Remodeling of extracellular matrix (ECM) is an important physiologic feature of normal growth and development. In addition to this critical function in physiology many diseases have been associated with an imbalance of ECM synthesis and degradation. In the kidney, dysregulation of ECM turnover can lead to interstitial fibrosis, and glomerulosclerosis. The major physiologic regulators of ECM degradation in the glomerulus are the large family of zinc-dependent proteases, collectively refered to matrix metalloproteinases (MMPs). The tight regulation of most of these proteases is accomplished by different mechanisms, including the regulation of MMP gene expression, the processing and conversion of the inactive zymogen by other proteases such as serine proteases and finally the inhibition of active MMPs by endogenous inhibitors of MMPs, denoted as tissue inhibitors of metalloproteinases (TIMPs). Namely, the MMP-9 has been shown to be critically involved in the dysregulation of ECM turnover associated with severe pathologic conditions such as rheumatoid arthritis or fibrosis of lung, skin and kidney. In the present work I searched for a possible modulation of MMP-9 expression and/or activity in glomerular mesangial cells which are thought as key players of many inflammatory and non-inflammatory glomerular diseases. I found that various structurally different PPARalpha agonists such as WY-14,643, LY-171883 and fibrates potently suppress the cytokine-induced MMP-9 expression in renal MC. Furthermore, I demonstrate that the inhibition of MMP-9 expression by PPARalpha agonists was paralleled by a strong increase of cytokine-induced iNOS expression and subsequent NO formation, suggesting that PPARalpha-dependent effects on MMP-9 expression level primarily result from alterations in NO production which in turn reduces the MMP-9 mRNA half-life. Searching for the detailed mechanism of NO-dependent effects on MMP-9 mRNA stability, I found that NO either given from exogenous sources or endogenously produced increases the MMP-9 mRNA degradation by decreasing the expression of the mRNA stabilizing factor HuR. Furthermore, I demonstrate a reduction in the RNA-binding capacity of HuR containing complexes to MMP-9 ARE motifs in cells treated with NO. Since the reduction of HuR expression can be mimicked by the cGMP analog 8-Bromo-cGMP, I suggest that NO reduces in a cGMP-dependent manner the expression of HuR. Finally, I elucidated the modulatory effect of extracellular nucleotides, mainly ATP, on cytokine-triggered MMP-9 expression. Interestingly, I found that in contrast to NO, gamma-S-ATP the stable analog of ATP potently amplifies the IL-beta mediated MMP-9 expression. The increase in mRNA stability was paralleled by an increase in the nuclear-cytosolic shuttling of the mRNA stabilizing factor HuR. Furthermore, I demonstrate an increase in the RNA-binding capacity of HuR containing complexes to the 3'-UTR of MMP-9 by ATP. In summary, the data presented here may help to find new targets (posttranscriptional regulation) that could be used to manipulate or modulate the expression of not only MMP-9 but also other genes regulated on the level of mRNA stability.
Reaktive Sauerstoffspezies (ROS) sind von großer Bedeutung für die Regulation des vaskulären Tonus und die intrazelluläre Signaltransduktion. Die ROS-produzierenden Enzyme der Gefäßwand sind bisher nur unzureichend charakterisiert. Wir untersuchten, ob eine ähnliche NADPH Oxidase wie in Leukozyten, mit den Untereinheiten p22phox und gp91phox auch in den Endothel- und glatten Muskelzellen der Gefäßwand existiert. Phorbol 12-myristate 13-acetat (PMA) aktiviert die leukozytäre NADPH Oxidase und steigert in endothelintakten Aortenringen die ROS-Produktion. PMA steigert auch die Radikalproduktion in Endothelzellen (EC); jedoch nicht in glatten Muskelzellen (SMC) und denudierten Aortenringen. ECs und SMCs exprimieren p22phox, während gp91phox nur in ECs nachgewiesen werden konnte. Mox1, ein Homolog von gp91phox konnte im Gegensatz dazu nur in SMCs und nicht in ECs gefunden werden. Die funktionelle Relevanz von gp91phox wurde mittels vergleichender Experimente mit Aorten-Segmenten von Wildtyp-, gp91phox-/--und eNOS-/--Mäusen analysiert. Die PMA-induzierte ROS-Produktion der Aortenring von Wildtyp und eNOS-/--Mäusen war ähnlich stark, während sie in den Aortenringen von gp91phox-/--Mäusen ausblieb. Der Radikalfänger Tiron verstärkte die endothelabhängige Relaxation in Aortensegmenten von Wildtyp-, jedoch nicht von gp91phox-/--Mäusen. Unsere Daten zeigen, dass ECs, ähnlich wie Leukozyten, eine gp91phox enthaltende NADPH Oxidase exprimieren. Dieses Enzym ist eine Hauptquelle arterieller ROS und beeinflusst die Bioverfügbarkeit von endothelialem NO.
