Background: Rates of permanent pacemaker implantation (PPI) have been low using the self‐expanding ACURATE neo device, but data regarding risk factors of PPI for this specific device are scarce.
Methods: The study cohort consisted of patients (n = 1000) with severe aortic stenosis undergoing transfemoral transcatheter aortic valve implantation (TAVI) using the ACURATE neo prosthesis in our center between May 2012 and December 2019. For the present analysis, we excluded patients with previous permanent pacemaker (n = 110), high‐grade AV block prior to TAVI (n = 3), and patients requiring conversion to surgical valve replacement (n = 4) or the implantation of a second prosthesis as valve‐in‐valve (n = 15). Preexisting conduction abnormalities were determined, and the implantation depth of the prosthesis was measured on final angiography. Differences across quartiles based on the original consecutive cohort were analyzed with respect to implantation depth and PPI rate. Predictors of PPI were identified using logistic regression.
Results: The PPI rate was 10%. Preexisting AV block I°, right bundle branch block (RBBB), and the implantation depth were independent predictors of PPI. Across quartiles, the implantation depth differed significantly with lowest values in the last quartile, whereas differences of PPI rates across quartiles were not statistically significant, but showed a notable decrease in the last quartile.
Conclusion: Preexisting RBBB, AV block I°, and low implantation depth were independent predictors of PPI following TAVI using the ACURATE neo device. Instead of deliberately aiming at a high position, avoidance of a low implantation depth may represent a reasonable compromise to reduce the rate of PPI without increasing the risk of malpositioning.
Begriffe sind häufig nicht eindeutig. Eine „Bank“ kann ein Finanzinstitut oder eine Sitzgelegenheit sein und die Stadt Frankfurt existiert mehr als einmal. Dennoch können sie in vielen Fällen problemlos von Menschen unterschieden werden. Computer sind noch nicht in der Lage, diese Leistung mit vergleichbarer Genauigkeit zu erfüllen.
Der in dieser Arbeit vorgestellte Ansatz baut auf dem für das Deutsche bereits gute Ergebnisse erzielenden fastSense auf und verwendet ein neuronales Netz, um Namen und Begriffe in englischen Texten mit Hilfe der Wikipedia zu disambiguieren. Dabei konnte eine Genauigkeit von bis zu 89,5% auf Testdaten erreicht werden.
Mit dem entwickelten Python-Modul kann das trainierte Modell in bestehende Anwendungen eingebunden werden. Die im Modul enthaltenen Programme ermöglichen es, neue Modelle zu trainieren und zu testen.
Autophagy, together with the ubiquitin-proteasome system, is the main quality control pathway responsible for maintaining cell homeostasis. There are several types of autophagy distinguished by cargo selectivity and means of induction. This thesis focuses on macroautophagy, hereafter autophagy, where a double-layered membrane is formed originating from the endoplasmatic reticulum (ER) engulfing cargo selectively or unselectively. Subsequently, a vesicle forms around the cargo, an autophagosome, and eventually fuses with the lysosome leading to degradation of the vesicle content and release of the cargo “building blocks”. Basal autophagy continuously occurs, unselectively engulfing a portion of the cytoplasm. However, autophagy can also be induced by stress such as starvation, protein aggregation, damaged organelles, intracellular pathogens etc. In this case, the cargo is selectively targeted, and the fate of the autophagosome is the same as in basal autophagy. In recent years, interest in identifying mechanisms of autophagy regulation has risen due to its importance in neurodegenerative diseases and cancer. Given the complexity of the process, its execution is tightly regulated from initiation, autophagosome formation, expansion, closure, and finally fusion with the lysosome. Each of the steps involves different protein complexes, whose timely activity is orchestrated by post-translational modifications. One of them is ubiquitination. Ubiquitin is a small, 76-amino acid protein conjugated in a 3-step reaction to other proteins, in a reversible manner, meaning undone by deubiquitinases. Originally described as a degradation signal targeting proteins to the proteasome, today it is known it has various additional non-proteolytic functions, such as regulating a protein’s activity, localization, or interaction partners. The role of ubiquitin in autophagy has already been shown. However, given the reversibility and fine-tuning of the ubiquitin signal, many expected regulators remain unidentified. This work aimed to identify novel deubiquitinating enzymes that regulate autophagy. We identified ubiquitin-specific protease 11 (USP11) as a novel, negative regulator of autophagy. Loss of USP11 leads to an increase in autophagic flux, whereas overexpression of USP11 attenuates it. Moreover, this observation was reproducible in model organism Caenorhabditis elegans, emphasizing the importance of USP11 in autophagy regulation. To identify the mechanism of USP11-dependent autophagy regulation, we performed a USP11 interactome screen after 4 hour Torin1 treatment and identified a plethora of autophagy-related proteins. Following the most prominent hits, we have investigated versatile ways in which USP11 regulates autophagy. USP11 interacts with the PI3KC3 complex, the role of which is phosphorylating lipids of the ER, thereby initiating the formation of the autophagosomal membrane. Phosphorylated lipids serve as a recruitment signal for downstream effector proteins necessary for the membrane expansion. The core components of the complex are VPS34, the lipid kinase, ATG14, the protein responsible for targeting the complex to the ER, VPS15, a pseudokinase with a scaffolding role, Beclin1, a regulatory subunit, and NRBF2, the dimer-inducing subunit. We have found USP11 interacts with the complex and, based on its activity, USP11 influences post-translational status of all the aforementioned subunits, except for ATG14. Moreover, we have found that loss of USP11 leads to an increase in NRBF2 levels, whereas it does not change the levels of the other proteins. Given that the dimerization of the complex leads to an increase in complex activity, we investigated if the complex is more tightly formed in the absence of USP11, and if it is more active. We have found both to be the case. Although the exact mechanism of USP11-dependent PI3KC3 complex regulation remains to be identified, we found that loss of USP11 stimulates the complex formation and activity, likely contributing to the general effect of USP11 on autophagy flux. Additionally, we found that USP11 modulates levels of mTOR, the most upstream kinase in autophagy initiation steps and general multifaceted metabolism regulator. Loss of USP11 led to downregulation of mTOR levels, suggesting USP11 may rescue mTOR from proteasome-mediated degradation. Furthermore, we found mTOR to be differentially modified depending on the activity of USP11. However, it remains to be shown if USP11-dependent mTOR regulation contributes to the observed autophagy phenotype. Taken together, USP11 is a novel, versatile, negative regulator of autophagy, and an important addition to our knowledge on the regulation of autophagy by the ubiquitin system.
