Die steigende Zahl von Pilzinfektionen, die Entwicklung und Einführung neuer anti-mykotischer Substanzen sowie die Möglichkeit der Resistenzentwicklung unter Therapie mit Antimykotikahaben in der Vergangenheit zu einem ständig wachsenden Bedarf an standardisierten Verfahren zur Empfindlichkeitstestung von pathogenen Pilzen geführt. Hierbei entstand unter anderem eine Vielzahl kommerzieller Testverfahren, bei denen mit Hilfe vorgefertigter Testkits eine einfache und schnelle Durchführung der Empfindlichkeitsprüfung erzielt werden soll. Eine dieser Methoden, welche in manchen Laboratorien in Deutschland angewendet wird, ist das so genannte LD 2Ring-Verfahren, welches auf dem Prinzip der Agardiffusion beruht unter Verwendung vorgefertigter, antimy-kotika-beschichteter Papierringe. In der vorliegenden Arbeit wird dieses Verfahren auf seine Reproduzierbarkeit bei der Testung von zehn Qualitätskontrollstämmen hin überprüft. Die Ergebnisse zeigen eine starke Schwankungsbreite und somit eine schlechte Reproduzierbarkeit, so dass dieses Verfahren zwar für die Bearbeitung von wissenschaftlichen Fragestellungen, nicht jedoch für die Routinetestung als geeignet angesehen werden kann. Des Weiteren erfolgte eine Untersuchung auf das Vor-liegen eines so genannten "minor error", man erhält für einen sensiblen Stamm das Ergebnis "resistent", "major error", man erhält für einen intermediären Stamm das Ergebnis "sensibel", und "very major error", man erhaält für einen resistenten Stamm das Ergebnis "sensibel". Hierbei kam es in 16,25% der untersuchten Fälle zum Vorliegen eines "minor errors". Ein "major error" wurde nicht beobachtet
An important goal is to identify the direct activation domain (AD)-interacting components of the transcriptional machinery within the context of native complexes. Toward this end, we first demonstrate that the multisubunit TFIID, SAGA, mediator, and Swi/Snf coactivator complexes from transcriptionally competent whole-cell yeast extracts were all capable of specifically interacting with the prototypic acidic ADs of Gal4 and VP16. We then used hexahistidine tags as genetically introduced activation domain-localized cross-linking receptors. In combination with immunological reagents against all subunits of TFIID and SAGA, we systematically identified the direct AD-interacting subunits within the AD-TFIID and AD-SAGA coactivator complexes enriched from whole-cell extracts and confirmed these results using purified TFIID and partially purified SAGA. Both ADs directly cross-linked to TBP and to a subset of TFIID and SAGA subunits that carry histone-fold motifs.
Biglycan, a nitric oxide-regulated gene, affects adhesion, growth, and survival of mesangial cells

