Since the late 1950's Paul Celan has been deeply interested in scientific questions in regard to history and society as well as in the terminology of scientific language. Therefore, since the publication of “Sprachgitter” up to “Schneepart”, many of his poems include terms out of texts that deal with topics as different as geology, biology, astronomy, nuclear physics, and medicine. This essay addresses the problem of this form of adaptation by a close reading of Celan's poem »In der Blasenkammer« (published in 1970). Celan's poems turn against any notion of ›communicative functionality‹ of language; especially the languages of the sciences are here a prominent challenge because of their claim to be semantically objective. So the mentioned adaptation is a serious problem indeed, because the turning of scientific terms into words of poetry does not imply that these terms are simply transferred from one determined semantic context into another. It is rather the question of semantics itself that is at stake. So it has to be stressed that Celan does neither intend to equip scientific terms with ›poetical connotations‹ nor does he turn them into simple metaphors nor poetic or metapoetic concepts. His poems rather deal with the dispensation of semantics and metaphoricity – the language of these poems is a language of abundance and privation at the same time.
Neue Synthesestrategien als Thema des Chemieunterrichts - Experimente zur Kombinatorischen Chemie

In der vorliegenden Dissertation wurde die experimentelle Umsetzung der Thematik Kombinatorische Chemie für den Chemieunterricht in der Sekundarstufe II ausgearbeitet. Dazu wurden geeignete Experimente erarbeitet, die einen Einblick in die Kombinatorische Chemie und somit in die neuen Arbeitmethoden der heutigen Chemie schaffen. Mit deren Hilfe gängige Synthesemethoden wie die Split and Combine-Synthese, Teabag-Methode, Spot-Methode und Pin-Methode aufgezeigt werden können und von den Schülern selbst durchgeführt werden. Als ein vorteilhaftes Modell zum Aufzeigen der Arbeitsweise der Kombinatorischen Chemie haben sich Farbstoffe erwiesen, da sie schnell darstellbar sind und keiner aufwendigen Detektion der entstandenen Produkte bedürfen. Als erstes Farbstoffsystem wurden Azofarbstoffe ausgewählt, da sie auch im fakultativen Themenbereich „Farben“ im Chemieunterricht thematisiert werden. Die Übertragung der normalerweise in Lösung hergestellten Azofarbstoffe auf die Festphasenchemie musste zuerst eine geeignete und kostengünstige Festphase gefunden werden. Hierbei haben sich im Handel erhältliche Ionenaustauscher (Harze ohne Linker), die auch zur Wasseraufbereitung verwendet werden, als geeignet herausgestellt. Die erforderlichen Reaktionsbedingungen der Lösungsreaktionen wurden erprobt, optimiert und auf die Festphasenchemie übertragen. Eine Übertragung der Syntheseprinzipien auf drei weitere Farbstoffsysteme wie der Fluoreszenzfarbstoffe der Pyrydin- und Pyrylium-Reihe sowie der Oxidationsfarbstoffe schloß sich an. Nunmehr stehen für den schulischen Einsatz, aber auch für die Ausbildung, einfache Systeme zur Verfügung, die anhand schnell und problemlos durchzuführender Experimente einen Zugang zu unterschiedlichen Methoden der Kombinatorischen Chemie vermitteln können. Dabei bieten die Experimente Anregungen für weitere Arbeiten. Als problematisch stellte sich jedoch der Zugang zu mehrstufigen kombinatorischen Synthesen auf dem Gebiet der Farbstoffe dar. Hier stößt man mit den aus didaktischen Überlegungen sehr einfach gewählten Beispielen an eine Grenze. Ergänzend wurde die Thematik Kombinatorische Chemie mit hochbegabten Schülern der Internatsschule Schloß Hansenberg und interessierten Schülern aus dem Raum Frankfurt, die an dem Projekt Science Camp teilnahmen, erprobt und das Verständnis für diese Thematik mittels eines Fragebogens untersucht. Ein direkter Vergleich beider Schülergruppen konnte aufgrund unterschiedlicher schulischer Bildung nicht vorgenommen werden, weshalb die Auswertung lediglich erste Eindrücke vermittelt. Danach ist das bisher ausgearbeitete Konzept in Lerngruppen mit hochbegabten Schülern gut einsetzbar. Ein Vergleich mit durchschnittlich leistungsstarken Lerngruppen steht noch aus. Eine wünschenswerte Erweiterung und ein Ausbau der Thematik aus Sicht der Schulchemie sollte anorganische Systeme mit einbeziehen und Strategien der Dekonvolution.
Der (...) Beitrag setzt sich strukturell auf drei Ebenen mit Rolf Hochhuths Dramenpoetik auseinander. Erstens wird die gegenläufige Beziehung zwischen ihrem historisch-adäquaten Gestaltungsanspruch und ihrer literarisch-symbolhaften Überformung analysiert. Zweitens wird Hochhuths Verhältnis zu den für ihn poetologisch bedeutsamen Vorgängern Lessing und Schiller untersucht. Drittens geht es um die Frage, welche Konsequenzen sich aus der in den »Guerillas« formulierten Darstellungsabsicht ziehen lassen, der realen Wirklichkeit durch die Projektion einen neuen (literarischen) Wirklichkeit entgegenzutreten.
The Birds of the Highlands of South-West Saudi Arabia and adjacent parts of the Tihama: July 2010

The objective of the survey was to compare habitats and bird life in the Asir region, particularly Jebal Souda and the Raydah escarpment protected area of the Saudi Wildlife Commission, and adjacent regions of the tihama, with those observed in July 1987 (Jennings, et al., 1988). The two surveys were approximately the same length and equal amounts of time were spent in the highlands and on the tihama. A number of walked censuses were carried out during 2010 on Jebal Souda, using the same methodology as walked censuses in 1987, and the results are compared. Broadly speaking the comparison of censuses revealed that in 2010 there were less birds and reduced diversity on the Jebal Souda plateau, compared to 1987. However in the Raydah reserve the estimates of breeding bird populations compiled in the mid 1990s was little changed as far as could be assessed in 2010. The highland region of south-west Saudi Arabia, especially Jebal Souda, has been much developed since the 1987 survey and is now an important internal recreation and resort area. This has lead to a reduction in the region’s importance for terraced agriculture. These changes may be a contributing factor to changes in bird numbers on the plateau. Subsidiary tasks that arose during the 2010 survey were to help locate satellite tagged Bald Ibises Geronticus eremita from Syria which were transiting Saudi Arabia at the time. Secondly to search for the Asir subspecies of the Eurasian Magpie Pica pica asirensis, which is endemic to the south-west Saudi Arabia highlands, and is reported to be in decline. A separate team searching for the Bald Ibis located some individuals and one was found dead. Few Magpies were located and it seems clear that this very scarce bird has declined further in numbers in recent years. A number of interesting records of birds were obtained, especially on the tihama, where two new birds for Saudi Arabia were observed, Black-headed Heron Ardea melanocephala and Painted Snipe Rostratula benghalensis and one species, Glossy Ibis Plegadis falcinellus was found breeding for the first time in the Arabian Peninsula. Some recommendations for the protection and management of the Raydah reserve are presented. A systematic list of all birds seen is provided.
During the period 24 May to 1 June 2010 observations were made of the flora, fauna and habitats of two study areas in the Rub al Khali in the Eastern Province of Saudi Arabia. Firstly the route of an existing pipeline, known as Shabab 1, which runs from the Jafurah desert south of Abqaiq to a point 211 km west of the Shaybah oil field, a distance of 422 km. Secondly the Shaybah oilfield area (administered by Saudi ARAMCO), paying particular attention to the sites listed at paragraph 6. (See also the map at Fig. 1.) In the following paragraphs the two sites are often identified simply as Shabab and Shaybah.
