Limb loss is a devastating disability and while current treatments provide aesthetic and functional restoration, they are associated with complications and risks. The optimal solution would be to harness the body's regenerative capabilities to regrow new limbs. Several methods have been tried to regrow limbs in mammals, but none have succeeded. One such attempt, in the early 1970s, used electrical stimulation and demonstrated partial limb regeneration. Several researchers reproduced these findings, applying low voltage DC electrical stimulation to the stumps of amputated rat forelimbs reporting "blastema, and new bone, bone marrow, cartilage, nerve, skin, muscle and epiphyseal plate formation". In spite of these encouraging results this research was discontinued. Recently there has been renewed interest in studying electrical stimulation, primarily at a cellular and subcellular level, and studies have demonstrated changes in stem cell behavior with increased proliferation, differentiation, matrix formation and migration, all important in tissue regeneration. We applied electrical stimulation, in vivo, to the stumps of amputated rat limbs and observed significant new bone, cartilage and vessel formation and prevention of neuroma formation. These findings demonstrate that electricity stimulates tissue regeneration and form the basis for further research leading to possible new treatments for regenerating limbs.
The human 5-lipoxygenase (5-LO), encoded by the ALOX5 gene, is the key enzyme in the formation of pro-inflammatory leukotrienes. ALOX5 gene transcription is strongly stimulated by calcitriol (1α, 25-dihydroxyvitamin D3) and TGFβ (transforming growth factor-β). Here, we investigated the influence of MLL (activator of transcript initiation), AF4 (activator of transcriptional elongation) as well as of the leukemogenic fusion proteins MLL-AF4 (ectopic activator of transcript initiation) and AF4-MLL (ectopic activator of transcriptional elongation) on calcitriol/TGFβ-dependent 5-LO transcript elongation. We present evidence that the AF4 complex directly interacts with the vitamin D receptor (VDR) and promotes calcitriol-dependent ALOX5 transcript elongation. Activation of transcript elongation was strongly enhanced by the AF4-MLL fusion protein but was sensitive to Flavopiridol. By contrast, MLL-AF4 displayed no effect on transcriptional elongation. Furthermore, HDAC class I inhibitors inhibited the ectopic effects caused by AF4-MLL on transcriptional elongation, suggesting that HDAC class I inhibitors are potential therapeutics for the treatment of t(4;11)(q21;q23) leukemia.
Cancer research has become a global enterprise, and the number of researchers, as well as the cost for their activities, has skyrocketed. The budget for the National Cancer Institute of the United States National Institutes of Health alone was US$5.2 billion in 2015. Since most of the research is funded by public money, it is perfectly legitimate to ask if these large expenses are worth it. In this brief commentary, we recapitulate some of the breakthroughs that mark the history of breast cancer research over the past decades and emphasize the resulting benefits for afflicted women. In 1971, only 40% of women diagnosed with breast cancer would live another 10 years. Today, nearly 80% of women reach that significant milestone in most developed countries. This dramatic change has afforded breast cancer patients many productive years and a better quality of life. Progress resulted largely from advances in the understanding of the molecular details of the disease and their translation into innovative, rationally designed therapies. These developments are founded on the revolution in molecular and cellular biology, an entirely new array of methods and technologies, the enthusiasm, optimism, and diligence of scientists and clinicians, and the considerable funding efforts from public and private sources. We were lucky to be able to spend our productive years in a period of scientific upheaval in which methods and concepts were revolutionized and that allowed us to contribute, within the global scientific community, to the progress in basic science and clinical practice.
An optochemokine tandem was developed to control the release of calcium from endosomes into the cytosol by light and to analyze the internalization kinetics of G-protein coupled receptors (GPCRs) by electrophysiology. A previously constructed rhodopsin tandem was re-engineered to combine the light-gated Ca2+-permeable cation channel Channelrhodopsin-2(L132C), CatCh, with the chemokine receptor CXCR4 in a functional tandem protein tCXCR4/CatCh. The GPCR was used as a shuttle protein to displace CatCh from the plasma membrane into intracellular areas. As shown by patch-clamp measurements and confocal laser scanning microscopy, heterologously expressed tCXCR4/CatCh was internalized via the endocytic SDF1/CXCR4 signaling pathway. The kinetics of internalization could be followed electrophysiologically via the amplitude of the CatCh signal. The light-induced release of Ca2+ by tandem endosomes into the cytosol via CatCh was visualized using the Ca2+-sensitive dyes rhod2 and rhod2-AM showing an increase of intracellular Ca2+ in response to light.
