The Japanese micropterigid moths are revised. Seventeen species in five genera are recognized from Japan, described or redescribed with the male and female genital figures. Of these, two genera, Issikiomartyria HASHIMOTO and Kurolkopteryx HASHIMOTO, and seven species, Issikiomartyria akemiae HASHIMOTO, Issikiomartylia plicata HASHIMOTO, Issihiomartyria distincta HASHIMOTO, Issihiomartyria bisegmentata HASHIMOTO, Kurokopteryx dolichocerata HASHIMOTO, Neomicropteryx hiwana HASHIMOTO, and Neomicropteryx redacta HASHIMOTO, are new to science. A new combination is given: Issikiomartyria nudata (Issuu). Biology and immature structures of the Japanese species are also described together with the keys to genera and to species provided on the basis of the adult characters. Phylogenetic relationships among the Northern Hemisphere genera are analyzed by the cladistic analysis using PAUP* (SWOFFORD, 2002) based on the morphological characters of adults. A monophyly of the Northern Hemisphere genera except for Micropterix is supported by nine apomorphies, but their immediate sister taxon remains unresolved.
"Neobiota" ist ein biologischer Begriff zur Bezeichnung von nicht-einheimischen Arten, welche infolge direkter oder indirekter menschlicher Einwirkungen in neue geografische Gebiete eindringen. Biologische Invasionen durch neue gebietsfremde Lebewesen (Neobiota), die sowohl Tierarten (Neozoa) als auch Pftanzenarten (Neophyta) betreffen, wurden in den letzten Jahren zunehmend von Interesse, da ihre Anzahl und Ausbreitung ständig zunimmt. Solche Fremdlinge (Aliens) verändern oft ihre neue Umwelt, die einheimischen bodenständigen Planzen- und Tierarten müssen sich an diese ungewohnte Umgebung anpassen. Insbesondere invasive fremde Arten werden als eine der Hauptursachen für Verluste an Biodiversität erachtet. In der vorliegenden Arbeit werden für Südtirol einige markante Beispiele rezent einschleppter Tier- und Pflanzenarten erörtert. Es wird der bestehende Trend einer rezenten Zunahme von Neobiota aufgezeigt, als Folge einer Zunahme von Verkehr und Warenaustausch, und auf die Notwenigkeit hingewiesen, die Einschleppungswege und Zeiträume genau zu verfolgen und zu registrieren. Insgesamt werden hier 44 Tierarten behandelt: davon 40 Invertebraten und 4 Vertebrata, sowie 5 Pflanzenarten. Davon sind einige Arten auch Neumeldungen für Südtirol: Diptera: Rhagoletis alternata, Rhagoletis completa; Heteroptera: Leptoglossus occidentalis; Pisces: Rhodeus amarus, Pseudorasbora parva; Crustacea: Procambarus klarkii; Araneae: Atea triguttata und Araniella displicata (beides heimische Arten), Tegenaria atrica (adventiv).
Die vorliegende Arbeit beschäftigt sich mit der Charakterisierung des Proteoglykans Biglycan und seiner Funktion als Signalmolekül in inflammatorischen und autoimmunen Prozessen. Die biologische Bedeutung der in vitro gewonnenen Ergebnisse in primären Makrophagen und dendritischen Zellen wurde durch in vivo Modelle der Pathogenvermittelten-und nicht-Pathogen-vermittelten Inflammation und der Autoimmun-Erkrankung Lupus Nephritis bestätigt. In primären Makrophagen und dendritischen Zellen induziert Biglycan die Produktion proinflammatorischer Zytokine und Chemokine durch Interaktion mit Toll-like Rezeptor (TLR) 2 und 4. Mit nucleotide-binding oligomerization like Rezeptorprotein3 (NLRP3)-,apoptosisassociated speck-like protein containing a CARD (ASC)- , Caspase-1- und TLR2/4- defizienten Mäusen und verschiedenen pharmakologischen Inhibitoren war es möglich in primären murinen peritonealen und Knochenmark-Makrophagen nachzuweisen, dass Biglycan die Caspase-1 NLRP3/ASC-abhängig aktivierte und damit die Prozessierung der Proform und Sekretion von reifem IL-1β induzierte. Durch Bindung an TLR2/TLR4 aktivierte Biglycan die NFκB, Erk und p38 mitogen-activated protein kinase (MAPK) Signalwege und stimulierte die Expression von Interleukin-1 beta (IL-1β). Biglycan aktivierte zudem den P2X7 Rezeptor (P2X7R) in Makrophagen und ist somit in der Lage auch ohne zusätzliche Ko-Stimulation, beispielsweise durch ATP, das NLRP3 Inflammasom zu stimulieren und die Prozessierung von aktivem IL-1β anzuregen. In einem Pathogenvermittelten (Lipopolysaccharid (LPS)-induzierte Sepsis) wie auch –nicht-athogenvermittelten (unilaterale Uretherobstruktion, UUO) Mausmodell der Inflammation wurde die biologische Relevanz dieser Prozesses gezeigt. Die Defizienz von Biglycan ging in diesen Modellen mit verminderter Aktivierung des NLRP3/Caspase-1 Inflammasomes, geringeren Spiegeln von reifem IL-1β und geringerer Organschädigung einher. Nachdem aufgezeigt werden konnte, dass die Biglycan-Konzentrationen in Nierenbiopsien und im Plasma von Patienten mit Lupus Nephritis stark erhöht waren, wurde seine Relevanz in Immunitätsreaktionen einschließlich autoinflammatorischen Prozessen genauer untersucht. Die Effekte von Biglycan in verschiedenen Stadien der Erkrankung wurden mit der MRL-Faslpr (kurz MRL/lpr) Maus, einem etablierten Modell der Lupus Nephritis (LN) und einem dafür generierten Modell der Defizienz (Bgn-/- MRL/lpr) und Überexpression von Biglycan (hBGN MRL/lpr) analysiert. In den verschiedenen Stadien der LN nahm die Konzentration von zirkulierendem und renalem Biglycan in MRL/lpr Mäusen zu und korrelierte gleichermaßen mit dem Fortschreiten der Erkrankung. Die Defizienz von Biglycan verminderte hingegen stark die renale Infiltration von Entzündungszellen, insbesondere B1-Zellen, außerdem die Zytokin-, Chemokin- und Immunglobulin-Konzentrationen und minderte die Progredienz der Niereninsuffizienz verglichen mit Lupus Mäusen gleichen Alters. In der Initialphase der Lupus Nephritis induzierte Biglycan in Mäusen, transient transgen für humanes Biglycan (hBGN MRL/lpr), vermehrte renale Zellinfiltration und Albuminurie als Zeichen nephrotischer Dysfunktion. Zudem konnte gezeigt werden, dass die Produktion des stark proinflammatorischen Zytokines IL-1β in jungen Lupus Nephritis Mäusen NLRP3/Caspase-1-abhängig ist und durch Biglycan verstärkt wurde. Die Mechanismen, durch die endogenes Biglycan die Leukozyteninfiltration in Lupus Mäusen induzierte und somit inflammatorische und autoimmune Vorgänge potenzierte, wurden insbesondere an B-Zellen untersucht. Es konnte gezeigt werden, dass Biglycan die renale Migration von einem besonderen B-Zell Subtyp, B1-Zellen, verantwortlich für die T-Zellenunabhängige Autoimmunglobulinproduktion beim LN, unterhielt. Dabei vermittelte Biglycan die Rekrutierung von B1-Zellen in die Niere durch Regulation der Expression und Synthese der B-Zell C-X-C Chemokin Ligand 13 (CXCL13) in der Niere und in residenten peritonealen Makrophagen. In vitro konnte zudem der Mechanismus aufgeklärt werden, über den Biglycan CXCL13 reguliert. In primären Makrophagen und dendritischen Zellen induziert Biglycan die Expression und Sekretion von CXCL13 über TLR2 und TLR4. Die Daten zeigen auf, dass Biglycan als endogenes Gefahrensignal starke proinflammatorische Reaktionen hervorruft. Über Rezeptoren des angeborenen Immunsystems, TLR2 und TLR4, aktiviert Biglycan des weiteren Zellen des adaptiven Immunsystems und inuziert die Rekrutierung weiterer Lymphozyten. Demnach kann postuliert werden, dass Biglycan als Brückenmolekül das anegborene und adaptive Immunsystem verbindet, und somit ein potenzielles neues „drug target“ in autoinflammatorischen, wie auch autoimmunen Vorgängen darstellt.
Die Gattung Phormictopuswurde im Jahre 1901 von POCOCK aufgestellt. In seine neue Gattung nahm er als Typusart Mygale cancerides LATREILLE, 1806 von der Insel Hispaniola auf, dazu kam Lasiodora cautus AUSSERER, 1875, eine Art, die ohne Angabe des locus typicus beschrieben worden war. Bisher waren 14 Arten und 2 Unterarten bekannt, von denen 5 aus Südamerika stammen. Die vorliegende Arbeit reduziert die Artenzahl auf 12, wobei 5 neue Arten beschrieben und 4 synonymisiert, 3 zu nomina dubia (Typus verschollen), und 3 "incertae sedis" (in andere Gattungen gehörig) erklärt werden.
La famille des Pyuridae n'est représentée en Nouvelle-Calédonie que par onze espèces, la plupart vivant dans les zones portuaires. Une seule espèce est nouvelle, Bolteniopsis pacificus, qui vit dans des sables grossiers et représente la première espèce de ce genre trouvée dans le littoral tropical. Les Molgulidae ne sont représentées que par une espèce nouvelle.
Dix-neuf espèces de Styelidae, pour la plupart coloniales, sont décrites ou signalées du lagon. Huit sont nouvelles. Sur les onze espèces connues, neuf sont présentes en Australie. Quatre espèces ont une repartitlon tres vaste couvrant dans certains cas toutes les mers chaudes. Les espèces nouvelles appartiennent toutes à des genres dont la diversité maximale est en zone tropicale.
Quinze espèces dont deux nouvelles pour la Science ont été récoltées en Nouvelle-Calédonie. La plupart d'entre elles sont aussi connues de l'Australie et de l'Indonésie. Une seule espèce est considérée comme faisant partie d'une faune tempérée australe. Toutes les autres sont d'affinités tropicales.
Quinze espèces d'ascidies Phlébobranches ont été récoltées dans le lagon de Nouvelle-Calédonie. Elles appartiennent à quatre familles: Corellidae, Ascidiidae, Perophoridae et Cionidae. Cette dernière n'avait pas encore été signalée de cette région. Quatre espèces sont nduvelles pour la science: Diazona textura (Cionidae), Ascidia dorsalis (Ascidiidae), Ecteinascidia aequale et E. koumaci (Perophoridae). Seules cinq espèces (une Corellidae et quatre Ascidiidae) étaient déjà connues de Nouvelle-Calédonie.
Revision of the millipede family Andrognathidae in the Nearctic region (Diplopoda, Platydesmida)

Homology of virtually all major components of facial anatomy is assessed in Archosauria in order to address the function of the antorbital cavity, an enigmatic structure that is diagnostic for the group. Proposed functions center on its being a housing for a gland, a muscle, or a paranasal air sinus. Homology is approached in the context of the Extant Phylogenetic Bracket method of reconstructing unpreserved aspects of extinct organisms. Facial anatomy and its ontogeny was studied in extant archosaurs (birds and crocodilians) to determine the osteological correlates of each soft-tissue component; resemblances between birds and crocodilians comprised the similarity test of homology. The congruence test of homology involved surveying phyiogenetically relevant fossil archosaurs for these bony signatures. The facial anatomy of extant birds and crocodilians is examined in detail to provide background and to discover those apomorphic aspects that contribute to the divergent specialization of these two groups and thus obscure homologies. Birds apomorphically show enlarged eyeballs, expanded nasal vestibules, and reduced maxillae, whereas crocodilian faces are dorsoventrally flattened (due to nasal rotation) and elongated. Most facial attributes of archosaurs are demonstrably homologous and in fact characterize much more inclusive groups. Special emphasis has been placed on the nasal conchae and paranasal air sinuses. Within Amniota, the following conchal structures are homologous, and all others are neomorphs: avian caudal concha, crocodilian concha + preconcha, Sphenodon caudal concha, squamate concha, and probably the mammalian crista semicircularis. The avian antorhital paranasal air sinus is homologous with the crocodilian caviconchal sinus; the maxillary sinus of placental mammals is not homologous with the archosaurian paranasal sinus. With regard to the function of the antorbital cavity, archosaurs possess homologous nasal glands, dorsal pterygoideus muscles, and paranasal air sinuses, but the osteological correlates of only the paranasal sinus involve the antorbital fenestrae and fossae. Thus, the antorbital cavity is best interpreted as principally a pneumatic structure.
An annotated checklist and bibliography of 197 species (representing 78 genera and 26 families) of non-marine polychaetes of the world is presented, including synonymies, information on ecology, distribution, habitat, and references to the taxonomic and biological literature. Over half (57%) of the checklist species are represented by just three families as follows: Nereididae (61 species including Namanereis, Namalycastis, Neanthes and Hediste), Aeolosomatidae (27 species, mostly Aeolosoma) and Sabellidae (24 species including Caobangia and Manayunkia). Other well-represented taxa are the epizoic histriobdel1id Stratiodrilus (11 species), the inland-sea-specialist ampharetid Hypania and related genera (5 species), and the freshwater-tolerant spionid Marenzelleria (5 species). One new combination is proposed for the nereidid Nereis tenuipalpa Pflugfelder, 1933, viz. Paraleonnates tenuipalpa n. comb.
Birds are characterized by pneumatization of their skeletons by epithelial diverticula from larger, air-filled cavities. The diverticula-or 'air sacs'-that invade the postcranium result from outgrowths of the lungs; poslcranial pneumaticity has been very well studied. Much more poorly understood are the air sacs that pneumatize the skull. Study or craniofacial pneumaticity in modern birds (Neornithes) indicates the presence of two separate systems: nasal pneumaticity and tympanic pneumaticity, The lacrimal and maxillary bones arc pneumatized by diverticula of the main paranasal cavity, the antorbital sinus. There are five tympanic diverticula in neornithines that pneumatize the quadrate, articulare and the bones of the braincase. The pneumatic features of the following six genera of Mesozoic birds are examined: Archaeopteryx, Ellaliornis, Baplomis, Parahesperornis, Hesperornis and lchthyornis. Despite the 'archaic' aspect of most of these birds, many of the pneumatic features of neornithines are found in .Mesozoic birds and are considered primitive for Aves. The phylogenetic levels at which most of the avian pneumatic features arose within Archosauria are uncertain. Until the phylogenetic levels at which homologous pneumatic features arose are determined, it is unwise to use most pneumatic characters in the discussion of avian origins. Within avian phylogeny, Ornithurae and Neornithes are well-supported by pneumatic synapomorphies. There is a trend towards reduction of craniofacial pneumaticity within Hesperornithiformes. Witthin Neornithes, four derived pneumatic characters suggest that the Palaeognathae (ratites and tinamous) is monophyletic.
Aus den vorhergehenden Untersuchungen erhellt, dass die fossilen Hölzer von König-Karls-Land ein aussergewöhnliches Interesse beanspruchen. Sie umfassen sechs Arten, die zu fünf verschiedenen Gattungen gehören, und zwar: Phyllocladoxylon sp., Xenoxylon phyllocladoides GOTHAN, Cupressinoxylon cf. McGeei KNOWLTON, Cedroxylon cedroides n. sp., Cedroxylon transiens n. sp., Protopiceoxylon exstinctum n. gen. et sp. und dazu noch ein paar nicht bestimmbare Arten.
After giving a brief historical account of the use and precise definitions of the various measurements and their indices in termites, the need for bringing together all such known measurements and indices in the fonn a monograph is explained. Precisely defined measurements obviate the necessity of using vague expressions for the comparison of allied taxa. Of the 88 measurements and 53 indices thus listed, 66 and 34 respectively have been used already in the published literature, and 22 and 19 new ones are added here.
Carnian (Upper Triassic) fishes from Polzberg bei Lunz have been known since 1886 but no comprehensive account has been published. Eleven species are described nine of which, Saurichthys calcaratus, Polzbergia brochatus, Peltoplellrus dinlmptus, Habroichthys gregarius, Nannolepis elegans, Phaidrosoma lunzensis, Elpistoichthys pectinatus, E. striolatus and Pholidophoretes salvus are new, and two others, Thoracopterus niederristi Bronn and Gigantopterus telleri Abel, previously little-known. New supraspecific taxa defined are: the order Polzbergiiformes, the family Thoracopteridae and the genera Polzbergia, Nannolepis, Phaidrosoma, Elpistoichthys and Pholidophoretes. Habroichthys. Thoracopterus, Gigantopterus and Nannolepis show an unusual skull-roof pattern and are included in the re-defined order Luganoiiformes. Two new ichthyokentemids considerably extend the known time-range of this family. The genus Pholidophoretes is intermediate between the Archaeomenidae Goodrich 1909, and the Pholidophoridae sensu stricto Nybelin 1966. The Polzberg assemblage was probably mainly marine with a small freshwater contribution; it shows less similarity to the Besano and Raibl assemblages than these do to each other. The Luganoiiformes are probably, but not certainly, monophyletic; relationships within the order are analyzed and a cladogram constructed. The Platysiagiformes, Peltopleuriformes, Luganoiiformes and Cephaloxeniformes could all have been derived from a common ancestor at the Perleidus level and are probably offshoots of the perleidid radiation.
The Opisthobranchia comprise highly specialized marine gastropods and have therefore been subject to diverse investigations covering various biological disciplines. However, a robust phylogeny of these gastropods is still lacking and several subclades have only been rarely studied. Furthermore, crucial aspects for the evolution of Opisthobranchia have not been comparatively analysed. Therefore, the aim of the present thesis is to gain new insights into the phylogeny of the Opisthobranchia with special focus on certain critical groups (Pleurobranchomorpha, Acteonoidea) and to assess several crucial features of the evolution of the investigated clades. The combination of four different gene markers (18S rDNA, 28S rDNA, 16S rDNA and CO1) and modern molecular systematic analysis tools were used to construct phylogenetic hypotheses focussing on Opisthobranchia as a whole as well as Pleurobranchomorpha and Acteonoidea in more detail. Intriguing new aspects of phylogeny and evolution of Opisthobranchia were revealed. First of all, monophyly of Opisthobranchia is definitely rejected based on the present data, while monophyly of Euthyneura (comprising Opisthobranchia and Pulmonata) is supported. Monophyly of opisthobranch subclades is confirmed for Nudipleura (as well as its constituting groups Nudibranchia and Pleurobranchomorpha), Umbraculida, Pteropoda (as well as subclades Thecosomata and Gymnosomata) and Acochlidiacea, for Cephalaspidea (if Runcinacea is regarded as a separate clade) and for Sacoglossa (if Cylindrobulla is accepted as an Oxynoacea). Aplysiomorpha are rendered paraphyletic due to the position of Akera bullata, but this result needs further investigation and should be considered with caution. The Nudipleura are found as the first single offshoot of the Euthyneura implying an early evolutionary separation of the last common ancestor of this clade. The remaining taxa form two main clades, one comprising the opisthobranch subgroups Umbraculida, Cephalaspidea, Aplysiomorpha and Pteropoda, while the other contains the pulmonate taxa and the opisthobranch Sacoglossa and Acochlidiacea. The interrelationships within these clades remain largely unresolved due to low statistical support values. However, a possible sister group relationship of Acochlidiacea and Eupulmonata receives statistical support. Opisthobranchia display various highly specific adaptations to diverse food sources. However, evolution of these specialized traits has never been assessed at an analytical level. The current thesis reconstructs the evolution of dietary preferences with novel methodologies based on the newly proposed phylogenetic hypothesis. Reconstruction of dietary evolution revealed herbivory as the ancestral condition in Euthyneura implying that carnivory evolved at least five times independently in the diverse lineages. The first comprehensive molecular phylogenetic hypothesis of the Pleurobranchomorpha could not reveal monophyly of the two main subclades Pleurobranchaeidae and Pleurobranchidae. This is due to the position of a single taxon (Euselenops luniceps) which is assigned to the Pleurobranchaeidae based on morphology but clusters within Pleurobranchidae in the current hypothesis. Furthermore, the tribe Berthellini and the genus Berthella are rendered paraphyletic by the current analyses. The results of molecular systematic analyses were used to reconstruct historical biogeography of Pleurobranchomorpha. Four different methodological approaches were applied yielding ambiguous results for Pleurobranchomorpha. However, the Pleurobranchidae comprising about 80% of the extant Pleurobranchomorpha most probably derived from an Antarctic origin. Dating of the phylogenetic tree via molecular clock methods yielded divergence of Pleurobranchidae into the Antarctic Tomthompsonia antarctica and the remaining species in Early Oligocene. Afterwards the latter underwent rapid radiation during Oligocene and Early Miocene. This divergence event coincides with two major geological events in the Antarctic region. On the one hand, the onset of glaciation and on the other hand the opening of the Drake Passage with concurrent formation of an Antarctic circumpolar current (ACC). I suppose that these sudden and dramatic changes in climate and palaeogeography probably accounted for migration of the last common ancestor of Pleurobranchidae (besides Tomthompsonia) into warmer regions via the Drake Passage to the Western Atlantic and Eastern Pacific and via the South Tasman Rise to the Indo-West Pacific. Furthermore, the ACC may have triggered larval dispersal to the Eastern Atlantic. The phylogenetic position of Acteonoidea has been a matter of debate for decades and they have long been considered as basal opisthobranchs. Results of the present thesis rather support placement in “Lower Heterobranchia” as sister group of Rissoelloidea. The current division of Acteonoidea into three families has never been investigated by means of phylogenetic methods. Thus, this thesis provides the first comprehensive investigation of this clade challenging present division into three families. The results rather support division into two main clades with the monogeneric Bullinidae clustering within Aplustridae doubting its separate status. Additionally, Rictaxis punctocaelatus which has been assigned to Acteonidae clusters basal to Aplustridae rendering Acteonidae paraphyletic. Since information on morphology of R. punctocaelatus was lacking until now, I conducted the first detailed investigation on morphology and histology of this species in order to reassess the unexpected molecular systematic placement. Character tracing analyses revealed similarities with both acteonoidean families implying an intermediate position of this species which might be assigned to a separate family in the future. Furthermore, the common features of Acteonidae and Rictaxis (massive shell, small foot, anterior mantle cavity opening, and absence of oral gland) are possibly plesiomorphic for the whole Acteonoidea. In summary, the results of the present thesis provide valuable novel insights into the phylogeny and evolution of the Opisthobranchia by employing state-of-the-art approaches of molecular systematics and evolutionary reconstruction. Thus, diverse hypotheses on opisthobranch phylogeny and evolution were either supported or rejected as well as novel hypotheses proposed which offer the basis for further research on these extraordinary gastropods.
Recently, the first part of the morphological revision of the Southeast Asian water monitor lizards of the Varanus salvator (Laurenti, 1768) species group provided a taxonomic overview over the members of this successful and widespread species complex (Koch et al. 2007). There, the Philippine taxa marmoratus, nuchalis and cumingi were reelevated to species status due to diagnostic morphological characteristics, e.g. significantly enlarged scales on the neck region. In this second part of the ongoing revision, these three species are re-investigated using additional voucher specimens and advanced statistical techniques including canonical variates analysis and principal component analysis. Our new investigations indicate that V. marmoratus represents a composite species, comprising at least three distinct taxa. Hence, the populations of the Sulu Archipelago (Tawi-Tawi Island) and those of the Palawan region are described as new species, viz. Varanus rasmusseni sp. nov. and V. palawanensis sp. nov., respectively. The allopatric island populations of V. cumingi inhabiting Samar, Leyte, and Bohol (the East Visayan subregion) show characteristic and geographically correlated colour patterns distinct from the type locality Mindanao (the second subregion of Greater Mindanao), warranting subspecific partition of this species. The new subspecies is named V. cumingi samarensis ssp. nov. In contrast, the taxonomic status of V. nuchalis remained unchanged, although this species shows some considerable variation in colour pattern. The systematic chapters are supplemented with notes about biology and conservation status. The hitherto underestimated diversity and zoogeography of Philippine water monitors is discussed in the light of Pleistocene sea level fluctuations. Finally, we introduce a scenario for the evolution and spread of Southeast Asian water monitor lizards and provide an identification key for the Philippine members of the V. salvatoI' complex.