Eine wichtige Eigenschaft des menschlichen Gehirns besteht in der Fähigkeit, flexibel auf eintreffende Reize zu reagieren und sich den Anforderungen und Veränderungen der Umwelt anzupassen. Anpassung oder Adaptation lässt sich in vielen Situationen beobachten. Beispielsweise kommt es in der Retina beim Übergang von einer sehr hellen Umgebung in eine dunkle Umgebung zu Anpassungsleistungen. Neuronale Adaptation wird in den Neurowissenschaften genutzt, um Aussagen über die Funktion bestimmter Hirnareale machen zu können. In sogenannten Adaptationsexperimenten werden Stimuli wiederholt dargeboten und die dadurch erzeugten neuronalen Antworten in verschiedenen Hirnarealen miteinander verglichen. Nimmt das Signal in einem Areal ab, dann wird daraus geschlossen, dass die Zellen in diesem Bereich an der Verarbeitung des Stimulus beteiligt waren. Wiederholte Reizdarbietung führte in zahlreichen Untersuchungen zu einer Abnahme der neuronalen Antwort. Daneben wurde jedoch auch der gegenteilige Effekt, eine Verstärkung der neuronalen Antwort, bei Wiederholung eines Reizes nachgewiesen. In der vorliegenden Arbeit wurde die Verarbeitung frequenzmodulierter Töne im auditorischen Kortex des Menschen mit Hilfe eines Wiederholungsparadigmas untersucht. Frequenzmodulationen sind eine beim Menschen noch wenig untersuchte Reizklasse, die in natürlichen Geräuschen und besonders in der menschlichen Sprache eine wichtige Rolle spielen. Ausgangspunkt dieser Arbeit war die Frage, ob sich im auditorischen Kortex des Menschen eine Sensitivität für die Richtung einer Frequenzmodulation nachweisen lässt. Dieser Frage wurde mit drei Magnetenzephalographie-Studien nachgegangen. In Studie 1 wurde ein Zwei-Ton-Paradigma angewendet. Dabei wurde in jedem Durchgang ein frequenzmodulierter Ton jeweils zwei Mal präsentiert. Lediglich die Richtung, in der der Ton abgespielt wurde, also von den niedrigen zu den hohen oder von den hohen zu den niedrigen Frequenzen, wurde variiert. Die beiden frequenzmodulierten Töne in Studie 1 hatten eine Dauer von 500 ms und wurden in einem Abstand von 1 Sekunde präsentiert. Mit dieser Versuchsanordnung sollte untersucht werden, ob es bei Wiederholung der Frequenzrichtung zu einer Abnahme des neuronalen Signals kommt. Diese Abnahme wurde vor allem in der N1m-Komponente aber auch in späteren Komponenten wie der N2m erwartet. Der Vergleich der N1m-Amplitude für den zweiten Ton zeigte jedoch nur geringe Unterschiede zwischen den Bedingungen. Die Wiederholung derselben Frequenzrichtung bewirkte nur eine schwache Abnahme des Signals. Deutliche Adaptationseffekte konnten nicht gefunden werden. Daneben zeigten sich Hemisphärenunterschiede bei der Verarbeitung der frequenzmodulierten Töne. Über den Sensoren der rechten Hemisphäre war die Antwort signifikant stärker ausgeprägt als über der linken Hemisphäre. Als mögliche Erklärung für die schwach ausgeprägten Adaptationseffekte in Studie 1 wurde der zeitliche Aufbau des Paradigmas herangezogen. In der zweiten Studie wurde daher sowohl die Dauer der präsentierten Stimuli als auch der zeitliche Abstand zwischen den beiden Tönen reduziert. Dieses Paradigma führte zu signifikanten Unterschieden in der Reaktion auf den zweiten Reiz. Entgegen der Erwartung einer Adaptation bei Reizwiederholung bewirkte die Wiederholung derselben Frequenzrichtung eine signifikant höhere neuronale Antwort im Vergleich zu der Präsentation einer abweichenden Frequenzrichtung. Diese Unterschiede traten auf der rechten Hemisphäre über einen Zeitraum von 150 bis 350 ms nach Beginn des zweiten Stimulus auf, während sich auf der linken Hemisphäre 200 bis 300 ms nach Beginn des zweiten Tons signifikante Unterschiede zwischen gleichen und unterschiedlichen Frequenzrichtungen zeigten. In der N1m-Amplitude zeigten sich dagegen keine Wiederholungseffekte. Ähnlich wie in Studie 1 traten auch in Studie 2 Hemisphärenunterschiede auf. Für die Sensoren der rechten Hemisphäre waren die Verstärkungseffekte stärker und über einen längeren Zeitraum zu beobachten. Das unerwartete Ergebnis von Studie 2 stellte die Motivation für den Aufbau der dritten Studie dar. Mithilfe dieser Studie sollte überprüft werden, welche Rolle das Inter-Stimulus-Intervall auf die Verarbeitung eines nachfolgenden Stimulus hat. Zu diesem Zweck wurde in Studie 3 die Länge des ISIs zwischen 100 und 600 ms variiert. Damit sollte zum einen überprüft werden, innerhalb welchen zeitlichen Bereichs es zu einer Verstärkung des Signals kommt und wann beziehungsweise ob es ab einem bestimmten zeitlichen Abstand zwischen den Stimuli zu Adaptationsprozessen kommt. Bei dem kürzesten ISI von 100 ms führte die Wiederholung derselben Frequenzrichtung zu einer signifikant stärkeren N1m-Amplitude als bei der Präsentation einer abweichenden Frequenzrichtung. Bei ISIs > 100 ms zeigte sich keine höhere N1m-Amplitude mehr bei Wiederholung derselben Frequenzrichtung. Deutliche späte Effekte wie sie in Studie 2 über einen Zeitbereich von 150 bis 300 ms nachgewiesen wurden, traten in Studie 3 nicht auf. Bei einem ISI von 300 bis 500 ms waren leichte Verstärkungseffekte in einem Zeitbereich von 200 bis 400 ms zu beobachten. Bei einem ISI von 600 ms zeigten sich keine Unterschiede zwischen den Bedingungen. Durch Studie 3 konnte der in Studie 2 gefundene Effekt in einen größeren Zusammenhang gestellt werden. Zu einer Verstärkung der N1m-Komponente kommt es lediglich bei einem ISI von 100 ms. Liegen die Stimuli 200 ms auseinander, findet eine Verstärkung der späteren Komponenten statt, die bei ISIs > 200 ms immer weiter abnimmt. Offen bleibt, welche Abläufe zu der Verstärkung des Signals bei Wiederholung der Frequenzrichtung geführt haben. Um die dem Verstärkungseffekt zugrunde liegenden Prozesse zu verstehen, sind weitere Studien nötig.
Qualität und Qualitätsmanagement in der universitären naturwissenschaftlichen Lehrerfortbildung

Vor dem Hintergrund der politischen Entwicklungen in Hessen und der allgemeinen Debatte über Qualität im Bildungsbereich sollte im Lehrerfortbildungszentrum Chemie (lfbz-Chemie) der Universität Frankfurt am Main eine systematische Qualitätsentwicklung etabliert und dieses Vorhaben wissenschatflich begleitet werden. Die wissenschaftliche Arbeit besteht aus drei Säulen: - Eine erste, qualitative Studie sollte Qualitätskriterien und -indikatoren liefern. - In einer Fallstudie am lfbz-Chemie Frankfurt sollte die Einführung von Instrumenten des Qualitätsmanagements aus dem gewerblichen Weiterbildungsbereich beobachtet und bewertet werden. - Eine bundesweite Umfrage unter universitären Fortbildungsanbietern im naturwissenschaftlichen Bereich sollte sowohl die Struktur der anbietenden Institutionen als auch die allgemeine Einstellung der Befragten zum professionellen Qualitätsmanagement (QM) nach einem im gewerblichen Bereich geläufigen Modell (LQW) beleuchten. Die Qualitätsindikatoren und -kriterien fielen sehr spezifisch für die naturwissenschaftliche Lehrerfortbildung aus. Allein die drei folgenden Bereiche erfassen zwei Drittel aller Nennungen: die „Qualität der Teamer bzw. Moderatoren“, die „Fortbildungsgestaltung“ und die „Zielgruppenorientierung (Schulbezug)“. Nimmt man die Anzahl der Indikatoren und Kriterien pro Bereich als Maßstab, fokussieren die Befragten sehr stark auf Anforderungen, die die Professionalität des Personals betreffen. Zur Orientierung für die Qualitätsarbeit am lfbz-Chemie (Fallstudie) wurde das QM-Modell LQW 2 gewählt; als kleinere Instrumente kamen die Tabellen nach dem Vorbild der Balanced Scorecard (BSC) nach Kaplan und Norton sowie die Stärken-Schwächen-Analyse zum Einsatz. Als erstes Ergebnis der Fallstudie kristallisierten sich drei Arbeitsbereiche heraus: Personalorganisation, Evaluation und Innovationen. Diese Arbeitsfelder ergänzten die identifizierten Anforderungen an Fortbildung somit um Bereiche, die eher der organisatorischen Ebene zuzuordnen sind. Es ließ sich in der Fallstudie feststellen, dass ein ganzes Qualitätsmanagementmodell wie LQW 2 nur als Anregung für den einen oder anderen, als wichtig erachteten Bereich dienen konnte, während sich kleinere Instrumente eher als handhabbar erwiesen. Außerdem konnte postuliert werden, dass - ein persönlicheres Eingehen auf die Ziele und Aufgaben der einzelnen Personen im Sinne eines Total Quality Managements vermutlich zu besseren Ergebnissen geführt hätte als die Konzentration auf allgemeine Themen und die Orientierung am eher abstrakten Modell, - Erfolge sehr nachdrücklich kommuniziert werden müssen und - möglichst eine Person aus dem Stammpersonal für den Bereich Qualität verantwortlich sein sollte. Um ergänzend zur Fallstudie zu allgemeineren Schlüssen gelangen zu können, wurde eine Charakterisierung der universitären Fortbildungsanbieter mittels Clusteranalyse mit Daten der bundesweiten Umfrage durchgeführt. Sie zeigte vier Typen von Anbietern auf. Insgesamt fanden sich neben der Gruppe der großen Anbieter, zu denen auch das lfbz-Chemie gehört, nur vergleichsweise kleine Anbieter. Die in der Fallstudie erhaltenen Ergebnisse können somit nur eingeschränkt auf die meisten Anbieter übertragen werden. Insgesamt konnten Stärken und Schwächen der universitären Anbieter identifiziert werden, mit denen sich die universitäre naturwissenschaftliche Lehrerfortbildung von der gewerblichen Weiterbildung abhebt. Stärken lagen vor allem in den besonderen Kompetenzen, die an der Hochschule anzutreffen sind. Problemfelder lagen ausgerechnet hauptsächlich im Personalbereich. Als spezifische Schwächen der universitären Anbieter im Personalbereich kann z. B. die Stellung der Fortbildung als Nebenaufgabe oder der Verlust von Know-how durch die Fluktuation des Personals gelten. Auch allgemein kann ein Modell wie LQW 2 als Anregung für die Qualitätsarbeit in der universitären Lehrerfortbildung dienen, nicht jedoch umfassend umgesetzt werden. Die „kleineren“ Instrumente können dagegen eher als allgemein geeignet betrachtet werden. Während die „großen“ Anbieter durch verbesserte personelle Ressourcen für Aufgaben wie die Qualitätsarbeit unterstützt werden könnten, bietet sich für die „kleinen“ Anbieter eher die Unterstützung durch eine zentrale Stelle der Universität an, um die Fortbildungsanbieter doch noch in ein professionelles Qualitätsmanagement einbinden zu können.