In vitro release testing as an alternative to establishing bioequivalence of drug products in vivo

Generische Arzneimittel werden als Arzneimittel definiert, die im Vergleich zu einem Referenzarzneimittel hinsichtlich der meisten pharmazeutischen Aspekte identisch sind.
Um die therapeutische Äquivalenz zum Referenzprodukt sicherzustellen, sind Bioverfügbarkeitsstudien erforderlich. Für Arzneimittel, die als feste, perorale, schnell freisetzende Darreichungsformen formuliert sind, kann auf den Nachweis der Bioäquivalenz in vivo zugunsten eines vergleichenden Freisetzungstests in vitro im Rahmen eines sogenannten Biopharmaceutics Classification System (BCS) basierten Biowaivers verzichtet werden.
Der BCS-basierte Biowaiver ist ein vielversprechendes Instrument, welches Kosteneinsparungen sowie eine Verringerung des regulatorischen Aufwands im Zuge der behördlichen Zulassung von Generika ermöglicht und dazu beitragen kann, die Zugänglichkeit unentbehrlicher Arzneimittel zu verbessern. Dabei gibt es jedoch auch Hürden, welche die weitläufige Anwendung des Verfahrens verhindern: Unklare Löslichkeits- und Permeabilitätsklassifizierungen von Wirkstoffen, Arzneimittel, welche die in vitro Freisetzungskriterien nicht erfüllen, sowie Zweifel an der Eignung der regulatorischen Spezifikationen, Freisetzungsunterschiede in vitro erfassen zu können, die für das Verhalten in vivo relevant sind.
In der vorliegenden Dissertation werden diese Probleme thematisiert, indem eine umfassende Bewertung der Anwendbarkeit und Einschränkungen des BCS-basierten Biowaivers in seinem aktuell regulatorisch vorgeschriebenen Ablauf vorgenommen wird. Mögliche Anpassungen des Verfahrens wurden auf der Grundlage experimenteller in vitro Daten untersucht, bewertet und mithilfe von in silico Simulationsmodellen auf die Situation in vivo extrapoliert.
Seit dem 1. Januar 2018 erhalten alle aktiven Beamt*innen, Richter*innen, Tarifbeschäftigte und Auszubildende des Landes Hessen eine Freifahrtberechtigung, sodass sie den öffentlichen Personennah- und Regionalverkehr im gesamten Bundesland kostenlos nutzen können. Diese Umstellung stellt den Anlass für die Fallstudie dar, die die Auswirkungen der Einführung eines solchen Tickets am Beispiel der Beschäftigten der Goethe-Universität Frankfurt untersucht. Im Frühjahr 2019 wurde eine quantitative Online-Befragung an der Goethe-Universität durchgeführt (n=1686). Als Grundlage und Vergleichsdatensatz diente eine Studie, die im Jahr 2015 an der Goethe-Universität durchgeführt worden ist.
Ziel der Befragung war es, Informationen über die Verkehrsmittelnutzung sowie die Einstellungen zu unterschiedlichen Mobilitätsangeboten zu erhalten. Der Schwerpunkt der Befragung lag dabei auf der Nutzung des hessischen Landestickets, sodass die Forschungsfrage untersucht wurde, welche mobilitätsbezogenen Verhaltensänderungen das hessische Landesticket hervorruft. Im Vergleich der Befragungen unter den Beschäftigten der Goethe-Universität 2015 und 2019 lässt sich in der Studie von 2019, ein Jahr nach der Einführung des hessischen Landestickets, ein signifikanter Anstieg der regelmäßigen Nutzung des öffentlichen Verkehrs erkennen. Im Hinblick auf eine erhoffte Verkehrswende weg von motorisiertem Individualverkehr hin zu ökologisch freundlicherem, öffentlichem Personennahverkehr lassen sich aus diesem Ergebnis politische Implikationen ableiten.

Kurt Goldstein zählt zu den herausragenden deutschen Neurologen des 20. Jahrhunderts. Bekannt wurde er u. a. durch einen eigenständigen Ansatz der ganzheitlichen Neurologie. Besondere Aufmerksamkeit schenkte er dabei den Kompensationsreaktionen des Gehirns und des ganzen Menschen. In der vorliegenden Studie wird erstmals der Lebensgang dieses jüdischen Arztes in den Jahren 1933 bis 1940 genauer betrachtet.
Ursprünglich sollte das vorliegende Buch nur eine Studie über die von der Kanzlei des Führers bzw. ihrem Umfeld nach Kriegsbeginn initiierten Entwürfe zu einem NS-„Euthanasie“-Gesetz werden. Dabei sollte vor allem der wichtige, allerdings mit zahlreichen Fehlern behaftete Beitrag von Roth/Aly(1984) revidiert werden. Da diese Fehler auch den Beginn der Debatte über die „Euthanasie“ im NS-Staat und den Beginn der Planung und Durchführung der „NS-Euthanasie“ (1939/1940) betrafen, musste weiter ausgeholt werden. Die Gesetzentwürfe aus der Zeit nach Kriegsbeginn machen unzweifelhaft deutlich, dass die am Krankenmord Beteiligten wussten, dass der auf den 1.9.1939 datierte „Euthanasie“-Erlass Hitlers in legaler Hinsicht nicht „ausreichte“.
Ziel dieser klinischen Studie war es, die Ergebnisse der operativen Behandlung eines idiopathischen Makulaforamens durch Vitrektomie, ggf. in Kombination mit einer Phakoemulsifikation, ILM-Peeling und einer Tamponade entweder mit Luft oder mit 20%-igem SF6-Gas zu vergleichen. Primärer Endpunkt war die Verschlussrate nach einer Vitrektomie und die Reoperationsrate bei primär nicht geschlossenen Makulaforamina. Sekundärer Endpunkt war die Visusentwicklung in der ersten postoperativen Woche und nach 3 Monaten.
