Capoeta damascina (Teleostei: Cyprinidae) is one of the most common freshwater fish species, found throughout the Levant, Mesopotamia, Turkey and Iran. According to the state of knowledge prior to this study, C. damascina, which is distributed over a wide range of isolated water bodies, was not a well-defined species. It was questionable whether it represents a single species or a complex of closely related species with high intraspecific and comparatively low interspecific variability. The goal of this study was to investigate the taxonomy, systematic position of the C. damascina species complex and the phylogenetic relationships among its members, based on morphological features as well as molecular phylogeny. Samples obtained from throughout the geographic range of this species complex were subjected to comparative morphological analyses in order to define, properly diagnose and separate species within the C. damascina complex. To elucidate phylogenetic relationships among members of the C. damascina species complex, samples were subjected to genetic analyses, using two molecular markers targeting the mitochondrial cytochrome oxidase I (COI, n = 103) and the two adjacent divergence regions (D1-D2) of the nuclear 28S rRNA genes (LSU, n = 65). Based on morphological and molecular genetic data, six closely related species were recognized within the C. damascina complex: C. buhsei, C. caelestis, C. damascina, C. saadii, C. umbla and an undescribed species, Capoeta sp.1. Analyses of the morphometric and meristic data obtained in this study revealed phenotypic variability among the various populations within a species and among the different species. Such differences in morphological characters reflect genetic differences, environmentally induced phenotypic variation or both, as the meristic phenotype of fish is sometimes a consequence of environmental parameters acting on the genotype. Based on phylogenetic analyses, two main lineages were identified within the C. damascina species complex: a western lineage represented by C. caelestis, C. damascina and C. umbla and an eastern lineage represented by C. buhsei, C. saadii and Capoeta sp.1. The close phylogenetic relationships between C. damascina and C. umbla and the sharing of same haplotypes between one specimen of C. damascina from Euphrates and another of C. umbla from Tigris reflect one of three possibilites: recent speciation, mitochondrial introgression or a combination of both. The results obtained in this study indicate that speciation of the above-mentioned six taxa is quite recent and that their dispersal and present-day distribution can be related to Pleistocene events. The drying out of the Persian Gulf, probably during one of the first glacials of the Pleistocene, led the ancestor of the C. damascina species complex in Mesopotamia to reach the rivers of the Gulf and of Hormuz basins and differentiate there, giving rise to the eastern lineage (ancestor of C. buhsei, C. saadii and Capoeta sp.1). As connections presumably existed among the different river drainages and basins in Iran during the wet periods of the Pleistocene, the ancestor of C. buhsei, C. saadii and Capoeta sp.1 was subsequently able to colonize the various Iranian drainages and differentiate there, giving rise to C. buhsei, C. saadii and Capoeta sp.1. After the separation from the eastern lineage, the western lineage, represented by the ancestor of C. damascina, C. umbla and C. caelestis, most likely reached the Levant from the Tigris-Euphrates system during the Pleistocene glacials, when river connections existed in the regions of the upper courses of Ceyhan Nehri (southern Turkey) and some western affluents to the Euphrates. From Ceyhan Nehri, it dispersed into other rivers in southern Turkey during Pleistocene periods of low sea levels until it reached Göksu Nehri and evolved into C. caelestis. The sister population differentiated into C. damascina and C. umbla. Based on the results obtained in this study, it is likely that C. damascina colonized the Levant and southern Turkey during the Pleistocene glacials. This is well supported by the low genetic variability among the C. damascina populations. Direct connections existed among the river drainages in the Levant during the Pleistocene periods of low sea level, thus serving as a pathway for the dispersal of C. damascina. The results of this study provide a coherent picture of the taxonomic position, phylogenetic relationships and evolutionary history of the C. damascina species complex and explain present patterns of distribution considering paleogeographic events.
