L’école de Frankfort abandonne progressivement les thèses de la philosophie de l’Histoire, dont l’acceptation fournissait une espèce de fondement pour la théorie critique, dans la mesure où l’on pouvait croire aux contenus émancipateurs de la raison inclus au sein même des idées de la société bourgeoise. Nous essayerons de montrer cequi est oublié dans les thèses de la philosophie de l’Histoire à partir de la dialectique du maître et de l’esclave de Hegel, et donc, pourquoi de telles thèses se sont montrées insuffisantes. Enfin, nous chercherons à démontrer que la figure théorique opposée aux thèses de la philosophie de l’Histoire se retrouve dans Über den Begriff der Geschichte de Benjamin. De plus, la récupération d’un tel fondement oublié dans les thèses de la philosophie de l’Histoire détermine, vraisemblablement, la considération morale de la théorie critique habermasienne.
Mit der vorliegenden Arbeit wurde die Nutzbarkeit morphologischer und anatomischer Merkmale aus den Bereichen des Fruchtknotens und der Samenanlangen für die Systematik und Taxonomie der Bromelioideae (Bromeliaceae) untersucht. Hierzu wurden 30 Merkmale im Bereich des Fruchtknotens und der Samenanlagen definiert und anhand anatomischer Schnittpräparate die Verteilung der Merkmalsausprägungen an 102 Arten aus 28 (von 32) Gattungen der Unterfamilie Bromelioideae sowie zwei Vertretern der Unterfamilie Pitcairnioideae als Vergleichgruppe ermittelt. Allein 41 Taxa entstammten der größten und als polyphyletisch anzunehmenden Gattung Aechmea mit Vertretern aus allen sieben Untergattungen. Die Auswertung der Merkmalsverteilung erfolgte einerseits im Hinblick auf eine funktionale Deutung beobachteter Kopplungen bestimmter Strukturen im ökologischen Kontext und zum anderen unter taxonomischen Gesichtspunkten im Hinblick auf Beantwortung der Fragen, wieweit derzeitige Gattungsumgrenzungen von den hier untersuchten Merkmalen unterstützt werden, bzw. wie weit Anregungen zur Abänderung bestehender Konzepte abgeleitet werden können, welche Beziehungen zwischen den Untergattungen von Aechmea und anderen Gattungen der Bromelioideae bestehen und wodurch sich basale Linien der Bromelioideae von den abgeleiteten Formen der „Eu- Bromelioideae“ im Sinne von SCHULTE (2007) unterscheiden lassen. Um diese Fragen zu beantworten, wurden zwei unterschiedliche Ansätze kombiniert. Die Verteilung der Merkmalszustände wurde einerseits auf Topologien geplottet, die aus publizierte Phylogenien mit genetischen Merkmalen als Datenquelle beruhen. Desweiteren wurden neue Parsimonieanalysen durchgeführt a) auf der Grundlage einer morphologischen Datenmatrix mit den 30 selbst erhobenen Merkmalen aus dem Fruchtknoten- und Samenbereich, b) einer weiteren Matrix, in die fünf weitere selbst erhobene Merkmale aus dem floralen Bereich eingehen und schließlich c) einem Datensatz, in den zusätzlich acht aus der Literatur entnommene Angaben zur Morphologie und Ökologie der untersuchten Pflanzen eingingen. Die aus den molekularen Analysen von SCHULTE et al. (2005) hervorgehende Gliederung der Unterfamilie in eine paraphyletische Gruppe von basalen Linien und der von SCHULTE (2007) als „Eu-Bromelioideae“ bezeichneten monophyletischen Gruppe abgeleiteterer Gattungen findet eine deutliche Entsprechung in Merkmalen des Fruchtknotenbereichs und der Samenanlagen. Die untersuchten Vertreter der basalen Linien besitzen alle ± rechtwinklig von der Fruchtknotenachse abspreizende Samenanlagen, diese sind auf mehr als 70% der Fruchtknotenachse verteilt und die Mikropyle ist stets relativ lang ausgebildet (>100 micro m). Eine der Kerninnovationen der Eu-Bromelioiden scheint die Entwicklung des chalazalen Samenanhängsels als Hilfsinstrument bei der Besiedlung glatter Oberflächen gewesen zu sein. Diese Struktur findet sich nur bei Eu-Bromelioideae und ist stets bereits an der Samenanlage vorgebildet. Sekundär scheint dieses chalazale Anhängsel innerhalb der Eu-Bromelioideae allerdings auch immer wieder verloren gegangen zu sein. Die Umgrenzung der meisten Gattungen sowohl der basalen Linien als auch der Eu-Bromelioideae konnten in ihrer jetzigen Form anhand der Merkmale des Fruchtknotenbereichs und der Samenanlagen nachvollzogen werden. Eine Ausnahme hiervon stellt lediglich die hochgradig polyphyletische Gattung Aechmea s.l. dar. Für keine der untersuchten Gattungen konnten synapomorphe Merkmalszustände erkannt werden, vielmehr gibt es jeweils gattungsspezifische Kombinationen von Merkmalen. Die Ausprägungen der Einzelmerkmale dagegen sind stets auch in anderen Verwandschaftsgruppen zu beobachten. Die hier untersuchten Merkmale unterstreichen das für die Bromelioideae seit langem bekannte Phänomen eines extrem hohen Homoplasiegrades in nahezu allen morphologischen Strukturen. Selbst für den Fruchtknoten lassen sich klare Abhängigkeiten der Merkmalsausprägungen von ökologischen Selektionsfaktoren erkennen. Insbesondere scheinen funktionale Notwendigkeiten im Kontext der Nektarsekretion und der Bestäubungsökologie für die Merkmalsausprägung des Fruchtknotens eine wichtige Rolle zu spielen. Die Samenanlage ist zwar zum Zeitpunkt der Anthese noch nicht direkten Selektionsdrücken ausgesetzt, ihre Merkmale sind aber nur dann zu verstehen, wenn die Funktion hier bereits angelegter Strukturen (wie z.B. der chalazalen Samenanhängsel) im Kontext mit den reifen Samen gedeutet wird.
During a 4-week run in October–November 2006, a pilot experiment was performed at the CERN Proton Synchrotron in preparation for the CLOUD1 experiment, whose aim is to study the possible influence of cosmic rays on clouds. The purpose of the pilot experiment was firstly to carry out exploratory measurements of the effect of ionising particle radiation on aerosol formation from trace H2SO4 vapour and secondly to provide technical input for the CLOUD design. A total of 44 nucleation bursts were produced and recorded, with formation rates of particles above the 3 nm detection threshold of between 0.1 and 100 cm−3s−1, and growth rates between 2 and 37 nm h−1. The corresponding H2SO4 concentrations were typically around 106 cm−3 or less. The experimentally-measured formation rates and H2SO4 concentrations are comparable to those found in the atmosphere, supporting the idea that sulphuric acid is involved in the nucleation of atmospheric aerosols. However, sulphuric acid alone is not able to explain the observed rapid growth rates, which suggests the presence of additional trace vapours in the aerosol chamber, whose identity is unknown. By analysing the charged fraction, a few of the aerosol bursts appear to have a contribution from ion-induced nucleation and ion-ion recombination to form neutral clusters. Some indications were also found for the accelerator beam timing and intensity to influence the aerosol particle formation rate at the highest experimental SO2 concentrations of 6 ppb, although none was found at lower concentrations. Overall, the exploratory measurements provide suggestive evidence for ion-induced nucleation or ion-ion recombination as sources of aerosol particles. However in order to quantify the conditions under which ion processes become significant, improvements are needed in controlling the experimental variables and in the reproducibility of the experiments. Finally, concerning technical aspects, the most important lessons for the CLOUD design include the stringent requirement of internal cleanliness of the aerosol chamber, as well as maintenance of extremely stable temperatures (variations below 0.1°C).
A taxonomic overview for the fritillary genus Speyeria Scudder (= Argynnis Fabricius) and its placement within the Heliconiinae (Nymphalidae) is presented. Taxonomic accounts, type images, and relevant literature for the 25 subspecies within the Speyeria atlantis (Edwards) and Speyeria hesperis (Edwards) complexes and the 16 nominate Speyeria species are included. Errors in nomenclature are identified, taxonomic, life history, and distributional information are updated, and type locality information is discussed. Images of primary type specimens for all 16 Speyeria species and the 25 subspecies in the atlantis - hesperis complexes appear together in color here for the first time. One new combination of a species-subspecies is created: Speyeria hesperis hanseni Emmel, Emmel, and Matoon, 1998, new combination, which was previously Speyeria atlantis hanseni Emmel, Emmel, and Matoon.

Este artigo enfoca a apropriação que Horkheimer e Adorno fazem da doutrina kantiana do esquematismo no sentido de apontar para o procedimento – característico da indústria cultural – de usurpar de seus consumidores a capacidade de “esquematizar” (referir intuições a conceitos) por si próprios. Considerando-se que os autores não dão outras indicações sobre como se dá esse processo em relação aos meios de massa, o texto procura explicar como a própria percepção em geral é atingida pela “usurpação do esquematismo” a partir de colocações do capítulo da Dialética do esclarecimento sobre o antisemitismo. Essas colocações são complementadas – e também comparadas – com as de Hans Lenk no seu livro O pensamento e o seu conteúdo.
