The mechanism by which the enzyme pyruvate decarboxylase from yeast is activated allosterically has been elucidated. A total of seven three-dimensional structures of the enzyme, of enzyme variants or of enzyme complexes from two yeast species (three of them reported here for the first time) provide detailed atomic resolution snapshots along the activation coordinate. The prime event is the covalent binding of the substrate pyruvate to the side chain of cysteine 221, thus forming a thiohemiketal. This reaction causes the shift of a neighbouring amino acid, which eventually leads to the rigidification of two otherwise flexible loops, where one of the loops provides two histidine residues necessary to complete the enzymatically competent active site architecture. The structural data are complemented and supported by kinetic investigations and binding studies and provide a consistent picture of the structural changes, which occur upon enzyme activation.
Fließgewässer und ihre angrenzenden Auen bieten vielfältige Lebensräume für zahlreiche Tier- und Pflanzenarten. Diese Vielfalt zu sichern und vorhandene Defizite abzubauen, ist Ziel der naturnahen Gewässerentwicklung, wie sie im Wassergesetz für Baden-Württemberg verankert ist. Als Grundlage hierfür nimmt die vorliegende Schrift eine Bewertung der baden-württembergischen Fließgewässer als Lebensraum für die Artengruppen Vögel, Laufkäfer, Libellen und Muscheln vor (Karten 1 5, siehe Anhang). Einen Überblick gibt die Karte "Fließgewässer in Baden-Württemberg als Lebensraum ausgewählter Artengruppen", 2001 H im Maßstab 1 :350.000 (Anhang). Diese erste Analyse im Landesmaßstab, die ausschließlich durch die Sammlung und Aufbereitung verstreut vorliegender Daten entstand, soll den Zustand der Fließgewässer als Lebensraum von Arten verdeutlichen, prioritäre Gewässerabschnitte für den Gewässerschutz aufzeigen, aber auch Hinweise für die Gewässerentwicklung geben. Auf der Grundlage der Einzelbewertungen für die vier Artengruppen wurde eine Gesamtbewertung der Gewässer vorgenommen (Karte 6, siehe Anhang). Entstanden ist also neben Einzelbewertungen für die Artengruppen eine überblicksartige Gesamtbewertung der baden-württembergischen Fließgewässer als Artenlebensräume.
Die Rahmenbedingungen für den Wasserbau haben sich in den letzten Jahren tiefgreifend verändert. Was gestern noch als "Kleines-Einmal-Eins" galt, ist heute aufgrund neuer Erkenntnisse über die Auswirkungen und die Zusammenhänge in der Natur völlig zurückgedrängt. Wir haben gelernt, daß Wasserbau nicht ausschließlich nach technischen, sondern in gleicher Weise auch nach ökologischen Gesichtspunkten zu beurteilen ist. Mit dieser Veröffentlichung liegt nunmehr eine Handreichung vor, die allen, die mit Wasserbau zu tun haben, die Entscheidungsfindung bei anstehenden Böschungs- und Ufersicherungen erleichtern soll. Dabei ist grundsätzlich zu überprüfen, ob und in welchem Umfang bauliche Sicherungsmaßnahmen notwendig sind. Teilweise können durch entsprechenden Grunderwerb bzw. durch Entschädigungsleistungen mit der "Bauweise Null" Böschungs- und Ufersicherungen stark eingeschränkt werden oder sogar ganz entfallen.
Gewässer sind die Lebensadern unserer Landschaft. Sie bestimmen wesentlich das Landschaftsbild und den Naturhaushalt. Eingriffe in die Natur und die naturnahe Kulturlandschaft haben dazu geführt. daß diese Funktionen gestört oder zerstört wurden. Wasserbau und Ökologie standen sich hierbei lange als Gegensätze gegenüber. Das Gesamtkonzept Naturschutz und Landschaftspflege Baden-Württemberg setzt nun das Ziel, Technik und Ökologie in gesamtschaulicher Betrachtung zusammenzubinden. Wesentliche Ziele der Naturschutzpolitik sind - Erhalten und Sichern naturnaher Auelandschaften und - Regeneration beeintrachtigter Gewasser und Auen. Im Rahmen eines Pilotvorhabens "Naturnahe Umgestaltung ausgebauter Fließgewässer", hat das Umweltministerium in Verbindung mit Umgestaltungsmaßnahmen umfangreiche Untersuchungen, Erhebungen und Dokumentationen veranlaßt, aus denen Anleitungen für das künftige Vorgehen gewonnen werden. Die wichtigsten der zwischenzeitlich abgeschlossenen Untersuchungsvorhaben wurden nun im Rahmen des Kolloquiums "Naturgemäße Gestaltung von Fließgewässern" der Fachwelt vor- und zur Diskussion gestellt. In den Fachvorträgen konnten die umfangreichen Ergebnisse der Untersuchungen nur in Auszügen dargestellt werden. Das Umweltministerium wird die Ergebnisse dieser Untersuchungen in weiteren Heften des Handbuchs Wasserbau veroffentlichen. Die Niederschrift dieses Kolloquiums gibt einen Überblick der derzeitigen Erkenntnisse.
Genetic engineering of baker’s and wine yeasts using formaldehyde hyperresistance-mediating plasmids

Yeast multi-copy vectors carrying the for maldehyde-resistance marker gene SFA have proved to be a valuable tool for research on industrially used strains of Saccharomyces cerevisiae. The genetics of these strains is often poorly understood, and for various reasons it is not possible to simply subject these strains to protocols of genetic engineering that have been established for laboratory strains of S. cerevisiae. We tested our vectors and protocols using 10 randomly picked baker’s and wine yeasts all of which could be transformed by a simple protocol with vectors conferring hyperresistance to formaldehyde. The application of formaldehyde as a selecting agent also offers the advantage of its biodegradation to CO2 during fermentation, i.e., the selecting agent will be consumed and therefore its removal during down-stream processing is not necessary. Er gibt als Nachschlagewerk einen schnellen Überblick über die Luftgüte und deren Langzeitentwicklung in Baden-Württemberg sowie über die sonstigen Erkenntnisse, die wir in den unterschiedlichen Aufgabenfeldern gefunden haben. Weiter zeigt er an Einzelbeispielen das Engagement der Mitarbeiterinnen und Mitarbeiter innerhalb unseres breiten Einsatz- und Aufgabenspektrums sowie die Erfolge der Tätigkeiten der UMEG auf.
Flexibility and constraints in migration and breeding of the barnacle goose Maximising fitness means, to a large extent, optimising management of time and energy. This thesis investigates aspects of timing and resource acquisition and utilisation in the barnacle goose Branta leucopsis during spring migration and reproduction. The barnacle goose population wintering along the Wadden Sea coast, and traditionally migrating via a stopover in the Baltic to its breeding sites in northern Russia, has shown some remarkable developments, among them, a geometric growth rate since the 1950s and a notable change in the timing of departure from the wintering grounds. Furthermore, long being regarded as an obligate Arctic breeder, within the past three decades this species has successfully colonised a wide variety of habitats at temperate latitudes, thereby shortening the migratory distances considerably or refraining from migration altogether. These remarkable changes raise questions about the flexibility of migratory and reproductive schedules. What are the costs and benefits of different migration strategies? And, given the context of global change, to what extent are animals capable of adapting to rapidly changing environments? In order to answer these questions we tracked migratory geese by satellite and with the help of data loggers and, furthermore, explored possible relationships of migratory behaviour and breeding performance. Another central thesis topic represents a within-species comparison of major life-history traits in populations breeding along a large ecological gradient from arctic to temperate environments. Fieldwork was conducted in arctic Russia, Sweden and The Netherlands.
Background Multidirectional interactions in social (or communication) networks can have a profound effect on mate choice behavior. For example, Poecilia mexicana males show weaker expression of mating preferences when being observed by an audience male. It was suggested that this behavior is an adaptation to reduce sperm competition risk, which arises because commonly preferred female phenotypes will receive attention also by surrounding males, and/or because the audience male can copy the focal male's mate choice. Do P. mexicana males indeed respond to perceived sperm competition risk? We gave males a choice between two females and repeated the tests under one of the following conditions: (1) during the 2nd part of the tests an empty transparent cylinder was presented (control); (2) an audience male inside the cylinder observed the focal male throughout the 2nd part, or (3) the audience male was presented only before the tests, but could not eavesdrop during the actual choice tests (non-specific sperm competition risk treatments); (4) the focal male could see a rival male sexually interacting with the previously preferred, or (5) with the non-preferred female before the 2nd part of the tests (specific sperm competition risk treatments). Results When comparing the strength of individual male preferences between the 1st and 2nd part of the tests (before and after presentation of an audience), male preferences declined slightly also during the control treatment (1). However, the decrease in strength of male preferences was more than two-fold stronger in audience treatment (2), i.e., with non-specific sperm competition risk including the possibility for visual eavesdropping by the audience male. No audience effect was found in treatments (3) and (5), but a weak effect was also seen when the focal male had seen the previously preferred female sexually interact with a rival male (treatment 4; specific sperm competition risk). Conclusions When comparing the two 'non-specific sperm competition risk' treatments (2 and 3), a very strong effect was found only when the audience male could actually observe the focal male during mate choice in treatment (2). This suggests that focal males indeed attempt to conceal their mating preferences in the visual presence of other males so as to avoid mate choice copying. When there is no potential for eavesdropping [treatment (3)], non-specific specific sperm competition risk seems to play a minor or no role. Congruent with studies on other poeciliid species, our results also show that P. mexicana males respond to perceived specific sperm competition risk, and tend to share their mating effort more equally among females when the resource value of their previously preferred mate decreases (after mating with a rival male). However, this effect is comparatively weak.
Background: Nitric oxide (NO) is an essential vasodilator. In vascular diseases, oxidative stress attenuates NO signaling by both chemical scavenging of free NO and oxidation and down-regulation of its major intracellular receptor, the alpha/beta heterodimeric heme-containing soluble guanylate cyclase (sGC). Oxidation can also induce loss of sGC's heme and responsiveness to NO.
Results: sGC activators such as BAY 58-2667 bind to oxidized/heme-free sGC and reactivate the enzyme to exert disease-specific vasodilation. Here we show that oxidation-induced down-regulation of sGC protein extends to isolated blood vessels. Mechanistically, degradation was triggered through sGC ubiquitination and proteasomal degradation. The heme-binding site ligand, BAY 58-2667, prevented sGC ubiquitination and stabilized both alpha and beta subunits.
Conclusion: Collectively, our data establish oxidation-ubiquitination of sGC as a modulator of NO/cGMP signaling and point to a new mechanism of action for sGC activating vasodilators by stabilizing their receptor, oxidized/heme-free sGC.
