The present work deals with the integration of variable renewable energy sources, wind and solar energy into the European and US power grid. In contrast to other networks, such as the gas supply mains, the electricity network is practically not able to store energy. Generation and consumption therefore always have tobe balanced. Currently, the load curve is viewed as a rigid boundary condition, which must be followed by the generation system. The basic idea of the approach followed here is that weather-dependent generation causes a shift of focus of the electricity supply. At high shares of wind and solar generation, the role of the rigid boundary condition falls to the residual load, that is, the remaining load after subtraction of renewable generation. The goal is to include the weather dependence as well as the load curve in the design of the future electricity supply.
After a brief introduction, the present work first turns to the underlying weather-, generation and load data, which form the starting point of the analysis. In addition, some basic concepts of energy economics are discussed, which are needed in the following.
In the main part of the thesis, several algorithms are developed to determine the load flow in a network with a high share of wind and solar energy and to determine the backup supply needed at the same time. Minimization of the energy needed from controllable power plants, the capacity variable power plants, and the capacity of storing serve as guiding principles. In addition, the optimization problem of grid extensions is considered. It is shown that it can be formulated as a convex optimization problem. It turns out that with an optimized, international transmission network which is about four times the currently available transmission capacity, much of the potential savings in backup energy (about 40%) in Europe can be reached. In contrast, a twelvefold increase the transmission capacity would be necessary for a complete implementation of all possible savings in dispatchable power plants.
The reduction of the dispatchable generation capacity and storage capacity, however, presents a greater challenge. Due to correlations in the generation of time series of individual countries, it may be reduced only with difficulty, and by only about 30%.
In the following, the influence of the relative share of wind and solar energy is illuminated and examined the interplay with the line capacitance. A stronger transmission network tends to lead to a higher proportion of wind energy being better integrated. With increasing line capacity, the optimal mix in Europe therefore shifts from about 70% to 80% wind. Similar analyses are carried out for the US with comparable results.
In addition, the cost of the overall system can be reduced. It is interesting at this point that the advantages for the network integration may outweigh higher production costs of individual technologies, so that it is more favourable from the viewpoint of the entire system to use the more expensive technologies.
Finally, attention is given to the flexibility of the dispatchable power plants. Starting from a Fourier-like decomposition of the load curve as it was a few years ago, when hardly renewable generation capacity was present, capacities of different flexibility classes of dispatchable power plant are calculated. For this purpose, it is assumed that the power plant park is able to follow the load curve without significant surplusses or deficits. From this examination, it is derived what capacity must at least be available without having to resort to a detailed database of existing power plants.
Assuming a strong European cooperation, with a stronger international transmission network, the dispatchable power capacity can be significantly reduced while maintaining security of supply and generating relatively small surplusses in dispatchable power plants.
Grundproblematik und Zielstellung: Masern- und Mumpsinfektionen sind als typische Kinderkrankheiten bekannt, die in der Regel komplikationslos verlaufen und eine lebenslange Immunität hinterlassen. Es gibt jedoch, vor allem bei der Erkrankung im Erwachsenenalter oder von kleinen Kindern und immungeschwächten Personen, auch schwerwiegendere Verläufe. Diese bringen häufiger Komplikationen (Masernpneumonie mit einer Häufigkeit von 1:1000, sowie das Enzephalitisrisiko) mit sich, können bleibende Schäden verursachen oder führen selten sogar zum Tod. Aus diesem Grund ist seit 1973 eine Impfung gegen Masern sowie seit 1976 eine Impfung gegen Mumps (in Kombination mit Röteln als MMR seit 1980) öffentlich empfohlen. Diese Ausarbeitung