Das aktuell diskutierte Modell der lichtabhängigen Magnetrezeption bei Vögeln beschreibt, dass die Richtung des Erdmagnetfelds durch Radikalpaarprozesse in spezialisierten Photorezeptoren wahrgenommen wird. Dabei werden Cryptochrome, eine Klasse photoaktiver Flavoproteine, als potentielle Kandidaten für das Radikalpaarmodell und damit als mögliches Magnetrezeptormolekül diskutiert. Verhaltensbiologische Experimente mit Zugvögeln zeigen, dass der Magnetrezeptionsprozess offensichtlich stark lateralisiert im rechten Auge stattfindet und dass dieser Prozess Licht aus dem blau-grünen Teil des Spektrums benötigt. Cryptochrome absorbieren Licht in diesem Bereich und besitzen darüber hinaus biochemische Eigenschaften, die für die Funktion des Radikalpaarmodells entscheidend sind. Vor dem Hintergrund dieser Überlegungen habe ich untersucht, ob Cryptochrom (CRY) in der Retina des Rotkehlchens, Erithacus rubecula, einem nachtziehenden Singvogel, zu finden ist. Mit molekulargenetischen Methoden konnte ich erstmals drei individuell exprimierte Cryptochrome aus der Rotkehlchenretina isolieren: Erithacus (e)-CRY1a, -CRY1b und -CRY2. Während eCRY1a und eCRY2 fast vollständig homolog zu Cryptochrom 1 und 2 in der Retina des Haushuhns sind, besitzt eCRY1b eine noch unbeschriebene C-terminale Domäne, die auf neue Proteinbindungseigenschaften und Interaktionspartner hindeutet. Die Identifizierung eines Kernlokalisierungssignals bei eCRY2 weist auf dessen Lokalisierung im Zellkern hin und macht seine Funktion als sensorischer Lichtrezeptor eher unwahrscheinlich. Ein mit Hilfe der Aminosäurensequenz erstelltes Proteinmodell von eCRY1 zeigt, dass dieser Typus zwei Cofaktoren besitzt: das katalytische Chromophor Flavinadenindinukleotid (FAD) und das lichtsammelnde Pterin Methenyltetrahydrofolat (MTHF). eCRY1a und eCRY1b sind Spleißprodukte des gleichen Gens; ihre C-terminalen Domänen sind auf unterschiedlichen Exonen codiert. eCRY1a und eCRY1b besitzen beide keine Membranbindedomäne und liegen zytosolisch vor. Weil jedoch laut Radikalpaartheorie die magnetosensitiven Rezeptoren zumindest im Moment der Photonenabsorption in einer bestimmten Anordnung und Raumrichtung vorliegen müssen, benötigen beide eCRY1-Varianten daher mindestens einen sphärisch fixierten Interaktionspartner. In meiner Arbeit diskutiere ich Opsine als mögliche Partner, da diese in den Aussensegmenten der Photorezeptoren stark geordnet und in konstanter Raumrichtung vorliegen. Die Genexpression von eCRY1a und eCRY1b unterscheidet sich sowohl zwischen den Augen als auch zwischen den CRY-Varianten. Im Mittel waren die Expressionswerte von eCRY1a in der Rotkehlchenretina fünf- bis zehnmal höher als die von eCRY1b. In der Dämmerung und nachts ist eCRY1a im linken Auge signifikant höher exprimiert als im rechten Auge. Im Gegensatz dazu ist die Expression von eCRY1b in beiden Augen gleich, zeigt jedoch einen signifikanten Anstieg zu Beginn des Zuges in der Dämmerung. Durch die unterschiedlichen Expressionsraten von eCRY1a und eCRY1b verschiebt sich zum Zeitpunkt der Richtungsentscheidung des Vogels in der Dämmerung das Verhältnis zwischen den beiden Cryptochrom1-Varianten im rechten Auge zugunsten von eCRY1b. Die Ergebnisse meiner Expressionsstudie deuten darauf hin, dass die in Verhaltensversuchen nachgewiesene Lateralisation des Magnetkompass bereits auf Rezeptorebene in der Retina beginnen könnte. mRNA in situ- als auch immunohistochemische Untersuchungen dieser Arbeit zeigen, dass bei Zugvögeln sowohl die mRNA als auch die Proteine beider eCRY1-Typen in den Innensegmenten der retinalen Photorezeptoren lokalisiert sind. Die räumliche Nachbarschaft zu Opsinen unterstützt die Annahme einer möglichen Interaktion zwischen diesen und Cryptochrom. Darüber hinaus lässt dieser Befund Spekulationen zu, dass die neuronale Verarbeitung der aus den Photorezeptoren stammenden magnetischen Richtungsinformation analog zum bildverarbeitenden Sehen stattfinden könnte. Modelle für eine mögliche Verschaltung und Signalverarbeitung der Cryptochromsignale werden in dieser Arbeit diskutiert. Insgesamt unterstützen die Befunde meiner Arbeit die im Radikalpaarmodell diskutierte Annahme, dass Cryptochrome eine entscheidende Rolle bei der Perzeption von magnetischer Kompassinformation bei Zugvögeln spielen und möglicherweise als Magnetrezeptormoleküle fungieren. Darüber hinaus könnten die Ergebnisse einen wichtigen Beitrag zu einem besseren Verständnis des Gesamtprozesses der Magnetrezeption bei Tieren leisten, und zwar von der Rezeptorzelle bis zur Verhaltensantwort. Meine Ergebnisse tragen möglicherweise auch dazu bei, die bekannten neurowissenschaftlichen und funktionell morphologischen mit den entsprechenden ethologischen Ergebnissen zu verknüpfen. Ich würde es sehr begrüßen, wenn meine Arbeit zu weiterführender Forschung auf diesem spannenden Gebiet anregt und zu einem besseren Verständnis der noch unbekannten Aspekte der Magnetrezeption beitrüge.
