Background: Approximately one in three patients suffers from preoperative anaemia. Even though haemoglobin is measured before surgery, anaemia management is not implemented in every hospital. Objective: Here, we demonstrate the implementation of an anaemia walk-in clinic at an Orthopedic University Hospital. To improve the diagnosis of iron deficiency (ID), we examined whether reticulocyte haemoglobin (Ret-He) could be a useful additional parameter. Material and Methods: In August 2019, an anaemia walk-in clinic was established. Between September and December 2019, major orthopaedic surgical patients were screened for preoperative anaemia. The primary endpoint was the incidence of preoperative anaemia. Secondary endpoints included Ret-He level, red blood cell (RBC) transfusion rate, in-hospital length of stay and anaemia at hospital discharge. Results: A total of 104 patients were screened for anaemia. Preoperative anaemia rate was 20.6%. Intravenous iron was supplemented in 23 patients. Transfusion of RBC units per patient (1.7 ± 1.2 vs. 0.2 ± 0.9; p = 0.004) and hospital length of stay (13.1 ± 4.8 days vs. 10.6 ± 5.1 days; p = 0.068) was increased in anaemic patients compared to non-anaemic patients. Ret-He values were significantly lower in patients with ID anaemia (33.3 pg [28.6–40.2 pg]) compared to patients with ID (35.3 pg [28.9–38.6 pg]; p = 0.015) or patients without anaemia (35.4 pg [30.2–39.4 pg]; p = 0.001). Conclusion: Preoperative anaemia is common in orthopaedic patients. Our results proved the feasibility of an anaemia walk-in clinic to manage preoperative anaemia. Furthermore, our analysis supports the use of Ret-He as an additional parameter for the diagnosis of ID in surgical patients.
USP22 controls necroptosis by regulating receptor-interacting protein kinase 3 ubiquitination

Dynamic control of ubiquitination by deubiquitinating enzymes is essential for almost all biological processes. Ubiquitin-specific peptidase 22 (USP22) is part of the SAGA complex and catalyzes the removal of mono-ubiquitination from histones H2A and H2B, thereby regulating gene transcription. However, novel roles for USP22 have emerged recently, such as tumor development and cell death. Apart from apoptosis, the relevance of USP22 in other programmed cell death pathways still remains unclear. Here, we describe a novel role for USP22 in controlling necroptotic cell death in human tumor cell lines. Loss of USP22 expression significantly delays TNFα/Smac mimetic/zVAD.fmk (TBZ)-induced necroptosis, without affecting TNFα-mediated NF-κB activation or extrinsic apoptosis. Ubiquitin remnant profiling identified receptor-interacting protein kinase 3 (RIPK3) lysines 42, 351, and 518 as novel, USP22-regulated ubiquitination sites during necroptosis. Importantly, mutation of RIPK3 K518 reduced necroptosis-associated RIPK3 ubiquitination and amplified necrosome formation and necroptotic cell death. In conclusion, we identify a novel role of USP22 in necroptosis and further elucidate the relevance of RIPK3 ubiquitination as crucial regulator of necroptotic cell death.
In situ investigation of membrane proteins is a challenging task. Previously we demonstrated that nitroxide labels combined with pulsed ESR spectroscopy is a promising tool for this purpose. However, the nitroxide labels suffer from poor stability, high background labeling, and low sensitivity. Here we show that Finland (FTAM) and OX063 based labels enable labeling of the cobalamin transporter BtuB and BamA, the central component of the β-barrel assembly machinery (BAM) complex, in E coli. Compared to the methanethiosulfonate spin label (MTSL), trityl labels eliminated the background signals and enabled specific in situ labeling of the proteins with high efficiency. The OX063 labels show a long phase memory time (TM) of ≈5 μs. All the trityls enabled distance measurements between BtuB and an orthogonally labeled substrate with high selectivity and sensitivity down to a few μm concentration. Our data corroborate the BtuB and BamA conformations in the cellular environment of E. coli.
Background and purpose: Superficial siderosis of the central nervous system is a sporadic finding in magnetic resonance imaging, resulting from recurrent bleedings into the subarachnoid space. This study aimed to determine the frequency of spinal dural cerebrospinal fluid (CSF) leaks amongst patients with a symmetric infratentorial siderosis pattern. Methods: In all, 97,733 magnetic resonance images performed between 2007 and 2018 in our neurocenter were screened by a keyword search for “hemosiderosis” and “superficial siderosis.” Siderosis patterns on brain imaging were classified according to a previously published algorithm. Potential causative intracranial bleeding events were also assessed. Patients with a symmetric infratentorial siderosis pattern but without causative intracranial bleeding events in history were prospectively evaluated for spinal pathologies. Results: Forty-two patients with isolated supratentorial siderosis, 30 with symmetric infratentorial siderosis and 21 with limited (non-symmetric) infratentorial siderosis were identified. Amyloid angiopathy and subarachnoid hemorrhage were causes for isolated supratentorial siderosis. In all four patients with a symmetric infratentorial siderosis pattern but without a causative intracranial bleeding event in history, spinal dural abnormalities were detected. Dural leaks were searched for in patients with symmetric infratentorial siderosis and a history of intracranial bleeding event without known bleeding etiology, considering that spinal dural CSF leaks themselves may also cause intracranial hemorrhage, for example by inducing venous thrombosis due to low CSF pressure. Thereby, one additional spinal dural leak was detected. Conclusions: Persisting spinal dural CSF leaks can frequently be identified in patients with a symmetric infratentorial siderosis pattern. Diagnostic workup in these cases should include magnetic resonance imaging of the whole spine.
Linking epigenetic signature and metabolic phenotype in IDH mutant and IDH wildtype diffuse glioma

Aims: Changes in metabolism are known to contribute to tumour phenotypes. If and how metabolic alterations in brain tumours contribute to patient outcome is still poorly understood. Epigenetics impact metabolism and mitochondrial function. The aim of this study is a characterisation of metabolic features in molecular subgroups of isocitrate dehydrogenase mutant (IDHmut) and isocitrate dehydrogenase wildtype (IDHwt) gliomas. Methods: We employed DNA methylation pattern analyses with a special focus on metabolic genes, large-scale metabolism panel immunohistochemistry (IHC), qPCR-based determination of mitochondrial DNA copy number and immune cell content using IHC and deconvolution of DNA methylation data. We analysed molecularly characterised gliomas (n = 57) for in depth DNA methylation, a cohort of primary and recurrent gliomas (n = 22) for mitochondrial copy number and validated these results in a large glioma cohort (n = 293). Finally, we investigated the potential of metabolic markers in Bevacizumab (Bev)-treated gliomas (n = 29). Results: DNA methylation patterns of metabolic genes successfully distinguished the molecular subtypes of IDHmut and IDHwt gliomas. Promoter methylation of lactate dehydrogenase A negatively correlated with protein expression and was associated with IDHmut gliomas. Mitochondrial DNA copy number was increased in IDHmut tumours and did not change in recurrent tumours. Hierarchical clustering based on metabolism panel IHC revealed distinct subclasses of IDHmut and IDHwt gliomas with an impact on patient outcome. Further quantification of these markers allowed for the prediction of survival under anti-angiogenic therapy. Conclusion: A mitochondrial signature was associated with increased survival in all analyses, which could indicate tumour subgroups with specific metabolic vulnerabilities.
Culturally adapted cognitive behavioral therapy (CA-CBT) is a well-evaluated, transdiagnostic group intervention for refugees that uses psychoeducation, meditation, and stretching exercises. In the current study, we added problem-solving training to CA-CBT and evaluated this treatment (i.e., CA-CBT+) in a randomized controlled pilot trial with a sample of Farsi-speaking refugees. Participants (N = 24) were male refugees diagnosed with DSM-5 PTSD, major depressive disorder, and anxiety disorders who were randomly assigned to either a treatment or waitlist control (WLC) condition. Treatment components were adapted both to the specific cultural background and the current social problems of asylum seekers. Assessments were performed pretreatment, 12-weeks posttreatment, and 1-year follow-up. The primary treatment outcome was the General Health Questionnaire (GHQ-28); secondary outcome measures included the Posttraumatic Stress Disorder Checklist, Patient Health Questionnaire, Somatic Symptom Scale, World Health Organization Quality of Life, and Emotion Regulation Scale. Eleven of 12 participants were randomized to CA-CBT+ completed treatment. Based on intent-to-treat data, large between-group effect sizes were seen at posttreatment in the GHQ-28, d = 3.0, and for most secondary outcome measures. Improvements for individuals in the treatment group decreased at 1-year follow-up, but effect sizes demonstrated continued large improvements on all measures as compared to pretreatment levels. In summary, CA-CBT+ led to large improvements in general psychopathological distress and quality of life, which were maintained in the long term. In addition, the dropout rate was very low, with delivery in group format. Thus, problem-solving training appears to be a promising addition to CA-CBT.
This article presents the findings from systematically reviewing 26 empirical research studies published from 2005 to 2014 on the use of GIS for learning and teaching. By employing methods of narrative synthesis and qualitative content analysis, the study gives evidence about the state of knowledge of competence-based GIS education. The results explain what factors and variables effect GIS learning in terms of technology use, major subject contents, learning contexts, and didactic and pedagogical aspects. They also show what facets of knowledge, process skills, and affect the research literature has investigated. The analysis of the type and quality of the methods used indicates that current GIS education research is a heterogeneous field that needs a systematic research framework for future efforts, according to empirical education research.
This paper explores the many interesting implications for oscillator design, with optimized phase-noise performance, deriving from a newly proposed model based on the concept of oscillator conjugacy. For the case of 2-D (planar) oscillators, the model prominently predicts that only circuits producing a perfectly symmetric steady-state can have zero amplitude-to-phase (AM-PM) noise conversion, a so-called zero-state. Simulations on standard industry oscillator circuits verify all model predictions and, however, also show that these circuit classes cannot attain zero-states except in special limit-cases which are not practically relevant. Guided by the newly acquired design rules, we describe the synthesis of a novel 2-D reduced-order LC oscillator circuit which achieves several zero-states while operating at realistic output power levels. The potential future application of this developed theoretical framework for implementation of numerical algorithms aimed at optimizing oscillator phase-noise performance is briefly discussed.
We report here the in-cell NMR-spectroscopic observation of the binding of the cognate ligand 2′-deoxyguanosine to the aptamer domain of the bacterial 2′-deoxyguanosine-sensing riboswitch in eukaryotic cells, namely Xenopus laevis oocytes and in human HeLa cells. The riboswitch is sufficiently stable in both cell types to allow for detection of binding of the ligand to the riboswitch. Most importantly, we show that the binding mode established by in vitro characterization of this prokaryotic riboswitch is maintained in eukaryotic cellular environment. Our data also bring important methodological insights: Thus far, in-cell NMR studies on RNA in mammalian cells have been limited to investigations of short (<15 nt) RNA fragments that were extensively modified by protecting groups to limit their degradation in the intracellular space. Here, we show that the in-cell NMR setup can be adjusted for characterization of much larger (≈70 nt) functional and chemically non-modified RNA.
We developed three bathochromic, green-light activatable, photolabile protecting groups based on a nitrodibenzofuran (NDBF) core with D-π-A push–pull structures. Variation of donor substituents (D) at the favored ring position enabled us to observe their impact on the photolysis quantum yields. Comparing our new azetidinyl-NDBF (Az-NDBF) photolabile protecting group with our earlier published DMA-NDBF, we obtained insight into its excitation-specific photochemistry. While the “two-photon-only” cage DMA-NDBF was inert against one-photon excitation (1PE) in the visible spectral range, we were able to efficiently release glutamic acid from azetidinyl-NDBF with irradiation at 420 and 530 nm. Thus, a minimal change (a cyclization adding only one carbon atom) resulted in a drastically changed photochemical behavior, which enables photolysis in the green part of the spectrum.
Die Multiple Sklerose (MS) gehört zu den häufigsten chronisch-entzündlichen Erkrankungen des zentralen Nervensystems in Deutschland und kann durch Sehstörungen, Paresen oder Sensibilitätsstörungen symptomatisch werden.
Konventionelle Magnetresonanztomographie (MRT)-Verfahren leisten in der Diagnostik der MS einen wichtigen Beitrag, da diese die Läsionslast der weißen Substanz gut darstellen können. Frühere Studien deuten an, dass kognitive und psychomotorische Symptome wie Fatigue sowie Konzentrations- und Gedächtnisstörungen bei der MS mit Schädigungen des zerebralen Kortex in Beziehung stehen könnten. Mit konventionellen MRT-Bildgebungsverfahren lässt sich zwar kortikale Atrophie, nicht jedoch die zugrundeliegenden mikrostrukturellen kortikalen Umbauprozesse erfassen. In der vorliegenden Studie wurden daher quantitative MRT(qMRT)-Verfahren verwendet, die eben diese diffusen kortikalen Gewebsveränderungen messen und quantifizieren können. Mithilfe der dabei genutzten Diffusions-Tensor-Bildgebung (DTI) als qMRT-Verfahren konnten Diffusionsanomalien analysiert und charakterisiert werden. Dabei wurden zwei Gewebsparameter im Gehirn bestimmt: die mittlere Diffusivität(MD) und die fraktionelle Anisotropie (FA). Da vorherige Studien uneinheitliche Ergebnisse hinsichtlich Änderungen von DTI-Parametern in der grauen Substanz bei der MS erbrachten, beschäftigten wir uns mit der Frage, ob kortikale MD- und FA-Veränderungen bei Patienten mit schubförmig-remittierender MS (RRMS) mithilfe optimierter DTI-Messtechniken zu detektieren sind, wie diese charakterisiert sind und wie sich diese im Kortex verteilen.
An der vorliegenden Studie nahmen 24 Patienten mit RRMS und 25 gesunde Kontrollprobanden teil. Der Schweregrad der Erkrankung wurde mithilfe des Expanded Disability Status Scale (EDSS) eingestuft.
Bei der MRT-Datenerfassung wurde eine optimierte DTI-Methode mit intrinsischer „Eddy-Current“-Kompensation verwendet. Die MD und die FA wurden für jeden Bildpunkt bestimmt. Kortikale Parameterwerte wurden ausgelesen und in Oberflächendatensätzen gespeichert. Es erfolgte ein oberflächenbasierter statistischer Gruppenvergleich. Kortikale Mittelwerte wurden für die MD und die FA bestimmt und zwischen den Gruppen verglichen.
Für Parameter mit nachgewiesenen globalen Gruppenunterschieden wurde die Korrelation mit dem klinischen Status (quantifiziert durch den EDSS) bestimmt.
Die Analyse kortikaler Mittelwerte zeigte eine Erhöhung der MD in der Patientengruppe. Die MD-Veränderungen waren räumlich ausgedehnt und es fanden sich Cluster mit erhöhten MD-Werten in der Patientengruppe, insbesondere in temporalen, okzipitalen und parietalen Regionen. Des Weiteren konnte eine signifikante positive Korrelation zwischen dem EDSS-Score und der kortikalen MD festgestellt werden. Außerdem ließen sich fokale FA-Erniedrigungen im Temporal- und Okzipitallappen nachweisen. Die MD quantifiziert das Ausmaß und die FA die Gerichtetheit der Diffusion.
Somit bietet die MD möglicherweise Hinweise auf die Intaktheit mikrostruktureller Barrieren und die FA auf die Integrität von Faserverbindungen. Unsere Ergebnisse könnten demnach darauf hinweisen, dass im Kortex von MS-Patienten der Abbau mikrostruktureller Barrieren räumlich ausgedehnter stattfindet als eine Störung axonaler Strukturen. Die Korrelation der MD mit dem klinischen Status legt die Möglichkeit der Quantifizierung klinisch relevanter kortikaler Gewebsveränderungen und somit eine mögliche Relevanz dieser Techniken für klinische Studien nahe.
The relationship between exhaustion and work engagement has received considerable attention during the past decades. Although the theoretical proposition exists that work engagement may increase exhaustion over time, previous research has been mixed. Drawing on the transactional stress model and applying latent growth modeling, we aim to provide a more comprehensive picture of the work engagement–exhaustion relationship over time. In two longitudinal studies, with four measurement points each, we found consistent evidence that a higher initial work engagement related to increased exhaustion over time. Consistent with our hypotheses, a higher initial work engagement also related to less initial exhaustion, and increases in work engagement related to decreases in exhaustion over time. However, contrary to our expectations, a higher initial exhaustion related to elevated work engagement over time. In conclusion, our findings suggest that engaged employees are less exhausted but face a higher risk of exhaustion over time. At the same time, exhausted employees are less engaged, but they have the potential to become more so over time. The theoretical and practical implications of these findings will be discussed in this paper.
Acinetobacter baumannii is an opportunistic pathogen, which has become a rising threat in healthcare facilities worldwide due to increasing antibiotic resistances and optimal adaptation to clinical environments and the human host. We reported in a former publication on the identification of three phopholipases of the phospholipase D (PLD) superfamily in A. baumannii ATCC 19606T acting in concerted manner as virulence factors in Galleria mellonella infection and lung epithelial cell invasion. This study focussed on the function of the three PLDs. A Δpld1-3 mutant was defect in biosynthesis of the phospholipids cardiolipin (CL) and monolysocardiolipin (MLCL), whereas the deletion of pld2 and pld3 abolished the production of MLCL. Complementation of the Δpld1-3 mutant with pld1 restored CL biosynthesis demonstrating that the PLD1 is implicated in CL biosynthesis. Complementation of the Δpld1-3 mutant with either pld2 or pld3 restored MLCL and CL production leading to the conclusion that PLD2 and PLD3 are implicated in CL and MLCL production. Mutant studies revealed that two catalytic motifs are essential for the PLD3-mediated biosynthesis of CL and MLCL. The Δpld1-3 mutant exhibited a decreased colistin and polymyxin B resistance indicating a role of CL in cationic antimicrobial peptides (CAMPs) resistance.
The genome of the halophilic archaeon Haloferax volcanii encodes more than 40 one-domain zinc finger µ-proteins. Only one of these, HVO_2753, contains four C(P)XCG motifs, suggesting the presence of two zinc binding pockets (ZBPs). Homologs of HVO_2753 are widespread in many euryarchaeota. An in frame deletion mutant of HVO_2753 grew indistinguishably from the wild-type in several media, but had a severe defect in swarming and in biofilm formation. For further analyses, the protein was produced homologously as well as heterologously in Escherichia coli. HVO_2753 was stable and folded in low salt, in contrast to many other haloarchaeal proteins. Only haloarchaeal HVO_2753 homologs carry a very hydrophilic N terminus, and NMR analysis showed that this region is very flexible and not part of the core structure. Surprisingly, both NMR analysis and a fluorimetric assay revealed that HVO_2753 binds only one zinc ion, despite the presence of two ZBPs. Notably, the analysis of cysteine to alanine mutant proteins by NMR as well by in vivo complementation revealed that all four C(P)XCG motifs are essential for folding and function. The NMR solution structure of the major conformation of HVO_2753 was solved. Unexpectedly, it was revealed that ZBP1 was comprised of C(P)XCG motifs 1 and 3, and ZBP2 was comprised of C(P)XCG motifs 2 and 4. There are several indications that ZBP2 is occupied by zinc, in contrast to ZBP1. To our knowledge, this study represents the first in-depth analysis of a zinc finger µ-protein in all three domains of life.
We report here the nuclear magnetic resonance 19F screening of 14 RNA targets with different secondary and tertiary structure to systematically assess the druggability of RNAs. Our RNA targets include representative bacterial riboswitches that naturally bind with nanomolar affinity and high specificity to cellular metabolites of low molecular weight. Based on counter-screens against five DNAs and five proteins, we can show that RNA can be specifically targeted. To demonstrate the quality of the initial fragment library that has been designed for easy follow-up chemistry, we further show how to increase binding affinity from an initial fragment hit by chemistry that links the identified fragment to the intercalator acridine. Thus, we achieve low-micromolar binding affinity without losing binding specificity between two different terminator structures.
Phytochrome photoreceptors operate via photoisomerization of a bound bilin chromophore. Their typical architecture consists of GAF, PAS and PHY domains. Knotless phytochromes lack the PAS domain, while retaining photoconversion abilities, with some being able to photoconvert with just the GAF domain. Therefore, we investigated the ultrafast photoisomerization of the Pr state of a knotless phytochrome to reveal the effect of the PHY domain and its “tongue” region on the transduction of the light signal. We show that the PHY domain does not affect the initial conformational dynamics of the chromophore. However, it significantly accelerates the consecutively induced reorganizational dynamics of the protein, necessary for the progression of the photoisomerization. Consequently, the PHY domain keeps the bilin and its binding pocket in a more reactive conformation, which decreases the extent of protein reorganization required for the chromophore isomerization. Thereby, less energy is lost along nonproductive reaction pathways, resulting in increased efficiency.
The structure and flexibility of RNA depends sensitively on the microenvironment. Using pulsed electron-electron double-resonance (PELDOR)/double electron-electron resonance (DEER) spectroscopy combined with advanced labeling techniques, we show that the structure of double-stranded RNA (dsRNA) changes upon internalization into Xenopus lævis oocytes. Compared to dilute solution, the dsRNA A-helix is more compact in cells. We recapitulate this compaction in a densely crowded protein solution. Atomic-resolution molecular dynamics simulations of dsRNA semi-quantitatively capture the compaction, and identify non-specific electrostatic interactions between proteins and dsRNA as a possible driver of this effect.
Purpose: Auditory functional MRI (fMRI) often uses silent inter-volume delays for stimulus presentation. However, maintaining the steady-state of the magnetization usually requires constant delays. Here, a novel acquisition scheme dubbed “pre-Saturated EPI using Multiple delays in Steady-state” (SEPIMS) is proposed, using spin saturation at a fixed delay before each volume to maintain steady-state conditions, independent of previous spin history. This concept allows for variable inter-volume delays and thus for flexible stimulus design in auditory fMRI. The purpose was to compare the signal stability of SEPIMS and conventional sparse EPI (CS-EPI). Methods: The saturation module comprises two non-selective adiabatic saturation pulses. The efficiency of the saturation and its effect on the SEPIMS signal stability is tested in vitro and in vivo. Results: Data show that SEPIMS yields the same signal stability as CS-EPI, even for extreme variations between inter-volume delay durations. However, dual saturation pulses are required to achieve sufficiently high saturation efficiency in compartments with long T1 values. Importantly, spoiler gradient pulses after the EPI readout have to be optimized to avoid eddy-current-induced image distortions. Conclusion: The proposed SEPIMS sequence maintains high signal stability in the presence of variable inter-volume durations, thus allowing for flexible stimulus design.
The C40A/C82A double mutant of barstar has been shown to undergo cold denaturation above the water freezing point. By rapidly applying radio-frequency power to lossy aqueous samples, refolding of barstar from its cold-denatured state can be followed by real-time NMR spectroscopy. Since temperature-induced unfolding and refolding is reversible for this double mutant, multiple cycling can be utilized to obtain 2D real-time NMR data. Barstar contains two proline residues that adopt a mix of cis and trans conformations in the low-temperature-unfolded state, which can potentially induce multiple folding pathways. The high time resolution real-time 2D-NMR measurements reported here show evidence for multiple folding pathways related to proline isomerization, and stable intermediates are populated. By application of advanced heating cycles and state-correlated spectroscopy, an alternative folding pathway circumventing the rate-limiting cis-trans isomerization could be observed. The kinetic data revealed intermediates on both, the slow and the fast folding pathway.
Background and Objective: Long-term tooth retention is the ultimate goal of periodontal therapy. Aim of this study was to evaluate tooth loss (TL) during 10 years of supportive periodontal therapy (SPT) in periodontal compromised patients and to identify factors influencing TL on patient level. Material and Methods: Patients were re-examined 120 ± 12 months after active periodontal therapy. TL and risk factors [smoking, initial diagnosis, SPT adherence, interleukin-1 polymorphism, cardiovascular diseases, age at baseline, bleeding on probing (BOP), change of practitioner, insurance status, number of SPT, marital and educational status] influencing TL on patient level were assessed. Results: One-hundred patients (52 female, mean age 65.6 ± 11 years) lost 121 of 2428 teeth (1.21 teeth/patient; 0.12 teeth/patient/y) during 10 years of SPT. Forty-two of these were lost for periodontal reasons (0.42 teeth/patient; 0.04 teeth/patient/y). Significantly more teeth were lost due to other reasons (P < .001). Smoking, baseline severity of periodontitis, non-adherent SPT, positive interleukin-1 polymorphism, marital and educational status, private insurance, older age at baseline and BOP, small number of SPT were identified as patient-related risk factors for TL (P < .05). Conclusion: During 120 ± 12 months of SPT, only a small number of teeth was lost in periodontally compromised patients showing the positive effect of a well-established periodontal treatment concept. The remaining risk for TL should be considered using risk-adopted SPT allocation.
The RHO gene encodes the G-protein-coupled receptor (GPCR) rhodopsin. Numerous mutations associated with impaired visual cycle have been reported; the G90D mutation leads to a constitutively active mutant form of rhodopsin that causes CSNB disease. We report on the structural investigation of the retinal configuration and conformation in the binding pocket in the dark and light-activated state by solution and MAS-NMR spectroscopy. We found two long-lived dark states for the G90D mutant with the 11-cis retinal bound as Schiff base in both populations. The second minor population in the dark state is attributed to a slight shift in conformation of the covalently bound 11-cis retinal caused by the mutation-induced distortion on the salt bridge formation in the binding pocket. Time-resolved UV/Vis spectroscopy was used to monitor the functional dynamics of the G90D mutant rhodopsin for all relevant time scales of the photocycle. The G90D mutant retains its conformational heterogeneity during the photocycle.
CD4+ T cell lymphopenia predicts mortality from Pneumocystis pneumonia in kidney transplant patients