soll nun den Verlauf der Immunitätslage der Bevölkerung in den Jahren 2001 bis 2005 darlegen und das Vorhandensein von etwaigen Immunitätslücken bezüglich dieser Krankheiten untersuchen. Methodik: Zur Bestimmung der Immunität wurden Serumproben ausgewertet, die am Institut für Medizinische Virologie des Universitätsklinikum Frankfurt am Main vom 1. Januar 2001 bis zum 31. Dezember 2005 auf Antikörper gegen Masern (n = 3.405) und Mumps (n = 2.875) untersucht wurden. Bei der Bestimmung der Seroprävalenzen wurde nach Altersgruppe und Geschlecht differenziert. Ergebnisse: Sowohl bei Masern als auch bei Mumps sind bei tendenziell abnehmender Immunität zum Teil erhebliche Immunitätslücken zu finden. Die masernspezifischen Antikörper stiegen zunächst bis zum Jahr 2003 auf 95,7 % Seropositive an, fielen jedoch bis 2005 stetig wieder auf 75,2 % ab. Die immunitätanzeigenden Antikörper gegen Mumps erreichten in keinem der Jahre Werte über 60 %. Im Bezug auf das Geschlecht wiesen in der Regel die Frauen höhere Immunitätsraten auf als die Männer. Im Vergleich der Altersgruppen zeichneten sich bei beiden Krankheiten deutliche Immunitätslücken im Kindesalter sowie bei den Jugendlichen und im jungen Erwachsenenalter ab. Bei den Masern waren nahezu kontinuierlich über den gesamten Untersuchungszeitraum Immunitätslücken bei den Altersgruppen der 1 bis 4 Jährigen und der 15 bis 19 Jährigen erkennbar, die zwischen 20 und 50 % lagen und von 2001 bis 2005 zunahmen. Die Immunität im Erwachsenenalter war grundsätzlich höher als im Kindesalter, zeigte jedoch ebenfalls über die Jahre fallende Tendenzen und nahm 2005 durchschnittliche Werte von knapp 90 % an. Ab dem 60. Lebensjahr sank der Anteil der Seropositiven jedoch wieder auf im Mittel 75 % ab. Bei Mumps lagen die Immunitätsraten im gesamten Kindesalter kaum höher als 70 %, 2005 sogar unter 50 %. Im jungen Erwachsenenalter stieg die Anzahl der Seropositiven dann zögerlich von Werten um die 60 % bei den 20 bis 29 Jährigen auf 80 bis 90 % bei den 40 bis 59 Jährigen an. Ab dem 60. Lebensjahr nahm der Anteil der Seronegativen ähnlich wie bei den Masern zu. Folgerungen: Die vorliegende Arbeit zeigt, dass weiterhin Immunitäts- und Impflücken bei Masern und Mumps bestehen und sich diese sogar bis zum Jahr 2005 in ihrer Größe ausgedehnt haben. Vielmehr als zuvor weisen nun auch höhere Altersgruppen größere Immunitätsdefizite auf. Folge hiervon sind die steigenden Erkrankungszahlen im Erwachsenenalter und die dadurch bedingte Erhöhung der Komplikationsraten im Krankheitsfall. Es gilt also, auch in Anbetracht des von der WHO gesetzten Ziels der Maserneradikation, verstärktes Augenmerk auf den Erhalt und den Ausbau der Populationsimmunität bei Masern und Mumps zu legen. Um weitere Ausbrüche dieser Krankheiten zu verhindern, muss das Impfverhalten in der Bevölkerung dringend verbessert werden. Ziel ist es hierbei, Impfraten von mindestens 95 % für die erste Applikation zu erreichen.
In this dissertation, we look at environmental effects in extreme and intermediate mass ratio inspirals into massive black holes. In these systems, stellar mass compact objects orbit massive black holes and lose orbital energy due to gravitational wave emission and other dissipative forces. We explore environmental interactions with dark matter spikes, stellar distributions, accretion disks, and combine and compare them. We discuss the existence and properties of dark matter spikes in the presence of these environmental effects. The signatures of the environmental effects, such as the phase space flow, dephasing, deshifting of the periapse, and alignment with accretion disks, are examined. These signatures are quantified in isolated spike systems, in dry, and in wet inspirals. We generally find dark matter effects to be subdominant to the other environmental effects, but their impact on the waveform is still observable and identifiable. Lastly, the rates of inspirals and the impact of spikes are estimated. All of these results are obtained with the help of a code imripy that is published alongside. If dark matter spikes exist, they should be observable with space-based gravitational wave observatories.
Die vorliegende Arbeit untersuchte die Chancen und Restriktivitäten einer künstlerischen Ausbildung. Welche Möglichkeiten stehen einer künstlerischen Ausbildung zur Verfügung und welchen Grenzen steht sie unüberwindlich gegenüber? Diese waren konstitutionstheoretisch zu entfalten und wissenschaftspraktisch anhand von mir erhobenem Datenmaterial (vornehmlich Interviews mit Studenten und Professoren von Kunsthochschulen in Deutschland) zu belegen bzw. zu verdichten...
