Background: European robins, Erithacus rubecula, show two types of directional responses to the magnetic field: (1) compass orientation that is based on radical pair processes and lateralized in favor of the right eye and (2) so-called 'fixed direction' responses that originate in the magnetite-based receptors in the upper beak. Both responses are light-dependent. Lateralization of the 'fixed direction' responses would suggest an interaction between the two magnetoreception systems. Results: Robins were tested with either the right or the left eye covered or with both eyes uncovered for their orientation under different light conditions. With 502 nm turquoise light, the birds showed normal compass orientation, whereas they displayed an easterly 'fixed direction' response under a combination of 502 nm turquoise with 590 nm yellow light. Monocularly right-eyed birds with their left eye covered were oriented just as they were binocularly as controls: under turquoise in their northerly migratory direction, under turquoise-and-yellow towards east. The response of monocularly left-eyed birds differed: under turquoise light, they were disoriented, reflecting a lateralization of the magnetic compass system in favor of the right eye, whereas they continued to head eastward under turquoise-and-yellow light. Conclusion: 'Fixed direction' responses are not lateralized. Hence the interactions between the magnetite-receptors in the beak and the visual system do not seem to involve the magnetoreception system based on radical pair processes, but rather other, non-lateralized components of the visual system.
Das Neuroblastom ist ein Tumor, der sich von sympathoadrenergen Vorläuferzellen ableitet und die häufigste solide Krebsform im Kindesalter darstellt. Das breite klinische Spektrum dieses Tumors, das von spontaner Regression zu fataler Progression reicht, spiegelt die außerordentliche biologische und genetische Heterogenität dieses Tumors wider. Polyploidie und Genexpressionsanalysen werden auf klinischer Seite zur Risiko- und Therapieeinschätzung eingesetzt. Genomweite Screeninganalysen identifizierten den Transkriptionsfaktor Phox2b als erstes Prädispositionsgen für NB. Frühere Arbeiten haben gezeigt, dass Phox2b absolut essentiell für die Entstehung aller Ganglien des autonomen NS aus Neuralleistenzellen ist. Ziel der Untersuchungen dieser Arbeit war es, die Auswirkungen der NB-assoziierten Phox2b-Mutationen auf Proliferations- und Differenzierungsverhalten sympathoadrenerger Vorläuferzellen zu untersuchen. Hierzu wurden paravertebrale, sympathische Grenzstränge aus Huhnembryonen des Embryonaltags 7 präpariert, dissoziiert und mit Expressionsplasmiden für Phox2bwt und NB-Phox2b-Mutationen transfiziert. Nach zwei Tagen in Kultur wurden mit molekularbiologische Methoden Veränderungen des Proliferations-und Differenzierungsverhalten untersucht. Die Analyse des Proliferationsverhaltens transfizierter Neurone mit BrdU-Proliferationsassays und Phospho-Histon-3-Antikörpern offenbarte einen stark antiproliferativen Effekt des Phox2bwt-Proteins, den die untersuchten NB-Phox2b-Mutationen nicht aufwiesen. NB-Phox2b-Patienten sind heterozygot, d.h. Träger eines gesunden und eines mutierten Phox2b-Allels. Um die genetische Situation im NB-Patienten nachzuahmen, wurde mit spezifischen siRNAs das