During glomerular inflammation mesangial cells are the major source and target of nitric oxide that pro-foundly influences proliferation, adhesion, and death of mesangial cells. The effect of nitric oxide on the mRNA expression pattern of cultured rat mesangial cells was therefore investigated by RNA-arbitrarily-primed polymerase chain reaction. Employing this approach, biglycan expression turned out to be down-regulated time- and dose-dependently either by interleukin-1beta-stimulated endogenous nitric oxide production or by direct application of the exogenous nitric oxide donor, diethylenetriamine nitric oxide. There was a corresponding decline in the rate of biglycan biosynthesis and in the steady state level of this proteoglycan. In vivo, in a model of mesangioproliferative glomerulonephritis up-regulation of inducible nitric-oxide synthase mRNA was associated with reduced expression of biglycan in isolated glomeruli. Biglycan expression could be normalized, both in vitro and in vivo, by using a specific inhibitor of the inducible nitric-oxide synthase, l-N6-(l-iminoethyl)-l-lysine dihydrochloride. Further studies showed that biglycan inhibited cell adhesion on type I collagen and fibronectin because of its binding to these substrates. More importantly, biglycan protected mesangial cells from apoptosis by decreasing caspase-3 activity, and it counteracted the proliferative effects of platelet-derived growth factor-BB. These findings indicate a signaling role of biglycan and describe a novel pathomechanism by which nitric oxide modulates the course of renal glomerular disease through regulation of biglycan expression.
Membrane-bound complex I (NADH:ubiquinone oxidoreductase) of the respiratory chain is considered the main site of mitochondrial radical formation and plays a major role in many mitochondrial pathologies. Structural information is scarce for complex I, and its molecular mechanism is not known. Recently, the 49-kDa subunit has been identified as part of the "catalytic core" conferring ubiquinone reduction by complex I. We found that the position of the 49-kDa subunit is clearly separated from the membrane part of complex I, suggesting an indirect mechanism of proton translocation. This contradicts all hypothetical mechanisms discussed in the field that link proton translocation directly to redox events and suggests an indirect mechanism of proton pumping by redox-driven conformational energy transfer.
Cardiolipin stabilized supercomplexes of Saccharomyces cerevisiae respiratory chain complexes III and IV (ubiquinol:cytochrome c oxidoreductase and cytochrome c oxidase, respectively), but was not essential for their formation in the inner mitochondrial membrane because they were found also in a cardiolipin-deficient strain. Reconstitution with cardiolipin largely restored wild-type stability. The putative interface of complexes III and IV comprises transmembrane helices of cytochromes b and c1 and tightly bound cardiolipin. Subunits Rip1p, Qcr6p, Qcr9p, Qcr10p, Cox8p, Cox12p, and Cox13p and cytochrome c were not essential for the assembly of supercomplexes; and in the absence of Qcr6p, the formation of supercomplexes was even promoted. An additional marked effect of cardiolipin concerns cytochrome c oxidase. We show that a cardiolipin-deficient strain harbored almost inactive resting cytochrome c oxidase in the membrane. Transition to the fully active pulsed state occurred on a minute time scale.
The renin-angiotensin-aldosterone system plays a pivotal role in the regulation of salt and water homeostasis. Here, we demonstrate the expression and functional role of cGMP-dependent protein kinases (PKGs) in rat adrenal cortex. Expression of PKG II is restricted to adrenal zona glomerulosa (ZG) cells, whereas PKG I is localized to the adrenal capsule and blood vessels. Activation of the aldosterone system by a low sodium diet up-regulated the expression of PKG II, however, it did not change PKG I expression in adrenal cortex. Both, activation of PKG II in isolated ZG cell and adenoviral gene transfer of wild type PKG II into ZG cells enhanced aldosterone production. In contrast, inhibition of PKG II as well as infection with a PKG II catalytically inactive mutant had an inhibitory effect on aldosterone production. Steroidogenic acute regulatory (StAR) protein that regulates the rate-limiting step in steroidogenesis is a new substrate for PKG II and can be phosphorylated by PKG II in vitro at serine 55/56 and serine 99. Stimulation of aldosterone production by PKG II in contrast to stimulation by PKA did not activate StAR gene expression in ZG cells. The results presented indicate that PKG II activity in ZG cells is important for maintaining basal aldosterone production.
Atrial natriuretic peptide (ANP) plays a key regulatory role in arterial blood pressure homeostasis. We recently generated mice with selective deletion of the ANP receptor, guanylyl cyclase-A (GC-A), in vascular smooth muscle (SMC GC-A knockout (KO) mice) and reported that resting arterial blood pressure was completely normal in spite of clear abolition of the direct vasodilating effects of ANP (Holtwick, R., Gotthardt, M., Skryabin, B., Steinmetz, M., Potthast, R., Zetsche, B., Hammer, R. E., Herz, J., and Kuhn M. (2002) Proc. Natl. Acad. Sci. U. S. A. 99, 7142–7147). The purpose of this study was to clarify mechanisms compensating for the missing vasodilator responses to ANP. In particular, we analyzed the effect of the endothelial, cGMP-mediated vasodilators C-type natriuretic peptide and nitric oxide (NO). In isolated arteries from SMC GC-A KO mice, the vasorelaxing sensitivity to sodium nitroprusside and the endothelium-dependent vasodilator, acetylcholine, was significantly greater than in control mice. There was no difference in responses to C-type natriuretic peptide or to the activator of cGMP-dependent protein kinase I, 8-para-chlorophenylthio-cGMP. The aortic expression of soluble GC (sGC), but not of endothelial NO synthase or cGMP-dependent protein kinase I, was significantly increased in SMC GC-A KO mice. Chronic oral treatment with the NO synthase inhibitor Nw-nitro-l-arginine methyl ester increased arterial blood pressure, the effect being significantly enhanced in SMC GC-A KO mice. We conclude that SMC GC-A KO mice exhibit a higher vasodilating sensitivity to NO. This can be attributed to an enhanced expression of sGC, whereas the expression and/or activity levels of downstream cGMP-effector pathways are not involved. Increased vasodilating responsiveness to endothelial NO contributes to compensate for the missing vasodilating effect of ANP in SMC GC-A KO mice.
Atovaquone is a substituted 2-hydroxynaphthoquinone that is used therapeutically to treat Plasmodium falciparum malaria, Pneumocystis carinii pneumonia, and Toxoplasma gondii toxoplasmosis. It is thought to act on these organisms by inhibiting the cytochrome bc1 complex. We have examined the interaction of atovaquone with the bc1 complex isolated from Saccharomyces cerevisiae, a surrogate, nonpathogenic fungus. Atovaquone inhibits the bc1 complex competitively with apparent Ki = 9 nm, raises the midpoint potential of the Rieske iron-sulfur protein from 285 to 385 mV, and shifts the g values in the EPR spectrum of the Rieske center. These results indicate that atovaquone binds to the ubiquinol oxidation pocket of the bc1 complex, where it interacts with the Rieske iron-sulfur protein. A computed energy-minimized structure for atovaquone liganded to the yeast bc1 complex suggests that a phenylalanine at position 275 of cytochrome b in the bovine bc1 complex, as opposed to leucine at the equivalent position in the yeast enzyme, is responsible for the decreased sensitivity of the bovine bc1 complex (Ki = 80 nm) to atovaquone. When a L275F mutation was introduced into the yeast cytochrome b, the sensitivity of the yeast enzyme to atovaquone decreased (Ki = 100 nm) with no loss in activity, confirming that the L275F exchange contributes to the differential sensitivity of these two species to atovaquone. These results provide the first molecular description of how atovaquone binds to the bc1 complex and explain the differential inhibition of the fungal versus mammalian enzymes.
Pin1 is a peptidyl-prolyl cis/trans isomerase (PPIase) essential for cell cycle regulation. Pin1-catalyzed peptidyl-prolyl isomerization provides a key conformational switch to activate phosphorylation sites with the common phospho-Ser/Thr-Pro sequence motif. This motif is ubiquitously exploited in cellular response to a variety of signals. Pin1 is able to bind phospho-Ser/Thr-Pro-containing sequences at two different sites that compete for the same substrate. One binding site is located within the N-terminal WW domain, which is essential for protein targeting and localization. The other binding site is located in the C-terminal catalytic domain, which is structural homologous to the FK506-binding protein (FKBP) class of PPIases. A flexible linker of 12 residues connects the WW and catalytic domain. To characterize the structure and dynamics of full-length Pin1 in solution, high resolution NMR methods have been used to map the nature of interactions between the two domains of Pin1. In addition, the influence of target peptides on domain interactions has been investigated. The studies reveal a dynamic picture of the domain interactions. 15N spin relaxation data, differential chemical shift mapping, and residual dipolar coupling data indicate that Pin1 can either behave as two independent domains connected by the flexible linker or as a single intact domain with some amount of hinge bending motion depending on the sequence of the bound peptide. The functional importance of the modulation of relative domain flexibility in light of the multitude of interaction partners of Pin1 is discussed.
Recently, we reported that in crude enzyme preparations, a monocyte-derived soluble protein (M-DSP) renders 5-lipoxygenase (5-LO) activity Ca2+-dependent. Here we provide evidence that this M-DSP is glutathione peroxidase (GPx)-1. Thus, the inhibitory effect of the M-DSP on 5-LO could be overcome by the GPx-1 inhibitor mercaptosuccinate and by the broad spectrum GPx inhibitor iodoacetate, as well as by addition of 13(S)-hydroperoxy-9Z,11E-octadecadienoic acid (13(S)-HPODE). Also, the chromatographic characteristics and the estimated molecular mass (80-100 kDa) of the M-DSP fit to GPx-1 (87 kDa), and GPx-1, isolated from bovine erythrocytes, mimicked the effects of the M-DSP. Intriguingly, only a trace amount of thiol (10 micro M GSH) was required for reduction of 5-LO activity by GPx-1 or the M-DSP. Moreover, the requirement of Ca2+ allowing 5-LO product synthesis in various leukocytes correlated with the respective GPx-1 activities. Mutation of the Ca2+ binding sites within the C2-like domain of 5-LO resulted in strong reduction of 5-LO activity by M-DSP and GPx-1, also in the presence of Ca2+. In summary, our data suggest that interaction of Ca2+ at the C2-like domain of 5-LO protects the enzyme against the effect of GPx-1. Apparently, in the presence of Ca2+, a low lipid hydroperoxide level is sufficient for 5-LO activation.
Nisin-producing Lactococcus lactis strains show a high degree of resistance to the action of nisin, which is based upon expression of the self-protection (immunity) genes nisI, nisF, nisE, and nisG. Different combinations of nisin immunity genes were integrated into the chromosome of a nisin-sensitive Bacillus subtilis host strain under the control of an inducible promoter. For the recipient strain, the highest level of acquired nisin tolerance was achieved after coordinated expression of all four nisin immunity genes. But either the lipoprotein NisI or the ABC transporter-homologous system NisFEG, respectively, were also able to protect the Bacillus host cells. The acquired immunity was specific to nisin and provided no tolerance to subtilin, a closely related lantibiotic. Quantitative in vivo peptide release assays demonstrated that NisFEG diminished the quantity of cell-associated nisin, providing evidence that one role of NisFEG is to transport nisin from the membrane into the extracellular space. NisI solubilized from B. subtilis membrane vesicles and recombinant hexahistidine-tagged NisI from Escherichia coli interacted specifically with nisin and not with subtilin. This suggests a function of NisI as a nisin-intercepting protein.
Na,K-ATPase mediates net electrogenic transport by extruding three Na+ ions and importing two K+ ions across the plasma membrane during each reaction cycle. We mutated putative cation coordinating amino acids in transmembrane hairpin M5-M6 of rat Na,K-ATPase: Asp776 (Gln, Asp, Ala), Glu779 (Asp, Gln, Ala), Asp804 (Glu, Asn, Ala), and Asp808 (Glu, Asn, Ala). Electrogenic cation transport properties of these 12 mutants were analyzed in two-electrode voltage-clamp experiments on Xenopus laevis oocytes by measuring the voltage dependence of K+-stimulated stationary currents and pre-steady-state currents under electrogenic Na+/Na+ exchange conditions. Whereas mutants D804N, D804A, and D808A hardly showed any Na+/K+ pump currents, the other constructs could be classified according to the [K+] and voltage dependence of their stationary currents; mutants N776A and E779Q behaved similarly to the wild-type enzyme. Mutants E779D, E779A, D808E, and D808N had in common a decreased apparent affinity for extracellular K+. Mutants N776Q, N776D, and D804E showed large deviations from the wild-type behavior; the currents generated by mutant N776D showed weaker voltage dependence, and the current-voltage curves of mutants N776Q and D804E exhibited a negative slope. The apparent rate constants determined from transient Na+/Na+ exchange currents are rather voltage-independent and at potentials above -60 mV faster than the wild type. Thus, the characteristic voltage-dependent increase of the rate constants at hyperpolarizing potentials is almost absent in these mutants. Accordingly, dislocating the carboxamide or carboxyl group of Asn776 and Asp804, respectively, decreases the extracellular Na+ affinity.
We report the first evidence for the formation of the "607- and 580-nm forms" in the cytochrome oxidase aa3/H2O2 reaction without the involvement of tyrosine 280. The pKa of the 607-580-nm transition is 7.5. The 607-nm form is also formed in the mixed valence cytochrome oxidase/O2 reaction in the absence of tyrosine 280. Steady-state resonance Raman characterization of the reaction products of both the wild-type and Y280H cytochrome aa3 from Paracoccus denitrificans indicate the formation of six-coordinate low spin species, and do not support, in contrast to previous reports, the formation of a porphyrin pi-cation radical. We observe three oxygen isotope-sensitive Raman bands in the oxidized wild-type aa3/H2O2 reaction at 804, 790, and 358 cm-1. The former two are assigned to the Fe(IV)[double bond]O stretching mode of the 607- and 580-nm forms, respectively. The 14 cm-1 frequency difference between the oxoferryl species is attributed to variations in the basicity of the proximal to heme a3 His-411, induced by the oxoferryl conformations of the heme a3-CuB pocket during the 607-580-nm transition. We suggest that the 804-790 cm-1 oxoferryl transition triggers distal conformational changes that are subsequently communicated to the proximal His-411 heme a3 site. The 358 cm-1 mode has been found for the first time to accumulate with the 804 cm-1 mode in the peroxide reaction. These results indicate that the mechanism of oxygen reduction must be reexamined.
Cholecystokinin (CCK) and related peptides are potent growth factors in the gastrointestinal tract and may be important for human cancer. CCK exerts its growth modulatory effects through G(q)-coupled receptors (CCK(A) and CCK(B)) and activation of extracellular signal-regulated protein kinase 1/2 (ERK1/2). In the present study, we investigated the different mechanisms participating in CCK-induced activation of ERK1/2 in pancreatic AR42J cells expressing both CCK(A) and CCK(B). CCK activated ERK1/2 and Raf-1 to a similar extent as epidermal growth factor (EGF). Inhibition of EGF receptor (EGFR) tyrosine kinase or expression of dominant-negative Ras reduced CCK-induced ERK1/2 activation, indicating participation of the EGFR and Ras in CCK-induced ERK1/2 activation. However, compared with EGF, CCK caused only small increases in tyrosine phosphorylation of the EGFR and Shc, Shc-Grb2 complex formation, and Ras activation. Signal amplification between Ras and Raf in a CCK-induced ERK cascade appears to be mediated by activation of protein kinase Cepsilon (PKCepsilon), because 1) down-modulation of phorbol ester-sensitive PKCs inhibited CCK-induced activation of Ras, Raf, and ERK1/2 without influencing Shc-Grb2 complex formation; 2) PKCepsilon, but not PKCalpha or PKCdelta, was detectable in Raf-1 immunoprecipitates, although CCK activated all three PKC isoenzymes. In addition, the present study provides evidence that the Src family tyrosine kinase Yes is activated by CCK and mediates CCK-induced tyrosine phosphorylation of Shc. Furthermore, we show that CCK-induced activation of the EGFR and Yes is achieved through the CCK(B) receptor. Together, our data show that different signals emanating from the CCK receptors mediate ERK1/2 activation; activation of Yes and the EGFR mediate Shc-Grb2 recruitment, and activation of PKC, most likely PKCepsilon, augments CCK-stimulated ERK1/2 activation at the Ras/Raf level.
Physiological conditions which lead to changes in total carotenoid content in tomato plantlets were identified. Carotenoid levels were found to increase after the onset of a dark period during a normal 24h cycle. This rapid initial increase is followed by a steady decrease in carotenoid content throughout the night. A decrease in the expression of several carotenogenic genes, namely pds, zds (carotenoid desaturases) and ptox (plastid terminal oxidase), was observed following the removal of the light (when carotenoid content is at its highest). An increase in gene expression was observed before the return to light for pds and zds (when carotenoid levels were at their lowest), or following the return to light for ptox. The phytoene desaturation inhibitor norflurazon leads to a decrease coloured carotenoid content and, in the light, this correlated with pds and zds gene induction. In the dark, norflurazon treatment led to only a weak decrease in carotenoid content and only a small increase in pds and zds gene expression. The striking absence of phytoene accumulation under norflurazon treatment in the dark suggests a down-regulation of carotenoid formation in darkness. However, prolonged dark conditions, or treatment with photosynthetic inhibitors, surprisingly led to higher carotenoid levels, which correlated with decreased expression of most examined genes. In addition to light, which acts in a complex way on carotenoid accumulation and gene expression, our results are best explained by a regulatory effect of carotenoid levels on the expression of several biosynthetic genes. In addition, monitoring of protein amounts for phytoene desaturase and plastid terminal oxidase (which sometimes do not correlate with gene expression) indicate an even more complex regulatory pattern.
Receptor tyrosine kinases of the epidermal growth factor (EGF) receptor family regulate essential cellular functions such as proliferation, survival, migration, and differentiation but also play central roles in the etiology and progression of tumors. We have identified short peptide sequences from a random peptide library integrated into the thioredoxin scaffold protein, which specifically bind to the intracellular domain of the EGF receptor (EGFR). These molecules have the potential to selectively inhibit specific aspects of EGF receptor signaling and might become valuable as anticancer agents. Intracellular expression of the aptamer encoding gene construct KDI1 or introduction of bacterially expressed KDI1 via a protein transduction domain into EGFR-expressing cells results in KDI1·EGF receptor complex formation, a slower proliferation, and reduced soft agar colony formation. Aptamer KDI1 did not summarily block the EGF receptor tyrosine kinase activity but selectively interfered with the EGF-induced phosphorylation of the tyrosine residues 845, 1068, and 1148 as well as the phosphorylation of tyrosine 317 of p46 Shc. EGF-induced phosphorylation of Stat3 at tyrosine 705 and Stat3-dependent transactivation were also impaired. Transduction of a short synthetic peptide aptamer sequence not embedded into the scaffold protein resulted in the same impairment of EGF-induced Stat3 activation.
There is evidence that endothelial nitric-oxide synthase (eNOS) is regulated by reciprocal dephosphorylation of Thr497 and phosphorylation of Ser1179. To examine the interrelationship between these sites, cells were transfected with wild-type (WT), T497A, T497D, S1179D, and T497A/S1179D eNOS and activity, NO release and eNOS localization were assessed. Although eNOS T497A, S1179D and T497A/S1179D eNOS had greater enzymatic activity than did WT eNOS in lysates, basal production of NO from cells was markedly reduced in cells transfected with T497A and T497A/S1179D eNOS but augmented in cells transfected with S1179D eNOS. Stimulating cells with ATP or ionophore normalized the loss of function seen with T497A and T497A/S1179D eNOS to levels observed with WT and S1179D eNOS, respectively. Despite these functional differences, the localization of eNOS mutants were similar to WT. Because both T497A and T497A/S1179D eNOS exhibited higher enzyme activity but reduced production of NO, we examined whether these mutations were “uncoupling” NO synthesis. T497A and T497A/S1179D eNOS generated 2-3 times more superoxide anion than WT eNOS, and both basal and stimulated interactions of T497A/S1179D eNOS with hsp90 were reduced in co-immunoprecipitation experiments. Thus, the phosphorylation/dephosphorylation of Thr497 may be an intrinsic switch mechanism that determines whether eNOS generates NO versus superoxide in cells.
Cytochrome P450-derived epoxyeicosatrienoic acids (EETs) stimulate endothelial cell proliferation and angiogenesis. In this study, we investigated the involvement of the forkhead box, class O (FOXO) family of transcription factors and their downstream target p27Kip1 in EET-induced endothelial cell proliferation. Incubation of human umbilical vein endothelial cells with 11,12-EET induced a time- and dose-dependent decrease in p27Kip1 protein expression, whereas p21Cip1 was not significantly affected. This effect on p27Kip1 protein was associated with decreased mRNA levels as well as p27Kip1 promoter activity. 11,12-EET also stimulated the time-dependent phosphorylation of Akt and of the forkhead factors FOXO1 and FOXO3a, effects prevented by the phosphatidylinositol 3-kinase inhibitor LY 294002. Transfection of endothelial cells with either a dominant-negative or an “Akt-resistant”/constitutively active FOXO3a mutant reversed the 11,12-EET-induced down-regulation of p27Kip1, whereas transfection of a constitutive active Akt decreased p27Kip1 expression independently of the presence or absence of 11,12-EET. To determine whether these effects are involved in EET-induced proliferation, endothelial cells were transfected with the 11,12-EET-generating epoxygenase CYP2C9. Transfection of CYP2C9 elicited endothelial cell proliferation and this effect was inhibited in cells co-transfected with CYP2C9 and either a dominant-negative Akt or constitutively active FOXO3a. Reducing FOXO expression using RNA interference, on the other hand, attenuated p27Kip1 expression and stimulated endothelial cell proliferation. These results indicate that EET-induced endothelial cell proliferation is associated with the phosphatidylinositol 3-kinase/Akt-dependent phosphorylation and inactivation of FOXO factors and the subsequent decrease in expression of the cyclin-dependent kinase inhibitor p27Kip1.
Tetrahydrobiopterin, a critical factor in the production and role of nitric oxide in mast cells