Genauso wie die zentralen materiellen Güter unserer Gesellschaft sozial ungleich verteilt sind, ist auf einer zeitlichen Ebene die Zeit sozial ungleich verteilt. Kann die Zeit als eine zentrale Ungleichheitsdimension bezeichnet werden? Um dieser Antwort auf die Spur zu kommen, wird ein besonderer Untersuchungsblickwinkel gewählt, der an eine bestehende Debatte in der Ungleichheitsforschung anschließt. An die Entstrukturierungsthese anknüpfend, die das Verschwinden vertikaler Strukturen beschreibt, wird davon ausgegangen, dass soziale Schichten als klassisches Konzept der Soziologie noch immer eine wichtige Bedeutung im gesellschaftlichen Leben aufweisen. In einer Sekundäranalyse mit einem Datensatz der amtlichen Statistik (Zeitbudgeterhebung 2001/02) wird deshalb die ungleiche Verteilung auf der Ebene der Alltagszeit mit einem Schichtungsansatz verbunden. Die Zeit äußert sich im Lebensalltag der Menschen vor allem als abstrakte Zeit, sie ist die in homogene Einheiten eingeteilte Zeit. Die zentrale Hypothese dieser Arbeit lautet: Die Zeiten der alltäglichen Routine - Schlafenszeit, Erwerbsarbeitszeit, Hausarbeit, Freizeit und Wegezeit - sind ungleich in verschiedenen sozialen Schichten verteilt. Neben Zeitlängen werden auch die Uhrzeiten, wann etwas getan wird, und der Ort, wo etwas getan wird, theoretisch und empirisch in die Analyse integriert. Geschlecht und Alter ergänzen als horizontale Dimensionen die Forschungsperspektive. Weil Frauen und Männer keine homogene Gesamtheit darstellen, werden schichtspezifische Differenzierungen innerhalb der Geschlechter bei der (un)gleichen Verteilung der Zeiten der alltäglichen Routine untersucht. Die mittleren sozialen Schichten weisen in der "Durée der Alltagserfahrung" oft Konvergenzen auf, während vor allem die Unterschicht und Oberschicht kontrastreich in vielen Aspekten der Zeiten im Alltag sind. Am Wochenende haben die Akteure der "Unterschicht und unteren Mittelschicht" die längsten Erwerbsarbeitszeiten, die Akteure der "Oberschicht und oberen Mittelschicht" die geringsten. Weil ihre Arbeitszeit in einer Zeitinstitution liegt, die für Regeneration steht, sind die Akteure der "Unterschicht und unteren Mittelschicht" bei der bezahlten Arbeitszeit in diesem Aspekt benachteiligt. Für Frauen gilt: Je niedriger die soziale Schichtzugehörigkeit, desto kürzer sind die bezahlten Erwerbsarbeitszeiten an einem Werktag. Die schichtspezifische Regionalisierung der bezahlten Arbeitszeit macht die soziale Ungleichheit deutlich: Die Arbeitszeiten sind für soziale Schichten in Zonen am Tag eingeteilt: Die erwerbstätigen Akteure der "Unterschicht und unteren Mittelschicht" beginnen die Alltagsroutinen der Erwerbsarbeit viel früher am Morgen - im Durchschnitt 2 Stunden früher - und beenden diese auch früher am Tag. Ihre Mittagspause ist in einer anderen Zeitzone verortet (12 Uhr) als die Mittagspause der Erwerbstätigen aus "Oberschicht und oberen Mittelschicht" (13 Uhr). Männer der Oberschicht haben weitläufigere Zonen bei Freizeiten und bezahlten Arbeitszeiten. Dadurch eröffnen sich ihnen neue Interaktionsrahmen, in denen Wissen und Macht vermehrt werden können. Im Vergleich zu anderen Dimensionen sozialer Ungleichheit lassen sich Vor- und Nachteile sozialer Zeit schwieriger bestimmen. Während mit höherem materiellem Wohlstand, zunehmender Macht, Bildung und zunehmendem Prestige die Vorteile und Begünstigungen in der Gesellschaft ansteigen, so gilt diese "Je mehr desto besser" Regel bei der Zeit nicht unbedingt.
Diseases such as cardiac arrhythmias, CPVT and other issues of the human heart still remain largely unexplored. To contribute to this field of research, it is necessary to create tools to control the spatial and temporal release and reuptake of Ca2+ from the sarcoplasmic/endoplasmic reticulum (SR/ER). Ca2+ release and uptake by the ryanodine receptor (RyR) and Sarcoplasmic/endoplasmic reticulum calcium ATPase (SERCA), respectively, are essential for the function of excitable cells. In this process, the rapid Ca2+ release from the SR/ER and the associated contraction in muscle cells is modulated by RyR. However, diseases due to calcium leakage, such as cardiac arrhythmias, seizures and contractile dysfunction, are also caused by RyR. The resting Ca2+ concentration in the cytosol, which is important for the cell, is kept in balance by Ca2+ release and reuptake into the SR/ER. This reuptake is controlled quite considerably by SERCA. SERCA is important for development and muscle function in both nematodes such as C. elegans and mammals, though there is also a great need for tools that can help study precise function.