Phytosociological syntaxa of North and Central Anatolia were reviewed almost at all categories and discussed from some methodological viewpoints. Totally 178 diumallepidopteran associations, inhabited on various vegetational units from planar to alpin zone were studied for the first time. The possibility of the application of some phytosociologica1 methods for the Lepidoptera associations were investigated. The selection of the character-species of the higher lepidopteran syntaxa, and the aspect concept, as well as importance of the distribution pattern in biogeography for the character-species to be selected were discussed. Faunistically, 334 diurnal species in 25 lepidopteran families were recorded. Among them, four species and one genus are new to the fauna of Turkey. Each species recorded were also investigated autoecologically. Their habitats were described using the phytosociological syntaxa, as well as the plant formations. Habitat preferences of the species were evaluated. Vertical and horizontal distributions, abundances, phenology, and food-plants of the adult stages were listed. Species recorded in various plant formations and vegetational units were given in separate lists.
1. Fomes applanatus, a perennial polypore of wide distribution and of common occurrence in North America and in Europe is responsible for the decay of very large quantities of wood annually. It attacks practically all deciduous species and several coniferous species, both dead wood and living trees. This fungus, heretofore commonly regarded as a pure saprophyte, has now for the first time been comprehensively studied; the investigations recorded in this paper have followed along three main lines of inquiry; (1) a study of the morphology and the ecology of F. applanatus and of the action on its host; (2) a determination of its etiological relationships by culture methods, and a testing of the applicability of such methods to a study of the timber destroying fungi; (3) an investigation of the possibility of finding criteria by which we may distinguish parasitic action on wood from saprophytic. 2. Fomes applanatus produces basidiospores only; conidia are not produced by the mycelium, nor, as is commonly affirmed, on the upper surface of the sporophore. The basidiospores are not of the ordinary type; each consists of a yellow, papillate, thick-walled chlamydospore within a thin hyaline wall. The so-called" truncate" base is in reality the apical end of the spore. Spore discharge is enormous and continues for by far the longest period recorded for fungi. It is continuous day and night for about six months-visible from vigorous fruiting bodies as spore clouds. Discharge is not affected by variations in light, humidity of the air, or temperature within very wide limits; frost causes an instant cessation and thereafter there is no further spore fall until a new set of pores is organized. The spores were not found to retain their viability for more than 6 1/2 months. They germinate in water and various other media within 48 hours after sowing, but the percentage of viable spores is very low and their behaviour with respect to germination is erratic. 3. No difficulty was experienced in culturing F. applanatus on artificial media or on wood. Three parallel series of cultures on wood were carried through to sporulation with mycelium obtained from (a) spores, (b) pieces of sporophore, (c) pieces of diseased wood. 4. Wood rotted by F. applanatus exhibits a mottled appearance sufficiently characteristic to permit of its recognition. when compared with other rots. In the case of living wood the area of attack is delimited by a wide dark band. Histologically, the decay is characterized by a perforation and corrosion of the elements culminating in disappearance of the tissues in localized pockets into which the mycelium gathers forming strands. Chemically, the change is one of delignification followed by a progressive solution of the resultant cellulose wall except for a few resistant fragments. In the later uniform white stage of decay, the threads of F. applanatus were always found to be accompanied by other destructive agentsbacteria or fungi. 5. The particular type of decay in timber due to Fomes applanatus has now been linked with its causal agent by actual culture. 6. Fomes applanatus has been proved to be a wound parasite, and in southern Ontario at least is one of the commonest and most destructive of this type. The proof rests on three grounds: (a) the conventional test applied to other such fungi-the mycelium works upward most readily by the way of the heartwood causing a characteristic decay and outward into the sapwood, eventual1y reaching the cambium, and is apparently the cause of the death of the tissues traversed by it; (b) a broad brown band is present in the wood of living trees along the advance line of the invading mycelium of this fungus. Within this band there is a copious production of brown wound gum and an excessive multiplication of tyloses; This band steadily moves fonvard with the advancing hyphae, the tyloses and wound gum being destroyed by the mycelium along its posterior margin as rapidly as they are formed along its antedor edge. The tyloses (and possibly the wound gum alsoJ certify to the living condition of the invaded tissues; their production can be ascribed only to the influence of the fungus, and the invasion of these tissues and their fate demonstrates directly its ability to act as a parasite; (c) inoculations with the spores and mycelium of F. applanatus into living trees resulted in an extensive browning of the inoculated wood with a multiplication of tyloses-both far in excess of similar phenomena due to traumatic stimulation. 7. The occurrence of wound gum and the multiplication of tyloses in a band marking the advance of a wood~destroying fungus in a living tree would appear to furnish an unerring criterion for the recognition of the fungus as a parasite.
Over 6260 fungi have been isolated from samples of feathers, nests, pellets, droppings, cloaca contents and visceral organs of 92 species of free-living birds in Czechoslovakia and Yugoslavia. Several species have been obtained of fungi pathogenic to homeotherms, poikilotherms and plants, and many fungi recorded belong to the toxinogenic species.Cellulolytic fungi have participated with 44.3 % in the total number of fungi, and keratinolytic fungi with 18.3 %. The thesis has been confirmed that habits (specific bionomics) of the birds influence,. to a certain degree, the distribution of the fungi among them. According to the results of this study, the fungi may conveniently be divided into three categories with respect to the character of their distribution and dispersal by free-living birds: (1) euryornithochous fungi , the occurrence of which in birds is undetermined by the birds' habits (ubiqitous species: Aspergillus flavus, Gliocladium roseum, Rhizopus. nigricans, Trichothecium roseum); (2) mesoornitohochorous fungi, the occurrence of which in birds is determined to a certain degree by the birds' habits (e. g., Aphanoascus fulvescens, Arthrodermcr curreyi, Chrysosporium keratinophilum, C. tropicum, Mucor hiemalis); (3) stenoornithochoruss fungi, which are associated with certain habit groups of the birds only (e. g., Aphanoascus terreus, Arthroderma ciferrii, A. tuberculatum,.Aspergillus fumigatus, Chrysosporium pannorum, Ctenomyces serratus). The importance of free-living birds in the spreading of pathogenic organisms has also been discussed from a more general point of view, particularly with regard to several epidemiologically important aspects of bird ecology such as synanthropism, migration (making possible a long-range carriage of pathogens or infected vectors), colonial breeding or mass roosting.
In 1080 samples (feathers, nesls, pellets, droppings, cloaca contents and visceral organs) collected from 92 species of free-living birds in many localities and habitats, a total of 6266 fungi was found; we identified 232 species and 112 genera of fungi (73.4 % Ascomycetes, 12.4 % Fungi imperfecti, 8.0 % Zygomycetes, 5.7 % Endomycetes, 0.3 % Oomycetes; and 0.2 % Basidiomycetes). The most common fungal genera were, in plumage, Alternaria. Cladosporium, Arthroderma, Chaetominum, Aspergillus, Penicillium and Chrllsosporium; in nests, Alternaria, Arthroderma, Penicillium, Aphanoascus and Scopulariopsis; on pellets, Penicillium, Mucor, Aspergillus, Chrysosporium and Rhizopus; in droppings, Scopulariopsis, Mucor, Aspergillus, Penicillium and Candida; from the cloaca contents, we isolated Penicillium, Candida, Aspergillus, Scopulariopsis, a. o., and from the visceral organs, Aspergillus, Penicillium, Rhodotorula, and some others. Of the fungi with a potential pathogenicity for homeotherms, we obtained, e. g., Absidia corymbifera, Aspergillus fumigatus, Candida albicans, Cryptococcus neoforrnans and Microsporum gypseum. We confirmed also the presence of several species of toxinogenic fungi (e. g., Aspergillus flavus), of the fungi with a potential pathogenicity for poikilotherms (e. g., Beauveria bassiana. Paecilomyces jarinosus, P. fumoso-roseus), and of plant parasites (Cylindrocarpon radicicola, Fusarium oxysponom and Verticillium albo-atrum). Aphanoascus reticulisporum has been described as comb. nov. (basionym: Anixiopsis: reticulispom ROUTIEN). 66 species of the fungi have not been recorded earlier in Czechoslovakia. Similarly, the occurrence of 111 species had, apparently, not been confirmed in free-living birds on a world-wide scale.
Compounds containing a carboxylic acid group may be metabolized along several routes. However, the most common pathways involve conjugation reactions with amino acids or glucuronic acid. Also, chain shortening due to beta-oxidation occurs when the carboxylic acid group is attached to a suitable aliphatic moiety. The various metabolic reactions of compounds containing the carboxylic acid group were reviewed by Caldwell. I Additional reactions of the carboxy group have been discovered which reveal that numerous I ipophilic conjugates may be formed. This subject was reviewed by CaldweW3 and Quistad and Hutson4 and the findings demonstrate that the acids may undergo chain extension or incorporation into triglycerides, cholesterol esters, and fatty acid derivatives. In addition to these reactions which occur in the tissues, the metabolism (e.g., decarboxylation or reduction) of some carboxylic acids may be carried out by the intestinal microflora.
In der vorliegenden monographischen Studie sollen die indomalayischen Arten der Gattung Pycnolejeunea Spruce einer kritischen Betrachtung unterzogen werden. Nach der Feststellung, welche systematische Bedeutung den Einzelmerkmalen innerhalb der Gattung zukommt. wird im speziellen Teil zuerst das untersuchte Herbarmaterial quellemnässig aufgeführt. Es folgt dann als Hauptteil die Behandlung der wichtigsten Fragen, die sich während der Arbeit herausstellten und von grundlegender Bedeutung für einzelne Arten sind. Im dritten Abschnitt wird die Abgrenzung sowohl der Arten untereinander als auch die der Gattung gegen andere klargestellt. Die Beschreibung der im Material vorhandenen neuen Arten beendet diesen Teil. Zum Schluss folgen einige geographische Einzelheiten der indomalayische Formen.
Colonies of up to 30 cats, Felis catus, which were partially dependent upon man for direct provisioning with food, were recorded on 82% of 775 English farms. They lived at a mean density of 6.3 per km2. Members of one such colony were observed intermittently from 1978-81. These cats' ranging, foraging and scent marking behaviours are described briefly as a background to observations of their social interactions. The adult male's home range was 83 ha, whereas those of three adult females averaged 13.1 (SD 7.2) ha. The cats visited the observation barn independently of each other. When in the barn each cat differed in how it positioned itself with respect to the others. Furthermore, the cats' social relationships were structured by differences in the numbers, rates and types of interactions with one another. Some individuals were classed as net initiators of interactions, whereas others were net recipients. The tendency to rub the perioral and cheek regions of the face on another cat was the clearest single indicator of initiator status. Kittens were initiators to adults, females to the adult male, and some adult females were initiators to others. On average, each adult female in the colony rubbed on another once every 25.3 h. Behaviour within the colony was generally amicable, whereas towards outsiders it was aggressive. All adult females in the colony gave birth to kittens each year, and used communal nests. Females tended, groomed and nursed kittens other than their own, and cooperated with each other during parturition. Although a female might nurse certain kittens preferentially, these preferences were not necessarily for her own kitten. The frequency with which a female nursed a kitten and the frequency with which it rubbed on her were positively correlated. A case of infanticide, when an unrelated adult male killed kittens, is described, together with circumstantial reports indicating that this incident was not unique. Farm cat society appears to be structured centripetally, with interactions flowing predominantly from socially (and, sometimes, spatially) peripheral individuals to socially central ones.
The 70 Frullania species (+ 1 subspecies, 9 varieties, and 3 formae) belonging to 9 subgenera are confirmed in China, among which 1 subgenus and 4 species are new to science, and 12 species are newly reported from China. A new name (nom. nov.) and 12 combinations are proposed and several species are reduced to synonyms of other species. This study reveals the Frullania flora of China, particularly Yunnan and the neighboring provinces, to be most highly differentiated, containing many phytogeographic ally interesting taxa, and may be regarded as a center of the Paleotropic Frullania flora.
In den vorliegenden Untersuchungen wurde anhand umfangreichen Herbarmateriala sowie über längere Zeit ausgedehnter Geländestudien versucht, zwei Problemen nachzugehen, einem allgemeinen und einem speziellen. Bei den Flechten werden seit langem relativ primitiv gebaute krustige und morphologisch wie anatomisch hochentwickelte laubige und strauchige Typen unterschieden. Anhand einer Gruppe, die am Übergang zwischen beiden Lebensformen steht, eben der als (Sektion oder) Untergattung Placodium des Genus Lecanora sensu amplo zusammengefaßten Arten, sollte nun geprüft werden, wie sich die morphologische Differenzierung von Randloben usw. im anatomischen Bau auswirkt. Die leitenden Prinzipien finden sich nun gleichsam in den verschiedenen systematisohen Einheiten manifestiert und können etwa folgendermaßen skizziert werden: Die primitiveren, rein krustigen oder auch schon primitiv gelappten Formen verfügen noch nicht über eine echte Rinde. Die sog. Rinde besteht bei ihnen aus absterbendem und kollabierendem Thallusgewebe mit Einschluß alter Algernüllen. Als nächsthöhere Stufe sind die Arten mit einfacher, echter Rinde aufzufassen, welche als spezielles Organ aufgebaut wird und keine toten Algenhüllen mehr enthält. Im weiteren Verlauf der angenommenen Entwicklung kommt es zu einer verschiedenartigen Differenzierung der Rinde unter gleichzeitiger Reduktion des Markes. Bei den als Gattung Squamarina ausgeschiedenen Arten wird die Oberrinde allein zum formgebenden Element des Fleohtenkörpers; sie ist auch durch auffällige Starrheit ausgezeichnet. Das streng abgesetzte Mark wird weit mehr als bei den vorher genannten Typen zum Ablagerungsplatz anorganischer Bestandteile. Bei den meisten Arten von § Placodium ist der Cortex dagegen in eine Vielzahl kegeliger, unter sich nur wenig zusammenhängender Teile aufgelöst die jeweils straff mit dem Mark verbunden sind und den Loben dadurch ihre flache Gestalt geben. Von hier, wie von den Arten mit einfacher echter Rinde, führt nun durch Ausdifferrenzierung einer Unterrinde, die zusammen mit der Oberrrinde als Außenskelett wirkt, der Weg zu blattflechtenähnlichen Typen, bei denen das Mark mehr und mehr zum Verschwinden kommt bzw. nur als sehr lockeres Füllorgan dient. Als hier anzuschließende Sonderform können die Schildflechten gelten, die bei gleicher Grundstruktur ihre Verbindung mit dem Substrat auf einen Punkt reduziert haben. Parallel zur anatomiechen Differenzierung der Pilzkomponente steht das Verhalten der Algen: im Anfang eine mehrminder regellose Verteilung in den oberen Partien des Flechtenkörpers, dann die Entstehung einer fest definierten schicht, schließlich wieder - bei den auch unterseits berindeten Formen, bei denen das Mark sehr aufgelockert ist - die Auflösung dieser Ordnung und das Wandern der Algen in die für die Assimilation günstig gelegenen, belichteten Teile des Flechtdnkörpers. Die höchstentwickelten Formen verschiedener Gruppen können bereits als echte Laubflechten gelten, womit auch in einem kleinen Bereich wieder der Beweis erbracht wäre, daß die Lebensformen der Flechten nur als Entwicklungsstufen, nicht aber als reelle systematische Einheiten höherer Ordnung aufzufassen sind. Als zweites Problem sollte, im Zusammenhang mit den eben kurz skizzierten Untersuchungen und unter Berüoksichtigung der gefundenen Tatsachen, eine systematische Gliederung und Bewertung der lobaten Arten der Sammelgattung Lecanara erarbeitet werden. Die großen Verschiedenheiten der entsprechenden Spezies führten dabei zu einer Auflösung der ganzen Einheit, also zur Erkenntnis der Polyphylie der Gruppe und damit zum Nachweis vielfältiger, parallel gerichteter Entwicklungstendenzen. Insgesamt werden -- einige species incertae sedis ausgenommen - 88 Arten behandelt und auf 4 Gattungen verteilt, von denen eine -- Squamarina mit 15 Arten in 2 Sektionen -- neu aufgestellt werden mußte. Die bei Leoanora verbleibenden Arten werden in 11, größtenteils aus Sektionen bewertete Gruppen gegliedert. Eine Anzahl bisher anerkannter Spezies mußte eingezogen werden; 14 neue aus den Alpen, dem Mittelmeergebiet, dem Himalaya und aus Innerasien werden beschrieben.
As a preliminary step towards a more intensive research on the diversity of macromycetes in Greece, an updated check-list of the Greek mycoflora is presented together with information on the host-substrates and geographic occurrence. The data originated from a thorough literature search and the authors' field observations. In total, 58 families, 214 genera and 811 species of fungi are recorded belonging to Basidiomycetes. The systematics and nomenclature of the relative bibliography have been updated and suitably revised. The large gaps in our knowledge on the existence and distribution of higher fungi in Greece are emphasized.
Echinococcosis/hydatidosis, caused by Echinococcus granulosus, is a chronic and debilitating zoonotic larval cestode infection in humans, which is principally transmitted between dogs and domestic livestock, particularly sheep. Human hydatid disease occurs in almost all pastoral communities and rangeland areas of the underdeveloped and developed world. Control programmes against hydatidosis have been implemented in several endemic countries, states, provinces, districts or regions to reduce or eliminate cystic echinococcosis (CE) as a public health problem. This review assesses the impact of 13 of the hydatid control programmes implemented, since the first was introduced in Iceland in 1863. Five island-based control programmes (Iceland, New Zealand, Tasmania, Falklands and Cyprus) resulted, over various intervention periods (from < 15 to > 50 years), in successful control of transmission as evidenced by major reduction in incidence rates of human CE, and prevalence levels in sheep and dogs. By 2002, two countries, Iceland and New Zealand, and one island-state, Tasmania, had already declared that hydatid disease had been eliminated from their territories. Other hydatid programmes implemented in South America (Argentina, Chile, Uruguay), in Europe (mid-Wales, Sardinia) and in East Africa (northwest Kenya), showed varying degrees of success, but some were considered as having failed. Reasons for the eventual success of certain hydatid control programmes and the problems encountered in others are analysed and discussed, and recommendations for likely optimal approaches considered. The application of new control tools, including use of a hydatid vaccine, are also considered.
Untersuchungen zum Target und Wirkmechanismus der eNOS-Transkriptionsverstärker AVE9488 und AVE3085

Die Substanzen AVE9488 und AVE3085 der neuen chemischen Klasse der eNOS-Transkriptionsverstärker führen zu einer signifikanten Aktivierung des eNOSPromotors, die in vitro und in vivo in einer Erhöhung der eNOS-Expression auf mRNAund Proteinebene resultiert. Als Folge davon kommt es zu einer signifikant gesteigerten Synthese von bioverfügbarem Stickstoffmonoxid (NO). Dieser Effekt führt in Kombination mit einer ebenfalls beobachteten Normalisierung der eNOS-Entkopplung zu einer deutlichen antiatherosklerotischen Wirkung in ApoE-Knockout-Mäusen. AVE9488 und AVE3085 wurden initial durch eine phänotypische Reihentestung chemischer Bibliotheken in einer stabilen eNOS-Promotor-Reporter-Endothelzelllinie identifiziert. Daher waren bis zum heutigen Zeitpunkt molekulare Targets und ihre Wirkweise unbekannt. Das Ziel der vorliegenden Arbeit bestand daher darin, Targetproteine der eNOS-Transkriptionsverstärker zu identifizieren, zu validieren und Hinweise auf den zugehörigen Wirkmechanismus zu sammeln. Zur Identifizierung potenzieller Zielproteine wurden zwei unterschiedliche Strategien verfolgt. In einem genomischen Ansatz sollten einzelne, für die Aktivierung des eNOSPromotors durch AVE9488 und AVE3085 essentielle Transkriptionsfaktoren identifiziert werden. Eine in silico-Analyse des für die Substanzwirkung notwendigen 300bp-Promotorbereichs vor Transkriptionsstart auf konservierte Transkriptionsfaktor-Bindemotive ergab potenzielle Bindungsstellen für insgesamt 105 Transkriptionsfaktoren. In Gel-Retardierungsexperimenten mit Oligonukleotiden, die aufbauend auf der bioinformatischen Analyse die konservierten cis-Elemente in diesem Sequenzbereich abdeckten, und Kernextrakten aus Endothelzellen, die mit AVE9488 behandelt worden waren, konnte eine Abschwächung der DNA-Protein-Komplexbildung beobachtet werden. Diese Effekte konnten jedoch mit dieser Technologie aufgrund der hohen Anzahl möglicher bindender Transkriptionsfaktoren nicht auf einzelne eingeengt werden. Im Folgenden wurden daher einzelne Transkriptionsfaktoren ausgewählt, die eine nachgewiesene Rolle bei der Regulation der eNOS-Transkription besitzen, und auf ihre Bedeutung bei der Substanz- ermittelten eNOS-Promotoraktivierung untersucht. Nach Ausschaltung durch siRNA von Sp1, GATA-2, Ets-1, MAZ und PATZ1 im eNOS-Transkriptionstest konnten bei Sp1, GATA-2 und MAZ lediglich eine Erniedrigung der basalen eNOS-Promotoraktivitätfestgestellt werden. Da die Substanz-induzierte eNOS-Transkriptionserhöhung bei keinem der untersuchten Transkriptionsfaktoren beeinflusst wurde, scheinen diese Proteine bei diesem Prozess keine Rolle zu spielen. Als zweite Strategie kamen Proteomik-Techniken zur Identifizierung möglicher Targetprot eine durch ihre direkte biochemische Affinität zum Pharmakophor der eNOS-Transkriptionsverstärker zum Einsatz. Durch die chromatographische Anreicherung bindender Proteine aus Endothelzelllysaten an spezifischen Affinitätsmaterialien und der Markierung rekombinanter, humaner Proteine auf Protein-Mikroarrays mit Biotinmarkierten eNOS-Transkriptionsverstärkern konnte eine Gesamtliste mit insgesamt 18 potenziellen Targetkandidaten ermittelt werden. Die weitere zelluläre Validierung dieser Kandidaten erfolgte durch ihre siRNA-vermittelte Ausschaltung im eNOS-Transkriptionstest. Innerhalb aller potenziellen Targets konnte nur nach Ausschalten von Häm-bindenden Protein 1 (HEBP1) der eNOS-Promotor durch AVE3085 nur noch signifikant abgeschwächt gegenüber der Kontrolle aktiviert werden. Dieses Protein stellte somit den einzigen Targetkandidaten dar, der bei der Aktivierung des eNOS-Promotors eine Rolle zu spielen scheint. Nach rekombinanter Expression des humanen HEBP1 konnte die Bindung der eNOS-Transkriptionsverstärker an dieses Protein durch zwei unabhängige biochemische Methoden konfirmiert werden. Bei Messungen der Tryptophanfluoreszenz des HEBP1 konnte der eNOS-Transkriptionsverstärker 9257 den Liganden Hämin, der eine Quenchung der Fluoreszenz bewirkt, aus der Bindung verdrängen. Weiterhin konnte mit Hilfe einer Fluoreszenz-markierten eNOS-Substanz ihre direkte Bindung an das HEBP1 durch Messung der Fluoreszenzpolarisation nachgewiesen und ein KD-Wert von 11,7μM ermittelt werden. Abschließend konnte in ersten Untersuchungen der Hebp1-Expression in einem Herz-Kreislauf-relevanten Tiermodell für chronische Herzinsuffizienz nach Myokardinfarkt die differentielle Hochregulation des Hebp1-Gens um den Faktor 2,5 unter pathologischen Bedingungen nachgewiesen werden, die jedoch nicht durch die Behandlung mit AVE3085 beeinflusst wurde.
