Vascular integrity is essential for organ homeostasis to prevent edema formation and infiltration of inflammatory cells. Long non-coding RNAs (lncRNAs) are important regulators of gene expression and often expressed in a cell type-specific manner. By screening for endothelial-enriched lncRNAs, we identified the undescribed lncRNA NTRAS to control endothelial cell functions. Silencing of NTRAS induces endothelial cell dysfunction in vitro and increases vascular permeability and lethality in mice. Biochemical analysis revealed that NTRAS, through its CA-dinucleotide repeat motif, sequesters the splicing regulator hnRNPL to control alternative splicing of tight junction protein 1 (TJP1; also named zona occludens 1, ZO-1) pre-mRNA. Deletion of the hnRNPL binding motif in mice (Ntras∆CA/∆CA) significantly repressed TJP1 exon 20 usage, favoring expression of the TJP1α- isoform, which augments permeability of the endothelial monolayer. Ntras∆CA/∆CA mice further showed reduced retinal vessel growth and increased vascular permeability and myocarditis. In summary, this study demonstrates that NTRAS is an essential gatekeeper of vascular integrity.
Due to an increasing awareness of the potential hazardousness of air pollutants, new laws, rules and guidelines have recently been implemented globally. In this respect, numerous studies have addressed traffic-related exposure to particulate matter using stationary technology so far. By contrast, only few studies used the advanced technology of mobile exposure analysis. The Mobile Air Quality Study (MAQS) addresses the issue of air pollutant exposure by combining advanced high-granularity spatial-temporal analysis with vehicle-mounted, person-mounted and roadside sensors. The MAQS-platform will be used by international collaborators in order 1) to assess air pollutant exposure in relation to road structure, 2) to assess air pollutant exposure in relation to traffic density, 3) to assess air pollutant exposure in relation to weather conditions, 4) to compare exposure within vehicles between front and back seat (children) positions, and 5) to evaluate "traffic zone"- exposure in relation to non-"traffic zone"-exposure. Primarily, the MAQS-platform will focus on particulate matter. With the establishment of advanced mobile analysis tools, it is planed to extend the analysis to other pollutants including including NO2, SO2, nanoparticles, and ozone.
Encouraging clinical results were reported on a novel cone-in-cone coupling for the fixation of dental implant-supported crowns (Acuris, Dentsply Sirona Implants, Mölndal, Sweden). However, the presence or absence of a microgap and a potential bacterial leakage at the conometric joint has not yet been investigated. A misfit and a resulting gap between the conometric components could potentially serve as a bacterial reservoir that promotes plaque formation, which in turn may lead to inflammation of the peri-implant tissues. Thus, a two-fold study set-up was designed in order to evaluate the bidirectional translocation of bacteria along conometrically seated single crowns. On conometric abutments filled with a culture suspension of anaerobic bacteria, the corresponding titanium nitride-coated (TiN) caps were fixed by friction. Each system was sterilized and immersed in culture medium to provide an optimal environment for microbial growth. Positive and negative controls were prepared. Specimens were stored in an anaerobic workstation, and total and viable bacterial counts were determined. Every 48 h, samples were taken from the reaction tubes to inoculate blood agar plates and to isolate bacterial DNA for quantification using qrt-PCR. In addition, one Acuris test system was subjected to scanning electron microscopy (SEM) to evaluate the precision of fit of the conometric coupling and marginal crown opening. Throughout the observational period of one week, blood agar plates of the specimens showed no viable bacterial growth. qrt-PCR, likewise, yielded a result approaching zero with an amount of about 0.53 × 10−4 µg/mL DNA. While the luting gap/marginal opening between the TiN-cap and the ceramic crown was within the clinically acceptable range, the SEM analysis failed to identify a measurable microgap at the cone-in-cone junction. Within the limits of the in-vitro study it can be concluded that the Acuris conometric interface does not allow for bacterial translocation under non-dynamic loading conditions.
Tagungsbericht zum Symposion an der Universität Potsdam vom 20.-22. Oktober 2000
Endlich ist es vollbracht: Nach verschiedensten juristischen, verlagstechnischen u.a. Peripetien, die Maria Sommer, die Leiterin des Gustav Kiepenheuer Bühnenvertriebs und Inhaberin der deutschen Pirandello-Werkrechte, bei der Präsentation plastisch schilderte, liegt nun die deutsche Pirandello-Ausgabe (Propyläen-Verlag) vollständig in 16 Bänden vor. Allemal ein Anlaß für den Herausgeber Michael Rössner (München), gemeinsam mit Helene Harth (Potsdam) und dem Istituto Italiano di Cultura (Berlin), dort sowie an der Universität Potsdam vom 20.-22. Oktober 2000 ein Symposion zu veranstalten.
In Javier Marias' Roman 'Tadas las almas' (1989), dessen Titel auf den Ort der Handlung, das Oxforder All Souls College, verweist, begrüßt der Portier Will den Ich-Erzähler jeden Morgen mit einem anderen Namen. Der fast neunzigjährige Portier wähnt sich nämlich, von Tag zu Tag wechselnd, in einer anderen Epoche seines langen Lebens und spricht den spanischen Gastprofessor dann jeweils als einen Kollegen aus jener Zeit an. So wird dieser innerhalb einer Woche für Will zu Personen aus den verschiedensten Jahrzehnten. Diese Verwechslungen führen zu dem Schluss, dass die Oxford-Professoren sich zumindest seit dem ersten Weltkrieg nicht verändert haben und letztlich austauschbar sind. Ganz anders verhält es sich mit den fünf Romanen, um die es im Folgenden geht: Sie sind alle in einer Epoche, nämlich in 'unserer postmodernen Moderne' entstanden, lassen sich unter dem Genre des Campusromans rubrizieren, weisen aber gleichzeitig derart eklatante Unterschiede auf, dass zunächst Kriterien bestimmt werden müssen, um einem solchen Vergleich Stringenz zu verleihen. Da der Publikumserfolg der als Beispiel ausgewählten Romane stark variiert - vom internationalen Bestseller über einen Goncourt-Preisträger bis zu einem nur in Erstauflage publizierten Buch - werden zunächst alle fünf in chronologischer Reihenfolge kurz vorgestellt. Es folgen einige Überlegungen zum Genre des Campusromans, um dann die Theorie der Intrige nach Peter von Matt zu applizieren und auf ihre Anwendbarkeit hin zu überprüfen. Abschließend werde ich die Funktion der Intertextualität in den einzelnen Texten untersuchen.
