In plants, a family of more than 20 heat stress transcription factors (Hsf) controls the expression of heat stress (hs) genes. There is increasing evidence for the functional diversification between individual members of the Hsf family fulfilling distinct roles in response to various environmental stress conditions and developmental signals. In response to hs, accumulation of both heat stress proteins (Hsp) and Hsfs is induced. In tomato, the physical interaction between the constitutively expressed HsfA1 and the hs-inducible HsfA2 results in synergistic transcriptional activation (superactivation) of hs gene expression. Here, we show that the interaction is strikingly specific and not observed with other class A Hsfs. Hetero-oligomerization of the two-component Hsfs is preferred to homo-oligomerization, and each Hsf in the HsfA1/HsfA2 hetero-oligomeric complex has its characteristic contribution to its function as superactivator. Distinct regions of the oligomerization domain are responsible for specific homo- and hetero-oligomeric interactions leading to the formation of hexameric complexes. The results are summarized in a model of assembly and function of HsfA1/A2 superactivator complexes in hs gene regulation.
Immunsuppressiva sind essentiell für die allogene Organtransplantation. Graff et al konnten in drei vorangegangenen Studien zeigen, dass Immunsuppressiva zu einer verstärkten Thrombozytenaktivierung führen. Diese ist für die verschiedenen Substanzen unterschiedlich stark ausgeprägt. Eine verstärkte Thrombozytenaktivierung könnte zu vermehrten kardiovaskulären Ereignissen führen. Insbesondere bei nierentransplantierten Patienten ist das kardiovaskuläre Risiko ohnehin deutlich erhöht. Es stellt sich die Frage ob diese medikamentös induzierte Plättchenaktivierung als Ko-Faktor kardiovaskulärer Ereignisse bei nierentransplantierten anzusehen ist. Im Vorfeld dieser Studie wurden in den Studien NTX, RAPA und PLANT bei insgesamt 176 nierentransplantierten Patienten mit einer Mono-Immunsuppression flowzytometrische Plättchenfunktionsmessungen durchgeführt, dabei wurden insbesondere der Degranulationsmarker CD62 und der aktivierte GP IIb/IIIa-Rezeptor PAC-1 gemessen. Diese Ergebnisse wurden in Relation zum Mittelwert der jeweiligen Kontrollgruppen gesetzt: Messwert(Patient) / Mittelwert(Gesunde_Kontrolle) Diese relativen Plättchenfunktionswerte waren zwischen den Studien vergleichbar. Des Weiteren wurden epidemiologische Eckdaten und klinische Endpunkte bis zu 5 Jahre nach der Thrombozytenmessung erfasst, im Mittel 3,7 Jahre. Aufgrund des Studiendesigns der vorangegangenen Studien wurde hier ein Kollektiv mit relativ niedrigem kardiovaskulärem Risiko untersucht, jedoch sollte durch diese Reduktion klassischer Risikofaktoren der Einfluss der Plättchenaktivität besser sichtbar sein. Die retrospektive Analyse untersuchte ob die Plättchenfunktionsparameter CD62 und PAC-1 als prädiktiver Marker für kardiovaskuläre Ereignisse geeignet sind. Insgesamt traten seit den Messungen 21 kardiovaskuläre Ereignisse auf, davon 9 akute. Es gab keine signifikanten Unterschiede zwischen den Werten bei Patienten mit Ereignissen und denen ohne Ereignis: Bei CD62 (baseline) lag der Mittelwert bei 2,82±1,9 mit Ereignis vs. 2,73±1,47 ohne, für PAC-1 bei 1,76 ±1,97 vs. 1,93±1,53. Nach Aktivierung mit TRAP lag der Mittelwert für CD62 bei Ereignispatienten bei 1,57±0,88 vs. 1,76±0,83, für PAC-1 bei 1,56±1,65 vs. 1,44±1,04. In einem weiteren Schritt wurden die Patienten nach dem Ausmaß der Plättchenaktivierung klassifiziert, als Cut-Off-Werte wurden die Mittelwerte und Mediane der jeweiligen Plättchenfunktion aller Patienten gewählt, ferner noch willkürlich eine 3- bzw 5-fache Aktivierung gegenüber den gesunden Kontrollen. Hierbei wurden in den Gruppen mit hohen Funktionswerten nicht signifikant mehr Ereignisse beobachtet als in den Gruppen mit niedrigen Werten. In einer daraufhin durchgeführten Time-to-event-Analyse nach Kaplan-Meyer zeigte sich allerdings für CD62 (baseline) > 5 ein früheres Eintreten kardiovaskulärer Ereignisse, im Mittel 47,6 vs. 69,9 Monate, p = 0,05. Als Risikofaktoren in dieser Studie erwiesen sich insbesondere das Alter und das Alter bei Transplantation, letzteres erwies sich in einer logistischen Regressionsanalyse als der wichtigster Faktor. Dies ist nach unserem Wissen die erste Untersuchung, die den prädiktiven Wert von Thrombozyten-gebundenem CD62 sowie von PAC-1 für kardiovaskuläre Ereignisse evaluiert. Es ergibt sich kein Hinweis, dass diese Marker einen prädiktiven Wert haben. Somit erscheint auch eine direkte pharmakologische Beeinflussung der Thrombozytenfunktion oder eine Veränderung der Immunsuppression nicht wegweisend, um die Inzidenz kardiovaskulärer Ereignisse bei nierentransplantierten Patienten zu senken.
Auswirkungen einer Mutation des Adhäsionrezeptors PSGL-1 bei humanen neutrophilen Granulozyten

Der VNTR Polymorphismus des PSGL-1 ist schon seit längerer Zeit Gegenstand wissenschaftlicher Untersuchungen. Der Rezeptor weist je nach Genotyp eine unterschiedliche Länge auf. Dabei ist der Rezeptor mit dem Allel A der längste, das C-Allel weist phänotypisch den kürzesten Rezeptor auf. Der Rezeptor mit dem B-Allel liegt mit der Länge in der Mitte der drei Rezeptoren. In dieser Studie wurde das Roll- und das Adhäsionsverhalten von Leukozyten mit den Genotypen AA bzw. BB mit Hilfe eines Flusskammermodells untersucht. Dieses Verhalten kommt im Rahmen der Bindung zustande, welche P-Selektin und PSGL-1 miteinander eingehen. Das Blut von acht Probanden wurde untersucht, vier Probanden wiesen den Genotyp AA, die anderen vier den Genotyp BB auf. Die Leukozyten wurden aus dem Vollblut isoliert, verdünnt und in das Flusskammermodell eingespeist. Die verdünnte Zellsuspension wurde mittels einer Pumpe über ein Schlauchsystem durch eine Messkammer befördert, welche im Vorfeld mit P-Selektin beschichtet wurde. Dort konnten die Zellen mikroskopisch untersucht und videotechnisch dokumentiert werden. Anschließend konnte die Rollgeschwindigkeit der neutrophilen Granulozyten und die Anzahl der adhärenten Zellen an der Oberfläche durch Computerprogramme ermittelt werden. Als Resultat ergab sich ein signifikanter Unterschied zwischen der Rollgeschwindigkeit der Zellen des Genotyps AA und der Zellen des Genotyps BB. Dabei wiesen die Zellen mit dem B-Allel wegen der geringeren Länge des PSGL-1 eine höhere Rollgeschwindigkeit auf, als die Zellen mit dem AAllel, deren PSGL-1 länger ist. Zudem blieben signifikant weniger Leukozyten mit dem BAllel an der Oberfläche haften als die Leukozyten der anderen Gruppe. Daraus lässt sich schließen, dass die Bindungskraft zwischen PSGL-1 und PSelektin von der Länge des PSGL-1 abhängig ist. Der kürzere Rezeptor (B-Allel des PSGL-1) weist also eine geringere Bindungskraft auf als der längere Rezeptor (A-Allel des PSGL-1). Die Bindung zwischen PSGL-1 und PSelektin lässt sich wahrscheinlich pharmakologisch beeinflussen. Es wurden Antikörper entwickelt, die in klinischen Studien Erfolg versprechende Ansätze bei der Entzündungshemmung gezeigt haben. Die aktuelle Literatur zu klinisch pharmakologischen Behandlungsansätzen wurde in dieser Arbeit diskutiert.
Very little is known about the occlusal wear pattern in the Neanderthal posterior dentition. Usually dental wear is closely related to the physical properties of the ingested food, and consequently can be used to obtain information about diet. Neanderthal dietary reconstructions have been mostly based on the analysis of accompanying faunal remains and isotopic signatures of bones and tooth enamel, suggesting that they exploited larger portions of animal proteins from large and medium-sized herbivores. Probably these studies may do not reflect the bulk diet, tending to underestimate plant consumption and to overestimate meat consumption. In the present work the occlusal wear pattern of maxillary molars of Homo neanderthalensis (N=19) and early Homo sapiens (N=12)have been analyzed, applying non-destructive methods based on virtual three-dimensional polygonal models generated from surface scanning of dental casts. The sample groups occupied different geographical areas at different chronological times. The 3D digital tooth models were analyzed using the “Occlusal Fingerprint Analysis” (OFA) method (Kullmer et al. 2009), describing and quantifying the occlusal wear pattern derived from two wear facet angles (dip and dip direction), wear facet area and occlusal relief index (ORI). The OFA method provides information about the dynamics of the occlusal relationships and their function, permitting the reconstruction of the mandibular movements responsible for the contacts created during the chewing cycle. Since jaw movements and diet are closely related, the results obtained, can be used to interpret the diet of the two Pleistocene hominin species. In order to evaluate how dietary differences influence the occlusal wear pattern, upper molars of modern hunter-gatherers (N=42) with known diet and different dietary habits, have been included in the sample and compared with those of Neanderthals and early Homo sapiens. Results show that within the modern hunter-gatherers sample, the occlusal wear pattern of carnivorous populations differs from those who relied on a mixed-diet. In particular, the study of relative facet areas clearly distinguish meat-eaters from mixed-diet hunter-gatherers, while ORI results and wear facet inclinations (dip angle) seem to reflect directly the abrasiveness of the diet, including the influence of exogenous materials during food preparation. The Neanderthal occlusal wear pattern is characterized by an ecogeographic variation, suggesting the exploitation of different food resources. In particular Neanderthals who inhabited relatively warm environments of southern Europe and the Near East exhibit an occlusal wear pattern different from those of meat-eaters hunter-gatherers from tempered and cooler regions, displaying some features similar to those of Bushmen. These results suggest the exploitation of a broad variety of food sources. The analysis of the occlusal wear pattern in Neanderthals and early Homo sapiens who inhabited Europe during the cooler Oxygen Isotope Stage 3 (OIS3) shows many similarities between the two hominid species. These results indicate the exploitation of similar and low-diversified food sources, based mostly on the consumption of animal proteins, as suggested through the clear similarities with the wear patterns found in modern meat-eaters hunter-gatherers. In both studied groups, Neanderthals and early Homo sapiens the occlusal wear pattern is characterized by high ORI and dip angle values, suggesting the intake of a low-abrasive diet, probably due to the absence of sophisticated food preparation techniques introducing external silica grains, e.g. from soil (grinding of seeds) or plant cells, as those, seen in modern hunter-gatherer populations. The analysis of the occlusal fingerprints in Neanderthal and early European Homo sapiens upper molars suggests that both species followed very similar adaptive dietary strategies, based on a distinctive versatility and flexibility in the daily diet, depending on availability of resources according to environmental circumstances.
The C-module-binding factor (CbfA) is a multidomain protein that belongs to the family of jumonji-type (JmjC) transcription regulators. In the social amoeba Dictyostelium discoideum, CbfA regulates gene expression during the unicellular growth phase and multicellular development. CbfA and a related D. discoideum CbfA-like protein, CbfB, share a paralogous domain arrangement that includes the JmjC domain, presumably a chromatin-remodeling activity, and two zinc finger-like (ZF) motifs. On the other hand, the CbfA and CbfB proteins have completely different carboxy-terminal domains, suggesting that the plasticity of such domains may have contributed to the adaptation of the CbfA-like transcription factors to the rapid genome evolution in the dictyostelid clade. To support this hypothesis we performed DNA microarray and real-time RT-PCR measurements and found that CbfA regulates at least 160 genes during the vegetative growth of D. discoideum cells. Functional annotation of these genes revealed that CbfA predominantly controls the expression of gene products involved in housekeeping functions, such as carbohydrate, purine nucleoside/nucleotide, and amino acid metabolism. The CbfA protein displays two different mechanisms of gene regulation. The expression of one set of CbfA-dependent genes requires at least the JmjC/ZF domain of the CbfA protein and thus may depend on chromatin modulation. Regulation of the larger group of genes, however, does not depend on the entire CbfA protein and requires only the carboxy-terminal domain of CbfA (CbfA-CTD). An AT-hook motif located in CbfA-CTD, which is known to mediate DNA binding to A+T-rich sequences in vitro, contributed to CbfA-CTD-dependent gene regulatory functions in vivo.
