Purpose. Status epilepticus (SE) is characterized by recurrent seizure activity and can be drug-resistant. Knowledge of neuronal and metabolic activity of the brain during SE may be helpful to improve medical care. We here report the effects of three anti-seizure drugs on changes of acetylcholine energy metabolites and oxidative stress during SE. Methods. We used the lithium-pilocarpine model in rats to induce SE and in vivo-microdialysis to monitor cholinergic and metabolic activity in the hippocampus. We measured extracellular concentrations of acetylcholine, glucose, lactate, pyruvate, glycerol and isoprostanes before and during SE, and after acute treatment with pregabalin, valproic acid, and levetiracteam. Results. Upon onset of SE, acetylcholine (ACh) release increased six- to eightfold. Glucose was increased only transiently by 30% but lactate levels rose four-fold, and extracellular concentrations of glycerol ten-fold. Isoprostanes are markers of oxidative stress and increased more than 20-fold. Two hours after pilocarpine adminstration, rats were treated with pregabalin (100 mg/kg), levetiracetam (200 mg/kg) or valproic acid (400 mg/kg) by i.p. injection. All three drugs stopped seizure activity in a delayed fashion, but at the doses indicated, only animals that received levetiracetam reached consciousness. All drugs reduced ACh release within 60-120 minutes. Lactate/pyruvate ratios, glycerol and isoprostanne levels were also reduced significantly after drug administration. Conclusions. Hippocampal ACh release closely follows seizure activity in SE and is attenuated when SE subsides. Pregabalin, valproic acid and levetiracetam all terminate seizures in the rat SE model and attenuate cholinergic and metabolic changes within two hours.
BACKGROUND: Ketone bodies are known to substitute for glucose as brain fuel when glucose availability is low. Ketogenic diets have been described as neuroprotective. Similar data have been reported for triheptanoin, a fatty oil and anaplerotic compound. In this study, we monitored the changes of energy metabolites in liver, blood, and brain after transient brain ischemia to test for ketone body formation induced by experimental stroke.
METHODS AND RESULTS: Mice were fed a standard carbohydrate-rich diet or 2 fat-rich diets, 1 enriched in triheptanoin and 1 in soybean oil. Stroke was induced in mice by middle cerebral artery occlusion for 90 minutes, followed by reperfusion. Mice were sacrificed, and blood plasma and liver and brain homogenates were obtained. In 1 experiment, microdialysis was performed. Metabolites (eg glucose, β-hydroxybutyrate, citrate, succinate) were determined by gas chromatography-mass spectrometry. After 90 minutes of brain ischemia, β-hydroxybutyrate levels were dramatically increased in liver, blood, and brain microdialysate and brain homogenate, but only in mice fed fat-rich diets. Glucose levels were changed in the opposite manner in blood and brain. Reperfusion decreased β-hydroxybutyrate and increased glucose within 60 minutes. Stroke-induced ketogenesis was blocked by propranolol, a β-receptor antagonist. Citrate and succinate were moderately increased by fat-rich diets and unchanged after stroke.
CONCLUSIONS: We conclude that brain ischemia induces the formation of β-hydroxybutyrate (ketogenesis) in the liver and the consumption of β-hydroxybutyrate in the brain. This effect seems to be mediated by β-adrenergic receptors.
Single long-chain omega-3 fatty acids (e.g. docosahexaenoic acid (DHA) or eicosapentaenoic acid (EPA)) are known for their neuroprotective properties associated with ischemic stroke. This pilot study aimed to test the effectiveness of an acute treatment with a long-chain omega-3 lipid emulsion (Omegaven 10%®, OGV) that contains fish oil (DHA 18 mg/ml; EPA 21 mg/ml) and α-tocopherol (0.2 mg/ml) in a transient middle cerebral artery occlusion (MCAO) model of ischemic stroke in mice. For this purpose, female CD-1 mice were anesthetized and subjected to 90 minutes of MCAO. To reflect a clinically relevant situation for an acute treatment, either after induction of stroke or after reperfusion, a single dose of OGV was injected intravenously into the tail vein (5 ml/kg b.w.). A neurological severity score was used to assess motor function and neurological outcome. Stroke-related parameters were determined 24 hours after MCAO. Microdialysis was used to collect samples from extracellular space of the striatum. Mitochondrial function was determined in isolated mitochondria or dissociated brain cells. Inflammation markers were measured in brain homogenate. According to control experiments, neuroprotective effects could be attributed to the long-chain omega-3 content of the emulsion. Intravenous injection of OGV reduced size and severity of stroke, restored mitochondrial function, and prevented excitotoxic glutamate release. Increases of pro-inflammatory markers (COX-2 and IL-6) were attenuated. Neurological severity scoring and neurochemical data demonstrated that acute OGV treatment shortly after induction of stroke was most efficient and able to improve short-term neurological outcome, reflecting the importance of an acute treatment to improve the outcome. Summarising, acute treatment of stroke with a single intravenous dose of OGV provided strong neuroprotective effects and was most effective when given immediately after onset of ischemia. As OGV is an approved fishoil emulsion for parenteral nutrition in humans, our results may provide first translational data for a possible early management of ischemic stroke with administration of OGV to prevent further brain damage.
