1. There are two classes of theories of Universal Grammar: (1) Formalist theories, such as the widespread varieties of generative grammar. These theories start from the assumption that certain strings of linguistic forms are grammatical while other strings are ungrammatical. A grammar of this type produces grammatical strings and does not produce ungrammatical ones. All theories of this class fail in the same respect: they do not account for the meaning of the strings. (2) Semiotactic theories, which describe the meaning of a string in terms of the meanings of its constituent forms and their interrelations. The only elaborate formalized theory of this class presently available is the one advanced by C.L. Ebeling (Syntax and Semantics, Leiden: Brill, 1978). I shall discuss some of its mathematical properties here.
Most scholars nowadays reconstruct a static root present with an alternation between lengthened grade in the active singular and full grade in the active plural and in the middle. I am unhappy about this traditional methodology of loosely postulating long vowels for the proto-language. What we need is a powerful theory which explains why clear instances of original lengthened grade are so very few and restrains our reconstructions accordingly. Such a theory has been available for over a hundred years now: it was put forward by Wackernagel in his Old Indic grammar (1896: 66-68). The crucial element of his theory which is relevant in the present context is that he assumed lengthening in monosyllabic word forms, such as the 2nd and 3rd sg. active forms of the sigmatic aorist injunctive.
The philosophy of language comes in three varieties. 1. The functionalist’s view: linguistic forms are instruments used to convey meaningful elements. This is the basis of European structuralism. 2. The formalist’s view: linguistic forms are abstract structures which can be filled with meaningful elements. This is the basis of generative grammar. 3. The parasitologist’s view: linguistic forms are vehicles for the reproduction of meaningful elements. This is the view which I advocated twelve years ago in a Festschrift (1985).
On Russenorsk
(2002)
The concept of mixed language has recently gained some popularity, to my mind for no good reason. It is unclear how a mixed language can be distinguished from the product of extensive borrowing or relexification. I therefore think that the concept only serves to provoke muddled thinking about linguistic contact and language change. Note e.g. that Munske adduces German as an example "because the author is a professor of German linguistics and because the phenomenon of language mixing can be explained better in relation to a language on which a large amount of research has been done than, for example, in relation to pidgin and creole languages" (1986: 81).
The origin of the Goths
(2004)
Witold Ma´nczak has argued that Gothic is closer to Upper German than to Middle German, closer to High German than to Low German, closer to German than to Scandinavian, closer to Danish than to Swedish, and that the original homeland of the Goths must therefore be located in the southernmost part of the Germanic territories, not in Scandinavia (1982, 1984, 1987a, 1987b, 1992). I think that his argument is correct and that it is time to abandon Iordanes’ classic view that the Goths came from Scandinavia. We must therefore reconsider the grounds for adopting the latter position and the reasons why it always has remained popular.
A correct evaluation of the Slavic evidence for the reconstruction of the Indo- European proto-language requires an extensive knowledge of a considerable body of data. While the segmental features of the Slavic material are generally of corroborative value only, the prosodic evidence is crucial for the reconstruction of PIE. phonology. Due to the complicated nature of Slavic historical accentology, this has come to be realized quite recently.1 As a result, much of the earlier literature has become obsolete to the extent that it is based upon an interpretation which does not take the multifarious accentual developments into account. I shall give one example.
The genetic code, the primary manifestation of life, and, on the other hand, language, the universal endowment of humanity and its momentous leap from genetics to civilization, are the two fundamental stores of information transmissible from the ancestry to the progeny, the molecular succession, which ensures the transfer of hereditary messages from the cells of one generation to the next generation, and the verbal legacy as a necessary prerequisite of cultural tradition. Divergent terminologies direct attention to different pattemings; and finding a logically convincing test, acceptable all around, that can determine whether one such system of terms is superior to its rivals, is often impossible. Yet the slow processes of evolution presumably apply to human societies and their symbolic systems as much as to human bodies, so that when logic cannot decide, survival eventually will.
A correct interpretation of the genitive plural forms in Slavic and related languages requires a detailed chronological analysis of the material. At every stage of development we have to reckon with both phonetically regular and analogical forms. Analogy operates quite often along the same lines in different periods. Explaining an analogic change amounts to indicating a model, a motivation, and a stage of development for its effectuation. If one of these cannot be indicated, we must look for a phonetic explanation.
Indo-European is a branch of Indo-Uralic which was radically transformed under the influence of a North Caucasian substratum when its speakers moved from the area north of the Caspian Sea to the area north of the Black Sea (cf. Kortlandt 2007b). As a result, Indo-European developed a minimal vowel system combined with a very large consonant inventory including glottalized stops, also grammatical gender and adjectival agreement, an ergative construction which was lost again but has left its traces in the grammatical system, especially in the nominal inflection, a construction with a dative subject which was partly preserved in the historical languages and is reflected in the verbal morphology and syntax, where it gave rise to new categories, and a heterogeneous lexicon. The Indo-Uralic elements of Indo-European include pronouns, case endings, verbal endings, participles and derivational suffixes. In the following I shall give an overview of the grammar of Proto-Indo-European as it may have been spoken around 4000 BC in the eastern Ukraine, shortly after the ancestors of the Anatolians left for the Balkans (for more recent developments I refer to Beekes 1995).
In his magnum opus (Syntax and Semantics, Leiden 1978, henceforth: S&S) C.L. Ebeling makes a distinction between temporal gradation (pp 301-308 and 337-339) and temporal limitation (pp 311-315). In the case of temporal gradation “p , q”, the meaning “q” specifies the time during which the referent carries the mean-ing “p”.
The Indo-Uralic verb
(2002)
C.C. Uhlenbeck made a distinction between two components of Proto-Indo-European, which he called A and B (1935a: 133ff.). The first component comprises pronouns, verbal roots, and derivational suffixes, and may be compared with Uralic, whereas the second component contains isolated words, such as numerals and most underived nouns, which have a different source. The wide attestation of the Indo-European numerals must be attributed to the development of trade resulting from the increased mobility which was the primary cause of the Indo-European expansions. Numerals do not belong to the basic vocabulary of a neolithic culture, as is clear from their absence in Proto-Uralic (cf. also Collinder 1965: 112) and from the spread of Chinese numerals throughout East Asia. Though Uhlenbeck objects to the term “substratum” for his B complex, I think that it is a perfectly appropriate denomination.