Development of a computational method for reaction-driven de novo design of druglike compounds

A new method for computer-based de novo design of drug candidate structures is proposed. DOGS (Design of Genuine Structures) features a ligand-based strategy to suggest new molecular structures. The quality of designed compounds is assessed by a graph kernel method measuring the distance of designed molecules to a known reference ligand. Two graph representations of molecules (molecular graph and reduced graph) are implemented to feature different levels of abstraction from the molecular structure. A fully deterministic construction procedure explicitly designed to facilitate synthesizability of proposed structures is realized: DOGS uses readily available synthesis building blocks and established reaction schemes to assemble new molecules. This approach enables the software to propose not only the final compounds, but also to give suggestions for synthesis routes to generate them at the bench. The set of synthesis schemes comprises about 83 chemical reactions. Special focus was put on ring closure reactions forming drug-like substructures. The library of building blocks consists of about 25,000 readily available synthesis building blocks. DOGS builds up new structures in a stepwise process. Each virtual synthesis step adds a fragment to the growing molecule until a stop criterion (upper threshold for molecular mass or number of synthesis steps) is fulfilled. In a theoretical evaluation, a set of ~1,800 molecules proposed by DOGS is analyzed for critical properties of de novo designed compounds. The software is able to suggest drug-like molecules (79% violate less than two of Lipinski’s ‘rule of five’). In addition, a trained classifier for drug-likeness assigns a score >0.8 to 51% of the designed molecules (with 1.0 being the top score). In addition, most of the DOGS molecules are deemed to be synthesizable by a retro-synthesis descriptor (77% of molecules score in the top 10% of the decriptor’s value range). Calculated logP(o/w) values of constructed molecules resemble a unimodal distribution centred close to the mean of logP(o/w) values calculated for the reference compounds. A structural analysis of selected designs reveals that DOGS is capable of constructing molecules reflecting the overall topological arrangement of pharmacophoric features found in the reference ligands. At the same time, the DOGS designs represent innovative compounds being structurally distinct from the references. Synthesis routes for these examples are short and seem feasible in most cases. Some reaction steps might need modification by using protecting groups to avoid unwanted side reactions. Plausible bioisosters for known privileged fragments addressing the S1 pocket of trypsin were proposed by DOGS in a case study. Three of them can be found in known trypsin inhibitors as S1-adressing side chains. The software was also tested in two prospective case studies to design bioactive compounds. DOGS was applied to design ligands for human gamma-secretase and human histamine receptor subtype 4 (hH4R). Two selected designs for gamma-secretase were readily synthesizable as suggested by the software in one-step reactions. Both compounds represent inverse modulators of the target molecule. In a second case study, a ligand candidate selected for hH4R was synthesized exactly following the three-step synthesis plan suggested by DOGS. This compound showed low activity on the target structure. The concept of DOGS is able to deliver synthesizable and bioactive compounds. Suggested synthesis plans of selected compounds were readily pursuable. DOGS can therefore serve as a valuable idea generator for the design of new pharmacological active compounds.