Regulation des matrizellulären Proteins SMOC-1 durch Zytokine und Stickoxid in Rattenmesangiumzellen

Zytokine stimulieren in Mesangiumzellen die Produktion und die Freisetzung großer Mengen entzündlicher Mediatoren. In dieser Arbeit wurden mit Hilfe der RAP-PCR („RNA arbitrarily primed polymerase chain reaction“), einer auf mRNA basierenden „Differential display“-Methode, die Effekte von Interleukin-1β (IL-1β) auf das Genexpressionsmuster in glomerulären Rattenmesangiumzellen untersucht. Dabei wurde das matrizelluläre Glykoprotein „Secreted modular calcium-binding protein-1“ (SMOC-1) identifiziert, welches in Mesangiumzellen durch IL-1β herunterreguliert wird. SMOC-1 wird von verschiedenen Zelltypen exprimiert und sezerniert, doch seine biologische Funktion konnte bisher nicht aufgedeckt werden. Weitere Experimente bestätigten, dass die mRNA- und Proteinexpression von SMOC-1 durch proinflammatorische Zytokine, wie IL-1β und Tumornekrosefaktor-α (TNF-α) herunterreguliert wird. Dieser Effekt wird zu einem großen Teil durch die endogene Freisetzung von Stickoxid (NO) aufgrund der Aktivität der induzierbaren NO-Synthase (iNOS) und der nachfolgenden Aktivierung der löslichen Guanylatzyklase (sGC) vermittelt. Außerdem tragen auch reaktive Sauerstoffver-bindungen (ROS), deren Bildung durch die Zytokine verstärkt wird, zur Herunter-regulierung der SMOC-1-Expression bei. Durch In-situ-Hybridisierungsexperimente konnte ferner gezeigt werden, dass die Hemmung der NO-Synthese durch den spezifischen iNOS-Inhibitor L-NIL in einem Rattenmodell der anti-Thy1.1-Glomerulonephritis die SMOC-1-Expression deutlich erhöhte. Dies belegt somit auch in vivo die biologische Relevanz von NO in der Modulierung der SMOC-1-Expression. Die funktionelle Rolle von SMOC-1 in Mesangiumzellen wurde durch die Hemmung der SMOC-1-Expression mit Hilfe einer spezifischen siRNA untersucht. Dabei zeigte sich, dass die Hemmung von SMOC-1 eine deutliche Inhibierung der mRNA-Expression von „Transforming growth factor β1“ (TGF-β1) sowie dessen Gesamtproteinspiegel zur Folge hat. Auch die Aktivität von TGF-β1 wurde reduziert, wie anhand der verringerten Spiegel an aktivem TGF-β1-Protein und der verringerten mRNA-Expression bekannter TGF-β-regulierter Gene, wie „Connective tissue growth factor“ (CTGF), „Plasminogen activator inhibitor-1“ (PAI-1) und Biglykan gezeigt wurde. Diese Ergebnisse deuten auf eine Rolle von SMOC-1 bei der Modulierung des TGF-β1-Signalwegs hin. Zusammenfassend betrachtet scheint NO die SMOC-1-Expression in der akuten glomerulären Entzündung zu vermindern und dadurch die TGF-β-getriebenen profibrotischen Signalprozesse zu limitieren. Der zweite Aspekt dieser Arbeit befasst sich mit der Rolle von Peroxisomen-Proliferator-aktivierten Rezeptoren (PPAR) in der IL-1β-vermittelten Expression der induzierbaren NO-Synthase. PPARα-Aktivatoren steigern in Mesangiumzellen die IL-1β-induzierte Aktivität der iNOS, während die Hemmung von PPARα durch spezifische Inhibitoren oder siRNA die iNOS-Expression/-Aktivität deutlich reduziert. Die Ergebnisse von Promotor-studien zeigten die essentielle Rolle einer möglichen PPAR-Bindestelle im iNOS-Promotor. IL-1β scheint die Bildung eines endogenen PPAR-Liganden zu induzieren, wodurch die Bindung von PPAR-Proteinkomplexen an das regulatorische DNA-Element im iNOS-Promotor verstärkt und dessen Aktivität gesteigert wird. Daneben scheinen jedoch auch Nebeneffekte der PPAR-Aktivatoren, wie die Freisetzung von ROS, zur synergistischen Wirkung auf die IL-1β-induzierte iNOS-Expression beizutragen. Die Wirkung von dualen PPARα/γ-Aktivatoren auf entzündliche Prozesse wird seit längerem diskutiert, daher wurden die biologischen Effekte von neu synthetisierten möglichen dualen PPARα/γ-Aktivatoren auf Entzündungsparameter, wie z. B. die iNOS-Expression, in Mesangiumzellen untersucht. Alle untersuchten Aktivatoren steigerten in der Form von Esterverbindungen die IL-1β-induzierte Expression der iNOS sowie der sekretorischen Phospholipase A2 (sPLA2), während die entsprechenden freien Säuren wenig Effekte zeigten. Diese proinflammatorische Wirkung scheint jedoch weniger auf einer Aktivierung des PPAR-Rezeptors zu beruhen als auf der Freisetzung von ROS, die durch die Aktivatoren teilweise deutlich erhöht wurde. Weitere Experimente zur Charakterisierung der PPAR-spezifischen Wirkung der Aktivatoren sowie zur optimalen Wirkkonzentration sind nötig, bevor der Effekt dieser Aktivatoren auf die inflammatorische Genexpression genau bewertet werden kann.
Pollen-based climate reconstructions were performed on two high-resolution pollen marines cores from the Alboran and Aegean Seas in order to unravel the climatic variability in the coastal settings of the Mediterranean region between 15 000 and 4000 years BP (the Lateglacial, and early to mid-Holocene). The quantitative climate reconstructions for the Alboran and Aegean Sea records focus mainly on the reconstruction of the seasonality changes (temperatures and precipitation), a crucial parameter in the Mediterranean region. This study is based on a multi-method approach comprising 3 methods: the Modern Analogues Technique (MAT), the recent Non-Metric Multidimensional Scaling/Generalized Additive Model method (NMDS/GAM) and Partial Least Squares regression (PLS). The climate signal inferred from this comparative approach confirms that cold and dry conditions prevailed in the Mediterranean region during the Oldest and Younger Dryas periods, while temperate conditions prevailed during the Bølling/Allerød and the Holocene. Our records suggest a West/East gradient of decreasing precipitation across the Mediterranean region during the cooler Late-glacial and early Holocene periods, similar to present-day conditions. Winter precipitation was highest during warm intervals and lowest during cooling phases. Several short-lived cool intervals (i.e. Older Dryas, another oscillation after this one (GI-1c2), Gerzensee/Preboreal Oscillations, 8.2 ka event, Bond events) connected to the North Atlantic climate system are documented in the Alboran and Aegean Sea records indicating that the climate oscillations associated with the successive steps of the deglaciation in the North Atlantic area occurred in both the western and eastern Mediterranean regions. This observation confirms the presence of strong climatic linkages between the North Atlantic and Mediterranean regions.

The Invertebrate section of the Museum of Zoology QCAZ at the Pontifical Catholic University of Ecuador in Quito maintains nearly two million curated specimens, and comprises Ecuador's largest collection of native taxa. We review 1902 type specimens from 6 subspecies and 320 species in 121 genera and 42 families, currently kept in the Museum. The list includes 116 holotypes, 10 allotypes, 1774 paratypes and 2 neoparatypes. The collection of type specimens is particularly strong in the Coleoptera (family Carabidae and Staphylinidae) and Hymenoptera. However, other insect orders such as Diptera and Lepidoptera and non-insect arthropods such as Acari, Aranea and Scorpiones, are moderately represented in the collection. This report provides original data from labels of every type specimen record. An analysis of the geographic distribution of type localities showed that collection sites are clustered geographically with most of them found. towards the northern region of Ecuador, in Pichincha, Cotopaxi and Napo provinces. Sites are mainly located in highly accessible areas near highways and towns. Localities with a high number of type species include the cloud forest reserve Bosque Integral Otonga and Parque Nacional Yasunf in the Amazon rainforest near PUCE's Yasuni Scientific Station. Type localities are not well represented in the Ecuadorian National System of Protected Areas. Future fieldwork Sllould include. localities in the southern region of Ecuador but also target less accessible areas not located near highways or towns. We discuss the value of the collection as a source of information for conservation and biodiversity policies in Ecuador.
Corporate borrowers care about the overall riskiness of a bank’s operations as their continued access to credit may rely on the bank’s ability to roll over loans or to expand existing credit facilities. As we show, a key implication of this observation is that increasing competition among banks should have an asymmetric impact on banks’ incentives to take on risk: Banks that are already riskier will take on yet more risk, while their safer rivals will become even more prudent. Our results offer new guidance for bank supervision in an increasingly competitive environment and may help to explain existing, ambiguous findings on the relationship between competition and risk-taking in banking. Furthermore, our results stress the beneficial role that competition can have for financial stability as it turns a bank’s "prudence" into an important competitive advantage.
We first report from the 5th Dry Grassland Meeting held from 28th to 30st of August 2008 in Kiel, Germany. Then we take stock of the achievements of the German Arbeitsgruppe Trockenrasen and the international Working Group on Dry Grasslands in the Nordic and Baltic Region towards establishment of vegetation databases of dry grasslands in these study regions. At the conference, the European Dry Grassland Group (EDGG) was founded as an international platform of dry grassland researchers and conservationists. As networking tools it provides a homepage on the internet, publishes a quarterly electronical bulletin, and will organise the future European Dry Grassland Meetings. In the last part of this contribution, we give a short introduction to the five articles of this Special Feature. Four of them make a major contribution to supra-national classification of Koelerio-Corynephoretea and Festuco- Brometea communities, respectively, by providing syntaxonomic overviews based on comprehensive data from eastern European countries for which only few data have been available until now. The other major focus of the Feature papers is on degradation and restoration of various types of dry grasslands.
Der vorliegende Artikel befasst sich mit dem aktuellen Thema des Anglizismengebrauchs in der deutschen Sprache. Er konzentriert sich ausschließlich auf den Anglizismengebrauch in der Zeitschrift Marketing Journal. Verglichen werden zwei Ausgaben, und zwar das zweite erschienene Exemplar aus dem Jahr 1969 und das erste Heft aus dem Jahr 2007. Es wird herausgestellt, welche qualitativen, quantitativen und lexikalischen Veränderungen der Ang-lizismengebrauch im Laufe von 39 Jahren erfahren hat. Darüber hinaus wird auf die Schreibweise der Anglizismen näher eingegangen.