Background: Pneumocystis jirovecii pneumonia (PcP) remains a life-threatening opportunistic infection after solid organ transplantation, even in the era of Pneumocystis prophylaxis. The association between risk of developing PcP and low CD4+ T cell counts has been well established. However, it is unknown whether lymphopenia in the context of post-renal transplant PcP increases the risk of mortality. Methods: We carried out a retrospective analysis of a cohort of kidney transplant patients with PcP (n = 49) to determine the risk factors for mortality associated with PcP. We correlated clinical and demographic data with the outcome of the disease. For CD4+ T cell counts, we used the Wilcoxon rank sum test for in-hospital mortality and a Cox proportional-hazards regression model for 60-day mortality. Results: In univariate analyses, high CRP, high neutrophils, CD4+ T cell lymphopenia, mechanical ventilation, and high acute kidney injury network stage were associated with in-hospital mortality following presentation with PcP. In a receiver-operator characteristic (ROC) analysis, an optimum cutoff of ≤200 CD4+ T cells/µL predicted in-hospital mortality, CD4+ T cell lymphopenia remained a risk factor in a Cox regression model. Conclusions: Low CD4+ T cell count in kidney transplant recipients is a biomarker for disease severity and a risk factor for in-hospital mortality following presentation with PcP.
Background: Cerebral O2 saturation (ScO2) reflects cerebral perfusion and can be measured noninvasively by near-infrared spectroscopy (NIRS). Objectives: In this pilot study, we describe the dynamics of ScO2 during TAVI in nonventilated patients and its impact on procedural outcome. Methods and Results: We measured ScO2 of both frontal lobes continuously by NIRS in 50 consecutive analgo-sedated patients undergoing transfemoral TAVI (female 58%, mean age 80.8 years). Compared to baseline ScO2 dropped significantly during RVP (59.3% vs. 53.9%, p < .01). Five minutes after RVP ScO2 values normalized (post RVP 62.6% vs. 53.9% during RVP, p < .01; pre 61.6% vs. post RVP 62.6%, p = .53). Patients with an intraprocedural pathological ScO2 decline of >20% (n = 13) had higher EuroSCORE II (3.42% vs. 5.7%, p = .020) and experienced more often delirium (24% vs. 62%, p = .015) and stroke (0% vs. 23%, p < .01) after TAVI. Multivariable logistic regression revealed higher age and large ScO2 drops as independent risk factors for delirium. Conclusions: During RVP ScO2 significantly declined compared to baseline. A ScO2 decline of >20% is associated with a higher incidence of delirium and stroke and a valid cut-off value to screen for these complications. NIRS measurement during TAVI procedure may be an easy to implement diagnostic tool to detect patients at high risks for cerebrovascular complications and delirium.
Cortical changes in epilepsy patients with focal cortical dysplasia: new insights with T2 mapping

Background: In epilepsy patients with focal cortical dysplasia (FCD) as the epileptogenic focus, global cortical signal changes are generally not visible on conventional MRI. However, epileptic seizures or antiepileptic medication might affect normal-appearing cerebral cortex and lead to subtle damage. Purpose: To investigate cortical properties outside FCD regions with T2-relaxometry. Study Type: Prospective study. Subjects: Sixteen patients with epilepsy and FCD and 16 age-/sex-matched healthy controls. Field Strength/Sequence: 3T, fast spin-echo T2-mapping, fluid-attenuated inversion recovery (FLAIR), and synthetic T1-weighted magnetization-prepared rapid acquisition of gradient-echoes (MP-RAGE) datasets derived from T1-maps. Assessment: Reconstruction of the white matter and cortical surfaces based on MP-RAGE structural images was performed to extract cortical T2 values, excluding lesion areas. Three independent raters confirmed that morphological cortical/juxtacortical changes in the conventional FLAIR datasets outside the FCD areas were definitely absent for all patients. Averaged global cortical T2 values were compared between groups. Furthermore, group comparisons of regional cortical T2 values were performed using a surface-based approach. Tests for correlations with clinical parameters were carried out. Statistical Tests: General linear model analysis, permutation simulations, paired and unpaired t-tests, and Pearson correlations. Results: Cortical T2 values were increased outside FCD regions in patients (83.4 ± 2.1 msec, control group 81.4 ± 2.1 msec, P = 0.01). T2 increases were widespread, affecting mainly frontal, but also parietal and temporal regions of both hemispheres. Significant correlations were not observed (P ≥ 0.55) between cortical T2 values in the patient group and the number of seizures in the last 3 months or the number of anticonvulsive drugs in the medical history. Data Conclusion: Widespread increases in cortical T2 in FCD-associated epilepsy patients were found, suggesting that structural epilepsy in patients with FCD is not only a symptom of a focal cerebral lesion, but also leads to global cortical damage not visible on conventional MRI. Background and Objectives: Patient blood (more accurately: haemoglobin, Hb) management (PBM) aims to optimize endogenous Hb production and to minimize iatrogenic Hb loss while maintaining patient safety and optimal effectiveness of medical interventions. PBM was adopted as policy for patients by the World Health Organization (WHO), and, all the more, should be applied to healthy donors. Materials and Methods: Observational data from 489 bone marrow (BM) donors were retrospectively analysed, and principles of patient blood management were applied to healthy volunteer BM donations. Results and Conclusion: We managed to render BM aspiration safe for donors, notably completely avoiding the collection of autologous blood units and blood transfusions through iron management, establishment and curation of high-yield aspiration technique, limitation of collection volume to 1·5% of donor body weight and development of volume prediction algorithms for the requested cell dose.
The exhaustive trichlorosilylation of hexachloro-1,3-butadiene was achieved in one step by using a mixture of Si2Cl6 and [nBu4N]Cl (7:2 equiv) as the silylation reagent. The corresponding butadiene dianion salt [nBu4N]2[1] was isolated in 36 % yield after recrystallization. The negative charges of [1]2− are mainly delocalized across its two carbanionic (Cl3Si)2C termini (α-effect of silicon) such that the central bond possesses largely C=C double-bond character. Upon treatment with 4 equiv of HCl, [1]2− is converted into neutral 1,2,3,4-tetrakis(trichlorosilyl)but-2-ene, 3. The Cl− acceptor AlCl3, induces a twofold ring-closure reaction of [1]2− to form a six-membered bicycle 4 in which two silacyclobutene rings are fused along a shared C=C double bond (84 %). Compound 4, which was structurally characterized by X-ray crystallography, undergoes partial ring opening to a monocyclic silacyclobutene 2 in the presence of HCl, but is thermally stable up to at least 180 °C.
The extraordinary desiccation resistance of the opportunistic human pathogen Acinetobacter baumannii is a key to its survival and spread in medical care units. The accumulation of compatible solute such as glutamate, mannitol and trehalose contributes to the desiccation resistance. Here, we have used osmolarity as a tool to study the response of cells to low water activities and studied the role of a potential inorganic osmolyte, K+, in osmostress response. Growth of A. baumannii was K+-dependent and the K+-dependence increased with the osmolarity of the medium. After an osmotic upshock, cells accumulated K+ and K+ accumulation increased with the salinity of the medium. K+ uptake was reduced in the presence of glycine betaine. The intracellular pools of compatible solutes were dependent on the K+ concentration: mannitol and glutamate concentrations increased with increasing K+ concentrations whereas trehalose was highest at low K+. After osmotic upshock, cells first accumulated K+ followed by synthesis of glutamate; later, mannitol and trehalose synthesis started, accompanied with a decrease of intracellular K+ and glutamate. These experiments demonstrate K+ uptake as a first response to osmostress in A. baumannii and demonstrate a hierarchy in the time-dependent accumulation of K+ and different organic solutes.
Genetic code expansion facilitates position-selective modification of nucleic acids and proteins