Die Hitzestresstranskriptionsfaktoren HsfA1 und HsfA2 repräsentieren wichtige transkriptionelle Regulatoren in der Regulation der Hitzestressantwort von Lycopersicon esculentum (Tomate). Unter Stressbedingungen induziert HsfA1 die Expression von HsfA2 und bildet heterooligomere HsfA1/HsfA2 Komplexe, die im Zusammenhang mit der erhöhten Expression von Hitzestressgenen stehen (Scharf et al., 1998b, Mishra et al., 2002, Port et al., 2004). Durch funktionelle Charakterisierungen der Wechselwirkung zwischen HsfA1 und HsfA2 werden neue Aspekte der spezifischen und synergistischen Aktivierung durch HsfA1 und HsfA2 erläutert. - Die Spezifität der funktionellen Interaktion zwischen HsfA1 und HsfA2 wird in Vergleich mit weiteren Klasse A Hsfs, HsfA3, HsfA4b und HsfA5 anhand von GUS Reporter Assays, Coimmunpräzipitationsanalysen und der interaktionsvermittelten Kernretention von GFP-HsfA2 verdeutlicht. Trotz des Potenzials von HsfA2, multiple Wechselwirkungen einzugehen, ist die Spezifität zwischen HsfA1 und HsfA2 am höchsten. Für die Analyse der synergistische Aktivierung durch HsfA1 und HsfA2 werden 3HA-HsfA1 und 3HA-HsfA2 in unterschiedlichen Mengenverhältnissen coexprimiert. Sowohl am Hsp17.3B-CI::GUS Reporter als auch an der induzierte, endogene Tabak Hsp17-CI Expression kann der spezifische Effekt der synergistischen Aktivierung durch HsfA1 und HsfA2 demonstriert werden. - Um die strukturellen Voraussetzungen der synergistischen Aktvierung zu definieren, werden Mutanten mit Defekten in der DNA Bindung, Oligomerisierung und Aktivierung in funktionellen Analysen der transkriptionellen Aktivität (GUS Reporter Assays, Induktion endogener Hsp17-CI Expression), Komplexbildung (Co-Immunpräzipitation) und der HsfA1 vermittelte Kernretention von HsfA2 (Immunfluoreszenz) untersucht. Die synergistische Aktivierung erfordert die Bildung heterooligomerer HsfA1/HsfA2 Komplexe, die über eine Kombination ihrer C-terminalen Aktivierungsdomänen kooperativ aktivieren. Dagegen hat die DNA Bindung durch die DBDs beider Hsfs einen geringen Anteil an der synergistischen Aktivierung. Zur Verifizierung der funktionellen Unterschiede zwischen HsfA1 und HsfA2 werden HsfA1-HsfA2 Hybride durch Coexpression mit HsfA1 und HsfA2 Wildtypformen analysiert. Heterooligomere Komplexe aus Wildtyp und Hybrid-Hsfs zeigen ausschließlich eine synergistische Aktivierung, wenn die C-terminalen Aktivierungsdomänen von beiden Hsf Typen stammen, während heterooligomere HsfA1/HsfA2 Komplexe mit typgleichen C-Termini nicht synergistisch aktivieren. Weiterhin wird gezeigt, dass Wildtyp- Hybridkomplexe mit identischen HR-A/B Regionen in der synergistischen Aktivierung abgeschwächt sind. - Die Bildungseigenschaften der DNA-Hsf Komplexe (DNP) von HsfA1 und HsfA2 werden in Hinblick auf qualitative Veränderungen unter Coexpressionsbedingungen betrachtet. Interessanterweise konnte die Bildung intermediärer DNPs sowie von Hsf-Komplexen mit intermediärer Größe in Gelfiltrationsanalysen als Indizien für qualitativ veränderte HsfA1/HsfA2 Komplexen nachgewiesen werden. Die funktionelle Analyse von HsfA1 C-terminalen Deletionsmutanten führt zur Identifizierung einer de-regulierten HsfA1 Mutante, die trotz de-regulierter Aktivität mit HsfA2 zur synergistische Aktivierung fähig ist. - Zur Verifizierung der interaktionsvermittelten synergistischen Aktivierung wird die Oligomerisierung partiell deletierter HsfA2 HR-A/B Mutanten ermittelt. Da diese Mutanten intermediäre Oligomerisierungszustände zeigen, werden durch die systematische Deletionsmutation der HR-A/B Region von HsfA2 strukturellen Voraussetzungen für die synergistische Aktivierung durch HsfA1 und HsfA2 charakterisiert. Co-Immunpräzipitationsversuche belegen, dass die Integrität der HR-A/B Region für die Bildung stabiler HsfA1/HsfA2 Komplexe benötigt wird, jedoch eine transiente und spezifische Interaktion über die C-terminalen L2 und HR-B Regionen für die synergistische Aktivierung ausreicht. - In der Charakterisierung der kooperativen, synergistischen Aktivierung durch beide CTADs werden Mutanten der vier vorhandenen AHA Motive von HsfA1 und HsfA2 durch Coexpression mit dem Wildtyp Hsf Partner getestet. Die Ergebnisse verdeutlichen, dass jedes der AHA Motive unterschiedlich zur synergistischen Aktivierung beitragen.