In his magnificent book on the language relations across Bering Strait (1998), Michael Fortescue does not consider Nivkh (Gilyak) to be a Uralo-Siberian language. Elsewhere I have argued that the Indo-European verbal system can be understood in terms of its Indo-Uralic origins (2001). All of these languages belong to Joseph Greenberg’s Eurasiatic macro-family (2000). In the following I intend to reconsider the grammatical evidence for including Nivkh into the Uralo-Siberian language family. The Indo-Uralic evidence is of particular importance because it guarantees a time depth which cannot otherwise be attained.
Twenty years ago (1983), I severely criticized Halle and Kiparsky’s review (1981) of Garde’s history of Slavic accentuation (1976). I concluded that Halle and Ki-parsky’s theoretical framework “rests upon an unwarranted limitation of the available evidence, obscures the chronological perspective, and yields results which are partly not new and partly incorrect. It is harmful because it does not give the facts their proper due and thereby blocks the road to empirical study, giving a free hand to unrestrained speculation” (1983: 40). As Halle has recently returned to the subject (2001), it may be interesting to see if there has been some progress in his thinking over the last two decades. In the following I shall try to avoid repeating what I have said in my earlier discussion.
Koivulehto and Vennemann have recently (1996) revived Posti’s theory (1953) which attributed Finnic consonant gradation to Germanic influence, in particular to the influence of Verner’s law. This theory disregards the major differences between Finnic and Saami gradation (cf. Sammallahti 1998: 3) and ignores the similar gradation in Nganasan and Selkup (cf. Kallio 2000: 92).
The history of Slavic accentuation is complex. As a result, the significance of the Slavic accentual evidence is not immediately obvious to the average Indo-Europeanist. In this contribution I intend to render the material more easily accessible to the non-specialist. I shall focus on the Serbo-Croatian dialectal area, where the Proto-Slavic accentual system is better preserved than elsewhere. The main point of reference will be the neo-Štokavian system which was codified in the 19th century as a basis for the standard languages.
Holger Pedersen’s "Études lituaniennes" reflects the issues under discussion at the time of its publication (1933). Its five unequal chapters deal with the following topics: I. The Lithuanian future and its Indo-European origins: the sigmatic formation, the 3rd person zero ending, the short root vowels e and a, the shortening and metatony in the 3rd person, and the future participle. II. The accentuation of nouns in Lithuanian: accentual mobility in the Indo- European consonant stems and its absence in the o-stems, the origins of accentual mobility in Lithuanian nominal paradigms, the accentuation of separate case forms, and accentual peculiarities of the adjective. III. The acute tone of the root in consonant stems. IV. The past active participle. V. Secondary vocalic alternations: new vowel length and new acute tone.
The highly successful conference on Balto-Slavic accentology organized by Mate Kapovic and Ranko Matasovic has given much food for thought. It has clarified the extent of fundamental disagreements as well as established areas of common interest where the evidence seems to be ambiguous. In the following I shall comment upon some of the papers presented at the conference which are directly relevant to my own research.
Since 1973 I have been advocating the view that the Balto-Slavic acute tone was in fact glottalic and has been preserved unchanged in originally stressed and unstressed syllables in Žemaitian and Latvian, respectively (e.g. 1975, 1977, 1985, 1998). Jay Jasanoff has now (2004) adopted the gist of my view, but with-out mentioning my name. It may therefore be useful to sketch the background of our differences and to point out the remaining discrepancies.
Hittite ammuk 'me'
(2005)
In the Indo-European department of Leiden University, Alwin Kloekhorst has initiated a discussion on Hittite ammuk ‘me’. The central question is: where did the geminate come from? This has led me to reconsider the origin of the Indo-European personal pronouns against the background of my reconstruction of Indo-Uralic (2002: 221-225). For the historical data I may refer to Schmidt (1978).
In an earlier study (1983) I argued that unlike aorists and athematic presents, Indo-European perfects and thematic presents originally had a dative subject, as in German mir träumt ‘me dreams’ for ich träume ‘I dream’, e.g. Greek oida ‘I know’ < ‘it is known to me’, édomai ‘I will eat’ < ‘it is eatable to me’. On the basis of Oettinger’s epoch-making book (1979), I proposed that the Hittite hi-flexion originated from a merger of the perfect, where *-i was added to 3rd sg. *-e in order to supply a new present, with the thematic flexion of causatives and iteratives, where the final *-e of 3rd sg. *-eie was dropped before the loss of intervocalic *-i- (1983: 315).
In his early years, C. C. Uhlenbeck was particularly interested in the problem of the Indo-European homeland (1895, 1897). He rejected Herman Hirt’s theory (1892) that the words for ‘birch’, ‘willow’, ‘spruce’, ‘oak’, ‘beech’ and ‘eel’ point to Lithuania and its immediate surroundings and returned to Otto Schrader’s view (1883, 1890) that the original homeland must rather be sought in southern Russia and may have included some of the later Germanic and Iranian territories. It is clear that the Mediterranean region and the area around the North Sea can safely be excluded because the arrival of the Indo-Europeans was comparatively recent here, as it was in Iran and the Indian subcontinent. It is difficult to be more specific within the limits of central and eastern Europe and central Asia. Uhlenbeck was impressed by the lexical correspondences between Indo-European and Semitic which had been adduced in favor of an eastern homeland but pointed out that borrowings from Semitic may have reached the Indo-Europeans through an intermediary. He agrees that the Indo-European words for trees and animals point to a moderate climate but questions the possibility of a more specific localization as well as the concept of homeland itself.