Transcription and translation obey to the genetic code of four nucleobases and 21 amino acids evolved over billions of years. Both these processes have been engineered to facilitate the use of non-natural building blocks in both nucleic acids and proteins, enabling researchers with a decent toolbox for structural and functional analyses. Here, we review the most common approaches for how labeling of both nucleic acids as well as proteins in a site-selective fashion with either modifiable building blocks or spectroscopic probes can be facilitated by genetic code expansion. We emphasize methodological approaches and how these can be adapted for specific modifications, both during as well as after biomolecule synthesis. These modifications can facilitate, for example, a number of different spectroscopic analysis techniques and can under specific circumstances even be used in combination.
Background: Visual exploration in autism spectrum disorder (ASD) is characterized by attenuated social attention. The underlying oculomotor function during visual exploration is understudied, whereas oculomotor function during restricted viewing suggested saccade dysmetria in ASD by altered pontocerebellar motor modulation. Methods: Oculomotor function was recorded using remote eye tracking in 142 ASD participants and 142 matched neurotypical controls during free viewing of naturalistic videos with and without human content. The sample was heterogenous concerning age (6–30 years), cognitive ability (60–140 IQ), and male/female ratio (3:1). Oculomotor function was defined as saccade, fixation, and pupil-dilation features that were compared between groups in linear mixed models. Oculomotor function was investigated as ASD classifier and features were correlated with clinical measures. Results: We observed decreased saccade duration (∆M = −0.50, CI [−0.21, −0.78]) and amplitude (∆M = −0.42, CI [−0.12, −0.72]), which was independent of human video content. We observed null findings concerning fixation and pupil-dilation features (POWER = .81). Oculomotor function is a valid ASD classifier comparable to social attention concerning discriminative power. Within ASD, saccade features correlated with measures of restricted and repetitive behavior. Conclusions: We conclude saccade dysmetria as ASD oculomotor phenotype relevant to visual exploration. Decreased saccade amplitude and duration indicate spatially clustered fixations that attenuate visual exploration and emphasize endogenous over exogenous attention. We propose altered pontocerebellar motor modulation as underlying mechanism that contributes to atypical (oculo-)motor coordination and attention function in ASD.
Acinetobacter baumannii is outstanding for its ability to cope with low water activities and therefore its adaptation mechanism to osmotic stress. Here we report on the identification and characterization of five different secondary active compatible solute transporters, belonging to the betaine-choline-carnitine transporter (BCCT) family. Our studies revealed two choline-specific and three glycine betaine-specific BCCTs. Activity of the BCCTs was differentially dependent to the osmolality: one choline and one betaine transporter were osmostress-independent. Addition of choline to resting cells of Acinetobacter grown in the presence of the co-substrate choline or with phosphatidylcholine as sole carbon source led to ATP synthesis in the wild type but not in the BCCT quadruple mutant. This indicates that the BCCTs are essential to transport the energy substrate choline. The role of the different BCCTs in osmostress resistance and in metabolic adaptation of A. baumannii to the human host is discussed.
We present the results of geochemical analysis of silver coinage issued by Rome and dated between the fourth and second century BCE, which are complemented by data of coinage issued by Carthage, the Brettii, and the Greek colony of Emporion. Each of these minting authorities represents one of the major parties involved in the struggle for hegemony in the fourth to second centuries BCE Western Mediterranean region. This study retraces how the metal supply shifts in response to the transforming power relations and how this change is related to Rome's rise to the virtually uncontested ruler of the region.
Sphingosine 1-phosphate (S1P) signaling influences numerous cell biological mechanisms such as differentiation, proliferation, survival, migration, and angiogenesis. Intriguingly, our current knowledge is based solely on the role of S1P with an 18-carbon long-chain base length, S1P d18:1. Depending on the composition of the first and rate-limiting enzyme of the sphingolipid de novo metabolism, the serine palmitoyltransferase, other chain lengths have been described in vivo. While cells are also able to produce S1P d20:1, its abundance and function remains elusive so far. Our experiments are highlighting the role of S1P d20:1 in the mouse central nervous system (CNS) and human glioblastoma. We show here that S1P d20:1 and its precursors are detectable in both healthy mouse CNS-tissue and human glioblastoma. On the functional level, we focused our work on one particular, well-characterized pathway, the induction of cyclooxygenase (COX)-2 expression via the S1P receptor 2 (S1P2). Intriguingly, S1P d20:1 only fairly induces COX-2 expression and can block the S1P d18:1-induced COX-2 expression mediated via S1P2 activation in the human glioblastoma cell line LN229. This data indicates that S1P d20:1 might act as an endogenous modulator of S1P signaling via a partial agonism at the S1P2 receptor. While our findings might stimulate further research on the relevance of long-chain base lengths in sphingolipid signaling, the metabolism of S1P d20:1 has to be considered as an integral part of S1P signaling pathways in vivo.
Molecular analysis of the ribosome recycling factor ABCE1 bound to the 30S post-splitting complex

Ribosome recycling by the twin-ATPase ABCE1 is a key regulatory process in mRNA translation and surveillance and in ribosome-associated protein quality control in Eukarya and Archaea. Here, we captured the archaeal 30S ribosome post-splitting complex at 2.8 Å resolution by cryo-electron microscopy. The structure reveals the dynamic behavior of structural motifs unique to ABCE1, which ultimately leads to ribosome splitting. More specifically, we provide molecular details on how conformational rearrangements of the iron–sulfur cluster domain and hinge regions of ABCE1 are linked to closure of its nucleotide-binding sites. The combination of mutational and functional analyses uncovers an intricate allosteric network between the ribosome, regulatory domains of ABCE1, and its two structurally and functionally asymmetric ATP-binding sites. Based on these data, we propose a refined model of how signals from the ribosome are integrated into the ATPase cycle of ABCE1 to orchestrate ribosome recycling.
The mammalian target of rapamycin and the integrated stress response are central cellular hubs regulating translation upon stress. The precise proteins and pathway specificity of translation targets of these pathways remained largely unclear. We recently described a new method for quantitative translation proteomics and found that both pathways control translation of the same sets of proteins.
Minimal residual disease (MRD) is the strongest predictor of relapse in B-cell precursor acute lymphoblastic leukemia (BCP-ALL). In BLAST study (NCT01207388), adults with BCP-ALL in remission with MRD after chemotherapy received blinatumomab, a CD19 BiTE® immuno-oncotherapy, 15 µg/m2/day for up to four 6-week cycles (4 weeks continuous infusion, 2 weeks off). Survival was evaluated for 110 patients, including 74 who received HSCT in continuous complete remission. With a median follow-up of 59·8 months, median survival (months) was 36·5 (95% CI: 22.0–not reached [NR]). Median survival was NR (29.5–NR) for complete MRD responders (n = 84) and 14.4 (3.8–32.3) for MRD non-responders (n = 23; p = 0.002); after blinatumomab and HSCT, median survival was NR (25.7–NR) (n = 61) and 16.5 (1.1–NR) (n = 10; p = 0.065), respectively. This final analysis suggests complete MRD response during blinatumomab treatment is curative. Post-hoc analysis of study data suggests while post blinatumomab HSCT may be beneficial in appropriate patients, long-term survival without HSCT is also possible.
Introduction: Dravet syndrome (DS), a prototypic developmental and genetic epileptic encephalopathy (DEE), is characterized by an early onset of treatment-refractory seizures, together with impairments in motor control, behavior, and cognition. Even with multiple conventional anti-epileptic drugs, seizures remain poorly controlled, and there has been a considerable unmet need for effective and tolerable treatments. Areas covered: This targeted literature review aims to highlight recent changes to the therapeutic landscape for DS by summarizing the most up-to-date, evidence-based research, including pivotal data from the clinical development of stiripentol, cannabidiol, and fenfluramine, which are important milestones for DS treatment, together with the latest findings of other pharmacotherapies in development. In phase III, double-blind, placebo-controlled randomized controlled trials stiripentol, cannabidiol, and fenfluramine have shown clinically relevant reductions in convulsive seizure frequency, and are generally well tolerated. Stiripentol was associated with responder rates (greater than 50% reduction in convulsive seizure frequency) of 67%-71%, when added to valproic acid and clobazam; cannabidiol was associated with responder rates of 43%-49% (48%-63% in conjunction with clobazam), and fenfluramine of 54%-68% across studies. Therapies in development include soticlestat, ataluren, verapamil, and clemizole, with strategies to treat the underlying cause of DS, including gene therapy and antisense oligonucleotides beginning to emerge from preclinical studies. Expert opinion: Despite the challenges of drug development in rare diseases, this is an exciting time for the treatment of DS, with the promise of new efficacious and well-tolerated therapies, which may pave the way for treatment advances in other DEEs.
Im vorliegenden Report wurde anhand von Daten aus dem IAB-Betriebspanel ein Schlaglicht auf die Innovationskraft hess- ischer Betriebe geworfen. Nur wenige Betriebe sind, in der Regel in Kooperation mit anderen Betrieben und Hochschulen, explizit mit Forschung und Entwicklung beschäftigt. Dennoch investiert ein großer Teil, nämlich ein Drittel der hessischen Betriebe, in die Weiterentwicklung und Verbesserung von Pro- dukten und Dienstleistungen. Acht Prozent der Betriebe haben darüber hinaus den Produktionsprozess verbessert. Diese Anteile haben sich in den letzten Jahren kaum verändert. 17 Prozent der Betriebe haben ihr Angebotsspektrum zuletzt erweitert, und immerhin fünf Prozent entwickeln gänzlich neue Produkte oder Dienstleistungen.Wie gezeigt wurde, konnten im Geschäftsjahr 2018 jedoch nicht sämtliche geplanten Innova-tionen umgesetzt werden. Jeder zwanzigste Betrieb gab an, dass dies nicht möglich war. Hauptgrund ist der Mangel an Fach-kräften. Daneben spielt aber auch die wirtschaftliche Situation der Betriebe eine Rolle: Gemäß der Ergebnisse des IAB-Betriebspanels befinden sich unter den Betrieben, die Produkte, Dienstleistungen oder betriebsinterne Prozesse weiterentwickelt haben, deutlich mehr mit einem positiven Jahresergebnis als solche mit einem negativen Jahresergebnis.
Der Einsatz von Robotern nimmt in den hessischen Betrieben zu. Zwar ist er, außer im Verarbeitenden Gewerbe, derzeit noch wenig verbreitet; die Zahl der Neuanschaffungen ist jedoch in den vergangenen Jahren – gerade auch in kleineren Betrieben – stark gestiegen. Die Corona-Krise könnte diese Entwicklung begünstigen, wenn es aufgrund der Unsicherheiten, die mit globalen Lieferketten verbunden sind, zu einer Verlagerung von Produktionsschritten nach Deutschland kommen sollte. Dies könnte mit einem stärkeren Einsatz von Robotern einhergehen, so die Expertin für Internationale Wirtschaftsbeziehungen Prof. Dr. Dalia Marin im Interview mit der Frankfurter Allgemeinen Zeitung (FAZ vom 01.03.2020). Andererseits könnten den Betrieben die Mittel für größere Investitionen aufgrund des Einbruchs der Wirtschaft fehlen. Auffällig ist, dass die Großbetriebe bei allen Auswertungen im vorliegenden Report als innovationskräftiger erscheinen als die kleinen und mittleren Betriebe. Sie investieren häufiger in Forschung und Ent- wicklung sowie in Produkt-, Dienstleistungs- und Verf- ahrensverbesserungen. Dabei ist jedoch auch anzumerken, dass jeweils ein Drittel der Kleinstbetriebe, der kleinen und mittleren Betriebe an der Verbesserung und Weiterentwicklung ihrer Produkte und Dienstleistungen arbeitet. Es kann hier also nicht von einem klaren Trend, dass die Innovativität mit steigender Betriebsgröße zunimmt, gesprochen werden. Der Innovationsforscher Everett Rogers vermutet, dass die Betriebsgröße eine „Stellvertretervariable“ ist, die leicht zu messen ist, die aber verdeckt, dass vielmehr Faktoren wie eine positive Einstellung von Führungskräften gegenüber Veränder- ungen, eine wenig zentralistische Betriebsstruktur und eine Betriebskultur mit kurzen, auch hierarchieübergreifenden Kommunikationswegen einen entscheidenden Einfluss auf die Innovativität eines Betriebs haben (Rogers 2003: 409ff.). Solche Voraussetzungen lassen sich in Betrieben aller Größenklassen finden. Die Ausstattung der Betriebe mit Hochgeschwindigkeits-Internet als eine technische Voraussetzung für Innovationen ist grundsätzlich als gut anzusehen, allerdings hat die Zufriedenheit damit abgenommen.
Womöglich ist die gestiegene Unzufriedenheit ein An zeichen für höhere Ansprüche. Im Zuge der Corona-Krise dürfte die Bedeutung von Hochgeschwindigkeits-Internet zugenommen haben; viele Beschäftigte arbeiten zuhause und greifen von dort bspw. auf Betriebsserver zu. Insbesondere im Wirtschaftszweig Handel und Reparatur ist ein recht hoher Anteil der Betriebe unzu- frieden mit der verfügbaren Bandbreite, also in einem Bereich, in dem der Druck zur Online-Vermarktung gerade durch die Corona-Krise wächst. Insgesamt zeigen die Auswertungen jedoch, dass die Innovationskraft der hessischen Betriebe als stabil zu bewerten ist. Im Vergleich zu den Ergebnissen aus dem Befragungsjahr 2017 ergeben sich keine klaren Hinweise einer zunehmenden oder abnehmenden Innovativität. Auffällig ist die ausgeprägte Kooperationsbereitschaft hessischer Betriebe mit Hochschulen und außerhochschulischen Forschungseinrichtungen, die zur Zukunftsfähigkeit der Betriebe beiträgt. Andererseits erweist sich der Fachkräftemangel bereits heute in einigen Betrieben als Innovationsbremse, und personelle Engpässe dürften in den kommenden Jahren an Bedeutung gewinnen. Aktivitäten zur Sicherung des Fachkräftebedarfs sind auch deshalb zu intensivieren, weil damit die Innovationsfähigkeit der hessischen Wirtschaft gestärkt wird
Im vorliegenden Report wurde anhand von Daten aus dem IAB-Betriebspanel ein Schlaglicht auf die Aktivitäten hessischer Betriebe zur Fachkräftebindung geworfen. Beispielhafte Anreize für die Beschäftigten, im Betrieb zu verbleiben, stellen das Angebot betrieblich geförderter Weiterbildungen, die Übernahme von Auszubildenden und die Beteiligung an Gewinn, Erfolg und Kapital eines Betriebs dar. Die Auswertungen zeigen, dass in Hessen gut die Hälfte aller Betriebe über Weiterbildungs- angebote Anreize zur Fachkräftebindung setzt. Allerdings liegt dieser Anteil noch etwas unter dem Anteil der Betriebe in Westdeutschland. Größere Betriebe sind praktisch alle in der Weiterbildung aktiv, jedoch profitiert davon ein geringerer Teil der Belegschaften als in kleineren Betrieben. Insbe- sondere Dienstleistungsbetriebe scheinen Weiterbildung syste-matisch zur Fachkräftebindung zu nutzen. In den allermeisten Betrieben gilt die Weiterbildungszeit als Arbeitszeit, und die Betriebe übernehmen die Weiterbildungskosten vollständig. Beides ist wichtig, damit Weiterbildung für die Beschäftigten attraktiv ist und tatsächlich als Anreiz zur Bindung an den Betrieb wirksam werden kann. Was die Weiterbildungsformate angeht, setzen die hessischen Betriebe nach wie vor insbe- sondere auf externe Kurse, Lehrgänge und Seminare. Weniger verbreitet scheinen arbeitsintegrierte Weiterbildungen zu sein. Insbesondere das selbstgesteuerte Lernen mit Medien wird erst bei gut einem Viertel der hessischen Betriebe genutzt. Allerdings nimmt der Anteil der Betriebe, die diese Formate einsetzen, zu. Es wird interessant sein, im Laufe dieses Jahres und darüber hinaus zu verfolgen, ob und wie sich die Arbeitserfahrungen während der Corona-Krise auf eine Ausweitung des selbstgesteuerten digitalen Lernens auswirken. Die Übernahme von Auszubildenden ist das wichtigste Instrument zur Fachkräftebindung, welches über alle Betriebsgrößen hinweg einen hohen Verbreitungsgrad in den hessischen Betrieben hat. Besonders stark scheint es in Kleinst- und Großbetrieben zum Einsatz zu kommen. Die zunehmend größere Bedeutung von Über- nahmeangeboten zur Fachkräftebindung zeigt sich auch in steigenden Übernahme- quoten. Dagegen setzen nur wenige Betriebe Gewinn-, Erfolgs- und Kapitalbeteiligung der Beschäftigten als gezielte Anreiz- systeme zur Mitarbeiter- bindung ein. Am ehesten unterbreiten Großbetriebe ihren Beschäftigten entsprechende Angebote. Der Report zeigt am Beispiel von drei Formen betrieblicher An- reize, dass bereits viele der hessischen Betriebe entsprechende Impulse setzen. Je nach Betriebsgröße und Branchenzugehörigkeit unterscheidet sich die Intensität der jeweiligen betrieblichen Aktivitäten. Interessant ist aber, dass es nicht nur Großbetriebe sind, die gezielt Anreize zur Fachkräftebindung setzen, sondern auch mittelgroße und selbst Kleinstbetriebe.
Seit 1996 wird das IAB-Betriebspanel jährlich bundesweit im Auftrag des Instituts für Arbeitsmarkt- und Berufsforschung der Bundesagentur für Arbeit (IAB) durchgeführt. In den Monaten Juli bis Oktober 2019 wurden die Betriebe erneut zu betrieblichen Bestimmungsgrößen der Beschäftigung befragt. Grundlage der Auswertungen für Hessen sind Befragungsdaten von insgesamt 976 Betrieben im Bundesland. Die befragten Betriebe repräsentieren die Grundgesamtheit der rund 162.000 hessischen Betriebe mit mindestens einem sozialversicherungspflichtig Beschäftigten. Im Folgenden werden die zentralen Ergebnisse der letztjährigen Befragungswelle dargestellt.
Über den Auswertungen der 2019er Befragung im IAB-Betriebspanel Hessen steht das Rahmenthema „Zukunftsorientierung hessischer Betriebe“. Der 1. Report beschäftigt sich mit dem Engagement der hessischen Betriebe in der betrieblichen Ausbildung, der 2. Report mit Anreizen zur Fachkräftebindung und der 3. Report mit dem Innovation- sgeschehen in den Betrieben. Im vorliegenden 4. Report greifen wir die Themenbereiche der Vorgängerreporte wieder auf und untersuchen, ob sich Betriebe mit jüngerer und solche mit älterer Belegschaft hinsichtlich der Rekrutierung von Fachkräften, bei den Aktivitäten zur Fachkräftebindung und der Umsetzung von Innovationen unterscheiden. Vor dem Hintergrund des immensen altersbedingten Ersatzbedarfs, welcher in Hessen laut Prognosen aus dem Projekt „regio pro“ bereits im Jahr 2024 insgesamt 19 Prozent des Beschäftigtenstandes des Jahres 2017 entsprechen wird (Demireva et al. 2019: 13), ist es sinnvoll, betriebliche Aktivitäten genauer nach Altersstruktur in den Blick zu nehmen (siehe auch Ossola-Haring et al. 2019: 268). Laut den Prognosen wird der altersbedingte Ersatzbedarf nämlich immensen Einfluss auf den Arbeitskräftebedarf in Hessen haben (Demireva et al. 2019: 19ff.). Mit dem Ausscheiden der sogenannten Baby Boomer aus dem Erwerbsleben könnte sich der Mangel an Fachkräften massiv verschärfen. Zudem geht den Betrieben wichtiges implizites Erfahrungswissen verloren, welches wesentlich zu einem reibungslosen Ablauf betrieblicher Prozesse beiträgt (Armutat 2018: 25f.; Piech 2020). Solches Wissen kann nicht ohne weiteres vom Arbeitsmarkt bezogen, geschweige denn schnell aufgebaut werden. Die Innovationskraft eines Betriebs kann durch den Verlust betriebsinternen Wissens stark beeinträchtigt werden. Zu erwarten ist, dass viele Betriebe auf die Heraus-forderung alternder Belegschaften bereits reagieren, indem sie unterschiedliche Strategien einsetzen: So könnten sie bewusst in die betriebliche Ausbildung investieren, um die Belegschaft zu verjüngen und den Transfer von Wissen an junge Fachkräfte zu ermöglichen. Darüber hinaus könnten Betriebe versuchen, ältere Beschäftigte länger im Betrieb zu halten. Mit der sukzessiven Erhöhung des gesetzlichen Rentenein- trittsalters ist ein Anreiz geschaffen, dass Beschäftigte länger berufstätig bleiben: Wer früher in den Ruhestand geht, erhält spürbar weniger Rente als jene, die länger arbeiten (Troger 2019a: 23). Nicht selten versuchen Betriebe allerdings auch weiterhin, ältere Beschäftigte in Frührente zu schicken, um sie durch jüngere (und günstigere) Arbeitskräfte zu ersetzen oder um bei wirtschaftlichen Engpässen Personal abzubauen (ebd.). Dies trifft durchaus auch den Wunsch vieler älterer Beschäftigter, die gerne lieber früher als später in Rente gehen würden, sofern die Höhe ihrer Bezüge dafür ausreicht (ebd.). Älteren Menschen haften immer noch besonders viele negative Vorurteile – wie beispielsweise eine eingeschränkte Leistungsfähigkeit – an (Schröder-Kunz 2019: 178ff.). Eine Folge solcher Vorurteile ist unter anderem, dass ältere Beschäftigte weit weniger an betrieblicher Weiterbildung teilnehmen als jüngere (BMBF 2019: 36ff.). Eine strategische Personalplanung, die dabei hilft, auf kommende Herausforderungen zu reagieren und zukunftsfähig zu bleiben, wird nicht in allen Betrieben umgesetzt. Gerade kleine und mittlere Unternehmen (KMU) tun sich schwer damit, ihren genauen Personalbedarf zu beziffern (Immerschitt & Stumpf 2019: 61). Vielerorts fehlen differenzierte Daten und Kennzahlen, beispielsweise zum Neu- oder Ersatzbedarf an Arbeitskräften je nach Qualifikation (Treier 2019: 94).
Im vorliegenden Bericht untersuchen wir anhand von Daten aus dem IAB-Betriebspanel, inwieweit Betriebe mit älterer Belegschaft anders agieren als Betriebe mit jüngerer Belegschaft. Dazu wird im ersten Kapitel zunächst ein Überblick über die Altersstruktur in hessischen Betrieben, differenziert nach Wirtschaftszweigen und Betriebsgrößen, gegeben. Die Betriebe werden einem von drei Betriebstypen zugeordnet: Unterschieden werden Betriebe mit vergleichsweise junger, Betriebe mit vergleichsweise Betriebliche Aktivitäten in Relation zur Altersstruktur der Beschäftigten alter und Betriebe mit mittelalter Belegschaft. Die Auswertungs- ergebnisse werfen an einigen Stellen womöglich mehr Fragen auf als sie beantworten können, und natürlich bilden Gleichaltrige nicht zwangsläufig eine homogene Gruppe, sondern unterscheiden sich in vielerlei Hinsicht voneinander (Schröder-Kunz 2019: 177). Der explorative Charakter des vorliegenden Reports ist allerdings bewusst gewählt, da es wenig Literatur zum betrieblichen Verhalten nach Altersstruktur der Beschäftigten gibt. Im zweiten Kapitel wird beschrieben, ob und wie sich diese drei Typen von Betrieben hinsichtlich der Umsetzung von Innovationen unterscheiden. Im dritten Kapitel werden betriebliche Aktivitäten zur Rekrutierung von Fachkräften verglichen und im vierten Kapitel Aktivitäten zur Fachkräft- ebindung. Der Bericht endet mit einem zusammenfassenden Resümee im fünften Kapitel.
The Spanish reproductive bioeconomy has bloomed in the last few decades. There are now over three hundred fertility clinics in Spain, which has become one of the main destinations for what is often called “reproductive tourism” in the European context. The phenomenon of assisted reproduction has been extensively studied within English-speaking countries of the global North, but not so much in the cluster of Spanish-speaking countries, with a few interesting exceptions. Following the invitation to collaborate in this special issue around reproduction in Latin America and Spain, we offer an analysis of how Spanish oocyte provision and domestic work function as part of global care chains (GCC). We will compare the results of two major projects: one focusing on domestic work and the other on egg donation programs, both in Spain. We will introduce different perspectives around care and GCC, discussing how transference of oocytes can be viewed as a type of feminized labor involving affective-care work, clinical work, and biological work. The framework of GCC, a concept used to unpack unjust power relations embedded in transferences of care in current neoliberal and globalized socio-economic arrangements, can help to enable a conversation on how transferences of reproductive capacity might be reinforcing the stratification of reproduction.
Support for innovative on-street parking policies: empirical evidence from an urban neighborhood

