The Alor-Pantar Archipelago is still one of the most under-researched areas of Indonesia. However, in January 2009 I was able to collect data on anthropomorphic sculptures in the Tanjung Muna area of Pantar. The study used photographs of objects that had been purchased recently by European private art collectors in order to elicit responses regarding the local mythology. I was also able to interview local traders as well as the dealer who sold the sculptures, which allowed me to document information regarding their origin and former use, as well as the circumstances under which they had found their way on to the international art market. Traditional marriage rules as well as black magic played a central role. Furthermore, in the course of my research by chance I learned of a contemporary ancestor cult connected to the construction of a new clan house and of traditional (pre-Protestant) forms of healing.
The ubiquinol:cytochrome c oxidoreductase is a key component of several aerobic respiratory chains in different organisms. It is an integral membrane protein complex, made up of three catalytic subunits (cytochrome b, cytochrome c1 and Rieske iron sulphur protein) and up to eight additional subunits in mitochondria. The complex oxidizes one quinol molecules and reduces two cytochrome c during the Q cycle, originally described by Peter Mitchell. Electrons are split between the low and the high potential chain and protons are released on the positive side of the membrane, increasing the protonmotive force needed by the ATP-synthase for energy transduction. The cytochrome bc1 complex from P. denitrificans is a perfect model for structural and functional studies. Bacteria are easy to grow and the genetic material is readily accessible for genetic manipulation. Moreover, the P. denitrificans aerobic respiratory chain is very close to the mitochondrial one: the complexes involved in electron transfer resemble the ones found in mitochondria, but lack most of the additional subunits. As a unique feature, P. denitrificans has a strongly acidic domain at the N-terminal region of the cytochrome c1, a sequence of 150 aminoacids which does not correlate with any known protein. An analogous composition can be found in the eukaryotic cytochrome bc1 complex as a part of an accessory subunit, proposed to be involved in facilitating electron transfer between the complex and the electron acceptor cytochrome c. In order to study the function of this domain in the P. denitrificans cytochrome bc1 complex, a deletion mutant has been previously cloned and modified with an affinity tag as a C-terminal extension of cytochrome b. The complex is purified by affinity chromatography and characterized by steady-state kinetics using not only horse heart cytochrome c but also the endogenous electron acceptor, the membrane bound cytochrome c552, employed here as a soluble fragment. Steady–state kinetics indicate that the deletion of the long acidic domain had effects neither on the turnover rate nor on the apparent affinity for the substrate. To understand wether the deletion affects the reaction between the cytochrome bc1 complex and the substrate, laser flash photolysis experiments are performed, showing that the interaction observed was not changed in the complex missing the acidic domain. The results presented in this work confirm the ones previously obtained by Julia Janzon using soluble fragments of the same interaction partners. The deletion, however, affected the oligomerization state of the complex, as shown by LILBID (Laser Induced Liquid Bead Ion Desorption) analysis. The wild type complex has a tetrameric structure, better described as a “dimer of dimers”. The deletion of the acidic domain on the cytochrome c1 results in the separation of the two dimers, yielding the canonical dimer. Therefore, the complex deleted in the acidic domain is used for cloning and expression of a heterodimeric complex, containing an inactivating mutation in the quinol oxidation site in only one monomer, thus allowing a selective switch-off for half the complex. Such a complex is needed for the verification of an internal regulation mechanism, the half-of-the-sites reactivity. According to it, the dimeric structure of the cytochrome bc1 complex has functional implications, since the two monomers can communicate and work in a coordinated manner. This approach confirms that substrate oxidation does effectively take place only in one of the two monomers constituting the dimer, and that the binding of substrate at the Qo and Qi site regulates the switch between active and inactive monomer. Moreover, this mechanism works also as an effective protection against the reaction of quinone intermediates with oxygen and the formation of reactive oxygen species (ROS), responsable for cellular aging. The motion of the ISP head domain is also addressed in this work; in particular the mechanism which regulates the movements towards the cytochrome c1 and the electron bifurcation at the quinol oxidation site. Laser flash kinetics in presence of several inhibitors and the substrate allow studying the response of the ISP to the binding of different species at the quinol oxidation site. The binding of ligand at the Qo site in the complex triggers the conformational switch in the ISP head domain, supporting the mechanism proposed in the literature according to which the Qo site is able to “sense” the presence of substrate and transfer the information to the ISP, regulating its mobility. The internal electron pathway between the ISP and the cytochrome c1 has been analyzed also by stopped-flow kinetics, in presence and absence of inhibitors. The results indicate that two kinetic phases describe the reduction of cytochrome c1 by the ISP, and a model for the simulation of the data is proposed.