A growing body of evidence suggests that macrophage polarization dictates the expression of iron-regulated genes. Polarization towards iron sequestration depletes the microenvironment, whereby extracellular pathogen growth is limited and inflammation is fostered. In contrast, iron release contributes to cell proliferation, which is important for tissue regeneration. Moreover, macrophages constitute a major component of the infiltrates in most solid tumors. Considering the pivotal role of macrophages for iron homeostasis and their presence in association with poor clinical prognosis in tumors, we approached the possibility to target macrophages with intracellular iron chelators. Analyzing the expression of iron-regulated genes at mRNA and protein level in primary human macrophages, we found that the iron-release phenotype is a characteristic of polarized macrophages that, in turn, stimulate tumor cell growth and progression. The application of the intracellular iron chelator (TC3-S)2 shifted the macrophage phenotype from iron release towards sequestration, as determined by the iron-gene profile and atomic absorption spectroscopy (AAS). Moreover, whereas the addition of macrophage supernatants to tumor cells induced tumor growth and metastatic behavior, the supernatant of chelator-treated macrophages reversed this effect. Iron chelators demonstrated potent anti-neoplastic properties in a number of cancers, both in cell culture and in clinical trials. Our results suggest that iron chelation could affect not only cancer cells but also the tumor microenvironment by altering the iron-release phenotype of tumor-associated macrophages (TAMs). The study of iron chelators in conjunction with the effect of TAMs on tumor growth could lead to an improved understanding of the role of iron in cancer biology and to novel therapeutic avenues for iron chelation approaches.
Estimates suggest that more than 25,000 to 125,000 people die annually from snakebite envenomation worldwide. In contrast to this major disease burden, thorough bibliometric studies do not exist so far that illustrate the overall research activity over a long time span. Therefore, the NewQIS-platform conducted an analysis on snakebite envenoming using the Thomson Reuters database Web of Science. To determine and assess changes regarding the scientific activities and to specifically address the more recent situation we analyzed two time intervals (t). During the first time interval from 1900 to 2007 (t1) 13,015 publications (p) were identified. In the following period (2008–2016 = t2) 4,982 publications were identified by the same search strategy. They originate from 114 (t1) respectively 121 countries (t2), with the USA (p = 3518), Brazil (p = 1100) and Japan (p = 961) being most productive in the first period, and the USA (p = 1087), Brazil (p = 991) and China (p = 378) in the second period, respectively. Setting the publication numbers in relation to GDP/capita, Brazil leads with 92 publications per 10,000 Int$GDP/capita, followed by India with 79 publications per 10000 Int$GDP/capita (t1). Comparing the country’s publication activity with the Human Development Index level indicates that the majority of the publications is published by highly developed countries. When calculating the average citation rates (citations per published item = CR) mainly European countries show the highest ranks: From 1900–2007 Sweden ranks first with a CR = 27, followed by the Netherlands (CR = 24.8), Switzerland (CR = 23), Spain, Austria and the USA (CR = 22). From 2008 to 2016 the highest rate achieves Switzerland with a value of 24.6, followed by Belgium (CR = 18.1), Spain (CR = 16.7), Costa Rica (CR = 14.9) and Netherlands (CR = 14). Compared with this, the USA was placed at rank 13 (CR = 9,5).
In summary, the present study represents the first density-equalizing map projection and in-depth scientometric analysis of the global research output on snakebites and its venoms. So it draws a sketch of the worldwide publication architecture and indicates that countries with a high incidence of snakebites and a low economical level still need to be empowered in carrying out research in this area.

BACKGROUND: Inflammatory events after kidney transplantation (Tx) may lead to activation of the tryptophane-catabolizing enzyme indoleamine 2,3-dioxygenase followed by the formation of kynurenine (KYN). Post-transplant KYN serum levels in kidney allograft recipients were analyzed for their diagnostic value.
MATERIAL AND METHODS: This was a retrospective analysis of KYN levels (normal value: 2.7±0.6 nmol/ml) measured in 4083 blood samples collected from 355 kidney graft recipients in connection with uncomplicated courses, acute rejections (ARs), infections, and type of immunosuppression. We performed descriptive data analysis and analysis of variance.
