Im Rahmen dieser Arbeit wurde die Funktion ausgewählter Gene näher charakterisiert und ihr globaler Einfluß auf die Regulation der Genexpression untersucht. Im Fokus der Untersuchungen stand ein Gen, welches im Rahmen der Promotion von Alexander Zaigler in der Arbeitsgruppe Soppa (Universität Frankfurt) zunächst als Transkriptionsregulator ähnlich zu rspA aus E. coli identifiziert wurde. Zu Beginn dieser Arbeit konnte durch in silico Analysen das entsprechende Genprodukt zur Enolase Superfamilie (COG1441) zugeordnet werden. Mit Hilfe der „pop-in/pop-out“ Methode wurde das Gen in H. volcanii in frame deletiert und durch Wachstumsversuche, Northern- sowie Westernblot Analysen näher charakterisiert. Bei der Untersuchung des Wachstums konnte ein bemerkenswerter Phänotyp entdeckt werden: nur der Wechsel der Nährstoffquelle von reichhaltigen zu armen Bedingungen resultierte in einer dreitägigen Lagphase. Darüber hinaus wurde die Genexpression im Laufe eines Wachstumzyklus mittels Northern- und Westernblot Analysen bestimmt. Während das Transkript in reichhaltigem Medium nur transient expremiert wurde, wurde es in nährstoffarmen Bedingungen in allen Wachstumsphasen sehr stark induziert. Durch die Ergebnisse konnte zudem eine translationale Regulation der Genexpression nachgewiesen werden. Die Resultate offenbarten eine wichtige Funktion bei der Transition von reichem zu ärmerem Nährstoffangebot und führten schließlich zur Genbezeichnung iftA („important for transition A“). Desweiteren wurden Transkriptomuntersuchungen der Deletionsmutante im Vergleich zum Wildtyp zum Zeitpunkt der höchsten transienten Expression von iftA durchgeführt. Dadurch konnte gezeigt werden, dass iftA etwa 1% aller Gene des Genoms beeinflußt und gleichzeitig die Zahl differentieller Funktionen dieser Gene sehr gering ist. Die Ergebnisse führten insgesamt zu der Annahme, dass iftA eine Doppelfunktion besitzt, sowohl als enzymatisches Protein im Energiestoffwechsel als auch als essentieller Regulator mit noch unbekannter Funktion. Neben iftA fiel das Augenmerk auf mehrere Gene, die im Rahmen der Promotion von Neta Altman-Price an der Universität in Tel-Aviv als Histon-Acetylasen und –Deacetylasen identifiziert wurden. Zudem konnte in H. volcanii ein essentielles Gen, welches für ein Histon kodiert, entdeckt werden. Das Histon besitzt konservierte Lysinreste, die im eukaryotischen Histon H3 Ziele für eine posttranslationale Acetylierung sind. Für die weiteren Untersuchungen wurden die Lysinreste irreversibel zum einen in Glutamin (acetylierter Zustand) und zum anderen in Arginin (deacetylierter Zustand) mutiert. Im Rahmen dieser Arbeit wurden die Funktionen und die Einflüsse der beiden Histonmutanten sowie einer Acetylase- (delta pat1) und einer Deacetylase-Deletionsmutante (delta sir2) auf die globale Genregulation analysiert. Dazu wurden zunächst Transkriptomuntersuchungen in der exponentiellen Wachstumsphase mittels Microarray Analysen an den Mutanten im Vergleich zum Wildytp durchgeführt. Die Ergebnisse zeigten einen erheblich stärkeren Einfluß auf die Regulation der Genexpression durch das acetylierte Histon sowie der Deacetylase-Deletionsmutante im Vergleich zu den Ergebnissen des deacetylierten Histons und der Acetylase-Deletionsmutante. In der exponentiellen Wachstumsphase liegt das Histon in H. volcanii daher in überwiegend deacetylierter Form vor. Durch die Analysen konnte auch demonstrieren werden, dass Sir2 und das Histon eine regulatorische Wirkung auf exakt die gleichen Gene ausüben und daher unmittelbar miteinander in Verbindung stehen. Die experimentelle Bestätigung dieses Zusammenhangs stellt im Reich der Archaea bisher ein absolutes Novum dar. Die Untersuchungen des Transkriptoms der delta sir2 Mutante enthüllte zudem einen positiven Einfluß von Sir2 auf ein größeres Gencluster (HVO_1201-25). Die Gene dieses Clusters konnten durch in silico Analysen der Chemotaxis und der Flagellenbiosynthese zugeordnet werden. Für die weitere Charakterisierung der Deletionsmutante wurden daher Untersuchungen zur Bestimmung der Motilität von H. volcanii durchgeführt. Es konnte gezeigt werden, dass die Deletion von Sir2 die Beweglichkeit und gerichtete Fortbewegung von H. volcanii in erheblichem Maße beeinflußte, während die Biosynthese der Flagellen nicht beeinträchtigt war. Die Deacetylierung spielt daher eine unmittelbar Rolle bei der Signaltransduktion und Motilität. Insgesamt konnte durch die Arbeit gezeigt werden, dass die Acetylierung und Deacetylierung von Proteinen durch Pat1 resp. Sir2 die Regulation der Genexpression beeinflußt. Dies geschieht in H. volcanii indirekt durch die posttranslationale Veränderung von internen Signalen oder direkt durch die Modulierung des Histons und die damit verbundene Änderung der DNA-Struktur.

Introduction: The Vbeta12-transgenic mouse was previously generated to investigate the role of antigen-specific T cells in collagen-induced arthritis (CIA), an animal model for rheumatoid arthritis. This mouse expresses a transgenic collagen type II (CII)-specific T-cell receptor (TCR) beta-chain and consequently displays an increased immunity to CII and increased susceptibility to CIA. However, while the transgenic Vbeta12 chain recombines with endogenous alpha-chains, the frequency and distribution of CII-specific T cells in the Vbeta12-transgenic mouse has not been determined. The aim of the present report was to establish a system enabling identification of CII-specific T cells in the Vbeta12-transgenic mouse in order to determine to what extent the transgenic expression of the CII-specific beta-chain would skew the response towards the immunodominant galactosylated T-cell epitope and to use this system to monitor these cells throughout development of CIA. Methods: We have generated and thoroughly characterized a clonotypic antibody, which recognizes a TCR specific for the galactosylated CII(260-270) peptide in the Vbeta12-transgenic mouse. Hereby, CII-specific T cells could be quantified and followed throughout development of CIA, and their phenotype was determined by combinatorial analysis with the early activation marker CD154 (CD40L) and production of cytokines. Results: The Vbeta12-transgenic mouse expresses several related but distinct T-cell clones specific for the galactosylated CII peptide. The clonotypic antibody could specifically recognize the majority (80%) of these. Clonotypic T cells occurred at low levels in the naïve mouse, but rapidly expanded to around 4% of the CD4+ T cells, whereupon the frequency declined with developing disease. Analysis of the cytokine profile revealed an early Th1-biased response in the draining lymph nodes that would shift to also include Th17 around the onset of arthritis. Data showed that Th1 and Th17 constitute a minority among the CII-specific population, however, indicating that additional subpopulations of antigen-specific T cells regulate the development of CIA. Conclusions: The established system enables the detection and detailed phenotyping of T cells specific for the galactosylated CII peptide and constitutes a powerful tool for analysis of the importance of these cells and their effector functions throughout the different phases of arthritis.
Das Wissen um eine antike Kultur ist nicht selten vom Stand der modernen Forschung geprägt. Auf dem Gebiet der Erforschung des antiken Westbalkan hat dieser keineswegs ein Optimum erreicht. Vielmehr haben der politische Umbruch in Osteuropa sowie die Kriege seit den 1990er Jahren in den Nachfolgestaaten des ehemaligen Jugoslawien ein negatives Licht auf jene Länder und seine Menschen geworfen. Diese Bewertung wird darüber hinaus bis in die heutigen Tage paradoxerweise auch auf die Kulturen der Antike übertragen. Dabei bildet die Region an der Adria mit ihren archäologischen Denkmälern aus verschiedenen Jahrtausenden der Menschheitsgeschichte eine der ältesten Kulturlandschaften Europas. In kaum einem anderen Teil des Kontinents finden sich Natur und Kultur derart eng beieinander. Die in albanischer, serbischer, kroatischer oder slowenischer Sprache verfassten wissenschaftlichen Abhandlungen sowie die von den Touristenrouten häufig abseits gelegenen Kulturgüter sind darüber hinaus in Westeuropa zumeist unbekannt. Aus diesem Grund soll dieser Aufsatz einen Beitrag zum kulturellen „Wiederaufbau“ des am Adriatischen Meer und im Dinarischen Gebirge gelegenen antiken Illyricum leisten. Zunächst wird zur allgemeinen Orientierung der geographische Rahmen abgesteckt. Des weiteren soll die geographische sowie topographische Situation der verschiedenen Kulturlandschaften entlang der Adria sowie des Dinarischen Hinterlandes exemplarisch aufgezeigt werden. Schließlich wird versucht, die sowohl historische als auch archäologische „Lücke“ zwischen Griechenland und Italien für die vorrömische Zeit zu schließen.