Wading the Tide

Wading the Tide is an expression of profound emotions touching on a wide range of issues-personal and political-from the birth of the Cameroon nation, her political meandering, until the state of emergency declared on the North West Province in 1992. Accordingly, Doh complains, ridicules, and pays tribute, even as he instructs and guides on timeless matters of life, all in an effort to draw attention to his country's gradual, downward spiral into anomy.

ORIKI'BADAN, is an entertaining, revealing, and equally didactic poem in which Doh, through an enchanting metaphorical backdrop, recaptures a memorable era-rich, diverse, challenging, yet gratifying-in the life of a distinguished institution-the University of Ibadan, Nigeria. Characteristically bitter about those in power and the socio-political state of affairs on the African continent, this is a rare shot of Doh paying glaring tribute to his alma mater along with the distinguished faculty and student body that gave Ibadan its character during his days there as a student.
Characteristically, Africans in any Western country are asked so many different questions about 'Africa,' as Westerners love to refer to the many countries that make up that huge continent, as if Africa were a single nation state. So one begins wondering why it is that Africans, on the other hand, do not refer to individual European countries as 'Europe' simply, then the trends and consequences of stereotyping begin setting in just as one is getting used to being asked if Africa has a president, or if one can say something in African. It is some of these questions that Emmanuel Fru Doh has collected over the years and has attempted answering them in an effort to shed some light on a continent that is in many ways like the rest of the world, when not better, but which so many love to paint as dark, backward, chaotic, and pathetic.
Joachim Wittstock presents three outstanding personalities from the Transylvanian Saxon and Romanian past – the theologian and reformer Johannes Honterus, the Saxon count and Royal Judge Johann Zabanius Sachs von Harteneck, and the Walachian prince Constantin Brâncovean – from very new, innovative and surprising points of view by combining fact and fiction in a subtle way. A special feature of Joachim Wittstock’s approach is the matching of form and content, naturally with varying emphasis on language construction or topic composition in the three pieces of prosa. In terms of the content, the author combines a circular perspective with a parallel one, thus creating a distinct up- and- down pattern. Linguistically, Joachim Wittstock uses cultivated everyday speech laced with telling word play, proverbs and idioms, foreign and archaic words, to ev oke the historic atmosphere of the past and making subtle political connotations. Vom 07.06. bis zum 28.08.1997 wurden in der Malkachan-Bucht (Region Magadan, Ochotskisches Meer, Russischer Ferner Osten) bei Pentadenzählungen die rastenden Limikolenarten erfasst. Die Erfassungen erfolgten im Rahmen einer Expedition der Universitäten Osnabrück und Jena in Kooperation mit dem Institute for Biological Problems of the North (Magadan). Insgesamt wurde mit 33 Limikolenarten, darunter 14-16 lokalen Brutvogelarten, eine sehr hohe Biodiversität festgestellt. Die höchsten Rastmaxima wurden beim Regenbrachvogel (Numenius phaeopus variegatus) (>2.000 Ind.) und beim Odinshühnchen (Phalaropus lobatus) (1.000 Ind.) erfasst. Für beide Arten hat die Malkachan-Bucht damit internationale Bedeutung als Rastgebiet, da mehr als 1 % der Flyway-Population rasten. Besonders bemerkenswert ist die Revierfeststellung und die Beobachtung eines gewissen Jungvogel-Durchzugs des weltweit gefährdeten Tüpfelgrünschenkels (Tringa gutiffer) in etwa 800 km Entfernung vom bisher bekannten Brutgebiet. Neben dem Regenbrachvogel weist das Gebiet auch höhere Rastzahlen anderer (mittel)großer Limikolenarten wie Uferschnepfe (Limosa limosa melanuroides) (383 Ind., auch Brutvogel), Pfuhlschnepfe (Limosa lapponica baueri) (262 Ind.) und Isabellbrachvogel (Numenius madagascariensis) (250 Ind., auch Brutvogel) auf. Demgegenüber hat die Malkachan-Bucht – zumindest im Jahr 1997 – keine so hohe Bedeutung für rastende Strandläuferarten aufgewiesen. Die Schutzwürdigkeit des Gebiets und seine internationale Bedeutung konnte auch mit Hilfe der Limikolen-Pentadenzählungen im Rahmen der Expedition unter Beweis gestellt werden.
Um eine kardiale Ursache für den Zustand klinisch instabiler Patienten auszuschließen, gilt der Einsatz einer fokussierten Echokardiographie als aussagekräftige orientierende Untersuchungsmaßnahme. Auch während einer kardiopulmonalen Reanimation kann eine ALS-konforme Echokardiographie eingesetzt werden. Dafür benötigt der Notfallmediziner ein standardisiertes Training, welches spezielle Schwerpunkte beinhaltet. Dabei muss neben der Vermittlung von theoretischen Kenntnissen und der praktischen Anwendung der Echokardiographie auch besonders auf das Trainieren von visuell perzeptiven Fähigkeiten geachtet werden, da ein schnelles Erkennen von spezifischen Diagnosen elementar für die Prognose klinisch instabiler Patienten ist. Diese Arbeit befasst sich mit der Entwicklung und Anwendung eines computerbasierten Testverfahrens, der dem Erfassen des Lernerfolges im Rahmen des Stufe-1-Training für Notfallechokardiographie. Dabei wird vor allem in Hinblick auf die schnelle Identifikation von Blickdiagnosen geachtet, indem eine Zeitmessung erfolgte. Bei der Entwicklung wurden, basierend auf die aktuellen Empfehlungen für den Einsatz von Echokardiographie in der Intensiv-und Notfallmedizin, die 10 wichtigsten Echokardiographiediagnosen als Filmsequenzen dargestellt. Dazu gehören Befunde wie die PEA, der Perikarderguss, Asystolie, reduzierte Pumpfunktion und Rechtsherzbelastung. Zu jeder Filmsequenz, die je nach Version 5 Sekunden oder bis zu 60 Sekunden dauern, wurde eine Freitexteingabe gefordert und eine Multiple-Choice-Aufgabe gestellt. Ergebnisse wurden im Rahmen von drei Kurskonzepten für Stufe-1-Noallechokardiographie gesammelt. Damit einhergehend wurde die Validität des Simulationstests Notfallechokardiographie bewiesen. Bei der Betrachtung der Ergebnisse zeigt sich, dass es möglich ist, mit dem Test den Wissensstand der Teilnehmer aufzuzeigen. Dabei wurde deutlich, dass Befunde wie die PEA, der Perikarderguss und die hochgradig eingeschränkte Pumpfunktion von der Mehrzahl der Teilnehmer korrekt in Freitexteingabe und Multiple-Choice-Aufgabe benannt werden konnten. Schwächen im Erkennen bestimmter Befunde, allen voran des Normalbefundes, wurden anhand der Testanwendung aufgedeckt. Somit kann der Simulationstest Notfallechokardiographie zu einer Überarbeitung der Kursinhalte beitragen. Bei einer Erfassung der Zeit, die für die Identifikation der Befunde benötigt wird, wird ersichtlich, dass definierte Diagnosen innerhalb von 10 Sekunden erkannt werden können. Vor allem der Befund der PEA wurde nach dem Kurs schnell erkannt. Andere Diagnosen, wie das normale Herz, zeigen, dass es nach einem Kursprogramm weiterhin schwierig sein kann, bestimmte Befunde innerhalb von 10 Sekunden zu stellen. Die gewonnenen Ergebnisse werden durch die erhobenen Daten bei der Anwendung des Simulationstest Notfallechokardiographie in der 5-Sekunden-Version unterstrichen. Spannend ist, dass im Rahmen eines spezialisierten Trainings kein Unterschied in den Ergebnissen festzustellen war. Es spielte dabei keine Rolle, ob die Teilnehmer die Filmsequenzen 5 Sekunden oder 60 Sekunden sehen konnten. Aus diesen Ergebnissen lässt sich der Schluss ziehen, dass nach einem achtstündigen Kurs für ALS-konforme Echokardiographie definierte Blickdiagnosen dass definierte Befunde innerhalb 5 Sekunden erkannt werden können.
Kaon and pion production in centrality selected minimum bias Pb+Pb collisions at 40 and 158A GeV

Results on charged kaon and negatively charged pion production and spectra for centrality selected Pb+Pb mininimum bias events at 40 and 158A GeV have been presented in this thesis. All analysis are based on data taken by the NA49 experiment at the accelerator Super Proton Synchrotron (SPS) at the European Organization for Nuclear Research (CERN) in Geneva, Switzerland. The kaon results are based on an analysis of the mean energy loss <dE/dx> of the charged particles traversing the detector gas of the time projection chambers (TPCs). The pion results are from an analysis of all negatively charged particles h- corrected for contributions from particle decays and secondary interactions. For the dE/dx analysis of charged kaons, main TPC tracks with a total momentum between 4 and 50 GeV have been analyzed in logarithmic momentum log(p) and transverse momentum pt bins. The resulting dE/dx spectra have been fitted by the sum of 5 Gaussians, one for each main particle type (electrons, pions, kaons, protons, deuterons). The amplitude of the Gaussian used for the kaon part of the spectra has been corrected for efficiency and acceptance and the binning has been transformed to rapidity y and transverse momentum pt bins. The multiplicity dN/dy of the single rapidity bins has been derived by summing the measured range of the transverse momentum spectra and an extrapolation to full coverage with a single exponential function fitted to the measured range. The results have been combined with the mid-rapidity measurements from the time-of-flight detectors and a double Gaussian fit to the dN/dy spectra has been used for extrapolation to rapidity outside of the acceptance of the dE/dx analysis. For the h- analysis of negatively charged pions, all negatively charged tracks have been analyzed. The background from secondary reactions, particle decays, and gamma-conversions has been corrected with the VENUS event generator. The results were also corrected for efficiency and acceptance and the pt spectra were analyzed and extrapolated where necessary to derive the mean yield per rapidity bin dN/dy. The mean multiplicity <pi-> has been derived by summing up the measured dN/dy and extrapolating the rapidity spectrum with a double Gaussian fit to 4pi coverage. The results have been discussed in detail and compared to various model calculations. Microscopical models like URQMD and HSD do not describe the full complexity of Pb+Pb collisions. Especially the production of the positively charged kaons, which carry the major part of strange quarks, cannot be consistently reproduced by the model calculations. Centrality selected minimum bias Pb+Pb collisions can be described as a mixture of a high-density region of multiply colliding nucleons (core) and practically independent nucleon-nucleon collisions (corona). This leads to a smooth evolution from peripheral to central collisions. A more detailed approach derives the ensemble volume from a percolation of elementary clusters. In the percolation model all clusters are formed from coalescing strings that are assumed to decay statistically with the volume dependence of canonical strangeness suppression. The percolation model describes the measured data for top SPS and RHIC energies. At 40A GeV, the system size dependence of the relative strangeness production starts to evolve from the saturation seen at higher energies from peripheral events onwards towards a linear dependence at SIS and AGS. This change of the dependence on system size occurs in the energy region of the observed maximum of the K+ to pi ratio for central Pb+Pb collisions. Future measurements with heavy ion beam energies around this maximum at RHIC and FAIR as well as the upgraded NA49 successor experiment NA61 will further improve our understanding of quark matter and its reflection in modern heavy ion physics and theories.