RESULTS: In 212 recipients with immediately functioning grafts, the KYN levels dropped from pre-Tx 13.3±5.9 nmol/ml to nearly normal values at day 5 (5.8±3.0 nmol/ml). In patients with delayed graft function, the KYN reduction started only after the last hemodialysis treatment. With respect to ARs in recipients with creatinine values <300 µmol/l pre-AR, the increase of KYN levels depended on the severity of ARs (steroid-sensitive ARs: from 4.5±1.4 to 6.0±6.1 nmol/ml; steroid-resistant ARs: from 6.1±3.1 to 12.9±7.1 nmol/ml; vascular rejections: from 5.8±3.0 to 16.9±9.1 nmol/ml). In patients with creatinine values ≥300 µmol/l pre-AR, a further increase of the KYN level (from 10.1 to 13.2 nmol/ml) was only observed in severe, steroid-resistant ARs. With respect to infections evaluated, the KYN levels before diagnosis/start of treatment were 5.7±3.4 nmol/ml in asymptomatic CMV infections, 7.5±4.4 nmol/ml in CMV diseases, 8.3±3.3 nmol/ml in pneumonia, and 10.4±6.5 nmol/ml in bacterial sepsis.
CONCLUSIONS: Serum KYN seems to be a reliable diagnostic tool for the assessment of post-transplant inflammatory complications, already in an early stage, and for monitoring the efficacy of therapeutic interventions. Prospective studies are recommended.
The P300/CBP-associated factor plays a central role in retroviral infection and cancer development, and the C-terminal bromodomain provides an opportunity for selective targeting. Here, we report several new classes of acetyl-lysine mimetic ligands ranging from mM to low micromolar affinity that were identified using fragment screening approaches. The binding modes of the most attractive fragments were determined using high resolution crystal structures providing chemical starting points and structural models for the development of potent and selective PCAF inhibitors.
Ceraceosorus bombacis is an early-diverging lineage of smut fungi and a pathogen of cotton trees (Bombax ceiba). To study the evolutionary genomics of smut fungi in comparison with other fungal and oomycete pathogens, the genome of C. bombacis was sequenced and comparative genomic analyses were performed. The genome of 26.09 Mb encodes for 8,024 proteins, of which 576 are putative-secreted effector proteins (PSEPs). Orthology analysis revealed 30 ortholog PSEPs among six Ustilaginomycotina genomes, the largest groups of which are lytic enzymes, such as aspartic peptidase and glycoside hydrolase. Positive selection analyses revealed the highest percentage of positively selected PSEPs in C. bombacis compared with other Ustilaginomycotina genomes. Metabolic pathway analyses revealed the absence of genes encoding for nitrite and nitrate reductase in the genome of the human skin pathogen Malassezia globosa, but these enzymes are present in the sequenced plant pathogens in smut fungi. Interestingly, these genes are also absent in cultivable oomycete animal pathogens, while nitrate reductase has been lost in cultivable oomycete plant pathogens. Similar patterns were also observed for obligate biotrophic and hemi-biotrophic fungal and oomycete pathogens. Furthermore, it was found that both fungal and oomycete animal pathogen genomes are lacking cutinases and pectinesterases. Overall, these findings highlight the parallel evolution of certain genomic traits, revealing potential common evolutionary trajectories among fungal and oomycete pathogens, shaping the pathogen genomes according to their lifestyle.
Measurements of the transverse momentum (pt) spectra of K0 s and Λ(Λ̄) in Pb–Pb and pp collisions at √sNN = 2.76TeV with the ALICE detector at the LHC at CERN up to pt = 20GeV/c and pt = 16GeV/c, respectively, are presented in this thesis. In addition, the particle rapidity densities at mid-rapidity and nuclear modification factors of K0 s and Λ(Λ̄) are shown and discussed. The analysis was performed using the Pb–Pb data set from 2010 and the pp data set from 2011. For the identification of K0 s and Λ(Λ̄), the on-the-fly V0 finder was employed on tracking information from the TPC and ITS detectors. The Λ and Λ̄ spectra were feed-down corrected using the measured published Ξ− spectra as input.
