Pulsed dipolar (PD) EPR spectroscopy is an established and reliable tool for the investigation of biomolecules. In terms of long distance and orientation measurements, it is one of the leading methods and further fields of application are constantly being explored. The distances that can be detected with PD EPR also correspond to the range in which almost all important biomolecule interactions occur. In the transition from in vitro spectroscopy to in-cell spectroscopy, the power of PD EPR spectroscopy is particularly evident. It is non-invasive, more sensitive than NMR, and does not exhibit background signals from diamagnetic molecules. In particular, the absence of background signals is of great importance given the high density of molecules within cellular environment. However, like any other spectroscopic method, PD EPR has certain limitations. Owing to the intrinsically fast electron spin echo dephasing at higher temperature, these experiments are commonly carried out in frozen solutions at about 50 K. This temperature is far away from the physiological conditions and the freezing additives used, e.g. glycols, can further influence the structure. To enable measurements with and within living organisms, it is therefore necessary to ascend from the cold depths of the frozen state. At the same time, one has to adapt the spin tags for the desired application. Established nitroxides commonly used for EPR studies are typically susceptible to reduction. Thus, for studies under physiological conditions, e.g. in the cell, one has to fight against the reductive environment in the cell and somehow protect the spin labels. Initial published in-cell experiments within the research group and investigations of homogeneously distributed labeled double-stranded (ds) ‐DNA samples in solid matrices showed promising results and enabled pulsed measurement in the temperature range of 50‐ 295 K. It could also be demonstrated that spherical shielded nitroxides have a significantly longer life span in cellular environments than non-protected ones and first nuclear acids were measured in cell. Based on these results, we have gone further to overcome the standing limitations and developed the use of PD EPR spectroscopy. This work addresses these challenges with the overall goal of advancing the applications of PD EPR spectroscopy for studying biomolecules under physiological conditions.
We have focused on four different approaches. The results of these studies were published in various publications. They are presented and discussed together with further studies and put into the context of research conducted before and after the authors' publications.
In approach 1, we fought against the two main obstacles for using pulsed dipolar spectroscopy at ambient conditions – minimizing phase memory time T2 and averaging of the anisotropic dipolar coupling by rotational diffusion. We focused on an immobilization approach, while using rigid spin labels at same time. Besidesto the distance information, the incorporated rigid spin labels will give additional angular constrains and information about the molecular dynamics.
In approach 2, we focused on the on-site and on-demand formation of nitroxide spin labels using light-sensitive alkyl protection groups. This a very mild and efficient procedure that will hardly interfere with sensitive functional groups present in oligonucleotides or peptides. By establishing this method and using coumarin protecting groups plus two-photon excitation, this property may offer the potential to generate spin labels with very high levels of spatial and temporal resolution.
For approach 3, we used paramagnetic Gd3+ -ions as intrinsically stable labels, which are not reducible within a cellular environment. Easy to mix and bound to encodable lanthanide binding tags within the molecule Interleucin 1β, we were able to measure distances between two tags with PELDOR spectroscopy. We tested the extent to which this system is suitable for in-cell measurements.
Finally, we focus on methods for easier labeling by using non-covalentlabeling techniques. One of these is the novel nitroxide G´ for site-directed spin labeling of nucleic acids, especially for RNA. This spin label is sterically hindered, easy to build and binding occurs in seconds by simply mixing the spin label with the target. For large RNAs, another easy-to-mix and noncovalent spin-labeling strategy will be experimentally accompanied and presented.
The approaches and results described here are intended to demonstrate that the study of the biological functions of biomolecules under physiological conditions by pulsed EPR spectroscopy is feasible and operational. In combination, they will enable the life sciences to make further and faster progress in the search for the molecular master plan.
Endocannabinoids (eCB) are signaling lipids and became known for their importance in the central nervous system as well as in immune defense. Beneficial effects of eCB are shown in processes of excitotoxic lesion, secondary damage and neuronal plasticity throughout the last years. Two canabinoid receptors, type 1 (CB1) and type 2 (CB2) as the respective endogenous ligands belong to the endocannabinoid system (eCBS). In 1990, the CB1 could be cloned and was localised mainly on neurons. Shortly thereafter in 1993, the CB2 was characterised and found primarily on cells belonging to the immune system. N-arachidonoylethanolamide (AEA), often called anandamide, and 2-arachidonoylglycerol (2-AG) are the best characterised eCB. N-palmitylethanolamide (PEA) and N-oleoylethanolamide (OEA) have no or only low affinity to CB1 but enhance the affinity of AEA significantly. This group is therefore often summarized as N-ethanolamides (NEA). ECB are derivates of arachidonic acid and are stored in membranes where they become hydrolysed on demand by specific enzymes. Traumatic brain injury altered the levels of eCB in the blood in vivo and when applied in vitro after neuronal damage, eCB could reduce the damaging burden. Further studies demonstrated that eCB are potent to down-regulate pro-inflammatory cytokines and most important to decrease neuronal excitation.
