Typ I Interferone sind bekannt für die durch sie vermittelten immunaktivierenden bzw. antiviralen Effekte. Nach ihrer Induktion, im Rahmen der angeborenen Immunantwort, vermitteln Interferone nicht nur einen systemischen anti-viralen Status, sondern können auch wichtige Effektormechanismen der adaptiven Immunität dahingehend beeinflussen, dass sie diese verstärken bzw. ermöglichen. Im Allgemeinen kann diese Eigenschaft als pro-inflammatorische Aktivität der Interferone bezeichnet werden. Allerdings gehört es ebenfalls zu den Eigenschaften der Interferone eine Verminderung der adaptiven Immunität bewirken zu können, was als anti-inflammatorische Aktivität verstanden werden kann. Insgesamt kann man die durch Interferone induzierten Effekte also als ambivalent bezeichnen.
Die Leber als Immunorgan besitzt, ähnlich wie die Interferone, eine zentrale Rolle in der Immunität und sollte in ihrer Funktion als Vermittler zwischen Immunaktivierung und Immuntoleranz nicht unterschätzt werden. Die Aufgaben der Leber können ebenfalls als ambivalent bezeichnet werden, da sie zum einen eine unnötige Aktivierung des Immunsystems verhindern muss um eine Schädigung der Leberzellen zu vermeiden (Immuntoleranz). Zum anderen muss auch in der Leber eine Immunaktivierung stattfinden können, um den Schutz vor Pathogenen zu gewährleisten.
In einem Leberschadenmodell, das künstliche Doppelstrang-RNA (poly(I:C)) zur Induktion von Typ I Interferonen verwendet, sollen im Rahmen der vorliegenden Arbeit Immunmodulationen, insbesondere in der Leber, untersucht werden. Hierbei liegt das Hauptaugenmerk auf den Interferon-vermittelten Effekten, die eine Schädigung der Leber verhindern.
Werden Interferonrezeptor-defiziente Tiere (IFNAR-/-) intraperitoneal mit poly(I:C) behandelt kann eine ausgeprägte Schädigung der Leber sowie Hepatitis in diesen Tieren beobachtet werden. Wildtyp (WT) Mäuse zeigen hingegen keinerlei Schädigungen der Leber, was für einen protektiven bzw. anti-inflammatorischen Effekt spricht, der über den IFNAR und damit über Typ I Interferone vermittelt wird. Unter Verwendung von Mäusen, die eine selektive Deletion des IFNAR auf bestimmten Immunzellen tragen (alle anderen Zellen der Maus exprimieren jedoch weiterhin den IFNAR), konnte der Immunzelltyp ermittelt werden, der beim IFNAR-vermittelten Schutz der Leber eine Schlüsselrolle übernimmt. Aus diesen Experimenten wird deutlich, dass es myeloide Zellen sind, die über den IFNAR durch Typ I Interferone stimuliert werden müssen, um im poly(I:C)-induzierten Leberschadenmodell einen Schutz der Leber zu bewirken. Ergänzend dazu konnte gezeigt werden, dass CD11b- und F4/80-doppelt positive Makrophagen nach poly(I:C)-Behandlung in die Leber von WT Mäusen infiltrieren. Zudem wurde in Experimenten mit Interferon-Reporter Mäusen deutlich, dass diese infiltrierenden Makrophagen über den IFNAR durch Typ I Interferone stimuliert sind. Nach poly(I:C)-Behandlung konnte gezeigt werden, dass Leber-infiltrierende Zellen in WT Mäusen anti-inflammatorischen Interleukin-1 Rezeptor Antagonisten (IL-1RA) sekretieren. In Abwesenheit eines funktionalen Interferonsystems hingegen (in IFNAR-/- Mäusen) konnte eine gestörte IL-1beta- und IL-1RA-Balance festgestellt werden. Für diese Zytokine, die sich gegenseitig regulieren, indem der anti-inflammatorische IL-1RA mit dem pro-inflammatorischen IL-1beta um die Bindung an den IL-1 Rezeptor konkurriert, konnte gezeigt werden, dass ihre Expression in der Leber Interferon-abhängig reguliert wird. In IFNAR-/- Mäusen und in Mäusen, deren IFNAR selektiv auf myeloiden Zellen deletiert war, konnte keine IL-1RA-Expression durch infiltrierende Zellen detektiert werden. Da in diesen Tieren nach poly(I:C)-Behandlung massive Leberschäden beobachtet wurden, kann vermutet werden, dass das Vorhandensein des anti-inflammatorischen IL-1RA unerlässlich für den Schutz der Leber ist.
Abschließend kann zusammengefasst werden, dass die Interferon-vermittelten Effekte, die eine Schädigung der Leber verhindern, zum einen auf der Stimulation und Rekrutierung von Makrophagen beruhen. Zum anderen beruhen diese Effekte auf der Induktion des anti-inflammatorischen Zytokins IL-1RA, und der dadurch blockierten Wirkung des pro-inflammatorischen IL-1beta.
Durch diese Ergebnisse werden neue Einblicke in die Interferon-vermittelte Hemmung von Virus- und Autoimmun-induzierten Erkrankungen der Leber ermöglicht. Genutzt werden könnten diese für die Optimierung IFN-basierter Therapien. Beispielsweise kann durch die gezielte Induktion anti-inflammatorischer Zytokine über IFNAR-induzierte Signalwege oder die direkte Gabe anti-inflammatorischer Zytokine (z.B. IL-1RA) eine Therapie entwickelt werden, die neben den vorteilhaften Eigenschaften der Zytokine eine verbesserte Aktivierung von Immunzellen ermöglicht.
Im Zentralen Nervensystem (ZNS) kommunizieren neuronale Synapsen über eine Kombination von chemischen und elektrischen Signalen, die in ihrer Umgebung eine spezifische Komposition von Ionen benötigen. Um eine strenge Kontrolle des ZNS-Milieus zu gewährleisten, hat sich in Säugetieren eine endotheliale Blut-Hirn-Schranke (BHS) entwickelt. Die BHS limitiert den parazellulären Molekül Transport und wird von den Kapillargefässen des Gehirns gebildet, wobei die physische Barrier von den Tight Junctions (TJs) des vaskulären Endothels generiert wird. Das Gehirnendothel ist Teil einer neurovaskulären Einheit (NVE), zu der auch Perizyten (PZ), Astrozyten (AZ), Mikroglia und Interneurone zählen. Fehlkommunikation oder defekte zelluläre Komponenten in der NVE führen in der Regel zu Störungen in der BHS Funktion und können schwerwiegende neuronale Erkrankungen zur Folge haben.
Vor einigen Jahren haben wir und andere Forschungsgruppen herausgefunden, dass der Wnt/β-Catenin Signalweg essentiell für die Vaskularisierung des Gehirns während der Embryonalentwicklung ist und darüber hinaus auch eine bedeutende Rolle in der Induktion der BHS spielt. Des Weiteren konnte im Zebrafischmodell eine Aktivierung des kanonischen Wnt Signalweges auch im adulten Organismus nachgewiesen werden. Allerdings ist die Quelle der Wnt Wachstumsfaktoren bis dato unbekannt. Der Wnt Signalweg ist eine hoch konservierte und komplexe zelluläre Signalkaskade, die in allen mehrzelligen Organismen vorkommt. Wnt Wachstumsfaktoren sind sekretierte, hydrophobe Signalmoleküle, die sowohl über lange als auch kurze Strecken entweder den β-Catenin-abhängingen („kanonischen“) oder β-Catenin-unabhängingen („nicht-kanonischen“) Wnt Signalweg aktivieren können.
Da die meisten ZNS Erkrankungen mit einem Zusammenbruch der BHS-Funktion assoziiert sind, ist die Forschung bestrebt die Mechanismen, die der Entstehung und Aufrechterhaltung der BHS zugrunde liegen, zu ermitteln und zu verstehen. Das Ziel meiner Doktorarbeit war es herauszufinden, ob AZ Wnts produzieren und ob deren Wirkung auf das Gehirnendothel an der Aufrechterhaltung der BHS beteiligt ist. Zu diesem Zweck, habe ich ein in vitro BHS Kokultivierungs-Modellsystem etabliert das erstmalig ausschliesslich auf der Verwendung von murinen AZ und Gehirnendothelzellen basiert. Zu Beginn der Studie wurden sowohl primäre AZ als auch eine murine Gehirnendothel-zelllinie (MBE) bezüglich ihrer zell-spezifischen Eigenschaften charakterisiert. Dabei konnte belegt werden, dass sowohl die primären AZ als auch die MBE Zelllinie, aufgrund ihrer Proteinexpressionsprofile als repräsentative Vertreter ihres Zelltyps eingestuft werden können. Die darauffolgenden Untersuchungen konnten zeigen, dass primäre AZ über mehrere Passagen hinweg fast alle 19 Wnt Liganden auf mRNA Ebene exprimierten. Ferner konnte in primären Gehirnendothelzellen und zwei Gehirnendothelzelllinien die korrespondierenden Frizzled (FZD) Rezeptoren und low density lipoprotein receptor-related protein (LRP) Korezeptoren nachgewiesen werden. Dieser Befund legte Nahe, dass AZ und Gehirnendothelzellen die basalen Eigenschaften besitzen, um über den Wnt Signalweg miteinander zu kommunizieren. Die Stimulation von pMBEs mit Astrozyten konditioniertem Medium (AKM) induzierte die Hochregulation von Claudin-3 einem bekannten kanonischen Wnt Zielgens. Interessanterweise konnte diese Regulation teilweise durch die Zugabe von dickkopf 1 (Dkk1), einem Wnt/β-Catenin Antagonisten, inhibiert werden.
Um die physiologische Rolle der Wnt Liganden zu bestimmen, habe ich mir die Eigenschaft des universellen Sekretionsmechanismus der Wachstumsfaktoren, welcher von dem Transmembranprotein evenness interrupted (Evi) abhängig ist, zu Nutze gemacht. Die Verpaarung von Evifl/fl mit hGFAP-Cre Mäusen erlaubt die AZ-spezifische Deletion des Evi Proteins (Evi KO), was zur Folge hat, dass die Astrozyten der Nachkommen keine Wnt Wachstumsfaktoren sekretieren können.
In vitro führte der Verlust von Wnts in AKM zu einer teilweisen Delokalisierung von Junction Proteinen. Während die Kokultivierung mit Evi WT AZ einen straken Anstieg im TEER und reduzierte Permeabilitätsmesswerte induzierten, konnten diese pro-BHS Eigenschaften bei Evi KO AZ nicht beobachtet werden. Diese Ergebnisse zeigten deutlich, dass Wnts sekretiert von AZ den BHS Phenotyp positive beeinflussen, indem sie die Zell-Zell-Verbindung verstärken, was wiederum zu erhöhtem Zellwiderstand und reduzierter transzellulärer Permeabilität führt. Die Analyse des in vivo Phänotyps von Evi KO Mäusen ergab, dass mit fortschreitendem, postnatalem Alter makroskopisch erkennbare zerebrale Blutungen auftraten. Ausserdem konnte ich zeigen, dass eine Subpopulation von Blutgefässen Malformationen aufwies, die mit reduzierter Astrozytenendfuss-Assoziierung einhergingen.
Das Wissen um die Beteiligung des Wnt Signalweges an der Regulation der BHS auch im adulten Organismus kann in Zukunft von wichtiger Bedeutung sein, da es potentielle therapeutische Anwendungen ermöglicht.
Bei Untersuchungen HBV-positiver Zellen konnte zunächst, anders als für HCV, eine deutlich gesteigerte Menge an TIP47 im Western Blot nachgewiesen werden. Da außerdem auch die zellulären mRNA-Spiegel von TIP47 erhöht waren, wurde in Promotorstudien der genaue Regulationsmechanismus untersucht. Für HBV sind zwei wichtige Faktoren bekannt, welche diverse zelluläre Signalkaskaden, wie z. B. die c-Raf/MAP-Kinase-Kaskade, modulieren, die PreS2-Aktivatordomäne des LHBsAg und das HBx-Protein [360]. Diese regulieren via c-Raf die Expression der unterschiedlichsten Gene. Nach eingehenden Analysen lässt sich dazu auch TIP47 zählen, dessen Expression durch HBx und LHBs gesteigert werden kann. Außerdem konnte in CLSM-Analysen eine partielle Colokalisation von LHBs und TIP47 beobachtet werden. Durch Modulation der TIP47-Expression in HBV-positiven Zellen konnte anschließend die Relevanz für die Virus-Sekretion untersucht werden. Durch gezielten knockdown von TIP47 durch spezifische siRNAs wurde die Freisetzung von viralen Partikeln gestört, wohingegen die Menge an freigesetzten subviralen Partikeln erhöht war. Die Überexpression von TIP47 hingegen konnte die Virus-Sekretion steigern, während das Niveau der subviralen Partikel nahezu gleich blieb. Des Weiteren konnte auch für HBV die Rab9-Bindung an TIP47 als essentielle Funktion Charakterisiert werden, da eine Inhibition dieser Interaktion eine Hemmung der Sekretion viraler Partikel zur Folge hatte. Auch hier konnte kein Einfluss auf die subviralen Partikel beobachtet werden. In Studien wurde a-Taxilin als neuer Bindungspartner von Proteinen der Syntaxin-Familie entdeckt. Es spielt daher eine wichtige Rolle im intrazellulären Vesikeltransport. Vor allem die Interaktion mit Syntaxin-4 ist gut untersucht [132]. Es wird vermutet, dass a-Taxilin durch die Bindung an freies Syntaxin-4 die v-SNARE-Bildung verhindert und so einen inhibitorischen Effekt auf den vesikulären Transport ausübt. Des Weiteren konnten Untersuchungen beim Hepatitis-B-Virus demonstrieren, dass die Expression von a-Taxilin durch die Virus-Replikation drastisch erhöht ist und die Sekretion der subviralen Partikel, welche mittels Vesikeln aus der Zelle transportiert werden, negativ beeinflusst. Andererseits interagiert a-Taxilin mit dem großen viralen Oberflächenprotein LHBs und dient so als Adapter zwischen LHBs und tsg101 beim ESCRT-vermittelten Export des Virus via MVBs - einem Zusammenschluss aus vielen späten Endosomen [126].Anders als für HBV, welches aktiv die Menge an intrazellulärem a-Taxilin erhöht, konnte in früheren RNA-Expressionsexperimenten mit transgenen Mäusen, welche Leberspezifisch das regulatorische HCV-Protein NS5A produzieren, eine deutlich verminderte Expression von a-Taxilin beobachtet werden [140]. Durch Analysen von Leberzelllysaten im Western Blot konnte dieser Effekt auch auf Proteinebene bestätigt werden. Dieanschließende Analyse HCV-replizierender Zellen in vitro ergab ebenfalls eine verminderte a-Taxilin-Expression und in der Folge eine reduzierte Proteinmenge. Weiterhin konnte diese Arbeit klären, dass HCV via NS5A den a-Taxilin-Promotor negativ beeinflusst und dafür den bereits für NS5A beschriebenen Mechanismus der c-Raf-Modulation nutzt [234]. Darüber hinaus wird a-Taxilin durch HCV destabilisiert, da in HCV-replizierenden Zellen die Proteinhalbwertszeit von a-Taxilin in etwa halbiert war. Der genaue Mechanismus hierfür muss jedoch noch genauer untersucht werden. Es kann aber aufgrund von anderen aktuellen Studien davon ausgegangen werden, dass a-Taxilin höchstwahrscheinlich durch HCV-Strukturproteine abgefangen wird, welche nicht am Aufbau neuer Virionen beteiligt sind. Diese werden dann, zusammen mit dem gebundenen a-Taxilin, im autophagosomalen Kompartiment recycelt. Gestützt wird diese Hypothese durch die Beobachtungen in CLSM-Analysen, dass die HCV- Strukturproteine E1, E2 und Core partiell mit a-Taxilin colokalisieren und auch durch Co-Immunpräzipitationen sowie yeast-2-hybrid-Analysen eine direkte Interaktion nachgewiesen werden konnte. Dabei konnten vor allem für das Core-Protein zwei unterschiedliche Fraktionen nachgewiesen werden, von denen nur die zytoplasmatisch lokalisierte Fraktion mit a-Taxilin colokalisierte, nicht aber mit dem an den lipid droplets gebundenen Core. Neben der Untersuchung der funktionellen Zusammenhänge wurde außerdem die Relevanz von a-Taxilin für den HCV-Lebenszyklus charakterisiert. Dabei wurde die Expression von a-Taxilin moduliert und der Einfluss auf die Freisetzung infektiöser HCV-Partikel untersucht. Durch die Überexpression von a-Taxilin konnte die Sekretion von Virionen verhindert werden, wohingegen die weitere Reduktion der a-Taxilin-Menge mittels spezifischer siRNA zu einer verstärkten Virus-Freisetzung führte. In einem parallel durchgeführten Projekt konnten durch die Modulation von Syntaxin-4 genau gegenteilige Beobachtungen gemacht werden. Demnach verstärkte eine Überexpression von Syntaxin-4 die HCV-Sekretion, während der knockdown zur Inhibition des Prozesses führte. Abschließend lässt sich festhalten, dass im Rahmen dieser Arbeit zwei zelluläre Proteine in Bezug auf die Morphogenese und Sekretion von HBV und HCV näher Charakterisiert wurden, denen zuvor für das jeweils andere Virus eine entscheidende Rolle im viralen Lebenszyklus zugeordnet werden konnte. TIP47 wurde somit als positiver Regulator für die HBV-Sekretion identifiziert, auch wenn die genaue funktionelle Relevanz bzw. der Funktionsmechanismus bisher noch nicht eindeutig geklärt werden konnte. So liegt jedoch der Schluss nahe, dass es nur die Freisetzung der viralen Partikel via MVBs beeinflusst und nicht an der Sekretion der subviralen Partikel beteiligt ist. Für HCV konnte mit a-Taxilin erstmals ein viraler Restriktionsfaktor beschrieben werden, da es entscheidend die Sekretion infektiöser Viruspartikel verhindert. Im Gegenzug hat HCV, durch die Deregulation des Promotors und durch das Abfangen von a-Taxilin, Mechanismen entwickelt, welche diesem restriktiven Effekt entgegen wirken.
In order to investigate the diversity of the western honeybee, Apis mellifera L., in West and Central Africa, a total of 204 colonies were sampled from 44 localities in four countries – Nigeria, Niger, Cameroon and Chad. 86 of these colonies, from 23 localities, were subjected to full morphometric analysis. In a principal component analysis (PCA) of the morphometric data, the colonies formed a single cluster. It also revealed that overall size of the body was the most important source of variation between the colonies. A hierarchical structure analysis, followed by a stepwise discriminant analysis, classified the colonies into three distinct morphoclusters; however, these clusters were not geographically demarcated. In another PCA carried out with the samples under investigation and reference samples of A. m. adansonii, A. m. jemenitica and A. m. scutellata, the colonies under investigation again formed one cluster which lying over and extended beyond the clusters of the reference subspecies. This is suggestive of a wider variation in size in the bees under investigation. In a stepwise DA, 94.2% of cross-validated grouped cases were correctly classified and the distances between group centroids were highly significant (p < 0.0005) according to F-statistic. 61 and 22 of the 83 colonies under investigation were assigned to A. m. jemenitica and A. m. adansonii, respectively. Mitochondrial DNA analysis was carried out on 148 colonies from 39 localities. Four mitochondrial haplotypes, previously reported from Africa and belonging to the African mitochondrial lineage, A, were detected: A1 (n = 62), A4 (n = 70), A4' (n = 15) and A14 (n = 1). The overall haplotype diversity was low (h = 0.478 ± S. E. 0.057). A chi-square test for association was conducted between haplotypes and type of vegetation, latitude, longitude, altitude, temperature and rainfall, severally. There was a statistically significant association between haplotype and each of the six variables and the association was strong with latitude, moderate with vegetation and rainfall and weak with the remaining variables. The neighbour-joining, maximum likelihood and maximum parsimony trees, obtained from sequence variation of the cytochrome b gene of mitochondrial DNA, showed that the samples, from the current study, unambiguously clustered with the reference sequences of A. m. scutellata from Kenya, but without showing further subdivision within this sub-Saharan cluster. 133 workers (one per colony) collected from 38 localities were subjected to microsatellite analysis. A total of 292 different alleles were recorded for the 15 microsatellite loci used. All microsatellite loci were polymorphic and the number of different alleles per locus ranged between 10, in locus At163, and 31, in locus A029. Heterozygosity (or gene diversity) was high in all loci. The unbiased expected heterozygosity, which is a better expression of gene diversity, was 0.861 ± S.E. 0.017. The overall FST value, which is a good estimate of genetic differentiation of populations, was very low: 0.007 ± S.E. 0.001 (0.001 - 0.014). AMOVA and Bayesian assignment showed no differentiation of the investigated populations. Based on morphometric analysis, the results of this study present the honeybees of western Africa as a single entity with an internal variation which lacks a geographical demarcation. Consequently the results do not support the splitting of the honeybees of the region into the two subspecies, A. m. adansonii and A. m. jemenitica, as reported in the literature. More morphometric, molecular, physiological and behavioural studies are required to confirm the taxonomic status of the honeybees of the region. Meanwhile, the use of A. m. adansonii, as the sole sub-specific name for the honeybees of West and Central Africa, is recommended.
Protein translocation across the chloroplast membrane is mediated by molecular machinery composed of protein complexes termed the TOC/TIC (the outer/inner envelope chloroplasts translocases). This translocation process is regulated by metabolic energy in form of GTP and ATP and is influenced by the lipid composition of the membrane. The ability to study the function of a single complex “TOC” in vitro using purified protein or purified chloroplast outer envelope vesicles has been instrumental for our understanding of the mechanism underlying this process.
Indeed, the TOC complex has been purified by previously established procedures. However its functional and structural analyses are impaired by the limited yield of purified protein. Therefore, protocols for native TOC complex purification are described here. The complex isolation is achieved by direct biochemical treatment of biological membrane hosting this complex or by tandem affinity purification of modified protein complex components from generated transgenic plants.
Furthermore, in this thesis, radioactive based in vitro import assays are described, namely those that allow monitoring translocation activity across the outer envelope of chloroplast. Based on the analysis of knock-out plants and isolated complexes it was previously suggested that lipid dependence of protein translocation might exist. Thus, the question was raised whether the lipid composition of the membrane has a direct influence on the behavior and functionality of the TOC translocon, or whether additional components of the chloroplast membrane account for the observed effect in vivo. To answer this question, a technique for vesicle fusion was developed. The principal aim was to explore the effect of an exchange of the lipid environment surrounding the complex translocon. This method helped to demonstrate that the SQDG and PI act stimulatory on the translocation across the outer envelope of chloroplast, whereas DGDG exhibits an inhibitory effect on TOC complex functionality.
CAP (c-Cbl assoziiertes Protein) ist ein Adapterprotein, welches zusammen mit ArgBP2 und Vinexin die SoHo-Protein-Familie bildet. Es besitzt in seinem N-terminalen Bereich eine SoHo-Domäne und im C-Terminus drei SH3-Domänen, über welche es mit einer Vielzahl von Proteinen interagieren kann. CAP spielt bei verschiedenen Signaltransduktionsvorgängen und der Reorganisation des Aktinzytoskelettes eine Rolle. So wurde ihm eine Funktion im PI3-Kinase-unabhängigen Insulinsignalweg zugeschrieben. In Zell-Zell-Kontakten ist CAP zusammen mit Nectin und Afadin Bestandteil des NAPSystems, welches parallel zu Cadherin-Catenin-Adhäsionen existiert. In Fokalkontakten bindet CAP an FAK, Paxillin und Vinculin, welche wichtig für die Regulation der Zell-Matrix-Adhäsionen sind. In der vorliegenden Arbeit sollte die Funktion von CAP bei der Assoziation mit dem Aktinzytoskelett näher untersucht werden. Es wurde gezeigt, daß die Kolokalisation von CAP mit Vinculin und Aktin dynamisch und vom Ausbreitungsgrad der Zelle und dem Expressionsniveau des Proteins abhängig ist. Ohne funktionelle SH3-Domänen lokalisiert CAP nicht mehr in Fokalkontakten, kann aber bei Überexpression noch eine Ausbildung von Streßfasern induzieren. Die Funktionalität der zweiten und der dritten SH3-Domäne von CAP ist für die Zelladhäsion von Epithelzellen von Bedeutung, da Konstrukte, die in konservierten Aminosäuren der Domänen mutiert worden sind, eine verringerte Zellausbreitung aufweisen. CAP wurde zudem als ein Interaktionspartner und Substrat der Tyrosinkinasen c-Abl und c-Src charakterisiert. Die Assoziation mit den Kinasen wird dabei vor allem über den C-Terminus von CAP vermittelt, und CAP kann direkt mit der Src-Kinase interagieren. CAP wird von c-Abl überwiegend an Tyrosin 360, und von c-Src bevorzugt an Tyrosin 326 phosphoryliert. Diese Tyrosine liegen innerhalb bekannter Konsensus-Motive der Kinasen. Phosphorylierungsdefiziente CAP-Konstrukte sind noch dazu in der Lage, mit Vinculin und Aktin zu kolokalisieren. Tyrosin 326 hat jedoch einen Einfluß auf die Zellausbreitung auf Fibronektin, da ein in dieser Aminosäure mutiertes Konstrukt einen Defekt hierbei aufweist. Den genauen Mechanismus, über welchen CAP seinen Einfluß auf die Zell-Matrix-Adhäsion ausübt, ist noch ungeklärt. Durch die hier erstmals aufgezeigte Phosphorylierung des Adapterproteins erweitern sich dessen Interaktionsmöglichkeiten durch mögliche Bindungen an SH2-Domänen-enthaltende Proteine. CAP könnte als Bindeglied zwischen c-Abl und c-Src und weiteren zytoskelettalen Komponenten dazu beitragen, die Kinasen an Fokalkontakten zu verankern und ihnen neue Substrate zuzuführen. Die Interaktion mit CAP könnte dabei die Aktivität der Kinasen erhöhen. Weiterführende Studien werden die in diesem Signalweg nachgeschalteten Komponenten untersuchen und auch eine putative Rolle der Phosphorylierung von CAP in den bereits bekannten zellulären Zusammenhängen wie dem Insulinsignalweg näher beleuchten.
Chemical contamination of the environment and thus of aquatic ecosystems is steadily increasing. Whenever environmental pollutants enter a water body, they affect not only the water, but also the sediment. Substances that bind to sediment particles can be stored for a long time, whereby sediments act as sinks for some contaminants. Therefore, sediment
assessments often more accurately describe the contamination of a water body than investigations of the water itself. Among environmental chemicals, endocrine disrupting compounds (EDCs) have gained more and more attention in recent years. Since they interfere with endocrine systems and may disturb reproduction, they endanger the survival of populations or even species. Hazardous substances enter the aquatic environment by different pathways, with sewage treatment plants (STPs) belonging to the most important contamination sources.The main objective of this work is a comprehensive sediment assessment of predominantly small surface waters in the German federal state of Hesse. The 50 study sites, located in 44 different creeks and small rivers, are situated in the densely populated and economically important Frankfurt/Rhine-Main area, as well as in rural and less urbanized regions.
Chemical analytical data, provided by the Hessian Agency for the Environment and Geology (HLUG), indicated different contamination levels of the study sites. In order to investigate the general toxicity of the sediment samples, the oligochaete Lumbriculus variegatus and the midge Chironomus riparius were exposed to whole sediments and apical endpoints regarding biomass, survival, and reproduction were determined. In further experiments, special attention was paid to the contamination with endocrine active compounds. For this purpose, the reproductive success of the New Zealand mudsnail Potamopyrgus antipodarum was analyzed after exposure to whole sediments. Additionally, a yeast-based reporter gene assay was applied with sediment eluates to assess the estrogenic and androgenic activity of the samples. Biotest results were compared with chemical analysis data to investigate whether the test organisms reflect the measured pollution of the study sites and if the observed effects can be explained by chemical contamination.
Five study sites, all located less than 1 km downstream of a STP discharger, were selected for further investigations based on the results of the sediment monitoring. The sediments from these sites were conspicuous due to their general toxic and/or estrogenic activity. In order to investigate whether the observed effects can be ascribed to the effluents, an active biomonitoring study was conducted with the mudsnail P. antipodarum and the zebra mussel Dreissena polymorpha, exposed at study sites located up- and downstream of the discharger.
