In der vorliegenden Arbeit wurden zwei motivationale Erklärungsmodelle, Zielorientierungen und das kognitiv-motivationale Prozessmodell, im Rahmen des selbstregulierten Lernens integriert. Selbstregulationsprozesse sind nach Carver und Scheier (1981) zyklisch angelegt. Zu den Bestandteilen der Selbstregulation gehören nach Boekaerts (1999)Regulation der Informationsverarbeitung, Regulation des Lernprozesses und Regulation des Selbsts. Auf dieser Ebene sind die Zielorientierungen angesiedelt. In dieser Arbeit das 2 x 2 Modell von Elliot und McGregor (2001) herangezogen. Es berücksichtigt zwei Dimensionen, die Valenz (Annäherungs- und Vermeidungskomponente) und die Kompetenz (Vergleichsmaßstab intern und extern). Hieraus resultieren: 1) Lern-Annäherungs-Ziele (positive Valenz & interner Vergleich, 2) Lern-Vermeidungs-Ziele (negative Valenz & interner Vergleich), 3) Leistungs-Annäherungs-Ziele (positive Valenz & externer Vergleich) und 4) Leistungs-Vermeidungs-Ziele (negative Valenz & externer Vergleich). Diese vier Zielorientierungen sind der erste Teil des integrierten Modells, der zweite zentrale Teil ist das kognitiv-motivationale Prozessmodell (Vollmeyer & Rheinberg, 1999, 2006). Ausgangspunkt ist die aktuelle Motivation, die aus 1) der Erfolgswahrscheinlichkeit, 2) der Misserfolgsbefürchtung, 3) dem Interesse und 4) der Herausforderung als unabhängige Faktoren besteht. Diese aktuelle Motivation wirkt nicht direkt auf die Leistung, sondern durch kognitive (hier: Metakognition) und motivationale (hier: Flow-Erleben) Mediatoren. Es wurden drei Fragestellungen bearbeitet. Die erste Fragestellung betraf das Zusammenspiel der Zielorientierungen und der aktuellen Motivation. Eine Überprüfung dieses integrierten Motivationsmodell erfolgte mittels Pfadanalyse. Die zweite und dritte Fragestellung befasste sich spezifisch mit dem kognitiv-motivationalen Prozessmodell. Es wurden Subgruppen auf der Basis der aktuellen Motivation identifiziert und ihre Bedeutung für die Mediatoren und die Leistung untersucht. Die dritte Fragestellung fokussierte den Prozesscharakter des Modells. Hier erfolgten Analysen über den Arbeitsprozess hinweg. Die Fragebögen (Achievement Goal Questionnaire (Elliot & McGregor, 2001), Metakognitionsfragebogen (aufbauend auf LIST (Wild, 2000) und Metacognitive Awareness Inventory (Schraw & Dennison, 1994), Fragebogen zur aktuellen Motivation (Rheinberg, Vollmeyer & Burns, 2001), Flow-Kurz-Skala (Rheinberg, Vollmeyer & Engeser, 2003)) und die Problemlöseaufgaben (Sudokus) wurden in einer Pilotstudie getestet. An der Hauptstudie nahmen 202 Personen teil (73% weiblich). Das integrierte Motivationsmodell geht davon aus, dass eine positiver Effekt der Zielorientierungen als Personenvariable auf die aktuelle Motivation besteht. Die aktuelle Motivation als Startpunkt des situationalen Geschehens wiederum wirkt positiv auf die Mediatoren Flow-Erleben und Metakognition, hat aber keinen direkten Effekt auf die Leistung. Dieser wird nämlich über die Mediatoren vermittelt. Deswegen wird ein positiver Effekt des Flow-Erlebens und der Metakognition auf die Leistung erwartet. Das Zusammenwirken der beiden Mediatoren kann aus dem kognitiv-motivationalen Prozessmodell nicht abgeleitet werden. Ob ein Zusammenhang besteht oder nicht wird geprüft. Für das Vorwissen wird ein positiver Effekt auf die aktuelle Motivation und die Leistung erwartet. Die Pfadanalyse zur Überprüfung des integrierten Motivationsmodells zeigte, dass eine Integration nicht nur theoretisch sinnvoll ist, sondern auch empirisch gestützt wird. Die Bedeutung der Metakognition als kognitiver Mediator wurde gezeigt. Die zweite Fragestellung fokussierte auf die aktuelle Motivation. Auf der Basis dieser wurden mittels Clusteranalyse drei distinkte Gruppen identifiziert: hoch Motivierte, niedrig Motivierte und ängstlich