Diese Dissertation befasst sich mit der massenspektrometrischen Analytik von Membranproteinen. Diese gestaltet sich aufgrund der hydrophoben Natur der Proteine und der damit verbundenen schlechten Löslichkeit in wässrigen Puffersystemen als deutlich schwieriger als die Untersuchung löslicher Proteine. Einige weitere Probleme entstehen durch die konsequente Verwendung der Referenzprotease Trypsin zur Hydrolyse der Proteine. Sie spaltet ausschließlich Peptidbindungen nach basischen Aminosäuren, die naturgemäß in den unpolaren Helixbereichen der Membranproteine nur vereinzelt anzutreffen sind. Im Rahmen dieser Arbeit wurden auf weniger spezifischen Proteasen basierende Protokolle zur Analyse von ganzen Membranproteomen entwickelt, welche einige der ursprünglichen Probleme umgehen oder zumindest vermindern. Die Proteolysereaktion fand in Anwesenheit von Methanol statt, welcher die Membranstruktur destabilisiert und so die Zugänglichkeit der in die Membran eingebetteten Proteinteile für das Enzym erhöht. Zusätzlich erhöhte sich dadurch die Löslichkeit gebildeter hydrophober Peptide im Puffer. Das neue Protokoll wurde an Bacteriorhodopsin, einem geläufigen Modellsystem für α helikale Membranproteine, erfolgreich etabliert. Bei Verwendung der Proteasen Elastase und Pepsin konnten nach nLC-Trennung und MALDI-MS/MS deutlich mehr Peptide des Membranproteins signifikant identifiziert werden, als es mit Trypsin möglich gewesen ist. Auch qualitativ ergaben sich Unterschiede zwischen beiden Enzymen. Mit den alternativen Enzymen ließen sich nicht nur die hydrophilen peripheren Proteinabschnitte nachweisen, sondern auch zahlreiche Peptide aus den hydrophoben Transmembranbereichen. Das am Modellprotein etablierte Elastase-Protokoll konnte problemlos auf die Analyse des komplexeren Membranproteoms von Corynebacterium glutamicum transferiert werden. So konnten ca. 150 verschiedene Proteine identifiziert werden, darunter auch zahlreiche Membranproteine. Die erzeugten Peptidmischungen wurden nicht nur mittels nLC-MALDI-TOF/TOF sondern auch mit einer nLC-ESI-Orbitrap-Plattform analysiert. Anhand einer detaillierten Analyse der physikochemischen Eigenschaften der entstandenen Peptide konnte gezeigt werden, dass der dabei beobachtete Vorteil des ESI-Instrumentes zu großen Teilen auf die bessere Detektion aliphatischer Neutralpeptide zurückzuführen war. Diese werden bei der MALDI vornehmlich aufgrund der schlechteren Ionisation bei Verwendung der Standardmatrix α-Cyano-4-Hydroxyzimtsäure diskriminiert. Ein weiterer Vorteil des verwendeten ESI-Gerätes ist seine bessere Massengenauigkeit durch den hochauflösenden Orbitrap-Massenanlysator. Wurden die MALDI-TOF/TOF-Fragmentierungsdaten durch, auf einem zweiten Instrument gemessene, genaue MALDI-Orbitrap-Vorläufermassen supplementiert, konnten über 30% mehr Peptide signifikant identifiziert werden. Die gesammelten Daten konnten zur Erhebung einiger Statistiken über Elastase und Pepsin herangezogen werden. Erstmals wurde eine umfassende Untersuchung der Spaltspezifität von Elastase vorgenommen. Das Enzym hydrolysiert in ca. 82% der Fälle nach den kleinen hydrophoben Aminosäuren Ala, Val, Ile, Leu, Ser und Thr. Diese Spezifität ist für proteolytische Schnitte innerhalb der hydrophoben Helixbereiche von Membranproteinen äußerst vorteilhaft. Sie reicht aus, um Peptide Mass Fingerprinting an Einzelproteinen durchzuführen, wie exemplarisch am Modellprotein Bacteriorhodopsin gezeigt wurde. Die für Pepsin erhaltene Spezifität ist hingegen zu undifferenziert für solche Experimente, weshalb hier MS/MS-basierte Strategien vorzuziehen sind. Weitergehende Verbesserungen für Datenbanksuchen mit weniger spezifischen Proteasen wurden vorgeschlagen. Hierzu wurde erstmals eine größere Fragmentierungsstatistik eines nichttryptischen Datensatzes auf einem MALDI-Instrument erstellt, deren Aussage zur Verifizierung von Suchergebnissen in die Algorithmen implementiert werden kann. Beim Einsatz weniger spezifischer Proteasen erhöht sich im Vergleich zu Trypsin die entstehende Peptidkomplexität. Zur Kompensation dessen wurde eine Fraktionierung mittels Off-gel IEF vor der nLC-MALDI-Analyse etabliert. Zwecks direkter Kompatibilität zur nLC wurde ein Off-gel IEF-Protokoll ohne die Probeninjektion störendes Glycerol entwickelt. So ließ sich die dreifache Menge an Peptiden nachweisen, als mit ausschließlich eindimensionaler nLC-Trennung. In Verbindung mit den durch Trypsin erzielbaren Resultaten lassen sich mit den weniger spezifischen Enzymen viele Membranproteine umfassender charakterisieren, da zusätzlich die hydrophoben Transmembranbereiche und somit mehr Informationen für die Analytik zugänglich sind. Der gezielte Einsatz von alternativen Enzymen verbessert die momentane Situation in der Membranproteom-Analytik deutlich und führt diese einen weiteren Schritt an die Analytik löslicher Proteine heran.