In addition to endocrine activity, genotoxic effects were investigated using the comet assay and the micronucleus assay. Endocrine activity was examined based on the reproductive output of P. antipodarum and the content of vitellogenin-like proteins in D. polymorpha. Yeast-based reporter gene assays were used to estimate the endocrine potential (estrogen, anti-estrogen, anti-androgen, dioxin-like) of sediment and water samples.
22% of the 50 sediments showed ecologically relevant effects in the biotests with L. variegatus and C. riparius. Only one sediment caused a relevant effect on both test organisms, while the other ten positively tested sediments affected either L. variegatus or C. riparius, probably due to differences in inter-species sensitivities. This suggests that a combination of different biotests is necessary for a comprehensive evaluation of sediment toxicity. 78% of the sediments caused a significantly increased number of embryos in P. antipodarum, which could be ascribed to estrogenic contamination of the sediment samples. An increase in the number of embryos by 60%, as observed in this study, and an associated increase in population size may result in the displacement of other, less competitive species.
In the in vitro tests, 66% of the sediments showed estrogenic activity and 68% showed androgenic activity. Maximum observed values were 40.9 ng EEQ/kg sediment (EEQ = estradiol equivalent) for estrogenic and 93.4 ng TEQ/kg sediment (TEQ = testosterone equivalent) for androgenic activity. Natural and synthetic hormones as well as alkylphenols were the major contributors to the total estrogenicity of environmental samples in several other studies, and are likely responsible for a large part of the estrogenic activity in this case as well. Similarly, androgenic activity is mainly due to natural steroids and their metabolites.
Bioassay results reflect the analytically measured contamination levels at the study sites only very infrequently. This can be ascribed to the occurrence of integrated effects of chemical mixtures present in the sediments. Additionally, effects of substances not included in the analytical program or of substances present in concentrations below the detection limit of the chemical analytical investigations as well as varying bioavailabilities might be relevant. The fact that a large part of the observed effects cannot be explained by the chemical contamination demonstrates the need for effect studies in ecotoxicological sediment assessments.
In order to identify possible causes for the effects observed in the sediment monitoring, e.g. contamination sources, the area types (urban fabrics, arable lands, pasturages, etc.) of the catchment areas belonging to the study sites were analyzed. No significant differences were found between the area profiles of the sampling sites with and without effects in the biotests.
The results indicate that the contamination responsible for the observed effects can be ascribed to different sources. Furthermore, study sites whose sediments exerted significant effects in biotests were located in anthropogenic as well as in predominantly natural areas. The active biomonitoring study at STPs revealed genotoxic and endocrine effects only sporadically.
However, in the in vitro tests considerable endocrine activities of sediment and water samples were determined. No conclusive picture emerges as to whether the observed effects occur more frequently downstream of the dischargers, and thus could be attributed to a contamination by sewage. This indicates that contamination sources other than STP dischargers, for example agricultural runoff, may contribute to the observed effects. Weaker effects and biological activities downstream of a discharger compared to an upstream site might be ascribed to a dilution effect by the effluents. A comparison of the measured in vitro estrogenicity with exposure studies described in the literature shows that adverse effects in aquatic organisms can be expected at the EEQ concentrations determined in the present study.
The results of the sediment monitoring and the STP study revealed a widespread endocrine pollution of small surface waters in Hesse. The fact that the bioassay results only rarely reflect study site contamination as determined by chemical analysis demonstrates the need for effect studies in comprehensive sediment assessments. In some cases STP dischargers increased, in other cases they decreased the observed in vivo effects and in vitro activity of environmental samples. Transferring the results obtained in laboratory studies to the field, adverse effects on aquatic ecosystems can be expected. The study illustrates the need for restrictive measures that contribute to the removal or reduction of environmental pollutants.
For the identification of substances that have so far not been linked to adverse effects on the environment, methods such as effect-directed analyses (EDA) or toxicity identification evaluation (TIE) should be increasingly applied in future studies. Furthermore, bioassays for the assessment of endocrine activity should be implemented in standardized monitoring programs.
Clathrin-mediated endocytosis (CME) involves spatially and temporally restricted molecular dynamics.
Although protein kinases and the actin cytoskeleton contribute to the process, whether and how
functions of kinases and actin are integrated remains unknown. Here, we demonstrate that neural
Wiskott-Aldrich syndrome protein (N-WASP) and protein kinase CK2 form a complex and localize on
clathrin-coated vesicles (CCVs). N-WASP binds to and is phosphorylated by CK2, thereby reducing the
kinase activity of CK2. By contrast, N-WASP-promoted actin polymerization is decreased upon both
phosphorylation and binding of CK2. Knockdown of N-WASP and CK2, alone or in combination, results
in impaired endocytosis of epidermal growth factor (EGF) and increased cell-surface levels of EGF
receptor (EGFR). In order to rescue the phenotype of N-WASP-CK2 knockdown cells, both N-WASP and
CK2 activities and abilities to assemble in a complex are required. In summary, this study shows that the
N-WASP-CK2 complex integrates in a single circuit different activities contributing to CME of EGFR and
that the interplay between the two proteins optimizes this process.
Synaptic plasticity is the basis for information storage, learning and memory and is achieved by modulation of the synaptic transmission. The amount of active AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazol-propionic acid) receptors at the synapse determines the transmission properties, therefore the regulation of AMPA receptor trafficking affects the synaptic strength. The protein GRIP (glutamate receptor interacting protein) binds to AMPA receptors and is one of the important regulators of AMPA receptor stability at the synapse (Dong et al., 1997; Osten et al., 2000). Previous studies have shown that the ablation of ephrinB2 or ephrinB3 in the nervous system leads to severe defects in hippocampal LTP (long term potentiation) and LTD (long term depression) (Grunwald et al., 2004). We found that ephrinB2 ligands play an important role in the stabilization of AMPA receptors at the cellular membrane (Essmann et al., 2008). Treating cultured hippocampal neurons with AMPA resulted in a robust AMPA receptor internalization, which could be inhibited by simultaneous ephrinB2 activation with soluble EphB4-Fc fusion proteins. Conditional hippocampal ephrinB2 knock-out (KO) neurons showed enhanced constitutive internalization of AMPA receptors. Interaction and interference experiments revealed that ephrinB ligands and AMPA receptors are bridged by GRIP. This interaction is regulated by phosphorylation of a single serine residue in close proximity to the C-terminal PDZ protein target site in ephrinB ligands (Essmann et al., 2008). To investigate the in vivo relevance of this previously undescribed feature of ephrinB reverse signaling, we generated ephrinB2 S-9>A knock-in mice, where the serine at position -9 was replaced by an alanine to prevent phosphorylation. The mutated ephrinB2 of this mouse line was expressed and able to form clusters following stimulation with the preclustered receptor EphB4-Fc. Surface ephrinB2 cluster size and cluster number was slightly smaller in comparison to wild type (WT) mice. Analyzing AMPA receptor internalization, we oserved an increased basal GluR2 endocytosis in cultured hippocampal neurons of ephrinB2 S-9>A mice. Dendrite and spine morphology was similar in pyramidal CA1 neurons of brain slices from adult ephrinB2 S-9>A and WT mice, suggesting a redundancy between the different ephrinB familily members.
Apart from regulating AMPA receptor stability at the synapse, GRIP1 also has an important role in the secretory pathway to deliver cargo proteins along microtubules to dendrites and synapses (Setou et al., 2002). Proteins involved in synaptic transmission and plasticity, as well as lipids required for the outgrowth and remodeling of dendrites and axons have to be transported. We showed in our laboratory with a directed proteomic analysis using the tandem affinity purification-mass spectrometry methodology (Angrand et al., 2006) and with immunoprecipitation assays with brain lysates that the small regulatory protein 14-3-3 interacts with GRIP1. Further immunoprecipitation assays with lysates from HeLa cells transfected with various parts and sequence mutants of GRIP1 revealed that threonine 956 in the linker region L2 between PDZ6 and PDZ7 of GRIP1 is necessary for the interaction with 14-3-3. GRIP1 has been postulated to influence dendritic arborization and maintenance in hippocampal neurons in culture due to defective kinesin-dependent transport along microtubules (Hoogenraad et al 2005). In order to address the role of the association of GRIP1 and 14-3-3 in dendritogenesis, we transfected rat hippocampal neurons with GRIP1-WT and GRIP1 mutants and performed Sholl analysis to evaluate dendritic arborization defects. We could observe striking increased formation and growth of dendrites in developing neurons as well as in mature neurons overexpressing GRIP1-WT. However, overexpression of GRIP1-T956A, where the threonine 956 was replaced by an alanine to prevent phosphorylation, did not show enhanced dendritogenesis, indicating a role for threonine 956 phosphorylation in dendrite branching. To investigate the importance of the interaction between GRIP1 and 14-3-3 in vivo, we generated transgenic mouse lines with a GRIP1-T956A transgene or a GRIP1-WT transgene as control. These mice were crossed with heterozygous GRIP1 mice and by further breedings we obtained some surviver mice carrying either the wild type or the mutated GRIP1 transgene in the usually embryonic lethal GRIP1-KO background (Bladt et al., 2002; Takamiya et al., 2004). In embryonic day (E) 14.5 cultured hippocampal GRIP1-KO neurons we could observe reduced dendritic growth. We also showed reduced GluR2 staining on the dendritic surface in cultured hippocampal neurons from GRIP1-KO and GRIP1-KO neurons containing the GRIP1-T956A transgene. GRIP1-KO neurons containing the GRIP1-WT transgene showed a similar surface GluR2 signal intensity as WT neurons. Reduced surface GluR2 staining in GRIP1-KO neurons and GRIP1-KO neurons with the GRIP1-T956A transgene might be a consequence of defective kinesin-dependent transport of GluR2 to dendrites, indicating an important role of threonine 956 phosphorylation of GRIP1 for GluR2 trafficking.
Due to recent technical developments, it became evident that the mammalian transcriptome is much more complex than originally expected. Alternative splicing(AS) and the transcription of long non-coding RNAs (lncRNAs) are two phenomenas which have been greatly underestimated in their frequency. Nowadays it is accepted that almost every gene has at least one alternative isoform and the number of lncRNAs exceeds the one of protein-coding genes.
We built user-friendly web interfaces which can process Affymetrix GeneChip Exon 1.0 ST Arrays (exon arrays) and GeneChip Gene 1.0 ST Arrays (gene arrays)for the analysis of alternative splicing events. Results are presented with detailed annotation information and graphics to identify splice events and to facilitate biological validations. Based on two studies using exon arrays, we show how our tools were used to profile genome-wide splicing changes under silencing of Jmjd6 and under hypoxic conditions. Since gene arrays are not intended for AS analysis originally, we demonstrated their applicability by profiling alternative splicing events during embryonic heart development.
To measure lncRNAs expressions with exon arrays, we completely re-annotation all probes and built a lncRNA specific annotation. To demonstrate the applicability of exon arrays in combination with our annotation, we profiled the expression of tens of thousands of lncRNAs. Further, our custom annotation allows for a detailed inspection of lncRNAs and to distinguish between isoforms, as we validated by RTPCR.
To allow for a general usage to the research community, we integrated the annotation in an easy-to-use web interface, which provides various helpful features for the analysis of lncRNAs.
In der vorliegenden Arbeit konnte die β-Carotin-Ketolase aus dem Cyanobakterium Synechocystis PCC 6803 nach heterologer Expression in E. coli gereinigt und enzymatisch charakterisiert werden. Die Funktion der β-Carotin-Ketolase wurde in vivo durch Komplementierung von β- Carotin-produzierenden E. coli-Transformanten überprüft. Die β-Carotin-Ketolase agierte hier auch als Diketolase und synthetisierte sowohl Echinenon als auch Canthaxanthin. Untersuchungen der Substratspezifität der β-Carotin-Ketolase in vivo und in vitro ergaben, daß nur Carotinoide erkannt werden, die einen β-Iononring ohne Hydroxygruppe in Position C3 aufwiesen. So wurden die Carotinoide β-Carotin, Echinenon, β-Cryptoxanthin und α-Carotin als Substrate erkannt und zu Echinenon, Canthaxanthin, 3’-Hydroxyechinenon und 4-Keto-α-Carotin umgesetzt. Zeaxanthin, 3’-Hydroxyechinenon, 4-Ketozeaxanthin sind keine Substrate der β-Carotin-Ketolase. Die β-Carotin-Ketolase kann einen ε-Iononring, wie in α-Carotin, nicht modifizieren. Die β-Carotin-Ketolase mit einer apparenten Molmasse von 61 kDa wurde durch pPQE30crtO und pPEU30crtO als rekombinantes Polypeptid mit sechs N-terminalen Histidinen in E. coli heterolog exprimiert. Die kinetischen Parameter der β-Carotin-Ketolase konnten in in vitro-Enzymaktivitätstests bestimmt werden. Der KM-Wert für das Substrat β-Carotin lag bei 41,6 μM und der dazugehörende Vmax-Wert bei 1,318 μmol mg-1 h-1. Für das Substrat Echinenon wurde ein KM-Wert von 35,3 μM und ein Vmax-Wert von 0,339 μmol mg-1 h-1 ermittelt. Die Spezifität der β-Carotin-Ketolase war für β-Carotin dreimal höher als für Echinenon. Es konnte keine Kofaktorabhängigkeit nachgewiesen werden, aber eine starke Abhängigkeit der β-Carotin-Ketolase von molekularem Sauerstoff. Die Zugabe des Detergenz Nonidet P-40 in in vitro-Enzymaktivitätstests erhöhte die enzymatische Aktivität der β-Carotin-Ketolase deutlich. Durch die Metallionen-Affinitätschromatographie konnte das Enzym annährend zur Homogenität (93%) unter Erhalt seiner enzymatischen Aktivität gereinigt werden. Dabei blieb die enzymatische Aktivität der β-Carotin-Ketolase nicht nur erhalten, sondern steigerte sich im Vergleich zur Aktivität in der cytosolischen Fraktion um den Faktor 4,5. Die funktionelle Expression der β-Carotin-Ketolase in höheren Pflanzen Nicotiana tabacum, N. tabacum CrtZ Linie U3, N. glauca und Solanum tuberosum Baltica 47-18 erfolgte unter der Kontrolle des konstitutiven CaMV 35S-Promotors. Außer in der Transformante N. glauca CrtO schien die Integration der Ketolase in das Genom der Pflanzen und die Expression von CrtO die Fitness der Transformanten, gemessen am Chlorophyllgehalt und der photosynthetischen Effizienz, nicht negativ beeinflußt zu haben. Der Gesamtcarotinoidgehalt in den Blättern von N. tabacum CrtO und N. tabacum CrtZ Linie U3 CrtO änderte sich trotz der Integration des crtO-Gens kaum im Vergleich zu den Wildtypen. In Blättern von S. tuberosum Baltica 47-18 CrtO konnte eine leichte Erhöhung des Gesamtcarotinoidgehaltes beobachtet werden. Dagegen kann es in Blättern von N. glauca CrtO zu einer Verdoppelung des Carotinoidgehaltes bei gleichzeitig halbiertem Chlorophyllgehalt. In den Blättern akkumulierten Ketocarotinoide mit Anteilen von 5% in N. tabacum CrtO, 12% in S. tuberosum Baltica 47-18 CrtO, 18% in N. tabacum CrtZ Linie U3 CrtO und 16-33% in N. glauca CrtO. Die Anteile der synthetisierten Ketocarotinoide setzten sich in den Blättern von N. tabacum CrtO und N. tabacum CrtZ Linie U3 CrtO aus Echinenon, 3’-Hydroxyechinenon und Ketolutein zusammen, während in Blättern von N. glauca CrtO und S. tuberosum Baltica 47-18 CrtO Echinenon, 3’-Hydroxyechinenon und 4-Ketozeaxanthin enthalten waren. In Nektarien von N. tabacum CrtO und N. tabacum CrtZ Linie U3 CrtO wurde der Gesamtcarotinoidgehalt verdoppelt bis verdreifacht im Vergleich zu den Nektarien des entsprechenden Wildtyps. Es akkumulierten Echinenon, 3’-Hydroxyechinenon, 4-Ketozeaxanthin und Ketolutein. Dabei enthielten die Nektarien von N. tabacum CrtZ Linie U3 CrtO die meisten Ketocarotinoide. Die Nektarien von N. glauca CrtO enthielten deutlich weniger Ketocarotinoidanteile, obwohl der Gesamtcarotinoidgehalt fast dreimal so hoch ist wie in N. tabacum CrtO und N. tabacum CrtZ Linie U3 CrtO. Es akkumulierten nur Echinenon, 3’-Hydroxyechinenon und Ketolutein. Die anderen Blütenorgane von N. glauca CrtO wiesen deutlich höhere Anteile an Ketocarotinoiden auf, zeigten aber wie die Nektarien gegenüber dem Wildtyp keine deutlichen Unterschiede im Gesamtcarotinoidgehalt. Für die Synthese von Astaxanthin war eine Interaktion zwischen der β-Carotin-Hydroxylase und der β-Carotin-Ketolase von entscheidender Bedeutung. Die Akkumulation von „Intermediaten“ der Astaxanthin-Biosynthese in N. tabacum CrtO, N. tabacum CrtZ Linie U3 CrtO und N. glauca CrtO wies auf eine erfolgreiche Interaktion hin. Einzig in den Knollen der CrtO-Transformanten von S. tuberosum Baltica 47-18 konnte Astaxanthin mit einem Anteil von 2% am Gesamtcarotinoidgehalt nachgewiesen werden. Die Nektarien N. tabacum CrtZ Linie U3 CrtO erwiesen sich neben den Knollen als am besten für die Produktion von ketolierten und hydroxylierten Carotinoiden. Die Transformation von N. glauca mit einem Gen der Carotinoidbiosynthese wurde in der vorliegenden Arbeit erstmals durchgeführt, zeigte aber nicht die erwartete Produktion größerer Mengen an Ketocarotinoiden in Kronblättern. Außerdem ist es in der vorliegenden Arbeit erstmals gelungen, in der Kartoffelknolle durch die Einführung einer cyanobakteriellen β-Carotin-Ketolase die Biosynthese von Ketocarotinoiden zu etablieren, um das für die Ernährung wichtige Ketocarotinoid Astaxanthin zu akkumulieren.

Due to its global significance related to food, energy and material production, photosynthesis research is one of the leading scientific fields in the contemporary world. Particular interest in photosynthesis research is focused on diatoms and as one of the major players of marine phytoplankton, diatoms have a huge impact on global photosynthesis.
Diatoms originated from a secondary endosymbiosis that took place between a putative photosynthetic red algal ancestor and a heterotrophic eukaryote. Secondary endosymbiosis resulted in the formation of chloroplasts with four membranes. Centric diatoms (e.g. Thalassiosira pseudonana or Cyclotella meneghiniana) usually possess many small chloroplasts, while pennates (e.g. Phaeodactylum tricornutum) have several larger ones, or even only one which can occupy half of the cell volume...
Die Chemokinrezeptoren CXCR3 und CXCR4 sowie deren spezifische Liganden, CXCL9, -10 und -11 bzw. CXCL12, sind in bedeutender Weise an den pathologischen Prozessen der Th1-/Th17-vermittelten (Typ1- und Typ17-T-Helferzelle) Autoimmunerkrankungen beteiligt. Die dabei auftretenden chronischen Entzündungen sind gekennzeichnet durch eine massive Infiltration von Th1-Gedächtniszellen. Ergebnisse sowohl von tierexperimentellen Studien als auch von in vitro Experimenten weisen deutlich auf eine spezifische Wechselwirkung zwischen den proentzündlichen CXCR3- und dem homöostatischen CXCR4-Liganden hin. Weiterführenden Ergebnisse zu der molekularen Wechselwirkung von CXCR3 und -4 wurden jedoch bislang nicht veröffentlicht. Die Untersuchungen dieser Dissertation konzentrierten sich auf die Kooperation der beiden Chemokinrezeptoren in murinen Th1-Gedächtniszellen. Dabei sollte insbesondere der potentielle Einfluss dieser Interaktion auf die einzelnen Teilprozesse der Extravasation der T-Lymphozyten in vitro analysiert werden. Eingesetzt wurden hierfür statische Chemotaxis- und dynamische Flusskammerexperimente, die zum einen sensitiv genug und zum anderen für einen hohen Probendurchsatz geeignet sein mussten. Die verwendeten Techniken wurden dazu im Rahmen der Dissertation etabliert und validiert. Zunächst musste die Präzision des statischen Migrationssytems mit einer hohen Standardabweichung von durchschnittlich ± 40% deutlich verbessert werden. Ein Wechsel auf ein Kammersystem der Firma Corning verringerte die Abweichung auf ± 25%, und sogar auf ± 9,9% bei einer optimierten Auswertung mittels Durchflusszytometrie. Als weitere Methode wurde ein dynamisches Flusskammersystem mit automatischer Videoanalyse zur Bestimmung der Geschwindigkeit von Zellen etabliert. Zur Validierung der neu entwickelten Analysesoftware Imagoquant® wurden identische Filmaufnahmen von Flusskammerexperimenten hinsichtlich Zellrollen und Zellgeschwindigkeit ausgewertet und mit den Ergebnissen von zwei etablierten Methoden, der Handzählung und einem halbautomatischen Tracking-Programm, verglichen. In der gesamten Validierung stimmten die Berechnungen von Imagoquant® mit Ergebnissen der verschiedenen Auswertemethoden qualitativ überein, wobei die Filme um ein Vielfaches (16- bzw. 20-fach) schneller analysiert werden konnten als mit den bisher verwendeten Methoden. Somit konnte erfolgreich eine computergestützte Analysemethode validiert und etabliert werden, die schnell und benutzerunabhängig arbeitet und folglich objektive Daten im Hochdurchsatz generiert. Die Untersuchungen in den statischen und dynamischen Migrationssystemen ergaben, dass die Stimulation von Th1-Zellen mit CXCL9 zu einer heterologen Desensitivierung verschiedener CXCL12-vermittelter Effekte führt. In statischen Migrationsexperimenten wurde sowohl durch eine synchrone als auch eine sequentielle Stimulation mit CXCL9 eine CXCL12-vermittelte Chemotaxis signifikant vermindert. Der auftretende Effekt war dabei lang anhaltend und konnte noch bei einer Stimulationsdauer von 20 h beobachtet werden, ohne an Intensität zu verlieren. Weitere funktionelle Experimente erfolgten in dynamischen Flusskammerexperimenten, um die desensitivierende Wirkung von CXCL9 auf CXCL12-abhängige Interaktionen der Adhäsionskaskade von Th1-Zellen zu untersuchen. In mit E-Selektin und ICAM-1 Fc-Chimära) beschichteten Flusskammern führte immobilisiertes CXCL12 zu vermehrtem integrinabhängigen Rollen, welches durch eine Vorinkubation der Th1-Zellen mit CXCL9 reduziert wurde. In Flusskammern mit murinen Endothelzellen bewirkte immobilisiertes CXCL12 eine rasche integrinabhängige Adhäsion der Zellen und verkürzte dadurch deren Rollphase signifikant. Eine Vorbehandlung der Zellen mit CXCL9 verminderte dagegen die CXCL12-vermittelte Adhäsion und führte damit zu längeren Rollphasen. Deutliche Effekte zeigte CXCL12 bezüglich einer gesteigerten intravasalen und transendothelialen Migrationsrate von T-Lymphozyten, die durch eine Vorstimulation mit CXCL9 aufgehoben wurden. Um die beteiligten Mechanismen dieser Desensitivierung zu entschlüsseln, wurde die Oberflächenexpression von CXCR3 und CXCR4 in dem Th1-Zellklon durchflusszytometrisch analysiert. Dabei zeigte sich, dass eine Stimulation mit CXCL9 neben der ligandenspezifischen Internalisierung von CXCR3 auch eine Kreuzregulation der CXCR4-Oberflächenexpression bewirkte. Im Weiteren wurde die Phosphorylierung bekannter Signalmoleküle der CXCR4-Signalwege analysiert. Eine Vorbehandlung der Zellen mit CXCL9 desensitivierte die CXCL12-induzierte Phosphorylierung von Akt signifikant und führte zu einer zeitlichen Modulation des Signals. Ferner verzögerte eine Vorbehandlung der Th1-Zellen mit CXCL9 das CXCL12-induzierte Calciumsignal erheblich, während dabei eine 3,5-fach höhere maximale Ca2+-Konzentration gemessen wurde. Ein abgeleiteter Mechanismus der CXCL9-abhängigen Desensitivierung von CXCR4-Signalwegen beeinflusst insbesondere die Signaltransduktion über den T-Zellrezeptor und dadurch auch die Regulation von Rac1. Des Weiteren führt CXCL9 zur Gi- oder ZAP-70-vermittelten Aktivierung der PKC, welche darauffolgend den CXCR4-Rezeptor phosphoryliert und damit zu dessen Internalisierung führt. Die in vitro beobachtete Desensitivierung verschiedener CXCL12/CXCR4-vermittelter Effekte durch CXCL9 wirkt potentiell in der in vivo Situation von Autoimmunerkrankungen auf unterschiedliche Weise proinflammatorisch. Zum einen wird die Mobilisierung von Th1-Zellen aus CXCL12 exprimierenden peripheren Gewebe gefördert und gleichzeitig verhindert, dass Th1-Zellen in nicht entzündetes peripheres Gewebe rekrutiert werden. Zum anderen wird im Entzündungsgebiet die Affinität der Th1-Zellen zu den CXCL12-exprimierenden Endothelzellen verringert und die Migration in tieferliegende Gebiete der Entzündung begünstigt. Ferner vermindern CXCR3-Liganden auch antiinflammatorische Effekte des CXCL12s, wie z.B. die Polarisierung der Th1-Zellen in regulatorische T-Zellen.
Gephyrin ist ein Protein mit strukturellen und enzymatischen Eigenschaften. Ein Aspekt der strukturellen Funktion ist die synaptische Verankerung inhibitorischer Rezeptoren an das Zytoskelett. Darüber hinaus besteht die Möglichkeit, dass ein Interaktionspartner von Gephyrin, Collybistin, die Organistion des Aktin-Zytoskeletts reguliert. Die enzymatische Funktion von Gephyrin besteht in der Synthese eines Kofaktors, welcher für die Aktivität von Molybdoenzymen notwendig ist. Das gezielte Ausschalten des für Gephyrin kodierenden Gens weist im Mausmodell einen letalen Phänotyp auf. Als Ursache hierfür kommen die fehlende synaptische Aggregation von Glyzin- und GABAA-Rezeptoren in neuronalem Gewebe, sowie die fehlende Aktivität von Molybdoenzymen in peripheren Organen in Frage. Da sowohl Molybdän-Kofaktor-Defizienzen beim Menschen, wie auch bestimmte Mutationen von Glyzin- und GABAA-Rezeptoren bei Mäusen letal sind, kann der beobachtete Phänotyp nicht eindeutig zugeordnet werden. Die vorliegende Arbeit verfolgt zwei Ziele. Das erste ist eine Einengung der Binderegion von Collybistin an Gephyrin. Dabei konnte diese durch immunzytochemische und biochemische Methoden auf wenige Aminosäuren genau beschrieben werden. Verschiedene Strukturvorhersageprogramme weisen auf eine helikale Struktur für diesen Sequenzabschnitt hin. Der Austausch der Aminosäuren R107, D108, R111 und E117 mit Alanin führt über den Verlust ionischer Ladungen zum Verlust der Bindungseigenschaft. Das zweite verfolgte Ziel ist die Einführung eines transgenen Konstrukts, welches nur die Molybdän-Kofaktor-Synthesefunktion wiederherstellen soll, in den genetischen Hintergrund der Gephyrin-„knock-out“ Maus. Ziel dieser Analyse ist die Herstellung eines Phänotyps, bei dem die inhibitorische Transmission in einer Weise gestört ist, welche Symptome der Molybdän-Kofaktor-Insufizienz ausschließt. So sollte die Ursache für die Letalität der Gephyrin-Mutation zugeordnet werden können. Dazu wurde das pflanzliche Gephyrin-Homolog Cnx1 in ein Expressionskonstrukt kloniert, und nach biochemischer, immunzytochemischer und funktioneller Analyse in heterologen Zellen in eine Wildtyp-Mauslinie injiziert. Diese wurde mit Geph-/--Mäusen gekreuzt und die daraus resultierenden homozygoten, transgenen Tiere analysiert. Diese Tiere sterben ebenso wie die Geph-/--Mäuse am Tag der Geburt. mRNA des Transgens konnte in Hirn und Leber nachgewiesen werden, das Protein allerdings nur in Hirnextrakten. Dementsprechend lag die Aktivität des für die Lebensfähigkeit von Säugetieren wichtigsten Molybdoenzyms, der Sulfitoxidase, in Leberextrakten nur bei 30%. Eine morphologische Untersuchung von Gehirnschnitten ergab keine Auffälligkeiten. Eine Immunhistochemische Analyse von Rückenmarkschnitten zeigte in Übereinstimmung mit Beobachtungen an Gephyrin-„knock out“-Mäusen diffus verteilte Glyzin-Rezeptoren. Somit wurde eine Maus mit beeinträchtigter inhibitorischer synaptischer Transmission und partiell wiederhergestellter Molybdän-Kofaktor Biosynthese hergestellt. Aufgrund der großen Schwankungsbreite der Aktivität der Sulfitoxidase in Leberextrakten verschiedener Geph-/- + cnx Tiere wäre ein Unterschied in der Lebenserwartung verschiedener Tiere zu erwarten gewesen, wenn die Defizienz dieses Molybdoenzyms die Ursache für den letalen Phänoyp gewesen wäre. Da die transgenen Tiere aber wie die Geph-/- Mäuse wenige Stunden nach der Geburt sterben, ist davon auszugehen, dass der beschriebene Phänotyp auf das Fehlen der strukturellen Eigenschaften von Gephyrin zurückzuführen ist, und nicht von einer durch Beeinträchtigung des Schwefelstoffwechsels hervorgerufene progressive Schädigung des ZNS.