Municipalities and planners often hesitate to implement restrictive parking policies because residents regularly oppose any changes to on-street parking space. Residential parking is one key factor of parking management because its location and availability influence a household's car ownership and use. Moreover, as more residents regularly use other means of transportation that need space and infrastructure in the urban realm, and as parking takes up a vast amount of land, municipalities are considering the reuse of on-street parking space for other purposes. As public acceptability is a precondition for the successful implementation of a proposed policy, our empirical analysis investigates to what extent residents support restrictive and demand-oriented on-street parking policies in the dense, highly urbanized neighborhood of Frankfurt-Bornheim, Germany (N = 1027). Surprisingly, despite some variations, the majority of the residents in our survey are in favor of the policy options suggested. Support for the demand-oriented policies (extension of bicycle infrastructure, improved sharing supply and mobility hubs, neighborhood garages, and improved public transit supply) is higher than the acceptance of the restrictive policies, and of policies that are a combination of restrictive and demand-oriented policies. However, surprisingly, a majority is still in favor of many of these (extension of parking fees and parking restrictions, and reuse of parking space for better livability). Furthermore, we classify residents who live in a household with private cars into the stage model of self-regulated behavior change to analyze their intention towards a reduction in private car use. Results from linear regression analyses indicate that residents who have intentions to change their behavior towards car use reduction assess the policy options more similarly to car-free households and regular bike users, and not like the other car-owning households. The findings suggest that while the residents support financial-related policies the least, they are more receptive towards parking policies than policy makers and planners assume if the reuse of parking lots creates space for other users or if it increases the quality of life, for instance, by adding bike lanes, wider sidewalks or greenery. Hence, a combination of restrictive and demand-oriented on-street parking policies results in high acceptance among residents, and the communication from municipalities regarding the implementation of the different policies needs to vary depending on the kind of household.
Die vorliegende Arbeit beschäftigt sich mit der außerhäuslichen Alltagsmobilität älterer Menschen, die eine zentrale Schlüsselfunktion in der Erhaltung von Lebensqualität und Gesundheit besonders im höheren Lebensalter einnimmt. Außerhäusliche Alltagsmobilität vollzieht sich stets in einem räumlichen Umweltausschnitt und kann aus ökogerontologischer Perspektive als Ergebnis eines gelungenen Person-Umwelt-Austauschs verstanden werden. Inwiefern psychologische Ressourcen im Sinne mobilitätsspezifischer Einstellungen zum Verständnis von zielgerichteter und habitualisierter Alltagsmobilität älterer Menschen beitragen können, ist Gegenstand der vorliegenden Arbeit. Altersspezifische, mobilitätsrelevante Einstellungen im außerhäuslichen Kontext werden sowohl in der sozialwissenschaftlichen Mobilitäts- und Alternsforschung als auch in der Praxis, etwa im Rahmen einer altersgerechten Stadtgestaltung, bislang noch zu wenig berücksichtigt. Die vorliegende Arbeit reagiert auf dieses Forschungsdesiderat, indem sie mobilitätsspezifische Einstellungen im höheren Lebensalter konzeptuell beschreibt, in den Kontext ökogerontologischer Theorien einbettet und ihre Bedeutung für den Erhalt eines aktiven und gelingenden Alterns untersucht. Im Rahmen der Dissertation wurde zunächst auf der Basis klassischer und neuer ökogerontologischer Modelle das Konstrukt der mobilitätsbezogenen Handlungsflexibilität und Routinen (MBFR) konzeptuell entwickelt. MBFR umfasst einerseits die individuelle Überzeugung, das eigene Mobilitätsverhalten an Herausforderungen außer Haus anpassen zu können (FLEX) und andererseits die Präferenz für mobilitätsbezogene Alltagsroutinen (ROU). Daraufhin wurde ein standardisiertes Messinstrument zur Erfassung des MBFR-Konzepts entwickelt, optimiert und hinsichtlich seiner psychometrischen Qualität untersucht. Die Formulierung der Testitems erfolgte in Anlehnung an bereits existierende Fragebögen zu verwandten Konstrukten. In der vorwiegend online durchgeführten Pilotstudie (Penger & Oswald, 2017) wurden die Items mittels explorativer Faktorenanalysen hinsichtlich ihrer dimensionalen Struktur untersucht. Die Stichprobe umfasste 265 Personen im Alter von 65 Jahren oder älter. Die Analysen des MBFR-Instruments ergaben nach Ausschluss von Items mit niedrigen und nicht eindeutigen Ladungen drei substanzielle Faktoren. Die Items der ersten Dimension bildeten die Überzeugung ab, flexibel mit personenbezogenen, altersassoziierten Herausforderungen (z. B. Schwierigkeiten im Gehen oder auf eine Gehhilfe angewiesen sein) umgehen zu können, um außerhäuslich mobil zu sein. Die Items der zweiten Dimension erfassten die Überzeugung, flexibel mit herausfordernden außerhäuslichen Umweltbedingungen (z. B. eine verlegte Haltestelle oder ein schlechter Zustand der Gehwege) umgehen zu können. Items, die auf den dritten Faktor luden, bildeten die Neigung zu Routinen im Mobilitätsalltag ab, z. B. bekannte Wege beizubehalten oder bei der Ausübung von außerhäuslichen Aktivitäten vertraute Orte aufzusuchen. Während die ersten beiden Faktoren mobilitätsbezogene Handlungsflexibilität (FLEX) messen, werden im dritten Faktor habitualisierte Verhaltensweisen (ROU) erfasst. Alle drei Faktoren wiesen eine akzeptable Reliabilität auf. Auf Basis von Rückmeldungen der Studienteilnehmer:innen wurde das MBFR-Instrument anschließend sprachlich angepasst und gekürzt. Der modifizierte Fragebogen wurde daraufhin in der empirischen Studie „MOBIL bleiben in Stuttgart“ (MBIS) eingesetzt. Dabei sollte die Frage beantwortet werden, ob das finale MBFR-Instrument die zugrundeliegenden Konstrukte valide und reliabel erfasst und die Testwerte somit ausreichende Gültigkeit hinsichtlich faktorieller, Konstrukt- und Kriteriumsvalidität bei älteren Menschen im urbanen Raum aufweisen (Penger & Conrad, eingereicht). Es wurden insgesamt 211 privatwohnende Stuttgarter:innen ab 65 Jahren in persönlichen Interviews und mithilfe eines 7-tägigen Wegetagebuchs zu verschiedenen Aspekten ihrer Mobilität im Wohnumfeld befragt. Statistische Analysen auf latenter Ebene erfolgten mittels Strukturgleichungsmodellen. Bivariate Zusammenhänge und Subgruppenanalysen wurden mittels Korrelations- und Regressionsanalysen berechnet. Die dreifaktorielle Struktur des MBFR-Fragebogens konnte im konfirmatorischen Modell empirisch bestätigt werden. Zudem fiel die interne Konsistenz aller drei Faktoren gut aus. Zusammenhänge zu konstruktverwandten Merkmalen – wie allgemeine und mobilitätsspezifische Einstellungen – deuten darauf hin, dass das MBFR-Instrument ausreichend konvergente Validität aufweist. Analysen auf latenter Ebene ergaben, dass Befragte durchschnittlich mehr außerhäusliche Wege zurückzulegten, wenn sie in stärkerem Maße überzeugt waren, flexibel auf mobilitätsbezogene Herausforderungen reagieren zu können (FLEX). Weiterhin ließen sich positive Zusammenhänge zwischen FLEX und der erlebten Selbstständigkeit sowie dem subjektiven Wohlbefinden aufzeigen. Die Befunde belegen somit hinreichende Übereinstimmungsvalidität der Testwerte. Differenzierte Analysen machten darüber hinaus deutlich, dass FLEX vor allem bei Befragten mit Mobilitätseinschränkungen bedeutsam zur Vorhersage des außerhäuslichen Mobilitätsverhaltens beitrug. ...
Die Befragung der Betriebe in der Region Rhein-Main liefert folgende zentrale Erkenntnisse: 61 Prozent der Betriebe und damit mehr als in den Jahren 2014 und 2017 spüren derzeit einen Arbeitskräfterückgang. Mit steigender Betriebsgröße steigt der Anteil der Betriebe, die einen Arbeitskräfterückgang wahrnehmen. Im Vergleich der Wirtschaftszweige nehmen Betriebe des Baugewerbes in besonderem Ausmaß einen Arbeitskräfterückgang wahr. Etwa ein Viertel der Betriebe hat Ausbildungsplätze für das beginnende Ausbildungsjahr angeboten. Mit steigender Betriebsgröße steigt der Anteil der Betriebe, die Ausbildungsplätze anbieten. In vielen Betrieben können nicht sämtliche angebotene Ausbildungsplätze besetzt werden. Insbesondere Kleinstbetriebe haben Schwierigkeiten bei der Stellenbesetzung. Die Hälfte der Betriebe macht Zugeständnisse bei der Besetzung von Ausbildungsplätzen. In etwa jedem dritten Betrieb kommt es zu Ausbildungsabbrüchen. In Großbetrieben erscheint das Abbruchrisiko größer als in kleinen und Kleinstbetrieben. Jeder fünfte Betrieb mit Ausbildungsplätzen wirbt nicht aktiv um Auszubildende. Großbetriebe sind in diesem Bereich aktiver als kleinere Betriebe
Um die Entgeltgleichheit zwischen Frauen und Männern gezielt und nachhaltig zu fördern, bedarf es einer hohen Transparenz, so dass passgenau Aktivitäten initiiert werden können. Die Neuauflage des Hessischen Lohnatlas knüpft an die Analysen von Entgeltdaten der Wohnbevölkerung zum Stand 2015 an (erstmals im Jahr 2017 vorgestellt) und zeigt transparent auf, wie sich die Lohnlücken bis zum Jahr 2018 verändert haben. Bei diesen Analysen finden ausschließlich sozialversicherungspflichtige Vollzeitbeschäftigte Berücksichtigung. Die Teilzeitbeschäftigten können in den Analysen nicht einbezogen werden, da keine Informationen zum Stundenumfang ihrer Teilzeitbeschäftigung vorliegen und damit nicht erfasst werden kann, in welchem Maße Entgeltunterschiede auch durch unterschiedliche Stundenzahlen zustande kommen. Zum Erfassen der Bruttomonatsentgelte wird auf öffentliche Daten (vor allem Stichtagsdaten 31.12.) zurückgegriffen, die vom Statistikservice Südwest der Regionaldirektion Hessen der
Bundesagentur für Arbeit und dem Hessischen Statistischen Landesamt stammen. Bei diesen Daten handelt es sich um sogenannte Populationsdaten. Dies bedeutet, dass alle Einwohner*innen der Kreise und kreisfreien Städte in Hessen, zu denen entsprechende Entgeltdaten vorliegen, in den Analysen einbezogen werden. Die Analysen werden für Hessen sowie für jeden Kreis und jede kreisfreie Stadt durchgeführt. Damit können auf allen Ebenen noch bestehende Entgeltunterschiede zwischen Frauen und Männern in der jeweiligen Wohnbevölkerung erfasst werden. Die damit geschaffene Transparenz schafft eine wichtige Grundlage zum Diskurs über Ansätze, die die Förderung der Entgeltgleichheit im Land, aber auch vor Ort in der Regionen begünstigen können. Mit der Neuauflage des Hessischen Lohnatlas wird darüber hinaus erstmals die Entgeltlage von Frauen und Männern in den Betrieben vor Ort, also in den Kreisen und kreisfreien Städten, erfasst. Damit werden Aussagen zur Lage der Entgeltgleichheit in der lokalen Wirtschaft möglich. Mit dem Fokus auf die Betriebe vor Ort, sind auch jene Beschäftigte einbezogen, die täglich in den Kreis oder die kreisfreie Stadt zur Erwerbsarbeit einpendeln. Berücksichtigung findet hier, wie bereits oben ausgeführt, ausschließlich die Entgeltlage der in den Betrieben in sozialversicherungspflichtiger Vollzeit Beschäftigten. Basis der Analysen bilden Daten zu den durchschnittlichen Tagesbruttoentgelten im Jahr 2017 aus verschiedenen Stichproben, die durch das Institut für Arbeitsmarkt- und Berufsforschung (IAB) zur Verfügung gestellt wurden. Anders als bei den obigen Populationsdaten können die Befunde aus der Analyse der Stichproben nicht einfach verallgemeinert werden. Die Ergebnisse sind als Trends zu interpretieren. Von besonderem Interesse ist bei diesen Analysen, in welchem Maße sich Merkmale wie beispielsweise die Betriebsgröße, die
Geschlechterzusammensetzung der sozialversicherungspflichtig Beschäftigten in den Betrieben oder die Branchenzugehörigkeit der Betriebe auf die Entgeltlücken zwischen Frauen und Männern auswirken können. Die mit diesen Analysen erzeugte Transparenz kann insbesondere den Akteuren der Wirtschaft deutlich machen, wo noch Handlungsbedarfe liegen, um die Entgeltgleichheit von Frauen und Männern in den Betrieben des Landes zu verbessern.
Im Folgenden erfolgt zunächst die Darstellung aller Befunde auf der Ebene des Landes Hessen. Im Anschluss wird für jeden Kreis und jede kreisfreie Stadt ein Dossier vorgelegt. Damit lassen sich Unterschiede und Gemeinsamkeiten zwischen den Kreisen und den kreisfreien Städten sowohl hinsichtlich der Entgeltlage der Wohnbevölkerung als auch in Bezug auf die Entgeltlage in den Betrieben vor Ort erfassen.
Für die Zukunftsfähigkeit des Mittelstandes ist es von großer Bedeutung, dass Betriebe über ausreichend und gut qualifizierte Fach- und Arbeitskräfte verfügen. Die Roadmap KMU. Kompetent. stellt ein Instrument dar, damit Geschäftsführer*innen und Personalverantwortliche für ihre wichtige Personalarbeit die passgenaue Unterstützung erhalten können. Das Instrument der Roadmap schafft in einem ersten Schritt Transparenz darüber, welche Angebote und Ansprechpartner*innen zur Unterstützung von Personalverantwortlichen schon vorhanden sind und wo noch Handlungsbedarfe bestehen. Daran kann vor Ort angeknüpft und weitere Angebote und Ansprechpartner*innen ergänzt werden.
Mithilfe dieser Roadmap sollen einerseits Bertriebe einen einfach zugänglichen Überblick über regionale und überregionale Angebote und Ansprechpartner*innen erhalten und andererseits regionale Akteure vernetzt darauf einwirken können, dass Personalverantwortliche in kleinen und mittelgroßen Betrieben auch zukünftig ihre wichtige Personalarbeit, gut unterstützt und begleitet, leisten können.
Für die Zukunftsfähigkeit des Mittelstandes ist es von großer Bedeutung, dass Betriebe über ausreichend und gut qualifizierte Fach- und Arbeitskräfte verfügen. Die Roadmap KMU. Aquatic ecosystems are globally contaminated with microplastics (MP). However, comparative data on MP levels in freshwater systems is still scarce. Therefore, the aim of this study is to quantify MP abundance in water and sediment of the German river Elbe using visual, spectroscopic (Fourier-transform infrared spectroscopy) and thermo analytical (pyrolysis gas chromatography mass spectrometry) methods. Samples from eleven German sites along the German part of the Elbe were collected, both in the water and sediment phase, in order to better understand MP sinks and transport mechanisms. MP concentrations differed between the water and sediment phase. Sediment concentrations (mean: 3,350,000 particles m−3, 125–5000 μm MP) were in average 600,000-fold higher than water concentrations (mean: 5.57 particles m−3, 150–5000 μm MP). The abundance varied between the sampling sites: In sediments, the abundance decreased in the course of the river while in water samples no such clear trend was observed. This may be explained by a barrage retaining sediments and limiting tidal influence in the upstream parts of the river. Particle shape differed site-specifically with one site having exceptionally high quantities of spheres, most probably due to industrial emissions of PS-DVB resin beads. Suspended MP consisted predominantly of polyethylene and polypropylene whereas sediments contained a higher diversity of polymer types. Determined MP concentrations correspond well to previous results from other European rivers. In a global context, MP levels in the Elbe relate to the lower (water) to middle section (sediment) of the global range of MP concentrations determined for rivers worldwide. This highlights that elevated MP levels are not only found in single countries or continents, but that MP pollution is an issue of global concern.
Zur Evolution der Hirnmorphologie und Anpassungen an Extremhabitate im Taxon Poecilia (Teleostei)