IL-18, a recently identified member of IL-1 family, is now recognized as an important regulator of innate and acquired immune responses. Therefore, the antitumor activities of IL-18 have been investigated. IL-18 has been shown to induce IFN-γ production by T, B, and NK cells, enhances NK cell activity, activates Fas ligandmediated apoptosis of the tumor cells, and improves the overall antitumor immunity. KG-1 cells were derived from a patient with acute myeloid leukemia (AML). IL-18 has been shown to induce IFN-γ production in those leukemic cells. TLR-3, in addition to its ability to recognize viral double stranded RNA, also can recognize the synthetic analogue poly(I:C) and induces type I IFN, inflammatory cytokine production, e.g TNF-α, and maturation of denderitic cells. In the present work the potential modulatory effect of PIC on IFN-γ and TNF-α production by KG-1 cells treated with IL-18 was investigated. Indeed, PIC strongly amplified the production of IFN-γ induced by IL-18 on mRNA and protein levels via NF-κB as well as p38 and JNK MAPK activation. Compared to IFN-γ, TNF-α showed different behaviour in KG-1 cells. On mRNA level I found only weak induction of TNF-α by IL-18 which was potentiated in the presence of PIC. Similarly, the release of TNF-α by IL-18 plus PIC required NF-κB as well as p38 and JNK MAPK activation. Furthermore, in the present work I found that TLR-3 is required for IFN-γ and TNF-α production. In addition, it is demonstrated by immunofluoresence that TLR-3 is localized in cytoplasm but not on the cell surface in KG-1 cells. Recently, it has been demonstrated that IFN-γ shows therapeutic potential as detected in AML blasts, specifically via inhibition of proliferation and induction of apoptosis. Thus our data could serve as a rationale for the clinical use of PIC and IL-18 in combination therapy. In search for new cytokines potentially modulated by the combination IL-18 plus PIC in KG-1 cells, cytokine antibody array analysis was performed. I found an upregulation of expected genes like IP-10 but most interestingly unexpected upregulation of PDGF-AA. Searching for detailed mechanisms of PDGF-AA induction, I found that neither p38 nor JNK is involved in PDGF-AA production but NF-κB is essential for the expression of PDGF-AA. Furthermore, I found that PDGF-AA is not able to increase the proliferation of KG-1 cells. PDGF and TGF-β are examples of signaling molecules which control the growth, survival, motility, and differentiation of cells. Therefore, the release of TGF-β by IL-18 plus PIC was monitored by ELISA. The level of TGF-β in cellular supernatants revealed that neither PIC nor IL-18 was able to significantly mediate release of TGF-β indicating that only PDGF-AA but not TGF-β is induced by PIC and IL-18 in KG-1 cells. To the best of our knowledge this is the first time that IL-18 or PIC is shown to induce the expression of PDGF-AA in KG-1 cells.
Seit einigen Jahren ist bekannt, dass Sphingolipide nicht nur eine strukturgebende Funktion in der Plasmamembran aufweisen, sondern ebenfalls als Botenstoffe intra- und extrazellulär aktiv sind. Sphingosin-1-Phosphat (S1P) bildet dabei einen Schlüssel-Metaboliten, da es verschiedene Zellfunktionen wie Wachstum und Zelltod beeinflusst. Es wird durch zwei Isoformen der Sphingosinkinasen, SK1 und SK2, gebildet. Die SK1 wurde bereits gut untersucht und es konnte gezeigt werden, dass sie eine wichtige Rolle beim Zellwachstum einnimmt und einen entscheidender Regulator bei inflammatorischen Erkrankungen und Krebs darstellt. Über die SK2 ist soweit wenig bekannt und die Ergebnisse sind zum Teil kontrovers. Sowohl pro-proliferative als auch anti-proliferative Funktionen der SK2 wurden beschrieben. Andererseits handelt meine Arbeit von Nierenfibrose, da beschrieben wurde, dass Sphingolipide einen wichtigen Einfluss auf die Entwicklung chronischer Nierenerkrankungen nehmen. Nierenfibrose stellt das Endstadium chronischer Nierenerkrankungen dar und führt zu einer Akkumulation der Extrazellulärmatrix, Organvernarbung und zum Verlust der Nierenfunktion. Die SK1 spielt dabei eine protektive Rolle bei der Entstehung von Nierenfibrose. Deshalb sollte in dieser Arbeit die Rolle der Sk2 bei der Entstehung von Nierenfibrose untersucht werden.