Poster presentation: Self-organized critical (SOC) systems are complex dynamical systems that may express cascades of events, called avalanches [1]. The SOC state was proposed to govern brain function, because of its activity fluctuations over many orders of magnitude, its sensitivity to small input and its long term stability [2,3]. In addition, the critical state is optimal for information storage and processing [4]. Both hallmark features of SOC systems, a power law distribution f(s) for the avalanche size s and a branching parameter (bp) of unity, were found for neuronal avalanches recorded in vitro [5]. However, recordings in vivo yielded contradictory results [6]. Electrophysiological recordings in vivo only cover a small fraction of the brain, while criticality analysis assumes that the complete system is sampled. We hypothesized that spatial subsampling might influence the observed avalanche statistics. In addition, SOC models can have different connectivity, but always show a power law for f(s) and bp = 1 when fully sampled. This may not be the case under subsampling, however. Here, we wanted to know whether a state change from awake to asleep could be modeled by changing the connectivity of a SOC model without leaving the critical state. We simulated a SOC model [1] and calculated f(s) and bp obtained from sampling only the activity of a set of 4 × 4 sites, representing the electrode positions in the cortex. We compared these results with results obtained from multielectrode recordings of local field potentials (LFP) in the cortex of behaving monkeys. We calculated f(s) and bp for the LFP activity recorded while the monkey was either awake or asleep and compared these results to results obtained from two subsampled SOC model with different connectivity. f(s) and bp were very similar for both the experiments and the subsampled SOC model, but in contrast to the fully sampled model, f(s) did not show a power law and bp was smaller than unity. With increasing the distance between the sampling sites, f(s) changed from "apparently supercritical" to "apparently subcritical" distributions in both the model and the LFP data. f(s) and bp calculated from LFP recorded during awake and asleep differed. These changes could be explained by altering the connectivity in the SOC model. Our results show that subsampling can prevent the observation of the characteristic power law and bp in SOC systems, and misclassifications of critical systems as sub- or supercritical are possible. In addition, a change in f(s) and bp for different states (awake/asleep) does not necessarily imply a change from criticality to sub- or supercriticality, but can also be explained by a change in the effective connectivity of the network without leaving the critical state.
Poster presentation: How can two distant neural assemblies synchronize their firings at zero-lag even in the presence of non-negligible delays in the transfer of information between them? Neural synchronization stands today as one of the most promising mechanisms to counterbalance the huge anatomical and functional specialization of the different brain areas. However, and albeit more evidence is being accumulated in favor of its functional role as a binding mechanism of distributed neural responses, the physical and anatomical substrate for such a dynamic and precise synchrony, especially zero-lag even in the presence of non-negligible delays, remains unclear. Here we propose a simple network motif that naturally accounts for zero-lag synchronization of spiking assemblies of neurons for a wide range of temporal delays. We demonstrate that when two distant neural assemblies do not interact directly but relaying their dynamics via a third mediating single neuron or population and eventually achieve zero-lag coherent firing. Extensive numerical simulations of populations of Hodgkin-Huxley neurons interacting in such a network are analyzed. The results show that even with axonal delays as large as 15 ms the distant neural populations can synchronize their firings at zero-lag in a millisecond precision after the exchange of a few spikes. The role of noise and a distribution of axonal delays in the synchronized dynamics of the neural populations are also studied confirming the robustness of this sync mechanism. The proposed network module is densely embedded within the complex functional architecture of the brain and especially within the reciprocal thalamocortical interactions where the role of indirect pathways mimicking direct cortico-cortical fibers has been already suggested to facilitate trans-areal cortical communication. In summary the robust neural synchronization mechanism presented here arises as a consequence of the relay and redistribution of the dynamics performed by a mediating neuronal population. In opposition to previous works, neither inhibitory, gap junctions, nor complex networks need to be invoked to provide a stable mechanism of zero-phase correlated activity of neural populations in the presence of large conduction delays.
Poster presentation: The analysis of neuronal processes distributed across multiple cortical areas aims at the identification of interactions between signals recorded at different sites. Such interactions can be described by measuring the stability of phase angles in the case of oscillatory signals or other forms of signal dependencies for less regular signals. Before, however, any form of interaction can be analyzed at a given time and frequency, it is necessary to assess whether all potentially contributing signals are present. We have developed a new statistical procedure for the detection of coincident power in multiple simultaneously recorded analog signals, allowing the classification of events as 'non-accidental co-activation'. This method can effectively operate on single trials, each lasting only for a few seconds. Signals need to be transformed into time-frequency space, e.g. by applying a short-time Fourier transformation using a Gaussian window. The discrete wavelet transform (DWT) is used in order to weight the resulting power patterns according to their frequency. Subsequently, the weighted power patterns are binarized via applying a threshold. At this final stage, significant power coincidence is determined across all subgroups of channel combinations for individual frequencies by selecting the maximum ratio between observed and expected duration of co-activation as test statistic. The null hypothesis that the activity in each channel is independent from the activity in every other channel is simulated by independent, random rotation of the respective activity patterns. We applied this procedure to single trials of multiple simultaneously sampled local field potentials (LFPs) obtained from occipital, parietal, central and precentral areas of three macaque monkeys. Since their task was to use visual cues to perform a precise arm movement, co-activation of numerous cortical sites was expected. In a data set with 17 channels analyzed, up to 13 sites expressed simultaneous power in the range between 5 and 240 Hz. On average, more than 50% of active channels participated at least once in a significant power co-activation pattern (PCP). Because the significance of such PCPs can be evaluated at the level of single trials, we are confident that this procedure is useful to study single trial variability with sufficient accuracy that much of the behavioral variability can be explained by the dynamics of the underlying distributed neuronal processes.
Poster presentation: Background To test the importance of synchronous neuronal firing for information processing in the brain, one has to investigate if synchronous firing strength is correlated to the experimental subjects. This requires a tool that can compare the strength of the synchronous firing across different conditions, while at the same time it should correct for other features of neuronal firing such as spike rate modulation or the auto-structure of the spike trains that might co-occur with synchronous firing. Here we present the bi- and multivariate extension of previously developed method NeuroXidence [1,2], which allows for comparing the amount of synchronous firing between different conditions. ...
Poster presentation: Introduction The ability of neurons to emit different firing patterns is considered relevant for neuronal information processing. In dopaminergic neurons, prominent patterns include highly regular pacemakers with separate spikes and stereotyped intervals, processes with repetitive bursts and partial regularity, and irregular spike trains with nonstationary properties. In order to model and quantify these processes and the variability of their patterns with respect to pharmacological and cellular properties, we aim to describe the two dimensions of burstiness and regularity in a single model framework. Methods We present a stochastic spike train model in which the degree of burstiness and the regularity of the oscillation are described independently and with two simple parameters. In this model, a background oscillation with independent and normally distributed intervals gives rise to Poissonian spike packets with a Gaussian firing intensity. The variability of inter-burst intervals and the average number of spikes in each burst indicate regularity and burstiness, respectively. These parameters can be estimated by fitting the model to the autocorrelograms. This allows to assign every spike train a position in the two-dimensional space described by regularity and burstiness and thus, to investigate the dependence of the firing patterns on different experimental conditions. Finally, burst detection in single spike trains is possible within the model because the parameter estimates determine the appropriate bandwidth that should be used for burst identification. Results and Discussion We applied the model to a sample data set obtained from dopaminergic substantia nigra and ventral tegmental area neurons recorded extracellularly in vivo and studied differences between the firing activity of dopaminergic neurons in wildtype and K-ATP channel knock-out mice. The model is able to represent a variety of discharge patterns and to describe changes induced pharmacologically. It provides a simple and objective classification scheme for the observed spike trains into pacemaker, irregular and bursty processes. In addition to the simple classification, changes in the parameters can be studied quantitatively, also including the properties related to bursting behavior. Interestingly, the proposed algorithm for burst detection may be applicable also to spike trains with nonstationary firing rates if the remaining parameters are unaffected. Thus, the proposed model and its burst detection algorithm can be useful for the description and investigation of neuronal firing patterns and their variability with cellular and experimental conditions.
Poster presentation: Coordinated neuronal activity across many neurons, i.e. synchronous or spatiotemporal pattern, had been believed to be a major component of neuronal activity. However, the discussion if coordinated activity really exists remained heated and controversial. A major uncertainty was that many analysis approaches either ignored the auto-structure of the spiking activity, assumed a very simplified model (poissonian firing), or changed the auto-structure by spike jittering. We studied whether a statistical inference that tests whether coordinated activity is occurring beyond chance can be made false if one ignores or changes the real auto-structure of recorded data. To this end, we investigated the distribution of coincident spikes in mutually independent spike-trains modeled as renewal processes. We considered Gamma processes with different shape parameters as well as renewal processes in which the ISI distribution is log-normal. For Gamma processes of integer order, we calculated the mean number of coincident spikes, as well as the Fano factor of the coincidences, analytically. We determined how these measures depend on the bin width and also investigated how they depend on the firing rate, and on rate difference between the neurons. We used Monte-Carlo simulations to estimate the whole distribution for these parameters and also for other values of gamma. Moreover, we considered the effect of dithering for both of these processes and saw that while dithering does not change the average number of coincidences, it does change the shape of the coincidence distribution. Our major findings are: 1) the width of the coincidence count distribution depends very critically and in a non-trivial way on the detailed properties of the inter-spike interval distribution, 2) the dependencies of the Fano factor on the coefficient of variation of the ISI distribution are complex and mostly non-monotonic. Moreover, the Fano factor depends on the very detailed properties of the individual point processes, and cannot be predicted by the CV alone. Hence, given a recorded data set, the estimated value of CV of the ISI distribution is not sufficient to predict the Fano factor of the coincidence count distribution, and 3) spike jittering, even if it is as small as a fraction of the expected ISI, can falsify the inference on coordinated firing. In most of the tested cases and especially for complex synchronous and spatiotemporal pattern across many neurons, spike jittering increased the likelihood of false positive finding very strongly. Last, we discuss a procedure [1] that considers the complete auto-structure of each individual spike-train for testing whether synchrony firing occurs at chance and therefore overcomes the danger of an increased level of false positives.
Poster presentation: An important challenge in neuroscience is understanding how networks of neurons go about processing information. Synapses are thought to play an essential role in cellular information processing however quantitative and mathematical models of the underlying physiologic processes that occur at synaptic active zones are lacking. We are generating mathematical models of synaptic vesicle dynamics at a well-characterized model synapse, the Drosophila larval neuromuscular junction. This synapse's simplicity, accessibility to various electrophysiological recording and imaging techniques, and the genetic malleability intrinsic to Drosophila system make it ideal for computational and mathematical studies. We have employed a reductionist approach and started by modeling single presynaptic boutons. Synaptic vesicles can be divided into different pools; however, a quantitative understanding of their dynamics at the Drosophila neuromuscular junction is lacking [4]. We performed biologically realistic simulations of high and low release probability boutons [3] using partial differential equations (PDE) taking into account not only the evolution in time but also the spatial structure in two dimensions (the extension to three dimensions will be implemented soon). PDEs are solved using UG, a program library for the calculation of multi-dimensional PDEs solved using a finite volume approach and implicit time stepping methods leading to extended linear equation systems be solvedwith multi-grid methods [3,4]. Numerical calculations are done on multi-processor computers for fast calculations using different parameters in order to asses the biological feasibility of different models. In preliminary simulations, we modeled vesicle dynamics as a diffusion process describing exocytosis as Neumann streams at synaptic active zones. The initial results obtained with these models are consistent with experimental data. However, this should be regarded as a work in progress. Further refinements will be implemented, including simulations using morphologically realistic geometries which were generated from confocal scans of the neuromuscular junction using NeuRA (a Neuron Reconstruction Algorithm). Other parameters such as glutamate diffusion and reuptake dynamics, as well as postsynaptic receptor kinetics will be incorporated as well.
