Background: Alzheimer's disease is a common debilitating dementia with known heritability, for which 20 late onset susceptibility loci have been identified, but more remain to be discovered. This study sought to identify new susceptibility genes, using an alternative gene-wide analytical approach which tests for patterns of association within genes, in the powerful genome-wide association dataset of the International Genomics of Alzheimer's Project Consortium, comprising over 7 m genotypes from 25,580 Alzheimer's cases and 48,466 controls.
Principal findings: In addition to earlier reported genes, we detected genome-wide significant loci on chromosomes 8 (TP53INP1, p = 1.4×10−6) and 14 (IGHV1-67 p = 7.9×10−8) which indexed novel susceptibility loci.
Significance: The additional genes identified in this study, have an array of functions previously implicated in Alzheimer's disease, including aspects of energy metabolism, protein degradation and the immune system and add further weight to these pathways as potential therapeutic targets in Alzheimer's disease.
Wenn es Ziel des literarischen Realismus ist, die Wirklichkeit darzustellen, dann stellt Theodor Storms Novelle 'Der Schimmelreiter' (1888) ein paradigmatisches Beispiel für diese Bewegung dar. Der Text ist nicht nur darin exemplarisch, dass er ein überlegenes Niveau an sozialer und psychologischer Wirklichkeitsnähe erreicht, sondern auch dadurch, dass er die eigentlichen Vorgänge und Begrenzungen, die jeder Darstellungsgeste eigen sind, dramatisch inszeniert. Storms Novelle legt die Sehnsüchte bloß, die Frustrationen und Opfer, die jeden Versuch, vorsprachliche Erfahrungen in sprachliche Form zu verwandeln, zwangsläufig begleiten. Anhand der unvergesslichen Schilderung seines Protagonisten, Hauke Haiens, der nahezu eigenhändig den Dorfbewohnern mehr bewohnbares Land verschafft, macht Storm die überzeitliche, mythische Konfrontation des Menschen mit dem unbändigen Meer zur Allegorie für das realistische Projekt. Vermittels eines allegorischen Schemas wird Hauke Haiens hydrotechnische Arbeit mit Storms literarischen Bemühungen parallelisiert. Diese miteinander verbundenen Aufgaben sind beide geographisch, insofern sie Einschreibungen in die Erde durch technē sind und das im doppelten Sinne des Wortes: als Technik und Kunst. Genauso wie der Deichgraf dem Meer mehr ertragreiches Land für seine Nachbarn entreißt, integriert der Schriftsteller mehr Wirklichkeit für seine Leser; genauso wie das neue Land durch den sorgfältigen Deichbau gesichert wird, ist die Erzählung durch eine Reihe vielschichtiger Rahmen geschützt; und genauso wie die erworbene Siedlung von Überschwemmungen bedroht bleibt, schließt die Geschichte, um beständig zu sein, ein widerspenstiges Element in sich ein, welches das ganze Unternehmen motiviert wie aber auch gefährdet. Dieser letzte Punkt wirkt sich vernichtend auf das realistische Darstellungsprojekt aus, das ja einen Sinngehalt artikulieren will, also eine Artikulation anvisiert, die sich direkt auf das Bild der Küste bezieht, genauer auf die Linie, welche Land und Meer voneinander trennt.
Hamilton vergleicht Freuds Flirt-Kritik mit der platonischen Auffassung von Mimesis. Ihr zufolge kann die Nachahmung nur abgeleitete, mildernde Darstellungen des Todes hervorbringen und so den Glauben an die eigene Unsterblichkeit stützen. Mit Rückgriff auf Roger Caillois' Schriften zur Mimikry lässt sich Mimesis jedoch auch als eine Praxis der Selbst-Verausgabung beschreiben: Wie im Flirt, so setzen wir uns auch in der Mimesis einer Verschwendung aus, in welcher Selbst-Verlust zu einer anderen Form der Gemeinschaft führen kann.
Background: Pythium ultimum (P. ultimum) is a ubiquitous oomycete plant pathogen responsible for a variety of diseases on a broad range of crop and ornamental species. Results: The P. ultimum genome (42.8 Mb) encodes 15,290 genes and has extensive sequence similarity and synteny with related Phytophthora species, including the potato blight pathogen Phytophthora infestans. Whole transcriptome sequencing revealed expression of 86% of genes, with detectable differential expression of suites of genes under abiotic stress and in the presence of a host. The predicted proteome includes a large repertoire of proteins involved in plant pathogen interactions although surprisingly, the P. ultimum genome does not encode any classical RXLR effectors and relatively few Crinkler genes in comparison to related phytopathogenic oomycetes. A lower number of enzymes involved in carbohydrate metabolism were present compared to Phytophthora species, with the notable absence of cutinases, suggesting a significant difference in virulence mechanisms between P. ultimum and more host specific oomycete species. Although we observed a high degree of orthology with Phytophthora genomes, there were novel features of the P. ultimum proteome including an expansion of genes involved in proteolysis and genes unique to Pythium. We identified a small gene family of cadherins, proteins involved in cell adhesion, the first report in a genome outside the metazoans. Conclusions: Access to the P. ultimum genome has revealed not only core pathogenic mechanisms within the oomycetes but also lineage specific genes associated with the alternative virulence and lifestyles found within the pythiaceous lineages compared to the Peronosporaceae.
Teleconnections of the Quasi-Biennial Oscillation in a multi-model ensemble of QBO-resolving models

The Quasi-biennial Oscillation (QBO) dominates the interannual variability of the tropical stratosphere and influences other regions of the atmosphere. The high predictability of the QBO implies that its teleconnections could lead to increased skill of seasonal and decadal forecasts provided the relevant mechanisms are accurately represented in models. Here modelling and sampling uncertainties of QBO teleconnections are examined using a multi-model ensemble of QBO-resolving atmospheric general circulation models that have carried out a set of coordinated experiments as part of the Stratosphere-troposphere Processes And their Role in Climate (SPARC) QBO initiative (QBOi). During Northern Hemisphere winter, the stratospheric polar vortex in most of these models strengthens when the QBO near 50 hPa is westerly and weakens when it is easterly, consistent with, but weaker than, the observed response. These weak responses are likely due to model errors, such as systematically weak QBO amplitudes near 50 hPa, affecting the teleconnection. The teleconnection to the North Atlantic Oscillation is less well captured overall, but of similar strength to the observed signal in the few models that do show it. The models do not show clear evidence of a QBO teleconnection to the Northern Hemisphere Pacific-sector subtropical jet.