A brief account of the present state of weevil taxonomy is followed by a detailed study of certain structures used in their classification, namely the venter, abdominal tergites, sternite 8 of the male, apex of the hind tibia and deciduous mandibular processes. A key to some 50 families and subfamilies of Curculionoidea is followed by a list of family-group taxa. The following changes are made: Brachyceridae, Erirhinidae. The primary subdivisions of the brain (telencephalon, diencephalon, mesencephalon, metencephalon, and myelencephalon) have similar relations and comparable functions in all vertebrates. Accordingly, the landmarksthat define their boundaries can be regarded as reliable for following their development. On the basis of a more complete series of well preserved embryos than has been available hitherto, we present evidence that the subdivisions of the adult brain can be traced back to neural-fold stages in which a series of growth centers can be recognized, differing from one another in form, size, and relations. The possibility of following the constrictions between the various subdivisions throughout development has been doubted by some, notably Hochstetter (1919). At present we are convinced that they can be distinguished if certain criteria are followed. These are: (a) constrictions involve the neural tube as a whole; (b) constrictions do not give rise primarily to any neural centers; (c) constrictions change in relative length and width, and in certain stages they become inconspicuous in models. The anatomical descriptions of progressive stages of development have important practical implications. It is known, for example, that congenital malformations of the central nervous system in man are common and that they are responsible for a substantial portion of fetal wastage as well as infant mortality and morbidity. In certain patients comprehensive clinical studies may indicate the underlying abnormality, such as dysraphism, arhinencephaly, hypoplasia of the cerebellum. or absence of the corpus callosum. In addition, anatomical examination of the affected brains may reveal in detail such abnormalities as lyssencephaly, polymicrogyria, or other cortical dysgeneses. These very complex cerebral malformations can only be understood and unraveled in the light of normal development. An investigation of early development of the brain must necessarily begin with a stage in which the major landmarks of the adult brain can be readily identified.As progressively younger stages are analyzed certain landmarks can no longer be recognized, although others persist at least to the third week of gestation. We believe that the evidence on which this study is based can be followed more satisfactorily in this inverted sequence, and the detailed description is so presented. It is followed by a summary of the sequence of events written in the conventional manner, as far as the eighth week of gestation.
Helicobacter pylori (H. pylori) ist ein weit verbreitetes Humanpathogen, welches den menschlichen Magen besiedelt und zu schwerwiegenden entzündlichen Erkrankungen des gastralen Traktes führen kann. Bereits 1994 wurde das Bakterium als ein Klasse 1 Karzinogen deklariert, da H. pylori im erwiesenen Maße mit der Entstehung von hochinvasivem Magenkrebs in Verbindung gebracht wird. In vitro induziert H. pylori eine starke Migration der infizierten Epithelzellen, die unter anderem mit der Auflösung der Zellkontakte einhergeht. Die zugrunde liegenden molekularen Zusammenhänge konnten bisher noch nicht vollständig aufgeklärt werden. Die Mechanismen der Auflösung der Zelladhäsion wurden in der vorliegenden Arbeit untersucht, um einen tieferen Einblick in die H. pylori vermittelten Virulenz zu erhalten. So konnte eine H. pylori-induzierte Dissoziation des E-Cadherin Komplexes, bestehend aus p120, 􀄮- und 􀈕-Catenin beobachtet werden, der in einem Verlust der Zelladhäsion resultierte. Es konnte darüber hinaus eine Spaltung der extrazellulären Domäne von ECadherin detektiert werden, die wahrscheinlich zu einer Destabilisierung und somit zur Auflösung des gesamten E-Cadherin Komplexes führte. Durch den Zerfall der Adhärenzverbindungen wurden Catenine in den zytoplasmatischen Pool freigegeben, von denen p120 in den Zellkern translozierte und die Transaktivierung von Zielgenen auslöste, die in diesem Zusammenhang mit Hilfe von Reportergenanalysen quantifiziert wurden. Diese Prozesse zeigten sich von dem Pathogenitätsfaktor CagA (cytotoxin associated gene A) unabhängig, der über das bakterielle Typ IV Sekretionssystem in die Wirtszellen transloziert wird und krebsassoziierte Signaltransduktionswege aktivieren kann. In weiteren Untersuchungen wurden deshalb die Auswirkungen löslicher H. pylori Faktoren auf die Spaltung von E-Cadherin und folglich auf die Zellmotilität und Morphologie epithelialer Zellen analysiert. Aufgrund dieser Beobachtungen wurden in weiteren Experimenten proteolytische Aktivitäten von H. pylori untersucht. Dabei konnte erstmalig die hypothetische Protease HtrA (high temperature requirement protein A) von H. pylori durch massenspektrometrischen Analysen als eine caseinolytisch aktive Protease gefunden werden. Nach der Klonierung und Aufreinigung von HtrA konnte darüber hinaus auch E-Cadherin als spezifisches biologisches Substrat der Wirtszellen für HtrA identifiziert werden. Diese selektive Fragmentierung von E-Cadherin durch HtrA fügt sich als ein neues Element in das Modell der H. pylori Pathogenese, die einen initialen Schritt in der frühen Phase der Infektion darstellen könnte.
Identifizierung und Charakterisierung neuartiger 5-Lipoxygenase-Inhibitoren – in silico und in vitro

Ziel dieser Arbeit war die Identifizierung und Charakterisierung neuer potenter 5-LO-Inhibitoren unter Verwendung sowohl computergestützter als auch experimenteller Methoden. Ausgangspunkt war ein ligandenbasiertes virtuelles Screening unter Verwendung der ladungsbasierten Deskriptoren Charge 3D und TripleCharge3D. Hierbei konnten zwei neue direkte 5-LO-Inhibitoren identifiziert werden. Jede dieser beiden Substanzen diente als Startpunkt weiterer virtueller Screenings mit dem Ziel, die Potenz der Substanzen zu verbessern bzw. eine SAR der Substanzklasse zu erhalten. Dabei zeigte sich für die Klasse der Thiazolinone, dass eine hohe Toleranz gegenüber unterschiedlichen Substituenten am Grundgerüst bezüglich der Auswirkung auf die Aktivität vorliegt: insbesondere werden relativ große Substituenten toleriert. Des Weiteren scheint der 2-Phenylsubstituent für die 5-LO-inhibitorische Aktivität essentiell zu sein, da Derivate, die einen Heterozyklus an dieser Position aufweisen, inaktiv sind. Eines der aktivsten Derivate dieser Klasse, C06 (Substanz 12), konnte weiter molekular-pharmakologisch charakterisiert werden. Die Substanz zeigt keine offensichtlichen zytotoxischen Effekte, ist unabhängig vom Stimulus der 5-LO-Aktivierung und zeigt nanomolare inhibitorische Aktivität sowohl in intakten PMNL (IC50-Wert 0,65 =M) als auch in PMNL-Homogenaten (IC50-Wert 0,66 =M) sowie in zellfreiem PMNL-S100 (IC50-Wert 0,26 =M) und am gereinigten Enzym (IC50-Wert 0,3 =M). C06 ist selektiv für die 5-LO, da andere arachidonsäurebindende Proteine (PPARs, cPLA2 und 12- und 15-LO) nicht beeinflusst werden. Auch Nager-5-LO (aus der Ratte und der Maus) wird inhibiert mit IC50-Werten im nanomolaren Bereich. Allerdings zeigte sich die Substanz inaktiv in einem menschlichen Vollblutassay in Gegenwart von Serum. C06 scheint nicht an die für die Interaktion der 5-LO mit der Membran verantwortliche C2-ähnliche Domäne der 5-LO zu binden. Ebenso hat der Membranbestandteil Phosphatidylcholin keinen bzw. nur geringen Einfluss auf das inhibitorische Potential von C06. Aktivitätstests mit steigender Substratkonzentration deuten auf einen allosterischen Bindemodus von C06 hin. Diese experimentellen Daten flossen in die Identifizierung des putativen putativen Bindemodus an der 5-LO ein. Hierzu wurde zunächst ein Homologie- Modell der 5-LO unter Verwendung der kürzlich neu veröffentlichten Daten der Röntgenkristallstruktur der Kaninchen-15-LO (PDB-Code: 2p0m [Choi et al., 2008]) erstellt. Durch eine Bindetaschenvorhersage mit dem Programm PocketPicker und einer pseudorezptorbasierten Auswahl der potentiellen Bindetasche mit anschliessendem Liganden-Docking konnten zwei Bindebereiche postuliert werden, beide an einer Oberflächenbindetasche der 5-LO ausserhalb des aktiven Zentrums. .....
In der vorliegenden Dissertation konnte gezeigt werden, dass Hyperforin wichtige Funktionnen von Keratinozyten beeinflusst. Hyperforin triggert die Ausdifferenzierung der epidermalen Zellen und inhibiert gleichzeitig ihre Proliferation. Diese Ergebnisse zeigen die physiologische Relevanz der Hyperforin-vermittelten Effekte, da diese beiden Prozesse in der Haut normalerweise gegenläufig ablaufen. Weiterhin war die von Hyperforin ausgeübte Ausdifferenzierungs-Zunahme und Proliferationshemmung vergleichbar mit den Effekten einer hohen [Ca2+]ex, die in vivo die Funktionen von Keratinozyten steuert. Die Frage, die aus diesen Ergebnissen resultierte, war, über welchen Mechanismus Hyperforin die Effekte in Keratinozyten beeinflusst. Die Untersuchung des Mechanismus der Hyperforin-induzierten Effekte zeigte, dass Hyperforin über die Aktivierung des TRPC6-Kanals einen Calcium-Influx in Keratinozyten bewirkt und resultierend daraus die Ausdifferenzierung von Keratinozyten anregt. Auch hier ergeben sich Parallelen zu der durch hohes [Ca2+]ex -induzierten Ausdifferenzierung, die ebenfalls einen Calcium-Einstrom in Keratinozyten auslöst. Eine Microarray-Analyse von Hyperforin-inkubierten Keratinozyten und anschließende Experimente mit selektiven Inhibitoren zeigte die Beteiligung der PKC/Ras/MEK/ERK-Signalkaskade. Interessanterweise gibt es hier ebenfalls Ähnlichkeiten zwischen Hyperforin und einer hohen [Ca2+]ex, da bekannt ist, dass eine hohe [Ca2+]ex die Ausdifferenzierung zumindest teilweise über diesen Signatransduktions-Weg steuert. Aufgrund der Parallelen zwischen der Hyperforin- und Ca2+-induzierten Ausdifferenzierung und den vermehrten Hinweisen auf die wichtige Rolle von TRPC-Kanälen in Keratinozyten, stellte sich die Frage, ob eine hohe [Ca2+]ex ebenfalls zur Aktivierung von TRPC-Kanälen führt. Tatsächlich konnten wir in dieser Arbeit zeigen, dass Calcium in Keratinozyten mehrere TRPC-Kanäle aktiviert und so einer Erhöhung der [Ca2+]i führt. Hierdurch konnte zum ersten Mal auch die Beteiligung von DAG-aktivierten TRPC-Kanälen an der Ca2+-induzierten Ausdifferenzierung gezeigt werden. Bei der Untersuchung von Psoriasis-Keratinozyten, die eine Hyperproliferation und erniedrigte Ausdifferenzierung in vivo aufweisen, konnte in der vorliegenden Dissertation eine grundlegende Störung des Calcium-Haushaltes festgestellt werden. Dabei zeigten die Psoriasis-Keratinozyten im Vergleich zu gesunden hPKs eine abnorme Antwort auf unterschiedliche Stimuli, die zu einer Erhöhung der [Ca2+]i führen. Sowohl eine hohe [Ca2+]ex, als auch Hyperforin führte zu einem deutlich erniedrigten Calcium-Einstrom in den Keratinozyten der Psoriasis-Patienten. Aus diesen Ergebnissen resultiert die Frage nach den Ursachen dieser Störungen. Da in dieser Arbeit bereits gezeigt werden konnte, dass Hyperforin und eine hohe [Ca2+]ex über TRPC-Kanäle die Ausdifferenzierung in Ketatinozyten beeinflussen, untersuchten wir die Expression von TRPC-Kanälen in Psoriasis-Keratinozyten. Es konnte gezeigt werden, dass die Expression aller TRPC-Kanäle signifikant erniedrigt war. Vor dem Hintergrund der wichtigen Rolle der TRPC-Kanäle für die Calcium-Homöostase und Ausdifferenzierung von epidermalen Zellen, ist dies durchaus eine denkbare und plausible Erklärung für die Defekte der Psoriasis-Keratinozyten. Weiterhin zeigten unsere Ergebnisse, dass der CaR in Psoriasis-Keratinozyten vermindert exprimiert wird. Da dem CaR eine wichtige Rolle in der Regulation der [Ca2+]i als Antwort auf eine Änderung der [Ca2+]ex zugeschrieben wird, könnte dies eine weitere mögliche Erklärung für die gefundenen Störungen der Psoriasis-Keratinozyten sein.
Delthyridoid spiriferids are characterized by a global abundance and fast evolution during Silurian and Devonian, and, therefore, are used as important biostratigraphical and palaeobiogeographical tools. In this work, delthyridoid brachiopod faunas from different regions of today’s world, resp., of different palaeobiogeographical units, are compared side-by-side to investigate their phylogenetic relationships and to improve, in a second step, the palaeobiogeography from Late Silurian to Early Eifelian time. A new systematics of Delthyridoidae is established which is more complicated than hitherto assumed. The results of this study are mainly based on direct comparison of articulated and isolated brachiopod shells, external and internal moulds, as well as latex casts and serial sections. The computer supported cladistic analyses have turned out not to be useful due to different kinds of preservation resulting in an incomplete matrix which is insufficient for reliable cladograms. A further problem in terms of cladistical analyses are various convergences during the evolution of spiriferids. Many characters evolved independently from each other at different times in each lineage so that autapomorphies are hardly or not at all recognizable. As a result, families and genera are only definable by a combination of characters rather than by a single or a few autapomorphies. As a new method, 3D reconstruction from serial sections is introduced which made it possible for the first time to compare directly mouldic and shelly material. Preliminary results are presented herein. Statistical analyses of measurements taken from new taxa are made but regarded as a descriptive argument rather than a deciding factor for taxonmy due to incomplete preservation and/or tectonic deformation. Brachiopods, especially type material, from collections of different institutions and museums are studied as well as personal material, whenever possible collected from topotype outcrops. Emended diagnoses, if necessary, from family to species level are given. During this work several new taxa have been erected: 7 new families: Australospiriferidae, Murchisonispiriferidae, Orientospiriferidae, Otospiriferidae, Patriaspiriferidae, Rostrospiriferidae, and Trigonospiriferidae; 6 new genera, 1 of these in open nomenclature: Cyclopterospirifer, Hallispirifer, Parlinispirifer, Murchisonispirifer, Shujiapingensispirifer, and gen. nov. B; and 3 new species: Patriaspirifer merriami, Patriaspirifer johnsoni, and Murchisonispirifer feldmani; 1 taxon is defined as nomen novum: Orientospirifer nakaolingensis wani. In the framework of this project, 2 families: Filispiriferidae and Multispiriferidae; 1 subfamily: Multiplicatispiriferinae, 6 genera, 1 of them in open nomenclature: Frequentispirifer, Leonispirifer, Multiplicatispirifer, Ovetensispirifer, Turcispirifer, and Gen. A; and 9 new species, 3 of them in open nomenclature: Filispirifer hamadae, Leonispirifer leonensis, Multiplicatispirifer foumzguidensis, Oventensispirifer novascotianus, Quiringites arensentiae, Turcispirifer turciae, Multiplicatispirifer cf. foumzguidensis, Quiringites cf. arensentiae, and ?Turcispirifer sp. A which have already been established are also described in this work. The brachiopod faunas studied consist of externally very similar spiriferids which have been identified as same genera, species, or even subspecies in earlier times. These forms are considered as 6 distinct morphotypes Howellella-, Arduspirifer-, Acrospirifer-, Euryspirifer-, Paraspirifer-, and Multiplicatispirifer-like morphotypes, which are briefly introduced. The new systematics is characterized by different clades, the European/North African delthyridoid spiriferid clade, the North American delthyridoid spiriferid clade, the Asian delthyridoid spiriferid clade, the Malvinokaffric delthyridoid spiriferid clade, and the delthyridoid multiplicated spiriferid clade. Each of them is described in a cladistic and in a phylogenetic way. Their phylogenetic relationship sheds new light on palaeobiogeographical interpretations for the different stages from Late Silurian to early Middle Devonian time. A tendency for increasing endemicity is seen until the end of the Early Emsian, which is interrupted by short term regional faunal exchange within a province or within a realm, followed by a loss of endemicity resulting in global distribution of brachiopod genera until the end of Givetian time. The Old World Realm is re-defined due to the lack of phylogenetic relationship between its faunas and subdivided into the European Realm, consisting of the Gondwanan and Avalonian provinces, and the Asian Realm, consisting of the Siberian, Sino, and Mongolian provinces. A reconstruction of Lower Devonian palaeobiographical map is introduced.
TeaABC from the halophilic bacterium Halomonas elongata belongs to the family of tripartite ATP-independent periplasmic (TRAP) transporters. It facilitates the uptake of the compatible solutes ectoine and hydroxyectoine which protect the cell from dehydration by accumulating in the cytoplasm during hyperosmotic stress. It is the only known TRAP transporter activated by osmotic stress. Ectoine and hydroxyectoine accumulation in H. elongata is regulated by the cytoplasmic universal stress protein TeaD. The gene encoding TeaD is located in the same operon as the TeaABC gene. TeaD regulates the cellular homeostasis of ectoine possibly by interacting directly or indirectly with TeaABC. All subunits of TeaABC and TeaD were expressed in E. coli and purified. With TeaD and the solute binding protein (SBP) TeaA high levels of expression suitable for crystallization could be obtained and their 3D structures solved. The small transmembrane protein TeaB and the transporter TeaC showed only moderate and low levels of expression respectively. Functional analysis on TeaA was performed using Isothermal Titration Calorimetry. The measurements demonstrate that TeaA is a high affinity ectoine-binding protein (Kd = 0.19 _M) that also has a significant affinity for hydroxyectoine (Kd = 3.8 _M). The structure of TeaA was solved using ab initio phase determination by MAD (multiple anomalous dispersion). TeaA structures were determined in three conformations: TeaA alone, TeaA in complex with ectoine and TeaA in complex with hydroxyectoine. The resolutions of the structures were 2.2, 1.55 and 1.80 Å, respectively. These represent the first structures of an osmolyte SBP associated to a TRAP transporter. The structures reveal similar ligand binding compared to osmolyte SBPs of ABC transporter pointing to coevolution of the ligand binding modes. Moreover, unique features such as the solvent-mediated specific binding of the ligands ectoine and hydroxyectoine could be observed for TeaA. The structure of TeaD in complex with its cofactor ATP was solved by molecular replacement at a resolution of 1.9 Å. Comparison with other structures of universal stress proteins shows striking oligomerization and ATP binding in TeaD. In conclusion, this work presents the first detailed analysis of the molecular mechanisms underlying ligand recognition of an osmoregulated transporter from the TRAP-transporter family.
Strukturelle Analyse des CusCBA-Systems von Escherichia coli Kupfer ist als Kofaktor in vielen Enzymen ein essentielles Spurenelement. Die Aufrechterhaltung der Kupferhomöostase ist für die Zelle enorm wichtig, da es sich um ein redox-aktives Übergangsmetall handelt, das selbst in geringsten Konzentrationen toxisch wirkt. Gewöhnlich ist in der Zelle kein einziges freies Kupferion nachweisbar, da die Zelle redundante Mechanismen für die Detoxifikation von Kupfer besitzt. Ein Mechanismus zur Detoxifikation von Kupfer und Silber in E. coli ist das Cus-System. Es handelt sich um einen vierteiligen Effluxkomplex, der sich aus dem inneren Membranprotein CusA, dem periplasmatischen Membranfusionsprotein CusB und dem TolC ähnlichen äußeren Membranprotein CusC zusammensetzt. Das vierte Protein dieses Systems, CusF, dient im Periplasma als Kupferchaperon. Dieser Komplex ermöglicht das Ausschleusen von Cu(I)- und Ag(I)-Ionen aus dem Cytoplasma über das Periplasma und die äußere Membran in einem einzigen Schritt. In dieser Arbeit sollte die Röntgenstruktur des periplasmatischen Proteins CusB geklärt werden, um anhand struktureller Daten analysieren zu können wie CusA und CusC über CusB miteinander verbunden sind und welche Konformationsänderungen dabei vonstatten gehen. CusB wurde dafür über Ni2+-Chelat-Affinitätschromatographie und Größenausschluss-Chromatographie bis zur Homogenität gereinigt. Das Protein lag in Lösung als Monomer vor. Kristalle von CusB wurden nach der Dampfdiffusionsmethode des hängenden Tropfens hergestellt, wobei Kristalle in nur einem von 500 verschiedenen Ansätzen entstanden sind. Röntgenstreuung wurde bis zu einer Auflösung von 8 Å am ESRF (European Synchrotron Radiation Facility) in Grenoble gemessen. Die Streuung der Kristalle ließ starke Anisotropie und hohe Mosaizität erkennen. Um die Qualität der Kristalle von CusB zu verbessern, wurden Kristalle des Proteins ohne Hexahistidinanhängsel hergestellt. Diese Kristalle zeigten in Röntgenstreuungsexperimenten keine Verbesserung der Auflösung und Qualität. Die Röntgenstrukturen und Analysen durch Protease-Verdau von den zu CusB verwandten Proteinen AcrA und MexA zeigten, dass in diesen die N-Termini und C-Termini unstrukturiert sind. Deswegen wurden zunächst Konstrukte von CusB hergestellt in denen verschieden lange Bereiche des N-Terminus deletiert wurden. Ein Konstrukt von CusB, in dem die ersten 20 Aminosäuren deletiert waren, konnte in 10 von 500 Ansätzen kristallisiert werden. Nach Feinabstimmung der initialen Ansätze wurde für Kristalle dieses Konstrukts eine Auflösung von 5,3 Å am ESRF in Grenoble gemessen. Allerdings wies die Röntgenstreuung ebenfalls ein starke Anisotropie und Mosaizität auf, so dass die Struktur dieses Proteins nicht gelöst werden konnte. Strukturelle Analyse des RNAi-Suppressors B2 des Nodamura Virus: RNAi (RNA-Interferenz) bezeichnet einen sequenzspezifischen RNA-Degradationsprozess, um die Synthese eines Proteins zu verhindern. Zwei RNA-Typen wirken als Auslöser der RNAi: Doppelsträngige RNA dient als Vorläufer von siRNAs (small interfering RNAs), während einzelsträngige RNA mit Stamm-Schleifen-Strukturen als Vorläufer der miRNA (microRNA) dient. SiRNA und miRNA werden durch die Typ III Endonuklease Dicer im Cytoplasma produziert, sind 21-30 Nukleotide lang mit charakteristischen 2-NukleotidÜberhängen am 3’-Ende. Über den Komplex aus Dicer und dem doppelsträngige RNA-bindenden Protein R2D2 werden diese kleinen RNAs an das Protein Argonaute (AGO) abgeben. Dieses baut einen Strang der doppelsträngigen, kleinen RNAs über seine RNAse-Aktivität ab und hält den anderen (Führungs-) Strang gebunden. Daraufhin wird entweder komplementäre mRNA abgebaut oder die Translation komplementärer mRNA verhindert. RNAi dient im Organismus unter anderem der Verteidigung gegen Viren, wobei die Expression viraler Proteine durch RNAi verhindert wird. Durch Koevolution haben Viren allerdings Mechanismen zur Unterdrückung der RNAi in den Wirtszellen entwickelt. Ein RNAi Suppressor ist das Protein B2 des Nodamura Virus (NMV B2). Um Mechanismen und Gemeinsamkeiten der RNAi Suppression durch Viren analysieren zu können, wurde in dieser Arbeit die Röntgenstruktur der RNA bindenden Domäne von NMV B2 gelöst. Hierfür wurde ein Konstrukt (Aminosäuren 1-79) bis zur Homogenität aufgereinigt. Kristalle wurden mit einer Proteinkonzentration von 15 mg/ml mittels der Dampfdiffusions-Methode des hängenden Tropfens hergestellt. Diese wuchsen innerhalb von zwei Tagen als lange Nadeln mit Ausmaßen von 200 x 10 x 10 μm. Bei Messungen am ESRF in Grenoble wurde eine Auflösung bis 2,5 Å erreicht. Das Protein kristallisierte mit einem Dimer pro asymmetrischer Einheit. Die Kristalle wuchsen in der Raumgruppe P212121 mit den Einheitszelldimensionen a = 32.2, b = 56.6, c = 98.6. Die Phase wurde über molekularen Ersatz mit der Struktur des homologen Proteins B2 des Flock House Virus (FHV B2) bestimmt. Die Struktur stellte sich als ein gestrecktes Dimer mit einer Größe von ca. 55 x 10 x 15 Å, bestehend aus drei Alpha-Helices pro Monomer dar. Trotz geringer Sequenzidentität von NMV B2 und FHV B2 zeigten beide Strukturen ein Vier-Helix-Bündel, das von einer sehr kurzen Helix am C-Terminus bedeckt ist. Bei einem Vergleich der RNA-bindenden Aminosäurereste der beiden Strukturen fällt ein hoher Grad an Konservierung auf. Von zehn RNA-interagierenden Resten sind fünf identisch. Die RNA bindenden Reste werden von beiden Monomeren des Dimers beigetragen. So ist wohl mindestens ein Dimer für die RNA-Bindung durch B2 Proteine notwendig.