Diese Dissertation befasst sich mit den Auswirkungen kontrastierender Umweltbedingungen auf die Gehirnmorphologie von neotropischen Fischen der Gattung Poecilia, welche unterschiedlichen abiotischen sowie biotischen Stressoren ausgesetzt sind. Da das Gehirn der Teleostei ein energetisch kostspieliges Organ und viel plastischer ist als z. B. bei Säugetieren, stellt sich die Frage, wie die Gehirnanatomie durch divergierende ökologische Faktoren in verschiedenen Umgebungen geformt wird, die ´extreme´, ´ressourcenbeschränkte und günstige´ Umgebungen repräsentieren. Zur Beantwortung dieser Frage wurden intraspezifische Studien an freilebenden und Laborindividuen von Poecilia-Arten durchgeführt, um die evolutionäre und ökologische Formgebung des Gehirns besser verstehen zu lernen. Im ersten Teil der Arbeit wurden Gehirnvolumina verglichen zwischen reproduktiv isolierten Populationen des neotropischen Fisches Poecilia mexicana (Ntotal = 95), die in Dunkelheit leben (Cueva Luna Azufre), in einem nahegelegenen Oberflächenhabitat (El Azufre), welcher giftigen Schwefelwasserstoff enthält und einer Kombination aus beiden Stressoren Dunkelheit und H2S (Cueva del Azufre). In einer zweiten Studie wurde auf anatomische („konvergente“) Veränderungen im Teleost-Gehirn entlang eines natürlichen Gradienten von Sulfidkonzentrationen getestet. Hierfür wurden Gehirne (Ntotal = 100) von P. mexicana verglichen, die in drei Flusssystemen im Süden Mexikos unabhängig voneinander eine erhöhte Toleranz gegenüber Schwefelwasserstoff (H2S) entwickelt haben. Dazu gehörten eine phylogenetisch alte H2S-adaptierte Form (P. sulphuraria) und zwei P. mexicana Formen, welche frühere Stufen der Anpassung an H2S darstellen. Zur Überprüfung des Einflusses anderer abiotischer und biotischer Faktoren auf die Morphologie der Gehirnregionen wurde eine weitere Studie durchgeführt. Hierbei wurden die phänotypischen Variationen der Gehirnregionen und der Körpermorphologie von Poecilia vivipara-Populationen (Ntotal = 211) aus Lagunen des Restinga de Jurubatiba Nationalpark untersucht, die sich in abiotischen Umgebungsbedingungen, insbesondere in Salzgehalt, Wassertransparenz, Phosphat und Nitrat sowie biotischen Faktoren wie Prädatorendichte unterschieden. Die erste Studie zeigte lebensraumabhängige Unterschiede bei freilebenden Fischen. Bei Fischen, die in Dunkelheit ohne H2S (LA) oder in Oberflächenhabitaten mit H2S lebten, wurden vergrößerte telenzephale Lappen, kleinere Augen und optische Tekta gefunden. Fische aus der sulfidischen Höhle (CA) zeigten zusätzlich vergrößerte Corpus cerebelli. Der Vergleich mit den Gehirnen von Labor aufgezogenen weiblichen Fischen (Ntotal = 25) zeigt eine allgemeine Verringerung der Gehirngröße sowie eine geringe Abweichung der Gehirngröße zwischen Labor aufgezogenen und freilebenden Fischen. Auch in der zweiten Studie zeigten alle in H2S-haltigen Lebensräumen lebenden Fische kleinere Augen, ein kleineres optisches Tektum und ein kleineres Gehirnvolumen, jedoch größere Corpus cerebelli und Hypothalamusvolumen als Fische aus nicht-sulfidischen Lebensräumen. Flusssystem-spezifische Effekte wurden für die telenzephalen Lappen, das gesamte Gehirn und die Augengröße festgestellt, da die Geschlechter je nach Quelle des Flusssystems unterschiedlich auf das Vorhandensein von H2S reagierten. Die dritte Studie zeigt auch, dass andere Umwelteinflüsse bemerkenswerte Verschiebungen im Gehirn und in den Gehirnregionen verursachen können. Fische, die im Süßwasser leben, zeigten eine verringerte Gesamthirngröße, telenzephale Lappen, Corpus cerebelli und Hypothalamusvolumen. Darüber hinaus zeigten Fische aus Salzwasserlagunen (hypersalin), ein verringertes Volumen des optischen Tektum, während telenzephale Lappen, Corpus cerebelli und Hypothalamusvolumen im Vergleich zu Süßwasserfischen vergrößert waren. Im Brackwasser lebende Fische wiesen im Vergleich zu Süß- und Salzwasserfischen die größten Gehirnregion-Volumen auf. Darüber hinaus zeigten die Ergebnisse über die Lagunen hinweg auch Unterschiede in der Morphologie der Kopf- und Augendurchmesser. Bei Augengröße, Kopfgröße, optischem Tektum Volumen, Hypothalamusvolumen und dem Gesamthirnvolumen wurde ein sexueller Dimorphismus beobachtet. Die dargestellten Ergebnisse verdeutlichen, dass die gefundenen Muster nahezu mit denen von H2S-Fischen identisch sind. Die ausgeprägten Unterschiede in den Hirnregionen zwischen freilebenden Fischen können als Teil der Mosaikentwicklung interpretiert werden. Die Ergebnisse der Laborpopulation zeigen jedoch eine hohe phänotypische Plastizität. Diese Studie unterstreicht damit die Bedeutung der Kombination der Untersuchung von freilebenden mit im Labor lebenden Individuen zur Beantwortung von Fragen der Gehirnentwicklung. Kleinere Augen und ein kleineres optisches Tektum, aber größere telenzephale Lappen wurden auch bei Fischen aus einem sulfidischen Oberflächenhabitat in der Nähe einer der Höhlen gefunden und sind den Ergebnissen zufolge das Resultat begrenzter Sehkraft in trüben sulfidischen Lebensräumen.

Die Zigarettenalben entwickelten sich in den 1930er Jahren zu einem vitalen Instrument der Wirtschafts- und Gesellschaftskommunikation. Da sowohl die Zigarettenindustrie als auch das NS-Regime manipulativ auf soziale Gruppen einwirken wollten, war es folgerichtig, dass Wirtschaft und Politik das beliebte Massenmedium als Kommunikationsinstrument einsetzten, um von dem propagandistischen Synergieeffekt zu profitieren. In den Zigarettenbildern mit NS-Inhalten manifestiert sich die Assimilation ökonomischer und propagandistischer Interessen, die seit Beginn der Professionalisierung von Werbung und Propaganda eingesetzt hatte. Die NS-Propaganda knüpfte bei der strategischen und operativen Planung an die Forschung der Weimarer Zeit an, die sich mit den sozialpsychologischen Grundlagen der Propaganda, Massenkommunikation und Werbewirkung sowie der Propaganda der Entente während des Ersten Weltkrieges auseinandergesetzt hatte.
Die kommunikationspolitische Allianz zwischen Politik und Wirtschaft beruhte nicht auf gesellschaftspolitischer Konformität, sondern auf einem vielschichtigen, beidseitigen Abhängigkeitsverhältnis. Die Beteiligung der Zigarettenindustrie an der NS-Propaganda war bis auf wenige Ausnahmen kein Ausdruck einer politischen Gesinnung, sondern rein monetären Motiven geschuldet oder wie bei Reemtsma auch unternehmenspolitische Strategie. Letztendlich profitierten die Zigarettenfabrikanten wirtschaftlich, denn sie konnten an dem gesellschaftpolitischen Interesse der Bevölkerung in Bezug auf NS-Themen im Rahmen der Sammelwerke partizipieren. Aber auch das NS-Regime profitierte wirtschaftlich. Neben den finanziellen Vorteilteilnahmen und der Multiplikation der NS-Ideologie profitierte das NS-Regime von den Zigarettenbildern auch als psychologisches Instrument der Truppenbetreuung, denn das Oberkommando der Wehrmacht erachtete die Zigarettenbilder für die Betreuung der Soldaten als unerlässlich.
Mit der Nutzung der Zigarettenalben begab sich aber auch das NS-Regime in Abhängigkeit zur Zigarettenindustrie, denn Populärkultur erfordert effektive Produktionsmittel und Distributionskanäle, die die Multiplikation der Medien gewährleisten sowie ambitionierte Unternehmer, die eine Gewinnmaximierung verfolgen. Das RMVP musste der Wirtschaft daher einen gewissen Freiraum bei der Themenwahl belassen, damit das Medium insgesamt nicht an Akzeptanz bei der Bevölkerung verlor. Angesichts der Tatsache, dass die so genannte Bekenntnisliteratur bei der Bevölkerung nie auf große Resonanz gestoßen und ab 1934 kaum noch nachgefragt war, bildeten die Zigarettenalben eine der wenigen Plattformen, auf der das NS-Regime über soziale Schichten hinweg Bevölkerung erreichen konnte. Die Einflussnahme des NS-Regimes war damit ebenso begrenzt, wie die der Unternehmen, denn beide mussten den sozialpsychologischen Bedürfnissen der Bevölkerung Rechnung tragen. Sowohl Wirtschaft als auch Politik mussten, um Akzeptanz zu finden, die Themen bedienen, die vom Rezipienten nachgefragt wurden.
Die Untersuchungen belegen, dass die Zigarettenalben, die in den 1930er Jahren publiziert wurden, nicht ausschließlich zur Verbreitung nationalsozialistischer Propaganda verwendet wurden, sondern auch von Gemeinschaften genutzt wurden, die ihre spezifischen politischen und ideologischen Interessen vertreten wollten. Neben den Vertretern des sozialistischen Lagers war es die SA, die die Zigarettenalben als Sprachrohr für ihre eigenen Interessen in Anspruch nahm. Bei der Parteiarmee war es insbesondere der sozialistisch geprägte Flügel um Ernst Röhm, der mit den Alben der sympathisierenden Firma Sturm seine Interessen vertrat, die primär darin bestanden, eine Partizipation an der Macht zu legitimieren und einzufordern. Darüber hinaus boten die Zigarettenalben der Parteiarmee die Möglichkeit, die eigene Historie in der deutschen Geschichte zu verorten und eine Traditionslinie bis zu den Freiheitskriegen zu ziehen. Damit konnte die SA mit den Sturm-Alben sowohl eine faschistische Bewegungskultur etablieren, als auch eine eigene Historie installieren. Die SA und ihre Mitglieder erhielten so einen Identifikationsraum, der ihnen die Möglichkeit bot, sich als selbstbewusste und eigenständige Organisation zu definieren.
Die Lenkungshoheit über die Medien und die nationalen Symbole erlaubte es dem NS-Regime, die kommunikationspolitischen Maßnahmen stringent nach den eigenen politischen Zielen auszurichten. Da die Autonomie des Öffentlichkeitssystems völlig aufgehoben und alle Publikationen der staatlichen Kontrolle unterlagen, mussten auch die Zigarettenfabrikanten die Inhalte der Sammelalben regimekonform ausrichten. Den-noch konnten weder Politik noch Industrie bei der Kommunikationspolitik völlig autark agieren, denn aufgrund der Wechselbeziehung zwischen Kommunikator und Rezipient waren beide Parteien gezwungen, die Bedürfnisse der Bevölkerung und ihre sozialpsychologischen Identifikationsräume zu berücksichtigen. Die Propaganda des Nationalsozialismus war daher, wie in den Kommunikationswissenschaften vielfach dargestellt, kein dispositionales Konzept, bei dem das Individuum einem Reiz-Reaktions-Schema folgt.
Die Berücksichtigung der sozialpsychologischen Bedürfnisse der Rezipienten wurde insbesondere bei der Integrationspropanda verfolgt.
This work describes development of a comprehensive methodology for analyzing vibro-acoustic and wear mechanisms in transmission systems. The thesis addresses certain gaps present in the fields of structure dynamics and abrasion mechanism and opens new areas for further research.
The paper attempts to understand new and relatively unexplored challenges like influences of wear on the dynamics of drive train. It also focuses on developing new techniques for analyzing the vibration and acoustic behavior of the drive unit structures and surrounding fluids respectively.
The developed methodology meets the requirements of both the complete system and component level modeling by using specially identified combination of different simulation techniques. Based on the created template model, a three-stage spur plus helical gearbox is constructed and simulated as an application example. In addition to the internal mechanical excitation mechanisms, the transmission model also includes the rotational and translational dynamics of the gears, shafts and bearings. It is followed by illustration of wear among the rotating components.
Different kinds of static and dynamic analyses are performed and coupled at various levels depending on the mechanical complexities involved. Furthermore, the structure dynamic vibration of the housing and the associated sound particle radiations are mapped into the surrounding fluid. Additionally, the approach for selection of the potential parameters for optimization is depicted. Final part focuses on the measurements of different system states used for validation of the model. In the end, results obtained from both simulations and experiments are analyzed and assessed for there respective performances.
In the present article we argue that all communication is medial in the sense that every human sign-based interaction is shaped by medial aspects from the outset. We propose a dynamic, semiotic concept of media that focuses on the process-related aspect of mediality, and we test the applicability of this concept using as an example the second presidential debate between Clinton and Trump in 2016. The analysis shows in detail how the sign processing during the debate is continuously shaped by structural aspects of television and specific traits of political communication in television. This includes how the camerawork creates meaning and how the protagonists both use the affordances of this special mediality. Therefore, it is not adequate in our view to separate the technical aspects of the medium, the ‘hardware’, from the processual aspects and the structural conditions of communication. While some aspects of the interaction are directly constituted by the medium, others are more indirectly shaped and influenced by it, especially by its institutional dimension – we understand them as second-order media effects. The whole medial procedure with its specific mediality is a necessary, but not a sufficient condition of meaning-making. We distinguish the medial procedure from the semiotic modes employed, the language games played and the competence of the players involved.

