Background The role of the Fcgamma receptor IIa (FcgammaRIIa), a receptor for C-reactive protein (CRP), the classical acute phase protein, in atherosclerosis is not yet clear. We sought to investigate the association of FcgammaRIIa genotype with risk of coronary heart disease (CHD) in two large population-based samples. Methods FcgammaRIIa-R/H131 polymorphisms were determined in a population of 527 patients with a history of myocardial infarction and 527 age and gender matched controls drawn from a population-based MONICA- Augsburg survey. In the LURIC population, 2227 patients with angiographically proven CHD, defined as having at least one stenosis [greater than or equal to]50%, were compared with 1032 individuals with stenosis <50%. Results In both populations genotype frequencies of the FcgammaRIIa gene did not show a significant departure from the Hardy-Weinberg equilibrium. FcgammaRIIa R(-131)->H genotype was not independently associated with lower risk of CHD after multivariable adjustments, neither in the MONICA population (odds ratio (OR) 1.08; 95% confidence interval (CI) 0.81 to 1.44), nor in LURIC (OR 0.96; 95% CI 0.81 to 1.14). Conclusion Our results do not confirm an independent relationship between FcgammaRIIa genotypes and risk of CHD in these populations.
In this thesis, we studied the single impurity Anderson model and developed a new and fast impurity solver for the dynamical mean field theory (DMFT). Using this new impurity solver, we studied the Hubbard model and periodic Anderson model for various parameters. This work is motivated by the fact that the dynamical mean field theory is widely used for the studies of strongly correlated systems, and the most frequently used methods, e.g. the quantum Monte-Carlo method (QMC), and the exact digonalization method are much CPU time consuming and usually limited by the available computers. Therefore, a fast and reliable impurity solver is needed. This new impurity solver was explored based on the equation-of-motion method (also called Green's function and decoupling method in some literature). Using the retarded Green's function, we first derived the equations of motion of Green's functions. Then, we employed a decoupling scheme to close the equations. By solving self-consistently the obtained closed set of integral equations, we obtained the single particle Green's function for the single impurity Anderson model. After that, the single impurity Anderson model was solved along with self-consistency conditions within the framework of DMFT. In this work, we studied and compared two decoupling schemes. Moreover, we also derived possible higher order approximations which will be tested in future work. Besides the theoretical work, we tested the method in numerical calculations. The integral equations are first solved by iterative methods with linear mixing and Broyden mixing, respectively. However, these two methods are not sufficient for finding the self-consistent solutions of the DMFT equations because converged results are difficult to obtain. Moreover, the computing speed of the two methods is also not satisfactory. Especially the iterative method with linear mixing costs always a lot of CPU time due to the required small mixing. Hence, we developed a new method, which is a combination of genetic algorithm and iterative method. This new method converges very fast and removes artifacts appearing in the results from the iterative method with linear and Broyden mixing. It can directly operate on the real axis, where no numerical error from the high frequency tail corrections and the analytical continuation is introduced. In addition, our new technique strongly improves the precision of the numerical results by removing the broadening. With this newly developed impurity solver and numerical technique, we studied the single impurity Anderson model, the single band Hubbard model and the periodic Anderson model with arbitrary spin and orbital degeneracy N on the real axis. For the single impurity Anderson model, the spectral functions are calculated for the infinite and finite Coulomb interaction strength. We also studied the spectral functions in dependence of the parameters of impurity position and hybridization. For the Hubbard model, we studied the bandwidth control and filling control Mott metal-insulator transition for spin and orbital degeneracy N = 2. It gives qualitatively the critical value of Coulomb interaction strength for the Mott metal-insulator transition, and the spectral functions which are comparable to those obtained in QMC and numerical renormalization group methods. We also studied the quasiparticle weight and the self-energy in metallic states. The latter shows almost Fermi liquid behavior. At last we calculated the densities of states for the Hubbard model with arbitrary spin and orbital degeneracy N. The periodic Anderson model (PAM) is also studied as another important lattice model. It was solved for various combinations of parameters: the Coulomb interaction strength, the impurity position, the center position of the conduction band, the hybridization, the spin and orbital degeneracy. The PAM results represents the physics of impurities in a metal. In short, our method works for the Hubbard model and the periodic Anderson model in a large range of parameters, and gives good results. Therefore, our impurity solver could be very useful in calculations within LDA+DMFT. Finally, we also made a preliminary investigation of the multi-band system based on the success in single band case. We first studied the two-band system in a simplified treatment by neglecting the interaction between the two bands through the bath. This has given promising numerical results for the two-band Hubbard model. Moreover, we have studied theoretically the two-band system with mean field approximation and Hubbard-I approximation in dealing with the higher order cross Green's functions which are related to both the two bands. In the mean field approximation, we even generalized the two-band system to arbitrary M=N/2 band system. Potential improvement can be carried out on the basis of this work.