Poster presentation: Characterizing neuronal encoding is essential for understanding information processing in the brain. Three methods are commonly used to characterize the relationship between neural spiking activity and the features of putative stimuli. These methods include: Wiener-Volterra kernel methods (WVK), the spike-triggered average (STA), and more recently, the point process generalized linear model (GLM). We compared the performance of these three approaches in estimating receptive field properties and orientation tuning of 251 V1 neurons recorded from 2 monkeys during a fixation period in response to a moving bar. The GLM consisted of two formulations of the conditional intensity function for a point process characterization of the spiking activity: one with a stimulus only component and one with the stimulus and spike history. We fit the GLMs by maximum likelihood using GLMfit in Matlab. Goodness-of-fit was assessed using cross-validation with Kolmogorov-Smirnov (KS) tests based on the time-rescaling theorem to evaluate the accuracy with which each model predicts the spiking activity of individual neurons and for each movement direction (4016 models in total, for 251 neurons and 16 different directions). The GLMs that considered spike history of up to 35 ms, accurately predicted neuronal spiking activity (95% confidence intervals for KS test) with a performance of 97.0% (3895/4016) for the training data, and 96.5% (3876/4016) for the test data. If spike history was not considered, performance dropped to 73,1% in the training and 71.3% in the testing data. In contrast, the WVF and the STA predicted spiking accurately for 24.2% and 44.5% of the test data examples respectively. The receptive field size estimates obtained from the GLM (with and without history), WVF and STA were comparable. Relative to the GLM orientation tuning was underestimated on average by a factor of 0.45 by the WVF and the STA. The main reason for using the STA and WVF approaches is their apparent simplicity. However, our analyses suggest that more accurate spike prediction as well as more credible estimates of receptive field size and orientation tuning can be computed easily using GLMs implemented in Matlab with standard functions such as GLMfit.
Poster presentation: Introduction Rhythmic synchronization of neural activity in the gamma-frequency range (30–100 Hz) was observed in many brain regions; see the review in [1]. The functional relevance of these oscillations remains to be clarified, a task that requires modeling of the relevant aspects of information processing. The temporal correlation hypothesis, reviewed in [2], proposes that the temporal correlation of neural units provides a means to group the neural units into so-called neural assemblies that are supposed to represent mental objects. Here, we approach the modeling of the temporal grouping of neural units from the perspective of oscillatory neural network systems based on phase model oscillators. Patterns are assumed to be stored in the network based on Hebbian memory and assemblies are identified with phase-locked subset of these patterns. Going beyond foregoing discussions, we demonstrate the combination of two recently discussed mechanisms, referred to as "acceleration" [3] and "pooling" [4]. The combination realizes in a complementary manner a competition for activity on a local scale, while providing a competition for coherence among different assemblies on a non-local scale. ...

Neuroscientists, surgeons, and engineers have sought to understand the impact of anesthetics on the information processing in the brain and to properly assess the level of anesthesia in an non-invasive manner. Studies have indicated a more reliable depth of anesthesia (DOA) detection if multiple parameters are employed. Poster presentation: Functional connectivity of the brain describes the network of correlated activities of different brain areas. However, correlation does not imply causality and most synchronization measures do not distinguish causal and non-causal interactions among remote brain areas, i.e. determine the effective connectivity [1]. Identification of causal interactions in brain networks is fundamental to understanding the processing of information. Attempts at unveiling signs of functional or effective connectivity from non-invasive Magneto-/Electroencephalographic (M/EEG) recordings at the sensor level are hampered by volume conduction leading to correlated sensor signals without the presence of effective connectivity. Here, we make use of the transfer entropy (TE) concept to establish effective connectivity. The formalism of TE has been proposed as a rigorous quantification of the information flow among systems in interaction and is a natural generalization of mutual information [2]. In contrast to Granger causality, TE is a non-linear measure and not influenced by volume conduction. ...
Poster presentation: Our work deals with the self-organization [1] of a memory structure that includes multiple hierarchical levels with massive recurrent communication within and between them. Such structure has to provide a representational basis for the relevant objects to be stored and recalled in a rapid and efficient way. Assuming that the object patterns consist of many spatially distributed local features, a problem of parts-based learning is posed. We speculate on the neural mechanisms governing the process of the structure formation and demonstrate their functionality on the task of human face recognition. The model we propose is based on two consecutive layers of distributed cortical modules, which in turn contain subunits receiving common afferents and bounded by common lateral inhibition (Figure 1). In the initial state, the connectivity between and within the layers is homogeneous, all types of synapses – bottom-up, lateral and top-down – being plastic. During the iterative learning, the lower layer of the system is exposed to the Gabor filter banks extracted from local points on the face images. Facing an unsupervised learning problem, the system is able to develop synaptic structure capturing local features and their relations on the lower level, as well as the global identity of the person at the higher level of processing, improving gradually its recognition performance with learning time. ...
Poster presentation: Introduction We study the problem of object recognition invariant to transformations, such as translation, rotation and scale. A system is underdetermined if its degrees of freedom (number of possible transformations and potential objects) exceed the available information (image size). The regularization theory solves this problem by adding constraints [1]. It is unclear what constraints biological systems use. We suggest that rather than seeking constraints, an underdetermined system can make decisions based on available information by grouping its variables. We propose a dynamical system as a minimum system for invariant recognition to demonstrate this strategy. ...

NeuroXidence: reliable and efficient analysis of an excess or deficiency of joint-spike events

Poster presentation: We present a non-parametric and computationally-efficient method named NeuroXidence (see http://www.NeuroXidence.com ) that detects coordinated firing within a group of two or more neurons and tests whether the observed level of coordinated firing is significantly different from that expected by chance. NeuroXidence [1] considers the full auto-structure of the data, including the changes in the rate responses and the history dependencies in the spiking activity. We demonstrate that NeuroXidence can identify epochs with significant spike synchronisation even if these coincide with strong and fast rate modulations. We also show, that the method accounts for trial-by-trial variability in the rate responses and their latencies, and that it can be applied to short data windows lasting only tens of milliseconds. Based on simulated data we compare the performance of NeuroXidence with the UE-method [2,3] and the cross-correlation analysis. An application of NeuroXidence to 42 single-units (SU) recorded in area 17 of an anesthetized cat revealed significant coincident events of high complexities, involving firing of up to 8 SUs simultaneously (5 ms window). The results were highly consistent with those obtained by traditional pair-wise measures based on cross-correlation: Neuronal synchrony was strongest in stimulation conditions in which the orientation of the sinusoidal grating matched the preferred orientation of most of the SUs included in the analysis, and was the weakest when the neurons were stimulated least optimally. Interestingly, events of higher complexities showed stronger stimulus-specific modulation than pair-wise interactions. The results suggest strong evidence for stimulus specific synchronous firing and, therefore, support the temporal coding hypothesis in visual cortex. ...
Poster presentation: Introduction We here focus on constructing a hierarchical neural system for position-invariant recognition, which is one of the most fundamental invariant recognition achieved in visual processing [1,2]. The invariant recognition have been hypothesized to be done by matching a sensory image of a particular object stimulated on the retina to the most suitable representation stored in memory of the higher visual cortical area. Here arises a general problem: In such a visual processing, the position of the object image on the retina must be initially uncertain. Furthermore, the retinal activities possessing sensory information are being far from the ones in the higher area with a loss of the sensory object information. Nevertheless, with such recognition ambiguity, the particular object can effortlessly and easily be recognized. Our aim in this work is an attempt to resolve such a general recognition problem. ...
Poster presentation: Introduction We here address the problem of integrating information about multiple objects and their positions on the visual scene. A primate visual system has little difficulty in rapidly achieving integration, given only a few objects. Unfortunately, computer vision still has great difficultly achieving comparable performance. It has been hypothesized that temporal binding or temporal separation could serve as a crucial mechanism to deal with information about objects and their positions in parallel to each other. Elaborating on this idea, we propose a neurally plausible mechanism for reaching local decision-making for "what" and "where" information to the global multi-object recognition. ...

Poster presentation: NO-sensitive guanylyl cyclases (sGCs) are cytosolic receptors for nitric oxide (NO) catalyzing the conversion of GTP to cGMP. sGCs are obligate heterodimers composed of one alpha and beta subunit each. The allosteric mechanism of sGC activation via NO is well understood, however, our knowledge about alternative mechanisms such as protein-protein interactions regulating activity, availability, translocation and expression of sGC is rather limited. In a search by the yeast two-hybrid system using the catalytic domain of the alpha1 subunit as the bait, we have identified two structurally related proteins AGAP1 [1] and MRIP2 as novel sGC interacting proteins. MRIP2 is a multi-domain protein of 75 kDa comprising a single PH and ArfGAP domain each and two ankyrin repeats. Co-immunoprecipitation experiments using COS1 cells overexpressing both proteins demonstrated the interaction of MRIP2 with both subunits of the sGC alpha1beta1. Confocal microscopical analysis showed a prominent plasma membrane staining of MRIP2. This membrane association is mediated through an N-terminal myristoylation site and through binding of its PH domain to phospholipids such as phosphatidylinositol-3,5-bisphosphate (PI(3,5)P2). We hypothesize that MRIP2 may represent an acceptor protein for sGC that mediates recruitment of cytosolic sGC to the plasma membrane or other subcellular compartments.
Poster presentation: NO-sensitive guanylyl cyclases (GC) are the principal receptors for nitric oxide (NO) and convert GTP into the second messenger cGMP. We showed that GC is prone to tyrosine phosphorylation in COS1 cells overexpressing the human holoenzyme. Similar results were obtained in PC12 cells and in rat aortic tissue slices. The major phosphorylation site was mapped to position 192 in the regulatory domain of the beta1 subunit. Tyrosine phosphorylation of GC was reduced in the presence of the inhibitors PP1 and PP2 indicating that Src-like kinases are critically involved in phosphorylation. Moreover, co-immunoprecipitation experiments revealed an interaction between Src and GC. To further analyse the relevance of this posttranslational modification we generated a phospho-specific antibody raised against pTyr192. This antibody clearly distinguishes between phosphorylated and non-phosphorylated GC and may be a powerful tool to analyse the subcellular localisation of the phosphorylated enzyme.
Poster presentation NO-sensitive guanylyl cyclases (soluble guanylyl cyclase, sGC) are among the key regulators of intracellular cGMP concentration. The mechanisms underlying NO-mediated activation of sGC are quite well understood, however, little is known about the fine-tuning of sGC activity through alternative mechanisms such as protein phosphorylation. Several reports have demonstrated the reversible phosphorylation of sGC on serine/threonine residues, and it has been speculated, though not experimentally proven, that sGC might also be phosphorylated on tyrosine residues. Using broad-spectrum phosphatase inhibitors we were able to demonstrate tyrosine phosphorylation at Tyr192 of the beta 1 subunit of human sGC in COS1 cells. This residue forms part of a sequence segment (YEDL) representing a preferential binding site for SH2 domains of Src-like kinases. Pull-down assays and co-immunoprecipitation experiments showed that Src can indeed bind via its SH2 domain to pTyr192 of beta 1 indicating that tyrosine phosphorylation of sGC may be followed by recruitment of Src-like kinases to the phosphorylated beta 1 subunit. In support of this hypothesis, immunofluorescence studies showed a colocalization of overexpressed sGC and Src at the plasma membrane of COS1 and Hela cells. Together, our results point to an unexpected crosstalk between tyrosine kinase pathway(s) and the NO/cGMP signalling cascade which may result in translocation of the predominantly cytosolic sGC to the cytosolic face of the plasma membrane.

Gehölze an Fließgewässern prägen entscheidend die vielfältigen Lebensräume im Übergangsbereich Gewässer und seinem Umfeld. Die linienhafte Erstreckung des Gehölzsaums entlang eines Gewässers bietet zahlreiche Verteilungs- und Verbreitungsmöglichkeiten für Arten. In den Übergangsbereichen zwischen Wasser, Land und Luft entstehen durch die variierenden Wasserstände Verzahnungen unterschiedlicher Pflanzen- und Tiergesellschaften, die wichtige Bausteine im großräumigen Biotopverbund sind.

Gewässer sind Lebensadern unserer Landschaft. Sie prägen wesentlich den Naturhaushalt und das Landschaftsbild; die Entwicklung der Kulturlandschaft ist eng verbunden mit dem Ausbau der Gewässer. Die naturnahe Entwicklung der Gewässer und Auen ist eine Schwerpunktaufgabe der Umweltpolitik des Landes Baden-Württemberg. Sie bedarf der Unterstützung aller Beteiligten. Voraussetzung für zielgerichtetes Handeln sind fundiertes fachliches Wissen und Engagement.
