Jede erfolgreiche Software muss in einer geeigneten Art und Weise mit der Person, die sie benutzt, in Verbindung treten. Diese Schnittstelle zwischen Mensch und Maschine ist ein zentraler Baustein in der Softwareentwicklung. Eine noch so mächtige und ausgereifte Software kann ihr Potential nicht ausschöpfen, wenn Probleme und Missverständnisse bei der Kommunikation mit dem Anwender auftreten.
Bei graphischen Benutzeroberflächen erfolgt die Interaktion zwischen Benutzer und technischem System mittels graphischer Symbole, die am Bildschirm dargestellt werden. Die Oberfläche setzt sich aus verschiedenen Menüs und Steuerelementen mit dem Ziel zusammen, die zugrunde liegende Software für den Anwender bedienbar zu machen. Als Eingabegeräte dienen vor allem Maus und Tastatur. Für die Human Computer Interaction oder abgekürzt HCI (Mensch-Computer Interaktion) sind spezielle Normierungen und Anforderungen erstellt worden, die den Entwicklungsprozess unterstützten.
In dieser Arbeit wird eine graphische Benutzeroberfläche für einen Shader Viewer entworfen und implementiert. Beginnend bei ersten Skizzen und Prototypen wird der Entwicklungsprozess bis zur fertigen graphischen Oberfläche dargestellt. Probleme bei der Erstellung werden aufgezeigt und Lösungsstrategien entwickelt. Vor allem spielen Design und Usablity eine entscheidende Rolle. Verschiedene Aspekte und Alternativen, die im Entwicklungsprozess zu beachten sind, werden näher beleuchtet.
Shader zur Bildbearbeitung

In den letzten Jahren haben Grafikkarten eine starke Veränderung erfahren. Anfangs war lediglich die Darstellung vorberechneter Primitive möglich, mittlerweile lassen sich Vertex- und Pixelshader komplett frei programmieren. Die Spezialisierung auf den Rendervorgang hat die GPUs (Graphics Processing Units) zu massiv-parallelen Prozessoren wachsen lassen, die unter optimaler Ausnutzung ein Vielfaches der Rechenleistung aktueller CPUs erreichen. Die programmierbaren Shader haben Grafikkarten in der letzten Zeit vermehrt als weiteren Prozessor für General Purpose-Programmierung werden lassen.
Aktuelle Bildbearbeitungsprogramme zeigen, dass sich die Tendenz Richtung GPU bewegt, so wird sich auch in dieser Arbeit die enorme Rechenleistung der GPU für die Bildbearbeitung zu nutzen gemacht. Bildfilter lassen sich als Pixelshader realisieren und ermöglichen so die Ausführung direkt auf der GPU. Das vorgestellte Framework SForge wurde mit dem Ziel entwickelt, zu einem bestehenden Framework kompatibel zu sein. Als bestehendes Framework wurde auf AForge zurückgegriffen. Mit SForge können bestehende und eigene Bildfilter direkt auf der GPU ausgeführt werden, aber auch die Konvertierung von Farbräumen und Farbsystemen wurden realisiert. Das Framework arbeitet floatbasierend. Somit können auch HDR-Daten verarbeitet werden, um beispielsweise Tonemapping anzuwenden. Filter mit Parametern lassen sich über einen optionalen Dialog interaktiv ändern und modifizieren das Resultat in Echtzeit.
The use of chemically synthesized short interfering RNAs (siRNAs) is currently the method of choice to manipulate gene expression in mammalian cell culture, yet improvements of siRNA design is expectably required for successful application in vivo. Several studies have aimed at improving siRNA performance through the introduction of chemical modifications but a direct comparison of these results is difficult. We have directly compared the effect of 21 types of chemical modifications on siRNA activity and toxicity in a total of 2160 siRNA duplexes. We demonstrate that siRNA activity is primarily enhanced by favouring the incorporation of the intended antisense strand during RNA-induced silencing complex (RISC) loading by modulation of siRNA thermodynamic asymmetry and engineering of siRNA 3-overhangs. Collectively, our results provide unique insights into the tolerance for chemical modifications and provide a simple guide to successful chemical modification of siRNAs with improved activity, stability and low toxicity.
Chapter 1 contains the general background of our work. We briefly discuss important aspects of quantum chromodynamics (QCD) and introduce the concept of the chiral condensate as an order parameter for the chiral phase transition. Our focus is on the concept of universality and the arguments why the O(4) model should fall into the same universality class as the effective Lagrangian for the order parameter of (massless) two-flavor QCD. Chapter 2 pedagogically explains the CJT formalism and is concerned with the WKB method. In chapter 3 the CJT formalism is then applied to a simple Z2 symmetric toy model featuring a one-minimum classical potential. As for all other models we are concerned with in this thesis, we study the behavior at nonzero temperature. This is done in 1+3 dimensions as well as in 1+0 dimensions. In the latter case we are able to compare the effective potential at its global minimum (which is minus the pressure) with our result from the WKB approximation. In chapter 4 this program is also carried out for the toy model with a double-well classical potential, which allows for spontaneous symmetry breaking and tunneling. Our major interest however is in the O(2) model with the fields treated as polar coordinates. This model can be regarded as the first step towards the O(4) model in four-dimensional polar coordinates. Although in principle independent, all subjects discussed in this thesis are directly related to questions arising from the investigation of this particular model. In chapter 5 we start from the generating functional in cartesian coordinates and carry out the transition to polar coordinates. Then we are concerned with the question under which circumstances it is allowed to use the same Feynman rules in polar coordinates as in cartesian coordinates. This question turns out to be non-trivial. On the basis of the common Feynman rules we apply the CJT formalism in chapter 6 to the polar O(2) model. The case of 1+0 dimensions was intended to be a toy model on the basis of which one could more easily explore the transition to polar coordinates. However, it turns out that we are faced with an additional complication in this case, the infrared divergence of thermal integrals. This problem requires special attention and motivates the explicit study of a massless field under topological constraints in chapter 8. In chapter 7 we investigate the cartesian O(2) model in 1+0 dimensions. We compare the effective potential at its global minimum calculated in the CJT formalism and via the WKB approximation. Appendix B reviews the derivation of standard thermal integrals in 1+0 and 1+3 dimensions and constitutes the basis for our CJT calculations and the discussion of infrared divergences. In chapter 9 we discuss the so-called path integral collapse and propose a solution of this problem. In chapter 10 we present our conclusions and an outlook. Since we were interested in organizing our work as pedagogical as possible within the narrow scope of a diploma thesis, we decided to make extensive use of appendices. Appendices A-H are intended for students who are not familiar with several important concepts we are concerned with. We will refer to them explicitly to establish the connection between our work and the general context in which it is settled.
Of central importance in the whole thesis is the concept of the generating functional and the partition function, respectively. In appendix A.1 we present the general context in which the partition function appears and its general definition within the operator formalism of second quantization. Alternatively, this definition can be rewritten via the path integral formalism. We restrict ourselves to scalar fields in this case. Furthermore, the understanding of the CJT formalism is based on knowledge about n-point functions (connected or disconnected, in the presence or in the absence of sources) and the context in which they arise. In appendix A.2 we give their definition taking account of the different modifications in which these quantities occur in this thesis, i.e., scalar field theory at zero or at nonzero temperature, respectively. From a didactic point of view, we believe that it is helpful if one can establish a relation between special cases and a general framework. Therefore, in appendix A.3 we want to keep an eye on the overall picture. We discuss the general concept of the generating functional for correlation functions, which also covers the partition function. We also briefly comment on the general concept of Feynman rules and we clarify the meaning of the terms Green’s function and propagator.
Die vorliegende Arbeit setzt sich mit dem Einfluss von Tetraquarkzuständen auf den chiralen Phasenübergang auseinander. Das Quarkmodell ist ein wirkungsvolles Instrument zum Verständnis des Verhaltens der Mesonen und Baryonen. Im Bereich von Energien unter 1:8 GeV gibt es in Bezug auf die Mesonen dennoch eine Vielzahl von Problemen. So ist bis dato nicht klar, wie sich die skalaren Resonanzen unter 1:8 GeV zusammensetzen. In der Vergangenheit wurde eine Vielzahl von Möglichkeiten diskutiert, wie beispielsweise die Existenz eines Glueballs, die von mesonischen Molekülen sowie die von Tetraquarkzuständen. Alle diese Ansätze sind mehr oder weniger in der Lage, die Phänomene bei T = 0 zu erklären. Die vorliegende Arbeit beschäftigt sich mit dem Ansatz, Tetraquarkzuständen leichte skalare Resonanzen zuzuordnen.
Im Bereich der nichtverschwindenden Temperaturen wurden durch Gitterrechnungen große Erfolge erzielt, die aaffetiven Modellen bei nichtverschwindender Temperatur als Referenz dienen. Gleichwohl fehlt ein affektives Modell bei T <> 0, das einen Tetraquarkzustand beinhaltet. Hier setzt diese Arbeit an und erweitert das Modell, das leichte skalare Resonanzen mittels Tetraquarkzuständen erklärt, zu nichtverschwindenden Temperaturen.
In dieser ersten Studie zum Thema wird das Lineare Sigma-Modell um den leichtesten Tetraquarkzustand ergänzt und sein Einfluss auf den chiralen Phasenübergang untersucht.
Es zeigt sich, dass dieser Ansatz nicht nur Fragen im Bereich der Vakuumphysik lösen kann, sondern auch solche hinsichtlich der Restaurierung der chiralen Symmetrie.
Atmospheric new particle formation is a general phenomenon observed over coniferous forests. So far nucleation is either parameterised as a function of gaseous sulphuric acid concentration only, which is unable to explain the observed seasonality of nucleation events at different measurement sites, or as a function of sulphuric acid and organic molecules. Here we introduce different nucleation parameters based on the interaction of sulphuric acid and terpene oxidation products and elucidate the individual importance. They include basic trace gas and meteorological measurements such as ozone and water vapour concentrations, temperature (for terpene emission) and UV B radiation as a proxy for OH radical formation. We apply these new parameters to field studies conducted at conducted at Finnish and German measurement sites and compare these to nucleation observations on a daily and annual scale. General agreement was found, although the specific compounds responsible for the nucleation process remain speculative. This can be interpreted as follows: During cooler seasons the emission of biogenic terpenes and the OH availability limits the new particle formation while towards warmer seasons the ratio of ozone and water vapour concentration seems to dominate the general behaviour. Therefore, organics seem to support ambient nucleation besides sulphuric acid or an OH-related compound. Using these nucleation parameters to extrapolate the current conditions to prognosed future concentrations of ozone, water vapour and organic concentrations leads to a significant potential increase in the nucleation event number.
Die Gattung Mycobacterium umfasst mehr als einhundert verschiedene Spezies und wird in zwei große Gruppen unterteilt; die Gruppe der typischen Mykobakterien (obligat pathogen) und die Gruppe der atypischen Mykobakterien (fakultativ pathogen, ubiquitär vorkommend). Die Klinik der Mykobakteriosen ist mannigfaltig. Je nach Erreger und Immunstatus des Erkrankten können diese Infektionen, v.a. bei immunkompromittierten Patienten, letal verlaufen. Die frühzeitige Diagnostik und spezifische Therapie kann somit maßgeblich zur Senkung der Letalität der Mykobakteriosen beitragen. Die derzeit zur Verfügung stehenden diagnostischen Methoden sind nicht immer ausreichend um in jedem Einzelfall den Erreger genau zu klassifizieren. Ziel der vorliegenden Promotionsarbeit war die Erarbeitung einer sensitiven und schnellen Methode, zur Identifizierung der typischen und atypischen Mykobakterien. Die Testungen erfolgten zunächst mittels kulturell gewonnenen Mykobakterien-Suspensionen. Anschließend wurde der Methode anhand von, in Formalin-fixierten und in Paraffin eingebetteten, Gewebeproben evaluiert. Methode Im Rahmen der Extraktion wurde ein kommerzieller Kit (Roche High Pure PCR Template Preparation Kit®) durch mehrere Schritte ergänzt. Der Gewebe-Verdau mit Proteinase K wurde durch einen Inkubationsschritt über Nacht bei 72°C erweitert. Zudem wurde die Aufspaltung der Hülle der Mykobakterien durch wiederholende Temperaturwechsel von 300°C (+100°C in kochendem Wasser; -196°C in Flüssigstickstoff) unterstützt. Als Zielsequenz der Nested-PCR wurde die 16S rRNA verwendet. Das äußere Primerpaar, mit dem Primer 285 (5´ GAG AGT TTG ATC CTG GCT CAG 3´) und dem Primer 264 (5´TGC ACA CAG GCC ACA AGG GA 3´), amplifiziert ein 1037 Basenpaar langes Fragment. Bei dem inneren Amplifikat der Nested-PCR kommen die Primer 245 (5´ TAA TAC ATG CAA GTC GAA CG 3´) und Primer 259 (5´ TTT CAC GAA CAA CGC GAC AA 3´) zum Einsatz, welche ein Fragment mit der Länge von 560 Basenpaaren amplifizieren. Zur Verringerung der Kontaminationsgefahr wurde die Nested-PCR durch ein UTP-Verfahren ergänzt. Im Rahmen dessen wurden der Primer 285 um 2 Nukleotide am 3´Ende (5´ GAG AGT TTG ATC CTG GCT C 3‘) und der Primer 264 ebenfalls um 2 Nukleotide am 3´Ende (5´TGC ACA CAG GCC ACA AGG 3´) verkürzt. Ergebnisse Mit Hilfe der standardisierten Mykobakterien-Suspensionen konnte eine Sensitivitäts-Steigerung, alleinig durch die erneuerte DNA-Extraktion, um den Faktor 10 gezeigt werden. Die Etablierung der Nested-PCR, im Vergleich zu einer herkömmlichen PCR, brachte nochmalig eine Sensitivitäts-Steigerung um den Faktor 100. Insgesamt wurden 118 Organbiopsien untersucht. Davon war bei 46 Proben klinisch eine Tuberkulose und bei 42 Proben eine atypischen Mykobakteriose nachgewiesen. Die verbleibenden 30 Proben gehörten dem Vergleichskollektiv (Negativkontrollen) an. Von insgesamt 23 kulturell und/oder mikroskopisch positiven Proben konnte bei 7 Proben (30,4%) M. tuberculosis mittels Nested-PCR nachgewiesen werden. Für die atypischen Mykobakteriosen lag die Zahl bei 6 von 23 Proben (26,1%). Damit ist die Nested-PCR für atypische Mykobakterien nicht signifikant schlechter als für M. tuberculosis. Die Spezifität der Nested-PCR mit anschließender Sequenzierung, sowohl für die Tuberkulose, als auch für die atypischen Mykobakteriosen, beträgt 100%. Der positive Vorhersagewert für die Diagnosesicherung einer Tuberkulose, als auch einer atypischen Mykobakteriose, mittels Nested-PCR ist P = 1. Diskussion Diese Nested-PCR ermöglicht mittels nur einer Methode den Nachweis von typischen und atypischen Mykobakterien. Das Verfahren wurde erfolgreich getestet und evaluiert. Mit Hilfe dieses Verfahrens kann innerhalb von 48 Stunden (und an schließender Sequenzierung) der gezielte und spezifische Erregernachweis aus Organbiopsien erfolgen. Zudem bietet es die Möglichkeit einer routinemäßigen Anwendung. Diese Methode kann demnach in Einzelfällen zu einer eindeutigen und schnellen Diagnosesicherung und zur frühzeitigen und gezielten antimykobakteriellen Therapie beitragen. Darüber hinaus beinhaltet sie die Möglichkeit zur Durchführung von retrospektiven Studien, da der Nachweis der Mykobakterien aus Formalinfixierten und in Paraffin eingebetteten Gewebeproben erfolgte.
Die vorliegende Studie führte einen Vergleich zwischen Lokalanästhesie (LA) und Allgemeinanäthesie (GA) hinsichtlich der Entwicklung früher postoperativer kognitiver Dysfunktion (POCD) nach Carotis-Thrombendarterektomie (Carotis- TEA) durch. Es handelt sich hierbei um eine prospektive, extern randomisierte Studie, die als Teil einer Subgruppenanalyse der europaweiten GALA-Studie (General Anaesthesia versus Local Anaesthesia) durchgeführt wurde. Es wurden insgesamt 40 Patienten in die Studie eingeschlossen (GA n = 23, LA n = 17). Die kognitive Leistungsgeschwindigkeit wurde mittels Zahlen-Verbindungs-Tests (ZVT) und die Befindlichkeit wurde durch die Befindnisskalierung durch Kategorie und Eigenschaftswörter (BSKE(EWL)) zu jeweils vier Messzeitpunkten (MZP) (am Vorabend der Operation, 5, 29 und 77 Stunden postoperativ) bestimmt. Das Ausmaß der neuronalen Schädigung wurde mittels des biochemischen Markers Protein S-100b zu drei MZPs bestimmt (präoperativ, vor Unclamping der A. carotis interna und am Ende der Operation). Die Geschwindigkeit im ZVT nahm in der GA-Gruppe postoperativ signifikant ab (p = 0,0001). Sowohl fünf (p = 0,023) als auch 29 Stunden (p = 0,043) postoperativ zeigten sich signifikante Gruppenunterschiede. Die S-100b-Konzentration zeigte in der GA-Gruppe am zweiten MZP einen signifikanten (p < 0,001) Anstieg von 0,086 ± 0,038 mikro g/l, im Vergleich zum Ausgangswert von 0,061 ± 0,024 mikro g/l. Signifikante Gruppenunterschiede konnten wir hier am Ende der Operation messen (p = 0,006). Bei der Befindlichkeitsskalierung waren keine signifikanten Gruppenunterschiede messbar. Anhand unserer Ergebnisse können wir eindeutig die Thesen widerlegen, dass das verwendete Anästhesieverfahren keinen Einfluss auf die Entwicklung früher POCD hat und dass es nicht das Ausmaß der neuronalen Schädigung beeinflusst. Wir schließen daraus, dass die Lokalanästhesie einen positiven Effekt auf die Entwicklung früher POCD und das Ausmaß der neuronalen Schädigung hat. Eingeschränkt wird die Aussagekraft dieser Ergebnisse durch eine kleine Fallzahl, ein kurzes Follow-up und ein nicht standardisiertes Neuromonitoring und Shunting unter Allgemeinanästhesie.
