected, quasi monodisperse particles with a mobility diameter of 300 nm were fed into LACIS and droplets grew on these particles such that each droplet contained a single particle. Temperature dependent frozen fractions of these droplets were determined in a temperature range between −40 °C ≤T≤−28 °C. The pure ATD particles nucleated ice over a broad temperature range with their freezing behavior being separated into two freezing branches characterized through different slopes in the frozen fraction vs. temperature curves. Coating the ATD particles with sulfuric acid resulted in the particles' IN potential significantly decreasing in the first freezing branch (T>−35 °C) and a slight increase in the second branch (T≤−35 °C). The addition of water vapor after the sulfuric acid coating caused the disappearance of the first freezing branch and a strong reduction of the IN ability in the second freezing branch. The presence of ammonia gas during water vapor exposure had a negligible effect on the particles' IN ability compared to the effect of water vapor. Heating in the thermodenuder led to a decreased IN ability of the sulfuric acid coated particles for both branches but the additional heat did not or only slightly change the IN ability of the pure ATD and the water vapor exposed sulfuric acid coated particles. In other words, the combination of both sulfuric acid and water vapor being present is a main cause for the ice active surface features of the ATD particles being destroyed. A possible explanation could be the chemical transformation of ice active metal silicates to metal sulfates. The strongly enhanced reaction between sulfuric acid and dust in the presence of water vapor and the resulting significant reductions in IN potential are of importance for atmospheric ice cloud formation. Our findings suggest that the IN concentration can decrease by up to one order of magnitude for the conditions investigated.

Explosive volcanism affects weather and climate. Primary volcanic ash particles which act as ice nuclei (IN) can modify the phase and properties of cold tropospheric clouds. During the Eyjafjallajökull volcanic eruption we have measured the highest ice nucleus number concentrations (>600 L) in our record of 2 years of daily IN measurements in central Germany. Even in Israel, located about 5000 km away from Iceland, IN were as high as otherwise only during desert dust storms. These measurements are the only ones available on the properties of IN in the Eyjafjallajökull plume. The measured high concentrations and high activation temperature (−8 °C) point to an important impact of volcanic ash on microphysical and radiative properties of clouds through enhanced glaciation.
This paper compares measurements of gaseous and particulate emissions from a wide range of biomass-burning plumes intercepted by the NASA DC-8 research aircraft during the three phases of the ARCTAS-2008 experiment: ARCTAS-A, based out of Fairbanks, Alaska, USA (3 April to 19 April 2008); ARCTAS-B based out of Cold Lake, Alberta, Canada (29 June to 13 July 2008); and ARCTAS-CARB, based out of Palmdale, California, USA (18 June to 24 June 2008). Approximately 500 smoke plumes from biomass burning emissions that varied in age from minutes to days were segregated by fire source region and urban emission influences. The normalized excess mixing ratios (NEMR) of gaseous (carbon dioxide, acetonitrile, hydrogen cyanide, toluene, benzene, methane, oxides of nitrogen and ozone) and fine aerosol particulate components (nitrate, sulfate, ammonium, chloride, organic aerosols and water soluble organic carbon) of these plumes were compared. A detailed statistical analysis of the different plume categories for different gaseous and aerosol species is presented in this paper.
The comparison of NEMR values showed that CH4 concentrations were higher in air-masses that were influenced by urban emissions. Fresh biomass burning plumes mixed with urban emissions showed a higher degree of oxidative processing in comparison with fresh biomass burning only plumes. This was evident in higher concentrations of inorganic aerosol components such as sulfate, nitrate and ammonium, but not reflected in the organic components. Lower NOx NEMRs combined with high sulfate, nitrate and ammonium NEMRs in aerosols of plumes subject to long-range transport, when comparing all plume categories, provided evidence of advanced processing of these plumes.
This paper compares measurements of gaseous and particulate emissions from a wide range of biomass-burning plumes intercepted by the NASA DC-8 research aircraft during the three phases of the ARCTAS-2008 experiment: ARCTAS-A, based out of Fairbanks, Alaska USA (3 April to 19 April 2008); ARCTAS-B based out of Cold Lake, Alberta, Canada (29 June to 13 July 2008); and ARCTAS-CARB, based out of Palmdale, California, USA (18 June to 24 June 2008). Extensive investigations of boreal fire plume evolution were undertaken during ARCTAS-B, where four distinct fire plumes that were intercepted by the aircraft over a range of down-wind distances (0.1 to 16 hr transport times) were studied in detail. Based on these analyses, there was no evidence for ozone production and a box model simulation of the data confirmed that net ozone production was slow (on average 1 ppbv h−1 in the first 3 h and much lower afterwards) due to limited NOx. Peroxyacetyl nitrate concentrations (PAN) increased with plume age and the box model estimated an average production rate of ~80 pptv h−1 in the first 3 h. Like ozone, there was also no evidence for net secondary inorganic or organic aerosol formation. There was no apparent increase in aerosol mass concentrations in the boreal fire plumes due to secondary organic aerosol (SOA) formation; however, there were indications of chemical processing of the organic aerosols. In addition to the detailed studies of boreal fire plume evolution, about 500 smoke plumes intercepted by the NASA DC-8 aircraft were segregated by fire source region. The normalized excess mixing ratios (i.e. ΔX/ΔCO) of gaseous (carbon dioxide, acetonitrile, hydrogen cyanide, toluene, benzene, methane, oxides of nitrogen (NOx), ozone, PAN) and fine aerosol particulate components (nitrate, sulfate, ammonium, chloride, organic aerosols and water soluble organic carbon) of these plumes were compared.