Lorenz von Stein und die Französische Revolution : Zeitgeschichte als Theorie der Gesellschaft

In der Therapie des Diabetes mellitus Typ 2 ist neben Patientenschulung, Fewichtsreduktion und körperlicher Aktivität die Therapie mit oralen Antidiabetika von besonderer Bedeutung, um langfristig die Blutzuckereinstellung zu verbessern und kardiovaskuläre wie andere Folgeerkrankungen zu minimieren. Allerdings sinkt im Verlauf die Effizienz dieser Therapie, so dass die dauerhafte Gabe von Insulin notwendig wird. Dabei können jedoch auch schwerwiegende Nebenwirkungen, wie z.B. Hypoglykämien und eine wiederrum mit einem erhöhten Risiko an kardiovaskulären Erkrankungen verbundene ausgeprägte Gewichtszunahme auftreten. Demgegenüber führte der GLP-1 Rezeptor-Agonist Exenatide in jüngsten Studien zu einer Verbesserung der Blutzuckereinstellung und Reduktion des Körpergewichts und kann somit als eine neuartige Therapiealternative zum bisher verwendeten Insulin angesehen werden. In der hier vorliegenden prospektiven Studie wurde Exenatide mit Insulin bei Typ 2-Diabetes Patienten mit einer unzureichenden Stoffwechseleinstellung unter Metformin in Bezug auf deren Blutzuckereinstellung und das Hypoglykämie-Risiko verglichen. In dieser 26-wöchigen, multizentrischen, kontrollierten, randomisierten und zweiarmigen Phase IIIb-Studie wurden insgesamt 494 Patienten aus ganz Deutschland untersucht. Die Patienten mit einer unzureichenden Metformin- (Primärkollektiv) oder Kombinationstherapie (exploratives Kollektiv: Metformin plus Sulfonylharnstoff) wurden im Verhältnis 3:1 auf zweimal täglich Exenatide oder zweimal täglich Mischinsulin Aspart 30/70 randomisiert. In einem hierarchischen Modell wurde zunächst die Nichtunterlegenheit von Exenatide gegenüber dem Mischinsulin im Hinblick auf das primäre Endziel der Blutzuckereinstellung (in Form des HbA1c-Wertes) untersucht und anschließend die Überlegenheit von Exenatide in Hinblick auf ein geringeres Hypoglykämie-Risiko. Sekundäre Studienziele waren die Häufigkeit von schweren und von nächtlichen Hypoglykämien, die Veränderung des Körpergewichts sowie des Body Mass Index. Ferner wurden 7-Punkt-Blutzuckertagesprofile und Patientenfragebögen in die Auswertung einbezogen. Bei der insgesamt in acht Visiten unterteilten Studie erfolgte nach zwei Wochen die Randomisierung auf die beiden Therapie-Arme. Während die Dosierung des Mischinsulins im Verlauf der gesamten Studiendauer vom Arzt individuell titriert werden konnte, wurde die Dosis von Exenatide nach vier Wochen von 2 x 5 μg/Tag auf 2 x 10 μg/Tag verdoppelt und bis zum Ende der Studie nicht verändert. Die hier vorliegende Arbeit ist die erste Studie, die das Hypoglykämie-Risiko unter Exenatide und Mischinsulin als primären Endpunkt prospektiv und konfirmatorisch vergleichend untersucht. Im Primärkollektiv konnte die Nicht-Unterlegenheit von Exenatide zum Mischinsulin in Bezug auf den HbA1c-Wert festgestellt werden. Bei der Vermeidung von Hypoglykämien war Exenatide dem Mischinsulin statistisch signifikant überlegen. Zusätzlich konnte unter der Exenatide-Behandlung ein signifikanter Gewichtsverlust festgestellt werden, wohingegen die Patienten im Mischinsulin-Arm signifikant an Gewicht zunahmen. Die gleichzeitige Erreichung dieser Therapieziele (HbA1c<7,0%, keine Gewichtszunahme und keine Hypoglykämien) wurde als Triple-Endpoint definiert und im Exenatide-Arm signifikant häufiger als unter der Mischinsulin-Therapie beobachtet. In den 7-Punkt-Blutzuckertagesprofilen konnte neben einer allgemeinen Blutzuckersenkung in beiden Therapiearmen eine zusätzliche postprandiale Senkung zu den Zeiten der morgendlichen und abendlichen Exenatide-Injektion beobachtet werden. Demgegenüber waren die präprandialen Blutzuckerwerte im Mischinsulin-Arm niedriger, so dass insgesamt eine vergleichbare Verbesserung des Flächenintegrals in den Blutzuckertageskurven erzielt wurde, was die vergleichbare HbA1c-Verbesserung erklärt. Entsprechend zum Gewicht nahmen auch BMI und Hüftumfang der Patienten unter Exenatide ab und unter Insulin zu. Das Gesamtcholesterin und die Triglyceride sanken unter beiden Therapien leicht ab, HDL-Cholesterin stieg leicht an, wohingegen nur unter Exenatide das LDL-Cholesterin abnahm. Ebenfalls sanken nur im Exenatide-Arm der systolische und diastolische Blutdruck. Die häufigsten unerwünschten Ereignisse unter Exenatide waren Übelkeit, Erbrechen, Verdauungsstörungen, Durchfall, Kopfschmerzen und Erkältungen, wobei Durchfall, Kopfschmerzen und Erkältungen auch unter Insulin zu beobachten waren. Bei der in den Patientenfragebögen DTSQ und SF-12 dokumentierten subjektiven Empfindung der Nebenwirkungen zeigten sich allerdings eine Kompensation der deutlicheren Nebenwirkungen von Exenatide durch dessen Vorteile, u.a. im Hinblick auf seine einfachere Dosierung und der als angenehm empfundenen Gewichtsreduktion. Insgesamt war in beiden Therapie-Armen eine ausreichende Patientenzufriedenheit festzustellen. Unter denjenigen Patienten, bei denen Antikörper gegen Exenatide festgestellt wurden, nahm die Prävalenz einer Antikörperbildung gegen Exenatide bis zum Ende der Studie ab. Die Ergebnisse der hier vorgelegten Studie lassen die Schlussforderung zu, dass der GLP-1 Agonist Exenatide eine therapeutische Alternative zum Mischinsulin in der Behandlung des Diabetes Mellitus Typ 2 darstellt. Da eine Restfunktion der pankreatischen ß-Zellen eine Voraussetzung für den Therapieerfolg darstellt, sollte Exenatide möglichst früh bei nachlassender Wirkung des oralen Antidiabetikums Metformin eingesetzt werden. Hier wirkt Exenatide durch eine Verbesserung der Blutzuckereinstellung, die zudem mit einem verminderten Hypoglykämie-Risiko und einer Gewichtsreduktion verbunden ist. Die Kombination dieser drei bedeutenden Effekte hat den klinisch relevanten Vorteil, das Risiko an kardiovaskulären Spätfolgen und somit die Morbidität und Mortalität von Typ 2-Diabetikern langfristig zu senken. Dieses sollte in einer prospektiven Langzeitstudie untermauert werden.
Immer wieder sind Vorschläge vorgebracht worden, die Herkunft des im Deutschen insgesamt marginalen s-Plurals nicht nur bei importierten Fremdwörtern, sondern auch bei den Eigennamen zu verorten […]. Konkretisiert und diachron rekonstruiert wurde dieser Gedanke indessen noch nicht. In diesem Beitrag vertreten wir die Position, dass der s-Plural im Deutschen und Niederländischen dort, wo er – seiner gestaltschonenden Funktion wegen – am stärksten benötigt wird, nämlich bei Eigennamen, Onomatopoetika, Kurzwörtern und Substantivierungen, auch entstanden ist, konkret bei den Eigennamen, genauer: den Familiennamen. Dort hat er sich aus dem ursprünglichen Genitiv-Singular-Flexiv entwickelt. Dabei liefert nicht nur die germanisch-kontrastive, sondern auch und vor allem die dialektale Perspektive deutliche Evidenz für diese neue Sicht, haben doch manche deutsche Dialekte eine Art sprachlichen Archäopteryx vom Typ 's Müllers sin do '(die) Müllers sind hier', wörtlich: 'des Müllers sind hier' bewahrt […]. Für das Niederländische lässt sich nachweisen, dass sich die dialektalen s- und en-Pluralgebiete auffallend deutlich mit s- und en-Genitiv-Singular-Gebieten patronymischer Familiennamen decken. Auch hier lassen sich folglich die Familiennamen als Keimzelle der Reanalyse vom Kasus- zum Numerusmarker identifizieren.
