Die Rechtsgeschichte hat dem vormodernen Asyl lange Zeit einen bestenfalls marginalen Platz eingeräumt und es häufig als Hindernis auf dem Weg zum staatlichen Gewalt- und Justizmonopol bewertet oder den angeblichen "Missbrauch" des Asyls betont. Gleiches gilt cum grano salis für die allgemeine Geschichte, die wenige, eng begrenzte lokale Fallstudien beigesteuert hat, während umfassendere Darstellungen zur Geschichte der "Menschenrechte" oder zur historischen Kriminalitätsforschung das vormoderne Asylrecht weitgehend ignorieren. Erst in jüngster Zeit nahm die Zahl der Arbeiten zu, die sich intensiver mit der Geschichte des Asyls beschäftigen und neue Erkenntnisse sowie Forschungsperspektiven beitragen. ...
Lange hat die Forschung die zehn Kreise des Heiligen Römischen Reiches deutscher Nation vernachlässigt. Ließen diese sich doch kaum in das Modell der europäischen (National-)Staatsbildung einordnen. Die Reichskreise bildeten eine separate, spezifische verfassungsrechtliche Ebene zwischen der im Reich nur schwach ausgeprägten "Zentralgewalt", repräsentiert durch Kaiser und Reichstag, und den einzelnen Reichsständen. Unter letzteren billigte die ältere Forschung nur den größeren Territorien staatliche Qualitäten zu. Das Reich wurde dagegen als ein zu moderner Staatsbildung unfähiges "Monstrum" abgetan, das insbesondere in der Gesetzgebung sowie in der Außen-, Wirtschafts-, Ordnungs- und Sicherheitspolitik versagt habe. Erst die jüngere Forschung hat gezeigt, dass das Alte Reich als Ganzes und die Reichsmitglieder durchaus staatliche Funktionen ausübten, und zwar auch im Bereich der frühneuzeitlichen Ordnungs- bzw. Policeygesetzgebung. Die Normenproduktion der Reichskreise und kleineren Reichsstände ist allerdings noch kaum erschlossen, und moderne Editionen gerade umfangreicherer, exemplarischer Policeyordnungen fehlen völlig. Die hier vorzustellenden, von Wolfgang Wüst herausgegebenen drei Bände zur "guten Policey im Reichskreis" bilden folglich nicht nur eine wertvolle Ergänzung zu dem im Max-Planck-Institut für europäische Rechtsgeschichte entstandenen Repertorium der frühneuzeitlichen Policeyordnungen und den in diesem Kontext entstandenen Fallstudien, sondern sie verbinden mit den Themen "Reichskreise" und "Policeygesetzgebung" zwei wichtige Felder der Frühneuzeitforschung und eröffnen damit eine neue Perspektive auf die Gesetzgebungsgeschichte und die Entwicklung frühmoderner Staatlichkeit. ...
In der vorliegenden in vitro-Studie wurde der Einfluß von zwei Insertionstechniken auf die zervikale Randqualität von Klasse-II-Kompositrestaurationen unter Zuhilfenahme von Kunststoffmatrizen und Lichtkeilen untersucht. Als weiteren Versuchsparameter wählte man zur Adaptation des Füllungsmaterials neben herkömmlichen Metallinstrumenten zusätzlich modifizierte Biberschwanzpinsel.
Fungi belonging to the Rhytismatales (Ascomycota) are parasites or endophytes of plants, some are saprophytes. Their fruiting bodies are localized in different organs of the host plants belonging to many different families of gymnosperms and angiosperms. Many species of Rhytismatales are known on species of Pinaceae, Ericaceae, and Poaceae. These fungi usually have ascomata that are more or less embedded in host tissue and open by longitudinal or radial splits. They have a more or less carbonized covering stroma, thin-walled, iodine negative asci, and ascospores usually covered by gelatinous sheaths.