Critical perspectives have become more visible in German human geography. Drawing on an analysis of the debate around the German reader "Kulturgeographie" published in 2003, we suggest that this case provides new insights into the "geography of critical geography". We briefly discuss the history of critical geography in Germany, leading to a comparison of the conditions of critical geography around 1980 and in recent years. The focus is on two factors in the changed role of critical perspectives in German geography: (1) the growing internationalisation of German geography, which opened new avenues and allowed new approaches to enter the discipline; and (2) the high citation indices of "critical" journals, which leads to an enhanced reputation and a high significance of international critical geography in the German discipline. However, we draw an ambiguous conclusion: the increased role of critical approaches in German geography is linked to a growing neoliberalisation of academia and a decline of critical approaches in other disciplines.

Abstract: Bcl-2 family proteins including the pro-apoptotic BH3-only proteins are central regulators of apoptotic cell death. Here we show by a focused siRNA miniscreen that the synergistic action of the BH3-only proteins Bim and Bmf is required for apoptosis induced by infection with Neisseria gonorrhoeae (Ngo). While Bim and Bmf were associated with the cytoskeleton of healthy cells, they both were released upon Ngo infection. Loss of Bim and Bmf from the cytoskeleton fraction required the activation of Jun-N-terminal kinase-1 (JNK-1), which in turn depended on Rac-1. Depletion and inhibition of Rac-1, JNK-1, Bim, or Bmf prevented the activation of Bak and Bax and the subsequent activation of caspases. Apoptosis could be reconstituted in Bim-depleted and Bmf-depleted cells by additional silencing of antiapoptotic Mcl-1 and Bcl-XL, respectively. Our data indicate a synergistic role for both cytoskeletal-associated BH3-only proteins, Bim, and Bmf, in an apoptotic pathway leading to the clearance of Ngo-infected cells. Author Summary: A variety of physiological death signals, as well as pathological insults, trigger apoptosis, a genetically programmed form of cell death. Pathogens often induce host cell apoptosis to establish a successful infection. Neisseria gonorrhoeae (Ngo), the etiological agent of the sexually transmitted disease gonorrhoea, is a highly adapted obligate human-specific pathogen and has been shown to induce apoptosis in infected cells. Here we unveil the molecular mechanisms leading to apoptosis of infected cells. We show that Ngo-mediated apoptosis requires a special subset of proapoptotic proteins from the group of BH3-only proteins. BH3-only proteins act as stress sensors to translate toxic environmental signals to the initiation of apoptosis. In a siRNA-based miniscreen, we found Bim and Bmf, BH3-only proteins associated with the cytoskeleton, necessary to induce host cell apoptosis upon infection. Bim and Bmf inactivated different inhibitors of apoptosis and thereby induced cell death in response to infection. Our data unveil a novel pathway of infection-induced apoptosis that enhances our understanding of the mechanism by which BH3-only proteins control apoptotic cell death.
Soziale Ökologie als Wissenschaft von den gesellschaftlichen Naturverhältnissen hat sich in den vergangenen Jahren als ein neues transdisziplinäres Wissenschaftsgebiet in außeruniversitären Forschungsbereichen und mehr und mehr auch im akademischen Feld etabliert. Der strukturelle Wandel der Hochschulen eröffnet neue Chancen für die Soziale Ökologie, auch in der Lehre einen festen Platz zu erhalten. Der Beitrag reflektiert anhand des neuen Masterstudiengangs Umweltwissenschaften an der Frankfurter Goethe-Universität und anhand von umweltwissenschaftlichen Lehrveranstaltungen an der TU Darmstadt die Anforderungen an die Lehre einer transdisziplinären Wissenschaft. Verdeutlicht wird dies am Problem der Integration unterschiedlicher Problemsichten, Fachkulturen und Wissensbestände.
Das Modifizierte Vacciniavirus Ankara (MVA) entstand nach mehr als 500 Passagen des Chorioallantois Vacciniavirus Ankara (CVA) auf Hühnerembryofibroblasten. Mit der Passagierung ging u.a. der Verlust zahlreicher Vacciniavirus Virulenzfaktoren einher, wodurch MVA einen hochattenuierten Phänotyp aufweist. Bei mangelnder Pathogenität für humane Organismen verfügt das MVA jedoch über eine vollständig erhaltene Genexpression inklusive integrierter Fremdgene. Hierdurch bedingt ist MVA ein vielversprechender Kandidat nicht nur als Pockenimpfstoff der dritten Generation, sondern auch als Vektorvakzine gegen zahlreiche Infektionskrankheiten sowie in der Tumor-assoziierten Immunotherapie. Ein wesentliches Charakteristikum der Attenuierung von MVA ist seine fehlende Replikationsfähigkeit in humanen Zellen. Bislang ist es jedoch noch nicht möglich gewesen, die hierfür verantwortlichen genetischen Veränderungen eindeutig zuzuordnen. Vorangegangene Arbeiten wiesen darauf hin, dass sich der beschriebene Effekt zumindest teilweise auf die Zeitspanne zwischen zwei Vorläufern des MVA eingrenzen lässt. So zeigten sich humane HeLa Zellen für das CVA 152 permissiv, während gegenüber dem CVA 386 nur noch eine Semipermissivität bestand. Mit Hilfe einer vergleichenden Sequenzanalyse von CVA 152 und CVA 386 konnten verschiedene Deletionen identifiziert werden, deren Auftreten potentiell verantwortlich für den veränderten Phänotyp von CVA 386 ist. In diesem Zusammenhang wurden im Rahmen dieser Arbeit die Gene C12L, C14L, C15L und C16L der Deletion I und die Gene M1L, M2L und der OLR 037 der Deletion II charakterisiert. Durch Transfektion von Expressionsplasmiden wurden die entsprechenden Proteine bezüglich ihrer Expression und posttranslationalen Modifikation, sowie ihrer subzellulären Verteilung untersucht. So ließ sich für das Protein M2 eine posttranslationale N-Glykosylierung und die Lokalisation innerhalb des trans-medialen Golgiapparats nachweisen. Zusätzlich konnte eine Sekretion des M2 Proteins im Zellüberstand transfizierter 293T-Zellen gezeigt, sowie die bereits publizierte Inhibition der NF-:B-Aktivierung durch extrazellulär zugegebenes M2 Protein eingeleitet werden. Die Proteine C14 und C15 zeigten sich in der Laserscanmikroskopie mitochondrial lokalisiert; C16 dagegen kolokalisierte neben einer hauptsächlich intranukleären Verteilung zusätzlich mit F-Aktin. Ferner ergaben sich für die Proteine C14 und C15 erste Hinweise auf eine inhibitorische Funktion gegenüber Staurosporin-induzierter Apoptose.

Advents- und Weihnachtsbräuche unterlagen und unterliegen wirtschaftlich mitbedingten soziokulturellen Moden. Zum weihnachtlichen “Bunten Teller” der Nachkriegzeit gehörten neben allerlei Süßwaren und Backwerk auch blank polierte rote Äpfel und vor allem die begehrten und bestaunten Südfrüchte Apfelsinen und Mandarinen. Zu Weihnachten durfte man dieses sonst eher knappe Obst nach Herzenslust genießen. Seit den 1950er Jahren haben sich Anbau und Verzehr von Zitrusfrüchten stetig ausgeweitet und so sind sie heute ein selbstverständlicher Anteil unseres ganzjährigen Obstkorbes. Dies liegt nicht nur an den gewachsenen Einkommen der Konsumenten, sondern auch an der Ausweitung der Anbauflächen und Produktionssteigerungen sowie an den optimierten Transport- bzw. Lagerbedingungen. Erste Zeugnisse von Orangen findet man um 500 v. Chr. bei Konfuzius. Von China aus gelangten die Früchte über die Malaiische Halbinsel nach Indien, zur Ostküste Afrikas und in den Mittelmeerraum. Seit dem 15. Jahrhundert sind Apfelsinen und Zitronen, mit allen weiteren Sorten und Kulturformen als "Agrumen" zusammengefasst, in Europa bekannt, wo sie wie die Oliven als Charakterarten des mediterranen Raumes gelten Bei der Beschäftigung mit der Gattung Citrus begibt man sich “in eines der undurchdringlichsten Dickichte der Kulturpflanzen-Terminologie” (BRÜCHER 1977). Ursächlich dafür ist, dass die Gattung wohl nirgends mehr wild vorkommt, sondern seit gut zwei Jahrtausenden gezüchtet wird. Alle Citrus-Arten und Hybriden lassen sich untereinander kreuzen. Mit der Gattung Fortunella (Kumquat) können selbst Gattungshybriden erzeugt werden, und die Gattung Poncirus (Bitterorange) dient als Propfunterlage für Arten der Gattung Citrus. So wundert man sich kaum noch, dass es mehr als 60 verschiedene Zitrusfruchtarten gibt, etwa ein Duzend davon haben kommerzielle Bedeutung.
Der Hohle Lerchensporn, dessen Name sich auf die Ähnlichkeit des Sporns mit der Zehe einer Haubenlerche bezieht, zählt zu den Frühlingsgeophyten. Er treibt aus einer ausdauernden unterirdischen Knolle aus. Diese entsteht aus der Verdickung des Wurzelhalses (wie beim Radieschen, sog. Hypokotylknollen). Beim Hohlen Lerchensporn stirbt das verbrauchte Speichergewebe jährlich von innen her ab, so dass die Höhle sich vergrößert. Außen lagert sich neues Speichergewebe an.
Die Stiftung Naturschutz Hamburg und Stiftung LOKI SCHMIDT zum Schutze gefährdeter Pflanzen hat zur "Blume des Jahres 2009" die Wegwarte (Cichorium intybus) ausgewählt. Sie bevorzugt einen ähnlichen Lebensraum wie die Nickende Distel, die Blume des Jahres 2008. Die Wegwarte ist aber häufiger zu finden, sie wächst an Weg- und Ackerrändern und auf bäuerlichen Schuttplätzen. Mich hat schon als Kind diese Pflanze fasziniert, deren strahlend blaue Blüten sich morgens öffnen, aber am frühen Nachmittag "schlafen gehen". Die Wegwarte gehört zu der Pflanzenfamilie der Korbblütler (Compositen). Sie kann bis 120 cm hoch werden. Der Stängel ist recht verzweigt. Die dunkelgrünen, etwas rauhaarigen Blätter sind am Grund etwas fiederspaltig, die mittleren und obenstehenden Blätter sind aber fast ganzrandig.
Der Beitrag veranschaulicht das Vorgehen zur Identifizierung von Wald- und Buschwald-Lebensraumtypen in der Nordwest-Türkei als Grundlage für eine räumliche Erweiterung der gesamteuropäischen strategischen Vision zur Erhaltung und nachhaltigen Nutzung biologischer und landschaftlicher Vielfalt (PEBLDS).
Borrelia recurrentis, the etiologic agent of louse-borne relapsing fever in humans, has evolved strategies, including antigenic variation, to evade immune defence, thereby causing severe diseases with high mortality rates. Here we identify for the first time a multifunctional surface lipoprotein of B. recurrentis, termed HcpA, and demonstrate that it binds human complement regulators, Factor H, CFHR-1, and simultaneously, the host protease plasminogen. Cell surface bound factor H was found to retain its activity and to confer resistance to complement attack. Moreover, ectopic expression of HcpA in a B. burgdorferi B313 strain, deficient in Factor H binding proteins, protected the transformed spirochetes from complement-mediated killing. Furthermore, HcpA-bound plasminogen/plasmin endows B. recurrentis with the potential to resist opsonization and to degrade extracellular matrix components. Together, the present study underscores the high virulence potential of B. recurrentis. The elucidation of the molecular basis underlying the versatile strategies of B. recurrentis to escape innate immunity and to persist in human tissues, including the brain, may help to understand the pathological processes underlying louse-borne relapsing fever.
Die Mauser- und Überwinterungsgebiete der süddeutschen Brutvögel des Großen Brachvogels liegen an der Atlantikküste Frankreichs und der Iberischen Halbinsel, individuelle Unterschiede sowie Populationsunterschiede sind jedoch nicht bekannt. Daher wurde in einem Pilotversuch am 20. Mai 2008 ein brütendes Weibchen in einem Brutgebiet am badischen Oberrhein mit einem Satellitensender versehen. Das Weibchen zog am 8. Juni 2008 vermutlich in einem Direktflug von 1020 km in west-süd-westlicher Richtung an die kantabrische Atlantikküste. Signale wurden bis zum 9. Oktober 2008 empfangen.
NASAs Stardust Mission ist die erste Mission, die - nach den Apollo Missionen zum Mond - Material von einem extraterrestrischen Körper erfolgreich für die Untersuchung auf der Erde, zurückgebracht hat. Desweiteren konnten erfolgreich Proben von einem Interstellaren Partikelstrom aufgesammelt werden, der das Sonnensystem derzeit passiert. Die Mission erlaubt einen Einblick in die Beschaffenheit der Kometenpartikel, die Rolle von Kometen im Sonnensystem sowie den Eintrag von Staub in die Zodiakalwolke. Desweiteren erlaubt die Analyse der Kometenpartikel den direkten Vergleich zu bereits untersuchten Meteoriten und Interplanetaren Staub Partikeln (IDPs) die auf der Erde bzw. deren Stratosphäre gesammelt wurden. Stardust ist die vierte ”Discovery” Mission und wurde am 7. Februar 1999 gestartet. Während des Fluges zum Kometen 81P/Wild 2 wurde ein interstellarer Partikelstrom beprobt und der Asteroid Annefrank passiert. Nach fünf Jahren kam es zum Zusammentreffen mit dem Kometen 81P/Wild 2 und über fünf Minuten, wurden Proben mit einer Auffangvorrichtung eingesammelt. Es dauerte weitere zwei Jahre bis die Stardust Sonde die Proben erfolgreich zur Erde zurückgebracht hat und zur Untersuchung freigegeben wurden. Interstellare und kometare Partikel wurden mit einer tennisschlägerartigen Auffangvorrichtung eingefangen, die aus einer Vielzahl von Aerogel Zellen aufgebaut ist. Das für die Stardust Mission verwendete Aerogel besteht aus SiO2 dessen dendritische Struktur zu 99,8 % aus Luft (Poren) besteht. Dadurch erscheint es nahezu transparent, was die optische Suche mit Mikroskopen nach den Einschlagsspuren der Körner vereinfacht. Die jeweiligen Partikel wurden auf unterschiedlichen Seiten der Vorrichtung eingefangen, da unterschiedliche Eigenschaften des Aerogels notwendig waren und um sie später voneinander unterscheiden zu können. Die Seite, in der die Kometenpartikel eingefangen wurde, musste Körner mit unterschiedlichen Grössen, Morphologien und niedrigeren Geschwindigkeiten abbremsen, während auf der interstellaren Seite die Körner von wesentlich höheren Geschwindigkeiten abgebremst werden mussten. Die Aerogelzellen haben ein variierendes Dichteprofil: an der Oberfläche ist die Dichte geringer (5 mg/ml) und erhöht sich mit der Tiefe auf 30-50 mg/ml. Dieses Dichteprofil ist notwendig, da beim Einschlag der Körner auf das Auffangmedium ein hoher Druck entsteht, der umso geringer ist, je niedriger die Dichte im Moment des Auftreffens ist. Die Aerogelzellen für die Kometenpartikel haben drei Lagen mit unterschiedlichen Dichten, die Zellen für die interstellaren Körner haben zwei unterschiedliche Dichten (Tsou et al., 2003)....
Der botanische Name der Erbse, Pisum sativum, stammt aus dem Lateinischen. "Pisum" wurde mit dem germanischen Wort 'Erbse' übersetzt, "sativus" bedeutet: angepflanzt, angebaut. Erbsen zählen zu den seit dem Neolithikum in Europa kultivieren Nutzpflanzen und zusammen mit Emmer, Einkorn und stellenweise auch Gerste zu den ältesten Grundnahrungsmitteln der frühen Ackerbauern. Seit der Bronzezeit, ca. 1800 v. Chr., kennt man auch die beiden anderen, heute ebenso noch eine bedeutende Rolle spielenden Hülsenfrucht-Arten Linsen und Bohnen. Erbsen sind reich an Kohlehydraten und Proteinen. Sie enthalten alle essentiellen Aminosäuren, die der menschliche Organismus nicht selbst herstellen kann, sondern über die Nahrung aufnehmen muss.