Introduction: Cancer patients tend to prefer oral instead of parenteral chemotherapy. To date, there is little evidence on the medication adherence in cancer patients. We investigated medication adherence to tyrosine kinase inhibitors in patients suffering from non-small cell lung cancer. Methods: Tyrosine kinase inhibitor adherence was measured electronically by MEMS® (medication event monitoring system) over at least six months. Adherence rates were calculated in terms of Dosing Compliance, Timing Compliance, Taking Compliance, and Drug Holidays. Patients were dichotomized as adherent when Dosing Compliance and Timing Compliance were ≥80%, Taking Compliance ranged between 90 and 110%, and <1 Drug Holiday was registered. Quality of life was assessed by two questionnaires (EORTC QLQ-C30 version 3.0, EORTC QLQ-LC13) at three time points. Adverse drug events were reported via patient diaries. Results: Out of 32 patients enrolled, data from 23 patients were evaluable. Median Dosing Compliance, Taking Compliance, and Timing Compliance adherence rates of tyrosine kinase inhibitor intake amounted to 100%, 98%, and 99%, respectively; Drug Holidays were observed in three patients. Four patients were dichotomized as non-adherent. Three of them had a twice-daily tyrosine kinase inhibitor regimen. Median quality of life scores amounted to 67 (max. 100) and remained unchanged over the study period. Fatigue and rash were the most frequently reported adverse drug events. Conclusion: Medication adherence of non-small cell lung cancer patients treated with tyrosine kinase inhibitors was extraordinarily high and is likely to support the effectiveness of tyrosine kinase inhibitor treatment and a good quality of life over a long period of time. Adherence facilitating information and education is especially relevant for patients taking tyrosine kinase inhibitors in a twice-daily regimen.
This essay explores the problem of legitimation crises in deliberative systems. For some time now, theorists of deliberative democracy have started to embrace a “systemic approach.” But if deliberative democracy is to be understood in the context of a system of multiple moving parts, then we must confront the possibility that that system’s dynamics may admit of breakdowns, contradictions, and tendencies toward crisis. Yet such crisis potentials remain largely unexplored in deliberative theory. The present article works toward rectifying this lacuna, using the 2016 Brexit and Trump votes as examples of a particular kind of “legitimation crisis” that results in a sequence of failures in the deliberative system. Drawing on recent work of Rainer Forst, I identify this particular kind of legitimation crisis as a “justification crisis.”
Locomotor activity patterns of laboratory mice are widely used to analyze circadian mechanisms, but most investigations have been performed under standardized laboratory conditions. Outdoors, animals are exposed to daily changes in photoperiod and other abiotic cues that might influence their circadian system. To investigate how the locomotor activity patterns under outdoor conditions compare to controlled laboratory conditions, we placed 2 laboratory mouse strains (melatonin-deficient C57Bl and melatonin-proficient C3H) in the garden of the Dr. Senckenbergische Anatomie in Frankfurt am Main. The mice were kept singly in cages equipped with an infrared locomotion detector, a hiding box, nesting material, and with food and water ad libitum. The locomotor activity of each mouse was recorded for 1 year, together with data on ambient
temperature, light, and humidity. Chronotype, chronotype stability, total daily activity, duration of the activity period, and daily diurnality indices were determined from the actograms. C3H mice showed clear seasonal differences in the chronotype, its stability, the total daily activity, and the duration of the activity period. These pronounced seasonal differences were not observed in the C57Bl. In both strains, the onset of the main activity period was mainly determinedby the evening dusk, whereas the offset was influenced by the ambient temperature. The actograms did not reveal infra-, ultradian, or lunar rhythms or a weekday/weekend pattern. Under outdoor conditions, the 2 strains retained their nocturnal locomotor identity as observed in the laboratory. Our results indicate that the chronotype displays a seasonal plasticity that may depend on the melatoninergic system. Photoperiod and ambient temperature are the most potent abiotic entraining cues. The timing of the evening dusk mainly affects the onset of the activity period; the ambient temperature during this period influences the latter’s duration. Humidity, overall light intensities, and human activities do not affect the locomotor behavior.
Competition over land is at the core of many sustainable development challenges in Myanmar: villagers, companies, governments, ethnic minority groups, civil society organisations and non-governmental organisations from local to the international level claim access to and decision-making power over the use of land. Therefore, this article investigates the actor interactions influencing land-use changes and their impacts on the supply of ecosystem services and human well-being. We utilise a transdisciplinary mixed-methods approach and the analytical lens of the social-ecological systems framework. Results reveal that the links between land-use changes, ecosystem services and human well-being are multifaceted; For example ecosystem services can decline, while human well-being increases. We explain this finding through three different pathways to impact (changes in the resource systems, the governance systems or the broader social, economic and political context). We conclude with implications of these results for future sustainable land governance.
Traumatische Verletzungen fordern jährlich über fünf Millionen Todesopfer. Sie sind bei unter 45-Jährigen die häufigste Ursache für Tod und körperliche Behinderung dar. Ein Polytrauma verursacht eine schwere Belastung für das Immunsystem und ist häufig von schweren Störungen der Immunregulation gekennzeichnet. Die Immunreaktion übersteigt bei schweren Traumata das für lokale Reparaturmechanismen notwendige Maß, und so kommt es je nach Ausmaß der Verletzungen innerhalb der ersten Minuten bis Stunden zu einer systemischen Hyperinflammation, dem sogenannten Systemischen Inflammatorischen Response- Syndrom (SIRS). Auch in nicht verletzten Organen verursacht SIRS Störungen in der Endothel-Funktion, wodurch die Mikrozirkulation in diesen Organgen beeinträchtigt ist. In der Folge kommt es zu interstitieller Ödembildung, zur Gewebsinfiltration durch Leukozyten und zu Zelluntergang. Diese Prozesse können zur Fehlfunktion von Organen bis hin zum Organversagen, und, da sie häufig in mehreren Organen gleichzeitig ablaufen, auch zum klinisch dann oft schwer beherrschbaren Multiorganversagen (MOV) führen. Auf der anderen Seite stoßen schwere Verletzungen antiinflammatorische Prozesse an, die zu einer ausgeprägten Immunsuppression führen können, dem Kompensatorischen Antiinflammatorischen Response-Syndrom (CARS), mit der Folge, dass polytraumatisierte Patienten erhöht anfällig für infektiöse Komplikationen sind. Die beschriebenen Funktionsstörungen des Immunsystems sind ein wichtiger Mortalitätsfaktor von polytraumatisierten Patienten. Während wir SIRS und seine Folgen über die letzten Jahre immer besser verstehen, mit signifikanten Fortschritten auch für die klinische Handhabung dieser Komplikationen des Polytraumas, ist CARS weit schlechter untersucht.
Während der post-traumatschen Immunantwort spielen nicht nur Zellen der angeborenen, sondern auch solche der erworbenen Immunabwehr eine wichtige Rolle. So sind regulatorische T-Zellen (Treg) entscheidend an der posttraumatischen Immunsuppression beteiligt. Treg beeinflussen die immunologische Homöostase Treg mit einem Arsenal immunsuppressiver Werkzeuge. Sie töten oder beeinflussen beispielsweise antigenpräsentierende Zellen oder T-Effektorzellen und verändern das Zytokinmilieu und metabolische Signalwege. Nach einem Trauma kann eine überschießende Aktivität von Treg die immunologische Balance so beeinträchtigen, dass eine posttraumatische Immunsuppression entsteht oder intensiviert wird. Die hier vorgestellte Studie Ziel dient daher dem besseren Verständnis der Dynamik von Treg nach einer stattgehabten traumatischen Verletzung. Dafür untersuchten wir die Verläufe verschiedener Subpopulationen von Treg im Blut schwer verletzter Patienten. Da der Forschung am Menschen in vivo enge ethische und methodologische Grenzen gesetzt sind, nehmen Tiermodelle in der Traumaforschung einen hohen Stellenwert ein. Daher verglichen wir die an Patienten erhobenen Daten über die posttraumatische Dynamik von Treg mit den Verläufen in einem adäquaten Tiermodell.
Aufgrund der guten anatomischen, physiologischen und genetischen Ähnlichkeit zum Menschen werden Tiermodelle am Schwein zunehmend beliebter. Ein Polytraumamodell am Schwein existiert erst seit wenigen Jahren. Über Treg wurde in diesem Rahmen bisher nicht geforscht. Die Charakterisierung ihres Immunphänotyps und ihrer Dynamik könnte die Anwendbarkeit des Schweine-Modells für Fragen der Trauma-Forschung verbessern und gleichzeitig unser Verständnis der Pathophysiologie posttraumatischer Komplikationen wir SIRS oder Sepsis erhöhen.
Bei 20 Traumapatienten (TP) mit einem Injury Severity Score (ISS) ≥ 16 wurde bei Ankunft in der Notaufnahme, nach einem und nach drei Tagen venöses Blut entnommen. Zehn gesunde Freiwillige (HV) fungierten in der Studie als Kontrollgruppe. Das Polytrauma im Großtiermodell am Schwein bestand aus einer Femurfraktur, einer Leberlazeration, einer Lungenkontusion und einem hämorrhagischen Schock, was einen ISS von 27 ergab. Auf die Traumainduktion folgte die Reanimationsphase und die chirurgische Versorgung der Femurfraktur nach dem damage-control-Prinzip. Die Blutentnahmen erfolgten bei den Versuchstieren vor und sofort nach Trauma, sowie nach 24 und 72 Stunden. Wir verglichen die Dynamik der Verläufe der Treg von TP mit denen von HV und mit Daten aus den Tierversuchen. Es herrscht noch kein wissenschaftlicher Konsens darüber, welche Kombination aus immunologischen Oberflächenmarkern die Identifikation von Treg zuverlässig gewährleisten kann. Dies liegt auch daran, dass Treg eine Gruppe verschiedener Unterpopulationen darstellen. Folglich analysierten wir verschiedene Kombinationen. Wir färbten Cluster of differentiation (CD) 4-positive und CD25-positive (CD4+CD25+), CD4+CD25+forkhead box P3 (FoxP3)+, CD4+CD25+CD127-negative (CD127−) und CD4+CD25+CD127−FoxP3+ Zellen mit Antikörpern und charakterisierten die jeweilige Gruppe mithilfe der Durchflusszytometrie. CD4+CD25+CD127− Treg sind beim Menschen bekannt. Beim Schwein werden sie in dieser Studie erstmalig beschrieben.

We show that CH embraces the synchronous pi-calculus. The formal foundations are contextual semantics in both languages, where may- as well as should-convergence are observed. We succeed in defining and proving smart properties of a particular translation mapping the synchronous pi-calculus into CH. This implies that pi-processes are error-free if and only if their translation is an error-free CH-program Our result shows that the chosen semantics is not only powerful, but can also be applied in concrete and technically complex situations. The developed translation uses private names. We also automatically check potentially correct translations that use global names instead of private names. As a complexity parameter we use the number of MVars introduced by the transformation, where MVars are synchronized 1-place buffers. The automated refutation of incorrect translations leads to a classification of potentially correct translations, and to the conjecture that one global MVar is insufficient.

Differentialdiagnostik der frühen primären Myelofibrose (präPMF) gemäß der neuen WHO-Klassifikation

Die vorliegende Arbeit stützt sich auf die retrospektive Begutachtung von 348 Knochenmarkbiopsien, welche anhand der WHO-Klassifikation von 2016 erneut reevaluiert wurden. Insbesondere widmeten wir uns der Differentialdiagnostik der „echten ET“ und der thrombozythämisch verlaufenden Form der PMF (präPMF). Die Einteilung erfolgte anhand morphologischer Kriterien, wie sie in der WHO-Klassifikation von 2016 aufgeführt sind.
Zusammengefasst ist eine richtungsweisende diagnostische Entscheidung der Patienten mit ET und präPMF nur möglich, wenn man sowohl die molekulargenetischen und klinischen Parameter in Kombination mit histologischen Kriterien und deren charakteristischen Mustern betrachtet. Eine repräsentative Knochenmarkbiopsie und deren standardisierte Befundung ist daher von absoluter diagnostischer Wichtigkeit für die Subtypisierung der MPN. Augenmerk sollte auf das typisch dargebotene morphologische Muster gelegt werden und nicht auf einzelne Merkmale. Die vorliegende Knochenmarkbiopsie muss nicht, wie früher praktiziert, anhand vieler einzelner Merkmale detailliert betrachtet und bewertet werden. Durch eine kombinierte Betrachtung von nur wenigen morphologischen Schlüsselparametern ist bereits eine Diagnose mit hoher Reproduzierbarkeit möglich. Mit dieser Arbeit wird der Stellenwert der WHO-Klassifikation unterstrichten, da die hier vorgegebenen Kriterien absolut essentiell und trotzdem in ihrem Umfang ausreichend sind, um die Differentialdiagnostik der MPN in Zukunft zu verbessern.
Machine Learning (ML) is so pervasive in our todays life that we don't even realise that, more often than expected, we are using systems based on it. It is also evolving faster than ever before. When deploying ML systems that make decisions on their own, we need to think about their ignorance of our uncertain world. The uncertainty might arise due to scarcity of the data, the bias of the data or even a mismatch between the real world and the ML-model. Given all these uncertainties, we need to think about how to build systems that are not totally ignorant thereof. Bayesian ML can to some extent deal with these problems. The specification of the model using probabilities provides a convenient way to quantify uncertainties, which can then be included in the decision making process.
In this thesis, we introduce the Bayesian ansatz to modeling and apply Bayesian ML models in finance and economics. Especially, we will dig deeper into Gaussian processes (GP) and Gaussian process latent variable model (GPLVM). Applied to the returns of several assets, GPLVM provides the covariance structure and also a latent space embedding thereof. Several financial applications can be build upon the output of the GPLVM. To demonstrate this, we build an automated asset allocation system, a predictor for missing asset prices and identify other structure in financial data.
It turns out that the GPLVM exhibits a rotational symmetry in the latent space, which makes it harder to fit. Our second publication reports, how to deal with that symmetry. We propose another parameterization of the model using Householder transformations, by which the symmetry is broken. Bayesian models are changed by reparameterization, if the prior is not changed accordingly. We provide the correct prior distribution of the new parameters, such that the model, i.e. the data density, is not changed under the reparameterization. After applying the reparametrization on Bayesian PCA, we show that the symmetry of nonlinear models can also be broken in the same way.
In our last project, we propose a new method for matching quantile observations, which uses order statistics. The use of order statistics as the likelihood, instead of a Gaussian likelihood, has several advantages. We compare these two models and highlight their advantages and disadvantages. To demonstrate our method, we fit quantiled salary data of several European countries. Given several candidate models for the fit, our method also provides a metric to choose the best option.
We hope that this thesis illustrates some benefits of Bayesian modeling (especially Gaussian processes) in finance and economics and its usage when uncertainties are to be quantified.
Across the entire animal kingdom, sociality, i.e. the tendency of individual animals to form a group with conspecifics, is a common trait. Environmental changes have to be met with corresponding, quick adaptations. For social species, the presence of conspecifics is important for survival and if social animals are deprived of access to conspecifics, this can lead to strong and lasting changes on a physiological level as well as behaviour. Gene expression changes responsible for these adaptations have so far not been understood in detail. As social isolation leads to changes on a neuronal level, it is important to investigate the gene expression changes that are induced in the brain. In this thesis, next-generation RNA-sequencing was applied to zebrafish, a well-established model organism characterized by its high degree of companionship. Within the entire brain, gene expression was analysed in zebrafish that were raised either with conspecifis or in isolation, ranging from 5 to 21 days post fertilization. Using this approach, several genes were identified that were downregulated by social isolation. In this thesis, I focused on one of these consistently downregulated genes, parathyroid hormone 2 (pth2). The expression of pth2 was demonstrated to be bidirectionally regulated by the number of conspecifics present and to be responsive to changes in the social environment within 30 minutes. Regulation of pth2 does not occur by visual or chemosensory access to conspecifcs, but is mediated by mechanosensory perception of other fish via the lateral line. In an experiment using an artificial mechanical stimulation paradigm, it was shown that the features necessary to elicit pth2 transcription closely mimick the locomotion of actual zebrafish. Other, similar stimulation paradigms are not capable to induce this transcriptional response.
Türkisch-russische Zentralasienpolitik : geopolitische Rivalität oder strategische Partnerschaft?