Background: The combination of high-throughput transcript profiling and next-generation sequencing technologies is a prerequisite for genome-wide comprehensive transcriptome analysis. Our recent innovation of deepSuperSAGE is based on an advanced SuperSAGE protocol and its combination with massively parallel pyrosequencing on Roche's 454 sequencing platform. As a demonstration of the power of this combination, we have chosen the salt stress transcriptomes of roots and nodules of the third most important legume crop chickpea (Cicer arietinum L.). While our report is more technology-oriented, it nevertheless addresses a major world-wide problem for crops generally: high salinity. Together with low temperatures and water stress, high salinity is responsible for crop losses of millions of tons of various legume (and other) crops. Continuously deteriorating environmental conditions will combine with salinity stress to further compromise crop yields. As a good example for such stress-exposed crop plants, we started to characterize salt stress responses of chickpeas on the transcriptome level. Results: We used deepSuperSAGE to detect early global transcriptome changes in salt-stressed chickpea. The salt stress responses of 86,919 transcripts representing 17,918 unique 26bp deepSuperSAGE tags (UniTags) from roots of the salt-tolerant variety INRAT-93 two hours after treatment with 25 mM NaCl were characterized. Additionally, the expression of 57,281 transcripts representing 13,115 UniTags was monitored in nodules of the same plants. From a total of 144,200 analyzed 26bp tags in roots and nodules together, 21,401 unique transcripts were identified. Of these, only 363 and 106 specific transcripts, respectively, were commonly up- or down-regulated (>3.0-fold) under salt stress in both organs, witnessing a differential organ-specific response to stress. Profiting from recent pioneer works on massive cDNA sequencing in chickpea, more than 9,400 UniTags were able to be linked to UniProt entries. Additionally, gene ontology (GO) categories over-representation analysis enabled to filter out enriched biological processes among the differentially expressed UniTags. Subsequently, the gathered information was further cross-checked with stress-related pathways. From several filtered pathways, here we focus exemplarily on transcripts associated with the generation and scavenging of reactive oxygen species (ROS), as well as on transcripts involved in Na+ homeostasis. Although both processes are already very well characterized in other plants, the information generated in the present work is of high value. Information on expression profiles and sequence similarity for several hundreds of transcripts of potential interest is now available. Conclusions: This report demonstrates, that the combination of the high-throughput transcriptome profiling technology SuperSAGE with one of the next-generation sequencing platforms allows deep insights into the first molecular reactions of a plant exposed to salinity. Cross validation with recent reports enriched the information about the salt stress dynamics of more than 9,000 chickpea ESTs, and enlarged their pool of alternative transcripts isoforms. As an example for the high resolution of the employed technology that we coin deepSuperSAGE, we demonstrate that ROS-scavenging and -generating pathways undergo strong global transcriptome changes in chickpea roots and nodules already 2 hours after onset of moderate salt stress (25mM NaCl). Additionally, a set of more than 15 candidate transcripts are proposed to be potential components of the salt overly sensitive (SOS) pathway in chickpea. Newly identified transcript isoforms are potential targets for breeding novel cultivars with high salinity tolerance. We demonstrate that these targets can be integrated into breeding schemes by micro-arrays and RT-PCR assays downstream of the generation of 26bp tags by SuperSAGE.
Während der vergangenen Jahrzehnte stieg die durchschnittliche Lebenserwartung der Bevölkerung in den westlichen Industrieländern durch die Verbesserung der allgemeinen Lebensbedingungen, insbesondere durch die Fortschritte in der Hygiene und der Medizin sowie stabile politische Verhältnisse, kontinuierlich an. Aufgrund dieser demographischen Entwicklung zu einer zunehmend älter werdenden Gesellschaft nimmt auch das Auftreten von progressiven, altersabhängigen Erkrankungen, wie zum Beispiel der Parkinson‟schen Krankheit zu. Dieser Trend stellt sowohl für die betroffenen Patienten und ihre Angehörigen als auch für die Gesundheits- und Sozialsysteme eine gewaltige und kostenintensive Herausforderung dar. Um wirkungsvolle Therapien entwickeln zu können, die früh im Krankheitsverlauf eingreifen und die Manifestation der Erkrankung verhindern oder verzögern beziehungs-weise die darauf abzielen, die Symptome der Erkrankung nach deren Manifestation zu lindern, ist es unerlässlich, die diesen progressiven, altersabhängigen Krankheiten zugrundeliegenden Mechanismen zu erforschen und entsprechende krankheitsspezifische, molekulare Biomarker zu identifizieren. Darüber hinaus stellt die Identifizierung solcher Biomarker einen wichtigen Ansatzpunkt für die klinische Diagnostik und Therapeutik sowie für die Entwicklung neuer therapeutischer Behandlungsstrategien dar. Das subzellulär vorwiegend präsynaptisch lokalisierte Protein alpha-Synuklein blieb in den Jahren nach seiner Erstbeschreibung 1988 durch Luc Maroteaux von der biomedizinischen Forschung