Background: Studies on the development of the nervous system and the musculature of invertebrates have become more sophisticated and numerous within the last decade and have proven to provide new insights into the evolutionary history of organisms. In order to provide new morphogenetic data on opisthobranch gastropods we investigated the neuromuscular development in the nudibranch Aeolidiella stephanieae Valdez, 2005 using immunocytochemistry as well as F-actin labelling in conjunction with confocal laser scanning microscopy (cLSM). Results: The ontogenetic development of Aeolidiella stephanieae can be subdivided into 8 stages, each recognisable by characteristic morphological and behavioural features as well as specific characters of the nervous system and the muscular system, respectively. The larval nervous system of A. stephanieae includes an apical organ, developing central ganglia, and peripheral neurons associated with the velum, foot and posterior, visceral part of the larva. The first serotonergic and FMRFamidergic neural structures appear in the apical organ that exhibits an array of three sensory, flask-shaped and two non-sensory, round neurons, which altogether disappear prior to metamorphosis. The postmetamorphic central nervous system (CNS) becomes concentrated, and the rhinophoral ganglia develop together with the anlage of the future rhinophores whereas oral tentacle ganglia are not found. The myogenesis in A. stephanieae begins with the larval retractor muscle followed by the accessory larval retractor muscle, the velar or prototroch muscles and the pedal retractors that all together degenerate during metamorphosis, and the adult muscle complex forms de novo. Conclusions: Aeolidiella stephanieae comprises features of the larval and postmetamorphic nervous as well as muscular system that represent the ground plan of the Mollusca or even the Trochozoa (e. g. presence of the prototrochal or velar muscle ring). On the one hand, A. stephanieae shows some features shared by all nudibranchs like the postmetamorphic condensation of the CNS, the possession of rhinophoral ganglia and the lack of oral tentacle ganglia as well as the de novo formation of the adult muscle complex. On the other hand, the structure and arrangement of the serotonergic apical organ is similar to other caenogastropod and opisthobranch gastropods supporting their sister group relationship.
Amphibians of Malawi : an analysis of their richness and community diversity in a changing landscape

This study summarizes the state of the knowledge of the amphibian diversity in Malawi highlighting the possible threats impending on this fauna correlated with human encroachment and land use change. New data about diversity, distribution and ecology have been gathered, whereas the old ones have been summarised, reviewed and commented. In order to put in context the responses of the amphibian communities to land use change, the main environmental characteristics of the country at a broad space and time scale have been explored. Furthermore, the original habitats and vegetation have been described, and their status in the present day Malawi discussed. In the same way, an overview of the actual state of the knowledge about the Malawian amphibians has been provided, and their ability to act as surrogate of environmental integrity in Sub-Saharan Africa commented on the basis of the available studies. Afterwards, the results of the study of the selected areas and samples have been analysed within this newly generated context. Different field and laboratory methods were applied for the quantitative analysis of the richness and diversity of the communities. Opportunistic search was used to detect species richness, whereas the visual encounter survey was applied to detect the relative abundance of species. Several indices of diversity and similarity, and extrapolations by means of true richness estimators were used for the analysis of the alpha and beta diversities. Additional information were gathered by means of pitfall traps with drift fence, and by the recording of the advertisement calls. Supplementary methods were applied for the analysis of the taxonomic composition of the collected material. In Malawi 84 amphibian species are recorded, two of which still undescribed (Leptopelis sp. and Phrynobatrachus sp.). Three further species need to be confirmed and might be possibly present too: Amietia viridireticulata, Hemisus guineensis, and Hyperolius minutissimus. Additionally, other unrecognised cryptic species — at least one — are present within the Hyperolius nasutus complex. Most of the species belong to the order Anura (82 species; 97.6%), whereas only two species belong to the Gymnophiona (2.4%). Anurans are divided into 12 families and 23 genera, whereas the two caecilians species into one family (Caecilidae) and two genera. The more diverse family is the Hyperoliidae (21 species, 25%) followed by the families Ptychadenidae (13 species, 15%), Arthroleptidae (11 species, 13%), Phrynobatrachidae (10 species, 12%), and Bufonidae and Pyxicephalidae (9 species, 11% respectively). The remaining high family diversity (seven families, Caecilidae included) is contrasted by a low number of species (11 species in total, 14%). Based on the available distribution data, the value of species richness of the anuran communities in Malawi is comprised between 5‒45 species. In average 16.8 ± 9.0 species (N=80) are to be found, 75% of the sites have less than 21 species, and only two sites have more than 25 species. Four hot spots of amphibian diversity were identified: the Nyika Plateau (24 species), Mangochi-Malombe (25 species), Zomba Plateau (32 species) and the Mulanje Massif (45 species). In the studied areas a mean of 14.7 ± 1.6 species was observed and extrapolations by means of the true richness estimators were in good agreement with this result. Among the studied areas the richest was Palm Forest Reserve (17 species), followed by Kaningina Forest Reserve (16 species) and Vinthukutu F. R., and Vwaza W. R (15 species). The poorest area was the Misuku Mountains with 12 species only and a slightly different ranking was generated by the true richness estimators. The mean of the