Als Teil der Atmungskette des hyperthermophilen Bakteriums Aquifex aeolicus besteht die NADH:Ubichinon-Oxidoreduktase aus 13 Untereinheiten, die durch 24 unterschiedliche Gene codiert werden. Im Rahmen dieser Arbeit wurde für den massenspektrometrischen Nachweis dieser Untereinheiten nach der Aufreinigung des Komplexes eine SDS-PAGE durchgeführt, an die sich eine tryptische Proteolyse und eine Messung per MALDI MS und MS/MS anschlossen. Mit dieser analytischen Strategie war es möglich, jede nicht-homologe Untereinheit nachzuweisen und 20 von 24 genombasierten Isoformen eindeutig zu identifizieren. Selbst kleine bzw. sehr hydrophobe Untereinheiten mit vielen Transmembranhelices konnten mit Hilfe dieser Methode analysiert werden. Die hohe Anzahl an präsenten, unterschiedlichen Isoformen und die vorherrschenden Verunreinigungen durch Fremdproteine (z.B. ATP-Synthase) könnten Indizien dafür sein, dass bis jetzt keine hinreichend stabile und reproduzierbare Enzympräparation gelungen ist, um eine Kristallstrukturanalyse durchzuführen. Im Allgemeinen kann der Ablauf eines solchen Proteomics-Experiments in drei Teile gegliedert werden: die Auftrennung eines Protein- oder Peptidgemischs zur Verringerung der Probenkomplexität, die Identifikation des Proteins per Massenspektrometrie und die damit verbundene bioinformatische Datenanalyse. Neben chromatographischen Methoden ist die Elektrophorese in Polyacrylamidgelen immer noch das am häufigsten angewandte Trennverfahren im Proteomics-Bereich. Vor der MS-Messung werden die aufgetrennten Proteine in der Regel direkt in der Gelmatrix verdaut. Die Qualität der Ergebnisse wird stark durch Proteinmenge und die auftretenden Probenverluste beeinflusst. Diese werden beispielsweise durch eine unzureichende Peptidextraktion aus dem Gel hervorgerufen. Im Rahmen dieser Dissertation wurde ein neues Gelsystem entwickelt, dass vor allem für die Proteolyseeffizienz, die Peptidextraktion und somit auch für die anschließende massenspektrometrische Messung von Vorteil ist. Das Monomer Acrylamid wird hierzu mit den beiden Quervernetzern N,N-Methylenbisacrylamid (MBA) und Ethylenglykoldiacrylat (EDA) polymerisiert. Es entsteht ein Gel, dessen Poren durch die basische Hydrolyse der Esterbindungen des EDAs vergrößert werden können, ohne die dreidimensionale Grundstruktur des Gels zu zerstören.   Die gemischten Gele mit MBA und EDA als Crosslinkern sind sowohl für Tris-Glycin- als auch Tris-Tricin-Puffersysteme einsetzbar. Die Evaluierung der Daten erfolgte meist gegen ein Standardgel mit T= 14% und C= 2,6%. Durch eine experimentell ermittelte Anpassung der Mischgele auf T+10% und C+45% mit einem MBA/EDA-Verhältnis von 0,5 verhalten sich die beiden Gelsysteme empirisch gleich und zeigen eine übereinstimmende elektrophoretische Trennung und Auflösung. Es gibt keine wesentlichen Nachteile in Bezug auf die Handhabung, die elektrophoretische Trennung und die anschließenden Analysen und Techniken, solange diese in sauren bis leicht basischen pH-Bereichen durchgeführt werden. So wurde das gemischte Gelsystem sowohl bei einer 2D IEF/SDS-PAGE als auch bei einem Western Blot-Experiment angewendet und für vollständig kompatibel befunden. Die gemessenen MS-Daten profitieren im Fall von Trypsin und Elastase erheblich von der besseren Zugänglichkeit des Enzyms zum Substratprotein und der verbesserten Peptidextraktion. Die Gelaufweitung führt zu signifikant besseren Ergebnissen, die größtenteils auf höhere S/N-Werte zurückzuführen sind. Die Signalintensitäten der Peptide sind um den Faktor 5 besser als die des Referenzgels. Der Nachweis von je 1 pmol der Proteine BSA, Serotransferrin und Alkoholdehydrogenase profitiert stark von den zusätzlich erhaltenen Daten, da die Zunahme an identifizierten Peptiden bei der PMF-Suche im Bereich von 40 bis 80% liegt. Auch bei geringen Proteinmengen wie 250 bzw. 50 fmol BSA ist die Anzahl der zugeordneten Signale um den Faktor 2-3 besser. Der in-Gel Verdau von 1 pmol Bacteriorhodopsin mit Elastase zeigt ebenfalls einen Anstieg der Peptidsignale um 60% im Vergleich zum Referenzgel. Da die Proteine im Gel alle mit kolloidalem Coomassie Brilliant Blue angefärbt wurden, kann davon ausgegangen werden, dass sich die guten MS-Ergebnisse auf jede mit diesem Farbstoff visualisierte Probe übertragen lassen. Für den Fluoreszenzfarbstoff RuPBS trifft dies ebenso zu, insgesamt wurden aber weniger Peptide als bei kolloidaler Coomassie Brilliant Blue-Färbung detektiert. Dementsprechend handelt es sich bei Acrylamidgelen mit MBA/EDA-Vernetzung um ein vielversprechendes alternatives Gelsystem, dass auf eine Vielzahl anderer Methoden angewendet werden kann.
Die vorliegende Arbeit analysierte die Behandlung von Patienten mit Infektionserkrankungen am Zentrum der Zahn-, Mund- und Kieferheilkunde des Klinikums der Johann Wolfgang Goethe-Universität, Frankfurt am Main (Carolinum). Dabei wurde vor allem untersucht, welche Auswirkungen die Reduzierung von Personalressourcen speziell für die Behandlung dieser Patienten ab dem Jahr 2000 hinsichtlich der Betreuung und Versorgung hatte. Die Studie war retrospektiv angelegt und wertete Daten aus den Jahren 1998 bis 2002 aus. Hierfür wurden alle im Archiv lagernden Karteikarten herangezogen und die Daten in eine dafür entwickelte FilemakerPro©-Datenbank übertragen. Im Untersuchungszeitraum nahmen 940 Patienten mit Infektionskrankheiten etwa 3700 Besuche wahr. Regelmäßig erschienen 25% der Patienten, auf sie entfielen 60% aller Besuche. Diese Gruppe wurde einer näheren Betrachtung unterzogen: Die Auswertung der erhobenen Daten zeigte, dass das Ziel der Absenkung der Patientenzahlen und die Anzahl der Behandlungstermine erreicht werden konnte, die systematische Betreuung der Patienten sich jedoch verschlechterte. Der Anteil der sanierten Patienten sank von 34% auf 18% ab, die Zahl der unsystematisch behandelten Patienten verdreifachte sich dagegen von 1999 auf 2000 und blieb bei diesem hohen Wert. Vorsorgebehandlungen nahmen maximal ein fünftel aller Behandlungen ein, mit abnehmender Systematik sank dieser Anteil gegen null. Begünstigender Faktor für eine Sanierung waren ein erstelltes OPG (ersatzweise ein Zahnfilmstatus) und die Erstellung einer Behandlungsplanung. Das Vorhandensein eines OPGs erhöhte aber nicht die Wahrscheinlichkeit für eine folgende Behandlungsplanung. Patienten die an das Carolinum von außerhalb überwiesen wurden, hatten eine größere Chance auf eine Sanierung. Letztlich wurden nur 4% aller regelmäßig erschienenen Patienten systematisch mit Recall betreut, zahnärztlich saniert wurden insgesamt jedoch 30% der behandelten Patienten. Entgegen allgemeiner Annahmen waren kurzfristig abgesagte oder nicht eingehaltene Termine die Ausnahme.