Modelling protein flexibility and plasticity is computationally challenging but important for understanding the function of biological systems. Furthermore, it has great implications for the prediction of (macro) molecular complex formation. Recently, coarse-grained normal mode approaches have emerged as efficient alternatives for investigating large-scale conformational changes for which more accurate methods like MD simulation are limited due to their computational burden. We have developed a Normal Mode based Simulation (NMSim) approach for efficient conformation generation of macromolecules. Combinations of low energy normal modes are used to guide a simulation pathway, whereas an efficient constraints correction approach is applied to generate stereochemically allowed conformations. Non-covalent bonds like hydrogen bonds and hydrophobic tethers and phi-psi favourable regions are also modelled as constraints. Conformations from our approach were compared with a 10 ns MD trajectory of lysozyme. A 2-D RMSD plot shows a good overlap of conformational space, and rms fluctuations of residues show a correlation coefficient of 0.78 between the two sets of conformations. Furthermore, a comparison of NMSim simulations starting from apo structures of different proteins show that ligand-bound conformations can be sampled for those cases where conformational changes are mainly correlated, e.g., domain-like motion in adenylate kinase. Efforts are currently being made to also model localized but functionally important motions for protein binding pockets and protein-protein interfaces using relevant normal mode selection criteria and implicit rotamer basin creation.
This article shows that investors financing a portfolio of projects may use the depth of their financial pockets to overcome entrepreneurial incentive problems. Competition for scarce informed capital at the refinancing stage strengthens investors’ bargaining positions. And yet, entrepreneurs’ incentives may be improved, because projects funded by investors with ‘‘shallow pockets’’ must have not only a positive net present value at the refinancing stage, but one that is higher than that of competing portfolio projects. Our article may help understand provisions used in venture capital finance that limit a fund’s initial capital and make it difficult to add more capital once the initial venture capital fund is raised. This paper shows that active investors, such as venture capitalists, can affect the speed at which new ventures grow. In the absence of product market competition, new ventures financed by active investors grow faster initially, though in the long run those financed by passive investors are able to catch up. By contrast, in a competitive product market, new ventures financed by active investors may prey on rivals that are financed by passive investors by “strategically overinvesting” early on, resulting in long-run differences in investment, profits, and firm growth. The value of active investors is greater in highly competitive industries as well as in industries with learning curves, economies of scope, and network effects, as is typical for many “new economy” industries. For such industries, our model predicts that start-ups with access to venture capital may dominate their industry peers in the long run. In der vorliegenden Arbeit wurden zwei motivationale Erklärungsmodelle, Zielorientierungen und das kognitiv-motivationale Prozessmodell, im Rahmen des selbstregulierten Lernens integriert. Selbstregulationsprozesse sind nach Carver und Scheier (1981) zyklisch angelegt. Zu den Bestandteilen der Selbstregulation gehören nach Boekaerts (1999)Regulation der Informationsverarbeitung, Regulation des Lernprozesses und Regulation des Selbsts. Auf dieser Ebene sind die Zielorientierungen angesiedelt. In dieser Arbeit das 2 x 2 Modell von Elliot und McGregor (2001) herangezogen. Es berücksichtigt zwei Dimensionen, die Valenz (Annäherungs- und Vermeidungskomponente) und die Kompetenz (Vergleichsmaßstab intern und extern). Hieraus resultieren: 1) Lern-Annäherungs-Ziele (positive Valenz & interner Vergleich, 2) Lern-Vermeidungs-Ziele (negative Valenz & interner Vergleich), 3) Leistungs-Annäherungs-Ziele (positive Valenz & externer Vergleich) und 4) Leistungs-Vermeidungs-Ziele (negative Valenz & externer Vergleich). Diese vier Zielorientierungen sind der erste Teil des integrierten Modells, der zweite zentrale Teil ist das kognitiv-motivationale Prozessmodell (Vollmeyer & Rheinberg, 1999, 2006). Ausgangspunkt ist die aktuelle Motivation, die aus 1) der Erfolgswahrscheinlichkeit, 2) der Misserfolgsbefürchtung, 3) dem Interesse und 4) der Herausforderung als unabhängige Faktoren besteht. Diese aktuelle Motivation wirkt nicht direkt auf die Leistung, sondern durch kognitive (hier: Metakognition) und motivationale (hier: Flow-Erleben) Mediatoren. Es wurden drei Fragestellungen bearbeitet. Die erste Fragestellung betraf das Zusammenspiel der Zielorientierungen und der aktuellen Motivation. Eine Überprüfung dieses integrierten Motivationsmodell erfolgte mittels Pfadanalyse. Die zweite und dritte Fragestellung befasste sich spezifisch mit dem kognitiv-motivationalen Prozessmodell. Es wurden Subgruppen auf der Basis der aktuellen Motivation identifiziert und ihre Bedeutung für die Mediatoren und die Leistung untersucht. Die dritte Fragestellung fokussierte den Prozesscharakter des Modells. Hier erfolgten Analysen über den Arbeitsprozess hinweg. Die Fragebögen (Achievement Goal Questionnaire (Elliot & McGregor, 2001), Metakognitionsfragebogen (aufbauend auf LIST (Wild, 2000) und Metacognitive Awareness Inventory (Schraw & Dennison, 1994), Fragebogen zur aktuellen Motivation (Rheinberg, Vollmeyer & Burns, 2001), Flow-Kurz-Skala (Rheinberg, Vollmeyer & Engeser, 2003)) und die Problemlöseaufgaben (Sudokus) wurden in einer Pilotstudie getestet. An der Hauptstudie nahmen 202 Personen teil (73% weiblich). Das integrierte Motivationsmodell geht davon aus, dass eine positiver Effekt der Zielorientierungen als Personenvariable auf die aktuelle Motivation besteht. Die aktuelle Motivation als Startpunkt des situationalen Geschehens wiederum wirkt positiv auf die Mediatoren Flow-Erleben und Metakognition, hat aber keinen direkten Effekt auf die Leistung. Dieser wird nämlich über die Mediatoren vermittelt. Deswegen wird ein positiver Effekt des Flow-Erlebens und der Metakognition auf die Leistung erwartet. Das Zusammenwirken der beiden Mediatoren kann aus dem kognitiv-motivationalen Prozessmodell nicht abgeleitet werden. Ob ein Zusammenhang besteht oder nicht wird geprüft. Für das Vorwissen wird ein positiver Effekt auf die aktuelle Motivation und die Leistung erwartet. Die Pfadanalyse zur Überprüfung des integrierten Motivationsmodells zeigte, dass eine Integration nicht nur theoretisch sinnvoll ist, sondern auch empirisch gestützt wird. Die Bedeutung der Metakognition als kognitiver Mediator wurde gezeigt. Die zweite Fragestellung fokussierte auf die aktuelle Motivation. Auf der Basis dieser wurden mittels Clusteranalyse drei distinkte Gruppen identifiziert: hoch Motivierte, niedrig Motivierte und ängstlich Motivierte (vergleichbar zu Vollmeyer & Rheinberg, 2004). Diese Gruppen unterschieden sich hinsichtlich ihres Flow-Erlebens, ihrer Metakognition und ihrer Leistung. Die hoch Motivierten erlebten am meisten Flow, berichteten mehr Metakognition und zeigten bessere Leistung. Der Prozesscharakter des kognitiv-motivationalen Prozessmodells stand im Mittelpunkt der dritten Fragestellung. Diese Analyse über die drei Sudokus hinweg zeigte eine ähnliche Entwicklung des Flow-Erlebens bei den hoch und niedrig Motivierten. Vom ersten zum zweiten Sudoku stieg es an, um dann wieder abzufallen. Dies war bei der Metakognition nicht der Fall. Die hoch Motivierten berichteten einen stärkeren Rückgang der Metakognition beim dritten Sudoku als die niedrig Motivierten. Bei der Leistung zeigte sich für die hoch Motivierten ein Anstieg beim zweiten Sudoku und dann ein Rückgang beim dritten, wohingegen die Leistung der niedrig Motivierten kontinuierlich abfiel. Abschließend wurden die Schwierigkeiten und Grenzen der vorliegenden Arbeit diskutiert und Implikationen der Ergebnisse für die Theorieentwicklung und ihre Bedeutung für den Anwendungskontext aufgezeigt.
We have investigated the role of reactive oxygen species and thiol-oxidizing agents in the induction of cell death and have shown that adenocarcinoma gastric (AGS) cells respond differently to the oxidative challenge according to the signaling pathways activated. In particular, apoptosis in AGS cells is induced via the mitochondrial pathway upon treatment with thiol-oxidizing agents, such as diamide. Apoptosis is associated with persistent oxidative damage, as evidenced by the increase in carbonylated proteins and the expression/activation of DNA damage-sensitive proteins histone H2A.X and DNA-dependent protein kinase. Resistance to hydrogen peroxide is instead associated with Keap1 oxidation and rapid translocation of Nrf2 into the nucleus. Sensitivity to diamide and resistance to hydrogen peroxide are correlated with GSH redox changes, with diamide severely increasing GSSG, and hydrogen peroxide transiently inducing protein-GSH mixed disulfides. We show that p53 is activated in response to diamide treatment by the oxidative induction of the Trx1/p38(MAPK) signaling pathway. Similar results were obtained with another carcinoma cell line, CaCo2, indicating that these findings are not limited to AGS cells. Our data suggest that thiol-oxidizing agents could be exploited as inducers of apoptosis in tumor histotypes resistant to ROS-producing chemotherapeutics.
Macrophages ingesting apoptotic cells attenuate inflammatory responses, such as reactive oxygen species (ROS) generation. In atherosclerosis, ongoing inflammation and accumulation of apoptotic/necrotic material are observed, suggesting defects of phagocytes in recognizing or responding to dying cells. Modified lipoproteins such as oxidized LDL (oxLDL) are known to promote inflammation and to interfere with apoptotic cell clearance. Here, we studied the impact of cells exposed to oxLDL on their ability to interfere with the oxidative burst in phagocytes. In contrast to apoptotic cells, cells dying in response to or in the presence of oxLDL failed to suppress ROS generation despite efficiently being taken up by phagocytes. In addition, apoptotic cells, but not oxLDL-treated cells, inhibited phosphorylation of extracellular signal-regulated kinase, which is important for NADPH oxidase activation. oxLDL treatment did not interfere with activation of the antiinflammatory transcriptional regulator peroxisome proliferator-activated receptor gamma by apoptotic cells. Moreover, cells exposed to oxLDL failed to suppress lipopolysaccharide- induced proinflammatory cytokine expression, whereas apoptotic cells attenuated these phagocyte responses. Thus, the presence of oxLDL during cell death impaired the ability of apoptotic cells to act antiinflammatory with regard to oxidative burst inhibition and cytokine expression in phagocytes.