species present in the samples was 4.8 ± 2.1 species, considerably less than the true species richness detected in the respective areas. Basing on the ranking generated by the K-dominance plot the most diverse samples were Palm F. R. and Misuku, whereas the less diverse were Kaningina F. R. and Fort Lister, confirmed by the values of the diversity indices. The main finding of this study was the observation of the lack of a clear match between environmental degradation and amphibian diversity, and the crucial importance of temporary water bodies for the preservation of the amphibian diversity. In fact, despite most of the original habitat formerly present in Malawi have been destroyed and replaced by cultivations, the amphibian communities of different areas showed a comparable diversity at both family and species richness level, and no evident match between environmental degradation and amphibian diversity was recognisable. Differences in species richness could mostly be explained by natural factors such the elevation gradient and the presence of temporary water bodies. However, it was not possible to exclude that the communities have changed during historical time and the shift in species composition already occurred together with the modification of their relative frequencies. Most of the species showed a remarkable ecological plasticity and several species were found in a variety of both natural and altered habitats. The classification of the Malawian amphibians on the basis of ecological guilds based on the available natural history data showed the preponderance (76%) of generalist pond breeders. As a consequence, most of these amphibians possessed a scarce capacity to act as surrogates of habitat integrity. Based on the result of this study the farm bush landscape with traditional agriculture practices bears a great potential to support amphibian diversity in terms of species richness, representing a compromise between local economic development and conservation. Furthermore, the results of this study indicate the outstanding importance of the southern-east region of Malawi for the conservation of the country’s amphibians.
Very little is known about the occlusal wear pattern in the Neanderthal posterior dentition. Usually dental wear is closely related to the physical properties of the ingested food, and consequently can be used to obtain information about diet. Neanderthal dietary reconstructions have been mostly based on the analysis of accompanying faunal remains and isotopic signatures of bones and tooth enamel, suggesting that they exploited larger portions of animal proteins from large and medium-sized herbivores. Probably these studies may do not reflect the bulk diet, tending to underestimate plant consumption and to overestimate meat consumption. In the present work the occlusal wear pattern of maxillary molars of Homo neanderthalensis (N=19) and early Homo sapiens (N=12)have been analyzed, applying non-destructive methods based on virtual three-dimensional polygonal models generated from surface scanning of dental casts. The sample groups occupied different geographical areas at different chronological times. The 3D digital tooth models were analyzed using the “Occlusal Fingerprint Analysis” (OFA) method (Kullmer et al. 2009), describing and quantifying the occlusal wear pattern derived from two wear facet angles (dip and dip direction), wear facet area and occlusal relief index (ORI). The OFA method provides information about the dynamics of the occlusal relationships and their function, permitting the reconstruction of the mandibular movements responsible for the contacts created during the chewing cycle. Since jaw movements and diet are closely related, the results obtained, can be used to interpret the diet of the two Pleistocene hominin species. In order to evaluate how dietary differences influence the occlusal wear pattern, upper molars of modern hunter-gatherers (N=42) with known diet and different dietary habits, have been included in the sample and compared with those of Neanderthals and early Homo sapiens. Results show that within the modern hunter-gatherers sample, the occlusal wear pattern of carnivorous populations differs from those who relied on a mixed-diet. In particular, the study of relative facet areas clearly distinguish meat-eaters from mixed-diet hunter-gatherers, while ORI results and wear facet inclinations (dip angle) seem to reflect directly the abrasiveness of the diet, including the influence of exogenous materials during food preparation. The Neanderthal occlusal wear pattern is characterized by an ecogeographic variation, suggesting the exploitation of different food resources. In particular Neanderthals who inhabited relatively warm environments of southern Europe and the Near East exhibit an occlusal wear pattern different from those of meat-eaters hunter-gatherers from tempered and cooler regions, displaying some features similar to those of Bushmen. These results suggest the exploitation of a broad variety of food sources. The analysis of the occlusal wear pattern in Neanderthals and early Homo sapiens who inhabited Europe during the cooler Oxygen Isotope Stage 3 (OIS3) shows many similarities between the two hominid species. These results indicate the exploitation of similar and low-diversified food sources, based mostly on the consumption of animal proteins, as suggested through the clear similarities with the wear patterns found in modern meat-eaters hunter-gatherers. In both studied groups, Neanderthals and early Homo sapiens the occlusal wear pattern is characterized by high ORI and dip angle values, suggesting the intake of a low-abrasive diet, probably due to the absence of sophisticated food preparation techniques introducing external silica grains, e.g. from soil (grinding of seeds) or plant cells, as those, seen in modern hunter-gatherer populations. The analysis of the occlusal fingerprints in Neanderthal and early European Homo sapiens upper molars suggests that both species followed very similar adaptive dietary strategies, based on a distinctive versatility and flexibility in the daily diet, depending on availability of resources according to environmental circumstances.