Many questions regarding gastropod phylogeny have not yet been answered like the molecular confirmation of the Heterobranchia concept based on morphological studies from Haszprunar (1985a; 1988). This taxon contains the “Lower Heterobranchia” with several “primitive” or “basal” members) and the Euthyneura (with the Opisthobranchia and Pulmonata). Phylogenetic relationships of subgroups within the Heterobranchia have not been satisfactorily resolved and monophyly of some taxa within the Heterobranchia (e.g. Opisthobranchia) is questionable. Moreover, most of the “Lower Heterobranchia” have not been included in former molecular studies. In order to resolve phylogenetic relationships within the Heterobranchia, I pursued a molecular systematic approach by sequencing and analysing a variety of genetic markers (including nuclear 28S rDNA + 18S rDNA and mitochondrial 16S rDNA + COI sequences). Maximum likelihood as well as Bayesian inference methods were used for phylogenetic reconstruction. The data were investigated a priori to tree reconstruction in order to find the most appropriate dataset for reconstructing heterobranch phylogeny. A variety of statistical tests (like Chi-Square-Test or Relative-Rate-Test) were applied and the substitution saturation was measured. The Relative-Rate-Test revealed the highest evolution rates within the “Lower Heterobranchia” (Omalogyra sp., Omalogyra fusca, Murchisonella sp., Ebala sp. and Architectonica perspectiva) and Opisthobranchia (Hyalocylis striata). Furthermore, many of the nucleotide positions show a high degree of substitution saturation. Additionally, bipartitions (splits) in the alignment were examined and visualized by split network analyses to estimate data quality. A high level of conflict indicated by many parallel edges of the same lengths could be observed in the neighbournet graphs. Moreover, several taxa with long terminal branches could be identified in all three datasets belonging to the Vetigastropoda, Caenogastropoda, “Lower Heterobranchia” or Opisthobranchia (Nudipleura). All phylogenetic analyses revealed a monophyletic Heterobranchia. Within the Heterobranchia several well supported clades could be resolved. However, the traditional classification based on morphological data could not be confirmed due to paraphyletic Euthyneura (because of the inclusion of the Pyramidellidae and Glacidorboidea) as well as paraphyletic Pulmonata and polyphyletic Opisthobranchia. Based on the phylogenetic inferred evolutionary trends regarding habitat colonisation or character complexes could be deduced. A case study was conducted in order to estimate divergence ages using a “relaxed” molecular clock approach with fossils as minimum age constraints. However, due to large 95% confidence intervals a precise dating of the nodes was not possible. Hence, the results are considered as preliminary. To test the plausibility of the newly obtained hypotheses, the results were evaluated a posteriori using a hypothesis test and secondary structures of the complete 18S rRNA and 28S rRNA. Secondary structure motifs were found within domain 43 and E23 2 &5 of the 18S rRNA as well as within domain E11 and G5_1 of the 28S rRNA, which contain phylogenetic signals to support various groups within the Heterobranchia. In addition, taxon specific motifs were found separating the Vetigastropoda from the Caenogastropoda and Heterobranchia, indicating a possible application of the secondary structure of 18S rRNA and 28S rRNA to reveal phylogenetic relationships at higher taxonomic levels such as Gastropoda or even Mollusca. The utility of the newly invented software RNAsalsa for the reconstruction of secondary structures was tested. The obtained structures were used to adjust evolutionary models specific to rRNA stem (paired basepairs) and loop (unpaired basepairs) regions with the intention of improving phylogenetic results. This approach proved unsuccessful. This molecular phylogenetic investigation provides the most comprehensive molecular study of Heterobranchia relationships to date. Substantial insights into the evolution and phylogeny of this enigmatic taxon have been gained.
Die vorliegende Arbeit befasst sich mit den nichtlinearen Zusammenhangsstrukturen in Form von Moderator- und quadratischen Effekten, die einen aktuellen Gegenstand der Forschung darstellen. In der Arbeit wurden zunächst die theoretischen Grundlagen von Strukturgleichungsmodellen sowie die Problematik von manifesten und latenten nichtlinearen Modellen erläutert. Die komplexeren Strukturgleichungsmodelle, die neben einem Moderatorterm auch quadratische Termen enthalten, wurden besprochen und danach sowohl die Produktindikatorverfahren vorgestellt, die zur Parameterschätzung die Maximum-Likelihood-Methode verwenden, als auch die neu entwickelten verteilungsanalytischen Verfahren, nämlich LMS (Klein, 2000; Klein & Moosbrugger, 2000) und QML (Klein & Muthén, 2007) vorgestellt. Das Thema wurde zuerst aus theoretischer Perspektive analysiert und unter "Fragestellung 1" dargestellt. Bei der Analyse von Moderator- und quadratischen Termen in multiplen Regressionsmodellen kann es bei steigender Prädiktorkorrelation wegen des zunehmenden gemeinsamen Varianzanteils dieser beiden Terme zu einer Fehlspezifizierung der tatsächlich vorhandenen nichtlinearen Effekte kommen. Um eine zuverlässige Aufdeckung dieser Effekte zu ermöglichen, wurde daher in der Vergangenheit mehrfach empfohlen, grundsätzlich Moderator− und quadratische Effekte simultan zu untersuchen. Da psychologische Variablen jedoch selten messfehlerfrei erfasst werden können, erwiesen sich entsprechende Untersuchungen, die lediglich auf manifester Variablenebene durchgeführt worden waren und somit, von perfekt reliablen Messungen ausgingen, als methodisch problematisch. Zur weiterführenden Untersuchung der simultanen Analyse nichtlinearer Effekte empfiehlt sich daher der Einsatz latenter Strukturgleichungsmodelle, die den Messfehler manifester Variablen explizit berücksichtigen. Im Anschluss daran wurden die neu entwickelten verteilungsanalytischen Verfahren, LMS und QML vorgestellt. Diese neuen Verfahren berücksichtigen erstmals explizit die multivariate nichtnormale Verteilung der Indikatorvariablen, die aus nichtlinearen Modellen resultiert. Unter "Fragestellung 2“ wurde ihre Leistungsfähigkeit im Vergleich zu LISRELML-Verfahren überprüft. Die Voraussetzungen zur Anwendung der beiden Methoden ist die Normalverteilung der Indikatoren der latenten Prädiktorvariablen. Da in der empirischen Forschung zu psychologischen oder sozialwissenschaftlichen Fragestellungen normalverteilte Variablen sehr selten sind, wurde unter "Fragestellung 3" die Frage untersucht, wie robust beide Verfahren gegenüber der Verletzung der Verteilungsvoraussetzung sind. Es wurden zahlreiche Simulationsstudien durchgeführt, anhand derer die Robustheit der verteilungsanalytischen Verfahren bei variierender Schiefe und Kurtosis der Variablen nachgewiesen werden konnte. Auch unter weiteren praxisrelevanten Bedingungen, z.B. sehr komplexen Modellen mit bis zu sechs nichtlinearen Effekten und relativ kleinen Stichproben, zeigten beide Methoden sehr gute Schätzeigenschaften. Diese Ergebnisse sind für Anwender sehr ermutigend, da sie zeigen konnten, dass für die Untersuchung von multiplen nichtlinearen Effekten unter verschiedenen simulierten Bedingungen die verteilungsanalytischen Verfahren LMS und QML sehr zuverlässig sind und eingesetzt werden können. Beide Methoden wiesen unter den realisierten Bedingungen bezüglich der Güte der Parameterschätzungen keine bedeutsamen Unterschiede auf. Beide Verfahren lieferten unabhängig von der Komplexität der untersuchten Modelle erwartungstreue Parameterschätzungen und unverzerrte Standardfehlerschätzungen. Anschließend konnte neben dem Nachweis der erweiterten Leistungsfähigkeit der verteilungsanalytischen Verfahren anhand eines empirischen Beispiels aus der Burnout-Forschung gezeigt werden, dass die Analyse von multiplen nichtlinearen Effekten eine praktische Relevanz für die Weiterentwicklung von Theorien haben kann.
The population diversity of Doranthes excelsa Corrêa (Doryanthaceae) was measured from nine distinct geographic populations across eastern Australia, using random amplified polymorphic DNA (RAPD) markers. An UPGMA dendrogram of individuals was derived from squared Euclidian distances based on the Dice (1945) algorithm. Three clusters corresponding to populations at Somersby, Newfoundland and Kremnos Creek populations were found to be distinct from the remainder of the sampled individuals. A ΦST value of 0.443 indicated that a significant diversity between geographic populations existed; this appeared to be a product of geographical distance and isolation between some of the populations. (PCR = Polymerase Chain Reaction; RAPD = Random Amplified Polymorphic DNA) The results suggest that there is lesser gene flow between the‘northern’ populations (Kremnos Creek and Newfoundland) when compared to the ‘southern’ populations and that they have a significant level of genetic isolation. The two ‘northern’ populations should therefore be regarded as being of considerable value for conservation authorities and the commercial breeding sector and should be given priority for conservation. The plants there appear to exhibit a smaller phenotype but confirming this requires further quantification.