Regarding the rapidity density at mid-rapidity, a suppression of the strange particle production in pp as compared to Pb–Pb collisions is observed at all centralities, whereas the production per pion rapidity density stays constant as a function of dNch/dη including both systems. Furthermore, the relative increase of the individual particle species in pp and AA collisions is compatible for non- and single-strange particles when going from RHIC (√sNN = 0.2TeV) to LHC energies. On the other hand, in case of multi-strange baryons, a stronger increase in the particle production in pp is seen. The Λ̄ and Λ production in Pb–Pb and pp collisions was found to be equal. Concerning the nuclear modification factors, at lower pt (pt <5GeV/c), an enhancement of the RAA of Λ with respect to that of K0 s and charged hadrons is observed. This baryon-to-meson enhancement appearing in central Pb–Pb collisions at RHIC and LHC is currently explained by the interplay of the radial flow and recombination as the dominant particle production mechanism in this pt sector. The effect of radial flow is thus also seen in the low and intermediate pt region of RAA, where a mass hierarchy is discovered among the baryons and mesons, respectively, with the heaviest particle being least suppressed. When comparing the results from RHIC and LHC, the RCP is found to be similar at low-to-intermediate pt, while a significantly smaller RAA of K0 s and Λ in central and peripheral events at the LHC is observed in this pt region as compared to the RHIC results. This can be attributed to the larger radial flow in AA collisions and to the harder spectra at the LHC. At high pt (pt > 8GeV/c), a strong suppression in central Pb–Pb collisions with respect to pp collisions is found for K0 s and Λ(Λ̄). A significant high-pt suppression of these hadrons is also observed in the ratio of central-to-peripheral collisions. The nuclear modification of K0 s and Λ(Λ̄) is compatible with the modification of charged hadrons at
high pt. The calculations with the transport model BAMPS agree with these results suggesting a similar energy loss for all light quarks, i.e. u, d and s. Moreover, a compatible suppression for c-quarks appears in the ALICE measurements via the D meson RAA as well as in the BAMPS calculations, which hints to a flavour-independent suppression if light- and c-quarks are regarded. Within this consideration, no indication for a medium-modified fragmentation is found yet.
To summarize, for the particle production in Pb–Pb collisions at the LHC relative to pp neither at lower pt (rapidity density) nor at higher pt (nuclear modification factor) a significant difference of K0 s and Λ(Λ̄) carrying strangeness to hadrons made of u- and d-quarks was found.
Mit der hier vorliegenden Arbeit ist das Entladungsverhalten einer Dielektrischen Barriere Entladung anhand der elektrischen Parameter untersucht worden. Dazu wurde ein planparallele Elektrodenkonfiguration entwickelt und aufgebaut, die mit einer Sinus-Spannungs von maximal 5000 Vss und einer Frequenz von 5 kHz-20 kHz angesteuert wurde. Als Arbeitsgas wurde Argon im Druckbereich von 100 mbar - 1000 mbar verwendet. Auf diese Weise konnte ein Plasma erzeugt werden, in das bei einer maximalen Transfereffiienz von 96%, eine mittlere Leistung von bis zu 845 mW eingekoppelt werden konnte.
Da die Dielektrische Barriere Entladung auf Grund der Abschirmung der Elektroden vom Gasraum einige Besonderheiten gegenüber eines klassischen kapazitiv eingekoppelten Plasmas aufweist, können keine Rückschlüsse mittels einer einfachen Strom-Spannungsmessung auf die Vorgänge im Plasma gemacht werden.
Um trotzdem Einblick in die Entladung zu erhalten, wurde das von [Tra08] vorgeschlagene Ladungstransportmodel für die Analyse herangezogen und an den hier vorliegenden experimentellen Aufbau angepasst. So konnte unter anderem der Ein
uss der auf den Dielektrika befindlichen Restladungsträger auf die Ausbildung der Entladung untersucht werden. Des Weiteren ist aus den gewonnen Größen die Elektronendichte des Plasmas bestimmt worden. Diese liegt bei der hier untersuchten dielektrischen Glimmentladung, je nach Gasdruck und Frequenz, im Bereich zwischen 5 X 10exp9 - 2 X 10exp10 1/cm3 . Auch konnten in diesem Zusammenhang qualitative Aussagen über die Entwicklung der Elektronentemperatur gemacht werden.
Zusammen mit Kurzzeitaufnahmen, die den Verlauf der Entladung dokumentieren und den in [Sch11] gemachten Untersuchungen zur Excimerstrahlung konnte so ein umfassendes Bild der Vorgänge im Plasma erstellt und Kriterien erarbeitet werden, die den für eine Dielektrische Barriere Entladung typischen Ubergang zwischen einer Glimmentladung zu einer filamentierten Entladung erklären können.