We investigate the utility of modern kernel-based machine learning methods for ligand-based virtual screening. In particular, we introduce a new graph kernel based on iterative graph similarity and optimal assignments, apply kernel principle component analysis to projection error-based novelty detection, and discover a new selective agonist of the peroxisome proliferator-activated receptor gamma using Gaussian process regression. Virtual screening, the computational ranking of compounds with respect to a predicted property, is a cheminformatics problem relevant to the hit generation phase of drug development. Its ligand-based variant relies on the similarity principle, which states that (structurally) similar compounds tend to have similar properties. We describe the kernel-based machine learning approach to ligand-based virtual screening; in this, we stress the role of molecular representations, including the (dis)similarity measures defined on them, investigate effects in high-dimensional chemical descriptor spaces and their consequences for similarity-based approaches, review literature recommendations on retrospective virtual screening, and present an example workflow. Graph kernels are formal similarity measures that are defined directly on graphs, such as the annotated molecular structure graph, and correspond to inner products. We review graph kernels, in particular those based on random walks, subgraphs, and optimal vertex assignments. Combining the latter with an iterative graph similarity scheme, we develop the iterative similarity optimal assignment graph kernel, give an iterative algorithm for its computation, prove convergence of the algorithm and the uniqueness of the solution, and provide an upper bound on the number of iterations necessary to achieve a desired precision. In a retrospective virtual screening study, our kernel consistently improved performance over chemical descriptors as well as other optimal assignment graph kernels. Chemical data sets often lie on manifolds of lower dimensionality than the embedding chemical descriptor space. Dimensionality reduction methods try to identify these manifolds, effectively providing descriptive models of the data. For spectral methods based on kernel principle component analysis, the projection error is a quantitative measure of how well new samples are described by such models. This can be used for the identification of compounds structurally dissimilar to the training samples, leading to projection error-based novelty detection for virtual screening using only positive samples. We provide proof of principle by using principle component analysis to learn the concept of fatty acids. The peroxisome proliferator-activated receptor (PPAR) is a nuclear transcription factor that regulates lipid and glucose metabolism, playing a crucial role in the development of type 2 diabetes and dyslipidemia. We establish a Gaussian process regression model for PPAR gamma agonists using a combination of chemical descriptors and the iterative similarity optimal assignment kernel via multiple kernel learning. Screening of a vendor library and subsequent testing of 15 selected compounds in a cell-based transactivation assay resulted in 4 active compounds. One compound, a natural product with cyclobutane scaffold, is a full selective PPAR gamma agonist (EC50 = 10 +/- 0.2 muM, inactive on PPAR alpha and PPAR beta/delta at 10 muM). The study delivered a novel PPAR gamma agonist, de-orphanized a natural bioactive product, and, hints at the natural product origins of pharmacophore patterns in synthetic ligands.
