Wir berichten im folgenden über histologische Befunde und physikalische Messungen, die zeigen, daß unter besonderen Beschallungsbedingungen in verschiedenen Säugetiergeweben Vorgänge ablaufen, die im Sinne einer Pseudokavitation gedeutet werden müssen. Die Einzelheiten der Untersuchungen sind teils in der Habilitationsschrift von O. Hug, Frankfurt a. M. 1953, teils in der Dissertation von R.Pape, Frankfurt a. M. 1953, niedergelegt.
Die Inaktivierung durch Röntgenstrahlen der an Lebermitochondrien gebundenen Bernsteinsäure-Oxydase wurde untersucht. Ihre Halbwertsdosis beträgt 3,5 · 106 r. Bernsteinsäure-Oxydase, die an Hepatommitochondrien gebunden ist, ist empfindlicher als die normaler Mitochondrien. Die Bernsteinsäure-Oxydase an kleinsten Partikeln zeigt dagegen in beiden Fällen eine größere Strahlenresistenz.
Die in der vorliegenden Arbeit beschriebenen Messungen von Dielektrizitätskonstanten und Leitfähigkeiten biologischer Substanzen im Bereich von 9 bis 180 cm Wellenlänge werden nach einem Resonanzverfahren durchgeführt. Dies ermöglicht trotz der starken Absorption der untersuchten Materialien eine genügende Meßgenauigkeit und einen relativ einfachen Aufbau der Meßanordnung. Die beschriebene Methode zur Auswertung der Messung ist so vereinfacht, daß sie auch von angelerntem Hilfspersonal leicht durchgeführt werden kann. Für den Wellenlängenbereich von 40 bis 180 cm wird eine Lecher-Leitung in Paralleldraht-Aus-führung benutzt, während sie für den Bereich von 9 bis 40 cm Wellenlänge konzentrisch aufgebaut ist. Die Meßfehler werden eingehend diskutiert und in Kurven anschaulich dargestellt. Anschließend werden die Ergebnisse der Messungen von DK und Leitfähigkeit an Blut, Leber, Muskel und Fettgewebe mitgeteilt. Bei allen Substanzen konnte unterhalb etwa 30 cm Wellenlänge eine Abnahme der DK und vor allem eine eindeutige Zunahme der Leitfähigkeit festgestellt werden. Diese Dispersion wird den polaren Molekülen in den Substanzen zugeschrieben und dürfte zumindest bei Blut und Leber im wesentlichen durch die Dispersion des Wassers verursacht werden.
Der Einfluß von Elektrodenpolarisation auf die Bestimmung der elektrischen Konstanten leitfähiger Substanzen wird untersucht; es wird gezeigt, daß der durch Polarisation bedingte Effekt auf die Kapazität um mehrere Zehnerpotenzen größer ist als der auf die Wirkwiderstandskomponente. Die Bestimmung der Dielektrizitätskonstante stark leitfähiger Materialien bei Niederfrequenz wird dadurch sehr erschwert. Die verschiedenen Möglichkeiten, Polarisationseinflüsse herabzusetzen, werden diskutiert und es wird gezeigt, daß einzig eine Messung mit verschiedenem Elektrodenabstand einwandfreie Ergebnisse gewährleistet, wenn Polarisation merklich auftritt. Eine bei biologischen Arbeiten öfter angewandte Methodik geht von der Voraussetzung aus, daß beim Austausch biologischen Materials gegen eine Salzlösung gleicher Beschaffenheit, wie sie intrazellular im biologischen Material vorliegt und in Kontakt mit den Elektroden steht, die Polarisationsimpedanz keiner Änderung unterliegt. Die Analyse eigener experimenteller Untersuchungen, über die berichtet wird, zeigt, daß diese Annahme nur berechtigt ist, wenn das biologische Material in so hinreichend großem Abstand von den Elektroden angeordnet wird, daß keine Schattenwirkung auftreten kann. In allen anderen Fällen ist sie falsch, und Arbeiten, die dem nicht Rechnung tragen, sind kritisch zu bewerten.
Es wurde der Einfluß von Röntgenbestrahlungen auf die Lebensfähigkeit und das Formbildungs-Vermögen kernloser Zellteile von Acetabularia mediterranea in Abhängigkeit von der Strahlendosis untersucht. Dabei erwies sich die Hutbildung stets als der am strahlenempfindlichste Prozeß. Die gefundenen Ergebnisse werden diskutiert und mit den entsprechenden Befunden nach UV-Bestrahlung verglichen.
Es wurde die Einwirkung monochromatischer ultravioletter Strahlen der Wellenlänge 254 mμ, 281 mμ und 297 mμ auf kernlose Zellteile von Acetabularia untersucht. Dazu wurden in Abhängigkeit von der Strahlendosis die mittlere Lebensdauer und das Formbildungs-Vermögen der kernlosen Teile bestimmt. Der 254-mμ-Strahlung kam dabei stets die größte biologische Wirksamkeit zu, während die 297-mμ-Strahlung im untersuchten Dosisbereich fast ohne Wirkung blieb. Die gefundene Wellenlängen-Abhängigkeit weist darauf hin, daß der UV-Absorption durch die Purine und Pyrimidine für den Wirkungsmechanismus der beobachteten UV-Schädigungen kernloser Zellteile besondere Bedeutung zukommen muß.
Es wird versucht, die Treffertheorie auf die indirekte Strahlenwirkung auszudehnen. Dazu wird angenommen, daß durch die Strahlung „Energieträger“ erzeugt werden, die durch Diffusion zu den „empfindlichen Bereichen“ gelangen und diese verändern können.
Der Berechnung des Wirkvolumens für derartige „indirekte Treffer“ folgt eine reaktionskinetische Betrachtung der indirekten Wirkung.
Durch die Einschaltung physikalisch-chemischer Prozesse zwischen Strahlenabsorption und „Treffer“ erscheint eine Berücksichtigung der physikalischen und chemischen Gegebenheiten im bestrahlten Objekt viel eher möglich als in der „klassischen“ Theorie der „direkten Trefferwirkung“.
Die Feldstärke- und Wärmequellenverteilung im ebenen Körper aus Muskel, Fett und Haut bei Anstrahlung wird für den Wellenbereich von 1 m bis 1,27 cm jeweils bei verschiedenen Fettschichtdicken berechnet und graphisch dargestellt. Die mit der Strahlungsfeldmethode erreichbare Tiefendosis-Leistung im Zusammenhang mit der Hautdosisleistung sowie die Frage einer geeigneten Anpassung zur Erreichung einer gleichmäßigen Tiefendosis-Leistung werden diskutiert.