weitgehend unbeachtet. Erst die Assoziationen von unterschiedlichen Mutationen des alpha-Synuklein-Gens mit seltenen, autosomal-dominant vererbten, monogenetischen Varianten der Parkinson‟schen Krankheit (PARK1 und PARK4) seit 1997 sowie die Identifizierung des Proteins im Jahre 1998 als Hauptbestandteil von intrazellulären Proteinaggregaten (Lewy-Körpern und Lewy-Neuriten), deren Vorkommen charakteristisch für progressive, neurodegenerative und unter dem Sammelbegriff „Synukleinopathien“ klassifizierte Erkrankungen (wie beispielsweise auch die häufigen, sporadischen Formen der Parkinson‟schen Krankheit) ist, ließen das alpha-Synuklein in den Fokus der biomedizinischen Forschung rücken. Trotz intensiver Bemühungen der weltweiten Forschungsgemeinschaft konnten seitdem in den vergangenen 13 Jahren die physiologischen Funktionen von alpha-Synuklein und die den unterschiedlichen Synukleinopathien zugrundeliegenden, molekularen pathophysiologischen Mechanismen nicht genau identifiziert werden. Stattdessen führte die intensive Forschung an alpha-Synuklein mit den unterschiedlichsten experimentellen Herangehensweisen und Modellsystemen zu verschiedenen und teilweise kontroversen Hypothesen und Theorien über dessen physiologische Funktion und pathophysiologische Wirkungsweisen. Die in dieser Dissertationschrift dargestellten experimentellen Untersuchungen wurden an zwei speziellen transgenen Mausmodellen durchgeführt, die entweder einen vollständigen Mangel (= „knockout“; KO) des alpha-Synuklein-Proteins oder eine transgene Überexpression von humanem, A53T-mutierten alpha-Synuklein aufwiesen. Das Hauptziel der dargestellten Studien war es, neue Erkenntnisse hinsichtlich der physiologischen Funktionen des alpha-Synuklein-Proteins, beziehungsweise der krankheits-relevanten, pathophysiologischen Mechanismen der den familiären PARK1- und PARK4-Varianten der Parkinson‟schen Krankheit zugrundeliegenden alpha-Synuklein-Mutationen (Substitution von Alanin durch Threonin an Position 53 der Aminosäuresequenz (A53T; PARK1) sowie Überexpression (Genduplikation/-triplikation; PARK4)) zu gewinnen...
Signal-dependent regulation of actin dynamics is essential for many cellular processes, including directional cell migration. In particular, cell migration is initiated by lamellipodia, actin-based protrusions of the plasma membrane. The formation of these protruding structures require incessant assembly and disassembly of actin filaments. The Arp2/3 complex and WAVE proteins are essential for both lamellipodium formation and its dynamics. WAVEs mediate the activation of the Arp2/3 complex downstream of the small GTPase Rac, thus being critical for Rac- and RTK-induced actin polymerization and cell migration. The WAVE-family proteins are always found associated with multiprotein complexes. The most abundant WAVE-based complex is referred to as the WANP (WAVE2-Abi-1-Nap1-PIR121) complex. IQGAP1 is a huge scaffolding protein with multiple protein-interacting domains. IQGAP1 participates in many fundamental activities, including regulation of the actin cytoskeleton, mitogenic, adhesive and migratory responses, as well as in cell polarity and cellular trafficking. IQGAP1 binds to N-WASP, thus raising the possibility that it might control actin nucleation by the Arp2/3 complex. In this study, IQGAP1 was found co-immunoprecipitated not only with WAVE, but also with the endogenous WANP-complex subunits. Correspondingly, IQGAP1 associated to both anti-WAVE and anti-Abi-1 immuno-complexes. Pull-down experiments proved that IQGAP1 binds directly to the WANP-complex subunits. Physical interaction between IQGAP1 and the reconstituted WANP complex could also be demonstrated. Together, these data indicate that IQGAP1 is an accessory component of the WANP complex. Interestingly, the IQGAP-WANP complex disassembled after either EGF stimulation or transfection with constitutively active Cdc42 and Rac1. HeLa cells devoid of IQGAP1 showed diminished and less persistent ruffling upon EGF, but not HGF, stimulation in comparison with the control. This phenotype was accompanied by a strong reduction in chemotaxis towards both growth factors, which was as dramatic as in WANP-complex knockdown (KD) cells. Moreover, GM130 and Giantin showed a polarized and flat ribbon-like pattern in control cells, as it is expected for cis- and cis/medial-Golgi markers. Conversely, small and dispersed vesicular structures were found in both IQGAP1 KD and WANP-complex KD cells. Importantly, Arp2/3-complex silencing resulted in the same phenotypes. Consistently, Brefeldin A-induced disassembly of the Golgi strongly inhibited the IQGAP1-WANP-complex interaction and chemotaxis towards EGF in wild-type cells. The re-expression of an RNAi-resistant wild-type IQGAP1 in IQGAP1 KD cells fully rescued both the ruffling abilities and Golgi structure. A constitutively active mutant, unable to bind to neither Rac1 /Cdc42 nor the WANP complex, could reconstitute only the former defect. Hence, this study shows that actin dynamics regulated by the IQGAP1-WANP complex controls Golgi-apparatus architecture and its contribution to cell chemotaxis. The working model here proposes that at the Golgi apparatus, recruitment of the WANP complex by IQGAP1 leads to the assembly of actin filaments required to maintain the appropriated Golgi morphology. The dissociation of the complex may be required to allow the remodeling of the Golgi membranes in order to respond following a chemoattractant gradient.