Motivierte (vergleichbar zu Vollmeyer & Rheinberg, 2004). Diese Gruppen unterschieden sich hinsichtlich ihres Flow-Erlebens, ihrer Metakognition und ihrer Leistung. Die hoch Motivierten erlebten am meisten Flow, berichteten mehr Metakognition und zeigten bessere Leistung. Der Prozesscharakter des kognitiv-motivationalen Prozessmodells stand im Mittelpunkt der dritten Fragestellung. Diese Analyse über die drei Sudokus hinweg zeigte eine ähnliche Entwicklung des Flow-Erlebens bei den hoch und niedrig Motivierten. Vom ersten zum zweiten Sudoku stieg es an, um dann wieder abzufallen. Dies war bei der Metakognition nicht der Fall. Die hoch Motivierten berichteten einen stärkeren Rückgang der Metakognition beim dritten Sudoku als die niedrig Motivierten. Bei der Leistung zeigte sich für die hoch Motivierten ein Anstieg beim zweiten Sudoku und dann ein Rückgang beim dritten, wohingegen die Leistung der niedrig Motivierten kontinuierlich abfiel. Abschließend wurden die Schwierigkeiten und Grenzen der vorliegenden Arbeit diskutiert und Implikationen der Ergebnisse für die Theorieentwicklung und ihre Bedeutung für den Anwendungskontext aufgezeigt.
Die spezifische Funktion des Sri Cakras in der beschriebenen externen Verehrung besteht in seiner doppelten Funktion zum einen als externes physisches Verehrungsobjekt und zum anderen als mentales Betrachtungsobjekt. Als externes physisches Verehrungsobjekt ermöglicht es dem Sadhaka, sich selbst als Göttin gegenüber zu treten und seine Identität mit der Göttin in der Zusammenführung von sich selbst mit der Göttin im Darbringen von materiellen Gaben, welche symbolisch für die Hingabe seiner selbst stehen, körperlich und sinnlich nachzuvollziehen. Hierbei richtet der Sadhaka seinen Geist und seine Handlung ganz auf die Göttin bzw. ihre Verehrung im Sri Cakra aus, so dass er sich selbst dabei vergisst und zur Göttin wird. Bei seiner externen Verehrung dient das Sri Cakra gleichzeitig auch als mentales Betrachtungsobjekt, dessen konzentrische Form den Verehrer in seine eigene Mitte, und somit zur Identifizierung mit der Göttin führt, und welches dem Sadhaka als Vorlage zur geistigen Durchschreitung des Kosmos und seines eigenen Inneren dient. Während der Sadhaka also seine Identität mit der Göttin in der körperlichen Verehrung nachvollzieht und realisiert, durchschreitet er es geistig und betrachtet hierbei ihre einzelnen Teilkräfte. In mentalen Verehrungsformen funktioniert das Sri Cakra, ebenso wie in der externen Verehrung, als mentales Betrachtungsobjekt, welches den Sadhaka in seine Mitte führt und in dessen Durchschreitung er den Teilkräften der Göttin begegnet, sich ihrer kosmische Bedeutung bewusst wird und sie in sich verwirklicht. Gleichzeitig dient das Sri Cakra als Verehrungsobjekt, während dessen Verehrung der Verehrer die Zusammenführung seiner selbst mit der Göttin jedoch nicht mit Körper und Sinnen nachvollzieht, sondern rein geistig. In der Bhavana-Verehrung wird die Verehrung als Zusammenführung der zwei Ebenen, der Teilgottheit und des Selbst, weder körperlich noch mental nachvollzogen, sondern die Identität der zwei zusammengeführten Ebenen durch Meditation dieselbe direkt verwirklicht. Das Sri Cakra dient hier als Vorlage für die Kräfte und die Reihenfolge, in welcher die Kräfte und ihre Identität mit ihren mikrokosmischen Entsprechungen meditiert werden. Die Identifizierung mit der Gottheit über die schrittweise Verwirklichung ihrer Teilkräfte beinhaltet das Erlangen von psychischen Kräften und Fähigkeiten, Siddhi. Diese Kräfte kann der Sadhaka im Rahmen magischer Rituale im Makrokosmos zum Wohle und zum Schaden anderer einsetzen. Magische Rituale beinhalten eine Verehrung, mittels welcher der Sadhaka sich mit seiner inneren Kraft verbindet und einen Akt, in welchem diese Kraft angewendet wird. In Anwendungen zur Heilung von