In the present study, the intrinsic regulation of the endocannabinoid system after neuronal damage over time was investigated in rat Organotypic Hippocampal Slice Cultures (OHSC). Temporal and spatial dynamics of eCB levels were analysed after transection of the perforant pathway (PPT) in originating neurons (enthorhinal cortex, EC), areas of deafferentiation/anterograde axonal degeneration (dentate gyrus, DG) and of the synaptically linked cornu ammonis region 1 (CA1) as well as after excitotoxic lesion in the respective regions.
A strong increase of all eCB was observed only in the denervation zone of the DG 24 hours post PPT. In excitotoxic lesioned OHSC all eCB were elevated, in the investigated regions up to 72 hours post lesion (hpl). The responsible enzyme for biosynthesis of the NEA, NAPE-PLD protein, was increased during the early timepoints of measurement (1-6 hpl). The responsible catabolizing enzyme, FAAH, and the CB1 receptor were up-regulated at a later timepoint, 48 hpl, explaining the eCB levels. In the present model, the inhibition of the enzyme responsible for 2-AG hydrolysis (MAGL) was neuroprotective as previously shown and a re-distribution within neurons and astrocytes during neuronal damage could be observed. In primary cell cultures microglia expressed the regulating enzymes of 2-AG and the enzyme responsible for NEA down-regulation, FAAH. Astrocytes expressed mainly the catalyzing enzymes, indicating the role for eCB break-down. All these findings together demonstrate the great capacity of the eCBS to control inflammatory processes and consequently neuronal cell death.
All effects of the known eCB could not be clarified by CB1/CB2 deficient mice. Several G-protein coupled receptors (GPR) are recently in discussion whether they might and should belong to the endocannabinoid system. The GPR55, the not yet cloned abnormal cannabidiol receptor and further GPRs are candidates as potential endocannabinoid receptors. Recently GPR55 has been discussed as a putative cannabinoid receptor type 3 (CB3). Quantitative PCR revealed that Gpr55 is present in primary microglia and the brain, but the exact regional and cellular distribution and the physiological/pathological effects downstream of GPR55 activation in the CNS still remain open. Therefore, the excitotoxic rat OHSC model, previously used to investigate the neuroprotective potency of eCB, was now used to investigate the neuroprotective potency of GPR55. Activation of GPR55 protected dentate gyrus granule cells in vitro after excitotoxic lesion, induced by NMDA. In parallel, GPR55 activation was able to reduce the number of microglia in the dentate gyrus. These neuroprotective effects vanished however in microglia depleted OHSCs as well as in OHSC transfected with Gpr55 siRNA, indicating a strong involvement of microglia in GPR55 mediated neuroprotection.
In summary, the present study found a strong time-dependent and anterograde mechanism of action of eCB after long-range projection damage and provided further evidence for the neuroprotective properties of eCB. The potential cannabinoid receptor 3 (GPR55) mediates neuronal protection on behalf of microglia.
Bei Entzündung oder Verletzung peripherer Gewebe und Nerven kommt es zur Bildung reaktiver Sauerstoffspezies (ROS) im schmerzleitenden System. Welche ROS-generierenden Systeme hierbei beteiligt sind, ist jedoch nur ansatzweise verstanden. In der vorliegenden Arbeit konnte gezeigt werden, dass die ROS-produzierende NADPH Oxidase 4 (Nox4) einen wichtigen ROS-Generator im nozizeptiven System darstellt. Nox4 wird in unmyelinisierten nicht-peptidergen sowie in myelinisierten primär afferenten Neuronen exprimiert. In Modellen für akute und inflammatorische Schmerzen zeigten Nox4-/--Mäuse ein ähnliches Verhalten wie ihre wildtypischen Wurfgeschwister, jedoch war ihr Schmerzverhalten in Modellen für neuropathische Schmerzen reduziert. Eine Microarray-Analyse des lumbalen Rückenmarks nach peripherer Nervenverletzung zeigte eine Hochregulation der Expression Myelin-spezifischer Gene in Wildtyp-, nicht aber in Nox4-/- Mäusen. Darüber hinaus wurden in Wildtyp-Mäusen Myelin-spezifische Proteine im N. ischiadicus nach peripherer Nervenverletzung herab reguliert, während in Nox4-/--Mäusen keine Regulation dieser Proteine beobachtet wurde. Diese Ergebnisse deuten darauf hin, dass Nox4 eine essentielle Rolle bei Myelinisierungsprozessen spielt und so die Verarbeitung neuropathischer Schmerzsignale beeinflusst.