Like its predecessor in Zagreb, the conference on Balto-Slavic accentology in Copenhagen was a great success. The enthusiasm of the organizers Adam Hyllested and Thomas Olander proved highly effective in stimulating discussion among the participants. While in Zagreb most papers dealt with Slavic data, in Copenhagen the emphasis was on Balto-Slavic problems.
Winter's law again
(2007)
Since I discussed the scholarly literature on Winter’s law twenty years ago (1988), several important articles on the subject have appeared (Young 1990, Campanile 1994, Matasovic 1995, Derksen 2002, Dybo 2002, Patri 2005, Derksen 2007). As the law evidently continues to be controversial, it is important to look into the nature of the evidence and counter-evidence which is adduced. It appears that doubts about Winter’s law are largely the result of four types of misunderstanding.
Last year Georg Holzer proposed a relative chronology of accentual developments in Slavic (2005). Here I shall compare his chronology with the one I put forward earlier (1975, 1989a, 2003) and discuss the differences. For the sake of convenience, I first reproduce the relevant parts of my chronology, omitting asterisks before pre-historic Slavic forms. 1. Proto-Indo-European. 2. Dialectal Indo-European. 3. Early Balto-Slavic. During this period the characteristic lateral mobility of Balto-Slavic accent patterns came into existence. 4. Late Balto-Slavic. During this period the Balto-Slavic accent patterns obtained their final shape.
Erdvilas Jakulis’ thorough, detailed and comprehensive study (2004) is an important contribution to our reconstruction of the Balto-Slavic verbal system. The following remarks are intended to complement his findings from a Slavic perspective. Jakulis demonstrates that the type of Lith. tekèti, teka ‘flow’ is largely of East Baltic provenance. He finds it difficult to identify the same type in Old Prussian.
It is gratifying to see that Jay Jasanoff has now (2004) adopted my theory that "the Balto-Slavic acute was a kind of stød or broken tone" (p. 172), which I have been advocating since 1973. Unfortunately, his acceptance of my view is not based on an evaluation of the comparative evidence (for which see Kortlandt 1985a) but on his desire to derive Balto-Slavic “acute” and "circumflex" syllables from the "bimoric" and "trimoric" long vowels which he assumes for Proto-Germanic as the reflexes of the Indo-European "acute" and "circumflex" tones of the neogrammarians. Since the original "circumflex" was limited to Indo-European VHV-sequences, Jasanoff proposes a whole series of additional lengthenings yielding "hyperlong" vowels in Germanic, Baltic and Slavic, which still do not suffice to eliminate the counter-evidence (cf. Kortlandt 2004b: 14). The reason for this failure is his unwillingness to recognize that lengthened grade vowels are circumflex in Balto-Slavic (cf. Kortlandt 1997a).
After the very well-organized Leiden conference for which we must be grateful to Tijmen Pronk, it seems appropriate for me to review some of the papers, as I did after the previous conferences in Zagreb and Copenhagen. The aim of this review is merely to point out some of the differences of opinion which require further debate.
The loss and restoration of the phoneme /j/ plays a major role in the development of Proto-Slavic. After vowel contraction in posttonic syllables, Dybo’s law, and the rise of new /j/ in east, South, and West Slavic, contracted and uncontracted forms may have coexisted during a considerable period of time. After Dybo’s law we have *voļȃ < *vòlja "will" but *rolьjà < *orlь̀ja "plowland", after contraction *roļá in Slovincian rolåu, Old Polish rolå. The loss of distinctive tone yielded merger of the two paradigms, as a result of which most nouns of the former type adopted the accentuation of the latter. Slavic deverbal ja-stems are original proterodynamic ī/jē-stems. The proterodynamic nouns *dūšà (c) "soul" and *zorjà (c) "dawn" have probably preserved the original accentuation. The other proterodynamic jā-stems evidently adopted the accent pattern of the deverbal ā-stems.
Keith Langston disagrees with my account of the Slovene neo-circumflex. He rejects compensatory lengthening as an explanation of the neo-circumflex, primarily on theoretical grounds. His "moraic analysis" is quite unacceptable to me because it starts from an a priori segmentation of the speech flow. In a strict autosegmental approach, the segmentation of the speech flow should be part of the analysis and not be given a priori. Langston's rejection of van Wijk's law, according to which the simplification of certain consonant clusters yielded lengthening of the following vowel, is based on a misguided theoretical interpretation which led him astray.
In a recent article of major importance (2013), Tijmen Pronk has treated the accentuation of l-participles of the type neslъ in western South Slavic. Pronk points out correctly that Dybo’s law did not shift the accent onto final jers, e.g. in *kòņь, *bòbъ, and that the short vowel was preserved in Slovak osem < *òsmь, oheň < *ògņь, mohol < *mòglъ. Contrary to what Pronk claims, Slovene nę́sǝlis the phonetic reflex of *néslъ < *neslъ̀, Slovak niesol. The Slovene doublets (v)ǫ̑gǝl < *ǫ̀glь and (v)ózǝl < *ǫ̀zlъ suggest an earlier paradigm with vǫ̑- < ǫ̑- in the nom.sg. form and ó- < *ǫ̀- in the oblique cases. The vowel of ógǝnj < *ògņь also stems from the oblique cases. The expected neo-circumflex in the nom.sg. form is actually attested in rę̑bǝr < *rèbrь beside rę́bǝr with the reflex of Stang’s law from the oblique cases. There is no reason to assume that the accent was not retracted at an early stage in *neslъ̀, nor is there any reason to assume that Dybo’s law shifted the accent to the final jer in *dòbrъ and *sèdmь, as Pronk claims.
Does it pay to invest in art? A selection-corrected returns perspective : [draft october 15, 2013]
(2013)
This paper shows the importance of correcting for sample selection when investing in illiquid assets with endogenous trading. Using a large sample of 20,538 paintings that were sold repeatedly at auction between 1972 and 2010, we find that paintings with higher price appreciation are more likely to trade. This strongly biases estimates of returns. The selection-corrected average annual index return is 6.5 percent, down from 10 percent for traditional uncorrected repeat sales regressions, and Sharpe Ratios drop from 0.24 to 0.04. From a pure financial perspective, passive index investing in paintings is not a viable investment strategy once selection bias is accounted for. Our results have important implications for other illiquid asset classes that trade endogenously.