Negation und Modalität

Der vorliegende Artikel behandelt die Problematik der Negation im Rahmen der Modalität. Die Aufmerksamkeit richtet sich auf formale und inhaltliche Aspekte der Negation auf dem modalen Feld. Nach dem Vergleich der Ausdrucksmöglichkeiten der Negation im Deutschen und Slowakischen werden die Auswirkungen der Negation auf die semantische Ebene der Modalverben (weiter nur MV) in beiden Sprachen konkret behandelt. Es gibt mehrere Verfahren zur Skopusfestlegung des Negators. Im Artikel wird ein Modell präsentiert, das die Diskrepanz zwischen der oberflächlichen und tatsächlichen Negation in beiden Sprachen zeigt.
Die Erkenntnis durch Illusion muss bei Harsdörffer, so könnte man probeweise formulieren, anders als bei Descartes, nicht überwunden werden. Vielmehr leistet sie durch den visuellen Effekt der Ungewissheit zugleich die [...] augenscheinliche Gewissheit, zumindest wenn man sich die Herstellbarkeit solcher Effekte bewusst macht.
Gegenstand dieses Aufsatzes sind die drei aufeinander aufbauenden Gedankenfiguren, die Descartes in den Meditationes (und teilweise in der Recherche de la vérité) einführt, um seine methodisch zu verstehende Theorie eines globalen Außenweltskeptizismus zu formulieren: Wahnsinn, Traum und Genius malignus. Ich werde argumentieren, dass einige der in der Forschung hervorgehobenen Bezüge, insbesondere die zur schönen Literatur und zur antiken Skepsis, diese Gedankenfiguren und ihre Verbindung untereinander nicht hinreichend historisch kontextualisieren. Vielversprechender scheint mir ein, ebenfalls in der Forschung vertretener, Erklärungsansatz zu sein, der auf die (spät-)mittelalterliche Debatte über die Potentia absoluta Gottes zurückgreift. Mit Bezug auf diese Traditionslinie lässt sich konstatieren, dass Descartes einen Sprung vom allmächtigen (und daher auch grundsätzlich der Täuschung fähigen) Gott des Mittelalters zum bösen Dämon vollzogen hat. In meinem Beitrag sollen nun dieser Sprung und vor allem der böse Dämon selbst ins Zentrum der Betrachtung gestellt werden. Dafür gilt es, auf die in der Frühen Neuzeit im Zusammenhang der Hexenverfolgung (bei Gegnern wie Verteidigern) diskutierte Dämonologie zurückzugreifen, genauer gesagt: auf einen universalen Topos über die Fähigkeit von Teufeln und Dämonen, mittels Eingriff in Fantasie und Sinne Sachverhalte vorzutäuschen. Berücksichtigt man, dass von diesen dämonischen Betrügereien gesagt wird, sie ähnelten einem Traum und funktionierten bei Schwachsinnigen am besten, so zeigt sich eine bemerkenswerte Parallele zu Descartes' Argumentation.
The YidC/Oxa1/Alb3 family of membrane proteins controls the insertion and assembly of membrane proteins in bacteria, mitochondria, and chloroplasts. Here we describe the molecular mechanisms underlying the interaction of Alb3 with the chloroplast signal recognition particle (cpSRP). The Alb3 C-terminal domain (A3CT) is intrinsically disordered and recruits cpSRP to the thylakoid membrane by a coupled binding and folding mechanism. Two conserved, positively charged motifs reminiscent of chromodomain interaction motifs in histone tails are identified in A3CT that are essential for the Alb3-cpSRP43 interaction. They are absent in the C-terminal domain of Alb4, which therefore does not interact with cpSRP43. Chromodomain 2 in cpSRP43 appears as a central binding platform that can interact simultaneously with A3CT and cpSRP54. The observed negative cooperativity of the two binding events provides the first insights into cargo release at the thylakoid membrane. Taken together, our data show how Alb3 participates in cpSRP-dependent membrane targeting, and our data provide a molecular explanation why Alb4 cannot compensate for the loss of Alb3. Oxa1 and YidC utilize their positively charged, C-terminal domains for ribosome interaction in co-translational targeting. Alb3 is adapted for the chloroplast-specific Alb3-cpSRP43 interaction in post-translational targeting by extending the spectrum of chromodomain interactions.