The NADH:ubiquinone oxidoreductase (complex I) is a large membrane bound protein complex coupling the redox reaction of NADH oxidation and quinone reduction to vectorial proton translocation across bioenergetic membranes. The mechanism of proton pumping is still unknown; it seems however that the reduction of quinone induces conformational changes which drive proton uptake from one side and release at the other side of the membrane. In this study the proposed quinone and inhibitor binding pocket located at the interface of the 49-kDa and PSST subunits was explored by a large number of point mutations introduced into complex I from the strictly aerobic yeast Yarrowia lipolytica. Point mutations were systematically chosen based on the crystal structure of the hydrophilic domain of complex I from Thermus thermophilus. In total, the properties of 94 mutants at 39 positions which completely cover the lining of the large putative quinone and inhibitor binding cavity are described and discussed here. A structure/function analysis allowed the identification of functional domains within the large putative quinone binding cavity. A possible quinone access path ranging from the N-terminal beta-sheet of the 49-kDa subunit into the pocket to tyrosine 144 could be defined, since all exchanges introduced here, caused an almost complete loss of complex I activity. A region located deeper in the proposed quinone binding pocket is apparently not important for complex I activity. In contrast, all exchanges of tyrosine 144, even the very conservative mutant Y144F, essentially abolished dNADH:DBQ oxidoreductase activity of complex I. However, with higher concentrations of Q1 or Q2 the dNADH:Q oxidoreductase activity was largely restored in the mutants with the more conservative exchanges. Proton pumping experiments showed that this activity was also coupled to proton translocation, indicating that these quinones were reduced at the physiological site. However, the apparent Km values for Q1 or Q2 were drastically increased, clearly demonstrating that tyrosine 144 is central for quinone binding and reduction. These results further prove that the enzymatically relevant quinone binding site of complex I is located at the interface of the 49-kDa and PSST subunits. The quinone binding pocket is thought to comprise the binding sites for a plethora of specific complex I inhibitors that are usually grouped into three classes. The large array of mutants targeting the quinone binding cavity was examined with a representative of each inhibitor class. Many mutants conferring resistance were identified which, depending on the inhibitor tested, clustered in well defined and partially overlapping regions of the large putative quinone and inhibitor binding cavity. Mutants with effects on type A (DQA) and type B (rotenone) inhibitors were found in a subdomain corresponding to the former [NiFe] site in homologous hydrogenases, whereby the type A inhibitor DQA seems to bind deeper in this domain. Mutants with effects on the type C inhibitor (C12E8) were found in a narrow crevice. Exchanging more exposed residues at the border of these well defined domains affected all three inhibitor types. Therefore, the results as a whole provide further support for the concept that different inhibitor classes bind to different but partially overlapping binding sites within a single large quinone binding pocket. In addition, they also indicate the approximate location of the binding sites within the structure of the large quinone and inhibitor binding cavity at the interface of the 49 kDa and the PSST subunit. It has been proposed earlier that the highly conserved HRGXE-motif in the 49-kDa subunit forms a part of the quinone binding site of complex I. Mutagenesis of the HRGXE-motif, revealed that these residues are rather critical for complex I assembly and seem to have an important structural role. The question why iron-sulfur cluster N1a is not detectable by EPR in many models organisms is not solved yet. Introducing polar and positively charged amino acid residues close to this cluster in order to increase its midpoint potential did not result in the appearance of the cluster N1a EPR signal in mitochondrial membranes from the mutants. Clearly, further research will be necessary to gain insights to the function of this iron-sulfur cluster in complex I. In an additional project, a new and simple in vivo screen for complex I deficiency in Y. lipolytica was developed and optimized. This assay probes for defects in complex I assembly and stability, oxidoreductase activity and also proton pumping activity by complex I. Most importantly, this assay is applicable to all Y. lipolytica strains and could be used to identify loss-of-function mutants, gain-of-functions mutants (i.e. resistance towards complex I inhibitors) and revertants due to mutations in both nuclear and mitochondrially encoded genes of complex I subunits.
Gegenstand dieser Arbeit sind Galoisoperationen auf quasiplatonischen Riemannschen Flächen mit einer Automorphismengruppe isomorph zu PSL(2,F(q)). Quasiplatonische Riemannsche Flächen werden durch torsionsfreie Normalteiler N in einer Dreiecksgruppe D uniformisiert, d.h. N ist die universelle Überlagerungsgruppe und die Flächen, die man auch als algebraische Kurven beschreiben kann, sind isomorph zu N\U, wenn U die obere Halbebene bezeichnet. Bzgl. der Größe der Automorphismengruppen bilden die quasiplatonischen Kurven die lokalen Maxima im Modulraum. Die absoluten Maxima liegen bei den Hurwitz-Kurven; hier hat die Automorphismengruppe die maximale Größe von 84(g-1), wenn g>1 das Geschlecht der Kurve ist. Der Normalisator in PSL(2,R) der Überlagerungsgruppe N ist dann die Dreiecksgruppe mit Signatur (2,3,7). Macbeath hat die Bedingungen dafür gefunden, wann PSL(2,F(q)) eine Hurwitz-Gruppe ist. Von besonderem Interesse ist dabei der Fall, dass q=p eine Primzahl kongruent +-1 mod 7 ist. Hier hat man drei nicht-isomorphe Kurven, die jedoch alle galoiskonjugiert zueinander sind. In der Arbeit werden Bedingungen angegeben, unter denen sich dieses Resultat auf Dreiecksgruppen D mit einer Signatur der Form (2,m_1,m_2) verallgemeinern lässt. Dabei gehen einerseits Ergebnisse von Frye ein, der die Anzahl der verschiedenen torsionsfreien Normalteiler N<D mit Quotienten PSL(2,F(q)) über die Spurtupel der Erzeugenden von D bestimmt hat. Andererseits wird eine Methode von Streit verwendet, mit der man die Galoisoperation auf den Kurven anhand des Verhaltens der Multiplikatoren der Erzeugenden in der Automorphismengruppe nachvollziehen kann. Es zeigt sich, dass sich Spur- und Multiplikatortupel entsprechen, woraus man die Anzahl und Länge der Galois-Orbits erhält. Außerdem lässt sich der Definitionskörper der Kurven bestimmen. Offen bleibt das genaue Verhalten bei Signaturen (m_0,m_1,m_2) mit m_i ungleich 2 für alle i. Hier gibt es zu jedem Multiplikatortupel zwei verschiedene Spurtupel. Kann man die Kurven durch die Multiplikatoren beschreiben, dann erhält man Projektionen D->>PSL(2,F(q)) auch über die Quaternionenalgebra, die die Dreiecksgruppe über ihrem Spurkörper erzeugt. Die Normalteiler erweisen sich dann als Schnitt der Dreiecksgruppe mit einer Hauptkongruenzuntergruppe nach einem Primideal P|char(F(q)) in der Norm-1-Gruppe einer Ordnung der Quaternionenalgebra. Dabei ist das Spurtripel in PSL(2,F(q)) gerade das Spurtripel aus D modulo P. Ändert man P, so erhält man ein anderes Spurtripel in PSL(2,F(q)), also auch einen anderen Normalteiler. Bilden die zugehörigen Kurven eine Bahn unter der Galoisoperation, dann ergeben sich alle Normalteiler auf diese Weise. Die Galoisoperation auf den Tripeln der Multiplikatoren, also die Galoisoperation auf den Kurven, ist verträglich mit der Operation, die die Primideale P|char(F(q)) permutiert. Wir erhalten also eine natürliche Korrespondenz zwischen der Galoisoperation auf den Kurven einerseits und der Operation auf den Primidealen andererseits.

Ziel dieser Nachuntersuchung war es zu zeigen, ob Patienten mit einer rheumatischentzündlichen Grunderkrankung und rein degenerativ veränderten Hüftgelenken im Langzeitverlauf von einer Hüft-Totalendoprothese über den dorsalen Zugang profitieren. Zugrunde lag ein Kollektiv von 109 degenerativ veränderten Patienten- Hüften (108 Patienten) und 22 rheumatische Patientenhüften (21 Patienten), die in den Jahren 1995 bis 1998 in der orthopädischen Universitätsklinik von einem einzigen Operateur eine Hüft-Totalendoprothese über den dorsalen Zugang erhielten. Der Untersuchungszeitraum beläuft sich auf durchschnittlich 9,2 Jahre. Neben der klinischen Untersuchung mittels des Harris-Hip-Scores wertete die Arbeitsgruppe radiologische Beckenübersichtsaufnahmen und Lauensteinaufnahmen der endoprothetisch versorgten Hüftgelenke aus. Hierfür eignete sich der Mayo-Hip- Score mit dem das Ablöseverhalten der Prothese vom Knochen in Form einer Saumbildung ermittelt wurde. Es wurden 42 rein degenerativ veränderte Patienten-Hüften klinisch nachuntersucht, von denen 23 radiologische Bilder ausgewertet werden konnten. Bei Patienten aus dem rheumatischen Formenkreis lagen von zehn klinisch nachuntersuchten Patienten sechs radiologische Bilder vor. Ein gutes Ergebnis (84%) wird für die Gruppe der Patienten mit einer rein degenerativen Hüftgelenkserkrankung (Arthrose) und ein mittleres Ergebnis (78%) für Patienten aus dem rheumatischen Formenkreis evaluiert. Der Zusammenhang zwischen einer besseren Funktionsfähigkeit und geringeren Schmerzen nach 10 Jahren post operationem bei einem minimalinvasiven dorsalen Zugang kann nicht nachgewiesen werden. Dennoch erscheint es früh-postoperativ einige Vorteile für das Verwenden des minimal-invasiven Zugangs zum Hüftgelenk zu geben: Laut o. a. Studien liegt der intraoperative Blutverlust unter den Mengen von konventionellen Schnitten und die mögliche frühere Rehabilitationsfähigkeit bei kleineren Zugängen spricht ebenfalls für das Verwenden eines minimal-invasiven Zugangs. Insgesamt sind 92% der Patienten mit dem heutigen postoperativen Ergebnis zufrieden. Eine kleine Operationsnarbe, die weit dorsal liegt, könnte laut Patientinnen-Angaben sehr gut durch die Badebekleidung verdeckt werden, was bei antero-lateralen Zugängen nicht der Fall zu sein scheint. Dies erhöht eindeutig die Lebensqualität von Patientinnen, insbesondere von jungen Rheumatikerinnen. In Folge einer Nachbeobachtungszeit von durchschnittlich 9,2 Jahren lässt sich sagen, dass die Implantation einer Hüft-Totalendoprothese über den dorsalen Zugang sowohl bei degenerativ veränderten als auch bei rheumatisch-entzündlichen Hüftgelenken eine gute Alternative zu den Operationstechniken über den anterioren oder antero-lateralen Zugang darstellt. Die anfänglich beschriebene Ablehnung den dorsalen Zugang zu nutzen - weil über einen dorsaler Zugang das stützende Gewebe traumatisiert und damit die dorsale Luxationstendenz auf lange Sicht begünstigt werden und zu einer chronischen Instabilität des Hüftgelenkes führen könnte – konnte in unserer Studie nicht nachgewiesen werden. Zwar traten Luxationen auf, die dann teilweise operativ korrigiert werden mussten, aber es kam in keinem Fall während der Nachbeobachtungszeit zu einer Revision aufgrund einer Luxation und folgender Instabilität. Aufgrund dieser Daten- und Informationslage kann der dorsale Zugang zur Implantation einer Hüft-Totalendoprothese bei Patienten mit Arthrose und Patienten des rheumatischen Formenkreises als gute und sichere Alternative betrachtet werden.
An der Universität Frankfurt entwickelte Online-Self-Assessment-Verfahren für die Studiengänge Psychologie und Informatik sollen Studieninteressierten noch vor Studienbeginn auf der Basis von Selbsterkundungsmaßnahmen und Tests eine Rückmeldung über ihre eigenen Fähigkeiten, Motive, personalen Kompetenzen und Interessen mit Blick auf den jeweiligen Studiengang geben. Sowohl die Befunde zur psychometrischen Güte der Verfahren als auch jene zur prognostischen Validität lassen ihren Einsatz zur Feststellung studienrelevanter Kompetenzen als geeignet erscheinen. Da die erfassten Kompetenzen und Merkmale substanzielle Beziehun-gen zu Studienleistungen aufweisen, könnten die Informationen über individuelle Stärken zur Wahl eines geeigneten Studienganges genutzt werden; Schwächen hingegen könnten frühzeitig Hinweise für geeignete Fördermaßnahmen liefern.
Background Parkinson's disease (PD) is an adult-onset movement disorder of largely unknown etiology. We have previously shown that loss-of-function mutations of the mitochondrial protein kinase PINK1 (PTEN induced putative kinase 1) cause the recessive PARK6 variant of PD. Methodology/Principal Findings Now we generated a PINK1 deficient mouse and observed several novel phenotypes: A progressive reduction of weight and of locomotor activity selectively for spontaneous movements occurred at old age. As in PD, abnormal dopamine levels in the aged nigrostriatal projection accompanied the reduced movements. Possibly in line with the PARK6 syndrome but in contrast to sporadic PD, a reduced lifespan, dysfunction of brainstem and sympathetic nerves, visible aggregates of alpha-synuclein within Lewy bodies or nigrostriatal neurodegeneration were not present in aged PINK1-deficient mice. However, we demonstrate PINK1 mutant mice to exhibit a progressive reduction in mitochondrial preprotein import correlating with defects of core mitochondrial functions like ATP-generation and respiration. In contrast to the strong effect of PINK1 on mitochondrial dynamics in Drosophila melanogaster and in spite of reduced expression of fission factor Mtp18, we show reduced fission and increased aggregation of mitochondria only under stress in PINK1-deficient mouse neurons. Conclusion Thus, aging Pink1 -/- mice show increasing mitochondrial dysfunction resulting in impaired neural activity similar to PD, in absence of overt neuronal death.
Mitochondrien sind die Kraftwerke unserer Zellen. In ihnen findet die Zellatmung statt, die unseren Körper mit lebenswichtiger Energie versorgt. Zusätzlich teilen sich die Zellorganellen und verschmelzen wieder miteinander im Minutentakt. Was aber passiert, wenn Teile dieses dynamischen Geflechts Defekte aufweisen? Die Antwort dazu könnte ein Protein sein, das auf zwei verschiedene Weisen in die Mitochondrien-Membranen eingebaut wird. Liegt keine kurze Form des Proteins vor, ist das ein Hinweis dafür, dass die Organellen defekt sind. Die Mitochondrien verbrennen die mit der Nahrung zugeführten Kohlenhydrate und Fette unter Verbrauch von Sauerstoff zu Kohlendioxid und Wasser. Bei diesem Vorgang, der Zellatmung, wird über eine Reihe von Proteinkomplexen ein elektrochemisches Potenzial aufgebaut, das zur Produktion des Energieträgers ATP (Adenosintriphosphat) genutzt wird. ATP kann aus den Mitochondrien abtransportiert werden und steht somit als eine Art Treibstoff für alle Stoffwechselprozesse zur Verfügung. Die Arbeit der Mitochondrien ist der Hauptgrund für unseren täglichen Sauerstoffbedarf. Außerdem tragen die Nano-Kraftwerke der Zelle dazu bei, unsere Körpertemperatur auf 37 °C aufrechtzuerhalten. Aufgrund dieser zentralen Funktionen ist es nicht verwunderlich, dass eine Reihe von Krankheiten beim Menschen durch den Funktionsverlust von Mitochondrien verursacht oder beeinflusst wird. Das sind in erster Linie neurologische oder muskuläre Erkrankungen, aber auch Diabetes, Fettleibigkeit, verschiedene Formen von Krebs und Alterungsprozesse. Folglich ist es von immenser Bedeutung zu verstehen, wie Mitochondrien funktionieren, wie sie ihre Funktionalität aufrechterhalten und gegebenenfalls repariert oder entsorgt werden können. Dem können wir am Wissenschaftsstandort Frankfurt hervorragend nachgehen, da sich einige international ausgewiesene Forschungsgruppen in den Fachbereichen Medizin, Biologie, Chemie und am Max-Planck-Institut für Biophysik mit verschiedenen Aspekten der mitochondrialen Biologie befassen. In zahlreichen interdisziplinären Kooperationen wird so versucht, dieses komplexe System besser zu verstehen.