Krankheiten funktionieren Yantras oft als Amulett, d. h. als Träger in welchen der Sadhaka seine innere Kraft einsetzt und von welchem aus sie auf den Amulett-Träger wirkt. Yantras können wie im Beispiel zum 37. Vers des Saundaryalaharis gezeigt, auch als Verehrungsobjekte funktionieren, durch dessen Verehrung die dem Yantra entsprechende Kraft im Verehrer aktiviert wird. Krankheit hat verschiedene ursächliche Ebenen und verschiedene Ebenen der Manifestation. Sie kann somit auf diesen verschiedenen Ebenen behandelt werden. Das Yantra, welches in einer heilenden Anwendung eingesetzt wird, wird jeweils in Abhängigkeit der Ebene, deren Kräfte mobilisiert werden sollen, ausgewählt. Der nach religiösem Heil strebende Sadhaka verwirklicht im Rahmen seiner schrittweisen Identifikation mit der Göttin nach und nach die Kräfte aller Seinsebenen, und somit auch die Kräfte zur Kontrolle der körperlichen und psychischen Ebenen, sowie Kräfte, die vor negativen Einflüssen schützen und Krankheit und Leiden ausgleichen. Der Weg zu religiösem Heil führt somit über das Erlangen von Gesundheit und die Auflösung von Krankheit und Leiden, sowie zum Schutz vor Gefahren und schädlichen äußeren Einflüssen. Diese auf dem Weg zur Erkenntnis der Göttin erworbenen inneren Kräfte kann der Sadhaka im Makrokosmos einsetzen. Oft dienen hierbei Yantras als Medien, über welche diese Kräfte weitergegeben werden.
Through the use of information about the biological target structure, the optimization of potential drugs can be improved. In this work I have developed a procedure that uses the quantitative change in the chemical perturbations (CSP) in the protein from NMR experiments for driving protein-ligand docking. The approach is based on a hybrid scoring function (QCSPScore) which combines traditional DrugScore potentials, which describe the interaction between protein and ligand, with Kendall’s rank correlation coefficient, which evaluates docking poses in terms of their agreement with experimental CSP. Prediction of the CSP for a specific ligand pose is done efficiently with an empirical model, taking into account only ring current effects. QCSPScore has been implemented in the AutoDock software package. Compared to previous methods, this approach shows that the use of rank correlation coefficient is robust to outliers. In addition, the prediction of native-like complex geometries improved because the CSP are already being used during the docking process, and not only in a post-filtering setting for generated docking poses. Since the experimental information is guaranteed to be quantitatively used, CSP effectively contribute to align the ligand in the binding pocket. The first step in the development of QCSPScore was the analysis of 70 protein-ligand complexes for which reference CSP were computed. The success rate in the docking increased from 71% without involvement of CSP to 100% if CSP were considered at the highest weighting scheme. In a second step QCSPScore was used in re-docking three test cases, for which reference experimental CSP data was available. Without CSP, i.e. in the use of conventional DrugScore potentials, none of the three test cases could be successfully re-docked. The integration of CSP with the same weighting factor as described above resulted in all three cases successfully re-docked. For two of the three complexes, native-like solutions were only produced if CSP were considered.Conformational changes in the binding pockets of up to 2 Å RMSD did not affect the success of the docking. QCSPScore will be particularly interesting in difficult protein-ligand complexes. They are in particular those cases in which the shape of the binding pocket does not provide sufficient steric restraints such as in flat protein-protein interfaces and in the virtual screening of small chemical fragments.