Unter der Voraussetzung, daß ein Strahler benutzt wird, der ein annähernd ebenes Wellenfeld liefert und in dem bestrahlten Körpergebiet ebene, parallele Schichten Haut, Fett, Muskel senkrecht getroffen werden, ergibt sich aus der Betrachtung der berechneten Feldstärke- und Wärmequellenverteilungen für die verschiedenen Wellenlängen etwa folgendes Bild:
Das Problem einer Fettüberlastung tritt bei dm-Bestrahlung bis herab zu 10 cm Wellenlänge nicht auf. Man erhält im Gegensatz zu UKW-Kondensatorfeld-Durchflutung eine starke Wärmeentwicklung in der Haut sowie in den oberen Schichten von Muskel oder inneren Organen. Der Abfall der Dosisleistung im Muskel nach der Tiefe zu wird ab λ = 30 cm mit kürzer werdender Wellenlänge zunehmend steiler. Es kann bei dm-Bestrahlung gegenüber UKW-C-Feld eine um das Mehrfache höhere Dosisleistung an der inneren Oberfläche des Körpers auch bei großen Dicken der Fettschicht erreicht werden, wenn gegebenenfalls durch zusätzliche äußere Maßnahmen (Kühlung) eine Überlastung der Haut bei bestimmten Dicken der Fettschicht (λF/4) vermieden wird.
Bei dm-Bestrahlung paralleler Schichten mit angepaßtem Sender lassen sich weitere qualitative Aussagen machen: Bei konstanter Intensität des Strahlers erfolgt bei längeren Wellen (1 m) mit wachsender Dicke der Fettschicht nur ein monotoner Abfall der Tiefendosis-Leistung auf etwa 60%. Im unteren dm-Bereich (30 cm, 10 cm) ergibt sich bei konstanter Intensität bei Vergrößerung der Fettschichtdicke von 0 bis λF/4 ein Abfall der Tiefendosis-Leistung auf etwa ⅓ und bei weiterer Zunahme der Fettschichtdicke von λF/4 bis λF/2 wieder eine Zunahme der Tiefendosis-Leistung im Verhältnis 1:2. Diese Zahlen gelten größenordnungsmäßig bei Anstrahlung aus einem Anpassungsmedium ε-Fett. Bei Anstrahlung aus Luft ist die absolute Tiefendosis-Leistung geringer und sind die Schwankungen größer. Diese Schwankungen der Tiefendosis-Leistung bei konstanter Intensität des Senders einer Wellenlänge können durch äußere Maßnahmen weitgehend verringert werden.
Wenn der Sender bei verschiedenen Fettschichtdicken jeweils auf gleiche Hautdosisleistung eingestellt wird (Dosierung nach der Hautempfindung), nimmt die Tiefendosis-Leistung bei Vergrößerung der Fettschichtdicke von 0 bis λF/4 stark ab auf etwa ⅙, die Tiefendosis-Leistungen bei weiterer Vergrößerung der Fettschichtdicke von λF/4 bis λF/2 verhalten sich dann etwa wie 1 : 3. Diese Schwankungen sind unabhängig von der Wahl des Anpassungsmediums. Eine Tiefendosierung nach der Hautempfindung ohne genauere Berücksichtigung der Fettschichtdicke wird daher bei Vorliegen von zu den Wellenflächen parallelen Schichten unzuverlässig sein. Es ergibt sich jedoch die Möglichkeit einer instrumentellen, brauchbaren Dosierung durch Einstellung der Intensität des Senders ohne Rücksichtnahme auf die Hautempfindung, wenn man die Schwankungen der Tiefendosis-Leistung mit der Fettschichtdicke bei konstanter Intensität durch die erwähnten Anpassungsmaßnahmen reduziert.
Die kurzen Wellenlängen von 3 cm und 1 cm sind für eine Wärmetherapie in der Tiefe wegen der hohen Absorptionsverluste in Haut und Fett kaum brauchbar, können jedoch zu einer Wärmetherapie der Haut und damit indirekten Beeinflussung innerer Erkrankungen oder zur Erwärmung oberflächiger bzw. weniger leitender Schichten herangezogen werden. Die Oberflächendosis-Leistung steigt bei derselben Intensität mit abnehmender Wellenlänge stark an.
Bezüglich der Angaben über die Dosisleistungs-Verteilung ist zu berücksichtigen, daß das Interferenz-Feld vor der Strahleröffnung bei der Rechnung vernachlässigt wurde. Ferner entspricht die Dosisleistungs-Verteilung nur dann der Temperaturverteilung während der Bestrahlung, wenn die Unterschiede in den Anfangstemperaturen und in den spezifischen Wärmen der biologischen Schichten außer Acht gelassen werden sowie intensiv und kurzzeitig bestrahlt wird, so daß noch kein merklicher Wärmetransport während der Bestrahlung stattfindet. Über die Temperaturverteilung bei Berücksichtigung des Wärmetransports, der bei schwacher, langdauernder Bestrahlung eine merkliche Erwärmung auch tieferer innerer Schichten, eine höhere Erwärmung des Fetts und eine geringere der Haut zur Folge haben kann, sind weitere Betrachtungen erforderlich.
Es wird eine einfache und billig herzustellende Resonanzanordnung beschrieben, mit der genaue Bestimmungen elektrischer Impedanzwerte und Materialkonstanten im Dezimeterwellenbereich durchführbar sind. Prinzip der Methode, Meßbereich, Genauigkeit und Einzelheiten des Aufbaues werden angegeben und an einigen Beispielen erläutert.
Die Primärwirkung von Röntgenstrahlen einer Dosis von etwa 0,5 bis 150 Millionen r auf die kristallisierte Trockensubstanz von Aminosäuren und Peptiden wurde mit Hilfe chemischer, biochemischer und physikalisch-chemischer Arbeitsmethoden untersucht. Es wurde gefunden, daß in allen prinzipiell möglichen Fällen folgende Reaktionen stets wiederkehren : Aminbildung infolge Decarboxylierung; Bildung einer α-Imino- bzw. α-Ketocarbonsäure infolge einer Dehydrierung in α-,β-Stellung; Bildung von β,γ- bzw. γ-δ-ungesättigten α-Aminocarbonsäuren oder deren γ- bzw. δ-Lactonen; Bruch und Vernetzung der aliphatischen Kohlenstoffketten. Bei Peptiden treten die gleichen Reaktionen wie bei den Aminosäuren auf, jedoch in einem anderen Verhältnis; hinzu kommt die strahlenchemische Dehydrierung einer Peptidbindung an der Aminogruppe zu einer energiereichen Iminoacyl-Bindung, welche bei Gegenwart von Wasser sofort hydrolysiert wird. Endprodukt namentlich bei längerkettigen Peptiden : Zwei Bruchstücke (daneben NH3); das eine mit alter amino-endständiger und neuer carboxyl-endständiger Aminosäure und das andere mit der alten carboxy-endständigen Aminosäure, statt Aminogruppe am anderen Ende jetzt Ketogruppe. — In fast allen Fällen wurden die Ionenausbeuten auch quantitativ bestimmt. Die lonenausbeuten für die Bildung von α-Ketosäuren aus α-Aminosäuren fallen exponentiell mit der eingestrahlten Dosis. Eine relativ einfache Funktion erklärt diese Verhältnisse. Die Ionenausbeute für die Bildung von Brenztraubensäure aus Serin ist dagegen unabhängig von der Dosis.