Im ersten Projekt der vorliegenden Arbeit wurden CD - 1 Mäuse mit drei unterschiedlichen Diäten für zwei Wochen ad libitum gefüttert. Die Diäten bestanden aus zwei kohlenhydratarmen, fettreichen Diäten und einer Standard Haltungsdiät. Die kohlenhydratarmen, fettreichen Diäten enthielten entweder Triheptanoin (dreifach mit Heptanoat verestertes Glycerol) oder Soja - Öl als Fettkomponente (jeweils 35 % der Gesamtkalorien). Nach zwei Wochen wurde ein ischämischer Schlaganfall für 90 min. mithilfe eines Silikonfadens induziert. Die Leber, das Blut und das Gehirn wurden nach dem Schlaganfall entnommen und die Konzentrationen der Metabolite β - Hydroxybutyrat, Glukose, Laktat und Citrat wurden mit der zuvor etablierten GC - MS-Methode ermittelt. Unter gleichen Bedingungen wurde eine Mikrodialysestudie durchgeführt.
Bei den Tieren, die die kohlenhydratarmen, fettreichen Diäten erhielten, konnte in den Leber - und Hirnhomogenaten, im Plasma sowie im Mikrodialysat eine Ketose festgestellt werden. Die BHB Konzentrationen durch eine Soja Diät erreichten im Leberhomogenat bis zu 4 mM, im Plasma bis zu 1,5 mM, im Hirnhomogenat bis zu 1,5 mM und im Mikrodialysat bis zu 30 µM. Um eine Aussage treffen zu können, ob das Gehirn die von der Leber produzierten Ketonkörper als Energiesubstrate nutzen kann, wurde eine Folgestudie (unter gleichen Bedingungen) durchgeführt. Bei dieser Studie wurde der Zeitpunkt der Gewebeentnahme 60 min. nach Entfernen des Fadens (Reperfusion) gewählt. In den Leber – und Hirnhomogenaten konnten erniedrigte Konzentrationen des Ketonkörpers BHB nachgewiesen werden. Die nicht operierten Tiere, die eine fettreiche Diät erhielten, hatten erhöhte Konzentrationen an Citrat in den genannten Geweben. Durch den Abbau des Ketonkörpers BHB können bei Verstoffwechslung in Geweben außerhalb der Leber, zwei Moleküle Acetyl - CoA gebildet werden. Diese gebildeten Acetyl - CoA Moleküle können in den Citratzyklus eingespeist werden.
Um diesen Befund mechanistisch besser verstehen zu können, wurde den Mäusen Propranolol (ein unselektiver β - Blocker) verabreicht, und zwar kurz nachdem der Faden die mittlere Zerebralarterie verschlossen hatte. Als Folge blieb bei den fettreich gefütterten Tieren die zuvor beobachtete Ketose, aus. Daraus wurde geschlossen, dass die auftretende Ketose bei den fettreich gefütterten Tieren durch adrenerge β - Rezeptoren vermittelt wurde. Zusammengefasst kann eine fettreiche bzw. ketogene Ernährung im Falle einer Ischämie die Versorgung des Gehirns durch die Bildung von Ketonkörper gewährleisten.
Die zu beobachtende hepatische Ketogenese aus dem ersten Projekt hat die Frage entstehen lassen, ob eine akute Gabe von β - Hydroxybutyrat (BHB) bei Entfernen des Fadens schützende Effekte auf das Verhalten bzw. die Mitochondrien als Kraftwerke der Zelle hat. Hierzu wurde BHB bei Reperfusion gegeben und die Wirkungen dieser Einmalgabe nach 24 h untersucht. Als erster Schritt wurde der Nachweis erbracht, dass eine exogene Gabe von BHB das Gehirn erreicht. Im zweiten Schritt wurde das Verhalten der Mäuse nach 24 h untersucht. Hierbei erbrachte die Gabe von BHB eine signifikante Verbesserung der sensorischen und motorischen Fähigkeiten der Mäuse. Die metabolischen Veränderungen nach 24 h wurden erneut in Leberhomogenaten und Plasma vermessen. Eine Einzelgabe von BHB bewirkte eine milde Ketose auch 24 h nach Reperfusion der mittleren Zerebralarterie. Um eine detailliertere Erkenntnis über die Wirkung von BHB zu erlangen, wurden die Mitochondrien als potentielles Ziel für BHB in den Fokus genommen. Die Einmalgabe von BHB verhinderte ein Absinken der Komplex – II Aktivität. Außerdem kann die Aktivität der Citratsynthase unter der Gabe von BHB erhalten werden, sodass die Mitochondrien vor allem im wichtigen Zeitraum nach der Reperfusion geschützt werden. Im Rahmen der Untersuchungen der Mitochondrien wurden unterschiedliche Substrateinflüsse auf die Respiration der isolierten Mitochondrien getestet. Bei Zugabe von BHB, Oxalacetat + Acetat oder Citrat zu dem Respirationsmedium stieg die Respiration der Mitochondrien an. Im Falle von Glukose, Propranolol oder Acetat wird die Respiration verringert. Bei Zugabe von Laktat, verbleibt die Respiration auf Ausgangsniveau. Abschließend ist festzustellen, dass die Einzelgabe von BHB nach 24 h das Verhalten der Mäuse verbessert, eine milde Ketose induziert, sowie Mitochondrien und die Citratsynthase gegen ischämische Ereignisse schützt.