Im ersten Teil meiner Arbeit wurde das Mausmodell der unilateralen Ureterobstruktion (UUO) verwendet, welches zur Entwicklung einer tubulointerstitiellen Nephritits und nachfolgender Fibrose führt. Es konnte dabei gezeigt werden, dass sowohl die Protein-Expression als auch die Aktivität der SK2 im fibrotischen Nierengewebe gesteigert wurden. Allgemein wiesen die SK2-/--Mäuse eine verminderte Fibrose in Folge des UUO auf im Vergleich zu den Wildtyp-Mäusen. Dies wurde bestätigt durch eine reduzierte Kollagenakkumulation, sowie eine verminderte Protein-Expression von Fibronektin-1, Kollagen-1, α-smooth muscle actin, connective tissue growth factor (CTGF) und Plasminogen-Aktivator-Inhibitor1 (PAI-1). Diese Effekte gingen einher mit einer gesteigerten Protein-Expression des inhibitorischen Smad7 und erhöhten Sphingosin-Spiegeln in SK2-/--UUO-Nieren. Auf mechanistischer Ebene vermindern die erhöhten Sphingosin-Spiegel die durch transforming growth factor-β (TGFβ) induzierte Kollagenakkumulation, die PAI-1- und CTGF-Expression, aber induzieren die Smad7-Expression in primären Nierenfibroblasten. In einem komplementären Versuch mit hSK2 tg-Mäusen wurde eine verstärkte Entstehung von Nierenfibrose mit erhöhter Kollagenakkumulation, sowie erhöhte Protein-Expressionen von Fibronektin-1, Kollagen-1, α-smooth muscle actin, CTGF und PAI-1 festgestellt. Die Smad7-Expression dagegen war vermindert.
Im zweiten Teil meiner Arbeit stand der glomeruläre Teil der Niere im Fokus und es wurde untersucht, ob die Überexpression der SK2 zu einer phänotypischen Veränderung der glomerulären Mesangiumzellen führt. Mesangiumzellen wurden dazu aus den hSK2 tg-Mäuse isoliert und charakterisiert. Es konnte gezeigt werden, dass hSK2 und mSK2 in den transgenen Zellen hauptsächlich in der zytosolischen Fraktion lokalisiert sind, während S1P ausschließlich im Kern akkumulierte. Weiterhin konnte eine verminderte Proliferation unter normalen Wachstumsbedingungen der hSK2 tg-Zellen im Vergleich zu den Kontrollzellen beobachtet werden. Die Zellen reagierten auch sensitiver auf Stress-induzierte Apoptose. Auf molekularer Ebene konnte dies durch eine reduzierte ERK- und Akt/PKB-Aktivierung erklärt werden. Nach Staurosporin-Behandlung wurde Apoptose durch den intrinsischen, mitochondrialen Apoptosesignalweg induziert. Dabei konnte eine reduzierte anti-apoptotische Bcl-xL-Expression und vermehrte Prozessierung von Caspase-9 und Caspase-3 und PARP beobachtet werden.
Zusammenfassend konnte in dieser Arbeit gezeigt werden, dass eine verminderte tubulointerstitielle Fibrose-Entstehung durch die Deletion der SK2, sowie anti-proliferative und Apoptose-induzierende Effekte durch die SK2 in Mesangiumzellen nachgewiesen werden konnten. Somit könnten SK2-Inhibitoren die Entstehung tubulointerstitieller Fibrose und mit Proliferation assoziierte Erkrankungen wie mesangioproliferative Glomerulonephritis positiv beeinflussen.