5-Lipoxygenase (5-LO) katalysiert die ersten beiden Schritte in der Biosynthese der Leukotriene, die an Reaktionen des Immunsystems beteiligt sind. Die 5-LO-Aktivität wird durch verschiedene Faktoren wie Ca2+, Phospholipide und den zellulären Redoxstatus beeinflusst. Die 5-LO besteht aus einer katalytischen und einer N-terminalen, C2-ähnlichen Domäne. Durch Oxidation des Eisens im aktiven Zentrum zur Fe3+-Form wird das Enzym aktiv. Die C2-ähnliche Domäne bindet Ca2+ und PC und ist für die Membranbindung der 5-LO verantwortlich. In der vorliegenden Arbeit wurde die Aktivierung der 5-LO durch das Diacylglycerid OAG untersucht. Bekannt war die Stimulation der Agonist-induzierten 5-LO-Aktivität in intakten Zellen durch OAG, die nicht auf den bekannten Aktivierungswegen wie Translokation, Phosphorylierung oder Ca2+-Mobilisierung beruht. Hier kann nun die direkte Aktivierung des 5-LO-Enzyms durch OAG in vitro gezeigt werden. Untersucht man den Einfluss von OAG auf die 5-LO-Aktivität in Anwesenheit von 1 mM Ca2+, lässt sich weder an Homogenat, S100 noch am partiell aufgereinigten Enzym aus PMNL ein Effekt beobachten. Entfernt man dagegen Ca2+ aus den Versuchsansätzen, induziert OAG (30 µM) bei Substratkonzentrationen unter 20 µM AA die 5-LO-Aktivität im S100 und am partiell aufgereinigten Enzym. Auch andere Glyceride wie DOG, OG und EAG aktivieren die 5-LO, nicht jedoch SAG. OAG stabilisiert die 5-LO-Aktivität vor der Hemmung durch die Glutathionperoxidase (GPx), dies vermögen ebenfalls andere Glyceride. Damit ähnelt der Einfluss von OAG auf die 5-LO dem bereits bekannten Effekt von Ca2+: es ist in der Lage, vor allem bei niedrigen Substratkonzentrationen die 5-LO-Produktmenge direkt zu erhöhen und kann die 5-LO-Aktivität gegen den inhibitorischen Effekt der Glutathionperoxidase (GPx) stabilisieren. Ähnlich wie Ca2+ scheint OAG die Affinität der 5-LO für das Substrat AA zu erhöhen und das Bedürfnis für aktivierende Hydroperoxide zu erniedrigen. Durch Untersuchungen im Rahmen dieser Arbeit kann eine Beteiligung der Ca2+-Bindungsstelle an der Interaktion zwischen 5-LO und OAG ausgeschlossen werden, da OAG auch an der loop2 mut-5LO die Aktivität zu steigern vermag. Der Anstieg der 5-LO-Produktbildung durch OAG in S100 und am partiell aufgereinigten 5-LO-Enzym lässt sich durch die Zugabe von PC und anderen Phospholipiden unterbinden. Damit erklärt sich gleichzeitig, weshalb OAG am Homogenat aus PMNL keine Effekte zeigt, da hier noch Zellmembran-Bestandteile enthalten sind. Der OAG-Effekt wird wohl über die drei Tryptophan-Reste Trp-13,-75 und -102 der Phospholipid-Bindungsstelle auf der C2-ähnlichen Domäne der 5-LO vermittelt. Dies zeigen auch Untersuchungen an der 3W mut-5LO, bei der die drei Tryptophan-Reste zu Alanin-Resten mutiert sind. Die Aktivität dieser Mutante lässt sich durch OAG unter den bereits beschriebenen Versuchsbedingungen nicht steigern. Ein weiterer Hinweis auf die Phospholipid-Bindungsstelle ist die Interaktion zwischen OAG und dem 5-LO-Inhibitor Hyperforin. Hyperforin selbst hemmt die 5-LO über diese Bindungsstelle, durch OAG wird der IC50-Wert von Hyperforin deutlich erhöht. Im Rahmen dieser Arbeit wurde ein Lipid Protein Overlay Assay für 5-LO ausgearbeitet, ein direkter Nachweis der Bindung zwischen OAG und 5-LO ist aber bisher nicht gelungen. OAG steigert nicht die Aktivität des 5-LO-Enzyms aus serumfrei kultivierten MM6-Zellen, BL41-E95-A Zellen und transfizierten HeLa-Zellen, auch die Produktmenge der Sojabohnen-LO kann nicht durch OAG beeinflusst werden. Untersucht man die Interaktion von OAG mit aufgereinigter regulatorischer Domäne (MBP-Fusionsprotein, MBP-5LO1-128), so zeigt sich eine weitere Steigerung der durch OAG-induzierten 5-LO-Aktivität, obwohl MBP-5LO1-128 bei höheren Konzentrationen eine Hemmung der 5-LO-Aktivität durch Reduktion der gebildeten 5-HPETE verursacht. Diese Ergebnisse lassen sich damit erklären, dass OAG das Bedürfnis der 5-LO für Hydroperoxide senkt und damit geringere Mengen an 5-HPETE zur Aktivierung der 5-LO ausreichen. Betrachtet man das 5-HETE/5-HPETE-Verhältnis, führt die Zugabe OAG in Abwesenheit von Ca2+ und bei 10 µg MBP-5LO1-128 zu einer geringen Abnahme des 5-HETE-Anteils. Im zweiten Teil der Arbeit wurden neue 5-LO-Inhibitoren identifiziert. Aus der Reihe der Pirinixinsäure-Derivate konnte durch entsprechende Modifikationen der potente 5-LO-Inhibitor LP 119 mit einem IC50-Wert von 0,6 µM in intakten PMNL-Zellen identifiziert werden. Durch systematisches Durchsuchen virtueller Datenbanken konnten mindestens zwei Substanzen gefunden werden, die sowohl an intakten Zellen wie auch am partiell aufgereinigten Enzym die 5-LO-Aktivität hemmen, weitere Untersuchungen ergaben, dass es sich wohl zumindest bei einer Strukturklasse um einen direkten 5-LO-Inhibitor handelt.