endogene Phox2b-Protein-Niveau herunter reguliert. NB-Phox2b-Mutationen mit mis- oder nonsense Mutation in der Homöodomäne zeigten unter Phox2b-Knockdown-Bedingungen einen proliferationsstimulierenden Effekt. Diese Experimente warfen die Frage auf, über welche Mechanismen Phox2b und NB-Phox2b-Mutationen Einfluss auf die Zellzykluskontrolle nehmen. Die quantitative Analyse der Expression bekannter Zellzyklusregulatoren wie Zyklin D1, D2 und D3 und der Zyklin-abhängigen Kinase-Inhibitoren p18, p21 und p57kip2 verlief ergebnislos. Die Überexpression des Zyklin-abhängigen Kinase-Inhibitors p27kip1 wirkte antiproliferativ auf Kulturen sympathoadrenerger Vorläuferzellen und das p27kip1-Epxressionsniveau korrelierte mit dem Phox2b-Proteinniveau. P27kip1 scheidet jedoch als alleiniger Vermittler des antiproliferativen Effekts von Phox2b aus, da die ebenfalls antiproliferativ wirkende Phox2-Homöodomäne keinen Einfluss auf das p27kip1-Expressionsniveau besitzt. Vielmehr wurde der bHLH-Transkriptionsfaktor Hand2 als Mediator der Proliferationseffekte von Phox2b identifiziert. Die Proliferation sympathoadrenerger Vorläuferzellen ist abhängig von der Hand2-Proteinmenge, und Hand2-Überexpression ist ausreichend, den antiproliferativen Effekt von Phox2b aufzuheben. Damit im Einklang geht der proliferationsstimulierende Effekt der Phox2bHDmut bei siRNA-vermitteltem Hand2-Knockdown verloren. Weiterhin führt Phox2b-Überexpression zu verringertem Hand2-Expressionsniveau und Phox2b-Knockdown zu vermehrter Hand2-Transkription. In Protein-Protein-Interaktionsexperimenten konnte eine direkte Bindung von Hand2 an Phox2bwt und die untersuchten NB-Phox2b-Mutanten nachgewiesen werden. Diese Ergebnisse zeigen, dass Hand2 in der Vermittlung der Phox2b-Proliferationseffekte auf transkriptioneller und posttranskriptioneller Ebene involviert ist. Der Wirkungsmechanismus dieser Phox2b-Varianten / Hand2-Komplexe konnte nicht endgültig geklärt werden und wird in Form verschiedener Modelle diskutiert. In NB-Patienten korreliert ein hohes Expressionsniveau von Differenzierungsmarkern mit mildem Krankheitsverlauf. Die Rolle von Phox2b als Schlüsselgen in der Entstehung und Differenzierung autonomer Neurone und die genetische Heterogenität des NB legen eine Funktion von NB-Phox2b-Mutationen auf den Differenzierungsstatus sympathoadrenerger Vorläuferzellen nahe. In quantitativen Analysen wurde die Expression von Phox2b-Zielgenen und in NB-Diagnostik involvierten Genen untersucht. Phox2bK155X, eine C-terminal trunkierte NB-Phox2b-Mutation, wirkte dominant-negativ auf die Expression der noradrenergen Markergene Th und Dbh, Tlx3 und die Neurotrophinrezeptoren trkA und p75, deren reduzierte Expression mit schlechter Prognose und damit aggressiven NB-Formen korreliert ist. Zusammenfassend konnte in dieser Arbeit gezeigt werden, das NB-Phox2b-Mutationen in sympathoadrenergen Vorläuferzellen, den potentiellen Tumorentstehungszellen des NB, proliferationsstimulierend wirken und zumindest die C-terminal trunkierte Phox2bK155X-Mutation zur Dedifferenzierung dieser Zellen führt. Hereditäre Mutationen in Phox2b könnten im Patienten nicht nur die terminale Differenzierung sympathischer Neurone stören, sondern auch durch eine verlängerte Phase der Neurogenese die Empfänglichkeit für weitere Tumor-initiierende Mutation erhöhen.