In the present study, two lists of species of Rhytismatales in China are presented. One is based on literature and includes 103 species in 15 genera. The second one contains the names of the species in the present study, 57 species in 20 genera based on 90 specimens I collected in the Yunnan and Anhui province in China during July to August in 2001. 31 species in the second list are new species or new records for China, so we presently know 134 species in 22 genera of Rhytismatales for China. 28 new species of Rhytismatales are proposed, 21 species from the Yunnan province and seven from the Anhui province. Among them, three new species are proposed in three new genera, Nematococcomyces, New Genus 1, and New Genus 2, respectively. The 28 new species are Cerion sp., Coccomyces spp. 1-2, Colpoma spp. 1-2, Hypoderma spp. 1-6, Lirula sp., Lophodermella sp., Lophodermium spp. 1-5, Nematococcomyces rhododendri C.-L. Hou, M. Piepenbr. & Oberw., Neococcomyces sp., New Genus 1 sp., New Genus 2 sp., Rhytisma spp. 1-2, Soleella sp., Terriera spp. 1-2, and Therrya sp. The genus Davisomycella is proposed as a synonym of Lophodermella based on observations of the morphology, ecology, and the infected organ. The four genera Cerion, Naemacyclus, Terriera, and Therrya, and three species, Hypoderma rubi, Lophodermium uncinatum, and Naemacyclus pinastri, are reported for the first time for China. All the new taxa, the newly recorded ones, as well as six species which had not been illustrated in detail before, are carefully described and illustrated by line drawings in the present study.
The results show that species of Rhytismatales are highly diverse especially in the natural vegetation in high mountainous areas in China. Most species of Rhytismatales are conspicuously host specific. The diversity of Rhytismatales is closely related to that of the preferred hosts, which are members of Pinaceae, Ericaceae, and Cupressaceae. Based on the detailed morphological observations, the significance of different morphological characteristics for a natural classification of Rhytismatales is discussed. Genera are traditionally defined by character states of a few characteristics, namely the opening patterns of ascomata, the depth of ascomata in the host tissue, and asci and ascospore shape. Data from collections in the field, detailed morphological investigation, and molecular data show, however, that the ecology, the infected organ, the host relationship, and many other characteristics have to be combined to circumscribe natural groups.
The discussion of the systematic significance of morphological characteristics is complemented by molecular data. In the present study, partial nuclear large subunit rDNA sequences of 52 specimens representing 38 species are used to analyse phylogenetic relationships for members of Rhytismatales.
Most species of Rhytismatales are placed in a monophyletic group corresponding to the Rhytismatales in the Maximum Parsimony analysis. The delimitation of the Rhytismatales from the Helotiales is, however, difficult. Cyclaneusma minus should be transferred from the Rhytismatales to the Helotiales, and Cudonia circinans and Spathularia flavida from the Helotiales to the Rhytismatales. These tranfers have previously been proposed based on SSU rDNA analysis by other authors. New Genus 1 sp. has morphological characteristics typical for species of Rhytismatales. In the LSU rDNA analysis, however, it is more closely related to Helotiales rather than toRhytismatales. Therefore New Genus 1 sp. is placed in the Helotiales.
Tryblidiopsis pinastri is morphologically intermediate between members of Rhytismataceae and Cudoniaceae. LSU rDNA sequences in the present study show that T. pinastri is more closely related to species of Cudoniaceae. Therefore, this species is removed from the Rhytismataceae to the Cudoniaceae. The delimitation of further families could not be resolved in the present analysis.
Though many new morphological, ecological, and molecular phylogenetic findings are contributed for the first time, the systematic conclusions at generic, family, and order level can only be fragmentary in the present study. With more collections and more molecular data of the worldwide 450 known and many more unknown species of Rhytismatales at hand, a natural system combining morphological and molecular analysis can be elaborated.