Flavins are employed to transform physical input into biological output signals. In this function, flavins catalyze a variety of light-induced reactions and redox processes. However, nature also provides flavoproteins with the ability to uncouple the mediation of signals. Such proteins are the riboflavin-binding proteins (RfBPs) with their function to store riboflavin for fast delivery of FMN and FAD. Here we present in vitro and in vivo data showing that the recently discovered archaeal dodecin is an RfBP, and we reveal that riboflavin storage is not restricted to eukaryotes. However, the function of the prokaryotic RfBP dodecin seems to be adapted to the requirement of a monocellular organism. While in eukaryotes RfBPs are involved in trafficking riboflavin, and dodecin is responsible for the flavin homeostasis of the cell. Although only 68 amino acids in length, dodecin is of high functional versatility in neutralizing riboflavin to protect the cellular environment from uncontrolled flavin reactivity. Besides the predominant ultrafast quenching of excited states, dodecin prevents light-induced riboflavin reactivity by the selective degradation of riboflavin to lumichrome. Coordinated with the high affinity for lumichrome, the directed degradation reaction is neutral to the cellular environment and provides an alternative pathway for suppressing uncontrolled riboflavin reactivity. Intriguingly, the different structural and functional properties of a homologous bacterial dodecin suggest that dodecin has different roles in different kingdoms of life.

Sprachtechnologie für übersetzungsgerechtes Schreiben am Beispiel Deutsch, Englisch, Japanisch

Wir [...] haben uns zur Aufgabe gesetzt, Wege zu finden, wie linguistisch basierte Software den Prozess des Schreibens technischer Dokumentation unterstützen kann. Dabei haben wir einerseits die Schwierigkeiten im Blick, die japanische und deutsche Autoren (und andere Nicht-Muttersprachler des Englischen) beim Schreiben englischer Texte haben. Besonders japanische Autoren haben mit Schwierigkeiten zu kämpfen, weil sie hochkomplexe Ideen in einer Sprache ausdrücken müssen, die von Informationsstandpunkt her sehr unterschiedlich zu ihrer Muttersprache ist. Andererseits untersuchen wir technische Dokumentation, die von Autoren in ihrer Muttersprache geschrieben wird. Obwohl hier die fremdsprachliche Komponente entfällt, ist doch auch erhebliches Verbesserungspotential vorhanden. Das Ziel ist hier, Dokumente verständlich, konsistent und übersetzungsgerecht zu schreiben. Der fundamentale Ansatz in der Entwicklung linguistisch-basierter Software ist, dass gute linguistische Software auf Datenmaterial basiert und sich an den konkreten Zielen der besseren Dokumentation orientiert.
Möglichkeiten einer wohnortnahen, gesundheitsbezogenen Bewegungsberatung für Senioren ab 65 Jahren

Körperliche Aktivität im Alter beugt gesundheitlichen Beschwerden physischer und psychischer Art vor (vgl. u. a. Martel et al., 1999; Puggaard et al., 2000; Stathi et al., 2002; Pedersen & Saltin, 2006). Mit Blick auf die demografische Situation Deutschlands gilt es, mittels gezielter Programme möglichst viele inaktive Senioren anzusprechen, um deren Bewegungsaktivitäten auf ein empfohlenes Minimum von 30 Minuten moderater aerober körperlicher Aktivität an fünf Tagen der Woche zu steigern und somit Gesundheitsressourcen zu erhalten (vgl. Nelson et al., 2007). In den letzten Jahren wurde vor allem im englischsprachigen Raum eine Reihe von Maßnahmen zur Bewegungsförderung für Erwachsene eingeführt und ausgewertet. Barrieren für die Aufnahme von Bewegung, wie z. B. infrastrukturelle oder gesundheitliche Hindernisse, sollten abgebaut werden, die Niedrigschwelligkeit von Bewegungsangeboten spielt dabei eine bedeutende Rolle. In Kapitel 2 dieser Arbeit wurde der Forschungsstand zu theoriegeleiteten und alltagsbezogenen Beratungsinterventionen vorgestellt sowie zu Programmen, in deren Rahmen Kooperationen mit Arztpraxen entstanden sind (vgl. Jakicic et al., 1999; Marshall & Biddle, 2001; Dapp et al., 2007). Forschungslücken im Hinblick auf die Übertragbarkeit vorhandener Modelle auf Senioren in deutschen Großstädten bildeten den Ausgangspunkt, um die vorgestellten Ansätze zu verknüpfen und ein Modell der individuellen theoriegeleiteten Bewegungsberatung mit maßgeschneiderten, wohnortnahen Aktivitätsangeboten in