Im Rahmen der vorliegenden Arbeit wurde anhand der Auswertung von persönlichen Fragebögen von TTP-Patienten und deren behandelnder Ärzte und der Sichtung stationärer und ambulanter Krankenakten der Krankheitsverlauf bei 67 TTP-Patienten zwischen 1982 und August 2004 retrospektiv untersucht. Neben der Beschreibung der wichtigsten klinischen Symptome und Begleiterkrankungen, möglicher Triggerfaktoren, dem Rezidivverhalten und der diversen Therapieverfahren wurden die Patienten hinsichtlich ihres Geschlechts gesondert betrachtet, da der weibliche Patientenanteil den männlichen deutlich übertrifft. Des Weiteren wurden die Patienten hinsichtlich ihres Rezidivverhaltens getrennt untersucht, da es in vielen Fällen noch immer unklar ist, wann und ob Rezidive auftreten. Das Verhältnis zwischen Frauen und Männern betrug annährend 3 : 1 und entspricht somit vielen aktuellen Literaturangaben. Der Median des Alters bei Erstmanifestation betrug insgesamt 35,0 Jahre (n=67), die insgesamt 49 weiblichen Patienten waren 31,0 Jahre alt (Median) und der Median des Alters der 18 männlichen Patienten betrug 47,0 Jahre. Dieser doch sehr große Altersunterschied zum Zeitpunkt der Erstmanifestation liegt höchstwahrscheinlich an hormonellen Einflüssen (Kontrazeptiva, Schwangerschaft), denen Frauen im gebärfähigen Alter ausgesetzt sind. Im Vergleich der Rezidivpatienten mit den Nicht-Rezidivpatienten zeigt sich einerseits eine deutliche Häufung der Erstmanifestation bei Rezidivpatienten in früheren Lebensabschnitten in beiden Geschlechtern und andererseits in beiden Gruppen ein höheres Manifestationsalter bei den männlichen TTP-Patienten. Auch bei diesen Beobachtungen könnten die bereits erwähnten hormonellen Einflüsse dafür verantwortlich gemacht werden. Der Median des Body-Mass-Index als Parameter des Ernährungszustandes betrug 25,9 (n=63). Bei den 46 weiblichen Patienten war der Median des BMI 26,0 und bei den 17 männlichen Patienten ergab sich ein Median von 25,6. Verglichen mit Daten der deutschen Normalbevölkerung kann in dieser Arbeit von einer geringfügig höheren präadipösen Patientengruppe ausgegangen werden. Inwieweit dies tatsächlich als Risikofaktor einzuschätzen ist, muss Gegenstand von Studien mit größeren Patientenzahlen sein. Im direkten Vergleich der Rezidivpatienten und der Patienten ohne Rezidiv zeigt sich eine erhöhter BMI bei Rezidivpatienten. Inwieweit eine vorbestehende Adipositas prädisponierend für eine TTP wirkt, müsste auch hier Gegenstand von Studien mit größeren Patientenzahlen sein. Unter den in dieser Arbeit untersuchten Triggerfaktoren konnten nur wenige Medikamente tatsächlich als Auslösefaktoren für eine TTP identifiziert werden. Darunter befanden sich Ticlopidin und Chinin. Andere genannte Medikamente konnten trotz intensiver Literaturrecherche nicht mit dem Auftreten einer TTP in Zusammenhang gebracht werden, zumal die Einnahme der Präparate nur bei jeweils einer Person erfolgte, und somit keine validen Aussagen getroffen werden können. Hier sollten Studien mit größeren Patientenzahlen folgen. Entzündliche Erkrankungen konnten, wie auch in der Literatur beschrieben, in dieser Arbeit als potentielle Triggerfaktoren gezeigt werden. Mehr als die Hälfte der untersuchten Patienten wiesen entweder einen grippalen oder gastrointestinalen Infekt auf. Eine weitere auffallende Triggerfaktorengruppe stellten östrogen- und/oder gestagenhaltige Hormonpräparate da, welche von 38,8% (19/49) der weiblichen Patienten angewandt wurden. Es konnte auch eine Assoziation von TTP mit dem Auftreten einer Schwangerschaft gesehen werden. Somit konnten bei insgesamt 53,1% der befragten Frauen (26/49) hormonelle Regulationsmechanismen als potentielle Auslösefaktoren verantwortlich gemacht werden. Im direkten Vergleich von Rezidivpatienten und Patienten ohne Rezidiv konnten keine relevanten Unterschiede hinsichtlich bestimmter Triggerfaktoren festgestellt werden. In dem untersuchten Patientenkollektiv wiesen 76,1% der Personen (51/67) eine neurologische Symptomatik auf. Es dominierten Cephalgie, Parästhesien, Paresen und Sprachstörungen bei den genannten Symptomen. Hinsichtlich der geschlechtsspezifischen Verteilung traten zum Teil unterschiedliche Verteilungsmuster auf, Ursachen hierfür konnten aufgrund der geringen Patientenzahl und der zum Teil retrospektiven Datenerhebung nicht festgestellt werden. Auch bei dem Vergleich der Rezidivpatientengruppe und der Gruppe der Nicht-Rezidivpatienten traten keine relevanten Unterschiede auf. Sonstige häufig gefundene Symptome waren Petechien, Hämatome, diverse Blutungserscheinungen (z.B. Epistaxis, Hypermenorrhoe oder Zahnfleischblutungen) und Allgemeinsymptome wie Blässe, Müdigkeit und Fieber. Auch hier fanden sich keine relevanten Unterschiede hinsichtlich der geschlechtsspezifischen Verteilung oder der Verteilung auf Rezidivpatienten und Patienten ohne Rezidiv. Als bestehende Grund- oder Begleiterkrankungen konnten als potentielle prädisponierende Faktoren bestehende Autoimmunerkrankungen, insbesondere ein SLE, diverse maligne Erkrankungen, eine gesteigerte Infektneigung und evt. eine bestehende Adipositas gesehen werden. Geschlechtsspezifische Unterschiede fanden wir nicht. Hinsichtlich der Differenzierung zwischen Rezidivpatienten und Nicht-Rezidivpatienten konnte eine gesteigerte Infekt- und Allergieneigung bei Patienten der letzteren Gruppe festgestellt werden. Des Weiteren traten maligne Erkrankungen in der Rezidivpatientengruppe häufiger auf. Bei Erstmanifestation betrieben 55,2% der Patienten (37/67) einen Nikotinabusus, und übertrafen somit deutlich den Anteil der rauchenden deutschen Gesamtbevölkerung. Ob das Rauchverhalten mit einem Auftreten einer TTP in Zusammenhang gebracht werden kann sollte Gegenstand von weiteren Untersuchungen sein. Geschlechtsspezifische Unterschiede fanden sich nicht. Rezidivpatienten hatten einen höheren Nikotinabusus als Nicht-Rezidivpatienten. Ein Zusammenhang zwischen Alkoholgenuss und TTP konnten in dem untersuchten Patientenkollektiv nicht gesehen werden, wobei Menge und Art des Alkohols in unserer Studie nicht genau differenziert wurden. Alle 67 Patienten dieser Arbeit erhielten FFP, 64 von 67 wurden mit eine Plasmapherese und 3 von 67 nur mit Plasmainfusion behandelt. Die Verträglichkeit der Plasmasubstitution war bei 91,0% der Patienten gut. Es gab keine nennenswerten Komplikationen. Die Katheter- / Shuntunverträglichkeit war bei ca. 75% der Patienten komplikationslos. Insgesamt erhielten 59,7% der Personen additiv Glucocorticoide. Immunsuppressiva wie z.B. Vincristin, Rituximab und Andere wurden bei insgesamt 46,3% der Befragten (31/67) angewandt. Weitere Therapieoptionen wurden nur bei einer sehr kleinen Patientenzahl angewandt, daher ergaben sich keine relevanten unterschiedlichen Untersuchungsbefunde. Hinsichtlich Geschlecht und Rezidivverhalten fanden sich keine größeren Unterschiede. 7,5% der befragten Patienten (5/67) litten an einer hereditären Form der TTP. Auffällig war, dass beide männliche Patienten relativ früh an TTP erkrankten, wohingegen die weiblichen Patienten deutlich später ihre Erstmanifestation erlitten. Es gab bezüglich der Triggerfaktoren, der Symptome, der Grundleiden, der Therapie und anderer Vergleiche keine Unterschiede zum restlichen Patientenkollektiv Die Rezidivhäufigkeit lag in dem befragten Patientenkollektiv bei 64,2% (43/67). Es fanden sich keine geschlechtsspezifischen Unterschiede. Die Mehrzahl der Patienten erlitten ein oder zwei Rezidive. Es zeigte sich in dieser Arbeit, dass Männer evt. zu mehr Rezidiven neigen. Von den 67 befragten Patienten konnten insgesamt 54 Fälle aufgrund einer ausreichend guten Dokumentation zur Untersuchung der rezidivfreien Intervalle herangezogen werden. Zwischen den TTP-Episoden lagen mindestens 4 Wochen, um eine Exazerbation der Erkrankung ausschließen zu können. Es blieben von dem untersuchten Patientengut 44,4 % (24/54) rezidivfrei. Der Median der Frauen mit 19 Monaten und der Median der Männer mit 26 Monaten unterschieden sich deutlich. Auch die Intervalle waren verschieden. Bei den weiblichen Patienten existierte eine deutlich längere Spannweite zwischen den einzelnen Rezidiven als bei den männlichen Patienten.

Orthopoxviruses are large DNA viruses that replicate within the cytoplasm of infected cells encoding over a hundred different proteins. The orthopoxviral 68k ankyrin‐like protein (68k‐ank) is highly conserved among orthopoxviruses, and this study aimed at elucidating the function of 68k‐ank. The 68k‐ank protein is composed of four ankyrin repeats (ANK) and an F‐box‐like domain; both motifs are known proteinprotein interaction domains. The F‐box is found in cellular F‐box proteins (FBP), crucial components of cellular E3 ubiquitin (Ub) ligases. With yeast‐two‐hybrid screens and subsequent co‐immunoprecipitation analyses, it was possible to identify S‐phase kinase‐associated protein 1a (Skp1a) as a cellular counterpart of 68k‐ank via binding to the F‐box‐like domain. Additionally, Cullin‐1 was co‐precipitated, suggesting the formation of a viral‐cellular SCF E3 Ub ligase complex. Modified Vaccinia virus Ankara (MVA) ‐ being attenuated and unable to replicate in most mammalian cell lines due to a block in morphogenesis – nevertheless, expresses its complete genetic information attributing to its properties as promising vector vaccine. Conservation of 68k‐ank as the only ANK protein encoded by MVA implied a substantial role of this viral factor. Hence, its function in the viral life cycle was assessed by studying a 68k‐ank knock‐out MVA. A mutant phenotype manifested in nonpermissive mammalian cells characterized by a block succeeding viral early gene expression and by a reduced ability of the virus to shutoff host protein synthesis. Studies with MVA encoding a 68k‐ank F‐box‐like domain truncated protein revealed that viral‐cellular SCF complex formation and maintenance of viral gene expression are two distinct, unrelated functions fulfilled by 68k‐ank. Moreover, K1, a well‐described VACV host range factor of the ANK protein family, is able to complement 68k‐ank function. This suggests that gene expression of MVA putatively depends on the ANKs encoded in 68k‐ank. In addition to the important findings in vitro, first virulence studies with the mouse pox agent, ectromelia virus (ECTV) deleted of the 68k‐ank ortholog (C11) suggested that this factor contributes to ECTV virulence in vivo.
Naphthalan ist ein natürliches mineralisches Schweröl mit einem hohen Anteil an gesättigten bi- bis pentazyklischen Kohlenwasserstoffen (KW). Es wird heutzutage erfolgreich in der Behandlung der Psoriasis, einer hyperproliferativen Hauterkrankung mit gleichzeitig mangelhafter terminaler Differenzierung der Keratinozyten eingesetzt. Seine Wirkungsweise ist bisher aber nicht bekannt. Ziel der vorliegenden Arbeit war es, den Einfluss von Naphthalan auf die Proliferation, Differenzierung und Toxizität von humanen Keratinozyten zu untersuchen und ggf. Rückschlüsse auf einen zu Grunde liegenden Wirkmechanismus zu ziehen. Für unsere Untersuchungen wurden zwei Fraktionen von Naphthalan verwendet: 1. eine durch Destillationsprozesse teilfraktionierte, bräunlich Subfraktion (NB) mit einem im Vergleich zur Ausgangssubstanz hohen Anteil an oligozyklischen gesättigten KW und einem sehr geringen Anteil an Aromaten und polaren KW. 2. eine farblose Subfraktion (NW), aus der die oligozyklischen gesättigten KW weitgehend entfernt wurden. Als Vergleich diente ein herkömmliches mineralisches Schweröl (MO). Die Effekte wurden an einer immortalisierten Zelllinie adulter humaner Keratinozyten (HaCaT) und an primären humanen Keratinozyten, die aus der Haut von Reduktionsplastiken isoliert wurden, untersucht, und zwar zeit- (24-72h) und konzentrationsabhängig (NB 0,001-1%; NW 0,001-1%, MO 0,01-10%). Die Untersuchung der Zellproliferation erfolgte mittels BrdU-ELISA, der Zytotoxizität mittels Trypanblau-Vitalfärbung/WST-1-Test und der Zelldifferenzierung mittels Durchflusszytometrie sowie mittels immunhistochemischer Färbung von Hautbiopsien von zuvor mit Naphthalan behandelten Patienten. Involucrin u. Cytokeratin 10/11 dienten als Zielantigene zur Untersuchung der Differenzierung. Für HaCaT- und primäre humane Keratinozyten ergab sich ein dosis- und zeitabhängiger Effekt von Naphthalan auf die Hemmung der Zellproliferation. Die max. Hemmung nach 48h betrug 89.82% (NB 1%) für HaCaT- und 71.62% (NB 1%) für primäre humane Keratinozyten (p􀂔0,0001). Bezüglich der Viabilität ergab sich im Trypanblau-Test lediglich nach prolongierter Inkubationsdauer von 48h unter NB 1% ein schwach signifikanter Effekt mit Nachweis von 14,68% avitalen HaCaT-Keratinozyten bzw. 9,54% avitalen primären humanen Keratinozyten. Hinsichtlich der Induktion der Zelldifferenzierung zeigten sich ebenfalls dosis- und zeitabhängige Effekte. Für den „frühen“ Differenzierungsmarker Cytokeratin 10/11 ergab sich für HaCaT-Keratinozyten ein max. Anstieg der Expression um 326,1% (NB 1%) und für primäre humane Keratinozyten um 264,18% (NB 1%) (p􀂔0,0002). Für den „späten“ Differenzierungsmarker Involucrin ergab sich erst nach einer prolongierten Inkubationsdauer von 48h ein signifikanter max. Anstieg der Expression um 4303,5% (NB 1%) für HaCaT-Keratinozyten und von 657,48% (NB 1%) für primäre humane Keratinozyten (p􀂔0,0002). Klinisch zeigte sich bei Patienten mit langjähriger therapierefraktärer Psoriasis nach 3-wöchiger topischer Naphthalan-Ganzkörpertherapie (1x/die 12min.) an einem Kollektiv von 5 bzw. 20 Patienten eine mittlere Reduktion des PASI-Scores von 20,2 auf 4,7 bzw. von 23,5 auf 8,3. Immunhistologisch zeigte sich eine Abnahme des Proliferationsindex von 29,3% auf 7,1%; die terminale Induktion der Keratinozytendifferenzierung konnte an der Zunahme der Cytokeratin 10/11- und Involucrin-Expression bestätigt werden. Histologisch zeigte sich eine weitgehende Normalisierung der durch Hyperproliferation, Akanthose, Hyper-/Parakeratose, Angiogenese, Vasodilatation und mononukleäre Zellinfiltration i.R. der Psoriasis gestörten Hautarchitektur. Zusammenfassend führt NB zu einer konzentrations- und zeitabhängigen hochsignifikanten Proliferationshemmung von HaCaT- und primären humanen Keratinozyten. Sie beruht nicht auf einem zytotoxischen Effekt auf die Zellen. Die Wirkung ist abhängig von den spezifischen Bestandteilen von NB, insbesondere den oligozyklischen gesättigten KW, i.e. Sterane und Hopane, die in NW und herkömmlichem Schweröl fast bzw. gar nicht vorkommen. NB führt weiterhin zu einer hochsignifikanten Zunahme der Cytokeratin 10/11- und Involucrin-Expression, zweier Markerproteine der terminalen Keratinozytendifferenzierung. Somit liegt die Ursache der Proliferationsinhibition und der klinischen Wirksamkeit von Naphthalan in der Induktion der terminalen Zelldifferenzierung. Aufgrund der einfachen topischen Anwendungsmöglichkeit, guter Verträglichkeit bei bisher nicht nachgewiesenen systemischen Nebenwirkungen, der erfolgreichen Anwendung auch in bisher therapierefraktären Fällen und aufgrund ökonomischer Betrachtungen erscheint Naphthalan als eine mögliche Therapieoption in der Behandlung der Psoriasis. Dies sollte in größeren, kontrollierten klinischen Studien überprüft werden. Gaschromatographische und massenspektrometrische Untersuchungen zeigten eine cluster-förmige Anhäufung von oligozyklischen KW aus der Gruppe der Sterane und Hopane. Der Vergleich mit strukturanalogen Substanzen (u.a. Vitamin D) legt einen Wirkmechanismus über eine MAP-Kinase-Kaskade vermittelte Modulation der Expression von Differenzierungsmarkern (Involucrin, Cytokeratin 10/11) über verschiedene Transkriptionsfaktoren, i.e. AP-1/-2, C/EBP, nahe. Hier sind zur genauen Klärung weitere Untersuchungen notwendig. Durch weitere Subfraktionierung könnte vielleicht das aktive Substrat einer möglicherweise neuen Substanzklasse isoliert werden.
In this work we present the setup and first tests of our new BIO IN detector. This detector is designed to classify atmospheric ice nuclei (IN) for their biological content. Biological material is identified via its auto-fluorescence (intrinsic fluorescence) after irradiation with UV radiation. Ice nuclei are key substances for precipitation development via the Bergeron–Findeisen process. The level of scientific knowledge regarding origin and climatology (temporal and spatial distribution) of IN is very low. Some biological material is known to be active as IN even at relatively high temperatures of up to –2°C (e.g. pseudomonas syringae bacteria). These biological IN could have a strong influence on the formation of clouds and precipitation. We have designed the new BIO IN sensor to analyze the abundance of IN of biological origin. The instrument will be flown on one of the first missions of the new German research aircraft ''HALO'' (High Altitude and LOng Range).
In the last decade, several sophisticated and accurate imaging methods such as positron emission tomography have been developed in order to evaluate malignant potential in enlarged mediastinal lymph nodes. This case illustrates an unusual presentation of sarcoidosis that mimicked lymphatic metastases of non small cell lung carcinoma. The reported high specificity and sensitivity of positron emission tomography-Computer Tomography regarding mediastinal staging could lead in same cases of false positives to a delaying of stage adapted therapy of non small cell lung carcinoma, showing that despite the recent advances of imaging techniques, such as positron emission tomography-computer tomography, several limitations of this imaging technique are still existing.