Das westphälische Modell für Staatsinstitutionen, einschließlich nationaler Exekutive, Legislative und Judikative, hat sich aus den Ereignissen europäischer Geschichte heraus entwickelt. Seit dem Ende des Kalten Krieges dient es als grundlegendes Paradigma für Internationale Interventionen zum Wiederaufbau von gescheiterten - oder zum Aufbau von neuen - Staaten. Für die internationale Gemeinschaft fungiert das westphälische Modell als Maß zur Beurteilung ihrer Interventionen, wie zum Beispiel in Somalia, Kambodscha oder den Balkanstaaten. In den meisten Fällen gilt eine durch sie beaufsichtigte oder gar durchgeführte ‚freie und faire’ Wahl als hauptsächliche Massnahme zur Bildung eines ‚westphälischen’ und demokratischen Staates. Die Erfolgsrate solcher internationalen Friedenseinsätze und ‚state-building operations’ ist jedoch enttäuschend. Bei näherer Betrachtung der Misserfolge des letzten Jahrzehnts wird deutlich, daß sich die lokalen Gesellschaftssysteme der betroffenen Bevölkerungen oft beträchtlich von liberaler Demokratie unterscheiden. Dies ist insbesondere der Fall in Gesellschaften deren Ordnung nicht auf Staatsinstiutionen basiert. Ihnen liegen sozio-politische Systeme zugrunde die sich oft mit dem Paradigma des westlichen Staatssystems nur schwer vereinen lassen. Um im Rahmen internationaler Friedenseinsätze erfolgreich Staatstrukturen zu etablieren, ist es daher notwendig lokale Sozialstrukturen und lokale Konzepte politischer Legitimität und Autorität zu addressieren. Erst mit solchem Verständnis ist es möglich einen Staatsapparat in den Augen der Bevölkerung zu legitimieren. Ist Letzteres nicht der Fall, so kann sich eine Regierung zwar in Übereinstimmung mit internationalen Menschenrechten befinden, oder alle wichtigen demokratischen Einrichtungen vorweisen, jedoch dennoch dem Prinzip der Partizipation durch die Bevölkerung widersprechen. Ist dies das Endresultat eines internationalen Friedenseinsatzes, so hat die internationale Gemeinschaft ihre eigenen Werte bestaetigt. Jedoch herrscht kein Vertrauen zwischen der Bevölkerung und Regierung, da letztere nicht kompatibel mit dem Versaendnis der Bürger ist. Der ‚demokratische’ Staat ist nur schwerlich funktionsfähig.Der internationale Einsatz in Osttimor illustriert dieses Problem. Hier wurden die Vereinten Nationen (VN) mit dem Wiederaufbau und der Verwaltung eines Staates betraut (UNTAET ‚Übergangsregierung der Vereinten Nationen in Osttimor’). Zum ersten mal in der Geschichte übernahm die international Gemeinschaft damit die Souveränität über ein territoriales Gebiet...
CFTR ist ein Chloridkanal, der bei der rezessiven Erbkrankheit Mukoviszidose defekt ist. Es ist bekannt, dass CFTR durch Proteinkinasen aktiviert und seine Aktivität durch Nukleotide reguliert wird. Die Regulation von CFTR wurde unter zwei verschiedenen Gesichtspunkten untersucht. Zum einen wurden Experimente durchgeführt, die Aufschluss über die Beteiligung der Nukleotidbindedomänen beim Öffnen und Schließen des Kanals und über die Notwendigkeit einer ATP-Hydrolyse geben sollten. Zum anderen wurde untersucht, ob neben der durch Proteinkinasen vermittelten Aktivierung von CFTR ein alternativer Prozess existiert. Hierbei wurde ein Regulationsmechanismus entdeckt, der eine Proteinkinase-unabhängige Aktivierung von CFTR durch Phosphatidylinositolphosphate ermöglicht.
Humaner CFTR wurde in Oozyten des Krallenfrosches Xenopus laevis heterolog exprimiert und mit der Patch-Clamp-Methode untersucht. Stationäre und zeitaufgelöste Ströme des CFTR-Wildtyps wurden mit mutierten CFTR-Kanälen verglichen. Das Lysin im Walker AMotiv ist an der Koordinierung des γ-Phosphats von MgATP bei der Hydrolyse beteiligt, so dass Walker A-Mutationen die ATP-Bindung und –Hydrolyse von ATPasen beeinflussen. In dieser Arbeit wurden Walker A-Mutanten untersucht, die eine Substitution des konservierten Lysins innerhalb der Walker A-Sequenz der NBD1 (K464A) oder beider Nukleotidbindedomänen (K464A/K1250A) aufwiesen. Da die Öffnungsgeschwindigkeit der Mutante K464A kaum einen Unterschied zu der des Wildtyps aufzeigte, die Mutante K1250A jedoch das Öffnen stark verlangsamte, wurde gefolgert, dass keine Hydrolyse von ATP an der NBD1 für die Öffnung nötig ist. Während Wildtyp-Kanäle auf eine gleichzeitige Applikation von ATP und AMP-PNP, einem nichthydrolysierbaren ATP-Analogon, mit einem verlängerten Offenhalten der Kanäle („locked open“–Effekt) reagierten, das sich in einem langsamen Schließen der Kanäle äußerte, konnte bei K464A-Mutanten dieser Effekt nicht beobachtet werden. Außerdem erfolgte das Schließen der Doppelmutante K464A/K1250A im Vergleich zur Einzelmutante K1250A nach MgATP-Entzug schneller. Daraus wurde geschlossen, dass die NBD1 auf das durch die NBD2 vermittelte Offenhalten des Kanals, möglicherweise durch eine direkte Interaktion, regulierend einwirkt, bevor letztere den Kanal wieder schließt. Da auch ein Öffnen und Schließen des CFTR-Kanals unter Mg2+-freien Bedingungen zu beobachten war, unter denen keine ATP-Hydrolyse erfolgen kann, konnte die Notwendigkeit einer ATP-Hydrolyse bezüglich des Kanalgatings ausgeschlossen werden. Ein Einwirken der NBD1 auf das Offenhalten der Kanäle durch die NBD2 war unter nicht-hydrolytischen Bedingungen anhand des Vergleichs der Schließkinetiken von WT und Mutante K464A nicht feststellbar, so dass eine direkte Interaktion beider Nukleotidbindedomänen wahrscheinlich ausgeschlossen werden kann.