Tens of thousands of man-made chemicals are in regular use and discharged into the environment. Many of them are known to interfere with the hormonal systems in humans and wildlife. Given the complexity of endocrine systems, there are many ways in which endocrine-disrupting chemicals (EDCs) can affect the body’s signaling system, and this makes unraveling the mechanisms of action of these chemicals difficult. A major concern is that some of these EDCs appear to be biologically active at extremely low concentrations. There is growing evidence to indicate that the guiding principle of traditional toxicology that “the dose makes the poison” may not always be the case because some EDCs do not induce the classical dose–response relationships. The European Union project COMPRENDO (Comparative Research on Endocrine Disrupters—Phylogenetic Approach and Common Principles focussing on Androgenic/Antiandrogenic Compounds) therefore aims to develop an understanding of potential health problems posed by androgenic and antiandrogenic compounds (AACs) to wildlife and humans by focusing on the commonalities and differences in responses to AACs across the animal kingdom (from invertebrates to vertebrates).
Polyploidy is common in higher eukaryotes, especially in plants, but it is generally assumed that most prokaryotes contain a single copy of a circular chromosome and are therefore monoploid. We have used two independent methods to determine the genome copy number in halophilic archaea, 1) cell lysis in agarose blocks and Southern blot analysis, and 2) Real-Time quantitative PCR. Fast growing H. salinarum cells contain on average about 25 copies of the chromosome in exponential phase, and their ploidy is downregulated to 15 copies in early stationary phase. The chromosome copy number is identical in cultures with a twofold lower growth rate, in contrast to the results reported for several other prokaryotic species. Of three additional replicons of H. salinarum, two have a low copy number that is not growth-phase regulated, while one replicon even shows a higher degree of growth phase-dependent regulation than the main replicon. The genome copy number of H. volcanii is similarly high during exponential phase (on average 18 copies/cell), and it is also downregulated (to 10 copies) as the cells enter stationary phase. The variation of genome copy numbers in the population was addressed by fluorescence microscopy and by FACS analysis. These methods allowed us to verify the growth phase-dependent regulation of ploidy in H. salinarum, and they revealed that there is a wide variation in genome copy numbers in individual cells that is much larger in exponential than in stationary phase. Our results indicate that polyploidy might be more widespread in archaea (or even prokaryotes in general) than previously assumed. Moreover, the presence of so many genome copies in a prokaryote raises questions about the evolutionary significance of this strategy.