Physical exercise has been shown to alter sensory functions, such as sensory detection or perceived pain. However, most contributing studies rely on the assessment of single thresholds, and a systematic testing of the sensory system is missing. This randomised, controlled cross-over study aims to determine the sensory phenotype of healthy young participants and to assess if sub-maximal endurance exercise can impact it. We investigated the effects of a single bout of sub-maximal running exercise (30 min at 80% heart rate reserve) compared to a resting control in 20 healthy participants. The sensory profile was assessed applying quantitative sensory testing (QST) according to the protocol of the German Research Network on Neuropathic Pain. QST comprises a broad spectrum of thermal and mechanical detection and pain thresholds. It was applied to the forehead of study participants prior and immediately after the intervention. Time between cross-over sessions was one week. Sub-maximal endurance exercise did not significantly alter thermal or mechanical sensory function (time × group analysis) in terms of detection and pain thresholds. The sensory phenotypes did not indicate any clinically meaningful deviation of sensory function. The alteration of sensory thresholds needs to be carefully interpreted, and only systematic testing allows an improved understanding of mechanism. In this context, sub-maximal endurance exercise is not followed by a change of thermal and mechanical sensory function at the forehead in healthy volunteers.
After our taxonomic revision of Ootheca Chevrolat, 1837, and the description of Oothecoides Kortenhaus & Wagner, 2011 and Ootibia Kortenhaus & Wagner, 2012, it became clear that a further four galerucine species, closely related to the above named taxa, form a distinct monophyletic group, that constitutes a new genus, Oosagitta gen. nov. with O. anningae sp. nov., O. geescheae sp. nov., O. melanopicta sp. nov. and O. thomasi sp. nov.. Exosoma angolensis Laboissière, 1939, the type species of the new genus, and Ergana minuta Laboissière, 1937 are newly transferred to Oosagitta gen. nov. All species of Oosagitta gen. nov. are characterized by a broad body and pronotum, a more or less convex dorsum and short legs, and as such are most similar to the other above named genera. The antennae of Oosagitta gen. nov. are distinctly longer than those of Ootheca, Oothecoides and Ootibia. Genital structures of the males allow a reliable identifi cation of the genus. (Re-) descriptions are given for all species, including semi-schematic illustrations depicting the habitus outline, shape of the basal antennomeres and the median lobe. Photographs of the name-bearing types and distribution maps are provided.
This dissertation explores the breadth and variation of authoritarian counter-terrorism strategies and their legitimacy-related origins to challenge prevailing assumptions in Terrorism Studies. Research and analysis are conducted in the form of a Structured Focused Comparison of domestic counter-terrorism strategies in two electoral autocracies. The first case is Russia’s domestic engagement against a mix of ethno-separatist and Islamist terrorism emanating from its North Caucasus republics between 1999 and 2018. The second case is China’s engagement vis-à-vis a similar type of terrorism in its Xinjiang Uyghur Autonomous Region between 1990 and 2018.
The comparison shows that, contrary to prevailing assumptions, the two strategies differ immensely from one another while containing significant if not predominant non-coercive elements. It further shows that the two strategies are closely related to the two states’ sources and resources of legitimacy, both in their original motivation to tackle the terrorist threat and in the design of counter-terrorism strategies. Drawing on David Beetham’s theory of The Legitimation of Power and on the Comparative Politics, Terrorism Studies and Civil War literatures, the dissertation explores the influence of five sources and (re)sources of legitimacy on the two counter-terrorism strategies: responsiveness, performance legitimacy, ideology, discursive power and co-optation. While governmental discursive power is discarded as a source of variation, findings are significant with respect to the influence of ideology and performance legitimacy. Reliance on ideology or related patterns for legitimation raise vulnerability to terrorism and constrain or facilitate the adoption of communicative and preventive measures that accommodate the grievances of potentially defective or even violently terrorist groups. Performance legitimacy is a key motivator in counter-terrorism and an influence on certain types of counter-terrorism policies. Responsiveness and co-optation are identified as potential sources of variation, based on idiosyncratic concurrence with policy choices.
Species commonly assigned to the cheilostome bryozoan genus Onychocella Jullien, 1882 are numerous in deposits of Late Cretaceous age. Among these are 15 species with wide stratigraphical and geographical distributions that are better placed in the genus Rhagasostoma Koschinsky, 1885. These are used here to show similarities between Late Cretaceous bryozoan associations from Western Europe and Central Asia. Type and additional material was examined of several species from the Turonian to the Maastrichtian of Western Europe, including material studied by R.M. Brydone, E. Voigt and T.A. Favorskaya and undescribed material from the Campanian and Maastrichtian of several localities in Eastern Europe and Central Asia. The new species Rhagasostoma brydonei sp. nov., R. aralense sp. nov. and R. operculatum sp. nov. are introduced. New and published data on the morphology and the stratigraphical and geographical distributions of R. inelegans (Lonsdale, 1850), R. gibbosum (Marsson, 1887), R. gibbosulum Brydone, 1936, R. rowei (Brydone, 1906) and R. mimosa (Brydone, 1930) is presented.
Bipolar disorder (BD) is a heritable mental illness with complex etiology. While the largest published genome-wide association study identified 64 BD risk loci, the causal SNPs and genes within these loci remain unknown. We applied a suite of statistical and functional fine-mapping methods to these loci, and prioritized 22 likely causal SNPs for BD. We mapped these SNPs to genes, and investigated their likely functional consequences by integrating variant annotations, brain cell-type epigenomic annotations, brain quantitative trait loci, and results from rare variant exome sequencing in BD. Convergent lines of evidence supported the roles of SCN2A, TRANK1, DCLK3, INSYN2B, SYNE1, THSD7A, CACNA1B, TUBBP5, PLCB3, PRDX5, KCNK4, AP001453.3, TRPT1, FKBP2, DNAJC4, RASGRP1, FURIN, FES, YWHAE, DPH1, GSDMB, MED24, THRA, EEF1A2, and KCNQ2 in BD. These represent promising candidates for functional experiments to understand biological mechanisms and therapeutic potential. Additionally, we demonstrated that fine-mapping effect sizes can improve performance and transferability of BD polygenic risk scores across ancestrally diverse populations, and present a high-throughput fine-mapping pipeline (https://github.com/mkoromina/SAFFARI).