Im zweiten Teil dieser Arbeit wurde der Effekt des Phospholipids Phosphatidylinositol-4,5-bisphosphat (PIP2) auf CFTR-Kanäle untersucht. Die Applikation von PIP2 und MgATP zu unphosphorylierten CFTR-Kanälen zeigte einen deutlichen Stromanstieg, der einem Chloridstrom entsprach. Einzelkanaluntersuchungen ergaben, dass durch PKA induzierte Kanäle und Einzelkanäle, die durch PIP2 aktiviert wurden, dieselbe Leitfähigkeit von ~5 pS besaßen. Somit konnte eine PIP2-induzierte Aktivität endogener Chloridkanäle ausgeschlossen und ein Einfluss des Phospholipids auf CFTR-Chloridkanäle bewiesen werden, der zudem ATP-abhängig war.
Neben PIP2, welches den stärksten Effekt auf die CFTR-Aktivität zeigte, konnten auch Phosphatidylinositol (PI) und Phosphatidylinositol-4-monophosphat (PIP), sowie Arachidonsäure unphosphorylierte CFTR-Kanäle aktivieren. Damit wurde gezeigt, dass der Effekt des Signalanstiegs durch Phosphatidylinositole abhängig von der Struktur des Moleküls war, also von der Anzahl der Phosphatgruppen am Inositolring und der Fettsäurezusammensetzung des Phospholipids.
Experimente, die unter Mg 2+-freien Bedingungen durchgeführt wurden, so dass eine Phosphorylierungsreaktion durch Kinasen ausgeschlossen werden konnte, zeigten dennoch eine PIP2-vermittelte Aktivierung von unphosphorylierten CFTR-Kanälen. Auch eine Substitution des nicht-hydrolysierbaren ATP-Analogons AMP-PNP anstelle von ATP erlaubte die Öffnung unphosphorylierter CFTR-Kanäle. Mit diesen beiden Ergebnissen wurde gezeigt, dass eine PIP2-vermittelte Aktivierung von unphosphorylierten CFTR-Kanälen unabhängig von einer Proteinphosphorylierung ist.
Physiologisch betrachtet könnte man sich vorstellen, dass über die Aktivierung von Lipidkinasen die Synthese von PIP2 über PI und PIP stimuliert wird, so dass das Phospholipid, wie für viele Ionenkanäle und Transporter gezeigt, eine direkte Interaktion mit dem Protein eingeht. Eine ATP-abhängige Synthese von PIP2 in Makropatches an Xenopus-Oozyten durch endogene Lipidkinasen könnte eine mögliche Erklärung für den gezeigten ATP-abhängigen Anstieg des CFTR-Signals sein.
In dieser Arbeit wurde bei CFTR-Kanälen zum ersten Mal ein alternativer Regulationsmechanismus über Phosphatidylinositolphosphate identifiziert, der Proteinkinaseunabhängig ist und der möglicherweise über eine direkte Interaktion zwischen dem Phospholipid und dem Protein vermittelt wird.