We show that average excess returns during the last two years of the presidential cycle are significantly higher than during the first two years: 9.8 percent over the period 1948 – 2008. This pattern in returns cannot be explained by business-cycle variables capturing time-varying risk premia, differences in risk levels, or by consumer and investor sentiment. In this paper, we formally test the presidential election cycle (PEC) hypothesis as the alternative explanation found in the literature for explaining the presidential cycle anomaly. PEC states that incumbent parties and presidents have an incentive to manipulate the economy (via budget expansions and taxes) to remain in power. We formulate eight empirically testable propositions relating to the fiscal, monetary, tax, unexpected inflation and political implications of the PEC hypothesis. We do not find statistically significant evidence confirming the PEC hypothesis as a plausible explanation for the presidential cycle effect. The existence of the presidential cycle effect in U.S. financial markets thus remains a puzzle that cannot be easily explained by politicians employing their economic influence to remain in power. Die Möglichkeiten zum Recherchieren von botanischer Literatur über Hessen in sieben Online-Datenbanken werden vergleichend betrachtet. Unter anderem werden Testrecherchen zu Beiträgen in den Zeitschriften Hessische Floristische Briefe und Botanik und Naturschutz in Hessen ausgewertet. Es stellt sich heraus, dass im Feld der getesteten Datenbanken die Hessische Bibliographie sowie Biological Abstracts jeweils eine besondere Stellung einnehmen; mehrere weitere, überwiegend lizenzfrei nutzbare Datenbanken liefern ebenfalls substanzielle Beiträge. Die von der Universitätsbibliothek Johann Christian Senckenberg in Frankfurt am Main aufgebaute Virtuelle Fachbibliothek Biologie (vifabio) erleichtert den Zugang zu einigen wesentlichen Ressourcen; unter anderem können Hessische Bibliographie und Biological Abstracts sowie weitere Datenbanken in vifabio mit einer einzigen Suchanfrage parallel durchsucht werden.
We estimate the risk and expected returns of private equity investments based on the market prices of exchange traded funds of funds that invest in unlisted private equity funds. Our results indicate that the market expects unlisted private equity funds to earn abnormal returns of about one to two percent. We also find that the market expects listed private equity funds to earn zero to marginally negative abnormal returns net of fees. Both listed and unlisted private equity funds have market betas close to one and positive factor loadings on the Fama-French SMB factor. Private equity fund returns are positively correlated with GDP growth and negatively correlated with the credit spread. Finally, we find that market returns of exchange traded funds of funds and listed private equity funds predict changes in self-reported book values of unlisted private equity funds.
This paper analyzes the impact of blockownership dispersion on firm value. Blockholdings by multiple blockholders is a widespread phenomenon in the U.S. market. It is not clear, however, whether dispersion among blockholder is preferable to having a more concentrated ownership structure. To test for the direction of the effect, we use a large dataset of U.S. firms that combines blockholder information, shareholder rights information, debt ratings, accounting information, and financial markets information. We find that a large fraction of aggregated block ownership negatively affects Tobin’s Q. The negative impact is larger if blockowners are more dispersed, suggesting that a concentrated ownership structure is to be preferred on average. Results are robust to controlling for blockholder type as well as proxies for shareholder rights. Our empirical findings are also confirmed if we study the impact of ownership dispersion on firm debt ratings rather than Tobin’s Q. JEL Classification: G3, G32
This study examines the legal environment of netting agreements covering financial contracts. It concludes that an international instrument should be developed capable of improving the effectiveness of netting agreements in mitigating systemic risk. To this end, two different aspects of the enforceability of netting agreements are considered: (i) the general enforceability of netting, and (ii) the possibility of precluding the operation of netting a mechanism by way of a regulatory moratorium for considerations of systemic stability. The first part of the study presents the use of netting and the various forms it may take before going on to explain the benefits and drawbacks of enforceable netting agreements. Benefits for individual firms consist in lower counterparty risk and more favourable capital requirements. Benefits for the financial market as a whole flow from greater financial market stability since the contagion of systemically relevant institutions by the default or insolvency of another institution is limited, thus helping to avoid systemic effects. Additionally, the use of netting arrangements can improve overall market liquidity. A potential drawback of enforceability of netting, in certain situations, is that the operation of a netting mechanism could actually work against the purpose of systemic stability where the transfer of parts of the business of an insolvent financial institution to a solvent bridge entity would enhance or maintain value to a greater extent than the operation of a netting agreement would. Regulatory authorities are considering under which conditions a moratorium to halt the netting mechanism until the situation is solved could avoid this threat to systemic stability. The second part of the study examines whether there is the potential to support the purpose of enhanced systemic stability by way of international harmonisation of private and insolvency law. As regards the issue of general enforceability, the global picture of netting legislation is heterogeneous. Given the great practical relevance of the matter, an international instrument could be very useful. As to the issue of private law consequences of regulatory moratoria, the absence of a harmonised framework appears to lead to actual cross-border inconsistency and legal uncertainty as regards financial contracts that are governed by a foreign law. Taking these to aspects into account, this paper recommends that work on developing an international instrument be undertaken. The final part of the study suggests a set of preliminary guidelines for the development of suchan instrument. In the light of the findings of the previous sections, a mixed, two-step approach is recommended. First, a non-binding instrument could be developed, serving as a benchmark and reservoir of legal solutions in respect of the relevant issues. Secondly, isolated aspects relating to both the general enforceability of netting and the accommodation of a regulatory moratorium in foreign private and insolvency law could be dealt with in an international Convention, in particular where cross-border situations involving netting require uniformity of applicable legal rules.
Nusa Tenggara Timor, a south-eastern province of Indonesia, is populated mainly by Christians. The Alor-Pantar Archipelago has a majority of Protestant inhabitants who were baptized by Dutch Calvinists in the first half of the twentieth century. In addition, there are some coastal enclaves that have been inhabited by Muslims for centuries. In some areas, such as in the headland of Muna (Tanjung Muna) forming the northeast of Pantar Island, there is an even greater diversity of monotheistic religions, with some Catholic families living next to Protestants and Muslims. All adherers of the three religious faiths living at Tanjung Muna share core elements of the local adat, which consists of core rules relating to social behavior. It is believed that the ancestors will notice transgressions of these rules, and may use their supernatural power to punish their human descendants. In Indonesia, the term adat was first used by Muslims to distinguish the non-Islamic practices from Muslim faith (Keane 1997:260-261). This is definitely not the case in the village of Pandai at the coast of Tanjung Muna, where Islam tolerates ancestral worship. The same is true for the Catholics in the inland village of Helangdohi, who do not only tolerate but even support such customs. Some villagers from Helangdohi had become acquainted with this kind of Catholicism on the nearby island of Flores, where ancestral worship is encouraged by the missionaries of the Societas Verbi Divini (SVD). The attitude of Protestantism, at least in the Alor Archipelago, is quite the contrary of the permissive views held by Catholicism and Islam. In the 1930s the Protestant-Calvinist missionaries banned any kind of ancestral worship and destroyed most relics (Dalen 1928: Picture 1). These drastic measures demanded the disavowal of the ancestors, including the destruction of heirlooms and omitting of rituals.
Das extrem thermophile Eubakterium Thermus thermophilus ist in den letzten Jahren zu einem Modell für thermophile Organismen geworden und verdankt seinen Statuszum Teil seiner hohe Wachstumsrate, den guten Zellerträgen und der konstitutivenExpression eines natürlichen Kompetenzapparates, der seine genetische Manipulation ermöglicht. Die Verfügbarkeit von kompatiblen Plasmiden und bis zu vier thermostabilen Antibiotikaresistenzmarkern konnten den Wert des Organismus in Hinblick auf biotechnologische Anwendungen noch weiter steigern und tatsächlich besteht nach wie vor ein ungebrochenes Interesse an der Struktur- und Funktionsaufklärung thermophiler Proteine. Der Focus der hier vorliegenden Arbeit richtete sich auf eine der insgesamt zwei terminalen Oxidasen der Atmungskette von T. thermophilus, die Cytochrom ba3 Oxidase. Es wurden verschiedene rekombinante Varianten des Proteins, die sich hinsichtlich der Position und Länge des verwendeten Histidin-Tags unterschieden, kloniert, exprimiert und aufgereinigt. Das Einfügen eines internen His12-Tags in einen periplasmatischen Loop zwischen den Transmembranhelices IV und V führte zu einer rekombinanten Version der Oxidase, die in ihren Eigenschaften dem nativen Wildtyp entsprach und sich durch die in dieser Arbeit etablierte Aufreinigungsstrategie relativ schnell, in guten Ausbeuten und hoher Reinheit aufreinigen ließ. Weiterhin konnten verschiedene Punktmutationen von möglicherweise am Elektronentransfer beteiligten Aminosäureresten generiert und die resultierenden Proteine aufgereinigt und über ihre enzymatische Aktivität charakterisiert werden. Eine weiterführende Charakterisierung der Mutanten erfolgte im Rahmen einer Kooperation und ist bisher noch nicht abgeschlossen. Das Herzstück dieser Arbeit machte jedoch die Definition der Transkriptionseinheit der Cytochrom ba3 Oxidase und die sich daraus ergebenden Fragestellungen aus. So konnte gezeigt werden, dass das ba3 Operon zusätzlich zu den Strukturgenen der dort kodierten Untereinheiten noch mindestens ein, höchst wahrscheinlich jedoch zwei zusätzliche Gene enthält: cbaX und cbaY. Bioinformatische Charakterisierungen ordneten CbaY der diversen Gruppe von sekundären Transportern zu, und es konnte experimentell gezeigt werden, dass seine Anwesenheit für die Expression der Cytochrom ba3 Oxidase förderlich ist. CbaX hingegen konnte über die durchgeführte Homologiesuche keine Funktion zugeordnet werden; uncharakterisierte Homologe waren einzig in der Thermaceae Gruppe zu finden. Durch Deletions- und Komplementationsstudien konnte dem Protein eine entscheidende Rolle in der Assemblierung der ba3 Oxidase bescheinigt werden. CbaX scheint eine zentrale Aufgabe bei der Häm a Insertion in Untereinheit I zu spielen und könnte, ohne Sequenzhomologie aufzuweisen, die Rolle des Surf1-Proteins übernehmen, welches in den sequenzierten Organismen der Thermaceae Gruppe nicht konserviert ist. Die homologe Expression und Aufreinigung von CbaX führte nicht zur erwarteten Ausbeute und Reinheit des Proteins, konnte aber durch immunologische Experimente eine potentielle Interaktion von CbaX und der Cytochrom ba3 Oxidase nachweisen.