Although it has been suggested that temperature increase may alter the toxic potential of environmental pollutants, few studies have investigated the potential risk of chemical stressors for wildlife under Global Climate Change (GCC) impact. We applied a bifactorial multigeneration study in order to test if GCC conditions alter the effects of low pesticide concentrations on life history and genetic diversity of the aquatic model organism Chironomus riparius. Experimental populations of the species were chronically exposed to a low concentration of the fungicide pyrimethanil (half of the no-observed-adverse-effect concentration: NOAEC/2) under two dynamic present-day temperature simulations (11.0–22.7°C; 14.0–25.2°C) and one future scenario (16.5–28.1°C). During the 140-day multigeneration study, survival, emergence, reproduction, population growth, and genetic diversity of C. riparius were analyzed. Our results reveal that high temperature and pyrimethanil act synergistically on the midge C. riparius. In simulated present-day scenarios, a NOAEC/2 of pyrimethanil as derived from a life-cycle toxicity test provoked only slight-to-moderate beneficial or adverse effects on C. riparius. In contrast, exposure to a NOAEC/2 concentration of pyrimethanil at a thermal situation likely for a summer under GCC conditions uncovered adverse effects on mortality and population growth rate. In addition, genetic diversity was considerably reduced by pyrimethanil in the future scenario, but only slightly under current climatic conditions. Our multigeneration study under near-natural (climatic) conditions indicates that not only the impact of climate change, but also low concentrations of pesticides may pose a reasonable risk for aquatic insects in future.
Previously undiagnosed anaemia is common in elective orthopaedic surgical patients and is associated with increased likelihood of blood transfusion and increased perioperative morbidity and mortality. A standardized approach for the detection, evaluation, and management of anaemia in this setting has been identified as an unmet medical need. A multidisciplinary panel of physicians was convened by the Network for Advancement of Transfusion Alternatives (NATA) with the aim of developing practice guidelines for the detection, evaluation, and management of preoperative anaemia in elective orthopaedic surgery. A systematic literature review and critical evaluation of the evidence was performed, and recommendations were formulated according to the method proposed by the Grades of Recommendation Assessment, Development and Evaluation (GRADE) Working Group. We recommend that elective orthopaedic surgical patients have a haemoglobin (Hb) level determination 28 days before the scheduled surgical procedure if possible (Grade 1C). We suggest that the patient's target Hb before elective surgery be within the normal range, according to the World Health Organization criteria (Grade 2C). We recommend further laboratory testing to evaluate anaemia for nutritional deficiencies, chronic renal insufficiency, and/or chronic inflammatory disease (Grade 1C). We recommend that nutritional deficiencies be treated (Grade 1C). We suggest that erythropoiesis-stimulating agents be used for anaemic patients in whom nutritional deficiencies have been ruled out, corrected, or both (Grade 2A). Anaemia should be viewed as a serious and treatable medical condition, rather than simply an abnormal laboratory value. Implementation of anaemia management in the elective orthopaedic surgery setting will improve patient outcomes.
Spatial and temporal structure of the spider community in the clay semi-desert of western Kazakhstan
(2011)
The spatial and temporal structure of spider communities was studied in the clay semi-desert of the north-western Caspian Lowland, western Kazakhstan (49°23' N, 46°47' E). The soils and vegetation are complex, being composed of a mosaic of desert and steppe plant communities. Besides the native associations, there are plantations of different tree species. The ground-dwelling spider assemblages in the native habitats are the most diverse. The number of species inhabiting forest plantations is three times as small. Gnaphosidae is the leading family in the ground layer. They show high abundance and diversity levels during the whole season. Thomisidae, Lycosidae, Philodromidae, and Salticidae are abundant as well. The species diversity of herbage-dwelling spiders in different open native habitats is very similar. The spectrum of dominant families (Thomisidae, Oxyopidae, Araneidae, and Salticidae) and the seasonal dynamics of their ratio in desert and steppe associations have much in common. Spider assemblages of native and artificial habitats are characterised by change from multispecies polydominant spring-summer communities to impoverished imbalanced autumn ones. Seasonal changes in the species structure of mature spider groupings in native habitats are well pronounced, while the impact of seasonal conditions is even stronger than between-habitat differences. Complexes of typical species with different levels of habitat preference are revealed.
The protected Mediterranean woodland habitats in Israel are undergoing tree encroachment, resulting in loss of open patches with herbaceous vegetation. We suggested that this process results in a ground spider community dominated by shade-loving species. At three Mediterranean woodland sites located along a rainfall gradient, we examined the effects on the ground-spider community of experimental removal of the woody vegetation in 1000 m2 plots by cutting and overall plant biomass reduction by grazing and browsing by livestock. Pitfall traps were placed in replicated plots of four treatments (control, cutting, grazing/browsing, and cutting together with grazing/browsing) and in two different habitat patch types (open, woody). ANOVA and multivariate analyses were performed on family abundance by treatment and habitat patch type. Tree-cutting reduced the number of families in plots at two of the three sites. Grazing did not have a significant effect on the number of families or on the ground spider community composition. The spider community of cut-woody patches was more similar to that of open patches than to that of uncut woody patches. Most spider families separated along an axis of open versus woody patches, with woody habitat families predominating at all sites. Families typical of open habitats were positively associated with cut-woody patches as well. The overall effect on ground spider diversity of such manipulations may depend on the scale of habitat changes.
Agrobiont spider species are well adapted to arable systems, which have fairly uniform vegetation structure and pest assemblages over continent-wide areas. We wanted to study, whether agrobiont spider subassemblages and the life history of the most prominent agrobiont, Pardosa agrestis, show any regional variation within Hungary, where only modest climatic differences exist between the NW and SE parts of the country. We studied agrobiont species of spider assemblages in 27 alfalfa and 21 cereal fields with suction sampling and pitfalls. The similarity structure of these agrobiont sub-assemblages (Sørensen distance measure) was congruent with the geographic distance matrices (Eucledian distance), as tested by Mantel tests. However, if we considered sub-assemblages consisting of the non-agrobiont species, this congruency was always higher. Thus, agrobionts responded only moderately to geographical variation if we compare them to non-agrobiont species. We studied the generation numbers and the occurrence of the first adult individuals in P. agrestis; the most common agrobiont spider in Hungary. This comparison involved comparing fields along a NW – SE gradient during 6 sampling years in pairwise comparisons, where in each year a northern and a southern population was compared with a minimum distance of 126 km in between. In generation numbers there was no difference; we found two generations across Hungary. In contrast, the first occurrence of adult individuals was on average 15 days earlier in both generations in the more southern populations. Thus, it can be concluded that agrobionts show a fairly stable and relatively low magnitude response over country-sized geographical ranges.