Im adulten Säugerhirn findet Neurogenese in der SVZ der Seitenventrikel kontinuierlich statt. Eine Vielzahl von Signalsystemen steuert in komplexer Weise zelluläre Antworten und reguliert die Proliferation, Differenzierug und Wanderung NSZ. Gegenwärtig ist nur wenig über die zugrundeliegenden Signalwege bekannt. Zunehmend gibt es Hinweise darauf, dass Nukleotide an diesen Prozessen beteiligt sind. Frühere Untersuchungen unserer Arbeitsgruppe zeigten, das die Nukleotide ADPbetaS und UTP in kultivierten NSZ der adulten SVZ einen schnellen Kalziumeinstrom induzieren und die Wachstumsfaktor-vermittelte NSZ-Proliferation steigern. In der vorliegenden Arbeit wurde ein System zur Kultivierung adhärenter adulter NSZ etabliert. Die Untersuchungen zeigen, dass adulte NSZ eine Vielzahl an P2Y- und P2X-Rezeptoren, sowie die Nukleotid-hydrolysierenden Enzyme NTPDase2 und TNAP exprimieren. Untersuchungen der ADPbetaS-, UTP- und EGF-vermittelten Signalwege zeigen, dass alle drei Agonisten eine ERK1/2- und CREB-Phosphorylierung induzieren, wobei sich die zeitlichen Charakteristika zwischen den Nukleotiden und EGF unterscheiden. Inhibierungsexperimente geben Einblicke in die dabei aktivierten Signalkaskaden und weisen auf eine ADPbetaS-induzierte Transaktivierung des EGF-Rezeptors hin. Während UTP über den P2Y2-Rezeptor wirkt, übt ADPbetaS seine Funktion über den P2Y1- und P2Y13-Rezeptor aus. Die Daten implizieren zudem, dass Nukleotide und EGF gleiche Zielproteine über verschiedene Signalwege induzieren und dass sie das Potenzial besitzen, bei der Kontrolle der Zellproliferation in der adulten Neurogenese synergistisch zu agieren. Vergleichende Analysen mit kultivierten NSZ aus Wildtyp-, P2Y1- und P2Y2-Rezeptor-Knockout-Mäusen belegen ein verändertes Antwortverhalten in Gegenwart von ADPbetaS, UTP und EGF und lassen kompensatorische Mechanismen vermuten. Die Resultate dieser Arbeit demonstrieren zudem, dass ATP, ADPbetaS, UTP und EGF die Migration von NSZ induzieren. Parallel dazu konnten Veränderungen des Aktinzytoskelletes, wie die Zunahme an F-Aktin, die Bildung von Stressfasern und eine Veränderung der Zellmorphologie gezeigt werden. Diese Prozesse gehen mit einer Aktivierung der Proteinkinasen Akt und FAK einher. Die Daten weisen darauf hin, dass Nukleotide und EGF für die Zytoarchitektur der SVZ und die Wanderung von Neuroblasten zum OB eine wichtige Rollen spielen könnten.
The transporter associated with antigen processing-like (TAPL) acts as a lysosomal ATP-dependent polypeptide transporter with broad length selectivity. To characterize in detail its substrate specificity, a procedure for solubilization, purification and functional reconstitution of human TAPL was developed. TAPL was expressed in Sf9 insect cells with the baculovirus expression system and solubilized from crude membranes. By intensive screening of detergents, the mild non-ionic detergents digitonin and dodecylmaltoside were found to be ideal for solubilization with respect to efficiency, long term stability, and functionality of TAPL. TAPL was isolated in a two-step procedure with a yield of 500 micro g/L cell culture and, subsequently, reconstituted into proteoliposomes. The KM(pep) for the peptide RRYCfKSTEL (f refers to fluorescence label) and KM(ATP) were determined to be 10.5 ± 2.3 micro M and 97.6 ± 27.5 micro M, respectively, which are in the same range as the Michaelis-Menten constants determined in the membranes. The peptide transport activity of the reconstituted TAPL strongly depends on the lipid composition. Interestingly, the E. coli lipids are prefered over other tested natural lipids extracts. Moreover, phosphatidylcholine, the most abundant phospholipid in eukaryotic cells influenced TAPL activity in a dose dependent manner. In addition, some negatively charged lipids like DOPA and DOPS increased peptide transport activity with preference for DOPS. However, DOPE or egg PG which are also negatively charged had no effect. It seems not only the charge but also the specific head group of phospholipids that has impact on the function of TAPL. With the help of combinatorial peptide libraries containing D-amino acid residues at defined positions as well as bulky fluorescein labeled peptides, the key positions of the peptides were localized to the N- and C-terminal residues with respect to peptide transport. The C-terminal position has the strongest selectivity since modification at this position shows strongest impact on peptide transport. Additionally, positions 2 and 3 of the peptide also have weak influence on peptide selectivity. Subsequently, the residue preferences at the key positions were systematically investigated by combinatorial peptide libraries with defined residues at certain positions. At both ends, TAPL favors positively charged, aromatic, or hydrophobic residues and disfavors negatively charged residues as well as asparagine and methionine. The residue preferences at the key positions are valid for peptide substrates with different length, indicating a general rule for TAPL selectivity. Besides specific interactions of both terminal residues, electrostatic interactions are important, since peptides with positive net charge are more efficiently transported than negatively charged ones. By size exclusion chromatography (SEC) and blue native PAGE, TAPL purified in the presence of digitonin or dodecylmaltoside had an apparent molecular weight of 200 kDa which is close to the theoretical molecular mass of the TAPL homodimer (172 kDa). The purified and reconstituted TAPL showed specific ATP hydrolysis activity which can be inhibited by orthovanadate. TAPL in proteoliposomes showed 6-fold higher ATP hydrolysis than digitonin solubilized protein, indicating the phospholipids impact on TAPL function. However, no peptide substrate stimulated ATPase activity was observed. For site-specific labeling of TAPL, eight cysteines in each half transporter were replaced by alanine or valine. The TAPL cys-less mutant showed the same peptide transport activity as TAPL wt. Based on the functional TAPL cys-less mutant, seven single cysteine mutants were introduced into strategic positions. All single cysteine mutants in the TMD did not influence peptide transport, whereas the mutant L701C, which is close to the conserved H-loop motif, displayed impaired transport. TAPL orthologs Haf-4 and Haf-9 from Caenorhabditis elegans possess around 40% sequence identities with TAPL and 50% with each other. Both proteins are putative half transporters and reported to be involved in the intestinal granule formation (Bauer, 2006; Kawai et al., 2009). To further understand the physiological functions of these two proteins, they were expressed in Sf9 insect cells. Haf-4 and Haf-9 showed weak but specific ATP- and peptide-dependent peptide transport activity for the given peptide RRYCfKSTEL. Therefore, it was proposed that the physiological roles for Haf-4 and Haf-9 might be related to their peptide transport activity. Besides forming functional homodimeric complex as estimated by the peptide transport activities, both half transporter could also form heteromers which was confirmed by coimmunoprecipitation. However, the heteromers showed decreased transport activity.
The Invertebrate section of the Museum of Zoology QCAZ at the Pontifical Catholic University of Ecuador in Quito maintains nearly two million curated specimens, and comprises Ecuador's largest collection of native taxa. We review 1902 type specimens from 6 subspecies and 320 species in 121 genera and 42 families, currently kept in the Museum. The list includes 116 holotypes, 10 allotypes, 1774 paratypes and 2 neoparatypes. The collection of type specimens is particularly strong in the Coleoptera (family Carabidae and Staphylinidae) and Hymenoptera. However, other insect orders such as Diptera and Lepidoptera and non-insect arthropods such as Acari, Aranea and Scorpiones, are moderately represented in the collection. This report provides original data from labels of every type specimen record. An analysis of the geographic distribution of type localities showed that collection sites are clustered geographically with most of them found. towards the northern region of Ecuador, in Pichincha, Cotopaxi and Napo provinces. Sites are mainly located in highly accessible areas near highways and towns. Localities with a high number of type species include the cloud forest reserve Bosque Integral Otonga and Parque Nacional Yasunf in the Amazon rainforest near PUCE's Yasuni Scientific Station. Type localities are not well represented in the Ecuadorian National System of Protected Areas. Future fieldwork Sllould include. localities in the southern region of Ecuador but also target less accessible areas not located near highways or towns. We discuss the value of the collection as a source of information for conservation and biodiversity policies in Ecuador.

Introduction: Lymphocyte infiltration (LI) is often seen in breast cancer but its importance remains controversial. A positive correlation of human epidermal growth factor receptor 2 (HER2) amplification and LI has been described, which was associated with a more favorable outcome. However, specific lymphocytes might also promote tumor progression by shifting the cytokine milieu in the tumor.
Methods: Affymetrix HG-U133A microarray data of 1,781 primary breast cancer samples from 12 datasets were included. The correlation of immune system-related metagenes with different immune cells, clinical parameters, and survival was analyzed.
Results: A large cluster of nearly 600 genes with functions in immune cells was consistently obtained in all datasets. Seven robust metagenes from this cluster can act as surrogate markers for the amount of different immune cell types in the breast cancer sample. An IgG metagene as a marker for B cells had no significant prognostic value. In contrast, a strong positive prognostic value for the T-cell surrogate marker (lymphocyte-specific kinase (LCK) metagene) was observed among all estrogen receptor (ER)-negative tumors and those ER-positive tumors with a HER2 overexpression. Moreover ER-negative tumors with high expression of both IgG and LCK metagenes seem to respond better to neoadjuvant chemotherapy.
Conclusions: Precise definitions of the specific subtypes of immune cells in the tumor can be accomplished from microarray data. These surrogate markers define subgroups of tumors with different prognosis. Importantly, all known prognostic gene signatures uniformly assign poor prognosis to all ER-negative tumors. In contrast, the LCK metagene actually separates the ER-negative group into better or worse prognosis.

In over 100 genera of tropical angiosperms, one or more species possess specialised structures for housing ants. The longevity and intimacy of these associations has often facilitated an increasing specialisation of both the ants and the plants, leading to a number of highly specific and obligate symbioses. Early literature contained only few anecdotal reports of the ant genus Cladomyrma WHEELER inhabiting (unidentified) plants. This work presents the new findings on Cladomyrma and its host plants that accumulated over the last two decades. My studies of Cladomyrma reveal that there is a largely overlooked community of south-east Asian plant-ants and their associated plants. Currently the genus consists of at least 12 species. Cladomyrma has been thought to be restricted to the ever-wet part of the West Malesian floristic region, comprising the Malay Peninsula, Borneo, and Sumatra, but recent collections from Thailand and Vietnam indicate that species of the genus penetrate the seasonal tropical forests of Continental Asia. Cladomyrma inhabits 24 plant species belonging to a surprisingly extensive range of plant taxa: Callerya, Saraca, Spatholobus (Fabaceae), Crypteronia (Crypteroniaceae), Drypetes (Putranjivaceae), Ryparosa (Achariaceae), Strychnos (Loganiaceae), Neonauclea (Rubiaceae), Luvunga (Rutaceae) and Sphenodesme (Verbenaceae). In terms of taxonomic diversity on the genus and family level the range of hosts utilised by Cladomyrma is one of the broadest ever recorded for any live stem-nesting plant-ant lineage worldwide. This work provides a species-level overview of all Cladomyrma host plants known from Borneo, the Malay Peninsula and Sumatra, including descriptions of ant-housing structures (domatia), ant inhabitant identity, onset of colonisation during plant ontogeny, nest structure, occupancy rate, and considerations of results obtained from herbarium specimens. Both the regularity of ant association and the degree of morphological specialisation toward myrmecophytism are assessed. The behavioural traits of Cladomyrma are compatible with traits exhibited by other protective plant-ants. This work demonstrates that all species of Cladomyrma investigated (dianeae, maschwitzi, yongi, petalae) confer antiherbivore protection to young leaves of its host. The ants also attack and repell or kill herbivorous insect larvae encountered on young foliage. Cleaning behaviour appears to be a trait shared by all members of the genus, and the two species tested (maschwitzi, petalae) successfully removed termite eggs experimentally placed onto young leaves. Another trait common to all known species of the genus is that the ants preferentially patrol young shoots and leaves ('neophily'). These behavioural traits of Cladomyrma likely reduce stem damage and pathogenic infection of their host. The ants prune encroaching vegetation (tested in dianeae maschwitzi, petalae, yongi, observed in crypteroniae) and attack paper tape used to mark host plants (observed in andrei, dianeae, hobbyi, nudidorsalis, maschwitzi, yongi, petalae). If these traits combined translate into a better reproductive success of the hosts has yet to be verified. Evidence for lifetime fitness benefits is particularly difficult to quantify for the long-lived woody host plants of Cladomyrma. The predominant food source of Cladomyrma appears to be the honeydew of scale insects (Coccidae and Pseudococcidae) which the ants tend inside their nest cavities. Observations on scale insect acquisition by Cladomyrma foundress queens show that hemipteran trophobionts are not transported by the queens on their nuptial flight but they nevertheless arrive on the host plant independently of the ants. Entry into nest chambers is facilitated by small holes kept open by the foundress queen. Most Cladomyrma species have been recorded from only one or two (three) host plant species (andrei, crypteroniae, hobbyi, maschwitzi, nudidorsalis, scopulosa, yongi), but two species, Cladomyrma petalae and C. dianeae, are more catholic in their host usage; the first being a 'generalist' plant-ant colonising hosts across a broad taxonomic range, the second inhabiting several members of the genus Neonauclea. First results of host-choice experiments with C. petalae are presented and the potential mechanisms promoting host specificity are discussed. My studies of the Cladomyrma/plant associations indicate that codiversification and host shifts or host expansions, rather than cospeciation, shape the pattern of species interactions in this system. Finally, I propose a scenario in which three key traits of Cladomyrma –access to live stems, utilisation of indirect food rewards via trophobionts and 'neophily'– are hypothesised to favour niche differentiation and the acquisition of new hosts over evolutionary time.
Anaphylactic shock is a severe allergic reaction involving multiple organs including the bronchial and cardiovascular system. Most anaphylactic mediators, like platelet-activating factor (PAF), histamine, and others, act through G protein – coupled receptors, which are linked to the heterotrimeric G proteins Gq /G 11 , G12/G13 , and Gi . The role of downstream signaling pathways activated by anaphylactic mediators in defi ned organs during anaphylactic reactions is largely unknown. Using genetic mouse models that allow for the conditional abrogation of G q /G 11 - and G 12 /G 13 -mediated signaling pathways by inducible Cre/loxP-mediated mutagenesis in endothelial cells (ECs), we show that Gq /G11 -mediated signaling in ECs is required for the opening of the endothelial barrier and the stimulation of nitric oxide formation by various infl ammatory mediators as well as by local anaphylaxis. The systemic effects of anaphylactic mediators like histamine and PAF, but not of bacterial lipopolysaccharide (LPS), are blunted in mice with endothelial G alpha q/G alpha 11 deficiency. Mice with endothelium-specific G alpha q /G alpha 11 deficiency, but not with G alpha 12/G alpha 13 deficiency, are protected against the fatal consequences of passive and active systemic anaphylaxis. This identifies endothelial Gq/G11 -mediated signaling as a critical mediator of fatal systemic anaphylaxis and, hence, as a potential new target to prevent or treat anaphylactic reactions.
Changes in the balance of cholesterol absorption and synthesis and moderately elevated plasma plant sterols have been suggested to be atherogenic. Measuring cholestanol, lathosterol, campesterol, and sitosterol, we investigated the relationships of cholesterol metabolism and plasma plant sterols with the severity of coronary artery disease (CAD) in 2,440 participants of the Ludwigshafen Risk and Cardiovascular health (LURIC) study. The coronary status was determined by angiography, and the severity of CAD was assessed by the Friesinger Score (FS). An increase in the ratio of cholestanol to cholesterol was associated with high FS (P = 0.006). In contrast, a high ratio of lathosterol to cholesterol went in parallel with low FS (P < 0.001). Whereas the campesterol to cholesterol ratio significantly correlated with the FS (P = 0.026), the relationship of the sitosterol to cholesterol ratio with the FS did not reach statistical significance in the whole group. Increased campesterol, sitosterol, and cholestanol to lathosterol ratios were associated high FS (P < 0.001). To conclude, there is a modest association of high cholesterol absorption and low cholesterol synthesis with an increased severity of CAD. An atherogenic role of plasma plant sterols themselves, however, seems unlikely in subjects without sitosterolaemia.
We developed a Monte Carlo event generator for production of nucleon configurations in complex nuclei consistently including effects of nucleon–nucleon (NN) correlations. Our approach is based on the Metropolis search for configurations satisfying essential constraints imposed by short- and long-range NN correlations, guided by the findings of realistic calculations of one- and two-body densities for medium-heavy nuclei. The produced event generator can be used for Monte Carlo (MC) studies of pA and AA collisions. We perform several tests of consistency of the code and comparison with previous models, in the case of high energy proton–nucleus scattering on an event-by-event basis, using nucleus configurations produced by our code and Glauber multiple scattering theory both for the uncorrelated and the correlated configurations; fluctuations of the average number of collisions are shown to be affected considerably by the introduction of NN correlations in the target nucleus. We also use the generator to estimate maximal possible gluon nuclear shadowing in a simple geometric model.
Based on a study of the characters used to define it, the genus Euphoriopsis Casey is placed in synonymy with Euphoria Burmeister. Euphoria punicea Janson is placed in synonymy with Euphoria steinheili Janson. Morphological characters supporting the synonymies, and the species' distributions and biological data are reviewed.
