Apoptotic cell (AC)-derived factors alter the physiology of macrophages (M Phi s) towards a regulatory phenotype that is characterized by enhanced production of anti-inflammatory mediators, an attenuated pro-inflammatory cytokine profile and reduced nitric oxide (NO) formation. Impaired NO production in response to ACs or AC-conditioned medium (CM) is facilitated by arginase II (ARG II) expression, which competes with inducible NO synthase for L-arginine. In this study, I investigated the signaling pathway that allowed CM to upregulate ARG II in M Phi s. A sphingolipid, further identified as sphingosine-1-phosphate (S1P), was required but authentic S1P alone only produced small effects. S1P acted synergistically with a so far unidentified factor to elicit high ARG II expression. S1P signaled through S1P receptor 2 (S1P2), since the S1P2-antagonist JTE013 and siRNA knock-down of S1P2 prevented ARG II upregulation. Further, inhibition and knock-down of extracellular signal-regulated kinase 5 (ERK5) attenuated CM-mediated ARG II protein induction. Exploring ERK5-dependent transcriptional regulation, promoter deletion and luciferase reporter analysis of the murine ARG II promoter (mpARG II) suggested the involvement of cyclic adenosine monophosphate (cAMP) responsive element binding protein (CREB). This was confirmed by EMSA analysis and decoyoligonucleotides scavenging CREB, thereby preventing it from activating target genes and thus, blocking ARG II expression. I concluded that AC-derived S1P binds to S1P2 and acts synergistically with other factors to activate ERK5 and concomitantly CREB. This signaling cascade shapes an anti-inflammatory M Phi phenotype by ARG II induction. Further investigations of ERK5-dependent CREB activation suggested an indirect mechanism implying that ERK5 inhibited phosphodiesterase 4 (PDE4) and thus, prevented hydrolysis of cAMP. Since S1P-dependent ERK5 activation presumably inhibited PDE4, subsequent cAMP accumulation led to enhanced PKA activity and CREB-mediated transcription. The unidentified factor(s) besides S1P probably provoked the required elevation of cAMP production in M Phi s. Indeed, pharmacological inhibition of cAMP-producing adenylyl cyclase with SQ22536 as well as cAMP-dependent protein kinase A (PKA) with KT5720 suggested cAMP to be involved in CM-mediated ARG II up-regulation. Furthermore, forskolin-dependent activation of adenyly cyclase and simultaneous rolipram-mediated inhibition of PDE4 mimicked CM-induced ARG II expression. Considering these findings, I propose that one or several unidentified factors in CM provoke cAMP production in M Phi s. In parallel, AC-derived S1P activates ERK5, which inhibits PDE4-dependent cAMP hydrolysis, further raising intracellular cAMP levels. Thus, unrestricted continuous cAMP signaling via PKA/CREB, results in a time-dependent and sustained ARG II induction.
European pea crabs - taxonomy, morphology, and host-ecology (Crustacea: Brachyura: Pinnotheridae)

Pinnotherids are small crabs symbiotic to a variety of invertebrates. The European species infest bivalves and sea squirts. Their way of life is parasitic and poses a threat to commercially exploited bivalves. While juveniles of both sexes still look very similar - being agile swimmers and partially free living - a metamorphosis takes place in the female after mating and results in a conspicuous sexual dimorphism. Thereafter, the female settles in its host definitely and is morphologically strongly adapted to the parasitic life phase. A very high reproductive output was demonstrated among several pea crab species infesting bivalves. Despite from that, hardly any information is present in the literature on the pinnotherids’ reproductive biology and the underlying morphology.
Due to their cryptic way of life, the sexual dimorphism, and the different morphotypes of the female, the taxonomy of the Pinnotheridae is a serious challenge. Two widely accepted species are recognized on European coasts: Pinnotheres pisum and Nepinnotheres pinnotheres. Pinnotheres pectunculi was so far only known from the bivalve Glycymeris glycymeris in its type locality Roscoff (France), while Pinnotheres ascidicola and Pinnotheres marioni were described as living exclusively in ascidians without careful comparison with the previously described species. In order to produce standardized comparative descriptions, pea crabs were collected and studied from different hosts and localities in the Northeast Atlantic and in the Mediterranean. Nepinnotheres pinnotheres and Pinnotheres pisum were redescribed with consideration to characters of female and male. According to our morphological analysis, Pinnotheres ascidicola and Pinnotheres marioni are junior synonyms of Nepinnotheres pinnotheres, whereas the status of Pinnotheres pectunculi as a valid species was ascertained. Important characters are the mouthparts, the male gonopods, and especially chelipeds that showed consistent characteristics among different crab stages of both sexes.