To advance towards the goal of developing tools for optogenetic stimulation of intracellular Ca2+ release from the SR/ER, the model organism C. elegans was chosen. Its advantages are the fully sequenced genome and the neural network connectome. In addition, the ease of maintenance, self-fertilisation, transparency and rapid generation cycles, as well as the fact that it is a eutelic animal, are advantages for the application of the optogenetic approach.
So far, tools for light-induced Ca2+ release (LICR) have already been developed, involving the creation of ChR2 versions with higher Ca2+ conductivity based on the "CatCh" variant and further improving their conductivity through several established mutations. In addition, the pharynx of C. elegans was modified to produce an optogenetically stimulated muscle pump that resembles mammalian cardiac muscle cells. In this work, both optoUNC-68 (optically excitable RyR) and SERCA/LOV2 were generated in different variants by CRISPR/Cas9 and plasmid-based genome editing to achieve light-driven manipulation of calcium homeostasis in C. elegans. Here, LICR was triggered by LOV2 domains in an opto-mechanical manipulation of RyR as well as SERCA. This approach was made possible by recently published high-resolution cryoEM structural images. In addition, alternative approaches using Ca2+ conductance-optimised channelrhodopsin variants were tested in C. elegans body wall muscle cells.
By inserting ChR-XXM into C. elegans and subsequent fluorescence microscopy of the co-introduced GFP, an expression in body wall muscle cells could be detected. Furthermore, in contraction assays, ChR-XXM was demonstrated to induce contractions of the animals of up to 16% compared to the original body length in both medium (0.8mW/mm²) and high (1.4mW/mm²) stimulation at 470nm. ChR-XXM was thus identified as an excellent candidate for the development of an optogenetic tool, as it exhibits significantly increased Ca2+ conductivity compared to other ChR2 variants.
The use of CRISPR/Cas9 to insert AsLOV2 domains (L404-L546) into different insertion sites of RyR allowed the generation of a transgenic strain of C. elegans that could be stimulated to elongate during 0.3mW/mm² photostimulation. This demonstrated that RyR can be manipulated by photostimulation, spatiotemporally through conformational changes in the LOV2 domain and the resulting disruption of the pore region.
The CRISPR/Cas9 method was also used to insert LOV2 domains into SERCA. Here it could be demonstrated that a conformational change of the LOV2 domains induced by photostimulation leads to a stop or impairment of Ca2+ ion translocation by SERCA from the cytosol into the SR/ER. In contrast to LOV2 in RyR, this resulted in a contraction of C. elegans body length.
The data presented here indicate that the intracellular Ca2+ cycle involving the SR/ER and cytosol can be successfully manipulated by the introduction of optogenetic tools. It turned out that the manipulation/impairment of individual components of this system, such as RyR or SERCA, is usually insufficient to achieve a clear response. Therefore, simultaneous manipulation of the two main actors RyR and SERCA is arguably the best way to take another step towards creating optogenetic tools for light-stimulated manipulation of Ca2+ release and reuptake from the SR/ER.
A generic drug product (World Health Organization (WHO) terminology: multisource product) is usually marketed and manufactured after the expiry date of the innovator’s patent. Generic drugs are less expensive than the innovator products because generic manufacturers do not have to amortize the investment costs of research, development, marketing, and promotion. Multisource products must contain the same active pharmaceutical ingredients (APIs) as the original formulation and have to be shown to be interchangeable with the original formulation. Multisource products have to be shown bioequivalent to the innovator counterpart with respect to pharmacokinetic and pharmacodynamic properties. Multisource products are therefore identical in dose, strength, route of administration, safety, efficacy, and intended use. Bioequivalence can be demonstrated by in vitro dissolution, pharmacokinetic, pharmacodynamic or clinical studies. Since 2000, the U.S. Food and Drug Administration (FDA) allows the approval of certain multisource products solely on the basis of in vitro studies, i.e. by waiving in vivo studies in humans (“Biowaiver”), based on the Biopharmaceutics Classification Scheme (BCS). The BCS characterizes APIs by their solubility and permeability in the gastrointestinal tract (GIT). The different BCS Classes I-IV (Class I: high solubility, high permeability; Class II: low solubility, high permeability; Class III: high solubility, low permeability and Class IV: low solubility, low permeability) result from all possible combinations of high and low solubility with high and low permeability. Since the adoption of the BCS by the FDA in 1995, the BCS criteria have been under continuous development. In 2006, the WHO has released the