In der vorliegenden Arbeit wurden hierzu retrospektiv 117 Augen von 117 konsekutiven Patienten analysiert. Es wurden die Ergebnisse von 2 Patientengruppen verglichen. In der ersten Gruppe wurden 66 Augen (m=27, w=39, Altersmedian 70 Jahre), bei denen am Ende der Vitrektomie eine Tamponade mit Luft erfolgte, untersucht. In der zweiten Gruppe wurden die Ergebnisse von 51 Augen (m=20, w=31, Altersmedian 71 Jahre), bei denen 20%-SF6-Gas als Tamponade verwendet wurde, ausgewertet. Bei etwa 70 % der Augen beider Gruppen erfolgte eine simultane Phakoemulsifikation. Der Verschluss des Makulaforamens wurde bereits in den ersten postoperativen Tagen mit einem modifizierten Fourier Domain-OCT untersucht. Sobald der Verschluss des Makulaforamens im OCT gesichert werden konnte, wurde die postoperative „Gesicht nach unten“-Lagerung („Bauchlage“) beendet. Die mediane Lagerungszeit betrug 1 Tag (Spanne 1-6 Tage). Der Zeitraum der Nachbeobachtung betrug 3 Monate (Median).
Die Verschlussrate nach einer Vitrektomie aller Makulaforamina betrug 87,2% (102/117 Augen). Die primäre Verschlussrate der Luft-Gruppe und der SF6-Gas-Gruppe betrug 83,3% (55/66 Augen) bzw. 92,2% (47/51 Augen). Bei Augen mit einem persistierenden Foramen erfolgte eine frühe Reoperation. Die Verschlussrate nach 3 Monaten lag insgesamt bei 99,1% (116/117 Augen), in der Luft-Gruppe bei 98,5% (65/66 Augen) und in der SF6-Gas-Gruppe bei 100% (51/51 Augen). Es zeigte sich kein statistisch signifikanter Unterschied der Verschlussrate bzw. der anatomischen Ergebnisse zwischen den beiden Endotamponade-Gruppen.
Es konnte jedoch ein statistisch signifikanter Unterschied des Fernvisus bei der Entlassung zwischen beiden Endotamponadegruppen nachgewiesen werden. Der logMAR-Visus bei Entlassung lag in der Luft-Gruppe mit 1,3 (Median) deutlich unter dem Visus der SF6-Gas-Gruppe mit 1,9 (Median). Die Anwendung von Luft führte somit im Vergleich zu SF6 zu einem schnelleren postoperativen Visusanstieg. In beiden Gruppen zeigte sich nach 3 Monaten eine signifikante Visusbesserung. Es ließ sich kein signifikanter Unterschied des Fernvisus zwischen den beiden Gruppen mehr nachweisen.
Die Makulaforamenchirurgie mit pars plana Vitrektomie und Peeling der ILM erzielte sowohl mit einer Luft- als auch mit einer 20%-SF6-Gas-Tamponade guten anatomischen und funktionellen Ergebnissen. Mit einer kurz wirksamen Endotamponade erscheint eine postoperative Lagerung essenziell, dürfte aber in den meisten Fällen nur wenige Tage erforderlich sein, um den Verschluss eines durchgreifenden Makulaforamens zu erreichen. Die Reoperations-Rate nach einer Lufttamponade war bei sehr großen Foramina (> 600 µm) signifikant höher als bei Foramina mit einem Durchmesser kleiner 600 µm. Daher scheint die Anwendung von Gasen, die länger als Luft wirken, nur bei sehr großen Foramina erforderlich zu sein. Vorteil einer kurz wirkenden Lufttamponade ist die postoperativ um einige Tage schneller einsetzende visuelle Rehabilitation, so dass die Patienten früher in ihren Alltag zurückkehren können. Eine frühe postoperative Untersuchung der Makula unter Einsatz der optischen Kohärenztomographie ist Voraussetzung für eine individualisierte Steuerung der Lagerung und für eine möglichst frühe Erkennung und Reoperation von nicht geschlossenen Makulalöchern.
Throughout their life cells of eukaryotic organisms can be confronted with a variety of proteotoxic stresses and in order to survive, corresponding resistance mechanisms had to evolve. Proteotoxic stresses can cause misfolding of proteins and accumulation of toxic protein aggregates. Failure to remove aggregates of misfolded proteins compromises cellular function and can ultimately cause cell death and disease. To deal with this challenge, cells utilize a complex network of protein quality control pathways, including chaperones, the ubiquitin-proteasome system and the autophagy system.
Another mechanism to cope with proteotoxic stresses is the stalling of translation initiation in order to save valuable resources and prevent faulty translation. Upon stress, intrinsically disordered RNA-binding proteins such as TIA-1 or G3BP1/2 are recruited to stalled preinitiation complexes and a network of multivalent interactions between RNAs and proteins is formed. These mRNP networks can merge with each other and phase separate into membraneless liquid-like structures called stress granules (SGs). Once stress is released, SGs are quickly resolved and translation continues. Yet, chronic stress or mutations of SG-associated proteins can cause persistent SGs, which can sequester misfolded proteins and have been linked to neurodegenerative diseases such as amyotrophic lateral sclerosis or frontotemporal dementia.
In mammalian cells, three isoforms of the small ubiquitin-related modifier (SUMO), SUMO1, SUMO2 and SUMO3 are covalently attached to lysine residues of target proteins. SUMO conjugation is catalyzed via an enzymatic cascade of an heteromeric E1 activating enzyme, the E2 conjugating enzyme Ubc9 and in some cases one of a limited number of E3 SUMO ligases. SUMOylation is a dynamic modification and can be reversed by SUMO isopeptidases, the best characterized of which belong to the SENP family. Cellular stresses such as heat or oxidative stress strongly induce SUMOylation resulting in increased numbers of poly-SUMOylation (formation of SUMO2/3 chains) on nuclear proteins.
The SUMO-targeted ubiquitin ligase (STUbL) RNF4 harbors four SUMO interaction motifs in its N-terminal domain. This feature allows RNF4 to specifically bind poly-SUMOylated proteins and catalyze their proteolytic or non-proteolytic ubiquitylation.