Bacterial and fungal toll-like receptor activation elicits type I IFN responses in mast cells
(2021)
Next to their role in IgE-mediated allergic diseases and in promoting inflammation, mast cells also have antiinflammatory functions. They release pro- as well as antiinflammatory mediators, depending on the biological setting. Here we aimed to better understand the role of mast cells during the resolution phase of a local inflammation induced with the Toll-like receptor (TLR)-2 agonist zymosan. Multiple sequential immunohistology combined with a statistical neighborhood analysis showed that mast cells are located in a predominantly antiinflammatory microenvironment during resolution of inflammation and that mast cell-deficiency causes decreased efferocytosis in the resolution phase. Accordingly, FACS analysis showed decreased phagocytosis of zymosan and neutrophils by macrophages in mast cell-deficient mice. mRNA sequencing using zymosan-induced bone marrow-derived mast cells (BMMC) revealed a strong type I interferon (IFN) response, which is known to enhance phagocytosis by macrophages. Both, zymosan and lipopolysaccharides (LPS) induced IFN-β synthesis in BMMCs in similar amounts as in bone marrow derived macrophages. IFN-β was expressed by mast cells in paws from naïve mice and during zymosan-induced inflammation. As described for macrophages the release of type I IFNs from mast cells depended on TLR internalization and endosome acidification. In conclusion, mast cells are able to produce several mediators including IFN-β, which are alone or in combination with each other able to regulate the phagocytotic activity of macrophages during resolution of inflammation.
Mast cells are long-lived tissue-resident leukocytes, located most abundantly in the skin and mucosal surfaces. They belong to the first line of defence of the body, protecting against invading pathogens, toxins and allergens. Their secretory granules are densely packed with a plethora of mediators, which can be released immediately upon activation of the cell. Next to their role in IgE-mediated allergic diseases and in promoting inflammation, potential anti-inflammatory functions have been assigned to mast cells, depending on the biological setting. The aim of this thesis was to contribute to a better understanding of the role of mast cells during the resolution of a local inflammation. Therefore, in a first of step a suitable model of a local inflammation had to be identified. Since comparison of the two Toll-like receptor (TLR)-agonists zymosan and lipopolysaccharide (LPS), which are most commonly used to locally induce inflammation, revealed a systemic response after LPS-injection and a local inflammation after zymosan-injection, the TLR2 agonist zymosan was chosen for the subsequent experiments. Multi epitope ligand cartography (MELC) combined with statistical neighbourhood analysis showed that mast cells are located in an anti-inflammatory microenvironment next to M2 macrophages during resolution of inflammation, while neutrophils and M1 macrophages are located in the zymosan-filled core of the inflammation. Furthermore, infiltrating neutrophils during peak inflammation and an increasing population of macrophages phagocytosing neutrophils during resolution of inflammation could be observed. MELC as well as flow cytometry analysis of mast cell-deficient mice revealed a decreased phagocytosing activity of macrophages in the absence of mast cells. As an untargeted approach to identify mast cell-derived mediators induced by zymosan, mRNA sequencing of bone marrow-derived mast cells (BMMCs) was performed. Gene ontology term analysis of the sequencing data revealed the induction of the type I interferon (IFN) pathway as the dominant response. Contradicting previous studies, I could validate the production of IFN-β by mast cells in response to zymosan and LPS in vitro. Furthermore IFN-β expression by mast cells was also detected in vivo. In accordance with previous studies regarding other cell types the release of IFN-β by mast cells depends on endosomal signaling. The potential of IFN-β to enhance the phagocytosing activity of macrophages has been demonstrated recently. Besides IFN-β, various other mediators with reported enhancing effects on macrophage phagocytosis were also induced by zymosan in BMMCs, including Interleukin (IL)-1β, IL-4, IL-13, and Prostaglandin (PG) E2. Thus, either one of these mediators alone or a combination of them could promote macrophage phagocytosis.
In conclusion, I herein present mast cells as a novel source for IFN-β induced by non-viral TLR ligands and demonstrate their enhancing effect on macrophage phagocytosis, thereby contributing to the resolution of inflammation.
einfaches lineares Modell inklusive Residuenanalyse
Anteil Diesjähriger in Invasionsjahren, während beim Erlenzeisig
Massenzug vorhergehenden Brutzeit vermutlich gute Nahrungsbedingungen
Fast alle Fundorte liegen in Rheinland-Pfalz, zwei in Hessen.
Device-related infections in recipients of left ventricular assist devices (LVAD) have been recognized as a major source of morbidity and mortality. They require a high level of diagnostic effort as part of the overall burden resulting from infectious complications in LVAD recipients. We present a multi-allergic patient who was treated for persistent sterile intrathoracic abscess formation and pericardial empyema following minimally invasive LVAD implantation including use of a sheet of e-polytetrafluoroethylene (ePTFE) membrane to restore pericardial integrity. Sterile abscess formation and pericardial empyema recurred after surgical removal until the ePTFE membrane was removed, suggesting that in disposed patients, ePTFE may be related to sterile abscess formation or sterile empyema.
Left ventricular non-compaction cardiomyopathy and left ventricular assist device: a word of caution
(2016)
BACKGROUND: In patients with left ventricular non-compaction (LVNC), implantation of a left ventricular assist device (LVAD) may be performed as a bridge to transplantation. In this respect, the particular characteristics of the left ventricular myocardium may represent a challenge.
CASE PRESENTATION: We report a patient with LVNC who required urgent heart transplantation for inflow cannula obstruction nine months after receiving a LVAD. LVAD parameters, echocardiography and examination of the explanted heart suggested changes of left ventricular configuration brought about by LVAD support as the most likely cause of inflow cannula obstruction.