Die türkisch-russische Geschichte ist eine Geschichte der Rivalitäten. Sie wird wegen 15 Kriege zwischen den beiden Staaten als konflikthaft bezeichnet. Ihren 1. Krieg führten die beiden Staaten wegen Zentralasien, um das Khanat Astrachan (1568–1570). Der Untersuchungszeitraum dieser Dissertation erstreckt sich von diesem Datum bis zum Ende 2019. In diesem Zeitraum rivalisierten die Türkei und Russland geopolitisch in Zentralasien. Diese Arbeit konzentriert sich auf die türkisch-russische Zentralasienpolitik, bzw. darauf, wie die Türkei und Russland auf ihre gegenseitige Zentralasienpolitik reagieren, warum sie in Zentralasien geopolitisch rivalisieren (1. Forschungsfrage) und ob in Zukunft eine türkisch-russische strategische Partnerschaft in Zentralasien möglich ist (2. Forschungsfrage). Politikwissenschaftlich sind diese Fragen von großer Relevanz, weil eine mögliche türkisch-russische strategische Partnerschaft die gesamten Machtverhältnisse der Welt verändern würde.
BH3 mimetics are novel anticancer therapeutics that induce apoptosis by targeting anti‐apoptotic BCL‐2 proteins. Highly specific inhibitors of the main anti-apoptotic proteins BCL-2, BCL‐XL and MCL‐1 promise new opportunities for the treatment of AML. However, it is currently unclear which of these anti-apoptotic BCL-2 proteins represents the most promising target in AML. Therefore, we investigated the effect of BH3 mimetics targeting either BCL-2 (ABT-199, S55746), BCL-XL (A-1331852) or MCL-1 (S63845) on eleven AML cell lines. Drug sensitivity screening revealed heterogeneous sensitivity towards the different BH3 mimetics, with the best responses observed upon targeting of MCL-1. Selected cell lines that displayed sensitivity towards the specific BH3 mimetics underwent intrinsic apoptosis, which was characterized by loss of mitochondrial membrane potential, exposure of phosphatidylserine and activation of caspases. Furthermore, S63845 turned out to displace BIMS and NOXA from MCL-1 to induce apoptotic cell death. Importantly, the translational relevance of this study was demonstrated by experiments in primary AML blasts, which displayed similar sensitivity towards BH3 mimetics as the cell lines did. Additionally, experiments with nonmalignant cells could confirm the clinical relevance of the MCL-1 inhibitor. There we could show, that S63845 does not cause cytotoxicity on HPCs at efficacious doses.
In conclusion, our findings reveal that the inhibition of BCL-2 proteins, especially MCL-1, by BH3 mimetics can be a promising strategy in AML treatment.
Two salts of the 6,6-difluoro-6H-dibenzo[c,e][1,2]oxaborinin-6-ide anion with different cations

The crystal structures are reported of the 6,6-difluoro-6H-dibenzo[c,e][1,2]oxaborinin-6-ide (or 9,9-difluoro-10-oxa-9-boraphenanthren-9-ide) anion with two different cations, namely, potassium 6,6-difluoro-6H-dibenzo[c,e][1,2]oxaborinin-6-ide, K+·C12H8BF2O−, (II), featuring a polymeric structure, and bis(tetraphenylphosphonium) bis(6,6-difluoro-6H-dibenzo[c,e][1,2]oxaborinin-6-ide) acetonitrile trisolvate, 2C24H20P+·2C12H8BF2O−·3CH3CN, (III), which is composed of discrete cations, anions and acetonitrile solvent molecules linked by C—H...O, C—H...N and C—H...F hydrogen bonds. There are only minor differences in the geometrical parameters of the anions in these structures.
Study design: Systematic review. Background and objectives: Preoperative neuromuscular function is predictive for knee function and return to sports (RTS) after reconstruction of the anterior cruciate ligament (ACL). The aim of this review was to examine the potential benefits of prehabilitation on pre-/postoperative objective, self-reported and RTS-specific outcomes. Methods: A systematic search was conducted within three databases. From the 1.071 studies screened, two randomized control trials (RCTs), two control trials (CTs) and two cohort studies (CS) met the inclusion criteria. Methodological quality rating adopted the PEDro- (RCT, CT) or Newcastle-Ottawa-Scale (CS). Results and conclusions: Methodological quality of the included studies was moderate (PEDro score: 6.5 ± 1.7; range 4 to 9). Two studies reported higher increases of the maximal quadriceps torque from baseline to pre-reconstruction: one study in the limb symmetry index (LSI), and one in both legs of the prehabilitation group compared to the controls. At 12-weeks post-reconstruction, one study (from two) indicated that the prehabilitation group had a lesser post-operative decline in the single-leg-hop for distance LSI (clinically meaningful). Similar findings were found in terms of quadriceps strength LSI (one study). At both pre-reconstruction (three studies) and two-year post-surgery (two studies), the prehabilitation groups reached significantly higher self-reported knee function (clinically meaningful) than the controls. RTS tended to be faster (one study). At two years post-surgery, RTS rates (one study) were higher in the prehabilitation groups. The results provide evidence for the relevance of prehabilitation prior to ACL-reconstruction to improve neuromuscular and self-reported knee function as well as RTS. More high quality confirmatory RCTs are warranted.
Die angespannte Lage am Wohnungsmarkt hat in vielen Städten eine neue Welle von Verdrängungsprozessen induziert und insbesondere die Situation einkommensschwacher Haushalte häufig prekär werden lassen. Angesichts dieser Entwicklungen haben sich vielerorts mietenpolitische Bewegungen konstituiert, die sich für eine Abkehr von einer neoliberalisierten und zunehmend finanzialisierten Wohnungsversorgung einsetzen. Lisa Vollmer nimmt in ihrer Forschungsarbeit zwei solcher Bewegungen in den Blick und fragt danach, wie sich politische Kollektivität in den alltäglichen Praktiken von Mieter*innen in Berlin bzw. New York formiert.
Class I and II histone deacetylases (HDAC) are considered important regulators of immunity and inflammation. Modulation of HDAC expression and activity is associated with altered inflammatory responses but reports are controversial and the specific impact of single HDACs is not clear. We examined class I and II HDACs in TLR-4 signaling pathways in murine macrophages with a focus on IκB kinase epsilon (IKKε) which has not been investigated in this context before. Therefore, we applied the pan-HDAC inhibitors (HDACi) trichostatin A (TSA) and suberoylanilide hydroxamic acid (SAHA) as well as HDAC-specific siRNA. Administration of HDACi reduced HDAC activity and decreased expression of IKKε although its acetylation was increased. Other pro-inflammatory genes (IL-1β, iNOS, TNFα) also decreased while COX-2 expression increased. HDAC 2, 3 and 4, respectively, might be involved in IKKε and iNOS downregulation with potential participation of NF-κB transcription factor inhibition. Suppression of HDAC 1–3, activation of NF-κB and RNA stabilization mechanisms might contribute to increased COX-2 expression. In conclusion, our results indicate that TSA and SAHA exert a number of histone- and HDAC-independent functions. Furthermore, the data show that different HDAC enzymes fulfill different functions in macrophages and might lead to both pro- and anti-inflammatory effects which have to be considered in therapeutic approaches.
Objectives: The ongoing coronavirus pandemic is challenging, especially in severely affected patients who require intubation and sedation. Although the potential benefits of sedation with volatile anesthetics in coronavirus disease 2019 patients are currently being discussed, the use of isoflurane in patients with coronavirus disease 2019–induced acute respiratory distress syndrome has not yet been reported. Design: We performed a retrospective analysis of critically ill patients with hypoxemic respiratory failure requiring mechanical ventilation. Setting: The study was conducted with patients admitted between April 4 and May 15, 2020 to our ICU. Patients: We included five patients who were previously diagnosed with severe acute respiratory syndrome coronavirus 2 infection. Intervention: Even with high doses of several IV sedatives, the targeted level of sedation could not be achieved. Therefore, the sedation regimen was switched to inhalational isoflurane. Clinical data were recorded using a patient data management system. We recorded demographical data, laboratory results, ventilation variables, sedative dosages, sedation level, prone positioning, duration of volatile sedation and outcomes. Measurements & Main Results: Mean age (four men, one women) was 53.0 (± 12.7) years. The mean duration of isoflurane sedation was 103.2 (± 66.2) hours. Our data demonstrate a substantial improvement in the oxygenation ratio when using isoflurane sedation. Deep sedation as assessed by the Richmond Agitation and Sedation Scale was rapidly and closely controlled in all patients, and the subsequent discontinuation of IV sedation was possible within the first 30 minutes. No adverse events were detected. Conclusions: Our findings demonstrate the feasibility of isoflurane sedation in five patients suffering from severe coronavirus disease 2019 infection. Volatile isoflurane was able to achieve the required deep sedation and reduced the need for IV sedation.
We explore the tilted-pulse-front excitation technique to control the superradiant emission of terahertz (THz) pulses from large-area photonconductive semiconductor switches. Two cases are studied. First, a photoconductive antenna emitting into free space, where the propagation direction of the optically generated THz beam is controlled by the choice of the tilt angle of the pump pulse front. Second, a THz waveguide structure with an integrated photoconductive window for the generation of THz radiation, where the injection of the THz radiation into a waveguide mode is optimized by the pulse front tilt. By providing long interaction lengths, such a waveguide-based optical-pump/THz-probe set-up may provide a new platform for the study of diverse short-lived optically induced excitations.
Motivated by the question of whether and how wave–wave interactions should be implemented into atmospheric gravity-wave parametrizations, the modulation of triadic gravity-wave interactions by a slowly varying and vertically sheared mean flow is considered for a non-rotating Boussinesq fluid with constant stratification. An analysis using a multiple-scale WKBJ (Wentzel–Kramers–Brillouin–Jeffreys) expansion identifies two distinct scaling regimes, a linear off-resonance regime, and a nonlinear near-resonance regime. Simplifying the near-resonance interaction equations allows for the construction of a parametrization for the triadic energy exchange which has been implemented into a one-dimensional WKBJ ray-tracing code. Theory and numerical implementation are validated for test cases where two wave trains generate a third wave train while spectrally passing through resonance. In various settings, of interacting vertical wavenumbers, mean-flow shear, and initial wave amplitudes, the WKBJ simulations are generally in good agreement with wave-resolving simulations. Both stronger mean-flow shear and smaller wave amplitudes suppress the energy exchange among a resonantly interacting triad. Experiments with mean-flow shear as strong as in the vicinity of atmospheric jets suggest that internal gravity-wave dynamics are dominated in such regions by wave modulation. However, triadic gravity-wave interactions are likely to be relevant in weakly sheared regions of the atmosphere.
Aim: The aim of this study is to utilize the niche measurement guidelines outlined by Jordans et al. in order to establish normal values and accurate description of caesarean section scars in a normal population. After defining the normal distribution, abnormal pregestational scar characteristics will be identified for predicting adverse pregnancy outcomes. Methods: This is a prospective observational multicenter clinical study where women with a history of only one caesarean section and yet open family planning are enrolled. The uterine length, cervical length, niche length, niche depth, niche width, residual myometrial thickness, endometrial thickness, scar to internal os distance, anterior myometrial thickness superior and inferior to the scar and the posterior myometrial thickness opposite the scar, superior and inferior to it are measured in a pregestational uterus. The lower uterine segment is measured over a length of 3 cm during subsequent pregnancy and followed up until delivery. Results: Data from 500 patients will yield normal distribution curves for all predefined measurements. Establishing a correlation between deviations from the normal measures and adverse events would be instrumental for counseling women regarding subsequent pregnancy and mode of delivery.
Conclusion: This study will demonstrate the changes of the post-caesarean scar from a non-pregnant uterus until delivery and can confirm the importance of the scar characteristics in predicting pregnancy outcome.
Background: Brodalumab is a fully human monoclonal immunoglobulin IgG2 antibody that binds to the human IL-17 receptor subunit A and by that inhibits the biologic action of IL-17A, IL-17F, IL-17C and IL-17E. Therapy with fumaric acid esters (FAE) is a well established and widely used first-line systemic treatment for subjects with moderate-to-severe plaque psoriasis. Objectives: To compare brodalumab to FAE in terms of clinical efficacy, patient-reported outcomes and safety in subjects with moderate-to-severe plaque psoriasis who were naïve to systemic treatment. Methods: Eligible subjects were randomized 1 : 1 to 210 mg brodalumab injections or oral FAE according to product label in this 24-week, open-label, assessor-blinded, multi-centre, head-to-head phase 4 trial. The primary endpoints were having PASI75 and having sPGA score of 0 or 1 (sPGA 0/1). Subjects with missing values for the primary endpoints were considered non-responders. Results: A total of 210 subjects were randomized. 91/105 subjects completed brodalumab treatment and 58/105 subjects completed FAE treatment. At Week 24, significantly more subjects in the brodalumab group compared to the FAE group had PASI75 (81.0% vs. 38.1%, P < 0.001) and sPGA 0/1 (64.8% vs. 20.0%, P < 0.001). In the brodalumab group, the median time to both PASI75 and to PASI90 was significantly shorter than in the FAE group (4.1 weeks vs. 16.4 weeks, and 7.4 weeks vs. 24.4 weeks, respectively, P < 0.0001 for both). The rate of adverse events was lower in subjects treated with brodalumab compared to subjects treated with FAE (616.4 vs. 1195.8 events per 100 exposure years). No new safety signals were detected for brodalumab. Conclusions: Brodalumab was associated with rapid and significant improvements in signs and symptoms of moderate-to-severe plaque psoriasis, with a superior efficacy profile to what was observed with FAE in systemic-naïve subjects over 24 weeks.
Die Sonografie wird als primäre Untersuchungsmethode zur Abklärung vielfältiger Krankheitsbilder eingesetzt und hat sich auch in Leitlinien etabliert. Die Rolle der Sonografie in der Geriatrie ist weniger bekannt und nicht systematisch untersucht.
Ziel der Studie war die Evaluierung der Sonografie als routinemäßig eingesetztes Verfahren und erweiterte körperliche Untersuchung bei geriatrischen Patienten in der medizinischen Akutversorgung. Alle sogenannten geriatrischen Assessments und auch die Sonografie als Screening-Untersuchung erfolgten unabhängig von der Symptomatik des geriatrischen Patienten. Die Befunde wurden mit einem hand-held ultrasound device (HHUSD) und einem high-end ultrasound (HEUS) erhoben und dann verglichen. Die HEUS-Ergebnisse wurden als Goldstandard angesehen. Die Untersuchungen mit HEUS erfolgten meist zu einem späteren Zeitpunkt als mit HHUSD; in der Regel in einem Zeitraum von bis zu sieben Tagen.
Es handelt sich um eine prospektive Studie, die 86 Patienten in einem Zeitraum von 10 Monaten eingeschlossen hat. Die Untersucherin und Doktorandin stellte Folgendes dar: Das Abdomen und die basalen Abschnitte des Thorax sowie die Schilddrüse. Zusätzlich erfolgte eine elastografische Untersuchung der Leber mittels FibroScan.
Die Ultraschalluntersuchung war bei 22/86 (25,6 %) Patienten aufgrund der Symptomatik indiziert und erfolgte bei 64/86 (74,4 %) Patienten als Screening, also ohne klinische Fragestellung. Die Indikationen der Untersuchungen waren: Tumorsuche (8/86 (9,3 %)), Anämie (5/86 (5,8 %)), Leberwert-Erhöhung (5/86 (5,8 %)), Dyspnoe (5/86 (5,8 %)), Frage nach Milzpathologien (2/86 (2,3 %)), Gewichtsverlust (1/86 (1,2 %)), Infektfokussuche (1/86 (1,2 %)), Durchfall (1/86 (1,2 %)), Frage nach intraabdominalem Hämatom (1/86 (1,2 %)) und Kontrolle bei bekanntem Bauchaortenaneurysma (1/86 (1,2 %)). Bei einigen Patienten kamen mehrere Fragestellungen vor.
Die Befunde, die am häufigsten gefunden wurden, waren: Cholezystolithiasis (28/86 (32,6 %); mit HHUSD 5 falsch negative Ergebnisse), rechtsseitiger Pleuraerguss (27/86 (31,4 %); 5 falsch positive Ergebnisse mit HHUSD), Schilddrüsenknoten (26/86 (30,2 %); 2 falsch negative und 1 falsch positiver Befunde mit HHUSD), Nierenzysten (24/86 (27,9 %); 6 falsch negative Ergebnisse mit HHUSD) und Fettleber (23/86 (26,7 %); 3 falsch negative Befunde mit HHUSD). Von 64/86 (74,4 %) untersuchten Patienten ergab sich bei 8/86 (9,3 %) eine therapeutische Konsequenz. Die wichtigsten Befunde, die mit HHUSD übersehen wurden waren: 2 zystische Formationen im Pankreas (2/86 (2,3 %); mit HEUS insgesamt (6/86 (7,0 %)), eine Lebezirrhose (mit HEUS 2/86 (2,3 %)), eine Choledocholithiasis (1/86 (1,2 %); mit HEUS 2/86 (2,3 %)) und ein Lungeninfiltrat (1/86 (1,2 %); mit HEUS 2/86 (2,3 %)). Alle übersehenen Befunde hatten keine dringende therapeutische Konsequenz, so dass die point of care Sonografie (POCUS) hier ausreichend war, um die „ja/nein“-Fragen zu klären.
Bei 65/86 (75,6 %) Patienten war eine Messung der Lebersteifigkeit mittels FibroScan erfolgreich.
Es gab keine Interobserver-Variabilität. Die Untersucherin war eine Fachärztin für Innere Medizin aber keine DEGUM-ausgebildete Ultraschallerin.
Insgesamt zeigte sich eine ausreichende bis gute Übereinstimmung der erhobenen Befunde zwischen HHUSD und HEUS. Die POCUS-Untersuchung lieferte in den meisten Fällen eine Antwort auf die häufigsten Fragestellungen in der Geriatrie (wie Hydratationsstatus, Harnverhalt). Weiterhin konnte die Behandlung bei zufällig gescreenten Patienten optimiert werden. Zur Bestimmung der Organgröße konnte ebenfalls eine gute Korrelation der beiden Geräte festgestellt werden, so dass sich die alleinige Untersuchung mit HHUSD in diesen Fällen ausreichend gezeigt hätte. Ein routinemäßiger Einsatz der Sonografie in der Geriatrie muss in weiteren Studien untersucht werden. Da es sich bei geriatrischen Patienten um multimorbide Patienten mit entsprechend angepassten therapeutischen Zielen handelt, hatten mehrere der erhobenen Befunde in dieser Studie zum Zeitpunkt der geriatrischen Behandlung keine therapeutische Konsequenz.