The title compound. C15H14N2O4, (I), has a gauche–gauche (O/C/C/C—O/C/C/C or GG) conformation and is a positional isomer of propane-1,3-diyl bis(pyridine-3-carboxylate), (II). The molecule of (I) lies on a twofold rotation axis, which passes through the central C atom of the aliphatic chain, giving one half-molecule per asymmetric unit. There is excellent agreement of the geometric parameters of (I) and (II). The most obvious differences between them are the O/C/C/C—O/C/C/C torsion angles [56.6 (2)° in (I) and 174.0 (3)/70.2 (3)° in (II) for GG and TG conformations, respectively] and the dihedral angle between the planes of the aromatic rings [80.3 (10)° in (I) and 76.5 (3)° in (II)]. The crystal structure is stabilized by weak C—H ... N and C—H ... O hydrogen bonding.

In the title compound, C17H11NO4, the dihedral angle between the two benzene rings is 8.66 (3)°. The nitro group is twisted by 4.51 (9)° out of the plane of the aromatic ring to which it is attached. The presence of intermolecular C—H ... O contacts in the crystal structure leads to the formation of chains along the c axis.
In the crystal of the title compound [systematic name: 2-(3,5-diamino-6-chloropyrazin-2-ylcarbonyl)guanidinium chloride methanol disolvate], C6H9ClN7O+·Cl-·2CH3OH , the components are connected by N—H ... N, N—H ... Cl, N—H ... O, O—H ... Cl and O—H ... O hydrogen bonds into a three-dimensional network. The dihedral angle between the aromatic ring and the guanidine residue is 6.0 (2)°.
In contrast to the previous structure determinations of the title structure, (NH4)2[MoS4], the present determination at 173 K localized the positions of the H atoms. The title structure belongs to the beta-K2SO4 family and all the ions are located on crystallographic mirror planes. The ions are held together by N—H ... S hydrogen bonds (some of which are bifurcated), forming a three-dimensional network. One of the N atoms has nine contacts to the S atoms shorter than 4 Å, and the other has ten.
The title compound (also know as azorellanone), C20H32O2, is built up from three fused carbocycles, one five-membered ring and two six-membered rings. The five membered-ring has an envelope conformation, whereas the six-membered rings have a distorted half-chair and a twist–boat conformation. In the crystal, molecules are linked by O—H ... O interactions into zigzag chains with graph-set notation C(8) along [010]. The absolute configuration was assigned on the basis of earlier chemical studies.
The dihydropyrimidine ring of the title compound, C13H15ClN2S, adopts an envelope conformation with five almost coplanar atoms (r.m.s. deviation = 0.054 Å) and the C atom bearing the two methyl substituents deviating from this plane by 0.441 (2) Å. The best plane through the five almost coplanar atoms forms a dihedral angle of 89.56 (5)° with the benzene ring. The title compound, [Na(CF3O3S)(C12H24O6)], features a sodium cation that is coordinated by eight O atoms in an irregular hexagonal bipyramidal environment. Acta Cryst. E64, o1209]. There are two molecules in the asymmetric unit, which are linked by two strong intramolecular O—H ... O hydrogen bonds with graph-set motif R 2 2(8). In both (I) and (II), the conformation of the three fused rings are almost identical. The five-membered ring has an envelope conformation, the six-membered ring has a chair conformation and the seven-membered ring has a boat conformation. The most obvious differences between the two compounds is the observed disorder of the acetoxymethyl fragments in both molecules of the asymmetric unit of (I). This disorder is not observed in (II). The crystal structure and the molecular conformation is stabilized by intermolecular C—H ... O hydrogen bonds. The ability to form hydrogen bonds is different in the two compounds. The crystal studied was a non-merohedral twin, the ratio of the twin components being 0.28 (1):0.72 (1)

In the crystal, two creatinine molecules form centrosymmetric hydrogen-bonded dimers linked by pairs of N—H[cdots, three dots, centered]N hydrogen bonds. In addition, creatinine is linked to a dimethyl sulfoxide molecule by an N—H[cdots, three dots, centered]O interaction. The packing shows layers parallel to (120).
The title compound, [Li3(C4F9O)3(C3H6O)3], features an open Li/O cube with an Li ion missing at one corner. Three of the four bridging O atoms of the cube carry a fluorinated tert-butyl residue, whereas the fourth is part of an acetone molecule. The analysis of biomolecular macrocomplexes requires certain preconditions to be fulfilled. The preparation of biomolecular samples usually results in low yields. Due to this constraint of low availability any method should provide a sufficient sensitivity to cope with typical sample amounts. Biomolecules also often show a reduced stability, i.e. a propensity for fragmentation upon ionisation, which requires reasonable soft methods for the investigation. Furthermore macromolecular complexes usually are composed by means of non-covalent interactions presenting additional demands on the softness. This holds true for specific complexes like protein-ligand or DNA double strand binding. For the formation of non-covalent, specific complexes the biomolecules’ native structure and environment are a basic prerequisite and hence crucial. Therefore it is desirable during analysis to keep the biomolecules in a native environment to preserve their structure and weak interactions. One suitable method for analysing biomolecules is mass spectrometry. Mass spectrometry is capable of high throughput screening as well as determining masses with high accuracy and high sensitivity. Especially since the availability of MALDI-MS and ESI-MS mass spectrometry evolved to a versatile tool to investigate biomolecular complexes. Both, MALDI- and ESI-MS are sufficiently soft methods to observe fragile biomolecules. Yet both methods have their advantages and disadvantages. During the recent years an alternative mass spectrometric approach has been developed in our group, termed LILBID-MS (Laser Induced Liquid Bead Ionisation/Desorption). In LILBID microdroplets of aqueous solution containing buffer, salt and further additives among the analyte molecules are injected into vacuum and irradiated one-by-one by mid-IR laser pulses. The absorption of the energy by the water leads to a rapid ablation of the preformed analyte ions. LILBID is highly tolerant for the addition of salts and detergents allowing to study biomolecular complexes in a native environment. As LILBID-MS is soft enough to avoid fragmentation, specific non-covalent complexes can be analysed directly from their native environment by this method. In addition dissociation can be induced on demand by increasing the laser intensity which allows for the study of subunit compositions. A further prominent property of LILBID is the possibility to study hydrophobic membrane proteins due to the tolerated use of detergents. During the course of this work, several instrumental improvements mostly concerning ion focussing and beam steering were introduced. Together with refinements of different modes of measurement the result is a significantly improved signal-to-noise ratio as well as a further improvement in sensitivity. In addition the accessible m/z range for a given flight time has been vastly increased. The new possibilities that LILBID now offers for the study of biomolecular complexes were investigated. The ability to detect specific binding in LILBID-MS was investigated by means of nucleic acids and their interaction with proteins. It could be shown that the stability of a 16bp dsDNA corresponds to that in solution phase regarding the dependency on concentration and type of the salts used. In addition a competitive experiment with the well-known transcription factor p50 was used to demonstrate the detection of sequence-specific binding with LILBID. The improved sensitivity allowed to detect single stranded DNA at nanomolar concentrations and even the 2686bp plasmid pUC19 could be easily detected without fragmentation using a concentration of only 80nM. In case of the transcription factor p63 the mass spectrometric analysis could help to identify a new model of activation and inhibition. For the first time known quarternary structures of membrane proteins like the light-driven proton pump bacteriorhodopsin and the potassium channel KcsA could be detected with mass spectrometry. For the light-driven proton pump proteorhodopsin the type and the concentration of the used detergents significantly influenced the stability of this protein as well as the preferred quarternary structure.