einem interdisziplinären Team umzusetzen und zu untersuchen. Gegenstand der Studie war die Evaluation der Angebotsnutzung, der Bereitschaft zur Verhaltensänderung und in diesem Zusammenhang der Steigerung körperlicher Aktivität ebenso wie die Überprüfung möglicher Indikatoren für eine erfolgreiche Teilnahme an der Beratung. Teilnehmer der neunwöchigen Studie waren insgesamt 181 Personen über 65 Jahre aus Frankfurt am Main und Umgebung. Die Rekrutierung erfolgte über Zeitungsannoncen und Ansprache von Seniorengruppenleitern. In Prätest und Posttest wurden allgemeine Anamnesedaten, die Bereitschaft zur Verhaltensänderung (Fragebogen zum Transtheoretischen Modell (TTM); Keller, 1999), das Aktivitätsniveau (International Physical Activity Questionnaire (IPAQ); Booth, 2000) sowie das subjektive Gesundheitsempfinden (SF-12; Ware et al., 1996) und in der Interventionsgruppe auch das Interesse an einer Beratung (Eintrag in Liste) erfasst. Den Teilnehmern der Interventionsgruppe (n = 84) wurde eine Bewegungsberatung angeboten. An individuellen Gesprächen mit der Bewegungsberaterin waren 80 % der Angesprochenen interessiert, eine Übungsstunde (Schnupperstunde) besuchten 40 %. Nach Ende der Beratung wollten 23 % eine neue Aktivität im Alltag einführen. In der Interventionsgruppe ist die Bereitschaft zur Verhaltensänderung in Bezug auf körperliche Aktivität, gemessen am Aufstieg in den Stadien des TTM, stärker gestiegen als in der Kontrollgruppe (p = 0,048). Zudem steigerte die Interventionsgruppe ihren mittleren Wochenumfang moderater bis intensiver körperlicher Aktivität mit p = 0,039 stärker als die Kontrollgruppe. Während die Aktivitäten der Interventionsgruppe von 336 ± 265 Min. / Woche (Median: 268 Min. / Woche) im Prätest auf 410 ± 278 Min. / Woche (Median: 433 Min. / Woche) im Posttest zunahmen, stiegen die Werte der Kontrollgruppe von Prätest (290 ± 224 Min. / Woche, Median: 228 Min. / Woche) zu Posttest (295 ± 201 Min. / Woche, Median: 245 Min. / Woche) kaum an. Des Weiteren war die Tendenz zu erkennen, dass ein schlechterer Gesundheitszustand sowie ein hohes Interesse zu Beginn der Beratung in Verbindung mit vermehrter Aktivität zu Beratungsende stehen. Diese Trends erreichten jedoch nicht das Signifikanzniveau, sodass für Bewegungsberater und Institutionen im Gesundheitswesen weitere Untersuchungen zu diesen potenziellen Indikatoren einer erfolgreichen Beratung wünschenswert sind. Die Ergebnisse der Studie sind vergleichbar mit den Resultaten internationaler Studien, deren Follow-up-Zeiten allerdings länger waren (¯ 6 Monate). Im Zuge der hier beschriebenen Maßnahme steigerten 47 % der 84 Personen aus der Interventionsgruppe ihre körperliche Aktivität. Bei Harland et al. (1999) und Märki et al. (2006) fanden sich Angaben von 38 bis 57 %. In Kapitel 6 Diskussion wurden neben den Untersuchungsergebnissen auch organisa- torische Aspekte der Beratungsintervention diskutiert. Die Problematik der präzisen Zielgruppenbestimmung und zweckmäßigen Rekrutierung kam zum Anklang, da viele Studienteilnehmer (48 %) bereits vor Beginn der Studie an mindestens drei Tagen der Woche 30 Minuten lang körperlich aktiv gewesen waren. Langzeitstudien sind nötig, um die Nachhaltigkeit von Beratungsprogrammen zu evaluieren, da die Veränderungs- und Aktivitätsbereitschaft im Laufe längerer Interventionen nachzulassen scheint (vgl. van der Bij et al., 2002 und Hillsdon et al., 2005). Außerdem sollten physische Kenngrößen objektiv erfasst werden, um gesundheitsfördernde Effekte einer Bewegungsberatung zu ermitteln (vgl. Ashworth et al., 2005). Durch das Zusammenwirken unterschiedlicher Fachrichtungen und Institutionen könnten in Zukunft ansprechende Beratungsangebote zur Vermittlung wohnortnaher Lebensstil-Aktivitäten für inaktive Ältere entstehen. Während ärztliche Empfehlungen oftabstrakt bleiben, kann ein Berater mit Hilfe des hier beschriebenen Modells konkrete Verbesserungsvorschläge für eine Steigerung körperlicher Aktivität im Alltag von Senioren unterbreiten.
Background: Nitric oxide (NO) is an essential vasodilator. In vascular diseases, oxidative stress attenuates NO signaling by both chemical scavenging of free NO and oxidation and down-regulation of its major intracellular receptor, the alpha/beta heterodimeric heme-containing soluble guanylate cyclase (sGC). Oxidation can also induce loss of sGC's heme and responsiveness to NO.