Case numbers of endemic Ca-deficiency rickets (CDR) have been reported to be alarmingly rising among children of subsistence farms in developing countries within the last 30 years. Fluoride toxicities in the environment are known to not be related to the disease. To investigate if, instead, CDR is caused by a nutrient deficiency in the environment, subsistence farms in an endemic CDR area near Kaduna, northern Nigeria, were investigated for bedrock, slope forms, soil types, and soil characteristics. The natural environment was investigated according to the World Reference Base, soil texture was analysed by pipette and sieving, and plant-available macronutrients were determined using barium-chloride or Ca-acetate-lactate extraction. The analyses showed that granite and slope deposits were the dominant parent materials. The typical slope forms and soil types were Lixisols and Acrisols on pediments, Fluvisols in river valleys, and Plinthosols and Acrisols on plains. Compared with West African background values, all of the soils had normal soil textures but were low in macronutrients. Comparisons to critical limits, however, showed that only the P concentrations were critically low, which are typical for savanna soils. A link between nutrient deficiency in soils and CDR in the Kaduna area was therefore considered unlikely.
Modifications of the gyromagnetic moment of electrons and muons due to a minimal length scale combined with a modified fundamental scale Mf are explored. First-order deviations from the theoretical SM value for g−2 due to these string theory-motivated effects are derived. Constraints for the fundamental scale Mf are given.
Eine in Verfassungslehre und Europarechtswissenschaft weit verbreitete Annahme lautet: "Europa muss, um eine Zukunft haben zu können, sich zu einer Geschichtsgemeinschaft entwickeln." Was man dabei übersieht: So wenig sich nationale Geschichtsgemeinschaften identifizieren lassen, so wenig wird sich eine europäische Geschichtsgemeinschaft konstituieren.
We study issues of duality in 3D field theory models over a canonical noncommutative spacetime and obtain the noncommutative extension of the self-dual model induced by the Seiberg–Witten map. We apply the dual projection technique to uncover some properties of the noncommutative Maxwell–Chern–Simons theory up to first-order in the noncommutative parameter. A duality between this theory and a model similar to the ordinary self-dual model is established. The correspondence of the basic fields is obtained and the equivalence of algebras and equations of motion are directly verified. We also comment on previous results in this subject.

Szene aus einer Montage-Fassung des Dramenfragments "Moloch" von Friedrich Hebbel. Die Szene steht als Einschub zwischen dem Ersten und Zweiten Akt der Tragödie, als eine Art Interludium. Im Laufe des Stückes wechselt die Perspektive mehrmals zwischen den Schauplätzen Germanischer Wald und Rom, dem Zentrum des Imperiums. Geplant ist eine Aufführung der Gesamtbearbeitung an der Volksbühne Berlin, voraussichtlich Ende 2004.
The majority of bacterial membrane-bound NiFe-hydrogenases and formate dehydrogenases have homologous membrane-integral cytochrome b subunits. The prototypic NiFe-hydrogenase of Wolinella succinogenes (HydABC complex) catalyzes H2 oxidation by menaquinone during anaerobic respiration and contains a membrane-integral cytochrome b subunit (HydC) that carries the menaquinone reduction site. Using the crystal structure of the homologous FdnI subunit of Escherichia coli formate dehydrogenase-N as a model, the HydC protein was modified to examine residues thought to be involved in menaquinone binding. Variant HydABC complexes were produced in W. succinogenes, and several conserved HydC residues were identified that are essential for growth with H2 as electron donor and for quinone reduction by H2. Modification of HydC with a C-terminal Strep-tag II enabled one-step purification of the HydABC complex by Strep-Tactin affinity chromatography. The tagged HydC, separated from HydAB by isoelectric focusing, was shown to contain 1.9 mol of heme b/mol of HydC demonstrating that HydC ligates both heme b groups. The four histidine residues predicted as axial heme b ligands were individually replaced by alanine in Strep-tagged HydC. Replacement of either histidine ligand of the heme b group proximal to HydAB led to HydABC preparations that contained only one heme b group. This remaining heme b could be completely reduced by quinone supporting the view that the menaquinone reduction site is located near the distal heme b group. The results indicate that both heme b groups are involved in electron transport and that the architecture of the menaquinone reduction site near the cytoplasmic side of the membrane is similar to that proposed for E. coli FdnI.