Mast cells (MC) are biologically potent, ubiquitously distributed immune cells with fundamental roles in host integrity and disease. MC diversity and function is regulated by exogenous nitric oxide; however, the production and function of endogenously produced NO in MC is enigmatic. We used rat peritoneal MC (PMC) as an in vivo model to examine intracellular NO production. Live cell confocal analysis of PMC using the NO-sensitive probe diaminofluorescein showed distinct patterns of intracellular NO formation with either antigen (Ag)/IgE (short term) or interferon-γ (IFN-γ) (long term). Ag/IgE-induced NO production is preceded by increased intracellular Ca2+, implying constitutive nitric-oxide synthase (NOS) activity. NO formation inhibits MC degranulation. NOS has obligate requirements for tetrahydrobiopterin (BH4), a product of GTP-cyclohydrolase I (CHI), IFN-γ-stimulated PMC increased CHI mRNA, protein, and enzymatic activity, while decreasing CHI feedback regulatory protein mRNA, causing sustained NO production. Treatment with the CHI inhibitor, 2,4-diamino-6-hydroxypyrimidine, inhibited NO in both IFN-γ and Ag/IgE systems, increasing MC degranulation. Reconstitution with the exogenous BH4 substrate, sepiapterin, restored NO formation and inhibited exocytosis. Thus, Ag/IgE and IFN-γ induced intracellular NO plays a key role in MC mediator release, and alterations in NOS activity via BH4 availability may be critical to the heterogeneous responsiveness of MC.
Different interaction modes of two cytochrome-c oxidase soluble CuA fragments with their substrates

Cytochrome-c oxidase is the terminal enzyme in the respiratory chains of mitochondria and many bacteria and catalyzes the formation of water by reduction of dioxygen. The first step in the cytochrome oxidase reaction is the bimolecular electron transfer from cytochrome c to the homobinuclear mixed-valence CuA center of subunit II. In Thermus thermophilus a soluble cytochrome c552 acts as the electron donor to ba3 cytochrome-c oxidase, an interaction believed to be mainly hydrophobic. In Paracoccus denitrificans, electrostatic interactions appear to play a major role in the electron transfer process from the membrane-spanning cytochrome c552. In the present study, soluble fragments of the CuA domains and their respective cytochrome c electron donors were analyzed by stopped-flow spectroscopy to further characterize the interaction modes. The forward and the reverse electron transfer reactions were studied as a function of ionic strength and temperature, in all cases yielding monoexponential time-dependent reaction profiles in either direction. From the apparent second-order rate constants, equilibrium constants were calculated, with values of 4.8 and of 0.19, for the T. thermophilus and P. denitrificans c552 and CuA couples, respectively. Ionic strength strongly affects the electron transfer reaction in P. denitrificans indicating that about five charges on the protein interfaces control the interaction, when analyzed according to the Brønsted equation, whereas in the T. thermophilus only 0.5 charges are involved. Overall the results indicate that the soluble CuA domains are excellent models for the initial electron transfer processes in cytochrome-c oxidases.
The proton-pumping NADH:ubiquinone oxidoreductase (complex I) couples the transfer of electrons from NADH to ubiquinone with the translocation of protons across the membrane. Electron transfer is accomplished by FMN and a series of iron-sulfur clusters. Its coupling with proton translocation is not yet understood. Here, we report that the redox reaction of the FeS cluster N2 located on subunit NuoB of the Escherichia coli complex I induces a protonation/deprotonation of tyrosine side chains. Electrochemically induced FT-IR difference spectra revealed characteristic tyrosine signals at 1,515 and 1,498 cm−1 for the protonated and deprotonated form, respectively. Mutants of three conserved tyrosines on NuoB were generated by complementing a chromosomal in-frame deletion strain with nuoB on a plasmid. Though the single mutations did not alter the electron transport activity of complex I, the EPR signal of cluster N2 was slightly shifted. The tyrosine signals detected by FT-IR spectroscopy were roughly halved in the mutants Y114C and Y139C while only minor changes were detected in the Y154H mutant. The enzymatic activity of the Y114C/Y139F double mutant was 80% reduced, and FT-IR difference spectra of the double mutant revealed a complete loss the modes characteristic for protonation reactions of tyrosines. Therefore, we propose that tyrosines 114 and 139 on NuoB were protonated upon reduction of cluster N2 and were thus involved in the proton-transfer reaction coupled with its redox reaction.
The ABC transporter Mdl1p, a structural and functional homologue of the transporter associated with antigen processing (TAP) plays an important role in intracellular peptide transport from the mitochondrial matrix of Saccharomyces cerevisiae. To characterize the ATP hydrolysis cycle of Mdl1p, the nucleotide-binding domain (NBD) was overexpressed in Escherichia coli and purified to homogeneity. The isolated NBD was active in ATP binding and hydrolysis with a turnover of 25 ATP per minute and a Km of 0.6 mm and did not show cooperativity in ATPase activity. However, the ATPase activity was non-linearly dependent on protein concentration (Hill coefficient of 1.7), indicating that the functional state is a dimer. Dimeric catalytic transition states could be trapped either by incubation with orthovanadate or beryllium fluoride, or by mutagenesis of the NBD. The nucleotide composition of trapped intermediate states was determined using [alpha-32P]ATP and [gamma-32P]ATP. Three different dimeric intermediate states were isolated, containing either two ATPs, one ATP and one ADP, or two ADPs. Based on these experiments, it was shown that: (i) ATP binding to two NBDs induces dimerization, (ii) in all isolated dimeric states, two nucleotides are present, (iii) phosphate can dissociate from the dimer, (iv) both nucleotides are hydrolyzed, and (v) hydrolysis occurs in a sequential mode. Based on these data, we propose a processive-clamp model for the catalytic cycle in which association and dissociation of the NBDs depends on the status of bound nucleotides.
The transporter associated with antigen processing (TAP) plays a key role in the adaptive immune response by pumping antigenic peptides into the endoplasmic reticulum for subsequent loading of major histocompatibility complex class I molecules. TAP is a heterodimer consisting of TAP1 and TAP2. Each subunit is composed of a transmembrane domain and a nucleotide-binding domain, which energizes the peptide transport. To analyze ATP hydrolysis of each subunit we developed a method of trapping 8-azido-nucleotides to TAP in the presence of phosphate transition state analogs followed by photocross-linking, immunoprecipitation, and high resolution SDS-PAGE. Strikingly, trapping of both TAP subunits by beryllium fluoride is peptide-specific. The peptide concentration required for half-maximal trapping is identical for TAP1 and TAP2 and directly correlates with the peptide binding affinity. Only a background level of trapping was observed for low affinity peptides or in the presence of the herpes simplex viral protein ICP47, which specifically blocks peptide binding to TAP. Importantly, the peptide-induced trapped state is reached after ATP hydrolysis and not in a backward reaction of ADP binding and trapping. In the trapped state, TAP can neither bind nor exchange nucleotides, whereas peptide binding is not affected. In summary, these data support the model that peptide binding induces a conformation that triggers ATP hydrolysis in both subunits of the TAP complex within the catalytic cycle.
Renal mesangial cells express high levels of matrix metalloproteinase 9 (MMP-9) in response to inflammatory cytokines such as interleukin (IL)-1β. We demonstrate here that the stable ATP analog adenosine 5′-O-(thiotriphosphate) (ATPγS) potently amplifies the cytokine-induced gelatinolytic content of mesangial cells mainly by an increase in the MMP-9 steady-state mRNA level. A Luciferase reporter gene containing 1.3 kb of the MMP-9 5′-promoter region showed weak responses to ATPγS but confered a strong ATP-dependent increase in Luciferase activity when under the additional control of the 3′-untranslated region of MMP-9. By in vitro degradation assay and actinomycin D experiments we found that ATPγS potently delayed the decay of MMP-9 mRNA. Gel-shift and supershift assays demonstrated that three AU-rich elements (AREs) present in the 3′-untranslated region of MMP-9 are constitutively bound by complexes containing the mRNA stabilizing factor HuR. The RNA binding of these complexes was markedly increased by ATPγS. Mutation of each ARE element strongly impaired the RNA binding of the HuR containing complexes. Reporter gene assays revealed that mutation of one ARE did not affect the stimulatory effects by ATPγS, but mutation of all three ARE motifs caused a loss of ATP-dependent increase in luciferase activity without affecting IL-1β-inducibility. By confocal microscopy we demonstrate that ATPγS increased the nucleo cytoplasmic shuttling of HuR and caused an increase in the cytosolic HuR level as shown by cell fractionation experiments. Together, our results indicate that the amplification of MMP-9 expression by extracellular ATP is triggered through mechanisms that likely involve a HuR-dependent rise in MMP-9 mRNA stability.
We use a simple hard-core gas model to study the dynamics of small exploding systems. The system is initially prepared in a thermalized state in a spherical container and then allowed to expand freely into the vacuum. We follow the expansion dynamics by recording the coordinates and velocities of all particles until their last collision points (freeze-out). We have found that the entropy per particle calculated for the ensemble of freeze-out points is very close to the initial value. This is in apparent contradiction with the Joule experiment in which the entropy grows when the gas expands irreversibly into a larger volume.
Mapping interactions between the Ca2+-ATPase and its substrate ATP with infrared spectroscopy