Interleukin-18-Bindungsprotein (IL-18BP) ist ein erst kürzlich entdeckter Gegenspieler von Interleukin-18 (IL-18). Aufgrund der Eigenschaft von IL-18BP mit hoher Affinität an IL-18 zu binden, wird IL-18 neutralisiert und seine biologischen Wirkungen durch IL-18BP inhibiert. Das Zytokin IL-18 ist ein multifunktioneller Botenstoff des Immunsystems, dessen Aktivität bei der Entstehung von Entzündungen, der Abwehr von Infektionen und der Rückbildung von Tumoren beteiligt sein kann. Eine der bedeutendsten Wirkungen von IL-18 ist insbesondere seine Fähigkeit die Produktion und Freisetzung von Interferon-gamma durch T-Helfer Typ 1 (Th1) Zellen, Natürliche Killer (NK) Zellen und CD8+ zytotoxische Zellen auszulösen. Bislang war lediglich bekannt, dass es sich bei IL-18BP um ein konstitutiv exprimiertes und sezerniertes Protein handelt. Die Zielsetzung dieser Promotionsarbeit war es zu untersuchen, ob eine Regulation der Genexpression von IL-18BP in Nicht-Immunzellen stattfindet. Dazu wurde im ersten Schritt eine semiquantitative RT-PCR Methode etabliert, mit Hilfe derer eine schwache konstitutive Expression der IL-18BP mRNA in Zellkulturen von humanen renalen Mesangiumzellen, epithelialen DLD-1 Kolonkarzinomzellen und Fibroblasten nachgewiesen wurde. Im Folgenden konnte als wesentliches Ergebnis festgestellt werden, dass eine Induktion der Genexpression von IL-18BP durch Interferon-gamma erfolgt. Mit RNase Protection Assays wurden nach Interferon-gamma Exposition 20 – 30fache relative Steigerungen der IL-18BP mRNA detektiert. In humanen Mesangiumzellen führte außerdem bakterielles Lipopolysaccharid zum Anstieg der IL-18BP Genexpression. Im zweiten Teil der Untersuchungen ließ sich unter Verwendung eines eigens hergestellten polyklonalen Antiserums nachweisen, dass durch Interferon-gamma auch eine starke Vervielfachung der Freisetzung bzw. Sekretion von IL-18BP stattfindet. Weiterhin wurden Kokulturen von IL-12/IL18 aktivierten humanen mononukleären Zellen aus dem peripheren Blut (PBMCs) mit entweder Mesangiumzellen oder DLD-1 Zellen durchgeführt. In diesen Kokulturen bewirkte die mittels ELISA gemessene Freisetzung von endogenem Interferon-gamma durch die PBMCs ebenfalls eine Induktion der Genexpression von IL-18BP in den Mesangiumzellen und DLD-1 Zellen. Darüber hinaus wurde in anderen Experimenten untersucht, ob die Regulation von IL-18BP gleichzeitig von Änderungen im Gehalt an IL-18 begleitet wird. Während in den humanen Mesangiumzellen kein IL-18 exprimiert wurde, konnte in den DLD-1 Zellen konstitutives proIL-18 detektiert werden. Jedoch hatte Interferon-gamma in DLD-1 Zellen keinen Einfluss auf die IL-18 Expression. Die hier zusammengetragenen Resultate belegen zum ersten Mal, dass es sich bei IL-18BP nicht nur um ein konstitutiv exprimiertes Protein, sondern vielmehr um einen spezifisch regulierten Immunmodulator handelt. Die Induktion der Freisetzung von IL-18BP durch Interferon-gamma stellt den entscheidenden Schritt eines bislang unbekannten negativen Rückkopplungsmechanismus zwischen Immunzellen und ortsständigen Nicht-Immunzellen dar: Nach der Freisetzung von IL-18 bei Entzündungen, Infektionen und Tumorerkrankungen führt das von Th1-, NK- und CD8+-Zellen produzierte Interferon-gamma zu einer Sekretion von IL-18BP durch Nicht-Immunzellen. Infolgedessen kommt es konsekutiv zur Limitierung der Aktivität von IL-18 mit Reduzierung seiner proinflammatorischen Wirkungen. Da ein Übermaß an IL-18 bei der Pathogenese von chronisch entzündlichen Erkrankungen wie beispielsweise der Rheumatoiden Arthritis und dem M. Chron eine Rolle zu spielen scheint, ist von besonderem Interesse welche natürlichen Wege für die Blockierung von IL-18 existieren. Die therapeutische Applikation von IL-18BP könnte sich in Zukunft als eine neue Strategie zur erfolgreichen Behandlung dieser Krankheiten erweisen.