The first fossil cyphophthalmid harvestman (Opiliones: Cyphophthalmi) from Palaeogene (Eocene) Baltic amber is described. This is only the third fossil example of this basal harvestman lineage; the others being from the probably slightly younger Bitterfeld amber and the much older, early Cretaceous, Myanmar (Burmese) amber. Although incomplete and lacking most of the appendages, the new Baltic amber fossil can be identified as a female. The somatic characters preserved, especially spiracle morphology and the coxo-genital region, allow it to be assigned with some confidence to the extant genus Siro Latreille, 1796 (Sironidae). This fossil is formally described here as Siro balticus sp. nov. It resembles modern North American Siro species more than modern European ones, and can be distinguished principally on its relatively large size and the outline form of the body.
Spiders are powerful predators, but the threats confronting them are numerous. A survey is presented of the many different arthropods which waylay spiders in various ways. Some food-specialists among spiders feed exclusively on spiders. Kleptoparasites are found among spiders as well as among Mecoptera, Diptera, Lepidoptera, and Heteroptera. Predators are found within spiders’ own population (cannibalism), among other spider species (araneophagy), and among different species of Heteroptera, Odonata, and Hymenoptera. Parasitoids are found in the orders Hymenoptera and Diptera. The largest insect order, Coleoptera, comprises a few species among the Carabidae which feed on spiders, but beetles are not represented among the kleptoparasites or parasitoids.
A comprehensive overview of plant, fungus and animal species of Austria revealed a total of 748 endemic and subendemic species, including, 11 harvestman and 46 spider species. Altogether two endemic harvestmen (Nemastoma bidentatum relictum, Nemastoma schuelleri) and 8 endemic spiders (Abacoproeces molestus, Collinsia (caliginosa) nemenziana, Mughiphantes severus, Mughiphantes styriacus, Pelecopsis alpica, Scotophaeus nanus, Troglohyphantes novicordis, Troglohyphantes tauriscus), beside 9 subendemic harvestman and 38 subendemic spider species have been recorded from Austria. Hot-spots of endemism in the Eastern Alps are the north-eastern (Ennstaler Alps) and southern Calcareous Alps (Karawanken, Karnische Alps) and the Central Alps (Hohe Tauern, Gurktaler Alps, Ötztaler and Stubaier Alps). Most of the endemic arachnid species occur from the nival down to the montane zone. Important habitats are rocky areas, caves and woodlands. High absolute numbers and percentages of endemics can be found within the harvestman families Cladonychiidae, Ischyropsalididae and Nemastomatidae and in the spider genera Lepthyphantes s. l. and Troglohyphantes. The conservation status of these highly endangered taxa – 85 % of the spider species and 100 % of the harvestman taxa are endangered in Austria – is poor.
Spiders were studied around the Pechenganikel smelter combine, Kola Peninsula, north-western Russia. The average spider density was 6-fold greater and the density of Linyphiidae specimens 11.5-fold higher at slightly polluted sites, compared with heavily polluted sites. Altogether, 18 species from 10 families were found at heavily polluted sites, the theridiid Robertus scoticus clearly dominating (23.3 % of identifiable specimens), also Neon reticulatus (9.6 %), Thanatus formicinus (9.6 %) and Xysticus audax (8.2 %) were abundant. The most numerous among 58 species found at slightly polluted sites were Tapinocyba pallens (18.5 %), Robertus scoticus (13.7 %), Maso sundevalli (9.5 %) and Alopecosa aculeata (8.2 %). The family Linyphiidae dominated at slightly polluted sites, 64 % of species and 60 % of individuals; compared with heavily polluted sites, 23 % and 38 % respectively.
In 2005 a team of Bulgarian zoologists started a project aiming to study the invertebrates inhabiting the deeper soil stratum (euedaphon) and the Superficial Underground Compartment (SUC) in Bulgaria. In the course of a four-year sampling, a total of 52 species of spiders were caught from 19 collecting sites and 9 geographical regions. They belong to the following families: Scytodidae (1), Segestriidae (1), Dysderidae (8), Nesticidae (1), Anapidae (1), Theridiidae (1), Linyphiidae (20), Agelenidae (3), Cybaeidae (1), Dictynidae (2), Amaurobiidae (2), Liocranidae (3), Corinnidae (1), Zodariidae (1), Gnaphosidae (5), and Salticidae (1). The family Anapidae, with the species Zangherella relicta (Kratochvíl, 1935) is recorded from three sites in the Pirin and Slavyanka mountains, and this represents the first record of the family, genus and species in Bulgaria. In spite of the active investigations of the epigean and cave spiders in these regions over the years Z. relicta was not found and it seems it occurs only in deeper subterranean habitats and nowhere else. Comparative study of almost topotypic specimens of Z. relicta from Montenegro with those collected from Bulgaria showed no variation in the shape of palp and female vulvae. Until the true identity of Z. apuliae (Caporiacco, 1949) from Italy is revealed, it remains unclear whether Z. relicta and Z. apuliae are conspecific, as it remains unclear whether the older records of Z. apuliae from the Balkan Peninsula refer to this species or to Z. relicta. Pelecopsis mengei (Simon, 1884) (Linyphiidae) and Scotolathys simplex Simon, 1884 (Dictynidae) are also reported from Bulgaria for the first time, the latter being also new to FYR of Macedonia. A faunistic overview of the spiders found in these underground environments is made, along with remarks on the distribution and ecology of some rare and interesting species. The presence of cave-dwelling and superficial spiders in the sampled sites indicates that SUC and euedaphon are inhabited by different ecotypes, e.g. litter-(tanathostromic), soil-(edaphic) and cave-(troglobitic) which at some places co-occur.