Primäre Hirntumore des Zentralen Nervensystems (gehen aus Nervenzellen, Gliazellen, Hirnhäute (Meningen) und auch Hirngefäße aus. Das anaplastische Astrozytom, Oligodendrogliom und Oligoastrozytom (WHO-Grad III) und auch die bösartigste Form, das Glioblastoma multiforme (WHO-Grad IV) bilden den wesentlichen Anteil der astrozytären Tumoren. Bedingt durch die ungünstige Prognose für Patienten mit Glioblastomen bedarf es einer Optimierung der therapeutischen Maßnahmen. Die momentane Therapie besteht nach Konstitution des Patienten und der Lokalisation des Tumors meist aus der Operation mit angeschlossener Strahlen- und eventueller adjuvanten Chemotherapie. Die Prognose ist abhängig von verschiedenen Faktoren, wie der Ausbreitung und der Lokalisation des Tumors und hat sich in den letzten Jahren leider nur unwesentlich verbessert, da immer noch von einer medianen Überlebenszeit des Patienten zwischen 8 und 15 Monaten im Falle eines Glioblastoms ausgegangen werden kann. Die Entstehung und Progression von malignen Tumorerkrankungen sind eng mit Störungen und Defekten in Signalwegen, die das Zellüberleben und den Zelltod regulieren, verknüpft. Aberrationen, wie die Blockade von Tumorsuppressorgenen, oder die Aktivierung bzw. Überexpression von Onkogenen, führen zur veränderten Expression von Zelltod-inhibitorischen Genen wie den anti-apoptotischen Bcl-2 Familienmitgliedern, und letztendlich zu Abweichungen der normalen zellulären Homöostase. Veränderungen dieser Art bewirken, dass Tumorzellen die Fähigkeit erhalten, unkontrolliert zu proliferieren, infiltrativ zu wachsen und eine eigene Vaskularisierung zu initiieren. Tumorzellen besitzen eine erhöhte Apoptose- und Zelltodresistenz, die maßgeblich durch die stark erhöhte Expression von anti-apoptotischen Bcl-2-Proteinen gekennzeichnet ist. In der vorliegenden Arbeit stand daher die Fragestellung im Mittelpunkt, inwieweit anti-apoptotische Proteine der Bcl-2-Familie zur Resistenz von malignen Gliomzellen gegen die Apoptose und den autophagischen Zelltod beitragen und wie diese Mechanismen gezielt überwunden werden können. Die Blockade der vier Familienmitglieder der Bcl-2-Subfamilie (Bcl-2, Bcl-xL, Mcl-1, Bcl-w) wurde über den Einsatz von spezifischen Inhibitoren, den sogenannten BH3-Mimetika, vermittelt. BH3-Mimetika besitzen eine hohe Affinität zur Bindungstasche der Bcl-2-Proteine, wobei durch die Bindung eine Blockade des Proteins hervorgerufen wird. Die eingesetzten BH3-Mimetika haben unterschiedliche Bindungsprofile und können daher ein, zwei oder mehrere Proteine der Bcl-2-Familie inhibieren. Die verwendeten Inhibitoren BH3I-2, HA14-1 und ABT-737, die nur eine limitierte Wirkung auf den Zelltod von Gliomzellen entfalteten, sind allesamt nicht in der Lage, Mcl-1 zu blockieren. Deshalb wurde zusätzlich ein Pan-Bcl-2-Inhibitor, das (-)-Gossypol, verwendet. Dieser Inhibitor hemmt alle anti-apoptotischen Bcl-2-Familienmitglieder, und steigerte über die Inhibition und Degradation von Mcl-1 effizient den Zelltod von Gliomzellen. Dagegen zeigte die (-)-Gossypol-Behandlung in nicht transformierte Astrozytenkulturen keine zytotoxischen Effekte. Zusätzlich wurde nachgewiesen, dass durch das BH3-Mimetikum (-)-Gossypol ein autophagischer Zelltod induziert wird. Unter der Verwendung eines charakteristischen Markerproteins, dem LC3-Protein, konnte nach Induktion des nicht-apoptotischen, autophagischen Zelltodes durch (-)-Gossypol eine Translokation von GFP-LC3 in die Autophagosomen und Autolysosomen festgestellt werden. Viele Gliome weisen eine Methylierung des MGMT-Promotors auf, die es den Zellen erschwert, schnell DNA-Schäden zu