Vorhofflimmern entsteht unter anderem durch ein Ungleichgewicht zwischen sympathischem und parasympathischem autonomen Nervensystem. Sowohl invasive Ansätze via Ablation oder Denervierung als auch minimalinvasive Ansätze via Elektrostimulation liefern erfolgsversprechende Ergebnisse in Bezug auf Beeinflussung eines bestehenden Vorhofflimmerns.
Interessanterweise konnte durch Akupunktur des Tragus eine Reduktion von Vorhofflimmer-Rezidiven nach Kardioversion erreicht werden. Ziel dieser Studie war die Evaluation von Akupunktur-induzierten kardialen (vegetativen) Effekten bei gesunden Probanden.
Herzgesunde männliche Probanden (n=24) wurden im Bereich des Vagus-Innervationsgebietes an der Concha inferior des Ohres (Herzpunkt 100, OHR) sowie an einem klassischen Akupunktur-Punkt des Herzens (ventralen Unterarm, P6/Neiguan, ARM) akupunktiert. Eine „Placebo“-Akupunktur eines Punktes, der gegen Gonarthrose-Schmerzen helfen soll (Magenpunkt 35), diente als Kontrolle. Die Hälfte der Probanden erhielt eine zusätzliche Messung ohne Akupunktur, um Effekte durch das alleinige Platzieren einer Nadel zu untersuchen.
Um eine Aktivierung des ANS zu bewirken und Veränderungen quantifizieren zu können, wurden 12-Kanal Langzeit-EKG Messungen wie folgt durchgeführt: 30 Minuten (min) in liegender Position unter Akupunktur, Entfernung der Akupunkturnadel, 5 min liegend, 5 min sitzend, 5 min stehend, 5 min sitzend.
Es erfolgten Analysen folgender Parameter der Herzfrequenzvariabilität: Herzfrequenz, SDNN, RMSSD, HF, LF, LF/HF. Des Weiteren wurden Periodic Dynamics Repolarization und Deceleration Capacity untersucht.
Verglichen mit Placebo-Akupunktur wurde sowohl durch Akupunktur am OHR als auch am ARM die Herzfrequenz in liegender und sitzender Körperpositionen signifikant gesenkt (Phasen 30L, 5S1, 5S2, ∆5L-5S1, ∆5S1-5ST).
Während die Akupunktur am OHR vor allem die SDNN signifikant erhöhte, führte die Akupunktur am ARM zu einer signifikanten Steigerung von RMSSD und dem parasympathischen Leistungsdichtespektrum-Parameter HF. Der Quotient LF/HF wurde ebenfalls signifikant gesenkt.
Kein Unterschied bestand zwischen Placebo-Akupunktur und Messung ohne Akupunktur, sodass postuliert werden kann, dass das alleinige Platzieren einer Nadel keinen nennenswerten Effekt auf das autonome Nervensystem besitzt.
Die Ergebnisse der Studie zeigen, dass eine Akupunktur an Orten, die mit dem autonomen Nervensystem verschaltet sind, zu einer Modulation des parasympathischen kardialen autonomen Nervensystems führen kann.
Eine Modulation der parasympathisch-sympathischen Balance durch minimalinvasive Zusatzbehandlung, wie in unserer Studie, könnte möglicherweise bei Patienten mit Vorhofflimmern zusätzlich zu medikamentöser oder nach invasiver Behandlung wie der Pulmonalvenenisolation das Therapieresultat verbessern und die Rezidivrate verringern.