Die chronische Hepatitis C Virus-Infektion ist eine der häufigsten Ursachen für Leberzirrhose und das hepatozelluläre Karzinom. CD81 ist ein Oberflächenprotein aus der Familie der Tetraspanine, welches auf Hepatozyten und Lymphozyten exprimiert ist. CD81 interagiert mit dem Hepatitis C Virus-Hüllprotein E2. Die Interaktion ist essentiell für die Infektion von Hepatozyten durch das Hepatitis C Virus und beteiligt an der Hepatitis C Virus assoziierten Immunmodulation, welche in Zusammenhang mit der Chronifizierung und dem Auftreten von autoimmunologischen Phänomenen steht. Verschiedene Untersuchungen haben gezeigt, dass CD81 von Lymphozyten sezerniert werden kann. Die Bedeutung der CD81-Serumkonzentration wurde bislang nicht untersucht. Um neue Erkenntnisse über die Rolle von CD81 innerhalb der Pathogenese der chronischen Hepatitis C zu erlangen, wurde in der vorliegenden Arbeit die CD81-Serumkonzentration bei Patienten mit chronischer Hepatitis C untersucht. Nach Anreicherung von CD81 mittels differentieller Zentrifugation konnte durch Anwendung des Western Blot-Verfahrens gezeigt werden, dass CD81 im Serum bei Patienten mit chronischer Hepatitis C im Vergleich zu gesunden Probanden signifikant erhöht ist (p = 0,001). Im Hinblick auf die CD81-vermittelte Immunmodulation wurde anschließend die CD81-Serumkonzentration mit der Höhe der Lebertransaminase GPT als Surrogatmarker für die entzündliche Aktivität der Leber verglichen. Die Untersuchungen ergaben eine signifikante Korrelation zwischen der CD81- Serumkonzentration und der Höhe der GPT (Korrelationskoeffizient r = 0,334, p = 0,016). Im nächsten Schritt wurde der Einfluss der antiviralen Therapie auf die CD81- Serumkonzentration untersucht. Bei Patienten mit chronischer Hepatitis C und erhöhten Transaminasen, die nach Therapie ein dauerhaftes Ansprechen entwickelten, zeigte sich unter Therapie ein signifikanter Abfall der CD81-Serumkonzentration im Vergleich zu der vor Therapie (p = 0,045). Zusammenfassend zeigen diese Ergebnisse der Promotionsarbeit, dass CD81 im Serum nachweisbar ist, dass die CD81-Serumkonzentration bei Patienten mit chronischer Hepatitis C erhöht ist und dass es einen Zusammenhang zwischen Leberentzündung und Therapieansprechen gibt. Diese Ergebnisse deuten daraufhin, dass CD81 für die Pathogenese der Hepatitis C Virus-Infektion möglicherweise von Bedeutung ist.
Employing NMR spectroscopy, it is not only possible to calculate the three dimensional structures of single proteins, but also to study dynamics and conformational changes of protein-complexes. In fact that is an important aspect, since the protein function depends on dynamics and interactions with other molecules. Therefore the study of protein-protein interactions is of highest importance for a better understanding of biological processes. Based on NMR methods, in this thesis we were able to determine protein-protein interactions within the enterobacterial Rcs signalling complex which is regulated via a phosphorelay. Originally identified as regulator of capsule synthesis, the Rcs phosphorelay is now considered to be implicated in stress response caused by disturbances in the peptidoglycan layer. Beyond that the Rcs system is involved in multiplex transcriptional networks including cell division, motility, biofilm formation and virulence. Because of such global nature and its extraordinary structural organisation involving membrane integrated sensor proteins (RcsC, RcsD), coactivators (RcsF, RcsA) and a transcription factor (RcsB), the Rcs system is one of the most remarkable phosphorelays in the family of enterobacteriacaea. During the complex phosphotransfer the histidine phosphotransferase (HPt) domain of the intermediary RcsD protein mediates the phosphotransfer between RcsC and RcsB, and probably modulates the phosphorylation state of the response regulator RcsB. Therefore the present work has been focused on the interface between RcsD and RcsB in more detail. In the first part of the thesis a new domain within the RcsD protein has been identified and structurally analysed by liquid NMR spectroscopy. RcsD is an inner membrane bound hybrid sensor like-kinase composed of a periplasmic sensor domain and a cytoplasmic portion. The cytoplasmic part contains the histidine like-kinase (HK) domain and the histidine phosphotransferase (HPt) domain. By analysis of the secondary structure in more detail, it was shown here that the two domains are intermitted by an additional 13.3 kDa domain. Corresponding to the position of the ABL (α−β−loop) domain of RcsC, located C-terminal to the RcsC-HK domain, the new identified domain was named RcsD-ABL. The central structural element of RcsD-ABL is a β-sheet composed of six strands with a β1−β2−β3−β4−β6−β5 topology and surrounded by two α-helices α1 and α2. In the second part of the thesis, RcsD-ABL is identified as a binding domain for the response regulator RcsB by NMR titration experiments. Such a binding domain for a response regulator has so far only been described for the histidine kinase CheA. In reportergene assays with β-galactosidase and ONPG as substrate it was shown that overexpression of RcsD-ABL in high amounts inhibited binding of RcsB to its target promoter. The β-galactosidase activity was reduced by 80 % with respect to cells carrying no plasmid encoding RcsD-ABL. The mapping of the binding interface was successfully achieved by chemical shift perturbations, a fast mapping protocol and selective labelling. It was shown that the interaction between RcsD-ABL and RcsB takes place via a binding interface comprising mainly the two α-helices of RcsD-ABL and the α-helices α7, α8 and α10 in the effector domain of RcsB. In the third part of the thesis, the interaction of RcsB with RcsD-ABL was related to that with RcsD-HPt. Using NMR titration experiments and ITC measurements, a comparison of the binding constants (Kd) of RcsB interacting either with the isolated RcsD-ABL (2 PM) or the isolated RcsDHPt domain (40 PM) revealed a higher affinity of RcsD-ABL to RcsB. A conjugate of RcsD-ABL-HPt interacting with RcsB decreased the Kd in the one-site fitting mode to 10 PM. However, the two-site fitting mode applied for RcsD-ABL-HPt/RcsB interaction resulted in a Kd (RcsD-ABL) of 2 PM and a Kd (RcsD-HPt) of 8 PM, indicating that RcsD-ABL enhances the binding of RcsD-HPt to RcsB. In the last part of the thesis, it was partly possible together with the data obtained from NMR titration experiments, PRE measurements and a HADDOCK protocol to develop a geometrical model for the interaction of RcsD with RcsB. In this model the receiver domain of RcsB interacts with the RcsD-HPt domain and the RcsB effector domain interacts with the RcsD-ABL domain. These results lead to surprising insights on the regulation of phosphorelays, since normally the effector domain binds to DNA. Here the effector domain is recognized by the newly identified RcsD-ABL domain. Prospectively, further investigations of phosphorylation affects and mutational studies will be of great interest.