Based on our sampling, we estimated the host-range of the European species. Nepinnotheres pinnotheres lives in ascidians and in the pen shell Pinna nobilis. Pinnotheres pisum infests numerous bivalve species - Pinna nobilis included. For Pinnotheres pectunculi novel host records are presented, all from the bivalve family Veneridae. Furthermore, feeding of the Pinnotheres-species was observed. They use a setae comb ventrally on the claw to brush mucus (and the accumulated food particles) from the bivalve gills. Feeding strategies and host-ecology will be thoroughly discussed in consideration to other Pinnotheridae.
We investigated the reproductive systems of European pinnotherids by histological methods, scanning and transmission electron microscopy, and confocal laser scanning microscopy.
The Eubrachyura have internal fertilization: paired vaginas enlarge into storage structures, the spermathecae, which are connected to the ovaries by oviducts. Sperm is stored until the oocytes are mature and transported into the spermathecae, where fertilization takes place. In the investigated pinnotherids, the vagina is of the ‘concave pattern’. Musculature is attached alongside flexible parts of the vagina-wall to control the dimension of its lumen. The genital opening is closed by a muscular mobile operculum.
The spermatheca can be divided into two distinct regions by function and morphology. The ventral part includes the connection with vagina and oviduct and is regarded as the zone where fertilization takes place. It is lined with cuticle except where the oviduct enters the spermatheca by the ‘holocrine transfer tissue’. At ovulation, the oocytes have to pass through this multi-layered glandular epithelium, which has a holocrine mode secretion. The dorsal part of the spermatheca is lined by a highly secretory apocrine glandular epithelium, which was to date only found in fiddler crabs of the genus Uca.
The male internal reproductive system consists of paired testes and corresponding vasa deferentia. The sperm morphology of pinnotherids conforms to other thoracotremes, with slight differences between Nepinnotheres pinnotheres and Pinnotheres pisum. Spermatozoa become enveloped into spermatophores in the secretory proximal vas deferens. The medial vas deferens is strongly enlarged and stores spermatophores embedded in seminal plasma. The distal vas deferens holds tubular appendices, which extend into the ventral cephalothorax and slightly into the pleon. These appendices produce and store vast quantities of seminal plasma. The copulatory system of the Brachyura is formed by paired penes and two pairs of gonopods, which function in sperm transfer. In pinnotherids, the long first gonopods transfers the sperm mass to the female. It holds the ejaculatory canal inside, which opens proximally and distally. The second gonopod is solid, short and conical. During copulation, the penis and the second gonopod are inserted into the base of the tubular first gonopod. The second gonopod functions in the transport of the sperm mass inside the ejaculatory canal towards its distal opening. The specific shape of the second gonopod is strongly adapted for a sealing of the tubular first gonopod with longitudinal cuticle foldings that interlock inside the first gonopod. The presented results are discussed concerning their function in reproduction and in respect of the systematic account.
The role of secretion in sperm transfer, storage and fertilization among the Brachyura is still under debate. It is notable that structure and function of secretion are more complex in pinnotherids and probably more efficient than in other brachyuran crabs, which will be discussed, in view of the parasitic way of life and the high fecundity of pinnotherids.
Obwohl zahlreiche zelluläre Funktionen von RNAs in direktem Zusammenhang mit Proteinen stehen, wurde auch eine Vielzahl von, unter anderem regulatorischen, RNA-Motiven identifiziert, die ihre Funktion ohne eine initiale Beteiligung von Proteinen ausüben. Das detaillierte Verständnis der zu Grunde liegenden Regulationsmechanismen beinhaltet die Charakterisierung von beteiligten RNA-Architekturen und deren funktionaler Stabilitäten, von dynamischen Aspekten der RNA-Faltungsprozesse sowie die Korrelation dieser Charakteristika mit zellulären Funktionen. Im Rahmen dieser Arbeit wurden strukturelle, thermodynamische und kinetische Aspekte der Ligand-bindenden Guanin Riboswitch-RNA Aptamerdomäne des xpt-pbuX Operons aus B. subtilis und eines Cofaktor-abhängigen katalytischen RNA-Motivs, des 'Adenin-abhängigen Hairpin Ribozyms', untersucht. ...