most recent bioequivalence guidance including relaxed criteria for bioequivalence studies based on modified BCS criteria. According to this guidance, APIs belonging to the BCS classes I – and under defined conditions - II and III – are eligible for a biowaiver-based approval. The principal objective of this work was to characterize the first-line anti tuberculosis APIs, isoniazid, pyrazinamide, ethambutol dihydrochloride and rifampicin, according to their physicochemical, biopharmaceutical, pharmacokinetic and pharmacological properties and to classify them according to the BCS. Ethambutol dihydrochloride and isoniazid were classified as borderline BCS class I/III APIs. Pyrazinamide was classified as a BCS class III and rifampicin as a BCS class II API. Based on the BCS classification and the additional criteria defined in the WHO bioequivalence guidance, the possibility of biowaiver-based approval for immediate release (immediate release) solid oral dosage forms containing the first-line antituberculosis drugs was evaluated. A biowaiver-based approval with defined constraints was recommended for immediate release solid oral dosage forms containing isoniazid (interaction with reducing sugars), pyrazinamide and ethambutol dihydrochloride (relative narrow therapeutic index). Rifampicin was classified as a BCS class II API, and it was concluded that rifampicin containing solid oral immediate release drug products as well as Scale-Up and Post-Approval Changes (SUPAC) changes should not be approved by a biowaiver on the following basis: (i) its solubility and dissolution are highly variable due to polymorphism and instability, (ii) concomitant intake of food and antacids reduces its absorption and bioavailability, (iii) no in vitro predictive dissolution test has been found which correlates to in vivo absorption and (iv) several publications reporting cases of non-bioequivalent and bioinequivalent rifampicin products have been located in the literature. Thus, it is recommended that bioequivalence of rifampicin containing solid oral immediate release drug products should be established by in vivo pharmacokinetic studies in humans. This risk-benefit benefit assessment of a biowaiver-based approval was presented as a poster at the American Association of Pharmaceutical Scientists (AAPS) 2005 and subsequently published as “Biowaiver Monographs” in the Journal of Pharmaceutical Sciences. Based on the assessment of the dissolution properties of the antituberculosis drugs for a biowaiver approval, quality control dissolution methodologies for the International Pharmacopoeia (Pharm. Int.) were developed, presented at the WHO expert meeting and adopted in the Pharm. Int. (http://www.who.int/medicines/publications/pharmprep/OMS_TRS_948.pdf). Additionally, preliminary biowaiver recommendations were also developed for four firstline antimalarial drugs listed on the WHO Essential Medicines List (EML): Quinine, as both the hydrochloride and sulphate, and proguanil hydrochloride were classified as borderline BCS class I/III APIs. Since quinine is a narrow therapeutic index drug and many cases of non-bioequivalence have been reported in the literature, a biowaiverbased approval was not recommended. For solid oral immediate release dosage forms containing proguanil a biowaiver-based approval was recommended under the condition that they dissolve very rapidly. Primaquine phosphate was classified as a BCS class I API. Therefore, a biowaiver-based approval was recommended for immediate release solid oral dosage forms containing primaquine phosphate. Mefloquine hydrochloride was classified as a basic, BCS class IV/II API, making it ineligible for the biowaiver. Additionally, reports of non-bioequivalence and a narrow therapeutic index were found in the scientific literature. Consequently, bioequivalence of solid oral immediate release dosage forms containing mefloquine hydrochloride should be established by in vivo pharmacokinetic studies. The results for quinine hydrochloride and sulphate, proguanil hydrochloride, primaquine diphosphate and mefloquine hydrochloride were presented as a poster at the Pharmaceutical Sciences World Congress (PSWC) 2007 and published as a WHO Collaborating Center Report in June 2006. The aim of this project was to collect, evaluate, generate and publish relevant information for a biowaiver-based approval of essential medicines in order to provide a summary to local regulatory authorities. This information complements the selected list of essential medicines by providing information about the biopharmaceutical properties and pharmaceutical quality of solid oral immediate release dosage forms containing these APIs. The aim of the biowaiver project, inspired by the WHO and brought in life by the International Pharmaceutical Federation (FIP), is to enable access to essential medicines in standardized quality at an affordable price. In this work, a significant contribution to this aim in the form of four biowaiver monographs for the antituberculosis drugs and several reports on the antimalarials has been achieved.