Specific functions of biological systems often require conformational transitions of macromolecules. Thus, being able to describe and predict conformational changes of biological macromolecules is not only important for understanding their impact on biological function, but will also have implications for the modelling of (macro)molecular complex formation and in structure-based drug design approaches. The “conformational selection model” provides the foundation for computational investigations of conformational fluctuations of the unbound protein state. These fluctuations may reveal conformational states adopted by the bound proteins. The aim of this work is to incorporate directional information in a geometry-based approach, in order to sample biologically relevant conformational space extensively. Interestingly, coarse-grained normal mode (CGNM) approaches, e.g., the elastic network model (ENM) and rigid cluster normal mode analysis (RCNMA), have emerged recently and provide directions of intrinsic motions in terms of harmonic modes (also called normal modes). In my previous work and in other studies it has been shown that conformational changes upon ligand binding occur along a few low-energy modes of unbound proteins and can be efficiently calculated by CGNM approaches. In order to explore the validity and the applicability of CGNM approaches, a large-scale comparison of essential dynamics (ED) modes from molecular dynamics (MD) simulations and normal modes from CGNM was performed over a dataset of 335 proteins. Despite high coarse-graining, low frequency normal modes from CGNM correlate very well with ED modes in terms of directions of motions (average maximal overlap is 0.65) and relative amplitudes of motions (average maximal overlap is 0.73). In order to exploit the potential of CGNM approaches, I have developed a three-step approach for efficient exploration of intrinsic motions of proteins. The first two steps are based on recent developments in rigidity and elastic network theory. Initially, static properties of the protein are determined by decomposing the protein into rigid clusters using the graph-theoretical approach FIRST at an all-atom representation of the protein. In a second step, dynamic properties of the molecule are revealed by the rotations-translations of blocks approach (RTB) using an elastic network model representation of the coarse-grained protein. In the final step, the recently introduced idea of constrained geometric simulations of diffusive motions in proteins is extended for efficient sampling of conformational space. Here, the low-energy (frequency) normal modes provided by the RCNMA approach are used to guide the backbone motions. The NMSim approach was validated on hen egg white lysozyme by comparing it to previously mentioned simulation methods in terms of residue fluctuations, conformational space explorations, essential dynamics, sampling of side-chain rotamers, and structural quality. Residue fluctuations in NMSim generated ensemble is found to be in good agreement with MD fluctuations with a correlation coefficient of around 0.79. A comparison of different geometry-based simulation approaches shows that FRODA is restricted in sampling the backbone conformational space. CONCOORD is restricted in sampling the side-chain conformational space. NMSim sufficiently samples both the backbone and the side-chain conformations taking experimental structures and conformations from the state of the art MD simulation as reference. The NMSim approach is also applied to a dataset of proteins where conformational changes have been observed experimentally, either in domain or functionally important loop regions. The NMSim simulations starting from the unbound structures are able to reach conformations similar to ligand bound conformations (RMSD < 2.4 Å) in 4 out of 5 cases of domain moving proteins. In these four cases, good correlation coefficients (R > 0.7) between the RMS fluctuations derived from NMSim generated structures and two experimental structures are observed. Furthermore, intrinsic fluctuations in NMSim simulation correlate with the region of loop conformational changes observed upon ligand binding in 2 out of 3 cases. The NMSim generated pathway of conformational change from the unbound structure to the ligand bound structure of adenylate kinase is validated by a comparison to experimental structures reflecting different states of the pathway as proposed by previous studies. Interestingly, the generated pathway confirms that the LID domain closure precedes the closing of the NMPbind domain, even if no target conformation is provided in NMSim. Hence, the results in this study show that, incorporating directional information in the geometry-based approach NMSim improves the sampling of biologically relevant conformational space and provides a computationally efficient alternative to state of the art MD simulations.
Background Gamma-aminobutyric acid (GABA) is an important inhibitory neurotransmitter which mainly mediates its effects on neurons via ionotropic (GABAA) and metabotropic (GABAB) receptors. GABAB receptors are widely expressed in the central and the peripheral nervous system. Although there is evidence for a key function of GABAB receptors in the modulation of pain, the relative contribution of peripherally- versus centrally-expressed GABAB receptors is unclear. Results In order to elucidate the functional relevance of GABAB receptors expressed in peripheral nociceptive neurons in pain modulation we generated and analyzed conditional mouse mutants lacking functional GABAB(1) subunit specifically in nociceptors, preserving expression in the spinal cord and brain (SNS-GABAB(1)-/- mice). Lack of the GABAB(1) subunit precludes the assembly of functional GABAB receptor. We analyzed SNS-GABAB(1)-/- mice and their control littermates in several models of acute and neuropathic pain. Electrophysiological studies on peripheral afferents revealed higher firing frequencies in SNS-GABAB(1)-/- mice compared to corresponding control littermates. However no differences were seen in basal nociceptive sensitivity between these groups. The development of neuropathic and chronic inflammatory pain was similar across the two genotypes. The duration of nocifensive responses evoked by intraplantar formalin injection was prolonged in the SNS-GABAB(1)-/- animals as compared to their control littermates. Pharmacological experiments revealed that systemic baclofen-induced inhibition of formalin-induced nociceptive behaviors was not dependent upon GABAB(1) expression in nociceptors. Conclusion This study addressed contribution of GABAB receptors expressed on primary afferent nociceptive fibers to the modulation of pain. We observed that neither the development of acute and chronic pain nor the analgesic effects of a systematically-delivered GABAB agonist was significantly changed upon a specific deletion of GABAB receptors from peripheral nociceptive neurons in vivo. This lets us conclude that GABAB receptors in the peripheral nervous system play a less important role than those in the central nervous system in the regulation of pain.
Diabetic nephropathy (DN) is a major cause of end-stage renal failure worldwide. Oxidative stress has been reported to be a major culprit of the disease and increased oxidized low density lipoprotein (oxLDL) immune complexes were found in patients with DN. In this study we present evidence, that CXCL16 is the main receptor in human podocytes mediating the uptake of oxLDL. In contrast, in primary tubular cells CD36 was mainly involved in the uptake of oxLDL. We further demonstrate that oxLDL down-regulated α3-integrin expression and increased the production of fibronectin in human podocytes. In addition, oxLDL uptake induced the production of reactive oxygen species (ROS) in human podocytes. Inhibition of oxLDL uptake by CXCL16 blocking antibodies abrogated the fibronectin and ROS production and restored α3 integrin expression in human podocytes. Furthermore we present evidence that hyperglycaemic conditions increased CXCL16 and reduced ADAM10 expression in podocytes. Importantly, in streptozotocin-induced diabetic mice an early induction of CXCL16 was accompanied by higher levels of oxLDL. Finally immunofluorescence analysis in biopsies of patients with DN revealed increased glomerular CXCL16 expression, which was paralleled by high levels of oxLDL. In summary, regulation of CXCL16, ADAM10 and oxLDL expression may be an early event in the onset of DN and therefore all three proteins may represent potential new targets for diagnosis and therapeutic intervention in DN.
We study a model of “information-based entrenchment” in which the CEO has private information that the board needs to make an efficient replacement decision. Eliciting the CEO’s private information is costly, as it implies that the board must pay the CEO both higher severance pay and higher on-the-job pay. While higher CEO pay is associated with higher turnover in our model, there is too little turnover in equilibrium. Our model makes novel empirical predictions relating CEO turnover, severance pay, and on-the-job pay to firm-level attributes such as size, corporate governance, and the quality of the firm’s accounting system.
This paper argues that banks must be sufficiently levered to have first-best incentives to make new risky loans. This result, which is at odds with the notion that leverage invariably leads to excessive risk taking, derives from two key premises that focus squarely on the role of banks as informed lenders. First, banks finance projects that they do not own, which implies that they cannot extract all the profits. Second, banks conduct a credit risk analysis before making new loans. Our model may help understand why banks take on additional unsecured debt, such as unsecured deposits and subordinated loans, over and above their existing deposit base. It may also help understand why banks and finance companies have similar leverage ratios, even though the latter are not deposit takers and hence not subject to the same regulatory capital requirements as banks.
Oral presentation from 4th International Conference of cGMP Generators, Effectors and Therapeutic Implications ; Regensburg, Germany. 19–21 June 2009 Background: An exaggerated pain sensitivity is the dominant feature of inflammatory and neuropathic pain both in the clinical setting and in experimental animal models. It manifests as pain in response to normally innocuous stimuli (allodynia), increased response to noxious stimuli (hyperalgesia) or spontaneous pain, and can persist long after the initial injury is resolved. Research over the last decades has revealed that several signaling pathways in the spinal cord essentially contribute to the pain sensitization. To test the contribution of cGMP produced by NO-sensitive guanylyl cyclase (NO-GC) to pain sensitization, we investigated the localization of NO-GC in the spinal cord and in dorsal root ganglia, and we characterized the nociceptive behavior of mice deficient in NO-GC (GC-KO mice). Results: We show that NO-GC (β1 subunit) is distinctly expressed in neurons of the mouse spinal cord, while its distribution in dorsal root ganglia is restricted to non-neuronal cells. GC-KO mice exhibited a considerably reduced nociceptive behavior in models of inflammatory or neuropathic pain, but their responses to acute pain were not impaired. Moreover, GC-KO mice failed to develop pain sensitization induced by spinal administration of drugs releasing NO. Surprisingly, during spinal nociceptive processing cGMP produced by NO-GC may activate signaling pathways different from cGMP-dependent protein kinase I (cGKI), while cGKI can be activated by natriuretic peptide receptor-B (NPR-B) dependent cGMP production. Conclusion: Taken together, our results provide evidence that NO-GC has a dominant role in the development of exaggerated pain sensitivity during inflammatory and neuropathic pain. Furthermore, beside the NO-mediated cGMP synthesis, cGMP produced by NPR-B contributes to pain sensitization by activation of cGKI.
The small leucine-rich proteoglycans (SLRPs) are biologically active components of the extracellular matrix (ECM), consisting of a protein core with leucine rich-repeat (LRR) motifs covalently linked to glycosaminoglycan (GAG) side chains. The diversity in composition resulting from the various combinations of protein cores substituted with one or more GAG chains along with their pericellular localization enables SLRPs to interact with a host of different cell surface receptors, cytokines, growth factors, and other ECM components, leading to modulation of cellular functions. SLRPs are capable of binding to: (i) different types of collagens, thereby regulating fibril assembly, organization, and degradation; (ii) Toll-like receptors (TLRs), complement C1q, and tumor necrosis factor-alpha (TNFα), regulating innate immunity and inflammation; (iii) epidermal growth factor receptor (EGF-R), insulin-like growth factor receptor (IGF-IR), and c-Met, influencing cellular proliferation, survival, adhesion, migration, tumor growth and metastasis as well as synthesis of other ECM components; (iv) low-density lipoprotein receptor-related protein (LRP-1) and TGF-β, modulating cytokine activity and fibrogenesis; and (v) growth factors such as bone morphogenic protein (BMP-4) and Wnt-I-induced secreted protein-1 (WISP-1), controlling cell proliferation and differentiation. Thus, the ability of SLRPs, as ECM components, to directly or indirectly regulate cell-matrix crosstalk, resulting in the modulation of various biological processes, aptly qualifies these compounds as matricellular proteins.
The manifestation of chronic back pain depends on structural, psychosocial, occupational and genetic influences. Heritability estimates for back pain range from 30% to 45%. Genetic influences are caused by genes affecting intervertebral disc degeneration or the immune response and genes involved in pain perception, signalling and psychological processing. This inter-individual variability which is partly due to genetic differences would require an individualized pain management to prevent the transition from acute to chronic back pain or improve the outcome. The genetic profile may help to define patients at high risk for chronic pain. We summarize genetic factors that (i) impact on intervertebral disc stability, namely Collagen IX, COL9A3, COL11A1, COL11A2, COL1A1, aggrecan (AGAN), cartilage intermediate layer protein, vitamin D receptor, metalloproteinsase-3 (MMP3), MMP9, and thrombospondin-2, (ii) modify inflammation, namely interleukin-1 (IL-1) locus genes and IL-6 and (iii) and pain signalling namely guanine triphosphate (GTP) cyclohydrolase 1, catechol-O-methyltransferase, μ opioid receptor (OPMR1), melanocortin 1 receptor (MC1R), transient receptor potential channel A1 and fatty acid amide hydrolase and analgesic drug metabolism (cytochrome P450 [CYP]2D6, CYP2C9).
Through the use of information about the biological target structure, the optimization of potential drugs can be improved. In this work I have developed a procedure that uses the quantitative change in the chemical perturbations (CSP) in the protein from NMR experiments for driving protein-ligand docking. The approach is based on a hybrid scoring function (QCSPScore) which combines traditional DrugScore potentials, which describe the interaction between protein and ligand, with Kendall’s rank correlation coefficient, which evaluates docking poses in terms of their agreement with experimental CSP. Prediction of the CSP for a specific ligand pose is done efficiently with an empirical model, taking into account only ring current effects. QCSPScore has been implemented in the AutoDock software package. Compared to previous methods, this approach shows that the use of rank correlation coefficient is robust to outliers. In addition, the prediction of native-like complex geometries improved because the CSP are already being used during the docking process, and not only in a post-filtering setting for generated docking poses. Since the experimental information is guaranteed to be quantitatively used, CSP effectively contribute to align the ligand in the binding pocket. The first step in the development of QCSPScore was the analysis of 70 protein-ligand complexes for which reference CSP were computed. The success rate in the docking increased from 71% without involvement of CSP to 100% if CSP were considered at the highest weighting scheme. In a second step QCSPScore was used in re-docking three test cases, for which reference experimental CSP data was available. Without CSP, i.e. in the use of conventional DrugScore potentials, none of the three test cases could be successfully re-docked. The integration of CSP with the same weighting factor as described above resulted in all three cases successfully re-docked. For two of the three complexes, native-like solutions were only produced if CSP were considered.Conformational changes in the binding pockets of up to 2 Å RMSD did not affect the success of the docking. QCSPScore will be particularly interesting in difficult protein-ligand complexes. They are in particular those cases in which the shape of the binding pocket does not provide sufficient steric restraints such as in flat protein-protein interfaces and in the virtual screening of small chemical fragments.