Results: sGC activators such as BAY 58-2667 bind to oxidized/heme-free sGC and reactivate the enzyme to exert disease-specific vasodilation. Here we show that oxidation-induced down-regulation of sGC protein extends to isolated blood vessels. Mechanistically, degradation was triggered through sGC ubiquitination and proteasomal degradation. The heme-binding site ligand, BAY 58-2667, prevented sGC ubiquitination and stabilized both alpha and beta subunits.
Conclusion: Collectively, our data establish oxidation-ubiquitination of sGC as a modulator of NO/cGMP signaling and point to a new mechanism of action for sGC activating vasodilators by stabilizing their receptor, oxidized/heme-free sGC.
Am 08.11.2009 beging Herr Klaus-Jürgen Seelig seinen 65. Geburtstag. Zu diesem Anlass gratulieren wir herzlich, erinnern uns an seinen Weg zur Ornithologie und seine Verdienste im ehrenamtlichen und beruflichen Naturschutz. In Magdeburg geboren, kam er schon in den Jugendjahren durch den Vater mit der Taubenzucht und der Singvogelhaltung in Kontakt. Von 1962 bis 1964 erlernte er den Beruf eines Forstfacharbeiters. Es entstanden erste Kontakte zu naturkundlichen Betätigungen, die sein Interesse an der Ornithologie weckten. 1967 schloss er die Fachschule für Pflanzenschutz in Halle als Staatlich geprüfter Pflanzenschutzagronom ab. Von 1970 bis 1975 absolvierte er ein Fernstudium an der Martin-Luther-Universität Halle-Wittenberg und schloss als Dipl.-Agraringenieur ab. Auch dieses Studium führte ihn an die Naturwissenschaften heran. Seit 1968 arbeitete Klaus-Jürgen Seelig in der Fachgruppe Ornithologie Magdeburg, im Kulturbund mit und wurde Mitglied des Ornithologischen Arbeitskreises Mittelelbe- Börde. 1978 legte er die Prüfung als Vogelberinger ab und wurde in die Bezirksarbeitsgruppe Artenschutz Magdeburg berufen, in der er die Bearbeitung der Vögel übernahm. Klaus-Jürgen Seelig leistete einen großen Beitrag für den Naturschutz, besonders in der heimischen Vogelwelt.
The mode of the antitumoral activity of multimutated oncolytic herpes simplex virus type 1 G207 has not been fully elucidated yet. Because the antitumoral activity of many drugs involves the inhibition of tumor blood vessel formation, we determined if G207 had an influence on angiogenesis. Monolayers of human umbilical vein endothelial cells and human dermal microvascular endothelial cells, but not human dermal fibroblasts, bronchial epithelial cells, and retinal glial cells, were highly sensitive to the replicative and cytotoxic effects of G207. Moreover, G207 infection caused the destruction of endothelial cell tubes in vitro. In the in vivo Matrigel plug assay in mice, G207 suppressed the formation of perfused vessels. Intratumoral treatment of established human rhabdomyosarcoma xenografts with G207 led to the destruction of tumor vessels and tumor regression. Ultrastructural investigations revealed the presence of viral particles in both tumor and endothelial cells of G207-treated xenografts, but not in adjacent normal tissues. These findings show that G207 may suppress tumor growth, in part, due to inhibition of angiogenesis.

Introduction Complex psychopathological and behavioral symptoms, such as delusions and aggression against care providers, are often the primary cause of acute hospital admissions of elderly patients to emergency units and psychiatric departments. This issue resembles an interdisciplinary clinically highly relevant diagnostic and therapeutic challenge across many medical subjects and general practice. At least 50% of the dramatically growing number of patients with dementia exerts aggressive and agitated symptoms during the course of clinical progression, particularly at moderate clinical severity. Methods Commonly used rating scales for agitation and aggression are reviewed and discussed. Furthermore, we focus in this article on benefits and limitations of all available data of anticonvulsants published in this specific indication, such as valproate, carbamazepine, oxcarbazepine, lamotrigine, gabapentin and topiramate. Results To date, most positive and robust data are available for carbamazepine, however, pharmacokinetic interactions with secondary enzyme induction limit its use. Controlled data of valproate do not seem to support the use in this population. For oxcarbazepine only one controlled but negative trial is available. Positive small series and case reports have been reported for lamotrigine, gabapentin and topiramate. Conclusions So far, data of anticonvulsants in demented patients with behavioral disturbances are not convincing. Controlled clinical trials using specific, valid and psychometrically sound instruments of newer anticonvulsants with a better tolerability profile are mandatory to verify whether they can contribute as treatment option in this indication.