Im ersten Teil dieser Arbeit wurde eine Variante des Anti-Thrombin-Aptamers HD1 entwickelt, die vor Belichten aktiv war und sich durch Belichten deaktivieren ließ. Dazu wurde das Wildtyp-Aptamer am 5'-Ende um eine GAAA-Schleife und eine Gegenstrangregion, bestehend aus vier Nukleotiden, erweitert. Dies reichte für eine vollständige Inaktivierung des Aptamers aus. In die Gegenstrangregion wurde ein photolabil geschütztes Nukleotid eingebaut, das die Bildung einer Haarnadelstruktur vorübergehend verhindert. Dazu wurde ein Desoxycytidin-Derivat synthetisiert, das an seiner N4-Position mit einer 1-(2-Nitrophenyl)ethyl-Gruppe modifiziert war. Durch die Maskierung der Antisense-Region wies das Aptamer vor Belichtung blutgerinnungshemmende Aktivität auf, allerdings in geringerem Maße als das Wildtyp-Aptamer. Durch Belichten wurde die Gegenstrangregion freigesetzt und dadurch die aktive Konformation des Aptamers zerstört, sodass es keine blutgerinnungshemmende Wirkung mehr besaß. In einem daran anknüpfenden Projekt sollte eine mit Licht ausschaltbare HD1-Variante mit verbessertem Schaltverhalten entwickelt werden, deren Aktivität vor dem Belichten mit der des Wildtyp-Aptamers vergleichbar ist. Tests zeigten, dass eine 5'-Erweiterung des Aptamers stets einen Aktivitätsverlust zur Folge hatte. Getestet wurden verschiedene Linker-Sequenzen, D-Spacer (Abasic Sites) und nicht nukleotidische Linker wie Glykollinker oder alkylische Linker. Eine Erweiterung am 3'-Ende brachte dagegen fast immer Aptamervarianten hervor, deren Aktivität die des Wildtypaptamers überstiegen. Um diese verbesserten Aptamervarianten zu deaktivieren, war eine Antisense-Region bestehend aus bis zu neun Nukleotiden nötig. Für eine photolabil geschützte Variante wurde zusätzlich ein Desoxyadenosinderivat mit N6-1-(2-Nitrophenyl)ethylmodifikation synthetisiert. Es zeigte sich, dass eine photolabile Schutzgruppe nicht ausreichte um die Antisense- Region zu neutralisieren. Aptamervarianten mit vier oder fünf photolabilen Schutzgruppen in der Antisenseregion waren vor dem Belichten aktiver als das Wildtyp-Aptamer HD1 und konnten durch Belichten vollständig deaktiviert werden. In einem weiteren Projekt dieser Arbeit wurde eine photolabil geschützte Glukosamin-6- phosphat-Variante synthetisiert, um eine lichtabhängige Spaltung des glmS-Ribozyms aus Bacillus subtilis zu induzieren. Dazu wurde GlcN6P an der Aminofunktion über eine Carbonyllinker mit einer 2-(2-Nitrophenyl) propylgruppe modifiziert. In vitro konnte gezeigt werden, dass mit dieser Verbindung durch Belichten die Spaltung eines glmS-EGFP-mRNA-Konstrukts induziert werden konnte. In HeLa-Zellen wurde untersucht, ob sich dieses System zur Regulation der EGFP-Expression eignet. Da erste Versuche erfolglos blieben, wurde eine lipophile, zellgängige Variante des photolabil geschützten GlcN6Ps synthetisiert. Versuche, in denen dieses Derivat getestet wird, werden zur Zeit von unseren Kooperationspartnern durchgeführt. In einem weiteren Projekt wurden Desoxyguanosinderivate für die DNA-Festphasensynthese synthetisiert, die an ihrer O6-Position mit einer p-Hydroxyphenacylgruppe bzw. mit einer 1-(3-Nitrodibenzofuran-2-yl)ethylgruppe modifiziert wurden. Diese wurden in ein Desoxyoligonukleotid eingebaut und es konnte gezeigt werden, dass die photolabilen Schutzgruppen durch Belichten abgespalten werden. Beide photolabilen Modifikationen waren allerdings unter den basischen DNA-Abspaltbedingungen zu instabil, als dass sie sich für den routinemäßigen Einsatz zur Herstellung lichtaktivierbarer Nukleinsäuren eignen würden. Im letzten Teil der Arbeit wurde eine photolabile Schutzgruppe entwickelt, die über einen zusätzlichen Aminolinker verfügt [2-(4-(Aminomethyl)-2-nitrophenyl)-propanol]. Die Aminofunktionalität war für die Dauer der DNA/RNA-Festphasensynthese mit einer Trifluoracetylgruppe geschützt, die unter den basischen Abspaltbedingungen ebenfalls entfernt wird. Mit dieser photolabilen Schutzgruppe wurden ein Thymidinderivat an der O4-Position und ein Desoxyguanosinderivat an der O6-Position modifiziert. Das Desoxyguanosinderivat wurde erfolgreich in der Oligonukleotidfestphasensynthese eingesetzt. Die photolabile Schutzgruppe konnte durch Belichten vollständig von der synthetisierten Nukleinsäure abgespalten werden. Darüber hinaus gelang es, über die Aminofunktionalität die heterobifunktionalen Crosslinker SMCC und SMPB mit der Nukleinsäure zu verknüpfen. Auf diese Weise ist eine reversible Vernküpfung der Nukleinsäure mit einem nahezu beliebigen Bindungspartner möglich. Durch Belichten kann die Nukleinsäure in ihrer ursprünglichen Form wiederhergestellt werden.