Das Nierenzellkarzinom (NZK) ist der häufigste maligne Tumor der Niere. In vielen Fällen sind bereits bei der Erstdiagnose Metastasen vorhanden oder entstehen im Verlauf der Therapie. Die Behandlungsmöglichkeiten für diese NZK-Patienten sind äußerst limitiert. Nahezu 2/3 der Betroffenen versterben an ihrer Erkrankung. Die Etablierung neuer Therapieansätze zur Behandlung des NZK ist dringend gefordert. Es wird postuliert, dass ein Therapiekonzept basierend auf dem Histondeacetylase (HDAC)-Inhibitor Valproat (VPA) kombiniert mit niedrig dosiertem Interferon (IFN)-alpha eine innovative und effiziente Behandlungsoption für austherapierte NZK-Patienten eröffnen könnte. In der vorliegenden Studie wurde der Einfluss einer VPA-Mono- versus VPA/IFN-alpha-Kombinationstherapie auf die malignen Eigenschaften verschiedener NZK-Zelllinien evaluiert. Mittels funktioneller Untersuchungen wurden Proliferations- und Adhäsionsphänomene unter den entsprechenden Therapien näher betrachtet. Fluorimetrische und molekularbiologische Studien dienten der detaillierten Aufklärung der den Veränderungen zugrunde liegenden Wirkmechanismen. Zur translationalen Gestaltung wurden zusätzlich tierexperimentelle Untersuchungen durchgeführt. VPA induzierte eine signifikante Reduktion der Proliferation von NZK-Zellen, die durch die additive Gabe von IFN-alpha weiter verstärkt wurde. Die anti-proliferativen Effekte korrelierten mit einer Zunahme der Zellen in der G0/G1-Phase und einer damit einhergehenden verminderten Anzahl an Zellen in der S-Phase. Die Verschiebung der Zellzyklusphasen war mit einer deutlichen Modulation relevanter regulatorischer Zellzyklusproteine assoziiert. Des Weiteren resultierte VPA und die korrespondierende Kombination mit IFN-alpha in einer signifikanten Inhibition der Adhäsion an Endothel und die extrazellulären Matrixproteine. VPA und die VPA/IFN-alpha-Kombination übten ihren Einfluss dabei offensichtlich über eine Modulation von Adhäsionsrezeptoren, implizit Integrine und CD44-Varianten, aus. Die antiproliferative und –adhäsive Wirkung war in der Regel nach längerer Inkubationszeit von 5 Tagen deutlich stärker als nach 3 Tagen. In analog behandelten normalen Nierenzellen zeigten sich im Vergleich keine solchen Effekte. Die Behandlung mit VPA und IFN-alpha scheint somit spezifisch maligne Zellen zu beeinflussen. VPA induzierte in den NZK-Zellen ferner die Reduktion von Protoonkogenen und MAP-Kinasen sowie die Zunahme von Tumorsuppressoren. Die zusätzliche Gabe von IFN-alpha resultierte in einer weiteren Wirkungsverstärkung gegenüber VPA allein. VPA und die Kombination mit IFN-alpha inhibierten zudem signifikant die HDAC6-Aktivität und -Proteinexpression der NZK-Zellen. Diese Hemmung ging mit einer Hyperacetylierung der Histone einher. Die epigenetische Modulation führte zur veränderten Genregulation und Transkription. So nahmen VPA und die korrespondierende Kombination neben den genannten funktionellen und molekularbiologischen Veränderungen maßgeblich Einfluss auf das Genexpressionsprofil der Tumorzellen. Die Expression negativer Regulatoren der Proliferation, Migration und Adhäsion sowie von Genen involviert in Differenzierung und Immunantwort wurden erhöht, wohingegen die Anzahl der Transkripte von Genen mitverantwortlich für die Ausbildung von Resistenzen und die Nährstoffversorgung der Tumoren reduziert wurde. Translationale tierexperimentelle Studien bestätigten die klinische Relevanz der VPA- und VPA/IFN-alpha-Behandlung, die in einer signifikanten Hemmung des Tumorwachstums resultierten. Die Wachstums-Inhibition war mit einer starken Modulation regulatorischer Proteine des Zellzyklus, der Apoptose und des HDAC-Systems assoziiert. Die vorliegenden Ergebnisse demonstrieren das viel versprechende Wirkungspotential von VPA und der korrespondierenden Kombination mit niedrig dosiertem IFN-alpha. VPAs anti-proliferative und -adhäsive Effekte in vitro und in vivo eröffnen die Perspektive für eine innovative Strategie in der Behandlung des NZK. Aufgrund der präsentierten Daten lässt sich postulieren, dass VPA und IFN-alpha die Grundlage für ein neues, effizientes Therapiekonzept bei austherapierten NZK-Patienten darstellen könnte.