Very-long-baseline interferometry (VLBI) observations of active galactic nuclei at millimetre wavelengths have the power to reveal the launching and initial collimation region of extragalactic radio jets, down to 10–100 gravitational radii (rg ≡ GM/c2) scales in nearby sources. Centaurus A is the closest radio-loud source to Earth. It bridges the gap in mass and accretion rate between the supermassive black holes (SMBHs) in Messier 87 and our Galactic Centre. A large southern declination of −43° has, however, prevented VLBI imaging of Centaurus A below a wavelength of 1 cm thus far. Here we show the millimetre VLBI image of the source, which we obtained with the Event Horizon Telescope at 228 GHz. Compared with previous observations, we image the jet of Centaurus A at a tenfold higher frequency and sixteen times sharper resolution and thereby probe sub-lightday structures. We reveal a highly collimated, asymmetrically edge-brightened jet as well as the fainter counterjet. We find that the source structure of Centaurus A resembles the jet in Messier 87 on ~500 rg scales remarkably well. Furthermore, we identify the location of Centaurus A’s SMBH with respect to its resolved jet core at a wavelength of 1.3 mm and conclude that the source’s event horizon shadow should be visible at terahertz frequencies. This location further supports the universal scale invariance of black holes over a wide range of masses.
In der vorliegenden Arbeit wird versucht, die sog. selektiven Effekte der Ultrakurzwellentherapie (UKWT) thermisch zu erklären, und zwar durch die Temperaturschwingungen, welche durch die pulsierende Wärmebildung der Hochfrequenz(HF)-Bestrahlung in Mikroorganismen hervorgerufen werden. Bei Annahme verschiedener Mechanismen der Energieumsetzung in Wärme innerhalb oder an der Oberfläche von Teilchen, die sich in einem Suspensionsmedium befinden, wird eine Art thermische Resonanz (Selektivität) aufgefunden. Die Lage der Selektivität, abhängig von der Frequenz des HF-Feldes, der Teilchengröße und der Wärmeleitfähigkeit des Teilcheninneren und das Verhältnis der Temperaturamplitude des Teilchenmittelpunkts zur Umgebungstemperatur lassen die Vermutung zu, in Übereinstimmung mit experimentellen Befunden bei der UKWT, daß der Effekt zur Erklärung der Wirksamkeit dieser medizinischen Behandlungsmethode beiträgt. Zurückgreifend auf ältere Arbeiten wird der Anlaufvorgang des Temperaturfeldes in Mikroorganismen, der durch das Einschalten des HF-Feldes entsteht, untersucht, durch einen Modellversuch gestützt und seine Bedeutung für die UKWT, die vielfach angenommen wurde, der Kürze der Anlaufzeiten wegen in Frage gestellt.
The extension of the nuclear two-centre-oscillator to three and four centres is investigated. Some special symmetry-properties are required. In two cases an analytical solution of the Schrödinger equation is possible. A numerical procedure is developed which enables the diagonalization of the Hamiltonian in a non-orthogonal basis without applying Schmidt's method of orthonormalization. This is important for calculations of arbitrary two-dimensional arrangements of the centres.
I. X-irradiation of isolated rat diaphragm with 10 to 200 kr produces a change in tissue metabolism which we schematize in two successive phases:
1st phase: Increase of oxygen comsumption, proportional to the dosage; an even greater increase of CO2 production; QCO2/QO2 > 1, that is, aerobic glycolysis; inhibition of anaerobic glycolysis.
2nd phase: Reduction of oxygen consumption, proportional to the dosage (over 65 kr the Qo2 decreases below the control); an even greater decrease of CO2 production: QCO2/QO2 > 1; a greater inhibition of anaerobic glycolysis.
With 200 kr or more no increase of respiration appears, but instead from the beginning there is a reduction of the metabolism as described in the second phase.
II. A similar effect is found in rat liver and in frog heart tissue.
III. When the tissue was incubated in the homologus serum no change in the quality of the described effect was observed. Under our experimental conditions the tissue was X-irradiated within a small quantity of incubation medium and immediately afterwards placed in a fresh medium; this limits the effect of oxidative radicals (arising in the X-irradiated water) upon the tissue.
IV. We set forth the experimental hypothesis that all the described changes in the metabolism of the cell after X-irradiation depend upon a primary alteration of electrolyte balance in the cell, especially of the potassium/sodium relationship. The well known decrease of glycolysis after X-irradiation is a consequence of the loss of potassium from the X-irradiated cell.
Die Primärwirkung von Röntgenstrahlung einer Dosis von 2 — 30 Millionen r auf kristallisiertes Lysozym wurde mit Hilfe physikalisch-chemischer (Elektrophorese, Ultrazentrifuge), chemischer, biochemischer und biologischer Arbeitsmethoden untersucht. Es wurde gefunden, daß durch Bestrahlung eine Reihe nah verwandter, jedoch weniger basischer Proteine verschiedenen Mol.-Gew. entsteht, deren Aminosäure-Bausteine als Folge der Bestrahlung teilweise in andere Verbindungen umgewandelt wurden. Bei der Untersuchung der amino- und carboxyl-endständigen Aminosäuren des bestrahlten Proteins wurden Unterschiede gegenüber Lysozym nur bei den carboxyl-terminalen Gruppen festgestellt. Die biologische Aktivität des Proteins blieb auch nach Bestrahlung mit einer Dosis von 5 Millionen r praktisch unverändert.
For experiments on fission-fragment induced desorption the detection of significant correlations between desorbed ions has been reported [1]. In this paper the method for the detection and quantitative description of these correlations will be described. The statistics of the desorption-process leads to equations for mass-line intensities of ion spectra. Using a time-to-amplitude-converter for flight-time measurements these intensities depend on interdependences of different ions desorbed by the same fission-fragment. The equations allow the computation of correlationcoefficients whose interdependence with desorption probabilities of the respective ions can be shown in Venn-diagrams. Results are given and an interpretation is suggested for fission-fragment desorbed thiamine molecular and fragment ions.
In this paper an instability calculation is given for an axially symmetric gas distribution which has a differential rotation and in which a magnetic field is present. It is a generalization of similar calculations given by CHANDRASEKHAR and BEL and SCHATZMAN. The generalization becomes necessary for the study of problems of the formation of planetary systems, and star formation.
The instability conditions and the critical wave lengths are calculated for plane-wave-like disturbances. For disturbances running perpendicularly to the axis of rotation instability can occur only if the gas density exceeds a critical value which depends on the differential rotation at the considered distance only as long as pressure gradients and gradients of the magnetic field strength are negligible. If the gas density exceeds this critical value the shortest unstable wave length is proportional to the square root of vT2+vB2, where vT means the velocity of sound and vB the ALFVÉN-velocity.
For disturbances running parallel to the axis of rotation in addition to the JEANS instability a new type of instability occurs due to the simultaneous action of the magnetic field and the differential rotation; for rigid rotation this instability vanishes.
The temporal development of macroobservables is described within a correlation-functionformalism. The results are exact for a certain class of initial ensembles. The same problem is discussed with the help of the linear-response-formalism. The results agree under certain conditions which should be fulfilled for macroobservables.
Die indirekte Wirkung von Röntgenstrahlen einer Dosis von 0,08 bis 7.5 Millionen r auf eine 2 · 10-2-m. wäßrige Tryptophanlösung wurde mit Hilfe chemischer und physikalisch-chemischer Arbeitsmethoden (Hochspannungs-Elektrophorese) untersucht.