Until today, from Greek caves a total of 109 species of spiders belonging to 25 families are known. One species, the linyphiid Porrhomma convexum (Westring, 1861) was recorded here for the first time in Greece. The 109 species are distributed in caves of different geographic territories as follows: Thrace – 8 species, Macedonia – 18, Epirus – 1, Thessaly – 6, Central Greece – 3, Attiki-Saronic Islands – 24, Peloponnese – 15, Evoia-Vories Sporades – 1, Eastern Aegean Islands – 5, Cyclades – 3, Dodecanese – 6, Ionian Islands – 23, Crete – 47. The largest fraction of troglobite species were encountered mainly in the territories of Crete – 15 species (5 of which are anophthalmic), the Ionian Islands – 4, Thrace – 2 (both anophthalmic), the Attiki-Saronic Islands – 2 (both anophthalmic), the Peloponnese – 2 (one anophthalmic), and Macedonia, Thessaly, and the Cyclades – each with 2 species. The richness of the troglobitic spidersin these regions strengthens the assumption that they were major centres of speciation and evolution for the species of this group. According to their current distribution, the established 109 species can be classified into 12 zoogeograpical categories, grouped into 4 complexes (widely distributed, European, Mediterranean, endemics). The largest number of species belong to the endemic complex (53.2 %) and are also the most characteristic and reflect the local character of the cave-dwelling spiders.
In this paper, three new spider species are described from the Greek Aegean island Lesbos: Tegenaria maelfaiti sp. nov. (Agelenidae), Amaurobius lesbius sp. nov. (Amaurobiidae) and Agroeca parva sp. nov. (Liocranidae), as well as the unknown male of Arabelia pheidoleicomes Bosselaers, 2009 (Corinnidae). Diagnostic features and notes on ecology and distribution of these species are given. Two new records for the island are provided, such that currently 300 spider species are known from Lesbos.
The spider fauna active on the bark of trees in forests on eight sites in different regions in Germany was investigated. Trunk eclectors at about 2-4 meters height on living trees were used in different regions of Germany (SW Bavaria, Hesse, Brandenburg) between 1990 and 2003. In Hesse eclectors were also used on dead beech trees (standing and lying). In this study data, mainly from beech (Fagus sylvatica) and spruce (Picea abies), from May to October are compared – whole year samples (including winter) are only available from Hesse. A total of 334 spider species were recorded with these bark traps, i.e. about one third of the spider species known from Germany. On average, each of the eight regions yielded 140.5 (± 26.2) species, each single tree 40.5 (± 12.2) species and 502 (±452) adult spiders per season (i.e. May to Oct.). The 20 most abundant species are listed and characterised in detail. Six of the 20 species were not known to be abundant on bark, three prefer conifers and three beech/broadleaf. Even in winter (December-March) there was a remarkably high activity on the trunks. However, only a few species occur exclusively or mainly in winter. Finally, the rarity of some bark spider species is discussed and details (all known records in Germany, phenology) of four of them are presented (Clubiona leucaspis, Gongylidiellum edentatum, Kratochviliella bicapitata, Oreonetides quadridentatus). The diversity and importance of the spider fauna on bark in Central Europe is still underestimated.
Die Gemeine Labyrinthspinne : Agelena labyrinthica (Araneae: Agelenidae) ; Spinne des Jahres 2011
(2011)
Notes on the biology of the unidentified invasive harvestman Leiobunum sp. (Arachnida: Opiliones)
(2011)
Since about the year 2000 an unidentified, introduced harvestman of the genus Leiobunum has been rapidly invading Europe. The published records are from the Netherlands, Germany, Austria and Switzerland. A population of Leiobunum sp. in the Netherlands was studied frequently during the day and night. Its life cycle, hunting strategy, diet and accompanying harvestman species were recorded, and mating, male-male fights and ovipositing behaviour studied, as well as the spider species preying on this Leiobunum species. Food items were collected, indicating that its food consists of a wide range of live as well as dead invertebrates including spent spider prey scavenged for at ground level. Vegetable matter like berries, as well as bird droppings were also consumed. The mating strategy is very complex. A male guards an egg depositing female and he defends her against other advancing males, resulting in male-male fights. The guarding male frequently mates. Also courtship behaviour has been observed, including nuptial feeding with a fluid, probably originating from the accessory penal glands and delivered by the male into the female’s stomotheca via sacs located on the distal part of the penis truncus. Eggs are deposited in holes and crevices of walls.
The present paper provides the first checklist of the spiders from coastal heathland on the Baltic Sea island of Hiddensee, Germany. A total of 171 species could be recovered by pitfall trapping in 2008 and 2009. The species inventory comprises several typical dune and heathland species. Ten species (Altella lucida, Centromerus capucinus, Dictyna latens, Drassodes cupreus, Hypsocephalus pusillus, Hypsosinga sanguinea, Micaria lenzi, Micrargus apertus, Philodromus histrio, Walckenaeria capito) are new to the arachnofauna of Mecklenburg-Western Pomerania.
Chthonius (Chthonius) hungaricus Mahnert, 1980 and Larca lata (Hansen, 1884) were recorded for the first time from Slovakia. The finding of C. hungaricus in Slovakia is the second known record since its description and the finding of L. lata is the first record of the family of Larcidae in Slovakia. The descriptions of the species offer an update on the variability of morphologic and morphometric characters. Indications about the habitats of C. hungaricus are also given for the first time.
The pseudoscorpions Chthonius (Chthonius) carinthiacus Beier, 1951 and Chthonius (Ephippiochthonius) tuberculatus Hadži, 1937, are recorded for the first time from Slovakia. An illustrated description of these species is provided based on their morphological and morphometric characters. The descriptions of the species offer an update on the variability of their morphological and morphometric characters.
