In der vorliegenden klinischen Studie wurde der Zusammenhang zwischen Hämatokrit und Mikrozirkulation bei Diabetikern mit und ohne Mikroangiopathie untersucht und in Relation gesetzt zu anderen Risikofaktoren, für die bereits eine Beeinflussung der Mikrozirkulation bei Diabetikern nachgewiesen wurde. Die Untersuchung erfolgte mittels Laser Doppler Fluxmetrie, Laser Doppler Scan und Laser Doppler Anemometrie. Als weiteres Ziel sollten diese erprobten Methoden bezüglich der Aussagekraft für die Fragestellung unserer Studie eingeordnet werden. Es zeigte sich, dass der Hämatokrit signifikant negativ mit der Ruhedurchblutung (für Mean DC: p<0,01, β = -0,411) der Hand und des Fußes (für Mean Flux: p<0,001, β=-0,531; für Mean DC: p<0,05, β= -0,359) korreliert. Dieser Einfluss lässt sich mit dem Laser Doppler Scan am Besten nachweisen. Eine signifikant positive Korrelation (p<0,05, β= 0,284) des Hämatokrits mit der Dauer der Hyperämie gemessen an der Hand besteht in der Laser Doppler Fluxmetrie, jedoch nicht in der Anemometrie. Die These, dass ein höherer Hämatokrit zu einer schlechteren Mikrozirku-lation beiträgt, wird durch eine höchst signifikante negative Korrelation der Zeit bis zum Erreichen der Maximalgeschwindigkeit mit dem Hämatokrit (p<0,001, β= -0,497) in Frage gestellt. Ob dieses Ergebnis tatsächlich neue Erkenntnisse bringt oder durch Begleitumstände beeinflusst wurde, sollte in weiteren Untersuchungen geklärt werden. Auch im Vergleich mit anderen Risikofaktoren findet sich der starke Einfluss des Hämatokrit vor allem auf die Ruhedurchblutung. Für die Dauer der Hyperämie in der Single Point Messung zeigt sich ebenfalls ein großer Einfluss von Laktatdehydrogenase (LDH) und Body Mass Index (BMI) (LDH: β= -0,311, BMI: β= -0,310, p<0,05). Unsere Ergebnisse stärken die Vermutung, dass multifaktorielle Ursachen für Änderungen der Mikrozirkulation verantwortlich sind. Der Hämatokrit spielt hierbei eine wichtige Rolle, mit der man sich weiterhin befassen und in größer angelegten Studien unter Ausschluss von Beeinflussung durch andere Variablen noch genauer untersuchen sollte. Bezüglich der beiden Messverfahren Anemometrie und Laser Doppler Fluxmetrie bleibt festzuhalten, dass es sich um ergänzende Methoden handelt. Der Laser Doppler Image Scan ist als Standardmethode für die Erfassung der Ruhedurchblutung anzusehen, während die Fluxmetrie und die Anemometrie beide verlässlich mit Provokationstests anzuwenden sind. Sie messen in verschiedenen Ebenen des Kapillarsystems und produzieren dadurch unterschiedliche Ergebnisse. Es konnte in der vorliegenden Studie nachgewiesen werden, dass ein deutlicher Einfluss des Hämatokrits auf die Mikrozirkulation besteht. Um aus unseren Ergebnissen klinische Konsequenzen für die Behandlung von Diabetikern zu ziehen, muss man sich vor allem der Frage des Zusammenhanges auf Höhe des nutritiven Kapillarsystems ausführlicher widmen und die Ursache der Veränderungen der Time to Peak in weiteren Studien abklären.
In der Reihe "Bildbände zur Archäologie" des Philipp von Zabern Verlags erschien 2015 der Band "Antike Felsgräber" von Stephan Steingräber. Ziel des Autors ist es, die monumentalen Felsfassadengräber Etruriens in einem größeren Kontext darzustellen, was im Untertitel "unter besonderer Berücksichtigung der etruskischen Felsgräbernekropolen" zum Ausdruck kommt. Mit 162 überwiegend sehr qualitätvollen Abbildungen auf 144 Seiten fügt sich der Band in die bekannte Reihe, die archäologische Denkmäler für ein breites, Kultur interessiertes Publikum erschließt. Der Autor gehört zu den Kennern dieser etruskischen Gräber und zieht im Vorwort die Verbindungslinien zu seiner wissenschaftlichen Tätigkeit. In 14 Kapiteln spannt der Verfasser einen weiten Bogen (S. 7-86) mit Schlaglichtern auf Felsgräber und Felsarchitektur außerhalb Südetruriens; letzteren ist ein ausführliches Kapitel gewidmet (S. 87-128). Ein Anhang mit einer nützlichen Zusammenstellung der wichtigsten Informationen zu den südetruskischen Felsgräbernekropolen (S. 131-137) und einem Glossar beschließen den Bildband. Die Bibliographie spiegelt in ihrer Auswahl die Schwerpunktsetzung des Bandes. Nur der Titel selbst – auf dem Umschlag steht der Untertitel nicht – sowie das zentrale Titelbild der Felsgrabnekropole von Myra in Kleinasien stehen in eigenartigem Widerspruch zur eindeutig regionalen Konzeption. Um es gleich vorwegzunehmen, diese ambivalente Zielsetzung wirkt sich im Verlauf der Darlegungen ungünstig aus. ...