Type 1 diabetes (T1D) is a chronic T cell-mediated autoimmune disorder that results in the destruction of insulin-producing pancreatic ß cells leading to life-long dependence on exogenous insulin. Attraction, activation and transmigration of inflammatory cells to the site of ß-cell injury depend on two major molecular interactions. First, interactions between chemokines and their receptors expressed on leukocytes result in the recruitment of circulating inflammatory cells to the site of injury. In this context, it has been demonstrated in various studies that the interaction of the chemokine CXCL10 with its receptor CXCR3 expressed on circulating cells plays a key role in the development of T1D. Second, once arrived at the site of inflammation adhesion molecules promote the extravasation of arrested cells through the endothelial cell layer to penetrate the site of injury. Here, the junctional adhesion molecule (JAM) JAM-C expressed on endothelial cells is involved in the process of leukocyte diabedesis. It was recently demonstrated that blocking of JAM-C efficiently attenuated cerulein-induced pancreatitis in mice. In my thesis I studied the influence of the CXCL10/CXCR3 interaction on the one hand, and of the adhesion molecule JAM-C on the other hand, on trafficking and transmigration of antigen-specific, autoaggressive T cells in the RIP-LCMV mouse model. RIP-LCMV mice express the glycoprotein (GP) or the nucleoprotein (NP) of the lymphocytic choriomeningitis virus (LCMV) as a target autoantigen specifically in the ß cells of the islets of Langerhans and turn diabetic after LCMV-infection. In my first project I found that pharmacologic blockade of CXCR3 during development of virus-induced T1D results in a significant delay but not in an abrogation of overt disease. However, neither the frequency nor the migratory properties of islet-specific T cells was significantly changed during CXCR3 blockade. In the second project I was able to demonstrate that JAM-C was upregulated around the islets in RIP-LCMV mice after LCMV infection and its expression correlated with islet infiltration and functional ß-cell impairment. Blockade with a neutralizing anti-JAM-C antibody slightly reduced T1D incidence, whereas overexpression of JAM-C on endothelial cells did not accelerate virus-induced diabetes. In summary, our data suggest that both CXCR3 as well as JAM-C are involved in trafficking and transmigration of antigen-specific autoaggressive T cells to the islets of Langerhans. However, the detection of only a moderate influence on the onset of clinical disease during CXCR3 or JAM-C blockade reflects the complex pathogenesis of T1D and indicates that several different inflammatory factors need to be neutralized in order to achieve a stable and persistent protection from disease.
Im Rahmen dieser Arbeit wurde die Funktion ausgewählter Gene näher charakterisiert und ihr globaler Einfluß auf die Regulation der Genexpression untersucht. Im Fokus der Untersuchungen stand ein Gen, welches im Rahmen der Promotion von Alexander Zaigler in der Arbeitsgruppe Soppa (Universität Frankfurt) zunächst als Transkriptionsregulator ähnlich zu rspA aus E. coli identifiziert wurde. Zu Beginn dieser Arbeit konnte durch in silico Analysen das entsprechende Genprodukt zur Enolase Superfamilie (COG1441) zugeordnet werden. Mit Hilfe der „pop-in/pop-out“ Methode wurde das Gen in H. volcanii in frame deletiert und durch Wachstumsversuche, Northern- sowie Westernblot Analysen näher charakterisiert. Bei der Untersuchung des Wachstums konnte ein bemerkenswerter Phänotyp entdeckt werden: nur der Wechsel der Nährstoffquelle von reichhaltigen zu armen Bedingungen resultierte in einer dreitägigen Lagphase. Darüber hinaus wurde die Genexpression im Laufe eines Wachstumzyklus mittels Northern- und Westernblot Analysen bestimmt. Während das Transkript in reichhaltigem Medium nur transient expremiert wurde, wurde es in