Background: The c-Cbl-associated protein (CAP), also known as ponsin, localizes to focal adhesions and stress fibers and is involved in signaling events. Phosphorylation has been described for the other two members of the sorbin homology family, vinexin and ArgBP2, but no data exist about the putative phosphorylation of CAP. According to previous findings, CAP binds to tyrosine kinase c-Abl. However, it is not known if CAP is a substrate of c-Abl or other tyrosine kinases or if phosphorylation regulates its localization.
Results: We here show that CAP is Tyr phosphorylated by and interacts with both c-Abl and c-Src. One major phosphorylation site, Tyr360, and two minor contributors Tyr326 and Tyr632 were identified as Abl phosphorylation sites, whereas Src preferentially phosphorylates Tyr326 and Tyr360. Phosphorylation of CAP was not necessary for its localization to focal adhesions and stress fibers, but Tyr326Phe substitution alters the function of CAP during cell spreading.
Conclusion: This is the first demonstration of phosphorylation of CAP by any kinase. Our findings suggest that coordinated action of Src and Abl might regulate the function of CAP and reveal a functional role especially for the Src-mediated Tyr phosphorylation of CAP in cell spreading.
Opting out of the great inflation: German monetary policy after the break down of Bretton Woods

During the turbulent 1970s and 1980s the Bundesbank established an outstanding reputation in the world of central banking. Germany achieved a high degree of domestic stability and provided safe haven for investors in times of turmoil in the international financial system. Eventually the Bundesbank provided the role model for the European Central Bank. Hence, we examine an episode of lasting importance in European monetary history. The purpose of this paper is to highlight how the Bundesbank monetary policy strategy contributed to this success. We analyze the strategy as it was conceived, communicated and refined by the Bundesbank itself. We propose a theoretical framework (following Söderström, 2005) where monetary targeting is interpreted, first and foremost, as a commitment device. In our setting, a monetary target helps anchoring inflation and inflation expectations. We derive an interest rate rule and show empirically that it approximates the way the Bundesbank conducted monetary policy over the period 1975-1998. We compare the Bundesbank´s monetary policy rule with those of the FED and of the Bank of England. We find that the Bundesbank´s policy reaction function was characterized by strong persistence of policy rates as well as a strong response to deviations of inflation from target and to the activity growth gap. In contrast, the response to the level of the output gap was not significant. In our empirical analysis we use real-time data, as available to policy-makers at the time. Artenreiche montane Rasengesellschaften auf Lawinenbahnen des Nationalparks Gesäuse (Österreich)

Das Ziel dieser Untersuchung war es, Erkenntnisse über die Bedeutung von Lawinenbahnen für die Biodiversität zu gewinnen. Dazu wurden im Nationalpark Gesäuse (Steiermark, Österreich) 16 Pflanzenbestände in drei ausgewählten Lawinenbahnen pflanzensoziologisch untersucht, vegetationsökologisch charakterisiert sowie mit Vegetationstabelle, bodenkundlichen Analysedaten und topographischen Parametern dokumentiert. Außerdem erfolgte eine Bestandsaufnahme der Schmetterlinge. Die Pflanzenartenvielfalt wurde mit verschiedenen Diversitäts-Indizes beurteilt. Die Ursachen für den Artenreichtum in den untersuchten Lawinenbahnen werden diskutiert.
Die Pflanzenbestände wurden überwiegend zum Origano-Calamagrostietum variae gestellt; eine Einzelaufnahme wurde dem Seslerio-Caricetum sem pervirentis zugeordnet. Im Ersteren dominieren Arten mit hemikryptophytischer Lebensform; Therophyten erreichen die zweithöchste Deckung. Die Pflanzengesellschaft wird von CSR-, CS- und C-Strategen aufgebaut; alle anderen Lebensstrategietypen haben keine Bedeutung. Das Origano-Calamagrostietum variae ist eine artenreiche, natürliche Dauergesellschaft, in der mäßiger Stress und schwache Störungen die Artenzusammensetzung maßgeblich bestimmen. Aktive Lawinenbahnen auf sehr flachgründigen, steinigen, nährstoffarmen, basenreichen Rendzinen in lokalklimatisch wärmebegünstigten steilen Hanglagen der montanen Höhenstufe zählen zu den arten-, blüten- und aspektreichsten und somit ökologisch wertvollsten Vegetationsformationen im Nationalparkgebiet. In den kräuterreichen Pflanzenbeständen wurden im Durchschnitt 71 Gefäßpflanzenarten und 5 Moosarten pro 20 m2 Aufnahmefläche gefunden. Mit dieser hohen Phytodiversität und dem Blütenreichtum ist auch eine große Artenvielfalt bei den Schmetterlingen verbunden. Die untersuchten Lawinenbahnen weisen nicht nur ein hohes Maß an Naturnähe auf, sie dürften auch der ursprüngliche Lebensraum einiger Pflanzenarten des Wirtschafts- und Extensivgrünlandes der Tallagen sein. Auf Grund ihrer hohen naturschutzfachlichen Bedeutung müssen Lawinenbahnen als Ganzes geschützt werden. Periodische oder episodische Lawinenabgänge sind Voraussetzung für die Existenz dieser störungsgeprägten Ökosysteme. Unterhalb von Lawinenbahnen dürfen keine Gebäude oder Infrastrukturen errichtet werden, weil diese Bautätigkeit Schutzobjekte erzeugt. Dadurch würde ein Bedarf für Schutzmaßnahmen entstehen, vor allem in Form von Lawinenverbauungen und Aufforstungen, wodurch Lawinenabgänge verhindert werden.
Die vorliegende Arbeit untersucht das nationale Selbstverständnis der gesamten Konföderation und behandelt insbesondere Aspekte der Innenpolitik sowie bestimmte soziokulturelle und sozioökonomische Charakteristika, aus denen sich die nationalistische Ideologie der Konföderierten konstituierte. Es bleibt zu betonen, dass sich der Fokus der hiesigen Untersuchungen auf innerstaatliche Ereignisse und Phänomene während des Bürgerkrieges richtet und somit vom großen Feld der konföderierten Außenpolitik beinahe vollständig absieht. Die Außenpolitik bildet zweifelsohne ein ebenso spannendes Forschungsgebiet, sie wird hier aber zu Gunsten einer expliziteren Betrachtung des innerstaatlichen Kontexts außen vor gelassen, da die inneren Umstände für die Genese des konföderierten Nationalismus unmittelbarer und somit von größerer Tragweite waren.
Solid state NMR is a emerging method for the study of membrane proteins, which has received much interest in recent years. Limiting the study of many pharmacologically relevant targets, are the often long measuring times, required to obtain especially higher dimensional solid state NMR spectra of good quality. To address this problem, multiple methods where developed in this work, which can be categorized into two groups. The first set of methods aims at the quality of certain spectra, by implementing a spectral filter, which increases the fidelity of the measured data. The second set of methods, addresses the problem of long measuring times directly, by increasing the sensitivity per unit time, as could be shown, for example, on homo- and heteronuclear singlequantum-singlequantum correlation experiments. The gains in measuring time for the latter group of methods are typically in the order of 2-3, but some experiments allow multiple methods to be employed simultaneously, which can lead to a decrease in measuring time of a factor of up to 8. It is important to mention, that none of the methods introduced in this work require any equipment in addition to the conventional setup present in most sold state NMR laboratories and no changes or addition to the samples under study are required. Therefore the gains reported in this work come at no extra cost and require only minimal implementation effort on the side of the user.
X-ray structure of the Na+-coupled Glycine-Betaine symporter BetP from Corynebacterium glutamicum

Cellular membranes are important sites of interaction between cells and their environment. Among the multitude of macromolecular complexes embedded in these membranes, transporters play a particularly important role. These integral membrane proteins perform a number of vital functions that enable cell adaptation to changing environmental conditions. Osmotic stress is a major external stimulus for cells. Bacteria are frequently exposed to either hyperosmotic or hypoosmotic stress. Typical conditions for soil bacteria, such as Corynebacterium glutamicum, vary between dryness and sudden rainfall. Physical stimuli caused by osmotic stress have to be sensed and used to activate appropriate response mechanisms. Hypoosmotic stress causes immediate and uncontrolled influx of water. Cells counteract by instantly opening mechanosensitive channels, which act as emergency valves leading to fast efflux of small solutes out of the cell, therebydiminishing the osmotic gradient across the cell membrane. Hyperosmotic stress, on the other hand, results in water efflux. This is counterbalanced by an accumulation of small, osmotically active solutes in the cytoplasm, the so-called compatible solutes. They comprise a large variety of substances, including amino acids (proline), amino acid derivatives (betaine, ectoine), oligosaccharides (trehalose), and heterosides (glucosylglycerol). Osmoregulated transporters sense intracellular osmotic pressure and respond to hyperosmotic stress by facilitating the inward translocation of compatible solutes across the cell membrane, to restore normal hydration levels. This work presents the first X-ray structure of a member of the Betaine-Choline-Carnitine-Transporter (BCCT) family, BetP. This Na+-coupled symporter from Corynebacterium glutamicum is a highly effective osmoregulated and specific uptake system for glycine-betaine. X-ray structure determination was achieved using single wavelength anomalous dispersion (SAD) of selenium atoms. Selenium was incorporated into the protein during its expression in methione auxotrophic E. coli cells, grown in media supplemented with selenomethionine. SAD data with anomalous signal up to 5 Å led to the detection of 39 selenium sites, which were used to calculate the initial electron density map of the protein. Medium resolution and high data anisotropy made the structure determination of BetP a challenging task. A specific strategy for data anisotropy correction and a combination of various crystallographic programs were necessary to obtain an interpretable electron density map suitable for model building. The crystal structure of BetP shows a trimer with glycine-betaine bound in a three-fold cation-pi interaction built by conserved tryptophan residues. The bound substrate is occluded from both sides of the membrane and aromatic side chains line its transport pathway. Very interestingly, the structure reveals that the alpha-helical C-terminal domain, for which a chemo- and osmosensory function was elucidated by biochemical methods, interacts with cytoplasmic loops of an adjacent monomer. These unexpected monomer-monomer interactions are thought to be crucial for the activation mechanism of BetP, and a new atomic model combing biochemical results with the crystal structure is proposed. BetP is shown to have the same overall fold as three unrelated Na+-coupled symporters. While these were crystallised in either the outward- or inward-facing conformation, BetP reveals a unique intermediate state, opening new perspectives on the alternating access mechanism of transport.
Both the genomes of the epsilonproteobacteria Wolinella succinogenes and Campylobacter jejuni contain operons (sdhABE) that encode for so far uncharacterized enzyme complexes annotated as ‘non-classical’ succinate:quinone reductases (SQRs). However, the role of such an enzyme ostensibly involved in aerobic respiration in an anaerobic organism such as W. succinogenes has hitherto been unknown. We have established the first genetic system for the manipulation and production of a member of the non-classical succinate:quinone oxidoreductase family. Biochemical characterization of the W. succinogenes enzyme reveals that the putative SQR is in fact a novel methylmenaquinol:fumarate reductase (MFR) with no detectable succinate oxidation activity, clearly indicative of its involvement in anaerobic metabolism. We demonstrate that the hydrophilic subunits of the MFR complex are, in contrast to all other previously characterized members of the superfamily, exported into the periplasm via the twin-arginine translocation (tat)-pathway. Furthermore we show that a single amino acid exchange (Ala86→His) in the flavoprotein of that enzyme complex is the only additional requirement for the covalent binding of the otherwise non-covalently bound FAD. Our results provide an explanation for the previously published puzzling observation that the C. jejuni sdhABE operon is upregulated in an oxygen-limited environment as compared with microaerophilic laboratory conditions.