Neben dem ROS-produzierenden System Nox4 wurde auch die Rolle des Peroxid-abbauenden Proteins Sestrin 2 (Sesn2) im nozizeptiven System untersucht. Nach peripherer Nervenverletzung wurde Sesn2-mRNA in den Spinalganglien und Sesn2-Protein im peripheren Nerv hochreguliert. Sesn2-/--Mäuse zeigten ein normales Verhalten in Modellen für akute und inflammatorische Schmerzen. Ihr Schmerzverhalten war jedoch im Formalin-Test und nach peripherer Nervenverletzung verstärkt.
Diese Ergebnisse lassen vermuten, dass sowohl Nox4 als auch Sesn2 bei der Verarbeitung neuropathischer Schmerzsignale wichtige Funktionen einnehmen. Während Nox4 pronozizeptiv wirkt, weist Sesn2 antinozizeptive Effekte auf. Die Produktion reaktiver Sauerstoffspezies scheint daher ein wichtiger endogener Faktor der Sensibilisierung im Rahmen von neuropathischen Schmerzen zu sein.
Präzise Zeitstrukturen der Aktivität der Hirnrinde haben sehr wahrscheinlich einen großen Einfluss auf die Verarbeitung von Sinnesreizen und damit auf kognitive Prozesse. Nach einer häufig zitierten Hypothese werden Einzelreize, die an verschiedenen Orten der Hirnrinde durch die jeweilige Aktivität lokaler Nervenzellpopulationen repräsentiert werden, dann als zusammengehörig wahrgenommen, wenn diese Populationen im sogenannten Gamma-Rhythmus (ca. 25-70 Hz) oszillieren und dabei die Zeitpunkte, zu welchen sie Aktionspotentiale feuern, millisekunden-präzise synchronisieren. Wenn synchrone Gamma-Oszillationen eine Rolle für kognitive Prozesse spielen, so ist zu erwarten, dass diese Zeitstrukturen durch das Vigilanzniveau beeinflusst werden. Ein Zustand aufmerksamer Wachheit führt zu einem verstärkten Auftreten synchroner Gamma-Oszillationen. Unter tiefer Narkose ist Wahrnehmung hingegen ausgeschaltet; ein solcher Zustand sollte sich demnach auch in stark reduzierten oder vollständig sistierten synchronen Gamma-Oszillationen ausdrücken. Die Regulation des Vigilanzniveaus steht unter der Kontrolle des aufsteigenden retikulären Aktivierungssystems. Dieses verteilte System innerviert unter Verwendung verschiedener Transmitter in diverse subkortikale Strukturen, unter anderem das basale Vorderhirn. Letzteres stellt die stärkste cholinerge Projektion in den Neokortex dar. Im ersten Teilprojekt dieser Arbeit wurde die Modulation “induzierter” (= nicht mit der Zeitstruktur des Stimulus assoziierter) synchroner Gamma-Oszillationen durch cholinerg wirksame Pharmaka untersucht. Hierzu wurden bei adulten Katzen Elektroden in den primären visuellen Kortex implantiert und in flacher Allgemeinanästhesie neuronale Antworten auf Sehreize aufgezeichnet. Gleichzeitig bestand die Möglichkeit, cholinerge Agonisten oder Antagonisten lokal zu applizieren. Es konnte gezeigt werden, dass cholinerge Agonisten einen fördernden Einfluss auf synchrone Gamma-Oszillationen haben, während cholinerge Antagonisten synchrone Gamma-Oszillationen hemmen und eine durch elektrische Stimulation von Teilbereichen des Aktivierungssystems ausgelöste Förderung der Oszillationen blockieren. Wahrnehmungsleistungen sind nicht nur abhängig vom Vigilanzniveau, sondern auch vom raum-zeitlichen Kontext. Das bedeutet, dass einzelne Reize nicht isoliert bewertet, sondern mit einer riesigen Menge von anderen Reizen in Beziehung gesetzt werden müssen. Der Kortex scheint im Falle der induzierten Gamma-Oszillationen sein komplexes Netzwerk für solche Interaktionen zu nutzen und in der Zeitstruktur kortikaler Signale Kontext zu kodieren. Die Aktivität kortikaler Neurone kann jedoch auch durch periodische sensorische Stimulation (zum Beispiel Flicker) zum Schwingen gebracht werden (“getriebene Oszillationen”). Es war bekannt, dass in der Aktivität des visuellen Kortex unter synchroner Stimulation Zeitstrukturen lokaler Stimuli sehr präzise abgebildet werden können. Gäbe es, wie oft angenommen, unter getriebenen Oszillationen keine Interaktion zwischen räumlich