The chiral critical surface is a surface of second order phase transitions bounding the region of
first order chiral phase transitions for small quark masses in the fmu;d;ms;mg parameter space.
The potential critical endpoint of the QCD (T;m)-phase diagram is widely expected to be part of
this surface. Since for m = 0 with physical quark masses QCD is known to exhibit an analytic
crossover, this expectation requires the region of chiral transitions to expand with m for a chiral
critical endpoint to exist. Instead, on coarse Nt = 4 lattices, we find the area of chiral transitions
to shrink with m, which excludes a chiral critical point for QCD at moderate chemical potentials
mB < 500 MeV. First results on finer Nt = 6 lattices indicate a curvature of the critical surface
consistent with zero and unchanged conclusions. We also comment on the interplay of phase
diagrams between the Nf = 2 and Nf = 2+1 theories and its consequences for physical QCD.
We perform a two-flavor dynamical lattice computation of the Isgur-Wise functions t1/2 and t3/2
at zero recoil in the static limit. We find t1/2(1) = 0.297(26) and t3/2(1) = 0.528(23) fulfilling
Uraltsev’s sum rule by around 80%. We also comment on a persistent conflict between theory and
experiment regarding semileptonic decays of B mesons into orbitally excited P wave D mesons,
the so-called “1/2 versus 3/2 puzzle”, and we discuss the relevance of lattice results in this
context.
We present a lattice QCD calculation of the heavy-light decay constants fB and fBs performed with Nf = 2 maximally twisted Wilson fermions, at four values of the lattice spacing. The decay constants have been also computed in the static limit and the results are used to interpolate the observables between the charmand the infinite-mass sectors, thus obtaining the value of the decay constants at the physical b quark mass. Our preliminary results are fB = 191(14)MeV, fBs = 243(14)MeV, fBs/ fB = 1.27(5). They are in good agreement with those obtained with a novel approach, recently proposed by our Collaboration (ETMC), based on the use of suitable ratios having an exactly known static limit.
We present first results from runs performed with Nf = 2+1+1 flavours of dynamical twisted mass fermions at maximal twist: a degenerate light doublet and a mass split heavy doublet. An overview of the input parameters and tuning status of our ensembles is given, together with a comparison with results obtained with Nf = 2 flavours. The problem of extracting the mass of the K- and D-mesons is discussed, and the tuning of the strange and charm quark masses examined. Finally we compare two methods of extracting the lattice spacings to check the consistency of our data and we present some first results of cPT fits in the light meson sector.

Para responder a esta questão, tentar-se-á, a seguir, tornar a teoria da hegemonia de antonio gramsci e seu modelo de direito hegemônico produtivos para o campo da teoria do direito. Tal tarefa tem de lidar com a dupla dificuldade de que, por um lado, gramsci não foi um teórico do direito no sentido mais estrito, razão pela qual o potencial de sua teoria para uma análise do direito raramente foi utilizada. Por outro lado, sua abordagem só pode ser empregada por meio de uma crítica às restrições relacionadas a seu tempo. isso se aplica especialmente à sua concepção de economia como a base e a núcleo essencialista oculto (laclau; mouffe, 2001:69), assim como à sua ideia de 'classismo' sob a forma de um enfoque unilateral das classes, em que há preferencialmente mais de um "pluralismo de poder" e inúmeras lutas (litowitz, 2000: 536). Recuperar-se-á, consequentemente, argumentos-chave, ampliando-os pela utilização das recentes descobertas feitas pelas abordagens feminista e neomaterialista da teoria jurídica, bem como as análises de foucault acerca das tecnologias de poder. por fim, uma interpretação da teoria sistêmica das autonomizações comunicativas.
O 11 de setembro acelerou o desenvolvimento de uma arquitetura transnacional de segurança que intervém profundamente nas liberdades civis individuais, tanto nos direitos básicos dos cidadãos dos Estados como nos direitos humanos dos cidadãos mundiais. O artigo delineia essa arquitetura, mostra como ela dissolve as categorias jurídicas tradicionais que preservam a liberdade e discute por que hoje se aceita amplamente a prioridade da segurança sobre a liberdade.
Os limites da tolerância

Este artigo apresenta os elementos constitutivos do conceito de tolerância e discute duas concepções diferentes do termo, como permissão e como respeito moral, que expressam modos diversos de demarcar os limites da tolerância. A tolerância é apresentada como um conceito que, para ganhar algum conteúdo, depende normativamente de um direito à justificação baseado na idéia de um uso público da razão segundo o qual as práticas e as instituições político-jurídicas que determinam a vida social dos cidadãos devem ser justificáveis à luz de normas que eles não podem recíproca e genericamente rejeitar.
Background: A growing number of German hospitals have been privatized with the intention of increasing cost effectiveness and improving the quality of health care. Numerous studies investigated what possible qualitative and economic consequences these changes issues might have on patient care. However, little is known about how this privatization trend relates to physicians' working conditions and job satisfaction. It was anticipated that different working conditions would be associated with different types of hospital ownership. To that end, this study's purpose is to compare how physicians, working for both public and privatized hospitals, rate their respective psychosocial working conditions and job satisfaction.
Methods: The study was designed as a cross-sectional comparison using questionnaire data from 203 physicians working at German hospitals of different ownership types (private for-profit, public and private nonprofit).
Results: The present study shows that several aspects of physicians' perceived working conditions differ significantly depending on hospital ownership. However, results also indicated that physicians' job satisfaction does not vary between different types of hospital ownership. Finally, it was demonstrated that job demands and resources are associated with job satisfaction, while type of ownership is not.
Conclusion: This study represents one of a few studies that investigate the effect of hospital ownership on physicians work situation and demonstrated that the type of ownership is a potential factor accounting for differences in working conditions. The findings provide an informative basis to find solutions improving physicians' work at German hospitals.
Die Arbeit analysiert den Begriff sowie den Wert der Freiheit in den Schriften des kanadischen Philosophen Charles Taylor, unter Bezugnahme auf dessen politische Philosophie und philosophische Anthropologie. Die begriffliche Klärung basiert auf einer Systematisierung der positiven Verwendung des Freiheitsbegriffes in Taylors Gesamtwerk. Die Wertanalyse interpretiert die Ergebnisse der Systematisierung in Bezug auf die Frage, ob Freiheit in Taylors Verständnis ein extrinsischer oder ein intrinsischer Wert ist.
In its admissibility decision in the Al-Saadoon case the ECtHR held that the United Kingdom had jurisdiction over the applicants, who had been arrested by British forces and kept in a British-run military prison in Iraq. Just before the respective mandate of the Security Council expired on 31 December 2008, the applicants were transferred to Iraqi custody at Iraqi request and thereby exposed to the risk of an unfair trial followed by capital punishment. In this respect, the case resembles the Soering case, although the applicants were, unlike Soering, not on British territory but on occupied Iraqi soil before they were handed over. This aspect raises the question of Iraqi sovereignty as a norm competing with the UK's human rights obligations. The authors trace back the ECtHR's case law concerning the extraterritorial application of the Convention and analyse the UK judgments and the ECtHR's admissibility decision in the Al-Saadoon affair from this angle. Furthermore they consider the doctrinal consequences of the ECHR's extraterritorial effect in cases like Soering and Al-Saadoon, where contracting parties violate guarantees of the Convention by exposing a person within their jurisdiction to a risk of a treatment contrary to these guarantees by a third state. Finally, they test the argument brought forward by the UK that not transferring the applicants would have violated Iraqi sovereignty and establish patterns how the ECtHR and the UK Courts did cope in the past with international law norms potentially competing with the Convention.

GoeTheo ist ein Projekt, welches der Transparenz des Fachbereichs und der Informationsweitergabe an Studierende, Förderer und Interessierte dient.
Die Fachbereichszeitschrift ist eine gemeinsame Publikation des Vereins der Freunde und Förderer der Evangelischen Theologie in Frankfurt/Main und des Fachbereichs Evangelische Theologie der Goethe-Universität. Seit Wintersemester 2009/10 erscheint GoeTheo immer zum Semesteranfang und informiert über die Lehrveranstaltungen des kommenden Semesters, sowie über aktuelle Entwicklungen des Fachbereichs in Forschung und Lehre.
In contrast to the US and recently Europe, Japan appears to be unsuccessful in establishing new industries. An oft-cited example is Japan's practical invisibility in the global business software sector. Literature has ascribed Japan's weakness – or conversely, America's strength – to the specific institutional settings and competences of actors within the respective national innovation system. It has additionally been argued that unlike the American innovation system, with its proven ability to give birth to new industries, the inherent path dependency of the Japanese innovation system makes innovation and establishment of new industries quite difficult. However, there are two notable weaknesses underlying current propositions postulating that only certain innovation systems enable the creation of new industries: first, they mistakenly confound context specific with general empirical observations. And second, they grossly underestimate – or altogether fail to examine – the dynamics within innovation systems. This paper will show that it is precisely the dynamics within innovation systems – dynamics founded on the concept of path plasticity – which have enabled Japan to charge forward as a global leader in a highly innovative field: the game software sector as well as the biotechnology industry.
European scholars, colonial administrators, missionaries, bibliophiles and others were the main collectors of Malay books in the nineteenth century, both in manuscript or printed form. Among these persons were many well-known names in the field of Malay literature and culture like Raffles, Marsden, Crawfurd, Klinkert, van der Tuuk, von Dewall, Roorda, Favre, Maxwell, Overbeck, Wilkinson and Skeat, to name only a few. Their collections were often handed over to public libraries where they form an important part of the relevant Oriental or Southeast Asian manuscript collections.
Therefore the knowledge of the intellectual culture of the Malay Peninsula and the Malay World in general depended very much on these manuscripts and printed books collected often by chance or in a rather unsystematic way. The collections reflect in a strong sense the interests of its administrative or philologist collectors: court histories, genealogies of aristocratic lineages, law collections (adat-istiadat as well as undangundang) or prose belles-lettres build a vast bulk of these collections, while Islamic religious texts and poetry forms popular in the 19th century (especially syair) are fairly underrepresented. Malay manuscripts and books located in religious institutions like mosques or pondok/pesantren schools have not been searched for; until today there are more or less no systematic studies of these collections. As in some statistics religious texts build about 20% of all existing Malay manuscripts, their neglect by Europeans scholars leads to a distorted view of the literary culture in the Malay language.
Following the discovery of context-dependent synchronization of oscillatory neuronal responses in the visual system, the role of neural synchrony in cortical networks has been expanded to provide a general mechanism for the coordination of distributed neural activity patterns. In the current paper, we present an update of the status of this hypothesis through summarizing recent results from our laboratory that suggest important new insights regarding the mechanisms, function and relevance of this phenomenon. In the first part, we present recent results derived from animal experiments and mathematical simulations that provide novel explanations and mechanisms for zero and nero-zero phase lag synchronization. In the second part, we shall discuss the role of neural synchrony for expectancy during perceptual organization and its role in conscious experience. This will be followed by evidence that indicates that in addition to supporting conscious cognition, neural synchrony is abnormal in major brain disorders, such as schizophrenia and autism spectrum disorders. We conclude this paper with suggestions for further research as well as with critical issues that need to be addressed in future studies.
The cerebral cortex presents itself as a distributed dynamical system with the characteristics of a small world network. The neuronal correlates of cognitive and executive processes often appear to consist of the coordinated activity of large assemblies of widely distributed neurons. These features require mechanisms for the selective routing of signals across densely interconnected networks, the flexible and context dependent binding of neuronal groups into functionally coherent assemblies and the task and attention dependent integration of subsystems. In order to implement these mechanisms, it is proposed that neuronal responses should convey two orthogonal messages in parallel. They should indicate (1) the presence of the feature to which they are tuned and (2) with which other neurons (specific target cells or members of a coherent assembly) they are communicating. The first message is encoded in the discharge frequency of the neurons (rate code) and it is proposed that the second message is contained in the precise timing relationships between individual spikes of distributed neurons (temporal code). It is further proposed that these precise timing relations are established either by the timing of external events (stimulus locking) or by internal timing mechanisms. The latter are assumed to consist of an oscillatory modulation of neuronal responses in different frequency bands that cover a broad frequency range from <2 Hz (delta) to >40 Hz (gamma) and ripples. These oscillations limit the communication of cells to short temporal windows whereby the duration of these windows decreases with oscillation frequency. Thus, by varying the phase relationship between oscillating groups, networks of functionally cooperating neurons can be flexibly configurated within hard wired networks. Moreover, by synchronizing the spikes emitted by neuronal populations, the saliency of their responses can be enhanced due to the coincidence sensitivity of receiving neurons in very much the same way as can be achieved by increasing the discharge rate. Experimental evidence will be reviewed in support of the coexistence of rate and temporal codes. Evidence will also be provided that disturbances of temporal coding mechanisms are likely to be one of the pathophysiological mechanisms in schizophrenia.
A suite of diagnostics is applied to in-situ aircraft measurements and one Chemistry-Climate Model (CCM) data to characterize the vertical structure of the Tropical Tropopause Layer (TTL). The diagnostics are based on vertical tracer profiles and relative vertical tracer gradients, using tropopause-referenced coordinates, and tracer-tracer relationships in the tropical Upper Troposphere/Lower Stratosphere (UT/LS).
Observations were obtained during four tropical campaigns performed from 1999 to 2006 with the research aircraft Geophysica and have been compared to the output of the ECHAM5/MESSy CCM. The model vertical resolution in the TTL (~500 m) allows for appropriate comparison with high-resolution aircraft observations and the diagnostics used highlight common TTL features between the model and the observational data.
The analysis of the vertical profiles of water vapour, ozone, and nitrous oxide, in both the observations and the model, shows that concentration mixing ratios exhibit a strong gradient change across the tropical tropopause, due to the role of this latter as a transport barrier and that transition between the tropospheric and stratospheric regimes occurs within a finite layer. The use of relative vertical ozone and carbon monoxide gradients, in addition to the vertical profiles, helps to highlight the region where this transition occurs and allows to give an estimate of its thickness. The analysis of the CO-O3 and H2O-O3 scatter plots and of the Probability Distribution Function (PDF) of the H2O-O3 pair completes this picture as it allows to better distinguish tropospheric and stratospheric regimes that can be identified by their different chemical composition.
The joint analysis and comparison of observed and modelled data allows to state that the model can represent the background TTL structure and its seasonal variability rather accurately. The model estimate of the thickness of the interface region between tropospheric and stratospheric regimes agrees well with average values inferred from observations. On the other hand, the measurements can be influenced by regional scale variability, local transport processes as well as deep convection, that can not be captured by the model.