Die vorliegende Schrift "Die heutige potentielle natürliche Vegetation an Fließgewässern " beinhaltet Informationen zur natürlichen Vegetationsentwicklung an Ufer- und in Auenbereichen der Fließgewässer. Es wird die Verbreitung der Vegetationsgesellschaften dargestellt, die unter den gegenwärtigen Standortbedingungen an den Ufern und in den Auen der Fließgewässer Baden-Württembergs ihr natürliches Vorkommen finden würden, würde der direkte menschliche Einfluß ab sofort nicht mehr gegenwärtig sein. Irreversible Veränderungen der standörtlichen Verhältnisse werden dabei aber berücksichtigt.
Einleitend wird ein Überblick über die Grundlagen typologischer Forschung an kleinen Fließgewässern in Baden-Württemberg gegeben. Der Ansatz einer regionalen Bachtypologie umschließt folgende Fachaspekte: Geologie und Geomorphologie, Gewässermorphologie und Abfluss, Hydrochemie, Gewässer- und Auenvegetation und Gewässerfauna. Zentraler Leitgedanke der typologischen Gliederung ist die Einteilung des Landes Baden-Württemberg in 11 Hauptfließgewässerlandschaften, die in erster Linie nach geologischgeomorphologischen Kriterien abgegrenzt wurden und naturräumliche Einheiten des Landes zusammenfassen. Von den 11 Hauptfließgewässerlandschaften wurden im Rahmen von zwei PW AB-Verbundforschungsvorhaben (" Projekt Wasser-Abfall-Boden") die folgenden vier auf methodisch einheitlicher Grundlage bearbeitet: Buntsandstein-Odenwald, Metamorphes Grundgebirge (Gneisschwarzwald), Berg- und Hügelland im Keuper, Oberrheinebene (holozäne Aufschüttungen und jungquartäre Schotterflächen).
Ein Schwerpunkt bei den Untersuchungen an der Enz ist die Dokumentation und Bewertung der Entwicklung der naturnahen Bauweisen gegenüber wechselnden hydraulischen Belastungen. Da weitere Umgestaltungsmaßnahmen in Baden-Württemberg geplant oder bereits ausgeführt worden sind, wird der Auswertung der Messungen und Beobachtungen durch Nachrechnungen zu den naturnahen Bauweisen an der Enz eine hohe Priorität eingeräumt. Die sich hieraus ergebenden allgemeingültigen Empfehlungen für die praxisbezogene Anwendung werden in dieser Schrift zusammengefasst.
Die Schaffung naturnaher Fließgewässer entzieht sich im Gegensatz zu technischen Bauwerken einer differenzierten, abschließenden Planbarkeit. Es können durch Planung und Ausführung lediglich günstige Vorraussetzungen für angestrebte Entwicklungen geschaffen werden. Innerhalb der von Maßnahmenträgern und Planern festgelegten Rahmenbedingungen soll sich die Natur entfalten können und dabei selbst gestaltend wirken.
Die vorliegende Schrift verfolgt das Ziel, die heterogene Diskussion um die ökologische Problematik der Verwendung von Hybrid-Pappeln im Gewässerbereich in Form einer Literaturstudie zusammenzufassen, um insbesondere für die Gewässerentwicklung und Gewässerunterhaltung Informationen, Argumente und Hinweise für den sachgerechten Umgang mit Pappeln zu liefern. Zitate zur historischen Behandlung der Pappeln werden in Erinnerung gebracht zur Vervollständigung des Themas in die Entwicklungszusammenhänge.

Fachliche Exzellenz und Bildungsnotstand – diese beiden Extreme beherrschen gegenwärtig die Diskussion um Schul- und Hochschulausbildung. Die Universität Frankfurt stellt sich der Elitediskussion und setzt auf Fokussierung und Schwerpunktbildung. Studiengänge werden modifiziert, die Art und Vielfalt möglicher Abschlüsse internationalen Standards angepasst. Die Universität will und wird wettbewerbsfähig sein, auch im internationalen Vergleich. Darüber sprach Dr. Monika Mölders mit Prof. Dr. Günther Wess, Honorarprofessor der Universität Frankfurt, Forschungsleiter Europa von Aventis und Mitglied der Geschäftsführung der Aventis Pharma Deutschland GmbH.
Im Zuge der steigenden Bedeutung der Proteomforschung und der »Molekularisierung« der Medizin werden neue, effizientere Plattformen zur Untersuchung von Proteinen und deren Wechselwirkungen notwendig. Hier bietet die Nanotechnologie, eine Wissenschaft mit Ursprüngen in der Physik und der Halbleiterindustrie, attraktive Lösungsperspektiven. Ein Bereich der Forschung am Institut für Biochemie der Universität Frankfurt um Prof. Dr. Robert Tampé widmet sich den Aspekten der Nanotechnologie zur Entwicklung von Protein-Chips für die Proteomforschung und Erzeugung von Mustern im Kleinstformat.
Die Depression gehört zu den häufigsten Volkskrankheiten. Derzeit sind rund vier Millionen Deutsche an einer behandlungsbedürftigen Depression erkrankt. Die Erkrankung verläuft typischerweise in Form von Episoden, die Wochen bis Monate, manchmal auch Jahre anhalten können. Wenn die Erkrankung unbehandelt bleibt, kann sie wiederkehren und einen chronischen Verlauf nehmen. Rund 75 Prozent der Betroffenen erleiden nach einer Ersterkrankung innerhalb von fünf Jahren mindestens eine neue depressive Phase. Zudem werden mit steigender Episodenzahl die episodenfreien Zwischenzeiten immer kürzer. Es gilt heute als unstrittig, dass mehr als die Hälfte aller Depressionen nicht diagnostiziert und allenfalls ein Fünftel adäquat behandelt werden. Das verursacht nicht nur enorme Kosten für die Volkswirtschaft, sondern ist für die Betroffenen auch mit erheblichem Leid und Lebensgefahr verbunden.
In der heutigen Zeit werden an die Beobachtung von Natur und Umwelt zunehmend vielfältigere Ansprüche gestellt. Datenerhebungsprojekte müssen heute nicht nur den sektoralen Ansprüchen von einzelnen Teildisziplinen des Naturschutzes innerhalb des Landes genügen. Immer häufiger ist eine Zusammenfassung verschiedenster Projektergebnisse zur Aufgabenerfüllung der Natur- und Umweltschutzverwaltung erforderlich. Dies zeigt sich beispielsweise bei der Auswahl von der nach der Fauna-Flora-Habitat-Richtlinie besonders zu schützenden Gebiete. Für die Gebietsauswahl mussten die Ergebnisse von Biotopkartierungen und Artenerhebungen sowie die Informationen über die geschützten Gebiete nach Naturschutzrecht zusammenfassend ausgewertet werden. Umfassende und interdisziplinäre Aussagen über den Zustand von Natur und Umwelt sind auch für die Umsetzung der Vereinbarungen der Konferenz der Vereinten Nationen für Umwelt und Entwicklung von Rio de Janeiro (Agenda 21, Konvention über die biologische Vielfalt) erforderlich. Für die Aufgabenerfüllung des Naturschutzes sind somit umfangreiche Datenbestände über Zustand und Entwicklung von unseren Landschaften und den darin lebenden Arten notwendig. Daten, die jedoch mit verschiedenen Schlüsseln erhoben werden, können zumeist nur mit erhöhtem Aufwand oder häufig überhaupt nicht zusammengeführt und ausgewertet werden. Um landesweite oder auch regionale Übersichten über den Zustand von Natur und Landschaft erstellen zu können, müssen daher die Informationen des Naturschutzes wie Vorkommen und Verbreitung von Arten und Biotopen oder Art und Grad der Landnutzung soweit wie möglich mit einheitlichen Erhebungsparametern erfasst werden. Nur hierdurch können die Erhebungsprojekte des Naturschutzes den Anforderungen der Naturschutzpolitik des Landes, des Bundes und den internationaler Verpflichtungen gerecht werden.
Background: Mitochondrial DNA sequencing increasingly results in the recognition of genetically divergent, but morphologically cryptic lineages. Species delimitation approaches that rely on multiple lines of evidence in areas of co-occurrence are particularly powerful to infer their specific status. We investigated the species boundaries of two cryptic lineages of the land snail genus Trochulus in a contact zone, using mitochondrial and nuclear DNA marker as well as shell morphometrics.
Results: Both mitochondrial lineages have a distinct geographical distribution with a small zone of co-occurrence. In the same area, we detected two nuclear genotype clusters, each being highly significantly associated to one mitochondrial lineage. This association however had exceptions: a small number of individuals in the contact zone showed intermediate genotypes (4%) or cytonuclear disequilibrium (12%). Both mitochondrial lineage and nuclear cluster were statistically significant predictors for the shell shape indicating morphological divergence. Nevertheless, the lineage morphospaces largely overlapped (low posterior classification success rate of 69% and 78%, respectively): the two lineages are truly cryptic.
Conclusions: The integrative approach using multiple lines of evidence supported the hypothesis that the investigated Trochulus lineages are reproductively isolated species. In the small contact area, however, the lineages hybridise to a limited extent. This detection of a hybrid zone adds an instance to the rare reported cases of hybridisation in land snails.
Breaking tolerance to the natural human liver autoantigen cytochrome P450 2D6 by virus infection

Autoimmune liver diseases, such as autoimmune hepatitis (AIH) and primary biliary cirrhosis, often have severe consequences for the patient. Because of a lack of appropriate animal models, not much is known about their potential viral etiology. Infection by liver-tropic viruses is one possibility for the breakdown of self-tolerance. Therefore, we infected mice with adenovirus Ad5 expressing human cytochrome P450 2D6 (Ad-2D6). Ad-2D6–infected mice developed persistent autoimmune liver disease, apparent by cellular infiltration, hepatic fibrosis, “fused” liver lobules, and necrosis. Similar to type 2 AIH patients, Ad-2D6–infected mice generated type 1 liver kidney microsomal–like antibodies recognizing the immunodominant epitope WDPAQPPRD of cytochrome P450 2D6 (CYP2D6). Interestingly, Ad-2D6–infected wild-type FVB/N mice displayed exacerbated liver damage when compared with transgenic mice expressing the identical human CYP2D6 protein in the liver, indicating the presence of a stronger immunological tolerance in CYP2D6 mice. We demonstrate for the first time that infection with a virus expressing a natural human autoantigen breaks tolerance, resulting in a chronic form of severe, autoimmune liver damage. Our novel model system should be instrumental for studying mechanisms involved in the initiation, propagation, and precipitation of virus-induced autoimmune liver diseases.

Früher wurden diese fliegenden Säugetiere deshalb mit Dämonen und Zauberei in Zusammenhang gebracht. Unsere heimischen Fledermäuse haben damit nichts gemein. Im Gegenteil – da sie sich ausschließlich von Insekten und anderen Gliedertieren ernähren, stehen sie sogar in unserem Dienst, denn viele der in der Dunkelheit fliegenden Insekten sind in der Land- und Forstwirtschaft nicht gern gesehen und den Menschen häufig lästig. Da die meisten der insektenfangenden Vögel nur bei Tage jagen, bleiben die Nachtinsekten von ihnen so gut wie verschont. Durch die nächtliche Jagd füllen die Fledermäuse diese „Lücke“ im Haushalt der Natur. In Baden-Württemberg gelten insgesamt 23 Fledermausarten als heimisch. Die Nymphenfledermaus als 23. Art wurde erst 2005 entdeckt. Derzeit sind von 19 Arten Fortpflanzungsvorkommen bekannt. Viele sind als sehr selten einzustufen. Innerhalb der letzten rund 50 Jahre sind die Fledermausbestände bei uns zum Teil dramatisch zurückgegangen – und dies, obwohl alle 23 Arten unter Naturschutz stehen. Dieses Heft möchte Ihnen die Lebensweise der Fledermäuse näher bringen und Schutzmöglichkeiten zu einer langfristigen Sicherung der Fledermausbestände erläutern.