A variety of substrate proteins have been shown to undergo SUMO-primed ubiquitylation by RNF4 in response to stress or DNA damage. RNF4-mediated ubiquitylation is often a signal for proteolytic degradation of these substrates.
In this work we aimed by identify novel RNF4 targets, in heat-stressed cells in order to gain a wider understanding of the nuclear proteotoxic stress response. Analysis by mass spectrometry revealed that a large fraction of RNF4-interacting proteins in heatstressed cells are nuclear RNA-binding proteins, many of which shuttle outside the nucleus and associate with SGs upon stress. We validated, that nuclear RNA-binding proteins, such as TDP-43 and hnRNP M are indeed heat-induced targets of SUMOprimed ubiquitylation by RNF4.
These initial results led us to further investigate the links between the SUMO/RNF4-mediated, nuclear protein quality control and the dynamics of cytosolic heat- or arsenite-induced SGs. SUMO2/3 and RNF4 are mainly nuclear proteins and we confirmed that they do not associate with SGs. Yet, we could demonstrate that depletion of SUMO2/3, the E3 SUMO ligase PML or RNF4 as well as chemical inhibition of SUMOylation strongly delayed SG clearance upon stress release, indicating that a functional STUbL pathway is essential for the timely clearance of SGs.
Next, we investigated how stress-induced poly-SUMOylation is regulated. Our data shows that SENP levels and activities are reduced in response to heat and arsenite stress, which allows the buildup of poly-SUMO chains on nuclear proteins. Limitation of poly-SUMOylation by overexpression of the SUMO chain-specific isopeptidases SENP6 and SENP7 induced SG formation. In contrast, poly-SUMO-priming by chemical depletion of SENP6 with the drug hinokiflavone drastically limited SG formation upon stress treatment. These results indicate a clear role of chain-specific SENPs in the regulation of stress-induced poly-SUMOylation and SG dynamics.
Last, we investigated whether the STUbL pathway could affect the phase separation of FUSP525 (an ALS-linked mutant of the SG-associated protein FUS) and observed that perturbations of the STUbL pathway lead to an increased phase separation of FUSP525L.
Thus, our work connects the SUMO/RNF4 protein quality control mechanism to the dynamics of SGs supporting the hypothesis that release of proteotoxic stress in the nucleus facilitates the clearance of cytosolic SGs. Thereby, we discovered a previously unknown link between the nuclear and cytosolic axis of proteotoxic stress response.
In the past decade, tissue-resident innate lymphoid cells (ILC) have become a central field of immunological research. ILC are a family of innate immune cells comprising cytotoxic Natural Killer (NK) cells and the non-cytotoxic helper like ILC1, ILC2 and ILC3. They mirror the functions and phenotypes of T cells, but do not require rearranged antigen-specific receptors for their rapid response to signals from injured or infected tissue. As potent cytokine producers being enriched in mucosal tissue, ILC play an essential role in tissue maintenance and regulating immunity to chronic inflammation and infection (Vivier et al., 2018). Although heterogeneity and plasticity of ILC complicates their classification, the pathophysiology of a broad variety of autoimmune and chronic inflammatory diseases have been associated with dysregulations in ILC subset distribution and functions (Dzopalic et al., 2019). This highlights their importance in human health and disease and accounts for the need for markers unambiguously describing the different ILC subtypes. This work introduces NKp65, a C-type lectin-like receptor (CTLR) encoded in the natural killer gene complex by the KLRF2 gene, as an exclusive marker for human ILC3. NKp65 expression especially discerns ILC3-like NK cell precursor from mature NK cells which express the NKp65-relative NKp80. Moreover, flow cytometric analysis of NKp65 expression aids in the demarcation of natural cytotoxicity receptor (NCR) expressing ILC3, from the closely related but functionally distinct RORt+ LTi cells and NCR- ILC3. This work further provides insights into NK cell development by in vitro differentiation studies in which NKp65 expressing cells are generated in presence of OP9 feeder cells and cytokines to support development. In such cultures, NKp65 expressing in vitro ILC (ivILC) acquire NKp80 expression in a Notch-dependent manner indicating their differentiation into mature NK cells. Acquisition of NK cell phenotypic markers is accompanied by NKp65 downregulation which leads to the mutually exclusive expression of NKp80 on NK cells and NKp65 on ILC3-like cells. Further insights are provided into the functional consequences of NKp65 engagement by its cognate high affinity ligand ‘keratinocyte-associated C-type lectin’ (KACL) which is selectively expressed on human keratinocytes (Bauer et al., 2015; Spreu et al., 2010). Expressed on ivILC, NKp65 mediates killing of KACL expressing target cells, suggesting that NKp65-KACL interaction promotes cellular cytotoxicity. In this context, the observed metalloproteinase dependent shedding of NKp65 might play a role in the termination of the cellular interaction. The findings on the regulation of NKp65 expression demonstrate the presence of a functional STAT5 response element in the KLRF2 promoter endowing a transcriptional control of NKp65 expression by IL-7 signaling. This provides an interesting link between the dependency of ILC3 on IL-7 signaling for their maintenance and the specific expression of NKp65 on these cells.
In summary, this study provides new insights into the physiologic expression of the CTLR NKp65 on human ILC3. The dependency of NKp65 surface expression on sustained STAT5 signaling provided by IL-7 underlines the connection of NKp65 expression and an ILC3 phenotype which might contribute to promote future research in discerning the interspersed pathways of ILC3 and NK cell development. The tissue and cell specific expression of NKp65 on ILC3 and its ligand KACL on keratinocytes of the human skin further suggests an important role of this genetically coupled receptor-ligand pair in tissue specific immunosurveillance.
Die Frage der Frauenrechte in Afghanistan diente nach dem Sturz des Taliban-Regimes als Legitimation für das militärische und entwicklungspolitische Engagement der internationalen Gemeinschaft. Jedoch existieren bis heute nur wenige empirische Arbeiten, die Aufschluss über Lebenslagen afghanischer Frauen geben und Interventionen lokaler Akteur*innen untersuchen, die diese adressieren. Die vorliegende Dissertation, die in der feministischen Ethnologie und in der Ethnologie der Menschenrechte verortet ist, trägt dazu bei, diese Forschungslücken zu schließen.