Die Wellen der Erregung haben sich inzwischen gelegt. Zu den ältesten Institutionen dieser Art gehören die Monumenta Germaniae Historica, kurz MGH. Für den im Mittelalter arbeitenden Rechtshistoriker ist unentbehrlich nicht nur die Reihe der Leges, sondern auch der Diplomata der deutschen Könige und Kaiser. Gerade in ihr ist in den letzten Jahrzehnten eine Herkulesarbeit an Materialbewältigung und Präzision mit der Edition der Diplome Friedrich I. Barbarossas in vier gewichtigen Bänden unter der Leitung von Heinrich Appelt bis 1990 geleistet worden. Begleitende Studien haben die Diplomatik bereichert und verfeinert, aber auch die Rolle des römischen Rechts genauer beleuchtet. Wichtige seither erschienene Monographien zu Barbarossa wären ohne diese Edition der Diplomata nicht denkbar gewesen. In der Reihe der Constitutiones liegt seit 1996 eine kritische Edition u. a. der Konstitutionen von Melfi Friedrichs II. vor, begleitet von einer zweibändigen Monographie des Herausgebers Wolfgang Stürner. ...

The purpose of this thesis was to investigate different aspects of the promotion of selfregulated learning in primary and secondary school education by focussing on its effectiveness, and on its assessment from different perspectives. Theoretically, the thesis is based on contemporary social-cognitive and constructivist theories of self-regulated learning. Two meta-analyses were conducted, an observation instrument was developed which was tested and employed in two observation studies, and a multi-method study was conducted to investigate different perspectives on the topic. Common to all studies is the evaluation of different aspects of the promotion of self-regulated learning among students. The results of this investigation are reported in four research articles (Studies 1-4), which have been accepted for publication (Study 1 and 2) or submitted to scientific peer reviewed journals (Study 3 and 4). The data are analyzed by applying a multi-method approach, using several sources of data (primary studies, self-reports, video data, interviews) and diverse methods (meta-analysis, observation analysis, survey analysis). The present data generally indicate that self-regulated learning can be enhanced both at primary and secondary school. The results of the first and the second study showed that primary and secondary school students partly benefit from different training characteristics. However, there were also common aspects of effective training characteristics that hold for both school levels. Moreover, the third study revealed that it was possible to develop an instrument to observe teachers’ promotion of self-regulated learning in a reliable way, which can be applied in several contexts. The results indicated that the stability of teachers’ promotion of selfregulated learning varies among the school subjects. Furthermore, the results showed that only little instruction of self-regulated learning takes place in primary and secondary school mathematics lessons. Yet, secondary school teachers showed more promotion of cognitive strategies than primary school teachers did, although the former included more constructivist characteristics in the learning environment. The observation studies produced a rich pool of data, serving as pilot studies for future studies with a larger sample sizes that are needed to further strengthen the results. As the fourth study indicated, teacher ratings differ significantly from video-based observations in perceiving their promotion of self-regulated learning. However, for some aspects they agree with their students’ perception. Finally, it was found that students’ perception on their teachers’ promotion of self-regulated learning had the highest impact on their self-regulation competence. In the future, it will be crucial to include the instruction of self-regulated learning from a theoretical and a practical perspective in the teacher training curriculum. Moreover, in future research the implementation of the promotion of self-regulated learning should be investigated, and in experimental settings different ways of supporting such an implementation should be examined. A close collaboration with teachers would be helpful to get deeper insights into teachers’ behaviour and attitudes. The promotion of self-regulated learning should start as early as in primary school as students are already able to learn it then and as it takes many years to develop it fully. In addition, when investigating teachers’ promotion of self-regulated learning, the school subject should be taken into account during assessment. Long-term measurements could acknowledge such a potential instability. Moreover, in further studies, observation data of a large sample of teachers should be gathered in order to get a representative overview of teachers’ promotion of self-regulated learning. Furthermore, research on the promotion of self-regulated learning should account for the impact of students’ perspectives referring on this.

Durch die Ästhetisierung und Regulierung der Gewalt im ritterlichen Kampf und insbesondere in seiner literarischen Darstellung spaltet sich der mehrdeutige deutsche Begriff ‘Gewalt’ in zwei seiner lateinischen Entsprechungen: Wofern die Regeln eingehalten werden, ist sie ‘vis’, wofern aber gegen die Regulierung verstoßen wird, ist sie ‘violentia’; Johannes Rothe nennt sie dann im ›Ritterspiegel‹ ‘bose gewalt’ (V. 2677). Nicht vergessen werden darf aber die dritte Komponente der Gewalt, die ebenfalls in diesem schillernden deutschen Wort enthalten ist: ‘potestas’, die auf ‘vis’ aufbaut. Die ‘êre’, die ein Ritter im Kampf erringt, ist mehr als nur ein guter Ruf, sie ist eine gesellschaftliche Anerkennung, die auch Elemente der Macht in sich trägt. Die ‘potestas’ aber des Herrschers stärkt sich, indem sie einige Formen der ‘vis’ anderer zur ‘violentia’ erklärt. Ein herausragendes Beispiel hierfür sind die Landfriedengebote, die insbesondere unter dem Stauferkaiser Friedrich I. häufig ausgesprochen und als Herrschaftsinstrument verwendet wurden. Durch sie sollte die Möglichkeit eines individuellen gewaltsamen Konfliktaustrags in der Fehde unterbunden werden, indem sich beide Parteien der richterlichen Gewalt, der ‘potestas’ des Herrschers, unterwerfen und es dem Richter überlassen wird, mit Gewalt denjenigen, der gegen Regeln des Zusammenlebens verstoßen hat, zu bestrafen. Die ‘peinlichen’, also ernsten körperlichen Strafen, die die hoheitliche Gerichtsbarkeit verhängen kann, sind Ausdruck dieser rechtlich kanalisierten, aber keineswegs abgeschafften Gewalt, auf der die Ordnung beruht.

Der Harz ist neben seinen Wäldern auch durch seine bunten Bergwiesen bekannt. Letztere sind schon seit längerer Zeit durch Nutzungsaufgabe oder intensivere Nutzung bedroht und gehören zu den besonders schutzbedürftigen Pflanzengesellschaften. Seit den 1980er Jahren werden zunehmend staatlich geförderte Pflegemaßnahmen zu ihrer Regeneration und Erhaltung durchgeführt. Im NSG „Bergwiesen bei St. Andreasberg“ sind seit 1987 wieder große Flächen durch einen ortsansässigen Landwirt gemäht worden. Zur Erfolgskontrolle und Erprobung anderer Pflegemaßnahmen wurden 1988 für ein Biomonitoring mehrere Dauerflächen eingerichtet, mit 10 x 10 m-Parzellen für jährliche Mahd und Brache, teilweise auch für Mahd alle zwei oder drei Jahre sowie Mulchen. Die Parzellen wurden bis 2002 auf 2x2 m-Kleinflächen erfasst. 2003 wurden die ganzen Parzellen aufgenommen, zusätzlich die Vertikalstruktur (Höhe der Schichten) und die oberirdische Biomasse gemessen.
Beschrieben werden hier vier Dauerflächen, zwei in Goldhaferwiesen (Geranio-Trisetetum potentilletosum erectae), je eine in einem Borstgrasrasen (Polygalo-Nardetum) und in einem Rubus idaeus-Brachestadium. Die Ergebnisse zeigen, dass sich durch jährliche Mahd mit Abfuhr des Mähgutes überall wieder eine artenreiche, bunt blühende Magerwiese mit offener Struktur und kleinwüchsigen Pflanzen eingestellt hat. Mahd alle zwei oder drei Jahre fördert etwas wuchskräftigere Arten. Mulchen erscheint wegen stärkerer Streubildung weniger geeignet. In den Brachen haben sich Hochstaudenfluren oder andere Dominanzstrukturen entwickelt. Eine Schlüsselart für die Regeneration ist Meum athamanticum, dessen Wuchskraft am besten durch jährliche Mahd eingedämmt wird. In der Diskussion wird deshalb als beste Erhaltungsmaßnahme eine jährliche Mahd ab Ende Juni empfohlen, möglichst im Mosaik mit seltener bis gar nicht gemähten Flächen. Auch eine extensive Beweidung wird diskutiert.

Der diesjährige Band 29 ist umfangreicher als der vorangegangene Band. Das Spektrum der Themen von 15 Beiträgen reicht inhaltlich und geografisch recht weit, sodass hoffentlich alle Leserinnen und Leser etwas Interessantes vorfinden werden. Auch gibt es mehrere Beiträge in englischer Sprache, was den internationalen Gebrauch fördern kann.
In Tuexenia 12 wurde erstmals über den European Vegetation Survey (EVS) berichtet (DlERSCHKE 1992). Der Name war zunächst Programm, ist inzwischen längst der Name einer aktiven Gruppe europäischer Pflanzensoziologen innerhalb der International Association for Vegetation Science (IAVS) geworden, die sich alljährlich zu einem Workshop trifft, zunächst immer in Rom, später auch alternierend an anderen Orten. Ziele sind internationale Präsentation von Daten und Methoden sowie Diskussion zur Entwicklung eines gesamteuropäischen Netzes von Fachleuten, nicht zuletzt auch zu Aufbau und Pflege persönlich-freundschaftlicher Kontakte.