Die vorliegende Arbeit bietet zunächst einen weiteren Beweis für die Existenz des neutralen Heliumdimers. Darüber hinaus konnten zwei verschiedene Prozesse identifiziert werden, über die die Absorbtion eines Photons zur Ionisation beider Atome des Dimers über sehr große Abstände führen kann. Oberhalb einer Photonenenergie von 65,4 eV konnte ein ICD Prozess beobachtet werden, der über Photoionisation mit gleichzeitiger Anregung von einem der beiden Atome realisiert wird. Bei 77,86 eV konnte ICD über elektronisch angeregte Zustände bis n=6 nachgewiesen werden. In der KER-Verteilung konnten zudem Strukturen gefunden werden, die auf Vibrationsanregungen im Zwischenzustand des Dimer-Ions schließen lassen. Eine vollständig quantenmechanische Rechnung von Sisourat et al. konnte dies schließlich hervorragend bestätigen. Es konnte also ein direkter Blick auf die Vibrationswellenfunktionen des Systems erlangt werden. In anderen Systemen ist dies in der Regel nicht möglich, da sich alle Zustände üblicherweise zu einer strukturlosen Verteilung überlagern. Weiterhin konnte gezeigt werden, dass sich die Winkelverteilungen von ICD- und Photoelektronen in verschiedenen Bereichen des KER mitunter stark voneinander unterscheiden. Dies konnte auf die unterschiedliche Besetzung von verschiedenen Potentialkurven zurückgeführt werden. Unterhalb der Photonenenergieschwelle zur Anregung und Ionisation eines Heliumatoms konnte ein weiterer, zweistufiger Ionisationsmechanismus gefunden werden. Hier wird zunächst durch Photoionisation ein Elektron aus einem der beiden Atome im Dimer freigesetzt. Dieses Photoelektron kann nun am neutralen Atom gestreut werden und dabei ausreichend viel Energie übertragen, um dieses ebenfalls zu ionisieren. Es konnte gezeigt werden, dass der Prozess einer Abhängigkeit von der Polarisation der Synchrotronstrahlung unterliegt, die man für Photoionisation erwarten würde. Die Energie- und Winkelverteilungen der Elektronen konnten daher mit vorangegangenen Elektronenstoß-Experimenten verglichen werden. Die gute Übereinstimmung mit diesen Daten rechtfertigt eine anschauliche Sichtweise des Prozesses als Analogon zum klassischen Billiard-Stoß. Der Two-Step-Prozess wurde bisher zwar schon in vielen Systemen als theoretisches Modell zur Doppelionisation beschrieben, allerdings konnten die einzelnen Unterprozesse bisher nicht gesondert gemessen werden. Die großen Abstände im Heliumdimer ermöglichen erstmals eine deutliche Trennung in Photoionisation an einem Atom und Elektronenstoß (e,2e) am Nachbaratom. Der Two-Step-Prozess konnte außerdem dazu verwendet werden, die ungewöhnliche Grundzustandswellenfunktion des Heliumdimers zu experimentell zu bestätigen. Eine Analyse des gemessenen KER konnte dabei deutliche Abweichungen zu einer klassischen Theorie aufzeigen. Erst eine vollständig quantenmechanische Rechnung des Übergangs von Sisourat et al. konnte die Messdaten beschreiben.
Von einem darwinschen Standpunkt aus kritisiere ich, dass zwar viele Bausteine geliefert werden, es jedoch an einem Fundament fehlt, auf welchem mit diesen Steinen zu bauen sei. Die Begriffe „Recht“ und „Verhalten“ seien durchgängig unbefriedigend, wenn überhaupt, defi-niert. Unrecht und Normbruch würden nicht hinlänglich unterschieden. Das Verhältnis von Sein und Sollen bleibe unklar. Schließlich seien durchgängig Ausbeutung, Unterdrückung und Manipulation von Präferenzen kein Thema. Die psychoanalytische Perspektive der „gesellschaftlichen Produktion von Unbewusstheit“ (Erdheim) fehle gänzlich.
Ich kritisiere die Konzeption Amstutz’ aus einer Perspektive, die eine kulturelle Evolution im strikten Sinne als gegeben ansieht. Gemessen daran verspielt Amstutz den Nutzen evolutionären Denkens, indem er sich nur metaphorisch auf Evolution bezieht, ohne zugleich seinen Rechtsbegriff zu entwickeln. Das führt zu wissenschaftlich unbefriedigenden Aussagen, zumal saltationistische, quasi-lamarckistische und gruppenselektionistische Modelle genutzt werden, die weder für die biologische noch die kulturelle Evolution brauchbar sind.
Ich setze voraus, dass Erfordernisse auf Wissenschaft beinhalten: (1) nicht-physische Ursachen für physische Wirkungen auszuschließen; (2) auch unbewiesene Annahmen und ungeprüfte Dogmen auszuschließen; (3) politisch autonom zu sein; (4) sich auf eine methodische Grundlage zu stützen und (5) im Rahmen der sog. vertikalen Konzeptintegration' zu arbeiten. Ich zeige, dass Strafrechtsdogmatik und Kriminologie, wenn sie Gesellschaft, Recht und die Verbindungen zwischen ihnen beschreiben, Subjekte behaupten, die es nicht gibt. Sie entkleiden Objekte ihrer natürlichen Qualitäten und schreiben ihnen außernatürliche Qualitäten zu. Außerdem behaupten sie Mittel im Kontext dieser Subjekt-Objekt-Beziehungen, die magisch sind, weil ihnen nachvollziehbare natürliche Wirkungen fehlen.