In nature, society and technology many disordered systems exist, that show emergent behaviour, where the interactions of numerous microscopic agents result in macroscopic, systemic properties, that may not be present on the microscopic scale. Examples include phase transitions in magnetism and percolation, for example in porous unordered media, biological, and social systems. Also technological systems that are explicitly designed to function without central control instances, like their prime example the Internet, or virtual networks, like the World Wide Web, which is defined by the hyperlinks from one web page to another, exhibit emergent properties. The study of the common network characteristics found in previously seemingly unrelated fields of science and the urge to explain their emergence, form a scientific field in its own right, the science of complex networks. In this field, methodologies from physics, leading to simplification and generalization by abstraction, help to shift the focus from the implementation's details on the microscopic level to the macroscopic, coarse grained system level. By describing the macroscopic properties that emerge from microscopic interactions, statistical physics, in particular stochastic and computational methods, has proven to be a valuable tool in the investigation of such systems. The mathematical framework for the description of networks is graph theory, in hindsight founded by Euler in 1736 and an active area of research since then. In recent years, applied graph theory flourished through the advent of large scale data sets, made accessible by the use of computers. A paradigm for microscopic interactions among entities that locally optimize their behaviour to increase their own benefit is game theory, the mathematical framework of decision finding. With first applications in economics e.g. Neumann (1944), game theory is an approved field of mathematics. However, game theoretic behaviour is also found in natural systems, e.g. populations of the bacterium Escherichia coli, as described by Kerr (2002). In the present work, a combination of graph theory and game theory is used to model the interactions of selfish agents that form networks. Following brief introductions to graph theory and game theory, the present work approaches the interplay of local self-organizing rules with network properties and topology from three perspectives. To investigate the dynamics of topology reshaping, coupling of the so called iterated prisoners' dilemma (IPD) to the network structure is proposed and studied in Chapter 4. In dependence of a free parameter in the payoff matrix, the reorganization dynamics result in various emergent network structures. The resulting topologies exhibit an increase in performance, measured by a variance of closeness, of a factor 1.2 to 1.9, depending in the chosen free parameter. Presented in Chapter 5, the second approach puts the focus on a static network structure and studies the cooperativity of the system, measured by the fixation probability. Heterogeneous strategies to distribute incentives for cooperation among the players are proposed. These strategies allow to enhance the cooperative behaviour, while requiring fewer total investments. Putting the emphasis on communication networks in Chapters 6 and 7, the third approach investigates the use of routing metrics to increase the performance of data packet transport networks. Algorithms for the iterative determination of such metrics are demonstrated and investigated. The most successful of these algorithms, the hybrid metric, is able to increase the throughput capacity of a network by a factor of 7. During the investigation of the iterative weight assignments a simple, static weight assignment, the so called logKiKj metric, is found. In contrast to the algorithmic metrics, it results in vanishing computational costs, yet it is able to increase the performance by a factor of 5.
Die Hämophile ist eine X-chromosomal vererbbare Blutungserkrankung, die durch einen funktionellen Mangel der Gerinnungsaktoren Faktor VIII oder Faktor IX im Fall der Hämo-philie A oder Hämophilie B hervorgerufen wird. Der niedrige therapeutische Schwellenwert in der Behandlung der Hämophilie (Faktorenspiegel >1%) macht diese Krankheit zu einem idealen Zielobjekt in der Gentherapie. Eine schwerwiegende Komplikation in der Behandlung der Hämophilen besteht in der Entwicklung von neutralisierenden Antikörpern gegen das the-rapeutisch substituierte Protein. Akute Blutungen in solchen Hemmkörper-Patienten werden derzeit durch FVIII- und FIX-unabhängigen Bypass-Agenzien kontrolliert. Im Rahmen dieser Arbeit wurde nach einer erfolgten Optimierung eines nicht-viralen Ansatzes zur Transgenex-pression von FIX in der Leber der Maus dieses Gentransfersystem dazu verwendet, um eine Entwicklung von neuen, auf FIX basierenden, FVIII-Bypass-Strategien in vivo zu ermöglichen. Im ersten Teil der vorliegenden Arbeit wurden Plasmidvektoren und Vektorrückgrat-freie Minicircle-Konstrukte auf ihre Effizienz zur Expression des FIX-Transgens nach hydrodyna-misch basiertem Gentransfer in die Leber der Maus untersucht. Der Gentransfer der Mini-circle führte hierbei zu einer höheren Transgenexpression und einem geringeren Abfall der FIX-Antigen-Spiegel über die Zeit. Darüber hinaus war das FIX-Protein selbst bei den höchs-ten Expressionsleveln (700% des physiologischen Levels) völlig funktional aktiv. Der Abfall der Transgenexpression über die Zeit konnte größtenteils auf den Verlust des Vektors zurück-geführt werden. Ein signifikanter Silencing-Effekt konnte nur in den Plasmidvektor- und nicht in den Minicircle-behandelten Gruppen beobachtet werden. Diesbezüglich wurde nach 100 Tagen Persistenz der nicht-viralen Vektoren in der Mausleber eine signifikante Akkumulation an CpG-Methylierungen in regulatorischen Sequenzen des hAAT-Promoters im Plasmidvek-tor gegenüber den Minicirclen ermittelt. Außerdem konnte ebenfalls eine Demethylierung von Cytosin-Nukleotiden in bakteriellen Dcm-Sequenzmotiven über die Zeit beobachtet werden, die eine signifikant höhere Rate für das Minicircle-Konstrukt im hAAT-Promoter aufwies. Die erprobten nicht-viralen Vektoren versprechen somit eine ausreichende Effizienz, um eine Translation in den Menschen, z. B. über eine spezifische Behandlung von Lebersegmenten, möglich erscheinen zu lassen. Die Methylierungsanalyse von Promoterelementen auf Sequenzebene erbrachte zudem konkrete Ansätze zum Design von nicht-viralen Vektoren mit einer noch stabileren FIX-Expression. Im zweiten Teil dieser Arbeit wurde mit Hilfe eines Screening-Systems zur Aktivitätstestung von rekombinant exprimierten FIX-Varianten die Mutation K265T als die entscheidende Sub-stitution im 99-loop identifiziert, welche die FVIII-unabhängige Aktivität im FIX-Protein unter physiologischen Bedingungen begünstigt. Das Wildtyp- IX-Protein braucht hingegen FVIII um eine 106-fache Steigerung seiner proteolytischen Aktivität zu erreichen. Durch Kombination der Mutation K265T mit V181I und I383V konnte diese spezifische FVIII-Bypass-Aktivität in der resultierenden FIX-Variante (ITV) auf 15,6% (verglichen zu FIX in Anwesenheit von FVIII) gesteigert werden. Durch Inkubation der FIX-Varianten mit Plasma-proben aus Hemmkörper-Patienten konnte die Funktionalität der FIX-Varianten selbst in der Anwesenheit von inhibitorischen FVIII-Antikörpern demonstriert werden. Durch Untersu-chungen des Aktivierungszustands von FIX konnte zudem gezeigt werden, dass die im Rah-men dieser Arbeit generierten FIX-Varianten nicht voraktiviert sind und die messbare FVIII-Bypass-Aktivität FIX spezifisch ist. Darüber hinaus konnte hier erstmals eine hämostatische Effizienz von FIX-Varianten mit einer FVIII-unabhängigen Aktivität in einem relevanten Krankheitsmodel demonstriert werden. Diesbezüglich resultierte der nicht-virale Gentransfer der FIX-Varianten in FVIII-K.O.-Mäusen in einer dosisabhängigen Verkürzung der aPTT-basierenden Gerinnungszeit und in einer Reduktion des Blutverlustes im tail-clip-Assay in An- oder Abwesenheit von inhibitorischen FVIII-Antikörpern. Desweiteren konnte zudem mittels Vakzinierung von FIX-WT-tolerierten Mäusen mit den FIX-Varianten demonstriert werden, dass die hier eingeführten Mutationen keine hoch-immunogenen Epitope des FIX-Proteins betreffen. Schließlich konnte die Expression der FIX-Varianten in hämostatisch normalen Mäusen nicht mit der Entstehung von thromboembolischen Ereignissen selbst in einem intakten Gerinnungssystem assoziiert werden. Die hier beschriebenen FIX-Varianten könnten somit einen sicheren und effizienten Therapieansatz zur Behandlung der Hämophilie A mit inhibitorischen Antikörpern darstellen und hinsichtlich der Halbwertszeit und des möglichen throboembolischen Risikos zu einer wesentlichen Verbesserung der bereits verfügbaren Bypass-Strategien beitragen.