Infrared spectroscopy has been used to map substrate-protein interactions: the conformational changes of the sarcoplasmic reticulum Ca(2+)-ATPase upon nucleotide binding and ATPase phosphorylation were monitored using the substrate ATP and ATP analogues (2'-deoxy-ATP, 3'-deoxy-ATP, and inosine 5'-triphosphate), which were modified at specific functional groups of the substrate. Modifications to the 2'-OH, the 3'-OH, and the amino group of adenine reduce the extent of binding-induced conformational change of the ATPase, with particularly strong effects observed for the latter two. This demonstrates the structural sensitivity of the nucleotide-ATPase complex to individual interactions between nucleotide and ATPase. All groups studied are important for binding and interactions of a given ligand group with the ATPase depend on interactions of other ligand groups. Phosphorylation of the ATPase was observed for ITP and 2'-deoxy-ATP, but not for 3'-deoxy-ATP. There is no direct link between the extent of conformational change upon nucleotide binding and the rate of phosphorylation showing that the full extent of the ATP-induced conformational change is not mandatory for phosphorylation. As observed for the nucleotide-ATPase complex, the conformation of the first phosphorylated ATPase intermediate E1PCa(2) also depends on the nucleotide, indicating that ATPase states have a less uniform conformation than previously anticipated.
Signal transducer and activator of transcription 5 (STAT5) is a transcription factor that activates prolactin (PRL)-dependent gene expression in the mammary gland. For the activation of its target genes, STAT5 recruits coactivators like p300 and the CREB-binding protein (CBP). In this study we analyzed the function of p300/CBP-associated members of the p160/SRC/NCoA-family in STAT5-mediated transactivation of β-casein expression. We found that only one of them, NCoA-1, acts as a coactivator for both STAT5a and STAT5b. The two coactivators p300/CBP and NCoA-1 cooperatively enhance STAT5a-mediated transactivation. For NCoA-1-dependent coactivation of STAT5, both the activation domain 1 and the amino-terminal bHLH/PAS domain are required. The amino-terminal region mediates the interaction with STAT5a in cells. A motif of three amino acids in an α-helical region of the STAT5a-transactivation domain is essential for the binding of NCoA-1 and for the transcriptional activity of STAT5a. Moreover we observed that NCoA-1 is involved in the synergistic action of the glucocorticoid receptor and STAT5a on the β-casein promoter. These findings support a model in which STAT5, in concert with the glucocorticoid receptor, recruits a multifunctional coactivator complex to initiate the PRL-dependent transcription.
Mitochondrial proton-translocating NADH:ubiquinone oxidoreductase (complex I) couples the transfer of two electrons from NADH to ubiquinone to the translocation of four protons across the mitochondrial inner membrane. Subunit PSST is the most likely carrier of iron-sulfur cluster N2, which has been proposed to play a crucial role in ubiquinone reduction and proton pumping. To explore the function of this subunit we have generated site-directed mutants of all eight highly conserved acidic residues in the Yarrowia lipolytica homologue, the NUKM protein. Mutants D99N and D115N had only 5 and 8% of the wild type catalytic activity, respectively. In both cases complex I was stably assembled but electron paramagnetic resonance spectra of the purified enzyme showed a reduced N2 signal (about 50%). In terms of complex I catalytic activity, almost identical results were obtained when the aspartates were individually changed to glutamates or to glycines. Mutations of other conserved acidic residues had less dramatic effects on catalytic activity and did not prevent assembly of iron-sulfur cluster N2. This excludes all conserved acidic residues in the PSST subunit as fourth ligands of this redox center. The results are discussed in the light of the structural similarities to the homologous small subunit of water-soluble [NiFe] hydrogenases.
String theory suggests the existence of a minimum length scale. An exciting quantum mechanical implication of this feature is a modification of the uncertainty principle. In contrast to the conventional approach, this generalised uncertainty principle does not allow to resolve space–time distances below the Planck length. In models with extra dimensions, which are also motivated by string theory, the Planck scale can be lowered to values accessible by ultra high energetic cosmic rays (UHECRs) and by future colliders, i.e., Mf≈ 1 TeV. It is demonstrated that in this novel scenario, short distance physics below 1/Mf is completely cloaked by the uncertainty principle. Therefore, Planckian effects could be the final physics discovery at future colliders and in UHECRs. As an application, we predict the modifications to the e+e−→f+f− cross-sections.
We calculate the antibaryon-to-baryon ratios, p̄/p,Λ̄/Λ,Ξ/Ξ, and Ω/Ω for Au+Au collisions at RHIC (sNN=200 GeV). The effects of strong color fields associated with an enhanced strangeness and diquark production probability and with an effective decrease of formation times are investigated. Antibaryon-to-baryon ratios increase with the color field strength. The ratios also increase with the strangeness content |S|. The netbaryon number at midrapidity considerably increases with the color field strength while the netproton number remains roughly the same. This shows that the enhanced baryon transport involves a conversion into the hyperon sector (hyperonization) which can be observed in the (Λ−Λ̄)/(p−p̄) ratio.
We study properties of compact stars with the deconfinement phase transition in their interiors. The equation of state of cold baryon-rich matter is constructed by combining a relativistic mean-field model for the hadronic phase and the MIT Bag model for the deconfined phase. In a narrow parameter range two sequences of compact stars (twin stars), which differ by the size of the quark core, have been found. We demonstrate the possibility of a rapid transition between the twin stars with the energy release of about 1052 ergs. This transition should be accompanied by the prompt neutrino burst and the delayed gamma-ray burst.
Recent calculations applying statistical mechanics indicate that in a setting with compactified large extra dimensions a black hole might evolve into a (quasi-)stable state with mass close to the new fundamental scale Mf. Black holes and therefore their relics might be produced at the LHC in the case of extra-dimensional topologies. In this energy regime, Hawking's evaporation scenario is modified due to energy conservation and quantum effects. We reanalyse the evaporation of small black holes including the quantisation of the emitted radiation due to the finite surface of the black hole. It is found that observable stable black hole relics with masses ∼1–3Mf would form which could be identified by a delayed single jet with a corresponding hard momentum kick to the relic and by ionisation, e.g., in a TPC.
We study the production of transversely polarized Λ hyperons in high-energy collisions of protons with large nuclei. The large gluon density of the target at saturation provides an intrinsic semi-hard scale which should naturally allow for a weak-coupling QCD description of the process in terms of a convolution of the quark distribution of the proton with the elementary quark–nucleus scattering cross section (resummed to all twists) and a fragmentation function. In this case of transversely polarized Λ production we employ a so-called polarizing fragmentation function, which is an odd function of the transverse momentum of the Λ relative to the fragmenting quark. Due to this kt-odd nature, the resulting Λ polarization is essentially proportional to the derivative of the quark–nucleus cross section with respect to transverse momentum, which peaks near the saturation momentum scale. Such processes might therefore provide generic signatures for high parton density effects and for the approach to the “black-body” (unitarity) limit of hadronic scattering.
Transverse activity of kaons and deconfinement phase transition in nucleus–nucleus collisions

We found that the experimental results on transverse mass spectrum of kaons produced in central Pb+Pb (Au+Au) collisions show an anomalous dependence on the colliding energy. The inverse slope of the spectrum increases with the energy in the low (AGS) and high (RHIC) energy domains, whereas it remains constant in the intermediate (SPS) energy range. We argue that this anomaly is probably caused by the modification of the equation of state in the transition region between confined and deconfined matter. This observation may be considered as a new signal, in addition to the previously reported anomalies in the pion and strangeness production, of the onset of deconfinement located in the low SPS energy domain.

After the introduction of the pottery tradition of La Hoguette and contemporaneous research on Earliest LBK about 10 to 15 years ago, research onthe spread of farming in Central Europe had somewhat stagnated; there were hardly any major advances in factual knowledge, nor could theoretical models be refined. In the last few years, however, an abundance of new data has appeared, partly deriving from botanical and anthropological analyses. Furthermore, newly available results from excavations in European Russia widenour understanding of the manifold and complex changes occurring during the latter 7th and 6th millennium cal BC.
El propósito de este ensayo es recorrer algunos de los secretos hilos que sitúan a Kant y a Adorno de un mismo lado en la pugna filosófica contemporánea. En el ensayo se trata de hacer patente cómo la “utopía de conocimiento” para Adorno supone, en el fondo, una reactualización de la idea de Kant de que las intuiciones sin conceptos son ciegas y los conceptos sin intuiciones son vacíos. Ello implica que tanto Kant como Adorno pretenden hallar un difícil equilibrio al mismo tiempo entre una objetividad que no olvide el papel activo del sujeto y una subjetividad que no agote en sí, al modo idealista postkantiano, la realidad del objeto.
O autor discute a importância da psicologia social de G. H. Mead para o pensamento social de Habermas. De especial interesse para o filósofo alemão é a confluência, em Mead, de duas linhas de reflexão do pragmatismo norte-americano – a filosofia analítica da linguagem e a teoria psicológica do comportamento – na crítica à filosofia da consciência. Apesar de denominar sua teoria ''behaviorismo social'', Mead se afasta das teorias do comportamento clássicas, ao enfatizar a experiência interna do indivíduo, e fazê-lo tendo como ponto de partida o ''todo social''. O artigo expõe, então, como a teoria da razão comunicativa pretende incorporar e, ao mesmo tempo, ultrapassar essa abordagem.
Semiformação (Halbbildung) é a determinação social da formação na sociedade contemporânea capitalista. Na perspectiva de Adorno, a sociedade deve ser apreendida em seu processo de reprodução material como reificação, mediação socialmente invertida. Cabe à teoria ir além do momento subjetivo da coisificação, ao decifrar as determinações objetivas da subjetividade. Não basta só revelar o sujeito por trás da reificação: ele é também socialmente determinado na adequação ao vigente, como sujeito que se sujeita e não experiencia as contradições sociais da produção efetiva da sociedade, ocultas ideologicamente na ordem social imposta pela indústria cultural. A educação não é idealista, para a emancipação, mas dialeticamente baseada na crítica à semiformação real e se orienta por possibilidades presentes, embora não concretizadas, na experiência das contradições da formação social efetiva.

Este artigo tem como objetivo mostrar a relação entre a concepção, de Adorno, da "Teoria da semiformação" e sua sugestão (juntamente com Horkheimer), na Dialética do esclarecimento, de que a indústria cultural usurpa dos indivíduos a capacidade de "esquematizar", isto é – de acordo com o ponto de vista kantiano na Crítica da razão pura –, referir sua percepção sensível a conceitos fundamentais. Já que no capítulo da Dialética do esclarecimento sobre a indústria cultural os autores não desenvolvem essa idéia, tento encontrar um desenvolvimento dela na parte intitulada "Elementos do anti-semitismo", particularmente na seção sobre a "falsa projeção". É possível mostrar, então, que a mesma concepção de semiformação, que na Dialética do esclarecimento liga a teoria sobre o anti-semitismo com a crítica à indústria cultural, poder ser considerada um conceito aplicado às questões educacionais.
O presente artigo trata da consciência filosófica expressa nas formulações de T. W. Adorno quanto ao desvanecimento da ética como possibilidade inerente ao ato educativo. Tendo em vista compreender o significado do termo "doutrina da vida reta" que, nas palavras de Adorno, teria caído em esquecimento na atualidade, o presente texto revisita o pensamento ético de Aristóteles, procurando elucidar o seu sentido imanente à educação. Por fim, extrairemos desse percurso algumas preocupações referentes ao problema da educação contemporânea sob a ótica das formulações de Adorno.
O eixo temático desta investigação trata de compreender que a obra de arte corporifica na sua forma interna uma autonomia relativa com relação à realidade empírica sobre a qual se torna reflexão crítica. Ao se caracterizar como mediação com a realidade social que a produziu, a arte é por isso mesmo a sua negação. É esse princípio de negação determinada, em que se condensam na obra de arte as antinomias e os antagonismos como antíteses da sociedade enquanto problema de sua forma interna, o elemento ao qual Theodor W. Adorno atribui dimensão epistemológica. Nessa categoria do conhecimento assim concebida, pela perspectiva estética, a razão instrumental como práxis brutal da sobrevivência é concretamente questionada na sua forma restritiva de conhecimento.
Este artigo é uma exposição do estudo analítico de um programa semanal da National Broadcasting Company (NBC), The Music Appreciation Hour, destinado a crianças e jovens escolares, realizado por Theodor W. Adorno durante sua participação no Princeton Radio Research Project (1938-1941). Tem duas partes: na primeira, Adorno analisou a pedagogia musical do programa; na segunda, apontou para as implicações culturais desse programa, que, embora não comercial e sustentado pela própria nbc, tinha todas as marcas da indústria do entretenimento comercial.
Este ensaio se propõe a rastrear, no transcurso de sua vida e de seus escritos, a seguinte asserção de Adorno: "Estudei filosofia e música. Em vez de me decidir por uma, sempre tive a impressão de que perseguia a mesma coisa em ambas", e mostrar como a relação contínua e dialética entre música e filosofia foi fecunda em sua formação educacional e científica, bem como na constituição de seu original pensamento filosófico.
O problema da universalização na filosofia moral não foi de forma alguma tematizado por Adorno. Contudo, há elementos nas suas obras que nos possibilitam refletir sobre esse tema. Tendo por base os escritos de Adorno, e esta é minha tese, pode-se estudar como a ambivalência das diretrizes normativas é integrada numa teoria crítica da moral sem que se renuncie a uma pretensão de validade crítico-normativa.