RNA-Interferenz (RNAi) erlangte eine herausragende Bedeutung zum Studium der Genfunktion, nachdem auch in Säugersystemen gezeigt wurde, daß durch Applikation von 21 nt langen siRNAs (small interfering RNAs) eine Sequenz-spezifische Degradierung der mRNA-Transkripte kognitiver Gene erreicht werden konnte. Im Gegensatz zur antisense-Technologie erwies sich die Wirkung von siRNA im Hinblick auf die Hemmung der Genexpression um ein Vielfaches potenter und hoch spezifisch. Für eine längerfristige Unterdrückung von Genen kristallisierte sich die Methode der Plasmid-Vektor-vermittelten intrazellulären Expression von shRNA (short hairpin RNA) heraus, welche transient oder stabil angewendet werden kann. Diese exprimierte shRNA wird intrazellulär enzymatisch zu wirksamer siRNA prozessiert, welche den eigentlichen Enzym-vermittelten RNAi-Mechanismus der Degradierung von mRNA-Transkripten kognitiver Gene auslöst. Die Anwendung von RNA-Polymerase III-abhängigen Promotoren für die stabile konstitutive Expression von shRNA stellte ein großes Problem für behandelte Zellen dar, wenn es sich bei dem zu unterdrückenden Zielgen um ein Gen mit essentiellen Funktionen für die Zelle handelte. Im Falle der Polo-like Kinase 1 (Plk1), einer in vielen Spezies hoch konservierten Serin/Threonin-Kinase mit essentiellen mitotischen Funktionen, bedeutete eine dauerhafte und stringente Unterdrückung einen veränderten Phänotyp beteiligter Zellen, welcher sich durch Defekte bei mitotischen Ereignissen bemerkbar machte. Plk1 ist in zahlreiche mitotische Prozesse, wie den Eintritt der Zellen in die Mitose, die Segregation der Chromosomen und die Aktivierung des APC/C (anaphase promoting complex / cyclosome), eingebunden. Darüber hinaus ist bekannt, daß Plk1 in nahezu allen Tumorarten überexprimiert vorliegt und die Prognose von Tumorwachstum und Metastasierungspotential über den Plk1-Gehalt definiert werden kann. Des weiteren bewirkte eine RNAi-vermittelte Unterdrückung der Plk1-Expression bei Tumorzellen eine Hemmung der Zellproliferation mit Auslösung der Apoptose. Hingegen konnte bei gesunden primären Zellen weder eine signifikante Hemmung der Proliferation noch die Auslösung der Apoptose beobachtet werden, was die große Bedeutung von Plk1 als Ansatzpunkt für eine Krebstherapie hervorhebt. Um die Funktion von Plk1 im Hinblick auf molekularbiologische Zusammenhänge besser studieren zu können, war es notwendig, den intrazellulären Plk1-Gehalt zu variieren. Im Rahmen dieser Promotionsarbeit wurden dazu induzierbare RNAi-Elemente entwickelt, mit deren Hilfe die intrazelluläre Plk1-Expression konditionell inhibiert werden konnte. Unter Verwendung des prokaryotischen Tet-Systems wurden auf Basis des RNA-Polymerase abhängigen H1-Promotors durch Insertion von einer oder zwei Operatorsequenzen (TetO) für den Tetrazyklin-Repressor (TetR) an verschiedene Orte innerhalb der Sequenz des H1-Promotors drei induzierbare Promotor-Derivate geschaffen. Die drei entwickelten H1-Promotor-Derivate wurden zur Expression von shRNA gegen Plk1 eingesetzt und in bezug auf die Auslösung der RNAi-Antwort getestet und untereinander verglichen. Zu diesem Zwecke wurde der endogene Plk1-Gehalt von HeLa-Tumorzellen auf Transkript- und auf Proteinebene bestimmt. Die Zellen wurden zuvor mit Plasmid-Vektoren für konstitutive TetR-Expression und jeweils einer der verschiedenen shRNA-Expressions-Kassetten ko-transfiziert. Als Kontrollen dienten dabei Wildtyp-H1-Promotoren, welche zur konstitutiven Expression von shRNA gegen Plk1 und einer unwirksamen Kontroll-shRNA eingesetzt wurden. Mit Hilfe des synthetischen Tetrazyklin-Analogons Doxyzyklin, welches einen potenten Aktivator für TetR darstellt, konnten die hergestellten Promotor-Derivate induziert werden, was durch einen reduzierten intrazellulären Plk1-Gehalt sichtbar wurde. Dabei fiel auf, daß alle drei Promotor-Typen unterschiedliche Eigenschaften im nichtinduzierten Zustand wie auch im induzierten Zustand unter Anwesenheit von Doxyzyklin aufwiesen. Für die Basalaktivität in Abwesenheit von Doxyzyklin (leakiness) war die relative Lage der TetO-Sequenz(en) innerhalb des Promotors verantwortlich. So veränderte die Insertion einer TetO-Sequenz in 3’-Richtung der TATA-Box die Eigenschaften des Wildtyp-H1-Promotors weniger als die Insertion einer TetO-Sequenz in 5’-Richtung der TATA-Box. Zum Studium von Plk1 als Zielgen in der Krebstherapie wurde das Proliferationsverhalten von HeLa-Tumorzellen als Antwort auf die Doxyzyklin-vermittelte Induktion der shRNAExpression unter Kontrolle eines der induzierbaren Promotoren ermittelt. Dabei konnte die Proliferation von Tumorzellen durch einen reduzierten Plk1-Gehalt, welcher durch die Doxyzyklin-induzierte Auslösung der RNAi-Antwort vermittelt