Innerhalb der letzten Jahre ist weltweit eine progrediente Zunahme der Zahl am Typ II-Diabetes erkrankter Personen beobachtet worden, die sich nicht mehr nur in hochentwickelten Industriestaaten, sondern zunehmend auch in Entwicklungsländern zu manifestieren scheint. Dabei geht das größte Gesundheitsrisiko eines Typ II-Diabetes von einer Amputation der unteren Extremitäten aus, die oft die finale therapeutische Maßnahme in der Behandlung eines diabetischen Fußulkus darstellt. Ein gemeinsames Charakteristikum diabetischer Ulzerationen und anderer chronischer kutaner Wundheilungsstörungen ist neben einer allgemein schlechten Heilungsprognose eine fehlregulierte Entzündungsreaktion. In der vorliegenden Arbeit ist daher die Rolle von SOCS3 als potentem Bestandteil entzündungsdämpfender Regelkreise während der kutanen Wundheilung zunächst in stoffwechselgesunden und diabetischen Mäusen untersucht worden, um später ein SOCS3-überexprimierendes transgenes Mausmodell als Basis weiterführender Untersuchungen zu etablieren und in die Arbeit zu integrieren. Dabei konnte gezeigt werden, dass SOCS3 in stoffwechselgesunden Mäusen während der entzündlichen Akutphase der Heilung deutlich, aber zeitlich begrenzt hochreguliert ist und somit offensichtlich eine wichtige Funktion in der intrazellulären Regulation proinflammatorischer Signalkaskaden übernimmt. Im Gegensatz dazu zeigten heilungsdefiziente Wunden diabetischer ob/ob-Mäuse eine deutlich verlängerte Expression dieses Proteins, wobei sich herausstellte, dass sich diese vorwiegend auf Keratinozyten der unterentwickelten Epithelränder und der atrophischen Neoepidermis beschränkte. Aus dieser Entdeckung heraus entstand die zentrale Fragestellung dieser Arbeit, die darauf abzielte zu klären, ob eine fehlregulierte SOCS3-Expression in Keratinozyten des Wundrandes als zentrale Komponente in der Kausalkette diabetisch-chronischer Wundheilungsstörungen aufgefasst werden kann. Tatsächlich konnte anhand des transgenen Mausmodells in vivo und in vitro nachgewiesen werden, dass eine erhöhte intrazelluläre Verfügbarkeit von SOCS3 in Keratinozyten negativ mit deren Potenzial zur Proliferation und Migration interferiert. Das Protein SOCS3 besetzt somit im Prozess der kutanen Wundheilung eine inhibitorische Schlüsselposition in der Regulation essentieller Kernkompetenzen von Wundrandkeratinozyten, und dies als eigenständiges Element isoliert von Komplikationen, die sich sekundär aus einem diabetischen Phänotyp ergeben. In der Konsequenz zeigten stoffwechselgesunde transgene Mäuse deutliche Defizite in der Reepithelialisierung des Wundareals, die in vergleichbarer Form aus Wunden diabetischer ob/ob-Mäuse bekannt sind. Darüber hinaus konnte in SOCS3-überexprimierenden Mäusen eine für chronische Wunden typische, gesteigerte Entzündungsantwort im Wundgewebe detektiert werden, die sich durch eine verlängerte und erhöhte Präsenz neutrophiler Granulozyten und Makrophagen sowie eine verstärkte Expression proinflammatorischer Zytokine und Entzündungsmarker auszeichnete. Obwohl immunhistochemische Färbungen und Messungen proentzündlicher Mediatoren im Wundgewebe auf eine kritische Rolle SOCS3-überexprimierender Keratinozyten in der Bereitstellung proentzündlicher Mediatoren hindeuteten, konnten in vitro-Untersuchungen keinen proinflammatorisch veränderten Phänotyp transgener Keratinozyten bestätigen. Dies impliziert, dass die unkontrollierte Entzündungsantwort im transgenen Mausmodell sehr wahrscheinlich sekundär aus der retardierten Reepithelialisierung hervorging. Demnach ist denkbar, dass die SOCS3-bedingte retardierte Epithelregenerierung auch während der diabetisch-gestörten Wundheilung in der ob/ob-Maus eine bislang unterschätzte, zentrale Position in der Ätiologie der Heilungsstörung einnimmt. Die interessanteste Beobachtung ergab sich nach systemischer Neutralisierung des antiinflammatorischen Wachstumsfaktors TGF-β, nachdem eine drastische, offenbar gegenregulatorische Steigerung der TGF-β1-Expression im Wundgewebe heilungsdefizienter, transgener Mäuse detektiert wurde. Überraschender Weise führte die Neutralisierung von TGF-β in der transgenen Maus trotz der nach wie vor massiven Entzündung zu einer deutlich verbesserten Heilungssituation, die in diesem Fall nicht - wie aus der Literatur bekannt - auf eine Verbesserung der Wundreepithelialisierung, sondern eher auf eine erhöhte Kontraktilität der Wunden zurückzuführen war. Einerseits verweist dies erneut auf den potenten proliferationshemmenden Einfluss der hohen SOCS3-Spiegel. Andererseits rückt damit die Entzündungsreaktion in den Bereich eines Epiphänomens ab, da gezeigt werden konnte, dass eine erhöhte Wundinflammation offensichtlich nicht zwingend ein Charakteristikum darstellt, das per definitionem negativ mit dem Heilungsfortschritt interferieren muss.