Sträucher und Bäume in der freien Landschaft zu pflanzen – dafür gibt es viele Gründe: Ufersicherung an Bächen und Flüssen, Böschungsbefestigung, Lärm- und Sichtschutz an Straßen und Wegen, Einbindung von Bauwerken, Belebung der Landschaft, Gliederung der Feldflur, Abschirmung sensibler Biotope oder die Entwicklung von Lebensstätten für Pflanzen- und Tierarten. So unterschiedlich wie die Gründe für eine Gehölzpflanzung, so unterschiedlich sind auch die Anforderungen an die zu pflanzenden Gehölzarten. Im einen Fall sollen es raschwüchsige, Sichtschutz bietende Gehölze sein, im anderen überflutungstolerante, im nächsten sollen Sträucher wichtige Nahrungspflanzen für die Tierwelt sein. Stets gilt aber der Grundsatz, dass die Pflanzung von Gehölzen nicht durch Einbringung naturraumfremder Arten zu einer Beeinträchtigung der heimischen Pflanzen- und Tierwelt führen darf oder die Eigenart der Landschaft verändert wird. Bei der Auswahl der Gehölzarten sind daher andere Aspekte von Bedeutung als im besiedelten Bereich, in Parks und Gärten, wo ästhetische oder individuelle Gesichtspunkte Vordergrund stehen können. Bei Pflanzungen in der offenen Landschaft ist dagegen ein besonderes Augenmerk auf die Verwendung naturraumtypischer Arten und auf die Herkunft des Pflanzguts zu werfen.
Welche Tiere und Pflanzen es auf unserer Erde überhaupt gibt und wie sie verbreitet sind, ist eine der Kardinalfragen biologischer Grundlagenforschung. Diese Erkenntnis hat sich gerade in den letzten Jahren auch in der Öffentlichkeit durchgesetzt und Biodiversität ist schon fast ein Modewort geworden. Die ungeheure Artenvielfalt der Insekten und hier auch ganz besonders der Käfer kann ohne die Mithilfe der sogenannten Liebhaber oder Amateure nicht erfasst werden. Solche Liebhaber sind eben nicht verschrobene Gestalten, die mit Schmetterlingsnetz und Botanisiertrommel durch Wald und Wiese streifen, sondern sie sind unverzichtbare Grundlagenforscher, die die Basis schaffen für alle weiterführenden biologischen Untersuchungen, wie beispielsweise über Bestandsveränderungen, Vergleich von Ökosystemen oder praktische Naturschutzarbeit. Dass dem so ist, haben Jürgen Frank und Eberhard Konzelmann mit dem vorliegenden Grundlagenwerk über die Käfer Baden-Württembergs eindruckvoll und beispielhaft bestätigt. Die Käfer sind eine der größten Organismen-Gruppen, die wir auf unserem Planeten kennen und sie besitzen eine große ökologische Plastizität in allen terrestrischen Lebensräumen, nur im Meer fehlen sie. Damit besitzen sie auch eine gute Indikatorfunktion und die Untersuchung der Käfergemeinschaften lässt auch Rückschlüsse auf andere Tier- und Pflanzengruppen zu. Das neue Verzeichnis belegt den aktuellen Bestand von rund 4800 Käferarten in Baden-Württemberg auf der Basis abgesicherter und nachprüfbarer Fundnachweise in den letzten 50 Jahren. Das sind mehr als 3/4 aller in Deutschland vorkommenden Arten. Die Arten werden nicht nur aufgelistet, sondern es wird die genaue Anzahl der Funde in den großen Naturräumen des Landes (Rheinebene, Schwarzwald, Neckarland, Schwäbische Alb und Oberschwaben) differenziert dokumentiert.
In vorliegender Veröffentlichung wird auf der Grundlage der bisherigen Erfahrungen beim Einsatz von Pferden im Naturschutz und in der Landschaftspflege die Eignung von Pferden in der Biotoppflege beschrieben. Nach einer kurzen Beschreibung der Einsatzbereiche von Pferden im Natur- und Landschaftsschutz (Verwertung von Extensivheu, Offenhaltung der traditionellen Kulturlandschaft, Einsatz in großflächigen Beweidungsprojekten, Biotoppflege mit dem Ziel des Arten- und Biotopschutzes) werden verschiedene Aspekte der Weidehaltung vorgestellt, die für den Einsatz von Pferden in der Biotoppflege wichtig sind. Dabei wird das arteigene Weideverhalten der Pferde (Selektivität, Geilstellen, Verbiss, Trittwirkung) beschrieben, auf Aspekte der Tierhaltung und Tiergesundheit eingegangen sowie die unterschiedliche Eignung der verschiedenen Pferderassen zur Biotoppflege dargestellt. Ausführlich werden spezielle Formen des Weidemanagements vorgestellt, bei denen eine kurzzeitige Umtriebsweide mit ein bis zwei Wochen Weidegang im Vordergrund steht. Daneben wird eine modifizierte Form der Portionsweide sowie eine ebenfalls zur Biotoppflege geeignete Form der Langzeitweide beschrieben. Auf die Bedeutung einer regelmäßigen Weidepflege in Form von Nachmahd, Mulchen und Entbuschung wird bei den aufgeführten Weideformen hingewiesen. Die Ergebnisse einer landesweiten Umfrage beim behördlichen Naturschutz Kreisbehörden, Regierungspräsidien), den Landwirtschaftsämtern und den Naturschutzverbänden werden kurz vorgestellt. Es zeigt sich, dass bisher nur sehr wenige konkrete Pflegeprojekte mit Pferden durchgeführt werden und trotz einiger Vorbehalte gegenüber Pferdebeweidung bei diesen Institutionen ein Interesse an ausführlicher Information besteht. Parallel zur Umfrage bei den Behörden wurde eine Umfrage bei einer Auswahl von Pferdehaltern durchgeführt, aus der abzuleiten ist, dass das Interesse dieser Gruppe an einer Teilnahme an Biotoppflege-Projekten in Baden-Württemberg groß ist. Auf der Grundlage der allgemein gültigen Richtlinien zum Weidemanagement sowie einer umfassenden Literatur- und Projekt-Recherche werden im zweiten Teil des Leitfadens für alle landwirtschaftlich nutzbaren Offenland-Grünland-Biotoptypen im Einzelnen Empfehlungen gegeben, ob und wie sich diese Biotope mit einer Pferdebeweidung erhalten und gegebenenfalls entwickeln lassen. Es wird auf die erforderlichen Besatzdichten, Weidezeiten und Maßnahmen der Weidepflege ebenso eingegangen wie auf Aspekte des speziellen Artenschutzes. Besonders berücksichtigt werden dabei die im Anhang I der Flora-Fauna-Habitat(FFH)-Richtlinie aufgeführten Lebensräume und die nach § 32 (bisher § 24a) Naturschutzgesetz Baden-Württemberg (NatSchG) besonders geschützten Biotope.
Im Zusammenhang mit dem agrarstrukturellen Wandel der letzten Jahrzehnte stellt sich aus Sicht des Naturschutzes immer wieder die Frage, wie dem Rückgang traditioneller Landnutzung an Grenzstandorten und in benachteiligten Gebieten begegnet werden kann. Um eine Offenhaltung der Landschaft und Pflege schutzwürdiger Biotope zu gewährleisten, ist die Beweidung mit Rindern oder Schafen eine vielfach eingesetzte Methode. Pferde wurden hingegen unter dem Aspekt von Naturschutz und Landschaftspflege kaum berücksichtigt, obwohl ihre Zahl in den letzten Jahrzehnten aufgrund steigender Freizeitnutzung in Deutschland erheblich zugenommen hat. Das vorliegende Merkblatt richtet sich vor allem an Landwirte und Pferdehalter, soll aber auch den unteren Naturschutzbehörden sowie Gemeindeverwaltungen, Planungsbüros und Verbänden als Arbeitshilfe dienen. Pferdehaltung lässt sich auf verschiedene Weise mit den Zielen von Naturschutz und Landschaftspflege vereinbaren. So wird spät geworbenes Heu extensiv genutzter Wiesen in der Pferdehaltung bereits vielfach und bevorzugt eingesetzt. Naturgemäße Pferdebeweidung trägt ebenso wie die Beweidung mit anderen Tierarten zur Offenhaltung der traditionellen Kulturlandschaft bei. Auch in den in jüngerer Zeit zunehmenden großflächigen Beweidungsprojekten vieler Bundesländer werden neben Rindern gerne ursprüngliche Pferderassen eingesetzt. Um den speziellen Anforderungen des Arten- und Biotopschutzes gerecht zu werden, genügt es jedoch nicht, eine naturverträgliche extensive Landnutzung und eine unter tierhalterischen Aspekten sachgemäße Beweidung durchzuführen. Es ist ein gezieltes Weidemanagement notwendig. Aus diesem Anlass wurde im Auftrag der Landesanstalt für Umweltschutz (heute LUBW Landesanstalt für Umwelt, Messungen und Naturschutz Baden-Württemberg) eine Studie erstellt. Die Ergebnisse wurden in einer umfangreichen Dokumentation im Internet veröffentlicht und bilden die Grundlage dieses Merkblattes.
Die hier vorgelegte Liste ist als Ergänzung des dreibändigen Grundlagenwerkes „Die Moose Baden-Württembergs“ zu sehen (NEBEL & PHILIPPI 2000, 2001, 2005). In diesem ausführlichen Werk wurde erstmals für alle im Bundesland vorkommenden Moose eine Einstufung in verschiedene Gefährdungskategorien vorgenommen. Diese Einstufungen sind in der vorliegenden Roten Liste zusammengefasst. Da das Erscheinen der ersten beiden Bände des Grundlagenwerks bereits einige Jahre zurückliegt, ergaben sich inzwischen zahlreiche neue Funde zu einzelnen Sippen. Einige Taxa wurden in der letzten Zeit neu für Baden-Württemberg entdeckt, sie fehlen daher in NEBEL & PHILIPPI (2000, 2001), andere mussten aus dem Arteninventar gestrichen werden. Mehrere Sippen, die im Grundlagenwerk noch landesweit als verschollen oder ausgestorben eingestuft wurden (RL 0), ließen sich neuerdings wieder nachweisen. Das Artenverzeichnis wurde daher entsprechend aktualisiert. Ebenso wurden die Gefährdungseinstufungen dem aktuellen Kenntnisstand angepasst. Veränderungen gegenüber dem Grundlagenwerk sind unter anderem in den „Anmerkungen zu einzelnen Sippen“ (siehe Kapitel 13) erläutert. Zu wichtigen neuen Funden werden außerdem genauere Daten mitgeteilt (Fundstelle oder Fundregion, Zeitpunkt der Beobachtung und Finder).