Der Einfluß der Dosis, Dosisleistung, Temperatur und die durch oxydierend wirkende Radikale (HO2) ausgelösten Reaktionsschritte bei Bestrahlung in Sauerstoffatmosphäre wurden in qualitativer Hinsicht geprüft.
Aus der Vielzahl der strahlenchemisch gebildeten Abbauprodukte konnten mit Sicherheit nachgewiesen werden: Glycin, α-Alanin, Asparaginsäure, Kynurenin, 3-Hydroxykynurenin, 3-Oxyanthranilsäure und Tryptamin.
Schließlich wurde versucht, die experimentell gewonnenen Ergebnisse mit Hilfe von Radikalwirkungen, die sich auf
a) Decarboxylierungen
b) und Veränderungen des Kohlenstoff-Gerüstes beziehen, zu deuten.
Elektronenresonanz-Untersuchungen von Nachreaktionen in einem röntgenbestrahlten Faserprotein

An improved two-center model has been used to describe the elastic scattering potentials for the collision of identical nuclei. The macroscopic-microscopic approach includes liquid drop (LD) deformation energies, shell corrections and pairing energy corrections. As basis for the microscopic part a two-center shell model has been used with a Hamiltonian including a Thomas-type spin-orbit potential and an l2 correction term. The model is applied to the sudden and adiabatic type of scattering process, including a compression energy term in the LD part for the former case. Results are given as potential energy surfaces for the adiabatic scattering process and potential energy curves for the sudden scattering process.
Die Aufgaben der biophysikalischen RöNTGEN-Mikrographie und ihre Besonderheiten werden erläutert. Die Notwendigkeit der Erweiterung des Wellenlängenbereiches der zur Anwendung kommenden RÖNTGEN-Strahlen in das extrem-weiche Gebiet bis zu Quantenenergien von etwa 13 eV wird gezeigt. Es wird über die entsprechenden Studien- und Entwicklungsarbeiten im Max-Planck-Institut für Biophysik in Frankfurt am Main berichtet. Insbesondere wird auf die letzte der entwickelten Konstruktionen eingegangen. Die neugebaute RÖNTGEN-Röhre gestattet einen kontinuierlichen Betrieb bei Röhrenspannungen von 50 Volt und Röhrenstromstärke bis 10 mA. Eine weitere Herabsetzung der Röhrenspannung bei etwas verkleinerter Stromstärke ist wahrscheinlich möglich. Einige Beispiele der mit den beschriebenen Typen von RÖNTGEN-Röhren erzielten RÖNTGEN-Mikrogramme biologischer Strukturen werden angegeben und kurz erläutert.
Es wird das Mikrowellenspektrum von Fluorwasserstoffassoziaten im X-und K-Band bei -70 °C und 0,01 Torr gemessen und analysiert. Dazu wird ein erstelltes Frequenzprogramm für den asymmetrischen Kreisel verwendet, sowie ein Extrapolationsprogramm, das eine in der Literatur angegebene druck-und temperaturabhängige Verteilung der Fluorwasserstoffassoziate auf für Mikrowellenspektroskopie geeignete Drücke und Temperaturen umzurechnen erlaubt. Es zeigt sich, daß planare hexamere und heptamere Fluorwasserstoffassoziate vorliegen mit F-F-F-Winkeln von etwa 104° und H-F-Bindungslängen von 0,9997 Å bzw. 0,9640 Å. Die Längen der Wasserstoff brücken sind 1,4998 Å bzw. 1,6105 Å. Ein Vergleich der Bindungslängen zeigt, daß bei Anlagerung von H-F an (HF)6 eine Kontraktion der Fluorwasserstoffbindung um 3,5% und eine Dilatation der Wasserstoffbrückenbindung um 1% stattfindet. Dieses Ergebnis steht im Einklang mit der oben erwähnten Assoziatverteilung, die eine Minderung der Kettenstabilität beim Übergang von hexamerer zu heptamerer Kette erwarten läßt.
We examine the possibility of reformulating quantum theory (QT) as a deterministic ensemble theory which (a) interprets observables as objective properties of physical systems and (b) coincides with QT in all quantitative statements. As will be demonstrated, such an Ensemble-Quantum-Theory (EQT) can only be constructed if (1) one accepts a modified observable-concept, and (2) as long as the theory of measurement is left out of account. A correct treatment of the measuring process is impossible within such an EQT. Consequently, there exist no Hidden-Variable Theories with the properties (a) and (b).
The meaning of a recently proposed formalism for quantization of interacting fields is discussed by studying the consequences of the time-dependent unitary transformation which is essential for this approach. It turns out that non-relativistic quantum electrodynamics in dipole approximation may serve as a useful, although rather singular, example for this method. In the relativistic case a different point of view is suggested in order to avoid inconsistent interpretation. It is further possible to give arguments for a reasonable choice of the unitary transformation concerned.
The often discussed question concerning the energy-momentum tensor of the electromagnetic field in matter can be answered using NOETHER'S theorem. The separation of the electromagnetic system from the mechanical system introduced here leads to the asymmetric expression for the energy momentum tensor. From covariance with respect to scale transformations one further concludes that the trace of the energy-momentum tensor vanishes.
We investigate the possibility of selfconsistent solutions for antiferromagnetism in the Hubbard model in the decoupling of the Greens functions introduced by Hubbard in his first paper. On the base of this approximation Arai has calculated the band splitting for antiferromagnetism, but, as will be shown in this paper, Hubbard's approach fails to yield antiferromagnetism for nearest neighbour hopping in the same way as it does not yield ferromagnetism, and no selfconsistent solutions of the problem beyond the well known paramagnetic solution do exist.
The nuclear charge form factgr from the high-energy elastic electron scattering on 6Li has been calculated from the modified independent-particle shell model (IPSM) wave function. The usual harmonic oscillator type IPSM wave function has been modified by the inclusion of a nucleon-nucleon correlation function which involves extra-core nucleons only. The technique is extremely simple and provides an excellent agreement with the experimental data.
The potential energy surface has been calculated by two methods which are compared with respect to spontaneous fission. In the first one essentially the sum of the single particle energies is computed as was done in a previous paper3 while in the second one the Strutinsky technique of renormalizing to a liquid drop model has been applied. Also the half-lives for electron capture are investigated together with the predictions of the half-lives for spontaneous fission and α-decay. The results support the existence of superheavy nuclei in the regions around Z = 114 and Z = 164.
A careful investigation of different corrections to binding energies of electrons in almost critical fields is performed. We investigate quantitatively the influence of the nuclear charge parameters, nuclear mass, degree of ionization on the value of the critical charge of the nucleus. Rather qualitative arguments are given to establish the contribution of the quantumelectrodynamic corrections, which are found to be small. Some phenomenological modifications of QED are quantitatively investigated and found to be of negligible influence on the value of the critical field. For heavy ion collisions with Z1+Z2>Zcr the critical separations between ions are given as results of precise solutions of the relativistic two coulomb center problem. Corrections due to electron-electron interaction are considered. We find (with present theoretical accuracy) Zcr=173±2, in the heavy ion collisions Rcr(U-U) = 34.7±2 fm and Rcr (U-Cf)=47.7±2 fm. We shortly consider the possibility of spontaneous muon production in muonic supercritical fields.