Dynamics of chaotic strings

The main topic of this thesis is the investigation of dynamical properties of coupled Tchebycheff map networks. At every node of the network the dynamics is given by the iteration of a Tchebycheff map, which shows strongest possible chaotic behaviour. By applying a coupling between the various individual dynamics along the links of the network, a rich structure of complex dynamical patterns emerges. Accordingly, coupled chaotic map networks provide prototypical models for studying the interplay between local dynamics, network structure, and the emergent global dynamics. An exciting application of coupled Tchebycheff map lattices in quantum field theory has been proposed Beck in Spatio-temporal chaos and vacuum fluctuations of quantized fields' (2002). In this so-called chaotic string model, the coupled map lattice dynamics generates the noise needed for the Parisi-Wu approach of stochastic quantization. The remarkable obversation is that the respective dynamics seems to reproduce distinguished numerical values of coupling constants that coincide with those observed in the standard model of particle physic. The results of this thesis give insights into the chaotic string model and its network generalization from a dynamical point of view. This leads to a deeper understanding of the dynamics, which is essential for a critical discussion of possible physical embeddings. Apart from this specific application to particle physics, the investigated concepts like synchronization or a most random behaviour of the dynamics are of general interest for dynamical system theory and the science of complex networks. As a first approach, discrete symmetry transformations of the model are studied. These transformations are formulated in a general way in order to be also applicable to similar dynamics on bipartite network structures. An observable of main interest in the chaotic string model is the interaction energy. In Spatio-temporal chaos and vacuum fluctuations of quantized fields' (2002) it has been observed that certain chaotic string couplings, corresponding to a vanishing interaction energy, coincide with coupling constants of the standard model of elementary particle physics. Since the interaction energy is basically a spatial correlation measure, an interpretation of the respective dynamical states in terms of a most random behaviour is tempting. In order to distinguish certain states as most random', or evoke another dynamical principle, a deeper understanding of the dynamics essential. In the present thesis the dynamics is studied numerically via Lyapunov measures, spatial correlations, and ergodic properties. It is shown that the zeros of the interaction energy are distinguished only with respect to this specific observable, but not by a more general dynamical principle. The original chaotic string model is defined on a one-dimensional lattice (ring-network) as the underlying network topology. This thesis studies a modification of the model based on the introduction of tunable disorder. The effects of inhomogeneous coupling weights as well as small-world perturbations of the ring-network structure on the interaction energy are discussed. Synchronization properties of the chaotic string model and its network generalization are studied in later chapters of this thesis. The analysis is based on the master stability formalism, which relates the stability of the synchronized state to the spectral properties of the network. Apart from complete synchronization, where the dynamics at all nodes of the network coincide, also two-cluster synchronization on bipartite networks is studied. For both types of synchronization it is shown that depending on the type of coupling the synchronized dynamics can display chaotic as well as periodic or quasi-periodic behaviour. The semi-analytical calculations reveal that the respective synchronized states are often stable for a wide range of coupling values even for the ring-network, although the respective basins of attraction may inhabit only a small fraction of the phase space. To provide analytical results in closed form, for complete synchronization the stability of all fixed points and period-2 orbits of all chaotic string networks are determined analytically. The master stability formalism allows to treat the ring-network of the chaotic string model as a special case, but the results are valid for coupled Tchebycheff maps on arbitrary networks. For two-cluster synchronization on bipartite networks, selected fixed points and period-2 orbits are analyzed.

Die Optimierung von Fuzzy-Zielfunktionen in Fuzzy-(Mehrziel-)LP-Systemen - Ein kritischer Überblick

Klassische Programmierungsmodelle benötigen eindeutig bestimmte Koeffizienten und exakt festgelegte Restriktionsgrenzen. Um eine Fehlmodellierung zu vermeiden, ist daher in der Regel eine umfangreiche Informationsaufnahme und -verarbeitung notwendig. Oft wird man dennoch bei Realproblemen einige der Modellparameter nur größenordnungsmäßig angeben können. Während in den klassischen Modellen nur der Weg bleibt, diese ungenauen Größen durch "Mittelwerte" zu ersetzen, bieten Fuzzy-Modelle die Möglichkeit, die subjektiven Vorstellungen eines Entscheiders so präzise zu modellieren, wie dieser es ausdrücken will und kann. Das Risiko, mit einem falschen Bild der Realität zu arbeiten und Lösungen auszuwählen, die nicht dem Realproblem entsprechen, wird somit deutlich reduziert. Beschränken wir die Betrachtung auf den am häufigsten benutzten Modelltyp, die Linearen Programmierungsmodelle, so lässt sich eine Fuzzy-Erweiterung allgemein durch das nachfolgende Fuzzy Lineare Programmierungs-Modelle (FLP-Modell) ausdrücken. ...