The joint analysis and comparison of observed and modelled data allows us to evaluate the capability of the model in reproducing the observed vertical structure of the TTL and its variability, and also to assess whether observations from particular regions on a monthly timescale can be representative of the fine scale mean structure of the Tropical Tropopause Layer.
The small leucine-rich proteoglycans (SLRPs) are biologically active components of the extracellular matrix (ECM), consisting of a protein core with leucine rich-repeat (LRR) motifs covalently linked to glycosaminoglycan (GAG) side chains. The diversity in composition resulting from the various combinations of protein cores substituted with one or more GAG chains along with their pericellular localization enables SLRPs to interact with a host of different cell surface receptors, cytokines, growth factors, and other ECM components, leading to modulation of cellular functions. SLRPs are capable of binding to: (i) different types of collagens, thereby regulating fibril assembly, organization, and degradation; (ii) Toll-like receptors (TLRs), complement C1q, and tumor necrosis factor-alpha (TNFα), regulating innate immunity and inflammation; (iii) epidermal growth factor receptor (EGF-R), insulin-like growth factor receptor (IGF-IR), and c-Met, influencing cellular proliferation, survival, adhesion, migration, tumor growth and metastasis as well as synthesis of other ECM components; (iv) low-density lipoprotein receptor-related protein (LRP-1) and TGF-β, modulating cytokine activity and fibrogenesis; and (v) growth factors such as bone morphogenic protein (BMP-4) and Wnt-I-induced secreted protein-1 (WISP-1), controlling cell proliferation and differentiation. Thus, the ability of SLRPs, as ECM components, to directly or indirectly regulate cell-matrix crosstalk, resulting in the modulation of various biological processes, aptly qualifies these compounds as matricellular proteins.
Mit der neuen Umgangspflegschaft verabschiedet sich der Gesetzgeber vom Grundsatz, dass Eingriffe in das elterliche Sorgerecht erst bei (nicht anders abwendbaren) erheblichen Kindeswohlgefährdungen zulässig sind. Bleibt zu hoffen, dass die Praxis mit größter Behutsamkeit von der Bestellung von Umgangspflegschaften Gebrauch macht, insbesondere dann, wenn Umgang gegen den Kindeswillen durchgesetzt werden soll, was sich aus rechtlichen, fachlichen und ethischen Gründen verbietet. Die massive Bedrohung der eigenständigen Interessenvertretung Minderjähriger – die bislang eine Erfolgsgeschichte zu werden schien – tritt am 01.09.2009 in Kraft. Verfahrenspfleger/-beistände werden sich auf die neue vergütungsrechtliche Situation einzustellen haben, mit welchen persönlichen Konsequenzen, das kann nur von ihnen beantwortet werden. Es wäre schade, insbesondere für die auf qualifizierte und einfühlsame erfahrenspfleger/-beistände angewiesenen Minderjährigen, wenn allzu viele dieser erfahrenen und bewährten Interessenvertreter Minderjähriger sich von diesem bedeutsamen, aber auch herausfordernden Arbeitsfeld abwenden müssten, geht es doch bei der Gruppe von Minderjährigen, die auf einen Verfahrenspfleger bzw. -beistand zur eigenständigen Wahrnehmung ihrer Interessen angewiesen sind, um Minderjährige, die bereits erheblich gefährdet sind oder waren bzw. denen eine solch massive Beeinträchtigung ihres Wohls droht. Sicherlich: Auf die Situation vor 1998 sind wir noch nicht zurückgeworfen, aber es gilt in mühsamer Arbeit die Rechtspolitik davon zu überzeugen, dass diese Fehlentscheidung alsbald korrigiert werden muss.
Dieser Band dokumentiert die Vorträge des ersten „Familienrechtlichen Forums Göttingen“, das am 28. Juni 2008 stattgefunden hat. Unmittelbar vor dem Abschluss des Gesetzgebungsverfahrens erörterten Experten aus Wissenschaft, Politik und Praxis das im September 2009 in Kraft tretende „Gesetz über das Verfahren in Familiensachen und in den Angelegenheiten der freiwilligen Gerichtsbarkeit“ (FamFG). Der Band enthält einen Überblick über die Grundzüge des neuen Familienverfahrensrechts sowie Analysen und Stellungnahmen zu einzelnen Bereichen. Dabei werden schwerpunktmäßig das neue Rechtsmittel- und Vollstreckungssystem, das Hinwirken auf Einvernehmen, das Vermittlungsverfahren, der Verfahrensbeistand, der Umgangspfleger und das Verfahren bei Kindeswohlgefährdung behandelt. Darüber hinaus wird eine interdisziplinäre Perspektive durch die soziologische Einordnung der derzeitigen Reformen in den Kontext der allgemeinen Familien- und Sozialpolitik eröffnet.
Background: During the last two decades the German hospital sector has been engaged in a constant process of transformation. One obvious sign of this is the growing amount of hospital privatization. To date, most research studies have focused on the effects of privatization regarding financial outcomes and quality of care, leaving important organizational issues unexplored. Yet little attention has been devoted to the effects of privatization on physicians' working routines. The aim of this observational real-time study is to deliver exact data about physicians' work at hospitals of different ownership. By analysing working hours, further impacts of hospital privatization can be assessed and areas of improvement identified.
Methods: Observations were made by shadowing 100 physicians working in private, for-profit or non-profit as well as public hospital departments individually during whole weekday shifts in urban German settings. A total of 300 days of observations were conducted. All working activities were recorded, accurate to the second, by using a mobile personal computer.
Results: Results have shown significant differences in physicians' working activities, depending on hospital ownership, concerning working hours and time spent on direct and indirect patient care.
Conclusion: This is the first real-time analysis on differences in work activities depending on hospital ownership. The study provides an objective insight into physicians' daily work routines at hospitals of different ownership, with additional information on effects of hospital privatization.
Background: Publications related to scoliosis have increased enormously. A differentiation between publications of major and minor importance has become difficult even for experts. Scientometric data on developments and tendencies in scoliosis research has not been available to date. The aim of the current study was to evaluate the scientific efforts of scoliosis research both quantitatively and qualitatively.
Methods: Large-scale data analysis, density-equalizing algorithms and scientometric methods were used to evaluate both the quantity and quality of research achievements of scientists studying scoliosis. Density-equalizing algorithms were applied to data retrieved from ISI-Web.
Results: From 1904 to 2007, 8,186 items pertaining to scoliosis were published and included in the database. The studies were published in 76 countries: the USA, the U.K. and Canada being the most productive centers. The Washington University (St. Louis, Missouri) was identified as the most prolific institution during that period, and orthopedics represented by far the most productive medical discipline. "BRADFORD, DS" is the most productive author (146 items), and "DANSEREAU, J" is the author with the highest scientific impact (h-index of 27).
Conclusion: Our results suggest that currently established measures of research output (i.e. impact factor, h-index) should be evaluated critically because phenomena, such as self-citation and co-authorship, distort the results and limit the value of the conclusions that may be drawn from these measures. Qualitative statements are just tractable by the comparison of the parameters with respect to multiple linkages. In order to obtain more objective evaluation tools, new measurements need to be developed.
Background: The increasing economic pressure characterizes the current situation in health care and the need to justify medical decisions and organizational processes due to limited financial resources is omnipresent. Physicians tend to interpret this development as a decimation of their own medical influence. This becomes even more obvious after a change in hospital ownership i.e. from a public to a private profit oriented organization. In this case each work procedure is revised.
To date, most research studies have focused mainly on differences between hospitals of different ownership regarding financial outcomes and quality of care, leaving important organizational issues unexplored. Little attention has been devoted to the effects of hospital ownership on physicians' working routines.
The aim of this observational real time study is to deliver exact data about physicians' work at hospitals of different ownership.
Methods: The consequences of different management types on the organizational structures of the physicians' work situation and on job satisfaction in the ward situation are monitored by objective real time studies and multi-level psycho diagnostic measurements.
Discussion: This study is unique in its focus. To date no results have been found for computer-based real time studies on work activity in the clinical field in order to objectively evaluate a physician's work-related stress. After a complete documentation of the physicians' work processes the daily work flow can be estimated and systematically optimized. This can stimulate an overall improvement of health care services in Germany.
The role of endocarditis, myocarditis and pericarditis in qualitative and quantitative data analysis

The current study is the first scientometric analysis of research activity and output in the field of inflammatory disorders of the heart (endo-, myo- and pericarditis). Scientometric methods are used to compare scientific performance on national and on international scale to identify single areas of research interest. Interest and research productivity in inflammatory diseases of the heart have increased since 1990. The majority of publications about inflammatory heart disorders were published in Western Europe and North America. The United States of America had a leading position in terms of research productivity and quality; half of the most productive authors in this study came from American institutions. The analysis of international cooperation revealed research activity in countries that are less established in the field of inflammatory heart disorder research, such as Brazil, Saudi Arabia and Tunisia. These results indicate that future research of heart inflammation may no longer be influenced predominantly by a small number of countries. Furthermore, this study revealed weaknesses in currently established scientometric parameters (i.e., h-index, impact factor) that limit their suitability as measures of research quality. In this respect, self-citations should be generally excluded from calculations of h-index and impact factor.
Objectives: Sepsis remains a disease with a high mortality in neonates. Microcirculatory impairment plays a pivotal role in the development of multiorgan failure in septic newborns. The hemodynamic effects of recombinant activated protein C (rhAPC) were tested in an animal model of neonatal septic shock focusing on intestinal microcirculation.
Materials and methods: Endotoxic shock was triggered by intravenous application of Escherichia coli lipopolysaccarides in newborn piglets. Thereafter, five animals received a continuous infusion of 24 µg/kg/h rhAPC, and five received vehicle for control. Over the course of three hours, intestinal microcirculation was assessed by intravital microscopy every 30 min. Macrocirculation and blood counts were monitored simultaneously.
Results: After a short hypotensive period in all animals, the arterial blood pressure returned to baseline in the rhAPC-treated piglets, whereas the hypotension became increasingly severe in the controls. By 90 min, mean blood pressure in the controls was significantly lower than in the treatment group. Similar observations were made regaring microcirculation. After an early impairment in all study animals, functional capillary density and intestinal microcirculatory red blood cell velocity and red blood cell flow recovered in the rhAPC group, but deteriorated further in the control piglets.
Conclusion: Recombinant activated protein C protects macro- and microcirculation from endotoxic shock.
EFP1 is an ER stress-induced glycoprotein which interacts with the pro-apoptotic protein Par-4

We have isolated the rat ortholog of EFP1 (EF-hand binding protein 1) as a novel interaction partner of the pro-apoptotic protein Par-4 (prostate apoptosis response-4). Rat EFP1 contains two thioredoxin domains, the COOH-terminal one harboring a CGFC motif, and has a similar protein domain structure as members of the protein disulfide isomerase (PDI) family. In REF52.2 and CHO cells, EFP1 colocalized with the endoplasmic reticulum (ER) marker PDI. Furthermore, EFP1 possesses catalytic activity as demonstrated by an insulin disulfide reduction assay. Western blot analysis revealed two EFP1 protein bands of approximately 136 and 155 kDa, representing different glycosylation states of the protein. Complex formation between EFP1 and Par-4 was confirmed in vitro and in vivo by co-immunoprecipitation, dot blot overlay and pull-down experiments. In CHO cells, coexpression of EFP1 and Par-4 resulted in enhanced Par-4-mediated apoptosis, which required the catalytic activity of EFP1. Interestingly, EFP1 was specifically upregulated in NIH3T3 cells after induction of ER stress by thapsigargin, tunicamycin, and brefeldin A, but not by agents that induce oxidative stress or ER-independent apoptosis. Furthermore, we could show that the induction of apoptosis by Ca2+ stress-inducing agents was significantly decreased after siRNA oligonucleotide-mediated knockdown of Par-4. Our data suggest that EFP1 might represent a cell-protective enzyme that could play an important role in the decision between survival and initiation of Par-4-mediated apoptosis.
Risikoabschätzung für das Auftreten Thrombolyse-assoziierter intrazerebraler Blutungen mittels MRT