Background: The potential anti-cancer effects of mammalian target of rapamycin (mTOR) inhibitors are being intensively studied. To date, however, few randomised clinical trials (RCT) have been performed to demonstrate anti-neoplastic effects in the pure oncology setting, and at present, no oncology endpoint-directed RCT has been reported in the high-malignancy risk population of immunosuppressed transplant recipients. Interestingly, since mTOR inhibitors have both immunosuppressive and anti-cancer effects, they have the potential to simultaneously protect against immunologic graft loss and tumour development. Therefore, we designed a prospective RCT to determine if the mTOR inhibitor sirolimus can improve hepatocellular carcinoma (HCC)-free patient survival in liver transplant (LT) recipients with a pre-transplant diagnosis of HCC. Methods: The study is an open-labelled, randomised, RCT comparing sirolimus-containing versus mTOR-inhibitor-free immunosuppression in patients undergoing LT for HCC. Patients with a histologically confirmed HCC diagnosis are randomised into 2 groups within 4-6 weeks after LT; one arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol and the second arm is maintained on a centre-specific mTOR-inhibitor-free immunosuppressive protocol for the first 4-6 weeks, at which time sirolimus is initiated. A 3-year recruitment phase is planned with a 5-year follow-up, testing HCC-free survival as the primary endpoint. Our hypothesis is that sirolimus use in the second arm of the study will improve HCC-free survival. The study is a non-commercial investigator-initiated trial (IIT) sponsored by the University Hospital Regensburg and is endorsed by the European Liver and Intestine Transplant Association; 13 countries within Europe, Canada and Australia are participating. Discussion: If our hypothesis is correct that mTOR inhibition can reduce HCC tumour growth while simultaneously providing immunosuppression to protect the liver allograft from rejection, patients should experience less post-transplant problems with HCC recurrence, and therefore could expect a longer and better quality of life. A positive outcome will likely change the standard of posttransplant immunosuppressive care for LT patients with HCC. Einleitung: Schwer verletzte Patienten nach Trauma (ISS > 16) sind häufig in Folge der Verletzungen über mehrere Tage beatmet. 40% dieser Patienten weisen eine Lungenkontusion auf. Mit zunehmender Beatmungsdauer steigt das Risiko einer Ventilator-assoziierten Pneumonie (VAP). Zeitgleich findet eine Reparation des Lungengewebes statt. Eine zeitnahe antiinfektive Therapie bei Verdacht auf eine VAP zu initiieren ist schwierig. Derzeit existiert kein validierter Parameter oder Score, der eine sichere Diskriminierung zwischen Infektion und Inflammation zulässt. Triggering receptor on myeloid cells (TREM-1) ist ein Rezeptor des angeborenen Immunsystems und wurde im Jahr 2000 erstmalig beschrieben. Sein löslicher Anteil, sTREM-1, ist in der bronchoalveolären Lavage (BAL) bei Patienten mit Pneumonie signifikant erhöht (> 200pg/ml). Es liegen keine Daten zu sTREM-1 bei Patienten nach Lungenkontusion vor. Unklar ist, ob sTREM-1 als Pneumonie-Marker nach Lungenkontusion geeignet ist. Material & Methoden: Nach Zustimmung der Ethikkommission und Einwilligung durch einen Angehörigen wurden prospektiv 42 Patienten mit Thoraxtrauma rekrutiert. Am ersten (im Median 15h nach dem Trauma) und an den Behandlungstagen zwei, drei, fünf, sechs und sieben wurden bei allen Patienten über den Tubus mit einem Aero-Jet Katheter BAL (20ml Spülung) gewonnen und zeitgleich Serumproben entnommen. Die Messung der sTREM-1-Konzentration erfolgte mittels Sandwich-ELISA in Doppelbestimmung (Quantikine sTREM-1 Immunoassay; Firma R&D Systems). Die Serum-Konzentrationen der Interleukine (IL) 6 und 10 sowie des Lipopolysaccharid bindenden Proteins (LBP) wurden mittels Immulite® bestimmt. Die Diagnose Pneumonie wurde retrospektiv mittels Clinical Pulmonary Infection Score (CPIS) gestellt: CPIS > 6 Pneumonie, ≤ 6 keine Pneumonie. Ergebnisse & Diskussion: 15 Stunden nach Trauma wurde der sTREM-1 Spiegel in der BAL, bei im Verlauf pulmonal klinisch unauffälligen Patienten, im Median mit 219pg/ml bestimmt. Im Weiteren stieg sTREM-1 im Median nach 24h auf 575pg/ml an und zeigte ähnliche Konzentrationen im Beobachtungszeitraum. Der Schweregrad der Lungenkontusion korreliert mit der Höhe des sTREM-1-Spiegels in der BAL 40h nach Trauma. Patienten mit schwerer Lungenkontusion (im Median 2240pg/ml) haben signifikant höhere Werte gegenüber Patienten ohne Kontusion (Median 217pg/ml), oder geringer Kontusion (Median 339pg/ml). Am Tag der Diagnosestellung Pneumonie (CPIS > 6, n= 9) zeigten die betroffenen Patienten einen signifikant erhöhten sTREM-1-Spiegel in der BAL (Median 2145pg/ml, p < 0,05) im Vergleich zum Tag vor der Pneumonie (Median 588pg/ml). Wird der cut off für sTREM-1 bei 800pg/ml festgelegt ergibt sich eine Sensitivität von 87% und eine Spezifität von 38%. Eine positive BAL weist im Vergleich zu einer negativen BAL signifikant höhere sTREM-1-Konzentrationen (Median 1492pg/ml vs. 971pg/ml, p < 0,05) auf. Die Sensitivität (85%) ist hoch, die Spezifität (51%) gering. Somit ist sTREM-1 nicht nur durch eine Infektion, sondern auch durch eine Gewebeschädigung mit Einblutung und Inflammation stimulierbar. sTREM-1 ist durch die kontusionsbedingte Stimulation in der ersten Woche nach Trauma ungeeignet, um sicher zwischen einer Pneumonie und einer kontusionsbedingten Inflammation zu unterscheiden. Zytokine und akute Phase Proteine (IL-6, LBP, Procalcitonin) sind bekanntermaßen ebenfalls nicht zur sicheren Diskriminierung einer Infektion geeignet. In Kombination mit sTREM-1 lassen sich jedoch zur Diagnosestellung einer Pneumonie vergleichbare Werte für Sensitivität und Spezifität erreichen wie mittels CPIS Score, wobei der CPIS nur retrospektiv ermittelt werden kann. Die Laborparameter liegen bereits am Tag des Verdachts auf eine Infektion vor. Die klinische Entscheidung zur Initiierung einer Antiinfekitvatherapie korrelierte weder mit dem CPIS noch mit den Inflammationsparametern. Drei von neun Patienten erhielten trotz steigenden Entzündungszeichen und einem CPIS > 6 keine Antiinfektiva. In der Konsequenz könnte eine Kombination aus IL-6 und LBP im Serum, sTREM-1 in der BAL und klinischen Parameter des CPIS eine sensitive und spezifische Entscheidungshilfe für eine antiinfektive Therapie bei Polytrauma und Verdacht auf eine VAP werden.