Mesenchymale Stammzellen (MSC) rücken in der regenerativen Medizin und im Tissue Engineering immer mehr in den Vordergrund. Im Gegensatz zu embryonalen Stammzellen bergen sie keine ethischen Probleme und sind leicht zu isolieren. Die ursprünglich aus Knochenmark gewonnenen MSC können inzwischen aus vielen verschiedenen Quellen wie Nabelschnurblut [Kern et al. 2006], Dentalgewebe [Huang et al., 2009], Plazenta [Huang et al., 2009], Haut [Salvolini et al., 2009] und aus Fettgewebe isoliert werden [Zuk et al., 2001]. Der Immunphänotyp variiert zwischen den aus verschiedenen Quellen gewonnenen MSC nur gering. Die Gewinnung aus Fettgewebe hat den Vorteil, dass eine minimal invasive Prozedur und eine hohe Ausbeute zusammenkommen. Die Stammzellen aus Fettgewebe (ASC) können in der Therapie eingesetzt werden und zu der Regeneration von Geweben nach Verletzungen beitragen [Wong et al., 2008; Poulsom et al., 2001]. Der genaue Mechanismus mit dem die Stammzellen wirken ist allerdings noch nicht geklärt. Sowohl die Integration der MSCs in das Gewebe als auch ein rein parakriner Einfluss wurden nachgewiesen. Klar ist nur, dass ein positiver Effekt von einer Therapie mit MSC ausgeht [Mizuno et al., 2009]. In vivo sind Zellen verschiedenen Einflüssen ausgesetzt, die den Zustand einer Zelle bestimmen. Lösliche Faktoren wie Wachstumsfaktoren, Hormone oder Vitamine wirken dabei ebenso wie die extrazelluläre Matrix und Zell-Zell-Kontakte auf die Zelle ein, die mit Wachstum, Zellform, Differenzierung oder Ähnlichem antwortet. In meiner Arbeit wurden daher drei unterschiedliche Ansätze für die in vitro Differenzierung von ASC in epitheliale Tubuluszellen untersucht: (1) die Wirkung von löslichen Faktoren, die dem Medium zugesetzt wurden, (2) der Einfluss der extrazellulären Matrix aus zum einen Tubuluszellen und zum anderen Matrigel und (3) die Co-Kultur, bei der auch direkter Zell-Zell-Kontakt untersucht wurde. Damit, und mit einer Kombination der einzelnen Bereiche, sollte das natürliche Umfeld der Tubuluszellen simuliert werden und epitheliale Differenzierung initiieren. Die Zugabe von ATRA, ActA und BMP-7 führte zu einer Differenzierung in die epitheliale Richtung, während die extrazelluläre Matrix aus Tubuluszellen nicht dafür ausreichte. Matrigel hingegen, konnte besonders in der Verbindung mit konditioniertem Medium eine Differenzierung induzieren. Die indirekte Co-Kultur über Membraneinsätze, über die u. a. der parakrine Einfluss der Tubuluszellen untersucht werden sollte, führte zu morphologischen Veränderungen der ASC, die aber nicht mit den hier verwendeten epithelialen Markern nachgewiesen werden konnte. Der direkte Zell-Zell-Kontakt zeigte eine Reduktion des Oberflächen Markers CD90 verbunden mit einer Erhöhung der Expression von CK18. Die Differenzierung von ASC in epitheliale Zellen ist also auf drei verschiedenen Wegen möglich. Zwischen verschiedenen Isolationen von ASC traten hohe Schwankungen bezüglich der Expression von Oberflächenmarkern und Proliferation auf, was auch einen Einfluss auf die Differenzierung haben könnte. Ein Grund dafür könnte die Heterogenität von ASC sein. Zur Reduzierung dieser wurden daher ein Waschschritt eine Stunde nach Kulturbeginn und eine immunomagnetische Isolation mit CD49a, CD90, CD105 oder CD271 durchgeführt. Die immunomagnetische Aufreinigung führte nur zu einer leichten Verbesserung der Heterogenität, aber zu einer sehr geringen Zellausbeute. Für den Waschschritt konnte gezeigt werden, dass die Expression der Stammzellmarker Nestin, oct4 und sall1 signifikant erhöht und Desmin und smA Expression reduziert wurden, was auf eine Reduktion der Heterogenität hindeutete. Der zusätzliche Waschschritt kann also schnell und unkompliziert die Heterogenität der ASC reduzieren. Die Differenzierung von ASC in epitheliale Tubuluszellen durch den Einfluss von Zell-Zell-Kontakten zeigt vielversprechende Ansätze und sollte weiter verfolgt werden. Eine verlängerte Kulturdauer sollte dabei angestrebt werden, da auch die adipogene Differenzierung zumeist erst nach 14 bis 21 Tagen nachweisbar war. Dafür müsste die Markierung mit CellTracker länger nachweisbar sein. Eine Inhibierung der Proliferation könnte die Grundlage dazu liefern. Um den Stand der Differenzierung in die epitheliale Richtung nachzuweisen, könnten andere epitheliale Marker, Ionenkanäle, die erst spät in den Tubuluszellen angelegt werden, und funktionelle Mechanismen untersucht werden. Das Zusammenspiel der verschiedenen Einflüsse auf die Zelle könnte ebenfalls noch genauer untersucht werden. Für die Regenerative Medizin ist es aus Gründen der GMP sinnvoller, die Zellen nur mit dem Zusatz an löslichen Faktoren zu differenzieren. Der Ansatz mit ATRA, ActA und BMP-7 scheint sehr vielversprechend zu sein und könnte in dieser Hinsicht weiter ausgebaut werden.