Das Burkitt Lymphom ist ein aggressives B-Zelllymphom, das in tropischen Regionen Afrikas und in Neu Guinea endemisch auftritt und vor allem bei Kindern vorkommt. Die sporadische Form des Burkitt Lymphoms tritt weltweit in geringerer Häufigkeit auf und betrifft alle Altersschichten. In nahezu allen endemischen Fällen ist das Epstein-Barr Virus in den Tumorzellen nachweisbar, jedoch nur in ca. 20 % der sporadischen Fälle. Der Beitrag von EBV zur Entstehung EBV-positiver Burkitt Lymphome ist seit über 50 Jahren EBV-Forschung ungeklärt. Im Jahr 2004 wurden im Genom des Epstein-Barr Virus eine Reihe von microRNAs entdeckt, die potentiell für die Pathogenese des EBV-positiven Burkitt Lymphoms relevant sein könnten. Da die Expression der viralen microRNAs seither für das Burkitt Lymphom nur unvollständig beschrieben worden sind, wurden sie in dieser Arbeit systematisch analysiert und dadurch ein vollständiges Expressionsprofil erstellt. Es konnte dabei keine Unterscheidung zwischen endemischen und sporadischen Fällen erreicht werden, jedoch wurden hierbei erstmals Fälle identifiziert, die trotz nachgewiesener EBV-Assoziation keine viralen microRNAs enthielten. Neben den viralen microRNAs könnten im Burkitt Lymphom auch die zellulären microRNAs für die Tumorentstehung von Bedeutung sein. Deshalb wurde in dieser Arbeit auch die Expression der zellulären microRNAs aus Burkitt Lymphom-Biopsien charakterisiert. Durch hierarchisches „Clustering“ bildeten sich drei Gruppen, die hauptsächlich durch An- und Abwesenheit von zwei microRNAs (miR21 und miR92a) definiert wurden, denen onkogenes Potential zugeschrieben wird. Die Expressionsmuster der einzelnen Gruppen weisen auf zelluläre Mechanismen der Pathogenese des Burkitt Lymphoms hin.
Die genetische Charakteristik des Burkitt Lymphoms ist eine Chromosomentranslokation, welche das Protoonkogen c MYC unter die Kontrolle von regulatorischen Elementen der Immunglobulingene bringt. Durch die somit erhöhte Transkription von c-MYC entfaltet das Genprodukt sein onkogenes Potential. Mutationen im offenen Leserahmen können dieses Potential zusätzlich verstärken. Da c MYC ein pleiotroper Transkriptionsfaktor ist und somit auf eine ganze Reihe zellulärer Prozesse Einfluss hat, bewirkt die Translokation massive Veränderungen in der Zelle. Vorangegangene Untersuchungen der Arbeitsgruppe zeigten, dass die antivirale Interferonantwort durch hohe c MYC-Expression unterdrückt wird. Diese Beobachtung liefert eine mögliche Erklärung für die Immunevasion von Burkitt Lymphom-Zellen, trotz Anwesenheit des EBV-Genoms. In Zelllinien, die aus Burkitt Lymphom-Biopsien generiert wurden, konnte gezeigt werden, dass EBV eine Interferoninduktion auslöst, die durch c-MYC unterdrückt wird. In dieser Arbeit konnte auch gezeigt werden, dass Epstein-Barr-virale Nukleinsäureprodukte durch den zytosolischen Rezeptor RIG-I Interferon induzieren, dieser aber durch die hohe c-MYC-Expression transkriptionell gehemmt wird. Neben RIG-I wurden weitere Rezeptoren und Mediatoren der Interferoninduktionskaskade identifiziert, die ebenfalls transkriptionell von c-MYC unterdrückt werden. Diese Ergebnisse stützen die Hypothese, dass c-MYC durch Unterdrückung der angeborenen Immunität die Immunevasion von Burkitt Lymphom-Zellen ermöglicht.
One of the key functions of blood vessels is to transport nutrients and oxygen to distant tissues and organs in the body. When blood supply is insufficient, new vessels form to meet the metabolic tissue demands and to re-establish cellular homeostasis. Expansion of the vascular network through sprouting angiogenesis requires the specification of ECs into leading (sprouting) tip and following (non-sprouting) stalk cells. Attracted by guidance cues tip cells dynamically extend and retract filopodia to navigate the nascent vessel sprout, whereas trailing stalk cells proliferate to form the extending vascular tube. All of these processes are under the control of environmental signals (e.g. hypoxia, metabolism) and numerous cytokines and peptide growth factors. The Dll4/Notch pathway coordinates several critical steps of angiogenic blood vessel growth. Even subtle alterations in Notch activity can profoundly influence endothelial cell behavior and blood vessel formation, yet little is known about the intrinsic regulation and dynamics of Notch signaling in endothelial cells. In addition, it remains an open question, how different growth factor signals impinging on sprouting ECs are coordinated with local environmental cues originating from nutrient-deprived, hypoxic tissue to achieve a balanced endothelial cell response. Acetylation of lysines is a critical posttranslational modification of histones, which acts as an important regulatory mechanism to control chromatin structure and gene transcription. In addition to histones, several non-histone proteins are targeted for acetylation reversible acetylation is emerging as a fundamental regulatory mechanism to control protein function, interaction and stability. Previous studies from our group identified the NAD+-dependent deacetylase SIRT1 as a key regulator of blood vessel growth controlling endothelial angiogenic responses. These studies revealed that SIRT1 is highly expressed in the vascular endothelium during blood vessel development, where it controls the angiogenic activity of endothelial cells. Moreover, in this work SIRT1 has been shown to control the activity of key regulators of cardiovascular homeostasis such as eNOS, Foxo1 and p53. The present study describes that SIRT1 antagonizes Notch signaling by deacetylating the Notch intracellular domain (NICD). We showed that loss of SIRT1 enhances DLL4-induced endothelial Notch responses as assessed by different luciferase responsive elements as well as transcriptional analysis of Notch endogenous target genes activation. Conversely, SIRT1 gain of function by overexpression of pharmacological activation decreases induction of Notch targets in response to DLL4 stimulation. We also showed that the NICD can be directly acetylated by PC AF and p300 and that SIRT1 promotes deacetylation of NICD. We have identified 14 lysines that are targeted for acetylation and their mutation abolishes the effects of SIRT1 of Notch responses. Furthermore, over-expression or activation of SIRT1 significantly reduces the levels of NICD protein. Moreover, SIRT1-mediated NICD degradation can be reversed by blockade of the proteasome suggesting a mechanism resulting from ubiquitin-mediated proteolysis. Indeed, we have shown that SIRT1 knockdown or pharmacological inhibition decreased NICD ubiquitination. We propose a novel molecular mechanism of modulation of the amplitude and duration of Notch responses in which acetylation increases NICD stability and therefore permanence at the promoters, while SIRT1, by inducing NICD degradation through its deacetylation, shortens Notch responses. In order to evaluate the physiological relevance of our findings we used different models in which the Notch functions during blood vessel formation have been extensively characterized. First, retinal angiogenesis in mice lacking SIRT1 activity shows decreased branching and reduced endothelial proliferation, similar to what happens after Notch gain of function mutations. ECs from these mice exhibit increased expression of Notch target genes. Second, these results were reproducible during intersomitic vessel growth in sirt1-deficient zebrafish. In both models, the defects could be partially rescued by inhibition of Notch activation. Third, we used an in vitro model of vessel sprouting from differentiating embryonic bodies in response to VEGF in a collagen matrix. Our results showed that Sirt1-deficient cells shows impaired sprouting which correlated with increased NICD levels. In addition, when in competition with wild-type cells in this assay, Sirt1-deficient cells are more prone to occupy the stalk cell position. Taken together, our study identifies reversible acetylation of NICD as a novel molecular mechanism to adapt the dynamics of Notch signaling and suggest that SIRT1 acts as a rheostat to fine-tune endothelial Notch responses. The NAD+-dependent feature of SIRT1 activity possibly links endothelial Notch responses to environmental cues and metabolic changes during nutrient deprivation in ischemic environments or upon other cellular stresses.
Life-attenuated measles virus (MV) vaccines have revealed their capacity to routinely induce life-long immunity against MV after just a single or two low-dose injections. Moreover, MV vaccines have been shown to be extensively safe and well tolerated, in general. Thus, MV is a prime candidate for a recombinant vaccine platform to protect also against other pathogens after vaccination. For this purpose, foreign genes can be inserted into additional transcription units (ATU) in recombinant MV genomes so that the encoded foreign proteins are co-expressed with MV proteins in infected cells. These so-called bivalent MV should protect against infection by MV or the pathogen, which the encoded foreign protein had been derived from. Bivalent MVs have already been shown to be effective vaccines against e.g. dengue virus or hepatitis B virus infections by inducing humoral and sometimes also cellular immune responses. In most of these studies, soluble or soluble versions of the pathogens' antigens were used for generation of bivalent MVs.
We hypothesized that the form of the antigen expressed by bivalent MVs is crucial for the potency and constitution of the induced immune responses. Therefore, three different forms of an antigen expressed by bivalent MVs were analyzed, here. The model antigen chosen for this purpose has been the envelope protein (Env) of SIVsmmPBj1.9. In its natural mature form, Env is composed of the surface unit gp120 and the transmembrane unit gp41, which stay non-covalently linked after proteolytic processing of the common precursor protein gp160. However, gp120 can be shed by infected cells or virus particles. Therefore, natural gp160 antigen was used as shedding form. Furthermore, stabilized covalently-linked gp160 variants and soluble gp140 variants were used in this thesis. These different antigen forms were inserted either behind the P or behind the H expression cassettes into the MV genome. The respective bivalent MVs were rescued and characterized. Expression of SIVsmmPBj1.9 Env variants by the bivalent MVs was confirmed by immuno blot and in situ immunoperoxidase assays. Replication curves of bivalent MV showed that growth of MVs expressing the different Env variants was slightly delayed by approximately 24 h compared to control viruses.
For immunization of transgenic, MV-susceptible IFNAR-/--CD46Ge mice, which are the current standard to analyze MV vaccines in a small animal model, an optimal dose of 1x105 TCID50 was determined. For the evaluation of humoral immune responses in transgenic mice, two ELISA systems for the detection of total α-MV and α-SIV antibodies and neutralization assays for detection of neutralizing antibodies against MV and SIV in sera of immunized mice were established. Mice immunized with any of the bivalent MVs showed significant humoral immune responses against MV comparable to those elicited by the parental MV vaccine strain without further genetic modifications. Mice immunized with MVvac2-gp140(P) expressing the soluble gp140 variant revealed highest α-SIV titers with a maximal OD of up to 0.4. Second highest levels of α-SIV antibodies were detected in mice that were immunized with the shedding variants or soluble Env in other positions. MVs expressing the stabilized variants induced only very low α-SIV antibody titers. Neutralizing antibodies directed against SIV could be detected in sera of mice immunized with MVs expressing the soluble or shedding variants, but not in sera of mice immunized with MVs expressing the stabilized variants. In sera of control mice immunized with PBS no antibodies could be detected, as expected. Thus, soluble and shedding antigens induced humoral immune responses, whereas stabilized antigens induced only weak humoral immune responses but no neutralizing antibodies. Analysis of cellular immune responses is still ongoing.
Besides Env, further SIV antigens could be tested for their potency to induce humoral as well as cellular immune responses.
Besides being used as a vaccine platform, recombinant MVs are evaluated as future agent for cancer therapy due to their significant inherent tumor-lytic, so-called oncolytic activity. Currently, the anti-tumoral activity of MV is analyzed in clinical phase I trials. MV strains with high fusion activity are used as oncolytic agents. The fusion protein F of MV strain NSe is highly fusogenic, in contrast to e.g. F of MVwt323, a clone of the pathogenic strain IC-B. Sequence analysis of these two proteins identified one coding nucleotide difference at aa 94 in the F2 domain: a valine (V) in FNSe and a methionine (M) in Fwt323. To evaluate impact of this difference, residues at aa 94 were exchanged. After transient-transfection of MV F and H expression plasmids in receptor-positive cells, V94 in the F2 subunit of FNSe or Fwt323 led to about 6-fold higher fusion activity compared to F proteins with M94. The co-expressed H protein (HNSe or Hwt323) did not influence fusion activity, indicating that the receptor (CD46 or SLAM) bound by H does not quantitatively affect the F proteins' activation. Analysis of F and H showed that formation and transport of MV glycoprotein complexes are not altered by substitution in aa 94 of FNSe or Fwt323.
Furthermore, recombinant MVNSe, MVNSe-F-M94, MVwt323, or MVwt323-F-V94 were rescued. Viral replication revealed slightly higher titers for recombinant MVs expressing M94 in F after 96 h of replication, compared to MVs expressing V94. MVs expressing V94 in F2 showed 2.5-fold higher fusion activity on CD46- and SLAM-positive Vero-hSLAM cells and 2-fold higher fusion activity on B95a cells expressing only SLAM compared to MVs expressing F with M94. Fusion activity of recombinant MVs can thus be modulated by substituting a single aa. V94 in the F protein results in highly fusion active MVs with possibly increased direct cytotoxicity in infected tumors, whereas M94 in F could be associated with decreased fusion activity for therapies, where higher virus titers are required.
Sponges are one of the major components of benthic communities and are considered to be a
key role organism in marine ecosystems. In addition to their importance in terms of
biodiversity, sponges are becoming increasingly attractive to the industry, as they themselves
or associated symbionts, produce various kinds of secondary metabolites of pharmaceutical
properties. Some of them have already been clinically applied.
The taxonomic characters of Porifera are limited to only a few morphological and
histological characters. In addition, sponges of the same species often show a wide
morphological variability, whereas the latter depends on different ecological parameters such
as water depth and current conditions. Thus, the taxonomic classification of sponges often
becomes a scientific challenge.
The fauna of the Yellow Sea rates among the least studied worldwide. At the same time,
according to the UN Atlas of the Ocean, the Yellow Sea is one of the most intensively
exploited marine areas in the world. This is not least due to the dense human population living
in the entire catchment area of the Yellow Sea region. In order to compile medium- and longterm
conclusions about the anthropogenic impact on biota of the Yellow Sea, the knowledge
of species and their distribution is of crucial importance, as these data form the baseline for all
future conservation efforts.
Until now the sponge fauna of the Chinese Yellow Sea is insufficiently investigated.
Thus, there is only one publication on sponges from this region that has been released
hitherto. This paper is dealing with only a view species. However, there is no reference
concerning the present location of the voucher material, on which this publication is based on.
Consequently, no scientific collection on Porifera from the Chinese part of the Yellow Sea
exists to date.
In order to compile a documentation of the recent sponge community of the Chinese
Yellow Sea, 12 study sites along the coast of the Liaoning Peninsula, China, Northeast
Yellow Sea, were investigated with focus on sponge distribution. The corresponding habitats
were characterized in regard to their topographical features, abiotic parameters, and common
composition of benthic megafaunal and macroalgal assemblages.
Due to the lack of comparable studies, a comprehensive literature research on sponges of the
shallow Northwest Pacific Ocean was required. As a result the first compilation of
publications is presented, dealing with sponges from shallow depths of the northwestern
Pacific Ocean.

In the course of this study, 31 sponge species in total were recorded, which are scientifically
processed. With the exception of four all specimens were determined to species- level.
Twelve out of the total number of species are new to science and are described and classified
according to the recent taxonomic system of the phylum Porifera.
The results of this study indicate considerable differences in species composition between
investigated sites. It is shown that physical factors (particularly current regime, sedimentation,
seasonally related variations in temperatures), as well the availability of suitable substrates are
directly related to the diversity and abundance of investigated sponge communities. In this
context possible adaptation strategies of the corresponding sponges were discussed in detail.
Two sponge species, Clathria (Clathria) asodes and Antho (Acarnia) lithophoenix, formerly
known exclusively from the northeastern Pacific Ocean, are now recorded from the Northwest
Pacific Ocean for the first time. Furthermore, Penares hongdoensis, Clathria (Clathria)
hongdoensis and Celtodoryx girardae were synonymized with Penares cortius, Clathria
(Clathria) acanthostyli, and Celtodoryx ciocalyptoides respectively. Moreover, the occurrence
of eight sponge species, which were known from previous records from the Yellow Sea, could
be confirmed.
As a result of this study the Asian origin of a sponge species that is invasive to the French and
Dutch coasts of the Northeast Atlantic Ocean since the 1990s could be established. Moreover,
it is demonstrated that Celtodoryx girardae from the northeastern Atlantic is in fact
conspecific with Cornulum ciocalyptoides described by Burton (1935) from the Posiet Bay,
Sea of Japan. Apart from taxonomic remarks, variations between populations from both
oceans are examined and discussed thoroughly in regard to possible ecological implications.
The community of documented sponges shows overlapping with the one from the Sea of
Japan. According to the results it is assumed that the endemic degree of the sponges from the
Chinese Yellow Sea is rather low to moderate.
The material obtained in the course of this study was integrated in the collection of the
Senckenbergischen Naturforschenden Sammlungen. Therefore, it is the first scientific
collection of sponges from the Chinese Yellow Sea that can be consulted as a basis for all
further studies on sponges of this region.
The present study is the only investigation of sponges from Dalian and adjacent waters before
the spill occurred in the Dalian harbour in July 2010. Therefore, it provides an essential
baseline needed to assess the impact of the oil spill on benthic communities.
Tumor hypoxia and nutrient starvation are common phenomena in cancerous tissue. Cells that resist this hostile environment are selected for a more aggressive phenotype, usually accompanied by therapy resistance. The hypoxia inducible factors HIF-1a and HIF-2a play a key role in the adaptive homeostatic responses to these challenging conditions inducing a number of target genes that are involved in the regulation of a variety of cellular processes such as angiogenesis, proliferation, metabolism, self-renewal and cell death/cycle arrest. Thus, the HIF pathway encompasses opposing adaptive responses on tumor growthgrowth promoting abilities on the one hand and growth inhibiting on the other. A recent study in our lab uncovered that this switch between cell death and cell survival critically depends on HIF-2a protein levels. Since PHDs (HIF prolyl hydroxylases) are the main regulators of HIF protein abundance and hypoxia drives the malignant phenotype of tumors, we wanted to characterize HIF regulatory functions of PHDs under hypoxic conditions. Our intention was to reveal the importance of PHD contribution to the opposing functions of HIFs under hypoxia. Characterization of PHD1-4 mRNA and protein expression levels under normoxic and hypoxic conditions in glioblastoma cell lines led to the identification of PHD2 and PHD3 as hypoxia inducible PHD isoforms and highlighted their predominant function under hypoxia. Mechanistically, we demonstrated that HIF mediates the hypoxic induction of PHD2 and 3 within a negative feedback loop, promoting its own degradation during prolonged hypoxia. The functional impact of PHD2 and 3 abundance on cell viability under hypoxic conditions was analyzed by disrupting PHD2 and PHD3 function either through a siRNA mediated approach or by application of the PHD inhibitor DMOG. These experiments uncovered that PHD2 and 3 are protective under hypoxic conditions and that PHD inhibition expedites cell death. Combined HIF and PHD suppression under hypoxic conditions abrogated this increased susceptibility to cell death, clearly showing that PHD2 and 3 act in a negative feedback regulatory loop to limit the HIF response under prolonged hypoxia. With respect to possible future therapeutical applications we co-treated cells with a PHD inhibitor and pro-apoptotic agents staurosporine or TRAIL. Co-challenging tumor cells even potentiated the cell death response, indicating a more widespread protective function of PHD. Taken together PHD2 and 3 protect tumor cells from cell death induction, functioning in a negative feedback regulatory loop to constrain the HIF dependent cell death responses under hypoxia. Interestingly, however, when assessing the role of PHD2 and PHD3 in in vivo tumor growth using an intracranial tumor model, we identified an exclusive tumor suppressor function for PHD3. Loss of PHD3 function enhanced tumor growth whereas increased PHD3 expression diminished the tumor burden. The accelerated tumor growth following PHD3 loss could be attributed to a decrease in the induction of apoptosis and an increase in proliferation. Tumor cells are frequently exposed to temporary and spatial depletion of nutrients. Interestingly, PHD3 loss conferred a growth advantage under growth factor deprivation. The growth regulatory function of PHD3 was isoform specific, HIF independent and importantly, did not require the hydroxylase function of PHD3. Previous reports have uncovered a regulatory function of the PHD system in NF-kB signaling. However, our results demonstrated that NF- kB signaling remained unaffected by alteration in the PHD3 status of the cell. Additionally, the PHD3 tumor suppressor function proved to be independent of two putative PHD3 downstream effectors, ATF4 and KIF1Bb. Mechanistically, PHD3 suppression reduced EGFR internalization, enhancing the amount of EGFR expressed on the cell surface. We further showed that the impaired EGFR internalization during PHD3 loss resulted in receptor hyperactivation under stimulated and growth factor deprived conditions. Importantly, PHD3 physcially associated with the EGFR complex as evidenced by co-immunoprecpitation. Consequently, this extended EGFR activation in PHD3 deficient cells resulted in enhanced downstream activation of EGFR signaling and increased proliferation. Consistent with the interpretation that PHD3 loss is beneficial for tumor growth, we found PHD3 promoter methylation in glioblastoma cell lines, hinting at a epigenetic mechanism to finetune PHD3 expression on top of the hypoxic driven gene regulation. Finally, we demonstrated that PHD3 tumor suppressor function is not restricted to glioblastomas since PHD3 suppression in lung adenocarcinoma accelerated subcutaneous tumor growth. With these findings, we expand the knowledge of PHD3 action from its oxygen sensing role to a regulatory function in growth factor signaling. This clearly discriminates PHD3 from the other isoforms and supports the exclusive tumor suppressor function in glioblastoma. Taken together our results identify a complex role of PHD signaling in cancer and delineate HIF dependent and HIF independent functions of the PHD system. We think that the HIF dependent protective effect of PHD2 and 3 and the HIF independent PHD3 tumor suppressor function are not mutually exclusive, but might be activated according to the heterogeneous intra-tumoral conditions. However, PHD3 hydroxylase activity is dispensable for its HIFindependent tumor suppressor function in glioma. This uncouples PHD3 function from co-factor and co-substrate requirements and allows it to act over a broader physiological range, since its influence on cellular processes is not constrained by the availability of rate limiting factors. It might explain, why the enzymatic independent functions of PHD3 predominate in vivo. Thus, therapeutic modulation of the PHD system to inhibit tumor growth has to be based on these contrasting functions of the PHD system. However, their differential dependence on the hydroxylase activity may facilitate a therapeutic strategy to specifically inhibit or promote the protective versus suppressive functions of the PHD system.
Die paravertebralen Grenzstränge entwickeln sich aus Neuralleistenzellen des Rumpf- und Lendenbereichs. Diese sammeln sich im Hühnerembryo an Embryonaltag 2,5-3 an der dorsalen Aorta und formen die primären sympathischen Ganglien. Die dorsale Aorta sezerniert Morphogene, welche einen Teil der Vorläuferzellen zur Differenzierung zu Neuroblasten anregt. Die sympathischen Neuroblasten sind, obgleich sie bereits neurale und noradrenerge Marker exprimieren, zur Zellteilung fähig. Sie unterscheiden sich darin von anderen Neuralleistenderivaten wie beispielsweise den Neuronen der parasympathischen Ziliarganglien und der sensorischen Hinterwurzelganglien. Schließlich wandern die primären sympathischen Ganglien weiter und bilden lateral zum Notochord die paravertebralen Grenzstränge (Rohrer, 2011).
Der Homöodomänen-Transkriptionsfaktor PROX1 wird im Laufe der Entwicklung höherer Vertebraten in vielen Geweben exprimiert. Welche Wirkung PROX1 dabei auf Überleben, Migration, Proliferation und Differenzierung hat, hängt davon ab, in welchem Zelltyp er aktiv ist (Dyer et al., 2003; Lavado et al., 2010). Im peripheren Nervensystem konnte PROX1 embryonal in den Hinterwurzelganglien und den sympathischen Ganglien nachgewiesen werden (Becker et al., 2009; Diplomarbeit Julia Holzmann, 2010). Zielsetzung dieser Dissertation war es, die Expression und die Funktion von PROX1 in sympathischen Ganglien von Hühnerembryonen zu analysieren.
Die Expressionsanalyse von PROX1 zeigte, dass der Anteil der PROX1-positiven Neurone an Embryonaltag 5 (E5) ein Maximum erreicht und danach im Laufe der Entwicklung stetig abnimmt. Dies gilt ebenso für die Population der proliferierenden Neuroblasten, welche ebenfalls im Laufe der Hühnerentwicklung erstmals detailliert untersucht wurde. Diese Korrelation führte zu der Vermutung, dass PROX1 hauptsächlich in proliferierenden Zellen exprimiert wird, welche anschließend experimentell bestätigt werden konnte. Die Population der PROX1-positiven und die der p27-negativen Neuroblasten haben in allen untersuchten Hamburger Hamilton-Stadien (HH-St 21-37) eine vergleichbare Größe. Dennoch ist PROX1 durchgehend in einem kleinen Teil der p27-positiven Neurone enthalten. Diese Population verändert sich im Laufe der Entwicklung kaum und das Fluoreszenzsignal eines oder beider Proteine ist bei doppelpositiven Zellen deutlich schwächer. Diese und andere Daten dieser Arbeit weisen darauf hin, dass es sich um Neuroblasten handelt, welche gerade aus dem Zellzyklus austreten. In postmitotischen Neuronen geht PROX1 verloren. Obwohl PROX1 in allen untersuchten HH-Stadien stark in der Population proliferierender Neurone exprimiert wird, zeichnet sich ab E7 eine kleinere Population von Neuroblasten in S-Phase ab, welche kein PROX1 enthalten.
Die Vorläuferzellen von Ziliarganglien werden, ähnlich wie die der sympathischen Ganglien, durch BMP-Proteine zur Differenzierung angeregt (Müller und Rohrer, 2002). Aufgrund der Ähnlichkeiten in der Entwicklung beider Neuralleistenderivate wurde die Expression von PROX1 in dieser Dissertation auch in Ziliarganglien untersucht: Der Transkriptionsfaktor wird dort nur an E4 und E5 vereinzelt in Neuronen exprimiert und nahezu gar nicht in Vorläuferzellen. In späteren HH-Stadien ist PROX1 in Ziliarganglien nicht mehr nachweisbar.