Chronic obstructive pulmonary disease (COPD) is a major global health problem and is predicted to become the third most common cause of death by 2020. Apart from the important preventive steps of smoking cessation, there are no other specific treatments for COPD that are as effective in reversing the condition, and therefore there is a need to understand the pathophysiological mechanisms that could lead to new therapeutic strategies. The development of experimental models will help to dissect these mechanisms at the cellular and molecular level. COPD is a disease characterized by progressive airflow obstruction of the peripheral airways, associated with lung inflammation, emphysema and mucus hypersecretion. Different approaches to mimic COPD have been developed but are limited in comparison to models of allergic asthma. COPD models usually do not mimic the major features of human COPD and are commonly based on the induction of COPD-like lesions in the lungs and airways using noxious inhalants such as tobacco smoke, nitrogen dioxide, or sulfur dioxide. Depending on the duration and intensity of exposure, these noxious stimuli induce signs of chronic inflammation and airway remodelling. Emphysema can be achieved by combining such exposure with instillation of tissue-degrading enzymes. Other approaches are based on genetically-targeted mice which develop COPD-like lesions with emphysema, and such mice provide deep insights into pathophysiological mechanisms. Future approaches should aim to mimic irreversible airflow obstruction, associated with cough and sputum production, with the possibility of inducing exacerbations.
We have studied the ubiquinone-reducing catalytic core of NADH:ubiquinone oxidoreductase (complex I) from Yarrowia lipolytica by a series of point mutations replacing conserved histidines and arginines in the 49-kDa subunit. Our results show that histidine 226 and arginine 141 probably do not ligate iron-sulfur cluster N2 but that exchanging these residues specifically influences the properties of this redox center. Histidines 91 and 95 were found to be essential for ubiquinone reductase activity of complex I. Mutations at the C-terminal arginine 466 affected ubiquinone affinity and inhibitor sensitivity but also destabilized complex I. These results provide further support for a high degree of structural conservation between the 49-kDa subunit of complex I and its ancestor, the large subunit of water-soluble [NiFe] hydrogenases. In several mutations of histidine 226, arginine 141, and arginine 466 the characteristic EPR signatures of iron-sulfur cluster N2 became undetectable, but specific, inhibitor-sensitive ubiquinone reductase activity was only moderately reduced. As we could not find spectroscopic indications for a modified cluster N2, we concluded that these complex I mutants were lacking most of this redox center but were still capable of catalyzing inhibitor-resistant ubiquinone reduction at near normal rates. We discuss that this at first surprising scenario may be explained by electron transfer theory; after removal of a single redox center in a chain, electron transfer rates are predicted to be still much faster than steady-state turnover of complex I. Our results question some of the central mechanistic functions that have been put forward for iron-sulfur cluster N2.
Teaching information literacy: substance and process This presentation explores the concept of information literacy within the broader context of higher education. It argues that, certain assertions in the library literature notwithstanding, the concepts associated with information literacy are not new, but rather very closely resemble the qualities traditionally considered to characterize a well-educated person. The presentation also considers the extent to which the higher education system does indeed foster the attributes commonly associated with information literacy. The term information literacy has achieved the immediacy it currently enjoys within the library community with the advent of the so-called "information age" The information age is commonly touted in the literature, both popular and professional, as constituting nothing short of a revolution. Academic librarians and other educators have of course felt called upon to make their teaching reflect both the growing proliferation of information formats and the major transformations affecting the process of information seeking. Faced with so much novelty and uncertainty, it is no surprise that many have felt that these changes call for a revolution in teaching. It is within this context that the concept of information literacy has flourished. It is argued in this presentation, however, that by treating information literacy as an essentially new specialty that owes much of its importance to the plethora of electronic information, we risk obscuring some of the most fundamental and enduring educational values we should be imparting to our students. Much of the literature on information literacy assumes - rather than argues - that recent changes in the way we approach education are indications of progress. Indeed, much of the self-narrative that institutions produce (in bulletins, mission statements, web sites, etc.) endorses an approach to education that will result in lifelong learners who are critical consumers of information. After critically examining the degree to which such statements of educational approach reflect reality, this presentation concludes by considering the effects of certain changes in the culture of higher education. It considers particularly the transformation - at least in North America - of the traditional model of higher education as a public good to a market-driven business model. It poses the question of whether a change of this significance might in fact detract from, rather than promote, the development of information literate students.
We suggest that the fluctuations of strange hadron multiplicity could be sensitive to the equation of state and microscopic structure of strongly interacting matter created at the early stage of high energy nucleus-nucleus collisions. They may serve as an important tool in the study of the deconfinement phase transition. We predict, within the statistical model of the early stage, that the ratio of properly filtered fluctuations of strange to non-strange hadron multiplicities should have a non-monotonic energy dependence with a minimum in the mixed phase region.