This thesis presents a 5.9 Å map of yeast FAS obtained by cryo-electron microscopy using single particle analysis (SPA). The EM-map has been analyzed both by quantitative and qualitative analysis to aid in understanding of the structure and dynamics of yeast FAS. This study approaches the factors limiting the resolution in EM (>20 Å) and further discusses the possibilities of achieving higher-resolutions (<10 Å) in cryo-EM by single particle analysis. Here, SPA is highlighted as a powerful tool for understanding the structure and dynamics of macro-molecular complexes at near native conditions. Though SPA has been used over the last four decades, the low-resolution range (20-30 Å) of the method has limited its use in structural biology. Over the last decade, sub nanometer resolution (<10 Å) structures solved by SPA have been reported --both in studies involving symmetric particles, such as GroEL (D7) and asymmetric particles, such as ribosomes (C1). Recently, near-atomic resolution in the range of 3.8-4.2 Å has been achieved in cases of highly symmetric icosahedral viral capsid structures as well. The yeast FAS structure (D3) presented here is one of two low symmetry structures submitted to the EM-database in a resolution range of 5-6 Å; the other being GroEL (D7). Fatty acid synthase (FAS) is the key enzyme for the biosynthesis of fatty acids in living organisms. There are two types of FAS, namely the type II FAS system in prokaryotes, consisting of a set of individual enzymes, and type I FAS found in eukaryotes as a multienzyme complex. Yeast fatty acid synthase (FAS) is a 2.6 MDa barrel-shaped multienzyme complex, which carries out cyclic synthesis of fatty acids. By electron cryomicroscopy of single particles we obtained a 3D map of yeast FAS at 5.9 Å resolution. Compared to the crystal structures of fungal FAS, the EM map reveals major differences and new features that indicate a considerably different arrangement of the complex in solution, as well as a high degree of variance inside the barrel. Distinct density regions in the reaction chambers next to each of the catalytic domains fit well with the substratebinding acyl carrier protein (ACP) domain. In each case, this resulted in the expected distance of ~18 Å from the ACP substrate binding site to the active site of the catalytic domains. The multiple, partially occupied positions of the ACP within the reaction chamber provide direct insight into the proposed substrate-shuttling mechanism of fatty acid synthesis in this large cellular machine.
Acute myeloid leukemia (AML) is a hematopoietic cell disorder characterized by a block in differentiation and increased proliferation and survival of malignant blasts. Expansion of the malignant cell clone effects the normal production of blood cells and – if left untreated – leads to death. Receptor tyrosine kinases (RTKs) play an important role in the pathogenesis of AML, as they are either often mutated or overexpressed. In normal hematopoiesis, RTK signal termination is tightly controlled, and involves ubiquitination, internalization, endocytosis and degradation. Cbl proteins are E3 ligases and have been shown to ubiquitinate several activated RTKs, including Flt3 and Kit, targeting them for degradation. Recently, several Cbl mutations have been identified: Cbl-R420Q was identified in an AML patient and Cbl-70Z was identified in a mouse lymphoma model. In this thesis work, the role of these Cbl mutants in Kit signaling and in a mouse transplantation model was studied. Cbl mutants (Cbl-R420Q, Cbl-70Z) have the ability to transform the myeloid 32D cell line in cooperation with Kit WT. Cbl mutants along with Kit promoted interleukin-3 (IL3)-independent proliferation and enhanced the cell survival of 32D cells. In contrast, expression of the Cbl mutants alone did not confer IL3-independent growth. Stem cell factor (SCF, the Kit ligand) dependent growth was enhanced in the presence of Cbl mutants and Cbl mutants promoted colonogenic growth in the presence of Kit. Furthermore, Cbl mutants inhibited the ubiquitination of the activated Kit receptor. In addition, Cbl mutants inhibited the endocytosis of the activated Kit receptor. Retroviral expression of Cbl mutants in transplanted bone marrow induced a generalized mastocytosis, a myeloproliferative disease and, in rare care cases, myeloid leukemia. Splenomegaly was observed in the presence of Cbl mutants. Furthermore, mast cells with variable range of infiltration were noticed in all the vital organs (spleen, liver, bone marrow, lung, kidney, heart) of Cbl (mutant) transplanted mice. Almost all recipients of bone marrow cells transduced with Cbl mutants developed a lethal hematologic disorder with a mean latency of 341 days in the Cbl-R420Q group and 395 days in the Cbl-70Z group. This is the first published report on a hematological disease with Cbl mutants in a mouse model. Co-immunoprecipitation studies indicated that Cbl-70Z binds to Kit, even in the absence of Kit ligand. Cbl-R420Q also bound to Kit in the absence of SCF, albeit to a lesser extent. Association of Cbl mutants to Kit was enhanced in the presence of SCF. Signaling studies demonstrated the constitutive activation of Akt and Erk in the presence of Cbl mutants and Kit. In addition, Cbl mutants enhanced the SCF-dependent Kit, Akt and Erk activation. Cbl-70Z, in association with kinase-dead Kit (Kit-KD) or kinase-dead Flt3 (Flt3-KD), conferred IL3-independent growth and survival to the myeloid 32D cell line. Cbl-R420Q provided only a slight growth advantage in the presence of Kit-KD. As demonstrated by pharmacological inhibition studies, Akt activation was necessary for the transformation mediated by Cbl-70Z and Kit-KD / Flt3-KD. Cbl mutants enhanced the Src family kinases (SFKs) activity. The pharmacological inhibition of SFK activity inhibited the proliferation and colonogenic growth. Interaction was found between Cbl-70Z, SFKs and Kit-KD. The SFK member Fyn was identified to bind to Cbl. In addition, kinase activity of SFKs was necessary for binding to Cbl, since SFKs inhibition by PP-2 abolished the binding between the complex-binding partners. Dasatinib and PP-2, both SFK inhibitors, inhibited the Cbl and Akt phosphorylation indicating that Fyn acts upstream of Akt. Inhibition of Kit with imatinib reduced the proliferation of cells overexpressing Kit WT and Cbl-70Z much stronger compared with cells expressing Kit-KD and Cbl-70Z, but much less than the dual KIT/SFK inhibitor dasatinib. This indicated that Kit kinase activity was required but not essential. The data presented in this thesis work implies that both RTK and SFK inhibition may have to be targeted, in order to effectively prevent transformation. In summary, the present thesis work indicates an important role of Cbl, Kit and SFKs in myeloid transformation and deregulated signal transduction.