CONCLUSIONS: We conclude that changes experienced by non-compacted myocardium during LVAD support may give rise to inflow cannula obstruction and flow reduction. Presence of LVNC mandates tight surveillance for changes in LV configuration and LVAD flow characteristics and may justify urgent transplantation listing status.
Background: Infection is a main cause of morbidity and mortality after heart surgery, with multi-resistant pathogens increasingly representing a challenge. Daptomycin provides bactericidal activity against gram-positive organisms that are resistant to standard treatment including vancomycin.
Methods: A cohort of cardiac surgical patients, treated with daptomycin for major infection at two tertiary care centers, were retrospectively studied with a particular focus on the type of infection, causative pathogens and co-infections, daptomycin dosage, adverse events and outcome in order to provide evidence for the efficiency and safety of daptomycin in a distinct high-risk patient population.
Results: Sixty-five patients (87.7 % males, 60.4 ± 13.5 years) who had undergone aortic surgery (20.0 %), ventricular assist device (VAD) implantation (21.5 %), combined procedures (21.5 %), coronary artery bypass grafting (12.3 %), isolated valve surgery (15.4 %) and heart transplantation (7.7 %) were diagnosed with catheter-related infection (26.1 %), valve endocarditis (18.8 %), sternal wound (13.0 %), VAD-associated (11.6 %), cardiac implantable electrophysiological device (CIED)-associated (4.1 %), respiratory tract (4.3 %), bloodstream (4.3 %) and other infection (4.3 %). In 13.0 %, no focus of infection was identified though symptoms of severe infection were present. The most frequent pathogens were Staphylococcus epidermidis (30.4 %), Staphylococcus aureus (23.1 %) and Enterococcus species (10.1 %). Daptomycin doses ranging from 3 mg/kg every 48 h to 10 mg/kg every 24 h were administered for 15.4 ± 11.8 days. 87.0 % of the cases were classified as success, 7.2 % as treatment failure and 5.8 as non-evaluable. Adverse events were limited to one case of mild and one case of moderate neutropenia with recovery upon termination of treatment.
Conclusion: Daptomycin proved safe and effective in major infection in high-risk cardiac surgical patients.
The family of scaffold attachment factor B (SAFB) proteins comprises three members and was first identified as binders of the nuclear matrix/scaffold. Over the past two decades, SAFBs were shown to act in DNA repair, mRNA/(l)ncRNA processing, and as part of protein complexes with chromatin-modifying enzymes. SAFB proteins are approximately-100-kDa-sized dual nucleic acid-binding proteins with dedicated domains in an otherwise largely unstructured context, but whether and how they discriminate DNA- and RNA-binding has remained enigmatic. We here provide the SAFB2 DNA- and RNA-binding SAP and RRM domains in their functional boundaries and use solution NMR spectroscopy to ascribe DNA- and RNA-binding functions. We give insight into their target nucleic acid preferences and map the interfaces with respective nucleic acids on sparse data-derived SAP and RRM domain structures. Further, we provide evidence that the SAP domain exhibits intra-domain dynamics and a potential tendency to dimerise, which may expand its specifically targeted DNA sequence range. Our data provide a first molecular basis of and a starting point towards deciphering DNA- and RNA-binding functions of SAFB2 on the molecular level and serve a basis for understanding its localization to specific regions of chromatin and its involvement in the processing of specific RNA species.

The current outbreak of the highly infectious COVID-19 respiratory disease is caused by the novel coronavirus SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2). To fight the pandemic, the search for promising viral drug targets has become a cross-border common goal of the international biomedical research community. Within the international Covid19-NMR consortium, scientists support drug development against SARS-CoV-2 by providing publicly available NMR data on viral proteins and RNAs. The coronavirus nucleocapsid protein (N protein) is an RNA-binding protein involved in viral transcription and replication. Its primary function is the packaging of the viral RNA genome. The highly conserved architecture of the coronavirus N protein consists of an N-terminal RNA-binding domain (NTD), followed by an intrinsically disordered Serine/Arginine (SR)-rich linker and a C-terminal dimerization domain (CTD). Besides its involvement in oligomerization, the CTD of the N protein (N-CTD) is also able to bind to nucleic acids by itself, independent of the NTD. Here, we report the near-complete NMR backbone chemical shift assignments of the SARS-CoV-2 N-CTD to provide the basis for downstream applications, in particular site-resolved drug binding studies.
The ongoing pandemic caused by the Betacoronavirus SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus-2) demonstrates the urgent need of coordinated and rapid research towards inhibitors of the COVID-19 lung disease. The covid19-nmr consortium seeks to support drug development by providing publicly accessible NMR data on the viral RNA elements and proteins. The SARS-CoV-2 genome encodes for approximately 30 proteins, among them are the 16 so-called non-structural proteins (Nsps) of the replication/transcription complex. The 217-kDa large Nsp3 spans one polypeptide chain, but comprises multiple independent, yet functionally related domains including the viral papain-like protease. The Nsp3e sub-moiety contains a putative nucleic acid-binding domain (NAB) with so far unknown function and consensus target sequences, which are conceived to be both viral and host RNAs and DNAs, as well as protein-protein interactions. Its NMR-suitable size renders it an attractive object to study, both for understanding the SARS-CoV-2 architecture and drugability besides the classical virus’ proteases. We here report the near-complete NMR backbone chemical shifts of the putative Nsp3e NAB that reveal the secondary structure and compactness of the domain, and provide a basis for NMR-based investigations towards understanding and interfering with RNA- and small-molecule-binding by Nsp3e.
The early Tournaisian (Early Carboniferous; Mississippian) ammonoids from the classical abandoned limestone quarry of Gattendorf (Upper Franconia) are revised, using the historical collections as well as so far undescribed material. The ammonoid assemblage is composed of prionoceratid ammonoids of the six genera Mimimitoceras, Paragattendorfia, Stockumites, Acutimitoceras, Gattendorfia and Gattenpleura, which indicate a stratigraphic position near the Devonian–Carboniferous boundary in the earliest Carboniferous. The new species Stockumites hofensis sp. nov. and S. nonaginta sp. nov. are described.