Recent experimental findings have reported the presence of unconventional charge orders in the enlarged (2 × 2) unit-cell of kagome metals AV3Sb5 (A = K, Rb, Cs) and hinted towards specific topological signatures. Motivated by these discoveries, we investigate the types of topological phases that can be realized in such kagome superlattices. In this context, we employ a recently introduced statistical method capable of constructing topological models for any generic lattice. By analyzing large data sets generated from symmetry-guided distributions of randomized tight-binding parameters, and labeled with the corresponding topological index, we extract physically meaningful information. We illustrate the possible real-space manifestations of charge and bond modulations and associated flux patterns for different topological classes, and discuss their relation to present theoretical predictions and experimental signatures for the AV3Sb5 family. Simultaneously, we predict higher-order topological phases that may be realized by appropriately manipulating the currently known systems.
Es werden Messungen beschrieben, die Übereinstimmung zwischen einer total absorbierenden Ionisationskammer und der Weichstrahl-Standardkammer des Max-Planck-Institutes für Biophysik ergeben. Damit wurde eine weitere Meßanordnung in Betrieb genommen, die besonders für die Absolutmessung der Dosis in „röntgen“ im Gebiet der sehr weichen Röntgenstrahlen geeignet ist.
Neutron stars are unique laboratories for the investigation of the high density properties of bulk matter. In this work, the astrophysical constraints for a phase transition from hadronic matter to deconfined quark matter are examined thoroughly. A scheme for relating known astrophysical observables such as mass, radius and tidal deformability to the parameter space of such a transition is devised and applied to the set of data currently available.
In order to span a wide parameter space, a highly parameterizable relativistic mean field equation in compliance with chiral effective field theory results is used, where the stiffness of the equation of state can be varied via the effective mass at saturation density. The phase transitions are modelled using a Maxwell construction and assumed to be of first order, with a constant speed of sound quark matter model. The resulting equations of state are analyzed and divided into four categories, which can be used to constrain the parameter space that allows phase transition. It is highlighted, that a subset of this parameter space would even be detectable without the need of higher precision measurements. A phase transition at high densities is shown to be particularly promising in this regard. Finally, the groundwork is laid to apply the equation of state used in this work for supernova or merger simulations, by extending it to non-zero temperatures.
Die Wirkungsweise des Protonen-Wendellinearbeschleunigers und sein prinzipieller Aufbau werden kurz beschrieben und Angaben über die erreichbare Parallelimpedanz gemacht. Es wird gezeigt, wie durch sektionsweisen Aufbau eine variable Endenergie erzielt und die Kühl- und Fokussierprobleme gelöst werden können. Zwei Konstruktionsbeispiele für kontinuierlichen und gepulsten Betrieb werden näher ausgeführt.
Light-particle accompanied fission is expected to yield results from which one hopes to learn more about binary scission configurations. As a step in this direction, we present a model that allows the calculation of the probabilities with which a given three-particle setup follows from different binary configurations. First results show the workability of the model.

Die Fermi-Verteilung gibt die Verteilung von Elektronen über die verschiedenen Energieniveaus in einem Festkörper unter der Voraussetzung an, daß sich ein thermodynamischer Gleichgewichtszustand eingestellt hat. Bei der Untersuchung von Leitfähigkeitsproblemen werden geringfügige Störungen dieser Gleichgewichtsverteilung durch elektrische Felder, Temperaturgefälle u. dgl. berücksichtigt. Der innere Photoeffekt bewirkt eine kräftige Störung der Gleichgewichtsverteilung, da fortgesetzt lichtelektrisch ausgelöste Elektronen aus einem niederen in einen höheren Energiezustand gehoben werden.
Im folgenden wird ein statistischer Ansatz für die Verteilung von Elektronen über dargebotene Energiestufen versucht für den Fall, daß laufend je Sekunde eine bestimmte Anzahl aus einem tieferen in ein höheres Niveau gehoben wird.
An alternative formulation is presented of the formal theory of multi-channel scattering in nonrelativistic quantum mechanics. We start by defining spaces of state vectors, where two particles either stay together or separate in the limit t →+∞ (or — ∞), when the state vector develops in time by e–i H t (H is the complete Hamiltonian of the n-particle system). A channel is defined as a space of state vectors with the following property: Developing in time by e-i H t they asymptotically describe a state of the n-particle system, where the particles are grouped in fragments. Defining a Hamiltonian Hγ for each channel, in which—compared to H—the interactions acting between particles from different fragments are missing, it is physically plausible that lim eiH e—iHt Ψ exists for vectors Ψ in the channel. Having discussed the limit vectors (asymptotic states), the S-matrix formalism can be introduced as usual. Finally the introduction of the exclusion principle is discussed.
Es wird eine massenspektrometrische Analysenmethode für Germanium angegeben, die den quantitativen Nachweis einiger Elemente in Germanium bis zu Relativkonzentrationen von 10-5—10-6 gestattet. Diese Methode wurde auf die Messung der Verteilungskoeffizienten von B und In in Ge angewandt. Die Analysen wurden an dem mit Hilfe der Zonenschmelze1 angereicherten Ende eines Regulus durchgeführt.
Die massenspektrometrischen Analysen des Germaniumdampfes zeigten einen molekularen Ge-Anteil in der Dampfphase. Es wurden Cluster-Ionen bis zu Ge8 nachgewiesen. Auch bei Zinn und Silicium fand sich ein Cluster-Anteil in der Dampfphase.
Nach einer kurzen Zusammenstellung von Verfahren, die die Herstellung von Thermoelementen mit geringer Wärmekapazität und -ableitung ermöglichen, werden Konstruktion und thermische Daten der vom Verfasser entwickelten mikroskopisch feinen „Thermonadeln“ wiedergegeben. Um die meßtechnischen Möglichkeiten dieser Thermonadeln beurteilen zu können, wird zunächst eine einfache Theorie des Thermoelementes als Wärmekraftmaschine entwickelt: Der Nutzeffekt wird definiert als der Quotient aus der durch Peltier- und Thomson-Effekt erzeugten elektrischen Energie und den durch die an der Meßstelle des Thermoelementes reversibel und irreversibel aufgenommenen Wärmemengen. Mit Hilfe eines „Arbeitsfaktors A“ (der ein Maß für die Qualität des Thermopaares ist) wird ein theoretischer und praktischer „Gütefaktor G bzw. G“ als das Verhältnis des Nutzeffektes eines Thermoelementes zum Nutzeffekt einer Carnot-Maschine definiert. Dieses Verhältnis gibt an, um welchen Faktor der Nutzeffekt eines Thermoelementes „theoretisch“ und „praktisch“ hinter dem thermodynamisch größtmöglichen Nutzeffekt zurückbleibt. Aus der Tatsache, daß G niemals größer als 1 werden kann, wird eine obere Grenze für die Arbeitsfaktoren A ≦ 1 und damit (in Abhängigkeit.