wurde, erfolgreich inhibiert werden. Zur Überprüfung der Eignung entwickelter Systeme, Tumorwachstum in vivo inhibieren zu können, wurden die entwickelten RNAi-Kassetten in das Genom von TetRexprimierenden HeLa-Zellen integriert und so stabile Klone geschaffen. Stabile HeLa-Klone zur induzierbaren Expression von Plk1-spezifischer shRNA, wie auch zur induzierbaren Expression von einer Kontroll-shRNA, wurden in die gegenüberliegenden Flanken von immunsupprimierten Nacktmäusen inokuliert, um anhand von Xenograft-Modellen einen direkten Vergleich des Tumorwachstums unter gleichen äußeren Bedingungen zu ermöglichen. Einem Teil der Mäuse wurde Doxyzyklin ins Trinkwasser gegeben, während Kontrollmäuse kein Doxyzyklin verabreicht bekamen. Das Tumorwachstum von Xenograft-Tumoren, welche aus Klonen zur Expression von shRNA gegen Plk1 hervorgingen, konnte in Doxyzyklin-behandelten Mäusen um 53% auf 47% an Tag 51 nach Inokulierung der Zellen inhibiert werden. Tumoren nicht-induzierter Mäuse, sowie Tumoren aus induzierten Mäusen, welche Kontroll-shRNA exprimierten, wuchsen dagegen unverändert in gleichem Maße. Anhand der in dieser Arbeit entwickelten induzierbaren H1-Promotor-Derivate zur konditionellen Auslösung von RNAi wurden wertvolle genetische Werkzeuge geschaffen, welche für das Studium der Genfunktion eingesetzt werden können. Im Falle der Unterdrückung von Plk1 können mit ihrer Hilfe sowohl grundlegende molekularbiologische Zusammenhänge studiert als auch die Bewertung von Plk1 als Zielgen in der Krebstherapie bewertet werden. Im Gegensatz zu kürzlich entwickelten Kinase-Inhibitoren, welche auf Proteinebene gegen Plk1 gerichtet sind und aufgrund ihrer bislang nicht nachgewiesenen Spezifität verwandte Kinasen in ihrer Wirkungsweise beeinflussen könnten, ist eine RNAibasierte Strategie hoch spezifisch und verspricht eine große Relevanz für zukünftige therapeutische Ansätze. Vorraussetzung für erfolgversprechende RNAi-basierte Strategien ist eine hohe Konservierung der Sequenz beteiligter Zielgene. Im Falle von Plk1 konnte eine hohe Konservierung durch Sequenzanalyse der Plk1-Gene von 15 Mamma-Karzinomen, 11 Ovarial-Karzinomen und mehrerer Tumorzellinien bestätigt werden.
Synaptopodin is the founding member of a family of actin-associated proline-rich proteins. It is present in a subset of telencephalic dendritic spines, where it is tightly associated with the dendritic spine apparatus, a putative calcium store. Synaptopodin-deficient mice lack the spine apparatus and show deficits in long-term potentiation and spatial memory. Thus, synaptopodin appears to play a role in synaptic plasticity. In the present thesis, three major questions were addressed: (1) What is the distribution of synaptopodin and the spine apparatus in identified hippocampal neurons? (2) Is the distribution of synaptopodin affected by denervation? (3) Is synaptopodin involved in the regulation of denervation-induced spine loss? The major findings of this thesis are: (1) Immunohistochemistry in the hippocampus of wildtype and EGFP-transgenic mice revealed significant layer-specific differences in the prevalence of synaptopodin at the level of individual neurons. (2) Light and electron microscopic analysis also revealed the presence of synaptopodin in axon initial segments of cortical and hippocampal principal neurons. There, it was found to be an essential component of the cisternal organelle, a putative axonal homologue of the dendritic spine apparatus. (3) Immunohistochemistry in the rat fascia dentata before and following entorhinal deafferentation revealed changes in synaptopodin expression in denervated and non-denervated layers of the hippocampus, suggesting that the distribution of synaptopodin in hippocampal neurons is regulated by presynaptic signals. (4) The dynamics of denervation-induced spine plasticity were studied in vitro using confocal live imaging of organotypic entorhino-hippocampal slice cultures. Whereas spines were remarkably stable under control conditions, spine loss and spine formation were seen following denervation. No significant differences were observed between cultures from wildtype and synaptopodin-deficient mice, suggesting that synaptopodin is not involved in lesion-induced spine plasticity. (5) Finally, a set of transgenic mice expressing fluorescently tagged synaptopodin were generated to facilitate future experiments on the dynamics and function of synaptopodin. In summary, this thesis presents novel findings on (1) the subcellular distribution of synaptopodin in spines and the axon initial segment, (2) the molecular composition of the cisternal organelle, and (3) the dynamics of spines and the spine apparatus organelle following deafferentation in vivo and in vitro.