Symptomatische intrazerebrale Blutungen (sICH) stellen die wichtigste Komplikation der Thrombolyse akuter ischämischer Schlaganfälle dar. Sie sind mit einer erhöhten Mortalität und sehr schlechten Langzeitergebnissen verbunden. Die Identifikation von Patienten mit einem erhöhten Blutungsrisiko stellt daher ein wesentliches Ziel aktueller Forschungsbemühungen dar.
In der vorliegenden Studie wurde die Bedeutung ausgewählter MR-Parameter (Leukoaraiosis, Mikroblutungen, Ausmaß der DWI-Läsion) als mögliche Prädiktoren für Thrombolyse-assoziierte sICH untersucht.
In die retrospektive, multizentrische Studie wurden prospektiv erhobene Daten von 475 Patienten aus sechs deutschen und zwei internationalen anerkannten Schlaganfallzentren eingeschlossen, die im Zeitfenster von <6 Stunden mit einem akuten ischämischen Schlaganfallereignis im Bereich der vorderen Zirkulation mit einer i.v. (n=367; 77%), i.a. (n=74; 16%) oder i.v./i.a. (n=34; 7%) Thrombolyse behandelt worden.
Alle Patienten wurden mit einem Schlaganfall-MR-Protokoll untersucht, welches neben Diffusions- und FLAIR-Sequenzen auch T2*-gewichtete Sequenzen enthielt.
Für die Analyse wurden die Patienten jeweils in eine Gruppe ohne und eine Gruppe mit schwerer LA (Fazekas 0-1 vs. 2-3) und je eine Gruppe mit kleinem (<1/3 des A. cerebri media-Stromgebietes (MCA)) und großem (>1/3 MCA) DWI-Läsionsausmaß dichotomisiert. T2*-Wichtungen wurden auf das Vorliegen und die Anzahl von zerebralen Mikroblutungen (CMB) untersucht und Patienten in eine Gruppe mit ≥1 CMB und ohne CMB dichotomisiert.
Eine sICH wurde als eine in der Verlaufsbildgebung (cCT oder MRT) dokumentierte (≥10 ml) Blutung definiert, die innerhalb von 36 Stunden nach Thrombolysebeginn mit einer klinischen Verschlechterung des Patienten (≥1 Punkt auf der NIHSS-Skale) einherging.
Insgesamt entwickelten 28 (5,9%) Patienten eine sICH. SICH waren signifikant häufiger in der Gruppe mit LA (Fazekas 2-3) (n=12/114; 10,5%) als in der Gruppe ohne LA (Fazekas 0-1) (n=13/335; 3,5%) zu finden. In einer logistischen Regressionsanalyse (Alter, National Institutes of Health Stroke Scale (NIHSS) bei Aufnahme, Art der Thrombolyse) blieb LA ein unabhängiger Risikofaktor (Odds ratio 2,7 (95% KI, 1,29-6,59), p=0,015).
Die Anzahl der sICH konnte auch in der Gruppe mit großen (>1/3 MCA) (n=6/32; 18,8%) signifikant häufiger als in der Gruppe mit kleinen DWI-Läsionen (<1/3 MCA) (n=12/319; 3,8%) erhoben werden, was umgerechnet einem 5,9 fachen Risiko (Odds ratio 5,9 (95% KI, 2,048-17,017, p=0,003) einer sICH für Patienten mit großen DWI-Läsionen im Vergleich zu Patienten mit kleinen DWI-Läsionen entspricht. Das Ausmaß der DWI-Läsion bleibt in einer multivariaten Regressionsanalyse (s.o.) ein unabhängiger Risikofaktor für sICH.
Die Ergebnisse einer großen Subgruppenanalyse der i.v.-behandelten Patienten (n=367/376, 77%) waren hinsichtlich der Bedeutung für LA und Ausmaß der DWI-Läsion vergleichbar mit den Ergebnisse der Gesamtmenge.
Die Anzahl der sICH unterschied sich nicht signifikant zwischen der Gruppe mit ≥1 CMB (n=7/79, 9%) und der Gruppe ohne CMB (n=20/367, 5%, p=0,295).
Zusammenfassend konnte die vorliegende Arbeit die MR-Parameter Leukoaraiose und das Ausmaß der initialen DWI-Läsion als unabhängige Risikofaktoren hinsichtlich der Enwicklung Thrombolyse-assoziierter sICH nachweisen. Hiermit kann einerseits die Sicherheit der Thrombolyse im 3h-Zeitfenster erhöht und der Behandlungzeitraum hoffentlich zukünftig erweitert werden. Der potentielle Behandlungsgewinn einer Thrombolyse sollte bei Einschluss in Betracht kommender Schlaganfallpatienten sorgfältig gegen das mit zunehmender LA und DWI-Läsionlast ansteigende Blutungsrisiko abgewogen werden. Eine unabhängige Assoziation zwischen sICH und CMB konnte nicht gezeigt werden, so dass das Blutungsrisiko bei Patienten mit CMB’s den Behandlungsvorteil wahrscheinlich nicht übersteigt.
Um die kontinuierlich auf uns einströmende Menge an Reizen zu verarbeiten, ist es wichtig, die Informationen genau zu selektieren. Ein hilfreicher Mechanismus ist hierbei die Aufmerksamkeit gezielt nur auf eine Informationsquelle zu richten und zu verarbeiten.
So ist es möglich, selbst in komplexen Situationen, wie zum Beispiel einer Feier mit vielen verschiedenen Gesprächen, ganz gezielt ein einzelnes herauszufiltern.
Der Effekt von Aufmerksamkeit auf auditory steady-state Responses (aSSR) wurde in der Vergangenheit schon in verschiedenen Studien mit unterschiedlichen Messverfahren und Stimuli untersucht. Dabei wurden teils widersprüchliche Ergebnisse gefunden, die entweder gar keine oder auf sehr kleine Frequenzbereiche beschränkte Aufmerksamkeitseffekte fanden.
Das Ziel dieser Studie war es, die Auswirkungen von Aufmerksamkeit auf die aSSR innerhalb eines größeren Frequenzspektrums (11 Hz, 23 Hz, 41 Hz, 73 Hz und 97 Hz) zu analysieren. Für diesen Zweck wurden den Probanden nach Instruktion der zu beachtenden Seite jeweils zwei benachbarte Modulationsfrequenzen während vier aufeinander folgenden Blöcken dichotisch präsentiert. Die Probanden wurden angewiesen, Trägerfrequenzänderungen auf der zu beachtenden Seite durch Betätigen einer Maustaste anzugeben. Die Registrierung der aSSR-Antworten geschah mittels Elektroenzephalogramm (EEG). Auch die durch die Stimulation hervorgerufene P300 wurde dargestellt.
Bezüglich des Effekts von Aufmerksamkeit auf die aSSRs zeigte sich nach Analyse der gewonnenen Daten im Frequenzbereich von 23 Hz eine Auswirkung. Diese stellte sich in Form einer Amplitudensteigerung auf der jeweils durch den Probanden beachteten Präsentationsseite dar. Bei einer Modulationsfrequenz von 41 Hz kam es bei Präsentation auf der rechten Seite zu höheren Amplituden als bei Präsentation auf der linken Seite. Bei 73 Hz und 97 Hz konnte keinerlei Auswirkung weder der Aufmerksamkeit noch der Präsentationsseite registriert werden. Auffällig war bei den präsentierten Tönen im 20 Hz und 70 Hz Bereich auch eine im Vergleich zu den anderen Frequenzbereichen (11 Hz, 41 Hz und 97 Hz) verlängerte Reaktionszeit der Probanden. In Kombination mit der Modulation der aSSR-Amplitude durch Aufmerksamkeit bei 23 Hz könnte dies ein Hinweis auf einen förderlichen Einfluss der Aufgabenschwierigkeit auf die Detektierbarkeit von Aufmerksamkeitseffekten sein.
Im Gegensatz hierzu zeigte die dargestellte P300 in allen präsentierten Blöcken einen deutlichen Effekt der Aufmerksamkeit. Dieser äußerte sich ebenfalls in einer Steigerung der Amplitude.
Es scheint also zumindest ein moderater Einfluss von Aufmerksamkeit auf die aSSRs zu existieren. Gleichzeitig wirkt dieser allerdings stark abhängig von gewähltem Stimulus und Messmethode. Der Effekt von Aufmerksamkeit auf die P300 konnte dagegen gut repliziert werden und scheint daher bei dichotischer Stimulation ein deutlicher Marker für Aufmerksamkeit zu sein.
In der vorliegenden Studie wurden Adoptierte auf Merkmale wie Selbstbewusstsein, Widerstandsfähigkeit und Bindungseinstellungen in Abhängigkeit verschiedener möglicher Einflussfaktoren wie beispielsweise Heimaufenthalten untersucht. Mittelpunkt der Forschung war die Exploration der Entwicklung von Persönlichkeitsmerkmalen in Abhängigkeit der verschiedenen Adoptionsformen, die den Kontakt zu den leiblichen Eltern möglich machen oder verhindern. Untersuchungen an erwachsenen Adoptierten sind noch selten, ebenso gibt es keine wissenschaftlichen Studien in Deutschland, die sich mit dem Thema der Adoption befassen. Auch die Forschung auf dem Gebiet der verschiedenen Adoptionsformen und ihrer Auswirkung auf die Entwicklung von Persönlichkeitsmerkmalen bei Adoptierten ist noch sehr jung. Dies wurde mit der vorliegenden Studie deutschlandweit erstmalig in Angriff genommen. Die Ergebnisse zeigten, dass Heimaufenthalte, je häufiger sie sind, zu einer verminderten Widerstandsfähigkeit der Adoptierten führten. Heimaufenthalte konnten als starker Prädiktor für verminderte Resilienz identifiziert werden. Ebenso sanken die Werte auf manchen Skalen der Multidimensionalen Selbstwertskala wie beispielsweise die Wertschätzung der eigenen Leistungsfähigkeit. Adoptierte wurden mit steigender Anzahl an Heimaufenthalten neurotischer. Leider konnte aufgrund mangelhafter Angaben die Dauer der Heimaufenthalte und Aufenthalte in Pflegefamilien nicht in die Auswertung mit einbezogen werden, darüber hinaus auch nicht die Qualität der Erinnerungen an diese Heimaufenthalte. Dennoch lässt sich feststellen, dass Heimaufenthalte das Selbstwertgefühl und die Selbstsicherheit Adoptierter nachhaltig beeinträchtigen können. Sie sollten so gering wie möglich gehalten werden. Auf die Bindungseinstellungen zeigten sie keine Auswirkungen. Hier könnte zukünftig eine genauere Untersuchung des Einflusses von Heimaufenthalten erfolgen, die unter anderem die Dauer und zusätzliche Wechsel von Pflegefamilien mit einbezieht. Bezüglich der Adoptionsformen offene Adoption vs. Inkognitoadoption konnte gezeigt werden, dass es für die Entwicklung der untersuchten Persönlichkeitsmerkmale keine Rolle zu spielen scheint, ob die Adoptierten die Möglichkeit des Kontaktes zu ihren leiblichen Eltern haben oder nicht. Dennoch führte die positive Bewertung dieses Kontaktes zu besseren Ergebnissen bezüglich des Selbstwertes als die negative Bewertung des Kontaktes zu den leiblichen Eltern. Interessant ist, dass Adoptierte, die keine Kontaktmöglichkeit zu ihren leiblichen Eltern hatten, bessere Werte bezogen auf Facetten des Selbstwertgefühles zeigten als Adoptierte, die diesen Kontakt negativ bewerteten. Es scheint, als wäre es für das eigene Selbstbewusstsein gesünder, keinen Kontakt zu haben als ihn letztendlich negativ zu bewerten. Positiv empfundener Kontakt konnte das Selbstwertgefühl und die Bindungseinstellungen nicht zusätzlich verbessern. Allerdings kann keine sinnvolle Konsequenz aus diesen Ergebnissen gezogen werden. Im Voraus ist selten zu beurteilen, wie der / die Adoptierte das Treffen und den Kontakt zu den leiblichen Eltern bewerten wird. Dies hängt mit Sicherheit nicht nur vom Adoptierten selbst, sonder mitunter von einer Anzahl an Faktoren ab, nicht zuletzt von dem Vorhandensein früher Vorurteile und der Unterstützung seitens des Adoptivelternhauses. Es ist nicht verantwortlich und angemessen, aus reiner Spekulation über den möglichen Ausgang eines Kontaktes diesen im Voraus zu verhindern und dem Kind diese Möglichkeit zu versagen. Laut den Ergebnissen ist ein anderes Ereignis im Leben eines Adoptierten für sein Selbstwertgefühl und sein Bindungsverhalten entscheidender. Dies bezieht sich auf die Kommunikation der Adoptiveltern mit ihrem Kind. Wichtigster Prädiktor für ein gesundes Selbstbewusstsein und günstige Bindungseinstellungen war laut den Ergebnissen das Geständnis der Eltern über die Adoption. Eltern, die diesbezüglich offen und ehrlich ihren Kindern gegenüber waren, wurden in ihrem Erziehungsverhalten von diesen als emotional wärmer und weniger kontrollierend bewertet als Eltern, die ihre Kinder nicht über die Adoption aufgeklärt hatten. Adoptierte, die über ihre eigene Adoption nicht aufgeklärt wurden, zeigten vermindertes Selbstwertgefühl, verminderte Widerstandsfähigkeit und schlechtere Bindungseinstellungen, wobei die Skala „Nähe“, das heißt die Messung zur Fähigkeit, andere Menschen an sich heran zu lassen, Werte außerhalb des Normbereiches aufzeigte und man hier sogar von einem pathologisch veränderten Persönlichkeitsmerkmal sprechen kann. Auch das Alter bei Aufklärung spielt eine wichtige Rolle. Frühe Aufklärung über die Adoption, in der vorliegenden Studie wurde ein Zeitpunkt bis zum neunten Lebensjahr als früh definiert, führte zu höherem allgemeinen Selbstwertgefühl und einer erhöhten Widerstandsfähigkeit. Auf die Bindungseinstellungen schien der Zeitpunkt der Aufklärung keine Auswirkung zu haben. Eltern, die ihre Kinder früh über ihre Adoption informierten, wurden als weniger überbehütend und kontrollierend bewertet. Wichtig zu erwähnen ist, dass die Werte bei den Rechnungen der Gruppenvergleiche, mit Ausnahme des oben erwähnten Wertes für die Skala „Nähe“ der Adult Attachment Scale, alle im Normbereich lagen. Es liegen also, auch im Vergleich mit den Normstichproben, keine Befunde vor, die bei der adoptierten Stichprobe auf Pathologien hinweisen. Zu finden sind jedoch leichte Abweichungen von der Norm, die statistisch gesehen von Signifikanz sind und auch inhaltlich wichtige Hinweise auf die Folgen von Adoption liefern. Obwohl der Großteil der Adoptierten (90.3%) über ihre Adoption aufgeklärt worden war und dies laut oben beschriebener Ergebnisse eine günstiger Faktor bezüglich der Entwicklung von Selbstwert und Bindung ist, lässt sich feststellen, dass Adoptierte im Vergleich zu den nicht adoptierten Normstichproben ein vermindertes Selbstwertgefühl und ungünstigere Bindungseinstellungen aufwiesen. Weitere Einflussfaktoren dieser Persönlichkeitsmerkmale im Leben eines Adoptierten müssten zukünftig identifiziert werden. Seelische Vorerkrankungen, wie beispielsweise eine Depression, könnten einen solchen Einflussfaktor darstellen. 