verteilter Aktivität, dann stünde die Zeitstruktur in neuronalen Antworten nicht mehr als unabhängiger Kodierungsraum für Kontext zur Verfügung, sondern wäre alleine durch den lokalen Stimulus determiniert. Dem gegenüber standen psychophysische Befunde, dass die Zeitstrukturen externer Reize zwar mit hoher Auflösung für die Wahrnehmung genutzt werden können, sie diese aber nicht zwingend determinieren. Im zweiten Teilprojekt dieser Arbeit wurde, ebenfalls im visuellen Kortex anästhesierter Katzen, die Modulation getriebener Oszillationen durch Zeitversatz zu räumlich entfernten Flickerstimuli untersucht. Es konnte demonstriert werden, dass lokale Antworten in getriebenen Oszillationen durch den zeitlichen Kontext zu Stimuli weit außerhalb des eigenen rezeptiven Feldes beeinflusst werden. Die zeitliche Streuung der Aktionspotentiale relativ zum Stimulus war in der synchronen Bedingung am kleinsten und wuchs mit steigendem Zeitversatz im Flicker. Die spektrale Analyse lokaler Feldpotentiale zeigte, dass für Antwortkomponenten im Gamma-Band die zeitliche Beziehung zum Stimulus durch asynchrones Flickern instabiler wurde, während niederfrequentere Komponenten (< 25 Hz) den gegenteiligen Effekt zeigten. Diese Änderungen waren auch abhängig von kortikaler Aktivierung. Die Ergebnisse beider Teile demonstrieren, dass präzise Zeitstrukturen kortikaler Aktivität frequenzspezifisch moduliert werden können. Die Befunde, die für den Gamma-Frequenzbereich erhoben wurden, stützen die Hypothese, dass kortikale Gamma-Oszillationen bei der Integration sensorischer Reize eine besonderen Rolle spielen. Die Beeinflussung synchroner Gamma-Oszillationen durch das cholinerge System könnte darüber hinaus eine Erklärung für den Zusammenhang zwischen cholinergem Defizit (wie beispielsweise beim Morbus Alzheimer oder beim Zentralen Anticholinergen Syndrom) und kognitiver Dysfunktion liefern.
Die Dorfschulen in Nassau-Usingen 1659 - 1806 : Schulregiment und Schulwirklichkeit auf dem Land

Die Suche nach einer umfassenden und vergleichenden Darstellung der Geschichte des niederen Schulwesens in Deutschland bleibt wenig erfolgreich. Das gilt insbesondere für die Geschichte des Schulwesens auf dem Land. Der Schwerpunkt der durchaus produktiven Forschung zur deutschen Bildungsgeschichte liegt auf universitärer, zunehmend auch auf gymnasialer Ebene, während das darunterliegende Schulwesen weniger Berücksichtigung findet. Ferner konzentriert sich die deutsche historische wie pädagogische Forschung mehr auf die Institutionen- oder aber Erziehungs-, Wissenschafts- und Ideengeschichte als auf sozialgeschichtlich-statistische Untersuchungen und die Geschichte der Schulwirklichkeit. Unter zahlreichen gleichlautenden Aussagen sei hier nur auf Lundgreen (1971) verwiesen, der den großen "Mangel an empirischen Untersuchungen des Bildungswesens" beklagt und auf Neugebauers Feststellung (1985), daß "die Erhellung der Schulwirklichkeit im 18. Jahrhundert noch immer ein Desiderat der Geschichtswissenschaft" sei. Die territoriale Zersplitterung Deutschlands in 1.774 Herrschaftseinheiten vor 1803 erschwert eine Zusammenschau der Situation des deutschen Schulwesens auf dem Land. Bisher wurden vorwiegend die großen oder bedeutenden deutschen Territorien erforscht. Dabei liegt ein deutlicher Schwerpunkt auf Preußen und auf der Geschichte der Volksschule seit den allgemein einsetzenden Schulreformen ab Mitte des 18. Jahrhunderts und insbesondere auf dem 19. Jahrhundert. In den letzten Jahren kamen wenige Einzeluntersuchungen zu Bayern und kleineren deutschen Territorien hinzu. Ein weiterer Grund für die vor allem in den vergangenen Jahrzehnten geringe Beachtung des deutschen Schulwesens auf dem Land könnte in den vielfältigen Untersuchungen zu diesem Thema im 19. und frühen 20. Jahrhundert liegen. Diese Darstellungen neigen jedoch häufig dazu, die Wirklichkeit idealistisch zu verkürzen. ...