The Opisthobranchia comprise highly specialized marine gastropods and have therefore been subject to diverse investigations covering various biological disciplines. However, a robust phylogeny of these gastropods is still lacking and several subclades have only been rarely studied. Furthermore, crucial aspects for the evolution of Opisthobranchia have not been comparatively analysed. Therefore, the aim of the present thesis is to gain new insights into the phylogeny of the Opisthobranchia with special focus on certain critical groups (Pleurobranchomorpha, Acteonoidea) and to assess several crucial features of the evolution of the investigated clades. The combination of four different gene markers (18S rDNA, 28S rDNA, 16S rDNA and CO1) and modern molecular systematic analysis tools were used to construct phylogenetic hypotheses focussing on Opisthobranchia as a whole as well as Pleurobranchomorpha and Acteonoidea in more detail. Intriguing new aspects of phylogeny and evolution of Opisthobranchia were revealed. First of all, monophyly of Opisthobranchia is definitely rejected based on the present data, while monophyly of Euthyneura (comprising Opisthobranchia and Pulmonata) is supported. Monophyly of opisthobranch subclades is confirmed for Nudipleura (as well as its constituting groups Nudibranchia and Pleurobranchomorpha), Umbraculida, Pteropoda (as well as subclades Thecosomata and Gymnosomata) and Acochlidiacea, for Cephalaspidea (if Runcinacea is regarded as a separate clade) and for Sacoglossa (if Cylindrobulla is accepted as an Oxynoacea). Aplysiomorpha are rendered paraphyletic due to the position of Akera bullata, but this result needs further investigation and should be considered with caution. The Nudipleura are found as the first single offshoot of the Euthyneura implying an early evolutionary separation of the last common ancestor of this clade. The remaining taxa form two main clades, one comprising the opisthobranch subgroups Umbraculida, Cephalaspidea, Aplysiomorpha and Pteropoda, while the other contains the pulmonate taxa and the opisthobranch Sacoglossa and Acochlidiacea. The interrelationships within these clades remain largely unresolved due to low statistical support values. However, a possible sister group relationship of Acochlidiacea and Eupulmonata receives statistical support. Opisthobranchia display various highly specific adaptations to diverse food sources. However, evolution of these specialized traits has never been assessed at an analytical level. The current thesis reconstructs the evolution of dietary preferences with novel methodologies based on the newly proposed phylogenetic hypothesis. Reconstruction of dietary evolution revealed herbivory as the ancestral condition in Euthyneura implying that carnivory evolved at least five times independently in the diverse lineages. The first comprehensive molecular phylogenetic hypothesis of the Pleurobranchomorpha could not reveal monophyly of the two main subclades Pleurobranchaeidae and Pleurobranchidae. This is due to the position of a single taxon (Euselenops luniceps) which is assigned to the Pleurobranchaeidae based on morphology but clusters within Pleurobranchidae in the current hypothesis. Furthermore, the tribe Berthellini and the genus Berthella are rendered paraphyletic by the current analyses. The results of molecular systematic analyses were used to reconstruct historical biogeography of Pleurobranchomorpha. Four different methodological approaches were applied yielding ambiguous results for Pleurobranchomorpha. However, the Pleurobranchidae comprising about 80% of the extant Pleurobranchomorpha most probably derived from an Antarctic origin. Dating of the phylogenetic tree via molecular clock methods yielded divergence of Pleurobranchidae into the Antarctic Tomthompsonia antarctica and the remaining species in Early Oligocene. Afterwards the latter underwent rapid radiation during Oligocene and Early Miocene. This divergence event coincides with two major geological events in the Antarctic region. On the one hand, the onset of glaciation and