Structured RNA regions are important gene control elements in prokaryotes and eukaryotes. Here, we show that the mRNA of a cyanobacterial heat shock gene contains a built-in thermosensor critical for photosynthetic activity under stress conditions. The exceptionally short 5´-untranslated region is comprised of a single hairpin with an internal asymmetric loop. It inhibits translation of the Synechocystis hsp17 transcript at normal growth conditions, permits translation initiation under stress conditions and shuts down Hsp17 production in the recovery phase. Point mutations that stabilized or destabilized the RNA structure deregulated reporter gene expression in vivo and ribosome binding in vitro. Introduction of such point mutations into the Synechocystis genome produced severe phenotypic defects. Reversible formation of the open and closed structure was beneficial for viability, integrity of the photosystem and oxygen evolution. Continuous production of Hsp17 was detrimental when the stress declined indicating that shutting-off heat shock protein production is an important, previously unrecognized function of RNA thermometers. We discovered a simple biosensor that strictly adjusts the cellular level of a molecular chaperone to the physiological need.
The p53-family member p73 plays a role in various cellular signaling pathways during development and growth control and it can have tumor suppressor properties. Several isoforms of p73 exist with considerable differences in their function. Whereas the functions of the N-terminal isoforms (TA and delta Np73) and their opposing pro- and antiapoptotic roles have become evident, the functional differences of the distinct C-terminal splice forms of TAp73 have remained unclear. Here, we characterized the global genomic binding sites for TAp73alpha and TAp73beta by chromatin immunoprecipitation sequencing as well as the transcriptional responses by performing RNA sequencing. We identified a specific p73 consensus binding motif and found a strong enrichment of AP1 motifs in close proximity to binding sites for TAp73alpha. These AP1 motif-containing target genes are selectively upregulated by TAp73alpha, while their mRNA expression is repressed upon TAp73beta induction. We show that their expression is dependent on endogenous c-Jun and that recruitment of c-Jun to the respective AP1 sites was impaired upon TAp73beta expression, in part due to downregulation of c-Jun. Several of these AP1-site containing TAp73alpha-induced genes impinge on apoptosis induction, suggesting an underlying molecular mechanism for the observed functional differences between TAp73alpha and TAp73beta.
The mfl-riboswitch regulates expression of ribonucleotide reductase subunit in Mesoplasma florum by binding to 2´-deoxyguanosine and thereby promoting transcription termination. We characterized the structure of the ligand-bound aptamer domain by NMR spectroscopy and compared the mfl-aptamer to the aptamer domain of the closely related purine-sensing riboswitches. We show that the mfl-aptamer accommodates the extra 2´-deoxyribose unit of the ligand by forming a more relaxed binding pocket than these found in the purine-sensing riboswitches. Tertiary structures of the xpt-aptamer bound to guanine and of the mfl-aptamer bound to 2´-deoxyguanosine exhibit very similar features, although the sequence of the mfl-aptamer contains several alterations compared to the purine-aptamer consensus sequence. These alterations include the truncation of a hairpin loop which is crucial for complex formation in all purine-sensing riboswitches characterized to date. We further defined structural features and ligand binding requirements of the free mfl-aptamer and found that the presence of Mg2+ is not essential for complex formation, but facilitates ligand binding by promoting pre-organization of key structural motifs in the free aptamer.
RNA thermometers are translational control elements that regulate the expression of bacterial heat shock and virulence genes. They fold into complex secondary structures that block translation at low temperatures. A temperature increase releases the ribosome binding site and thus permits translation initiation. In fourU-type RNA thermometers, the AGGA sequence of the SD region is paired with four consecutive uridines. We investigated the melting points of the wild-type and mutant sequences. It was decreased by 5°C when a stabilizing GC basepair was exchanged by an AU pair or increased by 11°C when an internal AG mismatch was converted to a GC pair, respectively. Stabilized or destabilized RNA structures are directly correlated with decreased or increased in vivo gene expression, respectively. Mg2+ also affected the melting point of the fourU thermometer. Variations of the Mg2+ concentration in the physiological range between 1 and 2 mM translated into a 2.8°C shift of the melting point. Thus, Mg2+ binding to the hairpin RNA is regulatory relevant. Applying three different NMR techniques, two Mg2+ binding sites were found in the hairpin structure. One of these binding sites could be identified as outer sphere binding site that is located within the fourU motif. Binding of the two Mg2+ ions exhibits a positive cooperativity with a Hill coefficient of 1.47. Free energy values delta G for Mg2+ binding determined by NMR are in agreement with data determined from CD measurements. Physiological Mg2+ concentrations reduce enthalpy and entropy values of uncorrelated base pair opening processes for almost all nucleobases.