The PANDA experiment at FAIR will perform world class physics studies using high-intensity cooled antiproton beams with momenta between 1.5 and 15 GeV/c. A rich physics program requires very good particle identification (PID). Charged hadron PID for the barrel section of the target spectrometer has to cover the angular range of 22-140° and separate pions from kaons for momenta up to 3.5 GeV/c with a separation power of at least 3 standard deviations. The system that will provide it has to be thin and operate in a strong magnetic field. A ring imaging Cherenkov detector using the DIRC principle meets those requirements. The design of the PANDA Barrel DIRC is based on the successful BABAR DIRC counter with several important changes to improve the performance and optimize the costs. The design options are being studied in detailed Monte Carlo simulation, and implemented in increasingly complex system prototypes and tested in particle beams. Before building the full system prototypes the radiator bars and lenses are measured on the test benches. The performance of the DIRC prototype was quantified in terms of the single photon Cherenkov angle resolution and the photon yield. Results for two full system prototypes will be presented. The prototype in 2011 aimed at investigating the full size expansion volume. It was found that the resolution for this configuration is at the level of in good agreement with ray tracing simulation results. A more complex prototype, tested in 2012, provided the first experience with a compact fused silica prism expansion volume, a wide radiator plate, and several advanced lens options for the focusing system. The performance of the baseline configuration of the prototype with a standard lens and an air gap met the requirements for the PANDA PID for most of the polar angle range but failed at polar angles around 90° due to photon loss at the air gap. Measurements with a prototype high-refractive index compound lens without an air gap at a polar angle of 128° beam angle showed a good resolution of σΘC = 11.8 ± 0.7 mrad and a high photon yield of Nph = 26.1 ± 0.4. Even at polar angles close to 90° the photon yield with this lens exceeded 15 detected photons per particle, meeting the PANDA Barrel DIRC PID requirements for the entire phase space and demonstrating that the compact focusing DIRC is a very promising option for PANDA.
Zielstellung: Es sollte im Rahmen dieser Arbeit geprüft werden ob (1) ein Zusammenhang zwischen einer radiologischen Beinlängendifferenz und einer veränderten Hüft- bzw. Kniegelenksbelastung bei Coxarthrosepatienten besteht. Des Weiteren galt es zu prüfen (2) ob die radiologisch-anatomische Beinachse bei Coxarthrosepatienten einen Einfluss auf die Gelenksbelastung von Hüfte und Knie habe und (3) ob sich in diesem Zusammenhang Unterschiede zwischen unilateral und bilateral betroffenem Patientenkollektiv darstellen ließe.
Hintergründe: Es konnte bereits gezeigt werden, dass Coxarthrosepatienten ein verändertes Gangbild aufweisen. Diese Veränderungen sind mitverantwortlich für eine veränderete Gelenkbelastung sowohl in Knie als auch Hüfte, sowohl im Vergleich zwischen betroffener und nicht betroffener Seite als auch im Vergleich zu hüftgesunden Normprobanden. Insbesondere bezüglich der Kniegelenksbelastung konnte bei den Betroffenen ein reduziertes Knieadduktionsmoment (KAM) während der Standphase nachgewiesen werden. Dies steht im Einklang mit der Beobachtung, dass Coxarthrosepatienten vermehrt eine laterale Kniearthrose auf der ipsilateralen Seite entwickeln. Coxarthrosepatienten zeigten darüber hinaus eine veränderte Beinachse, mit einer Tendenz hin zu einer Valgusfehlstellung. In wieweit insbesondere die proximale Beinachse einen Einfluss auf die alterierten Gelenkmomente hat, bleibt bislang jedoch offen. Zu bilateralen Patienten sind darüber hinaus nur wenige Daten vorliegend.