Es ist bekannt, dass die Aktivierung von S1P-Rezeptoren die Expression von profibrotischen Mediatoren, wie dem Bindegewebswachstumsfaktor CTGF, induzieren und deshalb auch eine Rolle bei der Entstehung der Nierenfibrose spielen kann. In diesem Kontext konnte unsere Arbeitsgruppe zeigen, dass die Aktivierung von S1P5 zur TGF-β2-induzierten CTGF-Expression in humanen glomerulären Mesangiumzellen beiträgt (Wünsche et al. 2015). Im Rahmen dieser Doktorarbeit wurde deshalb die Rolle von S1P5 in einem in vivo-Modell zur Nierenfibrose untersucht. Männliche S1P5-/--Mäuse und Wildtypmäuse mit C57BL/6J-Hintergrund wurden mit einer adeninreichen Diät für jeweils 7 und 14 Tage gefüttert, um eine tubulointerstitielle Fibrose hervorzurufen. Die Nieren von unbehandelten Mäusen des jeweiligen Genotyps dienten als Kontrolle. Die Ergebnisse zeigen, dass S1P5-/--Mäuse geringere Kreatininplasmaspiegel und weniger Schäden im Nierengewebe gegenüber Wildtypen zeigten. Darüber hinaus wurde festgestellt, dass die mRNA-Expression von mehreren Fibrosemarkern und proinflammatorischen Zytokinen in den S1P5-/--Mäusen schwächer war als in den Wildtypen. Die Auswertung von histochemischen Färbungen und Western Blots bestätigte diese Beobachtung. Zusammengefasst kann festgehalten werden, dass S1P5 eine wichtige Rolle bei der Entstehung von Adenin-induzierter Entzündung in der Niere und nachfolgender Pathogenese wie Gewebeschäden und Fibrose spielt.
Ceramide sind ein Bestandteil der Lipiddoppelschicht, in allen eukaryotischen Zellen vorhanden und zentrale Moleküle des Sphingolipidstoffwechsels. Die Synthese und der Abbau der Ceramide werden von vielen verschiedenen Enzymen reguliert. Neben ihrer Aufgabe als strukturelle Elemente der Zellmembranen wurde herausgefunden, dass Ceramide auch in verschiedenen Signalwegen involviert sind, die auch bei Nierenerkrankungen eine Rolle spielen. In Bezug auf die Kettenlänge der angehängten Fettsäure können so genannte kurz- und langkettige Ceramide Apoptose induzieren, wohingegen sehr langkettige Ceramide Zellproliferation fördern. In mehreren Studien wurden bereits Konzentrationsänderungen von kettenlängenspezifischen Ceramiden im Plasma und Serum von Patienten gemessen, die zu diesem Zeitpunkt an einer Nierenerkrankung litten. In dieser Arbeit wurde daher untersucht, ob solche Konzentrationsänderungen auch im Nierengewebe von Patienten und Mäusen mit einer Nierenfibrose, dem Kennzeichen nahezu aller chronischen Nierenerkrankungen, zu sehen sind. Zu diesem Zwecke wurden Biopsien der Nierenrinde und des Nierenmarks von fibrotischen Nieren aus Patienten, die an Hydronephrose und/oder Pyelonephritis litten, und von gesunden Gewebeproben untersucht. Letztere wurden durch Nephrektomien zur Behandlung von Nierenkarzinomen gewonnen und dienten als nichtfibrotische Kontrolle. Zum Vergleich mit fibrotischen Nieren aus Mäusen wurden männliche Mäuse der Linie C57BL/6J mit einer adeninreichen Diät für 14 Tage gefüttert. Die Konzentrationen der Sphingolipide wurden mittels Massenspektrometrie gemessen und die Level der fibrotischen Marker wurden mit Hilfe von RT-qPCR und histologischen Färbungen analysiert. Die Ergebnisse zeigen, dass die sehr langkettigen Ceramide Cer d18:1/24:0 und Cer d18:1/24:1 sowohl in fibrotischen Nierenrindenproben der Patienten als auch in fibrotischen Nieren der Mäuse im Vergleich zu den jeweiligen Kontrollproben signifikant geringer konzentriert waren. Diese Effekte korrelieren mit der Hochregulation der Fibrosemarker COL1α1, COL3α1 und αSMA in den fibrotischen Nieren. Es konnte gezeigt werden, dass nur bestimmte Ceramide in fibrotischem Nierengewebe in ihrer Konzentration verändert sind, was interessante Fragen hinsichtlich der Ursache dieser Veränderungen, ihrer funktionellen Aufgabe und zu möglichen Effekten einer Manipulation des Ceramidstoffwechsels mit dem Ziel der Behandlung der Nierenfibrose oder der Entdeckung neuer Biomarker aufwirft. Die hier präsentierten Ergebnisse zeigen zudem die Eignung von in vivo-Mausmodellen als translationalen Ansatz für das Verständnis der Beteiligung von Ceramiden in menschlichen Nierenerkrankungen.