Inhibition of the soluble epoxide hydrolase (sEH) has beneficial effects on vascular inflammation and hypertension indicating that the enzyme may be a promising target for drug development. As the enzymatic core of the hydrolase domain of the human sEH contains two tyrosine residues (Tyr383 and Tyr466) that are theoretically crucial for enzymatic activity, we addressed the hypothesis that the activity of the sEH may be affected by nitrosative stress. Epoxide hydrolase activity was detected in human and murine endothelial cells as well in HEK293 cells and could be inhibited by either authentic peroxynitrite (ONOO−) or the ONOO− generator 3-morpholino-sydnonimine (SIN-1). Protection of the enzymatic core with 1-adamantyl-3-cyclohexylurea in vitro decreased sensitivity to SIN-1. Both ONOO− and SIN-1 elicited the tyrosine nitration of the sEH protein and mass spectrometry analysis of tryptic fragments revealed nitration on several tyrosine residues including Tyr383 and Tyr466. Mutation of the latter residues to phenylalanine was sufficient to abrogate epoxide hydrolase activity. In vivo, streptozotocin-induced diabetes resulted in the tyrosine nitration of the sEH in murine lungs and a significant decrease in its activity. Taken together, these data indicate that the activity of the sEH can be regulated by the tyrosine nitration of the protein. Moreover, nitrosative stress would be expected to potentiate the physiological actions of arachidonic acid epoxides by preventing their metabolism to the corresponding diols.
Proline-rich tyrosine kinase 2 (PYK2) can be activated by angiotensin II (Ang II) and reactive oxygen species. We report that in endothelial cells, Ang II enhances the tyrosine phosphorylation of endothelial NO synthase (eNOS) in an AT1-, H2O2-, and PYK2-dependent manner. Low concentrations (1–100 µmol/liter) of H2O2 stimulated the phosphorylation of eNOS Tyr657 without affecting that of Ser1177, and attenuated basal and agonist-induced NO production. In isolated mouse aortae, 30 µmol/liter H2O2 induced phosphorylation of eNOS on Tyr657 and impaired acetylcholine-induced relaxation. Endothelial overexpression of a dominant-negative PYK2 mutant protected against H2O2-induced endothelial dysfunction. Correspondingly, carotid arteries from eNOS–/– mice overexpressing the nonphosphorylatable eNOS Y657F mutant were also protected against H2O2. In vivo, 3 wk of treatment with Ang II considerably increased levels of Tyr657-phosphorylated eNOS in the aortae of wild-type but not Nox2y/– mice, and this was again associated with a clear impairment in endothelium-dependent vasodilatation in the wild-type but not in the Nox2y/– mice. Collectively, endothelial PYK2 activation by Ang II and H2O2 causes the phosphorylation of eNOS on Tyr657, attenuating NO production and endothelium-dependent vasodilatation. This mechanism may contribute to the endothelial dysfunction observed in cardiovascular diseases associated with increased activity of the renin–angiotensin system and elevated redox stress.
Crista junctions (CJs) are important for mitochondrial organization and function, but the molecular basis of their formation and architecture is obscure. We have identified and characterized a mitochondrial membrane protein in yeast, Fcj1 (formation of CJ protein 1), which is specifically enriched in CJs. Cells lacking Fcj1 lack CJs, exhibit concentric stacks of inner membrane in the mitochondrial matrix, and show increased levels of F1FO–ATP synthase (F1FO) supercomplexes. Overexpression of Fcj1 leads to increased CJ formation, branching of cristae, enlargement of CJ diameter, and reduced levels of F1FO supercomplexes. Impairment of F1FO oligomer formation by deletion of its subunits e/g (Su e/g) causes CJ diameter enlargement and reduction of cristae tip numbers and promotes cristae branching. Fcj1 and Su e/g genetically interact. We propose a model in which the antagonism between Fcj1 and Su e/g locally modulates the F1FO oligomeric state, thereby controlling membrane curvature of cristae to generate CJs and cristae tips.
An efficient route for delivering specific proteins and peptides into neurons could greatly accelerate the development of therapies for various diseases, especially those involving intracellular defects such as Parkinson disease. Here we report the novel use of polybutylcyanoacrylate nanoparticles for delivery of intact, functional proteins into neurons and neuronal cell lines. Uptake of these particles is primarily dependent on endocytosis via the low density lipoprotein receptor. The nanoparticles are rapidly turned over and display minimal toxicity to cultured neurons. Delivery of three different functional cargo proteins is demonstrated. When primary neuronal cultures are treated with recombinant Escherichia coli beta-galactosidase as nanoparticle cargo, persistent enzyme activity is measured beyond the period of nanoparticle degradation. Delivery of the small GTPase rhoG induces neurite outgrowth and differentiation in PC12 cells. Finally, a monoclonal antibody directed against synuclein is capable of interacting with endogenous alpha-synuclein in cultured neurons following delivery via nanoparticles. Polybutylcyanoacrylate nanoparticles are thus useful for intracellular protein delivery in vitro and have potential as carriers of therapeutic proteins for treatment of neuronal disorders in vivo.
The cell division cycle protein 37 (Cdc37) and the 90-kDa heat shock protein (Hsp90) are molecular chaperones, which are crucial elements in the protein signaling pathway. The largest class of client proteins for Cdc37 and Hsp90 are protein kinases. The catalytic domains of these kinases are stabilized by Cdc37, and their proper folding and functioning is dependent on Hsp90. Here, we present the x-ray crystal structure of the 16-kDa middle domain of human Cdc37 at 1.88 angstroms resolution and the structure of this domain in complex with the 23-kDa N-terminal domain of human Hsp90 based on heteronuclear solution state NMR data and docking. Our results demonstrate that the middle domain of Cdc37 exists as a monomer. NMR and mutagenesis experiments reveal Leu-205 in Cdc37 as a key residue enabling complex formation. These findings can be very useful in the development of small molecule inhibitors against cancer.
The single nucleotide polymorphism 118A>G of the human micro-opioid receptor gene OPRM1, which leads to an exchange of the amino acid asparagine (N) to aspartic acid (D) at position 40 of the extracellular receptor region, alters the in vivo effects of opioids to different degrees in pain-processing brain regions. The most pronounced N40D effects were found in brain regions involved in the sensory processing of pain intensity. Using the mu-opioid receptor-specific agonist DAMGO, we analyzed the micro-opioid receptor signaling, expression, and binding affinity in human brain tissue sampled postmortem from the secondary somatosensory area (SII) and from the ventral posterior part of the lateral thalamus, two regions involved in the sensory processing and transmission of nociceptive information. We show that the main effect of the N40D micro-opioid receptor variant is a reduction of the agonist-induced receptor signaling efficacy. In the SII region of homo- and heterozygous carriers of the variant 118G allele (n=18), DAMGO was only 62% as efficient (p=0.002) as in homozygous carriers of the wild-type 118A allele (n=15). In contrast, the number of [3H]DAMGO binding sites was unaffected. Hence, the micro-opioid receptor G-protein coupling efficacy in SII of carriers of the 118G variant was only 58% as efficient as in homozygous carriers of the 118A allele (p<0.001). The thalamus was unaffected by the OPRM1 118A>G SNP. In conclusion, we provide a molecular basis for the reduced clinical effects of opioid analgesics in carriers of mu-opioid receptor variant N40D.
Thalassämia major und Sichelzellanämie sind hereditäre Erkrankungen, die zu der Gruppe der quantitativen bzw. qualitativen Hämoglobinsynthesestörungen gehören und in unterschiedlichem Maße mit einer chronischen Anämie einhergehen. Dabei besteht die Therapie der Anämie in regelmäßigen Bluttransfusionen. Im Falle der Thalassämia major sind regelmäßige Bluttransfusionen alle 2-4 Wochen notwendig. Dabei übersteigt die damit zugeführte Eisenmenge bei weitem die Eisenausscheidungskapazität des Körpers, die limitiert und passiver Natur ist. Es kann dadurch zur Eisenüberladung des Körpers mit Erschöpfung der Eisenbindungskapazität und Nachweisbarkeit von freiem Eisen kommen. Freies Eisen generiert über die Fenton-Reaktion freie Radikale und reaktive Sauerstoffspezies, die ihrerseits in der Lage sind, biologische Moleküle sowie Zellstrukturen zu schädigen. Der Organismus verfügt über Mechanismen um diese Schäden zu verhindern bzw. den Ausmaß der Schäden zu begrenzen, die als antioxidativen Abwehrmechanismen bezeichnet werden. In dieser vorliegenden Arbeit wurden Blutproben von 22 Patienten mit ß-Thalassämia major und 16 Patienten mit Sichelzellanämie Patienten untersucht. Bei ihnen wurde das Vorliegen der pathologischen Modellsituation einer Eisenüberladung angenommen. Als Kontrollgruppe wurden 16 phänotypisch gesunde Geschwister der beiden Patientenkollektive herangezogen. Hauptziele dieser Arbeit war, die Bleomycin-Methode im Stoffwechsellabor des Zentrums für Kinderheilkunde und Jugendmedizin der Johann Wolfgang Goethe-Universität zu etablieren und dabei die folgenden Fragen zu klären: 1. Entsteht freies Eisen bei polytransfundierten Patienten? 2. Ist die Bleomycin-Assay zur Bestimmung des freien Eisens geeignet? 3. Welche Zusammenhänge bestehen zwischen den Parametern des Eisenstoffwechsels und können diese zur Abschätzung des freien Eisens genutzt werden? Freies Eisen entsteht dann, wenn die Eisenbindungskapazität des Transferrins überschritten wird. Bei gesunden Menschen liegt eine Transferrinsättigung im Durchschnitt bei unter 30%. Es ist also eine wertvolle Reserve vorhanden, um effektiv die Enstehung des freien Eisens zu verhindern. Bei bestimmten pathologischen Situationen, wie sie auch bei polytransfundierten Patienten bei Thalassämie vorliegen, wird dem Körper massiv Eisen zugeführt. Wir konnten bei 18 von 22 Patienten in der Thalassämiegruppe freies Eisen nachweisen. Der Median der freien Eisenkonzentration lag bei 1,25 μmol/l bei einer Spannbreite von 5,3μmol/l. In der Sichelzellanämiegruppe konnte nur bei einem Patienten freies Eisen nachgewiesen werden. Dieser hatte ebenfalls häufig Bluttransfusionen erhalten. Zur Messung des freien Eisens wurde die Bleomycin Methode nach Gutteridge et al angewandt. Es ist eine nasschemische Methode (Messung erfolgt via Spektroskopie) und erfordert keinen hohen technischen Aufwand. Da bei der Bestimmung des freien Eisens im mikromolaren Bereich geschieht, ist das Hauptproblem die Kontamination der Reagenzien durch das ubiquitär vorkommende Eisen. Durch höchste Sorgfalt und genaues Arbeiten im staubfreien Milieu und Behandlung der Reagenzien mit einem geeigneten Eisenkomplexbildner, wie z.B. Chelex100®, ist dieses Problem beherrschbar. Dennoch erfordert die Methode einen enormen Zeitaufwand, weshalb nach Parametern gesucht wurde, die zur Abschätzung oder zur Vorselektion der geeigneten Blutproben zur Bestimmung des freien Eisens herangezogen werden können. Wie oben schon erwähnt, hatten 18 von 22 Thalassämiepatienten freies Eisen im Blut. Davon hatten 15 eine Transferrinsättigung über 100% und 2 knapp unter 100%. Lediglich ein Patient, bei dem aber auch nur freies Eisen von 0,05 μmol/l gemessen wurde, hatte eine Transferrinsättigung deutlich unter 100%. Ein Patient aus der Sichelzellanämiegruppe, bei dem auch freies Eisen gemessen wurde, hatte ebenfalls eine Transferrinsättigung über 100%. Zusammenfassend kann festgehalten werden, dass eine hoch signifikante positive Korrelation zwischen dem freien Eisen und der Transferrinsättigung festegestellt wurde (r = 0,63, p = 0,002). 95% der Patienten, die freies Eisen im Blut hatten, wiesen auch Transferrinsättigungswerte über bzw. knapp unter 100% auf. Es besteht auch ein statistisch signifikanter positiver Zusammenhang zwischen der Serumeisenkonzentration und freiem Eisen. Alle Patienten mit freiem Eisen im Blut hatten Serumeisenwerte über 170 μg/dl. Damit konnte die Arbeitshypothese bestätigt werden, dass es bei polytransfundierten Thalassämiepatienten zu einer Eisenüberladung mit in der Folge entstehendem freiem Eisen kommt; bekannterweise induziert zweiwertiges Eisen die Fenton-Reaktion bzw. Haber-Weiss-Reaktion und damit oxidativen Stress. Bei Sichelzellanämiepatienten , die keine regelmäßigen Hochregimebluttransfusionen erhalten, die ebenso, wie in der Literatur beschrieben, oxidativem Stress ausgesetzt sind, müssen auch andere Pathomechanismen angenommen werden.