Selektive COX-2-Inhibitoren wie Rofecoxib und Celecoxib haben sich im klinischen Alltag bewährt, da sie im Gegensatz zu den herkömmlichen NSAIDs wie ASS oder Ibuprofen weniger gastrointestinale Komplikationen verursachen. Jedoch wurde in klinischen Studien ein erhöhtes Risiko für das Entstehen kardiovaskulärer Ereignisse nachgewiesen, was zur Stigmatisierung der selektiven COX-2 Inhibitoren und zu einer strengenIndikationstellung von Celecoxib führte. Verschiedene Studien, welche den - 765G>C SNP des PTGS2-Promotorgens untersuchten, kamen zu konträren Ergebnissen. Einerseits wurde bei Trägern des SNP eine verminderte COX-2- Proteinexpression und im Rahmen dieser Studie ein protektiver Effekt für das Entstehen kardiovaskulärer Ereignisse atherosklerotischer Genese beschrieben. Andererseits gab es Ergebnisse klinischer Studien, die bei Trägern des SNP eine erhöhte COX-2-Expression zeigten. Das Ziel der Studie war es herauszufinden, ob bei Trägern der Mutation nach COX-2-Inhibition durch Celecoxib verglichen mit Trägern der Wildtypvariante ein erhöhter, verminderter oder kein Effekt auf Prostaglandinsynthese, COX-2-Proteinexpression und COX-2-mRNA-Expression zu beobachten ist. Je 10 gesunde homozygote Träger des PTGS2 -765GG-Genotyps sowie des PTGS2 -765CC-Genotyps erhielten 200 mg Celecoxib per os, nachdem Einflüsse der COX-1 auf die Prostaglandinsynthese 24 Stunden zuvor mit der Einnahme von 500 mg ASS ausgeschlossen wurden. Blutproben wurden vor der Applikation (Referenzwert) sowie 1,3,6,9 und 24 Stunden nach der Applikation entnommen. Mittels LC-MS/MS wurden die Plasmakonzentrationen von Celecoxib sowie die ex vivo PGE2-Produktion von LPS-stimulierten Monozyten des peripheren Blutes gemessen. m-RNA Expression des COX-2-Gens wurde mit real-time quantitativer PCR gemessen. Mithilfe von Western Blot-Analysen wurde die COX-2-Expression dargestellt. Ex vivo Stimulation der Blutproben führte zu einem statistisch signifikantem Anstieg der PGE2-Produktion (P<0,001) ohne Inhibition durch Celecoxib. In Anwesenheit von Celecoxib kam es zu einer reduzierten PGE2-Produktion (von 19,3±7,2 ng/ml vor Applikation [Referenzwert] zu 7,4±4,8 ng/ml nach 1 Stunde; P<0,001), welche bis 9 Stunden nach Applikation statistisch signifikant bestehen blieb (P 0,001). Celecoxib inhibierte die PGE2-Produktion (EC 50%) bei einer Konzentration von 155,1 ng/ml bei Trägern der homozygoten Wildtypvariante sowie bei einer Konzentration von 186,6 ng/ml bei Trägern der homozygoten Allelvariante, was statistisch nicht signifikant war (P=0,36). Die Referenzwerte der PGE2-Produktion sowie die AUCs der PGE2-Konzentration in Bezug zur Zeit, ähnelten sich zwischen den Genotypen (P>0,28). LPS führte zu einer Hochregulation der COX-2 mRNAExpression (P=0,016), was jedoch unabhängig vom Genotyp war. Die COX-2-Proteinexpression zeigte bei beiden Genotypen keinen Unterschied (P=0,63). Ein Unterschied zwischen den Genotypen konnte weder auf Ebene der Prostaglandinproduktion ex vivo, noch auf Ebene der COX-2-Proteinexpression und mRNA-Expression nachgewiesen werden. Das Resultat dieser Studie steht weder zu den Ergebnissen der einen, noch zu den Ergebnissen der anderen Seite in komplettem Kontrast. Dass zur Zeit ungenügende Wissen über die Rolle der PTGS2-Mutationen ist vielmehr Ausdruck dieser konträren Studienergebnisse.
Suicide genes have been broadly used in gene therapy. They can serve as safety tools for conditional elimination of infused cells or for directed tumor therapy. To date, the Herpes simplex virus thymidine kinase/ ganciclovir (HSVtk/GCV) system is the most prominent and the most widely used suicidegene/prodrug combination. Despite its promising performance, the system displays limitations, which include relatively slow killing kinetics and toxicity of the prodrug GCV. Consequently, several groups have either developed new suicide-gene/prodrug combinations or attempted to improve the established HSVtk/GCV suicide system. The present study also aimed towards optimization of the HSVtk/GCV system. To do so, a novel, codon-optimized point mutant (A168H) of HSVtk was developed. The novel mutant was named TK.007. It was extensively tested for its efficiency in two relevant settings: (1) control of severe graft-versus-host disease (GvHD) after adoptive immunotherapy with Tlymphocytes, and (2) direct elimination of targeted tumor cells. TK.007 was compared to the broadly used wild-type, splice-corrected scHSVtk and to a codon-optimized HSVtk (coHSVtk) not bearing the above point mutation. (1) For experiments related to the adoptive immunotherapy approach, HSVtkvariants were expressed from a γ-retroviral MP71 vector as a fusion construct with the selection and marker gene tCD34. Expression levels for TK.007 in transduced lymphoid and myeloid cell lines were significantly higher at initial transduction and over a 12 week period compared to the commonly used scHSVtk and coHSVtk indicating reduced toxicity of TK.007. Killing kinetics of transduced cell lines (PM1 and K562) and primary human T cells were significantly faster for TK.007 in comparison to scHSVtk and coHSVtk in vitro. In vivo-functionality of TK.007 was assessed in an allogeneic transplantation model. T cells derived from C57BL/6J.Ly5.1 donor mice were transduced with MP71 vectors expressing scHSVtk or TK.007. Transduced cells were selected and transplanted into Balb/c Rag2-/- γ-/- immune-deficient recipient mice. Acute, severe GvHD occurred and was effectively abrogated in all mice transplanted with TK.007- transduced T cells, and in five out of six mice transplanted with scHSVtk-transduced cells. In a slightly modified quantitative allogeneic transplantation mouse model, significantly faster and more efficient in vivo killing was demonstrated for TK.007 as compared to scHSVtk, especially at low doses of GCV. (2) In order to assess TK.007 functionality in cells derived from solid tumors, HSVtk-variants were expressed from lentiviral gene ontology (LeGO) vectors in combination with an eGFP/neo-opt selection cassette. Transduced and selected tumor cell lines that derived from several tissues were eliminated at significantly lower GCV doses and to higher extents when transduced with TK.007 compared to scHSVtk. Moreover, a significantly stronger bystander effect of TK.007 was demonstrated. The superior in vitro efficiency of TK.007 was confirmed in an in vivo subcutaneous xenograft mouse model for glioblastoma in NOD/SCID mice. Mice transplanted with TK.007 transduced cells stayed tumor-free after treatment with different GCV-doses. On the contrary, mice of the scHSVtk group either demonstrated only transiently reduced tumor growth in the low-dose GCV group (10 mg/kg) compared to the control groups or suffered from relatively fast relapses after initial tumor shrinking in the standarddose (50 mg/kg) GCV group. As a result, all mice in the scHSVtk group died from vigorous tumor growth. In summary, in two different applications for suicide gene therapy the present study has demonstrated superior functional performance of the novel suicide gene TK.007 as compared to the broadly used wild-type scHSVtk. Differences became particularly pronounced at low doses of GCV. It can be concluded that the new TK.007-gene represents a promising alternative to the commonly used scHSVtk for gene therapeutic applications.
The aim of this work is to develop an effective equation of state for QCD, having the correct asymptotic degrees of freedom, to be used as input for dynamical studies of heavy ion collisions. We present an approach for modeling an EoS that respects the symmetries underlying QCD, and includes the correct asymptotic degrees of freedom, i.e. quarks and gluons at high temperature and hadrons in the low-temperature limit. We achieve this by including quarks degrees of freedom and the thermal contribution of the Polyakov loop in a hadronic chiral sigma-omega model. The hadronic part of the model is a nonlinear realization of an sigma-omega model. As the fundamental symmetries of QCD should also be present in its hadronic states such an approach is widely used to describe hadron properties below and around Tc. The quarks are introduced as thermal quasi particles, coupling to the Polyakov loop, while the dynamics of the Polyakov loop are controlled by a potential term which is fitted to reproduce pure gauge lattice data. In this model the sigma field serves a the order parameter for chiral restoration and the Polyakov loop as order parameter for deconfinement. The hadrons are suppressed at high densities by excluded volume corrections. As a next step, we introduce our new HQ model equation of state in a microscopic+macroscopic hybrid approach to heavy ion collisions. This hybrid approach is based on the Ultra-relativistic Quantum Molecular Dynamics (UrQMD) transport approach with an intermediate hydrodynamical evolution for the hot and dense stage of the collision. The present implementation allows to compare pure microscopic transport calculations with hydrodynamic calculations using exactly the same initial conditions and freeze-out procedure. The effects of the change in the underlying dynamics - ideal fluid dynamics vs. non-equilibrium transport theory - are explored. The final pion and proton multiplicities are lower in the hybrid model calculation due to the isentropic hydrodynamic expansion while the yields for strange particles are enhanced due to the local equilibrium in the hydrodynamic evolution. The elliptic and directed flow are shown to be not sensitive to changes in the EoS while the smaller mean free path in the hydrodynamic evolution reflects directly in higher flow results which are consistent with the experimental data. This finding indicates qualitatively that physical mechanisms like viscosity and other non equilibrium effects play an essentially more important role than the EoS when bulk observables like flow are investigated. In the last chapter, results for the thermal production of MEMOs in nucleus-nucleus collisions from a combined micro+macro approach are presented. Multiplicities, rapidity and transverse momentum spectra are predicted for Pb+Pb interaction at different beam energies. The presented excitation functions for various MEMO multiplicities show a clear maximum at the upper FAIR energy regime making this facility the ideal place to study the production of these exotic forms of multistrange objects.