Ebenfalls konnte hier gezeigt werden, dass PROX1 in primären sympathischen Ganglien an E3 (HH-St 21) in Vorläuferzellen exprimiert wird, welche bereits begonnen haben, sich zu Neuroblasten zu differenzieren. Noch bevor die Differenzierung dieser Zellen jedoch abgeschlossen ist, wird PROX1 transient herunterreguliert. Die entstehenden Neuroblasten treten in dieser Phase kurzzeitig aus dem Zellzyklus aus. Da sich die Größe der p27-negativen und der PROX1-positiven Population auch an E3 stark ähnelt, kann man schließen, dass die Zellteilung in den Neuroblasten erst bei erneuter PROX1-Expression wieder aufgenommen wird. Ab E5 ist PROX1 fast ausschließlich in Neuroblasten nachweisbar.
Eine Funktionsanalyse von PROX1 unter Kulturbedingungen und im Hühnerembryo sollte durch Knockdown und Überexpression zeigen, welchen Einfluss der Transkriptionsfaktor auf die Proliferation der Neuroblasten nimmt. Die Manipulation der PROX1-Expression hatte in vitro einen proproliferativen Effekt. In vivo unterschieden sich Knockdown und Überexpression aber nicht von der Kontrolle.
Zusammenfassend wurde in dieser Doktorarbeit die Expression von PROX1 in sympathischen Ganglien von Hühnerembryonen im Detail analysiert. Der Transkriptionsfaktor ist sowohl in Vorläuferzellen als auch in Neuroblasten nur transient vorhanden. Zwar konnte eine klare Korrelation zwischen der Expression von PROX1 und der Proliferation der sympathischen Neuroblasten festgestellt werden, allerdings konnte eine Wirkung von PROX1 auf die Proliferation durch Funktionsanalysen nur teilweise bestätigt werden. Zusammen weisen die Daten darauf hin, dass PROX1 eine Rolle in der Feinregulation der Proliferation spielt.
Die noradrenergen Neurone der sympathischen Ganglien und die cholinergen Neurone der parasympathischen Ziliarganglien gehen aus den NLZ hervor. BMP-Signale induzieren die Differenzierung beider Neuronentypen, die mit der Expression von Ascl1 und Phox2a/b beginnt. Im Fall der sympathischen Ganglien werden dann Hand2 und GATA2/3 exprimiert, was wiederum zur Expression der noradrenergen Marker TH und DBH führt, die auch in differenzierten Neuronen weiterhin vorhanden sind. Im Gegensatz dazu werden während der Entwicklung der parasympathischen Ziliarneurone sowohl Hand2 als auch TH/DBH nur transient exprimiert, die differenzierten Neurone besitzen zum Großteil einen cholinergen Phänotyp (Goridis und Rohrer, 2002; Müller und Rohrer, 2002).
Thema dieser Arbeit war die Untersuchung der Rolle der Hox-Gene bei der Differenzierung des PNS. 14 der analysierten Hox-Gene werden in den sympathischen Ganglien exprimiert, wobei wir uns bei der näheren Analyse auf das HoxB-Cluster beschränkt haben. HoxB5, HoxB6, HoxB7, HoxB8 und HoxB9 werden zwischen E4 und E7 in den sympathischen und sensorischen Ganglien exprimiert, wobei nur HoxB8 und HoxB9 eine deutliche Expression in den sympathischen Ganglien zeigen. Die HoxB-Gene könnten dem Expressionsmuster nach also eine Rolle bei der frühen Entwicklung und auch bei der Aufrechterhaltung des noradrenergen Phänotyps der sympathischen Ganglien spielen.
Die differenzielle Expression der HoxB-Gene in den sympathischen Neuronen und den Ziliarneuronen und ihre mögliche Beteiligung bei der Aufrechterhaltung des noradrenergen Charakters waren Ausgangspunkt für die ektopische Expression eines Vertreters des HoxB-Clusters, HoxB8, in den Ziliarganglien. In der Normalentwicklung wird die Expression von Hand2, TH und DBH nach E4 in den Ziliarneuronen stark reduziert (Abb. 22A). Wird HoxB8 in den Vorläuferzellen der Ziliarneurone in vivo überexprimiert, wird die Hand2-, TH- und DBH-Expression weit über E4 hinaus, bis mindestens E8 auf einem signifikant höheren Niveau gehalten (Abb. 22B). HoxB8 kann diesen Effekt allerdings nur ausüben, wenn es in den noch undifferenzierten Vorläuferzellen exprimiert wird. Die HoxB8-Überexpression in Primärkulturen von Ziliarneuronen an E5 oder E8 führt nur noch zu einem Anstieg der Hand2-Expression, hat aber keinen Einfluss mehr auf die noradrenerge Genexpression (Abb. 22B).
HoxB8 zeigt zusätzlich im Vergleich mit den anderen analysierten Hox-Genen einen spezifischen Effekt auf die Hand2-, TH- und DBH-Expression, denn sowohl das paraloge Hox-Gen HoxC8 als auch das anterior-exprimierte HoxB-Gen HoxB1 erreichen nur an E5 eine signifikante Expression der drei Gene. Weder HoxC8 noch HoxB1 können die Expression von Hand2 und TH/DBH über E5 hinaus aufrechterhalten (Abb. 22C), während HoxB8 deren Expression auch noch an E8 auf einem hohen Niveau halten kann.
Die HoxB8-vermittelte Aufrechterhaltung der TH- und DBH-Expression in den Ziliarneuronen konnte allerdings nicht in einen direkten Zusammenhang mit der erhöhten Hand2-Expression gebracht werden, da die Überexpression von Hand2 nicht zu einer Aufrechterhaltung von TH und DBH an E5 und E6 führt (Abb. 22C).
Die Effekte von HoxB8 auf die Entwicklung der Ziliarneurone, die durch HoxB8 z.T. noradrenerge, sympathische Eigenschaften annehmen, unterstützen die Vorstellung, dass HoxB8 bei der Differenzierung und Ausbildung des noradrenergen Phänotyps in sympathischen Ganglien eine Rolle spielt. Es konnte also erstmals einem Vertreter der Hox-Gen-Familie eine mögliche Funktion bei der Differenzierung autonomer Neurone zugeordnet werden.
Die Gattung Palaua gehört zur Tribus der Malveae (Malvaceae, Malvoideae). Sie umfasst fünfzehn einjährige oder ausdauernde krautige Arten, die für die Nebeloasen („Lomas“, „Desierto Florido“) der Küstenwüste Perus und Chiles endemisch sind. Abweichend von den meisten anderen Gattungen der Malveae besitzt Palaua (mit Ausnahme von P. sandemanii) unregelmäßig übereinander angeordnete Merikapien. Dieses Merkmal ist ansonsten nur von den beiden altweltlichen Gattungen Kitaibela und Malope bekannt, weshalb diese früher mit Palaua in der Tribus Malopeae vereint wurden. Palynologische, cytogenetische und molekulare Analysen zeigten jedoch, dass die Malopeae eine polyphyletische Gruppe bilden und dass die in Südamerika verbreiteten Gattungen Fuertesimalva und Urocarpidium die nächsten Verwandten von Palaua sind. Ebenso wie im Aufbau des Gynözeums unterscheidet sich Palaua auch durch das Fehlen eines Epicalyx vom Großteil der Malveae, einschließlich ihrer Schwestertaxa. Seit der Erstbeschreibung der Gattung durch Cavanilles im Jahr 1785 sind nur zwei detaillierte Bearbeitungen der Gattung Palaua veröffentlicht worden. Die umfassendste davon stammt von Ulbrich (1909). Auf der Grundlage der umfangreichen Aufsammlungen von August Weberbauer beschrieb er mehrere neue Arten in seinem Werk „Malvaceae austro-americanae imprimis andinae“, das er in nachfolgenden Jahren (1916, 1932) vervollständigte. Die zweite bedeutsame Bearbeitung ist die Revision der Gattung durch Macbride (1956) in der „Flora of Peru“. Seit den 50er Jahren des vorherigen Jahrhunderts kamen jedoch zahlreiche Aufsammlungen hinzu, insbesondere durch den peruanischen Botaniker Ramón A. Ferreyra (1912-2005), sowie durch Ernesto Günther (1870-?) zusammen mit Otto Buchtien (1859-1946), Gerd K. Müller (1929-) und Michael O. Dillon (1947-), so dass eine Neubearbeitung von Palaua erforderlich wurde. Darin bestand das Hauptziel der hier vorgestellten Dissertation. Für die Revision der Gattung wurden 618 Herbarbelege der wichtigsten Herbarien morphologisch untersucht. In den Jahren 2002 und 2003 wurden während mehrmonatiger Geländearbeiten in den Lomas-Standorten Perus und Chiles eigene botanische Aufsammlungen durchgeführt sowie Daten zur Verbreitung der Arten und ihrer Ökologie erfasst. Des Weiteren wurde aus dem mitgebrachten Samenmaterial eine mehrere Arten einschließende Lebendsammlung angelegt, mit deren Hilfe detaillierte Untersuchungen zur Blütenmorphologie und Karyologie realisiert werden konnten. Besonders schwierig gestaltete sich die Bearbeitung nomenklatorischer Fragestellungen, da viele der in Berlin (B) aufbewahrten Typusbelege von Weberbauer im Zweiten Weltkrieg zerstört wurden und somit eine Identifizierung vieler Arten problematisch war. Auch die Ermittlung des Typusbelegs der Gattung, den Cavanilles für seine Beschreibung vorliegen hatte, war mühsam. Neben dem Studium der Originalbelege und Protologe mussten auch die historischen Begebenheiten rekonstruiert und Reiseberichte zu den Aufsammlungen durchgesehen werden, um unter anderem den Holotypus der Gattung identifizieren zu können. Die eigenen taxonomischen Studien führten zur Festlegung von insgesamt 8 Lectotypen, 3 Epitypen and 2 Ikonotypen. Im Rahmen der morphologischen Untersuchungen wurden sämtliche taxonomisch relevanten Merkmale detailliert erfasst, einschließlich der verschiedenen Behaarungstypen. Neben den für die Malvaceen bekannten Sternhaaren, sind hier auch Drüsenhaare für Palaua beschrieben und charakterisiert worden. Die anatomischen Studien konzentrierten sich auf Blatt- und Samenmerkmale. Zusätzlich zu den morphologisch-anatomischen Studien wurden molekularsystematische Analysen durchgeführt. Zwei Methoden kamen dabei zur Anwendung: DNA-Sequenzierung und Amplified Fragment Length Polymorphism (AFLP). Letztere wurde eingesetzt, um insbesondere die Verwandtschaftsverhältnisse junger Taxa, die sich mit DNA-Sequenzdaten kaum auflösen lassen, zu rekonstruieren. In umfangreichen Versuchen stellte sich jedoch heraus, dass diese Methode keine reproduzierbaren Ergebnisse hervorbrachte, vermutlich bedingt durch den sehr hohen Polysaccharidgehalt der DNA-Template, wie es von Malvaceen her bekannt ist. Selbst die Erprobung zahlreicher Reinigungsschritte und –methoden ergab kein zufriedenstellendes Resultat. Für die phylogenetische Rekonstruktion wurden daher ausschließlich DNA-Daten verwendet, und zwar Kern-DNA (Internal Transcribed Spacer, ITS) und Plastiden-DNA (psbAtrnH Intergenic Spacer). Andere getestete Marker, wie z.B. die trnL-F-Region, wiesen zu wenig phylogenetisch informative Merkmale auf. Die morphologischen Analysen ergaben, dass Merkmale wie die Behaarung der Kelch- und Laubblätter, die Blattform und die Größe der Blüten besonders hilfreich für die Abgrenzung der Arten sind. Im Gegensatz zu anderen nah verwandten Gattungen ist die Form der Merikarpien in Palaua relativ uniform und daher als diakritisches Merkmal ungeeignet. Die Größe der Blüte nimmt in der Regel mit der Anzahl der Staubgefäße und Merikarpien zu. Die Palaua-Arten zeigen einige Anpassungen an ihren extrem trockenen Lebensraum. Die meisten Arten sind Annuelle und vollziehen eine rasche Entwicklung während der kurzen Zeit, in der ausreichend Feuchtigkeit verfügbar ist. Bei solchen Pflanzen findet man als Anpassung häufig eine Tendenz zur vermehrten Samenproduktion. In diesem Zusammenhang ließe sich auch die innerhalb des Verwandtschaftskreises ungewöhnliche Stellung der Merikarpien bei Palaua interpretieren, mit der es den Arten gelingt, mehr Samen als bei Arten mit einreihiger Merikarpienanordnung zu produzieren. Als weitere Anpassung findet man bei den ausdauernden Arten größtenteils eine sehr dichte Behaarung, wobei die Sternhaare mehrjähriger Arten wesentlich mehr Strahlen besitzen als die bei den einjährigen Arten. In der hier vorgestellten Revision der Gattung werden 15 Arten anerkannt: P. camanensis, P. dissecta, P. guentheri, P. inconspicua, P. malvifolia, P. modesta, P. mollendoensis, P. moschata, P. rhombifolia, P. sandemanii, P. tomentosa, P. trisepala, P. velutina, P. weberbaueri sowie die neu zu beschreibende P. spec. nov. Die morphologisch abweichende P. sandemanii wird aufgrund der molekularen Analysen ebenfalls zu Palaua gestellt. Die auch in der jüngeren Literatur meist als getrennte Arten aufgefassten P. concinna und P. moschata lassen sich nach Durchsicht des umfangreichen Materials nicht mehr als eigenständige Arten aufrechterhalten. Die vormals als chilenischer Endemit behandelte P. concinna wird hier in die Synonymie von P. moschata gestellt. Auch die peruanische P. micrantha var. hirsuta wurde in die Synonymie der zuvor rein chilenischen P. modesta verwiesen, was bedeutet, dass sich das Vorkommen von P. modesta nun auch auf Peru ausdehnt. Auf infraspezifischem Niveau wurden einige Varietäten und eine Form neu beschrieben, um die im Sammlungsmaterial vorhandene morphologische Variabilität besser zu gliedern. Das ist der Fall bei P. dissecta (2 Varietäten), P. tomentosa (1 Varietät), P. weberbaueri (1 Varietät) und P. mollendoensis (1 Form). Die neuen Taxa werden an anderer Stelle gültig publiziert. Die von Baker (1890) and Ulbrich (1909) gewählte infragenerische Klassifikation mit der Einteilung in die Sektionen Annuae (einjährige Arten) und Perennes (mehrjährige Arten) erweist sich als nicht haltbar. Weder die morphologischen noch die molekularen Daten bieten hierfür Unterstützung. Auch die von Hochreutiner (1956) vorgeschlagene Ausgliederung von P. trisepala als eigene Untergattung Rauhia, aufgrund des Vorkommens von lediglich drei statt fünf Kelchblättern, erscheint nicht sinnvoll. Abgesehen von ihrer reduzierten Kelchblattzahl (3 statt 5 Kelchblätter) ist diese Art morphologisch P. moschata und P. velutina sehr ähnlich. Die Aufstellung einer eigenen Untergattung würde die tatsächlichen Verwandtschaftsverhältnisse verwischen und vermutlich eine paraphyletische Einheit schaffen. Im Vergleich zur Anzahl der Kelchblätter sind Merkmale wie der Aufbau der Infloreszenzen, die Blütengröße und -farbe, sowie die Blattmorphologie (geteilte vs. ungeteilte Blätter) nützlicher für eine infragenerische Unterteilung. Die Form der Stipeln, die von Ulbrich (1909) für eine weitere Unterteilung seiner Sektionen verwendet wurde, ist weniger für eine infragenerische Gliederung als für die Abgrenzung mancher Arten geeignet. Formell wurde in der hiesigen Arbeit auf eine infragenerische Unterteilung verzichtet, da zunächst abgewartet werden soll, ob weiterführende molekularsystematische Untersuchungen nicht doch zu einer besseren Auflösung und auch Unterstützung der basalen Knoten der Palaua-Phylogenie führen. Andernfalls steht zu befürchten, dass wiederum künstliche Sippen geschaffen werden. Nichtsdestotrotz, sprechen die eigenen morphologischen und zum Teil auch die molekularen Daten für eine Gliederung der Gattung in drei taxonomische Einheiten (siehe unten). Die Ergebnisse der molekularen Analysen (kombinierte Analyse von ITS- und psbA-trnHSequenzen) ergaben drei mehr oder weniger gut gestützte Kladen innerhalb einer sehr gut gestützten monophyletischen Palaua. Interessanterweise bildeten die Arten P. inconspicua und P. modesta eine Klade (88% Jackknife-Unterstützung, JK), die die Schwestergruppe zu den restlichen Arten der Gattung darstellt. Beide Arten haben eine von der restlichen Gattung abweichende Blütenmorphologie (kleine Petalen, weniger Merikarpien) und die razemösen Infloreszenzen enthalten neben Einzelblüten in den Achseln der Trägblätter auch 2-4-blütige Teilinfloreszenzen, an denen die Blüten kein Tragblatt aufweisen. Die zweite Klade (JK 97%) beinhaltet die Arten des P. dissecta-Komplexes, dessen Arten sich durch tief geteilte Blätter und große, auffällig rosarot bis violett gefärbte Blüten mit zahlreichen Merikarpien auszeichnen. In der dritten Klade (JK 73%) bildet P. guentheri die Schwestergruppe zu den restlichen Arten. Die hier vereinten Arten sind durch den Besitz ungeteilter Blätter und meist großer, auffällig rosarot bis violett gefärbter Blüten mit zahlreichen Merikarpien gekennzeichnet. Eine Ausnahme bildet P. guentheri, die geteilte Blätter hat und von daher Übereinstimmungen mit den Arten um P. dissecta aufweist. Sie weicht jedoch von den Arten des P. dissecta-Komplexes aufgrund ihrer geringeren Blütengröße und der geringeren Merikarpienanzahl ab. Außerdem sind die Blätter meist stärker reduziert und weniger regelmäßig geteilt als jene. Allerdings bedarf die Stellung von P. guentheri innerhalb der Gattung noch einer eingehenderen Überprüfung mit zusätzlichen (molekularen) Daten, da die Unterstützung für diese Klade vergleichsweise moderat ausfällt. Interessanterweise schließt diese Klade auch die aberrante P. sandemanii ein. Eine phylogenetische Rekonstruktion der Karpellanordnung ergab, dass die einreihige Anordnung der Karpelle in P. sandemanii vermutlich sekundär in Palaua entstanden ist. Allerdings zeigten Hypothesentests (Templeton-Test, Shimodaira-Hasegawa-Test), dass die Datengrundlage nicht ausreichend robust ist, um auch die Alternativhypothese einer sekundären Entstehung der unregelmäßig übereinander angeordneten Karpelle, wie sie die restlichen Arten der Gattung kennzeichnen, zu verwerfen. Innerhalb der Integrifolia-Klade lassen sich außerdem zwei Gruppen von Arten morphologisch deutlich unterscheiden. Die erste Gruppe besteht aus den einjährigen P. malvifolia und P. rhombifolia, die sich durch ihr fast kahles Indumentum auszeichnen und in Nord- bis Zentral-Peru vorkommen. Die zweite Gruppe, gebildet von den ausdauernden P. moschata, P. trisepala und P. velutina, ist durch ein samtiges Indumentum gekennzeichnet. Während sich P. moschata über das gesamte Verbreitungsgebiet der Gattung erstreckt, kommen die anderen Arten nur in Südperu vor. Die Chromosomenzahl von Palaua ist ein wichtiges Merkmal und diente Bates (1968) für deren Zuordnung zur Sphaeralcea-Allianz. Bis dato sind Chromosomenzählungen nur für zwei Arten bekannt gewesen: P. rhombifolia und P. moschata (beide mit 2n = 10 Chromosomen). In dieser Arbeit wurden weitere Zählungen durchgeführt und es wurde bestätigt, dass es neben diploiden auch tetraploide Arten mit 2n = 20 Chromosomen gibt. Polyploidie scheint dabei auf die ausdauernden Arten beschränkt zu sein. In manchen Arten, insbesondere in denjenigen des P. dissecta-Komplexes und in P. tomentosa, findet man eine ausgeprägte phänotypische Variabilität, die die Abgrenzung derselben stark erschwert. Ohne die Ursachen abschließend klären zu können, erscheint diese Variabilität zumindest teilweise als Ergebnis von Hybridisierung, Introgression und Polyploidisierung zu sein. In Bezug auf die Biogeographie der Gattung, zeigt sich, dass 11 Palaua-Arten endemisch für Peru sind und 4 Arten auch in Chile vorkommen. Das Verbreitungszentrum von Palaua ist das Gebiet der Lomas im Süden Perus (Departments Arequipa, Moquegua, Tacna), in dem 12 der 15 Arten auftreten. Die Blütezeit der Palaua-Arten variiert von Jahr zu Jahr, abhängig davon, wie viel Nebelfeuchtigkeit in der südhemisphärischen Winter-/Frühlingszeit für die Pflanzen zur Verfügung steht. Die Entstehung der Nebel variiert außerdem von Norden nach Süden, so dass sich die Blühphasen entlang dieses Gradienten verschieben. So liegt die Blütezeit in Nordperu zwischen Juli und August, in Zentral-Peru zwischen August und September und in Südperu und Chile zwischen Oktober und November. Abweichungen von diesem Schema entstehen vor allem in El Niño-Jahren, in denen auch während des südhemisphärischen Sommers die Lomaspflanzen blühen. Die Lomasvegetation ist eine bedrohte Pflanzenformation, deren Artenvielfalt bisher aber nur in Form eines recht kleinen Naturreservats geschützt wird. Da sich viele Lomasstandorte in der Nähe von Siedlungen befinden, sind etliche der lokal nur begrenzt vorkommenden Arten in ihrem Bestand bedroht. Dies betrifft insbesondere P. rhombifolia und P. malvifolia, deren Verbreitungszentrum im Gebiet der Hauptstadt Lima liegt. Eigene Beobachtungen am Standort haben zudem bestätigt, dass einige Populationen dieser Arten durch von Käfern verursachter Herbivorie nahezu vollständig zerstört werden. Weitere Schutzmaßnahmen zum Erhalt der Palaua-Arten (wie auch der anderen Lomas-Arten) wären daher dringend geboten.
The translocation of nuclear-encoded precursor proteins into chloroplasts is a highly ordered process involving the action of several components to regulate this molecular ensemble. Not only GTP hydrolysis and GDP release but also the phosphorylation of TOC GTPases is a widely discussed mechanism to regulate protein import. The receptor component (Toc34) and its isoform of A. thaliana (atToc33) were found to be regulated by phosphorylation. Although the phosphorylation of Toc33 is already known for several years, several questions regarding the molecular components involved in the regulation of the phosphorylation process, precisely what is the protein kinase and where this kinase is initially localized, so far remained unclear.
This thesis aimed at the defining of the phosphorylation status of TOC GTPases in monomeric and/or dimeric states, the identification of the nature of Toc33-PK (protein kinase), and in the same context it aimed at gaining first insights into the physiological significance of Toc33 phosphorylation. To this end, (I) An in vitro and in vivo system for investigating of TOC GTPases Phosphorylation (in monomeric or dimeric state) was developed. Since no information is available about the phosphorylation status of the Toc159 isoforms, the second receptor of the TOC complex, it was interesting to investigate whether these isoforms undergo phosphorylation or not. The results indicated that atToc159 isoforms are able to be phosphorylated by the kinase activity in purified outer envelope membranes (OEMs) of pea, but not atToc132. Moreover, an artificial dimer of psToc34 based on the interaction of a C-terminally fused leucine zipper was not phosphorylated. This result reflected the inability of the OEM kinase to phosphorylate the dimers of TOC GTPases. Also, In vivo labeling of atToc33 was developed and occurred in a dose-dependent manner. Therefore, this results evidenced that in vitro phosphorylation of atToc33 (both endogenous wild type and recombinant expressed proteins) is not artificial labeling but represents a physiological relevance. CD (circular dichroism) measurements revealed that recombinant GTPase domain of atToc33 is preferentially phosphorylated in its folded state. Therefore, it could be suggested that folding of atToc33rec is a prerequisite for its phosphorylation and the phosphorylation event occurs as a posttranslational modification most likely after insertion of Toc33 (Toc34) into the OE of chloroplasts.
Secondly, (II) Isolation and identification of Toc33-PK from OEMs of chloroplasts was performed. Four independent strategies were developed to identify the Toc33-protein kinase: UV-induced and chemically-based crosslinking, different applied chromatographic techniques, identification of PK-Toc33 interaction by means of HDN-PAGE (histidine- and deoxycholate-based native PAGE), and finally mass spectrometric approaches were performed on fractions including the potential kinase activity. UV-induced crosslinking procedure was developed and resulted in covalent bonding of nine proteins to [a-32P] ATP, while chemically-based one was not significant. The applied chromatographic and HDN-PAGE approaches, including mass spectrometry, have revealed the identification of 13 protein kinases. Of these identified kinases, phototropin2 (Phot2, AT5G58140), leucine-rich repeat PK (LRR-PK, AT4G28650.1), and receptor-like transmembrane PK (RLK, AT5G56040.2) were selected as the most promising candidates (ca. kinase type and one transmembrane helix for membrane localization).
(III) The physiological significance of Toc33 phosphoryation was shown to link this process with the environmental changes (especially, the light conditions). Identification of chloroplast OE-located PKs performed by nLC-MALDI-MS/MS resulted in the detection of Phot2. Furthermore, the subcellular localization of Phot2 in OEM of chloroplasts was confirmed by immunoblotting experiments using a-Phot2 antibody. The kinase activity of Phot2 towards TOC GTPases was characterized and revealed that fused GST-KD (kinase domain) protein able to specifically phosphorylate atToc33rec, but not atToc159rec. Also, endogenous atPhot2 was upregulated and heavily detected in the ppi1-S181A plant line (where serine to alanine exchange was performed to abolish the phosphorylation of atToc33). Hence, we suggested that certain signal cascades may directly or indirectly link Toc33 receptor phosphorylation, protein levels of Phot2 (as promising PK candidate), and irradiation conditions (as an inducing signal of the subsequent phosphorylation events). Light-dependent phosphorylation of Toc33 was shown either after de-etiolation conditions or after high light intensities of blue light was performed. Therefore, phosphorylation of Toc33 might be identified as an external regulatory signal to regulate preproteins import into chloroplasts in response to environmental conditions (e.g. light changes) or as a signal of chloroplast biogenesis.