Epidermal growth factor (EGF) receptor belongs to the broad family of enzymatic receptors called receptor tyrosine kinases (RTKs). Generally, the binding of a ligand to these receptors leads to activation of their intracellular kinase activity that sets in motion a cascade of signaling events. In order to ensure appropriate responses to physiological stimuli, the cell is endowed with the ability to regulate signal transduction via numerous mechanisms such as dephosphorylation of the RTK and its substrates as well as downregulation of the RTK. Activation of EGFR is a potent mitogenic (proliferative) and motogenic (cell motility) signal that plays crucial roles during embryonic development and maintenance of adult tissue. EGFR signaling is primarily regulated by ligand-induced receptor internalization with subsequent degradation in lysosomes. While the complex of proteins that are recruited to EGFR after its activation is well understood, proteins that interact with the receptor in the absence of ligand binding are still not systematically studied. With the goal of identifying novel binding partners of non-activated EGFR, a membrane based yeast-two hybrid screen (MYTH) was conducted. MYTH is based on the principle of in vivo reconstitution of the N-terminus (Nub) and C-terminus (Cub) halves of ubiquitin once brought into close proximity. A chimeric protein consisting of EGFR fused to Cub and a transcription factor was used as a bait to screen Nub-tagged cDNA library. Analysis of resultant yeast transformants revealed a total of 87 proteins to interact with EGFR. Of these only 11 were previously shown to bind to EGFR. A majority of the other proteins were shown to interact with the receptor by yeast retransformation. Fifteen were confirmed to bind to EGFR by coimmunoprecipitation assays in mammalian cells. One of the novel EGFR interactors identified in the screen was histone deacetylase 6 (HDAC6). This deacetylase is localized in the cytoplasm and known to deacetylate alpha-tubulin, HSP90 and cortactin. The juxtamembrane region of EGFR binds to the Cterminus of HDAC6. Functionally, overexpression of wild type HDAC6 stabilized ligand-induced degradation of the receptor. On the other hand, deacetylase deficient or EGFR binding compromised mutants of HDAC6 were able to stabilize EGFR only partially. Downmodulation of HDAC6 expression by RNAi markedly accelerated degradation of the receptor. Taken together, HDAC6 is a negative regulator of EGFR downregulation that is dependent on its deacetylase activity and ability to bind to the receptor. Imaging studies revealed that HDAC6 does not affect internalization of EGFR from the plasma membrane but rather influences the post-endocytic trafficking of the receptor-ligand complex to lysosomes. Pulse-chase experiments using fluorophoretagged EGF showed that EGFR is transported faster towards the peri-nuclear region and delivered to late endosomes rapidly in HDAC6 depleted cells. HDAC6 is demonstrated to act, at least partly, by regulating the acetylation of alpha-tubulin. Upon EGFR activation, acetylation of alpha-tubulin on lysine 40 is progressively increased as shown by mass spectrometry and immunoblotting. Forced expression of a dominant negative mutant of alpha-tubulin, but not wild type alpha-tubulin, led to reduced speed and processive movement of early endosomes in GFP-Rab5 expressing cells. In a surprising twist, EGFR is able to phosphorylate HDAC6 on Tyr570. Phosphorylation of Tyr570 and Ser568 leads to inactivation of the deacetylase function of HDAC6 as shown by in vivo and in vitro assays. In summary, HDAC6 diminishes EGFR downregulation by slowing the transport of intracellular vesicles. The inhibitory effect is removed once HDAC6 is phosphorylated on key residues. In line with these findings, two recent reports have shown that hyper-acetylation of alpha-tubulin induced by inhibition of HDAC6 increases the transport of brain derived neurotrophic factor and JNK interacting protein-1 in different cell systems. Acetylated microtubules are more efficient in recruiting motor proteins like kinesin-1 and dynein. These findings indicate that HDAC6 plays an important regulatory role in intracellular trafficking pathways. However, several outstanding issues still remain unresolved. How does acetylation of microtubules influence vesicular trafficking? In this regard, the temporal and spatial dynamics of alpha-tubulin acetylation following EGFR activation should be studied. Furthermore, whether HDAC6 affects the trafficking of other endocytic cargos and additional organelles is an interesting question to address.

Recent studies of single nucleotide polymorphisms (SNPs) within the endotoxin receptor and its signaling system showed an association with the risk of disease development. This study aims to examine the response associated with genetic variations of TLR4, the receptor for bacterial LPS, and a central intracellular signal transducer (TIRAP/Mal) on cytokine release and for susceptibility and course of severe hospital acquired infections in distinct patient populations. Methods: Three intensive care units in tertiary care university hospitals in Greece and Germany participated. 375 and 415 postoperative patients and 159 patients with ventilator associated pneumonia (VAP) were included. TLR4 and TIRAP/Mal polymorphisms in 375 general surgical patients were associated with risk of infection, clinical course and outcome. In two prospective studies, 415 patients following cardiac surgery and 159 patients with newly diagnosed VAP predominantly caused by Gram-negative bacteria were studied for cytokine levels in-vivo and after ex-vivo monocyte stimulation and clinical course. Results: Patients simultaneously carrying polymorphisms in TIRAP/Mal and TLR4 and patients homozygous for the TIRAP/Mal SNP had a significantly higher risk of severe infections after surgery (odds ratio (OR) 5.5; confidence interval (CI): 1.34 - 22.64; P = 0.02 and OR: 7.3; CI: 1.89 - 28.50; P < 0.01 respectively). Additionally we found significantly lower circulating cytokine levels in double-mutant individuals with ventilator associated pneumonia and reduced cytokine production in an ex-vivo monocyte stimulation assay, but this difference was not apparent in TIRAP/Mal-homozygous patients. In cardiac surgery patients without infection, the cytokine release profiles were not changed when comparing different genotypes. Conclusions: Carriers of mutations in sequential components of the TLR signaling system may have an increased risk for severe infections. Patients with this genotype showed a decrease in cytokine release when infected which was not apparent in patients with sterile inflammation following cardiac surgery.