Genes coding for membrane proteins make up 25%-30% of the genome in most organisms. Membrane proteins play an important role in cell functioning and their importance is enhanced by the fact that a large number of drugs are targeted at membrane proteins. Paradoxically, experimentally determined structures of membrane protein correspond to only about 1.7% of protein structures deposited in the protein data bank (PDB). This is largely due to the fact that membrane proteins are difficult to deal with owing to their amphipathic nature. The low abundance of membrane proteins in native tissue makes heterologous overexpression of these genes a necessity. This thesis work aimed at heterologous production of several secondary active transporter proteins for structural and functional characterizations and establishing alternative strategies to overcome the obstacles associated with heterologous overproduction. Four members of the heavy metal transporting cation diffusion facilitator (CDF) family from S. typhimurium and A. aeolicus were heterologously overproduced in E. coli and functionally characterized by an in vivo complementation assay using the zinc transport deficient E. coli GG48 strain. Out of these four, Aq_2073 from A. aeolicus was produced in large scale with substantial yield and purity sufficient to carry out structural studies. After extensive stability studies with different detergents, pHs and temperatures, the protein was subjected to 3D and 2D crystallization trials. Several C- terminal truncated constructs were made and the simultaneous crystallization screenings were carried out. These resulted in initial needle like crystals in 3D crystallization trials or optimum sized vesicles with crystalline patches in 2D crystallization trials but no obvious crystal. The protein showed significant increase in melting temperature in the presence of cadmium, when tested by differential scanning calorimetry. Another transporter, STM3880 of the potassium uptake permease (KUP) family from S. typhimurium, was heterologously overproduced in E. coli, purified by affinity chromatography, reconstituted into artificial liposome and functionally characterized by solid supported membrane based electrophysiology. In order to establish alternative expression strategies, continuous exchange cell free expression (CECF) of proteins from four different families was carried out. This method found to be aptly complementing the cell-based production approach. Targets from resistance to homoserine/threonine (RhtB) family not expressing in vivo could be expressed and purified using CECF. STM1781 of the sulfate permease (SulP) family was expressed, purified and characterized for stability while the cell-based production resulted in extensive degradation. PF0780 of multidrug/oligosaccharidyllipid/polysaccharide flippase (MOP) family was also purified to homogeneity and the stability was comparable to in vivo produced protein. Moreover, the effect of maltose binding protein (MBP) fusion at N-terminus on production and membrane integration was tested with three selected targets. The analysis revealed decreased yields in the presence of MBP if the protein had both termini in the cytoplasm. This work succeed in heterologously overproducing and establishing purification protocols for several secondary active transporters aiming at structural and functional characterization in a structural genomics framework. It also showed that integration of alternative strategies, like employing both cell-based and cell-free heterologous expression systems, expands the overall expression space coverage and in turn increases the chance of success of a structural genomics styled project.
Succinate:quinone oxidoreductases (SQORs) are integral membrane protein complexes, which couple the two-electron oxidation of succinate to fumarate (succinate → fumarate + 2H+ + 2e-) to the two-electron reduction of quinone to quinol (quinone + 2H+ + 2e- → quinol) as well as catalyzing the opposite reaction, the reduction of fumarate by quinol. In mitochondria and some aerobic bacteria, succinate:ubiquinone reductase, also known as complex II of the aerobic respiratory chain or as succinate dehydrogenase from the tricarboxylic acid (TCA or Krebs) cycle, catalyzes the oxidation of succinate by ubiquinone, which is mildly exergonic under standart conditions and not directly associated with energy storage in the form of a transmembrane electrochemical proton potential (Δp). Gram-positive bacteria do not contain ubiquinone but rather menaquinone, a quinone with significantly lower oxidation-reduction (“redox”) midpoint potential. In these cases, the catalyzed oxidation of succinate by quinone is endergonic under standard conditions. Consequently, these bacteria face a thermodynamic problem in supporting the catalysis of this reaction in vivo. Based on experimental evidence obtained on whole cells and purified membranes, it had previously been proposed that the SQR from Gram-positive bacteria supports this reaction at the expense of the protonmotive force, Δp. Nonetheless, it has been argued that the observed Δp dependence is not associated specifically with the activity of SQR because the occurrence of artifacts in experiments with bacterial membranes and whole cells can not be fully excluded. Clearly, definitive insight into the mechanism of catalysis of this intriguing reaction required a corresponding functional characterization of an isolated, membranebound SQR from a Gram-positive bacterium. The first aim of the present work addresses the question if the general feasibility of the energetically uphill electron transfer from succinate to menaquinone is associated specifically to a single enzyme complex, the SQR. The prerequisite to achieve this goal was stable preparation of this enzyme.
The nicotinamide-adenine-dinucleotide (NADH):ubiquinone oxidoreductase (complex I) from the strictly aerobic yeast Y. lipolytica contains at least 26 “accessory” subunits however the significance of most of them remains unknown. The aim of this study was to characterize the role of three accessory subunits of complex I, recently identified: two mitochondrial acyl carrier proteins, ACPM1 and ACPM2 and a sulfurtransferase (st1) subunit. ACPMs are small (approx. 10 kDa) acidic proteins that are homologous to the corresponding central components of prokaryotic fatty acid synthase complexes. Genomic deletions of the two genes ACPM1 and ACPM2 resulted in strains that were not viable or retained only trace amounts of assembled mitochondrial complex I, respectively, as assessed using two-dimensional blue native/sodium dodecyl sulfate polyacrylamide gel electrophoresis (BN/SDS) PAGE. This suggested different functions for the two proteins that despite high similarity could not be complemented by the respective other homolog still expressed in the deletion strains. To test whether complex I was affected by deletion of the ACPM2 gene, its activities in mitochondrial membranes were measured. Consequently, specific inhibitor sensitive dNADH: decylubiquinone (DBQ) oxidoreductase activity was lost completely and a strong decrease in dNADH: hexa-ammine-ruthenium (HAR) oxidoreductase activity was measured. Remarkably, the same phenotypes were observed if just the conserved serine carrying the phosphopantethein moiety was exchanged with alanine. Although this suggested a functional link to the lipid metabolism of mitochondria, using HPLC chromatography no changes in the lipid composition of the organelles were found. Proteomic analysis revealed that both ACPMs were tightly bound to purified mitochondrial complex I. Western blot analysis revealed that the affinity tagged ACPM1 and ACPM2 proteins were exclusively detectable in mitochondrial membranes but not in the mitochondrial matrix as reported for other organisms. Hence it has been concluded that the ACPMs can serve all their possible functions in mitochondrial lipid metabolism and complex I assembly and stabilization as subunits bound to complex I. A protein exhibiting rhodanese (thiosulfate:cyanide sulfurtransferase) activity was found to be associated with homogenous preparation of complex I. From a rhodanese deletion strain, functional complex I that lacked the additional protein but was fully assembled and displayed no functional defects or changes in EPR signature was purified. In contrast to previous suggestions, this indicated that the sulfurtransferase associated with Y. lipolytica complex I is not required for assembly of its iron–sulfur clusters.
Functional and structural characterization of Aquifex aeolicus sulfide:quinone oxidoreductase

This work presents the first complete structure of the membrane protein sulfide:quinone oxidoreductase (SQR), obtained by X-ray crystallography. Its description is complemented by the results of biochemical and functional experiments. SQRs are ubiquitous flavoprotein disulfide reductases (FDRs), present in all domains of life, including in humans. Their physiological role extends from sulfide detoxification to sulfide-dependent respiration and photosynthesis (in archaea and bacteria), to heavy metal tolerance (in yeast) and possibly to sulfide signalling (in higher eukaryotes). Until now understanding the function of SQRs was difficult because of the poor level of sequence conservation in this enzyme family, the limited functional characterization available and the absence of any structural data. SQR was identified in the native membranes of the hyperthermophilic bacterium Aquifex aeolicus by peptide mass fingerprinting (PMF) and by a spectrophotometric activity assay. The protein was solubilized in the detergent dodecyl-beta-D-maltoside (DDM) and purified to homogeneity in a functionally active state. It binds one FAD molecule per protein monomer and FAD is its only cofactor. Its structure was determined in the “as-purified”, substrate-bound and inhibitor-bound forms at resolutions of 2.3, 2.0 and 2.9 Å, respectively. It is composed of two Rossmann-fold domains and of one membrane-attachment region. Despite the overall monomeric architecture being similar to that of FDRs, the structure reveals properties that had not been observed in FDRs until now and that have strong implications for the SQR catalytic mechanism. Surprisingly, A. aeolicus SQR is trimeric in the crystal structure and in solution, as determined by density-matched analytical ultracentrifugation, cross-linking and single particle electron microscopy. The trimer creates an appropriate surface for binding lipids and thus ensures that SQR exclusively reduces hydrophobic quinones. SQR inserts to a depth of about 12 Å into the membrane as an integral monotopic membrane protein. The interaction is mediated by an amphipathic helix-turn-helix tripodal motif and two lipid clamps. A channel in the membrane-binding domain extends towards the si-side of FAD and represents the quinone-binding site. The quinone ring is sandwiched between the conserved amino acids Phe 385 and Ile 346 and is possibly protonated upon reduction via Glu 318, Lys 382 and/or neighboring solvent molecules. Sulfide polymerization occurs on the re-side of FAD, where the highly conserved Cys 156 and Cys 347 appear to be covalently bound to the putative product of the reaction, a polysulfur chain which takes the form of an S8 ring in some monomers. Finally, the structure shows that FAD is covalently connected to the protein in an unprecedented way, via a putative disulfide bridge between the 8-methyl group of the isoalloxazine moiety and Cys 124. The high resolution insight into the protein and all unexpected structural observations presented in this work suggest that the catalytic mechanism of SQRs is significantly different from that of FDRs. In agreement with the structural and functional data, two reaction schemes are proposed for A. aeolicus SQR. They both provide a detailed description of how sulfide and quinones reach and bind the active site, how electrons are transferred from sulfide to quinone via FAD and how the elongating polysulfur product is attached to the polypeptide and is finally released. The two hypotheses differ in defining the structure of the covalent protein-FAD intermediate that forms during the reaction cycle and whose identity still remains experimentally undetermined. Remarkably, the structure of the active site and the FAD-binding mode of A. aeolicus SQR are not conserved in another SQR structure which also became available recently, that of the archaeon Acidianus ambivalens. The variability in SQRs suggests that not all of these enzymes follow the same catalytic mechanism, despite having been considered homologous. Consequently, the currently available but contradictory sequence-based classifications of the SQR family were revised. A structure-based alignment calculated on the increasing number of available sequences allowed to define new SQR groups and their characteristic sequence fingerprints in agreement with the reported structural and functional data. In conclusion, the results obtained in this work offer for the first time a detailed look into the intriguing but complicated reactions catalysed by SQRs and provide a stimulus for further genetic, biochemical and structural investigation.