Since its recognition as an endothelium-derived relaxing factor, the control and consequences of nitric oxide (NO) production have been investigated intensely. We know now that NO is not simply a vasodilator or regulator of smooth muscle tone but is a potent anti-platelet agent, neuromodulator and regulator of gene expression. NO is synthesized from the amino acid Larginine by a family of enzymes termed NO synthases (NOS). The ‘endothelial’ (eNOS or NOS III) and ‘neuronal’ (nNOS, NOS I or bNOS) NOS isoforms, which were named after the tissues in which they were first identified, are expressed constitutively and are generally regulated by Ca2+/calmodulin (CaM). Endothelium-derived NO is thought to be responsible for maintaining the vasculature in an anti-atherosclerotic state and a decrease in the bioavailability of NO (a state generally referred to as endothelial dysfunction) results in “proatherosclerotic” alterations in vascular gene expression. Recently it has become clear that the activity of eNOS is largely determined by its association with regulatory proteins as well as by the phosphorylation of the enzyme on serine, threonine and possibly tyrosine residues. Moreover, the enzyme can be “uncoupled” i.e. transformed from a NO generating to a superoxide (O2-)-generating enzyme, which would be expected to attenuate vasodilator responses and enhance vascular inflammation. The aim of this thesis was to study the consequences of phosphorylation on specific serine, threonine and tyrosine residues on the activity and intracellular localisation of eNOS and in particular to determine whether a phospho-switch for eNOS uncoupling exists. eNOS is phosphorylated under basal conditions and its serine phosphorylation can be enhanced following cell stimulation with hemodynamic stimuli such as cyclic stretch and fluid shear stress as well as by hormonal stimuli such as histamine and bradykinin. Our group has previously demonstrated the importance of Ser1177 in the activation of eNOS and here I set out to determine the relative importance of phosphorylation on Ser633 and Ser114. By generating point mutants in which serine was replaced by either alanine (nonphosphorylatable mutants) or aspartate (phosphomimetic mutants) it was observed that the activity of the S633D and S114A eNOS mutants exhibited an 2-fold increase over the activity of the wild-type enzyme or either of the S633/634A or S114D eNOS mutants as determined by monitoring the conversion of L-arginine to L-citrulline. eNOS is basally phosphorylated on Thr495 and stimulation of endothelial cells with Ca2+-elevating agonists generally results in the transient dephosphorylation of this residue. The latter is essential to allow the binding of calmodulin to the enzyme and is the actually initiating step in the generation of NO. Correspondingly, the T495A eNOS mutant can be activated at lower Ca2+ and calmodulin concentrations than the T495D mutant. However, some eNOS mutants (T494A/S1177D and T495A) showed an enhanced ability to generate O2- in a NOS inhibitor-sensitive manner suggesting that the phosphorylation of the enzyme may also play a role in the uncoupling process. To determine the physiological relevance of eNOS dephosphorylation on Thr495 we assessed the consequences of treating cells with oxidised low-density lipoprotein (ox-LDL) on eNOS phosphorylation as well as on the eNOS-dependent generation of NO and O2-. Oxidised LDL concentration- and time-dependently decreased phosphorylation of eNOS on Thr495 and led to a concomitant decrease in cellular levels of cyclic GMP and an enhanced production of O2 - compared to cells treated with native LDL. Alterations in the activity of protein kinase C (PKC) were related to the change in eNOS Thr495 phosphorylation. There was not only the basal activity of PKCα inhibited by ox-LDL but the PKC activator phorbol-12-myristate-13-acetate also failed to elicit the phosphorylation of Thr495 in ox-LDL-treated endothelial cells. The dephosphorylation of eNOS on Thr495 in response to the addition of ox-LDL was not associated with an increase in the binding of calmodulin to eNOS, an association usually necessary for the activation of eNOS. Moreover, following treatment with ox-LDL for 24 hours eNOS was no longer detected at the plasma membrane but was redistributed to the cytosol indicating that ox-LDL may disrupt the eNOS signalling complex or signalosome. To date the role played by the tyrosine phosphorylation of eNOS in the regulation of its activity or intracellular association is controversial. However, during the preparation of this thesis we have been able to demonstrate a link between the tyrosine phosphorylation of eNO and the activation of the tyrosine kinases Src and PYK2. The application of fluid shear stress to endothelial cells resulted in the activation of Src and PYK2 as well as in the association of