on the other hand the opening of the Drake Passage with concurrent formation of an Antarctic circumpolar current (ACC). I suppose that these sudden and dramatic changes in climate and palaeogeography probably accounted for migration of the last common ancestor of Pleurobranchidae (besides Tomthompsonia) into warmer regions via the Drake Passage to the Western Atlantic and Eastern Pacific and via the South Tasman Rise to the Indo-West Pacific. Furthermore, the ACC may have triggered larval dispersal to the Eastern Atlantic. The phylogenetic position of Acteonoidea has been a matter of debate for decades and they have long been considered as basal opisthobranchs. Results of the present thesis rather support placement in “Lower Heterobranchia” as sister group of Rissoelloidea. The current division of Acteonoidea into three families has never been investigated by means of phylogenetic methods. Thus, this thesis provides the first comprehensive investigation of this clade challenging present division into three families. The results rather support division into two main clades with the monogeneric Bullinidae clustering within Aplustridae doubting its separate status. Additionally, Rictaxis punctocaelatus which has been assigned to Acteonidae clusters basal to Aplustridae rendering Acteonidae paraphyletic. Since information on morphology of R. punctocaelatus was lacking until now, I conducted the first detailed investigation on morphology and histology of this species in order to reassess the unexpected molecular systematic placement. Character tracing analyses revealed similarities with both acteonoidean families implying an intermediate position of this species which might be assigned to a separate family in the future. Furthermore, the common features of Acteonidae and Rictaxis (massive shell, small foot, anterior mantle cavity opening, and absence of oral gland) are possibly plesiomorphic for the whole Acteonoidea. In summary, the results of the present thesis provide valuable novel insights into the phylogeny and evolution of the Opisthobranchia by employing state-of-the-art approaches of molecular systematics and evolutionary reconstruction. Thus, diverse hypotheses on opisthobranch phylogeny and evolution were either supported or rejected as well as novel hypotheses proposed which offer the basis for further research on these extraordinary gastropods.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 Protein kinases are important targets for drug development. The almost identical protein folding of kinases and the common co-substrate ATP leads to the problem of inhibitor selectivity. Type II inhibitors, targeting the inactive conformation of kinases, occupy a hydrophobic pocket with less conserved surrounding amino acids. Human polo-like kinase 1 (Plk1) represents a promising target for approaches to identify new therapeutic agents. Plk1 belongs to a family of highly conserved serine/threonine kinases, and is a key player in mitosis, where it modulates the spindle checkpoint at metaphase/anaphase transition. Plk1 is over-expressed in all today analyzed human tumors of different origin and serves as a negative prognostic marker in cancer patients. The newly identified inhibitor, SBE13, a vanillin derivative, targets Plk1 in its inactive conformation. This leads to selectivity within the Plk family and towards Aurora A. This selectivity can be explained by docking studies of SBE13 into the binding pocket of homology models of Plk1, Plk2 and Plk3 in their inactive conformation. SBE13 showed anti-proliferative effects in cancer cell lines of different origins with EC50 values between 5 microM and 39 microM and induced apoptosis. Increasing concentrations of SBE13 result in increasing amounts of cells in G2/M phase 13 hours after double thymidin block of HeLa cells. The kinase activity of Plk1 was inhibited with an IC50 of 200 pM. Taken together, we could show that carefully designed structure-based virtual screening is well-suited to identify selective type II kinase inhibitors targeting Plk1 as potential anti-cancer therapeutics.