The plastids of cryptophytes, haptophytes, and heterokontophytes (stramenopiles) (together once known as chromists) are surrounded by four membranes, reflecting the origin of these plastids through secondary endosymbiosis. They share this trait with apicomplexans, which are alveolates, the plastids of which have been suggested to stem from the same secondary symbiotic event and therefore form a phylogenetic clade, the chromalveolates. The chromists are quantitatively the most important eukaryotic contributors to primary production in marine ecosystems. The mechanisms of protein import across their four plastid membranes are still poorly understood. Components of an endoplasmic reticulum-associated degradation (ERAD) machinery in cryptophytes, partially encoded by the reduced genome of the secondary symbiont (the nucleomorph), are implicated in protein transport across the second outermost plastid membrane. Here, we show that the haptophyte Emiliania huxleyi, like cryptophytes, stramenopiles, and apicomplexans, possesses a nuclear-encoded symbiont-specific ERAD machinery (SELMA, symbiont-specific ERAD-like machinery) in addition to the host ERAD system, with targeting signals that are able to direct green fluorescent protein or yellow fluorescent protein to the predicted cellular localization in transformed cells of the stramenopile Phaeodactylum tricornutum. Phylogenies of the duplicated ERAD factors reveal that all SELMA components trace back to a red algal origin. In contrast, the host copies of cryptophytes and haptophytes associate with the green lineage to the exclusion of stramenopiles and alveolates. Although all chromalveolates with four membrane-bound plastids possess the SELMA system, this has apparently not arisen in a single endosymbiotic event. Thus, our data do not support the chromalveolate hypothesis. Extending the carotenoid pathway to astaxanthin in plants is of scientific and industrial interest. However, expression of a microbial beta-carotene ketolase (BKT) that catalyses the formation of ketocarotenoids in transgenic plants typically results in low levels of astaxanthin. The low efficiency of BKTs in ketolating zeaxanthin to astaxanthin is proposed to be the major limitation for astaxanthin accumulation in engineered plants. To verify this hypothesis, several algal BKTs were functionally characterized using an Escherichia coli system and three BKTs were identified, with high (up to 85%), moderate (~38%), and low (~1%) conversion rate from zeaxanthin to astaxanthin from Chlamydomonas reinhardtii (CrBKT), Chlorella zofingiensis (CzBKT), and Haematococcus pluvialis (HpBKT3), respectively. Transgenic Arabidopsis thaliana expressing the CrBKT developed orange leaves which accumulated astaxanthin up to 2 mg g -1 dry weight with a 1.8-fold increase in total carotenoids. In contrast, the expression of CzBKT resulted in much lower astaxanthin content (0.24 mg g -1 dry weight), whereas HpBKT3 was unable to mediate synthesis of astaxanthin in A. thaliana. The none-native astaxanthin was found mostly in a free form integrated into the light-harvesting complexes of photosystem II in young leaves but in esterified forms in senescent leaves. The alteration of carotenoids did not affect chlorophyll content, plant growth, or development significantly. The astaxanthin-producing plants were more tolerant to high light as shown by reduced lipid peroxidation. This study advances a decisive step towards the utilization of plants for the production of high-value astaxanthin. Background: Multiple sclerosis (MS) is a chronic, inflammatory demyelinating disease of the central nervous system, believed to be triggered by an autoimmune reaction to myelin. Recently, a fundamentally different pathomechanism termed ‘chronic cerebrospinal venous insufficiency’ (CCSVI) was proposed, provoking significant attention in the media and scientific community.
Methods: Twenty MS patients (mean age 42.2±13.3 years; median Extended Disability Status Scale 3.0, range 0–6.5) were compared with 20 healthy controls. Extra- and intracranial venous flow direction was assessed by colour-coded duplex sonography, and extracranial venous cross-sectional area (VCSA) of the internal jugular and vertebral veins (IJV/VV) was measured in B-mode to assess the five previously proposed CCSVI criteria. IJV-VCSA≤0.3 cm2 indicated ‘stenosis,’ and IJV-VCSA decrease from supine to upright position ‘reverted postural control.’ The sonographer, data analyser and statistician were blinded to the patient/control status of the participants.