Over the last years there has been an increasing interest in the involvement of the MVA-pathway and of members of the small GTPases, in the development and progression of AD. Earlier investigations mainly focused on the role of cholesterol in disease pathology. This research was supported by retrospective cohort studies, initially showing beneficial effects of the long-term intake of cholesterol lowering statins, on the incidence of the development of sporadic AD. However, in more recent literature increasing attention has been paid to the isoprenoids, FPP and GGPP, due to their crucial role in the post-translational modifications of members of the superfamily of small GTPases. In AD, these proteins were amongst others shown to be involved in mechanisms affecting APP processing, ROS generation and synaptic plasticity. A major factor impeding the clarification of the role of the MVA-pathway intermediates in these mechanisms was the lack of a sensitive and accurate method to determine FPP and GGPP levels in brain tissue. Hence, a state of the art HPLC-FLD method for the quantification of the isoprenoids FPP and GGPP in brain tissue was successfully developed. After the introduction of a double clean-up step from complex brain matrix samples and the synthesis of an appropriate IS (DNP), the method was fully validated according to the latest FDA guideline for bioanalytical method validation. Furthermore, this method was transferred to a faster and more sensitive, state of the art UHPLC-MS/MS application. Additionally, the method was shown to be applicable for mouse brain tissue and data was generated from an in vivo mouse simvastatin study and for different mouse models. According to the aims of the thesis, the current work describes for the first time absolute isoprenoid concentrations in human frontal cortex white and grey matter. Furthermore, this is the first report of isoprenoid levels in the frontal cortex of human AD brains. Further results were shown from mouse brains originating from different mouse models, including the Thy-1 APP mouse model mimicking AD pathology in terms of Aβ formation or C57Bl/6 mice at different ages. AD prevalence can be clearly correlated with increasing age. Therefore, three different generations of mice were investigated. The study demonstrated constant isoprenoid and cholesterol levels in the first half of their life followed by a significant increase of FPP and GGPP in the second half (between 12 and 24 month of age). Cholesterol levels were also elevated in the aged group, but again the effect was less pronounced than shown for the isoprenoids. These results lead to the tentative conclusion that cerebral isoprenoid levels are elevated during aging and that this accumulation is amplified during AD leading to accelerated neuronal dysfunction. In a different mouse study, using the C57Bl/6 mice, in vivo drug intervention with the HMG-CoA reductase inhibitor simvastatin revealed strong inhibition of the rate limiting step of the mevalonate/isoprenoid/cholesterol pathway and resulted in the first report of significantly reduced FPP and GGPP levels in brain tissue of statin treated mice. These results open for the first time the possibility to monitor drug effects on cerebral isoprenoid levels and correlate these data with a modulation of APP processing, which was shown by our group in previous studies. Interestingly, apart from the isoprenoid reduction following statin treatment the reduction of brain cholesterol was also significant but to a lesser extent. These findings support the notion that isoprenoid levels are more susceptible to statin treatment than cholesterol levels. Furthermore, this suggests a strong cellular dependence on FPP and GGPP, as the pool seems to be easily depleted, which finally could lead to cell death. The first investigations of farnesylated Ras and geranylgeranylated Rac protein levels by means of immuno-blotting, substantiated the notion of a decreased abundance of prenylated small GTPases under statin influence as a consequence of reduced isoprenoid levels. These findings demonstrate for the first time a correlation of FPP and GGPP levels with the abundance of small GTPases. These findings together with the results from the AD study prove that isoprenoid levels are not strictly subject to the same regulation as cholesterol levels. To further understand the physiological regulation in the cell, in vitro experiments with different inhibitors of the mevalonate/isoprenoid/cholesterol pathway were conducted. These results confirmed the isoprenoid and cholesterol reducing effects of statin treatment as observed in the aforementioned in vivo mouse study. Interestingly, cholesterol synthesis inhibition targeted after FPP as the branch point, led to significantly elevated FPP levels. FTase inhibition led to significantly reduced FPP levels, whereas inhibition of the GGTase I did not show a significant change of either isoprenoid levels.
One of the key functions of blood vessels is to transport nutrients and oxygen to distant tissues and organs in the body. When blood supply is insufficient, new vessels form to meet the metabolic tissue demands and to re-establish cellular homeostasis. Expansion of the vascular network through sprouting angiogenesis requires the specification of ECs into leading (sprouting) tip and following (non-sprouting) stalk cells. Attracted by guidance cues tip cells dynamically extend and retract filopodia to navigate the nascent vessel sprout, whereas trailing stalk cells proliferate to form the extending vascular tube. All of these processes are under the control of environmental signals (e.g. hypoxia, metabolism) and numerous cytokines and peptide growth factors. The Dll4/Notch pathway coordinates several critical steps of angiogenic blood vessel growth. Even subtle alterations in Notch activity can profoundly influence endothelial cell behavior and blood vessel formation, yet little is known about the intrinsic regulation and dynamics of Notch signaling in endothelial cells. In addition, it remains an open question, how different growth factor signals impinging on sprouting ECs are coordinated with local environmental cues originating from nutrient-deprived, hypoxic tissue to achieve a balanced endothelial cell response. Acetylation of lysines is a critical posttranslational modification of histones, which acts as an important regulatory mechanism to control chromatin structure and gene transcription. In addition to histones, several non-histone proteins are targeted for acetylation reversible acetylation is emerging as a fundamental regulatory mechanism to control protein function, interaction and stability. Previous studies from our group identified the NAD+-dependent deacetylase SIRT1 as a key regulator of blood vessel growth controlling endothelial angiogenic responses. These studies revealed that SIRT1 is highly expressed in the vascular endothelium during blood vessel development, where it controls the angiogenic activity of endothelial cells. Moreover, in this work SIRT1 has been shown to control the activity of key regulators of cardiovascular homeostasis such as eNOS, Foxo1 and p53. The present study describes that SIRT1 antagonizes Notch signaling by deacetylating the Notch intracellular domain (NICD). We showed that loss of SIRT1 enhances DLL4-induced endothelial Notch responses as assessed by different luciferase responsive elements as well as transcriptional analysis of Notch endogenous target genes activation. Conversely, SIRT1 gain of function by overexpression of pharmacological activation decreases induction of Notch targets in response to DLL4 stimulation. We also showed that the NICD can be directly acetylated by PC AF and p300 and that SIRT1 promotes deacetylation of NICD. We have identified 14 lysines that are targeted for acetylation and their mutation abolishes the effects of SIRT1 of Notch responses. Furthermore, over-expression or activation of SIRT1 significantly reduces the levels of NICD protein. Moreover, SIRT1-mediated NICD degradation can be reversed by blockade of the proteasome suggesting a mechanism resulting from ubiquitin-mediated proteolysis. Indeed, we have shown that SIRT1 knockdown or pharmacological inhibition decreased NICD ubiquitination. We propose a novel molecular mechanism of modulation of the amplitude and duration of Notch responses in which acetylation increases NICD stability and therefore permanence at the promoters, while SIRT1, by inducing NICD degradation through its deacetylation, shortens Notch responses. In order to evaluate the physiological relevance of our findings we used different models in which the Notch functions during blood vessel formation have been extensively characterized. First, retinal angiogenesis in mice lacking SIRT1 activity shows decreased branching and reduced endothelial proliferation, similar to what happens after Notch gain of function mutations. ECs from these mice exhibit increased expression of Notch target genes. Second, these results were reproducible during intersomitic vessel growth in sirt1-deficient zebrafish. In both models, the defects could be partially rescued by inhibition of Notch activation. Third, we used an in vitro model of vessel sprouting from differentiating embryonic bodies in response to VEGF in a collagen matrix. Our results showed that Sirt1-deficient cells shows impaired sprouting which correlated with increased NICD levels. In addition, when in competition with wild-type cells in this assay, Sirt1-deficient cells are more prone to occupy the stalk cell position. Taken together, our study identifies reversible acetylation of NICD as a novel molecular mechanism to adapt the dynamics of Notch signaling and suggest that SIRT1 acts as a rheostat to fine-tune endothelial Notch responses. The NAD+-dependent feature of SIRT1 activity possibly links endothelial Notch responses to environmental cues and metabolic changes during nutrient deprivation in ischemic environments or upon other cellular stresses.
Die Bedeutung verschiedener CRASP-Proteine für die Komplementresistenz von Borrelia burgdorferi s.s.