PYK2 with eNOS. Co-expression of eNOS and PYK2 led to the putative identification of Tyr657 as a potential modulatory site. Mutating eNOS at Tyr657 to Asp or Glu resulted in the localisation of the mutant eNOS predominantly in the cytoskeleton and also in a complete inactivation of the enzyme. The Y657F mutants, on the other hand, did not demonstrate any marked alteration in the activity when compared with the wild-type eNOS. However, the In conclusion, the results describe in this thesis indicate that eNOS is regulated by phosphorylation at multiple sites. Depending on the phosphorylation site involved phosphorylation can inhibit or activate NO production or even uncouple the enzyme so that it generates O2-. While the phosphor-status of eNOS on Ser114 and Ser633 influenced NO release they did not contribute to O2 - production and the dephosphorylation of Thr495 seems sufficient to uncouple eNOS. Cell treatment with ox-LDL, which is known to increase eNOS-derived O2- output was correlated with a dephosphorylation of Thr495 as well as a decrease in the activity of the kinase that phosphorylates this site i.e., PKCα. The phosphorylation status of all the eNOS serine and threonine residues studied however did not influence the ability of the enzyme to dimerise, indicating that contrary to previously published reports the eNOS dimer is highly stable in endothelial cells. The tyrosine phosphorylation of eNOS was not initially expected to play a determinant role in the regulation but rather to facilitate the docking of associated regulatory proteins. However, Tyr657 seems to play a critical role in the generation of NO as its mutation resulted in the generation of a completely inactive enzyme as well as in an apparent intracellular mislocalisation of the protein. The physiological relevance of these findings remain to be further elucidated.
Compared to all other organisms with 1 to 3 heat stress transcription factors (Hsfs) or Hsf-related factors, plants have extraordinarily large Hsf families with more than 20 Hsfs. Plant Hsfs are classified into three classes according to their oligomerization domains which is built of hydrophobic heptad repeats (HR) in two parts, HR-A and HR-B. Both parts may be immediately adjacent (class B), or they are separated by insertion of 21 (class A) and 7 amino acid residues (class C). In plant Hsf family, detailed investigations are so far limited to Hsfs A1a, A2, A3, A4d, A9, and B1. They strongly indicate functional diversification to be the main reason for the coexistence of multiple Hsfs. As an example the functional triad of HsfA1a, HsfA2, and HsfB1 is essential for all three phases of the hs response, (i) the triggering of the response by HsfA1a as master regulator, (ii) the maintenance and high efficiency of hs gene transcription by cooperation of HsfA1a with Hsfs A2 and B1, and finally, (iii) the restoration of house-keeping gene transcription during the recovery phase mediated by HsfB1 in cooperation with house-keeping transcription factors. The results presented in this thesis for Hsfs A4 and A5 open completely different aspects of functional diversification and cooperation of Hsfs. HsfA4 and HsfA5 homooligomerize and bind to corresponding HSE motifs. But in contrast to the highly active HsfA4, HsfA5 is completely inactive as transcriptional activator. Yeast two hybrid and GST pull-down techniques showed that both Hsfs have strong tendency for heterooligomerization. Using fluorescence microscopy the HsfA4/A5 heterooligomers were found to localize in the nucleus. These complexes are transcriptionally inactive due to the impairment of DNA binding. The repressor function of HsfA5 requires only its OD and no additional factors, e.g. a putative co-repressor recruited by the C-terminal domain, are involved. Evidently, the repressor effect mainly results from the interference with the oligomeric state of HsfA4b, which is essential for efficient DNA binding and activator functions. EST database search revealed that plants have a single HsfA5 and usually two A4-type Hsfs. Using bioinformatics tools, Hsfs A4 and A5 were found to be phylogenetically closely related and clearly distinct from the other members of the Hsf family. On the basis of RT-PCR and Microarray data the representatives of the A4/A5 group are well expressed in different plant tissues albeit at very different levels which change with the developmental stages and stress conditions In rice and Arabidopsis, HsfA4 functions as an anti-apoptotic factor for stress induced oxidative damages. Based on my results, I hypothesize that HsfA5 functions as a novel type of selective repressor, regulating the function of A4-type Hsfs in plants. Considering the high sequence conservation with in plant Hsf family, it is tempting to speculate that this role of Hsf4/A5 pair is a fundamental feature of the Hsf system in plants.