Results: No participant showed retrograde flow of cervical or intracranial veins. IJV-VCSA≤0.3 cm2 was found in 13 MS patients versus 16 controls (p=0.48). A decrease in IJV-VCSA from supine to upright position was observed in all participants, but this denotes a physiological finding. No MS patient and one control had undetectable IJV flow despite deep inspiration (p=0.49). Only one healthy control and no MS patients fulfilled at least two criteria for CCSVI.
Conclusions: This triple-blinded extra- and transcranial duplex sonographic assessment of cervical and cerebral veins does not provide supportive evidence for the presence of CCSVI in MS patients. The findings cast serious doubt on the concept of CCSVI in MS.
Pulsed electron-electron double resonance (PELDOR) is a pulsed EPR method that can reliably and precisely provide structural information regarding duplex RNAs and DNAs by measuring long-range distances (1.5-7 nm) utilizing distance-dependent magnetic dipole-dipole interaction between two nitroxide spin labels. In this thesis the application field of PELDOR spectroscopy has been expanded. For the first time the global architecture of tertiary folded RNA has been mapped in vitro. Moreover, the first application of PELDOR for determining structural aspects of RNA and DNA molecules inside cells has been presented. RNA has the central role in cellular processes and gene regulation. It can adopt complex three dimensional structures, which in combination with its conformational dynamics is essential for its function as biological catalyst, structural scaffold and regulator of gene expression. Riboswitches are cis-acting RNA segments that modulate gene expression by direct binding of small molecules with high affinity and specificity. Neomycin-responsive riboswitch is an engineered riboswitch developed by combination of in vitro selection and in vivo screening. Upon insertion into the 5‟ untranslated region of mRNA and binding the cognate ligand it is able to inhibit translational initiation in yeast. Using enzymatic probing the secondary structure had been postulated comprising global stem-loop architecture with a terminal and an internal loop. In the first part of this thesis, the global conformational arrangement of this 27 nucleotides long RNA element has been studied by means of site-directed spin labeling and PELDOR spectroscopy. Spin-labeled neomycin-responsive riboswitch mutants were synthesized via a Sonogashira cross-coupling reaction between 5-membered pyrroline ring based nitroxide radical (TPA) and 5-iodo-uridine. The labeling positions were chosen outside of the binding pocket and UV melting curves revealed that spin-labeling neither disturbs the secondary structure nor interferes with ligand binding. Efficient ligand binding was proven by thermal stabilization of 20.3±3.3 oC upon addition of neomycin, as well as by cw EPR spectra. PELDOR time traces with long observation time windows and with good signal to noise ratio and modulation depth were recorded for all double-labeled samples allowing a reliable data analysis. The fact that there were no shifts in the measured distances upon addition of neomycin implied the existence of a prearranged tertiary structure of the neomycin-sensing riboswitch without a significant global conformational change induced by ligand binding. Measured distances were in very good agreement with the NMR structure of the ligand-bound state of the riboswitch indicating the intrinsic propensity of the global RNA architecture toward its energetically favored ligand-bound form at low temperature. The results harvested in this work represent the first application of PELDOR for mapping the global structure of a tertiary folded RNA. In the second part of this thesis the possibility of applying PELDOR on nucleic acids (NAs) in cellular environment has been investigated. It was shown before that global NA structure depends on matrix conditions, such as concentration of ions and small molecules, molecular crowding, viscosity and interactions with proteins. Therefore, PELDOR spectroscopy on a double-labeled 12-base pair DNA duplex, the 14-mer cUUCGg tetraloop hairpin RNA and the 27-mer neomycin-sensing riboswitch has been used to obtain long-range distance constraints on such systems in Xenopus laevis oocytes and to compare them with in vitro measurements. The reduced lifetime of nitroxide spin labels under cellular conditions has been a major challenge in these measurements. Investigation of nitroxide reduction kinetics in-cell has revealed that the 5-membered pyrrolidine and pyrroline rings are significantly slower reduced compared to 6-membered piperidine ring based nitroxides. Due to prolonged lifetime of the TPA nitroxides covalently attached to NA molecules PELDOR signals could be measured with good signal-to-noise ratios up to 70 minutes of incubation time. The partial loss of coupled spin labels due to nitroxide reduction only led to a decrease in the modulation depth upon increasing the incubation time. No alterations in the measured distances between in vitro and in-cell experiments implies the existence of stable overall conformations of the 14-mer cUUCGg tetraloop hairpin RNA and the 27-mer neomycin-sensing riboswitch, whereas the 12-bp duplex DNA experiences stacking in-cell but retaining the secondary structure. Thus, for the first time nanometer distance measurements were performed inside cells, clearly laying a foundation for the application of PELDOR spectroscopy to study biological processes in cells, such as diffusion, interaction with proteins and other factors or chemical reactions.