nährstoffarmen Bedingungen in allen Wachstumsphasen sehr stark induziert. Durch die Ergebnisse konnte zudem eine translationale Regulation der Genexpression nachgewiesen werden. Die Resultate offenbarten eine wichtige Funktion bei der Transition von reichem zu ärmerem Nährstoffangebot und führten schließlich zur Genbezeichnung iftA („important for transition A“). Desweiteren wurden Transkriptomuntersuchungen der Deletionsmutante im Vergleich zum Wildtyp zum Zeitpunkt der höchsten transienten Expression von iftA durchgeführt. Dadurch konnte gezeigt werden, dass iftA etwa 1% aller Gene des Genoms beeinflußt und gleichzeitig die Zahl differentieller Funktionen dieser Gene sehr gering ist. Die Ergebnisse führten insgesamt zu der Annahme, dass iftA eine Doppelfunktion besitzt, sowohl als enzymatisches Protein im Energiestoffwechsel als auch als essentieller Regulator mit noch unbekannter Funktion. Neben iftA fiel das Augenmerk auf mehrere Gene, die im Rahmen der Promotion von Neta Altman-Price an der Universität in Tel-Aviv als Histon-Acetylasen und –Deacetylasen identifiziert wurden. Zudem konnte in H. volcanii ein essentielles Gen, welches für ein Histon kodiert, entdeckt werden. Das Histon besitzt konservierte Lysinreste, die im eukaryotischen Histon H3 Ziele für eine posttranslationale Acetylierung sind. Für die weiteren Untersuchungen wurden die Lysinreste irreversibel zum einen in Glutamin (acetylierter Zustand) und zum anderen in Arginin (deacetylierter Zustand) mutiert. Im Rahmen dieser Arbeit wurden die Funktionen und die Einflüsse der beiden Histonmutanten sowie einer Acetylase- (delta pat1) und einer Deacetylase-Deletionsmutante (delta sir2) auf die globale Genregulation analysiert. Dazu wurden zunächst Transkriptomuntersuchungen in der exponentiellen Wachstumsphase mittels Microarray Analysen an den Mutanten im Vergleich zum Wildytp durchgeführt. Die Ergebnisse zeigten einen erheblich stärkeren Einfluß auf die Regulation der Genexpression durch das acetylierte Histon sowie der Deacetylase-Deletionsmutante im Vergleich zu den Ergebnissen des deacetylierten Histons und der Acetylase-Deletionsmutante. In der exponentiellen Wachstumsphase liegt das Histon in H. volcanii daher in überwiegend deacetylierter Form vor. Durch die Analysen konnte auch demonstrieren werden, dass Sir2 und das Histon eine regulatorische Wirkung auf exakt die gleichen Gene ausüben und daher unmittelbar miteinander in Verbindung stehen. Die experimentelle Bestätigung dieses Zusammenhangs stellt im Reich der Archaea bisher ein absolutes Novum dar. Die Untersuchungen des Transkriptoms der delta sir2 Mutante enthüllte zudem einen positiven Einfluß von Sir2 auf ein größeres Gencluster (HVO_1201-25). Die Gene dieses Clusters konnten durch in silico Analysen der Chemotaxis und der Flagellenbiosynthese zugeordnet werden. Für die weitere Charakterisierung der Deletionsmutante wurden daher Untersuchungen zur Bestimmung der Motilität von H. volcanii durchgeführt. Es konnte gezeigt werden, dass die Deletion von Sir2 die Beweglichkeit und gerichtete Fortbewegung von H. volcanii in erheblichem Maße beeinflußte, während die Biosynthese der Flagellen nicht beeinträchtigt war. Die Deacetylierung spielt daher eine unmittelbar Rolle bei der Signaltransduktion und Motilität. Insgesamt konnte durch die Arbeit gezeigt werden, dass die Acetylierung und Deacetylierung von Proteinen durch Pat1 resp. Sir2 die Regulation der Genexpression beeinflußt. Dies geschieht in H. volcanii indirekt durch die posttranslationale Veränderung von internen Signalen oder direkt durch die Modulierung des Histons und die damit verbundene Änderung der DNA-Struktur.