Plant parasitic species of Asterinaceae and Microthyriaceae (Dothideomycetes, Ascomycota, Fungi) are inconspicuous foliicolous fungi with a mainly tropical distribution. They form black colonies on the surface of living leaves. Members of Asterinaceae and Microthyriaceae are characterized by shield-shaped, flat ascomata (thyriothecia) which grow completely superficially on the leaf cuticle. Microthyriaceae, Asterinaceae and other families of thyriothecia-forming ascomycetes belong to the class Dothideomycetes due to the presence of bitunicate asci. However, until today no consistent taxonomic concept nor molecular phylogenetic studies exist for the families of thyriothecioid ascomycetes. In the present thesis, 42 species belonging to 13 different anamorphic and teleomorphic genera of Asterinaceae, Microthyriaceae and ‘Pycnothyriales’ recently collected in Western Panama, are identified, described in detail and illustrated with drawings, transmission and scanning electron microscopical photographs. Among the 42 species, 37 species belong to the Asterinaceae, four species to the Microthyriaceae and one species to the from group ‘Pycnothyriales’. Two species of Asterinaceae are new to sience: Asterina gaiadendricola with an Asterostomella anamorph and Asterina schlegeliae with a Mahanteshamyces anamorph. Among the remaining species of Asterinaceae, 28 species represent new records for Panama: Asterina cestricola, A. ciferriana, A. consobrina, A. corallopoda, A. davillae with anamorph, A. diplocarpa, A. diplopoda, A. ekmanii, A. fuchsiae, A. manihotis, A. phenacis, A. radiofissilis with anamorph, A. siphocampyli, A. sponiae, A. stipitipodia with anamorph, A. styracina, A. tonduzii with anamorph, A. weinmanniae, A. zanthoxyli, Asterostomella dilleniicola, Asterolibertia licaniicola, Asterolibertia nodulosa, Cirsosia splendida with its Homalopeltis chrysobalani anamorph and Prillieuxina winteriana with its Leprieurina winteriana anamorph. The remaining 11 species of Asterinaceae probably respresent new species: Asterina spp. 1-8, Asterolibertia sp., Halbanina sp. and Mahanteshamyces sp. The four species of Microthyriaceae are new records for Panama: Maublanica uleana, Platypeltella irregularis, Platypeltella smilacis and Xenostomella tovarensis. The species Hemisphaeropsis magnoliae in the form group ‘Pycnothyriales’ is a new record for Panama. During this study, voucher material of 44 additional species of plant parasitic thyriothecioid ascomycetes was examined. Thereby, the number of species of Asterinaceae known for Panama since 2006 raises from four to 30, for Microthyriaceae respectively from zero to four and for ‘Pycnothyriales’ from zero to one. 21 of the presented species are new records for Central America and two species are new records for the American Continent. The presented 42 species parasitize 47 host plant species in 39 genera belonging to 28 plant families. For 23 fungal species, new host plant species are discovered. From those, seven belong to host plant genera not reported before to be parasitized by a member of Asterinaceae and Microthyriaceae: Burmeistera (Campanulaceae), Curatella and Davilla (Dilleniaceae), Greigia (Bromeliaceae), Hirtella (Chrysobalanaceae), Oxandra and Xylopia (Annonaceae). In this study, the first molecular phylogenetic approach in Asterinaceae is provided. For the first time, DNA was isolated from fresh material of Asterina spp. and their respective anamorphic stages on leaves in Panama. The hypothesis derived from SSU and LSU rDNA neighbour-joining analysis supports the monophyly of the Asterinaceae and suggests a close relationship to Venturiaceae within the class Dothideomycetes. The data obtained from the ppMP project (plant parasitic microfungi of Panama) indicate a constant but low abundance of plant parasitic thyriothecioid ascomycetes in natural plant communities in Panama, with Asterinaceae as the most species-rich and diverse family. Further collection activities in tropical regions worldwide will certainly increase our knowledge about species diversity and ecology of tropical plant parasitic thyriothecioid ascomycetes.