Die Daten wurden in mehrmonatigen teilnehmenden Beobachtungen in zwei afghanischen Frauenrechts-NGOs und einem Frauenrechtsnetzwerk erhoben, ergänzt von 38 Interviews. Vier Interventionsformen gegen Gewalt an Frauen werden analysiert: Neben Frauenselbsthilfegruppen und Frauenhäusern sind dies Interaktionen mit religiösen und anderen Machtakteuren. Nach Merry (2006) werden die Interventionen als Aushandlungs- und Übersetzungsprozesse divergierender Vorstellungen über Geschlechternormen und soziale Praktiken innerhalb des afghanischen Normenpluralismus konzeptualisiert.
Die Studie liefert Ergebnisse auf mehreren Ebenen. Erstens werden sowohl spezifische Bedingungen für die Wirksamkeit der untersuchten Interventionsformen herausgearbeitet als auch ihre jeweiligen Begrenzungen, welche in den vorherrschenden Machtdynamiken begründet sind. Deutlich wird zweitens der Nutzen der universalen Menschenrechte als Bezugs- und Legitimationsrahmen für die Aktivist*innen, wobei sich gleichermaßen zeigt, wo sich der Bezug auf Menschenrechte als problematisch erweist. Um Rückschlüsse für zukünftige Interventionen von Frauenrechts-NGOs auch über den afghanischen Kontext hinaus ziehen zu können, wird drittens eine Erweiterung des Theorie-Praxis-Modells nach Merry (2006) vorgeschlagen, welche die Kontextualisierung in Machtverhältnisse miteinbezieht, sowie Begrenzungen als Indikatoren für Aushandlungsspielräume postuliert.

Ein großer Tag für das Institut für Informatik der Goethe-Universität: Es feiert sein 40-jähriges Jubiläum mit einem Festakt am Freitag, den 13. Oktober, ab 14 Uhr im Casino auf dem Campus Westend. Die Universität hat zusammen mit dem Alumni- und Förderverein hochkarätige Gastredner eingeladen, die ein Bild von der Bedeutung der Informatik und ihrer Forschung zeichnen werden.
Prof. Brigitte Geißel und ihr Team von der Forschungsstelle Demokratische Innovationen an der Goethe-Universität beschäftigen sich mit der Krise der repräsentativen Demokratie, wie sie im Augenblick unter dem Eindruck der letzten Bundestagswahl verstärkt auch in der Öffentlichkeit diskutiert wird. „Wahlen und Parteienwettbewerb sind lediglich die Form, die wir heute am besten kennen; dieses institutionelle Set ist aber nicht der innerste Kern von Demokratie“, so das Team der Forschungsstelle in seinem folgenden Beitrag.
Wie in allen Bereichen von Wirtschaft und Kultur gibt es auch im Buchwesen Kriminalität: Diebstahl, Betrug und Fälschung. Vor wenigen Jahren erregte ein Band mit angeblichen Zeichnungen Galileis weltweites Aufsehen. Im 19. Jahrhundert gab es mit Denis Vrain-Lucas einen „König der Fälscher“, der einem angesehenen Mathematiker angebliche Briefe Kleopatras an Marcus Antonius, Alexanders des Großen an Aristoteles und ein Schreiben des auferstandenen Lazarus an seine Angehörigen verkaufen konnte. Auch der Reformator Martin Luther fand seinen Imitator.
A recent global meta‐analysis reported a decrease in terrestrial but increase in freshwater insect abundance and biomass (van Klink et al., Science 368, p. 417). The authors suggested that water quality has been improving, thereby challenging recent reports documenting drastic global declines in freshwater biodiversity. We raise two major concerns with the meta‐analysis and suggest that these account for the discrepancy with the declines reported elsewhere. First, total abundance and biomass alone are poor indicators of the status of freshwater insect assemblages, and the observed differences may well have been driven by the replacement of sensitive species with tolerant ones. Second, many of the datasets poorly represent global trends and reflect responses to local conditions or nonrandom site selection. We conclude that the results of the meta‐analysis should not be considered indicative of an overall improvement in the condition of freshwater ecosystems.

Background: The aim of this study was to evaluate post‐irradiation changes in the central nervous system (CNS) detected using magnetic resonance (MR) imaging.
Methods: Magnetic resonance images of 15 children with CNS tumors treated through whole‐brain irradiation over 10 years were reviewed retrospectively. Variables such as age at the time of irradiation, total radiation dose, treatment length, and time interval between irradiation and MR changes, were evaluated.
Results: All patients included in the study had imaging abnormalities of the CNS. Eight patients (53%) developed CNS abnormalities within a short period of time – only a few months after irradiation (mean 4.8 months). Seven patients (47%) developed CNS abnormalities within a long time interval after treatment (mean 4.6 years). In almost all patients, a T2 increase in supra‐ and infratentorial white matter was observed. Follow‐up examinations showed nine patients (60%) with cerebellar atrophy.
Conclusions: In this sample of pediatric patients who underwent whole‐brain irradiation, the time receiving irradiation was not related to the severity of the MR changes. A correlation between the age of the child or the length of the radiotherapy and the extent of the changes could not be confirmed. However, we observed a trend towards stronger brain parenchymal degeneration with cystic changes in the younger age group of children in our sample. Older children who received irradiation seem to be more susceptible to vascular dysplasia with cavernous hemangiomas and microbleeding.