Das Auftreten von plötzlichem Herztod, das häufig durch ventrikuläre Tachyarrhythmien ausgelöst wird, stellt bis heute eine Herausforderung bei der Therapie der Patienten mit schwerer Herzinsuffizienz dar. Derartige Arrhythmien werden bei über 85% der Patienten mit schwerer Herzinsuffizienz beschrieben und über 50% der Todesursachen werden dabei auf das Auftreten von plötzlichem Herztod zurückgeführt. Es wird vermutet, dass das elektrische Remodeling als Teil der gesamten kardialen Umbauvorgänge bei der Entstehung einer Herzinsuffizienz die pathophysiologische Grundlage dieser Arrhythmien darstellt. Das Renin-Angiotensin-Aldosteron System spielt eine zentrale Rolle bei der Ausbildung des elektrischen Remodeling und insbesondere erhöhte Aldosteronkonzentrationen korrelieren mit dem Risiko kardiovaskulärer Zwischenfälle. Darüberhinaus konnte in zwei klinischen Studien (RALES und EPHESUS) gezeigt werden, dass die Therapie herzinsuffizienter Patienten mit den Aldosteronantagonisten Spironolacton und Eplerenon die Mortalität und Morbidität und insbesondere auch das Auftreten von plötzlichem Herztod signifikant senken konnte. Weitere Studien zeigen eine Verbindung zwischen dem Auftreten einer Herzinsuffizienz und Veränderungen in der Funktion und Expression kardiospezifischer repolarisierender K+-Kanäle. Neben den klinischen Daten, die einen protektiven Effekt der Aldosteronantagonisten bei plötzlichem Herztod belegen, ist wenig über die Auswirkungen von Aldosteron auf das elektrische Remodeling des Herzens bekannt. In dieser Arbeit sollte daher die Auswirkung einer chronischen Aldosteronexposition in Ratten auf die elektrophysiologischen Eigenschaften des Herzens untersucht werden. Dazu wurde Wistar-Ratten Aldosteron verabreicht und einigen Tieren die Aldosteronantagonisten Spironolacton und Eplerenon, um die Effekte der unspezifischen (Spironolacton) und spezifischen (Eplerenon) MR Blockade auf die elektrischen Eigenschaften der Kardiomyozyten zu untersuchen. Die Aldosteron exponierten Tiere entwickelten eine linksventrikuläre Hypertrophie, die sich unabhängig von Blutdruckveränderungen entwickelte, sowie ein signifikant verlängertes QT-Intervall, vermehrt auftretende ventrikuläre Extrasystolen und ventrikuläre Tachykardien. Die Elektrolytwerte (K+, Na+, Cl-) waren dabei nicht verändert. Die Aldosteronantagonisten Spironolacton und Eplerenon waren in der Lage, die unter Aldosteron auftretenden Veränderungen zu verhindern. Die Transkription der Untereinheiten kardiospezifischer K+-Kanäle (Ito, IKur, IK1) und des L-Typ Ca2+-Kanals war unter Aldosteronstimulation im linken Ventrikel signifikant erniedrigt. Auf Proteinebene konnte dies für die Kanaluntereinheiten Kv1.5 (IKur), Kir2.3 (IK1) und Cav1.2 (L-Typ Ca2+-Kanal) bestätigt werden. Die Untersuchung eventuell zugrunde liegender Signaltransduktionswege lieferte erniedrigte mRNA Expressionslevel der kardiospezifischen Proteinkinase C Isoformen PKC-α und PKC-ε, wohingegen die mRNA-Expression von PKC-δ unter Aldosteronstimulation unverändert war. Diese Veränderungen in der Transkription der PKC Isoformen wurden durch Behandlung der Tiere mit den Aldosteronantagonisten inhibiert, was für einen MR vermittelten Effekt spricht. Weiterhin zeigte eine chronische Aldosteronstimulation eine erniedrigte mRNA Expression von Calcineurin Aß (PPP3CB) sowie Calcineurinaktivität in linksventrikulärem Gewebe der Tiere. Dieser Effekt konnte durch die Aldosteronantagonisten nicht aufgehoben werden, so dass ein Signaltransduktionsweg, der nicht über den MR vermittelt wird, zugrunde liegen könnte. Insgesamt konnte in dieser Arbeit gezeigt werden, dass chronisch erhöhte Aldosteronkonzentrationen im Rattenherz blutdruckunabhängig zu strukturellen und elektrischen Veränderungen führen, die das Auftreten maligner ventrikulärer Arrhythmien begünstigen. Beide Aldosteronantagonisten Spironolacton und Eplerenon sind in der Lage, die durch Aldosteron vermittelten Effekte in gleicher Weise zu inhibieren. Die Ergebnisse zeigen pathophysiologische Zusammenhänge auf, die die Bedeutung von Aldosteron und der Therapie mit Aldosteronantagonisten für die Behandlung der Herzinsuffizienz und in Zukunft möglicherweise der Hypertrophie unterstreichen.