reparieren. Aufgrund dieser Methylierung sind diese Tumoren sensitiv für eine Behandlung mit alkylierenden Substanzen (Temozolomid). Auf Grundlage dieser Beobachtungen wurden die kombinatorischen Effekte von BH3 Mimetika und Temozolomid in MGMT-positiven (U87) und MGMT-negativen (U343) Gliomzelllinien verglichen, wobei sich lediglich in MGMT-negativen U343-Gliomzellen signifikante, kombinierte Effekte ergaben. Dieser Zelltod war mit einer Potenzierung der Autophagie assoziiert, nicht jedoch mit einer Aktivierung von Caspasen und einer lysosomalen Dysfunktion. Die Depletion des endogenen Autophagieinhibitors mTOR bewirkte nach den Behandlungen mit (-)-Gossyol und TMZ einen zusätzlichen zytotoxischen Effekt. Im Gegensatz dazu wurde durch lentivirale RNA-Interferenz gegen die Autophagiegene ATG5 und Beclin1 eine potente Reduktion der zellulären Autophagie und des autophagischen Zelltodes erreicht. Insgesamt unterstreichen diese Daten die zentrale Rolle von Bcl-2 Proteinen für die Regulation der Autophagie und legen nahe, dass durch (-)-Gossypol und (-)-Gossypol in Kombination mit Temozlomid in Gliomzellen zytotoxische Autophagie induziert wird.
In dieser Arbeit wurden die physiologische Funktion innerhalb der Ribosomenbiogenese und die physikalischen Interaktionen des nukleolären, essentiellen Proteins Nep1p in der Hefe Saccharomyces cerevisiae untersucht. Durch Hefe-Zwei-Hybrid-Experimente und biochemische Analysen konnte eine Homodimerisierung des Proteins festgestellt sowie eine strukturabgeleitete Dimerisierungsmutante identifiziert werden. Ebenfalls aus der Struktur des Nep1p-Homologs aus Methanocaldococcus jannaschii konnte eine Nop14p-Bindungsregion auf der der Dimerkontaktfläche abgewandten Seite des Hefeproteins vorhergesagt und nach in vitro-Mutagenese bestätigt werden. Innerhalb des Nop14-Proteins wurden zwei Domänen charakterisiert, die im Zwei-Hybrid-System mit Nep1p interagieren. Aus Strukturdaten in Kombination mit Hefe-Drei-Hybrid-Experimenten konnte die RNA-Bindungsregion an der Dimerkontaktfläche des Nep1-Proteins lokalisiert werden. In Drei-Hybrid-Selektionen wurden RNA-Sequenzen mit hoher Affinität zu dem M. jannaschii Nep1p identifiziert, die auf eine Bindung des Proteins bei Helix 35 der 16S rRNA schließen lassen. Aufgrund der hohen Konservierung dieser rRNA-Region ist eine Bindung des Hefeproteins an die 18S rRNA-Schleife von Nukleotid 1189-1196 sehr wahrscheinlich. Da Nep1p eine große Ähnlichkeit zu Proteinen der SPOUTFamilie von Methyltransferasen aufweist, war von einer rRNA-Methylierung im Verlauf der Ribosomenbiogenese als katalytische Funktion des Proteins auszugehen. Aus verschiedenen Drei-Hybrid-Experimenten zur RNA-Bindungungsspezifität ergab sich als mögliche Reaktion die N1-Methylierung des Nukleotids 1-Methyl-3-(3-Amino-3-Carboxypropyl)-Pseudouridin (m1acp3Y) 1191 der 18S rRNA. Durch eine spezifische radioaktive Markierung der acp-Gruppe konnte gezeigt werden, dass Nep1p keinen Einfluss auf die spätere Aminocarboxypropylmodifizierung hat. Diese findet auch bei einer Deletion der snoRNA35 statt, also auch an einem Uridin, und ist unabhängig von dem cytoplasmatischen Protein Tma20p. In RP-HPLC-Experimenten konnte nachgewiesen werden, dass die 18S rRNA einer Dnep1Dnop6-Doppelmutante ein Aminocarboxypropyl-modifiziertes Nukleosid enthält, dass sich in seinem Retensionsverhalten von dem m1acp3Y eines Wildtyps unterscheidet. Bei dem in diesem Stamm detektierten acp-modifizierten Nukleosid handelt es sich vermutlich um ein nicht-methyliertes acpY, was eine Funktion von Nep1p als N1-Methyltransferase des Nukleotids Y1191 der 18S rRNA höchst wahrscheinlich