Feral cats (Felis catus), introduced into Australia with European settlers in the 19th century, colonized the entire Australian continent in less than 100 years, including the Australian arid zone which covers more than 70% of the continent. Feral cats are responsible for the decline and extinction of a number of native species and the failure of a number of reintroduction attempts, especially in the arid zone. Many ecological studies on feral cats have been conducted on home range size and movement patterns in different environments, abundance and diet, with the aim of gaining a better understanding about their successful invasion of the Australian continent. There are no physiological studies on the feral cat to date. However, there is evidence that there is a strong interrelation between physiology and abiotic factors such as climate. Thus, distribution, habitat, and dispersal of species can not fully be understood without background knowledge of physiology. This PhD aims to contribute to a better understanding of three physiological parameters: metabolism, body mass and body temperature patterns. These parameters may possibly identify physiological adaptation to different climate zones, seasonal conditions and island isolation.
Im Rahmen dieser Arbeit wurde die Funktion ausgewählter Gene näher charakterisiert und ihr globaler Einfluß auf die Regulation der Genexpression untersucht. Im Fokus der Untersuchungen stand ein Gen, welches im Rahmen der Promotion von Alexander Zaigler in der Arbeitsgruppe Soppa (Universität Frankfurt) zunächst als Transkriptionsregulator ähnlich zu rspA aus E. coli identifiziert wurde. Zu Beginn dieser Arbeit konnte durch in silico Analysen das entsprechende Genprodukt zur Enolase Superfamilie (COG1441) zugeordnet werden. Mit Hilfe der „pop-in/pop-out“ Methode wurde das Gen in H. volcanii in frame deletiert und durch Wachstumsversuche, Northern- sowie Westernblot Analysen näher charakterisiert. Bei der Untersuchung des Wachstums konnte ein bemerkenswerter Phänotyp entdeckt werden: nur der Wechsel der Nährstoffquelle von reichhaltigen zu armen Bedingungen resultierte in einer dreitägigen Lagphase. Darüber hinaus wurde die Genexpression im Laufe eines Wachstumzyklus mittels Northern- und Westernblot Analysen bestimmt. Während das Transkript in reichhaltigem Medium nur transient expremiert wurde, wurde es in nährstoffarmen Bedingungen in allen Wachstumsphasen sehr stark induziert. Durch die Ergebnisse konnte zudem eine translationale Regulation der Genexpression nachgewiesen werden. Die Resultate offenbarten eine wichtige Funktion bei der Transition von reichem zu ärmerem Nährstoffangebot und führten schließlich zur Genbezeichnung iftA („important for transition A“). Desweiteren wurden Transkriptomuntersuchungen der Deletionsmutante im Vergleich zum Wildtyp zum Zeitpunkt der höchsten transienten Expression von iftA durchgeführt. Dadurch konnte gezeigt werden, dass iftA etwa 1% aller Gene des Genoms beeinflußt und gleichzeitig die Zahl differentieller Funktionen dieser Gene sehr gering ist. Die Ergebnisse führten insgesamt zu der Annahme, dass iftA eine Doppelfunktion besitzt, sowohl als enzymatisches Protein im Energiestoffwechsel als auch als essentieller Regulator mit noch unbekannter Funktion. Neben iftA fiel das Augenmerk auf mehrere Gene, die im Rahmen der Promotion von Neta Altman-Price an der Universität in Tel-Aviv als Histon-Acetylasen und –Deacetylasen identifiziert wurden. Zudem konnte in H. volcanii ein essentielles Gen, welches für ein Histon kodiert, entdeckt werden. Das Histon besitzt konservierte Lysinreste, die im eukaryotischen Histon H3 Ziele für eine posttranslationale Acetylierung sind. Für die weiteren Untersuchungen wurden die Lysinreste irreversibel zum einen in Glutamin (acetylierter Zustand) und zum anderen in Arginin (deacetylierter Zustand) mutiert. Im Rahmen dieser Arbeit wurden die Funktionen und die Einflüsse der beiden Histonmutanten sowie einer Acetylase- (delta pat1) und einer Deacetylase-Deletionsmutante (delta sir2) auf die globale Genregulation analysiert. Dazu wurden zunächst Transkriptomuntersuchungen in der exponentiellen Wachstumsphase mittels Microarray Analysen an den Mutanten im