Krebszellen zeichnen sich häufig durch Expression von tumorassoziierten Antigenen aus, über die grundsätzlich eine spezifische Erkennung durch das Immunsystem möglich ist. Ansätze zur Immuntherapie von Krebserkrankungen zielen darauf ab, das Potential der körpereigenen Immunabwehr zur Tumorabwehr auszunutzen. Für die Aktivierung von tumorspezifischen T-Zellen ist die Präsentation von Peptidepitopen eines Tumorantigens zusammen mit effizienter Kostimulierung durch professionelle antigenpräsentierende Zellen (APCs), insbesondere dendritische Zellen (DCs), entscheidend. In der vorliegenden Doktorarbeit wurden zur Induktion einer spezifischen Immunantwort gegen das tumorassoziierte Antigen ErbB2/HER2 neuartige zelluläre Vakzine generiert und ihre Aktivität in der Therapie bestehender ErbB2-exprimierender Tumoren analysiert. ErbB2 ist ein Mitglied der epidermalen Wachstumsfaktorrezeptorfamilie und wird von vielen humanen Tumoren epithelialen Ursprungs überexprimiert. Als Rezeptortyrosinkinase ist ErbB2 direkt an der Tumorpathogenese beteiligt und stellt eine wichtige Zielstruktur für unterschiedliche immuntherapeutische Ansätze dar. In Vorarbeiten der Arbeitsgruppe wurde ein chimäres Tumorvakzin entwickelt, das über die extrazelluläre Domäne des humanen B7-Liganden CTLA-4 die spezifische Beladung von B7-exprimierenden APCs mit einem immunogenen Abschnitt des humanen ErbB2 (HER2/neu) in vivo ermöglicht. Die Immunisierung von naiven Mäusen mit DNA-Vektoren, die für sekretierte CTLA-4-ErbB2 Fusionspoteine kodieren, induzierte in den vorangegangenen Arbeiten eine protektive Immunität gegen anschließend injizierte ErbB2-exprimierende murine Nierenkarzinomzellen (Renca-lacZ/ErbB2). Allerdings war eine therapeutische Vakzinierung mit den DNA-Vektoren gegenüber bereits bestehenden Tumoren nicht mehr wirksam. Zur Erhöhung der therapeutischen Wirksamkeit des CTLA-4-ErbB2 Tumorvakzins wurde in dieser Doktorarbeit eine alternative Behandlungsstrategie entwickelt, bei der das APC-spezifische Fusionsprotein durch ein zelluläres Vakzin in vivo zur Verfügung gestellt wird. Durch stabile Transfektion der aus BALB/c Mäusen stammenden nicht tumorigenen Mammaepithelzelllinie HC11 mit einem CTLA-4-ErbB2 kodierenden DNA-Konstrukt wurde dazu das klonale zelluläre Vakzin HC11/CTLA-4-ErbB2 abgeleitet und funktionell charakterisiert. Von diesem Zellvakzin wurde anhaltend CTLA-4-ErbB2 Fusionsprotein in das umgebende Milieu sezerniert, das in der Lage war, effizient an B7-exprimierende Zellen zu binden. Die dreimalige Immunisierung mit dem zellulären HC11/CTLA-4-ErbB2 Vakzin im wöchentlichen Intervall löste im immunkompetenten BALB/c Mausmodell eine ErbB2-spezifische Immunantwort aus. Mit Hilfe des bereits zuvor in den prophylaktischen DNA-Vakzinierungsexperimenten eingesetzten BALB/c Renca-lacZ/ErbB2 Tumortransplantationsmodells wurde zunächst untersucht, ob das zelluläre HC11/CTLA-4-ErbB2 Vakzin einen Einfluss auf das Wachstum von etablierten ErbB2-exprimierenden Tumoren hat. Die Inokulation des Vakzins in die Nähe von subkutan wachsenden Renca-lacZ/ErbB2 Tumoren führte in BALB/c Mäusen zu einer kompletten Tumorregression in der Mehrzahl der behandelten Tiere. Dabei war der antitumorale Effekt von der starken Induktion ErbB2-spezifischer Antikörper und einer mäßigen ErbB2-spezifischen Aktivität systemischer zytotoxischer T-Zellen begleitet. Durch Vakzinierung und Tumorabstoßung wurde ein Langzeitschutz induziert, der die erneute Abstoßung einer zwei Monate nach dem Primärtumor systemisch applizieren letalen Dosis von Renca-lacZ/ErbB2 Tumorzellen bewirkte. Die Vakzinierung mit HC11/CTLA-4-ErbB2 hatte keinen Einfluss auf das Wachstum ErbB2-negativer Tumorzellen. Ebenso führte die Behandlung mit HC11 Zellen, die ein irrelevantes CTLA-4 Fusionsprotein sezernieren, nicht zur Abstoßung von RencalacZ/ ErbB2 Tumoren. Diese Resultate bestätigen die antigenspezifische Wirkung des zellulären HC11/CTLA-4-ErbB2 Vakzins. Für die Evaluierung von ErbB2-spezifischen Immuntherapien stehen eine Reihe transgener Mausmodelle zur Verfügung, welche die Untersuchung des Einflusses einer bestehenden immunologischen Toleranz gegenüber humanem ErbB2 auf die therapeutische Aktivität zulassen. Ein gut etabliertes und häufig verwendetes Tiermodell sind dabei transgene WAP-Her-2 Mäuse, für die der humane ErbB2-Rezeptor ein Selbstantigen darstellt. In dieser Arbeit wurden durch Kreuzung mit BALB/c WAP-Her-2 F1 Mäuse mit einem genetischen CB6F1 Hintergrund erzeugt. Diese Tiere wiesen eine ausgeprägte immunologische Toleranz gegenüber humanem ErbB2 auf und eigneten sich somit für die Analyse der Aktivität des zellulären HC11/CTLA-4-ErbB2 Vakzins in einem physiologisch relevanten Kontext. Eine therapeutische Vakzinierung mit dem ursprünglichen HC11/CTLA-4-ErbB2 Vakzin führte in diesen WAP-Her-2 F1 Tieren nicht zu einer Abstoßung von Renca-lacZ/ErbB2 Tumoren. Zur Erhöhung der Effektivität von HC11/CTLA-4-ErbB2 in der Therapie von ErbB2-positiven Tumoren in immunologisch toleranten Mäusen wurde daher ein ähnliches zelluläres Vakzin generiert, das zusätzlich das immunmodulatorische Zytokin IL-15 sezerniert. Im Gegensatz zu HC11/CTLA-4-ErbB2 war das optimierte zelluläre HC11/CTLA-4-ErbB2/IL-15 Vakzin auch in immuntoleranten WAP-Her-2 F1 Mäusen therapeutisch wirksam und verzögerte signifikant das Wachstum von ErbB2-exprimierenden Tumoren. Die Ergebnisse dieser Arbeit zeigen, dass die andauernde Expression und Sekretion des APC-spezifischen CTLA-4-ErbB2 Fusionsproteins durch peritumoral injizierte Epithelzellen eine wirksame antigenspezifische Immunantwort und antitumorale Aktivität gegen etablierte Tumoren induzieren kann. Es ist wahrscheinlich, dass sich ähnliche Zellvakzine auch in menschlichen Krebspatienten als wirksam und nützlich erweisen können. Eine weitere Entwicklung dieses Ansatzes hin zu einer klinisch anwendbaren Immuntherapie erscheint daher sinnvoll.