Nanocarbon structures, such as fullerenes and nanotubes, have generated considerable interest and research, due to their unique properties and potential applications. In this thesis, we present a study of the phase transition properties of nanocarbon clusters,in particular, we pay special consideration to fullerenes. The work presented in this thesis is largely theoretical and computational in nature, employing as a tool, molecular dynamics simulations to probe the dynamic stability of fullerenes and associated nanocarbon structures such as graphenes and nanotubes.
Blood vessels form de novo through the tightly regulated programs of vasculogenesis and angiogenesis. Both processes are distinct but one of the steps they share is the formation of a central lumen, when groups of cells organized as vascular cords undergo complex changes to achieve a tube-like morphology. Recently, a protein termed epidermal growth factor-like domain 7 (EGFL7) was described as a novel endothelial cell-derived factor involved in the regulation of the spatial arrangement of cells during vascular tube assembly. With its impact on tubulogenesis and vessel shape EGFL7 joined the large family of molecules governing blood vessel formation. Only recently, the molecular mechanisms underlying EGFL7's effects have been started to be elucidated and shaping of the extracellular matrix (ECM) as well as Notch signaling might very well play a role in mediating its biological effects. Further, findings in knock-out animal models suggest miR-126, a miRNA located within the egfl7 gene, has a major role in vessel development by promoting VEGF signaling, angiogenesis and vascular integrity. This review summarizes our current knowledge on EGFL7 and miR-126 and we will discuss the implications of both bioactive molecules for the formation of blood vessels.
Background: Up to the 1950s, there was an ongoing debate about the diversity of hereditary optic neuropathies, in particular as to whether all inherited optic atrophies can be ascribed to Leber's hereditary optic neuropathy (LHON) or represent different disease entities. In 1954 W. Jaeger published a detailed clinical and genealogical investigation of a large family with explicit autosomal dominant segregation of optic atrophy thus proving the existence of a discrete disease different from LHON, which is nowadays known as autosomal dominant optic atrophy (ADOA). Since the year 2000 ADOA is associated with genomic mutations in the OPA1 gene, which codes for a protein that is imported into mitochondria where it is required for mitochondrial fusion. Interestingly enough, the underlying mutation in this family has not been identified since then. Results: We have reinvestigated this family with the aim to identify the mutation and to further clarify the underlying pathomechanism. Patients showed a classical non-syndromic ADOA. The long term deterioration in vision in the two teenagers examined 50 years later is of particular note 5/20 to 6/120. Multiplex ligation probe amplification revealed a duplication of the OPA1 exons 7-9 which was confirmed by long distance PCR and cDNA analysis, resulting in an in-frame duplication of 102 amino acids. Segregation was verified in 53 available members of the updated pedigree and a penetrance of 88% was calculated. Fibroblast cultures from skin biopsies were established to assess the mitochondrial network integrity and to qualitatively and quantitatively study the consequences of the mutation on transcript and protein level. Fibroblast cultures demonstrated a fragmented mitochondrial network. Processing of the OPA1 protein was altered. There was no correlation of the OPA1 transcript levels and the OPA1 protein levels in the fibroblasts. Intriguingly an overall decrease of mitochondrial proteins was observed in patients' fibroblasts, while the OPA1 transcript levels were elevated. Conclusions: The thorough study of this family provides a detailed clinical picture accompanied by a molecular investigation of patients' fibroblasts. Our data show a classic OPA1-associated non-syndromic ADOA segregating in this family. Cell biological findings suggest that OPA1 is regulated by post-translational mechanisms and we would like to hypothesize that loss of OPA1 function might lead to impaired mitochondrial quality control. With the clinical, genetic and cell biological characterisation of a family described already more than 50 years ago, we span more than half a century of research in optic neuropathies.
Background: Vaccinia virus strain Lister Elstree (VACV) is a test virus in the DVV/RKI guidelines as representative of the stable enveloped viruses. Since the potential risk of laboratory-acquired infections with VACV persists and since the adverse effects of vaccination with VACV are described, the replacement of VACV by the modified vaccinia Ankara strain (MVA) was studied by testing the activity of different chemical biocides in three German laboratories. Methods: The inactivating properties of different chemical biocides (peracetic acid, aldehydes and alcohols) were tested in a quantitative suspension test according to the DVV/RKI guideline. All tests were performed with a protein load of 10% fetal calf serum with both viruses in parallel using different concentrations and contact times. Residual virus was determined by endpoint dilution method. Results: The chemical biocides exhibited similar virucidal activity against VACV and MVA. In three cases intra-laboratory differences were determined between VACV and MVA - 40% (v/v) ethanol and 30% (v/v) isopropanol are more active against MVA, whereas MVA seems more stable than VACV when testing with 0.05% glutardialdehyde. Test accuracy across the three participating laboratories was high. Remarkably inter-laboratory differences in the reduction factor were only observed in two cases. Conclusions: Our data provide valuable information for the replacement of VACV by MVA for testing chemical biocides and disinfectants. Because MVA does not replicate in humans this would eliminate the potential risk of inadvertent inoculation with vaccinia virus and disease in non-vaccinated laboratory workers.