20.6% der Adoptierten der vorliegenden Studie gaben an, seelisch erkrankt zu sein, darunter waren 12.1% depressiv, was zu zusätzlichen Rechnungen veranlasste. Bei diesen Berechnungen mit dem Vergleich von depressiven Adoptierten versus seelisch gesunden Adoptierten fiel auf, dass die depressive Gruppe niedrigere Werte auf Skalen des Selbstwertes und der Resilienz aufwies, sowie schlechtere Bindungseinstellungen hatte als die seelisch gesunde Gruppe. Depressive Adoptierte waren zusätzlich neurotischer und bewerteten ihre Elternhäuser als emotional kühler. Ebenso hatten sie mehr Ablehnung und Strafe durch die Mütter erfahren. Besonders hervorzuheben sind jedoch die Ergebnisse auf den Skalen der emotionalen Selbstwertschätzung und der Bindungseinstellung Angst. Hierbei lagen die Werte der depressiven adoptierten Gruppe deutlich außerhalb des Normbereichs und wiesen auf pathologisch veränderte Persönlichkeitsmerkmale hin. Depressive hatten somit deutlich schlechtere Einstellungen gegenüber sich selbst als Adoptierte, die nicht depressiv waren. Ebenso hatten sie eine sehr schlechte Selbstachtung und litten vermehrt unter Ängsten. Es bleibt also die Frage offen, inwiefern seelische Vorerkrankungen, die in der vorliegenden Studie zu einem hohen Prozentsatz vertreten waren, zu den schlechteren Ergebnissen bezüglich Selbstwert und Bindung Adoptierter im Vergleich zu Normstichproben beitragen. Es bleibt offen, ob Adoption alleine ein Risikofaktor für vermindertes Selbstbewusstsein und schlechte Bindungseinstellungen ist oder ob diese Ergebnisse nicht vielmehr durch andere Faktoren, wie seelische Erkrankungen, beeinflusst werden. Abschließend lässt sich feststellen, dass die Ehrlichkeit der Adoptiveltern und der offene Umgang mit dem Thema der Adoption entscheidend zur Bildung eines gesunden Selbstwertes und günstiger Bindungseinstellungen ihres Kindes beitragen. Ebenso sollten Adoptierte nicht lange in Unwissenheit über ihre Herkunft gelassen werden. Ein früher Zeitpunkt der Aufklärung über die eigene Adoption hat positiven Einfluss auf die Entwicklung des Selbstwertes, der Widerstandsfähigkeit und der Bindungseinstellung. Weiterhin ist zu sehen, dass die reine Kontaktmöglichkeit zu den leiblichen Eltern für die Entwicklung von Selbstwertgefühl, Resilienz und Bindung keine entscheidende Rolle zu spielen scheint. Eine offene Adoptionsform alleine, in der der Kontakt zwischen Adoptivfamilie und leiblichen Eltern von Anfang an aufrechterhalten bzw. zu einem späteren Zeitpunkt wieder aufgenommen wird, scheint für die Entwicklung des Adoptivkindesnicht die entscheidende Rolle zu spielen. Interessant bleibt die Frage nach dem Einfluss von Adoption als Risikofaktor für die Entwicklung von seelischen Erkrankungen und hierunter insbesondere Depression. Ein hoher Prozentsatz der Adoptierten dieser Studie gab an, seelisch erkrankt zu sein (20.6%), die häufigste genannte Diagnose darunter war die Depression (12.1%). Die Diagnosehäufigkeiten und Prädiktoren für die Entwicklung von Depressionen müssen zukünftig genauer untersucht werden. Da es sich bei der vorliegenden Studie um eine anonymisierte Fragebogenstudie ohne zusätzliche Führung von Interviews handelte, war es nicht möglich, Fragen zu Angaben der Probanden zu beantworten und Missverständnisse zu klären. Manches Ergebnis, wie die Angabe der Probanden, depressiv zu sein, oder die Untersuchung der Anzahl von Heimaufenthalten schien sehr fraglich. Zukünftige Studien sollten zusätzlich Interviews mit den Teilnehmern beinhalten, da sich so Missverständnisse am leichtesten klären lassen und Rückfragen möglich sind. Zur Erfassung von seelischen und körperlichen Erkrankungen können Testverfahren wie das Brief Symptom Inventory (Franke 2000) und die Hospital Anxiety and Depression Scale (Lingen, Buss, Snaith 2005) verwendet werden. Die Teilnehmerzahl der Studie belief sich auf 165 Probanden. Bei 75.8% der Probanden handelte es sich um weibliche Teilnehmerinnen. Da Studien bereits belegt haben, dass das Geschlecht einen entscheidenden Einfluss auf die Ergebnisse bei Untersuchungen zu Persönlichkeitsmerkmalen hat (Freeark, Rosenberg et al.2005), müsste man die Unterteilung in männliche und weibliche Probandengruppen vornehmen. Aufgrund der zu kleinen Teilnehmerzahl an Männern konnte diesin der vorliegenden Studie nicht geschehen. Ebenso betrifft dies die Einteilung der Probanden in die Altersgruppen bei Aufklärung über die Adoption. Hier konnte aufgrund der kleinen Teilnehmerzahl nur eine Unterteilung in ein Alter bis zum neunten Lebensjahr und ab dem zehnten Lebensjahr erfolgen. Bis zum neunten Lebensjahr durchläuft das Kind jedoch wichtige Entwicklungsschritte auf den Gebieten der Bindung zu Bezugspersonen, Entwicklung eines Selbstbildes und Integration in eine Gemeinschaft, in denen das Kind die Information über die eigene Adoption unterschiedlich verarbeitet. Anhand größerer Teilnehmerzahlen kann eine genauere Einteilung in verschiedene Altersklassen auch vor dem neunten Lebensjahr erfolgen und so der optimale Zeitpunkt der Aufklärung über die Adoption genauer festgelegt werden. Ebenso wichtig wäre eine weitere Einteilung in Inlands- vs. Auslandsadoptionen. Kinder, die aus dem Ausland adoptiert wurden, haben vermehrt Integrationsschwierigkeiten aufgrund ihres andersartigen Erscheinungsbildes, die sich auch auf die Entwicklung von Selbstbewusstsein auswirken können (von Borczyskowski, Hjern et al. 2006). Dies müsste man anhand der Unterteilung in Inlands –und Auslandsadoptionen genauer untersuchen. Die Probanden der vorliegenden Studie gaben an, ein emotional wärmeres und empathischeres Elternhaus gehabt zu haben als die Normstichprobe. Bei der Bewertung des Elternhauses handelt es sich jedoch lediglich um die Einschätzung der Teilnehmer, es ist eine subjektive Bewertung. Über die Qualität der Elternhäuser an sich kann man aus objektiver Sicht keine Aussage machen. Weitere Testverfahren zur Messung der Beziehungsqualitäten zwischen den Adoptierten und ihren Eltern und/oder der Persönlichkeitsmerkmale der Adoptiveltern müssten hierfür herangezogen werden. Ebenso wenig kann mit der Adult Attachment Scale das tatsächliche Bindungsverhalten gemessen werden. Vielmehr gibt dieses Testverfahren einen Hinweis auf die Bindungseinstellungen der Probanden. Zur Untersuchung von Bindungsverhalten liegen keine Testverfahren in Fragebogenform vor, dies müsste auf andere Weise erfolgen, beispielsweise mit Hilfe des Adult Attachment Interviews. Es lässt sich also feststellen, dass zur genaueren Untersuchung von Adoptierten eine größere Teilnehmerzahl erforderlich wird. Eine erweiterte Testbatterie und die Führung von Interviews können Missverständnisse und offene Fragen aus dem Weg räumen und ermöglichen so eine differenziertere Auswertung der Ergebnisse.
Ziel dieser Arbeit ist es, Art, Häufigkeit und Dauer der Komplikationen nach sekundärer Kieferspaltosteoplastik mit Beckenkammtransplantation bei Patienten mit Lippen-, Kiefer-, Gaumenspalten aufzuzeigen. Dazu wurden in dieser retrospektiven Studie die Daten von 236 Patienten, die im Zeitraum zwischen Juli 1986 und August 1997 im Klinikum „Rechts der Isar“ der Technischen Universität München eine Spanentnahme am vorderen Beckenkamm zur autologen Transplantation im Mund-Kiefer-Gesichtsbereich erhielten, ausgewertet. Die Datenerhebung erfolgte durch Analyse der OP-Berichte, Krankenblätter, persönliche Befragung und körperliche Untersuchung aller 236 Patienten. Der Focus dieser Arbeit liegt bei der Darstellung der Ergebnisse der 182 LKG-Patienten, welche mit den anderen Indikationsgruppen verglichen wurden. Das durchschnittliche OP-Alter der LKGPatienten betrug 13,8 Jahre, die Daten wurden im Mittel 5,3 Jahre nach der Entnahme-OP erfasst. Chirurgische Frühkomplikationen konnten bei 15 LKG-Patienten (8,2 %) beobachtet werden, die bei zwei Patienten (1,1 %) einer chirurgischen Nachbehandlung (Hämatomausräumung) bedurften. Die Komplikationsrate war bei den Patienten mit der Diagnose LKG (8,2 %) geringer als bei den übrigen Indikationsgruppen zur Knochenentnahme. Diese reichte von 11 % bei Tumorpatienten bis 26 % bei Patienten mit Dysgnathie. Eine Gehbehinderung war mit 91,7 % die häufigste nichtchirurgische Komplikation. Die Gehbehinderung war bei 13,7 % der LKG-Patienten so stark, dass sie auf Gehhilfen angewiesen waren. 78 % waren ebenfalls gehbehindert, benötigten jedoch keine Gehhilfe. Starke postoperative Schmerzen in der Entnahmeregion nannten 48,4 %. Über postoperative Sensibilitätsstörungen klagten 28,6 % der LKG-Patienten. Zum Untersuchungszeitpunkt, im Durchschnitt 5,3 Jahre nach dem operativen Eingriff, wurden immerhin noch bei 6,0 % der LKG-Patienten persistierende Schmerzen als Spätfolge gefunden. Sensibilitätsstörungen persistierten bei 9,9 % aller Spalt-Patienten. Bei der subjektiven Bewertung fanden sich desweiteren bei 7,8 % nichtschmerzhafte bleibende Beschwerden wie Wetterfühligkeit. Mit der Ästhetik der Narbe im Entnahmebereich waren 86,3 % zufrieden, unzufrieden nur 7,1 %. Bei entsprechender Indikation würden sich 91,8% der LKG-Patienten wieder für die gleiche Therapieform (Beckenkammtransplantation) entscheiden, immerhin 14 Patienten (7,9 %) würden diesen Eingriff unter keinen Umständen wiederholen bzw. nur Fremdmaterial als Augmentat verwenden, als sich erneut einer Entnahme-OP zu unterziehen. Beim isolierten Vergleich der Schmerzhaftigkeit werteten 60,4% die Becken-Operation und 18,1 % die Operation im Mund-Kiefer-Gesichtsbereich als schmerzhafter. Beim Vergleich der Gesamtbefindlichkeit des postoperativen Zustandes hingegen wurden der Mund-Kiefer-Gesichtsbereich von 56,0 % und der Beckenbereich nur von 19,8 % als der insgesamt unangenehmere Befund bezeichnet. Für 29,7 % war die Entnahme-Operation schlimmer als erwartet, für 25,3 % deckte sich die Erfahrung durch die Operation mit ihrer Erwartungshaltung und 13,7 % erwarteten stärkere Beschwerden, als sie letztendlich auftraten. 31,3 % hatten keine Erwartungshaltung oder machten keine Angabe. Zusammenfassend lässt sich feststellen, dass mit der autologen Beckenkammtransplantation für Patienten mit Lippen-Kiefer-Gaumenspalten eine Therapieform zur Verfügung steht, die ästhetisch und funktionell sehr gute Ergebnisse ermöglicht. Dennoch sind mit der Entnahmeoperation Komplikationen und Langzeitschäden verbunden, über die der Patient aufgeklärt werden muss und die insgesamt das Operationsergebnis schmälern. Um zukünftig die Entnahme von Beckenkammspongiosa zu umgehen, sollten zukünftig vermehrt Knochenersatzstoffe zur Defektauffüllung zum Einsatz kommen. Dies ist Inhalt momentan laufender, intensiver Forschung.
CD95 co-stimulation blocks activation of naive T cells by inhibiting T cell receptor signaling