Die Vorstellungen des Wilsonschen Halbleitermodells werden quantitativ durchgeführt und die Abhängigkeit der Fermischen Grenzenergie von Temperatur, Konzentration und Art der Störstellen berechnet. Es ergibt sich, daß die Grenzenergie das elektrische Verhalten des Halbleiters entscheidet. Bisher ungeklärte experimentelle Ergebnisse werden gedeutet.
Nach neueren Messungen besitzt das Auge eine definierte Empfindlichkeit noch bis etwa 1.0 μ. Es wird hier eine Formel mitgeteilt, die die Messungsresultate befriedigend darstellt. Ihre Auslegung deutet darauf hin, daß der Abfall der Augenempfindlichkeit in diesem "ultraroten" Bereich gerade hinreichend rasch erfolgt, daß keine Dunkelreaktion im Auge den Sehakt stört.
Eine Erweiterung des Differenzen-Verfahrens zur Berechnung von kugelsymmetrischen Temperaturfeldern

Mit dem Differenzenverfahren können in einfacher Weise beliebige Anheiz- und Abkühlungsvorgänge, soweit sie kugelsymmetrisch verlaufen, behandelt werden. Sowohl der Fall, daß eine Kugel von einem Medium konstanter Außentemperatur umgeben wird, wie auch der Fall, daß die Temperatur ohne Störung durch Konvektion in der Umgebung der geheizten Kugel sich einstellt, wird erörtert und führt auf leicht durchführbare Rechnungen und Konstruktionen. Zum Schluß wird gezeigt, wie sich aus den angegebenen Rekursionsformeln die analytischen Gesetzmäßigkeiten der stationären Temperaturverteilung leicht elementar ableiten lassen.
Based on an e+e− collision data sample corresponding to an integrated luminosity of 2.93 fb−1 collected with the BESIII detector at √s=3.773 GeV, the first amplitude analysis of the singly Cabibbo-suppressed decay D+→K+K0Sπ0 is performed. From the amplitude analysis, the K∗(892)+K0S component is found to be dominant with a fraction of (57.1±2.6±4.2)%, where the first uncertainty is statistical and the second systematic. In combination with the absolute branching fraction B(D+→K+K0Sπ0) measured by BESIII, we obtain B(D+→K∗(892)+K0S)=(8.69±0.40±0.64±0.51)×10−3, where the third uncertainty is due to the branching fraction B(D+→K+K0Sπ0). The precision of this result is significantly improved compared to the previous measurement. This result also differs from most of theoretical predictions by about 4σ, which may help to improve the understanding of the dynamics behind.

An Hand neuerer paläoklimatologischer Forschungsergebnisse wird nachgewiesen, daß sowohl die Polwanderungstheorie als auch die astronomische Theorie von Milankovitch in bezug auf die Verhältnisse während des Känozoikums und namentlich während des Alluviums versagen. Es wird weiterhin ausgeführt, daß die Nebelveränderlichkeit nicht etwa durch rein optische Bedeckungseffekte, sondern durch die Bildung einer dichten, den Stern umgebenden Hülle, welche den Energietransport in der Sternatmosphäre stört, hervorgerufen wird. Die kleineren Schwankungen innerhalb der Eiszeiten (Interstadialzeiten) und die kurzdauernden Schwankungen des Alluvialklimas können durch die faserige (Filament-) Struktur der Dunkelwolken erklärt werden.
Im ersten Teil werden kurz die entscheidenden Schwierigkeiten der beiden bekanntesten Versuche zur Erklärung der Eiszeiten, und zwar der Pol-bzw. Kontinentalver-schiebungen (Epeirophorese) und der sog. astronomischen Theorie (Schwankungen der Schiefe der Ekliptik und der Erdbahnelemente) aufgezeigt. - Im zweiten Teil wird wahrscheinlich gemacht, daß eine schon 1921 vorgetragene Hypothese von Shapley, nach der die Sonne in der diluvialen Eiszeit eine von kosmischen Dunkelwolken verursachte schwache Veränderlichkeit ihrer Strahlung zeigte, nach neueren, in erster Linie astrophysikalischen Forschungsergebnissen eine brauchbare Erklärung zu liefern imstande ist, aus folgenden drei Gründen:
1. In allen dichteren kosmischen Dunkelwolken findet man unregelmäßige veränderliche Sterne eines ganz besonderen Typus; im Gegensatz zu fast allen übrigen Typen veränderlicher Sterne handelt es sich bei diesen Sternen um Zwergsterne, wie die Sonne oder noch schwächer.
2. Unser Sonnensystem befindet sich gegenwärtig innerhalb dunkler kosmischer Materie, und es ist höchstwahrscheinlich, daß es in naher Vergangenheit auch dichtere Teile von solchen Dunkelwolken durchquert hat.
3. Die Form der Dunkelwolken, ihre armförmigen, zirrus-oder zirrostratusähnlichen Anordnungen und Verästelungen lassen eine zwanglose Erklärung der mehrfachen Wiederholung der Eiszeiten zu; es wird hier also nicht nur die Eiszeit, sondern aus der gleichen Hauptursache auch deren Gliederung erklärt, was sonst nicht gelungen zu sein scheint.
Die Mitwirkung anderer, in erster Linie astronomischer und geographischer Faktoren, letztere insbesondere in der älteren Erdgeschichte, ist durchaus möglich.
During collisions of heavy ions with heavy targets below the Coulomb barrier, adiabatic molecular orbitals are formed for the inner electrons. Deviations from adiabaticity lead to coupling between various states and can be treated by time-dependent perturbation theory. For high charges ( Z1+Z2 ≧ 60) the molecular electrons are highly relativistic. Therefore, the Dirac equation has to be used to obtain the energies and wave functions. The Dirac Hamiltonian is transformed into the intrinsic rotating coordinate system where prolate spheroidal coordinates are introduced. A set of basis functions is proposed which allows the evaluation of all matrix elements of the Dirac Hamiltonian analytically. The resulting matrix is diagonalized numerically. The finite nuclear charge distribution is also taken into account. Results are presented and discussed for various characteristic systems, e. g. Br-Br, Ni-Ni, I-I, Br-Zr, I-Au, U -U, etc.
We present a unified formulation of the interaction of electrons with the electromagnetic field in heavy ion collisions, based on quantized interacting fields. This reduces the effort in treating many-electron systems substantially, as compared with the usual S-matrix theory. Both formalisms are shown to be equivalent. The simplification achieved by our new approach is demonstrated in detail for the example of quasi-molecular radiation.
A model for the quantum yield of the coloration caused by UV-light in spiropyran layers is described. This model allows to calculate the sensitivity of layers having different compositions. The mechanism concerning the stability of the coloration is essentially clarified. Calculations of the stability for layers of different compositions are possible by a model describing the mechanism approximately.
A scattering theory for reactions with three-particle channels above the two-particle threshold is developed. The S-matrix-technique is used for the calculation of the extended S-matrix. Correlated two-particle wave functions in the exit channels are employed to describe the exact two-particle continuum. For the usual shell model only a few partial waves dominate. The cross section depends on the energy-distribution between the two outgoing nucleons. Numerical results are presented for the model (d, 2n)-reaction exciting 0+ -states in O16 without Coulomb-effects. The treatment is restricted to three (2p2h)-states with the particles in the (sd)-shell and holes in the p-shell.