Die Rolle des cAMP/CREB-Signalwegs in der noradrenergen Differenzierung sympathischer Nervenzellen

Um die sympathische Differenzierung in Abwesenheit von CREB zu untersuchen, wurden zunächst Embryonen von CREB-Null-Mäusen analysiert. In diesen Mäusen wiesen die sympathischen Ganglien an Embryonaltag E10,5 und E12 keine Unterschiede in der Differenzierung zu heterozygoten und wildtyp-Ganglien auf. Es wurde deshalb vermutet, dass die mit CREB interagierenden Transkriptionsfaktoren CREM und ATF1 den Verlust von CREB im sympathischen Ganglion funktionell kompensieren. Da Mäuse mit einem Knock-out von CREB und CREM bereits vor E10 sterben, konnte die Entwicklung sympathischer Ganglien in diesen Mäusen nicht weiter untersucht werden. Durch Immunfärbung gegen ATF1 in CREB-deletierten Mäusen und in situ-Hybridisierung gegen CREM und ATF1 wurde gezeigt, dass im Rumpf der CREB-KO-Maus keine drastische Hochregulation der Expression beider Gene stattfindet. Eine geringfügige Hochregulation konnte aber nicht ausgeschlossen werden kann. Um die cAMP/CREB-Signaltransduktion in Ganglienvorläuferzellen auszuschalten, und Kompensation durch CREM und ATF1 zu umgehen, wurden die dominant-negative Effektoren ACREB und PKA-R(I)mut verwendet. ACREB blockiert die Funktion von CREB, CREM und ATF1 (Ahn et al., 1998). Durch PKA-R(I)mut sollte die Aktivität von PKA inhibiert werden. Die Funktionalität beider Effektoren wurde in differenzierenden Zellkulturen bestätigt. Um in vivo geeignete Expression der Inhibitoren zu erzielen, wurden verschiedene Methoden des Gentransfers im Huhnembryo erprobt. Durch Infektion der Neuralplatte des Embryos mit Retroviruskonzentrat konnte frühe embryonale Expression der dominant-negativen Effektoren in Neurallleistenzellderivaten erzielt werden. Die Expression von ACREB im Embryo zeigte, dass CREB in der initialen adrenergen Differenzierung bis E4 erforderlich ist, die sympathischen Ganglien wiesen reduzierte adrenerge Genexpression auf. In späteren Embryonalstadien ist dieser Effekt nicht mehr nachzuweisen. Im Gegensatz zur Inhibition von CREB, hatte die Inhibition von PKA durch PKAR(I)mut keine Auswirkungen auf die Ganglienentwicklung in vivo. Durch pharmakologische Inhibition von PKA wurden kleinere Ganglien erzielt, ein selektiver Effekt auf die adrenerge Differenzierung wurde auch in diesem in vivo Experiment nicht beobachtet. Dies widerspricht den Hinweisen aus Zellkultur auf die selektive Funktion von PKA in der adrenergen Differenzierung. Der Befund, dass die Funktion von CREB in vivo auf die initiale adrenerge Differenzierung sympathischer Neurone beschränkt ist, und nicht für den Erhalt adrenerger Differenzierung erforderlich ist, wurde in vitro in differenzierten sympathischen Neuronen des Embryo bestätigt. ACREB und PKA-R(I)mut hatten keine bzw. nur geringe Auswirkungen auf den Anteil TH-positiver Neurone in Kulturen sympathischer Neurone aus Embryonen der Embryonaltage E7 und E12. Somit konnte erstmals in vivo eine physiologische Rolle für CREB in der Differenzierung sympathischer Neurone gezeigt werden: CREB ist für die initiale Expression von TH erforderlich.