Dans un carton des archives saint-simoniennes, qui plus est consacré au "dogme", figure le manuscrit d'un compte rendu de l'essai publié en 1834, à Leipzig, par Moritz Veit, sous le titre ainsi traduit: "Saint-Simon et le saint-simonisme. Alliance générale des peuples et paix èternelle." Bien avant la prétendue révélation faite par Leroux en 1842, c'est là, sauf erreur, la toute premiëre trace en français d'une rumeur accréditée en Allemagne par Carové dès 1831. Quelles que soient l'origine et la destination de ce curieux texte (il y a lieu de le croire traduit de l'allemand ou rédigé par un Allemand de Paris), sa présence atteste que les intéressés n'ignorërent pas les soupçons d'allogénéité suscités en
France et en Allemagne par l'étrangeté de leur doctrine. L'auteur, en effet, s'appuie sur la présentation hégélianisante du saint-simonisme par Veit pour trouver confirmation de sa propre intuition, formée, dit-il, bien antérieurement, que la "clé" de toutes ces bizarreries idéologiques françaises serait "une ressemblance décidée avec la philosophie de Hegel".
Als Georg Büchner sein hintergründig ironisches Porträt eines leibhaftigen "St. Simonisten" gegen Ende Mai 1833 per Brief an die Eltern schickt, hat die "école saint-simonienne" den Gipfel ihrer ebenso sensationellen wie kurzlebigen Popularität bereits überschritten. Amüsiert erinnern Büchners Zeilen an die Eltern an den Erfolg jener weit über die Grenzen Frankreichs hinaus bekannt gewordenen Bewegung, die kurz vor und kurz nach der Julirevolution 1830 "wie ein Wunder" auf der restaurationspolitischen Schaubühne erschien und binnen weniger Jahre ein Spektakel von europäischem Ruf veranstaltete. Die Geschichte der saint-simonistischen Bewegung von der enthusiastisch begrüßten neuen Religion auf der deutschen Seite hin zur heimlichen Bewunderung der nationalchristlichen Sittlichkeitsdoktrin seitens
der Saint-Simonisten soll im Folgenden anhand ausgewählter Stationen der Aufnahme der Doktrin in Deutschland betrachtet werden. Der Treffpunkt in der deutsch-französischen Debatte über den Saint-Simonismus liegt in der Reflexion auf die Krise der Arbeit in der postrevolutionären kapitalistischen Moderne, deren Lösungsversuche an den SaintSimonismus-Texten von Friedrich Buchholz, Ludwig Börne und entlang
des berüchtigten Artikels "Über die Rehabilitation des Fleisches" in der "Evangelischen Kirchenzeitung" diskutiert werden soll.

Angesichts der fortgeschrittenen sozialen, politischen und theoretischen Verhältnisse in Paris mußte die gedankenschwere nach-hegelsche deutsche Philosophie, die in Ruges und Marx' Person zum Zwecke einer Allianz nach Paris gekommen war, in einem für sie ungewohnten Tempo eine welthistorische Entscheidung treffen. Das haben die beiden Herausgeber auch getan. Arnold Ruge entschied sich für das bürgerliche Lager, für gesetzliche Opposition, für den Idealismus, Karl Marx für die Arbeiterklasse, für die Revolution, für den
Materialismus. Diese Begriffe sind jedoch damals zwischen den beiden Beteiligten mit Gewißheit nicht gefallen, ja es ist nicht einmal sicher, ob der von der Grunddifferenz abgeleitete Streit über den Charakter der junghegelianischen Bewegung und ihrer Erneuerung thematisiert worden ist. Wir kennen Marx' Absagebrief an Ruge vom 26. März 1844 nicht, aber daß er, in welchen Worten auch immer, den grundsätzlichen Bruch
aussprach, ist sicher. Nachdem dies geschehen war, konnte man nicht mehr gemeinsam eine Zeitschrift redigieren. Gescheitert ist nicht eine persönliche Freundschaft (oder ein Zweckbündnis), sondern der (vielleicht schon unrealistische) Versuch einer gemeinsamen zeitgemäßen Erneuerung des Junghegelianismus. Während aber nun die Entscheidung von Marx in einer unübersehbaren Literatur seit Jahrzehnten kommentiert wird und sein erster Aufenthalt in Paris bis ins einzelne erforscht ist, kam Ruge weit stiefmütterlicher davon. Geht man jedoch auch seinen einzelnen Schritten nach, werden die Umstände und Hintergründe des Geschehens von Herbst
1843 bis Frühjahr 1844 mit seinen welthistorischen Folgen deutlicher. Dabei spielen Briefe, die bisher oft zu wenig herangezogen bzw. noch nicht publiziert wurden, eine große Rolle. In vielen Fällen sind sie die einzige Quelle.
Ruges Aufenthalt in Paris ist auch deshalb für die geistesgeschichtliche Forschung relevant, weil er die Peripetie in der Entwicklungskurve seiner politischer Radikalität bildet. Schon nach einem halben Jahr der Zusammenarbeit fand er Marx' Redaktion der Deutsch-Französischen Jahrbücher unpassend und überwarf sich mit allen seinen "kommunistischen" Freunden.
Die Geschichte der französischen Philosophie im 19. Jahrhundert beginnt als akademische Fachrichtung mit Victor Cousin, der weit mehr als seine blassen Vorgänger Laromiguière oder Royer-Collard die akademische Disziplin geprägt hat. Cousin stellte als offizieller Philosoph
der Julimonarchie, als Minister und als Vorstand des Ausschusses zur Auswahl der Gymnasiallehrer eine Art Galionsfigur der Geisteswissenschaften dar, und hatte einen entscheidenden Einfluss sowohl auf die Personalpolitik wie auf die Definition der Lehr- und Forschungsgegenstände. Er bekannte sich zu einer als Eklektizismus umschriebenen
Doktrin, die einerseits die Bewahrung der gegebenen Ordnung der Dinge auch in religiöser Hinsicht anstrebte, andrerseits die Philosophie von jeder Form religiösen Bekenntnisses emanzipierte und gewissermaßen den neutralen Geist der späteren "Laïcité" ankündigte. Das sogenannte
Regiment von Victor Cousin, d.h. die Philosophielehrer, deren Ernennung und Beförderung von seinem Gutdünken abhing, konnte sich auf seine Rückendeckung verlassen, wenn sie in Konflikt zu der religiösen Obrigkeit gerieten. Andrerseits mussten sie sich wie die Vertreter einer einzigen Doktrin, seiner Doktrin verhalten. Nun findet Cousins Doktrin ihre Wurzeln wenn nicht in der deutschen Philosophie - Heine bemerkte ironisch, dass Cousins Unkenntnis des Deutschen ihn gegen jeden
Verdacht philosophischer Deutschtümelei schützte -, so wenigstens in einem privilegierten Deutschlandbezug.
"Wahlverwandtschaften" : Programme einer deutsch-französischen Allianz von Heine bis Ruge und Marx

Nichts zeichnet den interkulturellen Transfer im Vormärz so aus wie der Versuch, die fortschrittlichsten Eliten Deutschlands und Frankreichs enger als je zu verknüpfen. Nie zuvor, und wohl nie danach, haben sich deutsche und französische Dichter und Denker so zahlreich für die geistige Entwicklung ihres Nachbarlandes begeistert - bereit, alles national Trennende zurück- und alles kulturell Verbindende voranzustellen. Mehr noch: Nie zuvor und niemals später haben sich führende Schriftsteller und Intellektuelle beider Länder derart intensiv damit beschäftigt, die Grundlagen einer informellen, geistigen Allianz zu erarbeiten. Man nenne eine Epoche, in der die Verwirklichung dieser wahrhaft europäischen Idee so greifbar nah war wie etwa 1844!
Wer dieser verschollenen Randfigur der Literatur des 19. Jahrhunderts nachspüren will, wird Mühe haben. Einschlägige deutsche Lexika widmen ihr keine Zeile, und ihr Name wird nur in älteren französischen Nachschlagewerken überliefert, die allerdings die wohl unvermeidlichen Lücken und Fehler aufweisen. Die trockenen Fakten deuten ihre Verdienste kaum an, und die Tatsache, dass sie in Frankreich posthum zu
einer Steadyseller-Autorin wurde, zwar nicht mit eigenen Werken, sondern mit Übersetzungen, ist nicht zu erkennen.
Der Versuch, dieser Randfigur Relief zu verleihen, muss infolge mangelnder Vorarbeiten und der wenigen entdeckten Quellen ein Unterfangen bleiben, das vorerst kein erschöpfendes Ergebnis zeitigen kann, eher ein Provisorium, das vielleicht zu weiterer Forschung anreizt.
Nach seiner Erfindung um die Wende vom 18. zum 19. Jahrhundert wurde der romantische Rhein in Deutschland bald als Abgrenzungskriterium zwischen Deutschland und Frankreich instrumentalisiert. Eine Ursache ist selbstverständlich die politische Situation: Deutschland ist zersplittert und steht unter französischer Besatzung, als sich
Clemens Brentano und Achim von Arnim 1802 auf ihre berühmte
Rheinfahrt begeben und Friedrich Schlegel 1803 auf der Reise nach Frankreich, über die er in seiner Zeitschrift Europa berichtet, den Rhein als Sinnbild Deutschlands entdeckt.
Tragfähig konnte die politische Instrumentalisierung des Rheins allerdings nur deshalb sein, weil sie auf einer seit der Jahrhundertwende schnell angewachsenen und von breitesten Bevölkerungsschichten in Deutschland mitgetragenen Tradition beruhte. Solche Traditionen zu
stiften und damit überhaupt erst so etwas wie eine nationale deutsche Kultur, in Absetzung und Überbietung von anderen, bereits bestehenden Nationalkulturen wie etwa der französischen zu begründen, war eines der Hauptanliegen der romantischen Bewegung. Insofern ist selbst den
Gründervätern des Rheinmythos zumindest indirekt der deutsch-französische Gegensatz als Antrieb nicht fremd.
Während sich die Regierung Louis Philippes konsolidiert, lässt der revolutionäre Enthusiasmus unter den Literaten merklich nach. Innerhalb des neuen historischen Kontexts,[...] gewinnen zwei Avantgarde-Bewegungen Kontur: La Jeune France und das Junge Deutschland. Beide prägen die öffentlich geführte Auseinandersetzung um das Funktionsverständnis von Literatur nach 1830. Ihre Vertreter opponieren gegen die sowohl in Frankreich als auch in Deutschland restriktive (Kultur-)Politik, setzen ihr künstlerische Vorstellungen entgegen, die der veränderten Lebenswirklichkeit einerseits und der jeweils favorisierten
authentischen Ausdrucksweise andererseits, ihrer subjektiven Weltsicht und ihren individuellen ästhetischen Vorlieben, entsprechen. Mag eine solch kurze Charakterisierung wie auch die ähnlich klingenden Bezeichnungen den Gedanken nahelegen, es könne sich um homogene Gruppierungen mit ähnlichen Zielsetzungen handeln, so ist dies nur teilweise richtig. Ihre unterschiedlichen Auffassungen von der Rolle des
Künstlers innerhalb der postrevolutionären Gesellschaft, seiner Themenwahl und Schreibweise, können vielmehr als paradigmatisch für die allmähliche Herausbildung der Dichotomie von autonomer und engagierter Literatur angesehen werden. Ihr vergleichbarer Ausgangspunkt ist die Kritik am Bestehenden, sowohl in politischer wie auch in künstlerischer Hinsicht.
Da Frankreich und dessen jüngste Geschichte für Börne mehr bedeuteten als ein Motiv seines schriftstellerischen Interesses und seiner Lebenswelt unter anderen, weil ihm sowohl Frankreich wie Deutschland, ob ausgesprochen oder latent, Welt und Gegenwelt waren, in denen sich Existenz wie Denken abspielten und abspiegelten, kann in diesem Rahmen nur auf wenige Facetten seines Schreibens eingegangen werden. Es wird sich daher im Wesentlichen um die Rolle handeln, die Frankreich und besonders Paris für seine Arbeit als Journalist und als Zeithistoriker
wie für den Ansatz seines geschichtsphilosophischen Denkens überhaupt gespielt haben.