Methoden: Um die oben genannten Hypothesen zu prüfen wurden bei insgesamt 29 Patienten, bei welchen die Indikation für eine Hüfttotalendoprothese aufgrund einer fortgeschrittenen Coxarthrose gestellt wurde, eine instrumentelle Bewegungsanalyse sowie eine biplanare EOSRöntgenaufnahme, welche in diesem Zusammenhang die konventionelle präoperative Planungsaufnahme ersetzte, durchgeführt. Anschließend wurden die Daten, die aus der 3D EOS Rekonstruktion über die Beinachse gewonnen wurden, sowie die Ergebnisse der Bewegungsanalyse mit bereits zuvor erhobenen Normdaten sowie untereinander verglichen und statistisch analysiert.
Ergebnisse: Es zeigte sich, dass die Patienten eine bis auf MFA nicht von der Norm abweichende Beinachse hatten. Die bilateralen zeigten eine signifikant größere Beinlängendifferenz als die Norm, die unilateralen Patienten jedoch diesbezüglich keine Abweichung. Insgesamt zeigten die Patienten ein verändertes Gangbild, welches weitgehend den bereits in der Literatur beschriebenen Veränderungen entsprach.
Bezüglich der Gelenkmomente zeigte sich ein im Vergleich zur Norm erniedrigtes KAM 2, die übrigen Gelenkmomente zeigten keine Abweichungen von der Norm.
Es zeigten sich weder für die Beinachse noch für die Gelenkmomente signifikante Unterschiede zwischen unilateralem und bilateralem Patientenkollektiv.
Die unilateralen Patienten als gesamtes zeigten keine Korrelationen mit der absoluten Beinlängendifferenz. In einer unilateralen Subgruppe, in der die betroffene Seite länger war, konnte eine inverse Korrelation mit HAM 1 und HAM 2 gefunden werden.
KAM 1 und KAM 2 korrelierten signifikant mit allen Beinachsenparametern. In einer schrittweisen Regressionsanalyse mit HKA, FO und MFA konnten damit 68% der KAM 1 Alterationen und 57,5 % der Veränderungen von KAM 2 erklärt werden. HAM 1 und HAM 2 zeigten jeweils eine signifikante Korrelation mit MFA.
Schlussfolgerung: Die vorliegenden Ergebnisse zeigen, dass die Beinachse, insbesondere das Offset, einen Teil der Veränderungen der Kniegelenksbelastung erklären. Eine intraoperative Vergrößerung des Offsets und damit eine Varisierung der Beinachse könnte möglicherweise so zu einer Normalisierung der Kniegelenksbelastung führen.
Ein neues kombiniertes pulse-wave Doppler- und M-Mode Ultraschallgerät zur semiinvasiven Messung des arteriellen Blutflusses (ABF) in der thorakalen Aorta descendens bei koronaren Revaskularisationseingriffen ohne Einsatz des kardiopulmonalen Bypass (off-pump coronary artery bypass grafting (OPCAB)) wurde untersucht und mit der Thermodilutionsmethode zur Bestimmung des cardiac output (CO) verglichen. Bei 45 Patienten, die sich einem OPCAB-Eingriff unterzogen, wurden ABF und CO gleichzeitig mittels des kombinierten Ultraschall-Dopplergerätes und der Thermodilutionsmethode bestimmt zu den folgenden fünf Messzeitpunkten: nach Anästhesieeinleitung (1), nach Sternotomie (2), nach Eröffnung des Perikards (3), nach Luxation des Herzens mittels eines speziellen Sperrersystems (4), nach Entfernen des Thoraxsperrers (5). Die Teilnehmer der Studie wurden in zwei Subgruppen unterteilt, entsprechend dem Typ des Thoraxsperrers, der eingesetzt wurde. Infolge einer Änderung des Operationsverfahrens wurden 15 Teilnehmer mittels des Octopus-Sperrers, 30 mittels des CTS-Sperrers revaskularisiert. Wir führten insgesamt 225 Messungen durch. Die Korrelationskoeffizienten und die linearen Regressionsgleichungen wurden berechnet. Alle Messungen und die Messwerte der Subgruppen getrennt wurden im Bland-Altman-Diagramm dargestellt. Der Korrelationskoeffizient für alleMesswerte war r = 0,66 (CO = 0,6508xABF+2,386) bzw. r = 0,66 (CO= 0,7499xABF+2,386) für dieOctopus-Sperrer Subgruppe und r = 0,65 (CO = 0,5583xABF+2,5281) für die CTS-Sperrer Subgruppe. Der Bias (Mittelwert der Differenzen) aller Messwerte betrug 0,8l/min (0,88l/min in der Octopus-Sperrer Subgruppe und 0,75l/min in der CTS-Sperrer Subgruppe) und die „limits of agreement“ (Mittelwert der Differenzen +-2SD)+-2,38l/min (2,07l/min in der Octopus-Sperrer Subgruppe und 2,52l/ min in der CTS-Sperrer Subgruppe). Demzufolge misst das Ultraschall-Dopplergerät den ABF während eines OPCAB-Eingriffes nicht mit ausreichender Genauigkeit. Weitere Untersuchungen in anderen klinischen Situationen sollten durchgeführt werden.