Die Photodynamische Therapie (PDT) wird mittlerweile bei einer Vielzahl von Erkrankungen eingesetzt. Ziel dieser Dissertation war die nähere Untersuchung der Kinetik und der Wirkmechanismen der Photosensibilisatoren Methylenblau und disulfoniertem Aluminiumphthalocyanin. Zuerst klärten wir die Frage der Toxizität des Methylenblaus. Wir ermittelten dabei die für die nachfolgenden Versuche nötigen Dosen und Höchstdosen des Methylenblaus in Bezug auf die humane Keratozyten-Linie HaCat und periphere mononukleäre Zellen. Für disulfoniertes Aluminiumphthalocyanin stützten wir uns auf vorhandene Publikationen. Als Lichtquelle benützten wir die PDT Lampe der Firma Waldmann, die ein homogenes Lichtspektrum von 600 bis 700 nm erzeugt, so dass das Wirkungsmaximum aller gängigen Photosensibilisatoren abgedeckt ist. Ausserdem liefert diese Lampe eine gleichmässige Energiedichte über eine größere Fläche, die die Reproduzierbarkeit der Ergebnisse gewährleistet.
In der Arbeit konnte gezeigt werden, dass es für die photodynamische Therapie mit Methylenblau und disulfoniertem Aluminiumphthalocyanin eine Dosis gibt mit der man sowohl Keratinozyten als auch Leukozyten in ihrer Proliferation hemmen kann, ohne eine zytotoxische Wirkung auszulösen. Für Keratinozyten ergab sich dabei ein Anstieg der Proliferationshemmung bei 5 µM Methylenblau und 2stündiger Inkubationszeit bei 200 J/cm², die Toxizität zeigte sich bei 5µM Methylenblau und 4stündiger Inkubationszeit und bereits bei 100 J/cm² maximal. Demgegenüber ergab sich bei stimulierten Leukozyten bereits bei 1µM Methylenblau und 2 Stunden Inkubationszeit ein starker proliferativer Effekt, bei 5µM Methylenblau und 2 Stunden Inkubationszeit zeigte sich dagegen ein deutliche Toxizität. Hierbei fand sich ab 0,5 J/cm² eine zunehmende Proliferationshemmung und Toxizität. Insgesamt war bei Keratinozyten die Differenz bzgl. antiproliferativer und zytotoxischer Dosis geringer als bei Leukozyten. Letztere zeigten sich dabei auch empfindlicher, besonders wenn man die Leukozyten zuvor stimulierte. Daraus ergibt sich ein Potential für den therapeutischen Einsatz der Photodynamischen Therapie bei entzündlichen Dermatosen.
Als mögliche Wege indirekt toxischer Wirkung wurde in der Folge die Stimulation des nukleären Transkriptionsfaktors NF-ΚB, die Bildung von Stickstoffmonoxid (NO) und der protektive Effekt von α-Liponsäure untersucht. Dass der nukleäre Transkriptionsfactor NF-ΚB durch Photodynamische Therapie mit Methylenblau aktiviert werden kann, ist bereits gezeigt worden, so dass wir diese Versuche nicht wiederholten. Die Photodynamische Therapie mit dem Photosensibilisator Methylenblau wirkt also sowohl direkt als auch indirekt toxisch. In unseren Versuchen beschränkten wir uns im weiteren auf die Wirkung des Photosensibilisators disulfoniertes Aluminiumphthalocyanin auf den nukleären Transkriptionsfaktor NF-ΚB. Mittels Gelelektrophorese konnten wir keine Aktivierung von NF-ΚB zeigen. Die Photodynamische Therapie mit dem Photosensibilisator disulfoniertem Aluminiumphthalocyanin wirkt also nur auf direkt toxischem Weg. Bezüglich der Stickstoffmonoxid-Bildung fand sich bei beiden Photosensibilisatoren in den von uns verwendeten Konzentrationen und Inkubabionszeiten kein Nitritnachweis. Auch bei α-Liponsäure ergab sich bei Keratinozyten weder ein pro- noch antiproliferativer Effekt und somt kein Anhalt auf eine indirekte toxische Wirkung.
Der klinische Einsatz der Photodynamischen Therapie erscheint vor dem Hintergrund der erarbeiteten Daten bei entzündlichen Dermatosen möglich, weil infiltrierende aktivierte Leukozyten sensibler gegenüber PDT sind als das umliegende Gewebe, wie hier beispielhaft für Keratinozyten gezeigt wurde.
