Bei der Bekleidungsmodellierung geht es um den Entwurf von Bekleidung von Personen, die beispielsweise in Szenen dargestellt werden können. Dabei stützt sich der Entwurf auf Informationen aus einer Datengrundlage. Die Darstellung von Szenen, in denen Personen dargestellt werden, stellt sich grundsätzlich als Zusammenspiel komplexer Teilaspekte dar. Dabei wird die Nachvollziehbarkeit einer modellierten Szene oder modellierter Avatare im Auge des Betrachters ganz wesentlich durch den Faktor passend gewählter Kleidung bestimmt.
In dieser Arbeit werden Ansätze und Verfahren vorgestellt, die zur Bekleidungsmodellierung auf Grundlage von Textdokumenten basieren. Dafür werden Möglichkeiten erörtert, die es erlauben Informationen aus Texten zu extrahieren und für die Modellierung einzusetzen.
Zur Bearbeitung der Aufgabenstellung wird zunächst ein aus dem Machine Learning bekanntes kontextuelles Modell hinsichtlich einer Mehrklassen-Klassifizierung trainiert und angewendet. Daraufhin wird die Erstellung einer eigenen Wissensressource, die sich auf textlicher Ebene mit dem Thema der Bekleidung auseinandersetzt, aufgebaut und mit zahlreichen Informationen aus bereits bestehenden Ressourcen popularisiert. Die neue Ressource wird in Form einer Graphdatenbank entworfen. Dabei werden Relationen zwischen den einzelnen Elementen mithilfe von statischen Modellen sowie einem kontextuellen Modell, dem BERT-Modell, erstellt. Schließlich wird auf Grundlage der entwickelten Graphdatenbank ein in der Programmiersprache Python entwickeltes Programm vorgestellt, dass Eingabetexte unter Hinzunahme der Informationen und Relationen innerhalb der Graphdatenbank verarbeitet und Kleidungsstücke detektiert.
Nach der theoretischen Aufarbeitung der entwickelten Ansätze werden die daraus resultierenden Ergebnisse diskutiert und bestehende Problematiken bei der Bearbeitung der Aufgabenstellung angesprochen. Abschließend wird die Arbeit zusammengefasst und Anregungen für die weitere Bearbeitung dieser Thematik vorgestellt.

Hereditary angioedema (HAE) in children and adolescents : a consensus on therapeutic strategies

Hereditary angioedema due to C1 inhibitor (C1 esterase inhibitor) deficiency (types I and II HAE-C1-INH) is a rare disease that usually presents during childhood or adolescence with intermittent episodes of potentially life-threatening angioedema. Diagnosis as early as possible is important to avoid ineffective therapies and to properly treat swelling attacks. At a consensus meeting in June 2011, pediatricians and dermatologists from Germany, Austria, and Switzerland reviewed the currently available literature, including published international consensus recommendations for HAE therapy across all age groups. Published recommendations cannot be unconditionally adopted for pediatric patients in German-speaking countries given the current approval status of HAE drugs. This article provides an overview and discusses drugs available for HAE therapy, their approval status, and study results obtained in adult and pediatric patients. Recommendations for developing appropriate treatment strategies in the management of HAE in pediatric patients in German-speaking countries are provided.Conclusion Currently, plasma-derived C1 inhibitor concentrate is considered the best available option for the treatment of acute HAE-C1-INH attacks in pediatric patients in German-speaking countries, as well as for short-term and long-term prophylaxis.
Although vaccination campaigns are currently being rolled out to prevent coronavirus disease (COVID-19), antivirals will remain an important adjunct to vaccination. Antivirals against coronaviruses do not exist, hence global drug repurposing efforts have been carried out to identify agents that may provide clinical benefit to patients with COVID-19. Itraconazole, an antifungal agent, has been reported to have activity against animal coronaviruses. Using cell-based phenotypic assays, the in vitro antiviral activity of itraconazole and 17-OH itraconazole was assessed against clinical isolates from a German and Belgian patient infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Itraconazole demonstrated antiviral activity in human Caco-2 cells (EC50 = 2.3 µM; 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay). Similarly, its primary metabolite, 17-OH itraconazole, showed inhibition of SARS-CoV-2 activity (EC50 = 3.6 µM). Remdesivir inhibited viral replication with an EC50 = 0.4 µM. Itraconazole and 17-OH itraconazole resulted in a viral yield reduction in vitro of approximately 2-log10 and approximately 1-log10, as measured in both Caco-2 cells and VeroE6-eGFP cells, respectively. The viral yield reduction brought about by remdesivir or GS-441524 (parent nucleoside of the antiviral prodrug remdesivir; positive control) was more pronounced, with an approximately 3-log10 drop and >4-log10 drop in Caco-2 cells and VeroE6-eGFP cells, respectively. Itraconazole and 17-OH itraconazole exert in vitro low micromolar activity against SARS-CoV-2. Despite the in vitro antiviral activity, itraconazole did not result in a beneficial effect in hospitalized COVID-19 patients in a clinical study (EudraCT Number: 2020-001243-15).

Wir haben ihm vorab einige Fragen gestellt – seine mitunter forschen Antworten deuten jedenfalls an, dass der Autor sein Publikum bestimmt nicht langweilen wird.
Für die anstehende Runde der Exzellenzstrategie des Bundes und der Länder bewirbt sich die Goethe-Universität Frankfurt mit vier neuen Clustern zu den Forschungsthemen Vertrauen im Konflikt (CONTRUST), Infektion und Entzündung (EMTHERA), Ursprung der Schweren Elemente (ELEMENTS) und zelluläre Architekturen (SCALE). Die Anträge vereinen die Kompetenzen und zukunftsweisenden Ideen der Goethe-Universität mit denen der Kolleg:innen des Verbunds der Rhein-Main-Universitäten (RMU) und weiterer Partner der vier großen Organisationen der außeruniversitären Forschung. Der seit 2019 bestehende Exzellenzcluster Cardiopulmonary Institute (CPI) wird im kommenden Jahr direkt einen Vollantrag einreichen.
Although vaccines are currently used to control the coronavirus disease 2019 (COVID-19) pandemic, treatment options are urgently needed for those who cannot be vaccinated and for future outbreaks involving new severe acute respiratory syndrome coronavirus virus 2 (SARS-CoV-2) strains or coronaviruses not covered by current vaccines. Thus far, few existing antivirals are known to be effective against SARS-CoV-2 and clinically successful against COVID-19.
As part of an immediate response to the COVID-19 pandemic, a high-throughput, high content imaging–based SARS-CoV-2 infection assay was developed in VeroE6-eGFP cells and was used to screen a library of 5676 compounds that passed phase 1 clinical trials. Eight candidates (nelfinavir, RG-12915, itraconazole, chloroquine, hydroxychloroquine, sematilide, remdesivir, and doxorubicin) with in vitro anti–SARS-CoV-2 activity in VeroE6-eGFP and/or Caco-2 cell lines were identified. However, apart from remdesivir, toxicity and pharmacokinetic data did not support further clinical development of these compounds for COVID-19 treatment.