Funktionelle Analyse der Helper-Component Proteinase (HC-Pro) aus Zucchini Yellow Mosaic Virus

In dieser Arbeit wurde der Einfluss einer Punktmutation der ZYMV HC-ProFINK auf die RNA Silencing Suppressor-Aktivität und die miRNA-Akkumulation in N. benthamiana-Pflanzen untersucht. Dabei konnte eine RNA Silencing Suppressor-Aktivität der HC-ProFINK nachgewiesen werden. Sowohl die HC-ProFRNK als auch die HC-ProFINK zeigten in vivo keinen Einfluss auf die Änderung der miRNA-Mengen in N. benthamiana-Pflanzen. Untersuchungen der in vitro sRNA-Bindung mit rekombinanten HC-Pro-Proteinen aus Pflanzen führte zu einer Bindung von 21 bp siRNAs und miRNAs durch die HC-ProFRNK, wobei kein Einfluss zwischen der Anzahl und Lage der Basenfehlpaarungen der miRNAs und der Bindungskapazität identifiziert werden konnte. Diese Bindung ist vermutlich abhängig von der Sequenz der miRNAs. Die Mutation von HC-ProFRNK zu HC-ProFINK bewirkte dagegen den Verlust der Bindung von kleinen RNA-Molekülen. Die HC-Pro Proteine konnten rekombinant mit einem N-terminalen Fusionsprotein exprimiert und gereinigt werden. Die funktionelle Analyse des MBP-HA-HC-ProFRNK-Proteins wies eine längenspezifische Bindung von 21 bp siRNAs auf. Die Analyse der in vitro Bindung von unterschiedlichen miRNAs aus A. thaliana und Mensch durch das rekombinante HC-ProFRNK-Protein aus Bakterien zeigte keinen Zusammenhang zwischen der Anzahl und Lage der Basenfehlpaarungen in den miRNAs und der Bindekapazität. Die Mutation im MBP-HA-HC-ProFINK-Protein führte zum Verlust der sRNA-Bindung. Diese Ergebnisse bestätigten die Beobachtungen der Gelshift-Analysen mit den rekombinanten HC-Pro-Proteinen aus Pflanzen. Durch die Fraktionierung eines A. thaliana-Proteinextrakts konnte ein nicht näher beschriebenes Protein unbekannter Funktion, welches eine Cupin-Domäne (QP) besitzt, identifiziert werden. Dies hat einen Einfluss auf die in vitro Bindung von sRNA-Molekülen durch die HC-Pro. Eine funktionelle Analyse des Trx-QP-His-Proteins mit Hilfe von Gelshift-Analysen nach der Expression in Bakterien und Reinigung zeigte einen konzentrationsabhängigen verstärkenden Effekt der siRNA Bindung durch das rekombinante MBP-HA-HC-Pro-Protein. Die Zugabe eines fraktionierten N. benthamiana-Proteinextrakts zur in vitro Bindungsreaktion führte ebenfalls zu einer verstärkten siRNA-Bindung durch die HC-Pro; Proteinextrakte von N. tabacum und der ZYMV Wirtspflanze Zucchini zeigten jedoch keinen Effekt. Die ZYMV HC-Pro besitzt eine von der TEV HC-Pro abweichende proteolytisch aktive Domäne. Durch eine rekombinante Expression des MBP-HA-HC-Pro-GFP-Fusionsproteins in Bakterien und Deletionsanalysen konnten zwei kritische Aminosäuren im C-terminalen Bereich der HC-Pro identifiziert werden. Eine Deletion der AS Asn-353 oder Glu-356 führte zum vollständigen Verlust der autoproteolytischen Aktivität des Proteins. Ein Austausch der AS Gly-456 innerhalb der Schnittstelle sowie eine N-terminale Deletion von 93 AS der HC-Pro hatten dagegen keinen Einfluss auf die autoproteolytische Aktivität. Mit Hilfe einer N-terminalen Sequenzierung des C-terminalen Spaltproduktes des MBP HC Pro mut C7-GFP, welches vermutlich in Folge einer Spaltung durch bakterielle Proteasen entsteht, sollten durch Deletionsmutanten die kritischen Aminosäuren für die Spaltung untersucht werden. Eine Deletion der konservierten AS Thr-146 von ZYMV HC-Pro sowie der flankierenden AS Val-145 bzw. Gln-147 hatte jedoch keinen Einfluss auf die Spaltung der HC-Pro. Dies deutet darauf hin, dass die Schnittstelle der Protease von deren Erkennungssequenz abweicht. Die in dieser Arbeit erzielten Ergebnisse zeigen, dass die ZYMV HC-Pro neben der sRNA-Bindung einen weiteren Mechanismus besitzt, um die Funktion als RSS auszuüben. Weitere Analysen sind nötig, um die Interaktion mit pflanzlichen Komponenten zu identifizieren und den Einfluss auf den RNA Silencing-Mechanismus aufzuklären.
In the adult mammalian central nervous system, two defined neurogenic regions retain the capacity to generate new neurons throughout adulthood, namely the subependymal zone (SEZ) at the lateral ventricles and the subgranular layer of the hippocampus (SGL). Adult neurogenesis consists of a whole set of events including proliferation, fate specification, migration, survival and finally synaptic integration of newly born neurons. Each of these events is controlled by the interplay of numerous factors. In this study two signalling systems were analysed with regard to their functional role in adult neurogenesis in vivo, namely the purinergic system and the growth factor EGF. Neither short- nor long-term application of the P2Y receptor agonists UTP and ADPβS and the P2Y receptor antagonist suramin into the lateral ventricle of adult mice altered cell responses as compared to vehicle controls in vivo. In contrast, analysis of the expansion rates of cultured neural stem cells (NSCs) from knockout mice revealed a strong increase in the number of NSCs from NTPDase2-/- mice, whereas cell numbers of NSCs from P2Y1-/- and P2Y2-/- mice were significantly reduced in comparison to wildtype levels. Notably, in vivo proliferation rates were potently elevated in the SGL and the SEZ of NTPDase2-deficient mice. However, in vivo proliferation in both neurogenic niches of the single receptor knockout mice P2Y1-/- and P2Y2-/- and P2Y1-/- P2Y2-/-double-knockout mice did not differ significantly from the wildtype. In mice lacking the P2Y2 receptor the survival of newly born neurons in the hippocampal granule cell layer was significantly increased. These data provide the first line of evidence that purinergic signalling is involved in the control of neural stem cells behaviour not only in vitro but also in vivo. In order to further characterise the role of epidermal growth factor (EGF) in adult neurogenesis, transit amplifying precursors (TAPs) and type B astrocytes were identified as EGF-responsive cell populations following ventricular EGF injection, whereas ependymal cells, neuroblasts and NG2-positive cells did not or only to a minor extent respond to EGF injection. These EGF-responsive cell populations were found on both, the septal as well as striatal lateral ventricle walls. Long-term ventricular EGF infusion for 6d, 1. increased cell proliferation of both ventricle walls revealing a gradient along the rostro-caudal axis, 2. altered the balance between neuronal and macroglial cell fates to generate oligodendrocyte precursors and 3. lead to an entire remodelling of the classical architecture of the SEZ.