macht. Diese katalytische Funktion konnte in Zusammenarbeit mit Prof. Wöhnert auch für das M. jannaschii Nep1p gezeigt werden. Dass sowohl eine snr35- Deletion als auch eine 18S rRNA-Mutation des Nukleotids 1191 nicht letal sind, machte deutlich, dass die N1-Methylierung nicht die essentielle Funktion von Nep1p darstellen kann. Weiterhin konnte nachgewiesen werden, dass die Suppression der nep1-1ts-Mutante durch S-Adenosylmethionin nicht auf der Unterstützung der Methyltransferase-Aktivität des Proteins, sondern vermutlich eher auf einer generellen Stabilisierung des temperatursensitiven Proteins beruht. Sowohl im Hefe-Nep1p als auch im humanen Homolog wurden durch biochemische und genetische Experimente mehrere Phänotypen der Bowen-Conradi-Mutation (Aspartat 90 zu Glycin in ScNep1p) nachgewiesen. Diese lassen auf eine Aggregation des mutierten Proteins sowie eine dadurch bedingte Fehllokalisation innerhalb der Zelle schließen. Zusätzlich ist aber auch ein RNA-Bindungsdefekt durch den Aminosäureaustausch wahrscheinlich. Nichtsdestotrotz liegt offensichtlich ausreichend Nep1p-Protein vor, dass seine essentielle Funktion erfüllen kann, da die Mutation selbst zu keinem Wachstumsphänotyp führt. Erst bei einer partiellen Translationsrepression des mutierten Proteins unter Verwendung des artifiziellen Tetrazyklin-Aptamer-Systems ist ein verlangsamtes Wachstum von Hefezellen zu beobachten, was dieses System geeignet zur Analyse von möglichen Therapeutika macht.
A new lizard genus and species is descnbed from the World Hentage Messel Pit Fossil Site (Middle Eocene: basal Lutetian, Geiseltalium) near Darmstadt, Germany. It is placed within the Scincoidea (Scincidae, Cordyliformes + Paramacellodidae), but no attribution to a distinct family can be made. Its phylogenetic position is problematic as it probably represents the descendant of a stem group taxon of the Scincoidea or an interesting „link" between Scincidae and Cordyliformes. Extensive phylogenetic analyses using numerical taxonomy are undertaken. Unaltered and modified matnces from different authors are analysed in order to generate a vanety of phylogenetic hypotheses. These are discussed with the results of comparative anatomy, the latter having a higher impact on the final classification. Key characters of the new taxon are as follows: The dorsal aspect of the skull is covered with a characteristic osteodermal pattem, the jugal has a huge posterior process, the retroaricular process and teeth show a typical scincoid morphology. Six well preserved specimens can safely be attributed to the new species which is not yet known from any other fossil site. The very long prehensile tail of the new genus and species, its curved claws, limb proportions and missing body osteodems strongly suggest an arboreal mode of life. The long tail could secure support by grabbing branches, but probably was too fragile to hold the entire body weight. The locomotion and microhabitat exploitation of the new taxon can best be compared to the arboreal emerald monitors Varanus prasinus and Varanus beccarii with their long tails and strongly curved claws. In three of the fossil specimens intestinal contents are still preserved. They contain plant remains and a few fiagments of insect cuticula. Due to its Cover of osteoderms, the skull was largely akinetic, suggesting the new taxon not to be an effective and agile predator, but a rather large herbivore and occasional insectivore, which was probably capable of strenuous biting. Its presence in the Lower Middle Eocene of Messel neither supports nor contradicts the current opinions of Scincoid Paleobiogeography.