Abstract
The mineralogy, chemical composition, and physical properties of cratonic mantle eclogites with oceanic crustal protoliths can be modified by secondary processes involving interaction with fluids and melts, generated in various slab lithologies upon subduction (auto‐metasomatism) or mantle metasomatism after emplacement into the cratonic lithosphere. Here we combine new and published data to isolate these signatures and evaluate their effects on the chemical and physical properties of eclogite. Mantle metasomatism involving kimberlite‐like, ultramafic carbonated melts (UM carbonated melts) is ubiquitous though not pervasive, and affected between ~20% and 40% of the eclogite population at the various localities investigated here, predominantly at ~60–150 km depth, overlapping cratonic midlithospheric seismic discontinuities. Its hallmarks include lower jadeite component in clinopyroxene and grossular component in garnet, an increase in bulk‐rock MgO ± SiO2, and decrease in FeO and Al2O3 contents, and LREE‐enrichment accompanied by higher Sr, Pb, Th, U, and in part Zr and Nb, as well as lower Li, Cu ± Zn. This is mediated by addition of a high‐temperature pyroxene from a UM carbonated melt, followed by redistribution of this component into garnet and clinopyroxene. As clinopyroxene‐garnet trace‐element distribution coefficients increase with decreasing garnet grossular component, clinopyroxene is the main carrier of the metasomatic signatures. UM carbonated melt‐metasomatism at >130–150 km has destroyed the diamond inventory at some localities. These mineralogical and chemical changes contribute to low densities, with implications for eclogite gravitational stability, but negligible changes in shear‐wave velocities, and, if accompanied by H2O‐enrichment, will enhance electrical conductivities compared to unenriched eclogites.

Oceanic crust formed at spreading ridges is recycled in subduction zones and undergoes metamorphism to eclogite. Some of this material is captured in the overlying lithospheric mantle, where it is exhumed by passing magmas. Having formed in spreading ridges, these eclogites have proven invaluable archives for the onset of plate tectonics, for the construction of cratons during subduction/collision, as probes of the convecting mantle from which their precursors formed, and as generators of heterogeneity upon recycling into Earth's convecting mantle. During subduction and until exhumation, interaction with fluids and melts (called metasomatism) can change the mineralogy, chemical composition, and physical properties of mantle eclogites, complicating their interpretation, but a comprehensive study of these effects is lacking so far. We investigated mantle eclogites from ancient continents (cratons) around the globe in order to define hallmarks of metasomatism by subduction‐related fluids and small‐volume ultramafic carbonated mantle melts. We find that the latter is pervasive and occurs predominantly at midlithospheric depths where seismic discontinuities are detected, typically causing diamond destruction and a reduction in density. This has consequences for their gravitational stability and for the interpretation of shearwave velocities in cratons.
Primary determinants of communities in deadwood vary among taxa but are regionally consistent

The evolutionary split between gymnosperms and angiosperms has far‐reaching implications for the current communities colonizing trees. The inherent characteristics of dead wood include its role as a spatially scattered habitat of plant tissue, transient in time. Thus, local assemblages in deadwood forming a food web in a necrobiome should be affected not only by dispersal ability but also by host tree identity, the decay stage and local abiotic conditions. However, experiments simultaneously manipulating these potential community drivers in deadwood are lacking. To disentangle the importance of spatial distance and microclimate, as well as host identity and decay stage as drivers of local assemblages, we conducted two consecutive experiments, a 2‐tree species and 6‐tree species experiment with 80 and 72 tree logs, respectively, located in canopy openings and under closed canopies of a montane and a lowland forest. We sampled saproxylic beetles, spiders, fungi and bacterial assemblages from logs. Variation partitioning for community metrics based on a unified framework of Hill numbers showed consistent results for both studies: host identity was most important for sporocarp‐detected fungal assemblages, decay stage and host tree for DNA‐detected fungal assemblages, microclimate and decay stage for beetles and spiders and decay stage for bacteria. Spatial distance was of minor importance for most taxa but showed the strongest effects for arthropods. The contrasting patterns among the taxa highlight the need for multi‐taxon analyses in identifying the importance of abiotic and biotic drivers of community composition. Moreover, the consistent finding of microclimate as the primary driver for saproxylic beetles compared to host identity shows, for the first time that existing evolutionary host adaptions can be outcompeted by local climate conditions in deadwood.
Background: Conditions during blood product storage and transportation should maintain quality. The aim of this in vitro study was to investigate the effect of interruption of agitation, temporary cooling (TC), and pneumatic tube system transportation (PTST) on the aggregation ability (AA) and mitochondrial function (MF) of platelet concentrates (PC).
Study Design and Methods: A PC was divided equally into four subunits and then allocated to four test groups. The control group (I) was stored as recommended (continuous agitation, 22 ± 2°C) for 4 days. The test groups were stored without agitation (II), stored as recommended, albeit 4°C for 60 minutes on day (d)2 (III) and PTST (IV). Aggregometry was measured using Multiplate (RocheAG; ADPtest, ASPItest, TRAPtest, COLtest) and MF using Oxygraph‐2k (Oroboros Instruments). The basal and maximum mitochondrial respiratory rate (MMRR) were determined. AA and MF were measured daily in I and II and AA in III and IV on d2 after TC/PTST. Statistical analysis was performed using tests for matched observations.
Results: Eleven PCs were used. TRAP‐6 induced AA was significantly lower in II when compared to I on d4 (P = 0.015*). In III the ASPItest was significantly lower (P = 0.032*). IV showed no significant differences. The basal and MMRR were significantly reduced over 4 days in I and II (for both rates in both groups: P = <0.0001*). No significant differences occurred on d4 (P = 0.495).
Conclusion: Our results indicate that ex vivo AA and MF of PCs are unaffected, even in no‐ideal storage and transport circumstances with respect to agitation, temperature, and force.
We analyze limit order book resiliency following liquidity shocks initiated by large market orders. Based on a unique data set, we investigate whether high‐frequency traders are involved in replenishing the order book. Therefore, we relate the net liquidity provision of high‐frequency traders, algorithmic traders, and human traders around these market impact events to order book resiliency. Although all groups of traders react, our results show that only high‐frequency traders reduce the spread within the first seconds after the market impact event. Order book depth replenishment, however, takes significantly longer and is mainly accomplished by human traders’ liquidity provision.