In dieser Arbeit wird der Frage nachgegangen, wie sich eine Darstellung des Nichts durch die Darstellung von Etwas vollzieht. Wie Arnold P. Hinchliffe richtig feststellt zerstört das Konzept von Nichts sich selbst, denn es produziert Etwas , dennoch haben wir eine Vorstellung von dem mit Nichts bezeichneten. Diese Arbeit stellt es sich zur Aufgabe die Leerstellen in der Handlung von Samuel Becketts En attendant Godot und Franz Kafkas Der Prozess zu analysieren. Hierfür ist es notwendig verschiedene Motive zu betrachten, die Lücken in ihren Definitionen aufweisen. Es sind diese Leerstellen auf denen gerade die Handlung aufbaut und sie nehmen einen großen Teil dabei ein, die Handlung zum Voranschreiten zu bewegen. Anhand von Becketts En attendant Godot und Kafkas Der Prozess sollen die Darstellungsweisen der nicht vorhandenen Begebenheiten betrachtet und analysiert werden. Das sich äußernde Nicht-sein durch Etwas führt zum Sein der Schriften selbst. Sie thematisieren ihre eigene nicht vorhandene Handlung, eine Leerstelle und handeln eben dadurch von etwas. Sowohl En attendant Godot als auch Der Prozess können infolge dessen als von sich selbst handelnde Schriften gelesen werden, was sich diese Arbeit zum Ziel setzt. Sie sind intratextuell, gehen jedoch auf Grund der Art dieser intratextuellen Darstellung weit über einen bloßen Intratext hinaus. In Becketts En attendant Godot stellt sich das Fehlen einer eigentlichen Handlung sehr deutlich dar. Das Erwartete, das Eintreffen von Godot, bleibt aus. Selbst die Gewissheit der Verabredung und der Existenz Godots verschwindet. Es bleibt nichts außer der Hoffnung der Charaktere, Vladimir und Estragon, die keine herkömmlichen Rollen haben, also eigentlich Nicht-Rollen sind. Vor anderem Hintergrund (dem der Gesetze) stellt sich das Nichts bei Kafkas Prozess dar. Josef K. wird eines Morgens verhaftet, doch die Verhaftung ist keine, die ihn an seiner gewöhnlichen Lebensweise hindert. Es ist eine Verhaftung ohne Festnahme, ein hängen bleiben an etwas Unbestimmtem, ein festhalten, ohne vor etwas zu bewahren.