In this thesis we report on the high pressure synthesis, crystal growth, structural characterisation and magnetic properties of the cubic vanadate pyrochlores A2V2O7 (with A = Y, Er and Dy). We have found that high pressure is requisite for the stabilization of the selected compounds. For this purpose, a multianvil high pressure apparatus was built in our laboratory and a new multianvil inset (i.e., a ceramic pressure medium and the interior parts) was developed. The multianvil press is based on a hydraulic press with a maximum force of 7.73 MN (corresponds to 788 tons), a Walker type module and a specially designed hydraulic and electric control. Pressure calibration of the multianvil setup was performed by high pressure fixed points (i.e. solid-solid transformation of Bi I-II (2.55 GPa) and Bi II-III (3.15 GPa)). A maximum pressure of 6 GPa was attained using hardened metal anvils (tungsten carbide) with truncation edge length (TEL) of 14 mm and a sample volume of ~ 70 mm3. Heating of the sample in our current multianvil setup (TEL = 14 mm) was achieved by resistive heating of a graphite furnace. Temperatures up to 1500 °C could be obtained at pressures up to 6 GPa. By systematic variation of the synthesis conditions (for instance the operation temperature or the choice of the crucible material) under high pressure and taking into account the well known ternary compounds, when accessing the phase diagram, the cubic vanadate pyrochlores A2V2O7 (with A = Y, Er and Dy) were synthesized successfully. It was found that the oxygen partial pressure is crucial for the formation of the desired pyrochlore phase. Gas-tight platinum crucibles were used as container material for the synthesis of the vanadate pyrochlores. We have investigated, that pressures of the order of 5.0 GPa and temperatures of approximately 1200 °C are necessary for the stabilization of the monophasic samples of the vanadate pyrochlores. Lu2V2O7 could be synthesized under ambient pressure conditions and is used in our studies for comparison purposes. A special graphite furnace was developed for the high pressure crystal growth of the vanadate pyrochlores. For the first time, A2V2O7 (with A = Y, Er and Dy) single crystals with a maximum size of 0.4 mm were grown by using the grain growth method at high pressure and high temperature conditions. The samples (i.e., powders and single crystals) were characterised by single crystal Xray diffraction, X-ray powder diffraction method, Laue method and scanning electron microscopy (SEM). Complementary to the X-ray diffraction methods, infrared absorsoption spectroscopy was used to distinguish between the fluorite and pyrochlore structure. It has been shown that all samples crystallize in a well-ordered cubic structure with the space group F d 3m. The vanadium (+4) content in the samples was determined by oxidative weight gain in air using a thermogravimetric (TG) balance. A structural phase transformation of cubic to tetragonal was observed by differential thermal analysis (DTA) in conjunction with high temperature diffractometry. The magnetic characterisation of the vanadate pyrochlores A2V2O7 (Y, Lu, Er and Dy) was performed by Katarina Removic-Langer in the laboratory of Prof. Dr. M. Lang. All materials studied are ferromagnetic. The ferromagnetic critical temperatures are between 70 and 73 K. In case of Er2V2O7 and Dy2V2O7 an additional increase in the magnetization was observed below 20 K. The increase in the magnetization below 20 K exhibited by Er2V2O7 and Dy2V2O7 originates from the interactions between the two magnetic sublattices (i.e., the rare earth- and the vanadium sublattice).