Die zellfreie Proteinsynthese hat sich in den letzten Jahren zu einem potenten Werkzeug – auch in der Produktion von Membranproteinen – entwickelt. Da keine lebenden Zellen genutzt werden, kann der Prozess der präparativen Membranproteinproduktion vereinfacht und individuell optimiert werden. Im Gegensatz zu konventionellen zellbasierten Expressionssystemen gewährleistet die zellfreie Proteinsynthese die direkte Zugänglichkeit zum Reaktionsort und damit die Möglichkeit der unmittelbaren Kontrolle. Dies ermöglicht eine genaue Anpassung der Reaktionsbedingungen auf das Zielprotein. Die Verbesserung und Entwicklung neuer Modi der zellfreien Membranproteinsynthese war ein Teil der vorliegenden Arbeit. Setzt man dem Zellfrei-System von Außen keine hydrophobe Umgebung zu, so präzipitiert das neu-synthetisierte Membranprotein im Reaktionsmix (P-CF). Interessanter Weise unterscheiden sich diese Präzipitate von den aus der E.coli zellbasierten Proteinproduktion bekannten Einschlußkörperchen, da sie sich teilweise leicht in mildem Detergenz resolubilisieren lassen. Zudem konnte für verschiedene Transportproteine, die aus Präzpitat resolubilisiert und danach in Liposomen rekonstituiert wurden, spezifische Transportaktivität gezeigt werden (z.B. eukaryotische Ionentransporter, Multi-Drug Resistenzproteine von E.coli). Alternativ können die Membranproteine direkt, durch die Zugabe von Detergenzien in den Reaktionsmix, solubilisiert werden (D-CF). Um die einzelnen Expressionsmodi zu optimieren wurden 24 gebräuchliche Detergenzien auf ihre Eigenschaft hin gestestet, strukturell sehr unterschiedliche Membranproteine zu solubilisieren. Die Familie der langkettigen Polyoxyethylen-alkyl Ether hat sich dabei als sehr geeignet erwiesen um das prokaryotische α-helikale Multi-Drug Resistenzprotein EmrE, den bakteriellen vornehmlich aus ß-sheets bestehenden Transporter Tsx und den eukaryotischen G-Protein gekoppelten Vasopressin Rezeptor V2R direkt im D-CF Modus zu solubilisieren. Zudem konnte eine Abhängigkeit der spezifischen Aktivität von Tsx vom verwendeten Expressionsmodus bzw. des verwendeten Detergenz mit Hilfe der Black Lipid Membrane´ Methode gezeigt werden. Die Expression eines repäsentativen Teils von 134 Zielproteinen des inneren Membranproteoms von E.coli wurde in drei verschiedenen Zellfrei-Expressionsmodi getestet. Ein an jedes Zielprotein des Membranroteoms C-terminal fusioniertes GFP diente der Konzentrationsbestimmung im D-CF Expressionsmodus. Die Faltung von GFP ist in Anwesenheit von Detergenz signifikant reduziert. Zunächst wurden alle Zielproteine in einem batch´ System im D-CF Modus im Mikrotiterplatten Maßstab mit Hilfe eines Roboters hergestellt. Die Etablierung einer robotergestützten Plattform, welche das Pipettieren, Inkubieren und Detektieren kombiniert, diente als Grundlage für den Herstellungsprozess des Membranroteoms von E.coli in einem Medium-Durchsatz Verfahren in batch´ Konfiguration. In dieser ersten Stufe des Screens im D-CF Modus konnten 84 Zielproteine (63%) aufgrund der detektierten GFP-Fluoreszens in Mengen von 1 bis 60μg pro mL Reaktion als erfolgreich produziert identifiziert werden. Zudem wurde das Membranproteom in dem effektiveren continous exchange (CE) Verfahren im P-CF, wie auch im D-CF Modus wiederholt exprimiert. Im Vergleich zur batch´ Konfiguration konnten im CE D-CF Modus deutlich mehr Zielproteine (75%) als positiv identifiziert werden. 16 Zielproteine wurden dabei bereits in Expressionsmengen von mehr als 100μg solubilisierte Membranproteinfusion pro mL Reaktionsmix gewonnen. 99 Zielproteine (74%) konnten als positiv identifiziert werden, nachdem die unlösliche Fraktion der CE P-CF Reaktion elektrophoretisch getrennt und angefärbt wurde. Für 66 Kandidaten (49%) stellt das produzierte Protein nach Coomassie-Färbung eine dominante Bande, und damit (semi-)präparative Proteinmengen, dar. Der Erhalt von Detergenz-solubilisierten Membranproteinproben von hoher Qualität ist ein wichtiger Schritt zur Gewinnung struktureller sowie biochemischer Daten. Das E.coli α-helikale Multi- Drug Resistenz Protein SugE konnte im CE P-CF Verfahren in präparativen Mengen von mehr als 2mg Protein pro mL des Reaktionsansatzes gewonnen werden. Durchgeführte analytische Größenausschlusschromatographie zeigte, dass der Transporter unter optimierten Reaktionsbedingungen in einem homogenen, schlanken Peak eluiert. Mittels elektronenmikroskopischer Gefrierbruchanalysen konnte eine effiziente und homogene Rekonstitution von SugE in E.coli Liposomen gezeigt werden. Bindungsstudien unter der Verwendung fluoreszensbasierter Anisotropie-Messungen haben gezeigt dass Proflavin – im Gegenteil zu Ethidium – ein Substrat von SugE ist. YedZ ist ein 24kDa leucinreiches Membranprotein mit sechs putativen Transmembransegmenten und enthält zwei Kofaktoren, ein Häm b und ein Flavin-mononukleotid (FMN). Im P-CF Modus exprimiertes YedZ kann effizient in den Detergenzien LMPG, LPPG, SDS und DPC resolubilisiert werden. Analytische Größenausschlusschromatographie zeigte einen symmetrischen Elutionspeak der apo-Form. Mittels CD-Spektroskopie des gereinigten apo-YedZ in 0.02% DDM wurde ein α-helikaler Sekundärstrukturanteil von 55% ermittelt. Zur Gewinnung von holo-YedZ wurde anstatt Hämb das chemisch verwandte Hemin eingesetzt. Die aufgenommenen UV/Vis Spektren der zellfrei produzierten holo-YedZ Proteinprobe in ihrer oxydierten und reduzierten Form, zeigen zu einer in vivo exprimierten Vergleichsprobe identische Absorptionsmaxima. Für sechs G-Protein gekoppelten Rezeptoren konnte die zellfreie Expression in präparativen Mengen gezeigt werden. Das Steroid-Derivat Digitonin, sowie einzelne Mitglieder der Detergenzfamilie der langkettigen Polyoxyethylen-alkyl Ether, wurden als am geeignetsten für die lösliche Expression der GPCRs im CE D-CF Verfahren ermittelt. Löslich in Anwesenheit von Brij78 produzierter GPCR Proben, zeigten nach Negativfärbung in elektronenmikroskopischen Einzelpartikelanalysen eine homogene Probenpräparation und geben Hinweis auf eine strukturelle Dimerisierug der Rezeptoren. Detergenzsolubilisierte Rezeptoren konnten in Liposomen, basierend auf E.coli Lipid-Mischungen, rekonstituiert werden. Elektronen-mikroskopische Gefrierbruchanalysen zeigten eine homogene Rekonstitution, welche auf eine funktionelle Faltung der Rezeptoren schließen lässt.
NADPH-Oxidasen der Nox-Familie sind eine wichtige Quelle für reaktive Sauerstoffspezies (ROS, reactive oxygen species) in verschiedenen Geweben und Zellen. Die Isoformen der NADPH-Oxidase unterscheiden sich dabei in ihrer physiologischen Funktion: Während Nox1 und Nox2 eher akute, Agonisten-induzierte ROS-Signaltransduktion vermitteln, sind Nox4-abhängig produzierte ROS an chronischen Prozessen wie Differenzierung beteiligt. Die Isoformen der NADPH-Oxidase unterscheiden sich darüber hinaus in ihrer intrazellulären Lokalisation, der Art der Aktivierung und der Spezies der abgegebenen ROS. Nox1 muss durch die Interaktion mit zytosolischen Untereinheiten (NoxA1 und NoxO1) aktiviert werden und produziert dann hauptsächlich Superoxidanionradikale (O2-). Nox4 dagegen ist unabhängig von zytosolischen Untereinheiten und somit konstitutiv aktiv und setzt eher Wasserstoffperoxid (H2O2) in den Extrazellulärraum frei. Zwischen diesen Unterschieden, deren strukturelle Ursachen weitgehend unbekannt sind, und den unterschiedlichen Funktionen von Nox1 und Nox4 besteht wahrscheinlich ein Zusammenhang. In transfizierten HEK293-Zellen konnte zunächst durch Immunofluoreszenz-Mikroskopie und subzelluläre Fraktionierung gezeigt werden, dass Nox1 in der Plasmamembran lokalisiert ist und Nox4 in der Membran des endoplasmatischen Retikulums (ER) verbleibt. Um die strukturellen Unterschiede zwischen Nox1 und Nox4, die eine Rolle bei der intrazellulären Lokalisation, den Aktivierungsmechanismen und der Art der abgegebenen ROS spielen könnten, zu identifizieren, wurden chimäre Proteine aus Nox1 und Nox4 konstruiert und analysiert. Zunächst konnte gezeigt werden, dass die konstitutive Aktivität von Nox4 durch den zytosolischen Bereich vermittelt wird. Dafür ist allerdings der komplette zytosolische Bereich ab der 6. Transmembrandomäne nötig, chimäre Konstrukte mit einem kürzeren Anteil des zytosolischen Bereichs von Nox4 waren nicht aktiv. Für die Aktivierung von Nox1 dagegen ist der zytosolische Bereich nicht ausreichend, vermutlich spielen weitere Interaktionen mit Bereichen im transmembranen Teil des Proteins ebenfalls eine Rolle. Für die korrekte intrazelluläre Lokalisation benötigen Membranproteine ein N-terminales Signalpeptid. Wenn das vorhergesagte Signalpeptid von Nox1 durch das von Nox4 ausgetauscht wurde, zeigte dieses chimäre Protein keine Plasmamembran-Lokalisation mehr, es war stattdessen in vesikelähnlichen Strukturen unterhalb der Plasmamembran lokalisiert. Das Signalpeptid von Nox1 war nicht dazu in der Lage, Nox4 zur Plasmamembran zu transportieren, das Protein war weiterhin im ER lokalisiert, was darauf hindeutet, dass Nox4 noch weitere Mechanismen besitzt, die seine ER-Lokalisation bedingen. Der Austausch des Signalpeptids von Nox4 gegen das von Nox1 führte dazu, dass das chimäre Protein statt H2O2 O2- produzierte. Dieses Ergebnis spricht dafür, dass der N-Terminus von Nox4 bei der H2O2-Produktion eine Rolle spielt. Experimente mit Nox1 und Nox4 mit N-terminalem Myc-Tag deuteten darauf hin, dass nur der N-Terminus von Nox1 prozessiert wird, also dass das Signalpeptid während der co-translationalen Translokation abgespalten wird. Ohne Signalpeptid waren sowohl Nox1 als auch Nox4 inaktiv. Die dritte extrazytoplasmatische Schleife von Nox4 enthält 28 zusätzliche Aminosäuren verglichen mit Nox1, die sich auf zwei Bereiche aufteilen. Eine Deletion der Aminosäuren, die nur in Nox4 vorhanden sind, führte dazu, dass das Protein O2- anstelle von H2O2 produzierte, ohne dass die intrazelluläre Lokalisation verändert wurde. Zwei konservierte Cysteine innerhalb der deletierten Bereiche scheinen bei diesem Prozess eine Rolle zu spielen, vermitteln den Effekt aber nicht alleine, da nach Mutation dieser Cysteine die Umkehr der ROS-Produktion nicht ganz so stark war wie bei der Deletion der kompletten Bereiche. In Nox1 sind die Cysteine wahrscheinlich in die Aktivierung des Proteins involviert, da deren Mutation die Aktivität von Nox1 reduzierte. Der N-Terminus und die dritte extrazytoplasmatische Schleife von Nox4 sind somit an einem Prozess beteiligt, der die H2O2-Produktion von Nox4 vermittelt. Die entsprechenden Abschnitte in Nox1 scheinen andere Funktionen zu erfüllen. Das Signalpeptid von Nox1 ist für die korrekte intrazelluläre Lokalisation in der Plasmamembran verantwortlich; die dritte extrazytoplasmatische Schleife ist wahrscheinlich an der Aktivierung des Proteins beteiligt. In dieser Arbeit konnten also strukturelle Unterschiede zwischen Nox1 und Nox4 in verschiedenen Bereichen der Proteine identifiziert werden, die für die unterschiedliche intrazelluläre Lokalisation und die Art der produzierten ROS verantwortlich sind.
Neuropathische Schmerzen werden durch Läsionen im zentralen oder peripheren Nervensystem ausgelöst. Sie treten z.B. bei der diabetischen Polyneuropathie oder nach einem Bandscheibenvorfall auf und sind durch die Symptome Allodynie und Hyperalgesie gekennzeichnet. Bislang lassen sich neuropathische Schmerzen nur unzureichend behandeln, weshalb ein großer Bedarf an neuen, besser wirksamen und verträglicheren Arzneimitteln besteht. Die Voraussetzung für die Entwicklung neuer Arzneimittel ist die Erforschung der molekularen Mechanismen von Neuropathien. Im ersten Projekt dieser Dissertation sollten deshalb mit Hilfe der differenziellen Gelelektrophorese Proteine identifiziert werden, die nach Induktion neuropathischer Schmerzen im Lumbalmark von Ratten reguliert sind. Durch vergleichende Proteomanalyse konnte gezeigt werden, dass 55 Proteine 7 Tage nach Induktion neuropathischer Schmerzen im Lumbalmark der Ratte differenziell exprimiert werden. 54 Proteine zeigten ein verminderte, und ein Protein eine gesteigerte Expression im Modell der „spared nerve injury“ (SNI). Durch massenspektrometrische Analyse der Proteinspots konnten 38 Proteine eindeutig identifiziert werden. Einige dieser Proteine sind möglicherweise an zentralen Sensibilisierungsvorgängen beteiligt und könnten somit als neue Zielmoleküle für die Schmerztherapie fungieren. Die meisten der deregulierten Proteine stehen im Zusammenhang mit dem Energiestoffwechsel und dem zellulären Metabolismus. Aufgrund der verminderten Expression dieser Proteine deutet vieles darauf hin, dass es 7 Tage nach SNI auf spinaler Ebene zu degenerativen Prozessen wie z.B. dem Abbau der Myelinscheide kommt. Mit einer Reihe ausgewählter Proteine wurden zusätzlich Genexpressionsanalysen mittels RT-PCR durchgeführt. Die Ergebnisse zeigten, dass zahlreiche Proteine, die im DIGE Experiment auf Proteinebene eine reduzierte Expression aufwiesen, schon auf Transkriptionsebene reguliert sind. Einige Gene waren in ihrer mRNA-Expression jedoch nicht verändert, was ein Hinweis darauf ist, dass die beobachteten Regulationen auf translationale bzw. posttranslationale Modifikationen zurückzuführen sind. Eines der Proteine, welches aufgrund der peripheren Nervenläsion eine deutlich geringere spinale Expression zeigte, wurde anhand des Massenspektrums als Latexin identifiziert. Latexin ist ein Inhibitor der Carboxypeptidase A Aktivität und spielt bei der Schmerzweiterleitung und Schmerzverarbeitung eine Rolle. Darüber hinaus wurde der Abbau von Latexin in der Literatur mit der neurodegenerativen Erkrankung Morbus Alzheimer in Verbindung gebracht. In der vorliegenden Arbeit wurde die periphere und zentrale Expression von Latexin mit Hilfe verschiedener molekularbiologischer Methoden untersucht. Dabei zeigte sich, dass Latexin unter neuropathischen Bedingungen im Lumbalmark sowohl auf Transkriptionsebene, als auch auf Translationsebene signifikant reduziert vorliegt. Zusätzlich wurde mit Hilfe eines Aktivitätsassays nachgewiesen, dass eine verminderte Latexinexpression zu einer signifikanten Erhöhung der Carboxypeptidase-Aktivität führt. Im peripheren Nervensystem konnte im Gegensatz zum zentralen Nervensystem keine Modulation von Latexin beobachtet werden. Um die funktionelle Relevanz von Latexin bei neuropathischen Schmerzen zu charakterisieren, wurden am Ende der Dissertation rekombinante Adenoviren hergestellt, mit deren Hilfe eine spinale Latexinüberexpression erreicht werden sollte. Die dazugehörigen Tierversuche wurden erst nach Abschluss dieser Arbeit durchgeführt. Im Vorfeld dieser Arbeit wurden mit NF-kappaB p50 Knockout-Mäusen mehrere Schmerztests durchgeführt, die gezeigt haben, dass p50 Knockout-Mäuse im Vergleich zu Wildtyp-Tieren signifikant weniger akute und chronische Entzündungsschmerzen hatten. In einem zweiten Projekt sollte deshalb untersucht werden, welche molekularen Mechanismen dem verminderten Schmerzverhalten der Knockout Tiere zugrunde liegen. Anhand von RT-PCR Experimenten konnte gezeigt werden, dass das NF-kappaB abhängige, proinflammatorische Gen COX-2 8 Stunden nach Zymosaninjektion in eine Hinterpfote bei den Wildtyp-Tieren im Lumbalmark signifikant erhöht war, während bei den Knockout-Mäusen keine signifikante Veränderung zu beobachten war. Weiterhin wurden Änderungen der Proteinexpression und der Genexpression im Lumbalmark von NF-kappaB Knockout- und Wildtyp-Tieren mittels 2D-Gelelektrophorese bzw. RNA-Microarray untersucht. Beim Vergleich der Proteinexpressionsmuster zeigten sich bei 5 Proteinen Regulationen infolge der p50 Deletion, während mit Hilfe des RNA-Microarrays 7 differenziell exprimierte Gene identifiziert werden konnten, die auf das Fehlen der p50 Untereinheit zurückzuführen sind. Zusammenfassend lässt sich aus diesen Befunden schlussfolgern, dass die p50 Untereinheit des Transkriptionsfaktors NF-kappaB als Zielmolekül für die Therapie akuter und chronischer Entzündungsschmerzen geeignet sein könnte. Es wird vermutet, dass die p50 Untereinheit von NF-kappaB bei chronischen Entzündungsschmerzen auf zentraler Ebene an einer Aktivierung proinflammatorischer Stimuli beteiligt ist, während p50 bei akuten Schmerzen wahrscheinlich eine konstitutive Rolle spielt.
Mitochondria are dynamic organelles indispensible for viability of eukaryotic cells. Diffusion of proteins in mitochondrial membranes is a prerequisite for the correct functionality of the organelles. However, its study is made complicated due to the nontrivial geometry, small size and positional instability of the organelle, restricting the usability of regular experimental methods and theoretical understanding of acquired data. Therefore, here the molecular transport along the main mitochondrial axis was investigated using highly accurate computational methods combining them with traditional experimental approaches. Using recently reported electron microscopic tomography data concerning the constitution of mitochondria [Fre02], a lattice model of the inner mitochondrial membrane (IM) reproducing its structure in great details was built up. With Monte Carlo (MC) simulations of particle dynamics on this model, it was found that the membrane geometry induces nonlinear effects in the motion of molecules along the mitochondrial axis, which in turn lead to a transient violation of the 2nd Fick?s equation. We show that mere curvature of the IM resulting from the presence of cristae is sufficient for the emergence of transient anomalous diffusion (TAD) in the membrane. The MC calculations have enabled an accurate estimation of regularities in the extent of deviations from the normal regime, therefore allowing us to propose non-homogenous power law as a suitable generalization of the current approach to the analysis of experimental data for the transient dynamics. The general cause of TAD resulting from the membrane curvature alone, without any involvement of specific inter-particle interactions prompted us to predict the similar dynamical effect also for other curved cellular membranes, be it diffusion in endoplasmic reticulum or in plasma membrane of cells possessing dense microvilli. The data indicate that the geometry-induced anomalous diffusion should be easily detectable with current experimental methods, but only in the restricted range of time scales corresponding to high temporal resolution. Until now, experimental measurements of molecular diffusion in biological membranes indiscriminately assumed either pure normal or pure anomalous diffusion schemes for the analysis of data acquired in very wide range of temporal resolutions, which often lead to ambiguities in the interpretation of diffusion parameters. The MC calculations have clearly illustrated the necessity for a more subtle treatment of experimental conditions: the assumption of pure Gaussian diffusion model is justified only if the applied temporal resolution is sufficiently low (as is often the case when using scanning techniques exemplified further); otherwise, the transient regime should be tested for by means of the non-homogenous power function. In the second part of the study the Fluorescence Recovery after Photobleaching (FRAP) with the laser scanning microscope is introduced as a method of choice for studying protein mobility within mitochondrial membranes. The conventional FRAP methodology [Axe76] was extended to enable its application for the determination of confined diffusion with conventional laser scanning microscopes which allowed us to communicate for the first time the direct measurement of protein diffusion in mitochondrial membranes of living cells. This is achieved through adaptation of FRAP data analysis to account for the spatial dimensions of the organelle and the spatiotemporal pattern of light pulses induced by the microscope. The experimental circumstances existing during the particular measurement session are computationally recreated and this way the best suited values of diffusion parameters are found. The method is validated experimentally for four FP-tagged mitochondrial membrane proteins: the IM OxPhos complexes F1F0 ATPase and cytochrome c oxidase and for Tom7 and hFis1 - components of the mitochondrial protein import and fission machineries respectively localized in the outer membrane. We find that for all proteins simple normal diffusion is not a sufficient description. In the inner membrane, diffusion coefficient of F1F0 ATPase expressed in HeLa cell line is found to be 0.2 ?m2/s, with more than 1/3 of the protein molecules being immobilized, while cytochrome c oxidase (in CEF primary cells) demonstrated a similar diffusivity pattern (0.4 ?m2/s, 30% immobile). In the outer membrane, the D (0.7 ?m2/s) and immobile fraction (7-8%) of GFP-Tom7 and GFP-hFis1 (both in HeLa cells) are identical, which designates a substantial difference in comparison to the IM protein mobility. Diffusion coefficients of mitochondrial membrane proteins studied here lay in the intermediate region between those measured in artificial bilayers and in plasma membranes. Protein crowding and intermolecular interactions will be among the major causes responsible for the detected slowdown of diffusion.