The distribution of the poorly known Central Asian wolf-spider Oculicosa supermirabilis Zyuzin, 1993 is clarified, discussed and mapped on the basis of both original and literature-derived data. The species is currently known from the Turan Lowland between the 41st and 43rd degrees of latitude north; its distribution coincides with that of the grey-brown desert soil and lies within the geobotanical sub-zone of southern deserts. Both sexes are also illustrated and diagnosed.
Dendrochernes cyrneus (Arachnida: Pseudoscorpiones: Chernetidae) in Brandenburg. The pseudoscorpion Dendrochernes cyrneus (L. Koch, 1873) is recorded for the first time in Brandenburg (Germany). The specimen was found in the ancient park of Sanssouci under the bark of a branch of a beech tree. The suitability of D. cyrneus as an indicator species for old-growth woodlands is discussed.
This paper presents new data, characteristic features, standard body measurements and illustrations of the rare European linyphiid spider Glyphesis taoplesius Wunderlich, 1969; which is recorded here for the first time in Slovakia. The species was found with high abundance in pitfall traps exposed in a floodplain forest near a water reservoir in the lowland Podunajská rovina.
Trogulus banaticus Avram, 1971 is characterised and recorded as new for Slovenia. This species was previously mistaken for T. coriziformis C. L. Koch, 1839 and T. graecus Dahl, 1903 which were later rejected from the Slovenian fauna. T. banaticus is compared with the similar, and partly sympatric, T. tingiformis C. L. Koch, 1847 with which it has often been confused. A table of distinguishing characters for both species is provided, and the ecology of T. banaticus and its general distribution are discussed.
A female specimen of the cribellate spider species Badumna longinqua (L. Koch, 1867) was found in a 'do-it-yourself-store' in Berlin. The species is of Australian origin and has been introduced into New Zealand, Japan, Uruguay and California. This is the first record of a representative of the family Desidae from Europe. B. longinqua lives in and around houses and is apparently capable of establishing itself in Europe.
Nephronectin regulates atrioventricular canal differentiation via Bmp4-Has2 signaling in zebrafish
(2011)
The extracellular matrix is crucial for organogenesis. It is a complex and dynamic component that regulates cell behavior by modulating the activity, bioavailability and presentation of growth factors to cell surface receptors. Here, we determined the role of the extracellular matrix protein Nephronectin (Npnt) in heart development using the zebrafish model system. The vertebrate heart is formed as a linear tube in which myocardium and endocardium are separated by a layer of extracellular matrix termed the cardiac jelly. During heart development, the cardiac jelly swells at the atrioventricular (AV) canal, which precedes valve formation. Here, we show that Npnt expression correlates with this process. Morpholino-mediated knockdown of Npnt prevents proper valve leaflet formation and trabeculation and results in greater than 85% lethality at 7 days post-fertilization. The earliest observed phenotype is an extended tube-like structure at the AV boundary. In addition, the expression of myocardial genes involved in cardiac valve formation (cspg2, fibulin 1, tbx2b, bmp4) is expanded and endocardial cells along the extended tube-like structure exhibit characteristics of AV cells (has2, notch1b and Alcam expression, cuboidal cell shape). Inhibition of has2 in npnt morphants rescues the endocardial, but not the myocardial, expansion. By contrast, reduction of BMP signaling in npnt morphants reduces the ectopic expression of myocardial and endocardial AV markers. Taken together, our results identify Npnt as a novel upstream regulator of Bmp4-Has2 signaling that plays a crucial role in AV canal differentiation.
The genome sequence of Haloferax volcanii is available and several comparative genomic in silico studies were performed that yielded novel insight for example into protein export, RNA modifications, small non-coding RNAs, and ubiquitin-like Small Archaeal Modifier Proteins. The full range of functional genomic methods has been established and results from transcriptomic, proteomic and metabolomic studies are discussed. Notably, Hfx. volcanii is together with Halobacterium salinarum the only prokaryotic species for which a translatome analysis has been performed. The results revealed that the fraction of translationally-regulated genes in haloarchaea is as high as in eukaryotes. A highly efficient genetic system has been established that enables the application of libraries as well as the parallel generation of genomic deletion mutants. Facile mutant generation is complemented by the possibility to culture Hfx. volcanii in microtiter plates, allowing the phenotyping of mutant collections. Genetic approaches are currently used to study diverse biological questions–from replication to posttranslational modification—and selected results are discussed. Taken together, the wealth of functional genomic and genetic tools make Hfx. volcanii a bona fide archaeal model species, which has enabled the generation of important results in recent years and will most likely generate further breakthroughs in the future.
We analyze the implications of the governance structure in academic faculties for their recruitment decisions when competing for new researchers. The value to individual members through social interaction within the faculty depends on the average status of their fellow members. In recruitment decisions, incumbent members trade off the effect of entry on average faculty status against alternative uses of the recruitment budget if no entry takes place. We show that the best candidates join the best faculties but that they receive lower wages than some lesser ranking candidates. We also study the allocation of surplus created by the entry of a new faculty member and show that faculties with symmetric status distributions maximize their joint surplus under majority voting.
Riboswitch RNAs fold into complex tertiary structures upon binding to their cognate ligand. Ligand recognition is accomplished by key residues in the binding pocket. In addition, it often crucially depends on the stability of peripheral structural elements. The ligand-bound complex of the guanine-sensing riboswitch from Bacillus subtilis, for example, is stabilized by extensive interactions between apical loop regions of the aptamer domain. Previously, we have shown that destabilization of this tertiary loop–loop interaction abrogates ligand binding of the G37A/C61U-mutant aptamer domain (Gswloop) in the absence of Mg2+. However, if Mg2+ is available, ligand-binding capability is restored by a population shift of the ground-state RNA ensemble toward RNA conformations with pre-formed loop–loop interactions. Here, we characterize the striking influence of long-range tertiary structure on RNA folding kinetics and on ligand-bound complex structure, both by X-ray crystallography and time-resolved NMR. The X-ray structure of the ligand-bound complex reveals that the global architecture is almost identical to the wild-type aptamer domain. The population of ligand-binding competent conformations in the ground-state ensemble of Gswloop is tunable through variation of the Mg2+ concentration. We quantitatively describe the influence of distinct Mg2+ concentrations on ligand-induced folding trajectories both by equilibrium and time-resolved NMR spectroscopy at single-residue resolution.