It is more than forty seven years ago that the Federation of black African students in France (FEANF) organised its first seminar in Paris on the relationship between black African literature and politics. The significance of the event came from the fact that literature served as a vehicle for unmasking traitors in Africa. This was also an opportunity for African students to define the role of literature in political struggles and to appreciate correctly and objectively the commitments of African writers in French. At no time was it a question of over emphasising the importance of this type of work in relation to the immense political challenges in the liberation struggle of African countries. Despite their ideological, religious and philosophical differences, African intellectuals were all committed to African independence and unity, and the need for a critical appraisal of the contribution of African literature in this regard. Participants at this seminar accomplished this task in serenity and with much lucidity. The young generation of pupils and students have the right to know the opinions of their elders who took part, in various degrees and for various reasons, in the struggles for independence on the African continent.
Mit der vorgelegten Dissertation wird ein Beitrag zur Diskussion um Beschäftigungssicherung unter veränderten globalen und betrieblichen Beziehungen geleistet. Aufgezeigt werden betriebliche Strategien im Umgang mit Beschäftigungskrisen in der Metall- und Elektroindustrie und der Beitrag einer aktiven Arbeitsmarktpolitik zur Sicherung von Beschäftigung. Hintergrund bilden die massiven Beschäftigungseinbrüche, insbesondere Anfang der 1990er Jahre, in der Metall- und Elektroindustrie. Veränderte wirtschaftliche Rahmenbedingungen im Verlauf des Strukturwandels führten zu einer Umorientierung bei den Unternehmens- und Managementstrategien, um auf veränderte Marktanforderungen und verschärfte Wettbewerbsbedingungen zu reagieren und auch weiterhin Gewinnspannen realisieren zu können. Neue Reorganisationskonzepte in den Unternehmen und der zunehmende Druck der von Shareholder Value-Strategien ausgeht, führen immer wieder zu unternehmerischen Anpassungsstrategien, die mit Stelleneinsparungen einhergehen. Am Ende betrieblicher Reorganisation steht häufig der Verlust von Arbeitsplätzen. Allein der Blick auf das Endresultat Personalabbau reicht allerdings nicht aus, um die Auswirkungen dieser Maßnahmen auf Betrieb, Beschäftigte und nicht zuletzt den Arbeitsmarkt erfassen zu können. Ziel der Arbeit ist es, anhand der Analyse von vier Fallstudien Ansatzpunkte dafür zu finden, wie Prozesse im Rahmen von Beschäftigungsabbau beeinflusst und gestaltet werden können. Dabei werden neben betriebsinternen Möglichkeiten der Beschäftigungssicherung ebenfalls Beschäftigungsperspektiven in der Region in den Blick genommen. Für diese Studie wurde ein disziplinenübergreifender Ansatz gewählt, in dem Elemente der Industriesoziologie und der Arbeitsmarktforschung integriert sind. Im Bereich der Industriesoziologie wird auf die Arbeitspolitik und damit einen handlungsorientierten Ansatz der Industriesoziologie zurückgegriffen. Aus dem Bereich der Arbeitsmarktforschung dient der segmentationstheoretische Ansatz als theoretische Grundlage, um die Strukturierung des Arbeitsmarktes und ihre Verfestigung aufzeigen zu können. Präventive Beschäftigungssicherung zu untersuchen setzt voraus, die Grenzen zwischen industriesoziologischen und arbeitsmarktpolitischen Fragestellungen zu überschreiten. Diese Forschungsperspektive ermöglicht es, den in der Regel zu engen Blick auf die Bestandssicherung der vom Abbau bedrohten Arbeitsplätze zu erweitern und ihn für die Beschäftigungsperspektiven im Unternehmen selbst und über das Unternehmen hinaus zu öffnen.
In der vorliegenden Arbeit wurden zehn Substanzen, die im Rahmen des Projekts INTAFERE analytisch in verschiedenen Gewässersystemen des Hessischen Rieds nachgewiesen werden konnten, ökotoxikologisch charakterisiert. Neben der Bestimmung der Akut- und der chronischen Toxizität wurde auch das endokrine Potential mit Hilfe eines rekombinanten Hefe-Assays ermittelt.Die akute Toxizität zeigt zwischen den verschiedenen Substanzen große Differenzen. Die drei Organophosphate TCPP, TBEP und TCEP zeigen selbst bei hohen Konzentrationen keine oder nur sehr geringe Effekte, während 4-NP, 4-t-OP, BPA, TDCPP, AHTN und Terbutryn mit LC50-Werten bis zu 5 mg/l eine höhere Toxizität besitzen.Im Hefe-Assay kann in mehreren Versuchswiederholungen das östrogene Potential von 4-NP (MW: 6,71x10-6 M), 4-t-OP (MW: 7,16x10-6 M) und BPA (MW: 4,88x10-6 M), sowie die antiandrogene Wirkung des Bisphenols (5,29x10-5 M) bestätigt werden. Im Gegensatz dazu können im Yeast Antiestrogen Screen zum ersten Mal Hinweise auf die antiöstrogene Wirkung von TCPP (MW: 6,86x10-5 M), Terbutryn (MW: 3,99x10-5 M), TDCPP (MW: 2,65x10-6 M) und TBP (MW: 2,28x10-5 M) aufgezeigt werden.TBP und TDCPP führen auch in der chronischen Exposition bei Potamopyrgus antipodarum zu einer Reduktion in der Embryonenzahl (mehrfach beobachtete NOEC beider Substanzen: 6,25 mg/kg), ein Effekt, der zunächst nicht von einer toxischen Wirkung unterschieden werden kann. Allerdings scheint sich ein antiöstrogener Wirkmechanismus auf die Schnecken bei den Versuchen zur Mischtoxizität zu bestätigen, da die gleichzeitige Exposition gegenüber BPA und 4-tOP zu einer geringfügigen Aufhebung des Effekts führt. Potamopyrgus reagiert ebenfalls mit einer Reduktion der Embryonenzahl bei der Exposition gegenüber AHTN (mehrfach beobachtete NOEC: 2 mg/kg), zeigt sich aber insensitiv gegenüber der Belastung mit Terbutryn.Die Exposition von Chironomus riparius gegenüber den verschiedenen Substanzen führt bis auf eine Ausnahme zu keinen signifikanten substanzbedingten Beeinträchtigungen. Einzig das s-Triazin Terbutryn verursacht eine hohe Mortalität mit einer NOEC (28 d) von 250 μg/kg. Eine Toxizität auf den Anneliden Lumbriculus variegatus zeigt sich lediglich bei derExposition gegenüber BPA (NOEC: 10 mg/kg; Biomasse, 28 d) und 4-NP (EC10: 6,88 mg/kg; 95% KI: 3,97-11,9; Reproduktion, 28 d). Die durchgeführten Versuche zur Toxizität von Mischungen zeigen eine größere Gefährdung auf als bei Vorliegen der Einzelsubstanzen in Konzentrationen unterhalb ihres Schwellenwertes zu vermuten wäre. Allerdings lassen sich die mit dem Hefe-Assay erzielten Ergebnisse aufgrund großer Variabilitäten zwischen den einzelnen Versuchswiederholungen nicht immer eindeutig mit Hilfe des additiven oder des unabhängigen Modells beschreiben, zeigen dabei jedoch trotzdem ein gegenüber den Einzelsubstanzergebnissen verändertes Risiko auf. Um diesem in der Risikobewertung Rechnung zu tragen, wird ein Verfahrensvorschlag entwickelt, in dem durch zusätzliche Sicherheitsfaktoren für PNECs ein Überschreiten des risikoanzeigenden Werts von 1 des PEC/PNEC-Quotienten einer Mischung verhindert werden kann.