Vergleich zum Wildytp durchgeführt. Die Ergebnisse zeigten einen erheblich stärkeren Einfluß auf die Regulation der Genexpression durch das acetylierte Histon sowie der Deacetylase-Deletionsmutante im Vergleich zu den Ergebnissen des deacetylierten Histons und der Acetylase-Deletionsmutante. In der exponentiellen Wachstumsphase liegt das Histon in H. volcanii daher in überwiegend deacetylierter Form vor. Durch die Analysen konnte auch demonstrieren werden, dass Sir2 und das Histon eine regulatorische Wirkung auf exakt die gleichen Gene ausüben und daher unmittelbar miteinander in Verbindung stehen. Die experimentelle Bestätigung dieses Zusammenhangs stellt im Reich der Archaea bisher ein absolutes Novum dar. Die Untersuchungen des Transkriptoms der delta sir2 Mutante enthüllte zudem einen positiven Einfluß von Sir2 auf ein größeres Gencluster (HVO_1201-25). Die Gene dieses Clusters konnten durch in silico Analysen der Chemotaxis und der Flagellenbiosynthese zugeordnet werden. Für die weitere Charakterisierung der Deletionsmutante wurden daher Untersuchungen zur Bestimmung der Motilität von H. volcanii durchgeführt. Es konnte gezeigt werden, dass die Deletion von Sir2 die Beweglichkeit und gerichtete Fortbewegung von H. volcanii in erheblichem Maße beeinflußte, während die Biosynthese der Flagellen nicht beeinträchtigt war. Die Deacetylierung spielt daher eine unmittelbar Rolle bei der Signaltransduktion und Motilität. Insgesamt konnte durch die Arbeit gezeigt werden, dass die Acetylierung und Deacetylierung von Proteinen durch Pat1 resp. Sir2 die Regulation der Genexpression beeinflußt. Dies geschieht in H. volcanii indirekt durch die posttranslationale Veränderung von internen Signalen oder direkt durch die Modulierung des Histons und die damit verbundene Änderung der DNA-Struktur.
Background: Studies on the development of the nervous system and the musculature of invertebrates have become more sophisticated and numerous within the last decade and have proven to provide new insights into the evolutionary history of organisms. In order to provide new morphogenetic data on opisthobranch gastropods we investigated the neuromuscular development in the nudibranch Aeolidiella stephanieae Valdez, 2005 using immunocytochemistry as well as F-actin labelling in conjunction with confocal laser scanning microscopy (cLSM). Results: The ontogenetic development of Aeolidiella stephanieae can be subdivided into 8 stages, each recognisable by characteristic morphological and behavioural features as well as specific characters of the nervous system and the muscular system, respectively. The larval nervous system of A. stephanieae includes an apical organ, developing central ganglia, and peripheral neurons associated with the velum, foot and posterior, visceral part of the larva. The first serotonergic and FMRFamidergic neural structures appear in the apical organ that exhibits an array of three sensory, flask-shaped and two non-sensory, round neurons, which altogether disappear prior to metamorphosis. The postmetamorphic central nervous system (CNS) becomes concentrated, and the rhinophoral ganglia develop together with the anlage of the future rhinophores whereas oral tentacle ganglia are not found. The myogenesis in A. stephanieae begins with the larval retractor muscle followed by the accessory larval retractor muscle, the velar or prototroch muscles and the pedal retractors that all together degenerate during metamorphosis, and the adult muscle complex forms de novo. Conclusions: Aeolidiella stephanieae comprises features of the larval and postmetamorphic nervous as well as muscular system that represent the ground plan of the Mollusca or even the Trochozoa (e. g. presence of the prototrochal or velar muscle ring). On the one hand, A. stephanieae shows some features shared by all nudibranchs like the postmetamorphic condensation of the CNS, the possession of rhinophoral ganglia and the lack of oral tentacle ganglia as well as the de novo formation of the adult muscle complex. On the other hand, the structure and arrangement of the serotonergic apical organ is similar to other caenogastropod and opisthobranch gastropods supporting their sister group relationship.