The railway cutting near Oberrödinghausen at the northern margin of the Rhenish Mountains is the cardinal section for the investigation of Early Tournaisian (Early Carboniferous; Mississippian) ammonoids. Early Carboniferous coiled nautiloids from North Africa are virtually unknown. An assemblage of nine species, all from the family Trigonoceratidae, from the Dalle à Merocanites (Tournaisian-Viséan boundary interval) of Timimoun in western Algeria is described, being the most diverse Carboniferous nautiloid assemblage known from North Africa but much less diverse than the time-equivalent assemblages from Belgium and Ireland. The assemblage consists of the species Maccoyoceras pentagonum sp. nov., Lispoceras orbis sp. nov., Thrincoceras devolvere sp. nov., Rineceras multituberculatum sp. nov., Rineceras rectangulatum sp. nov., Vestinautilus padus sp. nov., Vestinautilus concinnus sp. nov., Planetoceras destrictum sp. nov. and Planetoceras transforme sp. nov. A morphometric analysis of Maccoyoceras pentagonum sp. nov. and Lispoceras orbis sp. nov. shows that the intraspecific variation in these species is within rather narrow limits.
Viséan coiled nautiloids from North Africa are only poorly known. From the Mougoui Ayoun, Zrigat and Hamou-Rhanem formations of the eastern Anti-Atlas, we describe coiled nautiloids, which belong to the genera Rineceras, Stroboceras, Temnocheilus, Vestinautilus, Maccoyoceras, Endolobus, Epidomatoceras, Liroceras, Ephippioceras, and Solenochilus. The new species Temnocheilus imazighenorum sp. nov., Temnocheilus aubrechtovae sp. nov., Vestinautilus kesslerae sp. nov., Endolobus rota sp. nov., Epidomatoceras ebbighausenorum sp. nov., Liroceras vermis sp. nov., Liroceras karaouii sp. nov., Ephippioceras pygops sp. nov., Solenochilus lucynae sp. nov. and Solenochilus pohlei sp. nov. are described; six taxa are kept in open nomenclature. The assemblage is composed of the three superfamilies Trigonoceratoidea, Clydonautiloidea and Aipoceratoidea and shows a wide spectrum of conch morphologies, ranging from widely umbilicate compressed forms to involute compact forms, reflecting a broad ecological variation.

This synonymic list of the flat bugs (Aradidae) ofthe world enumerates 1,798 species in 211 genera. Names of eight fossil species are given in their original combination in modern genera. The list is introduced by brief discussions of habits, food, ecology and distribution. Previous studies document a relationship between gambling activity at the aggregate level and investments in securities with lottery-like features. We combine data on individual gambling consumption with portfolio holdings and trading records to examine whether gambling and trading act as substitutes or complements. We find that gamblers are more likely than the average investor to hold lottery stocks, but significantly less likely than active traders who do not gamble. Our results suggest that gambling behavior across domains is less relevant compared to other portfolio characteristics that predict investing in high-risk and high-skew securities, and that gambling on and off the stock market act as substitutes to satisfy the same need, e.g., sensation seeking.
Background: High-intensity focused ultrasound (HIFU) allows to inflict intracorporal thermal lesions without penetrating the skin or damaging the surrounding tissue. This analysis intends to assess the magnitude of HIFU-induced ablations within benign thyroid nodules using scintigraphic imaging with 99mTc.
Methods: Ten cold, hot, or indifferent nodules were treated using multiple pulses of HIFU to induce temperatures of around 85°C within the ablation zone. Pre- and posttreatment, uptake values of 99mTc pertechnetate or 99mTc-MIBI were recorded. The pre-post reduction of nodular uptake was evaluated to assess ablation magnitude.
Results: Relative nodular uptake in relation to total thyroidal uptake decreased after one session of HIFU in all cases. Median 99mTc-MIBI uptake reduction was 35.5% (ranging from 11% to 57%; p < 0.1), while 99mTc-pertechnetate scintigraphy showed a median uptake reduction of 27% (range 10% to 44%; p < 0.1). No major complications were observed.
Conclusions: HIFU appears to be safe and is an easy to perform means of thermal ablation. This study shows that HIFU treatment in thyroidal nodules can be evaluated by scintigraphic means shortly after the intervention. Due to small sample size, the exact magnitude of HIFU ablation efficiency in thyroidal nodules remains a value to beassessed in a larger study.
Background: Because Endomyocardial Biopsy has low sensitivity of about 20%, it can be performed near to myocardium that presented as Late Gadolinium Enhancement (LGE) in cardiovascular magnetic resonance (CMR). However the important issue of comparing topography of CMR and histological findings has not yet been investigated. Thus the current study was performed using an animal model of myocarditis. Results: In 10 male Lewis rats Experimental Autoimmune myocarditis was induced, 10 rats served as control. On day 21 animals were examined by CMR to compare topographic distribution of LGE to histological inflammation. Sensitivity, specificity, positive and negative predictive values for LGE in diagnosing myocarditis were determined for each segment of myocardium. Latter diagnostic values varied widely depending on topographic distribution of LGE and inflammation as well as on the used CMR sequence. Sensitivity of LGE was up to 76% (left lateral myocardium) and positive predictive values were up to 85% (left lateral myocardium), whereas sensitivity and positive predictive value dropped to 0 - 33% (left inferior myocardium). Conclusions: Topographic distribution of LGE and histological inflammation seem to influence sensitivity, specifity, positive and negative predictive values. Nevertheless, positive predictive value for LGE of up to 85% indicates that Endomyocardial Biopsy should be performed "MR-guided". LGE seems to have greater sensitivity than Endomyocardial Biopsy for the diagnosis of myocarditis.