Die in vitro-Untersuchungen zu der Rolle des S1P-Transporters Spns2 haben gezeigt, dass Spns2 die Expression von CTGF nach Stimulation von Mausmesangiumzellen mit TFG-β2 verstärkt. 24 Stunden nach Stimulation war im Zellkulturüberstand von Spns2-/--mMC im Gegensatz zu Spns2+/+-mMC keine Akkumulation des pro-fibrotischen Zytokins CTGF gegenüber der unstimulierten Kontrolle detektierbar. Nach 48 Stunden Stimulation mit TFG-β2 war die Menge an CTGF im Zelllysat als auch im Zellkulturüberstand von Spns2-/--mMC genauso hoch wie in der unstimulierten Kontrolle. Im Zelllysat und im Überstand der Spns2-/--mMC war die Expression von CTGF weiterhin deutlich höher im Vergleich zu den Proben der unstimulierten Zellen. Die basale und TFG-β2-induzierte Genexpression von S1P-synthetisierenden und S1P-degradierenden Enzymen, sowie die Konzentrationen an S1P und Sphingosin in den Zellen unterschieden sich zwischen Spns2-/--mMC und Spns2+/+-mMC nicht.

Caspase-2 is the evolutionary most conserved member of the caspase family and was shown to be involved in genotoxic stress induced apoptosis, control of aneuploidy, and ageing related metabolic changes. However, its role in apoptosis seems redundant due to the observation, that knockout does not inhibit apoptotic signalling exclusively. Instead, knockout of caspase-2 leads to tumor susceptibility in vivo, which led to the assumption, that caspase-2 has non-apoptotic functions and can act as a tumor suppressor. The underlying mechanism of the tumor suppressor activity of caspase-2 has not been clarified so far. Furthermore, caspase-2, has a prominent, and as pro-enzyme exclusive localisation in the nucleus and other subcellular compartments, implicating a distinct and location specific role.
In this study, a novel caspase-2 specific substrate, termed p54nrb, was identified. P54nrb is harbouring a caspase-2 specific cleavage site at the aspartate residue D422, and cleavage of p54nrb leads apparently to disruption of its putative DNA binding domain at the C-terminus.
P54nrb is a nuclear multifunctional RNA and DNA binding protein, known for roles in transcriptional regulation, DNA unwinding and repair, RNA splicing, and retention of defective RNA. Overexpression of p54nrb has been observed in several human cancers, such as cervix carcinoma, melanoma, and colon carcinoma.
Data from this study revealed, that depletion of p54nrb in tumor cell lines results in a loss of resistance to drug induced cell death and to reduced capability of anchorage independent growth, which is functionally equivalent to a reduced tumorigenic potential. Meanwhile, p54nrb depletion alone is not cytotoxic.
The investigation of p54nrb dependent gene regulations by high resolution quantitative proteomics uncovered an altering expression of multiple tumorigenic genes. For two of these candidates, the tumorigenic protease cathepsin-Z and the anti-apoptotic gelsolin, p54nrb dependent expression was detected universally in all three investigated tumor cell lines, cervix carcinoma, melanoma, and colon carcinoma. Additionally, a direct interaction of p54nrb with the cathepsin Z and gelsolin encoding DNA, but not with their corresponding mRNA, could be demonstrated.
Conjointly, this study unveils a novel mechanistic feature of caspase-2 as a tumor suppressor. The caspase-2—p54nrb axis can orchestrate the levels of several tumorigenic proteins and thereby determine the cell death susceptibility and long-term tumor survival. These findings might be of great value for future therapeutic interventions and for overcoming drug resistance of tumors.