Autism spectrum disorder (ASD) is a complex neurodevelopmental disorder that is accompanied by neurodevelopmental differences in regional cortical volume (CV), and a potential layer‐specific pathology. Conventional measures of CV, however, do not indicate how volume is distributed across cortical layers. In a sample of 92 typically developing (TD) controls and 92 adult individuals with ASD (aged 18–52 years), we examined volumetric gradients by quantifying the degree to which CV is weighted from the pial to the white surface of the brain. Overall, the spatial distribution of Frustum Surface Ratio (FSR) followed the gyral and sulcal pattern of the cortex and approximated a bimodal Gaussian distribution caused by a linear mixture of vertices on gyri and sulci. Measures of FSR were highly correlated with vertex‐wise estimates of mean curvature, sulcal depth, and pial surface area, although none of these features explained more than 76% variability in FSR on their own. Moreover, in ASD, we observed a pattern of predominant increases in the degree of FSR relative to TD controls, with an atypical neurodevelopmental trajectory. Our findings suggest a more outward‐weighted gradient of CV in ASD, which may indicate a larger contribution of supragranular layers to regional differences in CV.
We show that financial advisors recommend more costly products to female clients, based on minutes from about 27,000 real-world advisory meetings and client portfolio data. Funds recommended to women have higher expense ratios controlling for risk, and women less often receive rebates on upfront fees for any given fund. We develop a model relating these findings to client stereotyping, and empirically verify an additional prediction: Women (but not men) with higher financial aptitude reject recommendations more frequently. Women state a preference for delegating financial decisions, but appear unaware of associated higher costs. Evidence of stereotyping is stronger for male advisors.
Als die Existenz der Psychotherapeutischen Beratungsstelle für Studierende an der Goethe-Universität bedroht schien, da machte sich die Ehrensenatorin Renate von Metzler für diese wichtige Anlaufstelle für Studierende in schwierigen Lebenssituationen stark – mit Erfolg: Inzwischen nutzen jährlich mehr als 400 Ratsuchende dieses Angebot, damit hat sich ihre Zahl in kurzer Zeit verdoppelt. Ohne die „Anschubfinanzierung“ der Freunde und Förderer von 35.000 Euro wäre dies nicht so einfach möglich gewesen.

Anknüpfend an Untersuchungen von Nachwirkungserscheinungen vor allem an der Taenia coli des Meerschweinchens, bei denen Nachdehnung und Relaxation mit einer Hyperbelfunktion beschrieben wurden, gehörte es zu den Zielen dieser Arbeit, Gesetzmäßigkeiten im zeitlichen Verlauf der Nachdehnung bei dem anders strukturierten Uterushorn in verschiedenen Badlösungen zu untersuchen und insbesondere zu prüfen, ob die Hyperbelfunktion auch darauf angewendet werden kann und welche Rückschlüsse auf die funktionelle Struktur möglich sind.
Nach Voruntersuchungen an 30 Uterushörnern, bei denen auch Fragen der Tierwahl und der Standardisierung des Hormoneinflusses untersucht wurden, folgten nach ausführlicher biomathematischer Planung Hauptuntersuchungen an 58 Uterushörnern. Ein weiteres Ziel der Arbeit, eine wesentliche Verbesserung der Datenerfassung, konnte durch Einsatz eines Kurvenfolgers in Verbindung mit einer EDV-Anlage und eigenen Datenerfassungsprogrammen erreicht werden, wobei besonderer Wert darauf gelegt wurde, die Reproduzierbarkeit der Datenerfassung näher zu untersuchen, um die erzielte Genauigkeit, die höher als bei allen vergleichsweise zitierten Arbeiten liegt, nachprüfbarangeben zu können. Die erweiterten Möglichkeiten der Datenerfassung lieferten den Anstoß zum Erkennen von Problemen in der bisher üblichen Form der Datenauswertung, der Wertestandardisierung und der Nachdehnungsdefinition; hierfür konnten Lösungswege erarbeitet werden: u.a. ein EDV-Programm zur iterativen Hyperbelanpassung, Definition neuer Nachdehnungsgrößen und eine Aufgliederung in eine modellunabhängige und eine modellabhängige Auswertung.
Nach einem ausführlichen Überblick Uber die Physiologie glatter Muskulatur unter den Aspekten von Funktion und Struktur sowie einer Auseinandersetzung mit dem Begriff des Modells erfolgte eine zusammenfassende Datenauswertung: eine univariate Mehrwegkovarianzanalyse bestätigte nicht nur die Bedeutung des Faktors „Badlösung”, sondern ermöglichte auch unerwartete Aussagen über Kovariablen und Wechselwirkungen, die in früheren Arbeiten nicht in die Fragestellungen mit einbezogen worden waren; iterative Kurvenanpassungen zeigten die Überlegenheit mehrerer einfacher logarithmischer und exponentieller Funktionen im Vergleich zur Hyperbelfunktion. Außerdem waren Vergleiche zwischen verschieden strukturierten Muskeln und zwei Tierarten möglich.
Aus den gewonnenen Ergebnissen werden einige Schlußfolgerungen gezogen und Deutungen für die funktionelle Struktur des glatten Muskels gegeben.
Hintergrund: Die Einnahme vieler Antidepressiva und Antipsychotika in der frühen Schwangerschaft kann nach aktuellem Forschungsstand als sicher bezüglich des Risikos für Fehlbildungen erachtet werden. Daten hinsichtlich der Sicherheit einer Psychopharmakotherapie während der Stillperiode sind hingegen rar. Die meisten Betroffenen würden einerseits gerne ihre Kinder stillen, andererseits wollen sie aber auch sicher sein, dass sie ihrem Nachwuchs dadurch nicht schaden.
Ziele: Unser Ziel war es herauszufinden ob eine Korrelation zwischen der Tagesdosis der Psychopharmaka und der Medikamentenkonzentration im Serum sowie in der Muttermilch (MM) besteht. Weiterhin untersuchten wir, ob es eine allgemeingültige Regeln gibt, wie sich Muttermilchmedikamentenkonzentrationen im Tagesverlauf verhalten. Außerdem wollten wir herausfinden ob die Medikamentenexposition in der Schwangerschaft und/oder Stillperiode einen Einfluss auf die Entwicklung des Kindes in den ersten zwölf Lebensmonaten hat.