Neutron stars are very dense objects. One teaspoon of their material would have a mass of five billion tons. Their gravitational force is so strong that if an object were to fall from just one meter high it would hit the surface of the respective neutron star at two thousand kilometers per second. In such dense bodies, different particles from the ones present in atomic nuclei, the nucleons, can exist. These particles can be hyperons, that contain non-zero strangeness, or broader resonances. There can also be different states of matter inside neutron stars, such as meson condensates and if the density is height enough to deconfine the nucleons, quark matter. As new degrees of freedom appear in the system, different aspects of matter have to be taken into account. The most important of them being the restoration of the chiral symmetry. This symmetry is spontaneously broken, which is a fact related to the presence of a condensate of scalar quark-antiquark pairs, that for this reason is called chiral condensate. This condensate is present at low densities and even in vacuum. It is important to remember at this point that the modern concept of vacuum is far away from emptiness. It is full of virtual particles that are constantly created and annihilated, being their existence allowed by the uncertainty principle. At very high temperature/density, when the composite particles are dissolved into constituents, the chiral consensate vanishes and the chiral symmetry is restored. To explain how and when chiral symmetry is restored in neutron stars we use a model called non-linear sigma model. This is an effective quantum relativistic model that was developed in order to describe systems of hadrons interacting via meson exchange. The model was constructed from symmetry relations, which allow it to be chiral invariant. The first consequence of this invariance is that there are no bare mass terms in the lagrangian density, causing all, or most of the particles masses to come from the interactions with the medium. There are still other interesting features in neutron stars that cannot be found anywhere else in nature. One of them is the high isospin asymmetry. In a normal nucleus, the amount of protons and neutrons is more or less the same. In a neutron star the amount of neutrons is much higher than the protons. The resulting extra energy (called Fermi energy) increases the energy of the system, allowing the star to support more mass against gravitational collapse. As a consequence of that in early stages of the neutron star evolution, when there are still many trapped neutrinos, the proton fraction is higher than in later stages and consequently the maximum mass that the star can support against gravity is smaller. This, between many other features, shows how the microscopic phenomena of the star can reflect into the macroscopic properties. Another important property of neutron stars is charge neutrality. It is a required assumption for stability in neutron stars, but there are others. One example is chemical equilibrium. It means that the number of particles from each kind is not conserved, but they are created and annihilated through specific reactions that happen at the same rate in both directions. Although to calculate microscopic physics of neutron stars the space-time of special relativity, the Minkowski space, can be used, this is not true for the global properties of the star. In this case general relativity has to be used. The solution of Einstein's equations simplified to static, spherical and isotropic stars correspond to the configurations in which the star is in hydrostatic equilibrium. That means that the internal pressure, coming mainly from the Fermi energy of the neutrons, balances the gravity avoiding the collapse. When rotation is included the star becomes more stable, and consequently, can be more massive. The movement also makes it non-spherical, what requires the metric of the star to also be a function of the polar coordinate. Another important feature that has to be taken into account is the dragging of the local inertial frame. It generates centrifugal forces that are not originated in interactions with other bodies, but from the non-rotation of the frame of reference within which observations are made. These modifications are introduced through the Hartle's approximation that solves the problem by applying perturbation theory. In the mean field approximation, the couplings as well as the parameters of the non-linear sigma model are calibrated to reproduce massive neutron stars. The introduction of new degrees of freedom decreases the maximum mass allowed for the neutron star, as they soften the equation of state. In practice, the only baryons present in the star besides the nucleons are the Lambda and Sigma-, in the case in which the baryon octet is included, and Lambda and Delta-,0,+,++, in the case in which the baryon decuplet is included. The leptons are included to ensure charge neutrality. We choose to proceed our calculations including the baryon octet but not the decuplet, in order to avoid uncertainties in the couplings. The couplings of the hyperons were fitted to the depth of their potentials in nuclei. In this case the chiral symmetry restoration can be observed through the behavior of the related order parameter. The symmetry begins to be restored inside neutron stars and the transition is a smooth crossover. Different stages of the neutron star cooling are reproduced taking into account trapped neutrinos, finite temperature and entropy. Finite-temperature calculations include the heat bath of hadronic quasiparticles within the grand canonical potential of the system. Different schemes are considered, with constant temperature, metric dependent temperature and constant entropy. The neutrino chemical potential is introduced by fixing the lepton number in the system, that also controls the amount of electrons and protons (for charge neutrality). The balance between these two features is delicate and influenced mainly by the baryon number conservation. Isolated stars have a fixed number of baryons, which creates a link between different stages of the cooling. The maximum masses allowed in each stage of the cooling process, the one with high entropy and trapped neutrinos, the deleptonized one with high entropy, and the cold one in beta equilibrium. The cooling process is also influenced by constraints related to the rotation of the star. When rotation is included the star becomes more stable, and consequently, can be more massive. The movement also deforms it, requiring the metric of the star to include modifications that are introduced through the use of perturbation theory. The analysis of the first stages of the neutron star, when it is called proto-neutron star, gives certain constraints on the possible rotation frequencies in the colder stages. Instability windows are calculated in which the star can be stable during certain stages but collapses into black holes during the cooling process. In the last part of the work the hadronic SU(3) model is extended to include quark degrees of freedom. A new effective potential to the order parameter for deconfinement, the Polyakov loop, makes the connection between the physics at low chemical potential and hight temperature of the QCD phase diagram with the height chemical potential and low temperature part. This is done through the introduction of a chemical potential dependency on the already temperature dependent potential. Analyzing the effect of both order parameters, the chiral condensate and the Polyakov loop, we can drawn a phase diagram for symmetric as well as for star matter. The diagram contains a crossover region as well as a first order phase transition line. The new couplings and parameters of the model are chosen mainly to fit lattice QCD, including the position of the critical point. Finally, this matter containing different degrees of freedom (depending on which phase of the diagram we are) is used to calculate hybrid star properties.
Protein S-100B als Serummarker der zerebralen Schädigung bei pädiatrischen Schädel-Hirn-Verletzungen

Protein S-100 ist ein Calcium-bindendes Protein, das die Aktivität seiner Zielproteine moduliert. Der Aufgabenbereich umfasst die Regulation von Zellwachstum und Zellstrukturbildung. Entdeckt wurde es im Jahre 1965 von B.W. Moore, der es erstmals aus Rinderhirn isolierte. Seinen Namen erhielt es aufgrund seiner Löslichkeit in 100% gesättigtem Ammoniumsulfat bei neutralem ph-Wert. Protein S-100 besteht aus mehreren Untereinheiten, von denen S-100B nur im Hirngewebe vorkommt, hauptsächlich im Zytosol von Gliazellen. Bei astroglialer Zerstörung und konsekutiver Störung der Blut-Hirn-Schranke können erhöhte Konzentrationen von Protein S-100B im Serum gemessen werden. Durch seine spezifische Herkunft besitzt Protein S-100B einen potentiellen Wert als biochemischer Marker einer strukturellen Hirnschädigung. Ziel dieser Studie ist die Überprüfung des posttraumatischen Protein S-100B Serumspiegels bezüglich seiner Korrelation mit dem Schweregrad kindlicher Schädel-Hirn-Verletzungen und deren Prognose. In Form einer prospektiven Studie wurden insgesamt 45 Kinder (23 Jungen und 22 Mädchen) zwischen 4 Monaten und 17 Jahren (Mittelwert 7,64 Jahre) mit Hirnverletzungen unterschiedlicher Schweregrade untersucht, bei denen innerhalb der ersten 20 posttraumatischen Stunden mittels eines immunoluminometrischen Assays die Protein S-100B Serumkonzentration bestimmt wurde. Der Normalbereich für Protein S-100B liegt bei < 0,15 µg/l. Werte zwischen 0,15 und 0,5 µg/l gelten als leichtgradig erhöht, Werte über 0,5 µg/l als deutlich erhöht. Die S-100B-Werte wurden mit den klinischen Daten und dem posttraumatischen Outcome (Nachbeobachtungszeit mindestens 6 Monate) korreliert. Die statistischen Signifikanzberechnungen erfolgten mittels des Fisher-Exact-Tests.Die gemessenen S-100B Werte lagen zwischen 0,05 und 3,05 µg/l (Mittelwert 0,46 µg/l). Die mittlere Latenzzeit der posttraumatischen Protein S-100B Bestimmung betrug 5 Stunden. 38 % der Kinder zeigten leichtgradige Erhöhungen (>= 0,2-0,5 µg/l), 29 % deutliche Erhöhungen (> 0,5 µg/l). Niedrige Punktwerte in der initialen Glasgow-Coma-Scale (GCS) bedeuteten im Mittel höhere Werte für Protein S-100B. Ein initialer GCS-Wert von unter 8 ging in allen Fällen mit S-100B Werten über 0,5 µg/l einher (p=0,005). In Fällen ohne Verlust des Bewusstseins, bzw. maximal einer leichten Eintrübung des Bewusstseins (Lange-Cosack I-II) lagen die S-100B Werte im Mittel niedriger als bei Vorliegen eines Bewusstseinsverlustes (III-IV). Ein Bewusstseinsverlust ging immer mit einem S-100B Wert von mindestens 0,2 µg/l einher (p=0,04), meist lag der Wert über 0,5 µg/l (p=0,015). 25 Kinder erhielten eine Computer- und /oder Magnetresonanztomographie. Hier zeigten sich erhöhte S-100B Werte vor allem bei schweren Kontusionen und Subduralblutungen. Den höchsten Wert (3,05 µg/l) wies ein Kind mit ausgedehnten diffusen axonalen Verletzungen auf. Bei 38 der 45 Patienten konnte nach einer durchschnittlichen Nachbeobachtungszeit von 27,5 Monaten das neurologische Outcome (Glasgow Outcome Scale, GOS) erhoben werden. 8 Kinder (21 %) wiesen neurologische Residuen auf, vor allem Koordinationsstörungen und Verlangsamungen im Sinne eines Psychosyndroms, 1 Kind verstarb im Akutverlauf an einem malignen Hirnödem. Die Kinder mit posttraumatischen Residuen / Behinderungen wiesen im Mittel tendenziell höhere Werte für Protein S-100B auf. Die unterschiedlichen Beschwerdebilder und Residualzustände nach leichten und schweren Schädel-Hirn-Verletzungen lassen sich anhand der etablierten klinischen, neurophysiologischen und neuroradiologischen Prognosemarker bislang nur bedingt vorhersagen. Die Ergebnisse dieser Untersuchung zeigen gute Korrelation des Serum S-100B Wertes mit der Schwere und Dauer der initialen Bewusstseinsstörung sowie dem Ausmaß der hirnparenchymatösen Läsionen. Die Aussagekraft bezüglich des posttraumatischen Outcomes ist aufgrund der Fallzahl und der zu einem großen Teil einbezogenen leichten Schädel-Hirn-Verletzungen begrenzt. Die Untersuchungsergebnisse unterstützen die bei Erwachsenen erhobenen Daten und zeigen, dass Protein S-100B auch im Kindesalter einen viel versprechenden Parameter zur Objektivierung einer akuten Hirnschädigung darstellt und bereits zu einem frühen posttraumatischen Zeitpunkt einen Beitrag zur Beurteilung des Schweregrades und der Prognose von Schädel-Hirn-Verletzungen leisten kann. Der posttraumatische Protein S-100B Serumwert bringt hier als biochemischer Marker eine zusätzliche Information und kann so bei der Einschätzung und Initiierung von diagnostischen, therapeutischen, rehabilitativen und Follow-up-Maßnahmen hilfreich sein.