In plants, a family of more than 20 heat stress transcription factors (Hsf) controls the expression of heat stress (hs) genes. There is increasing evidence for the functional diversification between individual members of the Hsf family fulfilling distinct roles in response to various environmental stress conditions and developmental signals. In response to hs, accumulation of both heat stress proteins (Hsp) and Hsfs is induced. In tomato, the physical interaction between the constitutively expressed HsfA1 and the hs-inducible HsfA2 results in synergistic transcriptional activation (superactivation) of hs gene expression. Here, we show that the interaction is strikingly specific and not observed with other class A Hsfs. Hetero-oligomerization of the two-component Hsfs is preferred to homo-oligomerization, and each Hsf in the HsfA1/HsfA2 hetero-oligomeric complex has its characteristic contribution to its function as superactivator. Distinct regions of the oligomerization domain are responsible for specific homo- and hetero-oligomeric interactions leading to the formation of hexameric complexes. The results are summarized in a model of assembly and function of HsfA1/A2 superactivator complexes in hs gene regulation.
Immunsuppressiva sind essentiell für die allogene Organtransplantation. Graff et al konnten in drei vorangegangenen Studien zeigen, dass Immunsuppressiva zu einer verstärkten Thrombozytenaktivierung führen. Diese ist für die verschiedenen Substanzen unterschiedlich stark ausgeprägt. Eine verstärkte Thrombozytenaktivierung könnte zu vermehrten kardiovaskulären Ereignissen führen. Insbesondere bei nierentransplantierten Patienten ist das kardiovaskuläre Risiko ohnehin deutlich erhöht. Es stellt sich die Frage ob diese medikamentös induzierte Plättchenaktivierung als Ko-Faktor kardiovaskulärer Ereignisse bei nierentransplantierten anzusehen ist. Im Vorfeld dieser Studie wurden in den Studien NTX, RAPA und PLANT bei insgesamt 176 nierentransplantierten Patienten mit einer Mono-Immunsuppression flowzytometrische Plättchenfunktionsmessungen durchgeführt, dabei wurden insbesondere der Degranulationsmarker CD62 und der aktivierte GP IIb/IIIa-Rezeptor PAC-1 gemessen. Diese Ergebnisse wurden in Relation zum Mittelwert der jeweiligen Kontrollgruppen gesetzt: Messwert(Patient) / Mittelwert(Gesunde_Kontrolle) Diese relativen Plättchenfunktionswerte waren zwischen den Studien vergleichbar. Des Weiteren wurden epidemiologische Eckdaten und klinische Endpunkte bis zu 5 Jahre nach der Thrombozytenmessung erfasst, im Mittel 3,7 Jahre. Aufgrund des Studiendesigns der vorangegangenen Studien wurde hier ein Kollektiv mit relativ niedrigem kardiovaskulärem Risiko untersucht, jedoch sollte durch diese Reduktion klassischer Risikofaktoren der Einfluss der Plättchenaktivität besser sichtbar sein. Die retrospektive Analyse untersuchte ob die Plättchenfunktionsparameter CD62 und PAC-1 als prädiktiver Marker für kardiovaskuläre Ereignisse geeignet sind. Insgesamt traten seit den Messungen 21 kardiovaskuläre Ereignisse auf, davon 9 akute. Es gab keine signifikanten Unterschiede zwischen den Werten bei Patienten mit Ereignissen und denen ohne Ereignis: Bei CD62 (baseline) lag der Mittelwert bei 2,82±1,9 mit Ereignis vs. 2,73±1,47 ohne, für PAC-1 bei 1,76 ±1,97 vs. 1,93±1,53. Nach Aktivierung mit TRAP lag der Mittelwert für CD62 bei Ereignispatienten bei 1,57±0,88 vs. 1,76±0,83, für PAC-1 bei 1,56±1,65 vs. 1,44±1,04. In einem weiteren Schritt wurden die Patienten nach dem Ausmaß der Plättchenaktivierung klassifiziert, als Cut-Off-Werte wurden die Mittelwerte und Mediane der jeweiligen Plättchenfunktion aller Patienten gewählt, ferner noch willkürlich eine 3- bzw 5-fache Aktivierung gegenüber den gesunden Kontrollen. Hierbei wurden in den Gruppen mit hohen Funktionswerten nicht signifikant mehr Ereignisse beobachtet als in den Gruppen mit niedrigen Werten. In einer daraufhin durchgeführten Time-to-event-Analyse nach Kaplan-Meyer zeigte sich allerdings für CD62 (baseline) > 5 ein früheres Eintreten kardiovaskulärer Ereignisse, im Mittel 47,6 vs. 69,9 Monate, p = 0,05. Als Risikofaktoren in dieser Studie erwiesen sich insbesondere das Alter und das Alter bei Transplantation, letzteres erwies sich in einer logistischen Regressionsanalyse als der wichtigster Faktor. Dies ist nach unserem Wissen die erste Untersuchung, die den prädiktiven Wert von Thrombozyten-gebundenem CD62 sowie von PAC-1 für kardiovaskuläre Ereignisse evaluiert. Es ergibt sich kein Hinweis, dass diese Marker einen prädiktiven Wert haben. Somit erscheint auch eine direkte pharmakologische Beeinflussung der Thrombozytenfunktion oder eine Veränderung der Immunsuppression nicht wegweisend, um die Inzidenz kardiovaskulärer Ereignisse bei nierentransplantierten Patienten zu senken.
Auswirkungen einer Mutation des Adhäsionrezeptors PSGL-1 bei humanen neutrophilen Granulozyten

Der VNTR Polymorphismus des PSGL-1 ist schon seit längerer Zeit Gegenstand wissenschaftlicher Untersuchungen. Der Rezeptor weist je nach Genotyp eine unterschiedliche Länge auf. Dabei ist der Rezeptor mit dem Allel A der längste, das C-Allel weist phänotypisch den kürzesten Rezeptor auf. Der Rezeptor mit dem B-Allel liegt mit der Länge in der Mitte der drei Rezeptoren. In dieser Studie wurde das Roll- und das Adhäsionsverhalten von Leukozyten mit den Genotypen AA bzw. BB mit Hilfe eines Flusskammermodells untersucht. Dieses Verhalten kommt im Rahmen der Bindung zustande, welche P-Selektin und PSGL-1 miteinander eingehen. Das Blut von acht Probanden wurde untersucht, vier Probanden wiesen den Genotyp AA, die anderen vier den Genotyp BB auf. Die Leukozyten wurden aus dem Vollblut isoliert, verdünnt und in das Flusskammermodell eingespeist. Die verdünnte Zellsuspension wurde mittels einer Pumpe über ein Schlauchsystem durch eine Messkammer befördert, welche im Vorfeld mit P-Selektin beschichtet wurde. Dort konnten die Zellen mikroskopisch untersucht und videotechnisch dokumentiert werden. Anschließend konnte die Rollgeschwindigkeit der neutrophilen Granulozyten und die Anzahl der adhärenten Zellen an der Oberfläche durch Computerprogramme ermittelt werden. Als Resultat ergab sich ein signifikanter Unterschied zwischen der Rollgeschwindigkeit der Zellen des Genotyps AA und der Zellen des Genotyps BB. Dabei wiesen die Zellen mit dem B-Allel wegen der geringeren Länge des PSGL-1 eine höhere Rollgeschwindigkeit auf, als die Zellen mit dem AAllel, deren PSGL-1 länger ist. Zudem blieben signifikant weniger Leukozyten mit dem BAllel an der Oberfläche haften als die Leukozyten der anderen Gruppe. Daraus lässt sich schließen, dass die Bindungskraft zwischen PSGL-1 und PSelektin von der Länge des PSGL-1 abhängig ist. Der kürzere Rezeptor (B-Allel des PSGL-1) weist also eine geringere Bindungskraft auf als der längere Rezeptor (A-Allel des PSGL-1). Die Bindung zwischen PSGL-1 und PSelektin lässt sich wahrscheinlich pharmakologisch beeinflussen. Es wurden Antikörper entwickelt, die in klinischen Studien Erfolg versprechende Ansätze bei der Entzündungshemmung gezeigt haben. Die aktuelle Literatur zu klinisch pharmakologischen Behandlungsansätzen wurde in dieser Arbeit diskutiert.
Very little is known about the occlusal wear pattern in the Neanderthal posterior dentition. Usually dental wear is closely related to the physical properties of the ingested food, and consequently can be used to obtain information about diet. Neanderthal dietary reconstructions have been mostly based on the analysis of accompanying faunal remains and isotopic signatures of bones and tooth enamel, suggesting that they exploited larger portions of animal proteins from large and medium-sized herbivores. Probably these studies may do not reflect the bulk diet, tending to underestimate plant consumption and to overestimate meat consumption. In the present work the occlusal wear pattern of maxillary molars of Homo neanderthalensis (N=19) and early Homo sapiens (N=12)have been analyzed, applying non-destructive methods based on virtual three-dimensional polygonal models generated from surface scanning of dental casts. The sample groups occupied different geographical areas at different chronological times. The 3D digital tooth models were analyzed using the “Occlusal Fingerprint Analysis” (OFA) method (Kullmer et al. 2009), describing and quantifying the occlusal wear pattern derived from two wear facet angles (dip and dip direction), wear facet area and occlusal relief index (ORI). The OFA method provides information about the dynamics of the occlusal relationships and their function, permitting the reconstruction of the mandibular movements responsible for the contacts created during the chewing cycle. Since jaw movements and diet are closely related, the results obtained, can be used to interpret the diet of the two Pleistocene hominin species. In order to evaluate how dietary differences influence the occlusal wear pattern, upper molars of modern hunter-gatherers (N=42) with known diet and different dietary habits, have been included in the sample and compared with those of Neanderthals and early Homo sapiens. Results show that within the modern hunter-gatherers sample, the occlusal wear pattern of carnivorous populations differs from those who relied on a mixed-diet. In particular, the study of relative facet areas clearly distinguish meat-eaters from mixed-diet hunter-gatherers, while ORI results and wear facet inclinations (dip angle) seem to reflect directly the abrasiveness of the diet, including the influence of exogenous materials during food preparation. The Neanderthal occlusal wear pattern is characterized by an ecogeographic variation, suggesting the exploitation of different food resources. In particular Neanderthals who inhabited relatively warm environments of southern Europe and the Near East exhibit an occlusal wear pattern different from those of meat-eaters hunter-gatherers from tempered and cooler regions, displaying some features similar to those of Bushmen. These results suggest the exploitation of a broad variety of food sources. The analysis of the occlusal wear pattern in Neanderthals and early Homo sapiens who inhabited Europe during the cooler Oxygen Isotope Stage 3 (OIS3) shows many similarities between the two hominid species. These results indicate the exploitation of similar and low-diversified food sources, based mostly on the consumption of animal proteins, as suggested through the clear similarities with the wear patterns found in modern meat-eaters hunter-gatherers. In both studied groups, Neanderthals and early Homo sapiens the occlusal wear pattern is characterized by high ORI and dip angle values, suggesting the intake of a low-abrasive diet, probably due to the absence of sophisticated food preparation techniques introducing external silica grains, e.g. from soil (grinding of seeds) or plant cells, as those, seen in modern hunter-gatherer populations. The analysis of the occlusal fingerprints in Neanderthal and early European Homo sapiens upper molars suggests that both species followed very similar adaptive dietary strategies, based on a distinctive versatility and flexibility in the daily diet, depending on availability of resources according to environmental circumstances.
The C-module-binding factor (CbfA) is a multidomain protein that belongs to the family of jumonji-type (JmjC) transcription regulators. In the social amoeba Dictyostelium discoideum, CbfA regulates gene expression during the unicellular growth phase and multicellular development. CbfA and a related D. discoideum CbfA-like protein, CbfB, share a paralogous domain arrangement that includes the JmjC domain, presumably a chromatin-remodeling activity, and two zinc finger-like (ZF) motifs. On the other hand, the CbfA and CbfB proteins have completely different carboxy-terminal domains, suggesting that the plasticity of such domains may have contributed to the adaptation of the CbfA-like transcription factors to the rapid genome evolution in the dictyostelid clade. To support this hypothesis we performed DNA microarray and real-time RT-PCR measurements and found that CbfA regulates at least 160 genes during the vegetative growth of D. discoideum cells. Functional annotation of these genes revealed that CbfA predominantly controls the expression of gene products involved in housekeeping functions, such as carbohydrate, purine nucleoside/nucleotide, and amino acid metabolism. The CbfA protein displays two different mechanisms of gene regulation. The expression of one set of CbfA-dependent genes requires at least the JmjC/ZF domain of the CbfA protein and thus may depend on chromatin modulation. Regulation of the larger group of genes, however, does not depend on the entire CbfA protein and requires only the carboxy-terminal domain of CbfA (CbfA-CTD). An AT-hook motif located in CbfA-CTD, which is known to mediate DNA binding to A+T-rich sequences in vitro, contributed to CbfA-CTD-dependent gene regulatory functions in vivo.
Modelling protein flexibility and plasticity is computationally challenging but important for understanding the function of biological systems. Furthermore, it has great implications for the prediction of (macro) molecular complex formation. Recently, coarse-grained normal mode approaches have emerged as efficient alternatives for investigating large-scale conformational changes for which more accurate methods like MD simulation are limited due to their computational burden. We have developed a Normal Mode based Simulation (NMSim) approach for efficient conformation generation of macromolecules. Combinations of low energy normal modes are used to guide a simulation pathway, whereas an efficient constraints correction approach is applied to generate stereochemically allowed conformations. Non-covalent bonds like hydrogen bonds and hydrophobic tethers and phi-psi favourable regions are also modelled as constraints. Conformations from our approach were compared with a 10 ns MD trajectory of lysozyme. A 2-D RMSD plot shows a good overlap of conformational space, and rms fluctuations of residues show a correlation coefficient of 0.78 between the two sets of conformations. Furthermore, a comparison of NMSim simulations starting from apo structures of different proteins show that ligand-bound conformations can be sampled for those cases where conformational changes are mainly correlated, e.g., domain-like motion in adenylate kinase. Efforts are currently being made to also model localized but functionally important motions for protein binding pockets and protein-protein interfaces using relevant normal mode selection criteria and implicit rotamer basin creation.
This article shows that investors financing a portfolio of projects may use the depth of their financial pockets to overcome entrepreneurial incentive problems. Competition for scarce informed capital at the refinancing stage strengthens investors’ bargaining positions. And yet, entrepreneurs’ incentives may be improved, because projects funded by investors with ‘‘shallow pockets’’ must have not only a positive net present value at the refinancing stage, but one that is higher than that of competing portfolio projects. Our article may help understand provisions used in venture capital finance that limit a fund’s initial capital and make it difficult to add more capital once the initial venture capital fund is raised. This paper shows that active investors, such as venture capitalists, can affect the speed at which new ventures grow. In the absence of product market competition, new ventures financed by active investors grow faster initially, though in the long run those financed by passive investors are able to catch up. By contrast, in a competitive product market, new ventures financed by active investors may prey on rivals that are financed by passive investors by “strategically overinvesting” early on, resulting in long-run differences in investment, profits, and firm growth. The value of active investors is greater in highly competitive industries as well as in industries with learning curves, economies of scope, and network effects, as is typical for many “new economy” industries. For such industries, our model predicts that start-ups with access to venture capital may dominate their industry peers in the long run. In der vorliegenden Arbeit wurden zwei motivationale Erklärungsmodelle, Zielorientierungen und das kognitiv-motivationale Prozessmodell, im Rahmen des selbstregulierten Lernens integriert. Selbstregulationsprozesse sind nach Carver und Scheier (1981) zyklisch angelegt. Zu den Bestandteilen der Selbstregulation gehören nach Boekaerts (1999)Regulation der Informationsverarbeitung, Regulation des Lernprozesses und Regulation des Selbsts. Auf dieser Ebene sind die Zielorientierungen angesiedelt. In dieser Arbeit das 2 x 2 Modell von Elliot und McGregor (2001) herangezogen. Es berücksichtigt zwei Dimensionen, die Valenz (Annäherungs- und Vermeidungskomponente) und die Kompetenz (Vergleichsmaßstab intern und extern). Hieraus resultieren: 1) Lern-Annäherungs-Ziele (positive Valenz & interner Vergleich, 2) Lern-Vermeidungs-Ziele (negative Valenz & interner Vergleich), 3) Leistungs-Annäherungs-Ziele (positive Valenz & externer Vergleich) und 4) Leistungs-Vermeidungs-Ziele (negative Valenz & externer Vergleich). Diese vier Zielorientierungen sind der erste Teil des integrierten Modells, der zweite zentrale Teil ist das kognitiv-motivationale Prozessmodell (Vollmeyer & Rheinberg, 1999, 2006). Ausgangspunkt ist die aktuelle Motivation, die aus 1) der Erfolgswahrscheinlichkeit, 2) der Misserfolgsbefürchtung, 3) dem Interesse und 4) der Herausforderung als unabhängige Faktoren besteht. Diese aktuelle Motivation wirkt nicht direkt auf die Leistung, sondern durch kognitive (hier: Metakognition) und motivationale (hier: Flow-Erleben) Mediatoren. Es wurden drei Fragestellungen bearbeitet. Die erste Fragestellung betraf das Zusammenspiel der Zielorientierungen und der aktuellen Motivation. Eine Überprüfung dieses integrierten Motivationsmodell erfolgte mittels Pfadanalyse. Die zweite und dritte Fragestellung befasste sich spezifisch mit dem kognitiv-motivationalen Prozessmodell. Es wurden Subgruppen auf der Basis der aktuellen Motivation identifiziert und ihre Bedeutung für die Mediatoren und die Leistung untersucht. Die dritte Fragestellung fokussierte den Prozesscharakter des Modells. Hier erfolgten Analysen über den Arbeitsprozess hinweg. Die Fragebögen (Achievement Goal Questionnaire (Elliot & McGregor, 2001), Metakognitionsfragebogen (aufbauend auf LIST (Wild, 2000) und Metacognitive Awareness Inventory (Schraw & Dennison, 1994), Fragebogen zur aktuellen Motivation (Rheinberg, Vollmeyer & Burns, 2001), Flow-Kurz-Skala (Rheinberg, Vollmeyer & Engeser, 2003)) und die Problemlöseaufgaben (Sudokus) wurden in einer Pilotstudie getestet. An der Hauptstudie nahmen 202 Personen teil (73% weiblich). Das integrierte Motivationsmodell geht davon aus, dass eine positiver Effekt der Zielorientierungen als Personenvariable auf die aktuelle Motivation besteht. Die aktuelle Motivation als Startpunkt des situationalen Geschehens wiederum wirkt positiv auf die Mediatoren Flow-Erleben und Metakognition, hat aber keinen direkten Effekt auf die Leistung. Dieser wird nämlich über die Mediatoren vermittelt. Deswegen wird ein positiver Effekt des Flow-Erlebens und der Metakognition auf die Leistung erwartet. Das Zusammenwirken der beiden Mediatoren kann aus dem kognitiv-motivationalen Prozessmodell nicht abgeleitet werden. Ob ein Zusammenhang besteht oder nicht wird geprüft. Für das Vorwissen wird ein positiver Effekt auf die aktuelle Motivation und die Leistung erwartet. Die Pfadanalyse zur Überprüfung des integrierten Motivationsmodells zeigte, dass eine Integration nicht nur theoretisch sinnvoll ist, sondern auch empirisch gestützt wird. Die Bedeutung der Metakognition als kognitiver Mediator wurde gezeigt. Die zweite Fragestellung fokussierte auf die aktuelle Motivation. Auf der Basis dieser wurden mittels Clusteranalyse drei distinkte Gruppen identifiziert: hoch Motivierte, niedrig Motivierte und ängstlich Motivierte (vergleichbar zu Vollmeyer & Rheinberg, 2004). Diese Gruppen unterschieden sich hinsichtlich ihres Flow-Erlebens, ihrer Metakognition und ihrer Leistung. Die hoch Motivierten erlebten am meisten Flow, berichteten mehr Metakognition und zeigten bessere Leistung. Der Prozesscharakter des kognitiv-motivationalen Prozessmodells stand im Mittelpunkt der dritten Fragestellung. Diese Analyse über die drei Sudokus hinweg zeigte eine ähnliche Entwicklung des Flow-Erlebens bei den hoch und niedrig Motivierten. Vom ersten zum zweiten Sudoku stieg es an, um dann wieder abzufallen. Dies war bei der Metakognition nicht der Fall. Die hoch Motivierten berichteten einen stärkeren Rückgang der Metakognition beim dritten Sudoku als die niedrig Motivierten. Bei der Leistung zeigte sich für die hoch Motivierten ein Anstieg beim zweiten Sudoku und dann ein Rückgang beim dritten, wohingegen die Leistung der niedrig Motivierten kontinuierlich abfiel. Abschließend wurden die Schwierigkeiten und Grenzen der vorliegenden Arbeit diskutiert und Implikationen der Ergebnisse für die Theorieentwicklung und ihre Bedeutung für den Anwendungskontext aufgezeigt.