Channelrhodopsin-2, or ChR2, is a light-gated inward rectifying cation channel. Ever since its first characterisation (Nagel et al., 2003), it has been used extensively in the light-activated control of neural cells in culture as well as in living animals like mice, Caenorhabditis elegans and Drosophila melanagaster. Despite its broad application in the field of neuroscience, little is known about the properties of this ion channel. The aim of this thesis is to elucidate the single channel conductance under different conditions using stationary noise analysis on whole cell recordings of a HEK293 cell line that stably expresses the truncated ChR2 (amino acids 1-315), which behaves identically to the full length protein (Nagel et al., 2003). Stationary noise analysis is based on the fact that the ion channel noise due their opening and closing has a characteristic form of a plateau at low frequency points and a following decrease of power with 1/f² in difference power spectra, which are composed of the difference of fast Fourier transformed (FFT) stationary whole-cell recordings with and without illumination. From the parameters yielded by an approximation of the power spectra with a Lorentzian function the single channel conductance can be estimated. The single channel conductance of ChR2 was determined at -60 mV applied for different cations, yielding values of 91 ± 25 fS (Guanidine+), 42 ± 7 fS (Na+), 61 ± 18 fS (Li+) and 37 ± 14 fS (Methylammonium+). With 200 mM Guanidine+ outside of the cells and measurements between 0 mV and -60 mV applied, it could be shown that the inward rectification is still present on the scale of the single channel. Noise Analysis with concentrations between 40 and 200 mM Guanidine+ showed a saturation of the single channel conductance with high Guanidine+ concentrations with a maximal conduction of 129 ± 9 fS (Michaelis Menten approximation: Km = 82 ± 14 mM). Activation Energies of the rate constants k (2πfc, with fc = corner frequency of the Lorentzian function) and koff (1/τoff, with τoff = closing time of the channel at -60 mV) were determined to be 75 ± 23 kJ/mol and 64 ± 11 kJ/mol, respectively, which are similar to the value determined for the Channelrhodopsin-1 closing times (~60 kJ/mol; Nagel et al., 2002). The activation energy of the ChR2 single channel conductance was determined to be 21.2 ± 20.8 kJ/mol, which also is similar to the activation energy of the ChR1 current amplitude (20 kJ/mol; Nagel et al., 2002). The amount of active ChR2 channels in the membrane (160,000 or 226 ChR2/μm²) as well as the single channel current (-7.5 ± 0.6 fA) could be determined by variation of the light intensity (0.05 mW mm-2 to 5.3 mW mm-2). In the course of this thesis, the single channel parameters of the ChR2 mutant H134R were also determined. H134R had been previously published as a “gainof- function” mutant (Nagel et al., 2005a). The increased macroscopic current amplitude of H134R could be explained by an increased lifetime of the channel in comparison to the wildtype ChR2. Within the margin of error both single channel conductances in the presence of 200 mM Guanidine+ of the wildtype (91.1 ± 24.9 fS) and the H134R (89.4 ± 30.7 fS) are the same. In the presence of 200 mM Lithium+ values of 60.6 ± 17.8 fS for the wildtype ChR2 and 50.8 ± 9.6 fS for the H134R mutant were determined. This thesis marks the first in depth analysis of the single channel conductance of ChR2. Using stationary noise analysis the single channel conductance of Channelrhodopsin-1 as well as interesting Channelrhodopsin-2 mutants can also be analysed in the future.
Im Rahmen der vorliegenden Arbeit wurde die Funktion des in der Membran des Endoplasmatischen Retikulum lokalisierten Proteins Gsf2 der Hefe Saccharomyces cerevisiae näher charakterisiert. Gsf2 ist ein 46 kDa großes ER-Transmembran-Protein mit zwei membrandurchspannenden Domänen, wobei C- und N-Terminus cytosolisch orientiert sind. Zudem besitzt Gsf2 C-terminal ein klassisches Dilysin-Motiv. Dies deutet daraufhin, dass Gsf2 über den retrograden Transportweg mittels COPI-Vesikel recycelt wird. Dies impliziert auch einen Transport von Gsf2 über den anterograden Transportweg [COPII]. Bisherige Befunde deuteten darauf hin, dass Gsf2 in das sekretorische System involviert ist. Eine Deletion des GSF2-Gens resultiert in einer Retention der Hexosetransporter Hxt1, Hxt3 und Gal2 im ER. Für die Identifikation von potentiellen Interaktionspartnern von Gsf2 im sekretorischen System wurden Protein-Protein-Interaktionsstudien mit Hilfe des Split-Ubiquitin-Systems [SUS] durchgeführt. Dabei konnten Interaktionen zwischen den Proteinen Sar1, Sec12, Hxt1 und dem Sec61-Translokations-Komplex mit dem vermeintlichen Verpackungschaperon Gsf2 identifiziert werden. Zusätzlich konnte unter Anwendung einer Pull-Down-Analyse die Interaktion zwischen Gsf2 und Hxt1 biochemisch bestätigt werden. Zur Aufklärung von funktionellen Domänen, wurde ein in Gsf2 identifiziertes potentielles ER-Export-Motiv [RR](F)[RR] durch den Austausch durch fünf Alanine modifiziert. Die Veränderung der Aminosäuresequenz [RR](F)[RR] (354-358 AS) in Gsf2 bewirkte einen partiellen Funktionsverlust bei einer restriktiver Temperatur von 37°C. Des Weiteren deuteten mehrere Anhaltspunkte darauf hin, dass Gsf2 mit Hxt1 in COPII Vesikel verpackt wird. Eine Aufreinigung von konstitutiven COPII-Vesikel aus in vivo erwies sich experimentell als nicht durchführbar. Deshalb wurde ein in vitro Ansatz [COPII vesicle budding assay] ausgewählt. Dafür wurden die für die Vesikelbildung benötigten Komponenten aufgereinigt und mit ER-Donormembranen inkubiert. Mittels Western-Blot-Analyse konnte Gsf2 in COPII Vesikeln nachgewiesen werden. Das Vorhandensein eines klassischen Rücktransport-Motivs (KKSN) am C-Terminus von Gsf2 weist zudem daraufhin, dass das Protein über den retrograden Transportweg [COPI] zurück zum ER transportiert wird. Durch die Blockade des retrograden Transportweges mit anschließender Lokalisationsuntersuchungen mittels Saccharosedichtegradienten-Zentrifugation und Fluoreszenzmikroskopie konnte eine Fehlverteilung von Gsf2 an der Plasmamembran festgestellt werden. Somit wird Gsf2 möglicherweise über den retrograden Transportweg recycelt. Postuliert wird ein Modell bei dem Gsf2 für die Aufkonzentration spezifischer Cargomoleküle [Hxt1] an ER-Exit-Sites zuständig ist und deren Verpackung in COPII Vesikel gewährleistet. Anschließend wird es über den retrograden Transportweg zurück zu ER transportiert.
A recently completed field survey of springs throughout the Great Basin yielded collections of hydrobiid snails from more than 500 sites, and revealed a wealth of undescribed diversity of these small gastropods. In this, the first or a two-part taxonomic series treating this material, 58 new species of Pyrgulopsis Call & Pilsbry, 1886, are described; and new records are provided for 10 previously described members of this genus. Assignment of these novelties to Pyrgulopsis is done with the acknowledgement that this large genus, as currently constituted, is probably not monophyletic, but a more refined classiffcation of these snails reflecting evolutionary relationships must await preparation of a phylogenetic analysis, which is beyond the scope of this work. Pyrgulopsis occur in a variety of spring-fed water bodies in the Great Basin, including brackish and/or thermal habitats. Although a few species are widespread in the region, local endemism is prevalent lind 22 of the new species are known only from single localities. Several areas contain concentrations of locally endemic snails which may represent species flocks, notably Duckwater Valley (seven species) and southern Steptoe Valley (five species). This fauna is hugely distributed in an allopatric fashion, although a few springs harbor two or three species. Most of the springs inhabited by hydlrobiids in the region are small, fishless, and have been ignored by state and federal land management agencies. However, many of these sites are degraded by livestock grazing, water withdrawal, anti other activities and will require protection in order to conserve snails and other native aquatic biota. Two of the novellies described herein have become extinct during the past two decades.
This second and final part of a taxonomic treatment of hydrobiid snails of the Great Basin region in the western United States (based principally on material collected during a recently completed field survey) focuses on fauna other than the genus Pyrgulopsis. A new genus of small amnieoline snails, Colligyrlls, is proposed for Hydroia greggi Pilsbry, 1935, together with a new species from the Harney Lake basin of Oregon. This group is strongly differentiated from other amnicolines by a unique female genitalic groundplan. New records are provided for three species of Fluminicola, and two new congeners are described from the northwest Great Basin, both of which had previously been confused with F. turbiniformis (Tryon, 1865). A new genus of cochliopine snails, Eremopyrgus, is erected for a new species from Steptoe Valley, Nevada. Eremopyrgus is distinguished from other cochliopines by unique aspects of its glandular penial lobes and other genitalic features. New records are provided for two species of Tyronia, and a new congener is described from thermal springs in central Nevada. Several new records of Pristincola hemphilli (Pilsbry, 1890) from the extreme northwest Great Basin are provided
Das große therapeutische Potential der RNA Interferenz wird dadurch deutlich, dass sich wenige Jahre nach der Entdeckung die ersten potentiellen RNAi Medikamente in den klinischen Teststudien befinden. Jedoch müssen bis zur therapeutischen Anwendung im großen Maße noch einige Hindernisse überwunden werden. Um eine optimale Wirkung der siRNAs gewährleisten zu können müssen folgende Punkte beachtet werden: - Erhöhte Nuclease-Resistenz - Effiziente zelluläre Aufnahme - Keine Off-Target Effekte - in vivo geringe Toxizität Das richtige Design der siRNAs ist somit von enormer Bedeutung. Hierfür bedient man sich verschiedener Modifikationsmöglichkeiten wie z.B. Basenmodifikationen, Modifikationen an der 2´-O-Position sowie am Phosphatrückrat. Im Rahmen der vorliegenden Doktorarbeit ist es gelungen verschiedene Syntheserouten zur Darstellung von Nucleosiden mit kationischen sowie neutralen 2´-O-Modifikationsmotiven zu erarbeiten und zu optimieren. ...
1. Der Abgleich der Genprodukte von PA0119, PA0120 und PA0121 ergab, dass PA0119 68%ige Ähnlichkeit mit dem DctA-Transporter von S. meliloti besitzt. Das Genprodukt PA0120 zeigt eine 46%ige Ähnlichkeit mit dem Transkriptionsregulator LctR von E. coli und trägt konservierte Domänen der GntR- sowie der FCDSuperfamilie. Das Genprodukt PA0121 weist eine 44%ige Ähnlichkeit mit einem hypothetischen Transkriptionsregulator von Streptomyces ambofaciens auf. 2. Es konnte gezeigt werden, dass die drei offenen Leserahmen (ORFs) PA0119, PA0120 und PA0121 von P. aeruginosa in mRNA transkribiert werden und somit Gene sind. In den späten CF-Isolaten M25 und M26 ist deren Transkriptmenge im Vergleich zum frühen CF-Isolat M1 sowie zum Referenzstamm P. aeruginosa PAO1 um das 14fache erhöht. 3. Mit Hilfe von RT-PCR Analysen konnte gezeigt werden, dass die drei Gene PA0119, PA0120 und PA0121 eine Transkriptionseinheit bilden und somit in einem Operon P0119-PA0121 organisiert sind. Die angrenzenden ORFs PA0118 und PA0122 konnten dieser Transkriptionseinheit nicht zugeordnet werden. 4. Analysen der Promotor-Reportergenfusion führten zu dem Schluss, dass die Strukturgene PA0119, PA0120 und PA0121 des Operons von einer stromaufwärts von PA0119 lokalisierten Promotorsequenz transkribiert werden. Der Promotor liegt ca. 237 bp vor dem Start von PA0119. Darüber hinaus konnte eine FadR-ähnliche Bindestelle in der Promotorregion P119-121 abgeleitet werden. 5. Der Promotor P119-121 zeigt in mit Dicarboxylaten supplementiertem Minimalmedium M9 eine 2-3fach erhöhte Promotoraktivität gegenüber dem mit Glukose supplementierten Minimalmedium M9. In Gegenwart von Dicarboxylaten erfährt der Promotor P119-121 somit eine Induktion bzw. durch Glukose eine Repression. 6. Der Promotor P119-121 wird wachstumsphasenabhängig reguliert. Die Abhängigkeit der Promotoraktivität von RhlI, RhlR und RpoS indiziert eine Quorum Sensingsowie RpoS-abhängige Regulation des Promotors P119-121. 7. Eine mit PA0119 durchgeführte heterologe Komplementation einer DctA-Mutante von S. meliloti führte zu einer partiellen Wiederherstellung der Fähigkeit der DctAMutante, auf den Dicarboxylaten Succinat und Fumarat wachsen zu können. Dieser Befund indiziert zusammen mit den vier konservierten Motiven, die bisher in charakterisierten Dicarboxylat-Transportern gefunden wurden, dass PA0119 potentiell einen Dicarbonsäure-Transporter in P. aeruginosa darstellt. 8. Mutantenstudien mit den generierten Mutanten ΔPA0119Ω, PA0120Ω und ΔPA0121Ω zeigten im Vergleich zum Wildtyp allerdings kein abweichendes Kulturwachstum; weder in den nährstoffreichen Medien ASM und LB noch im Minimalmedium M9, das jeweils mit 40 mM Succinat bzw. Glutamat supplemiert wurde. Dies wurde durch MicroArrayTM-Analysen mit den Biolog-Platten PM1 und PM2 bestätigt. Daraus wird deutlich, dass keine der hier angebotenen C-Quellen (siehe Anhang) ausschließlich von PA0119 transportiert wird. Auch die Verwertung dieser C-Quellen wird nicht durch die in den Mutanten PA0120::Ω und ΔPA0121::Ω ausgeschalteten Genprodukte beeinflusst. 9. In der Biolog-Platte PM10a schien ΔPA0119::Ω gegenüber PAO1 in den Kavitäten C1 (pH5.5, Methionin) und C10 (pH 5.5, Ornithin) eine leicht abweichende Stoffwechselaktivität zu besitzen. Dies indiziert eine mögliche pH-Abhängigkeit des potentiellen PA0119-Transporters. 10. Im Minimalmedium M9, supplemiert mit Glutamat, zeigt die Mutante ΔPA0119::Ω unter unter erhöhten osmotischen Bedingungen gegenüber dem Wildtyp PAO1 eine reduzierte Wachstumsrate. Dies indiziert, dass PA0119 unter erhöhten osmotischen Bedingungen am Transport von Dicarboxylaten, wie Glutamat, in die Zelle beteiligt sein könnte. 11. Biofilmbildung stellt die bevorzugte Wachstumsform von P. aeruginosa in der CF-Lunge dar. Das Genprodukt PA0120 zeigt zudem eine Ähnlichkeit mit dem Regulator LctR, und es wurde gezeigt, dass eine LctR-Mutante von E. coli in ihrer Fähigkeit zur Biofilmbildung beeinträchtigt ist. Die Untersuchung der ΔPA0119::Ω- und der PA0120::Ω-Mutanten zeigt im Vergleich zum Wildtyp im LB-Medium eine Beeinträchtigung in der Anheftung an Mikrotiterplatten. Dies indiziert, dass sowohl der potentielle Transporter PA0119 als auch der potentielle Transkriptionsregulator PA0120 an der Biofilmbildung von P. aeruginosa beteiligt sind. 12. Die Messung der Promotoraktivität in der ΔPA0119::Ω-Mutante zeigt eine Repression des Promotors an. In der PA0120::Ω-Mutante dagegen findet sich eine zweifache Induktion des Promotors vor PA0119-PA0121. Dies Ergebnis wird durch die Ergebnisse der Real-Time-PCR-Analysen untermauert, da in der PA0120::Ω-Mutante signifikant erhöhte Transkriptmengen des PA0119-Gens detektiert wurden. Bei der ΔPA0121::Ω-Mutante liegt die Promotor-Aktivität im Bereich von PAO1. Diese Ergebnisse lassen den Schluss zu, dass das Genprodukt von PA0119 auf den Promotor P119-121 aktivierend wirkt, während das PA0120-Genprodukt die P119-121- Promotoraktivität negativ reguliert. 13. Zur Identifizierung weiterer PA0120-regulierter Gene wurde eine Transkriptomanalyse der PA0120::Ω-Mutante durchgeführt. Diese ergab, dass neben PA0119 vier weitere Gene, und zwar PA0656, PA0810, PA1852 und PA5261, signifikant hoch reguliert wurden. Vermutlich sind diese Genprodukte zusammen mit dem Regulator PA0120 Teile eines regulatorischen Netzwerks zur Anpassung von P. aeruginosa an die CF-Lunge.

The present catalogue is an attempt to bring together the genera and species of Trematoda currently known to parasitize Chiroptera Blumenbach, 1774, throughout their world distribution, as published in various journals. Since many of these are difficult to obtain for consultation, it is hoped that this catalogue may be of some utility in facilitating the work of helminthologists working with trematodes from bats.
Die 4. Fassung der Roten Liste der Brutvögel Deutschlands wurde durch das "Nationale Gremium Rote Liste Vögel" erarbeitet, in dem die wissenschaftlichen Institutionen der Ornithologie und Avifaunistik in Deutschland vertreten sind. Die Rote Liste ersetzt die 3. Fassung aus dem Jahr 2002 (BAUER et al. 2002); sie wurde erstmalig nach dem für alle Tier- und Pflanzenartengruppen sowie den Pilzen in Deutschland entwickelten Kriterienschema (5. LUDWIG et al. 2007) erarbeitet. Somit wird ein direkter Vergleich der Gefährdungssituation zwischen diesen Gruppen ermöglicht. Bestandsgröße, kurzfristiger (25 Jahre) und langfristiger (50-150 Jahre) Bestandstrend sind die wichtigsten Parameter zur Gefahrdungseinstufung der einzelnen Arten. Zusätzlich wurde jeweils die Wirksamkeit von Risikofaktoren artspezifisch identifiziert und berücksichtigt. Alle Einstufungen werden transparent vorgenommen und in der Anhangsliste publiziert. Der Dachverband Deutscher Avifaunisten (DDA) hat zur Erstellung der Datengrundlagen mit Stand 2005 für die Gefahrdungseinstufung ein neues Abfrageschema entwickelt, in dem die relevanten Informationen aus den nationalen Vogelmonitoring-Programmen aufgearbeitet und als Hintergrunddaten für die Einschätzungen von Bestandstrend und -größe auf Landesebene bereitgestellt wurden. Dadurch gewinnen die Einstufungen an Verlässlichkeit und Nachvollziehbarkeit. Der langfristige Trend wurde vom "Nationalen Gremium Rote Liste Vogel" ermittelt. Vor der Einstufung der Brutvogelarten wurde je Art eine Statuszuordnung vorgenommen, von denen nur die regelmäßig brütenden einheimischen Arten den weiteren Weg der Rote Liste-Erstellung durchlaufen. In der Roten Liste 2007 werden insgesamt 260 regelmäßige einheimische Brutvogelarten in Deutschland berücksichtigt, 25 weitere Arten brüteten nur unregelmäßig ('vermehrungsgäste': Status II), zudem wurden 29 Neozoen-Arten ermittelt (Status III), von denen 20 regelmäßig brüten. Dies ergibt zusammen 314 Arten, die höchste Zahl an Brutvogelarten, die je für eine Rote Liste zu Grunde gelegt wurde. Insgesamt befinden sich 110 regelmäßige Brutvogelarten in den Kategorien der Roten Liste 2007 (0 = ausgestorben, 1 = vom Aussterben bedroht, 2 :: stark gefährdet, 3 :: gefährdet und R = extrem selten), das entspricht 42,3 % der Arten, was einer minimal geringeren Gefahrdungsquote gegenüber der Vorgängerliste entspricht. Erfreulich ist, dass mit dem Bruchwasserläufer und dem Steinrötel zwei ehemals in Deutschland ausgestorbene Arten zwischen 2000 und 2005 wieder regelmäßig gebrütet haben. Dem entgegen ist mit der Blauracke eine weitere Art ausgestorben. Schwarzstorch, Wanderfalke, Seeadler und Uhu sind hier erstmals seit der ersten deutschen Roten Liste 1971 nicht mehr aufgeführt - ein Erfolg jahrzehntelanger direkter Schutzmaßnahmen der ehrenamtlichen und amtlichen Vogelschützer und gleichzeitig ein Beweis, dass sich Vogelschutz bei stark gefährdeten Arten lohnen kann. Andererseits sind mit Schreiadler, Zwergseeschwalbe oder Großem Brachvogel Alien in die höchste Gefährdungskategorie eingestuft worden, die zwar auch im Fokus des Vogelschutzes standen und stehen, bei denen aber bislang Maßnahmen nicht ausreichend erfolgreich umgesetzt werden konnten. Gerade die Kategorie "vom Aussterben bedroht" umfasst mit nunmehr 30 Arten den höchsten Wert seit Erscheinen der gesamtdeutschen Roten Liste. Der Analyse der aktuellen deutschen Brutvogelfauna zufolge sind die Boden brütenden Vogelarten, Großinsektenfresser und Langstreckenzieher am stärksten von Gefährdungen betroffen. Vogelgruppen mit einem hohen Anteil gefährdeter Arten sind demzufolge Hühnervögel, Rallen, Limikolen und Würger, während Eulen und Schnäpperverwandte derzeit vergleichsweise wenig gefährdet sind. Zudem erfolgt deutschlandweit ein weiteres Ausdünnen der typischen Vögel in der Normallandschaft, was sich vor allem in den Trendanalysen manifestiert, aber in der Roten Liste noch nicht sehr stark zum Ausdruck kommt. Die Nutzungsintensivierungen von Land- und Forstwirtschaft in jüngster Zeit geben hier großen Anlass zur Sorge in diesen Großlebensräumen. Diese Rote Liste stellt erneut ein kritisches Zeugnis über den Zustand der deutschen Vogelwelt aus. Aufgrund der in jüngster Zeit stark ausgeweiteten Monitoringprogramme wird es in Deutschland zukünftig noch besser möglich sein, die Gefahrdung aufzuzeigen. Um den dauerhaften Rückgang der Vogelbestände zu stoppen oder wenigstens zu verlangsamen, müssen die wirksamen Gefahrdungsfaktoren reduziert und minimiert werden. Dem gezielten Vogelartenschutz stellen sich dabei folgende vordringliche Aufgaben: Erhaltung der offenen Kulturlandschaft, Erhaltung strukturreicher Walder, Erhaltung nährstoffarmer Lebensräume, Sicherung der Schutzgebiete - insbesondere Natura 2000, Stärkung der internationalen Zusammenarbeit im Vogelschutz, Reduktion der Populationsverluste durch Unfälle und menschliche Verfolgung sowie Förderung des vogelkundlichen Nachwuchses.
The genus Maculinea van Eecke, 1915 (Lepidoptera: Lycaenidae) from the East Palaearctic Region

We revise the classification of taxa belonging to the genus Maculinea from the East Palaearctic Region. In this region, in addition to the well-known three species: M. arion (Linnaeus, 1758), M. ationides (Staudinger, 1887) and M. teleius (Bergstriisser, [1779] 1778-1780), two additional species occur: M. alcon ([Denis & Schiffermiiller], 1775) (upper and middle Amur River, Primor'e, China Northeast/Manchuria and North Korea) and M. kurentzovi sp. nov. (upper and middle Amur River, Primor'e, China Northeast and North Korea). Lycaena kondakovi (Kurentzov, 1970) described from Primor'e is a composite species: the lectotype if' designated here represents an East-Asian subspecies of M. alcon, but its single paralectotype is a female to be assigned to M. kurentzovi sp. nov. Only limited numbers of specimens have been known with M. alcon kondakovi from lowlands of "Far-Eastern" Russia and China Northeast, but in North Korea we found a conspicuous allied taxon arirang nov. (female unknown), which we treat here as a highland subspecies of M. alcon but which may actually represent a good species. Of kurentzovi, we have found a series of specimens which have so far been mostly confused with M. teleius in various collections. We treat Glaucopsyche xiaheana Murayama, 1991 from western Gansu as a subspecies of M. arion along with other subspecies from the central and western parts of China: M. adon philidor (Fruhstorfer, 1915) from the east end of the Qilian Range as well as Mongolia, the type locality, and M. arion inferna nom. nov., a replacement name for Lycaena talsienluica (OberthUr, 1910) (praeoccupied) from Tibet, Sichuan and Qinghai. Because of the similarity of male genitalia and existence of intermediate forms, we regard M. sinalcon Murayama, 1992 described from Qinghai as a subspecies of M. teleius despite a few significant characteristics of the holotype. East continental Asia may be regarded as the headquarter of the genus Maculinea.