Die Integration von Dienstgüte-Vorkehrungen in objektorientierte Verteilungsinfrastrukturen befähigt Anwendungsentwickler, den Verteilungs-induzierten Problemen verteilter Systeme zu begegnen. Im Rahmen dieser Arbeit wurde die generische Einbettung von Dienstgüte-Vorkehrungen in verteilte Objektsysteme untersucht und ein Lösungsansatz präsentiert. Zunächst wurde eine Analyse der für das Dienstgüte-Management notwendigen Aufgaben vorgestellt. Ausgehend von einem verteilten Objektmodell wurde untersucht, wie Dienstgüte-Vorkehrungen integriert werden können. Dienstgüte-Vorkehrungen stellen bei einem zugrundeliegenden Ob- jektmodell nicht-einkapselbare Verantwortlichkeiten dar. Die enge Bindung der Dienstgüte-Vorkehrungen an einen Dienst führt so zu Vermaschungen in den Strukturen der Implementierung. Damit ist die getrennte Wieder- verwendung beider erschwert. Zusätzlich werden unterschiedliche Abstrak- tionen vermischt. Die aspektorientierte Programmierung (AOP) behandelt solche Vermaschungen. Dienstgüte wurde bei der Integration in ein verteil- tes Objektmodell als ein Aspekt im Sinne der AOP klassifiziert. Ausgehend von den Anforderungen an das Dienstgüte-Management wur- de ein Rahmenwerk auf Basis eines verteilten Objektmodells entworfen. Der in dieser Arbeit dargestellte Schwerpunkt liegt auf der Spezifikation von Dienstgüte-Charakteristiken und deren Umsetzung in die Implementie- rungssprache der Anwendungsobjekte. Für die Unterstützung der Ende-zu- Ende-Dienstgüte-Erbringung ist der Einbezug von Dienstgüte-Vorkehrun- gen des Netzwerks, Betriebssystems oder spezieller Bibliotheken notwendig. Die resultierende Hierarchie von Dienstgüte-Mechanismen wird durch die vorgestellte Integration in eine Verteilungsinfrastruktur unterstützt. Durch die Integration der Dienstgüte-Spezifikation in die Schnittstel- lenbeschreibungssprache erlaubt das Rahmenwerk einen aspektorientierten Ansatz ohne die Einführung weiterer Sprachen zur Spezifikation oder Im- plementierung. Die Spezifikation von Dienstgüte-Charakteristiken in der erweiterten IDL wird in spezielle Entwurfsmuster in der Zielsprache umge- setzt. Diese Entwurfsmuster separieren die Anwendungsobjekte weitgehend von den Dienstgüte-Vorkehrungen. Die auf der Ebene der Anwendungsobjekte generierten Vorlagen für die Dienstgüte-Vorkehrungen können durch einen modifizierten bzw. schon da- für ausgelegten Verteilungsinfrastrukturkern in das System integriert wer- den. Eine einheitliche statische Schnittstelle erlaubt einen einfachen re- effektiven Ansatz. So ist der Zugriff auf Dienstgüte-Vorkehrungen tieferer Schichten wie auch die Integration anwendungsspezifischer Dienstgüte-Vor- kehrungen auf der Netzwerkschicht möglich. Das Rahmenwerk bietet somit eine klare Trennung der Verantwortlich- keiten, die sowohl Anwendungsentwickler wie auch Dienstgüte-Implemen- tierer unterstützt. Die aus der Schnittstellenbeschreibungssprache generier- ten Einheiten stellen für die Anwendungsobjekte eine Abstraktion dar, die sowohl die Verteilungsaspekte wie auch die Dienstgüte-Vorkehrungen ein- fach nutzbar anbietet und von der zugrundeliegenden Plattform isoliert. Eine sich aus dieser Arbeit ergebende Fragestellung besteht in der Er- weiterung und Verallgemeinerung des aspektorientierten Ansatzes. Die im Rahmen der Analyse betrachteten Dienstgüte-Charakteristiken sind aus dem systemnahen Bereich und insbesondere aus der Betrachtung typi- scher Probleme in verteilten Systemen und den daraus erwachsenen Anwen- dungsanforderungen gewonnen. Nicht-funktionale Aspekte der Dienster- bringung lassen sich weiter fassen. So kann ausgehend von den bereitge- stellten Abstraktionen untersucht werden, inwieweit auf Anwendungsebe- ne nicht-funktionale Eigenschaften in ähnlicher Weise einbettbar sind. Im Rahmen dieser Arbeit wurde beispielsweise eine Dienstgüte-Charakteristik zur Parallelisierung von Berechnungen realisiert. Eine anwendungsbezogene Dienstgüte-Charakteristik könnte numerische Optimierungen realisieren, die von den reinen mathematischen Operationen zu trennen ist. Andere Beispiele aus der Multimedia-Kategorie sind durch die Qualität einer Au- dio-Übertragung gegeben. So kann bei einer geringen Bandbreite durch die Kompression der Daten eine bessere Qualität der Audiowiedergabe ereicht werden, als durch Übertragung der Rohdaten. Die Kompressionsrate kann von der Anwendung isoliert und durch entsprechende