Die vorliegende Veröffentlichung umfasst zwei Grundbausteine. Zum einen die offizielle Rote Liste mit Nennung der Gefährdungskategorien, zum anderen ein revidiertes systematisches Gesamtartenverzeichnis der Mollusken Baden-Württembergs. Die Rote Liste dient zum schnellen Feststellen der jeweiligen Gefährdungskategorien der einzelnen Arten in Baden-Württemberg und ist wie üblich alphabetisch nach Gattungen geordnet. Sehr großer Wert wurde auf die sorgfältige Analyse der Ergebnisse gelegt (Kapitel 7). Das Gesamtartenverzeichnis dient der aktuellen systematischen Einordnung aller Arten, weshalb hier die Taxa im Kontext des wissenschaftlichen Systems der Mollusken aufgeführt werden. Im systematischen Artenverzeichnis soll der momentane Kenntnisstand über die Mollusken Baden-Württembergs in knapper Darstellung zum Ausdruck kommen. Hier sind auch die bekannten Unterarten aufgeführt und es werden zusätzliche Informationen zum Verbreitungstyp, zur Verbreitung (Vorkommen in den Naturräumen 3. Ordnung) sowie zur Ökologie (Zuordnung einzelner Arten zu bestimmten Biotoptypen) gegeben. Mit diesen Zusatzinformationen werden Rote Listen und Artenverzeichnisse zu Gradmessern der Biodiversitätsforschung. In über 130 ‚Anmerkungen‘ werden die entsprechenden Angaben zur Systematik, Verbreitung und Ökologie präzisiert und es wird auf die hierfür zu Grunde liegende Literatur verwiesen. Alle Angaben der Roten Liste sind auch im ausführlichen systematischen Artenverzeichnis enthalten. In beiden Listen sind die Arten mit ihrer laufenden Nummer aufgeführt. Damit ist ein problemloser Wechsel von der Roten Liste zu den Angaben im systematischen Artenverzeichnis gewährleistet. Der Forschungsstand findet sich vielfach in der historischen Literatur, die deshalb eine sorgfältige und kritische Berücksichtigung erfuhr (siehe Anmerkungen und Literaturverzeichnis). Einen unschätzbaren Wert haben in diesem Zusammenhang die zahlreichen Veröffentlichungen David Geyer‘s, die den Beginn der modernen Regionalfaunisik in Baden-Württemberg kennzeichnen. Ein eigenes Kapitel zur Forschungsgeschichte hätte jedoch den vorgegebenen Rahmen dieser Arbeit gesprengt.
In der jüngsten Vergangenheit hat die Anwendung der inzwischen veralteten Liste zunehmend Probleme bereitet und zwar nicht nur im Zusammenhang mit der Begründung von Naturschutzmaßnahmen, sondern auch bei Bewertungen von Flächen z.B. im Rahmen von Umweltverträglichkeitsstudien und Eingriffsplanungen. Die Diskrepanz zwischen einer früheren Rote-Liste-Einstufung und der heutigen, auf erweiterten Erkenntnissen beruhenden Bewertung der Bestandssituation bestimmter Arten muss unweigerlich zu falschen Schlüssen hinsichtlich der Wertigkeit der zu beurteilenden Flächen führen (vgl. WESTRICH & SCHWENNINGER 1997). Es war deshalb sehr zu begrüßen, dass die Landesanstalt für Umweltschutz Karlsruhe eine Neubearbeitung ermöglicht hat. Dabei sollte gleichzeitig auch den zwischenzeitlich vom Bundesamt für Naturschutz modifizierten Gefährdungskategorien und -kriterien Rechnung getragen und die Fülle an neuen wissenschaftlichen Erkenntnissen integriert werden. Diese waren, soweit möglich, bereits in der Roten Liste der Bienen Deutschlands (WESTRICH et al. 1998) berücksichtigt worden. Da eine Rote Liste stets das im Bezugsraum nachgewiesene Artenspektrum zur Grundlage haben muss, war es erforderlich, auch die Faunenliste im Vorfeld zu aktualisieren, zumal seit dem Erscheinen des Grundlagenwerks zahlreiche Arten erstmals in Baden-Württemberg aufgefunden werden konnten.
In Baden-Württemberg sind in dem 30-jährigen Zeitraum einschließlich dervorliegenden insgesamt fünf Fassungen der Roten Liste der gefährdeten Vogelarten erschienen, die jeweils auf den neuesten Stand der Erforschung der Vogelwelt Baden-Württembergs gebracht wurden. Die einzelnen Fassungen der Roten Liste sind 1973 (1. Fassung, Berthold, Ertel & Hölzinger 1974, 1975), 1977 (2. Fassung, Berthold, Ertel, Hölzinger, Kalchreuter & Ruge 1977), 1981 (3. Fassung, Hölzinger, Berthold, Kroymann & Ruge 1981), 1996 (4. Fassung, Hölzinger, Berthold, König & Mahler 1996) und 2007 (5., vorliegende Fassung) herausgegeben worden. In diesem über 30-jährigen Zeitraum wurden die Kriterien für die Roten Listen entsprechend dem Fortschritt der ornithologischen Forschung zunehmend mehr auf quantitative Grundlagen gestellt. Die Roten Listen waren und sind das Ergebnis systematischer und programmatisch orientierter Bestandsaufnahmen der Vogelwelt abseits emotionaler und naturschutzpolitischer Beurteilung.
Hiermit liegt nun nach 16 Jahren die 3. Fassung der Roten Liste der Farn- und Blütenpflanzen vor. Für diese Aktualisierung gibt es vor allem zwei Gründe: Die Gefährdungssituation hat sich bei vielen Sippen seit 1983 verändert und das Wissen zur Flora Baden-Württembergs ist deutlich angewachsen, vor allem durch die Bearbeitung des achtbändigen Grundlagenwerks „Die Farn- und Blütenpflanzen Baden-Württembergs“ (SEBALD & al. 1990-1998). Die 3. Fassung der Roten Liste enthält einige Neuerungen: Erstmals wurden zusätzlich Gefährdungseinstufungen für naturräumliche Regionen vorgenommen und es wurden alle in Baden-Württemberg etablierten Arten und Unterarten aufgelistet, auch die ungefährdeten. Um die Vergleichbarkeit mit der entsprechenden bundesdeutschen Roten Liste zu gewährleisten (KORNECK, SCHNITTLER & VOLLMER 1996; SCHNITTLER & LUDWIG 1996), waren einige Änderungen bei den Definitionen von Gefährdungskategorien, vor allem aber die Einführung neuer Kategorien notwendig. Eigene Kategorien für Sippen, für die nicht genügend Informationen zur Beurteilung der Bestandessituation vorliegen, ermöglichen es, den Wissensstand genauer als bisher zu dokumentieren. Die Autoren hoffen, dadurch den Gebrauchswert der Roten Liste erhöht zu haben, möchten zugleich aber anregen, mehr als bisher neben der Gefährdung auch andere Aspekte, beispielsweise pflanzengeographischer Art, bei der Bewertung von Pflanzenvorkommen zu berücksichtigen, denn nicht nur Bestände gefährdeter Arten sind schützenswert!
The C-module-binding factor (CbfA) is a multidomain protein that belongs to the family of jumonji-type (JmjC) transcription regulators. In the social amoeba Dictyostelium discoideum, CbfA regulates gene expression during the unicellular growth phase and multicellular development. CbfA and a related D. discoideum CbfA-like protein, CbfB, share a paralogous domain arrangement that includes the JmjC domain, presumably a chromatin-remodeling activity, and two zinc finger-like (ZF) motifs. On the other hand, the CbfA and CbfB proteins have completely different carboxy-terminal domains, suggesting that the plasticity of such domains may have contributed to the adaptation of the CbfA-like transcription factors to the rapid genome evolution in the dictyostelid clade. To support this hypothesis we performed DNA microarray and real-time RT-PCR measurements and found that CbfA regulates at least 160 genes during the vegetative growth of D. discoideum cells. Functional annotation of these genes revealed that CbfA predominantly controls the expression of gene products involved in housekeeping functions, such as carbohydrate, purine nucleoside/nucleotide, and amino acid metabolism. The CbfA protein displays two different mechanisms of gene regulation. The expression of one set of CbfA-dependent genes requires at least the JmjC/ZF domain of the CbfA protein and thus may depend on chromatin modulation. Regulation of the larger group of genes, however, does not depend on the entire CbfA protein and requires only the carboxy-terminal domain of CbfA (CbfA-CTD). An AT-hook motif located in CbfA-CTD, which is known to mediate DNA binding to A+T-rich sequences in vitro, contributed to CbfA-CTD-dependent gene regulatory functions in vivo.
Mammalian retinae have rod photoreceptors for night vision and cone photoreceptors for daylight and colour vision. For colour discrimination, most mammals possess two cone populations with two visual pigments (opsins) that have absorption maxima at short wavelengths (blue or ultraviolet light) and long wavelengths (green or red light). Microchiropteran bats, which use echolocation to navigate and forage in complete darkness, have long been considered to have pure rod retinae. Here we use opsin immunohistochemistry to show that two phyllostomid microbats, Glossophaga soricina and Carollia perspicillata, possess a significant population of cones and express two cone opsins, a shortwave-sensitive (S) opsin and a longwave-sensitive (L) opsin. A substantial population of cones expresses S opsin exclusively, whereas the other cones mostly coexpress L and S opsin. S opsin gene analysis suggests ultraviolet (UV, wavelengths <400 nm) sensitivity, and corneal electroretinogram recordings reveal an elevated sensitivity to UV light which is mediated by an S cone visual pigment. Therefore bats have retained the ancestral UV tuning of the S cone pigment. We conclude that bats have the prerequisite for daylight vision, dichromatic colour vision, and UV vision. For bats, the UV-sensitive cones may be advantageous for visual orientation at twilight, predator avoidance, and detection of UV-reflecting flowers for those that feed on nectar.
The CUG-binding protein 1 (CUG-BP1) is a member of the CUG-BP1 and ETR-like factors (CELF) family or the Bruno-like family and is involved in the control of splicing, translation and mRNA degradation. Several target RNA sequences of CUG-BP1 have been predicted, such as the CUG triplet repeat, the GU-rich sequences and the AU-rich element of nuclear pre-mRNAs and/or cytoplasmic mRNA. CUG-BP1 has three RNA-recognition motifs (RRMs), among which the third RRM (RRM3) can bind to the target RNAs on its own. In this study, we solved the solution structure of the CUG-BP1 RRM3 by hetero-nuclear NMR spectroscopy. The CUG-BP1 RRM3 exhibited a noncanonical RRM fold, with the four-stranded b-sheet surface tightly associated with the N-terminal extension. Furthermore, we determined the solution structure of the CUG-BP1 RRM3 in the complex with (UG)3 RNA, and discovered that the UGU trinucleotide is specifically recognized through extensive stacking interactions and hydrogen bonds within the pocket formed by the b-sheet surface and the N-terminal extension. This study revealed the unique mechanism that enables the CUG-BP1 RRM3 to discriminate the short RNA segment from other sequences, thus providing the molecular basis for the comprehension of the role of the RRM3s in the CELF/Bruno-like family.
Background Although current molecular clock methods offer greater flexibility in modelling historical evolutionary events, calibration of the clock with dates from the fossil record is still problematic for many groups. Here we implement several new approaches in molecular dating to estimate evolutionary ages of Lacertidae, an Old World family of lizards with a poor fossil record and uncertain phylogeny. Four different models of rate variation are tested in a new program for Bayesian phylogenetic analysis called TreeTime, based on a combination of mitochondrial and nuclear gene sequences. We incorporate paleontological uncertainty into divergence estimates by expressing multiple calibration dates as a range of probabilistic distributions. We also test the reliability of our proposed calibrations by exploring effects of individual priors on posterior estimates. Results According to the most reliable model, as indicated by Bayes factor comparison, modern lacertids arose shortly after the K/T transition and entered Africa about 45 million years ago, with the majority of their African radiation occurring in the Eocene and Oligocene. Our findings indicate much earlier origins for these clades than previously reported, and we discuss our results in light of paleogeographic trends during the Cenozoic. Conclusions This study represents the first attempt to estimate evolutionary ages of a specific group of reptiles exhibiting uncertain phylogenetic relationships, molecular rate variation and a poor fossil record. Our results emphasize the sensitivity of molecular divergence dates to fossil calibrations, and support the use of combined molecular data sets and multiple, well-spaced dates from the fossil record as minimum node constraints. The bioinformatics program used here, TreeTime, is publicly available, and we recommend its use for molecular dating of taxa faced with similar challenges.