Innerhalb des adaptiven Immunsystems spielt der Major Histocompatibility Complex (MHC)-Klasse I-Weg der Antigenpräsentation eine essenzielle Rolle bei der Erkennung und Zerstörung Virus-infizierter Zellen. Ein grundlegender Schritt innerhalb dieses Prozesses ist die Translokation endogener Peptide durch den transporter associated with antigen processing (TAP) in das ER-Lumen. Der TAP-Transporter ist zusammen mit verschiedenen Chaperonen und weiteren Faktoren in einem Peptidbeladungskomplex (PLC) assoziiert. Insbesondere Herpesviren, die durch eine lebenslange Persistenz im Wirt und wiederkehrende Reaktivierung unter Stresssituationen gekennzeichnet sind, interferieren direkt mit dem PLC und dem TAP-Transporter. Das varicellovirale Typ-I-Membranprotein UL49.5 inhibiert den TAP-Komplex, wobei das Protein des Rinderherpesvirus (Bovines Herpesvirus 1, BHV-1) zusätzlich die proteasomale Degradation verschiedener Komponenten des PLCs einleitet. Dieser Mechanismus wird durch die C-terminale Domäne des UL49.5-Proteins vermittelt und ist von keinem anderen Virusprotein bekannt. Welche Aminosäuren des Virusproteins jedoch für diese Inhibition und Degradation essenziell sind, wurde bisher nicht aufgeklärt. Ziel der vorliegenden Doktorarbeit war es, die Funktionsweise des BHV-1 UL49.5-Proteins zu verstehen und insbesondere zu analysieren, welche Bereiche des Proteins für die proteasomale Degradation des TAP-Komplexes verantwortlich sind. Das UL49.5-Protein wurde im Rahmen der vorliegenden Arbeit erfolgreich in Insektenzellen und in HeLa-Zellen exprimiert. Mittels Coimmunpräzipitation (Co-IP) wurde daraufhin die Bindung verschiedener UL49.5-Varianten an den TAP-Komplex analysiert. Unterstützt wurden diese Daten durch einen in vivo Interaktionsscreen (BiFC) und in vitro translatiertes UL49.5. Hierbei stellte sich heraus, dass das UL49.5-Protein in Abwesenheit sämtlicher Komponenten des Immunsystems an beide Untereinheiten des TAP-Transporters bindet. Die Bindung erfolgt sowohl an vollständige TAP-Untereinheiten als auch an den sogenannten coreTAP-Komplex, der nur die inneren sechs Transmembranhelices besitzt. Weiterhin wurden systematisch verkürzte UL49.5-Varianten generiert, um wichtige Reste für TAP-Inhibition und proteasomale Degradation zu identifizieren. Interessanterweise sind weder die N-terminale noch die C-terminale Domäne von UL49.5 für die Bindung an den TAP-Komplex zwingend notwendig. Die Bindung an den TAP-Transporter wird demnach über die Transmembrandomäne von UL49.5 vermittelt. Mit Hilfe von Peptidtransport-Analysen wurde die inhibitorische Aktivität verschiedener UL49.5-Mutanten eingehend untersucht. Zusätzlich wurde eine Untersuchung der MHC I-Oberflächenexpression in transient transfizierten HeLa-Zellen etabliert. In diesen Zellen wurde nach Sortierung eine drastisch reduzierte TAP-Konzentration nachgewiesen, die auf proteasomale Degradation des TAP-Komplexes zurückzuführen war. Die Untersuchung von C-terminal verkürzten UL49.5-Mutanten zeigte, dass die letzten zwei C-terminalen Aminosäuren essenziell für die Induktion der TAP-Degradation sind. Die C-terminale Domäne von UL49.5 konnte jedoch, nach Übertragung auf andere Proteine, keine proteasomale Degradation des TAP-Komplexes einleiten. Demnach ist ein weiterer Bereich des Proteins für diesen Prozess zwingend notwendig. Erstaunlicherweise waren auch N-terminal verkürzte UL49.5-Proteine deutlich in ihrer inhibitorischen Funktion beeinträchtigt. Bereits nach der Deletion von 10 N-terminalen Aminosäuren war das Protein nicht mehr in der Lage, eine proteasomale Degradation des TAP-Komplexes einzuleiten. Demnach spielt auch die ER-luminale Domäne von UL49.5 eine wichtige Rolle bei der UL49.5-induzierten TAP-Degradation. Somit wurde ein bisher noch nicht beschriebener neuartiger Inhibitionsmechanismus für das BHV-1 UL49.5-Protein entdeckt. Nach Bindung von UL49.5 über die Transmembrandomäne an beide Untereinheiten des TAP-Transporters scheint die ER-luminale Domäne von UL49.5 ein Signal über die ER-Membran an die zytoplasmatische Domäne zu übertragen, die dann die proteasomale Degradation des TAP-Komplexes einleitet. Es konnte im Rahmen dieser Doktorarbeit erstmals gezeigt werden, dass additive Effekte eines sehr kleinen Virusproteins auf zwei unterschiedlichen Seiten der ER-Membran zu einer proteasomalen Degradation eines sehr großen Membran-Komplexes führen.