Dienstgüte-Mecha- nismen realisiert werden. Qualitätsunterschiede ergeben sich durch mögli- che verlustbehaftete Kompression und de notwendigen Anforderungen an Hardware- oder Software-Unterstützung. Andere Kriterien für die Qualität lassen sich weniger leicht vor der Anwendung verbergen. Die Wiedergabe von Stereo- oder Mono-Audiodaten erfordert entsprechende Anwendungen und auch Ausstattungen der Endgeräte. Im Kontext dieser Arbeit wurde ein Objektmodell betrachtet, das eine starke Bindung zwischen Schnittstellen und Objekten besitzt. Insbeson- deren wurde bei der Umsetzung der Schnittstellenbeschreibungssprache in die Zielsprache eine Umsetzung gewählt, die Dienste als Objekte reprä- sentiert. Involviert die Diensterbringung verschiedene Objekte, kann nur ein Objekt als Stellvertreter all dieser Dienste den Service anbieten. Dieses Objekt ist für die Einhaltung von Dienstgüte-Vereinbarungen mit Klien- ten verantwortlich. Innerhalb der Objekte, die den Service realisieren, sind für die Dienstgüte-Erbringung dann ggf. weitere interne Dienstgüte-Vor- kehrungen zu etablieren. Komponentenmodelle versprechen hier einen all- gemeineren Ansatz, der die Integration von Dienstgüte-Vorkehrungen loh- nenswert erscheinen lässt. Zum einen unterstützen Komponentenmodelle definierte Schnittstellen zur Interaktion zwischen den beteiligten Objek- ten einer Komponente, und zum anderen bieten Komponenten eine über die Schnittstellenbeschreibungssprache hinausgehende Beschreibung ihrer Funktionalität in einer Komponentenspezifikation. Diese Komponentenspe- zifikation verspricht einen guten Ansatz, um Dienstgüte-Spezifikationen der Komponenten zu integrieren. Neben den beiden bislang beschriebenen Forschungsrichtungen, die je- weils ein Rahmenwerk für das Dienstgüte-Management voraussetzen und darauf aufbauen, existieren innerhalb des in der Arbeit vorgestellten Rah- menwerkes weitere offene Forschungsfragen. Die Ausgestaltung von Preisen bei der Vergabe von Ressourcen und die damit verbundenen Richtlinien für die Vergabe und auch den Entzug stellen noch kein abgeschlossenes Gebiet dar. Hier ist der Einbezug anderer Disziplinen vielversprechend. Preisrichtlinien für manche Ressourcen, die bei Nicht-Nutzung verfallen wie Netzwerkkapazität sind Gegenstand der Forschung in der Betriebs- wirtschaftslehre. Die Gestaltung von Vergaberichtlinien, insbesondere aber die Festlegung von Vergütungen bei Nichterbringung eines festgesetzten Dienstgüte-Niveaus oder Kompensationen bei dem Entzug von Ressourcen mit einer damit einhergehenden Verletzung der Dienstgüte-Vereinbarung, wirft rechtliche Fragen über die Gültigkeit solcher Richtlinien auf. Weitere, nicht-interdisziplinäre Fragestellungen, ergeben sich aus der Frage der Wiederverwendbarkeit und Dokumentation von Dienstgüte-Vor- kehrungen im Rahmenwerk. Die Erstellung eines Katalogs mit einem ein- heitlichen Aufbau wie es bei Entwurfsmustern üblich ist verspricht eine geeignete Dokumentationsform. Allerdings muss eine solche Dokumentati- on zwei Zielgruppen gerecht werden. Zum einen sind dies Anwendungsent- wickler, die eine gegebene Dienstgüte-Implementierung anwenden wollen und Informationen für die Nutzung und Anpassung der Anwendung benö- tigen und zum anderen Dienstgüte-Entwickler, die auf bereits existierende transportspezifische Dienstgüte-Mechanismen aufbauen. Für die hier skizzierten Forschungsrichtungen ist ein Rahmenwerk für das Dienstgüte-Management unerlässlich. Das in dieser Arbeit vorgestellte Rahmenwerk bietet eine gute Ausgangsbasis.
European pea crabs - taxonomy, morphology, and host-ecology (Crustacea: Brachyura: Pinnotheridae)

Pinnotherids are small crabs symbiotic to a variety of invertebrates. The European species infest bivalves and sea squirts. Their way of life is parasitic and poses a threat to commercially exploited bivalves. While juveniles of both sexes still look very similar - being agile swimmers and partially free living - a metamorphosis takes place in the female after mating and results in a conspicuous sexual dimorphism. Thereafter, the female settles in its host definitely and is morphologically strongly adapted to the parasitic life phase. A very high reproductive output was demonstrated among several pea crab species infesting bivalves. Despite from that, hardly any information is present in the literature on the pinnotherids’ reproductive biology and the underlying morphology.