Coupled channel calculations for Coulomb and nuclear excitation of the systems 136Xe-238U and 238U-238U have been performed using the rotation-vibration model. The impact parameter-, energy-and spin-dependence of the excitation probabilities are discussed for the ground state-, β-and γ-band up to Jπ = 36+. It is shown that the energy levels and quadrupole matrix elements are strongly influenced by the rotation-vibration interaction. Analytic expressions for the elastic and coupling potentials are presented.
Introducing correlated continuum wave functions for the two- and re-particle-continuum a microscopic theory of nuclear reactions based on a method of Fano is developed. The S-matrix-elements are given by the matrix-elements between correlated continuum wave functions and bound state wave functions. The antisymmetrization of the continuum wave functions with more than one particle in the continuum is included. The theory can be straightforwardly applied on the n-nucleon-emission process following photo- and particle excitations.
The master operators B which cause the entropy production dH/dt = - k-1 dS/dt to become extremal for fixed statistical operators W are constructed and discussed. There are boundaries of the set B of master operators, B = {B | Σ B2vu = b} for which the problem is solvable yielding minimal entropy production, while no solution exists in the set B without any constraints. Operators with maximal entropy production must be extremal points of B.
The spike protein (S) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is required for cell entry and is the primary focus for vaccine development. In this study, we combined cryo–electron tomography, subtomogram averaging, and molecular dynamics simulations to structurally analyze S in situ. Compared with the recombinant S, the viral S was more heavily glycosylated and occurred mostly in the closed prefusion conformation. We show that the stalk domain of S contains three hinges, giving the head unexpected orientational freedom. We propose that the hinges allow S to scan the host cell surface, shielded from antibodies by an extensive glycan coat. The structure of native S contributes to our understanding of SARS-CoV-2 infection and potentially to the development of safe vaccines.

We propose that the hinges allow S to scan the host cell surface, shielded from antibodies by an extensive glycan coat. The structure of native S contributes to our understanding of SARS-CoV-2 infection and the development of safe vaccines. The large scale tomography data set of SARS-CoV-2 used for this study is therefore sufficient to resolve structural features to below 5 Ångstrom, and is publicly available at EMPIAR-10453.
Transport of lipids across membranes is fundamental for diverse biological pathways in cells. Multiple ion-coupled transporters take part in lipid translocation, but their mechanisms remain largely unknown. Major facilitator superfamily (MFS) lipid transporters play central roles in cell wall synthesis, brain development and function, lipids recycling, and cell signaling. Recent structures of MFS lipid transporters revealed overlapping architectural features pointing towards a common mechanism. Here we used cysteine disulfide trapping, molecular dynamics simulations, mutagenesis analysis, and transport assays in vitro and in vivo, to investigate the mechanism of LtaA, a proton-dependent MFS lipid transporter essential for lipoteichoic acid synthesis in the pathogen Staphylococcus aureus. We reveal that LtaA displays asymmetric lateral openings with distinct functional relevance and that cycling through outward- and inward-facing conformations is essential for transport activity. We demonstrate that while the entire amphipathic central cavity of LtaA contributes to lipid binding, its hydrophilic pocket dictates substrate specificity. We propose that LtaA catalyzes lipid translocation by a ‘trap-and-flip’ mechanism that might be shared among MFS lipid transporters.

Phosphatidic acid (PA) is an important intermediate at the branch point of these pathways and is continuously monitored by the transcriptional repressor Opi1 to orchestrate lipid metabolism. In this study, we report on the mechanism of membrane recognition by Opi1 and identify an amphipathic helix (AH) for selective binding of PA over phosphatidylserine (PS). The insertion of the AH into the membrane core renders Opi1 sensitive to the lipid acyl chain composition and provides a means to adjust membrane biogenesis. By rational design of the AH, we tune the membrane-binding properties of Opi1 and control its responsiveness in vivo. Using extensive molecular dynamics simulations, we identify two PA-selective three-finger grips that tightly bind the PA phosphate headgroup while interacting less intimately with PS. This work establishes lipid headgroup selectivity as a new feature in the family of AH-containing membrane property sensors.

The SLC26 family of transporters maintains anion equilibria in all kingdoms of life. The family shares a 7 + 7 transmembrane segments inverted repeat architecture with the SLC4 and SLC23 families, but holds a regulatory STAS domain in addition. While the only experimental SLC26 structure is monomeric, SLC26 proteins form structural and functional dimers in the lipid membrane. Here we resolve the structure of an SLC26 dimer embedded in a lipid membrane and characterize its functional relevance by combining PELDOR/DEER distance measurements and biochemical studies with MD simulations and spin-label ensemble refinement. Our structural model reveals a unique interface different from the SLC4 and SLC23 families. The functionally relevant STAS domain is no prerequisite for dimerization. Characterization of heterodimers indicates that protomers in the dimer functionally interact. The SLC26 family of transporters maintains anion equilibria in all kingdoms of life. The family shares a 7 + 7 transmembrane segments inverted repeat architecture with the SLC4 and SLC23 families, but holds a regulatory STAS domain in addition. While the only experimental SLC26 structure is monomeric, SLC26 proteins form structural and functional dimers in the lipid membrane. Here we resolve the structure of an SLC26 dimer embedded in a lipid membrane and characterize its functional relevance by combining PELDOR distance measurements and biochemical studies with MD simulations and spin-label ensemble refinement. Our structural model reveals a unique interface different from the SLC4 and SLC23 families. The functionally relevant STAS domain exerts a stabilizing effect on regions central in this dimer. Characterization of heterodimers indicates that protomers in the dimer functionally interact. The combined structural and functional data define the framework for a mechanistic understanding of functional cooperativity in SLC26 dimers.
Molecular mechanisms of inorganic-phosphate release from the core and barbed end of actin filaments

The release of inorganic phosphate (Pi) from actin filaments constitutes a key step in their regulated turnover, which is fundamental to many cellular functions. However, the molecular mechanisms underlying Pi release from both the core and barbed end of actin filaments remain unclear. Here, we combine cryo-EM with molecular dynamics simulations and in vitro reconstitution to demonstrate how actin releases Pi through a ‘molecular backdoor’. While constantly open at the barbed end, the backdoor is predominantly closed in filament-core subunits and only opens transiently through concerted backbone movements and rotameric rearrangements of residues close to the nucleotide binding pocket. This mechanism explains why Pi escapes rapidly from the filament end and yet slowly from internal actin subunits. In an actin variant associated with nemaline myopathy, the backdoor is predominantly open in filament-core subunits, resulting in greatly accelerated Pi release after polymerization and filaments with drastically shortened ADP-Pi caps. This demonstrates that the Pi release rate from F-actin is controlled by steric hindrance through the backdoor rather than by the disruption of the ionic bond between Pi and Mg2+ at the nucleotide-binding site. Our results provide the molecular basis for Pi release from actin and exemplify how a single, disease-linked point mutation distorts the nucleotide state distribution and atomic structure of the actin filament.