Glyptostrobus Endlicher is well represented in early Early Cretaceous to Pleistocene deposits in the middle to high latitudes of North America and Eurasia. Although the taxonomy and nomenclature of the genus is complicated, the fossil record indicates Glyptostrobus was represented by a small number of species. The genus first appears in Aptian age deposits from western Canada and Greenland, and achieved a wide distribution early in its evolutionary history. Exchange of Glyptostrobus between Asia and North America occurred across the Spitsbergen and Beringian corridors, which were functional about 110 and 100 million years ago, respectively The Late Cretaceous fossil record of Glyptostrobus shows that the genus had spread into Russia, China and the shores of the Turgai Strait. By the early Tertiary, Glyptostrobus was a prominent constituent of the polar broad-leaved deciduous forests. Paleocene age deposits across western Canada and the United States indicate the genus was present in great abundance in the lowland warm temperate and subtropical forests east of the Rocky Mountains. The broad distribution in North America and Russia during the Paleocene and Eocene indicates that Glyptostrobus grew and reproduced under a diverse range of climatic and environmental conditions, including the cold and unique lighting conditions of the polar latitudes. The presence of Glyptostrobus in Europe indicates the North Atlantic land bridges that extended between North America and Eurasia (Fennoscandia) and Europe during the early Tertiary were used. In Europe, extensive Glyptostrobus dominated swan1ps occupied the Central European Depression during the late Tertiary. Increasing global aridity and cooling, as well as landscape stabilization together with increasing competition for resources and habitat by representatives of the Pinaceae, seem to have forced the genus out of North America, Europe and most of Asia during the Miocene and Pliocene. In Japan, Glyptostrobus persisted until the early Pleistocene. After the early Pleistocene extinction in Japan, Glyptostrobus reappeared in southeastern China. Details of the taxonomic and biogeographic history of Glyptostrobus are examined.
The regular or obligate aphytophagy of certain lycaenid butterflies (Lepidoptera) is discussed within the framework of the most recent general classification of the family. A summary survey of all Lycaenidae known to be aphytophagous is presented, together with a brief account of cannibalism and other opportunistic aphytophagy exhibited by normally phytophagous butterflies. The range of food sources (plants, animals, excretions and regurgitations) exploited by lycaenids is reviewed with emphasis falling on the ecology of myrmecophilous early stages and the significance of their ant-related adaptations. Adult feeding and oviposition behaviour reveal further associations with ants. Specificity oflycaenid/ant relationships and the possible biological effects ofaphytophagy on the Lycaenidae are discussed. Finally, speculations concerning the evolution of aphytophagy by these bulterflies are critically presented.
Epilabidocera amphitrites is one of the most common copepods in the deep waters adjacent to Friday Harbor and shows characteristic swarming behavior in the surface film of the water from later spring through early summer. That the swarms are composed mainly, up to 99 %, of adult males appears to be due to difference in phototaxis to a weak light. This species, at least in copepodid stages, is omnivorous, but seems to prefer an animal diet rather than diatoms. Reproduction takes place continuously from early spring through autumn. The external anatomy of both the female and male has been described in detail. The cuticle forming the arthrodial membrane and the lining of the esophagus, hindgut, and hypostomal and labral troughs appears to be of the same nature throughout, consisting of a single stratum. The cuticle on the general body surface, however, consists of two main strata. The endoskeletal structures consist of two categories, the endoskeleton proper and the endoskeletal tendons. The former involves apodemes and apophyses. Of these the major ones are described in detail. The latter consist of two median tendinous endosternites in the « head », four pairs of ventral intersegmental thoracic tendons, and a pair of dorsal longitudinal tendons in the metasome. The endosternites are well developed, serving as origins for dilators to the atrium oris and esophagus and also for a number of extrinsic muscles to the head appendages. The skeletomusculature may be divided into longitudinal trunk and limb muscles. The paired dorsal and ventral longitudinal trunk muscles in the metasome extend, respectively, from the levels of the cervical groove and the post-maxillulary apodeme to the end of the metasome. The longitudinal trunk muscles in the urosome origate at the anterior end and run most of its length. They are arranged as paired dorsal and ventral groups and a pair of lateral muscles. The extrinsic limb muscles are described in detail. They originate either from the lateral to dorsal exoskeleton or from the endosternites. The digestive tract starts with the atrium oris in the oral cone, followed by the mouth proper, esophagus, midgut, and finally by the hindgut which opens as the anus at the end of the urosome. The oral cone consisting of the three lobed labrum and the paired paragnaths has a longitudinal groove, the oral groove, which is covered ventrally by the spinulose setae of the maxillae and laterally by the gnathobasal endites of the maxillules, these together forming an effective feeding apparatus. The midgut is produced anteriorly into a diverticulum which is higly secretory. In the middle portion of the midgut the epithelial cells are highly vacuolated. As they pass through this vacuolated region the gut contents are cemented into fecal pellets by a mucous secretion and they acquire a peritrophic membrane. There is a strong valve between the midgut and the hindgut. Peristalsis in the midgut is irregular but powerful and primarily in the reverse direction. The circulatory system involves a single heart, enclosed in a large pericardial space, and an anteriorly directed aorta terminating in an anterodorsal aortic sines. The latter communicates through three paires of openings with the sinuses in the head, which are in turn continuous with the perivisceral cavity, from which blood is returned to the pericardium. The heart has the form of a flask with an aortic valve at the tapered anterior end and a posterior ostium. The aortic wall is continued posteriorly over the heart and wraps around the anterior three-fifths as an outer membrane. This outer membrane is extended dorsally at three places to attach the heart to the dorsal exoskeleton; and it is also drawn out ventrally to form the anterior and lateral walls of the pericardium. These walls are continuous with the pericardial floor which seals the pericardia! cavity from the perivisceral cavity. The heart-beat and the blood flow through the system have been discussed. The excretory system consists of a pair of maxillary glands, each comprising a coelomic end-sac, a coelomic secretory tubule and an ectodermal excretory duct. The end-sac communicates with the tubule through a valvular opening. Antennary glands are not gound either in the nauplius stage or in the adult. The male reproductive system consists of a single testis and a single genital duct which is divided into four differentiated sections, the vas deferens, the seminal vesicle, the spermatophore sac, and the ductus ejaculatorius. The vas deferens is a thick-walled glandular tube secreting the various constituents of the spermatophore. The seminal vesicle serves mainly as a reservoir for the various components of a definitive spermatophore, and it is here that these take up their final positions. The spermatophore sac is highly glandular and is mainly responsible for formation of the coupling apparatus of the spermatophore. The spermatophore is not open directly to the outside but is connected with a canal system in the coupling apparatus. When transferred to the female genital segment at copulation, the central secretion of the spermatophore is discharged through the canal system of the coupling apparatus to glue down the spermatophore. A duct through which the spermatozoa can pass from the spermatophore to the spermathecae of the female appears to be formed later by an action of the female, possibly secretion of an enzyme or lysin. The discharge of the contents of the spermatophore is effected by swelling of Q-spermatozoa in the distal region of the spermatophore. The functional spermatozoa are spherical or polygonal and nonmotile. The female reproductive system consists of a single ovary, two oviducts, each with several diverticula, leading to the paired opnenings into the vaginal vacity, a pair of spermathecae and a pair of glands which open into the oviducts. In the mature female the oviducts are wide and sac-like, expanded by growing oocytes. However, the last portion of the oviduct is usually empty of eggs and is highly secretory. The oldest oocytes in the oviducts are usually at the metaphase of the first maturation division. The evidence points to the conclusion that the eggs are laid in this stage, and they are fertilized when they pass through the vaginal cavity. Oogenesis has been studied in detail. There are two periods of yolk formation: the first immediately after the dispersion of the mitochondrial bodies and the second in the last phase of the oocyte growth when the vacuoles in the cytoplasm are gradually replaced by yolk. Two dorsal ocelli, in the copepodid stages, are placed dorsolaterally against the exoskeleton and highly developed, each with a perfectly spherical, cuticular lens, while a single ventral ocellus remains unspecialized through the copepodid stages. Each dorsal ocellus proper is suspended in the head sinus by several connective tissue stands in addition to an aye muscle and consists of a large, syncytial pigmented cup occupied by a cellular sphere which is composed of 9 retinular and 4 crystalline cells. Each of the 9 retinular cells gives off an axon which leaves the ocellar cup at one of three places to proceed to the nauplius eye center in the protocerebrum. The ventral ocellus consists of two multinucleated pigmented cells, a cup-shaped tapetum, 6 retinular cells and about 8 conjunctival cells. Each of the 6 retinular cells sends an axon which loops over the posterior rim of the ocellar cup in common with the others to course to the nauplius eye center in the protocerebrum. The ventral ocellus is innervated by two afferent nerve fibers. There is also found a pair of conspicuous nerve fibers, possibly afferent, associated with the dorsal and ventral ocelli. A pair of accessory retinular groups, each consisting of three retinular cells, is found posterior to the dorsal ocelli. Three efferent aXOl1S from each group form a nerve running to the nauplius eye center in the protocerebrum. A pair of frontal organs, each innervated by a frontal nerve, lies in the anterior end of the head. The frontal nerves can be traced up to a pair of neuropiles immerdiately ventral to the nauplius eye center in the proto cerebrum. A pair of suprafrontal nerves branched off from the frontal nerves is found to innervate a pair of sensory filaments, the suprafrontal sensiIla, at the lower anterior end of the head. The central nervous system, consisting of a well developed brain connected by massive circumesophageaI connectives to the ventral nerve cord, has been described in detail. The ganglion cells are found throughout the nerve cord, and they are arranged into ganglia in the thoracic segments bearing the swimming legs. The stomatogastric nervous system has two pairs of labral and a single gastric ganglia. The medial pair of the labral ganglia forms anteriorly a single ganglion which is connected to the brain by three small nerves. The giant fiber system, consisting of giant motor fibers and giant interneurons, has been studied in detail, and it appears to constitute the effector portion of an escape reflex. The cutaneous glands opening through small pores in the cuticle of the metasome, urosome, and the appendages have been described. Chromatophores, unicellular or syncytial with several nuclei, are scattered deep in the body and are responsible for the metachrosis.
During the last decade, three new acidophilous forests associations were detected in the Mecsek Mts (SW Hungary), and described as acidophilous beech wood (Sorbo torminalis-Fagetum (A. O. Horvat 1963a) Borhidi et Kevey in Kevey 2001), acido-mesophilous oak wood (Luzulo forsteri-Quercetum petraeae (A. O. Horvat 1963a) Borhidi et Kevey 1996) and acido-xerophilous oak shrubland (Genisto pilosae-Quercetum polycarpae (A. O. Horvat 1967) Borhidi et Kevey 1996). In this article two further new associations are described: the acidophilous oakwood of the Mecsek (Viscario-Quercetum polycarpae Kevey, ass. nova) and the acido-mesophilous oakwood of western Hungary (Campanulo rotundifoliae-Quercetum petraeae (Csapody 1964) Kevey, ass. nova). These associations are related to the acidophilous forests of the Balkan Peninsula based on the infrequent presence of sub-Mediterranean species. A detailed comparative study of these new associations with the earlier known ones permitted to develop a reshaped classification of the syntaxonomy of these units, creating four new suballiances: within the frame of Quercion farnetto I. Horvat 1938 the suballiances Luzulo forsteri-Quercenion polycarpae Kevey, suball. nova and the typical Quercenion farnetto Kevey, suball. nova, in the frame of Quercion petraeae Zolyomi et Jakucs 1957 the suballiances Luzulo multiflorae-Quercenion petraeae Kevey, suball. nova and the Quercenion petraeae Kevey, suball. nova.
5-lipoxygenase (5-LO) is the key enzyme in the formation of inflammatory leukotrienes, which are mediators of inflammation and allergy. The 5-LO catalyses the oxidation of arachidonic acid to 5-HPETE and subsequently to LTA4. The leukotrienes are involved in the development and maintenance of inflammatory diseases, like asthma and allergic rhinitis. Additionally, 5-LO is overexpressed in some cancer types, although its relevance is still not fully understood. 5-LO expressing cells are B- lymphocytes and cells of myeloid origin like monocytes, macrophages and granulocytes. The 5-LO promoter lacks a TATA or CCAT box and covers two CpG islands. These are characteristics of a housekeeping gene, but as the 5-LO is not expressed ubiquitiously, the expression of the 5-LO is tightly regulated. Epigenetic mechanisms were known to be involved in the control of the 5-LO expression. The HDAC inhibitor TsA significantly induced the transcriptional activity of the 5-LO promoter in reporter gene assays as well as on 5-LO mRNA transcript level in MM6 cells. The GC-boxes GC4 and GC5 in the proximal 5-LO promoter were identified to be essential for the TsA effect, as deletion of these element led to an attenuated TsA effect in reporter gene assay. Recruitment of the transcription factors Sp1 and Sp3 and the RNA polymerase II to the 5-LO promoter was detectable after TsA treatment in MM6 cells by chromatin immunoprecipitation assays (ChIP), while the acetylation status of histone H4 remained unchanged. Likewise it is known that DNA methylation leads to silencing of 5-LO expression in-vitro and in-vivo. The 5-LO promoter is densely methylated in the cell line U937, but unmethylated in HL-60 cells and - elucidated in this study - also in MM6 cells. Reporter gene assays with in-vitro methylated 5-LO promoter containing plasmids revealed that the frequency of methylated CpGs is directly proportional to reduction of 5-LO promoter activity. Incubation of U937 cells with 5-AdC, an inhibitor of DNA methyltransferases, was able to reactivate 5-LO transcription and to demethylate CpG dinucleotides. In the first part of this study the mechanism of TsA induced promoter activation was further investigated. I elucidated the mechanism of Sp1 and Sp3 recruitment to the 5-LO promoter after TsA treatment. Immnoprecipitation assay was used to detect a transcription factor complex containing Sp1 or Sp3 interacting with HDAC proteins, which might change its composition after TsA treatment. Besides the posttranslational modifications of the transcription factors Sp1 and Sp3 after TsA treatment were investigated, potentially causing an increased interaction of the proteins with the 5-LO promoter. Both aspects and their response in HDAC inhibition have been described. TsA did not affect the composition of the Sp1/HDAC1/HDAC2 complex. Sp3 was not located in a complex with the HDAC enzymes. Acetylation of Sp1 and Sp3 was detectable, but no change occurred after TsA treatment. Since neither release of the transcription factors off a complex, nor alterations in posttranslational modifications of Sp1 and Sp3 are the reason for the increased Sp1 and Sp3 binding to the 5-LO promoter, I elucidated alterations in the chromatin structure. The acetylation status of the histone proteins H3 and H4, as well as the chromatin marks H3K4me3, representing active chromatin, and H3K9me, representative for repressive state, were investigated. Additionally, the time course of the TsA effect was determined on 5-LO mRNA level using real-time PCR. The acetylation status of the histone proteins on the 5-LO core promoter correlated with the basal 5-LO mRNA transcript expression in MM6, HL-60 and U937 cells. The highest 5-LO mRNA level was detectable in MM6 cells, followed by HL-60 cells. The lowest 5-LO mRNA level was detected in 5-LO promoter methylated U937 cells. The order of the basal 5-LO mRNA expression of the three cell lines correlates with the basal acetylation status of histone proteins H3 and H4. In MM6 cells the highest basal levels in acH3 and acH4 were detected, followed by HL-60 and U937 cells. Moreover, the data obtained in U937 cells revealed that the correlation between DNA methylation and histone hypoacetylation is alike on the 5-LO promoter. TsA treatment induced the 5-LO mRNA level in the three cell lines with different intensity: 5-LO mRNA level in MM6 cells was induced 11-fold, in HL-60 cells 6- fold and in U937 cells 4- fold. The histone acetylation and methylation levels on the 5-LO promoter after TsA incubation were investigated. No increase in acH3 and acH4, but in H3K4me3 was detectable in MM6 cells by ChIP assay. HL-60 cells showed an increase in acH3 and acH4 as well as in H3K4me3. H3K9me was only detectable in untreated U937 cells, but disappeared after TsA treatment, while acH3, acH4 and H3K4me3 increased constantly after TsA treatme nt. A strong correlation between the histone modifications and the time course of the mRNA expression was detectable in all three cell lines. The combination of the posttranslational modifications acH3, acH4 and H3K4me3 led to a fast effect in transcriptional activation and the maxima of acH3 and acH4 were usually associated with the maximum in 5-LO mRNA transcript level. An increase in H3K4me3 alone, as detected in MM6 cells, led to continuous increase in the 5-LO mRNA expression with a late maximum. Additionally, we detected a slight overall decrease in 5-LO promoter methylation in U937 cells after TsA treatment. This fact taken together with the observed histone modifications could explain the 4- fold response in 5-LO mRNA level to TsA treatment of the methylated cell line U937. Another aim of the present study was to identify the specific HDAC enzymes involved in the 5-LO promoter regulation. Reporter gene assays and real-time PCR with selective HDAC inhibitors revealed that HDACs of class I are involved in 5-LO promoter regulation, namely HDAC 1, 2 and 3. The influence of each of the enzymes seemed to depend on the cell type, as inhibition of HDACs 2, 3 strongly induced 5-LO promoter activity in reporter gene assay in HeLa cells, whereas in MM6 cells HDACs 1 and 2, 3 seemed to be responsible for the 5-LO promoter regulation, measured as 5-LO mRNA level. The HDACs of class IIa and class III are not involved in the regulation of 5-LO mRNA expression. The second part of this study investigated the influence of MBD proteins on the methylated 5-LO promoter and the 5-LO mRNA expression. ChIP assays revealed MBD1, 2 and MeCP2 protein binding to the proximal 5-LO promoter in U937 cells. MBD1 was detectable on the 5-LO promoter in unmethylated HL-60 cells, while no MBD protein was located on the 5-LO promoter in MM6 cells. To elucidate the functional role of the MBD proteins, stable knocked down of MBD proteins was established in U937 cells. 5-LO mRNA transcript level was determined in the knock down clones by real-time PCR. The 5-LO transcript level was increased in all knock down samples. MBD2 knock down clones showed the highest effect in activating 5-LO with a 3- and 4.4-fold increase in the 5-LO mRNA level, followed by MBD1 (3.5- fold) and MeCP2 (2.5-fold) knock down clones. A combined participation of these three enzymes in the corepression of the methylated 5-LO promoter is indicated. Taken together, the data reveal that epigenetic mechanisms are strongly involved in the regulation of 5-LO transcription and might function as a crucial control mechanism of 5-LO expression.
Westwoodilla Bate. 1862 is revised and 12 species are recognized as distinct, 2 of which, W. tone and W. helle. are described as new. New characters of taxonomic importance are described and illustrated. The many variable characters that have led to taxonomic confusion in the pas! are converted to morphometries and the intra- and interspecific variation are illustrated. This has led to a new understanding of the genus and a clarification of the "W. caecula complex". The type species Westwoodia caecula Bate. 1857 is redescribed and a neotype is designated. W. megalops (Sars, 1883) is resurrected. The junior synonymy of W. longidactyla Carausu, 1949 with W. caecula is supported by the find of Carausu's original unpublished notes and drawings. Notes on undescribed species are given. New drawings are provided for W. manta Griffiths, 1974, W. longimana Shoemaker, 1934, W. brevicalcar (Goes, 1865) and W. megalops. All species are diagnosed; new distributions are given and plotted on a map; and a key to the named species is included.
Amphibians of Malawi : an analysis of their richness and community diversity in a changing landscape

This study summarizes the state of the knowledge of the amphibian diversity in Malawi highlighting the possible threats impending on this fauna correlated with human encroachment and land use change. New data about diversity, distribution and ecology have been gathered, whereas the old ones have been summarised, reviewed and commented. In order to put in context the responses of the amphibian communities to land use change, the main environmental characteristics of the country at a broad space and time scale have been explored. Furthermore, the original habitats and vegetation have been described, and their status in the present day Malawi discussed. In the same way, an overview of the actual state of the knowledge about the Malawian amphibians has been provided, and their ability to act as surrogate of environmental integrity in Sub-Saharan Africa commented on the basis of the available studies. Afterwards, the results of the study of the selected areas and samples have been analysed within this newly generated context. Different field and laboratory methods were applied for the quantitative analysis of the richness and diversity of the communities. Opportunistic search was used to detect species richness, whereas the visual encounter survey was applied to detect the relative abundance of species. Several indices of diversity and similarity, and extrapolations by means of true richness estimators were used for the analysis of the alpha and beta diversities. Additional information were gathered by means of pitfall traps with drift fence, and by the recording of the advertisement calls. Supplementary methods were applied for the analysis of the taxonomic composition of the collected material. In Malawi 84 amphibian species are recorded, two of which still undescribed (Leptopelis sp. and Phrynobatrachus sp.). Three further species need to be confirmed and might be possibly present too: Amietia viridireticulata, Hemisus guineensis, and Hyperolius minutissimus. Additionally, other unrecognised cryptic species — at least one — are present within the Hyperolius nasutus complex. Most of the species belong to the order Anura (82 species; 97.6%), whereas only two species belong to the Gymnophiona (2.4%). Anurans are divided into 12 families and 23 genera, whereas the two caecilians species into one family (Caecilidae) and two genera. The more diverse family is the Hyperoliidae (21 species, 25%) followed by the families Ptychadenidae (13 species, 15%), Arthroleptidae (11 species, 13%), Phrynobatrachidae (10 species, 12%), and Bufonidae and Pyxicephalidae (9 species, 11% respectively). The remaining high family diversity (seven families, Caecilidae included) is contrasted by a low number of species (11 species in total, 14%). Based on the available distribution data, the value of species richness of the anuran communities in Malawi is comprised between 5‒45 species. In average 16.8 ± 9.0 species (N=80) are to be found, 75% of the sites have less than 21 species, and only two sites have more than 25 species. Four hot spots of amphibian diversity were identified: the Nyika Plateau (24 species), Mangochi-Malombe (25 species), Zomba Plateau (32 species) and the Mulanje Massif (45 species). In the studied areas a mean of 14.7 ± 1.6 species was observed and extrapolations by means of the true richness estimators were in good agreement with this result. Among the studied areas the richest was Palm Forest Reserve (17 species), followed by Kaningina Forest Reserve (16 species) and Vinthukutu F. R., and Vwaza W. R (15 species). The poorest area was the Misuku Mountains with 12 species only and a slightly different ranking was generated by the true richness estimators. The mean of the species present in the samples was 4.8 ± 2.1 species, considerably less than the true species richness detected in the respective areas. Basing on the ranking generated by the K-dominance plot the most diverse samples were Palm F. R. and Misuku, whereas the less diverse were Kaningina F. R. and Fort Lister, confirmed by the values of the diversity indices. The main finding of this study was the observation of the lack of a clear match between environmental degradation and amphibian diversity, and the crucial importance of temporary water bodies for the preservation of the amphibian diversity. In fact, despite most of the original habitat formerly present in Malawi have been destroyed and replaced by cultivations, the amphibian communities of different areas showed a comparable diversity at both family and species richness level, and no evident match between environmental degradation and amphibian diversity was recognisable. Differences in species richness could mostly be explained by natural factors such the elevation gradient and the presence of temporary water bodies. However, it was not possible to exclude that the communities have changed during historical time and the shift in species composition already occurred together with the modification of their relative frequencies. Most of the species showed a remarkable ecological plasticity and several species were found in a variety of both natural and altered habitats. The classification of the Malawian amphibians on the basis of ecological guilds based on the available natural history data showed the preponderance (76%) of generalist pond breeders. As a consequence, most of these amphibians possessed a scarce capacity to act as surrogates of habitat integrity. Based on the result of this study the farm bush landscape with traditional agriculture practices bears a great potential to support amphibian diversity in terms of species richness, representing a compromise between local economic development and conservation. Furthermore, the results of this study indicate the outstanding importance of the southern-east region of Malawi for the conservation of the country’s amphibians.