Due to their cryptic way of life, the sexual dimorphism, and the different morphotypes of the female, the taxonomy of the Pinnotheridae is a serious challenge. Two widely accepted species are recognized on European coasts: Pinnotheres pisum and Nepinnotheres pinnotheres. Pinnotheres pectunculi was so far only known from the bivalve Glycymeris glycymeris in its type locality Roscoff (France), while Pinnotheres ascidicola and Pinnotheres marioni were described as living exclusively in ascidians without careful comparison with the previously described species. In order to produce standardized comparative descriptions, pea crabs were collected and studied from different hosts and localities in the Northeast Atlantic and in the Mediterranean. Nepinnotheres pinnotheres and Pinnotheres pisum were redescribed with consideration to characters of female and male. According to our morphological analysis, Pinnotheres ascidicola and Pinnotheres marioni are junior synonyms of Nepinnotheres pinnotheres, whereas the status of Pinnotheres pectunculi as a valid species was ascertained. Important characters are the mouthparts, the male gonopods, and especially chelipeds that showed consistent characteristics among different crab stages of both sexes.
Based on our sampling, we estimated the host-range of the European species. Nepinnotheres pinnotheres lives in ascidians and in the pen shell Pinna nobilis. Pinnotheres pisum infests numerous bivalve species - Pinna nobilis included. For Pinnotheres pectunculi novel host records are presented, all from the bivalve family Veneridae. Furthermore, feeding of the Pinnotheres-species was observed. They use a setae comb ventrally on the claw to brush mucus (and the accumulated food particles) from the bivalve gills. Feeding strategies and host-ecology will be thoroughly discussed in consideration to other Pinnotheridae.
We investigated the reproductive systems of European pinnotherids by histological methods, scanning and transmission electron microscopy, and confocal laser scanning microscopy.
The Eubrachyura have internal fertilization: paired vaginas enlarge into storage structures, the spermathecae, which are connected to the ovaries by oviducts. Sperm is stored until the oocytes are mature and transported into the spermathecae, where fertilization takes place. In the investigated pinnotherids, the vagina is of the ‘concave pattern’. Musculature is attached alongside flexible parts of the vagina-wall to control the dimension of its lumen. The genital opening is closed by a muscular mobile operculum.
The spermatheca can be divided into two distinct regions by function and morphology. The ventral part includes the connection with vagina and oviduct and is regarded as the zone where fertilization takes place. It is lined with cuticle except where the oviduct enters the spermatheca by the ‘holocrine transfer tissue’. At ovulation, the oocytes have to pass through this multi-layered glandular epithelium, which has a holocrine mode secretion. The dorsal part of the spermatheca is lined by a highly secretory apocrine glandular epithelium, which was to date only found in fiddler crabs of the genus Uca.
The male internal reproductive system consists of paired testes and corresponding vasa deferentia. The sperm morphology of pinnotherids conforms to other thoracotremes, with slight differences between Nepinnotheres pinnotheres and Pinnotheres pisum. Spermatozoa become enveloped into spermatophores in the secretory proximal vas deferens. The medial vas deferens is strongly enlarged and stores spermatophores embedded in seminal plasma. The distal vas deferens holds tubular appendices, which extend into the ventral cephalothorax and slightly into the pleon. These appendices produce and store vast quantities of seminal plasma. The copulatory system of the Brachyura is formed by paired penes and two pairs of gonopods, which function in sperm transfer. In pinnotherids, the long first gonopods transfers the sperm mass to the female. It holds the ejaculatory canal inside, which opens proximally and distally. The second gonopod is solid, short and conical. During copulation, the penis and the second gonopod are inserted into the base of the tubular first gonopod. The second gonopod functions in the transport of the sperm mass inside the ejaculatory canal towards its distal opening. The specific shape of the second gonopod is strongly adapted for a sealing of the tubular first gonopod with longitudinal cuticle foldings that interlock inside the first gonopod. The presented results are discussed concerning their function in reproduction and in respect of the systematic account.
The role of secretion in sperm transfer, storage and fertilization among the Brachyura is still under debate. It is notable that structure and function of secretion are more complex in pinnotherids and probably more efficient than in other brachyuran crabs, which will be discussed, in view of the parasitic way of life and the high fecundity of pinnotherids.