Crystallization and X-ray diffraction studies of a complete bacterial fatty-acid synthase type I

While a deep understanding of the fungal and mammalian multi-enzyme type I fatty-acid synthases (FAS I) has been achieved in recent years, the bacterial FAS I family, which is narrowly distributed within the Actinomycetales genera Mycobacterium, Corynebacterium and Nocardia, is still poorly understood. This is of particular relevance for two reasons: (i) although homologous to fungal FAS I, cryo-electron microscopic studies have shown that bacterial FAS I has unique structural and functional properties, and (ii) M. tuberculosis FAS I is a drug target for the therapeutic treatment of tuberculosis (TB) and therefore is of extraordinary importance as a drug target. Crystals of FAS I from C. efficiens, a homologue of M. tuberculosis FAS I, were produced and diffracted X-rays to about 4.5 Å resolution.
Little evidence exists on the financing decisions of newly founded firms or on the financing dynamics of these firms over their life cycle. We aim to help filling this gap by investigating the financing dynamics of 2,456 French manufacturing firms founded between 2004 and 2006 through their legally required and reported financial statements. Because we observe significant heterogeneity in the financing decision in the firms' founding year, we focus on analyzing whether these differences widen, persist, or converge by using different convergence concepts. We identify a persistence-cum-convergence pattern. We find the existence of ß-convergence (implying that e.g. firms with lower initial levels of debt accumulate more debt over time) but not of σ-convergence (i.e. we observe an increase in the cross-sectional dispersion of the financing structure). We also show that the dynamics of financing matter for the growth path of the firms.
We compared outcomes from a single-arm study of blinatumomab in adult patients with B-precursor Ph-negative relapsed/refractory acute lymphoblastic leukemia (R/R ALL) with a historical data set from Europe and the United States. Estimates of complete remission (CR) and overall survival (OS) were weighted by the frequency distribution of prognostic factors in the blinatumomab trial. Outcomes were also compared between the trial and historical data using propensity score methods. The historical cohort included 694 patients with CR data and 1112 patients with OS data compared with 189 patients with CR and survival data in the blinatumomab trial. The weighted analysis revealed a CR rate of 24% (95% CI: 20–27%) and a median OS of 3.3 months (95% CI: 2.8–3.6) in the historical cohort compared with a CR/CRh rate of 43% (95% CI: 36–50%) and a median OS of 6.1 months (95% CI: 4.2–7.5) in the blinatumomab trial. Propensity score analysis estimated increased odds of CR/CRh (OR=2.68, 95% CI: 1.67–4.31) and improved OS (HR=0.536, 95% CI: 0.394–0.730) with blinatumomab. The analysis demonstrates the application of different study designs and statistical methods to compare novel therapies for R/R ALL with historical data.
This article reviews the most recent results concerning second harmonic generation (SHG) experiments of non-phase matchable and phase matchable powder samples at high pressures and explains the pressure dependence of the intensity of the SHG signal by correlating it to the ratio between the average coherence length and the average particle size. The examples discussed here include pressure-induced structural changes in quartz, ZnO, ice VII and KIO3. It is shown that the second harmonic generation technique is a unique tool for the detection of pressure-induced structural phase transitions. It is laboratory based and allows fast measurements. It is complementary to X-ray diffraction and provides additional information about the presence of an inversion center for unknown or controversially discussed structures at high pressure.
During CNS development and adult neurogenesis, immature neurons travel from the germinal zones towards their final destination using cellular substrates for their migration. Classically, radial glia and neuronal axons have been shown to act as physical scaffolds to support neuroblast locomotion in processes known as gliophilic and neurophilic migration, respectively (Hatten, 1999; Marin and Rubenstein, 2003; Rakic, 2003). In adulthood, long distance neuronal migration occurs in a glial-independent manner since radial glia cells differentiate into astrocytes after birth. A series of studies highlight a novel mode of neuronal migration that uses blood vessels as scaffolds, the so-called vasophilic migration. This migration mode allows neuroblast navigation in physiological and also pathological conditions, such as neuronal precursor migration after ischemic stroke or cerebral invasion of glioma tumor cells. Here we review the current knowledge about how vessels pave the path for migrating neurons and how trophic factors derived by glio-vascular structures guide neuronal migration both during physiological as well as pathological processes
Im Rahmen dieser Diplomarbeit haben Arbeiten am Elektronen-Streuexperiment von Prof. J. Jacoby und Dr. R. Berezov an der Goethe-Universität Frankfurt stattgefunden. Am Experiment wurden durch Møller-Streuung verschränkte Elektronen auf ihre Spin-Abhängigkeit untersucht. Die Untersuchung erfolgte als Koinzidenzmessung, dessen Genauigkeit von der zeitlichen Auflösung der Detektoren abhängt. Als Koinzidenzlogik diente ein von Julian Schunk entwickeltes Datenerfassungsprogramm, das auf einem Oszilloskop installiert wurde...