Im adulten Säugerhirn findet Neurogenese in der SVZ der Seitenventrikel kontinuierlich statt. Eine Vielzahl von Signalsystemen steuert in komplexer Weise zelluläre Antworten und reguliert die Proliferation, Differenzierug und Wanderung NSZ. Gegenwärtig ist nur wenig über die zugrundeliegenden Signalwege bekannt. Zunehmend gibt es Hinweise darauf, dass Nukleotide an diesen Prozessen beteiligt sind. Frühere Untersuchungen unserer Arbeitsgruppe zeigten, das die Nukleotide ADPbetaS und UTP in kultivierten NSZ der adulten SVZ einen schnellen Kalziumeinstrom induzieren und die Wachstumsfaktor-vermittelte NSZ-Proliferation steigern. In der vorliegenden Arbeit wurde ein System zur Kultivierung adhärenter adulter NSZ etabliert. Die Untersuchungen zeigen, dass adulte NSZ eine Vielzahl an P2Y- und P2X-Rezeptoren, sowie die Nukleotid-hydrolysierenden Enzyme NTPDase2 und TNAP exprimieren. Untersuchungen der ADPbetaS-, UTP- und EGF-vermittelten Signalwege zeigen, dass alle drei Agonisten eine ERK1/2- und CREB-Phosphorylierung induzieren, wobei sich die zeitlichen Charakteristika zwischen den Nukleotiden und EGF unterscheiden. Inhibierungsexperimente geben Einblicke in die dabei aktivierten Signalkaskaden und weisen auf eine ADPbetaS-induzierte Transaktivierung des EGF-Rezeptors hin. Während UTP über den P2Y2-Rezeptor wirkt, übt ADPbetaS seine Funktion über den P2Y1- und P2Y13-Rezeptor aus. Die Daten implizieren zudem, dass Nukleotide und EGF gleiche Zielproteine über verschiedene Signalwege induzieren und dass sie das Potenzial besitzen, bei der Kontrolle der Zellproliferation in der adulten Neurogenese synergistisch zu agieren. Vergleichende Analysen mit kultivierten NSZ aus Wildtyp-, P2Y1- und P2Y2-Rezeptor-Knockout-Mäusen belegen ein verändertes Antwortverhalten in Gegenwart von ADPbetaS, UTP und EGF und lassen kompensatorische Mechanismen vermuten. Die Resultate dieser Arbeit demonstrieren zudem, dass ATP, ADPbetaS, UTP und EGF die Migration von NSZ induzieren. Parallel dazu konnten Veränderungen des Aktinzytoskelletes, wie die Zunahme an F-Aktin, die Bildung von Stressfasern und eine Veränderung der Zellmorphologie gezeigt werden. Diese Prozesse gehen mit einer Aktivierung der Proteinkinasen Akt und FAK einher. Die Daten weisen darauf hin, dass Nukleotide und EGF für die Zytoarchitektur der SVZ und die Wanderung von Neuroblasten zum OB eine wichtige Rollen spielen könnten.
Recently a first genome-wide analysis of translational regulation using prokaryotic species had been performed which revealed that regulation of translational efficiency plays an important role in haloarchaea. In fact, the fractions of genes under differential growth phase-dependent translational control in the two species Halobacterium salinarum and Haloferax volcanii were as high as in eukaryotes. However, nothing is known about the mechanisms of translational regulation in archaea. Therefore, two genes exhibiting opposing directions of regulation were selected to unravel the importance of untranslated regions (UTRs) for differential translational control in vivo. Differential translational regulation in exponentially growing versus stationary phase cells was studied by comparing translational efficiencies using a reporter gene system. Translational regulation was not observed when 5'-UTRs or 3'-UTRs alone were fused to the reporter gene. However, their simultaneous presence was sufficient to transfer differential translational control from the native transcript to the reporter transcript. This was true for both directions of translational control. Translational regulation was completely abolished when stem loops in the 5'-UTR were changed by mutagenesis. An “UTR-swap” experiment demonstrated that the direction of translational regulation is encoded in the 3'-UTR, not in the 5'-UTR. While much is known about 5'-UTR-dependent translational control in bacteria, the reported findings provide the first examples that both 5'- and 3'-UTRs are essential and sufficient to drive differential translational regulation in a prokaryote and therefore have to functionally interact in vivo. The current results indicate that 3'-UTR-dependent translational control had already evolved before capping and polyadenylation of transcripts were invented, which are essential for circularization of transcripts in eukaryotes.
Anliegen: Der hausärztlich-ambulanten Versorgung von depressiven Patienten kommt eine zentrale Bedeutung zu. Ein wichtiges Symptom von Depressionen ist Suizidalität. Suizidgedanken bleiben im Hausarztkontakt jedoch oftmals unerkannt. Eine verbesserte Kenntnis von Prädiktoren für Suizidgedanken speziell bei depressiven Patienten in der Primärversorgung kann dazu beitragen, die Erkennung von Suizidgedanken durch den Hausarzt zu verbessern und dadurch eine rechtzeitige Intervention zu ermöglichen. Mit Prädiktoren für Suizidgedanken explizit bei depressiven Patienten in der Primärversorgung haben sich bislang nur wenige Studien befasst, mit teilweise heterogenen Ergebnissen.