In Deutschland und in fast allen Industrieländern finden sich mittlerweile Medikamentenwirkstoffe in nahezu allen Gewässern und vereinzelt auch im Trinkwasser. Auch wenn die Konzentrationen in der Regel sehr gering sind, lassen sich erste Anzeichen für Auswirkungen auf Wasserlebewesen nachweisen. Akute Folgen für die menschliche Gesundheit sind bisher nicht erwiesen. Es kann allerdings nicht ausgeschlossen werden, dass sich Langzeitfolgen dieser Niedrigstkonzentrationen entwickeln und unerwartete Effekte durch die Wechselwirkung zwischen verschiedenen Wirkstoffen (Cocktaileffekt) entstehen. Besonders gefährdet sind dabei sensible Bevölkerungsgruppen wie Kinder und chronisch Kranke. Es besteht daher nicht zuletzt aus Vorsorgegründen Handlungsbedarf. Das Problem der Medikamentenreste im Wasser ist bei den wichtigsten Akteuren des Gesundheitssystems weitgehend unbekannt. Auch wenn Wissen nicht mit Bewusstsein gleichgesetzt werden kann – denn es gibt auch das Phänomen des Nicht-Wissen-Wollens – geht es in einem ersten Schritt darum, fundiertes Wissen zu erzeugen. Nur auf Basis dieser Sensibilisierung können weitere Strategien umgesetzt und letztendlich Aufklärung und Verhaltensänderungen erreicht werden. Dabei geht es um die gesamte Alltagspraxis im Umgang mit Medikamenten. Diese umfasst Fragen der Verschreibung, der Compliance, der nichtmedikamentösen Krankheitsvorsorge bis hin zum Arzt-Patienten-Verhältnis. Das ist nämlich häufig von Missverständnissen und mangelnder Kommunikation über – vermeintliche – Verschreibungsnotwendigkeiten geprägt. Der erste Teil der Strategie für die Bevölkerung soll über unterschiedliche Kanäle und Medien drei unterschiedliche Zielgruppen ansprechen, die in einer empirischen Untersuchung vom ISOE identifiziert wurden und auf das angesprochene Problem ganz unterschiedlich reagieren: · ‚Die Verleugner/Relativierer‘ · ‚Die Aufklärungsinteressierten‘ · ‚Die Hypersensiblen‘ Jede Zielgruppe soll in der passenden sprachlichen und argumentativen Art und Weise durch spezifische Medien und mit dem richtigen Grad der Differenziertheit angesprochen werden. Dabei spielen „die Aufklärungsinteressierten“ eine Opinionleader-Rolle. Sie können über anspruchsvolle Medien mit sehr differenzierten Informationen versorgt werden und geben dieses Wissen dann in angemessener Form an ihre Gesprächspartner weiter. Der zweite Teil der Strategie für die Bevölkerung bezieht sich auf die Kommunikation richtiger Entsorgungswege für Altmedikamente. Ziel ist es, dass Medikamentenreste nur noch in der Apotheke, keinesfalls aber in der Spüle oder in der Toilette entsorgt werden. Auf Grundlage einer Analyse typischer Fehler in bereits bestehenden Kommunikationsmedien zu diesem Thema hat das ISOE Empfehlungen zur richtigen Konzeption von Infomaterialien erarbeitet. Bei der Ansprache der Apotheker geht es in einem ersten Schritt um die Vermittlung von Faktenwissen: Wir schlagen dazu eine PR-Kampagne vor, die Artikel in den wichtigsten Fachmedien platziert. Gleichzeitig soll das Thema auch Teil der Aus- und Fortbildung werden. Zusätzlich soll die Beraterfunktion der