During the past several years, ceramide has emerged as an important second messenger triggering cell responses including proliferation, differentiation, growth arrest and apoptosis. This thesis has focused on the regulation of neutral ceramidase which critically determines, in concert with ceramide generating sphingomyelinases, the intracellular ceramide levels. In the first part it is reported that besides a rapid and transient increase in neutral sphingomyelinase activity a second delayed peak of activation occurs after hours of IL-1beta treatment. This second phase of activation is first detectable after 2 h of treatment, and steadily increases over the next two hours reaching maximal values after 4 h. In parallel, a pronounced increase in neutral ceramidase activity is observed, which accounts for a constant or even decreased level of ceramide after long-term IL-1beta treatment, despite continuous sphingomyelinase activation. The increase in neutral ceramidase activity is due to expressional up-regulation, as detected by an increase in mRNA level and enhanced de novo protein synthesis. The increase of neutral ceramidase protein levels and activity can be blocked dosedependently by the p38- mitogen-activated protein kinase (p38-MAPK) inhibitor, SB 202190, whereas the classical MAPK pathway inhibitor U0126, and the PKC inhibitor Ro 31-8220 were ineffective. Moreover, co-treatment of cells for 24 h with IL-1~ and SB 202190 leads to an increase in ceramide formation. Interestingly, IL-1beta-stimulated neutral ceramidase activation is not reduced in mesangial cells isolated from mice deficient in MAPK-activated protein kinase 2 (MAPKAPK-2), which is one possible downstream substrate of the p38-MAPK, thus suggesting that the p38-MAPK-mediated induction of neutral ceramidase occurs independently of MAPKAPK-2. The results suggest a biphasic regulation of sphingomyelin hydrolysis in cytokine-treated mesangial cells with a delayed de novo synthesis of neutral ceramidase counteracting sphingomyelinase activity and apoptosis. Neutral ceramidase may thus represent a novel cytoprotective enzyme for mesangial cells exposed to inflammatory stress conditions. In a second part, the effect of NO on neutral ceramidase was studied. Ceramide levels are strongly increased in a delayed fashion by stimulation of renal mesangial cells with NO. This effect is due to a dual action of NO, comprising an activation of sphingomyelinases and an inhibition of ceramidase activity. The inhibition of neutral ceramidase activity correlates with the decrease of neutral ceramidase protein. A complete loss of neutral ceramidase protein is obtained after 24h of NO stimUlation. Moreover, the NO-induced degradation is reversed by the protein kinase C (PKC) activator, 12-0-tetradecanoylphorbol-13-acetate (TPA) , but also by the physiological PKC activators platelet-derived growth factor-BB (PDGF-BB), angiotensin II and ATP, resulting in a normalisation of neutral ceramidase protein as well as activity. In vivo phosphorylation studies using 32Pj-labelled mesangial cells, reveal that TPA, PDGF-BB, angiotensin II and ATP trigger an increased phosphorylation of the neutral ceramidase, which is blocked by the broad-spectrum PKC inhibitor Ro-31 8220, but not by CGP 41251, which has a preferential action on Ca2+-dependent PKC isoforms, thus suggesting the involvement of a Ca2+-independent PKC isoenzyme. In vitro phosphorylation assays using recombinant PKC isoenzymes and neutral ceramidase immunoprecipitated from unstimulated mesangial cells, show that particularly the PKC-alpha isoform, and to a lesser extent the PKC-a isoform, are efficient in directly phosphorylating neutral ceramidase. The data show that NO is able to induce degradation of neutral ceramidase thereby promoting accumulation of ceramide in the cell. This effect is reversed by PKC activation, most probably by the PKC-delta isoenzyme, which may directly phosphorylate and thereby, prevent neutral ceramidase degradation. In the third chapter it is demonstrated that the NO-triggered degradation of neutral ceramidase involves activation of the ubiquitin/proteasome complex. The specific proteasome inhibitor, lactacystin, completely reverses the NO-induced degradation of ceramidase protein and neutral ceramidase activity. As a consequence, the cellular amount of ceramide, which drastically increases by NO stimulation, is reduced in the presence of lactacystin. Furthermore, ubiquitinated neutral ceramidase accumulates after NO stimulation. The data clearly show that the ubiquitin/proteasome complex is an important determinant of neutral ceramidase activity and thereby regulates the availability of ceramide. In a last part, the cellular localisation of neutral ceramidase was investigated using green fluorescent protein (GFP) as fusion protein to examine cellular distribution and translocation of neutral ceramidase. Unstimulated HEK 293 cells reveal after transient transfection experiments that neutral ceramidase is preferentially localized in the cytoplasm. PKC activation led to an accumulation of neutral ceramidase at the nuclear membrane. In summary, this work demonstrates that the neutral ceramidase is a fine regulated protein that plays a critical role in regulating intracellular ceramide levels and thereby the cell's fate to undergo apoptosis or survive. Regulation of neutral ceramidase can be achieved on all levels, i.e. on the mRNA level, the protein level or posttranslationally by phosphorylation and subcellular translocation. Future work will reveal whether neutral ceramidase can serve as a therapeutic target in the development of novel antiinflammatory and anti-tumour drugs.