Die funktionelle Integrität des Endothels ist von essentieller Bedeutung für den Organismus. Die Entstehung und Progression vaskulärer Erkrankungen, wie z.B. der Atherosklerose, ist daher oftmals ursächlich mit einer Dysfunktion des Endothels verbunden. Vor diesem Hintergrund ist insbesondere die Aufklärung der molekularen Grundlagen der Regulation von Endothelzellfunktionen, ein zentraler Aspekt heutiger Forschung. Homeobox- (Hox) Transkriptionsfaktoren nehmen eine Schlüsselposition bei der Regulation einer Vielzahl zellulärer Prozesse, wie Proliferation, Migration und Gewebe-spezifischer Differenzierung ein. Die Identifikation sowie die Analyse der Funktion und Regulation von Hox-Transkriptionsfaktoren in Endothelzellen, leistet deshalb einen wichtigen Beitrag zum Verständnis der Endothelzellbiologie. Als ein zentraler Befund dieser Arbeit, konnte mit der Histon-Methyltransferase MLL erstmals die funktionelle Rolle eines epigenetischen Hox-Regulators auch in differenzierten Endothelzellen nachgewiesen werden. MLL erwies sich hierbei von essentieller Bedeutung für pro-angiogene Endothelzell-Funktionen. Die bedeutende Rolle von MLL bei der Migration von Endothelzellen konnte mit der transkriptionellen Regulation der beiden Hox-Transkriptionsfaktoren HoxA9 und HoxD3 in Verbindung gebracht werden, die hier erstmals als direkte Zielgene von MLL in Endothelzellen beschrieben wurden. Als funktionelle Mediatoren der MLLabhängigen Migration konnten zudem der EphB4-Rezeptor sowie die Integrine αVβ3 und α5β1, als Zielgene von HoxA9 bzw. HoxD3 nachgewiesen werden. Neben der Migration konnte für MLL auch eine essentielle Rolle für das Sprouting von Endothelzellen nachgewiesen werden, die sich im Gegensatz zur Migration, nicht auf die Regulation von HoxA9 oder HoxD3 zurückführen ließ. Diese Beobachtung lässt auf die Involvierung zusätzlicher MLLabhängiger Faktoren schließen, und verdeutlicht damit die zentrale Rolle von MLL bei der Regulation komplexer, pro-angiogener Prozesse in Endothelzellen. Über die genannte Rolle von MLL hinaus konnte im Rahmen dieser Arbeit das Wissen um Hox-Transkriptionsfaktoren mit funktioneller Relevanz für Endothelzellen, um die beiden Hox-Transkriptionsfaktoren HoxB4 und HoxB5 erweitert werden. Hier konnte für HoxB4 eine Rolle für die Fähigkeit von Endothelzellen zur Ausbildung zwei- und 3-dimensionaler Gefäßstrukturen nachgewiesen werden, während HoxB5 in die Proliferation, die Expression des endothelialen Markergens eNOS sowie die morphologische Beschaffenheit von Endothelzellen eingreift. Zusätzlich konnte die Rolle von transkriptionellen Hox Co-Faktoren, als Modulatoren von Hox-Funktionen, am Beispiel der Interaktion von Meis1 und HoxA9 bei der Transaktivierung des eNOS-Promoters aufgezeigt werden. Zusammenfassend leisten die hier gezeigten Daten einen Beitrag zum Verständnis der Rolle von Hox-Transkriptionsfaktoren als molekulare Regulatoren endothelialer Zellfunktionen.