We study electron transport through a single-molecule magnet (SMM) and the interplay of its anisotropic spin with quantized vibrational distortions of the molecule. Based on numerical renormalization group calculations we show that, despite the longitudinal anisotropy barrier and small transverse anisotropy, vibrational fluctuations can induce quantum spin-tunneling (QST) and a QST-Kondo effect. The interplay of spin scattering, QST and molecular vibrations can strongly enhance the Kondo effect and induce an anomalous magnetic field dependence of vibrational Kondo side-bands.
The IrIII atom of the title compound, [Ir(C11H8N)2Cl(CH3CN)], displays a distorted octahedral coordination. The pyridyl groups are in trans positions [N—Ir—N = 173.07 (10)°], while the phenyl groups are trans with respect to the acetonitrile and chloride groups [C—Ir—N = 178.13 (11) and C—Ir—Cl = 176.22 (9)°]. The pyridylphenyl groups only show a small deviation from planarity, with the dihedral angle between the planes of the two six-membered rings in each pyridylphenyl group being 5.6 (2) and 5.8 (1)°. The crystal packing shows intermolecular C—H[cdots, three dots, centered]Cl, C—H[cdots, three dots, centered]π(acetonitrile) and C—H[cdots, three dots, centered]π(pyridylphenyl) contacts.
We compute the phase and the modulus of an energy- and pressure-free, composite, adjoint, and
inert field φ in an SU(2) Yang-Mills theory at large temperatures. This field is physically relevant in describing part of the ground-state structure and the quasiparticle masses of excitations. The field φ possesses nontrivial S1-winding on the group manifold S3. Even at asymptotically high temperatures, where the theory reaches its Stefan-Boltzmann limit, the field φ, though strongly power suppressed, is conceptually relevant: its presence resolves the infrared problem of thermal perturbation theory.
Protein translocation across the chloroplast membrane is mediated by molecular machinery composed of protein complexes termed the TOC/TIC (the outer/inner envelope chloroplasts translocases). This translocation process is regulated by metabolic energy in form of GTP and ATP and is influenced by the lipid composition of the membrane. The ability to study the function of a single complex “TOC” in vitro using purified protein or purified chloroplast outer envelope vesicles has been instrumental for our understanding of the mechanism underlying this process.
Indeed, the TOC complex has been purified by previously established procedures. However its functional and structural analyses are impaired by the limited yield of purified protein. Therefore, protocols for native TOC complex purification are described here. The complex isolation is achieved by direct biochemical treatment of biological membrane hosting this complex or by tandem affinity purification of modified protein complex components from generated transgenic plants.
Furthermore, in this thesis, radioactive based in vitro import assays are described, namely those that allow monitoring translocation activity across the outer envelope of chloroplast. Based on the analysis of knock-out plants and isolated complexes it was previously suggested that lipid dependence of protein translocation might exist. Thus, the question was raised whether the lipid composition of the membrane has a direct influence on the behavior and functionality of the TOC translocon, or whether additional components of the chloroplast membrane account for the observed effect in vivo. To answer this question, a technique for vesicle fusion was developed. The principal aim was to explore the effect of an exchange of the lipid environment surrounding the complex translocon. This method helped to demonstrate that the SQDG and PI act stimulatory on the translocation across the outer envelope of chloroplast, whereas DGDG exhibits an inhibitory effect on TOC complex functionality.
Sponges are one of the major components of benthic communities and are considered to be a
key role organism in marine ecosystems. In addition to their importance in terms of
biodiversity, sponges are becoming increasingly attractive to the industry, as they themselves
or associated symbionts, produce various kinds of secondary metabolites of pharmaceutical
properties. Some of them have already been clinically applied.
The taxonomic characters of Porifera are limited to only a few morphological and
histological characters. In addition, sponges of the same species often show a wide
morphological variability, whereas the latter depends on different ecological parameters such
as water depth and current conditions. Thus, the taxonomic classification of sponges often
becomes a scientific challenge.
The fauna of the Yellow Sea rates among the least studied worldwide. At the same time,
according to the UN Atlas of the Ocean, the Yellow Sea is one of the most intensively
exploited marine areas in the world. This is not least due to the dense human population living
in the entire catchment area of the Yellow Sea region. In order to compile medium- and longterm
conclusions about the anthropogenic impact on biota of the Yellow Sea, the knowledge
of species and their distribution is of crucial importance, as these data form the baseline for all
future conservation efforts.
Until now the sponge fauna of the Chinese Yellow Sea is insufficiently investigated.
Thus, there is only one publication on sponges from this region that has been released
hitherto. This paper is dealing with only a view species. However, there is no reference
concerning the present location of the voucher material, on which this publication is based on.
Consequently, no scientific collection on Porifera from the Chinese part of the Yellow Sea
exists to date.
In order to compile a documentation of the recent sponge community of the Chinese
Yellow Sea, 12 study sites along the coast of the Liaoning Peninsula, China, Northeast
Yellow Sea, were investigated with focus on sponge distribution. The corresponding habitats
were characterized in regard to their topographical features, abiotic parameters, and common
composition of benthic megafaunal and macroalgal assemblages.
Due to the lack of comparable studies, a comprehensive literature research on sponges of the
shallow Northwest Pacific Ocean was required. As a result the first compilation of
publications is presented, dealing with sponges from shallow depths of the northwestern
Pacific Ocean.
Abstract
2
In the course of this study, 31 sponge species in total were recorded, which are scientifically
processed. With the exception of four all specimens were determined to species- level.