Um die in dieser Arbeit gezeigten Daten über metabolische Veränderungen zeigen zu können, musste eine vorherige Etablierung der GC – MS Analytik vollzogen werden. Auf der einen Seite musste die Probenvorbereitung, aber auch die gesamte Vermessung der Proben aufgebaut werden. Es wurden insgesamt 11 Analyte in vier unterschiedlichen Kompartimenten quantifiziert. Die Nachweisgrenze lag bei diesen 11 Analyten bei 0,01 - 1 ng/µl, was einer umgerechneten Stoffmengenkonzentration von 0,5 - 10 µM entspricht. Mithilfe dieser Methode können optional weitere Substanzen aus verschiedenen Geweben zugänglich gemacht werden. Diese Arbeit bietet hierzu eine Anleitung, wie die Etablierung erfolgen kann. Im Rahmen der Probenvorbereitung wurden alle Schritte systematisch verbessert. Dazu wurden Wiederholungsmessungen für unterschiedliche Modalitäten vollzogen. Die Abundance und die Zeitbeständigkeit waren die wesentlichen Beurteilungskriterien. So wurden die Daten für die Extraktionseffektivität, die Lösungsmittelabhängigkeit der Silylierung, der Zusatz von Hünig - Base sowie die Temperatur und Zeitabhängigkeit der Silylierung in dieser Arbeit erarbeitet. Die Quantifizierung wurde anhand von internen Standardverbindungen durchgeführt. Die jeweiligen Response – Faktoren blieben über die gesamte Zeit nach der Etablierung konstant und erlaubten die Quantifizierung mit geringen Fehlern. Die Beurteilung der ermittelten Daten über die Validierung wurden anhand von geltenden Regelwerken der pharmazeutischen Industrie entschieden. Es wurde ein Protokoll entwickelt, das im Rahmen der universitären Forschung eine vertrauenswürdige Aussage über Veränderungen von Metabolitenspiegeln in vielen Geweben der Maus und der Ratte geben kann.
ß-Hydroxybutyrate (BHB) is a ketone body formed in high amounts during lipolysis and fasting. Ketone bodies and the ketogenic diet were suggested as neuroprotective agents in neurodegenerative disease. In the present work, we induced transient ischemia in mouse brain by unilaterally occluding the middle cerebral artery for 90 min. BHB (30 mg/kg), given immediately after reperfusion, significantly improved the neurological score determined after 24 h. In isolated mitochondria from mouse brain, oxygen consumption by the complexes I, II and IV was reduced immediately after ischemia but recovered slowly over 1 week. The single acute BHB administration after reperfusion improved complex I and II activity after 24 h while no significant effects were seen at later time points. After 24 h, plasma and brain BHB concentrations were strongly increased while mitochondrial intermediates (citrate, succinate) were unchanged in brain tissue. Our data suggest that a single administration of BHB may improve mitochondrial respiration for 1–2 days but not for later time points. Endogenous BHB formation seems to complement the effects of exogenous BHB administration.