We have investigated the role of reactive oxygen species and thiol-oxidizing agents in the induction of cell death and have shown that adenocarcinoma gastric (AGS) cells respond differently to the oxidative challenge according to the signaling pathways activated. In particular, apoptosis in AGS cells is induced via the mitochondrial pathway upon treatment with thiol-oxidizing agents, such as diamide. Apoptosis is associated with persistent oxidative damage, as evidenced by the increase in carbonylated proteins and the expression/activation of DNA damage-sensitive proteins histone H2A.X and DNA-dependent protein kinase. Resistance to hydrogen peroxide is instead associated with Keap1 oxidation and rapid translocation of Nrf2 into the nucleus. Sensitivity to diamide and resistance to hydrogen peroxide are correlated with GSH redox changes, with diamide severely increasing GSSG, and hydrogen peroxide transiently inducing protein-GSH mixed disulfides. We show that p53 is activated in response to diamide treatment by the oxidative induction of the Trx1/p38(MAPK) signaling pathway. Similar results were obtained with another carcinoma cell line, CaCo2, indicating that these findings are not limited to AGS cells. Our data suggest that thiol-oxidizing agents could be exploited as inducers of apoptosis in tumor histotypes resistant to ROS-producing chemotherapeutics.
Macrophages ingesting apoptotic cells attenuate inflammatory responses, such as reactive oxygen species (ROS) generation. In atherosclerosis, ongoing inflammation and accumulation of apoptotic/necrotic material are observed, suggesting defects of phagocytes in recognizing or responding to dying cells. Modified lipoproteins such as oxidized LDL (oxLDL) are known to promote inflammation and to interfere with apoptotic cell clearance. Here, we studied the impact of cells exposed to oxLDL on their ability to interfere with the oxidative burst in phagocytes. In contrast to apoptotic cells, cells dying in response to or in the presence of oxLDL failed to suppress ROS generation despite efficiently being taken up by phagocytes. In addition, apoptotic cells, but not oxLDL-treated cells, inhibited phosphorylation of extracellular signal-regulated kinase, which is important for NADPH oxidase activation. oxLDL treatment did not interfere with activation of the antiinflammatory transcriptional regulator peroxisome proliferator-activated receptor gamma by apoptotic cells. Moreover, cells exposed to oxLDL failed to suppress lipopolysaccharide- induced proinflammatory cytokine expression, whereas apoptotic cells attenuated these phagocyte responses. Thus, the presence of oxLDL during cell death impaired the ability of apoptotic cells to act antiinflammatory with regard to oxidative burst inhibition and cytokine expression in phagocytes.
Specific functions of biological systems often require conformational transitions of macromolecules. Thus, being able to describe and predict conformational changes of biological macromolecules is not only important for understanding their impact on biological function, but will also have implications for the modelling of (macro)molecular complex formation and in structure-based drug design approaches. The “conformational selection model” provides the foundation for computational investigations of conformational fluctuations of the unbound protein state. These fluctuations may reveal conformational states adopted by the bound proteins. The aim of this work is to incorporate directional information in a geometry-based approach, in order to sample biologically relevant conformational space extensively. Interestingly, coarse-grained normal mode (CGNM) approaches, e.g., the elastic network model (ENM) and rigid cluster normal mode analysis (RCNMA), have emerged recently and provide directions of intrinsic motions in terms of harmonic modes (also called normal modes). In my previous work and in other studies it has been shown that conformational changes upon ligand binding occur along a few low-energy modes of unbound proteins and can be efficiently calculated by CGNM approaches. In order to explore the validity and the applicability of CGNM approaches, a large-scale comparison of essential dynamics (ED) modes from molecular dynamics (MD) simulations and normal modes from CGNM was performed over a dataset of 335 proteins. Despite high coarse-graining, low frequency normal modes from CGNM correlate very well with ED modes in terms of directions of motions (average maximal overlap is 0.65) and relative amplitudes of motions (average maximal overlap is 0.73). In order to exploit the potential of CGNM approaches, I have developed a three-step approach for efficient exploration of intrinsic motions of proteins. The first two steps are based on recent developments in rigidity and elastic network theory. Initially, static properties of the protein are determined by decomposing the protein into rigid clusters using the graph-theoretical approach FIRST at an all-atom representation of the protein. In a second step, dynamic properties of the molecule are revealed by the rotations-translations of blocks approach (RTB) using an elastic network model representation of the coarse-grained protein. In the final step, the recently introduced idea of constrained geometric simulations of diffusive motions in proteins is extended for efficient sampling of conformational space. Here, the low-energy (frequency) normal modes provided by the RCNMA approach are used to guide the backbone motions. The NMSim approach was validated on hen egg white lysozyme by comparing it to previously mentioned simulation methods in terms of residue fluctuations, conformational space explorations, essential dynamics, sampling of side-chain rotamers, and structural quality. Residue fluctuations in NMSim generated ensemble is found to be in good agreement with MD fluctuations with a correlation coefficient of around 0.79. A comparison of different geometry-based simulation approaches shows that FRODA is restricted in sampling the backbone conformational space. CONCOORD is restricted in sampling the side-chain conformational space. NMSim sufficiently samples both the backbone and the side-chain conformations taking experimental structures and conformations from the state of the art MD simulation as reference. The NMSim approach is also applied to a dataset of proteins where conformational changes have been observed experimentally, either in domain or functionally important loop regions. The NMSim simulations starting from the unbound structures are able to reach conformations similar to ligand bound conformations (RMSD < 2.4 Å) in 4 out of 5 cases of domain moving proteins. In these four cases, good correlation coefficients (R > 0.7) between the RMS fluctuations derived from NMSim generated structures and two experimental structures are observed. Furthermore, intrinsic fluctuations in NMSim simulation correlate with the region of loop conformational changes observed upon ligand binding in 2 out of 3 cases. The NMSim generated pathway of conformational change from the unbound structure to the ligand bound structure of adenylate kinase is validated by a comparison to experimental structures reflecting different states of the pathway as proposed by previous studies. Interestingly, the generated pathway confirms that the LID domain closure precedes the closing of the NMPbind domain, even if no target conformation is provided in NMSim. Hence, the results in this study show that, incorporating directional information in the geometry-based approach NMSim improves the sampling of biologically relevant conformational space and provides a computationally efficient alternative to state of the art MD simulations.
Background Gamma-aminobutyric acid (GABA) is an important inhibitory neurotransmitter which mainly mediates its effects on neurons via ionotropic (GABAA) and metabotropic (GABAB) receptors. GABAB receptors are widely expressed in the central and the peripheral nervous system. Although there is evidence for a key function of GABAB receptors in the modulation of pain, the relative contribution of peripherally- versus centrally-expressed GABAB receptors is unclear. Results In order to elucidate the functional relevance of GABAB receptors expressed in peripheral nociceptive neurons in pain modulation we generated and analyzed conditional mouse mutants lacking functional GABAB(1) subunit specifically in nociceptors, preserving expression in the spinal cord and brain (SNS-GABAB(1)-/- mice). Lack of the GABAB(1) subunit precludes the assembly of functional GABAB receptor. We analyzed SNS-GABAB(1)-/- mice and their control littermates in several models of acute and neuropathic pain. Electrophysiological studies on peripheral afferents revealed higher firing frequencies in SNS-GABAB(1)-/- mice compared to corresponding control littermates. However no differences were seen in basal nociceptive sensitivity between these groups. The development of neuropathic and chronic inflammatory pain was similar across the two genotypes. The duration of nocifensive responses evoked by intraplantar formalin injection was prolonged in the SNS-GABAB(1)-/- animals as compared to their control littermates. Pharmacological experiments revealed that systemic baclofen-induced inhibition of formalin-induced nociceptive behaviors was not dependent upon GABAB(1) expression in nociceptors. Conclusion This study addressed contribution of GABAB receptors expressed on primary afferent nociceptive fibers to the modulation of pain. We observed that neither the development of acute and chronic pain nor the analgesic effects of a systematically-delivered GABAB agonist was significantly changed upon a specific deletion of GABAB receptors from peripheral nociceptive neurons in vivo. This lets us conclude that GABAB receptors in the peripheral nervous system play a less important role than those in the central nervous system in the regulation of pain.
Diabetic nephropathy (DN) is a major cause of end-stage renal failure worldwide. Oxidative stress has been reported to be a major culprit of the disease and increased oxidized low density lipoprotein (oxLDL) immune complexes were found in patients with DN. In this study we present evidence, that CXCL16 is the main receptor in human podocytes mediating the uptake of oxLDL. In contrast, in primary tubular cells CD36 was mainly involved in the uptake of oxLDL. We further demonstrate that oxLDL down-regulated α3-integrin expression and increased the production of fibronectin in human podocytes. In addition, oxLDL uptake induced the production of reactive oxygen species (ROS) in human podocytes. Inhibition of oxLDL uptake by CXCL16 blocking antibodies abrogated the fibronectin and ROS production and restored α3 integrin expression in human podocytes. Furthermore we present evidence that hyperglycaemic conditions increased CXCL16 and reduced ADAM10 expression in podocytes. Importantly, in streptozotocin-induced diabetic mice an early induction of CXCL16 was accompanied by higher levels of oxLDL. Finally immunofluorescence analysis in biopsies of patients with DN revealed increased glomerular CXCL16 expression, which was paralleled by high levels of oxLDL. In summary, regulation of CXCL16, ADAM10 and oxLDL expression may be an early event in the onset of DN and therefore all three proteins may represent potential new targets for diagnosis and therapeutic intervention in DN.
We study a model of “information-based entrenchment” in which the CEO has private information that the board needs to make an efficient replacement decision. Eliciting the CEO’s private information is costly, as it implies that the board must pay the CEO both higher severance pay and higher on-the-job pay. While higher CEO pay is associated with higher turnover in our model, there is too little turnover in equilibrium. Our model makes novel empirical predictions relating CEO turnover, severance pay, and on-the-job pay to firm-level attributes such as size, corporate governance, and the quality of the firm’s accounting system.
This paper argues that banks must be sufficiently levered to have first-best incentives to make new risky loans. This result, which is at odds with the notion that leverage invariably leads to excessive risk taking, derives from two key premises that focus squarely on the role of banks as informed lenders. First, banks finance projects that they do not own, which implies that they cannot extract all the profits. Second, banks conduct a credit risk analysis before making new loans. Our model may help understand why banks take on additional unsecured debt, such as unsecured deposits and subordinated loans, over and above their existing deposit base. It may also help understand why banks and finance companies have similar leverage ratios, even though the latter are not deposit takers and hence not subject to the same regulatory capital requirements as banks.
Oral presentation from 4th International Conference of cGMP Generators, Effectors and Therapeutic Implications ; Regensburg, Germany. 19–21 June 2009 Background: An exaggerated pain sensitivity is the dominant feature of inflammatory and neuropathic pain both in the clinical setting and in experimental animal models. It manifests as pain in response to normally innocuous stimuli (allodynia), increased response to noxious stimuli (hyperalgesia) or spontaneous pain, and can persist long after the initial injury is resolved. Research over the last decades has revealed that several signaling pathways in the spinal cord essentially contribute to the pain sensitization. To test the contribution of cGMP produced by NO-sensitive guanylyl cyclase (NO-GC) to pain sensitization, we investigated the localization of NO-GC in the spinal cord and in dorsal root ganglia, and we characterized the nociceptive behavior of mice deficient in NO-GC (GC-KO mice). Results: We show that NO-GC (β1 subunit) is distinctly expressed in neurons of the mouse spinal cord, while its distribution in dorsal root ganglia is restricted to non-neuronal cells. GC-KO mice exhibited a considerably reduced nociceptive behavior in models of inflammatory or neuropathic pain, but their responses to acute pain were not impaired. Moreover, GC-KO mice failed to develop pain sensitization induced by spinal administration of drugs releasing NO. Surprisingly, during spinal nociceptive processing cGMP produced by NO-GC may activate signaling pathways different from cGMP-dependent protein kinase I (cGKI), while cGKI can be activated by natriuretic peptide receptor-B (NPR-B) dependent cGMP production. Conclusion: Taken together, our results provide evidence that NO-GC has a dominant role in the development of exaggerated pain sensitivity during inflammatory and neuropathic pain. Furthermore, beside the NO-mediated cGMP synthesis, cGMP produced by NPR-B contributes to pain sensitization by activation of cGKI.
The small leucine-rich proteoglycans (SLRPs) are biologically active components of the extracellular matrix (ECM), consisting of a protein core with leucine rich-repeat (LRR) motifs covalently linked to glycosaminoglycan (GAG) side chains. The diversity in composition resulting from the various combinations of protein cores substituted with one or more GAG chains along with their pericellular localization enables SLRPs to interact with a host of different cell surface receptors, cytokines, growth factors, and other ECM components, leading to modulation of cellular functions. SLRPs are capable of binding to: (i) different types of collagens, thereby regulating fibril assembly, organization, and degradation; (ii) Toll-like receptors (TLRs), complement C1q, and tumor necrosis factor-alpha (TNFα), regulating innate immunity and inflammation; (iii) epidermal growth factor receptor (EGF-R), insulin-like growth factor receptor (IGF-IR), and c-Met, influencing cellular proliferation, survival, adhesion, migration, tumor growth and metastasis as well as synthesis of other ECM components; (iv) low-density lipoprotein receptor-related protein (LRP-1) and TGF-β, modulating cytokine activity and fibrogenesis; and (v) growth factors such as bone morphogenic protein (BMP-4) and Wnt-I-induced secreted protein-1 (WISP-1), controlling cell proliferation and differentiation. Thus, the ability of SLRPs, as ECM components, to directly or indirectly regulate cell-matrix crosstalk, resulting in the modulation of various biological processes, aptly qualifies these compounds as matricellular proteins.
The manifestation of chronic back pain depends on structural, psychosocial, occupational and genetic influences. Heritability estimates for back pain range from 30% to 45%. Genetic influences are caused by genes affecting intervertebral disc degeneration or the immune response and genes involved in pain perception, signalling and psychological processing. This inter-individual variability which is partly due to genetic differences would require an individualized pain management to prevent the transition from acute to chronic back pain or improve the outcome. The genetic profile may help to define patients at high risk for chronic pain. We summarize genetic factors that (i) impact on intervertebral disc stability, namely Collagen IX, COL9A3, COL11A1, COL11A2, COL1A1, aggrecan (AGAN), cartilage intermediate layer protein, vitamin D receptor, metalloproteinsase-3 (MMP3), MMP9, and thrombospondin-2, (ii) modify inflammation, namely interleukin-1 (IL-1) locus genes and IL-6 and (iii) and pain signalling namely guanine triphosphate (GTP) cyclohydrolase 1, catechol-O-methyltransferase, μ opioid receptor (OPMR1), melanocortin 1 receptor (MC1R), transient receptor potential channel A1 and fatty acid amide hydrolase and analgesic drug metabolism (cytochrome P450 [CYP]2D6, CYP2C9).