Twelve out of the total number of species are new to science and are described and classified
according to the recent taxonomic system of the phylum Porifera.
The results of this study indicate considerable differences in species composition between
investigated sites. It is shown that physical factors (particularly current regime, sedimentation,
seasonally related variations in temperatures), as well the availability of suitable substrates are
directly related to the diversity and abundance of investigated sponge communities. In this
context possible adaptation strategies of the corresponding sponges were discussed in detail.
Two sponge species, Clathria (Clathria) asodes and Antho (Acarnia) lithophoenix, formerly
known exclusively from the northeastern Pacific Ocean, are now recorded from the Northwest
Pacific Ocean for the first time. Furthermore, Penares hongdoensis, Clathria (Clathria)
hongdoensis and Celtodoryx girardae were synonymized with Penares cortius, Clathria
(Clathria) acanthostyli, and Celtodoryx ciocalyptoides respectively. Moreover, the occurrence
of eight sponge species, which were known from previous records from the Yellow Sea, could
be confirmed.
As a result of this study the Asian origin of a sponge species that is invasive to the French and
Dutch coasts of the Northeast Atlantic Ocean since the 1990s could be established. Moreover,
it is demonstrated that Celtodoryx girardae from the northeastern Atlantic is in fact
conspecific with Cornulum ciocalyptoides described by Burton (1935) from the Posiet Bay,
Sea of Japan. Apart from taxonomic remarks, variations between populations from both
oceans are examined and discussed thoroughly in regard to possible ecological implications.
The community of documented sponges shows overlapping with the one from the Sea of
Japan. According to the results it is assumed that the endemic degree of the sponges from the
Chinese Yellow Sea is rather low to moderate.
The material obtained in the course of this study was integrated in the collection of the
Senckenbergischen Naturforschenden Sammlungen. Therefore, it is the first scientific
collection of sponges from the Chinese Yellow Sea that can be consulted as a basis for all
further studies on sponges of this region.
The present study is the only investigation of sponges from Dalian and adjacent waters before
the spill occurred in the Dalian harbour in July 2010. Therefore, it provides an essential
baseline needed to assess the impact of the oil spill on benthic communities.
gesundheitspolitischen Herausforderungen unserer Zeit.
Wissenschaftssprache. Veröffentlichungen erfolgen bevorzugt in den weltweit
124
führenden virologischen, tropen- oder präventivmedizinischen sowie infektiologischen
und entomologischen Fachzeitschriften, gelegentlich auch in renommierten
medizinischen Periodika ohne Spezialisierung, z.B. Lancet.
koennen. Die Daten zeigen auch, dass SVA-vermittelte 5-Tansduktionsereignisse
alternatives RNA-Spleissen an putativen Spleissstellen involvieren.
SVA-Elemente fuer diesen Prozess hoechst relevant.

A series of novel mono-1,2,3-triazole and bis-1,2,3-triazole acyclonucleoside analogues of 9-(4-hydroxybutyl)guanine was prepared via copper(I)-catalyzed 1,3-dipolar cycloaddition of N-9 propargylpurine, N-1-propargylpyrimidines/as-triazine with the azido-pseudo-sugar 4-azidobutylacetate under solvent-free microwave conditions, followed by treatment with K2CO3/MeOH, or NH3/MeOH. All compounds studied in this work were screened for their antiviral activities [against human rhinovirus (HRV) and hepatitis C virus (HCV)] and antibacterial activities against a series of Gram positive and negative bacteria.
PaCATB : a secreted catalase protecting Podospora anserina against exogenous oxidative stress
(2011)
A differential mass spectrometry analysis of secreted proteins from juvenile and senescentPodospora anserina cultures revealed age-related differences in protein profiles. Among other proteins with decreased abundance in the secretome of senescent cultures a catalase, termed PaCATB, was identified. Genetic modulation of the abundance of PaCATB identified differential effects on the phenotype of the corresponding strains. Deletion of PaCatB resulted in decreased resistance, over-expression in increased resistance against hydrogen peroxide. While the lifespan of the genetically modified strains was found to be unaffected under standard growth conditions, increased exogenous hydrogen peroxide stress in the growth medium markedly reduced the lifespan of the PaCatB deletion strain but extended the lifespan of PaCatB over-expressors. Overall our data identify a component of the secretome of P. anserina as a new effective factor to cope with environmental stress, stress that under natural conditions is constantly applied on organisms and influences aging processes.
Arrangement of electron transport chain components in bovine mitochondrial supercomplex I1III2IV1
(2011)
The respiratory chain in the inner mitochondrial membrane contains three large multi-enzyme complexes that together establish the proton gradient for ATP synthesis, and assemble into a supercomplex. A 19-Å 3D map of the 1.7-MDa amphipol-solubilized supercomplex I1III2IV1 from bovine heart obtained by single-particle electron cryo-microscopy reveals an amphipol belt replacing the membrane lipid bilayer. A precise fit of the X-ray structures of complex I, the complex III dimer, and monomeric complex IV indicates distances of 13 nm between the ubiquinol-binding sites of complexes I and III, and of 10–11 nm between the cytochrome c binding sites of complexes III and IV. The arrangement of respiratory chain complexes suggests two possible pathways for efficient electron transfer through the supercomplex, of which the shorter branch through the complex III monomer proximal to complex I may be preferred.
Background: Hydrolysates of plant biomass used for the production of lignocellulosic biofuels typically contain sugar mixtures consisting mainly of D-glucose and D-xylose, and minor amounts of L-arabinose. The yeast Saccharomyces cerevisiae is the preferred microorganism for the fermentative production of ethanol but is not able to ferment pentose sugars. Although D-xylose and L-arabinose fermenting S. cerevisiae strains have been constructed recently, pentose uptake is still a limiting step in mixed sugar fermentations.