CD95 is a multifunctional receptor that induces cell death or proliferation depending on the signal, cell type, and cellular context. Here, we describe a thus far unknown function of CD95 as a silencer of T cell activation. Naive human T cells triggered by antigen-presenting cells expressing a membrane-bound form of CD95 ligand (CD95L) or stimulated by anti-CD3 and -CD28 antibodies in the presence of recombinant CD95L had reduced activation and proliferation, whereas preactivated, CD95-sensitive T cells underwent apoptosis. Triggering of CD95 during T cell priming interfered with proximal T cell receptor signaling by inhibiting the recruitment of ζ-chain–associated protein of 70 kD, phospholipase-γ, and protein kinase C-θ into lipid rafts, thereby preventing their mutual tyrosine protein phosphorylation. Subsequently, Ca2+ mobilization and nuclear translocation of transcription factors NFAT, AP1, and NF-κB were strongly reduced, leading to impaired cytokine secretion. CD95-mediated inhibition of proliferation in naive T cells could not be reverted by the addition of exogenous interleukin-2 and T cells primed by CD95 co-stimulation remained partially unresponsive upon secondary T cell stimulation. HIV infection induced CD95L expression in primary human antigeen-presenting cells, and thereby suppressed T cell activation, suggesting that CD95/CD95L-mediated silencing of T cell activation represents a novel mechanism of immune evasion.

Abstract: Quer zur Standardrezeption analysiert Claudia Bruns in ihrer Dissertation Blühers Konzeption von Mann, Männlichkeit und mann-männlicher Beziehung unter besonderer Berücksichtigung damit einhergehender Ausschlüsse. Zentral legt sie das Konzept des Männerbundes zugrunde, das den Dreh- und Angelpunkt der vorliegenden Untersuchung bildet, und zieht die Theorien Blühers als paradigmatisches Beispiel dafür heran. Dieses Vorgehen ist aus verschiedenen Gründen klug gewählt und vorteilhaft. Bislang ist kaum der ideologisch vielschichtige Gehalt von Diskussionen, wie sie Blüher führte und führen konnte, in die Geschichts- und Gesellschaftswissenschaften eingegangen. Zudem gab es bislang kein Gesamtverzeichnis der Schriften Blühers, wodurch die Rezeption erschwert wurde, und das sich nun im Anhang der Arbeit dankenswerterweise findet.

Abstract: Die Idee der Gleichheit bei den beiden Klassikern der politischen Philosophie Hobbes und Rousseau soll vor dem Hintergrund der neueren feministischen Debatten um Gleichheit und Differenz betrachtet werden. Doch die Ausführungen verlaufen stark entlang der bloßen Wiedergabe der Originalliteratur und deren Rezeptionsgeschichte – unter besonderer Berücksichtigung von Lacan. Die vielfältigen und großen Diskussionen in der feministischen Theorie finden dabei viel zu wenig Beachtung.
Background: Human primary monocytes are refractory to infection with the human immunodeficiency virus 1 (HIV-1) or transduction with HIV-1-derived vectors. In contrast, efficient single round transduction of monocytes is mediated by vectors derived from simian immunodeficiency virus of sooty mangabeys (SIVsmmPBj), depending on the presence of the viral accessory protein Vpx.
Methods and Findings: Here we analyzed whether Vpx of SIVsmmPBj is sufficient for transduction of primary monocytes by HIV-1-derived vectors. To enable incorporation of PBj Vpx into HIV-1 vector particles, a HA-Vpr/Vpx fusion protein was generated. Supplementation of HIV-1 vector particles with this fusion protein was not sufficient to facilitate transduction of human monocytes. However, monocyte transduction with HIV-1-derived vectors was significantly enhanced after delivery of Vpx proteins by virus-like particles (VLPs) derived from SIVsmmPBj. Moreover, pre-incubation with Vpx-containing VLPs restored replication capacity of infectious HIV-1 in human monocytes. In monocytes of non-human primates, single-round transduction with HIV-1 vectors was enabled.
Conclusion: Vpx enhances transduction of primary human and even non-human monocytes with HIV-1-derived vectors, only if delivered in the background of SIVsmmPBj-derived virus-like particles. Thus, for accurate Vpx function the presence of SIVsmmPBj capsid proteins might be required. Vpx is essential to overcome a block of early infection steps in primary monocytes.