Apotheken gestärkt werden. Die spezielle Zielgruppe der umweltsensiblen Kunden würde durchaus positiv darauf reagieren, wenn sie auf die Problematik der Medikamentenreste im Wasser hingewiesen würde. Bei allen anderen Kunden können und sollen die Apotheker ihre Rolle als Berater wahrnehmen: Sie betonen, wie wichtig die korrekte Einnahme (Compliance) und adäquate Packungsgrößen sind und warnen ihre Kunden, insbesondere die älteren, auch vor potenziellen Fehleinnahmen. Bei der Kommunikationsstrategie für Ärzte geht es im ersten Schritt ebenfalls um Wissen. Dabei muss aber deren Selbstverständnis als Wissenschaftler bei gleichzeitig niedrigem Wissensstand in diesem speziellen Feld berücksichtigt werden. Hier muss der Weg einer ‚diskursiven Selbstaufklärung‘ beschritten werden. Das Thema Medikamentenreste im Wasser kann somit nicht von Laien von außen an die Ärzte herangetragen werden, sondern muss in wichtigen Medien der Ärzteschaft und durch Verbandsfunktionäre angenommen und kommuniziert werden (top-down). Wenn es im zweiten Schritt um eine Problemsensibilisierung geht, muss mit starkem Widerstand eines Teils der Ärzteschaft gerechnet werden. Sie könnten fürchten, dass eine Einmischung in Heilungspläne aus Umweltsicht droht und betonen, dass Ärzte nicht für Umweltfragen zuständig seien. Letztlich steht – das haben empirische Untersuchungen des ISOE gezeigt – hinter dieser Problemabwehr ein Tabu: Es soll nicht darüber gesprochen werden, dass in zahlreichen Praxen zu viel verschrieben wird. Diese Problematik kann tatsächlich nicht aus der Umweltperspektive angegangen werden. Doch decken sich hier die Ziele des Gewässerschutzes mit den ökonomischen Zielen eines sparsamen Umgangs mit Arzneimitteln. Bei jeder Kommunikationsmaßnahme für diese Zielgruppe muss berücksichtigt werden, dass sich die Ärzte von dem, was sie als ‚Dauergesundheitsreform‘ aller Regierungen wahrnehmen, gegängelt fühlen. Sie sind keinesfalls bereit, eine neue Form der Regulierung, diesmal aus Umweltgründen, hinzunehmen. Ganz anders wird das Problem von ‚kritischen Ärzten‘ wie Umweltmedizinern und von Ärzten mit Naturheilschwerpunkt gesehen. Sie interessieren sich für die Problematik, weil sie einen Zusammenhang zwischen Umweltqualität und Gesundheit sehen. Außerdem haben sie Patienten, die an möglichst wenig Medikamentenverschreibungen, dafür aber an einer ‚sprechenden Medizin‘ interessiert sind. Wenn eine Kommunikationsstrategie also auch das schwierige Problem der übermäßigen Verschreibungen angehen will, empfiehlt es sich, die Erfahrungen dieser Mediziner einzubeziehen und zusätzlich auf eine ‚Bottom-up-Strategie‘ abzuzielen. Mit der Umsetzung der strategischen Kommunikation sollte eine Agentur beauftragt werden, die Erfahrungen im ‚Issue Management‘ vorweisen kann. Weiterhin sollte die Agentur Kenntnisse im Social Marketing und der Beeinflussung von Verhalten haben. Alle wichtigen Entscheidungen sollten von einem Konsens-Gremium (Runder Tisch ‚MeriWa‘1) verabschiedet werden, in dem die Ärzteschaft, die Apotheker sowie die Verbraucherinnen und Verbraucher angemessen repräsentiert sind.