Through the use of information about the biological target structure, the optimization of potential drugs can be improved. In this work I have developed a procedure that uses the quantitative change in the chemical perturbations (CSP) in the protein from NMR experiments for driving protein-ligand docking. The approach is based on a hybrid scoring function (QCSPScore) which combines traditional DrugScore potentials, which describe the interaction between protein and ligand, with Kendall’s rank correlation coefficient, which evaluates docking poses in terms of their agreement with experimental CSP. Prediction of the CSP for a specific ligand pose is done efficiently with an empirical model, taking into account only ring current effects. QCSPScore has been implemented in the AutoDock software package. Compared to previous methods, this approach shows that the use of rank correlation coefficient is robust to outliers. In addition, the prediction of native-like complex geometries improved because the CSP are already being used during the docking process, and not only in a post-filtering setting for generated docking poses. Since the experimental information is guaranteed to be quantitatively used, CSP effectively contribute to align the ligand in the binding pocket. The first step in the development of QCSPScore was the analysis of 70 protein-ligand complexes for which reference CSP were computed. The success rate in the docking increased from 71% without involvement of CSP to 100% if CSP were considered at the highest weighting scheme. In a second step QCSPScore was used in re-docking three test cases, for which reference experimental CSP data was available. Without CSP, i.e. in the use of conventional DrugScore potentials, none of the three test cases could be successfully re-docked. The integration of CSP with the same weighting factor as described above resulted in all three cases successfully re-docked. For two of the three complexes, native-like solutions were only produced if CSP were considered.Conformational changes in the binding pockets of up to 2 Å RMSD did not affect the success of the docking. QCSPScore will be particularly interesting in difficult protein-ligand complexes. They are in particular those cases in which the shape of the binding pocket does not provide sufficient steric restraints such as in flat protein-protein interfaces and in the virtual screening of small chemical fragments.
Background: The c-Cbl-associated protein (CAP), also known as ponsin, localizes to focal adhesions and stress fibers and is involved in signaling events. Phosphorylation has been described for the other two members of the sorbin homology family, vinexin and ArgBP2, but no data exist about the putative phosphorylation of CAP. According to previous findings, CAP binds to tyrosine kinase c-Abl. However, it is not known if CAP is a substrate of c-Abl or other tyrosine kinases or if phosphorylation regulates its localization.
Results: We here show that CAP is Tyr phosphorylated by and interacts with both c-Abl and c-Src. One major phosphorylation site, Tyr360, and two minor contributors Tyr326 and Tyr632 were identified as Abl phosphorylation sites, whereas Src preferentially phosphorylates Tyr326 and Tyr360. Phosphorylation of CAP was not necessary for its localization to focal adhesions and stress fibers, but Tyr326Phe substitution alters the function of CAP during cell spreading.
Conclusion: This is the first demonstration of phosphorylation of CAP by any kinase. Our findings suggest that coordinated action of Src and Abl might regulate the function of CAP and reveal a functional role especially for the Src-mediated Tyr phosphorylation of CAP in cell spreading.
Opting out of the great inflation: German monetary policy after the break down of Bretton Woods

During the turbulent 1970s and 1980s the Bundesbank established an outstanding reputation in the world of central banking. Germany achieved a high degree of domestic stability and provided safe haven for investors in times of turmoil in the international financial system. Eventually the Bundesbank provided the role model for the European Central Bank. Hence, we examine an episode of lasting importance in European monetary history. The purpose of this paper is to highlight how the Bundesbank monetary policy strategy contributed to this success. We analyze the strategy as it was conceived, communicated and refined by the Bundesbank itself. We propose a theoretical framework (following Söderström, 2005) where monetary targeting is interpreted, first and foremost, as a commitment device. In our setting, a monetary target helps anchoring inflation and inflation expectations. We derive an interest rate rule and show empirically that it approximates the way the Bundesbank conducted monetary policy over the period 1975-1998. We compare the Bundesbank´s monetary policy rule with those of the FED and of the Bank of England. We find that the Bundesbank´s policy reaction function was characterized by strong persistence of policy rates as well as a strong response to deviations of inflation from target and to the activity growth gap. In contrast, the response to the level of the output gap was not significant. In our empirical analysis we use real-time data, as available to policy-makers at the time. Artenreiche montane Rasengesellschaften auf Lawinenbahnen des Nationalparks Gesäuse (Österreich)

Das Ziel dieser Untersuchung war es, Erkenntnisse über die Bedeutung von Lawinenbahnen für die Biodiversität zu gewinnen. Dazu wurden im Nationalpark Gesäuse (Steiermark, Österreich) 16 Pflanzenbestände in drei ausgewählten Lawinenbahnen pflanzensoziologisch untersucht, vegetationsökologisch charakterisiert sowie mit Vegetationstabelle, bodenkundlichen Analysedaten und topographischen Parametern dokumentiert. Außerdem erfolgte eine Bestandsaufnahme der Schmetterlinge. Die Pflanzenartenvielfalt wurde mit verschiedenen Diversitäts-Indizes beurteilt. Die Ursachen für den Artenreichtum in den untersuchten Lawinenbahnen werden diskutiert.
Die Pflanzenbestände wurden überwiegend zum Origano-Calamagrostietum variae gestellt; eine Einzelaufnahme wurde dem Seslerio-Caricetum sem pervirentis zugeordnet. Im Ersteren dominieren Arten mit hemikryptophytischer Lebensform; Therophyten erreichen die zweithöchste Deckung. Die Pflanzengesellschaft wird von CSR-, CS- und C-Strategen aufgebaut; alle anderen Lebensstrategietypen haben keine Bedeutung. Das Origano-Calamagrostietum variae ist eine artenreiche, natürliche Dauergesellschaft, in der mäßiger Stress und schwache Störungen die Artenzusammensetzung maßgeblich bestimmen. Aktive Lawinenbahnen auf sehr flachgründigen, steinigen, nährstoffarmen, basenreichen Rendzinen in lokalklimatisch wärmebegünstigten steilen Hanglagen der montanen Höhenstufe zählen zu den arten-, blüten- und aspektreichsten und somit ökologisch wertvollsten Vegetationsformationen im Nationalparkgebiet. In den kräuterreichen Pflanzenbeständen wurden im Durchschnitt 71 Gefäßpflanzenarten und 5 Moosarten pro 20 m2 Aufnahmefläche gefunden. Mit dieser hohen Phytodiversität und dem Blütenreichtum ist auch eine große Artenvielfalt bei den Schmetterlingen verbunden. Die untersuchten Lawinenbahnen weisen nicht nur ein hohes Maß an Naturnähe auf, sie dürften auch der ursprüngliche Lebensraum einiger Pflanzenarten des Wirtschafts- und Extensivgrünlandes der Tallagen sein. Auf Grund ihrer hohen naturschutzfachlichen Bedeutung müssen Lawinenbahnen als Ganzes geschützt werden. Periodische oder episodische Lawinenabgänge sind Voraussetzung für die Existenz dieser störungsgeprägten Ökosysteme. Unterhalb von Lawinenbahnen dürfen keine Gebäude oder Infrastrukturen errichtet werden, weil diese Bautätigkeit Schutzobjekte erzeugt. Dadurch würde ein Bedarf für Schutzmaßnahmen entstehen, vor allem in Form von Lawinenverbauungen und Aufforstungen, wodurch Lawinenabgänge verhindert werden.
Die vorliegende Arbeit untersucht das nationale Selbstverständnis der gesamten Konföderation und behandelt insbesondere Aspekte der Innenpolitik sowie bestimmte soziokulturelle und sozioökonomische Charakteristika, aus denen sich die nationalistische Ideologie der Konföderierten konstituierte. Es bleibt zu betonen, dass sich der Fokus der hiesigen Untersuchungen auf innerstaatliche Ereignisse und Phänomene während des Bürgerkrieges richtet und somit vom großen Feld der konföderierten Außenpolitik beinahe vollständig absieht. Die Außenpolitik bildet zweifelsohne ein ebenso spannendes Forschungsgebiet, sie wird hier aber zu Gunsten einer expliziteren Betrachtung des innerstaatlichen Kontexts außen vor gelassen, da die inneren Umstände für die Genese des konföderierten Nationalismus unmittelbarer und somit von größerer Tragweite waren.
Solid state NMR is a emerging method for the study of membrane proteins, which has received much interest in recent years. Limiting the study of many pharmacologically relevant targets, are the often long measuring times, required to obtain especially higher dimensional solid state NMR spectra of good quality. To address this problem, multiple methods where developed in this work, which can be categorized into two groups. The first set of methods aims at the quality of certain spectra, by implementing a spectral filter, which increases the fidelity of the measured data. The second set of methods, addresses the problem of long measuring times directly, by increasing the sensitivity per unit time, as could be shown, for example, on homo- and heteronuclear singlequantum-singlequantum correlation experiments. The gains in measuring time for the latter group of methods are typically in the order of 2-3, but some experiments allow multiple methods to be employed simultaneously, which can lead to a decrease in measuring time of a factor of up to 8. It is important to mention, that none of the methods introduced in this work require any equipment in addition to the conventional setup present in most sold state NMR laboratories and no changes or addition to the samples under study are required. Therefore the gains reported in this work come at no extra cost and require only minimal implementation effort on the side of the user.
X-ray structure of the Na+-coupled Glycine-Betaine symporter BetP from Corynebacterium glutamicum

Cellular membranes are important sites of interaction between cells and their environment. Among the multitude of macromolecular complexes embedded in these membranes, transporters play a particularly important role. These integral membrane proteins perform a number of vital functions that enable cell adaptation to changing environmental conditions. Osmotic stress is a major external stimulus for cells. Bacteria are frequently exposed to either hyperosmotic or hypoosmotic stress. Typical conditions for soil bacteria, such as Corynebacterium glutamicum, vary between dryness and sudden rainfall. Physical stimuli caused by osmotic stress have to be sensed and used to activate appropriate response mechanisms. Hypoosmotic stress causes immediate and uncontrolled influx of water. Cells counteract by instantly opening mechanosensitive channels, which act as emergency valves leading to fast efflux of small solutes out of the cell, therebydiminishing the osmotic gradient across the cell membrane. Hyperosmotic stress, on the other hand, results in water efflux. This is counterbalanced by an accumulation of small, osmotically active solutes in the cytoplasm, the so-called compatible solutes. They comprise a large variety of substances, including amino acids (proline), amino acid derivatives (betaine, ectoine), oligosaccharides (trehalose), and heterosides (glucosylglycerol). Osmoregulated transporters sense intracellular osmotic pressure and respond to hyperosmotic stress by facilitating the inward translocation of compatible solutes across the cell membrane, to restore normal hydration levels. This work presents the first X-ray structure of a member of the Betaine-Choline-Carnitine-Transporter (BCCT) family, BetP. This Na+-coupled symporter from Corynebacterium glutamicum is a highly effective osmoregulated and specific uptake system for glycine-betaine. X-ray structure determination was achieved using single wavelength anomalous dispersion (SAD) of selenium atoms. Selenium was incorporated into the protein during its expression in methione auxotrophic E. coli cells, grown in media supplemented with selenomethionine. SAD data with anomalous signal up to 5 Å led to the detection of 39 selenium sites, which were used to calculate the initial electron density map of the protein. Medium resolution and high data anisotropy made the structure determination of BetP a challenging task. A specific strategy for data anisotropy correction and a combination of various crystallographic programs were necessary to obtain an interpretable electron density map suitable for model building. The crystal structure of BetP shows a trimer with glycine-betaine bound in a three-fold cation-pi interaction built by conserved tryptophan residues. The bound substrate is occluded from both sides of the membrane and aromatic side chains line its transport pathway. Very interestingly, the structure reveals that the alpha-helical C-terminal domain, for which a chemo- and osmosensory function was elucidated by biochemical methods, interacts with cytoplasmic loops of an adjacent monomer. These unexpected monomer-monomer interactions are thought to be crucial for the activation mechanism of BetP, and a new atomic model combing biochemical results with the crystal structure is proposed. BetP is shown to have the same overall fold as three unrelated Na+-coupled symporters. While these were crystallised in either the outward- or inward-facing conformation, BetP reveals a unique intermediate state, opening new perspectives on the alternating access mechanism of transport.
Both the genomes of the epsilonproteobacteria Wolinella succinogenes and Campylobacter jejuni contain operons (sdhABE) that encode for so far uncharacterized enzyme complexes annotated as ‘non-classical’ succinate:quinone reductases (SQRs). However, the role of such an enzyme ostensibly involved in aerobic respiration in an anaerobic organism such as W. succinogenes has hitherto been unknown. We have established the first genetic system for the manipulation and production of a member of the non-classical succinate:quinone oxidoreductase family. Biochemical characterization of the W. succinogenes enzyme reveals that the putative SQR is in fact a novel methylmenaquinol:fumarate reductase (MFR) with no detectable succinate oxidation activity, clearly indicative of its involvement in anaerobic metabolism. We demonstrate that the hydrophilic subunits of the MFR complex are, in contrast to all other previously characterized members of the superfamily, exported into the periplasm via the twin-arginine translocation (tat)-pathway. Furthermore we show that a single amino acid exchange (Ala86→His) in the flavoprotein of that enzyme complex is the only additional requirement for the covalent binding of the otherwise non-covalently bound FAD. Our results provide an explanation for the previously published puzzling observation that the C. jejuni sdhABE operon is upregulated in an oxygen-limited environment as compared with microaerophilic laboratory conditions.
Inhibition of the soluble epoxide hydrolase (sEH) has beneficial effects on vascular inflammation and hypertension indicating that the enzyme may be a promising target for drug development. As the enzymatic core of the hydrolase domain of the human sEH contains two tyrosine residues (Tyr383 and Tyr466) that are theoretically crucial for enzymatic activity, we addressed the hypothesis that the activity of the sEH may be affected by nitrosative stress. Epoxide hydrolase activity was detected in human and murine endothelial cells as well in HEK293 cells and could be inhibited by either authentic peroxynitrite (ONOO−) or the ONOO− generator 3-morpholino-sydnonimine (SIN-1). Protection of the enzymatic core with 1-adamantyl-3-cyclohexylurea in vitro decreased sensitivity to SIN-1. Both ONOO− and SIN-1 elicited the tyrosine nitration of the sEH protein and mass spectrometry analysis of tryptic fragments revealed nitration on several tyrosine residues including Tyr383 and Tyr466. Mutation of the latter residues to phenylalanine was sufficient to abrogate epoxide hydrolase activity. In vivo, streptozotocin-induced diabetes resulted in the tyrosine nitration of the sEH in murine lungs and a significant decrease in its activity. Taken together, these data indicate that the activity of the sEH can be regulated by the tyrosine nitration of the protein. Moreover, nitrosative stress would be expected to potentiate the physiological actions of arachidonic acid epoxides by preventing their metabolism to the corresponding diols.
Proline-rich tyrosine kinase 2 (PYK2) can be activated by angiotensin II (Ang II) and reactive oxygen species. We report that in endothelial cells, Ang II enhances the tyrosine phosphorylation of endothelial NO synthase (eNOS) in an AT1-, H2O2-, and PYK2-dependent manner. Low concentrations (1–100 µmol/liter) of H2O2 stimulated the phosphorylation of eNOS Tyr657 without affecting that of Ser1177, and attenuated basal and agonist-induced NO production. In isolated mouse aortae, 30 µmol/liter H2O2 induced phosphorylation of eNOS on Tyr657 and impaired acetylcholine-induced relaxation. Endothelial overexpression of a dominant-negative PYK2 mutant protected against H2O2-induced endothelial dysfunction. Correspondingly, carotid arteries from eNOS–/– mice overexpressing the nonphosphorylatable eNOS Y657F mutant were also protected against H2O2. In vivo, 3 wk of treatment with Ang II considerably increased levels of Tyr657-phosphorylated eNOS in the aortae of wild-type but not Nox2y/– mice, and this was again associated with a clear impairment in endothelium-dependent vasodilatation in the wild-type but not in the Nox2y/– mice. Collectively, endothelial PYK2 activation by Ang II and H2O2 causes the phosphorylation of eNOS on Tyr657, attenuating NO production and endothelium-dependent vasodilatation. This mechanism may contribute to the endothelial dysfunction observed in cardiovascular diseases associated with increased activity of the renin–angiotensin system and elevated redox stress.
Crista junctions (CJs) are important for mitochondrial organization and function, but the molecular basis of their formation and architecture is obscure. We have identified and characterized a mitochondrial membrane protein in yeast, Fcj1 (formation of CJ protein 1), which is specifically enriched in CJs. Cells lacking Fcj1 lack CJs, exhibit concentric stacks of inner membrane in the mitochondrial matrix, and show increased levels of F1FO–ATP synthase (F1FO) supercomplexes. Overexpression of Fcj1 leads to increased CJ formation, branching of cristae, enlargement of CJ diameter, and reduced levels of F1FO supercomplexes. Impairment of F1FO oligomer formation by deletion of its subunits e/g (Su e/g) causes CJ diameter enlargement and reduction of cristae tip numbers and promotes cristae branching. Fcj1 and Su e/g genetically interact. We propose a model in which the antagonism between Fcj1 and Su e/g locally modulates the F1FO oligomeric state, thereby controlling membrane curvature of cristae to generate CJs and cristae tips.