Maximum likelihood estimates of diffusion coefficients from single-particle tracking experiments

Single-molecule localization microscopy allows practitioners to locate and track labeled molecules in biological systems. When extracting diffusion coefficients from the resulting trajectories, it is common practice to perform a linear fit on mean-squared-displacement curves. However, this strategy is suboptimal and prone to errors. Recently, it was shown that the increments between the observed positions provide a good estimate for the diffusion coefficient, and their statistics are well-suited for likelihood-based analysis methods. Here, we revisit the problem of extracting diffusion coefficients from single-particle tracking experiments subject to static noise and dynamic motion blur using the principle of maximum likelihood. Taking advantage of an efficient real-space formulation, we extend the model to mixtures of subpopulations differing in their diffusion coefficients, which we estimate with the help of the expectation–maximization algorithm. This formulation naturally leads to a probabilistic assignment of trajectories to subpopulations. We employ the theory to analyze experimental tracking data that cannot be explained with a single diffusion coefficient. We test how well a dataset conforms to the assumptions of a diffusion model and determine the optimal number of subpopulations with the help of a quality factor of known analytical distribution. To facilitate use by practitioners, we provide a fast open-source implementation of the theory for the efficient analysis of multiple trajectories in arbitrary dimensions simultaneously.
The interaction between the Heat Shock Proteins 70 and 40 is at the core of the ATPase regulation of the chaperone machinery that maintains protein homeostasis. However, the structural details of the interaction remain elusive and contrasting models have been proposed for the transient Hsp70/Hsp40 complexes. Here we combine molecular simulations based on both coarse-grained and atomistic models with coevolutionary sequence analysis to shed light on this problem by focusing on the bacterial DnaK/DnaJ system. The integration of these complementary approaches resulted in a novel structural model that rationalizes previous experimental observations. We identify an evolutionarily conserved interaction surface formed by helix II of the DnaJ J-domain and a structurally contiguous region of DnaK, involving lobe IIA of the nucleotide binding domain, the inter-domain linker, and the β-basket of the substrate binding domain.

TriMem: A parallelized hybrid Monte Carlo software for efficient simulations of lipid membranes

Lipid membranes are integral building blocks of living cells and perform a multitude of biological functions. Currently, molecular simulations of cellular-scale membrane remodeling processes at atomic resolution are extremely difficult, due to their size, complexity, and the large times-scales on which these processes occur. Instead, elastic membrane models are used to simulate membrane shapes and transitions between them and to infer their properties and functions. Unfortunately, an efficiently parallelized open-source simulation code to do so has been lacking. Here, we present TriMem, a parallel hybrid Monte Carlo simulation engine for triangulated lipid membranes. The kernels are efficiently coded in C++ and wrapped with Python for ease-of-use. The parallel implementation of the energy and gradient calculations and of Monte Carlo flip moves of edges in the triangulated membrane enable us to simulate large and highly curved membrane structures. For validation, we reproduce phase diagrams of vesicles with varying surface-to-volume ratios and area difference. We also compute the density of states to verify correct Boltzmann sampling. The software can be used to tackle a range of large-scale membrane remodeling processes as a step toward cell-scale simulations. Additionally, extensive documentation make the software accessible to the broad biophysics and computational cell biology communities.
Abstract
The primary immunological target of COVID-19 vaccines is the SARS-CoV-2 spike (S) protein. S is exposed on the viral surface and mediates viral entry into the host cell. To identify possible antibody binding sites, we performed multi-microsecond molecular dynamics simulations of a 4.1 million atom system containing a patch of viral membrane with four full-length, fully glycosylated and palmitoylated S proteins. By mapping steric accessibility, structural rigidity, sequence conservation, and generic antibody binding signatures, we recover known epitopes on S and reveal promising epitope candidates for structure-based vaccine design. We find that the extensive and inherently flexible glycan coat shields a surface area larger than expected from static structures, highlighting the importance of structural dynamics. The protective glycan shield and the high flexibility of its hinges give the stalk overall low epitope scores. Our computational epitope-mapping procedure is general and should thus prove useful for other viral envelope proteins whose structures have been characterized.

The SARS-CoV-2 virus has caused a global health crisis. The spike protein exposed at its surface is key for infection and the primary antibody target. However, spike is covered by highly mobile glycan molecules that could impair antibody binding. To identify accessible epitopes, we performed molecular dynamics simulations of an atomistic model of glycosylated spike embedded in a membrane. By combining extensive simulations with bioinformatics analyses, we recovered known antibody binding sites and identified several epitope candidates as targets for further vaccine development.
More than 75% of surface and secreted proteins are modified by covalent addition of complex sugars through N- and O-glycosylation. Unlike proteins, glycans do not typically adopt specific secondary structures and remain very mobile, influencing protein dynamics and interactions with other molecules. Glycan conformational freedom impairs complete structural elucidation of glycoproteins. Computer simulations may be used to model glycan structure and dynamics. However, such simulations typically require thousands of computing hours on specialized supercomputers, thus limiting routine use. Here, we describe a reductionist method that can be implemented on personal computers to graft ensembles of realistic glycan conformers onto static protein structures in a matter of minutes. Using this open-source pipeline, we reconstructed the full glycan cover of SARS-CoV-2 Spike protein (S-protein) and a human GABAA receptor. Focusing on S-protein, we show that GlycoSHIELD recapitulates key features of extended simulations of the glycosylated protein, including epitope masking, and provides new mechanistic insights on N-glycan impact on protein structural dynamics.
Transient receptor potential (TRP) ion channels are among the most well-studied classes of temperature-sensing molecules. Yet, the molecular mechanism and thermodynamic basis for the temperature sensitivity of TRP channels remains to this day poorly understood. One hypothesis is that the temperature-sensing mechanism can simply be described by a difference in heat capacity between the closed and open channel states. While such a two-state model may be simplistic it nonetheless has descriptive value, in the sense that it can be used to to compare overall temperature sensitivity between different channels and mutants. Here, we introduce a mathematical framework based on the two-state model to reliably extract temperature-dependent thermodynamic potentials and heat capacities from measurements of equilibrium constants at different temperatures. Our framework is implemented in an open-source data analysis package that provides a straightforward way to fit both linear and nonlinear van ‘t Hoff plots, thus avoiding some of the previous, potentially erroneous, assumptions when extracting thermodynamic variables from TRP channel electrophysiology data.

Am Teilchenbeschleuniger in Darmstadt werden die extremen Bedingungen unseres Universums im Labor erforscht. Dabei gelang es den Physikerinnen und Physikern, eine Technologie zu entwickeln, die Energie zur Teilchenbeschleunigung wiederverwendet und einspart. Der Teilchenbeschleuniger ist eingebunden in das Clusterprojekt ELEMENTS, das gemeinsam von der Goethe-Universität Frankfurt und der TU Darmstadt geleitet wird.
Using 10.1 × 109 J/ψ events produced by the Beijing Electron Positron Collider (BEPCII) at a center-of-mass energy √s = 3.097 GeV and collected with the BESIII detector, we present a search for the rare semi-leptonic decay J/ψ → D−e+νe + c.c. No excess of signal above background is observed, and an upper limit on the branching fraction ℬ(J/ψ → D−e+νe + c. c.) < 7.1 × 10−8 is obtained at 90% confidence level. This is an improvement of more than two orders of magnitude over the previous best limit.