Due to the possession of huge contort strongyles, and a lack of triaenes in an otherwise 'astrophorine' spicule complement, the phylogenetic position of the endemic, monospecific New Zealand sponge genus, Lamellomorpha Bergquist, 1968, has remained enigmatic. The genus was established within Jaspidae de Laubenfels, 1968 (in the abandoned order Epipolasida Sollas, 1888), but it was not until 2002 that the genus was transferred formally to Astrophorina Sollas, 1887, albeit incertae sedis, by Hooper & Maldonado (2002). In this study, we recognise specimens of Lamellomorpha from the Subantarctic New Zealand region and Chatham Rise, considered by Bergquist to be conspecific with the type species, L. strongylata Bergquist, 1968, first described from the Three Kings-Spirits Bay region of Northland, as the new species, L. australis Kelly & Cárdenas sp. nov. These two species of Lamellomorpha have differences in external morphology and colour, skeletal architecture and spicules, natural products, geographical distribution, and depth ranges. Sequencing of the COI Folmer barcode/mini-barcode and of 28S (C1–C2 domains) of these two species suggests phylogenetic affinities of Lamellomorpha with the tetractinellid suborder Astrophorina and the family Vulcanellidae Cárdenas et al., 2011. Two Subantarctic New Zealand species of the vulcanellid genus Poecillastra Sollas, 1888, P. ducitriaena Kelly & Cárdenas sp. nov. and P. macquariensis Kelly & Cárdenas sp. nov., provide further support for the close relationship of Lamellomorpha and Poecillastra.
The Arp2/3 complex nucleates and cross-links actin filaments at the leading edge of motile cells, and its activity is stimulated by C-terminal regions of WASP/Scar proteins, called VCA domains. VCA domains contain a verprolin homology sequence (V) that binds monomeric actin and central (C) and acidic sequences (A) that bind the Arp2/3 complex. Here we show that the C domain binds to monomeric actin with higher affinity (K(d) = 10 microm) than to the Arp2/3 complex (K(d) > 200 microm). Nuclear magnetic resonance spectroscopy reveals that actin binds to the N-terminal half of the C domain and that both the V and C domains can bind actin independently and simultaneously, indicating that they interact with different sites. Mutation of conserved hydrophobic residues in the actin-binding interface of the C domain disrupts activation of the Arp2/3 complex but does not alter affinity for the complex. By chemical cross-linking the C domain interacts with the p40 subunit of the Arp2/3 complex and, by fluorescence polarization anisotropy, the binding of actin and the Arp2/3 complex are mutually exclusive. Our results indicate that both actin and Arp2/3 binding are important for C domain function but that the C domain does not form a static bridge between the two. We propose a model for activation of the Arp2/3 complex in which the C domain first primes the complex by inducing a necessary conformational change and then initiates nucleus assembly by bringing an actin monomer into proximity of the primed complex.

Containment problems belong to the classical problems of (convex) geometry. In the proper sense, a containment problem is the task to decide the set-theoretic inclusion of two given sets, which is hard from both the theoretical and the practical perspective. In a broader sense, this includes, e.g., radii or packing problems, which are even harder. For some classes of convex sets there has been strong interest in containment problems. This includes containment problems of polyhedra and balls, and containment of polyhedra, which have been studied in the late 20th century because of their inherent relevance in linear programming and combinatorics.
Since then, there has only been limited progress in understanding containment problems of that type. In recent years, containment problems for spectrahedra, which naturally generalize the class of polyhedra, have seen great interest. This interest is particularly driven by the intrinsic relevance of spectrahedra and their projections in polynomial optimization and convex algebraic geometry. Except for the treatment of special classes or situations, there has been no overall treatment of that kind of problems, though.
In this thesis, we provide a comprehensive treatment of containment problems concerning polyhedra, spectrahedra, and their projections from the viewpoint of low-degree semialgebraic problems and study algebraic certificates for containment. This leads to a new and systematic access to studying containment problems of (projections of) polyhedra and spectrahedra, and provides several new and partially unexpected results.
The main idea - which is meanwhile common in polynomial optimization, but whose understanding of the particular potential on low-degree geometric problems is still a major challenge - can be explained as follows. One point of view towards linear programming is as an application of Farkas' Lemma which characterizes the (non-)solvability of a system of linear inequalities. The affine form of Farkas' Lemma characterizes linear polynomials which are nonnegative on a given polyhedron. By omitting the linearity condition, one gets a polynomial nonnegativity question on a semialgebraic set, leading to so-called Positivstellensaetze (or, more precisely Nichtnegativstellensaetze). A Positivstellensatz provides a certificate for the positivity of a polynomial function in terms of a polynomial identity. As in the linear case, these Positivstellensaetze are the foundation of polynomial optimization and relaxation methods. The transition from positivity to nonnegativity is still a major challenge in real algebraic geometry and polynomial optimization.
With this in mind, several principal questions arise in the context of containment problems: Can the particular containment problem be formulated as a polynomial nonnegativity (or, feasibility) problem in a sophisticated way? If so, how are positivity and nonnegativity related to the containment question in the sense of their geometric meaning? Is there a sophisticated Positivstellensatz for the particular situation, yielding certificates for containment? Concerning the degree of the semialgebraic certificates, which degree is necessary, which degree is sufficient to decide containment?
Indeed, (almost) all containment problems studied in this thesis can be formulated as polynomial nonnegativity problems allowing the application of semialgebraic relaxations. Other than this general result, the answer to all the other questions (highly) depends on the specific containment problem, particularly with regard to its underlying geometry. An important point is whether the hierarchies coming from increasing the degree in the polynomial relaxations always decide containment in finitely many steps.
We focus on the containment problem of an H-polytope in a V-polytope and of a spectrahedron in a spectrahedron. Moreover, we address containment problems concerning projections of H-polyhedra and spectrahedra. This selection is justified by the fact that the mentioned containment problems are computationally hard and their geometry is not well understood.