This study focuses on structural features of a particular GPCR type, the family C GPCRs. Structure- and ligand-based approaches were adopted for prediction of novel mGluR5 binding ligand and their binding modes. The objectives of this study were: 1. An analysis of function and structural implication of amino acids in the TM region of family C GPCRs. 2. The prediction of the TM domain structure of mGluR5. 3. The discovery of novel selective allosteric modulators of mGluR5 by virtual screening. 4. The prediction of a ligand binding mode for the allosteric binding site in mGluR5. GPCRs are a super-family of structurally related proteins although their primary amino acid sequence can be diverse. Using sequence information a conservation analysis of family C GPCRs should be applied to reveal characteristic differences and similarities with respect function, folding and ligand binding. Using experimental data and conservation analysis the allosteric binding site of mGluR5 should be characterized regarding NAM and PAM and selective ligand binding. For further evaluation experimental knowledge about family A GPCRs as well as conservation between vertebrate rhodopsins was planned to be compared to results obtained for family C GPCRs (Section 4.1 Conservation analysis of family C GPCRs). Since no receptor structure is available for any family C GPCR, discussion of conserved sequence positions between family A and C GPCRs requires the prediction of a receptor structure for mGluR5 using a family A receptor as template. In order to predict the mGluR5 structure a sequence alignment to a GPCR template protein will have to be proposed and GPCR specific features considered in structure calculation (Section 4.1.4 Structure prediction of mGluR5). The obtained structure was intended to be involved in ligand binding mode prediction of newly discovered active molecules. For discovery of novel selective mGluR modulators several ligand-based virtual screening protocols were adapted and evaluated. Prediction models were derived for selection of possibly active molecules using a diverse collection of known mGluR binding ligands. For that purpose a data collection of known mGluR binding ligands should be established and this reference collection analyzed with respect to different ligand activity classes, NAM or PAM and selective modulators. The prediction of novel NAMs and PAMs using several combinations of 2D-, 3D-, pharmacophore or molecule shape encoding methods with machine learning techniques and similarity determining methods should be tested in a prospective manner (Section 4.2 Virtual screening for novel mGluR modulators). In collaboration with Merz Pharmaceuticals (Merz GmbH & Co. KGaA, Frankfurt am Main, Germany) the modulating effect of a few hundred molecules should be approved in a functional cell-based assay. With the objective to predict a binding mode of the discovered active molecules, molecule docking should be applied using the allosteric binding site of the modeled mGluR5 structure (Section 4.2.4 Modeling of binding modes). Predicted ligand binding modes are to be correlated to conservation profiles that had resulted from the sequence-based entropy analysis and information from mutation experiments, and shall be compared to known ligand binding poses from crystal structures of family A GPCRs.
Epidermal growth factor (EGF) receptor belongs to the broad family of enzymatic receptors called receptor tyrosine kinases (RTKs). Generally, the binding of a ligand to these receptors leads to activation of their intracellular kinase activity that sets in motion a cascade of signaling events. In order to ensure appropriate responses to physiological stimuli, the cell is endowed with the ability to regulate signal transduction via numerous mechanisms such as dephosphorylation of the RTK and its substrates as well as downregulation of the RTK. Activation of EGFR is a potent mitogenic (proliferative) and motogenic (cell motility) signal that plays crucial roles during embryonic development and maintenance of adult tissue. EGFR signaling is primarily regulated by ligand-induced receptor internalization with subsequent degradation in lysosomes. While the complex of proteins that are recruited to EGFR after its activation is well understood, proteins that interact with the receptor in the absence of ligand binding are still not systematically studied. With the goal of identifying novel binding partners of non-activated EGFR, a membrane based yeast-two hybrid screen (MYTH) was conducted. MYTH is based on the principle of in vivo reconstitution of the N-terminus (Nub) and C-terminus (Cub) halves of ubiquitin once brought into close proximity. A chimeric protein consisting of EGFR fused to Cub and a transcription factor was used as a bait to screen Nub-tagged cDNA library. Analysis of resultant yeast transformants revealed a total of 87 proteins to interact with EGFR. Of these only 11 were previously shown to bind to EGFR. A majority of the other proteins were shown to interact with the receptor by yeast retransformation. Fifteen were confirmed to bind to EGFR by coimmunoprecipitation assays in mammalian cells. One of the novel EGFR interactors identified in the screen was histone deacetylase 6 (HDAC6). This deacetylase is localized in the cytoplasm and known to deacetylate alpha-tubulin, HSP90 and cortactin. The juxtamembrane region of EGFR binds to the Cterminus of HDAC6. Functionally, overexpression of wild type HDAC6 stabilized ligand-induced degradation of the receptor. On the other hand, deacetylase deficient or EGFR binding compromised mutants of HDAC6 were able to stabilize EGFR only partially. Downmodulation of HDAC6 expression by RNAi markedly accelerated degradation of the receptor. Taken together, HDAC6 is a negative regulator of EGFR downregulation that is dependent on its deacetylase activity and ability to bind to the receptor. Imaging studies revealed that HDAC6 does not affect internalization of EGFR from the plasma membrane but rather influences the post-endocytic trafficking of the receptor-ligand complex to lysosomes. Pulse-chase experiments using fluorophoretagged EGF showed that EGFR is transported faster towards the peri-nuclear region and delivered to late endosomes rapidly in HDAC6 depleted cells. HDAC6 is demonstrated to act, at least partly, by regulating the acetylation of alpha-tubulin. Upon EGFR activation, acetylation of alpha-tubulin on lysine 40 is progressively increased as shown by mass spectrometry and immunoblotting. Forced expression of a dominant negative mutant of alpha-tubulin, but not wild type alpha-tubulin, led to reduced speed and processive movement of early endosomes in GFP-Rab5 expressing cells. In a surprising twist, EGFR is able to phosphorylate HDAC6 on Tyr570. Phosphorylation of Tyr570 and Ser568 leads to inactivation of the deacetylase function of HDAC6 as shown by in vivo and in vitro assays. In summary, HDAC6 diminishes EGFR downregulation by slowing the transport of intracellular vesicles. The inhibitory effect is removed once HDAC6 is phosphorylated on key residues. In line with these findings, two recent reports have shown that hyper-acetylation of alpha-tubulin induced by inhibition of HDAC6 increases the transport of brain derived neurotrophic factor and JNK interacting protein-1 in different cell systems. Acetylated microtubules are more efficient in recruiting motor proteins like kinesin-1 and dynein. These findings indicate that HDAC6 plays an important regulatory role in intracellular trafficking pathways. However, several outstanding issues still remain unresolved. How does acetylation of microtubules influence vesicular trafficking? In this regard, the temporal and spatial dynamics of alpha-tubulin acetylation following EGFR activation should be studied. Furthermore, whether HDAC6 affects the trafficking of other endocytic cargos and additional organelles is an interesting question to address.
Am Anfang der folgenden Auseinandersetzung mit der Queer-Theorie wäre sicherlich die einschlägige Definition von queer zu erwarten und dennoch könnte hier solche Erwartung enttäuscht werden, da sich um den genannten Begriff teilweise die Aura des Undefinierbaren, Wandelbaren und Uneindeutigen gebildet hat, die ihn zugleich als fließendes Feld von vielen Möglichkeiten erscheinen lässt. Es widerstrebt sogar manchen Theoretikerinnen und Theoretikern, diesen Begriff in eine Zwangsjacke von terminologischer Einengung zu bringen und ihn somit erstarren zu lassen. Auf der einen Seite birgt diese Einstellung Gefahr, dass infolge der Absenz einer exakten Definition die Queer-Theorie nur als intuitive und unzulänglich ausgearbeitete Theorie angesehen wird, und dass sie um so mehr um Akzeptanz ringen muss (vgl. Jagose 2005: 124). Auf der anderen Seite ist es eben die Unbestimmtheit, die den "vielbeschworenen Charme" von queer ausmacht.
This essay is concerned with the state of the concept of »work« in the realm. of literature. Around 1800, "work" becomes a central reflective category in the field of literature, as well as in philosophical and literary aesthetics. In the last two decades of the 18th century, especially within Goethe's and Schiller's classicist concepts of literature, "work" and "art"/"poetry" are generally considered unconnected, as is an economic and aesthetic attitude towards nature. Where the spheres of "art"/"poetry" and "work" are linked to each other this relationship is defined as one of opposition, as can be seen especially in Goethe's novel Wilhelm Meisters Lehrjahre and in Schiller's essay Über die ästhetische Erziehung des Menschen. This changes significantly with the re-conceptualization of literature by the German Early Romantics. Not only does the sphere of "work" - now regarded as something meaningful - become at this moment in the course of the· evolution of the literary system an eminent topic within literature and literary aesthetics, the poetic activity itself becomes - suddenly - conceptualized as "work" and the writer thought of as a "worker". This essay first raises the question of what cultural changes might follow the approaching end of the "working society" in Western societies before proceeding to this important turning point in the self-conceptualization of the literary system in relation to the modem concept of "work".
It has been shown that stem and progenitor cells are therapeutically effective after i.v application. Yet, many aspects regarding intracellular signaling pathways which are involved in the homing and local action of these cells still have to be elucidated. In this work, it was aimed to investigate the role of the small GTPase Rap1 in adhesion activation in Hematopoietic Stem and Progenitor Cells (HSC/HPC) and in Mesenchymal Stem Cells (MSC). The potential role of Rap1 was assessed in, mice which were homozygote negative for the expression of the Rap1a gene. Peripheral blood lymphocyte counts as well as numbers of HPCs in the blood were decreased in Rap1a-/- mice compared to wild-type controls. Additionally the adhesion capability of HPCs from Rap1a-/- to the endothelial ligand, Vascular Cell Adhesion Molecule – 1 under shear stress was decreased. The hematopoietic repopulation potential of Rap1a-/- HPC was however not decreased in a competitive bone marrow transplantation model, indicating that deficiency of Rap1a in HSC/HPC does not negatively affect their ability to interact with the bone marrow microenvironment. In contrast, the isolation of MSC was not possible from Rap1a-/- bone marrow, indicating an altered situation in the bone marrow niche through changed stromal cell behaviour. Instead, Rap1a+/- MSC could be isolated and showed an adhesion deficit under shear stress. In contrast, no differences were noted in their differentiation potential. In a mouse homing model, the overall ability of the Rap1a+/- MSC to home to different tissues was found preserved. Finally, in a murine subcutaneous carcinoma model, cells with an HPC phenotype were observed to be present in the tumor microenvironment, and it was shown that they home directly to tumors. Since HPC isolated from bone marrow were able to differentiate into cells with a pro-angiogenic phenotype in vitro, HPC may be of relevance for neovascularization, as tumor-infiltrating progenitor cells. The results of the study should contribute to the understanding of the regulation of progenitor cell homing behaviour in situations simulating cell therapy approaches in preclinical situations.