In dieser Arbeit wurde die Verteilung von Glykolyseenzymen in Kulturzellen unter normalen und die Glykolyse stimulierenden und hemmenden Bedingungen untersucht. Die Hauptfunktion, die der Strukturbindung und -Bildung von Glykolyseenzymen zugeschrieben wird, ist das so genannte metabolite-channelling bzw. substratechannelling, also die effiziente, lokale Bereitstellung von ATP. Im Mittelpunkt dieser Untersuchungen stand Aldolase (EC 4.1.2.13). Unter allen Bedingungen liegt Aldolase, wie die meisten anderen Glykolyseenzyme gleichzeitig sowohl in strukturgebundener, als auch in gelöster Form im Cytoplasma vor. Unter normalen Kulturbedingungen wurde die strukturgebunde Aldolase an Stressfasern sowie dem fibrillären Aktin-Netzwerk des Zellkortex vorgefunden. Diese Beobachtungen wurden an fixierten Zellen gemacht; an lebenden Zellen konnte die Bindung an Stressfasern durch Mikroinjektion fluorochromierter Aldolase direkt nachgewiesen werden. Ferner wurde beobachtet, dass Aldolase selbst, unabhängig vom Vorhandensein von Aktin in der Zelle netzwerkartige Strukturen bilden kann. Das Hinzufügen von Aldolase zu in vitro polymerisierten Aktinfilamenten führte zu einer Bündelung der Aktinfilamente, die wahrscheinlich auf eine quervernetzende Wirkung dieses Enzyms zurückzuführen ist. Durch Injektionen mit Gemischen unterschiedlich markierter Glykolyseenzyme konnte gezeigt werden, dass diese Enzyme auch in gelöster Form nicht völlig homogen in den Zellen verteilt sind. In Gegenwart hoher Substratkonzentrationen wurde entdeckt, dass Aldolase in großem Ausmaß an Intermediärfilamente gebunden wird. Da dieses Verhalten zwar unabhängig von der Präparationsmethode auftrat, in vitro durch Kosedimentation aber nicht bestätigt werden konnte, ist anzunehmen, dass neben dem Substrat weitere Faktoren bei diesem Phänomen eine Rolle spielen. Die Applikation von Insulin führte zu keinerlei mikroskopisch beobachtbaren Änderungen im Bindungsverhalten der Aldolase. Dennoch konnte eine geringe Steigerung der Aktivität in der unlöslichen Fraktion des Zellproteins nachgewiesen werden. Die Mikroinjektion unphysiologisch hoher Mengen von Aldolase führte zu einem drastischen Abbau von Aktinfilamenten und Stressfasern in den injizierten Zellen. Andere Glykolyseenzyme (GAPDH, PGM, LDH und TIM) zeigten dagegen keinerlei Wirkung. Der selbe Effekt ist bereits für PFK beschrieben worden, wo er durch eine starke Assoziation der PFK mit Gelsolin hervorgerufen wird. Der Wirkungsmechanismus bei der Aldolase ist dagegen bislang nicht bekannt. Durch die Inkubation fixierter Zellen mit fluorochromierter Aldolase wurden eine große Zahl freier Bindungsstellen an Mikrotubuli entdeckt. Allerdings konnten keine Bedingungen ermittelt werden, unter denen die intrazelluläre Aldolase an Mikrotubuli bindet. Weiterhin wurde ein Experiment entworfen, um die lokale Bereitstellung von Energie in Form von ATP und metabolite-channelling nachzuweisen. Dazu wurden Monolayerkulturen durch Hemmung von Atmung und Glykolyse ATP-entleert und durch Permeabilisierung alle löslichen Bestandteile freigesetzt. Die Gabe verschiedener ATP-Konzentrationen führte in diesem Fall zu einer Kontraktion der verbleibenden Zellbestandteile, wobei die Kontraktionsgeschwindigkeit abhängig von der gegebenen ATPKonzentration war. Durch die Gabe von FBP und ADP konnte diese Kontraktion ebenfalls ausgelöst werden, was für die Konversion von ADP zu ATP wodurch ein von FBP und den strukturgebundenen Enzymen unterhaltener glykolytischen FluSS und somit metabolite-channelling nachgewiesen wurde.
Background Identification and evaluation of surface binding-pockets and occluded cavities are initial steps in protein structure-based drug design. Characterizing the active site's shape as well as the distribution of surrounding residues plays an important role for a variety of applications such as automated ligand docking or in situ modeling. Comparing the shape similarity of binding site geometries of related proteins provides further insights into the mechanisms of ligand binding. Results We present PocketPicker, an automated grid-based technique for the prediction of protein binding pockets that specifies the shape of a potential binding-site with regard to its buriedness. The method was applied to a representative set of protein-ligand complexes and their corresponding apo-protein structures to evaluate the quality of binding-site predictions. The performance of the pocket detection routine was compared to results achieved with the existing methods CAST, LIGSITE, LIGSITEcs, PASS and SURFNET. Success rates PocketPicker were comparable to those of LIGSITEcs and outperformed the other tools. We introduce a descriptor that translates the arrangement of grid points delineating a detected binding-site into a correlation vector. We show that this shape descriptor is suited for comparative analyses of similar binding-site geometry by examining induced-fit phenomena in aldose reductase. This new method uses information derived from calculations of the buriedness of potential binding-sites. Conclusions The pocket prediction routine of PocketPicker is a useful tool for identification of potential protein binding-pockets. It produces a convenient representation of binding-site shapes including an intuitive description of their accessibility. The shape-descriptor for automated classification of binding-site geometries can be used as an additional tool complementing elaborate manual inspections.