Over the past few decades, changes in market conditions such as globalisation and deregulation of financial markets as well as product innovation and technical advancements have induced financial institutions to expand their business activities beyond their traditional boundaries and to engage in cross-sectoral operations. As combining different sectoral businesses offers opportunities for operational synergies and diversification benefits, financial groups comprising banks, insurance undertakings and/or investment firms, usually referred to as financial conglomerates, have rapidly emerged, providing a wide range of services and products in distinct financial sectors and oftentimes in different geographic locations. In the European Union (EU), financial conglomerates have become part of the biggest and most active financial market participants in recent years. Financial conglomerates generally pose new problems for financial authorities as they can raise new risks and exacerbate existing ones. In particular, their cross-sectoral business activities can involve prudentially substantial risks such as the risk of regulatory arbitrage and contagion risk arising from intra-group transactions. Moreover, the generally large size of financial conglomerates as well as the high complexity and interconnectedness of their corporate structures and risk exposures can entail substantial systemic risk and can therefore threaten the stability of the financial system as a whole. Until a few years ago, there was no supervisory framework in place which addressed a financial conglomerate in its entirety as a group. Instead, each group entity within a financial conglomerate was subject to the supervisory rules of its pertinent sector only. Such silo supervisory approach had the drawback of not taking account of risks which arise or aggravate at the group level. It also failed to consider how the risks from different business lines within the group interrelate with each other and affect the group as a whole. In order to address this lack of group-wide prudential supervision of financial conglomerates, the European legislator adopted the Financial Conglomerates Directive 2002/87/EC8 (‘FCD’) on 16 December 2002. The FCD was transposed into national law in the member states of the EU (‘Member States’) by 11 August 2004 for application to financial years beginning on 1 January 2005 and after. The FCD primarily aims at supplementing the existing sectoral directives to address the additional risks of concentration, contagion and complexity presented by financial conglomerates. It therefore provides for a supervisory framework which is applicable in addition to the sectoral supervision. Most importantly, the FCD has introduced additional capital requirements at the conglomerate level so as to prevent the multiple use of the same capital by different group entities. This paper seeks to examine to what extent the FCD provides for an adequate capital regulation of financial conglomerates in the EU while taking into account the underlying sectoral capital requirements and the inherent risks associated with financial conglomerates. In Part 1, the definition and the basic corporate models of financial conglomerates will be presented (I), followed by an illustration of the core motives behind the phenomenon of financial conglomeration (II) and an overview of the development of the supervision over financial conglomerates in the EU (III). Part 2 begins with a brief elaboration on the role of regulatory capital (I) and gives a general overview of the EU capital requirements applicable to banks and insurance undertakings respectively. A delineation of the commonalities and differences of the banking and the insurance capital requirements will be provided (II). It continues to further examine the need for a group-wide capital regulation of financial conglomerates and analyses the adequacy of the FCD capital requirements. In this context, the technical advice rendered by the Joint Committee on Financial Conglomerates (JCFC) as well as the currently ongoing legislative reforms at the EU level will be discussed (III). The paper finally closes with a conclusion and an outlook on remaining open issues (IV).

Dezember 2009 noch verbliebenen Einzelweisungsbefugnisse der Behörden gegenüber Privaten und gegenüber nationalen Aufsichtsbehörden sind rechtlich kaum abgesichert. (5) Wenn die hoheitlichen Befugnisse weitgehend oder vollständig beseitigt werden, bestehen Bedenken im Hinblick auf die Geeignetheit und Erforderlichkeit der Einrichtungen. (6) Die weitreichenden Unabhängigkeitsgarantien sind nicht mit den Anforderungen demokratischer Aufsicht und Kontrolle zu vereinbaren. Der Aufsatz untersucht die Frage, inwiefern die theatrale Konstruktion von christlichen Märtyrerfiguren eine Dynamik der Konversion zum bzw. des Abfalls vom Glauben voraussetzt, die in Figuralität und Ostentation zu begründen ist. Background: Vaccinia virus strain Lister Elstree (VACV) is a test virus in the DVV/RKI guidelines as representative of the stable enveloped viruses. Since the potential risk of laboratory-acquired infections with VACV persists and since the adverse effects of vaccination with VACV are described, the replacement of VACV by the modified vaccinia Ankara strain (MVA) was studied by testing the activity of different chemical biocides in three German laboratories. Methods: The inactivating properties of different chemical biocides (peracetic acid, aldehydes and alcohols) were tested in a quantitative suspension test according to the DVV/RKI guideline. All tests were performed with a protein load of 10% fetal calf serum with both viruses in parallel using different concentrations and contact times. Residual virus was determined by endpoint dilution method. Results: The chemical biocides exhibited similar virucidal activity against VACV and MVA. In three cases intra-laboratory differences were determined between VACV and MVA - 40% (v/v) ethanol and 30% (v/v) isopropanol are more active against MVA, whereas MVA seems more stable than VACV when testing with 0.05% glutardialdehyde. Test accuracy across the three participating laboratories was high. Remarkably inter-laboratory differences in the reduction factor were only observed in two cases. Conclusions: Our data provide valuable information for the replacement of VACV by MVA for testing chemical biocides and disinfectants. Because MVA does not replicate in humans this would eliminate the potential risk of inadvertent inoculation with vaccinia virus and disease in non-vaccinated laboratory workers.

V němčině se můţeme setkat s různými druhy výpůjček z angličtiny. Nejčastějším druhem výpůjčky je přímá výpůjčka, která bývá převzata do němčiny beze změny významu a pravopisu (vyjma velkého písmene u podstatných jmen). Anglická slovesa, která mají v němčině v infinitivu koncovku -en a časují se, jsou zpravidla rovněţ začleņována mezi přímé výpůjčky. Zajímavé jsou pseudoanglicismy, které často způsobují u rodilých mluvčích anglického jazyka nedorozumění. U výpůjček, které nejsou sloţeny z anglických morfémů, se někdy pochybuje o jejich správném zařazení mezi výpůjčky. Tyto výrazy jsou však rovněţ součástí vlivu anglického jazyka na německý.
Background: Leishmaniasis is a chronic disease that is found in various countries of the world. The aim of the current study was to investigate the impact of leishmaniasis on the world's research output. The present study assessed benchmarking of research output for the period between 1957 and 2006. Using large database analyses, research in the field of leishmaniasis was evaluated. Furthermore, cooperation between different countries was identified.
Results: The number of publications increased with time. Most publications came from Western countries such as the US, UK or Germany. Interestingly, countries like Brazil and India had a high research output. We found a substantial amount of cooperation between countries.
Conclusion: Although leishmaniasis is of limited geographic distribution it attracts a wide research interest. The central hub of research cooperation is the USA.
Die vorliegende Arbeit hat die Charakterisierung und Untersuchung des Stabilitätsverhaltens von
Parvulustat (PL), einem Homologen des α-Amylase-Inhibitors Tendamistat, zum Inhalt. Zur
weitreichenden Charakterisierung wurden verschiedene Proteinregionen des Parvulustats der CTerminus,
das hydrophobe Cluster, die Disulfidbrücken sowie die Proline auf ihren jeweiligen
Einfluss auf die Struktur und die thermodynamische Stabilität untersucht. In der vorliegenden
Zusammenfassung werden die Ergebnisse dieser Studien komprimiert präsentiert:
· Charakterisierung des Parvulustat-Wildtyps
Es galt vorweg herauszufinden, wie sich der Inhibitor unter nativen und denaturierten
Bedingungen verhält, um Rückschlüsse auf seine Merkmale und Strukturen zu ziehen. Die
erzielten Ausbeuten und die hohe Reinheit des isolierten Parvulustats erlaubten eine umfassende
Charakterisierung, einschließlich zahlreicher Kristallisationsexperimente, die vermuten lassen,
dass eine Kristallisation möglich sein sollte. Aufgrund der Tatsache, dass die Struktur des
Parvulustats bis 2009 unbekannt war, wurde die sekundäre Struktur mittels Circulardichroismus
und Fluoreszenzspektroskopie untersucht. Die Analyse des fernen UV-CD-Spektrums bei pH 7,0
und 25°C offenbarte eine „all-b-sheet“ Protein-Struktur. Mittels Fluoreszenzspektroskopie wurde
deutlich, dass die aromatischen Aminosäuren exponiert vorliegen. Um zunächst einen Einblick in
die Strukturveränderungen und die thermodynamische Stabilität zu erhalten, wurde der
temperaturinduzierte Entfaltungsübergang mittels CD-Spektroskopie verfolgt. Das Angleichen der
bei 230 nm gemessenen CD-Daten nach der linearen Extrapolations-Methode für eine
Zweizustands-Faltung ergab einen Tm-Wert von 82°C und D H(Tm) von 201,6 kJ/mol. Die
beträchtlichen Werte veranschaulichen die hohe Stabilität des Parvulustats. Eine aus den CDMessungen
bei 50° ergebende Übergangskurve zeigte, dass sich die Sekundärstruktur mit einem
Übergangsmittelpunkt bei 5,62 M GdnHCl kooperativ und reversibel entfaltet. Das Protein
entfaltet sogar infolge einer pH-Wert-Senkung bis auf pH 1 nicht vollständig, sondern es wechselt
direkt in einen Säure-Zustand („acid-state“). Dieser Zustand zeigt spektroskopisch die gleichen
Eigenschaften wie das native Protein, wobei die volle Inhibierungs-Aktivität nicht erhalten bleibt.