Überlegungen, die sich einem der bedeutendsten Kapitel der Beziehung zwischen der deutschen und französischen Literatur widmen, können an der Gestalt und dem Werk Heinrich Heines nicht vorbei gehen. Oft genug gibt er gar das Modell ab für politische und soziale Funktionen der Literatur in ihrer die Grenzen überschreitenden Möglichkeit. Heine selbst liefert dafür immer wieder Beispiele der Vermittlung in unterschiedlichster Weise. Den Franzosen die deutsche Eigenart wie Geisteswelt und den Deutschen die künstlerischen, intellektuellen wie revolutionären Errungenschaften der Franzosen nahe zu bringen, betrachtete er als Lebensaufgabe seiner letzten 25 Jahre von 1831 bis 1856, die er in Frankreich bzw. Paris verbracht hat.
Die immer leise, aber immer vernehmbare Stimme der Vernunft ließ sich trotz aller wiederkehrender Dramatik nie zum Verstummen bringen, selbst in Kriegsphasen nicht. Der Kulturtransfer, um den es hier geht, konnte vielmehr seinem eigenen Rhythmus folgen und ungeachtet aller Krisen so etwas wie eine selbständige Gesetzmäfligkeit entwickeln. Unter Schriftstellern und Intellektuellen gab es Schulstreite,
aber nie Krieg! So wurden besonnene und fortschrittlich eingestellte Franzosen und Deutsche nie müde, sich für Gedankenaustausch und Vermittlung, für Freundschaft und Verständigung, kurz für Ideentransfer einzusetzen - um damit den kreativen Eliten beider Länder Inspirationsquellen offen zu halten. Den Herausgebern dieses Jahrbuchs will scheinen, diese gemäfligten, die Rheingrenze ständig überschreitenden Gegenstimmen hätten wie ein vernunftbestimmtes Korrektiv zur lauten Staatenpolitik gewirkt. Eine Epoche wie der Vormärz bietet nun die große Chance, diese These einmal in einer kurzen, aber scharf konturierten Phase bilateraler Beziehungen der Probe aufs Exempel zu unterwerfen. Es erschien anregend, die verschiedenen Wirkungsweisen dieses Korrektivs in einer Zeit näher zu untersuchen, in der sich die herrschenden Kreise auf der einen Seite durch Gallophobie "auszeichneten", während ihnen auf der
anderen Seite - rares Phänomen - eine durch nichts getrübte Germanophilie gegenüberstand.

Nie zuvor und nie danach haben sich französische und deutsche Kultur so tiefgreifend beeinflusst und durchdrungen wie in der ersten Hälfte des 19. Jahrhunderts. Wer heute von einer deutsch-französischen Synthese spricht, sollte auf die Zeit von Julimonarchie und Vormärz zurückgehen: Dort konnte sich diese Synthese in bedeutenden Ansätzen verwirklichen.
Die Anordnung der in diesem Jahrbuch versammelten Beiträge folgt drei thematischen Schwerpunkten: dem literarischen, dem künstlerischen und dem philosophischen Transfer.
A publicação deste polêmico artigo é uma homenagem de Lua Nova aos oitenta anos de História e Consciência de Classe, de Georg Lukács. Seu autor faz um resgate do jovem Lukács, identificando os pressupostos e idéias que tornam esse livro de 1923 não tanto filosoficamente, mas politicamente, um feito revolucionário, em sintonia com o ''Evento de 1917'' na Rússia. Zizek entende que é justamente seu teor político, o qual aponta para uma crítica radical dos regimes liberal-democráticos predominantes no Ocidente, que o mantém atual, nesse sentido ultrapassando os limites auto-impostos dos autores da Dialética do esclarecimento.
Es war eine heißer, schwüler Sommerabend, als John Rawls im Hörsaal H der Frankfurter Universität einen Vortrag hielt. Er sprach leise, fast schleppend, und er hatte sich vorgenommen, den Text in einer deutschen Übersetzung vorzulesen, was für einen amerikanischen Professor ungewöhnlich war und deshalb Bewunderung verdiente. Doch war die angespannte Konzentration spürbar, die Rawls aufbringen musste, um deutscheWorte mit so wenig amerikanischer Phonetik wiemöglich zu sprechen, und der Vortrag wurde dadurch noch langsamer, die Stimme noch leiser. Außerdem funktionierte das Mikrofon nicht richtig. Deshalb wurde es ihm von seinem Übersetzer, Wilfried Hinsch, mit ausgestrecktem Arm so nahe an denMund gehalten, dass wenigstens ein paar Worte zu verstehen waren. Nach kurzer Zeit verließen die ersten Zuhörer den Hörsaal. Der ausgestreckte Arm des Helfers wurde sichtbar schwerer; Anstrengung und Hitze ließen Schweißbäche rinnen und das Oberhemd nass werden.Der Vortragwar nicht einfach. Rawls machte, wie gewohnt, keinerlei Konzessionen, sondern diktierte einen komplexen Satz nach dem anderen. Wer etwas verstehen wollte, musste von der komischen Situation absehen, alle Kräfte gegen die von der schwülen Hitze geführten Ermüdungsattacken aufbieten und sich irgendwie konzentrieren. Der einzige, der sich davon nicht beirren ließ, sondern hartnäckig Satz für Satz in den schwülen Sommerabend hämmerte, war der kleine, schmächtige, blasse, sein Gesicht hinter einer riesigen Brille verbergende, aber Respekt heischende Professor Rawls. Wenn Geist so unmittelbar präsent ist, wird eben alles andere banal. ...

Die juristische Europakarte bestand bis vor kurzem aus dem common law, dem romanistischen civil law und dem sozialistischen Recht. Angesichts dessen Niedergangs und des Zusammenwachsens Westeuropas vereint man die beiden ersteren unter dem altsowjetologischen Schild der western legal tradition, wodurch das Ostrecht aus der europäischen Rechtsgeschichte ausgegrenzt wird. Coings Weitsicht, Osteuropa einschließlich Russlands mindestens teilweise in die Disziplin einzubeziehen, findet praktisch keine Nachahmer. Immer noch versteht man, in Savignys Fußstapfen tretend, unter der "europäischen" mehr oder minder stillschweigend die westeuropäische Rechtsgeschichte. Die "Reformstaaten Mittel- und Osteuropas" tauchen in manchem Traktat der Rechtsvergleichung wie deus ex machina auf. Während man die Zäsur zwischen civil law und common law vernachlässigt, wird die europäische Natur Osteuropas, besonders Russlands und der Balkanländer, angezweifelt. ...
Das "Office for Strategic Studies" (OSS) wurde erst 1942, ein Jahr nach Kriegseintritt der USA geschaffen, es war ein Vorläufer des CIA. Seine Aufgaben bestanden nicht allein in der Beschaffung von Informationen über den Zustand der feindlichen Streitkräfte in Europa und Asien, sondern unter anderem auch in der Entwicklung von Möglichkeiten und Vorstellungen für die – nach erfolgreicher Beendigung der Kämpfe – einsetzende Aufbauarbeit in den zu besetzenden Ländern. Dieser Aufgabe diente auch die Beauftragung Carl Zuckmayers mit einem informativen Bericht über die politische und moralische Zuverlässigkeit, oder wenigstens "Brauchbarkeit" von Angehörigen der künstlerischen Elite, soweit sie im Deutschen Reich geblieben war; brauchbar nämlich für Beteiligung am kulturellen Leben in einem neuen, demokratischen Deutschland. An dieser Aufgabe arbeiteten in der Zentrale des OSS unter anderen HerbertMarcuse und Franz Neumann, die beide dem nach den USA ausgewanderten Frankfurter Institut für Sozialforschung angehört hatten. Diese linken Wissenschaftler legten in ihren Arbeiten für das OSS mehr Wert auf die Entmachtung der in Nazideutschland einflussreichen und besitzenden Eliten und auf eine von den Alliierten zu initiierende "re-education". Im Unterschied dazu dienen die Berichte Zuckmayers lediglich der differenzierten Beurteilung von Autoren, Regisseuren und vor allem Schauspielern, die in Deutschland geblieben waren und sich –mehr oder weniger – dem Regime angepasst hatten. ...
Journalisten fragen, Politiker antworten. Kurz nach Beginn des Ersten Weltkrieges, im November 1914, hat der Strafrechtler und Völkerrechtler Franz von Liszt auf Fragen des Chefredakteurs Helmer Key vom "Svenska Dagbladet" in Stockholm geantwortet – in bemerkenswert enger Anlehnung an die alte konservative Theorie von der deutschen Friedfertigkeit, die erst durch die Einkreisungspolitik Eduards VII. zur Wehrhaftigkeit habe werden müssen.

Ein beunruhigendes Gefühl, ausgelöst durch die knappe Mitteilung auf dem Anrufbeantworter: Walter Wilhelm ist gestorben. Das Gefühl, dass es mir zukomme, über diesen Toten einen Text zu schreiben. Weil anders wahrscheinlich niemand über ihn schreiben würde. Was freilich gleichgültig ist, wegen der Kraftlosigkeit von Geschriebenem, das sich einer Person zu bemächtigen sucht, und angesichts der Flüchtigkeit von Texten im Gedächtnis. Wozu also schreiben – zumal Walter Wilhelm ein emphatischer Gegner akademischer Nachrufe war, grimmiger Verächter sentimental verklärter Lügengeschichten und ins Grab geschleuderter professoraler Hochrufe, die dem einsamen Leben nicht gegönnt waren? Aber manche Worte lassen sich nicht auf Dauer bändigen, bohren sich hartnäckig in die Geschäfte des Tages, winken und starren abends mit vorwurfsvollen Augen auf eine beginnende Bequemlichkeit. Man muss ihnen nachgeben und hoffen, dass sie einen nicht unversehens im Stich lassen.

Die 1968 gegründete Zeitschrift "Kritische Justiz" stand anfangs am äußerst linken Rand der juristischen Publikationen. Auch der Rezensent, der in einer Ausgabe des Jahrgangs 1970 die Neuauflage der Lebenserinnerungen des KommunistenMax Hoelz "Vom 'Weißen Kreuz' zur roten Fahne" besprach, machte aus seinem dezidiert linken Standpunkt kein Geheimnis: die Neuauflage sei "wichtiges Lehrmaterial für die gegenwärtigen Klassenkämpfe" (Kritische Justiz 1970, 363–365). ...
In Band 1 dieser Zeitschrift wurde ein "Vorschlag" gemacht, wie – mit welchem Erkenntnisziel und welchem theoretischen Gerüst – man Rechtsgeschichte betreiben könnte. Dieser Vorschlag wurde von zwei Kollegen kommentiert: Simon Roberts zeigte sich skeptisch, ob ein systemtheoretischer Ansatz für vormoderne Gesellschaften überhaupt brauchbar sei, und besorgt um den Verlust des Menschen sowie konkurrierender Theorien in der Geschichtsschreibung. Marc Amstutz hingegen drängte auf erhöhte theoretische Genauigkeit, insbesondere was die Bedeutung exogener und endogener Faktoren von evolutiven Prozessen betrifft. Beiden sei herzlich gedankt. ...

"Falls Europa erwacht", werde es sich in ein post-imperiales "Reich" der Mitte verwandeln, damit zugleich den zweitausendjährigen Verwandlungsreigen europäischer Imperien überwinden. Sloterdijks Europa-Vision entspringt jedoch keiner neuen Utopievorstellung, vielmehr setzt der Philosoph aus Karlsruhe auf bereits vorhandene, kulturell tief verankerte Potentiale. Gemeint ist die Fähigkeit der Verwandlung, die Kraft der Metamorphose. ...