The Alborz Mountains are forming a ~100 km wide, E-W trending mountain chain where individual summits are up to 5000 m in elevation. The Alborz Mountains range are part of the Alpine orogen and are straddling a 2000 km wide area S of the Caspian Sea. The rocks of the Alborz Mountains consist of Neogen sediments, which are affected by folding and faulting. In the western part of the Alborz Mountains the folds and faults are trending NW-SE, whereas in the eastern part they are trending NE-SW. GPS data confirm N-S shortening including dextral strike-slip along ESE-WNW trending faults, and sinistral strike-slip along ENE-WSW trending faults. The present thesis is focusing on the active Garmsar salt nappe, the fragmented roof of which is pierced by rock salt which extruded near the front of the Alborz Mountains Range. During the past 5 m.y. the front of the Alborz chain migrated towards SSW on top of the salt of the Garmsar basin. The salt was squeezed towards SSW and took place at the Great Kavir. The extruded salt is forming the Eyvanekey plateau between the cities of Eyvanekey and Garmsar. Both the Garmsar salt nappe and the Eyvanekey plateau are dextrally displaced for ca. 9 km along the Zirab-Garmsar fault. Structural analyses of the Garmsar salt nappe indicate three different groups of joints which are trending perpendicular and parallel to the local mechanical anisotropy. The folds of the study area are congruent (type 2 and 3 after Ramsay) resulting from viscose inhomogeneous flow. InSAR-Investigations suggest the Alborz Mountains to be lifted up by ca. 1 cm/a, while horizontal shortening is active at a rate of 8 ±2 mm/a. These values are consistent with GPS data. Based on nine „Advanced Synthetic Aperture Radar“ (ASAR) scenarios, produced by the ENVISAT satellite of the European space agency between 2003 and 2006, we used interferograms to map the displacement via 22 increments during 2 – 18 months. The results suggest that the topographic height of the surface of the salt is changing at a rate which is controlled by the season. The displacement ranges from subsidence at -40 to -50 mm/a to uplift of 20 mm/a. In order to investigate the time-dependent deformation with high spatial resolution, we used algorithms which are based on data of small base lines (SBAS). The resulting interferometric SAR time series analyses also suggest that the study area is largely subsiding at a rate that is controlled by the seasons. The map with the averaged LOS deformation velocities, on the other hand, suggests the subsidence to increase from the upper part of the salt nappe towards deeper topographic 5 levels of the agricultural lowlands. The major part of subsidence is probably caused by the annual rainfall which results in subrosion of salt. The spatial changes in the subsidence rate are probably controlled by the distribution of fountains, mining activity at the margin of the salt glacier, and faults and fractures inside the salt. Striking seasonal imprints are obvious along the agricultural areas which are surrounding the Garmsar salt nappe. These areas are rapidly subsiding in summer and spring when groundwater is used for irrigations. The maximum rate of subsidence (40-50 mm/a) is located E and W of the Eyvanekey plateau, where large areas are irrigated. The maximum displacement is 20 mm/a in the farmland and 5 mm/a in the center of the salt nappe. Depth estimates using Euler deconvolution method for gravimetric and magnetic data suggest the salt to extrude from a depth less than ca. 2000 m. The gravity field of the study area is characterized by strong anomalies in the SW and weak anomalies in the NE. A considerable negative anomaly in the N indicates that the northern part subsided, whereas the southern part was lifted up. The seismic data show three major horizons inside the Miocene sediments: the Lower Red Formation, the Qom Formation, and the Upper Red Formation. The western part of the study area seems to be free from salt domes. The layers of the upper part of the Qom Formation show thinning along the NE and NW trending faults. In some areas the seismic reflectors indicate steep faults close the saddle of the folds. NE-SW-, NW-SE and E-Wtrending faults prevail. Analogue experiments have been carried out to extend our knowledge about the evolution of the Garmsar salt dome. We used a scaled model (34 cm * 25 cm * 2.5 cm) that was shortened perpendicular to its long side. The wedge shape of the Alborz Mountains was simulated by a wedge consisting of Styrofoam. Rock salt was simulated using Polydimethylsiloxan (PDMS), a linear viscous material with a viscosity of 2.3*104 Pa s and a density of 0.96 g/cm3 at room temperature. Other sediments were modeled using dry quartz sand. The experimental results can be used to simulate the structural evolution of the study area: The Alborz deformation front was emplaced on top of the salt rocks in the Garmsar area while migrating towards SSW. A salt basin and a salt extrusion have also been produced in the model. Cross sections through the wedge shaped analogue model indicate N- and S-dipping reverse faults, which are in line with the wedge shape of the Alborz chain. Moreover, ENE-WSW trending sinistral and ESE-WNW trending dextral strike-slip faults led to N-S shortening during the Miocene. Structural marker horizons, 6 which have been turned into Z-folds on the western fold limbs and to S-folds on the eastern fold limbs, are comparable with the folds of the study area. Solving the problem of waste is one of the central tasks of environmental protection. It is becoming increasingly difficult to find suitable sites that are acceptable to the public. Salt and salt formations have relevant properties to be utilizing as a repository for each kind of waste. The favorable properties make rock salt highly suitable as a host rock, in particular for nonradioactive and radioactive wastes. The Qom and Garmsar basins are the nearest salt diapirs to the Tehran province, and there are suitable repositories for waste disposal. Based on surface and subsurface data, the Garmsar salt diapir has been investigated as a case example for its suitability as a host and repository for various types of waste. The data used are based on field studies, interferometry, and geophysical investigations. The results of this study suggest the deep bedded salt of the Garmsar Salt Basin to be an appropriate host for the deposition of industrial waste. Rock salt of surficial layers or domes, on the other hand, is not regarded as an appropriate candidate for waste disposal.