Die vorliegende Arbeit liefert einen wichtigen Beitrag zum Verständnis des molekularen Mechanismus der Immunevasion von B. burgdorferi s.s., insbesondere der Bedeutung einzelner CRASP-Proteine für die Komplementresistenz. Sie trägt dazu bei, die Relevanz dieser Proteine für die Pathogenese dieses Erregers zu untermauern. Im Rahmen dieser Arbeit gelang es, verschiedene Vektoren mit den ursprünglichen oder mutierten CRASP-kodierenden Genen cspA, cspZ, erpP und erpA aus B. burgdorferi s.s. zu generieren und diese in das CRASP-negative Isolat B. garinii G1 zu transformieren. Die Expression der speziesfremden Gene als auch der Transport der CRASP-Moleküle auf die Zelloberfläche von B. garinii G1 konnten nachgewiesen werden. Für die konstitutiv CRASP-1- oder CRASP-2-produzierenden Borrelienzellen konnte gezeigt werden, dass diese, auf der Zelloberfläche lokalisierten CRASP-Moleküle mit Faktor H und FHL-1 interagieren, die gebundenen Komplementregulatoren ihre funktionelle Aktivität zur C3b-Inaktivierung aufrechterhalten und die Zellen in Gegenwart von Komplement überleben. Damit wurde erstmals der Nachweis erbracht, dass beide CRASP-Moleküle unabhängig voneinander Schutz vor komplementvermittelter Lyse verleihen. Untersuchungen mit den veränderten CRASP-1-Molekülen ergaben, dass die Transformante G1/pCRASP-1 E147K eine verringerte Bindung von Faktor H und FHL-1 aufwies, welche sich jedoch nicht auf die Komplementresistenz der Zellen auswirkte. Im Gegensatz dazu führte eine Aminosäuresubstitution im C-Terminus des CRASP-1-Moleküls an Position 240 zum Verlust der Bindung von Faktor H und einer stark verminderten Bindung von FHL-1, so dass auch keine Kofaktoraktivität nachgewiesen werden konnte. Trotz des Bindungsverlustes beider Komplementregulatoren zeigte die Transformante G1/pCRASP-1 Y240A nur geringe Ablagerungen des lytischen, terminalen Komplementkomplexes (TCC) auf der Zelloberfläche und Wachstum in Gegenwart von aktiven Komplement. Mittels eines Hämolyse-Assays wurde schließlich festgestellt, dass CRASP-1 direkt mit Komponenten des Komplementsystems interagiert und dadurch die Assemblierung des TCC verhindert. Die Bedeutung der Aminosäuren an den Positionen 81, 139, 207 und 211 im CRASP-2-Molekül für die Faktor H / FHL-1-Bindung und die daraus resultierenden Auswirkungen auf die Komplementresistenz der Borrelien wurde gleichfalls nachgewiesen. Dabei wies insbesondere die Transformante G1/pCRASP-2 Y211A ein inhibiertes Wachstum in Humanserum und verstärkt Komplementablagerungen auf der Zelloberfläche auf, was auf den Verlust bzw. der sehr schwachen Bindung von FHL-1 und Faktor H zurückzuführen ist. Im Gegensatz zu den Transformanten, welche ein CRASP-2-Molekül mit nur einem Aminosäureaustausch produzierten, zeigten die Transformanten, deren CRASP-2-Molekül zwei Aminosäuresubstitutionen aufwies (G1/pCRASP-2 R139A-Y207A, G1/pCRASP-2 R139A-Y211A, G1/pCRASP-2 Y207A-Y211A) keine Bindung der beiden Regulatorproteine und keinen Schutz der Zellen vor der lytischen Wirkung von Komplement. Neue, unerwartete Erkenntnisse ergaben sich aus den Untersuchungen mit Borrelienzellen, welche das CRASP-3- oder CRASP-5-kodierende erpP- bzw. erpA-Gen enthielten. Obwohl gereinigtes als auch denaturiertes CRASP-3 und RASP-5 in der Lage war, Faktor H zu binden, wiesen die vitalen Zellen der Transformanten G1/pCRASP-3 und G1/pCRASP-5 keine Bindung von Faktor H und keinen Schutz der Zellen vor komplementvermittelter Lyse auf. Aus den durchgeführten Untersuchungen konnten für gereinigtes CRASP-3 und CRASP-5 als auch für die CRASP-3- und CRASP-5-produzierenden Transformanten neue Liganden, nämlich CFHR-2 und CFHR-5, aus Humanserum identifiziert werden. Zusammenfassend lassen sich folgende Aussagen hinsichtlich des molekularen Mechanismus der Komplementresistenz bei B. burgdorferi s.s. aus den erhobenen Daten dieser Arbeit mit transformierten Borrelienzellen formulieren: *Die Komplementresistenz der Borrelien wird durch die Faktor H- und FHL-1-bindenden Proteine CRASP-1 und CRASP-2, jedoch nicht durch CRASP-3 und CRASP-5 determiniert, *CRASP-1 als multifunktionelles Protein ist zusätzlich in der Lage, direkt mit Komplement zu interagieren, *Die C-terminalen Domänen von CRASP-1 und CRASP-2 sind für die Bindung der beiden Komplementregulatoren Faktor H und FHL-1 relevant, *CRASP-3 und CRASP-5 auf der Borrelienoberfläche lokalisiert, interagieren mit CFHR-1, CFHR-2 und CFHR-5, aber nicht mit Faktor H.
Das vorgestellte Projekt verfolgt das Ziel, die in den Bibliotheken und Archiven Rumäniens befindlichen mittelalterlichen deutschen Handschriften zu erschließen, um auf dieser Basis und in einer regional vornehmlich auf Siebenbürgen bezogenen Perspektive die Geschichte der deutschen Literatur des Mittelalters auf dem Gebiet des heutigen Rumänien in Form von Fallstudien zu beschreiben. Methodisch gilt es dabei […] einem ausschließlich dem Autor und Werk verpflichteten Konzept von Literaturgeschichtsschreibung eine stärker überlieferungsorientierte und regional ausgerichtete Literaturhistoriographie entgegen[zu]setzen, um auf dieser Weise den Literaturbetrieb einer bestimmten Region zu erfassen. Eine regional vornehmlich auf Siebenbürgen ausgerichtete Literaturgeschichte wird sich indes nicht nur auf die deutschsprachige Literatur beschränken, sondern sich für einen interdisziplinären Zugang öffnen und auch die ungarische und vor allem die lateinische Literaturproduktion und -rezeption im untersuchten Gebiet in ihre Überlegungen mit einbeziehen, um möglichen literarischen Interferenzen Rechnung zu tragen. Der Vorteil einer solchen, nicht einer bestimmten ‚Nationalliteratur’ verpflichteten Sicht auf die Literaturgeschichte ist, dass sie ermöglicht, Aussagen über den Literaturbetrieb einer Region (hier vor allem Siebenbürgen) als Ganzes zu treffen.
With the help of miniaturized GPS recorders I recorded 167 tracks of 48 individual pigeons during their flight from 6 different sites around Frankfurt. The experiments consisted of two main series of repeated releases from two sites 30 km north and south from the pigeons' home loft. From the site in the south the pigeons homed 12 times and from the site in the north 16 times. After the final release from these sites, the pigeons were released at 60 km distance from home. These additional sites were selected so that the pigeons would presumably fly over the previous release site with which they were highly familiar. After conclusion of the main series two additional releases were performed, one within the magnetic anomaly of the Vogelsberg and one in a magnetically quiet region. To make these releases comparable, both release sites were selected so that the distance from the home loft was 40 km. All data obtained during these experiments were subjected to a threefold analysis, mostly based on methods that I had developed by myself or adapted for this specific study. In the first step, data were analyzed traditionally, evaluating variables similar to those that can be found in current literature. I therefore calculated values that correspond to those obtained by visual observation, like virtual vanishing bearings and intervals after one minute and after 2.5 km. Additionally I calculated the efficiency of the flights and efficiencies for specific portions of each flight, to derive variables that describe the behavior after vanishing. In the second step, which served also as a preparation for the mathematical analysis, the flight of the pigeons was separated into distinctive phases of the flight by the so-called points of decision. The flight of the pigeon can usually be separated into an initial phase of flying about, a departure and/or final homing phase. In more complex cases, however, several points of decision and a multitude of intermediary phases can be defined. Yet, the initial phase, the departure phase and the final homing phase can be defined for all tracks and therefore have been selected as appropriate candidates for a thorough analysis. In the last step I employed the so-called method of time lag embedding to reconstruct the underlying navigational process of the pigeons' homing flight. This method is based on the principles of chaos theory and is regularly employed for the analysis of dynamic systems. Its application allows the reconstruction of the underlying processes from experimentally recorded data without any a priori knowledge of the underlying system itself. For these reconstructed systems I calculated characteristic properties which are unique for each system. These are the so-called correlation dimension, describing the complexity of the system, and the so-called largest Lyapunov exponent, describing its predictability. Based on the knowledge gathered from these reconstructions, I used a variation of the previous methods to identify navigational phases, by calculating the correlation dimension as a sliding mean over the complete track. From these data I then derived further characteristics of the underlying process, such as its precision and differences in complexity depending on the pigeon's current position. ...
This dissertation connects two independent fields of theoretical neuroscience: on the one hand, the self-organization of topographic connectivity patterns, and on the other hand, invariant object recognition, that is the recognition of objects independently of their various possible retinal representations (for example due to translations or scalings). The topographic representation is used in the presented approach, as a coordinate system, which then allows for the implementation of invariance transformations. Hence this study shows, that it is possible that the brain self-organizes before birth, so that it is able to invariantly recognize objects immediately after birth. Besides the core hypothesis that links prenatal work with object recognition, advancements in both fields themselves are also presented. In the beginning of the thesis, a novel analytically solvable probabilistic generative model for topographic maps is introduced. And at the end of the thesis, a model that integrates classical feature-based ideas with the normalization-based approach is presented. This bilinear model makes use of sparseness as well as slowness to implement "optimal" topographic representations. It is therefore a good candidate for hierarchical processing in the brain and for future research.