Results: Here we described the cloning and characterization of two sugar transporters, AraT from the yeast Scheffersomyces stipitis and Stp2 from the plant Arabidopsis thaliana, which mediate the uptake of L-arabinose but not of D-glucose into S. cerevisiae cells. A yeast strain lacking all of its endogenous hexose transporter genes and expressing a bacterial L-arabinose utilization pathway could no longer take up and grow with L-arabinose as the only carbon source. Expression of the heterologous transporters supported uptake and utilization of L-arabinose especially at low L-arabinose concentrations but did not, or only very weakly, support D-glucose uptake and utilization. In contrast, the S. cerevisiae D-galactose transporter, Gal2, mediated uptake of both L-arabinose and D-glucose, especially at high concentrations.
Conclusions: Using a newly developed screening system we have identified two heterologous sugar transporters from a yeast and a plant which can support uptake and utilization of L-arabinose in L-arabinose fermenting S. cerevisiae cells, especially at low L-arabinose concentrations.
Small bowel endoscopy is crucial for diagnosing small bowel Crohn’s disease, and capsule endoscopy is complemented by balloon-assisted enteroscopy to take biopsies and by magnetic resonance imaging to visualize enteral and extra-intestinal involvement. Recently, imaging has also become a key instrument to manage Crohn’s disease patients. Treatment control is advised for patients who have undergone bowel resections and is increasingly used to testify treatment success in non-operated patients, too. In this review we present the modern imaging methods to diagnose and to manage Crohn’s disease with a special focus on the small bowel. Moreover, current knowledge on the impact of diagnostic methods on the patients’ outcome is reported.
NOSTRIN belongs to the recently defined F-BAR protein family. F-BAR proteins are
multi-domain proteins, which serve as adaptors between plasma membrane and
cytoskeleton components in processes such as membrane protrusion formation,
endocytosis and migration. NOSTRIN encompasses a F-BAR domain at the N-terminus,
which mediates membrane association, followed by a HR1 motif and an intermediate
domain (ID) domain in the middle, and a SH3 domain at the C-terminus. The domain
architecture and ability to form oligomers enable NOSTRIN to coordinate several
interaction partners namely dynamin, caveolin, N-WASP and endothelial nitric oxide
synthase (eNOS) in the process of eNOS trafficking. In this context NOSTRIN was
originally identified and hence termed eNOS traffick inducer. NOSTRIN is expressed in
vascularized tissues (e.g. liver and lung) and in primary endothelial cells.
Aims of the present work were (1) to investigate if NOSTRIN is involved in other
processes besides eNOS trafficking, (2) to analyse the function of NOSTRIN in vivo
through knockdown of NOSTRIN in developing zebrafish and (3) to study the
consequences of the loss of NOSTRIN on signal transduction in a primary cell culture
model derived from NOSTRIN knockout mice.
To study the possible involvement of NOSTRIN in other processes besides eNOS
trafficking a yeast two-hybrid screen was performed in which fibroblast growth factor
receptor 1 (FGFR1) was identified as a putative novel interaction partner of NOSTRIN. In
a series of yeast two-hybrid, pulldown and co-immunoprecipitation experiments the
interaction between NOSTRIN and FGFR1 was confirmed to occur between
endogenously expressed proteins and determined to be direct and to depend on the ID
domain of NOSTRIN and the 130 C-terminal amino acid residues of FGFR1. FGFR1 is
activated by binding of fibroblast growth factors (FGFs) and induces several different
signal transduction pathways (e.g. MAPK and Akt pathway). Overexpression of
NOSTRIN in HeLa cells specifically enhanced FGF2-dependent MAPK activation.
Accordingly, depletion of NOSTRIN attenuated FGF2-dependent MAPK activation and
did not affect FGF2-induced Akt activation.
In summary, NOSTRIN has been identified as a novel interaction partner of FGFR1
involved in FGF2-dependent signal transduction.
The morpholino oligonucleotide-mediated knockdown of NOSTRIN in developing
zebrafish caused vascular leakage and irregular vascular patterning e.g. a loss of the
proper trajectory of intersegmental vessel and interruptions of the dorsal longitudinal
anastomotic vessel. The vascular phenotype was consistent upon use of two different
morpholinos and could be rescued in a dose dependent manner by the injection of
zebrafish NOSTRIN mRNA. Detailed analysis involving confocal and time lapse
microscopy in zebrafish with endothelial specific expression of EGFP revealed that the
knockdown of NOSTRIN impacts in vivo on the migration and morphology of endothelial
tip cells and leads to a reduction of filopodia number and length.
Additionally a NOSTRIN knockout mouse was generated. The analysis of FGFR1 signal
transduction in primary mouse lung endothelial cells (MLECs) from NOSTRIN knockout
and wild type mice revealed that FGF2-dependent MAPK activation was attenuated in
MLECs isolated from NOSTRIN knockout mice when compared to MLECs isolated from
wild type mice. The effect of NOSTRIN on FGF2-dependent signal transduction seems to
be specific, since VEGF-induced MAPK activation was not affected in NOSTRIN
knockout MLECs. The importance of NOSTRIN for FGF2 signal transduction in vivo is
demonstrated by the greatly impaired angiogenic response to FGF2 in NOSTRIN
knockout mice in matrigel plug assay. In a detailed biochemical analysis it was
discovered that NOSTRIN interacts with the activated small GTPase Rac1 and that
overexpression of NOSTRIN enhances Rac1 activation. Furthermore, the interactions of
NOSTRIN with both Rac1 and its GEF Sos1 are required for NOSTRIN-mediated
activation of Rac1. In accordance, activation of Rac1 was not detected upon FGF2
stimulation in NOSTRIN knockout MLECs.
In conclusion, the present work describes a novel function of the F-BAR protein
NOSTRIN in FGFR1 signal transduction. Data presented in this work demonstrate that
NOSTRIN is required for the assembly of a complex consisting of FGFR1, Sos1 and
Rac1 and subsequently for the FGF2-dependent activation of Rac1 in endothelial cells.