Die vorliegende Rote Liste sowie das Artenverzeichnis der Köcherfliegen (Trichoptera) Baden-Württembergs basieren im Wesentlichen auf den Ergebnissen einer seit 1993 im Auftrag der Landesanstalt für Umweltschutz Baden-Württemberg durchgeführten landesweiten Erfassung der Köcherfliegenfauna sowie einer umfangreichen Literaturrecherche. Die Einstufung in die Gefährdungskategorien der Roten Liste ist damit das Resultat einer Bewertung der Bestandssituation der in Baden-Württemberg vorkommenden Köcherfliegenarten.
Vor über 10 Jahren wurde die erste Fassung einer Roten Liste und Checkliste gefährdeter Laufkäferarten Baden-Württembergs publiziert (TRAUTNER 1992), eine zweite Fassung, getrennt in Rote Liste und Artenverzeichnis, folgte 1996 (TRAUTNER 1996a, TRAUTNER & BRÄUNICKE 1996). Seit dieser Zeit hat sich der Kenntnisstand zu Verbreitung, Habitatansprüchen und Gefährdungssituation der Laufkäferarten in Baden-Württemberg wesentlich verbessert. Eine Neufassung der Roten Liste als wichtiges Instrument für die Praxis in Naturschutz und Landschaftsplanung war daher dringend erforderlich.
Schwebfliegen kommen in großer Zahl in fast allen terrestrischen Lebensräumen vor. Dennoch spielen sie bisher in der praktischen Naturschutzarbeit eine untergeordnete Rolle. Verglichen mit anderen Tiergruppen, wie den Tagfaltern, Laufkäfern oder Wildbienen, werden sie nur selten im Rahmen raumrelevanter Planungen berücksichtigt. Dabei decken sie wie keine andere der häufiger untersuchten Gruppen ein breites Spektrum unterschiedlicher Lebensweisen ab. Während die Imagines der meisten Arten eifrige Blütenbesucher sind und eine wichtige Funktion als Bestäuber ausüben, zeichnen sich die Larven durch eine hohe Diversität von Lebensstrategien aus. Die phytophagen Arten minieren in Stängeln, Wurzeln oder Blättern, befallen unterirdische Speicherorgane von Pflanzen oder zapfen das Kambium von Nadelbäumen an. Manche Arten leben in den Fruchtkörpern von Pilzen. Saprophage Arten nutzen abgestorbene feuchte Pflanzen, an organischem Material reiche Gewässer (Pfützen, Teiche, wassergefüllte Baumhöhlen, etc.), leben in Schleimflüssen von Bäumen, in sich zersetzendem Holz, in Säugerkot oder ernähren sich vom Abfall in Wespen- und Hummelnestern. Die zoophagen Vertreter fressen Blattläuse, Raupen, Wespen- oder Ameisenbrut. Schwebfliegen findet man in praktisch allen terrestrischen Lebensräumen, im Wald ebenso wie auf Äckern und im Grünland, auf Sandrasen wie im Hochmoor. In Mitteleuropa sind die meisten Arten eher in frischen bis feuchten Lebensräumen zu finden, Wälder und andere gehölzreiche Lebensräume sind artenreicher als ganz offene Biotope. Einen für interessierte Laien geschriebenen Überblick über die Lebensweise der Schwebfliegen hat SCHMID (1996) veröffentlicht.
Benthische Algen sind ein wesentlicher Teil des Ökosystems der Fließgewässer. Verschiedene Verfahren nutzen sie zur Bioindikation. Eines davon ist das PHYLIB-Verfahren (SCHAUMBURG et al. 2004, 2005) zur Bewertung des ökologischen Zustandes. In diesem Verfahren wird die Qualitätskomponente Makrophyten und Phytobenthos in drei Teilkomponenten gegliedert. Neben Makrophyten incl. Charales und Diatomeen werden die anderen Algenklassen als "übriges" Phytobenthos einbezogen. Das "übrige" Phytobenthos zeichnet sich in mehrerlei Hinsicht durch eine besondere Vielfalt aus. Dieser Feldführer versucht, eine erste Orientierung zu vermitteln. Dazu gehört eine kurz gehaltene Darstellung der relevanten Algenklassen im systematischtaxonomischen Teil, in dem auch die jeweils wichtigen Bestimmungswerke aufgeführt werden. Dabei werden grundlegende Begriffe erläutert, die für die Verwendung der Bestimmungsliteratur nützlich sind. Der Schwerpunkt liegt jedoch in dem Bemühen, den Blick für das Erkennen der Algen im Gewässer zu schärfen. Die für eine Indikation wichtigen Algenbestände sind oft wenig auffällig und leicht zu übersehen. Erfahrung und Wissen sind erforderlich, um sie zu erkennen. Auch offensichtliche Massenentwicklungen bestehen in der Regel aus Mischbeständen mehrerer Arten, die differenziert betrachtet werden müssen. Der hier vorliegende Feldführer stellt das dafür notwendige "Handwerkszeug" zur Verfügung. Dafür werden zum einen die möglichen Habitate benthischer Algen charakterisiert und mit Bildern vorgestellt. Zum anderen wird die Vielfalt der Lager- bzw. Wuchsformen benthischer Algen mit ihren Charakteristika hinsichtlich Farbe, Konsistenz und mitunter auch Geruch beschrieben. Um diese Vielfalt für die praktische Arbeit zu gliedern, wurden neun Kategorien aufgestellt. Aufgrund der im Gelände sichtbaren Wuchs- bzw. Lagerformen wurden zahlreiche Taxa diesen Kategorien zugeordnet. Die Taxa werden mit zahlreichen Abbildungen dargestellt. Anschließend wird die Vorgehensweise bei der Probenahme entsprechend den Vorgaben des PHYLIB-Bewertungsverfahren erklärt und anhand von zwei Beispielen illustriert. Hinweise auf Verfahren in anderen Ländern und ein Ausblick auf die EN-Norm stellen das PHYLIB-Verfahren in einen größeren Zusammenhang. Damit bietet dieser Feldführer eine gute Orientierung hinsichtlich der Formen- und Farbenvielfalt, der systematischen Vielfalt sowie der entsprechenden Vielfalt der Bestimmungsliteratur der Vertreter des „übrigen“ Phytobenthos. Anwender des PHYLIB-Bewertungsverfahrens finden in diesem Buch hilfreiche Anleitungen für die praktische Arbeit.
We radio-tracked two male and one female Virginia northern flying squirrels (Glaucomys sabrinus fuscus) in the Allegheny Mountains of West Virginia at Snowshoe Mountain Resort, in winter 2003 and Canaan Valley National Wildlife Refuge in winter 2004, respectively, to document winter home range and habitat use in or near ski areas. Male home range size in the winter was larger than that reported for males during summer and fall, whereas the female home range we observed was smaller than those reported for summer and fall. However, winter habitat use was similar to summer and fall habitat use reported in other studies. Virginia northern flying squirrels foraged and denned in both red spruce (Picea rubens)-dominated forests and northern hardwood forests; however, selection of red spruce-dominated forests and open areas was greater than expected based on availability. Use of northern hardwood forest occurred less than expected based on availability. Male squirrels denned near, and routinely crossed, downhill ski slopes and unimproved roads during foraging bouts, whereas the female approached, but did not cross forest edges onto roads or trails.
Stetig wächst das Bewusstsein, dass Spinnen schützenswerte Lebewesen mit einer höchst interessanten Lebensweise sind und dass sie hohe Bedeutung im Naturhaushalt besitzen. Sie werden nicht mehr so häufig wie früher als lästiges, ekeliges und totzuschlagendes "Ungeziefer" empfunden. Nicht zuletzt die faszinierenden Darstellungen von KULLMANN & STERN (1981) haben das Interesse an dieser Tiergruppe geweckt. Darüber hinaus steht heute mit BELLMANN (1997) ein Werk mit hervorragenden Abbildungen und weiteren wichtigen Grundinformationen zur Verfügung, in dem sich auch Nichtfachleute rasch und gründlich über Spinnen informieren können. In Baden-Württemberg kommen nach dem aktuellen Kenntnisstand 738 Spinnenarten vor. Bedingt durch seine Lage im Südwesten Deutschlands ist hier der Anteil der Arten mit Hauptverbreitung im Süden Europas im Ländervergleich besonders groß. Arten mit Hauptverbreitung im Osten Europas finden sich ebenfalls. Im Schwarzwald und auf der Schwäbischen Alb sind Arten des Gebirges zu Hause. Viele Menschen halten Spinnen irrtümlich für Insekten. Spinnen bilden jedoch zusammen mit Skorpionen, Weberknechten, Milben und anderen Tierordnungen, wie zum Beispiel den Pseudoskorpionen, eine eigene Tierklasse.
Seit 1984 untersucht die Landesanstalt für Umweltschutz Baden-Württemberg auf den Wald-Dauerbeobachtungsflächen des Ökologischen Wirkungskatasters die Schwermetallbelastung von Regenwürmern. Das Ziel dieser Untersuchung ist es, mit Hilfe des Bioindikators "Regenwurm" Schadstoffwirkungen auf den Lebensraum Boden mit seiner Biozönose (Bodenlebewelt) zu ermitteln. Aufgrund ihrer Lebensweise und Akkumulationsfähigkeit für Schwermetalle, ihrer weiten Verbreitung und Häufigkeit in Böden, ihrer relativen Ortstreue und nicht zuletzt wegen ihrer Relevanz für ökosystemare Prozesse eignen sich Regenwürmer grundsätzlich gut als Bioindikatoren. Darüber hinaus ist eine ökotoxikologische Beurteilung der Wald-Dauerbeobachtungsflächen von besonderem Interesse, da bislang nur wenig Informationen über die toxikologische Einschätzung der Metallgehalte und die mögliche Übertragbarkeit (Indikationswert) auf andere Ökosystemkomponenten vorliegen.
Detailed mass balance food web models were constructed to compare ecosystem characteristics for three Alaska regions: the eastern Bering Sea (EBS), the Gulf of Alaska (GOA), and the Aleutian Islands (AI). This paper documents the methods and data used to construct the models and compares ecosystem structure and indicators across models. The common modeling framework, including biomass pool and fishery definitions, resulted in comparable food webs for the three ecosystems which showed that they all have the same apex predator—the Pacific halibut longline fishery. However, despite the similar methods used to construct the models, the data from each system included in the analysis clearly define differences in food web structure which may be important considerations for fishery management in Alaska ecosystems. The results showed that the EBS ecosystem has a much larger benthic influence in its food web than either the GOA or the AI. Conversely, the AI ecosystem has the strongest pelagic influence in its food web relative to the other two systems. The GOA ecosystem appears balanced between benthic and pelagic pathways, but is notable in having a smaller fisheries catch relative to the other two systems, and a high biomass of fish predators above trophic level (TL) 4, arrowtooth flounder and halibut. The patterns visible in aggregated food webs were confirmed in additional more detailed analyses of biomass and consumption in each ecosystem, using both the single species and whole ecosystem indicators developed here.