The documentation of life on Earth, that is, the inventorization of nature and the naming and classification of organisms found therein, is a major task for biologists today and a fundamental precondition for nature conservation efforts. This study aimed at contributing to the inventory of amphibians and reptiles in selected, previously understudied ecoregions of Bolivia. I strove to document diversity patterns and seek possible ecological and historical reasons for these patterns. Special attention was paid to the Chiquitano Region situated in the eastern lowlands of Bolivia in a climatic transition zone between the humid evergreen Amazon Forests and the deciduous thorn-scrub vegetation of the Gran Chaco. In congruence with its location in the transition zone, the Chiquitano Region displays a mosaic of habitats: The vegetation is dominated by the endemic Chiquitano Dry Forest, which is probably the largest extant patch of Seasonal Dry Tropical Forest, with enclaves of savanna, the western outliers of the Cerrado biome of central Brazil. Taxonomic revisions: The taxonomic data in this study are used as a tool to measure biodiversity, to assess biogeographic relationships, and to evaluate conservation needs. Since all is predicated on the taxonomic decisions made, an adequate taxonomy is essential, and taxonomy can be regarded as the foundation of this study. The methodology encompassed a variety of herpetological field techniques, such as different survey methods, preparation and documentation of voucher specimens, recording of frog calls, and herpetological laboratory techniques, such as morphology, molecular procedures with mtDNA, phylogenetic analyses, and bioacoustic analysis and descriptions of frog calls. A total of 1251 specimens belonging to 200 species were obtained during this study, including 87 amphibian and 123 reptile species. This constitutes about 36% of the herpetofauna currently known for Bolivia, about 34% of the amphibians currently known for Bolivia and about 40% of the reptiles, respectively. In the course of this study, a new species of frog was described from the study site Caparu in the eastern lowlands of Bolivia; this species, Hydrolaetare caparu Jansen, Gonzales & G. Köhler 2007, differs from the other two congeners in external morphology (e.g., lateral fringes and relative length of fingers, size of palmar tubercle, webbing of toes, and colouration) and advertisement call. Two new colubrid snake species were also described from the study site San Sebastián. Thus far, both are known only from the Chiquitano Region, Provincia Ñuflo de Chávez. Phalotris sansebastiani Jansen & G. Köhler 2008 differs from all the other species in the genus in having a triangular projection of the red snout colouration reaching onto the parietals. Xenopholis werdingorum Jansen, Gonzales & G. Köhler 2009 can be identified as a member of the genus Xenopholis by its vertebral morphology. It differs from the other two species of Xenopholis in having a unique uniform dorsal colour pattern, and from X. scalaris in having two prefrontals and a narrow septum within the neural spine and perpendicular to its long axis as evident in the x-ray images. A review of a small collection of pitvipers from different lowland localities and from the Inter-Andean dry valleys of the region of Pampagrande revealed one new species of Bothrops and one of Bothrocophias (both to be formally described elsewhere). The two pitviper species differ morphologically and genetically from their congeners. The results of a brief review of a small collection of frogs of the genus Scinax (Anura: Hylidae) from different localities in the lowlands, together with analyses of their bioacoustics, suggest an unknown cryptic diversity in Bolivian species of Scinax cf. fuscomarginatus and allies. However, further studies are necessary to clarify the taxonomic status of these populations. In addition, this study provides new data on the morphology (e.g., pholidosis) of snakes, many of them previously known only from few museum specimens. Keys to the Bolivian lizard species of Cercosaura and the Bolivian snake species of Chironius, Clelia, Liophis, Lystrophis, Phalotris, and Xenodon are presented here for the first time. New information on distribution includes many range extensions of amphibian and reptile species, such as five new country records (one frog species, four snake species) and six new departmental records (two frog species, four snake species). Observations on ecology and natural history: Several observations on ecology and natural history were made during field work. Visual signaling, an aspect of territorial behavior that was already known for several species of the genus Phyllomedusa, could be described for the first time for Phyllomedusa boliviana (Jansen & J. Köhler 2007). Furthermore, during audio surveys of an anuran community at the study site San Sebastián from 2005 to 2007, a decline of certain amphibian populations was observed in the rainy season 2006/2007 (Jansen et al., in press). This is possibly related to an extreme drought in the dry season of 2006 where 158 consecutive days without rainfall were recorded. In addition, a new method for measuring intensity of anuran choruses by means of a continuous sound pressure metre was developed (Jansen 2009). The method was suitable to detect calling phenology (during one night), as well as differences in calling activity (between two nights). Biodiversity and biogeographical relationships: Species lists were compiled at the six study sites Pampagrande, Los Volcanes, San Sebastián, Caparú, El Espinal und El Corbalan. The total amphibian and reptile species numbers observed ranged from 37 to 101 with the highest species numbers in San Sebastián (101) and Caparú (89) and the lowest in Los Volcanes (37) and El Espinal (41). A preliminary species list of the herpetofauna of the Chiquitano Region was presented, including 60 amphibian and 84 reptile species. The majority of the amphibians of the Chiquitano Region are classified predominantly as inhabitants of open formations (41 species, 68.3%). Interestingly, even the majority of species recorded from the Chiquitano Dry Forest (32 species) are usually associated with open formations (22 species, 66.7%), followed by the number of species associated with open and forest formations (8 species, 24.4%). Only two of the observed species (6.0%) are predominant forest dwellers. The amphibian assemblage of the Chiquitano Region is most similar in composition to that of the Cerrado biome: 46 species (76.7%) occur in the Cerrado as well, and three species are regarded as Cerrado endemics (5.0%). The Chiquitano Region shares considerably fewer amphibian species with the other biomes (Amazon: 22 species, 36.7%; Gran Chaco: 13 species, 21.7%; Caatinga: 16 species, 26.7%). The reptile assemblage also has significant affinities to the Cerrado, which can be seen in the high proportion of reptile species distributed in that biome (68 species; 81.0%). Affinities to the other biomes are as follows: Amazon (48 species, 57.1%), Chaco (37 species, 40.1%), and Caatinga (30 species, 35.7%). When arranged in mutually exclusive biome categories, reptiles and amphibians showed similar patterns so that the majority of both amphibians and reptiles of the Chiquitano Region can be regarded as widespread. The high proportion of reptile species probably endemic to this region (5 species, 6.0%) is remarkable (i.e. Tropidurus xanthochilus, Apostolepis phillipsi, Phalotris sansebastiani, Xenopholis werdingorum, and Micrurus diana). In an analysis of the biodiversity patterns and biogeographical relationships of the herpetofauna of the study sites, these sites were compared with literature data from 37 localities and included in a presence/absence matrix with a total of 657 amphibian and reptile species in the surrounding South American biomes Amazon, Cerrado and Gran Chaco. The biogeographic relationships between these sites were evaluated using the Coefficient of Biogeographic Resemblance (CBR), cluster analysis, and multidimensional scaling (MDS) of sites. The analyses were first conducted on amphibians and reptiles combined, and than group-specific each for amphibians, reptiles, lizards, and snakes, separately. A “bias-reduced analysis” was developed for a better understanding of the affinities of the amphibians. In this analysis, e.g., the distinct habitat types of the Chiquitano Region, the Chiquitano Dry Forest and the Cerrado were taken into account. Analyses of the biodiversity patterns revealed that the sites in the Amazon comprise highest species numbers, as expected, followed successively by the sites in the Cerrado biome and sites in-between the two biomes. Within the eastern lowlands of Bolivia, the Chiquitano Region is the most rich in species. Comparing it with the other South American sites, the Chiquitano Region has a surprisingly high alpha diversity, especially in amphibians. The microgeographic variation in species composition (beta diversity) in the Chiquitano Region is also remarkably high and obviously related to the mosaic character of the vegetation and habitats. However, the bias-reduced analysis revealed that the amphibian fauna of the open areas and savannas at Hacienda San Sebastián (with 36 species in the Cerrado and pastureland) was one of the most species-rich savanna sites known for amphibians in South America. Considering that the Hacienda San Sebastián site is only ca. 3300 ha (= 1.29 amphibian species per km2), this outcome is particularly suprising. The results of the analyses of the biogeographical relationships suggest that the herpetofauna of Bolivia’s lowlands, including the Beni, the Pantanal and the Chiquitano Region, is as distinct from the herpetofauna of the Gran Chaco, Amazon, and Cerrado as these biomes are from each other. The Chiquitano herpetofauna in particular represents a unique and well-defined herpetofaunal assemblage when compared to all surrounding localities and biomes. This is supported by high CBR-values, findings from the cluster analysis, as well as a clear separation of the Chiquitano sites in the MDS. Biogeographic relations exist in all the surrounding biomes, but are strongest to Cerrado, followed by the Amazon. This study strongly suggests that the Chiquitano herpetofauna is composite and has multiple affinities. This is congruent with a well-defined Chiquitano flora, avifauna and mammalian fauna, suggesting a similar history. The bias-reduced analysis revealed a more detailed picture of the biogeographic relations of the Chiquitano Region, especially the Chiquitano Dry Forest. I argue here that the Chiquitano Dry Forest herpetofauna is a “young”, and “former savanna herpetofauna”. Whereas the Chiquitano Dry Forest is rather poor in amphibian and reptile species, and endemics are lacking from this forest type, the isolated Cerrado enclaves are especially diverse in species and probably contain locally endemic species, such as Phalotris sansebastiani and Xenopholis werdingorum. The colonization of the young Chiquitano Dry Forest may have taken place from savannas by mainly open area species, and only briefly through the Amazon. The results emphasise the importance of bias-reduction in studies of biogeography, e.g., by using group-specific analyses or by taking into account criterias as area size and heterogeneity of compared sites. The different biogeographic patterns of reptiles and amphibians of the Andean valleys indicate a different history of these two groups. In regard to reptiles, dispersals and withdrawals into the valleys in warm humid and dry cool periods in the Pleistocene seem likely, supported by a relation between the valleys and the dry lowland (e.g., Chaco). However, it is more plausible that, during these climatic fluctuations, amphibians migrated to adjacent, more humid regions, such as Yungas. The study verified the known patterns of sister-species pairs in the Inter-Andean Dry Forest and the lowlands. Additionally, pairs of populations with slight differences in morphology were found in the valleys and in the lowlands (Cercosaura parkeri and Xenodon rhapdocephalus). Further studies must test the taxonomic status of these populations. The discovery of new species of Bothrops and Bothrocophias from the Andean valleys has several implications, and possible reasons for the high endemism in the dry valleys are discussed. Conservation and outlook: The high local alpha and beta diversity of the Chiquitano herpetofauna shows that this is a region of complex faunal interaction, which reflects the present heterogeneity of the region, but which is possibly also related to a complex geological and environmental history. The Chiquitano Region can be assessed as a region of distinct regional herpetofaunal diversity charaterised by small scale diversity patterns. It therefore merits recognition as a unique ecoregion, and conservation effort should be increased. Further research is necessary to solve the taxonomic problems addressed in this study. Moreover, future work should be directed towards the development and institution of longterm monitoring programs to evaluate the effects of climate change and changes in land-use on biodiversity, especially that of the Chiquitano Region.
Elefanten sind die größten landlebenden Säugetiere und werden schon seit Jahrhunderten in Menschenobhut gehalten. In der heutigen Zeit liegt der Schwerpunkt der Elefantenhaltung auf dem Unterbringen dieser anspruchsvollen Tiere in verhaltensgerechten Bedingungen, die auch das Wohlbefinden der Tiere berücksichtigen. Es mangelt jedoch an langfristigen Studien, die Veränderungen im Verhalten und im Wohlbefinden von Elefanten in Menschenobhut erforschen. Vor allem das nächtliche Verhalten fand bisher wenig Beachtung, obwohl Studien im natürlichen Lebensraum als auch in Menschenobhut zeigen, dass Elefanten den größten Teil der Nacht aktiv sind. Die vorliegende Studie konzentriert sich daher auf eine langfristige Überwachung des nächtlichen Verhaltens von Afrikanischen Elefanten und stellt, unter Anwendung chronoethologischer Methoden, die haltungsbedingten Einflüsse auf das Verhaltensmuster dar. Es wurden insgesamt 16 Afrikanische Elefanten (Loxodonta africana) mit Zeitraffer-Videoaufnahmen überwacht, im Opel-Zoo in Kronberg 600 Nächte, im Tiergarten Schönbrunn in Wien 300 Nächte und im Wuppertaler Zoo 70 Nächte. Dies ergibt bei einer Erfassungszeit von jeweils 16:00 Uhr bis 8:00 Uhr für alle Elefanten zusammen eine Summe von 64.320 Stunden Verhaltensregistrierung. Es konnte nachgewiesen werden, dass das nächtliche Verhalten von Elefanten durch die Haltungsbedingungen beeinflusst wird. Drei Haltungssysteme konnten zum ersten Mal in einer Studie direkt miteinander verglichen und Unterschiede aufgezeigt werden. Auch eine saisonale Abhängigkeit des nächtlichen Verhaltens konnte beobachtet werden. Es stellte sich heraus, dass Elefanten im Winter mehr und früher schlafen als im Sommer. Dies muss im nächtlichen Management berücksichtigt werden. Soziale Kontakte beeinflussen das nächtliche Verhalten ebenfalls. Es konnte erstmals beschrieben werden, dass Elefanten sich gegenseitig aus dem „Schlaf im Liegen“ aufwecken und dieses „Aufwecken“ einen Einfluss auf die Schlafdauer im Liegen hat. Die Verfügbarkeit von Nahrung ist ebenfalls ein wichtiger Faktor. Es konnte gezeigt werden, wie Elefanten zu unterschiedlichen Zeiten der Nacht auf zusätzliche Futtergaben reagieren und dass sie zu bestimmten Zeiten durch das zusätzliche Nahrungsangebot gestört werden. Unter Anwendung chronoethologischer Methoden konnte herausgearbeitet werden, dass Störungen im nächtlichen Verhaltensmuster durch vermehrtes „Weben“ erhöhte „Lokomotion“ und Reduzierung des Schlafverhaltens angezeigt werden. Beim Auftreten von Krankheiten mit Schmerzen wird die schmerzende Stelle gekühlt, indem sie mit z.B. Matsch beworfen wird. Die in dieser Studie dargestellten Einflüsse auf das nächtliche Verhalten von Afrikanischen Elefanten wurden vorher noch nicht beschrieben oder systematisch untersucht. Sie stellen wichtige Erkenntnisse für die zukünftige Haltung und das Management von Elefanten dar, sowohl im Hinblick auf eine weitere Optimierung als auch in Bezug auf die Beurteilung ihres Wohlbefindens.
Molekulare Regulation der UDP-Zucker-Biosynthese Untersuchungen anhand der myo-Inositoxygenase

Der Nukleotidzucker UDP-Glucuronsäure ist die prinzipielle Zuckervorstufe für UDP-Galacturonsäure, -Xylose, -Arabinose und –Apiose, welche alle in die Zellwandpolymere der pflanzlichen Zellwand einfließen. UDP-Glucuronsäure kann in Arabidopsis über zwei alternative, funktionelle Synthesewege gebildet werden, wobei entweder die UDP-Glucose-Dehydrogenase oder die myo-Inositoxygenase als Schlüsselenzym, welche den Kohlehydratfluss in Richtung Zellwandbiosynthese katalysiert, involviert ist. In dieser Arbeit wurden die Gene für die Enzymisoformen der Inositoxygenase (MIOX) aus Arabidopsis thaliana analysiert. Sie repräsentieren eine kleine Genfamilie bestehend aus vier Isoformen. Studien mittels Promotor::GUS-Konstrukten und RT-PCR zeigten, dass die Transkription der MIOX-Gene eine sehr transiente und organspezifische Genexpression ist. Die Isoformen MIOX1 und MIOX2 ließen sich in nahezu allen Geweben nachweisen wogegen die Isoformen MIOX4/5 nur in den generativen Geweben aufzufinden waren. Es konnte eine deutliche Präsenz aller vier MIOX-Isoformen in den generativen Geweben nachgewiesen werden und die Nukleotidzucker, die während der Samenentwicklung benötigt werden, scheinen überwiegend über die MIOX zur Verfügung gestellt zu werden. So zeigen T-DNA-Insertionslinien von ΔMIOX1 & 2 eine reduzierte Schleimhülle um die Samen und eine erhöhte Ausfallrate bei der Samenentwicklung auf. Ansonsten zeigen die untersuchten T-DNA-Insertionslinien einen ähnlichen Wuchs wie der Wildtyp auf. Auch konnten keine Unterschiede über die durchgeführten Zellwandanalysen, mittels GC-MS, MALDI und Dionex-HPLC verifiziert werden, was durch die Redundanz der MIOX- und UGD-Isoformen erklärt werden könnte. Nichtsdestotrotz konnte bei den Isoformen ΔMIOX1 & 2 eine dramatische Reduktion beim Einbau von 3H-Inosit in Zellwandpolymere von Keimlingen verzeichnet werden, was einen klaren Beweis für eine funktionelle MIOX liefert, da in diesem Gewebe diese beiden Isoformen die einzig aktiven sind. Außerdem konnten über Promotor-Deletions-Analysen potentielle cis-Elemente für die Promotoren von MIOX2 und MIOX4 aufgedeckt werden.
BMPs control postnatal dendrite growth and complexity in sympathetic neurons / von Afsaneh Majdazari

The vertebrate nervous system is a complex network of billions of neurons connected by dendrites and axons, integrated to functional circuits and areas/organs in the central and peripheral nervous system. The cells of the nervous system origin from common progenitors, which take on different cell fates based on intrinsic and extrinsic factors. These factors determine general neuronal traits, but also the morphology and the type of connections made to other cells. Mechanisms underlying axonal and dendritic growth are well described in contrast to the initiation of neurite growth, which remains to be fully elucidated, especially concerning dendrite formation. Recently BMPs have been identified as candidate dendrite inducing factors in sympathetic, cortical and hippocampal neurons. Here we focus on the in vivo role of BMPs on dendrite growth in sympathetic neurons as their development and differentiation processes have been analyzed in detail.
Sowohl die Gifte der Kegel- (Conidae) als auch die der Schraubenschnecken (Terebridae) enthalten eine Vielzahl pharmakologisch aktiver Peptide. Vor allem die Conopeptide bzw. Conotoxine aus den Giften der Kegelschnecken werden aufgrund ihrer Selektivität für Ionenkanäle und Rezeptoren seit langem als Werkzeuge in der neuropharmakologischen Forschung eingesetzt. Hier rücken gerade neuronale nikotinische Acetylcholinrezeptoren immer mehr in den Fokus der medizinischen Forschung, da sie vermutlich an der Entwicklung neurodegenerativer Erkrankungen wie Alzheimer, Parkinson, Demenz, Schizophrenie und Epilepsie beteiligt sind. Ziel dieser Dissertation war es daher, neue Inhibitoren in den Giften Kegelschnecken (Conidae) und der Schraubenschnecken (Terebridae) für nikotinische Acetylcholinrezeptoren, vor allem der neuronalen Subtypen, zu identifizieren. Es erfolgte die:
1. Identifizierung neuer αD-Conotoxine
Aus den Giften von Conus capitaneus und C. mustelinus konnten zwei native αDConotoxine (αD-CAP und αD-MUS) isoliert und charakterisiert werden. Beide Toxine sind Homodimere mit Molekulargewichten von 11 kDa und inhibieren nikotinische ACh-Rezeptoren. Sie blockieren die Subtypen α7>α3β2>α4β2, wobei sich αD-MUS als potenter als αD-CAP erweist (IC50-Werte von αD-MUS: α7 0,12 nM, α3β2 1,08 nM, α4β2 4,5 nM; IC50-Werte von αD-CAP α7 0,25 nM, α3β2 2,8 nM, α4β2 28,6 nM). Hingegen haben die αD-Conotoxine auf die Rezeptorsubtypen α3β4, α4β4 und α1β1γδ keinen hemmenden Einfluss. Zusätzlich konnten drei weitere αD-Conotoxine mit Hilfe der cDNA von C. vexillum und C. betulinus identifiziert werden. Eine Besonderheit hierbei war, dass innerhalb der Familie der αD-Conotoxine zwei unterschiedliche Signalsequenzen vorkommen und somit diese Sequenzen nicht stark konserviert sind.
2. Charakterisierung des α-Conotoxins SI aus dem Gift von C. striatus
Im Gift der Kegelschnecke Conus striatus wurde ein Peptid mit inhibierender Wirkung an α7-Rezeptoren nachgewiesen. Molekulare Masse (1.352,5 Da) und Aminosäuresequenz entsprachen dem α-Conotoxin SI, das als Antagonist muskulärer nACh-Rezeptoren bekannt ist. Da die Ergebnisse mehrere Jahre zurück lagen und bisher keine Analysen im Oozytenexpressions-System durchgeführt wurden, wurdeeine mögliche Aktivität sowohl an neuronalen als auch an muskulären nACh-Rezeptoren vermutet. Voltage Clamp-Messungen bestätigten die spezifische Wirkung am Muskeltyp, wodurch die Aktivität am α7-Rezeptorsubtyp einem anderen Conopeptid, zugewiesen werden muss, das als Beiprodukt isoliert wurde.
3. Identifizierung neuer Conotoxine der A-Superfamilie Mit molekularbiologischen Methoden unter Nutzung von cDNA-Bibliotheken gelang es, 27 Conotoxine (17 neue und 10 bekannte) aus der A-Superfamilie zu identifizieren: drei α- und zwei κA-Conotoxine aus Conus striatus, zwei α-Conotoxine aus C. betulinus, zwei α- und zwei κA-Conotoxine aus C. carinatus, drei α-Conotoxine aus C. catus, drei α- und zwei κA-Conotoxine aus C. circumcisus, ein α-Conotoxin aus. C. geographus, zwei aus C. imperialis, jeweils eines aus C. lividus, C. quercinus, C. sponsalis sowie zwei aus C. terebra Die Vielzahl der identifizierten α-Conotoxine belegt die hohe Diversität dieser Toxine in den Giften der Kegelschecken. Anhand von Vergleichen mit bereits bekannten Toxinen werden die möglichen Wirkungsweisen einiger neuer α-Conotoxine diskutiert. Für einen Teil der α-Conotoxine wurden 3D-Strukturmodelle erstellt, die Einblicke in die Bindung der Toxine an den Rezeptor geben können.
4. Untersuchung der Gifte der Terebridae
Die inhibierende Aktivität einiger Gifte (Terebra consobrina, T. argus, Myurella affinis, Acus felina, A. chlorata, A. maculata und Hastulopsis pertusa) an nACh-Rezeptoren (α7, α3β2, α4β2, α3β4, α4β4 und α1β1γδ) wurde erstmals nachgewiesen. An Kalium- und Natriumkanälen zeigten die Giftextrakte keine Wirkung. Die Giftextrakte von Myurella affinis und Acus maculata waren am potentesten und blockierten alle untersuchten nACh-Rezeptoren. Dies ist besonders ungewöhnlich, da diese Terebriden-Arten nach der Literatur (Puillandre & Holford, 2010) keinen Giftapparat besitzen sollen. Eine weitere Auffälligkeit aller Terebriden-Giftextrakte war neben der Selektivität für a7-Rezeptorsubtypen, eine hohe Aktivität gegenüber α4-enthaltenden Rezeptoren. In den Giften und mit Hilfe von cDNA-Bibliotheken von Kegelschnecken konnte eine Vielzahl neuer Inhibitoren für neuronale nikotinische Acetylcholinrezeptoren identifiziert werden. Sie zeigen ein breites Wirkungsspektrum, das die unterschiedlichsten nAChRSubtypen einschließt, was ihre Verwendung als pharmaklogische Werkzeuge begrenzt. Hingegen zeigen die Gifte der Schraubenschnecken ein Selektivitätsspektrum, das die Analyse ihrer Peptide als vielversprechend erscheinen lässt.
Helicobacter pylori (H. pylori) ist ein gram-negatives Bakterium, das die menschliche Magenmukosa kolonisieren kann. Ca. 50 % der Weltbevölkerung sind mit diesem Erreger infiziert, wobei er ohne medizinische Behandlung über Jahrzehnte in seinem Wirt persistieren kann. Das Bakterium gilt als eine der häufigsten Ursachen bei der Entwicklung von schweren gastrointestinalen Erkrankungen wie chronischer Gastritis und Gastral- oder Duodenalulkus. Darüber hinaus kann eine Infektion aber auch zu einem gastralen Adenokarzinom oder dem MALT- („mucosa-associated lymphoid tissue“) Lymphom führen. Bestimmte H. pylori-Stämme können über ein Typ IV Sekretionssystem (T4SS) das bakterielle Protein CagA („cytotoxin-assoziiertes Gen A“) in die Wirtszelle injizieren und dadurch Signaltransduktionswege stören, die die Morphologie und Mobilität der infizierten Wirtszelle drastisch verändern. In diesem Zusammenhang wurde die putative Phosphorylierung der Proteine VASP („Vasodilator-stimuliertes Phosphoprotein“) und α-Actinin-4 untersucht, welche beide regulatorische Funktionen im Zytoskelett der Wirtszelle ausüben. Im Rahmen dieser Doktorarbeit sollte der Einfluss von H. pylori auf diese Proteine analysiert werden, sowie die daraus resultierenden zellulären Auswirkungen. Es konnte verifiziert werden, dass H. pylori die Phosphorylierung der drei bekannten Phosphorylierungsstellen von VASP, bzw. eine Tyrosin-Phosphorylierung von α-Actinin-4 (ACTN4) induziert. Weiterhin zeigte sich, dass beide Proteine nach einer Infektion mit dem Bakterium in fokalen Kontaktstellen der Zellen lokalisieren. Zusätzlich lies sich zeigen, dass VASP und α-Actinin-4 eine entscheidende Rolle bei der durch H. pylori-induzierten Zellelongation spielen, da eine Herunterregulation der Genexpression von beiden Proteinen über siRNA zu einer Inhibition der morphologischen Veränderungen führte. Die genaueren Studien über VASP zeigten, dass hauptsächlich die Phosphorylierungsstelle VASPSer239 für die H. pylori-induzierte Zellelongation verantwortlich ist und die Phosphorylierung durch die Proteinkinase G (PKG) vermittelt wird. Für α-Actinin-4 konnte in dieser Arbeit des Weiteren gezeigt werden, dass die Kinase c-Abl eine Tyrosin-Phosphorylierung vermitteln kann, wobei die genaue Phosphorylierungsstelle im Protein noch ermittelt werden muss. Durch den Einsatz von H. pylori-Mutanten lies sich darüber hinaus noch zeigen, dass die Phosphorylierung von VASPSer239 und VASPThr278 durch das bakterielle Protein CagA deutlich verstärkt wird. Für die Tyrosin-Phosphorylierung von α-Actinin-4 war CagA sogar ausschlaggebend. Diese neu entdeckten zellulären Zielproteine bei einer H. pylori-Infektion ermöglichen weitere Einblicke in die Deregulation des eukaryotischen Zytoskeletts und möglichen Mechanismen bei der Krebsentstehung.
Hitze-Stress-Transkriptionsfaktoren (Hsfs) stellen die zentralen regulatorischen Komponenten einer Signal-Transduktionskette dar, welche die Aktivierung von Hitze-Stress-induzierbaren Genen bewirken. Die Sequenzierung des Genoms von Arabidopsis thaliana (Arabidopsis) führte zu der Entdeckung von 21 Genen, die für putative Hsfs codieren. Aufgrund struktureller Charakteristika und phylogenetischer Analysen wurden die 21 Hsfs in die Klassen A, B und C aufgeteilt. Der Hauptteil der vorliegenden Arbeit beschäftigt sich mit der funktionellen Charakterisierung der Hsf Familie aus Arabidopsis. Um generelle Konzepte zur Funktion von pflanzlichen Hsfs als Aktivatoren zu unterstützen, wurden als Ausgangspunkt ausgewählte Hsfs aus Lycopersicon peruvianum (Tomate) zu detaillierten Analysen herangezogen. Hsfs besitzen, ähnlich anderen Transkriptions -aktivierenden Proteinen, eine modulare Struktur. In der vorliegenden Arbeit wurden funktionelle Module der 21 Arabidopsis Hsfs untersucht, die für den Oligomerisierungs-Zustand, die intrazelluläre Lokalisation sowie das Transkriptions-aktivierende Potential von Bedeutung sind. Essentiell für die Funktion von Klasse A Hsfs als Transkriptions-Aktivatoren sind kurze Peptidmotive, die durch aromatische und große hydrophobe Aminosäure-Seitenketten geprägt sind, die in einer sauren Umgebung eingebettet sind (AHA Motive: aromatic, large hydrophobic and acid amino acid residues). Es wurde mit GST pull-down Assays gezeigt, dass AHA Motive mit ausgewählten Transkriptions-Komplexen interagieren. Das generelle Konzept für AHA Motive als kohäsive Elemente zur Interaktion mit der Transkriptions- Maschinerie wurde durch Mutationsanalysen verifiziert, besonders detailliert am Beispiel von LpHsfA2 und LpHsfA1 aus Tomate in Reporter-Assays in Tabak-Protoplasten und Hefe als auch GST pull-down Experimenten gezeigt. Im Kontrast zu den Klasse A Hsfs besitzen Klasse B und C Hsfs keine AHA Motive. Diese zeigten auch keine eigene Aktivator-Funktion aber Experimente deuten darauf hin, daß sie Coregulatoren für Klasse A Hsfs darstellen. Es wurde gezeigt, dass diese pflanzliche Besonderheit bei Tomate als auch Arabidopsis existiert. Alle 21 Arabidopsis Hsfs wurden als Transkripte nachgewiesen, ihre Expression verhält sich sehr dynamisch in bezug auf Hitze-Stress als auch Entwicklungssignale. Die präsentierte funktionelle Charakterisierung der Hsf Familie von Arabidopsis gibt erste Einsichten in das komplexe Netzwerk der Hsfs und demonstriert bereits, dass im Vergleich zu anderen Organismen, wie z.B. Drosophila und Hefe mit einem Hsf und Säugern mit drei Hsfs, das komplexe pflanzliche Hsf System eine fein regulierte und effiziente Voraussetzung für Pflanzen darstellt, um schnell und vor allem erfolgreich auf Umwelteinflüsse zu reagieren, denen sie nicht entfliehen können.