Biodegradation and elimination of industrial wastewater in the context of whole effluent assessment

The focus of this thesis is on the assessment of the degradability of indirectly discharged wastewater in municipal treatment plants and on assessing indirectly discharged effluents by coupling the Zahn-Wellens test with effect-based bioassays. With this approach persistent toxicity of an indirectly discharged effluent can be detected and attributed to the respective emission source. In the first study 8 wastewater samples from different industrial sectors were analysed according to the “Whole-Effluent Assessment“ (WEA) approach developed by OSPAR. In another study this concept has been applied with 20 wastewater samples each from paper manufacturing and metal surface treating industry. In the first study generally low to moderate ecotoxic effects of wastewater samples have been determined. One textile wastewater sample was mutagenic in the Ames test and genotoxic in the umu test. The source of these effects could not be identified. After treatment in the Zahn-Wellens test the mutagenicity in the Ames test was eliminated completely while in the umu test genotoxicity could still be observed. Another wastewater sample from chemical industry was mutagenic in the Ames test. The mutagenicity with this wastewater sample was investigated by additional chemical analysis and backtracking. A nitro-aromatic compound (2-methoxy-4-nitroaniline) used for batchwise azo dye synthesis and its transformation products are the probable cause for the mutagenic effects analysed. Testing the mother liquor from dye production confirmed that this partial wastewater stream was mutagenic in the Ames test. The wasteweater samples from paper manufacturing industry of the second study were not toxic or genotoxic in the acute Daphnia test, fish egg test and umu test. In the luminescent bacteria test, moderate toxicity was observed. Wastewater of four paper mills demonstrated elevated or high algae toxicity, which was in line with the results of the Lemna test, which mostly was less sensitive than the algae test. The colouration of the wastewater samples in the visible band did not correlate with algae toxicity and thus is not considered as its primary origin. The algae toxicity in wastewater of the respective paper factory could also not be explained with the thermomechanically produced groundwood pulp (TMP) partial stream. Presumably other raw materials such as biocides might be the source of algae toxicity. In the algae test, often flat dose–response relationships and growth promotion at higher dilution factors have been observed, indicating that several effects are overlapping. The wastewater samples from the printed circuit board and electroplating industries (all indirectly discharged) were biologically pre-treated for 7 days in the Zahn–Wellens test before ecotoxicity testing. Thus, persistent toxicity could be discriminated from non-persistent toxicity caused, e.g. by ammonium or readily biodegradable compounds. With respect to the metal concentrations, all samples were not heavily polluted. The maximum conductivity of the samples was 43,700 micro S cm -1 and indicates that salts might contribute to the overall toxicity. Half of the wastewater samples proved to be biologically well treatable in the Zahn–Wellens test with COD elimination above 80%, whilst the others were insufficiently biodegraded (COD elimination 28–74%). After the pre-treatment in the Zahn–Wellens test, wastewater samples from four companies were extremely ecotoxic especially to algae. Three wastewater samples were genotoxic in the umu test. Applying the rules for salt correction to the test results following the German Wastewater Ordinance, only a small part of toxicity could be attributed to salts. In one factory, the origin of ecotoxicity has been attributed to the organosulphide dimethyldithiocarbamate (DMDTC) used as a water treatment chemical for metal precipitation. The assumption, based on rough calculation of input of the organosulphide into the wastewater, was confirmed in practice by testing its ecotoxicity at the corresponding dilution ratio after pre-treatment in the Zahn–Wellens test. The results show that bioassays are a suitable tool for assessing the ecotoxicological relevance of these complex organic mixtures. The combination of the Zahn–Wellens test followed by the performance of ecotoxicity tests turned out to be a cost-efficient suitable instrument for the evaluation of indirect dischargers and considers the requirements of the IPPC Directive.