Von den Canaren waren nur wenige Thysanopteren bekannt. Es ist daher um so mehr zu begrüßen, daß Titschack auf seiner Canaren-Reise im Frühjahre 1931 den Thysanopteren seine besondere Aufmerksamkeit schenkte. Ich freue mich, hervorheben zu können, daß dieses Material nicht nur durch seinen Reichtum an Individuen zu einer besonders exakten Bearbeitung geeignet war, sondern daß Titschack auch selbst das ganze Material in Canadabalsam-Präparate übertragen, dadurch der Zerstörung entrissen und damit das Hamburger Museum um eine prächtige Sammlung tadelloser Präparate bereichert hat. ... Die Ausbeute umfaßt 13 bereits bekannte und 14 für die Wissenschaft neue Arten; nur einige zweifelhafte Stücke wurden hier nicht behandelt. Die bekannten Arten sind 3 Kosmopoliten: Limothrips cerealium Hal., Thrips tabaci Lind. und Haplothrips gowdeyi Frkl. Eine Anzahl - wohl auch der größte Teil der neuen Arten - gehört dem Mediterranen Faunengebiet an, einzelne sind weiter verbreitet. Haplothrips retamae (Enderl.) ist nur von den Canaren bekannt. Regulation des matrizellulären Proteins SMOC-1 durch Zytokine und Stickoxid in Rattenmesangiumzellen

Zytokine stimulieren in Mesangiumzellen die Produktion und die Freisetzung großer Mengen entzündlicher Mediatoren. In dieser Arbeit wurden mit Hilfe der RAP-PCR („RNA arbitrarily primed polymerase chain reaction“), einer auf mRNA basierenden „Differential display“-Methode, die Effekte von Interleukin-1β (IL-1β) auf das Genexpressionsmuster in glomerulären Rattenmesangiumzellen untersucht. Dabei wurde das matrizelluläre Glykoprotein „Secreted modular calcium-binding protein-1“ (SMOC-1) identifiziert, welches in Mesangiumzellen durch IL-1β herunterreguliert wird. SMOC-1 wird von verschiedenen Zelltypen exprimiert und sezerniert, doch seine biologische Funktion konnte bisher nicht aufgedeckt werden. Weitere Experimente bestätigten, dass die mRNA- und Proteinexpression von SMOC-1 durch proinflammatorische Zytokine, wie IL-1β und Tumornekrosefaktor-α (TNF-α) herunterreguliert wird. Dieser Effekt wird zu einem großen Teil durch die endogene Freisetzung von Stickoxid (NO) aufgrund der Aktivität der induzierbaren NO-Synthase (iNOS) und der nachfolgenden Aktivierung der löslichen Guanylatzyklase (sGC) vermittelt. Außerdem tragen auch reaktive Sauerstoffver-bindungen (ROS), deren Bildung durch die Zytokine verstärkt wird, zur Herunter-regulierung der SMOC-1-Expression bei. Durch In-situ-Hybridisierungsexperimente konnte ferner gezeigt werden, dass die Hemmung der NO-Synthese durch den spezifischen iNOS-Inhibitor L-NIL in einem Rattenmodell der anti-Thy1.1-Glomerulonephritis die SMOC-1-Expression deutlich erhöhte. Dies belegt somit auch in vivo die biologische Relevanz von NO in der Modulierung der SMOC-1-Expression. Die funktionelle Rolle von SMOC-1 in Mesangiumzellen wurde durch die Hemmung der SMOC-1-Expression mit Hilfe einer spezifischen siRNA untersucht. Dabei zeigte sich, dass die Hemmung von SMOC-1 eine deutliche Inhibierung der mRNA-Expression von „Transforming growth factor β1“ (TGF-β1) sowie dessen Gesamtproteinspiegel zur Folge hat. Auch die Aktivität von TGF-β1 wurde reduziert, wie anhand der verringerten Spiegel an aktivem TGF-β1-Protein und der verringerten mRNA-Expression bekannter TGF-β-regulierter Gene, wie „Connective tissue growth factor“ (CTGF), „Plasminogen activator inhibitor-1“ (PAI-1) und Biglykan gezeigt wurde. Diese Ergebnisse deuten auf eine Rolle von SMOC-1 bei der Modulierung des TGF-β1-Signalwegs hin. Zusammenfassend betrachtet scheint NO die SMOC-1-Expression in der akuten glomerulären Entzündung zu vermindern und dadurch die TGF-β-getriebenen profibrotischen Signalprozesse zu limitieren. Der zweite Aspekt dieser Arbeit befasst sich mit der Rolle von Peroxisomen-Proliferator-aktivierten Rezeptoren (PPAR) in der IL-1β-vermittelten Expression der induzierbaren NO-Synthase. PPARα-Aktivatoren steigern in Mesangiumzellen die IL-1β-induzierte Aktivität der iNOS, während die Hemmung von PPARα durch spezifische Inhibitoren oder siRNA die iNOS-Expression/-Aktivität deutlich reduziert. Die Ergebnisse von Promotor-studien zeigten die essentielle Rolle einer möglichen PPAR-Bindestelle im iNOS-Promotor. IL-1β scheint die Bildung eines endogenen PPAR-Liganden zu induzieren, wodurch die Bindung von PPAR-Proteinkomplexen an das regulatorische DNA-Element im iNOS-Promotor verstärkt und dessen Aktivität gesteigert wird. Daneben scheinen jedoch auch Nebeneffekte der PPAR-Aktivatoren, wie die Freisetzung von ROS, zur synergistischen Wirkung auf die IL-1β-induzierte iNOS-Expression beizutragen. Die Wirkung von dualen PPARα/γ-Aktivatoren auf entzündliche Prozesse wird seit längerem diskutiert, daher wurden die biologischen Effekte von neu synthetisierten möglichen dualen PPARα/γ-Aktivatoren auf Entzündungsparameter, wie z. B. die iNOS-Expression, in Mesangiumzellen untersucht. Alle untersuchten Aktivatoren steigerten in der Form von Esterverbindungen die IL-1β-induzierte Expression der iNOS sowie der sekretorischen Phospholipase A2 (sPLA2), während die entsprechenden freien Säuren wenig Effekte zeigten. Diese proinflammatorische Wirkung scheint jedoch weniger auf einer Aktivierung des PPAR-Rezeptors zu beruhen als auf der Freisetzung von ROS, die durch die Aktivatoren teilweise deutlich erhöht wurde. Weitere Experimente zur Charakterisierung der PPAR-spezifischen Wirkung der Aktivatoren sowie zur optimalen Wirkkonzentration sind nötig, bevor der Effekt dieser Aktivatoren auf die inflammatorische Genexpression genau bewertet werden kann.
In der vorliegenden Arbeit wurden zehn Substanzen, die im Rahmen des Projekts INTAFERE analytisch in verschiedenen Gewässersystemen des Hessischen Rieds nachgewiesen werden konnten, ökotoxikologisch charakterisiert. Neben der Bestimmung der Akut- und der chronischen Toxizität wurde auch das endokrine Potential mit Hilfe eines rekombinanten Hefe-Assays ermittelt.Die akute Toxizität zeigt zwischen den verschiedenen Substanzen große Differenzen. Die drei Organophosphate TCPP, TBEP und TCEP zeigen selbst bei hohen Konzentrationen keine oder nur sehr geringe Effekte, während 4-NP, 4-t-OP, BPA, TDCPP, AHTN und Terbutryn mit LC50-Werten bis zu 5 mg/l eine höhere Toxizität besitzen.Im Hefe-Assay kann in mehreren Versuchswiederholungen das östrogene Potential von 4-NP (MW: 6,71x10-6 M), 4-t-OP (MW: 7,16x10-6 M) und BPA (MW: 4,88x10-6 M), sowie die antiandrogene Wirkung des Bisphenols (5,29x10-5 M) bestätigt werden. Im Gegensatz dazu können im Yeast Antiestrogen Screen zum ersten Mal Hinweise auf die antiöstrogene Wirkung von TCPP (MW: 6,86x10-5 M), Terbutryn (MW: 3,99x10-5 M), TDCPP (MW: 2,65x10-6 M) und TBP (MW: 2,28x10-5 M) aufgezeigt werden.TBP und TDCPP führen auch in der chronischen Exposition bei Potamopyrgus antipodarum zu einer Reduktion in der Embryonenzahl (mehrfach beobachtete NOEC beider Substanzen: 6,25 mg/kg), ein Effekt, der zunächst nicht von einer toxischen Wirkung unterschieden werden kann. Allerdings scheint sich ein antiöstrogener Wirkmechanismus auf die Schnecken bei den Versuchen zur Mischtoxizität zu bestätigen, da die gleichzeitige Exposition gegenüber BPA und 4-tOP zu einer geringfügigen Aufhebung des Effekts führt. Potamopyrgus reagiert ebenfalls mit einer Reduktion der Embryonenzahl bei der Exposition gegenüber AHTN (mehrfach beobachtete NOEC: 2 mg/kg), zeigt sich aber insensitiv gegenüber der Belastung mit Terbutryn.Die Exposition von Chironomus riparius gegenüber den verschiedenen Substanzen führt bis auf eine Ausnahme zu keinen signifikanten substanzbedingten Beeinträchtigungen. Einzig das s-Triazin Terbutryn verursacht eine hohe Mortalität mit einer NOEC (28 d) von 250 μg/kg. Eine Toxizität auf den Anneliden Lumbriculus variegatus zeigt sich lediglich bei derExposition gegenüber BPA (NOEC: 10 mg/kg; Biomasse, 28 d) und 4-NP (EC10: 6,88 mg/kg; 95% KI: 3,97-11,9; Reproduktion, 28 d). Die durchgeführten Versuche zur Toxizität von Mischungen zeigen eine größere Gefährdung auf als bei Vorliegen der Einzelsubstanzen in Konzentrationen unterhalb ihres Schwellenwertes zu vermuten wäre. Allerdings lassen sich die mit dem Hefe-Assay erzielten Ergebnisse aufgrund großer Variabilitäten zwischen den einzelnen Versuchswiederholungen nicht immer eindeutig mit Hilfe des additiven oder des unabhängigen Modells beschreiben, zeigen dabei jedoch trotzdem ein gegenüber den Einzelsubstanzergebnissen verändertes Risiko auf. Um diesem in der Risikobewertung Rechnung zu tragen, wird ein Verfahrensvorschlag entwickelt, in dem durch zusätzliche Sicherheitsfaktoren für PNECs ein Überschreiten des risikoanzeigenden Werts von 1 des PEC/PNEC-Quotienten einer Mischung verhindert werden kann.
En este catálogo provisional se recogen las malaeofaunas arqueológicas asociadas a ocupaciones humanas de la Península Ibérica. Se incluyen un total de 142 análisis malacológicos, y un anexo de 68 yacimientos donde los conjuntos son estrictamente ornament.tles o se encuentran en fase de estudio. Se encuentran reseñadas casi 200 especies de moluscos marinas, de agua dulce y terrestres. Además se ofrecen datos inéditos de 6 yacimientos (Abrigo de la Peila del Perro, Tennas romanas de Gijón, Cabezo Pequeño del Estaño, Pico Ramos, Almontc y La Viña). Consideramos que un trabajo de estas características resulta imprescindible en el actual estado de conocimiento arqueozoológico en la Península Ibérica y confiamos en que el catálogo se convierta en una referencia básica en estudios futuros.
Die oftmals apostrophierten Schwierigkeiten bei der taxonomischen Behandlung der Gattung Hylaeus F. können spätestens seit den genitalmorphologischen Untersuchungen von MEHELY (1935) als prinzipiell bewältigt gelten. Auf die beträchtlichen Unterschiede im Bau des Kopulationsapparates der Männchen hatte bereits E. SAUNDERS (1882a,b) unter Beifügung genauet Zeichnungen hingewiesen. Damit war der Schlüssel zur zuverlässigen Bestimmung dieser kleinen und relativ uniformen Bienen gegeben, aber leider hat in Europa bisher nur ELFVING (1951) versucht, diese Merkmale mit Bestimmungstabellen zu verbinden, wie es etwa in der Neuen Welt längst unumgänglich ist (vgl. z. B. METZ 1911, SNELLING 1966-1970). POPOV (1939) gründete seine subgenerische Einteilung der Gattung auf die Erkenntnisse von MEHELY, und diese hat sich trotz einseitiger Anlage, obwohl nur die 33 berücksichtigt wurden, bis heute weitgehend bewährt. Die Erfahrung lehrt, daß einzelne Insektengruppen erst über geeignete Bestimmungsliteratur auch mehr Bearbeiter finden und einem breiteren, kooperativen Studium zugänglich werden. Im gegenwärtigen Stand bedürfen die Hj/acus vor allem eingehender biologischer Bearbeitung von vielen Seiten. Insbesondere sollte unsere Kenntnis ihrer natürlichen Variation, Bionomie und Verbreitung ergänzt werden, bevor taxonomische Konsequenzen gezogen werden. Das vorliegende Manuskript ist deshalb in erster Linie als Arbeitsmaterial zur Verständigung und Diskussion angelegt und berücksichtigt praktische Erfordernisse, insbesondere die der Determinationspraxis. Trotzdem hat sich daraus gleichwohl von selbst eine Revision der Untergattungen (unter Einbeziehung der ... ) und eine nicht geringe Anzahl systematischer Anmerkungen ergeben. Durch die freundliche Vermittlung von Herrn Dr. KÖNIGSMANN (Berlin) konnte aus dem Nachlaß BLÜTHGENS ein umfangreiches, wenn auch unvollständiges Manuskript mit dem Arbeitstitel "Die mitteleuropäischen Arten der Gattung Prosopis F." mit ausgewertet werden. Für den Druck ist dieser Text, an dem BLÜTHGEN vor und noch neben seiner Vespoiden-Arbeit (1961) schrieb, viel zu breit angelegt. Von seiner Intention dürfte aber viel in die vorliegende Untersuchung eingeflossen sein. Außerdem habe ich seine sorgfältig recherchierte Synonymie weitgehend übernommen. Sie wurde nach Möglichkeit kontrolliert und ergänzt, ohne jedoch Vollständigkeit anzustreben. Die langen Listen von LEcLERcg (I964) und WARNCKE (1972 ) konnten nur insoweit überprüft werden, als es Fragen von akuter nomenklatorischer Relevanz betraf. Der Praktiker benötigt ohnehin zunächst nur die Angaben, die ihm den Anschluß an die (wenige) Spezialliteratur der jüngeren Vergangenheit erlauben. Diese Synonymie ist vollständig. Die Tabellen beziehen sich auf das gesamte Europa von Gibraltar bis zum Ural und schließen die großen Mittelmeerinseln ein; Zypern, wiewohl Kleinasien vorgelagert, wurde zusätzlich aufgenommen, denn die wenigen Formen (meist Endemiten) lassen sich nahtlos anfügen. Die Aufteilung der europäischen geographischen Regionen folgt dem allgemeinen Verständnis: Es werden West-, Ost-, Mittel-, Nord- und Südeuropa unterschieden (Abkürzungen: W, E, C, N, S). SE-Europa bezieht sich insonderheit auf den Balkan und die S-Ukraine. Zoogeographische Grenzen bestehen vor allem zwischen W- und C-Europa, wobei sich die Iberische Halbinsel noch einmal absetzt, sowie zwischen S- und C-Europa mit den Alpen als Barriere. Der Kaukasus gehört nur teilweise zu Europa, er wurde und wird aber gern besammelt und deshalb mit verlangt. In seinem westlichen Teil ist die Fauna noch sehr zentraleuropäisch, im östlichen zeigen sich verstärkt auch klein- und mittelasiatische Arten, die, soweit bekannt, ~inbegriffen wurden. Es sind alle Arten berücksichtigt, die nach meiner Kenntnis bisher für das Gesamtgebiet als gesichert gelten bzw. noch zu erwarten sind. Daß auch weiterhin mit neuen Arten zu rechnen ist, ergaben vorbereitende Studien (DATHE 1977, 1979b). Wenig an Fundangaben und Material lag mir aus dem europäischen Teil der RSFSR (E-Europa) vor. Die Bearbeitung für die europäische UdSSR von OSYCNJUK (1978) stellt erst einen Anfang dar. Von allen verzeichneten Arten werden Abbildungen der Gesichtsmaske und des Kopulationsapparates (...) gegeben. Der Verweis auf eine bestimmte Abbildungsnummer steht in eckigen Klammern. Alle Beschreibungen wurden in standardisierter Form völlig neu angefertigt. Dazu stund das umfangreiche Material des Zoologischen Museums Berlin (ZMB) zur Verfügung, insbesondere die Sammlungen von ALFKEN und BLÜTHGEN.
Die Gattung Aspilota FOERSTER ist als solche verhältnismäßig leicht zu erkennen. Die Bestimmung der Arten hingegen stößt auf ungewöhnliche Schwierigkeiten. Es ist fast sicher, daß erst ein kleiner Teil der existierenden Arten entdeckt ist. Der vorliegende Beitrag hat jedoch nicht das Ziel, neue Arten zu beschreiben, sondern stellt einen ersten Versuch dar, wenigstens den Großteil der schon bekannten Spezies in ein zusammenfassendes taxonomisches System zu bringen.
Several generic schemes used in classifying species belonging to Prionospio Malmgren, 1867 sensu lato have been reviewed; three taxa have been retained at the generic level, and three additional taxa at the subgeneric level. The following genera and subgenera are recognized: Prionospio Malmgren, 1867, including the subgenera Minuspio Foster, 1971, Aquilaspio Foster, 1971 and Prionospio Malmgren (sensu striclo); Apoprionospio Foster, 1969, and Paraprionospio Caullery, 1914. Prionospio sensu lata includes species with various combinations of branchiae which are smooth (apinnate), wrinkled, or with digitiform pinnules, beginning on setiger 2. Apoprionospio includes species having branchiae from setiger 2, with at least one pair having plate-like pinnules. Paraprionospio includes species with branchiae from setiger 1, with all pairs having platelike pinnules. Fifteen species, including seven new species, belonging to the genus Prionospio are described. Prionospio (Prionospio) steenstrupi Malmgren, 1867 is described from the syntype series, and is newly synonymized with P. fallax Söderström, 1920. The validity of P. bocki Söderström, 1920 as a separate species is discussed, as is the indeterminable nature of P. malmgreni Claparède, 1869. Prionospio (Prionospio) dubia Day, 1961 originally described as a new variety of P. malmgreni from S Africa, is raised to full species status. Prionospio (P.) cristata Foster, 1971 and P. (P.) heterobranchia Moore, 1907 are redescribed. Prionospio (P.) tripinnata, a new species with three pairs of pinnate branchiae, is described from the Mediterranean Sea and compared with P. plumosa Sars, 1872. A new synonymy is proposed for P. lobulata Fauchald, 1972 with P. (P.) ehlersi Fauvel, 1928. Two new species, P. (Minuspio) fauchaldi and P. (M.) laciniosa, are described in which the apinnate branchiae are distinctly wrinkled or sculptured, rather than smooth. P. (M.) laciniosa is also distinguished by dorsal crests modified into semicircular flaps. Several species previously referred to Prionospio (Minuspio) cirrifera Wirén, 1883 are reviewed and described. The seven species recognized by Foster in the genus Minuspio are considered; several are retained, and new species and new synonymies are proposed. Because the types are apparently lost, a description of P. (M.) cirrifera is given based on specimens from near the type locality. Prionospio (M.) aluta new species is separated from P. (M.) cirrifera on the basis of the presence of lateral pouches in P. aluta and their absence in P. cirrifera. Prionospio (M.) delta Hartman, 1965 is retained as a valid species; P. (M.) longibranchiata Reish, 1968 and P. (M.) minor Fauchald & Hancock, 1981 are newly synonymized with P. delta. P. (M.) multibranchiata Berkeley, 1927 is also retained as a valid species, and P. (M.) perkinsi, P. (M.) lighti and P. (M.) wireni are newly described from shallow water. Two species are recognized as belonging to Apoprionospio Foster: A. pygmaea (Hartman, 1961) and A. dayi Foster, 1969. New records and range extensions are given for both species. New records are also presented for Paraprionospio pinnata (Ehlers, 1901). The specimens examined as part of this study are based primarily on deep-sea materials collected in the Atlantic Ocean, but also include shallow-water specimens from the east, west, and gulf coasts of N America.
The theoretical concept of the biological species and the multidimensional species category, as currently applied by a majority of ornithologists and by many other biologists, replaced the typological-morphological species concept during the first half of this century and became a central tenet of the synthetic theory of evolution. The concept of biospecies is a 'horizontal' concept referring to contemporary reproductive communities at any particular period, e.g. the Recent period or any other time level of the geological past. Historical 'species' concepts as applied by cladists and palaeontologists refer to artificially delimited portions of 'vertical' phyletic lineages for which the application of the term 'species' causes severe problems. Discussions would be simplified if the concept and term 'species' was to be restricted to cross sections of phyletic lineages at any time level and a separate taxonomy outside the Linnaean system of genera and species was to be conceived to deal with phyletic lineages. Under each of the theoretical species concepts, species taxa are assigned broadly to intermediate or narrowly defined taxonomic species categories. Ornithologists of the 19th century applied morphological species concepts, emphasizing morphological character differences between species (rather than distinctness) and the fertility of con specific individuals (rather than the isolation from non-conspecific populations). Nearly all leading museum ornithologists in 19th-century Europe delineated monotypic Linnaean species, whereas the explorer-naturalists of the Gloger-Middendorff school (including Panas, Faber, Gloger, Nordmann, Middendorff, Schrenck, Radde, as well as Schlegel and Blasius) delimited widely circumscribed species taxa. Their researches in the vast territories of eastern Europe, Siberia and the Far East from the late 18th century to the 1880s and, in particular, their rich specimen material, demonstrated direct intergradation of many taxa (geographical varieties) of birds, thus revealing the conspecific nature of numerous narrowly conceived morphospecies previously described by museum workers. The ornithologists of the Gloger-Middendorff school also studied several conspicuous phenomena of geographical character variation in birds (and mammals) across Eurasia, especially plumage colouration (and pelage) and body size, but none of them was an evolutionist. They an adhered to a typological-creationist theoretical species concept. During the late 19th century, the museum specialists' taxonomic notion of narrow morphospecies dominated systematic ornithology in Europe, overtaking the work of the naturalists of the Gloger-Middendorff school, which fell into oblivion. The ornithologists of the Bairdian school in North America (Baird, Coues, Allen, Ridgway) further developed the concept of subspecies after the 1850s and especially from the 1870s onward. Their views were fully in accord with Darwin's theories of evolution' thus they defined the subspecies in a somewhat simplified manner as 'nascent species': These ornithologists were able to base their studies on collections of extensive specimen material which they had obtained during a series of exploring expeditions across the North American continent. Their studies led to the discovery of many aspects of both individual and geographic variation in birds. There are interesting historical similarities between the coinciding taxonomic interpretations and the comparable application of fairly broad limits of morphospecies by the North American ornithologists and the earlier exploring ornithologists in Europe, arrived at Independently by these, research groups, The study of specimens in 'series' (,suites'), beginninng with the naturalists of the Gloger-Middendorff school and, in particular, with the naturalists of the Bairdian school in North America, eventually led to the overcoming of the prevating typological view of variation and the development of 'population thinking'. Influenced by the work of Henry Seebohm in Britain and that of the North American ornithologists, Hartert in England and Kleinschmidt in Germany jointly succeeded in overcoming the strong opposition of the leadi.ng ornithologists in Europe during the 1890s and early 19008 and introduced a concept which soon developed into the biological species concept through the work of Stresemann, Rensch, and in particular, Ernst Mayr. Hopefully, ornithologists will continue the study of taxa at low, intermediate and high levels of microtaxonomic differentiation and will identify the subspecies groups, biological species and the biogeographical species in the world's avifaunas. Cladistic analyses will provide historical {'vertical'} overviews of phyletic lineages at different taxonomic levels.