NOSTRIN belongs to the recently defined F-BAR protein family. F-BAR proteins are
multi-domain proteins, which serve as adaptors between plasma membrane and
cytoskeleton components in processes such as membrane protrusion formation,
endocytosis and migration. NOSTRIN encompasses a F-BAR domain at the N-terminus,
which mediates membrane association, followed by a HR1 motif and an intermediate
domain (ID) domain in the middle, and a SH3 domain at the C-terminus. The domain
architecture and ability to form oligomers enable NOSTRIN to coordinate several
interaction partners namely dynamin, caveolin, N-WASP and endothelial nitric oxide
synthase (eNOS) in the process of eNOS trafficking. In this context NOSTRIN was
originally identified and hence termed eNOS traffick inducer. NOSTRIN is expressed in
vascularized tissues (e.g. liver and lung) and in primary endothelial cells.
Aims of the present work were (1) to investigate if NOSTRIN is involved in other
processes besides eNOS trafficking, (2) to analyse the function of NOSTRIN in vivo
through knockdown of NOSTRIN in developing zebrafish and (3) to study the
consequences of the loss of NOSTRIN on signal transduction in a primary cell culture
model derived from NOSTRIN knockout mice.
To study the possible involvement of NOSTRIN in other processes besides eNOS
trafficking a yeast two-hybrid screen was performed in which fibroblast growth factor
receptor 1 (FGFR1) was identified as a putative novel interaction partner of NOSTRIN. In
a series of yeast two-hybrid, pulldown and co-immunoprecipitation experiments the
interaction between NOSTRIN and FGFR1 was confirmed to occur between
endogenously expressed proteins and determined to be direct and to depend on the ID
domain of NOSTRIN and the 130 C-terminal amino acid residues of FGFR1. FGFR1 is
activated by binding of fibroblast growth factors (FGFs) and induces several different
signal transduction pathways (e.g. MAPK and Akt pathway). Overexpression of
NOSTRIN in HeLa cells specifically enhanced FGF2-dependent MAPK activation.
Accordingly, depletion of NOSTRIN attenuated FGF2-dependent MAPK activation and
did not affect FGF2-induced Akt activation.
In summary, NOSTRIN has been identified as a novel interaction partner of FGFR1
involved in FGF2-dependent signal transduction.
The morpholino oligonucleotide-mediated knockdown of NOSTRIN in developing
zebrafish caused vascular leakage and irregular vascular patterning e.g. a loss of the
proper trajectory of intersegmental vessel and interruptions of the dorsal longitudinal
anastomotic vessel. The vascular phenotype was consistent upon use of two different
morpholinos and could be rescued in a dose dependent manner by the injection of
zebrafish NOSTRIN mRNA. Detailed analysis involving confocal and time lapse
microscopy in zebrafish with endothelial specific expression of EGFP revealed that the
knockdown of NOSTRIN impacts in vivo on the migration and morphology of endothelial
tip cells and leads to a reduction of filopodia number and length.
Additionally a NOSTRIN knockout mouse was generated. The analysis of FGFR1 signal
transduction in primary mouse lung endothelial cells (MLECs) from NOSTRIN knockout
and wild type mice revealed that FGF2-dependent MAPK activation was attenuated in
MLECs isolated from NOSTRIN knockout mice when compared to MLECs isolated from
wild type mice. The effect of NOSTRIN on FGF2-dependent signal transduction seems to
be specific, since VEGF-induced MAPK activation was not affected in NOSTRIN
knockout MLECs. The importance of NOSTRIN for FGF2 signal transduction in vivo is
demonstrated by the greatly impaired angiogenic response to FGF2 in NOSTRIN
knockout mice in matrigel plug assay. In a detailed biochemical analysis it was
discovered that NOSTRIN interacts with the activated small GTPase Rac1 and that
overexpression of NOSTRIN enhances Rac1 activation. Furthermore, the interactions of
NOSTRIN with both Rac1 and its GEF Sos1 are required for NOSTRIN-mediated
activation of Rac1. In accordance, activation of Rac1 was not detected upon FGF2
stimulation in NOSTRIN knockout MLECs.
In conclusion, the present work describes a novel function of the F-BAR protein
NOSTRIN in FGFR1 signal transduction. Data presented in this work demonstrate that
NOSTRIN is required for the assembly of a complex consisting of FGFR1, Sos1 and
Rac1 and subsequently for the FGF2-dependent activation of Rac1 in endothelial cells.
Das Y-Chromosom war, mit Ausnahme der Pseudoautosomalen Regionen, während der Genomevolution einer enormen Veränderung unterworfen, die zu einem weitesgehenden Verlust oder der Inaktivierung von Genen führte, die nicht in den Prozess der Geschlechtsdetermination bzw. –differenzierung, oder Spermatogenese involviert sind. Gleichzeitig kam es durch Amplifikation von funktionellen Kopien der Spermatogenesespezifischen Genen zu sog. amplifikonischen Genfamilien, die fast ausschliesslich im Testis exprimiert werden. TSPY (Testis Specific Protein Y-encoded) ist nach bisherigen Untersuchungen mit 35 Kopien, bestehend aus funktionellen Kopien und inaktiven Pseudogenen, die grösste und homogenste Protein-kodierende amplikonische Genfamilie auf dem Y-Chromosom und wurde im Rahmen dieser Arbeit genauer analysiert. Es wird unter normalen Bedingungen in den Spermatogonien des Testis exprimiert, aber es ist auch eine aberrante Expression in malignen Geweben, wie Gonadoblastom, Carcinoma in situ (CIS) des Testis oder in einer Fraktion von Prostatakarzinomen detektierbar. Aufgrund der Expression und der korrelierten Kopplungsanalyse wurde postuliert, dass es sich bei TSPY um ein Kandidatengen für den sog. Gonadoblastom-Locus auf dem Y-Chromosom (GBY) handelt. TSPY gehört ausserdem aufgrund einer evolutionär konservierten Domäne zur Familie der SET/NAP-Proteine. Andere Mitglieder dieser Familie sind z.B. in Proliferation, Zell-Zyklus-Regulation, Chromatin-Umbau und Kerntransport involviert. Durch die Analyse der TSPY-Transkription konnte anhand des LNCaP-Zellkultursystems die postulierte Androgen-abhängige TSPY-Transkription widerlegt werden. Zusätzlich wurden zwei TSPY-Transkripte unterschiedlicher Grösse und Transkriptionsraten detektiert, wobei das grössere Transkript TSPY-S stärker exprimiert wird als das kleinere Transkript TSPY-L. Ursache hierfür ist alternatives Splicing in Intron 4 des TSPY-Gens, das zur Expression der beiden Protein-Varianten mit unterschiedlichen C-Termini führt. Die durchgeführte Haplotyp-Analyse zur Identifizierung transkribierter TSPY-Gene konnte aufzeigen, dass das einzige funktionelle Gen des sog. TSPY minor Clusters (mit dem Haplotyp G-G-18) als einziges nicht alternativ gespliced wird und andererseits das häufigste TSPY-S Transkript (44 %) darstellt. Hierfür wird die Möglichkeit einer „locus-control-region“ diskutiert. Um Interaktionspartner von TSPY-S zu identifizieren und dadurch möglicherweise Hinweise auf die Funktion von TSPY zu erhalten, wurde das Hefe-Zwei-Hybrid-System eingesetzt. Vorversuche lokalisierten eine mögliche Transkriptions-Aktivierungsdomäne im N-Terminus von TSPY, weshalb eine N-terminale Deletion von TSPY-S, die einen grossen Teil der NAP Domäne und den C-Terminus enthält, als Köder eingesetzt wurde, um eine Testis-cDNABibliothek nach interagierenden Proteinen zu durchsuchen. Die Interaktion von TSPY mit den drei interessantesten Kandidaten hTid-1, dem menschlichen Homolog eines Tumor-Suppressor-Gens aus Drosophila, einem Mitglied der Ubiquitin-like-Proteinfamilie Ubiquilin 3 (UBQLN3), das aufgrund der Protein-Struktur eine Funktion bei der Regulation des Zellzyklus haben könnte und Isopetidase T (USP5), einer Ubiquitin-spezifische Protease, die an der proteasomalen Protein-Degradation beteiligt ist wurde durch Co-Transformation verifiziert und genauer analysiert. Immunhistochemische Experimente bestätigten die ausschliessliche zelluläre Lokalisation von TSPY in den Spermatogonien. Es konnten jedoch leichte entwicklungsspezifische Unterschiede gezeigt werden. In post-pubertärem und adulten Testis-Gewebe wird TSPY im Kern und Cytoplasma der mitotischen Spermatogonien exprimiert, aber in einem sehr grossen Anteil der ruhenden Spermatogonien im früh-kindlichen Testis (6 Monate) fehlt ein Kernsignal. Expression der EGFP (enhanced green fluorescent protein) -markierten Wildtyp cDNAs von TSPY-S und –L in HEK-293 Zellen bestätigte eine Protein-Lokalisation in Cytoplasma und Kern. Gezielte Deletion des C-Terminus der EGFP-TSPY-Fusionsproteine führte zur Degradation der Proteine, was die Bedeutung des variablen C-Terminus hervorhebt. Selektive Mutation einer mit den Computer vorhergesagten und durch einen Phosphorylierungs-Assay experimentell bestätigten CK2-Phosphorylierungs-Stelle im C-Terminus von TSPY-S verhinderte die Kern-Lokalisation, was darauf hindeutete, dass die C-terminale Phosphorylierung für die Lokalisation im Zellkern notwendig ist. Für TSPY-L konnte dieses Muster nicht bestätigt werden, da Mutationen im direkten C-Terminus von TSPY-L zur Protein-Degradation führten. Aufgrund der Datenlage wurde die Arbeitshypothese aufgestellt, dass TSPY im Cytoplasma durch CK2 phosphoryliert und in den Zellkern transportiert wird. Nach Androgen-Stimulus und Signal-Transduktion kommt es zur Kern-Lokalisation von CK2 und dadurch zur verstärkten Phosphorylierung von TSPY im Kern. Dort führt die Interaktion von TSPY mit den einzelnen identifizierten Proteinen zur Regulation des Zellzyklus und der mitotischen Proliferation der Spermatogonien, aber auch zur pathologischen Funktion von TSPY bei der Entstehung von Keimzelltumoren. Diese Arbeitshypothese müsste überprüft und verbessert werden, um die Funktion von TSPY bei der Proliferation der Spermatogonien und der Krebsentstehung aufzuklären.
Juvenile Neuronal Ceroid Lipofuscinosis (JNCL) is a rare inherited childhood neurodegenerative disease that is caused by a mutation in the gene CLN3. The function of the protein produced by the gene has remained elusive, and therefore the disease mechanism of JNCL is as of yet unknown. The disease is fatal, and no cure is currently available. We believe that simvastatin shows promise as a possible treatment. Simvastatin is well tolerated in children, and as currently no other viable, less invasive treatment for JNCL exists, at least pilot-scale clinical trials for this new off-label use of simvastatin are warranted.
The protein CLN3 has been indicated to have several different subcellular localizations and functions, but conclusive evidence about its role in cellular metabolism is lacking. It is also unclear why the mutation causes the distinct phenotype of the JNCL disease. In order to bring lucidity to the issue, we set out to identify metabolic pathways related to the phenotype of JNCL by using Multi-Epitope Ligand Cartography (MELC) and the related field of toponomics. Toponomic methods are required to process the massive amount of data generated by the MELC runs in order to extract information from them.
Our disease model of choice was the CLN3Δex7/8 knock-in mouse. To separate cause from effect, we compared embryonal wild type and mutant mouse brains to their adult counterparts. The first analyses revealed progressively abnormal Combinatorial Molecular Patterns (CMPs, an unit of toponomic data) related to cholera toxin/ganglioside 1 (Ctx/GM1), which is a membrane microdomain marker.
Cholesterol is an essential part of microdomains, so we utilized filipin staining to see if there were actual changes in cholesterol concentration and localization between healthy and diseased animals. After the disturbance in cholesterol metabolism was verified, we investigated the metabolic pathway that synthesizes cholesterol, the mevalonate pathway. Simvastatin is a drug that specifically down-regulates the mevalonate pathway. Fish oil affects lipid homeostasis and has some effects similar to those of simvastatin, and both of these drugs have previously been studied for their effects on neurodegenerative diseases. After treatment of mice with these drugs, highperformance liquid chromatography (HPLC) measurements on the brain homogenate showed a decrease in levels of farnesyl pyrophosphate (FPP) and geranyl-geranyl pyrophosphate (GGPP), products of the mevalonate pathway, confirming the effect of these drugs on the brains of the animals. Analyses of motor function of the mice further supported the notion that simvastatin had a positive effect on the condition of the diseased animals.
CMP analyses from the simvastatin treated mice showed a rescue of the Ctx/GM1 CMPs, suggesting at least a partial restoration of membrane microdomain homeostasis. Filipin staining revealed reversion of the apparent cholesterol depletion in the adult mutant mouse hippocampus by simvastatin. Interestingly, an additional effect of the treatment was found: simvastatin also affected glutamate receptor homeostasis, especially as regarding to N-methyl-D-aspartate (NMDA) and alphaamino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA) receptors. This finding suggested that excitotoxicity could be a part of the disease process, and pointed towards glutamate receptors as possible therapy targets. This is in line with previous studies that have shown that attenuation of AMPA receptors and L voltage-dependent channels improve the phenotype of a JNCL mouse and cell model, respectively.
Simvastatin mediates many of its effects via downregulation of the mevalonate pathway products, such as isoprenoids and cholesterol. However, simvastatin also has multiple pleiotropic effects that include suppression of excitotoxicity and granting neuroprotection. It is apparent that simvastatin treatment has a positive effect on JNCL mice, but if its effects are mediated via cholesterol (and membrane microdomains), isoprenoids (and isoprenylated proteins) or via a fully cholesterol independent mechanism remains to be solved.
In this study we have shown that with the MELC method and toponomics it is possible to approach rare diseases with confounded disease mechanisms with a hypothesis-free approach, to identify possible drug targets, and to monitor the effects of the drugs on treated individuals. This should open up a new avenue in the research of the many diseases that so far have avoided all attempts at discerning their nature.
Cell-cell adhesion is an essential process during the development of multicellular organisms. It is based on various cellular junctions and ensures a tight contact between neighboring cells, enabling interactive exchanges necessary for morphological and functional differentiation and maintaining the homeostasis of healthy tissue organization. Two important types of cell-cell adhesions are the adherens junction (AJ) and the desmosome which link the actin cytoskeleton and intermediate filaments to cadherin-based adhesion sites. The core of these structures is composed of single-span transmembrane proteins of the cadherin superfamily which include, among other members, the classical cadherins, e.g. E-cadherin, as well as the desmosomal cadherins, e.g. desmoglein-3. The cytoplasmic domains of the desmosomal and classical cadherins enable interactions with proteins of the catenin family. Classical cadherins preferentially associate with β-catenin and p120-catenin, whereas desmosomal cadherins bind to γ-catenin and plakophilins. Intriguingly, γ-catenin, also known as plakoglobin, is so far the only protein known to be present both in the AJ and the desmosome.
In this study, we showed that the two homologous, membrane raft-associated proteins flotillin-1 and flotillin-2 associate with core proteins of the AJ and the desmosome in vitro and in vivo. In confluent human, non-malignant epithelial MCF10A cells and human skin cryosections, flotillin-2 colocalized with E-cadherin, desmoglein-3 and γ-catenin at cell-cell contact sites, whereas flotillin-1 showed barely any overlap with these proteins. In addition, we detected a colocalization of both flotillins with the actin-binding protein α-actinin in membrane ruffles in subconfluent and at cell-cell contact sites in confluent MCF10A cells as well as in human skin cryosections. The interaction with α-actinin was later shown to be flotillin-1 dependent by performing indirect GST pulldown experiments with purified α-actinin-1-GST in MCF10A cell lysates.
Since flotillin-2 strongly colocalized with cell-cell junctions, this suggested that flotillins might be found in complex with cell adhesion proteins. Thus, we performed coimmunoprecipitation experiments in murine skin lysates and various cell lines of epithelial origin, such as human breast cancer MCF7 cells, human keratinocyte HaCaT cells and primary mouse keratinocytes. These experiments demonstrated that flotillins, especially flotillin-2, coprecipitated with E-cadherin, desmosomal cadherins and γ-catenin in relation to the respective cell type and the maturation status of these cell-cell adhesion structures. However, since γ-catenin is so far the only protein known to be present in the AJ and the desmosome, we further assumed that the complex formation of flotillins with cell adhesion structures is mediated by γ-catenin. For this, we performed indirect GST pulldown experiments in MCF10A cell lysates with bacterially expressed, purified flotillin-1-GST, flotillin-2-GST and γ-catenin-GST and were able to verify the complex formation of adhesion proteins and flotillins in vitro. To further test if the interaction of γ-catenin and flotillins is a direct one, we used purified flotillin-1-GST or flotillin-2-GST and γ-catenin-MBP fusion proteins. Both flotillins directly interacted with γ-catenin in this in vitro assay. In addition, mapping of the interaction domains in γ-catenin by using GST fusion proteins carrying different parts of γ-catenin suggested that flotillins bind to a discontinuous γ-catenin binding domain which consists of a Major determinant around ARM domains 6-12, most likely with a major contribution of the ARM domain 7, and possibly including the NT part of γ-catenin.
To study the effect of flotillin depletion on cell-cell adhesion, we generated stable MCF10A cell lines in which flotillins were knocked down by means of lentiviral shRNAs. Staining of E-cadherin and γ-catenin in these cells showed that the localization at the cell-cell borders was significantly altered after flotillin-2 depletion, which pointed to a role for flotillin-2 in the formation of cell-cell adhesion structures in epithelial cells. Furthermore, isolation of detergent resistant membranes (DRMs) from these cells demonstrated that upon depletion of flotillin-2, a significant amount of E-cadherin and γ-catenin shifted into raft fractions. On the contrary, no change was detected in flotillin-1 knockdown cells. These observations point to a functional role of flotillin-2 in the regulation of raft association of cell-cell adhesion proteins. To gain more insight into the in vivo relevance of our findings, we next studied the function of flotillins in the skin of Flot2-/- knockout mice. Analysis of lysates prepared from the skin of one year old female animals revealed an increased expression of E-cadherin, desmoglein-1 and γ-catenin but not β-catenin, implicating that specific adhesion proteins are upregulated in flotillin-2 knockout skin.
Since flotillins are tightly associated with membrane microdomains we next studied the interaction of flotillin-2 with membrane cholesterol. Using the photoreactive cholesterol analog azocholestanol, we were able to show that flotillin-2 and cholesterol directly interacted. In addition, previous studies speculated that flotillin-2 interacts with cholesterol via two putative cholesterol recognition/interaction amino acid consensus (CRAC) motifs. Analysis of the flotillin-2 sequence revealed that flotillin-2 actually contains four putative CRAC motifs. However, using various flotillin-2 CRAC mutant GFP fusion proteins, we were able to show that none of the putative CRAC motifs is functional, which suggested that flotillin-2 interacts with membrane cholesterol, e.g., via posttranslational modifications, such as myristoylation and palmitoylation which were previously shown to be essential for membrane association of flotillin proteins.
Während der vergangenen Jahrzehnte stieg die durchschnittliche Lebenserwartung der Bevölkerung in den westlichen Industrieländern durch die Verbesserung der allgemeinen Lebensbedingungen, insbesondere durch die Fortschritte in der Hygiene und der Medizin sowie stabile politische Verhältnisse, kontinuierlich an. Aufgrund dieser demographischen Entwicklung zu einer zunehmend älter werdenden Gesellschaft nimmt auch das Auftreten von progressiven, altersabhängigen Erkrankungen, wie zum Beispiel der Parkinson‟schen Krankheit zu. Dieser Trend stellt sowohl für die betroffenen Patienten und ihre Angehörigen als auch für die Gesundheits- und Sozialsysteme eine gewaltige und kostenintensive Herausforderung dar. Um wirkungsvolle Therapien entwickeln zu können, die früh im Krankheitsverlauf eingreifen und die Manifestation der Erkrankung verhindern oder verzögern beziehungs-weise die darauf abzielen, die Symptome der Erkrankung nach deren Manifestation zu lindern, ist es unerlässlich, die diesen progressiven, altersabhängigen Krankheiten zugrundeliegenden Mechanismen zu erforschen und entsprechende krankheitsspezifische, molekulare Biomarker zu identifizieren. Darüber hinaus stellt die Identifizierung solcher Biomarker einen wichtigen Ansatzpunkt für die klinische Diagnostik und Therapeutik sowie für die Entwicklung neuer therapeutischer Behandlungsstrategien dar. Das subzellulär vorwiegend präsynaptisch lokalisierte Protein alpha-Synuklein blieb in den Jahren nach seiner Erstbeschreibung 1988 durch Luc Maroteaux von der biomedizinischen Forschung weitgehend unbeachtet. Erst die Assoziationen von unterschiedlichen Mutationen des alpha-Synuklein-Gens mit seltenen, autosomal-dominant vererbten, monogenetischen Varianten der Parkinson‟schen Krankheit (PARK1 und PARK4) seit 1997 sowie die Identifizierung des Proteins im Jahre 1998 als Hauptbestandteil von intrazellulären Proteinaggregaten (Lewy-Körpern und Lewy-Neuriten), deren Vorkommen charakteristisch für progressive, neurodegenerative und unter dem Sammelbegriff „Synukleinopathien“ klassifizierte Erkrankungen (wie beispielsweise auch die häufigen, sporadischen Formen der Parkinson‟schen Krankheit) ist, ließen das alpha-Synuklein in den Fokus der biomedizinischen Forschung rücken. Trotz intensiver Bemühungen der weltweiten Forschungsgemeinschaft konnten seitdem in den vergangenen 13 Jahren die physiologischen Funktionen von alpha-Synuklein und die den unterschiedlichen Synukleinopathien zugrundeliegenden, molekularen pathophysiologischen Mechanismen nicht genau identifiziert werden. Stattdessen führte die intensive Forschung an alpha-Synuklein mit den unterschiedlichsten experimentellen Herangehensweisen und Modellsystemen zu verschiedenen und teilweise kontroversen Hypothesen und Theorien über dessen physiologische Funktion und pathophysiologische Wirkungsweisen. Die in dieser Dissertationschrift dargestellten experimentellen Untersuchungen wurden an zwei speziellen transgenen Mausmodellen durchgeführt, die entweder einen vollständigen Mangel (= „knockout“; KO) des alpha-Synuklein-Proteins oder eine transgene Überexpression von humanem, A53T-mutierten alpha-Synuklein aufwiesen. Das Hauptziel der dargestellten Studien war es, neue Erkenntnisse hinsichtlich der physiologischen Funktionen des alpha-Synuklein-Proteins, beziehungsweise der krankheits-relevanten, pathophysiologischen Mechanismen der den familiären PARK1- und PARK4-Varianten der Parkinson‟schen Krankheit zugrundeliegenden alpha-Synuklein-Mutationen (Substitution von Alanin durch Threonin an Position 53 der Aminosäuresequenz (A53T; PARK1) sowie Überexpression (Genduplikation/-triplikation; PARK4)) zu gewinnen...
Evolutionary genetics of bears and red foxes over phylogenetic and phylogeographic time scales

Climatic fluctuations during the Pleistocene (2.6-0.01 million years) have played an important role during evolution of many species. Cyclic range contractions and expansions had demographic consequences within species, provided environmental conditions for population divergence and speciation and enabled secondary contact and interspecific hybridization. These and other evolutionary processes have left genetic signatures in the genomes of affected organisms. Comprehensive and unbiased estimates of evolutionary processes can be obtained using genetic markers from different parts of the genome and by integrating population genetic and phylogenetic concepts.
Suitable for studies on evolutionary processes and patterns over different evolutionary time scales are bears (Ursidae) and foxes (Vulpes), which occupy a wide range of habitats and evolved during the past few millions of years. In my thesis, I therefore used bears and red foxes as study species to investigate the genetic variation within and between species and to obtain estimates of evolutionary relationships and divergence times of populations and species that I interpreted in a climatic context. Further, I investigated population genetic processes during the evolution of bears. My thesis includes three publications and one submitted manuscript, spanning different evolutionary time scales - from evolutionary relationships and processes among species (phylogenetic time scales, Publications I & II), among populations and closely related species in a geographical context (phylogeographic time scales, Publications II & III), to ongoing processes within species (population genetic time scales, Publication IV).
In Publication I (Kutschera et al. 2014, Mol Biol Evol 31(8):2004-2017), I studied bears at several nuclear markers from several individuals per species, complemented with markers from the Y chromosome. Using approaches based on a population genetic concept (coalescent theory) I obtained a species tree with divergence time estimates. Further, I studied two evolutionary processes in bears, interspecific gene flow and incomplete lineage sorting (ILS). This study contributed to the growing evidence that population genetic processes can be relevant on time scales up to several millions of years.
In Publication II (Hailer, Kutschera et al. 2012, Science 336(6079):344-347), we complemented previous mitochondrial (mt) DNA-based inference of the evolutionary history of polar and brown bears with nuclear DNA. Coalescence-based species tree analyses of multiple nuclear markers from several individuals per species placed polar bears as sister lineage to brown bears and their divergence time to about 600 thousand years ago (ka). This contrasted previous mtDNA-based inference. We explained this discrepancy between mtDNA and nuclear DNA with interspecific gene flow between polar and brown bears.
In Publication III (Kutschera et al. 2013, BMC Evol Biol 13:114), I studied range-wide phylogeographic events and their timing in red foxes. A synthesis of newly generated and published mtDNA sequences was analyzed using a coalescence-based approach with multiple fossil calibration points. Thereby, I validated the identity and geographic distribution of several red fox lineages and showed that red foxes colonized North America and Japan several times independently during the late Pleistocene (126-11 ka) and around the last glacial maximum (26.5-19 ka). In a comparison of my results from red foxes to brown bears and grey wolves, I identified similar phylogeographic patterns.
In Publication IV (Kutschera et al., submitted to Biol Conserv), I found similar levels of genetic variability in vagrant polar bears that had reached Iceland compared to established subpopulations from across the range. Based on climate projections reported by the Intergovernmental Panel on Climate Change in 2014, polar bear habitat will markedly decline and become increasingly fragmented within the next decades. Dispersal will play an important role by connecting isolated subpopulations, thereby maintaining genetic diversity levels. My results indicate that vagrants could stabilize genetic variability when immigrating into established subpopulations.
In conclusion, my thesis provided a deeper understanding of evolutionary genetic processes and patterns and their timing in bears and red foxes in a climatic context, which can have conservation implications. Further, I showed that processes like ILS and interspecific gene flow can be relevant over different time scales and are important aspects of evolutionary history. Thereby, my thesis contributed to the knowledge on the evolutionary history of several carnivore species and on evolutionary processes acting within and between closely related species.