Katamnese und Lebenszufriedenheit von Kindern und Jugendlichen mit Geschlechtsidentitätsstörungen

Die Behandlung von Kindern und Jugendlichen mit Geschlechtsidentitätsstörungen (GIS) wird seit dem Beginn der pubertätshemmenden Hormontherapie in den neunziger Jahren international kontrovers diskutiert. Diese Störung scheint bei Kindern durch psychotherapeutische Intervention besser behandelbar als bei Jugendlichen. Erwartet wurden weniger psychopathologische Auffälligkeiten und eine höhere Lebenszufriedenheit bei umfassender psychotherapeutischer Begleitung. Des Weiteren wurden die Hypothesen geprüft, dass sich ein Geschlechtswechsel ebenfalls positiv auf Lebenszufriedenheit und Psychopathologie auswirkt.
Es nahmen insgesamt 37 Kinder, Jugendliche und Erwachsene an der schriftlichen Nachuntersuchung teil, die mindestens drei Jahre vor Studienbeginn aufgrund der Diagnose GIS des Kindes- und Jugendalters in der KJP Frankfurt vorgestellt wurden. Erfasst wurden Daten zur Geschlechtsidentität, zum Behandlungsverlauf und zur sexuellen Orientierung, zur Ausprägung der Psychopathologie laut altersangemessenem Screening-Inventar (CBCL, YSR, YASR) und zur Lebenszufriedenheit mithilfe des Inventars zur Erfassung der Lebensqualität bei Kindern und Jugendlichen (ILK). Außerdem wurden die Ausprägung der Psychopathologie (erfasst mit dem CBCL und oder YSR) bei der Erstvorstellung in der Klinik mit der jetzigen Einschätzung verglichen.
Die erhobenen Daten geben keinen Hinweis darauf, dass umfangreiche Psychotherapie oder ein Geschlechtswechsel zu einer höheren Lebenszufriedenheit und einer Verbesserung der Psychopathologie führen. Im Laufe der Zeit kam es zu einer signifikanten Reduktion der Werte der Syndromskalen soziale Probleme und aggressives Verhalten bei allen Studienteilnehmern. Allerdings wurde ein signifikanter Zusammenhang zwischen der Lebenszufriedenheit im Bereich seelische Gesundheit und der Zufriedenheit mit ihrer Psychotherapie gefunden. Die Drop-out-Analyse zeigte, dass Betroffene mit einer unbewältigten Problematik im Zusammenhang mit ihrer GIS eher nicht bereit waren, an dieser Studie teilzunehmen. Des Weiteren ergab sich, dass sich in der Gruppe der Studienteilnehmer signifikant mehr Personen für einen Geschlechtswechsel entschieden haben als in der Gruppe der Studienabbrecher.
Aus den Ergebnissen lässt sich ableiten, dass alle Teilnehmer der Nachuntersuchung mit ihrem Lebensweg gleichsam zufrieden sind, unabhängig vom Ausmaß der Psychotherapie und der Entscheidung für einen Geschlechtswechsel. Außerdem haben alle Personen der Stichprobe heute weniger Probleme mit aggressivem Verhalten und weniger soziale Probleme.
Da von einer Verzerrung der Stichprobe durch einen systematischen Ausfall von Teilnehmern auszugehen ist, sind weitere Nachuntersuchungen nötig, um die Hypothesen dieser Arbeit weiter zu überprüften. Darüber hinaus gilt es zu klären, ob bestimmte Psychotherapieformen bei einigen Patienten effektiver als andere sind.
Meine Überlegungen auf diesen Seiten drehen sich um ein von Walter Benjamin handbeschriebenes Blatt Papier, an dem sich die "performative" Dimension von Benjamins Poetik paradigmatisch illustrieren lässt. Das Manuskriptblatt (Benjamin-Archiv Ms 931) wurde von Gershom Scholem auf ca. 1935 datiert und gehört thematisch in den Umkreis des etwa zwei Jahre vorher verfassten sprachtheoretischen Essays "Lehre vom Ähnlichen". Dort heißt es, dass die "Einsicht in die Bereiche des 'Ähnlichen' [...] weniger im Aufweis angetroffener Ähnlichkeiten [zu gewinnen sei] als durch die Wiedergabe von Prozessen, die solche Ähnlichkeiten erzeugen" (GS II, 204). In Benjamins Beschriftung von Ms 931 wird dies in die Tat umgesetzt: wir können uns den Schreibprozess, dem sich der handschriftliche Text auf jenem Blatt verdankt, als Selbsterforschung eines solchen Prozesses denken. In dem vorliegenden Versuch scheint jedoch, neben der Erzeugung von Ähnlichkeiten, die "Überwindung des Mythos" (Ms 931) als zweiter Pol am Horizont auf. Zusammen stecken jene beiden Pole ein dialektisches Spannungsfeld ab, in welchem sich sowohl Benjamins Schreibprozess als auch der meinige bewegen, und dessen Gesetzlichkeit das eigentliche Objekt dieser Überlegungen ausmacht. ...