Water-filtered infrared-A (wIRA), as a special form of heat radiation with a high tissue penetration and a low thermal load to the skin surface, can improve the healing of acute and chronic wounds both by thermal and thermic as well as by non-thermal and non-thermic effects. wIRA increases tissue temperature (+2.7°C at a tissue depth of 2 cm), tissue oxygen partial pressure (+32% at a tissue depth of 2 cm) and tissue perfusion. These three factors are decisive for a sufficient supply of tissue with energy and oxygen and consequently also for wound healing and infection defense.
wIRA can considerably alleviate pain (without any exception during 230 irradiations) with substantially less need for analgesics (52–69% less in the groups with wIRA compared to the control groups). It also diminishes exudation and inflammation and can show positive immunomodulatory effects. The overall evaluation of the effect of irradiation as well as the wound healing and the cosmetic result (assessed on visual analogue scales) were markedly better in the group with wIRA compared to the control group. wIRA can advance wound healing (median reduction of wound size of 90% in severely burned children already after 9 days in the group with wIRA compared to 13 days in the control group; on average 18 versus 42 days until complete wound closure in chronic venous stasis ulcers) or improve an impaired wound healing (reaching wound closure and normalization of the thermographic image in otherwise recalcitrant chronic venous stasis ulcers) both in acute and in chronic wounds including infected wounds. After major abdominal surgery there was a trend in favor of the wIRA group to a lower rate of total wound infections (7% versus 15%) including late infections following discharge from hospital (0% versus 8%) and a trend towards a shorter postoperative hospital stay (9 versus 11 days).
Even the normal wound healing process can be improved.
The mentioned effects have been proven in six prospective studies, with most of the effects having an evidence level of Ia/Ib.
wIRA represents a valuable therapy option and can generally be recommended for use in the treatment of acute as well as of chronic wounds.

Der Grund für diesen Zellverlust durch Apoptose könnte sein, dass es im Alter zu erhöhtem
oxidativen Streß und zu einer Veränderung in der Konzentration der an der Apoptose beteiligten
Proteine und Faktoren kommt. Diese Studie befaßt sich mit der neuronalen Apoptose und den daran
beteiligten Proteinen und Regulatoren. Ziel war es, herauszufinden, ob es im Alter zu einer
gesteigerten Apoptoserate im Hippokampus und zu einer Veränderung der Konzentration von
Proteinen kommt, die das Auftreten bzw. die Hemmung der Apoptose begünstigen könnte.
Untersucht wurden die Hippokampi von Pavianen als mögliches Primaten-Modell für altersbedingte
Pathologien, die das menschliche Gehirn betreffen können.
Um apoptotische Zellen im Hippokampus der Paviane nachzuweisen, wurde mit der TUNEL-Methode
gearbeitet und mittels Immunhistochemie untersucht, welchen Effekt das Alter auf die Verteilung der
Immunreaktivität von Cytochrom C, AIF, Bax, Bcl-2, Caspase-3 sowie von XIAP, einem Inhibitor der
Apoptose, hat. Auch die Immunreaktivität der Enzyme PARP und ICAD wurde dargestellt. Die
Ergebnisse zeigen, dass in den Hippokampi der gealterten Paviane das anti-apoptotische Protein Bcl-2
in unveränderter Reaktivität vorlag und das pro-apoptotische Protein Bax hochreguliert war. Es gab
eine Umverteilung und möglicherweise Freisetzung von Cytochrom C, jedoch aber keine Aktivierung
von Caspase-3. Diese Ergebnisse zeigen eine gewebepezifische Änderung des Bcl-2/Bax-
Verhältnisses in den Hippokampi der alten Primaten, die zur Beeinflussung der Cytochrom CFreisetzung
beitragen, ohne jedoch zur Apoptose der Zelle zu führen. Apoptotische Zellen konnten
mittels TUNEL-Färbung nicht nachgewiesen werden. Das könnte damit zusammenhängen, dass
XIAP, ein inhibitorisches Regulatorprotein der Apoptose, bei den gealterten Pavianen hochreguliert
war, ebenso wie das DNA-Reparaturenzym PARP. Auch das Enzym ICAD, welches in aktivierter
Form während der Apoptose wesentlich zum Chromatinabbau beiträgt zeigte im Alter eine erhöhte
Aktivität. Da in einer Studie über Tau-Pathologie im Hippokampus von Pavianen für das Auftreten
dieser Pathologie ein Zusammenhang mit dem Alter nachgewiesen werden konnte, und man weiß,
dass Zellen in der Nähe der β-Amyloidablagerung bei M.Alzheimer durch Apoptose absterben, wurde
mit Hilfe der Immunfloureszenz die Reaktivität des Enzyms Neprelysin untersucht, welches für den
β-Amyloidablagerung zuständig ist. Dieses war in seiner Reaktivität in den Hippokampi
der älteren Paviane im Gegensatz zu der in den Hippokampi der jüngeren Primaten erniedrigt, was auf
eine altersbedingte Disposition zu diesen Plaques hinweist.

Drawing on an analysis of the debate around the German reader "Kulturgeographie" published in 2003, we suggest that this case provides new insights into the "geography of critical geography". We briefly discuss the history of critical geography in Germany, leading to a comparison of the conditions of critical geography around 1980 and in recent years. The focus is on two factors in the changed role of critical perspectives in German geography: (1) the growing internationalisation of German geography, which opened new avenues and allowed new approaches to enter the discipline; and (2) the high citation indices of "critical" journals, which leads to an enhanced reputation and a high significance of international critical geography in the German discipline. However, we draw an ambiguous conclusion: the increased role of critical approaches in German geography is linked to a growing neoliberalisation of academia and a decline of critical approaches in other disciplines.

This report focuses on the 3rd PrivacyOS conference, which was held in Vienna, October 26th and 27th 2009, co-located with the Austrian Big Brother Awards. 50 participants attended the conference and devised the agenda with 21 presentations in two parallel tracks. The topics of the presentations discussed included, amongst others: data protection awareness, data protection in healthcare, data protection in the Web 2.0, privacy-related technologies such as EnCoRe, TOR or Microformats as well as regulatory, cultural and sociological implications of data protection. Also at the 3rd PrivacyOS conference the software product “KiwiSecurity” was awarded the EuroPriSe Seal (European Privacy Seal, www.european-privacy-seal.eu). EuroPriSe is an initiative of the data protection authority Unabhängiges Landeszentrum für Datenschutz Schleswig-Holstein (ULD), Germany. It has been started as a European Project under the eTEN programme.
Chapter 1 contains the general background of our work. We briefly discuss important aspects of quantum chromodynamics (QCD) and introduce the concept of the chiral condensate as an order parameter for the chiral phase transition. Our focus is on the concept of universality and the arguments why the O(4) model should fall into the same universality class as the effective Lagrangian for the order parameter of (massless) two-flavor QCD. Chapter 2 pedagogically explains the CJT formalism and is concerned with the WKB method. In chapter 3 the CJT formalism is then applied to a simple Z(2) symmetric toy model featuring a one-minimum classical potential. As for all other models we are concerned with in this thesis, we study the behavior at nonzero temperature. This is done in 1+3 dimensions as well as in 1+0 dimensions. In the latter case we are able to compare the effective potential at its global minimum (which is minus the pressure) with our result from the WKB approximation. In chapter 4 this program is also carried out for the toy model with a double-well classical potential, which allows for spontaneous symmetry breaking and tunneling. Our major interest however is in the O(2) model with the fields treated as polar coordinates. This model can be regarded as the first step towards the O(4) model in four-dimensional polar coordinates. Although in principle independent, all subjects discussed in this thesis are directly related to questions arising from the investigation of this particular model. In chapter 5 we start from the generating functional in cartesian coordinates and carry out the transition to polar coordinates. Then we are concerned with the question under which circumstances it is allowed to use the same Feynman rules in polar coordinates as in cartesian coordinates. This question turns out to be non-trivial. On the basis of the common Feynman rules we apply the CJT formalism in chapter 6 to the polar O(2) model. The case of 1+0 dimensions was intended to be a toy model on the basis of which one could more easily explore the transition to polar coordinates. However, it turns out that we are faced with an additional complication in this case, the infrared divergence of thermal integrals. This problem requires special attention and motivates the explicit study of a massless field under topological constraints in chapter 8. In chapter 7 we investigate the cartesian O(2) model in 1+0 dimensions. We compare the effective potential at its global minimum calculated in the CJT formalism and via the WKB approximation. Appendix B reviews the derivation of standard thermal integrals in 1+0 and 1+3 dimensions and constitutes the basis for our CJT calculations and the discussion of infrared divergences. In chapter 9 we discuss the so-called path integral collapse and propose a solution of this problem. In chapter 10 we present our conclusions and an outlook. Since we were interested in organizing our work as pedagogical as possible within the narrow scope of a diploma thesis, we decided to make extensive use of appendices. Appendices A-H are intended for students who are not familiar with several important concepts we are concerned with. We will refer to them explicitly to establish the connection between our work and the general context in which it is settled.
We use observations of total particle number concentration at 36 worldwide sites and a global aerosol model to quantify the primary and secondary sources of particle number. We show that emissions of primary particles can reasonably reproduce the spatial pattern of observed condensation nuclei (CN) (R2=0.51) but fail to explain the observed seasonal cycle at many sites (R2=0.1). The modeled CN concentration in the free troposphere is biased low (normalised mean bias, NMB=−88%) unless a secondary source of particles is included, for example from binary homogeneous nucleation of sulfuric acid and water (NMB=−25%). Simulated CN concentrations in the continental boundary layer (BL) are also biased low (NMB=−74%) unless the number emission of anthropogenic primary particles is increased or an empirical BL particle formation mechanism based on sulfuric acid is used. We find that the seasonal CN cycle observed at continental BL sites is better simulated by including a BL particle formation mechanism (R2=0.3) than by increasing the number emission from primary anthropogenic sources (R2=0.18). Using sensitivity tests we derive optimum rate coefficients for this nucleation mechanism, which agree with values derived from detailed case studies at individual sites.
Bacterial porin disrupts mitochondrial membrane potential and sensitizes host cells to apoptosis

The bacterial PorB porin, an ATP-binding beta-barrel protein of pathogenic Neisseria gonorrhoeae, triggers host cell apoptosis by an unknown mechanism. PorB is targeted to and imported by host cell mitochondria, causing the breakdown of the mitochondrial membrane potential (delta psi m). Here, we show that PorB induces the condensation of the mitochondrial matrix and the loss of cristae structures, sensitizing cells to the induction of apoptosis via signaling pathways activated by BH3-only proteins. PorB is imported into mitochondria through the general translocase TOM but, unexpectedly, is not recognized by the SAM sorting machinery, usually required for the assembly of beta-barrel proteins in the mitochondrial outer membrane. PorB integrates into the mitochondrial inner membrane, leading to the breakdown of delta psi m. The PorB channel is regulated by nucleotides and an isogenic PorB mutant defective in ATP-binding failed to induce delta psi m loss and apoptosis, demonstrating that dissipation of delta psi m is a requirement for cell death caused by neisserial infection.
Neisseria meningitidis is a strictly human pathogen that has two facets since asymptomatic carriage can unpredictably turn into fulminant forms of infection. Meningococcal pathogenesis relies on the ability of the bacteria to break host epithelial or endothelial cellular barriers. Highly restrictive, yet poorly understood, mechanisms allow meningococcal adhesion to cells of only human origin. Adhesion of encapsulated and virulent meningococci to human cells relies on the expression of bacterial type four pili (T4P) that trigger intense host cell signalling. Among the components of the meningococcal T4P, the concomitantly expressed PilC1 and PilC2 proteins regulate pili exposure at the bacterial surface, and until now, PilC1 was believed to be specifically responsible for T4P-mediated meningococcal adhesion to human cells. Contrary to previous reports, we show that, like PilC1, the meningococcal PilC2 component is capable of mediating adhesion to human ME180 epithelial cells, with cortical plaque formation and F-actin condensation. However, PilC1 and PilC2 promote different effects on infected cells. Cellular tracking analysis revealed that PilC1-expressing meningococci caused a severe reduction in the motility of infected cells, which was not the case when cells were infected with PilC2-expressing strains. The amount of both total and phosphorylated forms of EGFR was dramatically reduced in cells upon PilC1-mediated infection. In contrast, PilC2-mediated infection did not notably affect the EGFR pathway, and these specificities were shared among unrelated meningococcal strains. These results suggest that meningococci have evolved a highly discriminative tool for differential adhesion in specific microenvironments where different cell types are present. Moreover, the fine-tuning of cellular control through the combined action of two concomitantly expressed, but distinctly regulated, T4P-associated variants of the same molecule (i.e. PilC1 and PilC2) brings a new model to light for the analysis of the interplay between pathogenic bacteria and human host cells.
Background: Intrauterine growth restriction is associated with an increased future risk for developing cardiovascular diseases. Hypoxia in utero is a common clinical cause of fetal growth restriction. We have previously shown that chronic hypoxia alters cardiovascular development in chick embryos. The aim of this study was to further characterize cardiac disease in hypoxic chick embryos. Methods: Chick embryos were exposed to hypoxia and cardiac structure was examined by histological methods one day prior to hatching (E20) and at adulthood. Cardiac function was assessed in vivo by echocardiography and ex vivo by contractility measurements in isolated heart muscle bundles and isolated cardiomyocytes. Chick embryos were exposed to vascular endothelial growth factor (VEGF) and its scavenger soluble VEGF receptor-1 (sFlt-1) to investigate the potential role of this hypoxia-regulated cytokine. Principal Findings: Growth restricted hypoxic chick embryos showed cardiomyopathy as evidenced by left ventricular (LV) dilatation, reduced ventricular wall mass and increased apoptosis. Hypoxic hearts displayed pump dysfunction with decreased LV ejection fractions, accompanied by signs of diastolic dysfunction. Cardiomyopathy caused by hypoxia persisted into adulthood. Hypoxic embryonic hearts showed increases in VEGF expression. Systemic administration of rhVEGF165 to normoxic chick embryos resulted in LV dilatation and a dose-dependent loss of LV wall mass. Lowering VEGF levels in hypoxic embryonic chick hearts by systemic administration of sFlt-1 yielded an almost complete normalization of the phenotype. Conclusions/Significance: Our data show that hypoxia causes a decreased cardiac performance and cardiomyopathy in chick embryos, involving a significant VEGF-mediated component. This cardiomyopathy persists into adulthood.
Background: Mild therapeutic hypothermia following cardiac arrest is neuroprotective, but its effect on myocardial dysfunction that is a critical issue following resuscitation is not clear. This study sought to examine whether hypothermia and the combination of hypothermia and pharmacological postconditioning are cardioprotective in a model of cardiopulmonary resuscitation following acute myocardial ischemia. Methodology/Principal Findings: Thirty pigs (28–34 kg) were subjected to cardiac arrest following left anterior descending coronary artery ischemia. After 7 minutes of ventricular fibrillation and 2 minutes of basic life support, advanced cardiac life support was started according to the current AHA guidelines. After successful return of spontaneous circulation (n = 21), coronary perfusion was reestablished after 60 minutes of occlusion, and animals were randomized to either normothermia at 38°C, hypothermia at 33°C or hypothermia at 33°C combined with sevoflurane (each group n = 7) for 24 hours. The effects on cardiac damage especially on inflammation, apoptosis, and remodeling were studied using cellular and molecular approaches. Five animals were sham operated. Animals treated with hypothermia had lower troponin T levels (p<0.01), reduced infarct size (34±7 versus 57±12%; p<0.05) and improved left ventricular function compared to normothermia (p<0.05). Hypothermia was associated with a reduction in: (i) immune cell infiltration, (ii) apoptosis, (iii) IL-1beta and IL-6 mRNA up-regulation, and (iv) IL-1beta protein expression (p<0.05). Moreover, decreased matrix metalloproteinase-9 activity was detected in the ischemic myocardium after treatment with mild hypothermia. Sevoflurane conferred additional protective effects although statistic significance was not reached. Conclusions/Significance: Hypothermia reduced myocardial damage and dysfunction after cardiopulmonary resuscitation possible via a reduced rate of apoptosis and pro-inflammatory cytokine expression.
Reciprocal t(9;22) ABL/BCR fusion proteins: leukemogenic potential and effects on B cell commitment