Thirteen species of skippers (six newly described; Lepidoptera: Hesperiidae: Hesperiinae: Hesperiini) from higher elevations of Mexico and Central America are reviewed. These are included in four genera (one newly described), some with proposed new combinations. Onespa Steinhauser, 1974, originally described as monotypic, is shown to include three species in addition to its type species, Onespa nubis Steinhauser, 1974. One of these, Atrytone gala Godman, 1900, that has been misplaced in several genera since its description, represents a new combination. The other two species, distributed in montane habitats in northwestern Mexico and in Costa Rica, are described as new. Buzyges Godman, 1900, distributed in Mexico and Central America and also formerly considered monotypic, is shown to embrace four species. Besides the type species, Buzyges idothea Godman, 1900, two species long placed in Poanes Scudder, 1872, Pamphila rolla Mabille, 1883, and Poanes benito Freeman, 1979, are included as new combinations. Another species, known only from Costa Rica, is described as new. These are united by several superficial characters, but especially by genital morphology of both sexes. Librita Evans, 1955, was described to include three species of which one, Librita raspa Evans, 1955, was subsequently removed. Augiades heras Godman, 1900 is here also removed from Librita and placed in a new genus with three previously undescribed species. This completes the disintegration of Librita, which is now monotypic. The four genera, although exhibiting similarities suggesting potential alliance, differ in their unique combinations of several superficial and genital traits from each other and other hesperiine skippers.
Sepsis is caused by infection and often followed by an overwhelming inflammatory response. This can lead to shock, organ failure and even death. Each year approximately 60,000 people die in Germany due to sepsis. There is good evidence that sepsis is associated with failure of the hypothalamic-pituitary-adrenal-axis. In patients with sepsis, glucocorticoids (e.g. corticosterone, cortisol) released from adrenal glands play an essential role in preventing an excessive pro-inflammatory response. Adrenal insufficiency occurs in a large number of patients with septic shock and is associated with an increased mortality. In the innate immune system, Toll-like receptors (TLRs) play a crucial role in its onset by recognizing pathogenassociated molecules. It is well known that there are interactions between the immune and endocrine stress systems; glucocorticoids and TLRs regulate each other in a bi-directional way. Therefore, a coordinated response of the adrenal and immune system is of vital importance for survival during severe inflammation. This experimental study focuses on the role of TLR-2, TLR-4 and TLR-9 during adrenal stress. The results show that in mice, the absence of TLR-2 and TLR-4, but not TLR-9 leads to altered adrenal morphology, relating to size and cellular structure. However, this alteration does not appear to compromise the phenotype of TLR knock-out mice. Mice deficient of TLR-2, 4 and 9 are not able to respond adequately to inflammatory stress induced by their potential ligands lipopolysaccharide (LPS), lipoteichoic acid (LTA) or cytidine phosphate guanosine-oligodeoxynucleotides (CpG-ODN). This impaired adrenal stress response appears to be associated with a decrease in systemic and intra-adrenal cytokine expressions. Taken together, these results suggest that TLR-2, 4 and 9 are key players in the immuno-endocrine response during inflammation and SIRS. In conclusion, TLRs play a crucial role in the immune-adrenal crosstalk. This close functional relationship needs to be considered in the treatment of inflammatory diseases where an intact adrenal stress response is required. Furthermore, TLR polymorphisms could contribute to the underlying mechanisms of impaired adrenal stress response in patients with bacterial sepsis
Diese Diplomarbeit hatte das Ziel ein Konzept zu erstellen, welches es ermöglicht, interessante und weniger interessante Artikel innerhalb eines Wikis zu erkennen und diese Informationen in verständlicher Form zur Recherche visuell bereitzustellen.
Dabei sollte das Konzept möglichst offen sein, so dass theoretisch jedes Wiki an die Visualisierung hätte angebunden werden können. Hier lag bereits das erste Problem, zum Zeitpunkt der Arbeit existieren bereits mehr als 100 unterschiedliche Wikis mit unterschiedlichen Architekturen. Wegen der Unterschiede der jeweiligen Wikisysteme entschloss man sich daher zwei Konzepte zu erarbeiten, ein allgemeines, welches wie in der gestellten Zielsetzung, das Einbinden jedes Wikis ermöglicht und ein Spezialfall, der die Vorteile einer API nutzt. Der Spezialfall wurde in ähnlicher Form in einer Implementierung umgesetzt.
Zu Beginn der Diplomarbeit mussten die unterschiedlichen Möglichkeiten der Extraktion von Informationen aus einem Wiki untersucht werden. Es hatte sich ziemlich früh herausgestellt das Links, Backlinks sowie Kategorien wichtige Indikatoren zur Bewertung eines Artikels darstellen. Damit die Bewertung der Informationen nicht nur alleine auf der Struktur eines Wikis beruht, wurde ein Thesaurus zur unterstützenden Bewertung miteinbezogen. Dieser lieferte durchgehend gute Ergebnisse, wobei - wie erwartet - der Thesaurus sehr schnell an seine Grenzen gekommen war, insbesondere wenn man die Anzahl der Artikel eines großen Wikis mit der Anzahl der Wörter die im Thesaurus gespeichert sind vergleicht.
Die extrahierten und gewichteten Informationen wurden im zweiten Schritt visualisiert, dabei hatte sich der Radial-Graph als eine gute Lösung zur Darstellung der Informationen herausgestellt. Neben einem Graphen mit gewichteten Knoten wurden in der Visualisierung unterschiedliche Ansichten der extrahierten Daten bereitgestellt: eine Autorenansicht, die zum gesuchten Artikel die Autoren darstellt, eine semantische Ansicht, die Wortbeziehungen veranschaulicht sowie eine Artikelansicht, die den Nutzer neben den gewichteten Artikeln auch wie gewohnt in einer Wiki lesen lässt.
Lernplattformen sind E-Learning-Systeme, deren Kernfunktionalität die Verwaltung und Verteilung von Lernmaterialien über das World Wide Web ist. In dieser Arbeit wurde untersucht, wie durch Aufzeichnung (Tracking), Auswertung und Visualisierung von Lernaktivitäten in Lernplattformen eine Verbesserung der Lernqualität erreicht werden kann. Der Ansatzpunkt dafür war, Informationen zu Lernaktivitäten in geeigneter Weise Lehrenden und Lernenden zu präsentieren, so dass diese Rückschlüsse ziehen können, um Lernprozesse eigenständig zu optimieren. Viele Lernplattformen verfolgen bereits diesen Ansatz und verfügen deshalb über entsprechende Funktionalität.
Es mussten zwei wesentliche Fragen beantwortet werden:
1. Was müssen Lernende und Lehrende über erfolgte Lernaktivitäten wissen?
2. Wie werden Lernaktivitäten in geeigneter Weise präsentiert?
Diese Fragen wurden durch Betrachtung existierender Lernplattformen (State of the Art) sowie Befragung von Experten in Form von Interviews beantwortet. Zur Beantwortung der 2. Frage wurden außerdem allgemeine Grundlagen der Auswertung und Visualisierung von Daten verwendet sowie (zu einem geringen Teil) Auswertungs- und Visualisierungsverfahren von Systemen, die keine Lernplattformen sind. Besondere Aufmerksamkeit wurde auch dem
Datenschutz gewidmet.
Beruhend auf den gewonnenen Erkenntnissen wurde dann ein Konzept für ein Auswertungs-/Visualisierungssystem entwickelt das in verschiedenen Punkten eine Verbesserung des State of the Art darstellt.
Teile des Konzepts wurden schließlich für das webbasierte Softwaresystem LernBar, das über einen Großteil der Funktionalität einer Lernplattform verfügt, prototypisch implementiert. Durch die Implementierung soll es ermöglicht werden, das Konzept im praktischen Einsatz zu evaluieren, was im Rahmen dieser Arbeit nicht möglich war.
Konzeption und Implementierung einer Kommunikation zwischen Second Life und Web 2.0 Anwendungen

Im Rahmen dieser Arbeit haben wir ein Konzept und einen Prototypen zur Kommunikation zwischen Second Life und Web 2.0 entwickelt.
Im Rahmen dieser Diplomarbeit wird ein Konzept und ein Prototyp zur Kommunikation zwischen Second Life und Web 2.0-Anwendungen entwickelt. In der Übung zur Veranstaltung "Einführung in das Projektmanagement" wurden Meetings in Second Life abgehalten. Dabei haben die Studierenden im Rahmen der Übung Protokolle erstellt, die sie im Internet veröffentlichten. Die Protokollierung musste immer manuell durchgeführt werden und war dadurch fehleranfällig und nicht ausfallsicher. Hier entstand der Wunsch, die Protokollierung zu automatisieren und somit den administrativen Aufwand zu reduzieren.
Im Kapitel 2 werden die Grundlagen behandelt, die für das Verständnis der Arbeit notwendig sind. Hierbei werden Second Life, sowie Blog und Wiki als Repräsentanten von Web 2.0 vorgestellt. Außerdem wird eine klare Abgrenzung zwischen diesen Technologien aufgezeigt.
Die Analyse dieser Diplomarbeit umfaÿt unter anderem die verschiedenen Möglichkeiten der Übertragung der Informationen zwischen Second Life und den Web 2.0-Anwendungen.
Im Konzept ist die Gesamtarchitektur zur Kommunikation zwischen Second Life und den Web 2.0 Anwendungen enthalten. Die Hauptsystemkomponenten, die dafür notwendig sind, stellen eine Ansammlung aus Second Life Skripten, dem HTTP Supervisor und den Wiki Bot Skripten dar. Die Second Life und die Wiki Bot Skripten sind eine Ansammlung von Unterprogrammen, die jeweils auf eine Aufgabe spezialisiert sind. Die genaue Erklärung wird im Kapitel 4 Aufgrund der Unterschiede der Web 2.0 Anwendungen wurde ein Bot eingesetzt, der eine Web 2.0 Anwendung bedient. Der HTTP Supervisor dient der Vermittlung dient der Vermittlung der Daten zwischen Second Life und der Web 2.0 Anwendung. Auÿerdem speichert er die Daten in temporäre Dateien. Der Second Life Teil des Programms dient der Steuerung des gesamten Systes.
Durch die prototypische Umsetzung ist die Durchführbarkeit bewiesen, die Daten aus Second Life herauszuführen und im MediaWiki zu speichern.
The supersilylated ethene trans-(tBu3Si)HC=CH(SitBu3) (triclinic, P ī) is accessible from the reaction of tBu3SiCHBr2 with nBuLi at −78 °C in THF or Et2 O. The reaction of Li(H2NCH2CH2NH2)C≡CH with tBu3SiBr leads to the formation of (tBu3Si)C≡CH and (tBu3Si)C≡C(SitBu3). X-Ray quality crystals of (tBu3Si)C≡C(SitBu3) (triclinic, P ī) were obtained by recrystallization from hexane. In contrast to the structures of the disilane tBu3Si-SitBu3 and the disiloxane tBu3Si-O-SitBu3, the sterically crowded ethene trans-(tBu3Si)HC=CH(SitBu3) and ethyne (tBu3Si)C≡C(SitBu3) feature dihedral angles of 60° in the solid-state structures.
[MesnacnacZn(μ-H)]2 (1) was synthesized by reaction of MesnacnacZnI with either an equimolar amount of KNH(iPr)BH3 or an excess of NaH and characterized by multinuclear NMR and IR spectroscopy as well as X-ray diffraction. Two polymorphs of 1 were found and their structures determined on single crystals.