In dieser Arbeit konnten durch chemische Mutagenese drei Pigmentmutanten erzeugt werden, die entsprechend ihrer Pigmentierung in drei Kategorien eingeteilt wurden:hellorange (OC-Mutante), gelb (YC-Mutante) und weiß (WH-Mutante). 2. Durch MS- und NMR-Analysen konnte die Struktur des von H. halophilus produzierten Carotinoids als Methylglykosyl-3,4-dehydro-apo-8’-lycopenoat aufgeklärt werden. Zusätzlich wurden auch die von der OC-Mutante produzierten Carotinoide als Hydroxy-3,4-dehydro-apo-8’-lycopin und Methylhydroxy-3,4-dehydro-apo-8’- lycopinoat identifiziert. Bei allen Pigmenten handelt sich um 8’-Apoderivate mit einer asymmetrischen Anordnung der Methylgruppen. Diese Art von Carotinoiden konnte zuvor nur in dem marinen Isolat P. maritimus gefunden werden (Shindo et al., 2008) 3. Auf Basis der genetischen und biochemischen Daten hinsichtlich der Carotinoide in H. halophilus konnte ein neuartiger Biosyntheseweg postuliert werden. Dabei beginnt die Synthese des C30-Carotinoids nicht wie bei allen bekannten Pigmenten dieser Art mit der Kondensation von zwei C15-Körpern, sondern durch die Kondensation von einem C10- mit einem C20-Molekül. Durch anschließende Desaturierungen, Hydroxylierungen, Methylierungen und Glykosylierungen entsteht das in H. halophilus identifizierte Carotinoid Methylglykosyl-3,4-dehydro-apo-8’-lycopenoat. Dieser Biosyntheseweg ist bisher einzigartig. 4. Die Carotinoide in H. halophilus sind essentiell für den Schutz der Zellen vor oxidativen Schäden. Während das Wachstum des pigmentierten Wildtyps nur wenig durch die Zugabe von bis zu 150 μM Duroquinon beeinträchtigt wurde, wurde das Wachstum der farblosen Pigmentmutante WH schon ab einer Duroquinonkonzentration von 120 μM vollständig gehemmt. Auch durch in vitro-Versuche konnte die antioxidative Eigenschaft von Methylglykosyl-3,4-dehydro-apo-8’-lycopenoat und der Derivate Hydroxy-,4-dehydro-apo-8’-lycopin und Methylhydroxy-3,4-dehydro-apo-8’-lycopinoat bestätigt werden. Dabei war die Wirksamkeit des Carotinoids aus H. halophilus sogar besser als die der biotechnologisch genutzten Pigmente ß-Carotin und Astaxanthin. 5. Durch Wachstumsexperimente konnte gezeigt werden, dass die Carotinoide auch eine Rolle in der Salzadaptation des Bakteriums spielen. Bei niedrigen Salinitäten zeigten der Wildtyp und die farblose Pigmentmutante keine Unterschiede im Wachstum. Eine hohe NaCl-Konzentration von 3,0 M NaCl führte jedoch zu einem verlangsamten Wachstum der Carotinoidmutante WH. 6. Im Genom von H. halophilus konnten anhand von Sequenzvergleichen mit bereits bekannten Kompetenzproteinen aus B. subtilis eine Reihe von putativen Genen identifiziert werden, die dem Bakterium theoretisch die Fähigkeit zur Ausbildung einer natürlichen Kompetenz verleihen. 7. Um die Fähigkeit von H. halophilus zur natürlichen DNA-Aufnahme zu testen, wurde eine Methode entwickelt, durch die es möglich war, viele unterschiedliche Bedingungen zu analysieren. Als Donor-DNA wurde die chromosomale DNA einer Erythromycin-resistenten Mutante von H. halophilus eingesetzt, die innerhalb dieser Arbeit erzeugt wurde. Insgesamt konnten so über 3000 Ansätze getestet werden. Jedoch konnte bei keiner dieser Bedingungen eine natürliche Kompetenz von H. halophilus festgestellt werden. 8. Innerhalb dieser Arbeit ist es gelungen eine Methode zur effizienten Transformation von H. halophilus zu etablieren. Dabei erfolgte die DNA-Aufnahme durch eine PEGvermittelte Protoplastenfusion. Mit dieser Methode konnten für H. halophilus Transformationseffizienzen von 2-102 Transformanden/μg DNA erreicht werden. 9. Durch die Möglichkeit H. halophilus transformieren zu können, ist es in dieser Arbeit erstmalig gelungen, ein System zur markerlosen Deletion von Genen in H. halophilus zu etablieren. Dabei wurde in einem ersten Schritt ein nicht-replizierendes Plasmid, das den gewünschten mutierten Locus enthält, durch Protoplastenfusion in H. halophilus transformiert. Durch einfach homologe Rekombination konnte das Plasmid in das Genom integrieren. Die so erhaltenen Integranten wurden im zweiten Schritt unter nicht-selektiven Bedingungen für etwa 90 Generationen kultiviert. Hierbei kam es zu einer zweiten homologen Rekombination, wodurch das Plasmid wieder aus dem Genom geschnitten wurde (Segregation). Dies resultierte entweder in den Wildtyp-Locus oder in den gewünschten mutierten Locus. Potentielle Integranten wurden auf einen CmS-Phänotyp getestet und mit Hilfe von Southern-Blot-Analysen verifiziert. 10. Mit Hilfe des in dieser Arbeit etablierten Systems zur markerlosen Mutagenese von H. halophilus wurde eine pro-Mutante generiert, deren proHJA-Operon deletiert war. 11. Überraschenderweise konnte die Mutante immer noch Prolin synthetisieren und war demnach nicht prolinauxotroph. Jedoch war H. halophilus pro nicht mehr in der Lage, Prolin als ein kompatibles Solut zu synthetisieren. Trotzdem zeigte das Wachstum der Mutante unter Hochsalzbedingungen keine Beeinträchtigung. Dies konnte auf eine gesteigerte Akkumulation von Glutamat, Glutamin und Ectoin zurückgeführt werden, wodurch der Verlust von Prolin als Osmolyt kompensiert wurde. 12. Die Expression der Gene glnA2, gltA und ectA, die für Biosyntheseenzyme von Glutamin, Glutamat und Ectoin kodieren, ist in H. halophilus pro induziert. Dabei hatte die Salinität einen stärkeren Einfluss auf die relativen RNA-Mengen von glnA2 und ectA in der pro-Mutante als im Wildtyp. gltA zeigte keine salzabhängige Expression im Wildtyp, der mRNA-Level des Gens stieg jedoch in H. halophilus pro bei hohen Salinitäten um das Doppelte im Vergleich zu Niedrigsalzbedingungen an. 13. Für die Glutaminsynthetase, dem Enzym der Glutaminsynthese, konnte eine gesteigerte spezifische Aktivität nachgewiesen werden. Dabei erhöhte sich die Aktivität mit steigender Salinität, sie war jedoch immer 30 bis 50% höher als im Wildtyp. 14. In dieser Arbeit wurde ein möglicher Biosyntheseweg zur salzunabhängigen Prolinsynthese aufgestellt. Hierbei dient Ornithin als Ausgangsmolekül, das entweder durch eine Ornithin-Aminotransferase (RocD) und eine Pyrrolin-5-Carboxylat-Reduktase (ProC und ComER) oder direkt durch eine Ornithin-Cyclodeaminase (ArcB) zu Prolin umgesetzt wird. Für alle Enzyme konnten die entsprechenden Gene im Genom von H. halophilus identifiziert werden. 15. Vor dieser Arbeit konnte bereits gezeigt werden, dass Prolin das dominante kompatible Solut unter Hochsalzbedingungen ist. Dabei war die Expression des pro-Operons sowohl von der Salzkonzentration als auch vom Anion Chlorid abhängig. Es zeigte sich, dass auch die zelluläre Konzentration von ProJ salzabhängig war, mit einem Maximum bei 2,0 – 3,0 M NaCl. Zudem war der ProJ-Gehalt wachstumsphasenabhängig. Während das Enzym besonders in frühexponentiellen Zellen detektiert werden konnte, sank die zelluläre ProJ-Konzentration kontinuierlich in stationären Zellen. Nach einem Salzschock von 0,8 auf 2,0 M NaCl stieg die ProJMenge nach 2 h leicht an und sank nach 4 h wieder kontinuierlich. Der ProJ-Level war in Gegenwart von NaCl maximal, sank aber nur leicht um 25%, wenn die Zellen mit 2 M Na-Glutamat oder Na-Nitrat inkubiert wurden. Die Prolinbiosynthese in H. halophilus wird demnach auch auf Ebene der Translation und/oder der Proteinstabilität reguliert. 16. Die Aminosäure Prolin wird nicht nur als kompatibles Solut in H. halophilus synthetisiert, sondern kann auch als C- und Energiequelle und als Stickstoffquelle genutzt werden. 17. Liegt Prolin als alleinige C- und Energiequelle im Medium vor, erfolgt eine salzabhängige Akkumulation des Osmolyts in H. halophilus, während der zelluläre mRNA-Level der pro-Gene stark reduziert ist. Prolin wird demnach nicht nur als Cund Energiequelle aufgenommen, sondern auch als kompatibles Solut. 18. Im Genom von H. halophilus konnten zwei Gene, die für die potentiellen Na+/Prolin- Symporter 3541 und 1381 kodieren, identifiziert werden, deren mRNA-Level bei Wachstum auf Prolin induziert war. 19. Durch Aminosäuresequenzvergleiche konnten in dieser Arbeit im Genom von H. halophilus jeweils zwei Isogene gefunden werden, die für potentielle Prolindehydrogenasen (prodh1/prodh2) und Pyrrolin-5-Carboxylat-Dehydrogenasen (p5cdh1/p5cdh2) kodieren. Mittels reverser Transkription von mRNA und anschließender PCR-Analysen konnte gezeigt werden, dass prodh2 und p5cdh2 ein Operon bilden (put-Operon). Eine Quantifizierung der Transkriptmengen der Abbaugene prodh1/p5cdh1 und prodh2/p5cdh2 mittels quantitativer PCR in Zellen, die in Gegenwart von Prolin als alleiniger C- und Energiequelle oder Stickstoffquelle gezogen wurden, zeigten deutlich, dass unter diesen Bedingungen nur die Expression von prodh2/p5cdh2 induziert wurde. Wurden die Zellen in Gegenwart von Glukose als C- und Energiequelle gezogen, stieg die mRNA-Menge von prodh2/p5cdh2 unter Hochsalzbedingungen um den Faktor 10 – 20 im Vergleich zu Zellen, die bei niedrigen Salinitäten kultiviert wurden, an. Die mRNA-Menge des put- Operons war in Glukose-gewachsenen Zellen wachstumsphasenabhängig mit einem Maximum in der stationären Wachstumsphase. In Gegenwart von 1,0 M NaCl stieg die mRNA-Menge um das 20-Fache, in Gegenwart von 3,0 M NaCl um das Doppelte. 20. Mit Hilfe des Systems zur markerlosen Mutagenese in H. halophilus konnte eine glnA2-Mutante generiert und verfiziert werden. H. halophilus glnA2 zeigte keinen Wachstumsphänotyp bei Anzucht in Gegenwart von unterschiedlichen NaCl-Konzentrationen. Die Deletion von glnA2 führte zu einer Inhibierung der salzabhängigen Prolinbiosynthese, während der Glutamat- und Glutaminpool bei allen getesteten Salinitäten im Vergleich zum Wildtyp nicht verändert war. Der Verlust von Prolin als kompatibles Solut konnte teilweise durch einen Anstieg des Ectoinlevels kompensiert werden. Die zelluläre Transkriptmenge von glnA1 und ectA war in H. halophilus glnA2 stärker von der Salinität beeinflusst als im Wildtyp, während die salzabhängige Regulation der proH-Expression aufgehoben wurde. Im Gegensatz zum Wildtyp wurde die spezifische Aktivität der Glutaminsynthetase in H. halophilus glnA2 durch steigende NaCl-Konzentrationen inhibiert.
The role of small leucine-rich proteoglycans, biglycan and decorin, in podocytopathy and albuminuria

Biglycan is a member of the small leucine-rich proteoglycan (SLRP) family and is involved in the assembly of extracellular matrix components. In macrophages soluble biglycan acts as an endogenous ligand of the innate immunity receptors TLR2 and TLR4. Data addressing the role of biglycan in renal pathology are surprisingly limited. In a normal kidney, biglycan is expressed mainly in the tubulointerstitium; however, in the course of various renal diseases its expression may be altered. The biological role and mechanisms of biglycan action in the pathology of renal diseases, especially those affecting glomeruli, remain poorly understood.
Albuminuria is the first detectable clinical abnormality in diabetic nephropathy. In this study we detected increased biglycan mRNA expression in glomeruli of renal biopsies of patients with incipient diabetic nephropathy, with predominant localization in podocytes. This novel finding raised the question about the role and mechanisms of biglycan action in diabetic podocyte injury and whether the mechanisms of biglycan signaling causing podocyte injury and albuminuria could be extrapolated to other glomerular diseases.
To investigate the role of biglycan in the cause of diabetic podocyte injury and albuminuria we used the murine model of STZ-induced diabetic nephropathy and wild type (Bgn+/0) and biglycan deficient (Bgn-/0) mice. We observed that biglycan was expressed on mRNA and protein levels in podocytes of diabetic Bgn+/0 mice and that diabetic Bgn+/0 mice also had significantly higher albuminuria compared to non-diabetic mice 6 and 12 weeks after disease induction. Biglycan deficiency was shown to be an important factor in albuminuria development. Namely, we observed that diabetic Bgn-/0 mice had significantly lower levels of urinary albumin compared to diabetic Bgn+/0 mice. We showed that less severe podocyte loss in the urine of diabetic Bgn-/0 mice was associated with significantly higher nephrin and podocin glomerular expression compared to diabetic Bgn+/0 mice. Our data suggested that biglycan deficiency was protective against podocyte loss into urine and might be beneficial against development of albuminuria in diabetes.
Biglycan contributed to podocyte actin rearrangement due to increased phosphorylation of Rac1 in vitro. Furthermore, biglycan induced caspase-3 activity and production of reactive oxygen species (ROS), thus enhancing apoptosis in cultured podocytes. Biglycan-induced ROS generation was TLR2/TLR4-dependent. Overexpression of soluble biglycan in wild type mice induced albuminuria under normal conditions and significantly increased albuminuria under pathological conditions (murine model of LPS-induced albuminuria). Inhibition of Rac1 activity in vivo decreased the albuminuria induced by biglycan overexpression. In patients with glomerular diseases, biglycan was detected in urine and was associated with nephrin appearance in the urine of these patients and with increased albuminuria. Collectively, our results elucidate a novel mechanism for biglycan-induced TLR2- and TLR4-dependent, Rac1- and ROS-mediated podocytopathy leading to podocyturia, albuminuria development and progression of glomerular diseases. Interfering with biglycan actions and blocking its signaling via TLR2 and TLR4 might be a potential therapeutic strategy against these diseases. To achieve this goal, the specific mechanisms for binding of biglycan to TLR2 and TLR4 must be elucidated and effective ways of preventing this binding must be developed. Nevertheless, biglycan remains the “danger signal” that activates innate immune receptors in non-immune cells and triggers the deleterious mechanisms leading to aggravation of renal injury.
Die Chloroplastenbewegung ist eine der wichtigsten Anpassungen, die Pflanzen entwickelt haben, um eine effiziente Ausbeute an Lichtquanten für die Photosynthese zu gewährleisten. Auch wenn der genaue Mechanismus und die Signalwege, die diesen Prozess vermitteln, noch nicht vollständig verstanden sind, konnten einige an der Chloroplastenbewegung beteiligte Proteine (phot1, phot2, chup1, Aktin, Profilin) in den letzten Jahren identifiziert werden. Chup1, das an der äußeren Chloroplastenmembran verankerte Protein, wird als putativer Linker zwischen den Chloroplasten und dem Aktin-Zytoskelett gesehen. Durch die Interaktion mit Profilaktin reguliert chup1 die Aktin-Polymerisierung und somit auch die Chloroplastenbewegung. Die Analyse der intra- und intermolekularen Interaktionen von chup1, die in dieser Studie ermittelt wurden, deutet auf eine Homodimerisierung von chup1 durch die Coiled coil Domäne sowie auf eine Assoziation des N- mit dem C-terminalen Leuzin- Zipper hin. Neben diesen Interaktionen konnte für einen der vier putativen Interaktionspartner von chup1, der wall associated kinase 3, eine mögliche Funktion bei der Vermeidungsbewegung der Chloroplasten gezeigt werden. In Anbetracht dieser Interaktion und der postulierten Phosphorylierungsstellen in chup1, könnte eine Regulierung von chup1 durch Phosphorylierung erfolgen. Um die Funktionen von chup1 und phot2 besser zu verstehen, wurden die Pflanzen mit knock-out in diesen Gene genauer charakterisiert und die T-DNA Insertionslinien von chup1 und phot2 zeigen keine Reduzierung in der photosynthetischen Aktivität. Im Gegensatz dazu, bedingt der knock out von beiden Genen eine deutliche Verminderung der photosynthetischen Leistung. Infolge der fehlenden Chloroplastenbewegung im chup1phot2 Doppel knock out führt das möglicherweise zu einem verringerten Schutz der Photosynthese. Dies bestätigt weiter die Verbindung zwischen der durch phot2 induzierten Signalkaskade und der Aktin-Polymerisierung, die durch chup1 reguliert wird. Um den Einfluss von chup1, phot1 und phot2 auf die transkriptionelle Regulierung nach BL zu analysieren, wurde das globale Expressionsmuster nach BL-Behandlung untersucht. Bei der Analyse der Mutanten-Pflanzen mit einem Defekt in der Chloroplastenbewegung (chup1, phot1, phot2) konnte keine Beeinflussung der Regulierung der Genexpression in BL-gesteuerten Signalkaskaden durch diese Proteine beobachtet werden. Die Regulierung der Expression geschieht eher auf posttranskriptioneller Ebene und wird mit Hilfe von microRNA gesteuert.
Ribosome biogenesis is best understood in the yeast Saccharomyces cerevisiae. In human or mammalian ribosome biogenesis, it has been shown that basic principles are conserved to yeast, but additional features have been reported. Our understanding about the interplay between proteins and RNA in human ribosome biogenesis is far from complete.
The present study focused on the analysis of the human ribosome biogenesis co-factors PWP2, EMG1 and Exportin 5 (XPO5) to understand the degree of conservation of ribosome biogenesis. The proteins were characterized in respect to their localization and interaction partners. For the early 90S co-factor, PWP2, it was possible to pull down and identify the human UTP-B complex with MALDI mass spectrometry. Besides the orthologues of the members of this complex known in yeast (TBL3, WDR3, WDR36, UTP6, UTP18), the human UTP-B complex is not only conserved from yeast to humans, but contains also additional components, like the DEAD-box RNA helicase DDX21, which lacks a yeast orthologue. DDX21 was localized to the nucleus, assembled to the native UTP-B complex and co-precipitated also with other UTP-B complex members, presumably extending the functions of this complex in ribosome biogenesis.
This phenomenon was also observed for the 90S co-factor EMG1, an RNA methyltransferase, whose mutant form causes the Bowen-Conradi syndrome, if aspartic acid is mutated to glycine at position 86. This study revealed that the mutant, EMG1-D86G, clearly lost its nucleolar localization and co-precipitated to histones for unknown reasons.
A participation of the nuclear export receptor XPO5 in human ribosome biogenesis was shown in this study. Pulldown analysis, sucrose density gradients and UV crosslinking and analysis of cDNAs of XPO5 revealed the involvement of XPO5 in pre-60S subunit maturation. Moreover, besides the known pre-miRNAs and tRNAs as substrates for nuclear export, XPO5 crosslinked to snoRNAs. XPO5 was further demonstrated to interact with the miRNA Let-7a, which has an important regulatory function for MYC, a transcription factor required for ribosome biogenesis.
All results support a role of these proteins in human ribosome biogenesis and therefore it seems that the biogenesis of ribosomes in human cells requires additional components, like DDX21 and XPO5.
Gepaarte assoziative Magnetstimulation (PAS) kann im primären menschlichen Motorkortex (M1) sowohl langzeitpotenzierungs- (LTP) als auch langzeitdepressionsähnliche (LTD) Erregbarkeitsveränderungen hervorrufen. Dies kann durch die Untersuchung magnetisch evozierter Potentiale (MEP) erfasst werden. Dagegen ist wenig über die Auswirkungen von PAS auf willkürliche Aktivität des motorischen Kortex bekannt. Im ersten Experiment haben wir bewegungsabhängige kortikale Potentiale (MRCP) bei zehn gesunden Probanden im EEG registriert, um die willkürliche Aktivität im Motorkortex während der Vorbereitung zweier motorischer Aufgaben zu erfassen. Die Probanden mussten dabei entweder den Daumen abduzieren (Hauptmuskel: Musculus abductor pollicis brevis, APB) oder das Handgelenk strecken (Hauptmuskel: Musculus extensor carpi radialis, ECR). Die Amplituden der motorisch evozierten Potentiale im APB wurden dabei durch PASLTP gesteigert, durch PASLTD vermindert und blieben bei PAScontrol unverändert. Im Gegensatz dazu wurden sie im ECR durch keine PAS-Bedingungen verändert. PASLTP verminderte die Negativität der MRCP während des späten Bereitschaftspotentials (-500 bis 0 ms vor Bewegungsbeginn) nur in der APB-Aufgabe. Diese Veränderungen zeigten sich hauptsächlich über zentralen Elektroden kontralateral zur bewegten Hand. Dieser Effekt korrelierte negativ mit dem durch PASLTP induzierten MEP-Anstieg im APB. PASLTD und PAScontrol hatten dagegen keinen Einfluss auf die MRCP Amplituden. Unsere Ergebnisse deuten auf eine spezifische Wechselwirkung von PAS mit willkürlicher Aktivität im Motorkortex während der Vorbereitung motorischer Aufgaben hin. Dies könnte durch ein Zusammenspiel aus erhöhter Exzitabilität von M1 und einer unterbrochenen effektiven Konnektivität zwischen prämotorischen Arealen und M1 erklärt werden. Die Modulation des dorsolateralen prämotorischen Kortex (PMd) durch repetitive transkranielle Magnetstimulation (rTMS) verändert die kortikospinale Erregbarkeit in M1. Die Auswirkungen von PMd-rTMS auf vorbereitende Prozesse für willkürliche Bewegungsabläufe sind jedoch unklar. Contingent negative variation (CNV) repräsentiert im EEG kortikale Vorbereitungsprozesse äußerlich getriggerter Bewegungen während das Bereitschaftspotential (BP) Vorbereitungsprozesse intern getriggerter Bewegungen repräsentiert. Im zweiten Experiment wurden CNV und BP jeweils vor und nach PMd-rTMS untersucht. Das Experiment bestand aus drei CNV-Versuchsblöcken mit insgesamt 243 Durchgängen. Dabei mussten die Probanden auf visuelle Anweisung hin eine zwei-Item Finger-Bewegungssequenz durchführen. RTMS wurde sowohl mit 1 Hz als auch mit 5 Hz bei einer Intensität von 110% der aktiven motorischen Schwelle (AMT) unter individueller MR-Navigation appliziert. Die Erfassung des BP erfolgte während der Durchführung derselben motorischen Aufgaben, allerdings bekamen die Probanden keine Anweisungen. Die Durchschnittsamplituden der frühen und späten Komponente von CNV (CNV1:1500-500 ms vor dem Startsignal (S2); CNV: 500-0 ms vor S2) und der frühen und späten Komponente des BP (BP1: 1500-500 ms vor EMG Beginn; BP2: 500-0 ms vor EMG-Beginn) wurden quantitativ für 25 zentrale Elektrodenpositionen verglichen. CNV2 zeigte eine signifikante Bahnung über dem frontal-zentralen Bereich nach 1 Hz PMd-rTMS, blieb aber unverändert nach 5 Hz PMd-rTMS. CNV1, BP1 und BP2 blieben durch 1 Hz und 5 Hz PMd-rTMS unbeeinflusst. Diese Ergebnisse lassen vermuten, dass der dominante PMd eine wichtigere Rolle in der Vorbereitung extern getriggerter Bewegungen spielt, als dies bei intern getriggerten Bewegungen der Fall zu sein scheint. Die CNV2-Antwort könnte eine intensive Interaktion innerhalb des menschlichen motorischen Kontrollnetzwerks anzeigen, die möglicherweise auf kompensationsähnlichen Mechanismen beruht.
The biogenesis and function of photosynthetically active chloroplasts relies on the import of thousands of nuclear encoded proteins via the coordinated actions of two multiprotein translocon machineries in the outer and inner envelope membrane. Trafficking of preproteins across the soluble compartment of InterMembrane Space (IMS) is currently envisioned to be facilitated by an IMS complex composed of outer envelope proteins Toc64 and Toc12, a soluble IMS component, Tic22 and an IMS-localized Hsp70. Among them, currently Tic22 is the only component that stands undisputed in terms of its existence. Having two closely related homologs in A. thaliana, their biochemical and functional characterization was still lacking. A critical analysis of Tic22 knockout mutants displayed growth phenotype reminiscent of ppi1, the mutant of Toc33. However, both the genes have similar expression patterns with no clear preference for photosynthetic or nonphotosynthetic tissues, which explained the absence of a detectable phenotype in single mutants. In addition, transgenic complementation study with either of the homolog affirmed the identical localization of both proteins in the IMS which characterizes the two homologs as functionally redundant. Based on the pale-yellow phenotype exhibited by the double mutant plants, an attempt to analyze the import capacity of a stromal substrate in the double mutant revealed threefold reduction when compared to wild-type acknowledging the essential role of Tic22 in the import mechanism. Initially, Tic22 was identified together with another protein, Tic20, which has been heavily discussed as a protein conducting channel in the inner membrane. Despite being characterized, in A. thaliana, two out of four homologs of Tic20 are differentially localized with one being additionally localized in mitochondria and the other, exclusively residing in the thylakoids.
According to in silico analysis, for all the Tic20 proteins, a four-helix transmembrane topology was predicted. Accordingly, its topology was mapped by employing the recently established selfassembling GFP-based in vivo experiments. Astonishingly, the expression of one of the inner envelope localized Tic20 homolog enforces inner membrane proliferation affecting the shape and organization of the membrane. Therefore this study focuses on analyzing the effects of high envelope protein concentrations on membrane structures, which together with the existing results, an imbalance in the lipid to protein ratio and a possible role of signaling pathway regulating membrane biogenesis is discussed.
ß1-integrins are essential for angiogenesis but the mechanisms regulating integrin function in endothelial cells (EC) and their contribution to angiogenesis remain elusive. BRAG2 is a guanine nucleotide exchange factor for the small Arf-GTPases Arf5 and Arf6. The role of BRAG2 in EC and angiogenesis and the underlying molecular mechanisms remains unclear. siRNA-mediated BRAG2-silencing reduced EC angiogenic sprouting and migration. BRAG2-siRNA-transfection differentially affected a5ß1- and aVß3-integrin function: specifically, BRAG2-silencing increased focal/fibrillar adhesions and EC adhesion on ß1-integrin-ligands (fibronectin and collagen), while reducing the adhesion on the aVß3-integrin-ligand, vitronectin. Consistent with these results, BRAG2-silencing enhanced surface expression of a5ß1-integrin, while reducing surface expression of aVß3-integrin. Mechanistically, BRAG2 mediated recycling of aVß3-integrins and endocytosis of ß1-integrins and specifically of the active/matrix bound a5ß1-integrin present in fibrillar/focal adhesions (FA), suggesting that BRAG2 contributes to the disassembly of FA via ß1-integrin-endocytosis. Arf5 and Arf6 are promoting downstream of BRAG2 angiogenic sprouting, ß1-integrin-endocytosis and the regulation of FA. In vivo silencing of the BRAG2-orthologues in zebrafish embryos using morpholinos perturbed vascular development. Furthermore, in vivo intravitral injection of plasmids containing BRAG2-shRNA reduced pathological ischemia-induced retinal and choroidal neovascularization. These data reveals that BRAG2 is essential for developmental and pathological angiogenesis by promoting EC sprouting through regulation of adhesion by mediating ß1-integrin internalization and associates for the first time the process of ß1-integrin endocytosis with angiogenesis.