Die Populationsgenetik beschäftigt sich mit dem Einfluss von zufälliger Reproduktion, Rekombination, Migration, Mutation und Selektion auf die genetische Struktur einer Population.
In dieser Arbeit mit dem englischen Titel "Ancestral lines under mutation and selection" wird das Zusammenspiel von zufälliger Reproduktion, gerichteter Selektion und Zweiwegmutation untersucht.
Dazu betrachten wir eine haploide Population in der jedes Individuum zu jedem Zeitpunkt genau einen von zwei Typen aus S:={0,1} trägt. Dabei sei 1 der neutrale und 0 der selektiv bevorzugte Typ. Im Diffusionslimes sehr großer Populationen modellieren wir den Prozess der Frequenz der Typ-0-Individuen durch eine Wright-Fisher-Diffusion X:=(X_t) mit Mutation und gerichteter Selektion.
Zu jedem Zeitpunkt s gibt es genau ein Individuum, dessen Nachkommen ab einem bestimmten zukünftigen Zeitpunkt t>s die gesamte Population ausmachen werden. Wir nennen dieses Individuum den gemeinsamen Vorfahren zum Zeitpunkt s, da alle Individuen zu allen Zeitpunkten r>t von ihm abstammen. Sei R_{s} dessen Typ zum Zeitpunkt s. Wir nehmen an, dass der Prozess X zum Zeitpunkt 0 im Gleichgewicht ist und definieren die Wahrscheinlichkeit, dass der gemeinsame Vorfahre zum Zeitpunkt 0 Typ 0 hat, durch h(x):= P(R_{0}=0|X_{0}=x). Eine Darstellung von h(x) wurde bereits von Fearnhead (2002) und Taylor (2007) gefunden und dort mit vorwiegend analytischen Methoden bewiesen. In dieser Arbeit entwickeln wir in Kapitel 3 ein neues Teilchenbild, den pruned lookdown ancestral selection graph (pruned LD-ASG), der für sich selbst genommen interessant ist und eine neue probabilistische Interpretation der Darstellung von h(x) liefert.
Durch Erweiterung des Teilchenbildes auf Nachkommenverteilungen mit schweren Tails und mit Hilfe einer Siegmund Dualität gelingt es uns in Kapitel 4 das Resultat für h(x) von klassischen Wright-Fisher-Diffusionen auf Lambda-Wright-Fisher-Diffuison zu erweitern.
Eine Verbindung zwischen Ideen von Taylor (2007), der den gemeinsamen Prozess (X,R) untersucht hat, und einem von Fearnhead (2002) betrachteten Prozess (R,V), der die Entwicklung des Typs R des gemeinsamen Vorfahren in einer Umgebung von V sogenannten virtuellen Linien beschreibt, stellen wir in Kapitel 6 her. Wir bestimmen die gemeinsame Dynamik des Tripels (X,R,V). In Kapitel 7 betrachten wir ein diskretes Bild mit endlicher Populationsgröße N und schlagen dort eine Brücke zu Resultaten von Kluth, Hustedt und Baake (2013).
Des Weiteren entwickeln wir in Kapitel 5 dieser Arbeit einen Algorithmus zur Simulation der Typen einer Stichprobe von m Individuen, die aus einer Wright-Fisher-Population mit Mutation und Selektion im Gleichgewicht gezogen wird. Mittels dieses Algorithmus illustrieren wir die Typenverteilung für verschiedene Parameterwerte und Stichprobengrößen.
Fungi are an important component of every ecosystem but hardly considered in biodiversity monitoring projects. This thesis aims at characterizing fungal diversity, with an emphasis on epigeous fungi, encompassing different biogeographic zones and points in time. A main sampling area was established in the Taunus mountain range in Germany, which was sampled monthly over three years.