In der vorliegenden Arbeit wurden zehn Substanzen, die im Rahmen des Projekts INTAFERE analytisch in verschiedenen Gewässersystemen des Hessischen Rieds nachgewiesen werden konnten, ökotoxikologisch charakterisiert. Neben der Bestimmung der Akut- und der chronischen Toxizität wurde auch das endokrine Potential mit Hilfe eines rekombinanten Hefe-Assays ermittelt.Die akute Toxizität zeigt zwischen den verschiedenen Substanzen große Differenzen. Die drei Organophosphate TCPP, TBEP und TCEP zeigen selbst bei hohen Konzentrationen keine oder nur sehr geringe Effekte, während 4-NP, 4-t-OP, BPA, TDCPP, AHTN und Terbutryn mit LC50-Werten bis zu 5 mg/l eine höhere Toxizität besitzen.Im Hefe-Assay kann in mehreren Versuchswiederholungen das östrogene Potential von 4-NP (MW: 6,71x10-6 M), 4-t-OP (MW: 7,16x10-6 M) und BPA (MW: 4,88x10-6 M), sowie die antiandrogene Wirkung des Bisphenols (5,29x10-5 M) bestätigt werden. Im Gegensatz dazu können im Yeast Antiestrogen Screen zum ersten Mal Hinweise auf die antiöstrogene Wirkung von TCPP (MW: 6,86x10-5 M), Terbutryn (MW: 3,99x10-5 M), TDCPP (MW: 2,65x10-6 M) und TBP (MW: 2,28x10-5 M) aufgezeigt werden.TBP und TDCPP führen auch in der chronischen Exposition bei Potamopyrgus antipodarum zu einer Reduktion in der Embryonenzahl (mehrfach beobachtete NOEC beider Substanzen: 6,25 mg/kg), ein Effekt, der zunächst nicht von einer toxischen Wirkung unterschieden werden kann. Allerdings scheint sich ein antiöstrogener Wirkmechanismus auf die Schnecken bei den Versuchen zur Mischtoxizität zu bestätigen, da die gleichzeitige Exposition gegenüber BPA und 4-tOP zu einer geringfügigen Aufhebung des Effekts führt. Potamopyrgus reagiert ebenfalls mit einer Reduktion der Embryonenzahl bei der Exposition gegenüber AHTN (mehrfach beobachtete NOEC: 2 mg/kg), zeigt sich aber insensitiv gegenüber der Belastung mit Terbutryn.Die Exposition von Chironomus riparius gegenüber den verschiedenen Substanzen führt bis auf eine Ausnahme zu keinen signifikanten substanzbedingten Beeinträchtigungen. Einzig das s-Triazin Terbutryn verursacht eine hohe Mortalität mit einer NOEC (28 d) von 250 μg/kg. Eine Toxizität auf den Anneliden Lumbriculus variegatus zeigt sich lediglich bei derExposition gegenüber BPA (NOEC: 10 mg/kg; Biomasse, 28 d) und 4-NP (EC10: 6,88 mg/kg; 95% KI: 3,97-11,9; Reproduktion, 28 d). Die durchgeführten Versuche zur Toxizität von Mischungen zeigen eine größere Gefährdung auf als bei Vorliegen der Einzelsubstanzen in Konzentrationen unterhalb ihres Schwellenwertes zu vermuten wäre. Allerdings lassen sich die mit dem Hefe-Assay erzielten Ergebnisse aufgrund großer Variabilitäten zwischen den einzelnen Versuchswiederholungen nicht immer eindeutig mit Hilfe des additiven oder des unabhängigen Modells beschreiben, zeigen dabei jedoch trotzdem ein gegenüber den Einzelsubstanzergebnissen verändertes Risiko auf. Um diesem in der Risikobewertung Rechnung zu tragen, wird ein Verfahrensvorschlag entwickelt, in dem durch zusätzliche Sicherheitsfaktoren für PNECs ein Überschreiten des risikoanzeigenden Werts von 1 des PEC/PNEC-Quotienten einer Mischung verhindert werden kann.
A highly efficient method for chromosomal integration of cloned DNA into Methanosarcina spp. was developed utilizing the site-specific recombination system from the Streptomyces phage PhiC31. Host strains expressing the PhiC31 integrase gene and carrying an appropriate recombination site can be transformed with non-replicating plasmids carrying the complementary recombination site at efficiencies similar to those obtained with self-replicating vectors. We have also constructed a series of hybrid promoters that combine the highly expressed M. barkeri PmcrB promoter with binding sites for the tetracycline-responsive, bacterial TetR protein. These promoters are tightly regulated by the presence or absence of tetracycline in strains that express the tetRgene. The hybrid promoters can be used in genetic experiments to test gene essentiality by placing a gene of interest under their control. Thus, growth of strains with tetR-regulated essential genes becomes tetracycline-dependent. A series of plasmid vectors that utilize the site-specific recombination system for construction of reporter gene fusions and for tetracycline regulated expression of cloned genes are reported. These vectors were used to test the efficiency of translation at a variety of start codons. Fusions using an ATG start site were the most active, whereas those using GTG and TTG were approximately one half or one fourth as active, respectively. The CTG fusion was 95% less active than the ATG fusion.