At the beginning of the 1980s, an increased frequency of immune deficiency was discovered in a population of homosexual men, which is nowadays known as the Acquired Immune Deficiency Syndrome (AIDS). A few years later, the retro virus Human Immunodeficiency Virus 1(HIV-1) has been discovered as the cause of AIDS. Since the beginning of the pandemic, more that 74 million people have become infected and more than 32 million people died. In 2018, it was estimated that 38 million people where living with HIV-1 of which 24.5 million had access to Highly Active Antiretroviral Therapy (HAART), which blocks viral replication and prevents the progression towards AIDS. In the most cases an HIV-1 infection leads to the patient’s death within a few years Without HAART.
Taken together, this thesis shows that hematopoietic stem and progenitor cells harbor the prerequisites and characteristics to form an HIV-1 reservoir in vivo. The subsets of HSCs, MPPs and CD34+CD38+ progenitors harbor CD4 & CXCR4 double-positive cells as well as a lower amount of CD4 & CCR5 doublepositive cells. In addition, the susceptibility to X4-tropic HIV-1 is shown in vitro. Susceptibility to R5-tropic HIV-1 is only seen to a very low amount for CD34+CD38+ progenitors. The results also show that transduced HSPCs are capable to pass on integrated viral genomes via proliferation and differentiation during in vitro colony formation. More over the experiments provide evidence that this can take place for long time span as the outcome of the replating assays shows. Ex vivo analysis of HSPCs isolated from PLHIV also suggests that these cells are susceptible to HIV-1. Proviral DNA detection using a nested PCR showed infection of Lin- cells of a single donor with an R5-tropic subtype B HIV-1 clone. However, the assay could not detect infection of CD34+ cells. The
received results of this thesis are in agreement with previously published results. Albeit the obvious susceptibility to HIV-1 and existing reports of viral survival within HSPCs for several years, the low frequency of detected in vivo infected HSPCs could be related to the cytopathic effects of HIV-1 during replication resulting in cell death of potentially infected CD34+ cells. Other reasons could be associated with assay sensitivity or the small number of available patient samples. This makes hematopoietic stem and progenitor cells a target, which can be infected by HIV-1. The role and the clinical relevance of hematopoietic stem and progenitor cells in contribution to the latent viral HIV-1 reservoir within an HIV-1 infected patient needs to be further analyzed.
Climate controls the broad-scale distribution of vegetation and change in climate will alter the vegetation distribution, biome boundaries, biodiversity, phenology and supply of ecosystem services. A better understanding of the consequences of climate change is required, particularly in under-investigated regions such as tropical Asia, i.e., South and South-east Asia, which is a host to 7 of the 36 global biodiversity hotspots. Conservation strategies would also require an in-depth understanding of the response of vegetation to climate change. Therefore, the main objective of this thesis was to investigate the impact of climate change and rising CO2 vegetation in tropical Asia. Dynamic global vegetation model (DGVMs) are the well-known tools to investigate vegetation-climate interactions and climate change impacts on ecosystems. In this thesis, I used a complex trait-based DGVM called adaptive dynamic vegetation model version 2 (aDGVM2).
In Chapter 1, I presented a brief background of the phytogeography and discussed the exiting knowledge gap on vegetation-climate interactions in the region. One major disadvantage for available DGVMs studies for the tropical Asia is that most of them have used fixed plant functional types (PFTs) and do not explicitly represent the distinct varieties of vegetation type of the region such as Asian savannas. In Chapter 2, I discussed at great length to improve DGVMs for South Asia and discussed ways to include them in the model for better representation of region vegetation-climate interaction.
I upgraded the current version of aDGVM2 and added a new vegetation type i.e., C3 grasses, and modified the sub-module to simulate photosynthesis for each individual plants to aDGVM2. In chapter 3, I used this updated version of aDGVM2 to simulate the current and future vegetation distribution in South Asia under RCP4.5 and RCP8.5 (RCP: representative concentration pathway). The model predicted an increase in biomass, canopy cover, and tree height under the presence of CO2 fertilization, which triggered transitions towards tree-dominated biomes by the end of the 21st century under both RCPs. I found that vegetation along the Western Ghats and the Himalayas are more susceptible to change due to climate change and open biomes such as grassland and savanna are prone to woody encroachment.
In Chapter 4, the study domain was extended to include South-east Asia to verify if the model configuration used in Chapter 3 can also simulate vegetation patterns in tropical Asia. The aDGVM2 simulations showed a robust trend of increasing vegetation biomass and transitions from small deciduous vegetation to taller evergreen vegetation across most of tropical Asia. Shifts in plant phenology also affect ecosystem carbon cycles and ecosystem feedback to climate, yet the quantification of such impacts remains challenging. The study showed increased biomass due to CO2 fertilization, indicates that the region can remain a carbon sink given there is no other resource limitation. However, nutrient limitations on CO2 fertilization effects were not included in the study, and carbon sink potential has to be seen with caution.
In Chapter 5, I focused on Asian savannas, which have been mismanaged since the colonial era due to misinterpretation as a degraded forest. I proposed a biome classification scheme to distinguish between degraded forest or woodland and savanna based on the abundance of grass biomass and canopy cover. I found that considering vegetation systems as woodland or degraded forest could easily be mistaken as a potential for forest restoration within a tree-centric perspective. This would put approximately 35% to 40% of a unique savanna biome at risk. Although projected woody encroachments may imply a transition toward the forest that benefits climate mitigation. This raises potential conflicts of interest between biodiversity conservation in open ecosystems, i.e., savanna and active afforestation, to enhance carbon sequestration. Proper management strategies should be taken into account to maintain a balance for both objective
In conclusion, the model predicted that vegetation in South and South-East Asia would significantly shift towards tree-dominated biomes due to CO2-induced fertilization of C3-photosynthesis. The simulation under fixed CO2 and rising CO2 scenarios clearly showed that rising level of atmospheric CO2 is responsible for most of the predicted change in biome properties. This study is an important step towards understanding ecosystems of South and Southeast Asia, specifically savannas. The aDGVM2 can serve as tools to inform decision making for climate adaptation and mitigation for savanna. The thesis, thus contributes to our ability to improve conservation strategies to mitigate the consequences of climate change.
Das Vorkommen von Kunststoffmaterialien <5 mm, sogenanntem Mikroplastik
(MP), in marinen Ökosystemen wurde bereits eingehend untersucht. Im Gegensatz dazu existieren erhebliche Wissenslücken hinsichtlich der Abundanz und der Auswirkung von MP in limnischen Ökosystemen. Vor diesem Hintergrund steht das Umweltvorkommen, mögliche Eintragspfade und die Auswirkungen von MP auf aquatische Invertebraten im Fokus dieser Arbeit. Zur Bestimmung der MP-Abundanz in Fließgewässern sind Sedimente der Elbe untersucht worden. Hierfür wurde zunächst eine Methode zur Extraktion und Identifizierung von MP aus Umweltproben entwickelt, optimiert und validiert. In der anschließenden Analyse konnten in elf Probenahmestellen 55–17400 MP kg-1 in den Sedimenten nachgewiesen werden. Der Einfluss der Gezeitenströmung wurde anhand der abnehmenden MP-Abundanz in der Tideelbe deutlich. Insgesamt weisen die Ergebnisse darauf hin, dass Sedimente von Fließgewässern eine Senke für MP darstellen. Für die Evaluation von Eintragspfaden von MP in Oberflächengewässer wurden die
Einleiter von fünf Kläranlagen beprobt und 240–897 MP m-3 in den Einleitern detektiert. Die Detailuntersuchung einer Kläranlage zeigte, dass >99% der MP-Fracht im Verlauf der Abwasseraufbereitung entfernt wird. Hierbei erfolgte die Hauptentfernung
bereits in der Vorklärung. Somit stellen Kläranlagen effektive Barrieren für den Eintrag von MP dar.
Insgesamt wird ersichtlich, dass die getesteten Arten C. riparius und G. pulex relativ insensitiv gegenüber einer MP-Exposition sind. So konnten bei G. pulex keine und bei C. riparius erst bei sehr hohen MP-Konzentrationen adverse Effekte detektiert werden. Hierbei ist die Autökologie der Spezies eine mögliche Erklärung für die Toleranz gegenüber partikulären Stressoren. Auf Basis dieser Daten sowie der ermittelten MPAbundanz kann das Umweltrisiko von MP in limnischen Ökosystemen vorläufig als
gering eingeschätzt werden. Hierbei gilt es jedoch zu beachten, dass eine abschließende
Bewertung aufgrund der nach wie vor existierenden Unsicherheiten nicht möglich ist. Diese Unsicherheiten betreffen die Umweltkonzentration von MP <80 μm, das Verhaltensowie das Wirkpotential dieser heterogenen und dynamischen Stressorenklasse
in umweltrelevanten Szenarien.
Zielstellung: Es sollte im Rahmen dieser Arbeit geprüft werden ob (1) ein Zusammenhang zwischen einer radiologischen Beinlängendifferenz und einer veränderten Hüft- bzw. Kniegelenksbelastung bei Coxarthrosepatienten besteht. Des Weiteren galt es zu prüfen (2) ob die radiologisch-anatomische Beinachse bei Coxarthrosepatienten einen Einfluss auf die Gelenksbelastung von Hüfte und Knie habe und (3) ob sich in diesem Zusammenhang Unterschiede zwischen unilateral und bilateral betroffenem Patientenkollektiv darstellen ließe.
Hintergründe: Es konnte bereits gezeigt werden, dass Coxarthrosepatienten ein verändertes Gangbild aufweisen. Diese Veränderungen sind mitverantwortlich für eine veränderete Gelenkbelastung sowohl in Knie als auch Hüfte, sowohl im Vergleich zwischen betroffener und nicht betroffener Seite als auch im Vergleich zu hüftgesunden Normprobanden. Insbesondere bezüglich der Kniegelenksbelastung konnte bei den Betroffenen ein reduziertes Knieadduktionsmoment (KAM) während der Standphase nachgewiesen werden. Dies steht im Einklang mit der Beobachtung, dass Coxarthrosepatienten vermehrt eine laterale Kniearthrose auf der ipsilateralen Seite entwickeln. Coxarthrosepatienten zeigten darüber hinaus eine veränderte Beinachse, mit einer Tendenz hin zu einer Valgusfehlstellung. In wieweit insbesondere die proximale Beinachse einen Einfluss auf die alterierten Gelenkmomente hat, bleibt bislang jedoch offen. Zu bilateralen Patienten sind darüber hinaus nur wenige Daten vorliegend.
Methoden: Um die oben genannten Hypothesen zu prüfen wurden bei insgesamt 29 Patienten, bei welchen die Indikation für eine Hüfttotalendoprothese aufgrund einer fortgeschrittenen Coxarthrose gestellt wurde, eine instrumentelle Bewegungsanalyse sowie eine biplanare EOSRöntgenaufnahme, welche in diesem Zusammenhang die konventionelle präoperative Planungsaufnahme ersetzte, durchgeführt. Anschließend wurden die Daten, die aus der 3D EOS Rekonstruktion über die Beinachse gewonnen wurden, sowie die Ergebnisse der Bewegungsanalyse mit bereits zuvor erhobenen Normdaten sowie untereinander verglichen und statistisch analysiert.
Ergebnisse: Es zeigte sich, dass die Patienten eine bis auf MFA nicht von der Norm abweichende Beinachse hatten. Die bilateralen zeigten eine signifikant größere Beinlängendifferenz als die Norm, die unilateralen Patienten jedoch diesbezüglich keine Abweichung. Insgesamt zeigten die Patienten ein verändertes Gangbild, welches weitgehend den bereits in der Literatur beschriebenen Veränderungen entsprach.
Bezüglich der Gelenkmomente zeigte sich ein im Vergleich zur Norm erniedrigtes KAM 2, die übrigen Gelenkmomente zeigten keine Abweichungen von der Norm.
Es zeigten sich weder für die Beinachse noch für die Gelenkmomente signifikante Unterschiede zwischen unilateralem und bilateralem Patientenkollektiv.
Die unilateralen Patienten als gesamtes zeigten keine Korrelationen mit der absoluten Beinlängendifferenz. In einer unilateralen Subgruppe, in der die betroffene Seite länger war, konnte eine inverse Korrelation mit HAM 1 und HAM 2 gefunden werden.
KAM 1 und KAM 2 korrelierten signifikant mit allen Beinachsenparametern. In einer schrittweisen Regressionsanalyse mit HKA, FO und MFA konnten damit 68% der KAM 1 Alterationen und 57,5 % der Veränderungen von KAM 2 erklärt werden. HAM 1 und HAM 2 zeigten jeweils eine signifikante Korrelation mit MFA.
Schlussfolgerung: Die vorliegenden Ergebnisse zeigen, dass die Beinachse, insbesondere das Offset, einen Teil der Veränderungen der Kniegelenksbelastung erklären. Eine intraoperative Vergrößerung des Offsets und damit eine Varisierung der Beinachse könnte möglicherweise so zu einer Normalisierung der Kniegelenksbelastung führen.
Background: Non-clear cell renal cell cancers (nccRCC) are rare entities, and the optimal therapy in metastatic disease has still to be defined. Methods: In this small prospectively randomized phase IIa multicenter trial, we investigated temsirolimus (TEM) versus sunitinib (SUN) as first-line therapy in patients with metastatic nccRCC. The patients were randomized 1:1 to either TEM in a dose of 25 mg i.v. once a week or SUN with 50 mg p.o. daily for 4 weeks on and 2 weeks off. Primary endpoint was progression-free survival (PFS). In total, 22 patients were included with predominantly papillary RCC (16/22) followed by chromophobe RCC and others. Results: The male to female ratio was 16:6. The tumor control rate (CR + PR + SD) was 58% for TEM and 90% for SUN-treated patients. There was also a trend for improved PFS with 9.3 versus 13.2 months (HR 1.64; 95% CI 0.65–4.18) in favor of SUN. There was no trend for overall survival. Conclusions: Despite this trial had to be terminated earlier due to low recruitment, the results match the other studies published so far with the mTOR inhibitor everolimus and SUN, which show a trend in favor of SUN for ORR and PFS.