Introduction: Amniotic fluid harbors cells indicative of all three germ layers, and pluripotent fetal amniotic fluid stem cells (AFSs) are considered potentially valuable for applications in cellular therapy and tissue engineering. We investigated whether it is possible to direct the cell fate of AFSs in vivo by transplantation experiments into a particular microenvironment, the mammary fat pad. This microenvironment provides the prerequisites to study stem cell function and the communication between mesenchymal and epithelial cells. On clearance of the endogenous epithelium, the ductal tree can be reconstituted by the transfer of exogenously provided mammary stem cells. Analogously, exogenously provided stem cells from other tissues can be investigated for their potential to contribute to mammary gland regeneration. Methods: We derived pluripotent murine AFSs, measured the expression of stem cell markers, and confirmed their in vitro differentiation potential. AFSs were transplanted into cleared and non cleared fat pads of immunocompromised mice to evaluate their ability to assume particular cell fates under the instructive conditions of the fat-pad microenvironment and the hormonal stimulation during pregnancy. Results: Transplantation of AFSs into cleared fat pads alone or in the presence of exogenous mammary epithelial cells caused their differentiation into stroma and adipocytes and replaced endogenous mesenchymal components surrounding the ducts in co-transplantation experiments. Similarly, transplantation of AFSs into fat pads that had not been previously cleared led to AFS-derived stromal cells surrounding the elongating endogenous ducts. AFSs expressed the marker protein α-SMA, but did not integrate into the myoepithelial cell layer of the ducts in virgin mice. With pregnancy, a small number of AFS-derived cells were present in acinar structures. Conclusions: Our data demonstrate that the microenvironmental cues of the mammary fat pad cause AFSs to participate in mammary gland regeneration by providing mesenchymal components to emerging glandular structures, but do not incorporate or differentiate into ductal epithelial cells.
This work reports on the study of the projectile x-ray emission in relativistic ion-atom collisions. Excitation of K-shell in He-like uranium ions, electron capture into H-like uranium ions and Simultaneous ionization and excitation of initially He-like uranium ions have been studied using the experimental storage ring at GSI. Information about the population of the excited states for the H- and He-like uranium ions, can be obtained by measuring the angular distribution of the decay radiation. Since the Ly_alpha2 transition is isotropic, the intensities of the Ly_alpha1 and K_alpha transitions were normalized to the Ly_alpha2 line. For the K_alpha1 and K_alpha2 transitions originating from the excitation of the He-like uranium ions, no alignment was observed. In contrast, the Ly_alpha1 radiation from the simultaneous ionization-excitation process of the He-like uranium ions shows a clear alignment. It is shown that the alignment of Ly_alpha1 was obtained by the Alignment parameter A_20. The experimental value leads to the inclusion of a magnetic term in the interaction potential. It is interesting to note that in the case of the Ly_alpha1 emission the small M2 contribution added coherently to the E1 transition amplitudes enhances the anisotropy. The capture process of target electrons into the highly-charged heavy ions was studied using H-like uranium ions at an incident energy of 220 MeV/u, impinging on N2 gas-target. It was shown that, the strongly aligned electrons captured in 2p3/2 level will couple with the available 1s1/2 electron which shows no initial directional preference. The magnetic sub-state population of the 2p3/2 electron will be redistributed according to the coupling rules to the magnetic sub-states of the relevant two-electron states. Consequently, the 1^P1 and 3^P2 states are corresponding to the the strongly aligned 2p3/2 state. This leads to the large anisotropy in the corresponding individual ground state transitions contributing to the K_alpha1 emission. Due to the fact that the 1^P1 --> 1^S0 and 3^P2 --> 1^S0 transitions are experimentally not resolved, a more detailed analysis of the angular dependence of the K_alpha1 radiation is required. From the K_alpha1/K_alpha2 ratio, the current results show that the incoherent addition of the E1 and M2 transition components yield to an almost isotropic emission of the total K_alpha1. In contrast to the radiative electron capture, the experimental results for the K-shell single excitation of He-like uranium ions indicate that only the 1^P1 level contributes to the K_alpha1 transition. For this case, the anisotropy parameter beta_20 was found to be -0.20 + 0.03 which is similar to that one calculated for pure E1 transition. This work also reports on the study of a two-electron process: the simultaneous ionization and excitation occurring in relativistic collisions of heavy highly-charged ions with gaseous targets. The investigation was performed on He-like uranium ions impinging upon xenon gas-target at an incident energy of 220 MeV/u. The measurements have been performed at the ESR gas-target using atomic xenon with a typical area density of 10^12 particles/cm^2. In contrast to the solid state target, the use of gas target offers the advantage of clear separation of the one step two-electron process due to the fact that the probability of two consecutive collision in such thin targets is negligible and the double step processes can be excluded. During the process of simultaneous ionization and excitation in He-like uranium ions, one of the ground-state electrons is promoted into the continuum and the other into the L-subshell states of the projectile. To select this process, the Lyman-series radiation has been measured at various observation angles in coincidence with up-charged projectiles (U^91+). From the yields of the Ly_alpha1 and Ly_alpha2 projectile radiation, the relative cross section for the process of simultaneous ionization and excitation was directly determined. The angle dependent measurement of the radiation yields provide information about the angular distributions of the emitted radiation and permits the determination of the alignment parameter A_{20}. This parameter gives information on the level population and the collision impact parameter. The present results (b^exp = 810 fm) show that the simultaneous ionization and excitation is a process which occurs at small impact parameter.
Poster Presentation from Nineteenth Annual Computational Neuroscience Meeting: CNS*2010 San Antonio, TX, USA. 24-30 July 2010 Statistical models of neural activity are at the core of the field of modern computational neuroscience. The activity of single neurons has been modeled to successfully explain dependencies of neural dynamics to its own spiking history, to external stimuli or other covariates [1]. Recently, there has been a growing interest in modeling spiking activity of a population of simultaneously recorded neurons to study the effects of correlations and functional connectivity on neural information processing (existing models include generalized linear models [2,3] or maximum-entropy approaches [4]). For point-process-based models of single neurons, the time-rescaling theorem has proven to be a useful toolbox to assess goodness-of-fit. In its univariate form, the time-rescaling theorem states that if the conditional intensity function of a point process is known, then its inter-spike intervals can be transformed or “rescaled” so that they are independent and exponentially distributed [5]. However, the theorem in its original form lacks sensitivity to detect even strong dependencies between neurons. Here, we present how the theorem can be extended to be applied to neural population models and we provide a step-by-step procedure to perform the statistical tests. We then apply both the univariate and multivariate tests to simplified toy models, but also to more complicated many-neuron models and to neuronal populations recorded in V1 of awake monkey during natural scenes stimulation. We demonstrate that important features of the population activity can only be detected using the multivariate extension of the test. ...