In sehr basischem Milieu bei pH 14 nimmt Parvulustat einen alkalisch denaturierten
Zwischenzustand IB an, der sich erheblich von dem GdnHCl-denaturierten, dem säurebehandelten
oder vom „molten globule“ Zustand unterscheidet. Allgemein behielt Parvulustat
über ein breites pH-Wert Spektrum (1,0-10,0) die native Struktur, bzw. eine „native like“ Struktur,
was erneut auf die enorme Stabilität des Proteins hindeutet. Die von Rehm et al. (Rehm et al.,

2009) aufgeworfene Hypothese des „induced fit“ Inhibierungsmechanismus des Parvulustats
konnte durch die in dieser Arbeit durchgeführten Experimenten bekräftigt werden. Mittels
Sekundärstrukturbestimmungen des Parvulustats unter Komplexbildung mit der a-Amylase
konnte eindeutig gezeigt werden, dass strukturelle Veränderungen am Inhibitor im Komplex
vorliegen. Durch zahlreiche Tests konnte festgestellt werden, dass die WRY-Region des
Parvulustats sich der Struktur der a-Amylase anpasst. Die Komplexierung des Parvulustats
bewirkte aber eine thermodynamische Destabilisierung der Inhibitor-Struktur.
· Einfluss des C-Terminus auf die Stabilität des Parvulustats
Um die Ursachen für die hohe Stabilität des Parvulustats auch im Vergleich zu Tendamistat (Tm:
79°C) zu finden, wurde der Einfluss des hoch flexiblen C-Terminus untersucht. Die Derivate mit
um zwei (PL-2AA), vier (PL-4AA) und sieben (PL-7AA) Aminosäuren verkürztem C-Terminus
wurden isoliert und analysiert. Die Entfaltungstemperaturen der verkürzten Derivate des
Parvulustats sinken mit abnehmender Zahl der Aminosäuren. Die Ergebnisse suggerieren, dass
der C-Terminus des Parvulustats eine entscheidende Rolle in der strukturellen Vollständigkeit des
Proteins während der thermischen Entfaltung spielt und damit auch in der Faltung (Tab.: 2.1). Der
Vergleich der Inhibitoraktivitäten der verkürzten Proteine mit dem nativen Parvulustat ergibt für
die Varianten PL-2AA und PL-4AA eine dem Wildtyp ähnliche Aktivität. Die Variante PL-7AA
weist eine leicht verringerte Aktivität auf.

Einfluss hydrophober Oberflächenclustern auf Stabilität und Faltung
des Parvulustats
Parvulustat besitzt in der Mitte des ersten b-Faltblatts, um die Position 22 liegend, einen
hydrophoben Oberflächencluster. Wie mit Hilfe von Substitutionsexperimenten in dieser Region
gezeigt werden konnte, ist die thermodynamische Stabilität des Parvulustats in hohem Maße von
der Bildung dieses kleinen aber wichtigen hydrophoben Kerns bestimmt. Demnach muss das b-
Faltblatt I und das b-Hairpin I eine entscheidende Rolle in der Faltung von Parvulustat spielen.
Position 22 ist in zweierlei Hinsicht für die Stabilität des Parvulustats wichtig: einerseits durch die
energetisch wichtigen Wasserstoffbrückenbindungen und zum zweiten steuert die Stelle zur
Formation des hydrophoben Kerns bei. Die Daten suggerieren, dass es auch eine
thermodynamische Kopplung zwischen dem hydrophoben Effekt und der Präsenz von
Wasserstoffbrückenbindungen geben könnte. Zumindest aus der Sicht der Stabilität ist es
eindeutig, dass interatomare Interaktionen wie Wasserstoffbrückenbindungen, van-der-Waalsund
Dipol-Dipol-Wechselwirkungen notwendig sind, um eine stabile Bildung von b-Faltblättern
in Parvulustat und seinen Derivaten zu verwirklichen.
· Der Effekt der Proline auf die Stabilität des Parvulustats
Der Effekt der Substitution des Prolins durch Alanin an verschiedenen Positionen des Parvulustats
wurde ebenfalls untersucht. Im Ganzen betrachtet führt auch die Mehrfachsubstitution von Prolin
zu keiner nennenswerten strukturellen Veränderung im Parvulustat. Die durchgeführten
thermischen Entfaltungsexperimente bestätigen diese Beobachtungen. Alle Einzelmutanten (P5A,
P42A, P48A, P72A) zeigten eine höhere thermische Stabilität als der Wildtyp (Abb. 2.1).

Die fast gleich bleibenden Tm-Werte bzw. die Erhöhung der Stabilität des Parvulustats sind durch
die strukturelle Fluktuationen zu erklären, denn die rigide XAS-Pro-Bindung wurde durch die
flexible XAS-Ala-Bindung ersetzt.
· Der Einfluss der Disulfidbindung auf die Stabilität des Parvulustats
Der Einfluss der zwei Disulfidbrücken des Parvulustats wurde bezüglich Aktivität, spektraler
Eigenschaften sowie Stabilität untersucht. Hierfür wurden 25 Inhibitorvarianten mittels gezielter
Mutagenese gewonnen, in denen jeweils zwei Cysteinreste, die im natürlich vorkommenden
Parvulustat Disulfidbrücken bilden, durch andere Aminosäuren ersetzt wurden. Die Ergebnisse
zeigten, dass die Faltung Parvulustats ein zwei Zustand Verhalten besitzt, das außer in
Tendamistat in keinem anderen disulfid-verknüpftem Protein gefunden wurde. Dieses Verhalten
wurde durch die Entfernung von Disulfidbrücken nicht beeinflusst. Wie durch die CD- und
fluoreszenzspektroskopischen Experimente belegt werden konnte, ist die native Struktur des
Parvulustats durch die Entfernung der C43-C70-Disulfidbindung tiefgreifend verändert worden.
Passend zu den Veränderungen der Struktur haben die Mutationen schwerwiegende
Destabilisierungseffekte auf das Protein verursacht, was auch an der Erniedrigung der Freien
Gibbs Energie der Denaturierung und der Tm-Werte zu erkennen ist. Die Untersuchung des
Einflusses der Aminosäure-Substitution in den Positionen 43 und 70 auf die thermodynamische
Stabilität des Parvulustats führt zum Ergebnis, dass die Hydrophobizität und Polarität des Restes
70 einen bedeutenden Effekt auf die Stabilität des Proteins besitzt. Die Betrachtung der
thermodynamischen Daten macht deutlich, dass der Beitrag der freien Energie zur Stabilisierung
nicht nur abhängig von der eingeführten Aminosäure ist, sondern zum Teil auch von dem
strukturellen Kontext abhängt. Die Daten zeigen, dass die Substitution des Alanins an der Stelle
70 durch Leucin oder Threonin die Struktur um 0,3 bzw. 1,7 kJ/mol stabilisieren. Zusätzliche
Beiträge zum Unterschied bezüglich des ΔG°-Werts zwischen den Varianten C43AC70L/T und
C43L/TC70A können sich auch durch die unterschiedliche lokale Umgebung, wie z.B. die
Seitenketten von benachbarten Aminosäuren um die zwei Mutationsstellen ergeben. Der Tm-Wert
des Wildtyp-Proteins beträgt 82°C. Die Vergleiche dieser Größe mit der von der stabilsten
Cystein-defizitären Doppelmutante C43AC70T (47,3°C) ergibt eine Differenz von 34,7°C. Dieses
Ergebnis untermauert, dass die Disulfidbindung 2 eine extrem wichtige strukturelle Komponente
in der ungewöhnlich hohen Stabilität des Parvulustats darstellt.
Das Ziel der Arbeit war es dennoch die Daten der Stabilitäten einzelner Disulfidmutanten zu
sammeln und zu erfassen, um dadurch allgemeine Grundregeln für eine rationale Gestaltung der
Elimination beider Disulfidbrücken im Sinne einer vorhersagbaren Auswirkung auf die
Proteinstabilität zu bekommen.

Der Versuch ein disulfidfreies Protein in großen Mengen aus S. lividans zu isolieren, stellte sich
aber als extrem schwierig dar. Nach der Änderung des Stamms des Wirtsorganismus
(Streptomyces lividans TK23), Erhöhung der Qualität der Protoplasten und der Expressions-
Bedingungen (19°C, 150 rpm) konnten auf dem SDS-Gel stärkere Banden des Derivats
C9AC25TC43AC70T-4AA beobachtet werden. Die Expression der Mutante
C9AC25TC43AC70L-4AA konnte hingegen nicht nachgewiesen werden. Die anschließende
Reinigung des Derivats C9AC25TC43AC70T-4AA des Parvulustats mittels Gelfiltration und RPHPLC
bzw. Isolierung des Proteins aus dem SDS-Gel brachte das erwünschte Produkt. Dieses
konnte mit der MALDI-Massenspektroskopie eindeutig nachgewiesen werden. Das aktive
Cystein-freie Derivat C9AC25TC43AC70T-4AA konnte auch auf dem a-Amylase Plattentest
nachgewiesen werden. Diese Ergebnisse zeigen, dass eine Expression und der Nachweis einer
cystein-freien Variante möglich sind, dennoch haben die Ausbeuten dieses Proteins für
weiterreichende Analytik nicht ausgereicht. Demnach sind die Disulfidbrücken für die Stabilität
und Struktur des Parvulustats von enormer Bedeutung dennoch sind sie nicht zwingend
erforderlich für die Aktivität und die Expression in S. lividans.