Geplünderte, rauchende Paläste, umgestürzte Statuen, ausgeweidete Museen – anders als die Ruinen von Bagdad, an die uns der TV-Jahresrückblick Ende 2003 erinnern wird, verweist die nur noch auf Bildern existierende Ruine der New Yorker Twin Towers nicht auf die Folgen eines Krieges, in dem auch das Völkerrecht ruiniert wurde, sondern auf die Folgen eines Terroranschlags, der das einstige stolze Symbol Manhattans in weniger als zwei Stunden in Schutt und Staub verwandelte. ...
So geht es einer Stadt, die Frieden schaffen sollte – selbst, aus eigner Kraft! – und für den Sieg gebetet hat. Als ob es Siege gäbe, wenn die Menschen sterben.
Wer diese Euripides-Verse in der Übertragung von Walter Jens im März 2003 am Vorabend, buchstäblich am Vorabend, eines Krieges gelesen hat, kann sich über den Titel des Buches, in dem sie stehen, nur wundern: "Ferne und Nähe der Antike". Wieso denn "Ferne"? Euripides- Jens ist so nah wie der Krieg nah war. Mehr gibt es dazu nicht zu sagen. ...
Wozu Rechtsgeschichte?

Es ist kaum noch zu ertragen. Immer wieder diese alten Fragen. Warum Aufklärung? Was können wir wissen? Wozu sollen wir das oder jenes wissen? Turfanforschung – schon das Sujet ist dem Normalsterblichen ein Rätsel. Ägyptologie – hier verspricht der Gegenstand wenigstens eine vorstellbare und noch heute erlebbare Exotik. Zwei Wochen Nilfahrt. Aber das alte Schreiben und Denken und Musizieren der Turkmenen? Wer will das kennen lernen? Wem mögen die Kenntnisse über die Turkmenenkultur nutzen? Dann doch lieber gleich Kulturwissenschaft tout court. Das kommt an. Also etwa die Kultur des Riechens, von der Toilette über das Parfum zu den Gerüchen der Städte. Damals und heute. Da kann, zu welchem Behufe auch immer, jeder mitriechen – auch der kultivierte Steuerzahler, der die Kulturwissenschaft bezahlt. Die ebenso zahlenden unkultivierten Bürger interessieren nicht weiter. Oder das sagenhafte Gehör der Wüstenspringmaus. Das ist Grundlagenforschung, bei der sich immerhin ahnen lässt, es könnte à la longue etwas dabei herauskommen, ein neues Hörgerät, nicht für Mäuse, sondern für Menschen. ...

Für Jean-Paul Sartre war die Rettungsfolter am 6. März 1958, als er diesen Satz schrieb, eine "Heuchelei". Zwei Wochen zuvor war "La Question" von Henri Alleg, dem Herausgeber des Alger Républicain, der einzigen freien Tageszeitung Algeriens, in den Éditions de Minuit erschienen: der vielleicht aufregendste, weil auf eigener Erfahrung beruhende und doch so kühl erzählte, Bericht über die Praxis in den französischen Folterkellern Nordafrikas. Sartre griff zur Feder und schrieb über die Folter im Zeitalter der Zivilisation, des 20. Jahrhunderts, das schon in seiner Mitte massenhaft Folterungen gesehen hatte, das die Folter zur Raserei werden ließ, das den Folterknecht in einen Sisyphos verwandelte, der immer wieder zur Tortur zurückkehren muss, in einem Jahrhundert, in dem die Folter endlich ihren Grund gefunden hat: sich selbst. Die letzten Jahrzehnte sahen, wie die ersten Jahrzehnte des 20. Jahrhunderts, die Globalisierung des Schmerzes der Gefolterten – der unterworfenen Untermenschen der Moderne. In dieser weltweiten Orgie der Qual – nicht nur dokumentiert in den Berichten von Amnesty International – geht es kaum um die Rettung von Menschen, um das Erzwingen von Geständnissen, um das Aufdecken einer Wahrheit. Es geht um die totale Herrschaft von Menschen über Menschen (Jan Philipp Reemtsma), um das Zusammenhalten der Gesellschaft durch die Drohung mit körperlicher Gewalt (Adorno), um den Blutkitt zwischen den Menschen, der diese beisammen hält, um die "Ich-Ausdehnung" der Peiniger (Lutz Ellrich). Die Folter des 20. Jahrhunderts – ob in Chile, in Indonesien oder im Deutschen Reich – kommt ohne Recht aus, interessiert sich nicht für in Körpern eingeschlossene Wahrheiten und feiert Feste der Angst. ...
Bartolus "down under"

In einem abgedunkelten Raum der Heidelberger rechtshistorischen Bibliothek am Friedrich-Ebert-Platz findet man separiert die wertvollen Rara-Bestände; darunter auch die letzte Ausgabe der Opera Omnia des Bartolus von Sassoferrato (1314–1357), die in Venedig 1615 für die Bedürfnisse juristischer Praktiker verlegt wurde. Friedlich vereint stehen die Folianten Rücken an Rücken, Regal an Regal, Wand an Wand – so als ob sie seit den Gründungsjahren der Universität im 14. Jahrhundert, mindestens aber seit der Zeit der Druckerpresse und damit ihrer eigenen Herstellungszeit, schon immer so gestanden hätten. Kommunizieren die Bücher etwa heimlich miteinander, wenn niemand sie beobachtet? ...
Hein Kötz hat es vor Jahren schon festgestellt: "Rechtsgeschichte und Rechtsvergleichung sind Holz vom gleichen Stamm. Sie sind Zwillingsschwestern." Und wie dies mit Zwillingen nun einmal so ist – sie lassen sich bisweilen nur schwer auseinander halten. So geht es auch dem Leser von Stefan Vogenauers fünfzehnhundert Seiten starker Dissertation zur Auslegung von Gesetzen in England und auf dem Kontinent: Mochte man als Rechtshistoriker zunächst noch glauben, es handele sich dabei um Rechtsvergleichung, wurde der "monumentalen Untersuchung Vogenauers" inzwischen bescheinigt, sie sei auch "als bedeutende rechtshistorische Leistung" zu würdigen. Aber macht es denn tatsächlich so viel aus, welche der beiden Disziplinen die Arbeit für sich vereinnahmen kann? ...

Moderne Selbstbeschreibungen können durchaus produktiv die historische Forschung zu neuen Paradigmen anregen. Die derzeit sehr populäre Vorstellung von "Netzwerken" lässt sich mit Gewinn sogar auf historische Gesellschaften übertragen. Die Folgen dieses Konzepts für Theorien der Herrschaft beschäftigten am 5. und 6. Dezember 2002 am Europäischen Hochschulinstitut in Florenz einen von Peter Becker und Raffaele Romanelli organisierten Workshop – sein Thema: "Governing through Networks: International Collaboration between Public Administrators". Der Veranstaltungstitel bildet nicht ab, dass es sich um ein dichtes Expertengespräch zum 19. Jahrhundert handelte. Thomas Nutz (München)berichtete über den sich internationalisierenden Gefängnisdiskurs in der ersten Hälfte des Jahrhunderts, Pietro Corsi (Paris) breitete imposantes Material über die bis heute fruchtlosen Versuche italienischer Geologen aus, eine vollständige Karte des Landes zu erstellen. Hier wie auch bei den Statistikern (Silvana Patriarca, Fordham) wurde deutlich, welch überragende Rolle internationalen Netzwerken von wissenschaftlichen Experten im Verlauf von nur wenigen Jahrzehnten zugewachsen war. Sie organisierten sich in privaten Vereinen oder halbstaatlichen Organisationen und bildeten grenzüberschreitende Wissenssysteme, denen zunehmend eine Schlüsselstellung bei der politischen Entscheidungsfindung zukam. ...
Brüssel im November. Drei Tage lang präsentiert die Europäische Union "Research Europe 2002", die Auftaktveranstaltung zum 6. Forschungsrahmenprogramm. Ort des Geschehens: die Messehallen von Heysel. An die zehntausend Teilnehmer waren angemeldet und Tausende sind gekommen, Forscher, Forschungspolitiker, Forschungsverwerter aus ganz Europa, mancher auch von noch weiter her – etwa ein Drittel aus Wirtschaft und Industrie, viele Osteuropäer, viele junge Leute mit Elan und European spirit. Drei Tage der Vorträge, der Podiumsdiskussionen, der Workshops, der Schulungsveranstaltungen – alles mit dem einen Ziel, das zu konstituieren, worum es aller Förderprogrammatik letztlich geht: den gemeinsamen europäischen Forschungsraum. ...
Rechtsgeschichte wozu? Die Frage drängt, diesen und jenen Nutzen anzuführen oder zu bestreiten. Doch wozu? Vor welchem Forum? Vor welchem Maßstab? Nützlich oder unnütz für wen: – für Laien, Juristen, Historiker, Wissenschaftler überhaupt, – für Jus-Studierende, Rechtsprofessoren, Juristen überhaupt, – für Rechtspolitiker, Hochschulpolitiker, Politiker überhaupt, – für Geschichtsspieler, Rechtsspieler, Spieler überhaupt, usw.? Für Quellenliebhaber, Exotikfreunde, Fällesammler, – usw.? ...
Es gibt sogenannte "Fakten" oder "Tatsachen" der Geschichte, die sich nach intensiver Überprüfung als Fiktionen erweisen. Es gibt Vorstellungen, die jahrhundertelang als gesichertes Wissen galten und bis heute in Enzyklopädien und einschlägigen Handbüchern zu finden sind. Ihre Faktizität gilt als gesichert; man sieht sie als "wirklich bestehende Sachverhalte" an. Und doch entpuppen sich immer wieder vermeintlich gesicherte Tatsachen als fiktiv. Jedoch können solche "fiktiven Tatsachen" in verschiedenen Zusammenhängen – und sei es "nur" in der Wissenschaftsgeschichte – ein Eigenleben entwickeln. Der traditionelle Begriff der Fälschung greift hier nicht mehr. Neuerdings verbreitet sich der Begriff der "imaginären Tatsache". ...
Jedem von uns ist das schon begegnet: ich will ein Wort verwenden. Es fällt mir einfach nicht ein. Mein Hirn macht sich auf die Suche und legt erst einmal alles andere lahm. Meine ganze Aufmerksamkeit peilt dieses eine Wort an, das sich entzieht. Ich kann mich auf nichts anderes konzentrieren. Ja, ich kann nicht einmal einschlafen, weil dieses unauffindbare Wort alle Funktionen, sogar vegetative, stört. Es hat sich mit dem ganzen Wortfeld eingekapselt, abgeschottet.
Wenn ich wenigstens ein Synonym finden könnte, irgendein Wort, das sinngemäß eine Richtung weist, ich könnte mich daran weiterhangeln, auch wenn mir der Boden unter den Wörtern abhanden gekommen ist. Grad noch das Wort davor ist verfügbar, ich klammere mich dran fest. Der Wortfluß, bisher moderat, schwemmt alles weg - und wo grad noch ein Meer von Wörtern war, nichts als eine wortlose Dürre. Nichts mehr da.

Die Worte Österreich, England, Europa, London, Wien habe ich das erste Mal im Jahr 1962 gehört, bei einem unserer Ausflüge zu den Hügeln draußen vor der Stadt und zu den Wäldern und Tieren. Ich hörte sie das erste Mal im Zusammenhang mit einem Brief, den die Mutter vorgelesen hat, laut gelesen hat, weil sie die Sprache in diesem Brief gerne hörte, laut, weil es ein englischer Brief war, der nicht auf Englisch geschrieben war, weil sie vielleicht dieses Aber-Englisch hören wollte, laut vielleicht, dass ich zuhören konnte. Es sei ein Brief, der von einem Kind namens Katharina erzählt und von einem Fisch. "Ich lese den englischen Brief vor", sagte die Mutter, dann las sie auf Deutsch. So hatte das Kind die Gewissheit, dass das Englische und Deutsche eins sind.