Harmonic decomposition of two particle angular correlations in Pb–Pb collisions at √sNN=2.76 TeV

Angular correlations between unidentified charged trigger (t) and associated (a) particles are measured by the ALICE experiment in Pb–Pb collisions at √sNN=2.76 TeV for transverse momenta 0.25<pTt,a<15 GeV/c, where pTt>pTa. The shapes of the pair correlation distributions are studied in a variety of collision centrality classes between 0 and 50% of the total hadronic cross section for particles in the pseudorapidity interval |η|<1.0. Distributions in relative azimuth Δϕ≡ϕt−ϕa are analyzed for |Δη|≡|ηt−ηa|>0.8, and are referred to as “long-range correlations”. Fourier components VnΔ≡〈cos(nΔϕ)〉 are extracted from the long-range azimuthal correlation functions. If particle pairs are correlated to one another through their individual correlation to a common symmetry plane, then the pair anisotropy VnΔ(pTt,pTa) is fully described in terms of single-particle anisotropies vn(pT) as VnΔ(pTt,pTa)=vn(pTt)vn(pTa). This expectation is tested for 1⩽n⩽5 by applying a global fit of all VnΔ(pTt,pTa) to obtain the best values vn{GF}(pT). It is found that for 2⩽n⩽5, the fit agrees well with data up to pTa∼3–4 GeV/c, with a trend of increasing deviation as pTt and pTa are increased or as collisions become more peripheral. This suggests that no pair correlation harmonic can be described over the full 0.25<pT<15 GeV/c range using a single vn(pT) curve; such a description is however approximately possible for 2⩽n⩽5 when pTa<4 GeV/c. For the n=1 harmonic, however, a single v1(pT) curve is not obtained even within the reduced range pTa<4 GeV/c.
The ALICE Collaboration has made the first measurement at the LHC of J/ψ photoproduction in ultra-peripheral Pb–Pb collisions at sNN=2.76 TeV. The J/ψ is identified via its dimuon decay in the forward rapidity region with the muon spectrometer for events where the hadronic activity is required to be minimal. The analysis is based on an event sample corresponding to an integrated luminosity of about 55 μb−1. The cross section for coherent J/ψ production in the rapidity interval −3.6<y<−2.6 is measured to be dσJ/ψcoh/dy=1.00±0.18(stat)−0.26+0.24(syst) mb. The result is compared to theoretical models for coherent J/ψ production and found to be in good agreement with those models which include nuclear gluon shadowing.
The ALICE Collaboration reports the measurement of the relative J/ψ yield as a function of charged particle pseudorapidity density dNch/dη in pp collisions at √s=7 TeV at the LHC. J/ψ particles are detected for pt>0, in the rapidity interval |y|<0.9 via decay into e+e−, and in the interval 2.5<y<4.0 via decay into μ+μ− pairs. An approximately linear increase of the J/ψ yields normalized to their event average (dNJ/ψ/dy)/〈dNJ/ψ/dy〉 with (dNch/dη)/〈dNch/dη〉 is observed in both rapidity ranges, where dNch/dη is measured within |η|<1 and pt>0. In the highest multiplicity interval with 〈dNch/dη(bin)〉=24.1, corresponding to four times the minimum bias multiplicity density, an enhancement relative to the minimum bias J/ψ yield by a factor of about 5 at 2.5<y<4 (8 at |y|<0.9) is observed.
A measurement of the multi-strange Ξ− and Ω− baryons and their antiparticles by the ALICE experiment at the CERN Large Hadron Collider (LHC) is presented for inelastic proton–proton collisions at a centre-of-mass energy of 7 TeV. The transverse momentum (pT) distributions were studied at mid-rapidity (|y|<0.5) in the range of 0.6<pT<8.5 GeV/c for Ξ− and Ξ¯+ baryons, and in the range of 0.8<pT<5 GeV/c for Ω− and Ω¯+. Baryons and antibaryons were measured as separate particles and we find that the baryon to antibaryon ratio of both particle species is consistent with unity over the entire range of the measurement. The statistical precision of the current data has allowed us to measure a difference between the mean pT of Ξ− (Ξ¯+) and Ω− (Ω¯+). Particle yields, mean pT, and the spectra in the intermediate pT range are not well described by the PYTHIA Perugia 2011 tune Monte Carlo event generator, which has been tuned to reproduce the early LHC data. The discrepancy is largest for Ω− (Ω¯+). This PYTHIA tune approaches the pT spectra of Ξ− and Ξ¯+ baryons below pT<0.85 GeV/c and describes the Ξ− and Ξ¯+ spectra above pT>6.0 GeV/c. We also illustrate the difference between the experimental data and model by comparing the corresponding ratios of (Ω−+Ω¯+)/(Ξ−+Ξ¯+) as a function of transverse mass.
The ALICE experiment has measured low-mass dimuon production in pp collisions at √s=7 TeV in the dimuon rapidity region 2.5<y<4. The observed dimuon mass spectrum is described as a superposition of resonance decays (η,ρ,ω,η′,ϕ) into muons and semi-leptonic decays of charmed mesons. The measured production cross sections for ω and ϕ are σω(1<pt<5 GeV/c,2.5<y<4)=5.28±0.54(stat)±0.49(syst) mb and σϕ(1<pt<5 GeV/c,2.5<y<4)=0.940±0.084(stat)±0.076(syst) mb. The differential cross sections d2σ/dydpt are extracted as a function of pt for ω and ϕ. The ratio between the ρ and ω cross section is obtained. Results for the ϕ are compared with other measurements at the same energy and with predictions by models.