Several classes of chemicals that are known or suspected contaminants were found in bed sediment in Rock Creek, including polyaromatic hydrocarbons (PAHs), phthalate esters, organochlorine pesticides, dioxins and furans, trace metals and metalloids (mercury, arsenic, cadmium, chromium, cobalt, copper, lead, nickel, silver, and zinc), and polychlorinated biphenyls (total PCBs and selected aroclors). Concentrations of many of these chemicals consistently exceeded thresholdor chronic-effects guidelines for the protection of aquatic life and often exceeded probable effects levels (PELs). Exceedance of PELs was dependent on the amount of total organic carbon in the sediments. Concurrent with the collection of sediment-quality data, white sucker (Catostomus commersoni) were evaluated for gross-external and internal-organ anomalies, whole-body burdens of chemical contaminants, and gut contents to determine prey. The histopathology of internal tissues of white sucker was compared to contaminant levels in fish tissue and bed sediment. Gut contents were examined to determine preferential prey and thus potential pathways for the bioaccumulation of chemicals from bed sediments. Male and female fish were tested separately. Lesions and other necroses were observed in all fish collected during both years of sample collection, indicating that fish in Rock Creek have experienced some form of environmental stress. No direct cause and effect was determined for chemical exposure and compromised fish health, but a substantial weight of evidence indicates that white sucker, which are bottom-eeding fish and low-order consumers in Rock Creek, are experiencing some reduction in vitality, possibly due to immunosuppression. Abnormalities observed in gonads of both sexes of white sucker and observations of abnormal behavior during spawning indicated some interruption in reproductive success.
Seabirds in the UK were generally more productive in 2005 than in 2004, when productivity for many species reached an all-time low. A presumed scarcity of sandeels in 2004, especially in the North Sea, led to widespread starvation of chicks in the Northern Isles and in many places along the east coast of Britain (there is also recent evidence that prey fish were of unusually low energy content in 2004 around SE Scotland). The likely knock-on effect for 2005 was that there were few larger sandeels present (those that hatched in 2004) and it is thought that feeding on these fish allow adults to attain breeding condition in spring. This food scarcity and a cold spring led to what was among the latest breeding seasons on record. However, a late appearance of young sandeels allowed some chicks to fledge, and alternative prey species (such as sprat and small haddock) were taken also. However, it is thought that some chicks starved in this late season, as sandeels become unavailable in late summer, when they settle on the seabed. Unusually, 2005 was a very poor breeding season for many species in NW Scotland, which was spared the food shortages of 2004 and previous years; preferred prey during chick rearing were scarce in this region in 2005.
Nestling growth and development studies have been a topic of interest for a greater part of the last century (Sutton 1935, Walkinshaw 1948) and continue to be of interest today. This is not surprising since studies on nestling growth can provide a wealth of biological information that has larger implications for avian management and conservation. Despite this history of studying nestling development, basic information is still limited or absent for many species. Many questions remain unanswered, and contradictory conclusions are often found in the literature (Starck and Ricklefs 1998a). Therefore, much information on aging and development can still be gained from studying the development patterns of similar species and from comparative studies, across avian orders (Minea et al. 1982, Saunders and Hansen 1989, Carsson and Hörnfeldt 1993). Additionally, nestling growth studies can yield insight into the effects of different nesting strategies on productivity (O’Connor 1978), as well as the impacts of parental effort and environmental variables on fitness (Ross 1980, Ricklefs and Peters 1981, Magrath 1991). Since low reproductive success may play a significant role in the declines of many North American passerines (Sherry and Holmes 1992, Ballard et al. 2003), a better understanding of the factors that influence reproductive success is a vital component of avian conservation (Martin 1992). Data on nestling aging can be used to improve nest survival estimates (Dinsmore 2002, Nur et al. 2004), providing information that can be used to more precisely age nests (Pinkowski 1975, Podlesack and Blem 2002), (Jones and Geupel 2007). Indeed, the relatively short time period young spend developing in the nest is a critical part of a bird’s life cycle and a nestling’s developmental path can affect its survival to independence, its survival as an adult, and its future reproductive success.
Background: Molecular phylogenies are being published increasingly and many biologists rely on the most recent topologies. However, different phylogenetic trees often contain conflicting results and contradict significant background data. Not knowing how reliable traditional knowledge is, a crucial question concerns the quality of newly produced molecular data. The information content of DNA alignments is rarely discussed, as quality statements are mostly restricted to the statistical support of clades. Here we present a case study of a recently published mollusk phylogeny that contains surprising groupings, based on five genes and 108 species, and we apply new or rarely used tools for the analysis of the information content of alignments and for the filtering of noise (masking of random-like alignment regions, split decomposition, phylogenetic networks, quartet mapping). Results: The data are very fragmentary and contain contaminations. We show that that signal-like patterns in the data set are conflicting and partly not distinct and that the reported strong support for a "rather surprising result" (monoplacophorans and chitons form a monophylum Serialia) does not exist at the level of primary homologies. Split-decomposition, quartet mapping and neighbornet analyses reveal conflicting nucleotide patterns and lack of distinct phylogenetic signal for the deeper phylogeny of mollusks. Conclusion: Even though currently a majority of molecular phylogenies are being justified with reference to the 'statistical' support of clades in tree topologies, this confidence seems to be unfounded. Contradictions between phylogenies based on different analyses are already a strong indication of unnoticed pitfalls. The use of tree-independent tools for exploratory analyses of data quality are highly recommended. Concerning the new mollusk phylogeny more convincing evidence is needed.

Mitochondrien sind die Kraftwerke unserer Zellen. In ihnen findet die Zellatmung statt, die unseren Körper mit lebenswichtiger Energie versorgt. Zusätzlich teilen sich die Zellorganellen und verschmelzen wieder miteinander im Minutentakt. Was aber passiert, wenn Teile dieses dynamischen Geflechts Defekte aufweisen? Die Antwort dazu könnte ein Protein sein, das auf zwei verschiedene Weisen in die Mitochondrien-Membranen eingebaut wird. Liegt keine kurze Form des Proteins vor, ist das ein Hinweis dafür, dass die Organellen defekt sind. Die Mitochondrien verbrennen die mit der Nahrung zugeführten Kohlenhydrate und Fette unter Verbrauch von Sauerstoff zu Kohlendioxid und Wasser. Bei diesem Vorgang, der Zellatmung, wird über eine Reihe von Proteinkomplexen ein elektrochemisches Potenzial aufgebaut, das zur Produktion des Energieträgers ATP (Adenosintriphosphat) genutzt wird. ATP kann aus den Mitochondrien abtransportiert werden und steht somit als eine Art Treibstoff für alle Stoffwechselprozesse zur Verfügung. Die Arbeit der Mitochondrien ist der Hauptgrund für unseren täglichen Sauerstoffbedarf. Außerdem tragen die Nano-Kraftwerke der Zelle dazu bei, unsere Körpertemperatur auf 37 °C aufrechtzuerhalten. Aufgrund dieser zentralen Funktionen ist es nicht verwunderlich, dass eine Reihe von Krankheiten beim Menschen durch den Funktionsverlust von Mitochondrien verursacht oder beeinflusst wird. Das sind in erster Linie neurologische oder muskuläre Erkrankungen, aber auch Diabetes, Fettleibigkeit, verschiedene Formen von Krebs und Alterungsprozesse. Folglich ist es von immenser Bedeutung zu verstehen, wie Mitochondrien funktionieren, wie sie ihre Funktionalität aufrechterhalten und gegebenenfalls repariert oder entsorgt werden können. Dem können wir am Wissenschaftsstandort Frankfurt hervorragend nachgehen, da sich einige international ausgewiesene Forschungsgruppen in den Fachbereichen Medizin, Biologie, Chemie und am Max-Planck-Institut für Biophysik mit verschiedenen Aspekten der mitochondrialen Biologie befassen. In zahlreichen interdisziplinären Kooperationen wird so versucht, dieses komplexe System besser zu verstehen.
Jeder Mensch kämpft täglich erfolgreich mit Krankheitserregern, ohne dass er sich der komplexen molekularen Vorgänge dabei bewusst wäre. Wie in einem Hollywood-Streifen geht es rasant zur Sache. Ist das Immunsystem angeschlagen oder trifft es auf starke Gegner, kann eine Infektion binnen weniger Tage außer Kontrolle geraten und lebensbedrohliche Reaktionen hervorrufen. Der menschliche Organismus benötigt eine effiziente Verteidigungsstrategie gegen die Eindringlinge und muss, ebenso wie der britische Geheimdienst im Bond-Film, in die Ausbildung geübter Agenten investieren, Agenten mit Doppel-Null-Status. Agenten wie James Bond.
Background, aim, and scope Food consumption is an important route of human exposure to endocrine-disrupting chemicals. So far, this has been demonstrated by exposure modeling or analytical identification of single substances in foodstuff (e.g., phthalates) and human body fluids (e.g., urine and blood). Since the research in this field is focused on few chemicals (and thus missing mixture effects), the overall contamination of edibles with xenohormones is largely unknown. The aim of this study was to assess the integrated estrogenic burden of bottled mineral water as model foodstuff and to characterize the potential sources of the estrogenic contamination. Materials, methods, and results In the present study, we analyzed commercially available mineral water in an in vitro system with the human estrogen receptor alpha and detected estrogenic contamination in 60% of all samples with a maximum activity equivalent to 75.2 ng/l of the natural sex hormone 17beta-estradiol. Furthermore, breeding of the molluskan model Potamopyrgus antipodarum in water bottles made of glass and plastic [polyethylene terephthalate (PET)] resulted in an increased reproductive output of snails cultured in PET bottles. This provides first evidence that substances leaching from plastic food packaging materials act as functional estrogens in vivo. Discussion and conclusions Our results demonstrate a widespread contamination of mineral water with xenoestrogens that partly originates from compounds leaching from the plastic packaging material. These substances possess potent estrogenic activity in vivo in a molluskan sentinel. Overall, the results indicate that a broader range of foodstuff may be contaminated with endocrine disruptors when packed in plastics. Ein funktionierendes Gedächtnis beruht darauf, dass die Kontakte zwischen den Milliarden Nervenzellen in unserem Gehirn sich ständig verändern und anpassen. Häufig verwendete Signalwege werden verstärkt und ausgebaut, wie eine Landstraße zu einer Schnellstraße. Weniger häufig benutze Signalwege können dagegen abgebaut werden. Die Signalübertragung verlangsamt sich wie der Verkehr auf einer lange nicht mehr instand gehaltenen Straße. Will man diese Prozesse auf molekularer Ebene verstehen, muss man die Synapsen näher betrachten. Das sind spezialisierte Kontaktstellen, die es den Nervenzellen ermöglichen, hochkomplexe Netzwerke, sogenannte Schaltkreise, zu knüpfen. Die Flexibilität dieser Schaltkreise ermöglicht es uns, Informationen zu verarbeiten und entsprechend zu reagieren. Inzwischen kennt man eine Fülle von Boten-Molekülen, Rezeptoren und Liganden, die diese Prozesse auf molekularer Ebene steuern.

Antibiotika-Resistenz: Die Tricks der Bakterien : Pumpsysteme werfen die Arzneistoffe aus der Zelle

Immer häufiger sind Bakterien resistent gegen ein bestimmtes Antibiotikum, oft auch gleich gegen mehrere. Eine Infektion, die von solchen multiresistenten Bakterien verursacht wird, kann nicht mehr mit Antibiotika bekämpft werden. Im schlimmsten Fall führt sie bei immungeschwächten Patienten zum Tod. Um zielgerichtet neue und wirkungsvolle Medikamente entwickeln zu können, ist es wichtig zu wissen, wie die Bakterienzelle sich gegen die Zerstörung durch Antibiotika wehrt. Ein inzwischen genau entschlüsselter Mechanismus ist die Efflux-Pumpe, die für die Zelle schädliche Substanzen wieder hinausbefördert.