This study comprises a survey on ecology, morphology and taxonomy of parasitic fungi infecting Pteridophytes and Orchidaceae found by the author on several field trips to Western Panama as part of the project plant parasitic micro-fungi of Western Panama (ppMP). In Panama, approximately 9500 species of vascular plants are found. Of these, Orchidaceae are with ca. 1150 (ca. 12%) species by far the most speciose family. The Pteridophytes in Panama comprise ca. 940 species in 31 families. Most fungal pathogens on Orchidaceae in tropical regions were described from plants in culture or from material intercepted at borders by plant quarantine services and not from their natural habitats. Therefore, little is known about distribution and ecology of these pathogens in their natural range. The author determined and classified several hundred Orchidaceae-species and Pteridophytes at the sites selected in the context of the project. This work facilitated the identification of many host plants (at least to genus-level) even in sterile condition in the field. About 65 species of Pucciniales are known to infest Orchidaceae and ca. 38% of them are described from tropical America. All available types of Pucciniales on Orchidaceae in tropical America were studied and compared with 91 specimens of rust fungi on orchids collected by the author in Panama. Several hundred additional specimens housed in the BPI, almost all intercepted from plant quarantine services, were used for comparison. As result of this work, it is suggested to combine Uromyces stenorrhynchi Henn. to Sphenospora and, as this is the oldest epithet, to synonymize S. kevorkianii Linder, S. mera Cumm. and S. saphena Cumm. with it. Further, it could be demonstrated that Uredo aurantiaca Montemartini, U. cyrtopodii Syd. & P. Syd., U. epidendri Henn., U. guacae Mayor, U. gynandrearum Corda, U. lynchii (Berk.) Plowr., U. neopustulata Cumm. (≡U. pustulata Henn.), U. nigropuncta Henn., U. oncidii Henn., U. ornithidii F. Kern., Cif. & Thurst., and presumably U. scabies Cke., are anamorphs of this variable species. U. gynandrearum is the oldest anamorph-name for all these taxa. Therefore, it can be established that this rust infects more than 80 species of Orchidaceae in three subfamilies. In total, the anamorph of this species was collected by the author on 17 different species of Orchidaceae in Panama which, apart from one species, are all new hosts to science. The molecular data obtained by the author confirm this view, although more data, especially from material from the whole range of distribution of U. gynandrearum, are necessary. Puccinia spiranthicola Cumm. was found to be a synonym of P. cinnamomea Diet. & Holw. and was found by the author on three different Orchidaceae in two subfamilies. Uredo pleurothallidis Keissl. is now considered a synonym of U. wittmackiana Henn. and the latter as the anamorph of Puccinia oncidii Cumm. In the anamorph genus Uredo, a new species was found infecting at least five different species of Sobralia and Elleanthus (Sobraliinae) at different localities. Molecular data indicate it to be related to the currently polyphyletic Phakopsoraceae. For the rusts with suprastomatal sori on Orchidaceae, now separated from Hemileia and placed in the genus Desmosorus (nom. inval.), the current concept with only one taxon is rejected and the establishment of three subspecies is suggested. The complicated taxonomy is discussed and makes it necessary to validate the genus-name and make a new combination. Another Hemileia-anamorph species was found by the author and is considered to be new to science. This is the first species of this alliance in America on Orchidaceae. Molecular data obtained by the author confirm the separation of Desmosorus from Hemileia and the position of the new species. For rusts on Pteridophytes, a new species of Milesia, (teleomorph: Milesina) and a new anamorphic species of Uredinopsis was found, both on hosts hitherto not known. In Calidion, the presumable anamorph-genus of Uncol, the species C. cf. cenicafeae Salazar & Buriticá was found on several new hosts. Further, the teleomorph was found. Morphologically, this teleomorph did not agree with the description of Uncol by the author of the genus, although the anamorph characteristics left no doubt that it is Calidion. Apparently, the description of Uncol is inadequate, but cannot be improved, as the type is unavailable. Molecular data obtained by the author show this species to be closest to Desmosorus. For Uredo superficialis Speg., the anamorph of Desmella, nine new hosts in eight different fern families were found by the author and the collaborators of the ppMP-project. Ecological data indicate that this species includes different host specific races, which, however could not be distinguished morphologically. For all these rusts, a thorough discussion of the ecology in their habitats is given. In total, 21 LSU rDNA sequences from 6 different rust species on Orchidaceae and Pteridophytes were obtained and analyzed with the Maximum Parsimony and Minimum Evolution method. Here, the position of several groups could be confirmed, and some anamorphs could be assigned to different teleomorphic relationships. Within the Ascomycota and their anamorphs, several hitherto unknown species and species not known from these hosts or not known from Panama were found and analyzed. On Orchidaceae, the following fungi belonging to the Ascomycota are described, illustrated and discussed: In the Phyllachorales, a hitherto not known Phyllachora sp. was found on Oncidium warszewiczii Rchb. f. and was compared with the other species of this order currently known from Orchidaceae. In the Asterinaceae s. l. Lembosia cf. epidendri Meir. Silva & O. R. Pereia was found on Maxillaria crassifolia (Lindl.) Rchb. f., which is a new host and new host alliance for this fungus hitherto only known from Brazil. The fungus is described and compared with all species of Asterinaceae currently known on Orchidaceae. In the Meliolaceae, Meliola orchidacearum Cif. was found on Camaridium biolleyi (Schltr.) Schltr. and an Epidendrum sp. which are new hosts and new host alliances of this fungus which was hitherto only known from the Caribbean Islands. It is described, illustrated and compared with the type. In the Glomerellaceae, Glomerella cingulata and its anamorph Colletotrichum gloeosporioides were found on several hosts. The species is illustrated, described and compared with data from literature. In the anamorphic Mycosphaerellaceae, Pseudocercospora odontoglossii (Prill. & Delacr.) U. Braun, a species currently only known from culture, was found on the new host Pleurothallis imraei Lindl. It is illustrated, described and compared with data from literature. On ferns, the following other fungi are described, illustrated and discussed: A conspicuous undescribed form of Polycyclus was found by the author on Elaphoglossum ciliatum (C. Presl.) T. Moore (Dryopteridaceae) and Serpocaulon loriceum (L.) A. R. Sm. (Polypodiaceae). A conspectus of Parmulariaceae infecting ferns is given and demonstrated that Polycyclina should be synonymized under Polycyclus. Summing up, it can be assessed, especially for the Pucciniales, that the most speciose plant family in Panama carries remarkable few species of specific parasites, and that many of them seem to be distributed over a wide range of species which often are not closely related. One reason amongst others seems to be that parasites need a minimum density of host plants in a habitat to survive. As orchid species often occur with only few (and often small) individual plants at a given locality, the probability for a specific pathogen to infect a plant gets too low, hence high diversity by low abundance of hosts might be an impediment for specific pathogens. In this case, unspecific parasites, or such which are infecting larger alliances, are in advantage. Other reasons could be specific traits of orchids, like succulence and mycotrophy which might hamper fungal infections.
Die Beobachtung, dass Tumorzellen häufig eine Abhängigkeit gegenüber einer einzelnen und treibenden Mutation entwickeln, obwohl sie zahlreiche Mutationen aufweisen, bildet die Grundlage der mittlerweile etablierten, zielgerichteten Tumortherapie (Weinstein, 2002). Mit der Identifikation verantwortlicher Signalwege sowie beteiligter Signalkomponenten, sind Ansatzpunkte für diese Therapieform geschaffen worden, die bereits zu einigen Erfolgen in der Leukämie-, Brustkrebs- oder Lungenkrebsbehandlung geführt haben (Druker et al., 2001; Slamon et al., 2001; Kwak et al., 2010) . In vielen Fällen stellt sich jedoch ein Rückfall aufgrund der Ausbildung von Resistenzen ein oder auch das Nichtanschlagen der Therapien wird beobachtet (Ramos & Bentires-Alj, 2015).
Verschiedenste Mechanismen kommen dabei in Frage, doch häufig werden kompensatorische Veränderungen in den Signalwegen beobachtet, die schließlich zur Umgehung der Inhibition führen (Holohan et al., 2013). Grundlage hierfür ist die Redundanz und Verknüpfung der Signalwege mit- und untereinander, die es der Zelle im Sinne der Homöostase ermöglichen sich flexibel an ihre Umgebung anzupassen (Rosell et al., 2013; Sun & Bernards, 2014) . Daher ist es von äußerster Wichtigkeit, die Mechanismen der Inhibition im Hinblick auf die Signalwege der Zellen genauer zu verstehen, und dabei nicht nur die direkten, sondern auch die indirekten Effekte der Inhibition zu analysieren. So lassen sich Rückschlüsse auf den Einsatz zielgerichteter Medikamenten ziehen, die in besseren Therapiekombinationen resultieren und dadurch die Entstehung von Resistenzen verhindern.
Eine Hyper-Aktivierung von STAT3 sowie das dadurch induzierte Genmuster sind als starkes onkogenes Signal identifiziert worden, und spielen darüber hinaus an der Vermittlung von Resistenzen gegenüber Tumortherapien eine entscheidende Rolle. Durch seine Rolle in diversen zellulären Prozessen, beeinflusst STAT3 die Proliferation und das Überleben von Tumorzellen, ihr migratorisches und invasives Verhalten sowie ihre Kommunikation mit Stroma- und Immunzellen. (Bromberg et al., 1999; Wake & Watson, 2015) Sehr selten ist die aberrante Aktivierung des Transkriptionsfaktors auf eigene Mutationen zurückzuführen, vielmehr sorgen Treiber überhalb für diese (Johnston & Grandis, 2011; Kucuk et al., 2015).
In der vorliegenden Arbeit wurden verschiedene STAT3-Inhibitionen in unterschiedlichen Modellen verglichen um darüber Rückschlüsse auf Kriterien einer Therapie zu ziehen. In einem Gliommodell aus der Maus, dem eine v-SRC-Expression als Treiber zu Grunde liegt (Smilowitz et al., 2007), wurde eine indirekte, BMX-vermittelte STAT3-Inhibition mit einer zielgerichteten STAT3-Hemmung verglichen. BMX, die zur TEC-Kinase-Familie gehört, wird als STAT3-aktivierende Kinase beschrieben. In letzter Zeit wurde ihr Einfluss bei der Tumorentwicklung immer deutlicher (Dai et al., 2006; Hart et al., 2011; Holopainen et al., 2012). Unter anderem konnte in Glioblastom-Stammzellen eine BMX-vermittelte STAT3-Aktivierung als Treiber für die Selbsterneurungskapazität und das tumorigene Potential identifiziert werden (Guryanova et al., 2011). Mit dem Tyrosinkinase-Inhibitor Canertinib ist es gelungen, in den murinen Tu-2449-Gliomzellen eine BMX-vermittelte STAT3-Aktivierung nachzuweisen und zu inhibieren. Dies ist damit die erste Arbeit, in der Canertinib als BMX-Inhibitor in einem endogenen Zellsystem getestet wurde. Die einmalige Canertinib-Gabe resultierte in einem Zellzyklusarrest der G1-Phase und die Aufrechterhaltung der Inhibitorwirkung im Zelltod. Im Vergleich dazu konnte eine RNAivermittelte STAT3-Stilllegung nicht das Absterben dieser Zellen induzieren. Mit der Suche weiterer Zielstrukturen von Canertinib, die die Grundlage dieser unterschiedlichen Phänotypen bilden, konnte eine zusätzliche AKT-Inhibition identifiziert werden. Sehr wahrscheinlich wird die AKT-Inhibition ebenfalls durch BMX vermittelt, da keine Inhibition der ERBB-Familie bestätigt werden konnte. Um die Effekte weiter abzugleichen wurden Canertinib-Versuche mit einem humanen Brustkrebsmodell durchgeführt, das als Treiber eine Überexpression des EGFR aufweist.
In MDA-MB-468-Zellen, in denen keine BMX-Aktivierung vorliegt, resultierte eine Canertinib-Behandlung in der sehr prominenten Inhibition des ERK-Signalweges und in einer weniger ausgeprägten Verminderung der STAT3- und AKT-Aktivierung. Auch in diesen Zellen führte die Canertinib-Behandlung zum Zelltod. Diese Effekte werden sehr wahrscheinlich durch die Inhibition des EGFR induziert, da Canertinib als pan-ERBBInhibitor beschrieben ist (Slichenmyer et al., 2001; Djerf Severinsson et al., 2011) .
Resultate die früher in der Arbeitsgruppe gewonnen wurden, beweisen, dass eine Herunterregulation von STAT3 in der Brustkrebszelllinie MDA-MB-468 ausreicht um ein Absterben der Zellen zu induzieren (Groner et al., 2008).
Die Ergebnisse dieser Arbeit zeigen, dass eine Canertinib-Behandlung über die Inhibition unterschiedlicher Signalwege den Zelltod in beiden Zelllinien induziert. Obwohl beide Zelllinien Treiber-vermitteltes, konstitutiv aktives STAT3 aufweisen, stellt nur in den Brustkrebszellen seine Inhibition eine ausreichende Therapiebedingung dar. Somit sind die Unterschiede zwischen den beiden Zelllinien essentiell für ein Überleben der Zellen nach einer STAT3-Inhibition. In Zukunft ist es wichtig, diese Unterschiede zu identifizieren um damit zu definieren, in welchen Patientengruppen eine STAT3-Inhibition zum Erfolg führt.
Signal-dependent regulation of actin dynamics is essential for many cellular processes, including directional cell migration. In particular, cell migration is initiated by lamellipodia, actin-based protrusions of the plasma membrane. The formation of these protruding structures require incessant assembly and disassembly of actin filaments. The Arp2/3 complex and WAVE proteins are essential for both lamellipodium formation and its dynamics. WAVEs mediate the activation of the Arp2/3 complex downstream of the small GTPase Rac, thus being critical for Rac- and RTK-induced actin polymerization and cell migration. The WAVE-family proteins are always found associated with multiprotein complexes. The most abundant WAVE-based complex is referred to as the WANP (WAVE2-Abi-1-Nap1-PIR121) complex. IQGAP1 is a huge scaffolding protein with multiple protein-interacting domains. IQGAP1 participates in many fundamental activities, including regulation of the actin cytoskeleton, mitogenic, adhesive and migratory responses, as well as in cell polarity and cellular trafficking. IQGAP1 binds to N-WASP, thus raising the possibility that it might control actin nucleation by the Arp2/3 complex. In this study, IQGAP1 was found co-immunoprecipitated not only with WAVE, but also with the endogenous WANP-complex subunits. Correspondingly, IQGAP1 associated to both anti-WAVE and anti-Abi-1 immuno-complexes. Pull-down experiments proved that IQGAP1 binds directly to the WANP-complex subunits. Physical interaction between IQGAP1 and the reconstituted WANP complex could also be demonstrated. Together, these data indicate that IQGAP1 is an accessory component of the WANP complex. Interestingly, the IQGAP-WANP complex disassembled after either EGF stimulation or transfection with constitutively active Cdc42 and Rac1. HeLa cells devoid of IQGAP1 showed diminished and less persistent ruffling upon EGF, but not HGF, stimulation in comparison with the control. This phenotype was accompanied by a strong reduction in chemotaxis towards both growth factors, which was as dramatic as in WANP-complex knockdown (KD) cells. Moreover, GM130 and Giantin showed a polarized and flat ribbon-like pattern in control cells, as it is expected for cis- and cis/medial-Golgi markers. Conversely, small and dispersed vesicular structures were found in both IQGAP1 KD and WANP-complex KD cells. Importantly, Arp2/3-complex silencing resulted in the same phenotypes. Consistently, Brefeldin A-induced disassembly of the Golgi strongly inhibited the IQGAP1-WANP-complex interaction and chemotaxis towards EGF in wild-type cells. The re-expression of an RNAi-resistant wild-type IQGAP1 in IQGAP1 KD cells fully rescued both the ruffling abilities and Golgi structure. A constitutively active mutant, unable to bind to neither Rac1 /Cdc42 nor the WANP complex, could reconstitute only the former defect. Hence, this study shows that actin dynamics regulated by the IQGAP1-WANP complex controls Golgi-apparatus architecture and its contribution to cell chemotaxis. The working model here proposes that at the Golgi apparatus, recruitment of the WANP complex by IQGAP1 leads to the assembly of actin filaments required to maintain the appropriated Golgi morphology. The dissociation of the complex may be required to allow the remodeling of the Golgi membranes in order to respond following a chemoattractant gradient.
Paleoecology is the study of organismal interactions with the environment in the geological past. Organisms are influenced in their distribution and abundance by abiotic factors such as temperature and precipitation. A change in these factors, for example by major climatic shifts, would then affect the communities of organisms. Studying this hypothesized causal link between climatic and faunal change is especially interesting for the Plio-Pleistocene of East Africa due to the fact that our own ancestors also inhabited these regions. Both the Turkana basin in Kenya and the Lake Albert region in Uganda offer unique opportunities to investigate these paleoecological issues. Their late Miocene through Pleistocene deposits provide a very good record of climatic, vegetation and faunal change in East Africa (Pickford et al. 1993, Leakey et al. 1995, 1998, McDougall & Feibel 2003, Wynn 2004). This study focuses on the mammal family Bovidae as they are good indicator of vegetation and environment (e.g. Vrba 1980, 1995, Shipman & Harris 1988, Bobe & Eck 2001, Bobe & Behrensmeyer 2004, Bobe et al. 2007). Bovidae are quite species-rich and inhabit a wide range of habitats from tropical rain forests to deserts which predicates their array of morphological adaptations (ecovariables) to these environments. Diet is the ecovariable that is most to climate and thus habitat change. Therefore, the fossil Bovidae are especially suitable for reconstructing past environments. The objective of this thesis is to test the hypothesis that, from the late Miocene through the Holocene, Africa has experienced an overall increase in aridity and concomitant pulses of habitat change. The hypothesis predicts that increasing aridity causes a likewise growth in the abundance of taxa adapted to open arid environments. In particular, an increase in bovid grazers should be observed in combination with a decrease of bovid browsers. To test this hypothesis, I examine the fossil bovid communities from each stratigraphic member of Lake Turkana (Lothagam, Kanapoi, West Turkana and Koobi Fora) and Lake Albert (Nkondo-Kaiso region) and through a taxonomic and a functional perspective reconstruct the paleoenvironments and -climates from approximately 8 to 0.6 Ma. This study is the first to use taxonomic and ecomorphological data together to reconstruct the paleoenvironments of the Turkana basin and the Nkondo-Kaiso region of Lake Albert. In a first analysis, mesowear, as introduced by Fortelius & Solounias (2000), is used to gather information about the diet of bovids. As a result of my preliminary investigations on upper vs. lower molars of recent species, the sample of fossil bovid specimens from the Turkana basin and Lake Albert were found to be unsuitable to reveal a meaningful diet reconstruction. Therefore, the bovids are assigned to diet categories based on literature. For each member of the time period from 8.0 to 0.6 Ma, I provide a detailed characterization of the bovid fauna in terms of α- and β- diversity both on tribe and diet level based on presence-absence as well as for the Turkana basin on abundance data. Statistical comparisons between the fossil bovid communities and those in modern protected areas with known vegetation and climatic conditions have yielded modern analogues for each stratigraphic member. Following that I provide paleoclimatic conditions such as assumed mean annual temperature for each member. Based on abundance of diet categories in the bovid communities, the paleoclimate of the Turkana basin was in general cooler and considerably more humid during the late Miocene to the Pleistocene than today. The mean annual temperature at Lothagam is assumed as 22.2 °C, the annual precipitation as 685 mm for 8.0 – 6.54 Ma and 4.9 – 3.4 Ma. The intervening time period is characterized by a slightly lower mean annual temperature and precipitation (20.3 °C, 583 mm). From 4.17 to 4.07 Ma Kanapoi faced 21.3 °C and 592 mm rainfall. In the eastern part of the basin the climate was warmer and more humid (3.4 – 2.68 Ma: 26.2, 961 mm; 2.68 – 1.3 Ma: 27.1 °C, 935 mm) from 3.4 to 1.3 Ma than in the preceeding eras. In the western part, the climate became warmer and more humid ~500,000 years later and was more variable than that in the eastern basin. From 2.94 to 2.52 Ma the mean annual temperature was 26.2 °C and the annual precipitation 961 mm. Between 2.34 and 1.6 Ma the climate again cooled and became drier as before 2.94 Ma. A second shift to higher temperature and precipitation occurred after 1.6 Ma (27.1 °C, 935 mm) lasted until 1.34 Ma. The results of the bovid community analyses do not support the hypothesis of increasing aridity in Eastern Africa during the late Mio- to Pleistocene. Instead, the results show that the bovid communities differed much over time and on a relatively small spatial scale. Regional paleovegetation and paleoclimate exhibit fluctuations through the studied time period at western Turkana and differences between the western and eastern part of the Turkana basin. This is indicative of a patchy habitat distribution both on temporal and spatial levels. Increased climate variability predicts an increase in landscape complexity as proposed by the ‘variability selection hypothesis’ (Potts 1998a+b). Therefore, this thesis research supports the hypothesis of increased landscape complexity on the spatial level. This study has important implications for future research. First, an analysis based on ecovariable characteristics such as diet may be preferred to a taxonomic analysis. Second, abundance data should be used for an ecovariable analysis because the results then provide more precise information on the paleovegetation and –climate than just the presence of these adaptations in the faunal community. Lastly, as this study is based on one mammal family, further studies on other mammal groups should be conducted to increase the database of exploited resource by the entire faunal community. Most significantly this study provides a basis for new interpretations of faunal community distributions. It also raises the question whether small scale spatial community variability is also to be expected at other fossil sites. If so then this methodology has important implications for reconstructions of paleovegetation and paleoclimate.
In dieser Arbeit wurden die physiologische Funktion innerhalb der Ribosomenbiogenese und die physikalischen Interaktionen des nukleolären, essentiellen Proteins Nep1p in der Hefe Saccharomyces cerevisiae untersucht. Durch Hefe-Zwei-Hybrid-Experimente und biochemische Analysen konnte eine Homodimerisierung des Proteins festgestellt sowie eine strukturabgeleitete Dimerisierungsmutante identifiziert werden. Ebenfalls aus der Struktur des Nep1p-Homologs aus Methanocaldococcus jannaschii konnte eine Nop14p-Bindungsregion auf der der Dimerkontaktfläche abgewandten Seite des Hefeproteins vorhergesagt und nach in vitro-Mutagenese bestätigt werden. Innerhalb des Nop14-Proteins wurden zwei Domänen charakterisiert, die im Zwei-Hybrid-System mit Nep1p interagieren. Aus Strukturdaten in Kombination mit Hefe-Drei-Hybrid-Experimenten konnte die RNA-Bindungsregion an der Dimerkontaktfläche des Nep1-Proteins lokalisiert werden. In Drei-Hybrid-Selektionen wurden RNA-Sequenzen mit hoher Affinität zu dem M. jannaschii Nep1p identifiziert, die auf eine Bindung des Proteins bei Helix 35 der 16S rRNA schließen lassen. Aufgrund der hohen Konservierung dieser rRNA-Region ist eine Bindung des Hefeproteins an die 18S rRNA-Schleife von Nukleotid 1189-1196 sehr wahrscheinlich. Da Nep1p eine große Ähnlichkeit zu Proteinen der SPOUTFamilie von Methyltransferasen aufweist, war von einer rRNA-Methylierung im Verlauf der Ribosomenbiogenese als katalytische Funktion des Proteins auszugehen. Aus verschiedenen Drei-Hybrid-Experimenten zur RNA-Bindungungsspezifität ergab sich als mögliche Reaktion die N1-Methylierung des Nukleotids 1-Methyl-3-(3-Amino-3-Carboxypropyl)-Pseudouridin (m1acp3Y) 1191 der 18S rRNA. Durch eine spezifische radioaktive Markierung der acp-Gruppe konnte gezeigt werden, dass Nep1p keinen Einfluss auf die spätere Aminocarboxypropylmodifizierung hat. Diese findet auch bei einer Deletion der snoRNA35 statt, also auch an einem Uridin, und ist unabhängig von dem cytoplasmatischen Protein Tma20p. In RP-HPLC-Experimenten konnte nachgewiesen werden, dass die 18S rRNA einer Dnep1Dnop6-Doppelmutante ein Aminocarboxypropyl-modifiziertes Nukleosid enthält, dass sich in seinem Retensionsverhalten von dem m1acp3Y eines Wildtyps unterscheidet. Bei dem in diesem Stamm detektierten acp-modifizierten Nukleosid handelt es sich vermutlich um ein nicht-methyliertes acpY, was eine Funktion von Nep1p als N1-Methyltransferase des Nukleotids Y1191 der 18S rRNA höchst wahrscheinlich macht. Diese katalytische Funktion konnte in Zusammenarbeit mit Prof. Wöhnert auch für das M. jannaschii Nep1p gezeigt werden. Dass sowohl eine snr35- Deletion als auch eine 18S rRNA-Mutation des Nukleotids 1191 nicht letal sind, machte deutlich, dass die N1-Methylierung nicht die essentielle Funktion von Nep1p darstellen kann. Weiterhin konnte nachgewiesen werden, dass die Suppression der nep1-1ts-Mutante durch S-Adenosylmethionin nicht auf der Unterstützung der Methyltransferase-Aktivität des Proteins, sondern vermutlich eher auf einer generellen Stabilisierung des temperatursensitiven Proteins beruht. Sowohl im Hefe-Nep1p als auch im humanen Homolog wurden durch biochemische und genetische Experimente mehrere Phänotypen der Bowen-Conradi-Mutation (Aspartat 90 zu Glycin in ScNep1p) nachgewiesen. Diese lassen auf eine Aggregation des mutierten Proteins sowie eine dadurch bedingte Fehllokalisation innerhalb der Zelle schließen. Zusätzlich ist aber auch ein RNA-Bindungsdefekt durch den Aminosäureaustausch wahrscheinlich. Nichtsdestotrotz liegt offensichtlich ausreichend Nep1p-Protein vor, dass seine essentielle Funktion erfüllen kann, da die Mutation selbst zu keinem Wachstumsphänotyp führt. Erst bei einer partiellen Translationsrepression des mutierten Proteins unter Verwendung des artifiziellen Tetrazyklin-Aptamer-Systems ist ein verlangsamtes Wachstum von Hefezellen zu beobachten, was dieses System geeignet zur Analyse von möglichen Therapeutika macht.
Studies in particular of the last decade showed that active neurogenesis continuously takes place in the subventricular zone (SVZ) of the lateral ventricles of the adult rodent brain. Neurogenesis in the SVZ leads to migration of neuroblasts within the rostral migratory stream (RMS) and mature neuron formation mainly in the olfactory bulb (OB). According to present understanding, glial cells with astrocytic properties represent the actual adult neural stem cells. The cell types representing the various cellular transition states leading to the formation of mature neurons as well as the mechanisms controlling adult neurogenesis and neuroblast migration are poorly understood. A previous study from this laboratory demonstrated that the ATP-hydrolyzing enzyme nucleoside triphosphate diphosphohydrolase 2 (NTPDase2) is associated with type B cells, the presumptive neural stem cells. NTPDase2 is a protein of the plasma membrane with its catalytic site facing the extracellular space. It hydrolyzes extracellular nucleoside triphosphates to their respective nucleoside diphosphates. This raises the possibility that the signaling pathway via extracellular nucleotides is involved in the control of adult neurogenesis. Neurons as well as glial cells express several subtypes of receptors (P2 receptors) that are responsive to the nucleotides ATP, ADP, UTP, or UDP. P2X receptors are ATP-gated Na+, K+ and Ca2+ permeable ion channels, P2Y receptors are coupled to trimeric G-proteins. In order to probe for a functional role of nucleotides in adult neurogenesis, the present study referred to an in vitro system (neurospheres). Neurospheres produced from isolates of the mouse SVZ and cultured in the presence of EGF and bFGF expressed the neural stem cell marker nestin and also GFAP, S100β, NTPDase2 and tissue non-specific alkaline phosphatase. Neurospheres generated from the cells of the subventricular zone were multipotenital. This was revealed by immunostaining of differentiated cells with markers for astrocytes, neurons and oligodendrocytes. The presence of ecto-nucleotidase was verified by analyzing the free phosphate released from nucleotides. The tissue non-specific form of alkaline phosphatase was the predominant enzyme. Both NTPDase2 and TNAP could be identified by immunocytochemistry and Western blotting. Hydrolysis was not observed for p-nitrophenyl thymidine monophosphate, a substrate of members of the ectonucleotide pyrophosphatase/phosphodiesterase family (NPP1 to NPP3). Since ecto-nucleotidases control the availability of extracellular nucleotide agonists, neurospheres were studied for the potential expression and functional role of nucleotide receptors. Neurospheres responded to extracellular nucleotides with a transient rise in Ca2+ (ATP = ADP > UTP). The rise in Ca2+ was due to P2Y receptors. The Ca2+ response was unaltered in the absence of extracellular Ca2+ and strongly reduced by thapsigargin, a blocker of internal Ca2+ stores. The P2Y1 antagonist MRS2179 strongly reduced the ATP- or ADP-induced increase in Ca2+, suggesting the involvement of a P2Y1 receptor. In addition, suramin and PPADS, non-selective antagonists for P2 receptors, inhibited most of the Ca2+ response. The agonistic activity of UTP and the lack of response to UDP implied the additional presence of a P2Y2 and/or a P2Y4 receptors and the absence of a functional P2Y6 receptor. RT-PCR experiments demonstrated that neurospheres expressed P2Y1 and P2Y2 receptors but not P2Y4 receptor. That the majority of the Ca2+ response to ATP was mediated via P2Y1 receptors was also confirmed by analysis of P2Y1 knockout mice and by application of the P2Y1 receptor-specific antagonist MRS2179. In addition, agonists of P2Y1 and P2Y2 receptors and low concentrations of adenosine augmented cell proliferation inspite of the presence of mitogenic growth factors. Neurosphere cell proliferation was attenuated after application of MRS2179 and in neurospheres from P2Y1 receptor knockout mice. These results infer a nucleotide receptor-mediated synergism that augments growth factor-mediated cell proliferation. Taken together these results suggest that P2Y-mediated nucleotidergic signalling is involved in neurosphere function and possibly also in adult neurogenesis in situ.