Driven by rapid technological advancements, the amount of data that is created, captured, communicated, and stored worldwide has grown exponentially over the past decades. Along with this development it has become critical for many disciplines of science and business to being able to gather and analyze large amounts of data. The sheer volume of the data often exceeds the capabilities of classical storage systems, with the result that current large-scale storage systems are highly distributed and are comprised of a high number of individual storage components. As with any other electronic device, the reliability of storage hardware is governed by certain probability distributions, which in turn are influenced by the physical processes utilized to store the information. The traditional way to deal with the inherent unreliability of combined storage systems is to replicate the data several times. Another popular approach to achieve failure tolerance is to calculate the block-wise parity in one or more dimensions. With better understanding of the different failure modes of storage components, it has become evident that sophisticated high-level error detection and correction techniques are indispensable for the ever-growing distributed systems. The utilization of powerful cyclic error-correcting codes, however, comes with a high computational penalty, since the required operations over finite fields do not map very well onto current commodity processors. This thesis introduces a versatile coding scheme with fully adjustable fault-tolerance that is tailored specifically to modern processor architectures. To reduce stress on the memory subsystem the conventional table-based algorithm for multiplication over finite fields has been replaced with a polynomial version. This arithmetically intense algorithm is better suited to the wide SIMD units of the currently available general purpose processors, but also displays significant benefits when used with modern many-core accelerator devices (for instance the popular general purpose graphics processing units). A CPU implementation using SSE and a GPU version using CUDA are presented. The performance of the multiplication depends on the distribution of the polynomial coefficients in the finite field elements. This property has been used to create suitable matrices that generate a linear systematic erasure-correcting code which shows a significantly increased multiplication performance for the relevant matrix elements. Several approaches to obtain the optimized generator matrices are elaborated and their implications are discussed. A Monte-Carlo-based construction method allows it to influence the specific shape of the generator matrices and thus to adapt them to special storage and archiving workloads. Extensive benchmarks on CPU and GPU demonstrate the superior performance and the future application scenarios of this novel erasure-resilient coding scheme.
During my thesis, I worked on two different membrane proteins. One is a bacterial secondary transporter and the second is a human mitochondrial calcium channel.
The first part of my thesis involves the structural and biochemical characterization of an L-carnitine/ γ-butyrobetaine antiporter from bacteria called CaiT. The aim of the project was to understand the Na+ independence of CaiT and to determine the crystal structures of CaiT in different conformations to expand the mechanistic understanding of substrate/ product antiport in CaiT.
The study revealed how a positively charged amino acid side chain (arginine 262) in CaiT could structurally and functionally mimic a sodium ion. Additionally, various crystal structures of CaiT obtained in this study demonstrate that the central substrate-binding site is highly dynamic and can accommodate the substrate in various orientations.
In the second part of my thesis, I was able to optimize the expression and purification conditions for the human mitochondrial calcium uniporter or the MCU. Understanding how this channel functions can help us unravel the mechanism of calcium uptake by mitochondria. Secondary structure prediction analysis in combination with mass spectrometry of degraded MCU products obtained during the purification of the full-length protein led to the identification of a stable MCU construct. This study resulted in the successful purification of milligram quantities of stable MCU protein for the first time. Further optimization may be required to obtain more homogenous protein that is amenable for crystallization.