Untersuchungen zur Wechselwirkung zwischen der Polo-like kinase 1 und dem CCCTC-binding factor

Die Polo-like kinase 1 ist ein Schlüsselprotein bei der Steuerung mitotischer, meiotischer und zytokinetischer Prozesse. Um den korrekten Ablauf dieser Prozesse zu gewährleisten, muss die Menge an PLK1 in den verschiedenen Phasen des Zellzykluses genauestens reguliert werden. Eine Vielzahl verschiedener Tumorentitäten weisen eine stark erhöhte Konzentration an PLK1-Protein auf. Für die Krebsforschung ist es daher von großer Relevanz Aufschluss über solche Proteine zu erhalten, die Einfluss auf die PLK1 Expression nehmen. Dieses Wissen könnte dazu beitragen, Strategien für eine noch gezieltere Krebstherapie zu entwickeln. Die vorliegende Arbeit zeigt einerseits erste Hinweise für eine Rolle des multivalenten Transkriptionsfaktors CCCTC-binding factor (CTCF) für die Regulation des PLK1-Promotors und andererseits erste Anhaltspunkte für einen Einfluss der PLK1 auf die transkriptionsregulatorische Funktion von CTCF. Grundlage dieser Arbeit war die Identifikation einer putativen Binderegion von CTCF im Bereich des PLK1-Promotors, die die Frage aufkommen ließ, ob CTCF einen transkriptionsregulatorischen Einfluss auf den PLK1-Promotor besitzt. Deshalb wurden initial Reportergenanalysen durchgeführt, bei denen gezeigt werden konnte, dass CTCF die Promotoraktivität einer PLK1-Promotorsequenz, welche die putative CTCF-Binderegion beinhaltet, steigern kann. Zudem ist es gelungen, die Bedeutung der putativen Binderegion für die Aktivitätssteigerung herauszuarbeiten, da eine 50%ige Deletion dieses Bereiches mit einer deutlichen Reduktion der CTCFbedingten Induktion der Reportergentranskription einherging. Interessanterweise konnten in den daraufhin durchgeführten in vitro-Interaktionsstudien erste Daten, die für eine Interaktion von CTCF und PLK1 sprechen, gesammelt werden, wobei die Kombination der Ergebnisse aus dem GST-Pull-Down- und dem Co-Immunopräzipitationsexperiment die Aussage zulassen, dass diese Wechselwirkung auf Seiten der PLK1 über die Polo-Box-Domäne und auf Seiten von CTCF über die aminoterminale Domäne vermittelt wird. Mit dem Nachweis der in vitro-Phosphorylierbarkeit von CTCF durch PLK1 konnte zudem ein mögliche Funktion dieser Interaktion herausgearbeitet werden. Durch den Einsatz von CTCF-Deletionsmutanten und eines punktmutierten CTCF-Fragments gelang es in weiteren Phosphorylierungsstudien Serin-224 als PLK1-Phosphorylierungsstellen in CTCF zu identifizieren. Abschließend ist es im Zuge von Reportergenanalysen, die den Effekt einer S224A Mutante von CTCF mit demjenigen von CTCF Wildtyp verglichen, gelungen, erste Hinweise für einen Rolle von Serin 224 im Hinblick auf die transkriptionsregulatorische Modulation von CTCF zu sammeln.
Auch in den arktischen Brutgebieten sind Wildgänse und ihre Brut durch Fressfeinde wie den Eisfuchs (Alopex lagopus) oder die Eismöwe (Larus hyperboreus) gefährdet. In diesem Bericht wird am Beispiel von Weißwangengänsen (Branta leucopsis) das Verhalten der Vermeidung von Prädation auf Svalbard mit dem der auf Kolguyev brütenden Artgenossen untersucht. Die Unterschiede und Gemeinsamkeiten werden diskutiert.
Im Rahmen dieser Arbeit wurden die 5´- und 3´-Enden 23 ausgewählter Transkripte des halophilen Archaeons Halobacterium salinarum bestimmt. Die Daten wurden dazu verwendet, die Längen der untranslatierten Bereiche zu ermitteln, Konsensussequenzen für die Transkriptionsinitiation und -termination abzuleiten und die Rolle haloarchaealer UTRs bei der Translationsinitiation und -regulation zu untersuchen. Der experimentelle Ansatz wurde mit einer bioinformatischen Analyse des Genoms von H. salinarum vervollständigt. Dabei konnten die Konsensussequenzen der basalen Promotorelemente genauer definiert werden und ein neues Promotorelement konnte entdeckt werden. Weiterhin wurde ein Konsensusmotiv gefunden, welches wahrscheinlich für die Termination der Transkription wichtig ist. Alle 23 analysierten Transkripte hatten eine 3´-UTR mit einer durchschnittlichen Länge von 48 Nukleotiden und ihre 3´-Enden waren nicht posttranskriptionell modifiziert. Die experimentellen Ergebnisse und bioinformatischen Daten ergaben, dass die Mehrheit der haloarchaealen Transkripte keine 5´-UTR besitzt. Die meisten Transkripte mit 5´-UTRs enthielten unerwarteterweise keine Shine-Dalgarno (SD)-Sequenz. Die Analyse des H. salinarum Genoms machte deutlich, dass weniger als 10% aller Gene eine SD Sequenz vorrausgeht und dass sogar bei Genen, die distal im Operon liegen, meistens keine SD-Sequenz zu finden ist. Weiterhin wurde der Einfluss einer ausgewählten 5´-UTR ohne SD-Sequenz und einer 3´-UTR auf die Transkriptstabilität und die Translationseffizienz untersucht. Das Transkript mit 5´ UTR ohne SD-Sequenz wurde in H. salinarum effizient translatiert. In einer Studie an H. volcanii konnte das Gleiche für verschiedene 5´ UTRs ohne SD-Sequenz in Verbindung mit einem Reportergen gezeigt werden (Brenneis et al., 2007). An diesen Transkripten kann die Translation also nicht über den „bakteriellen“ Mechansimus durch Basenpaarung mit dem 3´-Ende der 16S rRNA initiiert werden. Im weiteren Verlauf der Arbeit wurde untersucht, ob es sich bei dem Mechanismus der Translationsinitiation an Transkripten mit 5´-UTR ohne SD-Sequenz um einen Scanning-Mechanismus wie bei Eukaryonten oder um einen neuen Mechanismus handelt. Für die Experimente wurde das zuvor erwähnte Reportergensystem aus H. volcanii verwendet. Neben AUG wurden GUG und UUG effizient als Startkodons an einem Transkript mit 5´-UTR genutzt, wohingegen AUG das einzige funktionierende Startkodon bei einem Transkript ohne 5´-UTR war. Verschiedene Deletionsversionen der 20 Nukleotide langen 5´-UTR wurden im Gegensatz zur kompletten 5´-UTR nur sehr ineffizient translatiert. Das Einfügen zusätzlicher AUGs stromaufwärts des natürlichen Startkodons hatte keinen Einfluss auf die Translationseffizienz am internen AUG. Ein zusätzliches AUG am 5´-Ende im gleichen Leseraster des natürlichen AUGs führte zur gleichzeitigen Nutzung beider Startkodons auf derselben mRNA. Eine stabile Haarnadelstruktur am 5´-Ende inhibierte die Translation nur am ersten AUG und hatte keinen Einfluss auf die Translationseffizienz am internen AUG. Zusammenfassend konnte durch die erhaltenen Ergebnisse ausgeschlossen werden, dass der Mechanismus der Translationsinitiation an Transkripten mit 5´-UTR ohne SD-Sequenz ein Scanning-Mechansimus wie bei Eukaryonten ist. Neben der Translationsinitiation an Transkripten ohne 5´-UTR und an Transkripten mit SD-Sequenz existiert also noch ein dritter, bis jetzt unbekannter Mechansimus zur Translationsinitiation in Haloarchaea. In initialen Versuchen zur genaueren Charakterisierung der drei verschiedenen Translationsinitiationsmechanismen, wurden Konstrukte hergestellt, bei denen alle drei verschiedenen Startkodons auf einer mRNA vorhanden sind. Es wurden erste Hinweise darauf gefunden, dass Stressbedingungen einen Einfluss auf die unterschiedlichen Initiationsmechansimen haben. Anders als erwartet führte die Mutation einer natürlichen SD-Sequenz nicht zu einer verminderten Translationseffizienz am entsprechenden Startkodon. Die Bestimmung des 5´-Endes eines Transkripts mit in silico vorhergesagter SD-Sequenz offenbarte, dass das Transkript keine 5´-UTR und somit auch keine SD-Sequenz besaß. Beide Ergebnisse machen deutlich, dass SD-Sequenzen eine noch viel geringere Rolle bei der haloarchaealen Translationsinitiation spielen, als anhand der Sequenz und UTR Analysen angenommen werden konnte.
Der katheterinterventionelle Verschluss des persistierenden Foramen ovale ist heutzutage ein sicher durchzuführender Eingriff zur Sekundärprophylaxe einer paradoxen Embolie. Mit den derzeit zur Verfügung stehenden Verschlusssystemen werden komplette Verschlussraten von 86 % – 96 % erreicht. Mehrere Studien konnten zeigen, dass das Vorliegen eines Restshunts das Auftreten von erneuten paradoxen Embolien begünstigt. Mehrere Therapieoptionen für Patienten mit Restshunt stehen zur Verfügung: eine Operation mit Entfernung des PFO-Okkluders und Naht- oder Patchverschluss des PFO, eine medikamentöse Prophylaxe mit Thrombozytenaggregationshemmern oder oralen Antikoagulanzien sowie die Implantation eines zweiten Okkluders. Ziel dieser Arbeit war die Beurteilung der Wirksamkeit und Sicherheit der perkutanen Implantation eines zweiten Verschlusssystems bei Patienten mit mittlerem oder großem Restshunt. Zwischen Oktober 1997 und April 2008 wurde in unserem Zentrum bei 40 Patienten mit einem durchschnittlichen Alter von 51 ± 13 Jahren der katheterinterventionelle Verschluss eines Restshunts mit verschiedenen Verschlusssystemen versucht. Von den Patienten waren 22 weiblich und 18 männlich. Die Implantation eines zweiten Okkluder war bei 39 Patienten erfolgreich (98%). Nur bei einem Patienten war es nicht möglich den Restdefekt mit einem Katheter zu sondieren. Folgende Schirme wurden als Zweitokkluder implantiert: ein Premere-Okkluder bei 20 Patienten (51%), ein Amplatzer-Okkluder bei 13 Patienten (33%), ein STARFlex-Okkluder bei vier Patienten (10%), ein Helex-Okkluder bei einem Patienten (2,5%) und ein Angelwings-Okkluder bei einem Patienten (2,5%). Die mittlere Durchleuchtungszeit aller Eingriffe betrug 8,6 ± 8,6 Minuten. Während der Eingriffe und innerhalb von 30 Tagen nach Implantation eines zweiten Okkluders traten keine Komplikationen auf. Die Patienten wurden im Mittel 36 ± 29 Monate nachbeobachtet. In dieser Zeit wurde bei allen Patienten mindestens sechs Monate nach dem Eingriff eine transösophageale Echokardiographie mit Shuntdiagnostik durchgeführt. Hierbei zeigte sich bei 27 von 39 Patienten (69%) ein kompletter Verschluss des PFO. Bei neun Patienten verblieb nur noch ein kleiner Restshunt, bei einem Patienten ein mittlerer Restshunt und bei weiteren zwei Patienten ein großer Restshunt. Der Patient mit mittlerem Restshunt und ein Patient mit einem großen Restshunt erhielten einen dritten Okkluder. Letzterer verstarb 21 Tage nach Implantation des dritten Okkluders an einer akuten Perikardtamponade. Eine Patientin mit großem Restshunt entschied sich für eine Operation. Neben dem oben beschriebenen Todesfall nach Implantation eines dritten Okkluders traten während des ganzen Nachbeobachtungszeitraums keine weiteren Komplikationen auf. Diese Arbeit zeigt, dass die perkutane Implantation eines zweiten PFO-Okkluders sicher und technisch problemlos möglich ist. Ein kompletter Verschluss konnte in der Mehrzahl der Patienten erreicht werden. Außer einem Todesfall durch eine akute Perikardtamponade nach Implantation eines dritten Okkluders traten sowohl während den Eingriffen als auch im weiteren Verlauf keine Komplikationen auf.