Methoden: Wir untersuchten therapeutische Medikamentenspiegel parallel in Serum und MM zu verschiedenen Zeitpunkten um herauszufinden wann die höchsten/niedrigsten Konzentrationen ermittelt werden konnten. Neben dem Talspiegel, nach 12 h oder 24 h (T1), sammelten wir Proben 4 h (T5) und 8 h (T6) nach der Medikamenteneinnahme sowie direkt nach der Medikamenteneinnahme (T2), 1 h nach T2 (T3) und direkt beim nächsten Stillen (T4). Die Messungen wurden im TDM Labor im Zentrum für psychsiche Gesundheit, Universitätsklinikum Würzburg mittels isokratischer reversed-phase high performance liquid Chromatographie durchgeführt. Wir rekrutierten 25 Patientinnen, die in der ambulanten Spezialsprechstunde für psychiatrische Erkrankungen in Schwangerschaft und Stillzeit behandelt wurden. 68% der Patientinnen nahmen in der Schwangerschaft schon ein Psychopharmakon ein. Die Patientinnen litten an unipolaren Depressionen (n = 15), bipolaren Störungen (n = 2), schizoaffektiven Störungen (n = 2) sowie Angst- (n = 4) und Zwangsstörungen (n = 2). Das Spektrum von Antidepressiva und Antipsychotika erstreckte sich von Amitriptylin/Nortriptylin, Clomipramin/N-Desmethyl-Clomipramin, Mirtazapin, Escitalopram, Citalopram, Sertralin, Venlafaxin/ O-Desmethyl-Venlafaxin bis hin zu Lamotrigin und Quetiapin. Weiterhin erfassten wir ob es postnatale Auffälligkeiten sowie im Verlauf Entwicklungsauffälligkeiten bei den exponierten Kinder gab (Daten aus den routinemäßigen U-Untersuchungen der ersten 12 Lebensmonate). Um mehr Daten zur Kindesentwicklung zu sammeln, interviewten wir in einem retrospektiven Unterprojekt 14 weitere Patientinnen, die in der Vergangenheit von der Thematik betroffen waren.
Ergebnisse: Die parallel bestimmten Talspiegel in Serum und MM korrelierten nicht signifikant bei der Analyse aller Medikamente zusammen oder bei der Analyse der einzelnen Medikamente (Spearman Rho Korrelation, p > 0,05). Die Talspiegel im Serum korrelierten weder signifikant mit den Muttermilchkonzentration nach 4 h (T5), noch mit denen nach 8 h (T6) (Spearman Rho Korrelation, p = 0,46; p = 0,08). Die Tagesdosis korrelierte signifikant mit dem Talspiegel im Serum (p = 0,001), jedoch nicht mit den gleichzeitig bestimmten Muttermilchkonzentrationen (p = 0,88). Die Konzentrationen von Venlafaxin in der MM fielen relativ hoch aus. Es zeigte sich eine große interindividuelle Variation der Konzentrationen der unterschiedlichen Medikamente zu verschiedenen Zeitpunkten. Clomipramin und Quetiapin konnten gar nicht oder nur in sehr niedrigen Konzentrationen in der MM gemessen werden (n = 2, n = 4) wie auch Sertralin (ohne Ausreißer-Werte) und Escitalopram. Auch konnte Quetiapin nicht im Serum eines gestillten Kindes nachgewiesen werden, bei dem wir Blutproben gewinnen konnten (n = 1). Keines der gestillten Kinder zeigte negative Auswirkungen aufgrund der Medikation. Die höchste „concentration-bydose-ratio“ (C/D) in der MM wurden für Venlafaxin (n = 4, C/D MM = 5,11) und Lamotrigin (n = 1, C/D MM = 10,0) errechnet, während Clomipramin (n = 2, C/D MM = 0,0), Quetiapin (n = 4, C/D MM = 0,02) und Sertralin (n = 3, C/D MM = 0,03) die niedrigsten Quotienten zeigten. Wir kalkulierten die höchste „milk-to-plasma-ratio“ (M/P) für Mirtazapin (n = 3, M/P = 3,92) und Venlafaxin (n = 2, M/P = 2,4) sowie die niedrigsten für Clomipramin (n = 2, M/P = 0), Quetiapin (n = 3, M/P = 0,03) und Sertralin (n = 3, M/P = 0,08), was für ein niedriges Eindringungsvermögen vom Serum in die MM spricht.
Beim Exzellenzcluster „Normative Ordnungen“ eruiert man mit Hochdruck Möglichkeiten, wie es auch nach dem Ausscheiden aus der Exzellenzstrategie weitergehen kann. Das Land Hessen und die Goethe-Universität haben bereits ihre Unterstützung signalisiert, eine Liste von renommierten Forschern aus der ganzen Welt fordert die Weiterführung der international geschätzten Institution. Der UniReport konnte mit den beiden Sprechern des Clusters, Prof. Rainer Forst und Prof. Klaus Günther, direkt im Anschluss an die Jahreskonferenz des Clusters ein Gespräch über momentane Stimmungen und Perspektiven im Haus „Normative Ordnungen“ auf dem Campus Westend führen.
Michel Houellebecq oder Toni Morrison, Nadine Gordimer oder Jorge Semprún? Studierende an der Goethe-Uni müssen sich nicht mehr entscheiden, ob sie sich mit spanischsprachiger, französischer, italienischer oder englischsprachiger Literatur und Kultur beschäftigen möchten. Der junge Masterstudiengang Moving Cultures schlägt die Brücke zwischen den Sprachwelten und legt den Schwerpunkt gerade auf die wechselseitigen Einflüsse. Das lockt auch viele Externe nach Frankfurt.

Dr. Z. Ece Kaya, 1978 in Istanbul geboren, ist u. a. Diplom-Politologin und promovierte Erziehungswissenschaftlerin. Ihre Arbeitsschwerpunkte sind Erziehungswissenschaften in der NS-Zeit sowie pädagogische Antworten auf Rassismus und Nationalismus. Sie arbeitet auch als Übersetzerin, Sprachlehrerin und als Theaterpädagogin. Für ihre Dissertation »Kolonialpädagogische Schriften in der NS-Zeit: ,Eine spezifisch deutsche Theorie der Kolonisation‘ – Zur Geschichte des Kolonialrassismus in der deutschen Erziehungswissenschaft « wurde sie 2017 mit dem Werner-Pünder-Preis ausgezeichnet.