This study addresses the structure-function relationships of three essential membrane proteins: Porin from Paracoccus denitrificans, Porin OmpG from Eschericia coli and BetP from Corynobacterium glutamicum using Fourier transform infrared (FT-IR) spectroscopy and Attenuated Total Reflection (ATR) techniques. The structure of porin from P. denitrificans is known for more than a decade; however, the mechanism for loss of functionality together with the monomerization was not clear. In this study we have addressed the role of lipids for the functionality of porin using FT-IR. OmpF porin was found to interact with the lipid molecules via the aromatic girdles surrounding the protein for functionality. In this study, molecular bonds and groups of the lipids were established as reporter groups probing at different depths of the bilayer in order to understand the interaction partner of the aromatic girdles of porins. Monomerization of the trimeric assembly of OmpF porin reconstituted in lipids is induced by increasing the temperature. Porin (OmpF) was found to be extremely stable: The secondary structure of the protein was unaltered up to the temperature-induced main transition, around 80-90 °C, above which it is denatured. However, the interaction of the aromatic girdle with the lipid molecules exhibited distinct changes at much lower temperature values (40 - 50°) where, according to the previous functional studies, monomerization and the loss of function occurs. The results are compared with OmpG porin from E.coli, for which the functional unit is a monomer. The aromatic girdle-lipid interaction was monitored by the tyrosine aromatic ring C=C vibrational mode, a universal marker for the protein stability and interaction. We have also found that the aromatic girdles of porins are interacting with the interfacial region of the lipid bilayer instead of lipid headgroups. Lipid-protein interaction was found to be not only essential for the structural stability, but also for the functionality of OmpF porin. We have also studied the structural properties of OmpG from E.coli. The structure of OmpG at two pH values has been resolved using X-ray crystallography and the channel has been proposed to attain different states at different pH values as closed (pH < 5.5) and open (pH >7.5). This study, using IR spectroscopy, revealed that the pH-induced opening and closing of the channel is reflected by the frequency shifts of the ? sheet structure. OmpG has more rigid ? barrel properties upon opening of the channel. IR spectral analysis revealed multiple ? sheet signals with different hydrogen bond strengths. This enabled us to monitor the formation of hydrogen bridges between the extracellular loops upon opening of the channel. The conclusion that OmpG porin having two states at different pH values was also confirmed by the three mutants where the role of the histidine pair (H231 & H261) and loop 6 has been addressed. Temperature-profiling of the wild type (WT) protein and the mutants did not show pH dependent structural stability differences in detergent solution. However, the WT protein was found to be more stable in the open form in 2D crystals than the closed form. Reconstitution into lipids has increased the transition temperature value by ~20 °C in the closed state and ~25 °C in the open state. Therefore we conclude that the open and closed state of OmpG has structural stability differences that are only revealed in the lipid environment. A comparison of the transition temperature values of OmpG WT and the mutants suggested that the hydrogen bond network among S218-H231-H261-D267, together with the formation of 12 residue-long ?-sheet contributes to the structural stability of the open channel. In the process of closing and opening of the channel, the globular structure of the protein remains mainly unchanged, while there are changes in the side chain moieties. In addition to the role of the histidine pair and the loop L6, in situ opening/closing experiments showed that the negatively charged amino acids, i.e. Asp and Glu, and Arg residues also play an active role; possibly by interacting with each other inside the pore lumen. Therefore it could be concluded that the closure of the channel at acidic pH values is not only via closing the channel entrance by loop 6, but also via changing the electric potential inside the lumen due to the different states of charged amino acids in order to effectively block the gateway. BetP from C.glutamicum attains an active and inactive state in order to adjust its glycine betaine uptake rate to the osmotic conditions that the cell encounters. The structure of BetP is not yet available. The WT protein exhibited structural differences in the presence of excess K+, which is one of the activation conditions. In 2D crystals, increasing the ionic strength to 700 mM K+ was shown to induce changes in the ?-helical moiety with contributions from the ester groups and one Tyr residue using ATR-FTIR. An increase in ionic strength to 220 mM K+ was found to be the threshold value of potassium concentration ([K+]) where the protein exhibits structural alterations in detergent solution. The determined [K+] values are in good agreement with the previous functional studies. However, there are differences in the activation profile of BetP in 2D crystals and in detergent solution, which points out that the lipids are involved in the conformational transition from the inactive to the active state and their absence can lead to different structural properties. BetP WT was found to have ~65% alpha-helix, ~25% random coil and ~10% turn structure in detergent solution. In the presence of excess K+, the WT protein is found to adapt more unordered structure. Secondary structure analysis of the mutants revealed that both the N- and C-terminus are in ?-helical conformation. Reconstitution of WT protein in 2D crystals increased the main transition (denaturation) temperature value from ~62 °C to ~85 °C, a clear indication that the protein is more stable in lipid environment. Temperature-profiling of the two forms of the WT protein revealed that the structural breakdown is preceeded by monomerization of the trimeric assembly. Comparing the two forms of the WT protein and the mutant BetA, we conclude that the oligomeric status is stabilized via the interactions among hydrophilic regions involving the N terminus. H/D exchange and activation with excess K+ in D2O-buffer revealed that activation of the protein involves the interaction of Arg and Asp/Glu residues in the cytoplasmic region of the protein. BetP WT and the two mutants tested, i.e. BetA and BetP?C45, showed differences in protein packing upon activation. The WT protein and BetP?C45 mutant also show changes in the hydrogen bonding properties of turns. Since BetA does not show such a property in activation, we conclude that the N-terminus interacts with the loops in the inactive state via the interaction of charged amino acids for the WT protein and that this interaction is altered during the activation. It could be argued that the protein packing is affected via the changes in turns upon activation. We also have found experimental evidence that one Tyr residue has different orientations in the active and inactive state of BetP. Based on the previous functional studies, it could be one of the five Tyr residues in the cytoplasmic region of the protein (in loop 3, 6, 7 or C-terminus). The mutant BetP?C45, on the other hand, showed fewer differences between the active and inactive state conditions and based on the H/D exchange rates, the mutant shows the properties of an active WT protein, proving that the C-terminal truncation impairs the conformational transition between the active and inactive states.