For testing species richness on spatial scale, the Taunus transect was compared with four other areas, which were assessed with lower sampling effort. One of these areas was Bulau in Germany, in which four excursions were made. Furthermore, two sampling events were performed in Somiedo in Spain and one sampling event in Kleinwalsertal in Austria. Already existing data of a two-year monitoring project in Panama next to the river Majagua were additionally used for comparison.
All these areas were investigated with a standardized sampling protocol focusing on macroscopically evident fungi and vascular plants using a time-restricted transect design. The transects consisted of strips, which were 500 m long and about 20 m broad, and were sampled for 2 hours at each single sampling event....

Schlimmer noch, die Aussage „Ich kann nicht singen“ oder „Mathematik habe ich noch nie verstanden“ wird sie sicher nicht von einer erfolgreichen Karriere abhalten, noch wird es die Meinung anderer über sie ändern.
Das Projekt „European Music Portfolio – Sounding Ways into Mathematics“ (EMP-Maths) möchte dieses Verständnis ändern. Jeder kann singen und Musik machen und jeder kann Mathematik treiben. Beide Themen sind integraler Bestandteil unseres Lebens und unserer Gesellschaft. Was geändert werden muss, ist die Fähigkeit von Lehrpersonen, Lernenden die Gelegenheit zu geben, sie zu mögen.
Dieses Handbuch für Lehrpersonen stellt Aktivitäten mit unterschiedlichen mathematischen und musikalischen Inhalten vor, um Lehrpersonen ein möglichst breites Spektrum an Hilfsmitteln, Ideen und Beispielen anbieten zu können. Diese Aktivitäten sind so aufgebaut, dass sie erweiter- und anpassbar an unterschiedliche Kontexte sowie auf die Bedürfnisse einer jeden Lehrperson und deren Schülerinnen und Schülern sind. Ferner wurden diese Aktivitäten nicht nur entwickelt, um von der Lehrperson instruktiv ausgeführt zu werden, sondern, um sie gemeinsam mit der Lerngruppe zu nutzen und eventuell sogar gemeinsam zu verändern und weiter zu entwickeln.
Neben diesem Handbuch stellt das Projekt Weiterbildungsangebote zur Verfügung, eine Web-Seite (http://maths.emportfolio.eu), von der Materialien heruntergeladen werden können sowie eine Online Plattform zur interaktiven Zusammenarbeit. Ein Überblick über relevante Literatur und Forschung ist in einem separaten Dokument zusammengestellt. Zusätzliche Arbeitsbücher für Lehrpersonen enthalten weitere Aktivitäten sowie ausgewählte Aspekte aus dem hier zusammengestellten theoretischen Hintergrund. Diese Arbeitsbücher bilden die Grundlage der Weiterbildungskurse. Viele weitere Beispiele und Vorschläge sind bereits vorhanden (siehe Web-Seite des Projekts) und wir möchten jeden ermutigen, sie zu nutzen. Die Auswahl im Handbuch deckt einige zentralen Felder der Mathematik und der Musik ab: Singen, Tanzen, Hören, Probleme lösen, Zahlen, Messen, Raum und Form. Mit diesem Ansatz wollen wir das Projekt an die Kerncurricula der beteiligten Länder anbinden: Deutschland, Griechenland, Rumänien, Slowakei, Spanien, Schweiz und Großbritannien. Die Dokumentation der Beispiele erfolgt in einer Art von Didaktischen Design Patterns, deren Struktur an die Anforderungen des Projekts angepasst wurde.

Music and mathematics share an odd character: many people believe that they are not good at one or the other (or both). However, ‘I cannot sing’ or ‘I never understood mathematics’ will probably not keep them from having successful careers, and nor will it change the opinions others have about them.
The project ‘European Music Portfolio – Sounding Ways into Mathematics’ (EMP-Maths) aims towards a different understanding with regards to this character. Everyone can sing and make music, and everyone can do mathematics. Both topics are integral parts of our life and society. What needs to be improved is our ability to give students opportunities to like them.
This teacher’s handbook presents activities with different mathematical and musical content in order to offer teachers resources, ideas and examples. These activities are designed to be expandable, adaptable to different contexts, and adjustable to the needs of each teacher and their students. Furthermore, these activities are not just planned to be carried out individually; a teaching unit could be used to make sense of them, or they could even be developed in connection with each other.
Apart from this teacher’s handbook, the project provides a continuing professional development (CPD) course, a webpage (http://maths.emportfolio.eu) from which all materials can be downloaded, and an online collaboration platform. A general overview of related literature and research is available in separate documents. Additional teacher booklets provide related materials and a brief overview of the theoretical background, and are the basis for the CPD courses.