The purpose of the present paper is to explain the fake projective plane constructed by J. H. Keum from the point of view of arithmetic ball quotients. Beside the ball quotient associated with the fake projective plane, we also analize two further naturally related ball quotients whose minimal desingularizations lead to two elliptic surfaces, one already considered by J. H. Keum as well as the one constructed by M. N. Ishida in terms of p-adic uniformization.

The paper sketches out the framework of a transcultural model of language learning and teaching. In doing so it illuminates linguistic, psycholinguistic, hermeneutical und didactic aspects of the complex field of language learning rather than limiting itself to discussing mere methodological phenomena. The paper argues that the language learning and teaching profession can only advance by taking transcultural concepts of language acquisition, of linguistic systems, of language processing and of media use into account and by integrating them into a coherent system of language didactics.
To reach even language users not acquainted to the use of grammars the Institut für Deutsche Sprache in Mannheim (Germany) looked for new way to handle grammatical problems. Instead of confronting users with abstractions frequent difficulties of German grammar are introduced in form of exemplary questions like „Which form should be used or preferred: Anfang dieses Jahre or Anfang diesen Jahres?” Looking through the long list of such questions even laymen may find solutions of grammatical problems they might not be able to formulate as such.
The interest of this work devotes itself to the repeating linguistic actions of the students in the DaF conversation lessons. Repetitions in the lesson discourse are functionally different than repetitions in the daily discourse. The support of repetitions by the students in the class discourse is tried to be demonstrated here on the basis of examples. Recordings from the DaF conversation lessons were transcribed and reconstructed according to Hiat. The kinds of the repetitions and their functions in these DaF conversation lessons are limited with this study. The findings of the study should be concerned consciously in order to accomplish a better understanding and reacting to these repeating actions of the students like inquiry, correction, confirmation, precautionary self-control, verification and confirmation in the conversation lessons –most of which are accomplished by the students for a certain aim however unconsciously.
Integration and social advancement in our time without a solid language skills are no longer possible. What has not been done for decades, they now try through the integration abroad and in Germany make up very successful. But German is unfortunately only the first, though perhaps the most important step for a successful integration. The next question should now be: Lack of integration in spite of good knowledge of German - why?
The article addresses the growing importance of corpus-based research in the field of German foreign language acquisition. German corpora in general and learner corpora in particular are briefly introduced. A short overview of existing German learner corpora is followed by a detailed description of the error-annotated learner corpus Falko, a learner corpus of advanced learner German, which is accessible via internet (without any prior registration) and free of charge. Finally, a short example analysis demonstrates some of the functionalities of Falko. The aim of the article is to encourage researchers to employ corpora as helpful tools in their own work.
Even if translation has a long tradition within the conveyance of foreign languages, there has been a vehement discussion on its role since the 1970s – at least with respect to some languages, such as English. In the context of German as a foreign language this topic has been discussed only to some extent. With this in mind, the following article aims to examine the role of translation in the field of the German as a foreign language with specific focus on the advantages and limitations associated with its conveyance and the resultant consequences.
In terms of their functions and issues, the use of selection posters is possible in language teaching. Therefore, the present study aims to investigate the didactic potential of selection posters in German language teaching. Because of this reason, with this study, it is tried to show that the selection posters can be dealt with as materials in the courses in German Language teaching, which can be used parallel to the needs and interests. Accordingly, the alternative ways or approaches are tried to be made concrete throughout the courses. Consequently, the selection posters constitutes a wide range in German language teaching in terms of local culture, vocabulary knowledge, the processes of linguistic studies, visualization, authenticity, actuality, and spoken and written studies.
Speakers of Russian from the former Soviet Union and speakers of Turkish form the two biggest groups of immigrants in Germany. There is a number of surveys, that focus on early second language acquisition of kindergarten and primary school children in these ethnic groups. In this article, I will discuss differences and similarities of the second language acquisition process, that Russian and Turkish speaking children go through. I will compare not only the interlingual development (pronunciation, lexicon, syntax and morphology) but also the sociocultural context. For this purpose the data of my case studies will be contrasted with the other research results.
After giving an overview of the implementation of Business German in the curricula of German Departments outside of Germany and showing which place Business German has taken within these departments today, this article focuses on the teaching goals and contents as well as on the competences that ought to be achieved by the students in the German Department at Istanbul University in order to explain which chances and opportunities this study field opens up to students of German language and literature.
Gene therapy is a promising therapeutic strategy that emerged from the attractive idea of targeting therapy at the molecular level. For many patients who suffer from genetic and acquired diseases that cannot be effectively treated by conventional treatment approaches gene therapy remains a huge hope of cure in spite of the hurdles regarding efficacy and safety that need to be overcome. The development of efficient gene transfer vehicles, mainly retroviral vectors, led to the first successful gene therapy trial, to treat patients suffering from X-linked severe combined immunodeficiency syndrome (X-SCID) using gene modified stem cells (Hacein-Bey-Abina, Le Deist et al. 2002). Despite the success of this trial, it revealed the danger of retroviral insertional mutagenesis as a major adverse event of gene therapy using gene-modified stem cells (Hacein-Bey-Abina, von Kalle et al. 2003). In contrast to stem cells, T cells are relatively resistant to insertional mutagenesis and transformation even after transduction with potent oncogenes using retroviral vectors (Newrzela, Cornils et al. 2008). However, mature T cells can self-renew, proliferate and survive for long periods. These criteria are supposed to render T cells prone to transformation. Therefore, the questions of mature T cells transformability and the control mechanism limiting their transformation are still elusive.
Drug toxicity and viral resistance limit long-term efficacy of antiviral drug treatment for HIV
infection. Thus, alternative therapies need to be explored. Previously, group of “Prof. von Laer”
tested the infusion of T lymphocytes transduced with a retroviral vector (M87o) that expresses an
HIV entry inhibitory peptide (maC46). Gene-modified autologous T cells were infused into 10
HIV-infected patients with advanced disease and multidrug resistant virus during antiretroviral
combination therapy. T cell infusions were tolerated well with no severe side effects. A
significant increase of CD4 counts was observed post infusion. At the end of the one-year
follow-up, the CD4 counts of all patients were still around or above baseline. Gene-modified
cells could be detected in peripheral blood, lymph nodes and bone marrow throughout the oneyear
follow-up, whereby marking levels correlated with the cell dose. No significant changes of
viral load were observed during the first four months. Four of the seven patients that changed
their antiviral drug regimen thereafter responded with a significant decline in plasma viral load.
In conclusion, the transfer of gene-modified cells was safe, led to sustained levels of gene
marking and may improve immune competence in HIV-infected patients with advanced disease
and multidrug resistant virus. However, the low level of gene marking and the lack of substantial
long-term in vivo accumulation of gene-protected cells observed in this trial clearly demonstrate
the requirement for new vectors with new strategy.
In this thesis self‐inactivating lentiviral vectors harboring internal promoters and RNA elements
were therefore evaluated for their potential use in a clinical gene‐therapy trial. The results from
this work provide the basis for the selection of a suitable candidate vector for extensive
preclinical testing. Apart from being capable of transducing non‐dividing cells, lentiviral vectors
incorporate a number of additional features that are of potential value for gene therapeutic
applications. These include a larger packaging capacity, higher titers than γ‐retroviral vectors
and, most importantly, a reduced risk of deregulating cellular genes due to its natural integration

In order to find a suitable candidate for preclinical studies, two candidate therapeutic vectors
harboring different regulatory elements were selected based on results from pilot experiments.
The internal promoters used to drive expression of codon optimized maC46 were the PGK
promoter and MPSV promoter. This work focuses on the transgene expression levels in
lymphoid cells and antiviral activity. The issues of long term expression, propensity to
methylation mediated silencing of the promoters, and genotoxicity were also touched. In a first
step the performance of different vectors was evaluated in the human T cell lines. Based on
promising data from ex vivo human peripheral blood mononuclear cells, the vector carrying the
MPSV promoter along with intron were selected for in vivo transplantation experiments.
In summary, the ex vivo data suggested the long term survival of lentiviral gene modified cells,
along with maintained expression of introduced genes. It was observed that the expression of
these constructs depends strongly on the activation and differentiation status of the targeted T
cells. This regulation was not linked to any specific promotor. In vivo study shows that maC46
can be introduced into murine multiple hematopoietic lineages via lentiviral vector and expressed
at high levels in their mulilineage progeny, without altering the hematopoiesis. There was no
sign of any kind of hematopoietic or lymphoid malignancies. Although gene-modified
lymphocytes persisted in-vivo, the downregulation of transgene expression was consistent with
the ex-vivo observation. In contrast to that the T cells transplanted group showed delayed
engraftment of donor cells and there was no expression of C46 in blood and lymphatic organs. .
In conclusion, when considering HIV gene therapy focusing CD4+ T cells, potential problems of
T cell activation status as related to the desired clinical effect must be addressed. These results
might open the way for a gene therapy targeting mainly or exclusively activated T cells and
could be exploited for immunostimulatory as well as suppressive approaches.