In der vorliegenden Arbeit wurden die Angaben von 355 Patientinnen ausgewertet, die zwischen Januar 2000 und September 2007 aufgrund eines metastasierten Brustkrebses im onkologischen Fachzentrum Frankfurt Nord-Ost behandelt worden sind. Die 5-JÜR nach Kaplan-Meier liegt im untersuchten Kollektiv bei 76 %. Die Wahrscheinlichkeit, fünf Jahre nach Erstdiagnose einer Metastasierung überlebt zu haben beträgt 50 %. Die mittlere Überlebenszeit ab Metastasierung liegt bei 7,4 Jahren. Die Wahrscheinlichkeit für metastasenfreies Überleben nach fünf bzw. zehn Jahren beträgt 60 % bzw. 44 %. Im univariaten Log-Rank-Verfahren weisen Patientinnen mit folgenden Charakteristika eine bessere Prognose hinsichtlich der Gesamtüberlebenswahrscheinlichkeit auf: Prämeno-pausaler Status bei ED, Ablatio mammae, CMF-Chemotherapie sowie operative und antihormonelle Therapie der Fernmetastasen. Als prognostisch ungünstig gelten: Positiver Nodalstatus, niedriger Differenzierungsgrad, invasiv duktaler oder lobulärer Tumortyp, höheres UICC-Stadium, Stadium > 2 der St. Gallener Risikoklassifikation, First-Line-Chemotherapie, R1-resezierte Tumoren, adjuvante Chemotherapie, > 3 verschiedene Metastasenorte und zytostatische Metastasentherapie. Die Lokalisation der Fernmetastasen ermöglicht eine differenziertere prognostische Einschätzung: Viszerale Metastasen, vor allem in der Leber und im ZNS, beeinflussen die Überlebenszeit negativ. Im Verlauf auftretende Metastasen im Abdomen/ Peritoneum gehen ebenfalls mit einer schlechteren Prognose einher. Bei ossärer Metastasierung hingegen kann mit einem benigneren Krankheitsverlauf gerechnet werden, ebenso bei Erstmanifestation der Metastasen in der Haut und/oder den Weichteilen. In der multivariaten Cox-Analyse übt die Leber als primärer Metastasierungsort den größten negativen Einfluss auf die Überlebens-wahrscheinlichkeit aus. Auch eine hohe Anzahl befallener Lymphknoten, Metastasen an > 3 verschiedenen Lokalisationen sowie ein positiver Resektionsrand führen zu signifikant schlechteren Überlebensraten. Bessere Prognosen besitzen Patientinnen, die zum Diagnosezeitpunkt prämenopausal gewesen sind und deren Metastasen im Verlauf antihormonell therapiert werden konnten. In der Cox-Regressionsanalyse erwiesen sich das Grading und der Nodalstatus als unabhängige Prognoseparameter hinsichtlich des metastasenfreien Überlebens. Demnach gilt: Je schlechter der Differenzierungsgrad eines Tumors und je mehr Lymphknoten befallen sind, desto niedriger ist die Wahrscheinlichkeit nach zehn Jahren metastasenfrei überlebt zu haben.
The β-subunits of Na,K-ATPase and H,K-ATPase have important functions in maturation and plasma membrane targeting of the catalytic α-subunit but also modulate the transport activity of the holoenzymes. In this study, we show that tryptophan replacement of two highly conserved tyrosines in the transmembrane domain of both Na,K- and gastric H,K-ATPase β-subunits resulted in considerable shifts of the voltage-dependent E1P/E2P distributions toward the E1P state as inferred from presteady-state current and voltage clamp fluorometric measurements of tetramethylrhodamine-6-maleimide-labeled ATPases. The shifts in conformational equilibria were accompanied by significant decreases in the apparent affinities for extracellular K+ that were moderate for the Na,K-ATPase β-(Y39W,Y43W) mutation but much more pronounced for the corresponding H,K-ATPase β-(Y44W,Y48W) variant. Moreover in the Na,K-ATPase β-(Y39W,Y43W) mutant, the apparent rate constant for reverse binding of extracellular Na+ and the subsequent E2P-E1P conversion, as determined from transient current kinetics, was significantly accelerated, resulting in enhanced Na+ competition for extracellular K+ binding especially at extremely negative potentials. Analogously the reverse binding of extracellular protons and subsequent E2P-E1P conversion was accelerated by the H,K-ATPase β-(Y44W,Y48W) mutation, and H+ secretion was strongly impaired. Remarkably tryptophan replacements of residues in the M7 segment of Na,K- and H,K-ATPase α-subunits, which are at interacting distance to the β-tyrosines, resulted in similar E1 shifts, indicating their participation in stabilization of E2. Thus, interactions between selected residues within the transmembrane regions of α- and β-subunits of P2C-type ATPases exert an E2-stabilizing effect, which is of particular importance for efficient H+ pumping by H,K-ATPase under in vivo conditions.
Biogenesis of mitochondrial cytochrome c oxidase (COX) relies on a large number of assembly factors, among them the transmembrane protein Surf1. The loss of human Surf1 function is associated with Leigh syndrome, a fatal neurodegenerative disorder caused by severe COX deficiency. In the bacterium Paracoccus denitrificans, two homologous proteins, Surf1c and Surf1q, were identified, which we characterize in the present study. When coexpressed in Escherichia coli together with enzymes for heme a synthesis, the bacterial Surf1 proteins bind heme a in vivo. Using redox difference spectroscopy and isothermal titration calorimetry, the binding of the heme cofactor to purified apo-Surf1c and apo-Surf1q is quantified: Each of the Paracoccus proteins binds heme a in a 1:1 stoichiometry and with Kd values in the submicromolar range. In addition, we identify a conserved histidine as a residue crucial for heme binding. Contrary to most earlier concepts, these data support a direct role of Surf1 in heme a cofactor insertion into COX subunit I by providing a protein-bound heme a pool.