The 20 species of Neogene Scleractinia in the suborders Caryophylliina and Dendrophylliina known from the Dominican Republic are revised and illustrated. This research was based on 1590 specimens obtained primarily from the collections of the Naturhistorisches Museum, Basel, Switzerland; National Museum of Natural History, Smithsonian Institution, Washington, DC, U.S.A.; and Tulane University, New Orleans, LA, U.S.A. Eight new records are reported for the Neogene of the Dominican Republic, including four new species: Antillocyathus alatus, Trochocyathus chevalieri, T. duncani, and Paracyathus sinuosus. Special attention is given to the genus Asterosmilia, since half (five) of the known species in this genus occur in the Dominican Republic. Most species described herein are assumed to constitute a deep-water fauna by analogy to depth ranges of the same or similar species known from the Recent. Certain localities and parts of formations are inferred to represent deep-water (> 200 m) facies. These inferences may aid in the paleoecological interpretation of other fossils collected from these areas.
Multivariate statistical procedures are used to distinguish species in the reef-coral genus Stephanocoenia through a continuous Neogene sequence (five-million year time interval) in the Cibao Valley of the northern Dominican Republic. This genus is the only member of the family Astrocoeniidae that occurs in the sequence. The material consists of 56 colonies (17 of which are measured) from 24 localities in four river sections, the most important being Rio Gurabo and Rio Cana. Ten characters are measured on each of 10 corallites per colony. The data are analyzed using cluster and canonical discriminant analysis to group colonies into clusters representing species. Identical measurements on modem colonies collected near Discovery Bay, Jamaica are included for comparison. Two fossil species are defined in the analysis, one of which is new (Stephanocoenia duncani, n. sp.). Both species are significantly distinct from the single modem species (S. intersepla) that is the sole living representative of the genus. Study of collections from other reef localities shows that both fossil species occur only during Neogene time and only at a limited number of localities. Patterns within each species are traced up a composite stratigraphic section using nonparametric statistical analyses. One of the two fossil species (S. spongiformis) is found to remain stable through time, whereas the other (S. duncam) changes its morphology in a direction approaching the cluster for the modem species. Further study of patterns of variation within the one modern and two fossil clusters shows that intraspecific variation is unusually complicated in this genus. The clusters overlap, and colonies within each cluster differ widely. Variation between populations within the modem species occurs in the same characters as those which distinguish the modem species from the fossil species converging with it (S. duncam). However, these two species form a morphologic continuum that cannot be explained by environment alone. Therefore, they may represent two gradually intergrading chronospecies within one lineage. Of the two fossil species of Stephanocoenia defined, one species (s. spongiformis) exhibits an evolutionary pattem similar to that observed in the family Poritidae. In this pattern, species were found to have short durations and stable morphologies and to have become extinct during the mid- to late Pliocene through early Pleistocene mass extinction. In contrast, the second species of Slephanocaenia (S. duncam) may have evolved over a long time period, possibly forming chronospecies that survived the mass extinction. Unlike genera in the Poritidae, however, no radiation of taxa occurred in the genus after the extinction event. Since no consistent relationship has been discovered between morphology and environment in these corals with the data at hand, their paleoecologic value can only be determined after data on more taxa are collected, and their associations with other corals are studied. This study represents part of a multidisciplinary project on the stratigraphy and paleontology of the northern Dominican Republic, coordinated by P. Jung and J. B. Saunders of the Naturhistorisches Museum Basel, Switzerland.
Nach einer Betrachtung über die Geschichte und den. Stand der Erforschung der 2. Stufe des Mittleren Keupers, des "Schilfsandsteins", wurde der hisher bekannte Fossilinhalt der Stufe überprüft. Die in einer Tonstein-Grube von Eberstadt (Württ.) entdeckte neue, reiche Lamellibranchiaten-Fauna mit 47 sp. ssp. in 17 Gattungen wurde untersucht und beschriehen. Die Fauna ist marin und hat keine Beziehungen mehr zur Fauna der Germanischen Trias, im besonderen zu der verarmten Muschelkalk-Fauna des Lettenkeupers und Unteren Gipskeupers. Die Eberstadter Muschelfauna ist (vielleicht mit einer Ausnahme) nicht von S, aus dem alpinen Karn, eingewandert. Sie muß, als eigenständige Lamellibranchiaten-Fauna des Karn, von N, aus Richtung der heutigen Nordsee, gekommen sein; von dort floß Meerwasser in den Sedimentationsraum der Schilfsandstein-Stufe ein und nach S wieder aus. Einzelne Arten der Muschelfauna nehmen schon rhaetische und liassische Formen vorweg. Die Schilfsandstein-Stufe des süddeutschen Mittelkeupers ist mit ihren Peliten und ihrer autochthonen marinen Fauna als Ablagerung eines weiten, extrem flachen Binnen-Schelfmeeres zu verstehen, in das zusätzlich aus dem kontinentalen Raum im NO Sand eingeschüttet wurde.
Plastids are complex plant organelles fulfilling essential physiological functions, such as photosynthesis and amino acid metabolism. The majority of proteins required for these functions are encoded in the nuclear genome and synthesized on cytosolic ribosomes as precursors, which are subsequently translocated across the outer and inner membrane of the organelle. Their targeting to the organelle is ensured by a so called transit peptide, which is specifically recognized by GTP-dependent receptors Toc159 and Toc34 at the cytosolic side of outer envelope. They cooperatively regulate the insertion of the precursor protein into the channel protein Toc75, thereby initiating the translocation process. Toc34 is regarded as the primary receptor, while Toc159 probably provides the driving force for the insertion. Precursor transfer is achieved by the physical interaction between both receptors in the GTP loaded state. One translocon unit, also called the Toc core complex, is formed by four molecules Toc34, four molecules Toc75 and one molecule Toc159. In the GDP-loaded state, Toc34 preferably forms homodimers, whose physiological function was investigated in the presented study. It could be shown that the dissociation of GDP and therefore the nucleotide exchange are inhibited by the homodimeric state of Toc34. Dissociation of the homodimer is induced by the recognition of a precursor protein, which renders the binding of GTP and subsequent interaction with Toc159 possible. Thus, the homodimeric conformation could reflect an inactive state of the translocon, preventing GTP consumption in the absence of a precursor protein. Both homodimerization as well as heterodimerization of the receptor are regulated by phosphorylation, which could be demonstrated by in vitro and in vivo approaches using atToc33 from Arabidopsis thaliana as a model system. Since the phosphorylated form of Toc34 cannot be assembled with the Toc core complex, it can be concluded that the interactions between GTPase domains not only regulate the transfer of precursor proteins, but also warrant the integrity of the translocon.
The aim of the study was to investigate the role of the CX3C chemokine FKN in the role of platelet adhesion. The presence of the FKN receptor CX3CR1 in platelets is demonstrated and G-protein dependent activation of platelets with soluble FKN results in the increased adhesion of platelets to collagen and fibrinogen under flow 228 and adhesion of leucocytes to firmly attached platelets 231. Whether membrane-bound FKN is capable to promote the direct adhesion of platelets in flowing blood analogue to leucocytes was completely unknown. The adhesion mechanisms of FKN in mediating the adhesion of leucocytes under flow are well characterised and represent a novel unique mechanism of leucocyte capture and firm adhesion: FKN is responsible for immediate arrest of flowing CX3CR1 expressing leucocytes without the participation of additional adhesion receptors and ligands. This is in contrast to the classical leucocyte adhesion pathways, which are multistep processes involving leucocyte arrest, rolling and subsequent cell activation prior to firm arrest. In leucocytes, the FKN – CX3CR1 axis is sufficient to allow rapid arrest of leucocytes at low shear flow conditions 67, 101, 115, 122, 261. The set of data from this study demonstrates that immobilised FKN was capable to mediate the adhesion of platelets under low shear conditions, whereas there was no interaction in the absence of shear flow. In the presence of vWf in the adhesion matrix, FKN mediated the potent increased adhesion of platelets. This was in parts due to the activation of flowing platelets via CX3CR1 and the augmented translocation of platelets on FKN via the vWf receptor GPIbα. With respect to platelet activation, the function of endothelial FKN was comparable to leucocytes: in both cell types, the FKN dependent activation is mediated by its cognate receptor CX3CR1. This is in contrast to the adhesive capacity: in leucocytes, FKN dependent adhesion is mediated by CX3CR1, whereas in platelets, the adhesive capacity was mostly mediated by the vWf receptor GPIbα with only minor contribution from CX3CR1. In platelets, activation and adhesion by FKN were mediated by two distinct receptors, whereas in leucocytes, CX3CR1 is solely responsible for FKN dependent activation and adhesion. The presented results point out to a role of platelets in early stage of atherosclerosis. The in vivo expression of both, FKN and vWf is regulated by TNF-α, which is released in early stages of inflammation. The presence of vWf and FKN in the endothelial lining of blood vessels during these conditions is sufficient to initiate the capturing and translocation of platelets on the tunica interna. The rolling of platelets on the endothelium can induce endothelial damage and inflammation of the vessel, which might advance to the generation of clinically significant atherosclerotic plaques and fibrous atheroma.
Methanosarcina acetivorans kann Kohlenmonoxid (CO) als Kohlenstoff- und Energiequelle nutzen. Als Endprodukte entstehen bei der Verwertung von CO neben Methan signifikante Mengen an Azetat und Formiat sowie Dimethylsulfid (DMS). In dieser Arbeit sollten verschiedene Aspekte dieses außergewöhnlichen CO-Stoffwechsels analysiert werden. Folgende Ergebnisse wurden erzielt: 1) Weder die Methanogenese, noch die Bildung eines der anderen Metaboliten wird durch hohe CO-Partialdrücke gehemmt. Inhibitorstudien mit BES belegen, dass die CO-Oxidation und die Bildung von Azetat, Formiat und DMS nicht an die Methanogenese gekoppelt sind. Inhibitorstudien legen nahe, dass die Methanogenese aus CO am Aufbau eines Na+-Gradienten beteiligt ist und das Vorhandensein eines vom Protonenpotential-abhängigen Schrittes. 2) Eine neue, kostengünstige Transformationsmethode mittels Polyethylenglykol (PEG) konnte für M. acetivorans etabliert werden. Die Transformationshäufigkeit betrug ca. 1,1 x 107 Transformanden/μg DNA und liegt damit im Bereich von der der bisher etablierten Liposomen-vermittelten Transformationsmethode. 3) Mutantenanalysen und physiologische Studien belegen eine Beteiligung der Mts-Proteine in der DMS-Bildung und DMS-Verwertung, da in ihrer Abwesenheit kein DMS aus CO gebildet, kein Methan aus DMS produziert wird, und M. acetivorans nicht mehr auf DMS als Energiequelle wachsen kann. Die Mts-Proteine sind für das carboxidotrophe Wachstum jedoch nicht essentiell. Immunologische Analysen belegen eine substratabhängige Regulation von MtsF und weisen auf genetische Interaktionen der einzelnen Loci oder der Isoformen selbst hin. 4) Die monofunktionellen CODH-Isoformen von M. acetivorans sind am carboxidotrophen Wachstum beteiligt, jedoch nicht essentiell. Die beiden Isoformen der bifunktionellen CODH/ACS sind funktionell, und wenigstens eine von ihnen ist für autotrophes als auch carboxidotrophes Wachstum notwendig. Eine mögliche posttranslationale Modifikation von Cdh1 weist auf unterschiedliche physiologische Funktion und/oder Lokalisation hin. 5) Die F420H2-Dehydrogenase ist essentiell für methylotrophes, nicht jedoch für carboxidotrophes Wachstum.
Pflanzliche Biomasse bietet sich hervorragend als billiges und in großen Mengen verfügbares Ausgangssubstrat für biotechnologische Fermentationsprozesse an. Für die Herstellung von Bioethanol ist die Hefe Saccharomyces cerevisiae der wichtigste Produktionsorganismus. Allerdings kann S. cerevisiae die in Biomasse in großer Menge enthaltenen Pentosen Xylose und Arabinose nicht verwerten. Für einen ökonomisch effizienten Fermentationsprozess ist es daher essentiell, das Substratspektrum der Hefe entsprechend zu erweitern. Im Rahmen dieser Arbeit ist es gelungen, den bereits in Hefe etablierten bakteriellen Arabinose-Stoffwechselweg signifikant zu verbessern. Genetische und physiologische Analysen ergaben, dass eines der heterolog produzierten Enzyme, die L-Arabinose-Isomerase aus Bacillus subtilis, einen limitierenden Schritt innerhalb des Stoffwechselweges darstellte. In einem genetischen Screening konnte ein aktiveres Isoenzym aus Bacillus licheniformis gefunden werden. Zusätzlich wurde der Codon-Gebrauch aller heterologen bakteriellen Gene dem Codon-Gebrauch der hoch-exprimierten glykolytischen Gene von S. cerevisiae angepasst. Mit diesem rationalen Ansatz konnte die Ethanolproduktivität aus Arabinose um mehr als 250% erhöht werden, der Ethanolertrag wurde um über 60% gesteigert. Dies stellte die erste erfolgreiche Verbesserung eines heterologen Stoffwechselwegs in S. cerevisiae über Codon-optimierte Gene dar. In einem breit angelegten Screening wurde zum ersten Mal eine prokaryontische Xylose-Isomerase gefunden, die in S. cerevisiae eine hohe Aktivität aufweist. Durch das Einbringen des xylA-Gens aus Clostridium phytofermentans in verschiedene Hefe-Stämme wurden diese in die Lage versetzt, Xylose als alleinige Kohlenstoffquelle zu nutzen. Zusätzlich konnte damit die Vergärung von Arabinose und Xylose in einem einzigen S. cerevisiae-Stamm kombiniert werden. Vorherige Versuche, einen Pentose-vergärenden Stamm zu konstruieren, der einen bakteriellen Arabinose-Stoffwechselweg mit dem eukaryontischen Xylose-Reduktase/Xylitol-Dehydrogenase-Weg kombinierte, scheiterten an der unspezifischen Umsetzung der Arabinose durch die Xylose-Reduktase zu dem nicht weiter verstoffwechselbaren Arabitol. Für einen industriellen Einsatz der rekombinanten Hefen war es unerlässlich, die Eigenschaften für die Pentose-Umsetzung in Industrie-relevante Hefe-Stämme zu übertragen. Durch die Etablierung von genetischen Methoden und Werkzeugen ist es in dieser Arbeit gelungen, Industrie-Stämme zu konstruieren, die in der Lage sind, Arabinose oder Xylose zu metabolisieren. Dabei wurden die heterologen Gene stabil in die Chromosomen der Stämme integriert. Diese wurden mit Hilfe von „Evolutionary Engineering“ so optimiert, dass sie die Pentose-Zucker als alleinige Kohlenstoffquellen zum Wachstum nutzen konnten. Fermentationsanalysen zeigten eine effiziente Umsetzung der Pentosen zu Ethanol in diesen Stämmen. Damit ist ein neuer Startpunkt für die Konstruktion von industriellen Pentose-fermentierenden Hefe-Stämmen markiert, der zukünftig effizientere Bioethanol-Produktion ermöglichen wird.
Höhere Eukaryoten stellen ein Ensemble von Zellen dar, die in Kompartimente unterteilt sind. Somit sind intra- und interzelluläre Transportprozesse entscheidend für das Überleben dieser Zellverbände. In meiner Arbeit habe ich Evolution und Struktur von Translokationskomplexen untersucht, um einige Aspekte dieser komplexen Systeme zu untersuchen. Eingangs befassten wir uns mit Rezeptorsystemen am Beispiel des Proteintransports. Mittels phylogenetischer Analysen fanden wir heraus, dass Pex5 nicht der Urahn der anderen untersuchten 3-TPR-Domänen ist, obwohl Pex5 in allen eukaryotischen Organismen vorkommt. Ein Vergleich der 3-TPR-Domänen mit der restlichen Sequenz des Rezeptorproteins ergab, dass die 3-TPR-Domänen eine langsamere Evolutionsgeschwindigkeit aufweisen, was für eine Evolutionseinschränkung durch Interaktionspartner spricht. Sec72 ist möglicherweise aus einer TPR1 (Hop) Domäne entstanden und eine Funktion als Hsp70-erkennende Komponente des Sec-Komplexes für den post-translationalen Import kann daraus abgeleitet werden. „Recycling“ von 3-TPR-Domänen anderer Proteine konnten wir durch unsere phylogenetische Analyse auch für die zweite 3-TPR-Domäne von Tom34 nachweisen, die mit CYP40/FKBP51/52 clustert. Darüber hinaus war es uns möglich, die plastidär bzw. mitochondriell lokalisierten Formen von Toc64 phylogenetisch zu unterscheiden. Durch Erzeugung von Homologiemodellen konnten organellspezifische Aminosäuren strukturell eingeordnet werden. Dabei stellten wir fest, dass sich fast alle Positionen, die sich in der Aminosäurekomposition unterscheiden, auf der konvexen Seite der 3-TPR-Domäne befinden. Molekulardynamische Simulationen zeigten zudem deutliche Veränderung der Hauptbewegungen der 3-TPR-Domänen nach Komplexierung mit dem Hsp90-C-Terminus. Bei Bindung des Liganden werden intramolekulare Wasserstoffbrücken sowohl auf der konvexen als auch konkaven Seite der 3-TPR-Domäne „umgeschaltet“. Diese Erkenntnisse führen zu zwei Hypothesen: 1.) die Organellspezifität der Rezeptoren wird durch die Interaktion mit anderen Komplexpartnern garantiert und 2.) die Änderungen des Wasserstoffbrückennetzwerkes auf der konvexen Seite nach Hsp90-Bindung führen zur Ausbildung der Bindungsstelle für die andere Komplexkomponente. Beide Hypothesen erklären die experimentellen Beobachtungen bezüglich der Rezeptoren und warum keine phylogenetischen Hinweise für die Existenz von Vorstufenprotein-spezifischen Hsp70/90-Proteinen gefunden werden konnten. Nach dem Rezeptor haben wir uns mit dem Translokationsprozess befasst. Wir konnten phylogenetisch zeigen, dass sich Omp85 aus Proteobakterien im Vergleich zu Cyanobakterien und Eukaryoten insbesondere durch andersartige POTRA Domänen auszeichnet und fanden zwei konservierte Motive in der Porenregion. Zudem konnten wir im Heterokontophyten P. tricornutum ein vollständiges Omp85 identifizieren (bipartite Signalsequenz, 2 POTRAs, Pore mit langen Schleifen). Die Aminosäuresequenz weicht teils deutlich von den bekannten Omp85-Proteinen ab, was die Entdeckung erschwerte. Wir haben damit geklärt, dass auch im Translokationsapparat von komplexen Plastiden ein b-Fassprotein der Omp85 Familie die Kerneinheit bildet. Ebenfalls zu den Protein-transportierenden b-Fassproteinen gehört TolC, das aber im Gegensatz zu Omp85 auch andere Substanzen, wie zum Beispiel Siderophore transportiert. Alr2887 ist das einzige TolC-ähnliche Protein aus Anabaena sp. PCC7120. Vergleichende Phänotypuntersuchungen weisen auf eine Interaktion eines ABC-Transporters (DevBCA Operon) mit Alr2887 hin. Die Distanz zwischen äußerer Membran und Plasmamembran ist in Anabaena doppelt so groß wie in E. coli. Entsprechend fanden wir im Adapterprotein DevB eine stark verlängerte dimere Doppelwendel, die das von TolC gebildete a-Fass im Periplasma bis hin zum ABC-Transporter in der Plasmamembran theoretisch fortsetzen kann. Da verschiedenste in Anabaena existierende ABC-Transporter TolC als Abflusskanal benötigen, nehmen wir an, dass Alr2887 ein Rundumtalent in Bezug auf die zu transportierenden Substrate darstellt. Dieses ist auch aufgrund der basalen Einordnung im phylogenetischen Baum zu vermuten; es könnte somit auch in den „Multi-Drug-Efflux“ involviert sein. Nicht nur ABC-Transporter, auch TonB-abhängige Transporter stehen in funktionellem Zusammenhang mit TolC. Wir haben Aminosäuresequenzen von ~4600 TBDTs aus Gram-negativen Bakterien und Cyanobakterien zusammengetragen und nach ihrer paarweisen Ähnlichkeit geclustert. Anhand experimentell charakterisierter TBDTs mit bekannten Substraten und TBDTs mit vorhergesagten Substraten konnten wir sehr vielen Clustern ein Substrat zuordnen, das die in ihnen zusammengefassten TBDTs aller Wahrscheinlichkeit nach importieren. Wir konnten ferner feststellen, dass es noch eine Menge weiterer Cluster mit unbekannten Substratspezifitäten gibt und unsere Analysen stimulieren somit die Arbeiten an diesem System im Allgemeinen und in Cyanobakterien im Besonderen.
Die anaerobe Atmung mit Nitrat und Nitrit als terminalen Elektronenakzeptoren bildet einen wichtigen Teil des biologischen Stickstoff-Zyklus. Beispiele sind Denitrifikation und respiratorische Nitrat-Ammonifikation, wobei in beiden Fällen in einem ersten Schritt Nitrat zu Nitrit reduziert wird. In der Denitrifikation entstehen dann verschiedene gasförmige Produkte (NO, N2O, N2), wogegen Nitrit in der Ammonifikation ohne die Freisetzung weiterer Zwischenprodukte direkt zu Ammonium reduziert wird. Während die terminalen Reduktasen dieser Atmungsketten gut untersucht sind, ist das Wissen über die Zusammensetzung kompletter Elektronentransportketten sowie die Interaktion einzelner Proteine als auch zwischen den Proteinen und Chinonen in der Membran begrenzt. Ziel dieser Arbeit war die Charakterisierung der membranständigen Chinol-Dehydrogenasen NapGH und NrfH in der respiratorischen Nitrat-Ammonifikation von Wolinella succinogenes. Dieses Epsilonproteobakterium ist ein etablierter Modellorganismus der anaeroben Atmung und wächst durch respiratorische Nitrat-Ammonifikation mit Formiat oder H2 als Elektronendonoren. Als terminale Reduktasen werden dabei die periplasmatische Nitratreduktase NapA und die Cytochom c-Nitritreduktase NrfA benötigt. Die Genomsequenz weist keine weiteren typischen Nitrat- und Nitritreduktasen auf, und napA- und nrfA-defiziente Mutanten sind nicht in der Lage durch Nitrat- bzw. Nitritatmung wachsen. Das Operon des Nap-Systems (napAGHBFLD) von W. succinogenes kodiert Proteine, die an der Nitrat-Reduktion durch Menachinol beteiligt sind (NapA, -B, -G und -H) und Proteine, die für die Reifung und Prozessierung von NapA benötigt werden (NapF, -L und –D). Im Gegensatz zu vielen anderen Bakterien läuft die Nitrat-Atmung unabhängig von einem NapC-ähnlichen Protein ab, das als membrangebundenes Tetrahäm-Cytochrom c für die Chinol-Oxidation zuständig ist und Elektronen über den Elektronenüberträger NapB an die terminale Reduktase NapA liefert. Zwar sind im Genom zwei NapC-Homologe kodiert (FccC und NrfH), doch die Deletion beider Gene hatte keinen Einfluss auf die Nitrat-Atmung. Es wurde vermutet, dass die Funktion von NapC in W. succinogenes stattdessen durch die beiden Fe/S-Cluster Proteine NapG und NapH übernommen wird. Die Reduktion von Nitrit zu Ammonium wird durch den NrfHA-Komplex katalysiert. Das Pentahäm-Cytochrom c NrfA bildet dabei die katalytische Untereinheit, die über das membranständige Tetrahäm-Cytochrom c auf der periplasmatischen Seite der Membran gebunden ist. NrfH gehört zur NapC/NirT-Familie und überträgt Elektronen von Menachinol auf NrfA. Mittels gerichteter Mutagenese von nrfH wurden in früheren Arbeiten bereits Aminosäure-Reste identifiziert, die essentiell für die Elektronentransportaktivität von Formiat zu Nitrit sind.