Background: t(9;22) is a balanced translocation, and the chromosome 22 breakpoints (Philadelphia chromosome – Ph+) determine formation of different fusion genes that are associated with either Ph+ acute lymphatic leukemia (Ph+ ALL) or chronic myeloid leukemia (CML). The "minor" breakpoint in Ph+ ALL encodes p185BCR/ABL from der22 and p96ABL/BCR from der9. The "major" breakpoint in CML encodes p210BCR/ABL and p40ABL/BCR. Herein, we investigated the leukemogenic potential of the der9-associated p96ABL/BCR and p40ABL/BCR fusion proteins and their roles in the lineage commitment of hematopoietic stem cells in comparison to BCR/ABL. Methodology: All t(9;22) derived proteins were retrovirally expressed in murine hematopoietic stem cells (SL cells) and human umbilical cord blood cells (UCBC). Stem cell potential was determined by replating efficiency, colony forming - spleen and competitive repopulating assays. The leukemic potential of the ABL/BCR fusion proteins was assessed by in a transduction/transplantation model. Effects on the lineage commitment and differentiation were investigated by culturing the cells under conditions driving either myeloid or lymphoid commitment. Expression of key factors of the B-cell differentiation and components of the preB-cell receptor were determined by qRT-PCR. Principal Findings: Both p96ABL/BCR and p40ABL/BCR increased proliferation of early progenitors and the short term stem cell capacity of SL-cells and exhibited own leukemogenic potential. Interestingly, BCR/ABL gave origin exclusively to a myeloid phenotype independently from the culture conditions whereas p96ABL/BCR and to a minor extent p40ABL/BCR forced the B-cell commitment of SL-cells and UCBC. Conclusions/Significance: Our here presented data establish the reciprocal ABL/BCR fusion proteins as second oncogenes encoded by the t(9;22) in addition to BCR/ABL and suggest that ABL/BCR contribute to the determination of the leukemic phenotype through their influence on the lineage commitment.
Streptococcus agalactiae is a well-known pathogen for neonates and immunocompromized adults. Beyond the neonatal period, S. agalactiae is rarely found in the respiratory tract. During 2002–2008 we noticed S. agalactiae in respiratory secretions of 30/185 (16%) of cystic fibrosis (CF) patients. The median age of these patients was 3–6 years older than the median age CF patients not harboring S. agalactiae. To analyze, if the S. agalactiae isolates from CF patients were clonal, further characterization of the strains was achieved by capsular serotyping, surface protein determination and multilocus sequence typing (MLST). We found a variety of sequence types (ST) among the isolates, which did not substantially differ from the MLST patterns of colonizing strains from Germany. However serotype III, which is often seen in colonizing strains and invasive infections was rare among CF patients. The emergence of S. agalactiae in the respiratory tract of CF patients may represent the adaptation to a novel host environment, supported by the altered surfactant composition in older CF patients.
Borrelia recurrentis, the etiologic agent of louse-borne relapsing fever in humans, has evolved strategies, including antigenic variation, to evade immune defence, thereby causing severe diseases with high mortality rates. Here we identify for the first time a multifunctional surface lipoprotein of B. recurrentis, termed HcpA, and demonstrate that it binds human complement regulators, Factor H, CFHR-1, and simultaneously, the host protease plasminogen. Cell surface bound factor H was found to retain its activity and to confer resistance to complement attack. Moreover, ectopic expression of HcpA in a B. burgdorferi B313 strain, deficient in Factor H binding proteins, protected the transformed spirochetes from complement-mediated killing. Furthermore, HcpA-bound plasminogen/plasmin endows B. recurrentis with the potential to resist opsonization and to degrade extracellular matrix components. Together, the present study underscores the high virulence potential of B. recurrentis. The elucidation of the molecular basis underlying the versatile strategies of B. recurrentis to escape innate immunity and to persist in human tissues, including the brain, may help to understand the pathological processes underlying louse-borne relapsing fever.
Background: Glioblastoma is the most frequent and most malignant primary brain tumor with a poor prognosis. The translation of therapeutic strategies for glioblastoma from the experimental phase into the clinic has been limited by insufficient animal models, which lack important features of human tumors. Lentiviral gene therapy is an attractive therapeutic option for human glioblastoma, which we validated in a clinically relevant animal model. Methodology/Principal Findings: We used a rodent xenograft model that recapitulates the invasive and angiogenic features of human glioblastoma to analyze the transduction pattern and therapeutic efficacy of lentiviral pseudotyped vectors. Both, lymphocytic choriomeningitis virus glycoprotein (LCMV-GP) and vesicular stomatitis virus glycoprotein (VSV-G) pseudotyped lentiviral vectors very efficiently transduced human glioblastoma cells in vitro and in vivo. In contrast, pseudotyped gammaretroviral vectors, similar to those evaluated for clinical therapy of glioblastoma, showed inefficient gene transfer in vitro and in vivo. Both pseudotyped lentiviral vectors transduced cancer stem-like cells characterized by their CD133-, nestin- and SOX2-expression, the ability to form spheroids in neural stem cell medium and to express astrocytic and neuronal differentiation markers under serum conditions. In a therapeutic approach using the suicide gene herpes simplex virus thymidine kinase (HSV-1-tk) fused to eGFP, both lentiviral vectors mediated a complete remission of solid tumors as seen on MRI resulting in a highly significant survival benefit (p<0.001) compared to control groups. In all recurrent tumors, surviving eGFP-positive tumor cells were found, advocating prodrug application for several cycles to even enhance and prolong the therapeutic effect. Conclusions/Significance: In conclusion, lentiviral pseudotyped vectors are promising candidates for gene therapy of glioma in patients. The inefficient gene delivery by gammaretroviral vectors is in line with the results obtained in clinical therapy for GBM and thus confirms the high reproducibility of the invasive glioma animal model for translational research.
Ataxia represents a pathological coordination failure that often involves functional disturbances in cerebellar circuits. Purkinje cells (PCs) characterize the only output neurons of the cerebellar cortex and critically participate in regulating motor coordination. Although different genetic mutations are known that cause ataxia, little is known about the underlying cellular mechanisms. Here we show that a mutated axJ gene locus, encoding the ubiquitin-specific protease 14 (Usp14), negatively influences synaptic receptor turnover. AxJ mouse mutants, characterized by cerebellar ataxia, display both increased GABAA receptor (GABAAR) levels at PC surface membranes accompanied by enlarged IPSCs. Accordingly, we identify physical interaction of Usp14 and the GABAAR alpha 1 subunit. Although other currently unknown changes might be involved, our data show that ubiquitin-dependent GABAAR turnover at cerebellar synapses contributes to axJ-mediated behavioural impairment.
Background: There is currently no effective AIDS vaccine, emphasizing the importance of developing alternative therapies. Recently, a patient was successfully transplanted with allogeneic, naturally resistant CCR5-negative (CCR5 delta 32) cells, setting the stage for transplantation of naturally resistant, or genetically modified stem cells as a viable therapy for AIDS. Hematopoietic stem cell (HSC) gene therapy using vectors that express various anti-HIV transgenes has also been attempted in clinical trials, but inefficient gene transfer in these studies has severely limited the potential of this approach. Here we evaluated HSC gene transfer of an anti-HIV vector in the pigtailed macaque (Macaca nemestrina) model, which closely models human transplantation. Methods and Findings: We used lentiviral vectors that inhibited both HIV-1 and simian immunodeficiency virus (SIV)/HIV-1 (SHIV) chimera virus infection, and also expressed a P140K mutant methylguanine methyltransferase (MGMT) transgene to select gene-modified cells by adding chemotherapy drugs. Following transplantation and MGMT-mediated selection we demonstrated transgene expression in over 7% of stem-cell derived lymphocytes. The high marking levels allowed us to demonstrate protection from SHIV in lymphocytes derived from gene-modified macaque long-term repopulating cells that expressed an HIV-1 fusion inhibitor. We observed a statistically significant 4-fold increase of gene-modified cells after challenge of lymphocytes from one macaque that received stem cells transduced with an anti-HIV vector (p<0.02, Student's t-test), but not in lymphocytes from a macaque that received a control vector. We also established a competitive repopulation assay in a second macaque for preclinical testing of promising anti-HIV vectors. The vectors we used were HIV-based and thus efficiently transduce human cells, and the transgenes we used target HIV-1 genes that are also in SHIV, so our findings can be rapidly translated to the clinic. Conclusions: Here we demonstrate the ability to select protected HSC-derived lymphocytes in vivo in a clinically relevant nonhuman primate model of HIV/SHIV infection. This approach can now be evaluated in human clinical trials in AIDS lymphoma patients. In this patient setting, chemotherapy would not only kill malignant cells, but would also increase the number of MGMTP140K-expressing HIV-resistant cells. This approach should allow for high levels of HIV-protected cells in AIDS patients to evaluate AIDS gene therapy.
Background: The alpha-7 nicotinic acetylcholine receptor (alpha 7-nAChR) is well known as a potent calcium ionophore that, in the brain, has been implicated in excitotoxicity and hence in the underlying mechanisms of neurodegenerative disorders such as Alzheimer's disease. Previous research implied that the activity of this receptor may be modified by exposure to a peptide fragment derived from the C-terminal region of the enzyme acetylcholinesterase. This investigation was undertaken to determine if the functional changes observed could be attributed to peptide binding interaction with the alpha 7-nAChR, or peptide modulation of receptor expression. Methodology/Principal Findings: This study provides evidence that two peptides derived from the C-terminus of acetylcholinesterase, not only selectively displace specific bungarotoxin binding at the alpha 7-nAChR, but also alter receptor binding properties for its familiar ligands, including the alternative endogenous agonist choline. Of more long-term significance, these peptides also induce upregulation of alpha 7-nAChR mRNA and protein expression, as well as enhancing receptor trafficking to the plasma membrane. Conclusions/Significance: The results reported here demonstrate a hitherto unknown relationship between the alpha 7-nAChR and the non-enzymatic functions of acetylcholinesterase, mediated independently by its C-terminal domain. Such an interaction may prove valuable as a pharmacological tool, prompting new approaches for understanding, and combating, the process of neurodegeneration.
Die primär – und sekundärprophylaktische Versorgung durch einen plötzlichen Herztod bedrohter Patienten mittels Implantierbarer Cardioverter Defibrillatoren hat sich in mehreren groß angelegten Studien einer rein konservativ-medikamentösen Behandlung gegenüber als überlegen erwiesen. Ein Hauptproblem dieser Therapieform ist jedoch die mit 25 – 40% berichtete hohe Inzidenz inadäquater Therapieabgaben. Bisher weitgehend ungeklärt blieben die prognostische Bedeutung, eventuelle Risikofaktoren sowie die daraus abzuleitenden therapeutischen Implikationen des Auftretens inadäquater Therapieabgaben. Die vorliegende Arbeit analysiert die Daten 783 (630 Männer, 153 Frauen. Medianes Alter 65 Jahre) konsekutiv im Zeitraum von 1993 bis 2005 in der Medizinischen Klinik III – Kardiologie und Angiologie – Abteilung für Elektrophysiologie der Universitätsklinik der Johann Wolfgang Goethe-Universität implantierter Patienten über einen medianen Nachbeobachtungszeitraum von 37 Monaten. Implantiert wurden nach allgemein anerkannter Indikationsstellung Cardioverter Defibrillatoren der Firmen CPI/Guidant, Ela-Medical, Medtronic Inc. und St. Jude Medical. Zum Einsatz kamen hierbei Ein – Zwei – und Dreikammergeräte (Einkammer = 511, Zweikammer = 239, Dreikammer = 33). Die Sachkontrolluntersuchungen fanden im ersten Jahr nach Implantation dreimonatig, danach halbjährlich sowie bei Notwendigkeit außerplanmäßig statt. Hierbei wurden jeweils alle stattgehabten, von den Implantierbaren Cardioverter Defibrillatoren registrierten Ereignisse ausgedruckt und auf Speichermedien gesichert. Die archivierten Elektrogramme der in diesem Zeitraum aufgetretenen 11.942 tachykarden Ereignisse bilden die Grundlage der Analyse der vorliegenden Arbeit. Hierbei wurden alle aufgetreten inadäquaten Therapiegaben an Hand der Elektrogramme einer von neun ätiologischen Kategorien zugeordnet. Der aktuelle Status aller 783 Patienten wurde im Rahmen der vorliegenden Arbeit, soweit unbekannt und somit notwendig, telefonisch oder per Briefkontakt mit den weiterbehandelnden Ärzten und Zentren eruiert. Im Rahmen der Untersuchung wurden Parameter betreffend der Erkrankungen der Patienten (Herzinsuffizienz, Koronare Herzerkrankung, Dilatative Cardiomyopathie, Klappenvitien und vorbekannte atriale Tachykardien) sowie zu ihrer zum jeweiligen Untersuchungszeitpunkt aktuellen Medikation (Klasse-I-Antiarrhythmika, Sotalol, Amiodaron, Beta-Blocker, Klasse-IV-Antiarrhythmika, Digitalisglykoside, ACE-Inhibitoren/AT1-Antagonisten, Aldosteronantagonisten, Diuretika und CSE-Inhibitoren ) erfasst. Die statistische Auswertung bezüglich des Auftretens inadäquater Therapien sowie ihrer Ätiologie, der Versterbenshäufigkeit, der Überlebensdauer, des gefährdeten Auftretenszeitraums inadäquater Therapien, eines eventuellen prädilektiven Risikoalters, des Einflusses der verschieden Ätiologien und der Anzahl inadäquater Therapiegaben auf Überlebensdauer und Versterbenshäufigkeit, des Einflusses der Begleitmedikation, des Einflusses der Begleiterkrankungen, der Differenzen zwischen Ein – Zwei – und Dreikammergeräten, der Differenzen der Geräte der verschiedenen Hersteller sowie der erfolgten Programmierung der Implantierbaren Cardioverter Defibrillatoren erfolgte mittels des Leevene-Tests, des Kolmogoroff-Smirnoff-Tests, des Zweistichproben-t-Tests sowie Fisher’s exaktem Test. Im nachbeobachteten Zeitraum wurden 11.942 tachykarde Ereignisse registriert, deren Auswertung 2806 (23,5%) inadäquate Therapien ergab. Davon waren 27,9% Defibrillationen, 72,1% antitachykarde Überstimulationen. 200 der 783 Patienten (25,5%) erlitten inadäquate Therapieabgaben während der Nachbeobachtungsperiode. 29,1% der implantierten CPI/Guidant-Geräte waren von inadäquaten Therapien betroffen, 40,8% der Ela90 Geräte, 21,2% der Medtronic-Geräte und 25% der St. Jude-Geräte. Tachykard übergeleitetes Vorhofflimmern machte 45,6% der inadäquaten Therapien aus, Sinustachykardien 11,3%, Vorhofflattern 9,1%, supraventrikuläre Tachykardien 16,1%, Sondendefekte 14,1%, Muskelpotentialfehlwahrnehmungen 0,8%, T-Wellen-Fehlwahrnehmungen 1,6%, R-Wellen-Doppelwahrnehmungen 0,4% und nicht anhaltende Ventrikuläre Tachykardien 0,9%. Die Analyse der Differenzen der Patienten, welche inadäquate Therapien erlitten und der Patienten, welche keine inadäquaten Therapien erlitten, ergab signifikante Unterschiede. Die Nachbeobachtungsperiode der Patienten mit inadäquaten Therapien war mit 54,2 Monaten im Median länger als die der Patienten ohne inadäquate Therapien (Median hier 30 Monate, p<0,000001). Gleiches gilt für die Überlebensdauer der Patienten mit inadäquaten Therapien (mediane Überlebenszeit 56,9 Monate versus 33,4 Monate, p<0,0001). Weder das generelle Auftreten inadäquater Therapien noch die Zahl der erlittenen inadäquaten Therapien reduzierten die Überlebensdauer. Auch konnte keine Form von inadäquater Therapie identifiziert werden, die mit einer Übersterblichkeit einherging. Das Auftreten inadäquater Therapien an sich jedoch ging in einer Subgruppenanalyse der Patienten mit inadäquaten Therapien, der Patienten mit nur adäquaten Therapien und der Patienten ohne jegliche Therapien mit einer erhöhten Versterbenswahrscheinlichkeit einher (p<0,000003 bzw. p<0,01). Insofern kann an dieser Stelle ein negativer Einfluss inadäquater Therapien in der Zusammenschau der Befunde nur gemutmaßt, nicht aber eindeutig nachgewiesen werden. Bezüglich der Risikofaktoren für das Auftreten inadäquater Therapien ergab sich ein gefährdeter Zeitraum innerhalb der ersten sechs Monate nach Implantation, was auf die zu diesem Zeitpunkt noch nicht individuell optimierte Programmierung zurückzuführen ist. Patienten jeden Alters wiesen ein vergleichbares Risiko auf, inadäquate Therapien zu erleiden. Insofern konnte kein Risikoalter identifiziert werden. Mit einem erhöhten Risiko für die Entwicklung inadäquater Therapien gingen eine insuffiziente Versorgung mit Betablockern oder Amiodaron (p<0,000001 bzw. p<0,04) sowie vor der Implantation bekannte atriale Tachykardien (p<0,001) einher. Beide Umstände erklären sich mit der Dominanz atrialer Rhythmusstörungen als auslösende Ursache inadäquater Therapien (82,1% aller inadäquaten Therapien wurden durch atriale Rhythmusstörungen ausgelöst) sowie durch den reduzierenden Effekt der beiden Antiarrhythmika auf die Inzidenz dieser atrialen Rhythmusstörungen. Die restlichen Begleiterkrankungen blieben ohne nachweisbaren Einfluss auf das Auftreten inadäquater Therapieabgaben. Zwei- und Dreikammergeräte konnten, entgegen der Erwartungen, die Inzidenz inadäquater Therapieabgaben gegenüber den Einkammergeräten nicht signifikant reduzieren (p<0,1). Am ehesten zu erklären ist dies mit den bei Zwei- und Dreikammergeräten vermehrt auftretenden Wahrnehmungsproblemen sowie den zusätzlich benötigten, defektanfälligen Sonden. Die vorliegende Arbeit konnte die berichtete hohe Inzidenz inadäquater Therapiegaben sowie deren dominierende Ätiologien in einem breiten, den klinischen Routinebedingungen entsprechenden Patientenkollektiv innerhalb eines langen Zeitverlaufes bestätigen und eventuelle Risikofaktoren identifizieren. Weiterhin konnten Erkenntnisse bezüglich der prognostischen Bedeutung inadäquater Therapieabgaben gewonnen werden. Die Kenntnis der Risikofaktoren könnte dazu beitragen helfen, gefährdete Patienten frühzeitig zu identifizieren sowie durch prophylaktische Maßnahmen die Inzidenz inadäquater Therapiegaben zu mindern. Weiterhin können die in dieser Arbeit gewonnen Erkenntnisse als Grundlage der Hypothesengenerierung und Planung weiterer klinischer Studien dienen.
Die Nachhaltigkeit des Waldes wird heute im Allgemeinen als die gleichrangige Berücksichtigung ökonomischer, ökologischer und sozialer Waldfunktionen beschrieben. Insbesondere das Konzept des naturnahen Waldbaus "verbindet in hohem Maße die verschiedenen ökonomischen, ökologischen und sozialen Teilziele der Nachhaltigkeit" (Kleinschmit 2007: 133). Die in der Praxis teilweise zu beobachtende übermäßige Ausrichtung an ökonomischen Gesichtspunkten und an der Sicht auf den Wald als primären Holzproduzenten wird von vielen Akteuren und Akteurinnen kritisiert; im Zuge des Klimawandels und der Klimawandelforschung rücken zwar die ökologischen Funktionen des Waldes – bspw. als Kohlenstoffsenke, Biodiversitäts-Potential, aber auch als Energieressource – mehr in den Blickpunkt. Wie steht es jedoch um den dritten Pfeiler der Nachhaltigkeit, die sozialen Funktionen des Waldes? Ulrich Schraml (Forstpolitikwissenschaftler an der Universität Freiburg) zufolge "führen die sozialen Funktionen des Waldes neben Walderhaltung, Sicherung der Biodiversität, Rohstoffversorgung und den Klimaprogrammen in den laufenden Prozessen ein Schattendasein" (Schraml 2009: 2). Der vorliegende Text wird sich diesem "blinden Fleck" der aktuellen forstwissenschaftlichen Forschung zuwenden und die Frage stellen, (1) welche sozialen Funktionen der Wald erfüllen soll und (2) wie diese gewährleistet werden können. Während erstere Fragestellung auf den gesellschaftlichen Anspruch an Wald, d.h. auf die Art und Weise der sozialen Nutzung, abzielt, erfordert letztere den Einbezug forstpolitischer Rahmenbedingungen und deren Verhältnis zur Sicherstellung dieser Nutzungsformen. Die Frage lautet also, wie sich die Veränderung der forstpolitischen Rahmenbedingungen auf die sozialen Funktionen der Wälder und auf die Möglichkeiten sozialer Nutzungen auswirken oder in Zukunft auswirken könnten. ...
Das Biodiversität und Klima Forschungszentrum (BiKF) hat sich selbst das Ziel gesetzt, mit seinen Arbeiten wissenschaftliche Beiträge zum Management klimabedingter relevanter Biodiversitätsveränderungen sowie zur Erfüllung internationaler Übereinkommen wie der EU-Fauna-Flora-Habitat-Richtlinie (EU-FFH), der EU-Wasserrahmenrichtlinie (EU-WRRL), der internationalen Biodiversitätskonvention (CBD) und der Klimarahmenkonvention (UNFCCC) zu liefern. Voraussetzung dafür ist ein Ergebnis- und Wissenstransfer zwischen den Forschenden des Zentrums und den Anspruchsgruppen aus Zivilgesellschaft, Politik und Wirtschaft (vgl. Biodiversität und Klima Forschungszentrum (BiKF) 2008: 1 und 40ff.). Um gesellschaftliche Wirkungen zu erzeugen, müssen die Interessen, Positionen und praktischen Wissensbestände der Anspruchsgruppen in die Forschungen aufgenommen werden und die Forschungsergebnisse in die gesellschaftliche Praxis (rück-) vermittelt werden. Um für das Biodiversität und Klima Forschungszentrum (BiKF) ein besseres Verständnis über die Entstehung und den Transfer von Wissen zwischen Forschung und Gesellschaft vorzubereiten und Ansatzpunkte für Interventionen in öffentliche Debatten aufzuzeigen, wurde eine problemorientierte Diskursfeldanalyse (DFA) zum übergreifenden Themengebiet Klimabedingte Biodiversitätsveränderungen durchgeführt (zur Methode siehe Infobox). Dabei wurden prioritäre Themen identifiziert sowie relevante Akteure und ihre Positionen typisiert. Diese Analyse dient somit als Orientierungshilfe für die strategische Ausrichtung des Zentrums; sie bietet eine strukturierte Grundlage für eine zentrumsinterne Diskussion des weiteren Vorgehens im Wissens- und Ergebnistransfer. Die Diskursfeldanalyse speist sich aus drei Detailanalysen zu den Diskursarenen ‚9. Vertragsstaatenkonferenz der CBD‘, ‚Nationale Biodiversitätsstrategie‘ und ‚Millennium Ecosystem Assessment‘. Diese werden nach einer kurzen Einführung zur Methode vorgestellt. In einem zweiten Schritt werden die über diese drei Arenen hinweg relevanten und für BiKF mögliche Ansatzpunkte bietenden Diskursstränge aufgezeigt. Diese illustrieren die Vielfalt an Themen und Positionen der Akteure innerhalb des Diskursfelds. Abschließend werden Schlussfolgerungen für die Struktur von Wissenskonflikten im Diskursfeld gezogen, die Orientierung geben sollen für eine weitere Auseinandersetzung des Forschungszentrums mit möglichen Querschnittsthemen und Zielen von Interventionen in den Diskurs.