Ein weit verbreitetes Merkmal von Leukämien sind genetische Veränderungen, wobei die Entstehung der Leukämie häufig mit reziproken chromosomalen Translokationen assoziiert ist, welche zur Bildung chimärer Genprodukte führen. Eine Vielzahl dieser reziproken Translokationen basieren auf Translokationen des MLL Gens (Mixed Lineage Leukemia), die mit dem Krankheitstyp einer AML oder einer ALL verbunden sind. Die häufigste chromosomale Translokation ist die t(4;11) Translokation. Sie tritt vor allem bei Kleinkindern bzw. auch bei älteren Patienten mit einer Sekundärleukämie auf und resultiert in einer akuten lymphatischen Leukämie. Es handelt sich um eine Hochrisikoleukämie, welche aufgrund ihrer nahezu Therapie-resistenten Blasten mit einer besonders schlechten Prognose assoziiert ist. Der Mechanismus der Leukämieentstehung durch die MLL-Translokationen und der dabei entstehenden Fusionsproteine konnte bis heute nicht ausreichend geklärt werden. Für einige MLL-Translokationen, die mit einer myeloischen Leukämie verknüpft sind, konnte zunächst das onkogene Potenzial der Fusionsproteine im Mausmodell belegt werden. Im Bezug auf die t(4;11) Translokation blieben Ansätze zur Etablierung eines Tiermodells jedoch lang erfolglos. Erst 2006 konnten mittels einer knock-in-Strategie bzw. einem „inverter“-System Mausmodelle für MLL•AF4 entwickelt werden, in denen die Mäuse nach sehr langer Latenzzeit ein disseminiertes B-Zell-Lymphom aufwiesen. Ein neueres konditionales MLL•AF4 knock-in-Modell, in dem MLL•AF4 unter der Kontrolle des endogenen Zellzyklus-abhängigen MLL-Promotors steht, resultierte hingegen in einer ALL oder AML. Im Allgemeinen steht somit bislang das MLL•AF4-Fusionsprotein im Vordergrund der Erforschung des pathomolekularen Mechanismus der t(4;11) Translokation. Aufgrund der Tatsache, dass in der Regel jedoch neben dem MLL•AF4- ebenso ein AF4•MLL-Fusionstranskript nachgewiesen werden kann, befassten sich Studien unserer Arbeitsgruppe mit der Funktion des AF4•MLLFusionsproteins. Diese Untersuchungen zeigten, dass das AF4•MLL-Fusionsprotein in der Zelle akkumuliert und in der Entwicklung onkogener Effekte sowie der Wachstumstransformation der Zelle resultiert. Ergänzend belegten retrovirale Transduktions-/Transplantations-Experimente die Entwicklung einer akuten Leukämie im Mausmodell, wenn zuvor mit AF4•MLL bzw. mit AF4•MLL und MLL•AF4 transduzierte Stammzellen transplantiert wurden. Um nun die Funktion des AF4•MLL-Proteins sowie die molekularen Ursachen der beobachteten Eigenschaften besser verstehen zu können, wurde der AF4•MLL-Proteinkomplex mittels einer Strep-Tag-Affinitätschromatographie erfolgreich gereinigt und ein Molekulargewicht von ca. 2 MDa über Größenausschlusschromatographie bestimmt. Damit nicht nur ein Vergleich mit dem MLL- sondern auch mit dem AF4-Wildtyp- Proteinkomplex möglich war, wurden ebenfalls eine Größenbestimmung und eine Reinigung des AF4-Proteinkomplexes durchgeführt. Die folgenden Analysen der Komplexkomposition über Immunopräzipitationen, Western Blot-Analysen sowie massenspektrometrische Analysen zeigten, dass sich der AF4•MLL-Proteinkomplex aus Mitgliedern der beiden Wildtyp-Proteinkomplexe zusammensetzt; es wurden P-TEFb, HEXIM1, NFKB1, NPM1, DDX6 und das AF4-Wildtypprotein aus dem AF4-Komplex sowie ASH2L, RBBP5, WDR5, CREBBP, HCF-1 und HCF-2 aus dem MLL-Komplex nachgewiesen. Auf diese Weise werden im AF4•MLL-Proteinkomplex Eigenschaften bzw. Funktionen beider Wildtyp-Proteinkomplexe kombiniert. Um einen weiteren Hinweis auf die Funktion zu erhalten, wurde ergänzend ein in vitro Histon-Methyltransferase-Assay etabliert, der für beide gereinigten Proteinkomplexe eine Histonmethyltransferase-Aktivität zeigte. Basierend auf den vorliegenden Daten kann eine Konkurrenzsituation zwischen dem AF4•MLL-Proteinkomplex und den beiden Wildtyp-Proteinkomplexen um die entsprechenden Faktoren angenommen werden, welche die Assemblierung vollständiger und funktioneller Wildtyp-Proteinkomplexe verhindern könnte. Des Weiteren weisen die Ergebnisse auf Funktionen des AF4•MLL-Proteins in transkriptionellen Prozessen, Histonacetylierungen sowie der H3K4-Trimethylierung hin. Die Fehlregulation epigenetischer und transkriptioneller Prozesse durch die Anwesenheit des AF4•MLL-Proteinkomplexes spielt somit vermutlich eine entscheidende Rolle im pathomolekularen Mechanismus der t(4;11) Translokation.
A characterization of the ultra-fine aerosol particle counter COPAS (COndensation PArticle counting System) for operation on board the Russian high altitude research aircraft M-55 Geophysika is presented. The COPAS instrument consists of an aerosol inlet and two dual-channel continuous flow Condensation Particle Counters (CPCs) operated with the chlorofluorocarbon FC-43. It operates at pressures between 400 and 50 hPa for aerosol detection in the particle diameter (dp) range from 6 nm up to 1 micro m. The aerosol inlet, designed for the M-55, is characterized with respect to aspiration, transmission, and transport losses. The experimental characterization of counting efficiencies of three CPCs yields dp50 (50% detection particle diameter) of 6 nm, 11 nm, and 15 nm at temperature differences (DeltaT) between saturator and condenser of 17°C, 30°C, and 33°C, respectively. Non-volatile particles are quantified with a fourth CPC, with dp50=11 nm. It includes an aerosol heating line (250°C) to evaporate H2SO4-H2O particles of 11 nm<dp<200 nm at pressures between 70 and 300 hPa. An instrumental in-flight inter-comparison of the different COPAS CPCs yields correlation coefficients of 0.996 and 0.985. The particle emission index for the M-55 in the range of 1.4–8.4×10 16 kg -1 fuel burned has been estimated based on measurements of the Geophysika's own exhaust.
Das Gehirn als Hypoxie-empfindlichstes Organ des menschlichen Körpers ist auf eine konstante Blutversorgung angewiesen. Zur Gewährleistung der Unabhängigkeit von systemischen hypo- bzw. hypertensiven Blutdruckschwankungen, existieren verschiedene Regulationsmechanismen, welche den cerebralen Blutfluss (CBF) in einem weiten Bereich (arterieller Mitteldruck zwischen 60 - 150mmHg) konstant halten. Zu diesen Mechanismen gehören die Blutdruck-adaptierte Autoregulation, die metabolische Vasoreaktivität sowie die neurovasculäre Kopplung. Außerhalb des physiologischen Autoregulationsbereichs folgt der CBF druckpassiv den Schwankungen des systemischen Blutdrucks. Ischämische Hypoperfusion oder Gefäßwand-schädigende Blutdruckspitzen können die Folge sein. Zusätzlich führen verschiedene cerebrovasculäre Erkrankungen zu einer Einschränkung der cAR. Zu diesen gehören die cerebrale Mikroangiopathie, Stenosen bzw. Verschlüsse der A. carotis interna (ACI), der Schlaganfall sowie das Schädel-Hirn-Trauma (SHT). Eine valide Untersuchungsmethode zur Messung der cAR mit geringem apparativen Aufwand und fehlender Strahlenbelastung ist die transcranielle Doppler-Sonographie (TCD). Allerdings stellt eine Verfälschung der Messergebnisse durch verstärkte Streuung des Ultraschallsignals an der Schädelkalotte (schlechtes Schallfenster [sSF]) eine relevante Fehlerquelle dar. Mit steigendem Lebensalter finden sich in bis zu 30% der Bevölkerungen insuffiziente Schallbedingungen. Ziel der vorliegenden Studie war es daher festzustellen, ob die Dauerinfusion eines Ultraschallkontrastmittels (Levovist®) bei der TCD-Autoregulationsmessung an Probanden mit künstlich verschlechtertem Schallfenster eine valide Messung verschiedener Autoregulationswerte ermöglicht. 45 unselektierte Personen mit gutem Schallfenster aus dem Patientengut der Neurologie der J.W.G.-Universität Frankfurt am Main wurden in die Studie eingeschlossen. Die Doppler-Signalqualität, die Blutflussgeschwindigkeiten, der Pulsatilitätsindex nach Gosling und King (PI) sowie die Autoregulationsparameter Phasendifferenz (PD) und Kreuzkorrelations-Koeffizient (Mx) wurden anhand der über das transtemporale Schallfenster abgeleiteten A. cerebri media (ACM) beidseits bestimmt. Zur Imitation des sSF wurde eine Lage Aluminiumfolie ohne Veränderung der Sondenposition zwischen Ultraschallsonde und Haut eingebracht. Die Messungen erfolgten nach einem standardisierten Protokoll in drei Durchgängen (native Messbedingungen, künstliche Schallfensterverschlechterung, Infusion des Ultraschallkontrastmittels (KM) Levovist® [300 mg/min] bei bestehendem sSF) in einem Zeitraum von je 15 Minuten in Ruhe und je 3 Minuten während metronomischer Atmung. Mit künstlicher Verschlechterung des Schallfensters zeigte sich eine signifikante Verfälschung aller erhobenen Messparameter. Die mittlere Flussgeschwindigkeit in der ACM und die mittlere Energie des Doppler-Spektrums verringerten sich um 40%, bzw. um 22%, wohingegen sich der PI verdreifachte. Ebenso veränderten sich die Autoregulationsparameter mit einer Abnahme der PD um durchschnittlich 8 - 10° und des Kreuzkorrelations-Koeffizienten Mx von 0.308 +/- 0.170 auf 0.254 +/- 0.162. Die Verringerung der PD täuscht eine pathologische Einschränkung der cAR vor. Diese Einschränkung entspricht z.B. der einer 75 - 99%-igen Stenose der ACI und kann darüber hinaus auch bei einer Reihe weiterer cerebrovasculärer Erkrankungen (z.B. lakunäre Infarkte bei cerebraler Mikroangiopathie, Subarachnoidalblutung [SAB], maligne Hypertonie) beobachtet werden. Die Abnahme des Kreuzkorrelations-Koeffizienten Mx täuscht im Gegensatz dazu eine scheinbar intakte cAR (Mx < 0.3) vor. Bei Patienten mit schwerem Schädel-Hirn-Trauma (SHT), SAB oder Stenose der ACI besteht in diesem Fall die Gefahr, eine optimale Prävention von Sekundärschäden nicht gewährleisten zu können, da frühe Hinweise auf eine Schädigung des Gehirns, ausgelöst durch verminderte cerebrale Autoregulation, unter Umständen fehlen. Die Infusion des Ultraschallkontrastmittels Levovist® konnte diese Effekte der künstlichen Verschlechterung des Schallfensters auf die Autoregulationsparameter über den gesamten Messzeitraum fast vollständig ausgleichen (PD nativ vs. KM 38.2 +/- 10.0 vs. 37.2 +/- 12.3 und Mx nativ vs. KM 0.308 +/- 0.170 vs. 0.323 +/-0.163). Auch die übrigen erfassten Messparameter näherten sich durchweg den Werten unter nativen Messbedingungen an. Die Verwendung eines Ultraschallkontrastmittels bei der transcraniellen Doppler-Sonographie erhöht also die Validität der Messung der cerebralen Autoregulation bei Patienten mit schlechtem Schallfenster. Eine klinische Relevanz entsteht im besonderen, da sowohl ein schlechtes Schallfenster, als auch cerebrovasculäre Erkrankungen, welche im Zusammenhang mit einer verminderten cerebralen Autoregulation stehen, bevorzugt bei älteren Menschen anzutreffen sind.
The human immunodeficiency virus (HIV) had spread unrecognized in the human population as sexually transmitted disease and was finally identified by its disease AIDS in 1981. Even after the isolation of the causative agent in 1983, the burden and death rate of AIDS accelerated worldwide especially in young people despite the confection of new drugs capable to inhibit virus replication since 1997. However, at least in industrialised countries, this trend could be reversed by the introduction of combination therapy strategies. The design of new drugs is on going; besides the inhibition of the three enzymes of HIV for replication and maturation (reverse transcriptase, integrase and protease), further drugs inhibits fusion of viral and cellular membranes and virus maturation. On the other hand, viral diagnostics had been considerably improved since the emergence of HIV. There was a need to identify infected people correctly, to follow up the course of immune reconstitution of patients by measuring viral load and CD4 cells, and to analyse drug escape mutations leading to drug resistance. Both the development of drugs and the refined diagnostics have been transferred to the treatment of patients infected with hepatitis B virus (HBV) and hepatitis C virus (HCV). This progress is not completed; there are beneficial aspects in the response of the scientific community to the HIV burden for the management of other viral diseases. These aspects are described in this contribution. Further aspects as handling a stigmatising disease, education of self-responsiveness within sexual relationships, and ways for confection of a protective vaccine are not covered.
This book presents a study of the life history of Mtoro bin Mwinyi Bakari (c. 1869 - 1927). Mtoro bin Mwinyi Bakari grew up and studied Islamic Sciences in Bagamoyo, Tanzania. He became a Swahili lecturer and author in Germany and is known to have written Desturi za Wasuaheli, an important work in Swahili culture. The book introduces the wider historical context of his writings, and, in particular, reconstructs the racism and discrimination in both the colonial and metropolitan contexts, features which negatively influenced his career and his life as a whole. The study also offers insights into contributions of the colonized to the study of African languages and cultures during this same historical context.