Erwartungsgemäß wird Biodiversität sehr häufig thematisiert, besonders bezogen auf die Rotbuche. Bezogen auf die Klimaveränderung wird das Speicherpotential der Böden betont. Hinweise für weitere Aktivitäten im LOEWE Biodiversität und Klima Forschungszentrum BiKF wurden identifiziert.
Biodiversität und Klimawandel in der Debatte um den ökologischen Waldumbau – eine Diskursfeldanalyse

In der Umweltpolitik werden die Themenfelder Klimawandel und Biodiversität besonders betont. Klimaveränderung und Erhalt/Veränderungen der biologischen Vielfalt werden zumeist unabhängig voneinander betrachtet und nicht miteinander verknüpft. Debatten zum Klimawandel und zum Schutz der Biodiversität werden von Vertretern der Politik, Wirtschaft, Wissenschaft und Gesellschaft kontrovers geführt. Die im aktuellen Diskurs um den ökologischen Waldumbau erwähnten Zusammenhänge von Biodiversität und Klimawandel werden analysiert, da dieser Umbau darauf abzielt, Waldbestände mit naturnahen Strukturen und natürlichen Lebensabläufen zu schaffen. Aufgrund des Klimawandels sind trotz Wissenslücken Maßnahmen eines klimaanpassenden Umbaus sinnvoll. Erwartungsgemäß wird Biodiversität häufig thematisiert, etwa bezogen auf die Baumartenwahl und -mischung, aber auch die Verkürzung der Umtriebszeiten und Grenzen der Naturverjüngung. Hinweise für weitere Aktivitäten im LOEWE Biodiversität und Klima Forschungszentrum BiKF wurden identifiziert.
Das Ziel der vorliegenden Dissertationsarbeit ist die durchflusszytometrische Bestimmung Cytomegalievirus-spezifisch aktivierbarer CD4+T-Lymphozyten bei Patienten nach Lungentransplantation. Virusinfekte, insbesondere mit dem Cytomegalievirus (CMV), sind häufige Komplikationen nach Lungentransplantation. Bei Untersuchungen zu knochenmark-, leber-und nierentransplantierten Patienten konnte gezeigt werden, dass eine enge Korrelation zwischen der virusspezifischen, zellvermittelten T-Zellreaktion und der viralen Replikation besteht [Sesters, M. et. al., 2001 Transplantation 71, 1287-1294]. Zur Klärung der Frage, inwieweit ein Zusammenhang zwischen dem Auftreten einer CMVInfektion und dem Anteil CD4+ T-Zellen mit de-novo-Synthese von Interferon-γ (INF-γ) und CD69 bei Patienten nach einer Lungentransplantation existiert, wurde bei n=22 Patienten nach Lungentransplantation während 12 Monate der Anteil [%] virusspezifisch aktivierbarer CD4+ Lymphozyten mit de-novo-Synthese von INF-γ und CD69 gemessen. Dazu wurde peripheres venöses Blut mit CMV-KBR-Antigen inkubiert und der Anteil virusspezifisch stimulierbarer CD4+ Lymphozyten durchflusszytrometrisch quantifiziert. Bei n=7 CMV-positiven Patienten ohne akuten CMV-Infekt während des Beobachtungszeitraums (kein Nachweis von CMV-DNA und CMV-pp65) reagierten 6,13 % (median; Spannweite: 0,86-45,15%) der CD4+ T-Lymphozyten auf die Simulation mit dem Virusantigen mit einer de-novo-Synthese von INF-γ und CD69. Im Vergleich dazu zeigte sich bei n=5 CMV-positiven Patienten mit akuter CMV-Infektion im Beobachtungszeitraum ein deutlich verringerter Anteil CD4+ T-Zellen mit de-novo-Synthese von INF-γ und CD69 (0,99%; 0,01-3,20%). CMV-negative Patienten (n=10) wiesen im Vergleich zu den CMV-positiven Patienten mit und ohne CMV-Infekt im Untersuchungszeitraum einen geringeren Anteil CD4+ T-Zellen mit de-novo-Synthese von INF-γ und CD 69 (0,04%; 0,02-0,41%) auf. Bei der durchflusszytometrischen Bestimmung spezifisch aktivierbarer CD4+ T-Lymphozyten im peripheren Blut lungentransplantierter Patienten konnte ein Zusammenhang zwischen dem Anteil der virusspezifisch aktivierbaren T-Zellen und der viralen Replikation gezeigt werden. Die Methode ermöglicht eine Eingrenzung von Risikopatienten im Hinblick auf eine CMVReaktivation und kann das bestehende CMV-Monitoring sinnvoll ergänzen. Die gewonnenen Ergebnisse können Ausgangspunkt für weitere Studien zu virusspezifisch aktivierbaren T-Lymphozyten bei lungentransplantierten Patienten sein.
In mitochondrial respiration, the soluble protein cytochrome c accepts an electron from the membrane bound cytochrome bc1. The interaction between cytochrome bc1 and cytochrome c is highly transient in nature, enabling turnover numbers greater than 160 s-1. Yeast cytochrome bc1 has been successfully crystallised with bound cytochrome c with the help of an antibody fragment (Lange and Hunte 2002; Solmaz and Hunte 2008). In all crystal structures of the complex, the homodimeric cytochrome bc1 binds only one cytochrome c, with the binding site located on subunit cytochrome c1. Univalent cytochrome c binding is correlated with conformational changes of the Rieske protein head domain and subunit QCR6p. The interface of the complex is small. The haem moieties are centrally located in a mainly non-polar contact site that includes a cation–! interaction and is surrounded by complementary charged residues. The crystal structure is in agreement with the general architecture of the interfaces of transient redox complexes and also reveals several interesting features unique to the cytochrome bc1. On the basis of the crystal structures, an extensive thermodynamic and kinetic characterisation of the interaction was carried out in this work to challenge the static snapshot of the bound proteins in the crystal structure as the relevant physiological electron transfer. The thermodynamic parameters of the interaction between the redox partners were determined using isothermal titration calorimetry (ITC). The association constant for cytochrome bc1 and cytochrome c in oxidised state under physiological ionic strength of 120 mM at 25 °C, was determined to be 5 " 103 M-1 by direct ITC titration. So, the partners interact with an affinity of 200 #M. In spite of the low affinity the complex has a life time ($ = 1/koff) of 5 #second, sufficiently long to enable the theoretically calculated electron transfer rates of 1.0 " 106 to 2.6 " 107 s%1 with a lifetime ($ = 1/rate) of 1-0.04 μseconds and experimentally determined rate of 7.7 " 104 s%1 with a lifetime of 13 μseconds. The low affinity makes it difficult to ascertain the stoichiometry of binding. The enthalpy of the interaction is endothermic, which is consistent with the nature of an interface where hydrophobic interactions are dominant. The enthalpy and entropy is 3.6 kJmol-1 and 83 kJmol-1K-1, respectively. The importance of key interface residues was also investigated. The role of the interface residue G89 of cytochrome c which might have a role in the dissociation of the complex has been probed by site-directed mutagenesis. The interface contains a cation-! interaction between F230 of cytochrome bc1 and R19 of cytochrome c, which is thought to provide the specificity to the interaction between the otherwise promiscuous partners. To analyse the role of this interaction pair in electron transfer, F230L and F230W mutants were used to measure direct electron transfer rates by flash photolysis and steady state kinetics. The findings indicate that another ! system can work as functional substitution of F230, while deleting the ! system has a deleterious effect on the complex formation. The inability of F230L to achieve the transient and steady state turnover rates as wild type protein indicates a scenario where the variant achieves an altered bound state with inefficient electron transfer pathways and higher edge-to-edge distance. The role of supernumerary subunit QCR6p in complex formation was investigated by steady state kinetics measurements. Subunit QCR6p does not interact directly with cytochrome c but is positioned in such a way that it could electrostatically steer cytochrome c in a reactive ensemble. The highly acidic and disordered N-terminus of QCR6p could interact with a patch of conserved lysine residues on cytochrome c. The role of subunit QCR6p has been assessed using QCR6p deleted cytochrome bc1 and a lysine variant of cytochrome c. The results show that QCR6p not only affects the kinetics of the interaction but is also important for the stability of cytochrome bc1. The kinetic and thermodynamic data obtained during this study provide evidence for the functional importance of non-catalytic cytochrome bc1 subunit QCR6p, show that the entropy driven interaction is indeed of low affinity and highly transient in nature and indicate that the interface is well suited to ensure the high turnover of the electron transfer chain where cytochrome c interacts with multiple partners using overlapping interfaces. The suggested role of the cation-! interaction as a highly specific interaction has been validated.
The molecular conformation of the title compound, C18H18N2O3S, is stabilized by an intramolecular N—H ... O hydrogen bond. The crystal packing shows centrosymmetric dimers connected by N—H ... S hydrogen bonds. The terminal ethoxy substituents are statistically disordered [occupancy ratio 0.527 (5):0.473 (5)].
The title compound, C20H22O4S2, was synthesized by the reaction of 1,4-dibromobutene with methyl thiosalicylate. The aliphatic segment of this ligand is in an all-trans conformation. The bridging chain, –S-(CH2)4-S–, is almost planar (r.m.s. deviation for all non-H atoms: 0.056 Å) and its mean plane forms dihedral angles of 16.60 (7) and 5.80 (2)° with the aromatic rings. In the crystal, the molecules are linked by weak C—H ... O interactions into chains with graph-set notation C(14) along [0 0 1]. The crystal studied was a racemic twin, the ratio of the twin components being 0.27 (9):0.73 (9).
The title compound, C14H11NO4, crystallizes with two molecules in the asymmetric unit. The major conformational difference between these two molecules is the dihedral angle between the aromatic rings, namely 36.99 (5) and 55.04 (5)°. The nitro groups are coplanar with the phenyl rings to which they are attached, the O—N—C—C torsion angles being -1.9 (3) and 1.0 (3)° in the two molecules.
The 3,5-methoxy groups in the title compound, C16H23NO4, are almost coplanar with the aromatic ring, whereas the 4-methoxy group is bent out of this plane. The three CH3—O—C—C torsion angles are -1.51 (18), 0.73 (19) and 75.33 (15)°. The cyclohexane ring adopts a chair conformation. In the crystal, molecules are connected by intermolecular N—H ... O hydrogen bonds into chains running along the b axis.
The title compound, C6H5NO2·C6H6O2, crystallizes with one pyridinium-2-carboxylate zwitterion and one molecule of benzene-1,2-diol in the asymmetric unit. The crystal structure is characterized by alternating molecules forming zigzag chains running along the a axis: the molecules are connected by O—H ... O and N—H ... (O,O) hydrogen bonds.
Crystals of the title compound, C12H8N2·C7H8O2, were obtained during cocrystallization experiments of a compound with two hydrogen-bond donors (2-hydroxybenzyl alcohol) with another compound containing two hydrogen-bond acceptors (phenanthroline). Unexpectedly, the two molecules do not form dimers with two O—H ... N hydrogen bonds connecting the two molecules. However, one of the hydroxy groups forms a bifurcated hydrogen bond to both phenanthroline N atoms, whereas the other hydroxy group forms an O—H ... O hydrogen bond to a symmetry-equivalent 2-hydroxybenzyl alcohol molecule. In addition, the crystal packing is stabilized by Pi – Pi interactions between the two phenanthroline ring systems, with a centroid–centroid distance of 3.570 Å.
Der hereditäre Fibrinogen-Mangel ist ein seltener Hämostasedefekt, der in einer quantitativen (Hypofibrinogenämie und Afibrinogenämie) oder qualitativen Form (Dysfibrinogenämie) bzw. einer Kombination aus beiden Formen (Hypodysfibrinogenämie) vorliegt. Seltene Ausprägungen wie die Fibrinogen- Amyloidose und die hepatische Speicherkrankheit Endoplasmic Reticulum Storage Disease (ERSD) tragen zum heterogenen klinischen Bild der Erkrankungen bei, das neben einem asymptomatischen Verlauf Blutungen und Thrombosen umfassen kann. Ziel der vorliegenden Arbeit war es, das Spektrum genetischer Veränderungen, deren Auswirkungen auf die klinische Manifestation und gerinnungsphysiologische Ausprägung bei 101 Probanden mit angeborenem Fibrinogen-Mangel sowie bei 41 erstgradig verwandten Familienangehörigen zu untersuchen. Die vorliegende Arbeit stellt das bisher größte zusammenhängend untersuchte und genetisch charakterisierte Patientenkollektiv mit Fibrinogen-Mangel dar. Von den insgesamt 96 nachgewiesenen Sequenz-Varianten wurden 30 erstmals beschrieben. Hierdurch ergibt sich ein Zuwachs von 15 % an der Gesamtzahl der verschiedenen bisher in der Literatur beschriebenen genetischen Defekten. Bei acht nachgewiesenen Mutationen war bisher erst eine Familie mit dieser Sequenz-Variante beschrieben. Die Ergebnisse der vorliegenden Arbeit bestätigen, dass diese Mutationen kausal für den Phänotyp des Fibrinogen-Mangels sind. Die klinische Präsentation der Patienten umfasste, häufiger als in der Literatur bisher beschrieben, zu 50 % leichte bis moderate Blutungen in Form von Menorrhagien oder verlängerten Nachblutungen nach Verletzungen. Schwere Blutungen traten bei Patienten mit Afibrinogenämie oder selten bei heterozygoten Mutationsträgern in Assoziation mit Operationen, Schwangerschaften und Traumata auf. Probanden mit ERSD erlitten gegensätzlich zu den bisherigen Beschreibungen aus der Literatur sowohl Blutungen als auch Thrombosen. Die Vorhersage des klinischen Phänotyps nach traditioneller Einteilung des Fibrinogen-Mangels anhand laborchemischer Beschreibungen war nicht zuverlässig. Zudem lagen die hierfür erforderlichen Laborparameter häufig nicht vor. Die Mutationsdiagnostik konnte zu einer verbesserten Einschätzung des klinischen Phänotyps beitragen. Spezifische Mutationen wiesen eine Korrelation zu bestimmten klinischen Manifestationen auf. Hierbei zeigte sich eine ausgeprägte Assoziation der Varianten BbArg14Cys und gAsn319-Asp320del mit thromboembolischen Ereignissen. Für weitere Mutationen (AaArg19Gly, BbArg44Cys, gTyr262Cys, gAla327Thr und gLys380Asn) ist ebenfalls eine Assoziation mit Thromboseneigung anzunehmen, konnte jedoch aufgrund kleiner Fallzahlen nicht zweifelsfrei beurteilt werden. Die am häufigsten auftretenden Mutationen an Position AaArg16 waren mit einer moderaten Blutungs- (17 % bis 53 %) und einer nur geringen Thromboserate (3 % bis 13 %) assoziiert, die jedoch für die AaArg16Cys-Variante höher lag im Vergleich zur AaArg16His-Variante. Die Mutationen gGly333Ser und gArg375Trp zeigten eine Korrelation zur hepatischen Speicherkrankheit ERSD und gingen mit einer beeinträchtigten Leberfunktion einher. Die Verdachtsdiagnose des Fibrinogen-Mangels wurde bei einem Großteil der Indexpatienten (42 %) aufgrund pathologischer Gerinnungswerte gestellt. Vor allem „Null“-Mutationen gingen jedoch oftmals mit im Referenzbereich liegenden Fibrinogen-Werten einher, so dass sich neben der mehrmaligen Fibrinogen-Bestimmung die zusätzliche Testung weiterer Laborparameter, wie die Thromboplastinzeit (TPZ), die aktivierte partielle Thromboplastinzeit (aPTT) und die Thrombinzeit (TZ) als hilfreich erwiesen haben. Die Bestimmung des abgeleiteten Fibrinogens (Fib. QD) zeigte sich als nicht geeignet zur Detektion des kongenitalen Fibrinogen-Mangels, da die am häufigsten vorliegenden Mutationen an AaArg16 zu keiner Erniedrigung führten. Die Messung des funktionellen Fibrinogen nach Clauss (Fib. n. Cl.), der Fibrinogen- Konzentration (Fib. Ag) und die daraus resultierende Fibrinogen-Ratio (RaAg) bzw. die Bestimmung von RZ und TZ waren die Grundlage eines in der vorliegenden Arbeit entwickelten Stufenmodells für einer schnelle und kostengünstige Mutations-Detektion. Bei Anwendung der vorgeschlagenen Stufendiagnostik im Vergleich zur herkömmlichen Komplettsequenzierung ist eine Reduktion von Arbeitsaufwand und Kosten um bis zu 64 % zu erwarten.