Auf einem der Hügel auf dem Festland standen im Halbkreis Hütten, dort zogen englische Ferienfamilien ein, spielten Fußball und Tennis und Badminton, Leser saßen dazwischen und lasen The Times und The Daily Mirror und Kriminalromane und malaysischenglische Wörterbücher. "Achte einmal darauf, ich lese dir stückerlweise den Brief aus Österreich vor." Österreich? Österreich? "Ja, das ist ein Land, das weit weg in der Mitte von Europa liegt." Europa? Europa? "Ja, das ist eine Sammlung von Ländern, eine Art Asien wie hier, wo wir sind." Wer hat dir den Brief geschickt? "Den Brief hat mir niemand geschickt. Der Brief ist an Francis Bacon in England gerichtet worden, vor vielen hundert Jahren." Wer hat ihn geschrieben? "Das weiß ich nicht. Ein Mann namens Philipp Chandos, ich weiß nicht, wer der war. Ich weiß nur von einem anderen Chandos namens Grey, 5th Baron Chandos. ...

Briefe und Notizen, die erstmals in der Kritischen Ausgabe abgedruckt wurden, bleiben hier unberücksichtigt. Jede bibliographische Angabe erhält eine Ordnungsnummer, ausgenommen davon sind in der Regel Rezensionen. Einzelkritiken zu aktuellen Inszenierungen sind nur in Ausnahmefällen aufgenommen.
Obwohl Kafka den "Landarzt" besonders schätzte, hat sich die germanistische Forschung dieser Erzählung nur zögerlich angenähert. Angesichts der kompositorischen Komplexität und aufgrund der fast undurchdringlichen Hermetik dieser Geschichte schien es vielen Interpreten opportun, sich auf vage und allgemein gehaltene Deutungsmuster zurückzuziehen. Ältere Studien begnügten sich häufig damit, den "Landarzt" als symbolisch verschlüsselte Darstellung der conditio humana zu lesen oder als Variation der in Kafkas Werken allgegenwärtigen Todessehnsucht zu interpretieren. Seit den sechziger und siebziger Jahren widmete sich dann vor allem die psychoanalytische Literaturwissenschaft dem "Landarzt", da zentrale Handlungssequenzen in Kafkas Meistererzählung den Einfluss Sigmund Freuds verraten.
Hofmannsthal wurde auf das Motiv des Musikinstruments im "Jedermann" durch eine briefliche Mitteilung des Komponisten Clemens zu Franckenstein vom 12. April 1903 hingewiesen, der bei der Beschreibung einer Londoner Aufführung des "Everyman" besonders die musikalischen Elemente und Requisiten hervorhob: "Everyman" […] geht langsam, ein Serenadenartiges Lied singend und begleitet sich auf der Laute die er an einem Bande umgehängt trägt […] "Fellowship" […] tritt auf "Everyman" zu. "Everyman" reicht ihm beim Abschied seine Laute.
Die Idee, das Instrument zunächst in seiner Funktion als Begleitinstrument eines Liedes einzuführen und es zuletzt zum Zeichen für den endgültigen Abschied vom Freund zu setzen, dürfte Hofmannsthal dieser Bühnenbeschreibung Franckensteins entnommen haben. Die erotischen Konnotationen, die das Requisit im Prosa-Jedermann gewinnt, und vor allem die symbolische Zerschneidung seiner Saiten, als es am Ende dem Freund gegeben wird, hat Hofmannsthal neu eingebracht.
Hofmannsthals "Gespräch über Gedichte", nur ein Jahr nach dem Chandos-Brief konzipiert und 1904 erstmals, mit dem Titel "Über Gedichte", veröffentlicht, leidet unter einer fatalen Anschuldigung. "Hofmannsthal sucht im Georgedialog das ästhetische Symbol als Opferritual zu fassen", erklärt Theodor W. Adorno in einem Essay von 1939. Er zitiert dann die berühmte Erzählung Gabriels vom ersten Opferer, mit ein paar Auslassungszeichen allerdings, und kommt zu dem Schluß: "Diese blutrünstige Theorie des Symbols, welche die finsteren politischen Möglichkeiten der Neuromantik einbegreift, spricht etwas von ihren eigentlichen Motiven aus. Angst zwingt den Dichter, die feindlichen Lebensmächte anzubeten: mit ihr rechtfertigt Hofmannsthal den symbolischen Vollzug. Im Namen der Schönheit weiht er sich der übermächtigen Dingwelt als Opfer." Blutrünstig und politisch finster, Angst und Selbstopfer - da öffnet sich ein ästhetischer und gleich auch politischer Abgrund. Denn umstandslos setzt dieser Begriffswirbel Symbol (also auch Poesie) und Blutopfer, den Dichter und den Opferer gleich.
Nachfolgende Beobachtungen zum Chandos-Brief Hofmannsthals haben die im letzten Drittel des Briefes beschriebenen "guten Augenblicke" zum Thema, die als exemplarische Darstellungen einer ästhetischen bzw. poetischen Erlebnisweise ausgewiesen werden sollen. Getragen ist also die Lektüre von der vielleicht kontroversen Voraussetzung, daß in die Schilderung der guten Augenblicke ein poetologischer Vorstellungskreis eingeht, der auch über die Grenzen des Chandos-Briefs hinaus für Hofmannsthals Schreiben bestimmend bleibt. So wird sich die Argumentation auch auf spätere Texte beziehen, zumal auf solche wie "Das Gespräch über Gedichte" oder den Vortrag "Der Dichter und diese Zeit", deren Anliegen ein explizit poetologisches ist.
Den berühmten Text, um den es hier geht, schrieb Hugo von Hofmannsthal im Sommer 1902 und veröffentlichte ihn im selben Jahr in zwei Folgen in der Berliner Zeitschrift "Der Tag". Aus demselben Sommer sind viele persönliche Briefe Hofmannsthals erhalten, die dasselbe Thema der Sprach- und Schreibkrise behandeln. Hofmannsthal erlebte in dieser Zeit längere unproduktive Phasen, die mit einem Vertauensschwund in das Wort als künstlerisches Medium und Ausdruck von Realität zusammenhingen. Im OEuvre des Autors markiert der Brief eine Zäsur zwischen den Jugendwerken und denen des reiferen Alters, die zugleich auf einen Wechsel der Gattungen verweist. Hofmannsthal verkündet hier, so schreibt Gotthart Wunberg, "seine große Absage an die lyrische Produktion seiner frühen Jahre, an die Werke, die ihn berühmt gemacht haben." Es ist der Abschied von der Magie des lyrischen Worts und die Hinwendung zu einer Prosa, die er ebenfalls als als 'reine Form' zu binden erhofft. Das Prinzip der Form - wie immer wieder bemerkt worden ist, wurde dieser Essay über das Verstummen in einer meisterhaften Prosa artikuliert - überbrückt nicht nur den Abstand zwischen Lyrik und Prosa, sondern auch den zwischen den biographischen Briefen und dem literarischen Brief.
Die rhetorische Bravour des "Briefes" – eine Bravour, die bekanntlich ihr eigenes Dementi voraussetzt und damit zur glanzvollsten Präteritio ihrer selbst gerät –, diese Bravour also findet sich exemplarisch in einem seiner Sätze:
Es begegnete mir, daß ich meiner vierjährigen Tochter Katharina Pompilia eine kindische Lüge, deren sie sich schuldig gemacht hatte, verweisen und sie auf die Notwendigkeit, immer wahr zu sein, hinführen wollte, und dabei die mir im Munde zuströmenden Begriffe plötzlich eine solche schillernde Färbung annahmen und so ineinander überflossen, daß ich den Satz, so gut es ging, zu Ende haspelnd, so wie wenn mir unwohl geworden wäre und auch tatsächlich bleich im Gesicht und mit einem heftigen Druck auf der Stirn, das Kind allein ließ, die Tür hinter mir zuschlug und mich erst zu Pferde, auf der einsamen Hutweide einen guten Galopp nehmend, wieder einigermaßen herstellte.
Der als Sprachkrise inszeniertea "mystische Weg", den Hofmannsthal rückblickend als Weg des Lord Chandos beschworen hat, bezeichnet weniger den "Anstand des Schweigens", als vielmehr eine literarisch äußerst produktive und semiotisch folgenreiche Poetologie des inneren Sehens, der endogenen Bilder des Bewußtseins.
"Es ist mir völlig die Fähigkeit abhanden gekommen, über irgend etwas zusammenhängend zu denken oder zu sprechen." Wohl kein Leser hat es versäumt, den Widerspruch zu bemerken, daß der berühmte Satz einer Erfahrung gilt, die Lord Chandos macht oder erleidet, nicht aber den Sätzen, in denen er diese Erfahrung formuliert und mitteilt. Um die Paradoxie zu erklären, möchte ich vorschlagen, den Chandos-Brief als poetologisches Experiment zu lesen. Hofmannsthal zerstört - oder setzt für die Dauer eines Textes außer Kraft - , was man die Spielregel der kreativen Reproduktion nennen könnte. Seit der Goethezeit und das 19. Jahrhundert hindurch stellte sie eine unantastbare Einheit dar: als jene persönliche Instanz, die Welt vernimmt oder wahrnimmt und dem Vernommenen oder Wahrgenommenen wiederum Ausdruck gibt. Diese Instanz entfällt; nicht nur die Sprache, auch Lord Chandos selber zerfällt in Teile, in eine erlebende und in eine erzählende Person. Eine Anregung hierfür, das wäre mein zweiter Vorschlag, konnte Hofmannsthal den Schriften Ernst Machs entnehmen, zumal dem ebenso berühmt gewordenen Satz "Das Ich ist unrettbar". Was geschieht, wenn man diesen Satz auf die Autorschaft anwendet? Schließlich und drittens wäre zu bedenken, ob nicht die Erlebnisse eines Autors, dem sich der Ausdruck versagt, gleichwohl Manifestationen von Kreativität sein könnten. In diesem Sinne möchte ich besonders die "guten Augenblicke" (S. 50) des Lord Chandos erörtern. ...

es freut mich, daß Sie so zahlreich erschienen sind zu dieser Versammlung, und daß ein jeder von ihnen seinen Zehnt abgibt von seinem Herz, womit wir die Kosten des Mitleids mit Herrn Chandos tragen wollen. Von den Reden meiner zahlreichen Vorgänger kam viel Mitgefühl für sein Schicksal, daß der Tag davon nun bereits bis an den Rand gefüllt ist, und nur ein Seufzer noch fehlte, und schon schwappte etwas davon über und tropfte in mein Gemüt, und dann besitze auch ich ein paar Tränen. ... mit groszer Anteilnahme habe ich Ihre Klagen aufgenommen und möchte zu Ihrer Tröstung bekennen, dasz es mir vor einiger Zeit ähnlich ergangen ist: ich hatte in dieser meiner Krise aller Welt vorgemacht, ich sei an der Arbeit, muszte mir jedoch eingestehen, dasz ich in Wahrheit völlig ausgebrannt war, ja, dasz die Vorstellung mich verfolgte, ich hätte noch nie auch nur I Wort aufgeschrieben, ich würde nie wieder Pläne oder Ahnungen von Gedanken empfangen können, am allerwenigsten jene so wundertätigen Verbalträume, die mich jederzeit mitten hinein in die Produktion katapultiert hatten. Aber dann versuchte ich mich selbst aufzufangen, indem ich den Gründen meines Versagens nachzuforschen begann, und ich entdeckte, warum es mir so elend erging. …
"Ich bin Hofmannsthal das erstemal Anfang 1902 begegnet. In seinem Rodauner Haus, wohin mich Hermann von Keyserling brachte. Jahre also, nachdem ich schon der Bezauberung durch seine Gedichte und einige Aufsätze unterlegen war. Ich sage Gedichte, denn auch seine kleinen Dramen nahm ich für solche. Seinen Aufsätzen hatte ich unter anderem den ersten Hinweis auf englische Dichter wie A. C. Swinburne und den Ästheten und Essayisten Walter Pater verdankt, auch auf andere, von denen später mein erstes Buch gehandelt hat, das im übrigen auch die Brücke war, die mich, da mir jede andere Verbindung mit ihm gefehlt hat, zu Hofmannsthal hinüberführen sollte."
Mit diesen Worten gedenkt Kassner nach mehr als einem Menschenalter, im Jahre 1946, der Anfänge dieser Freundschaft und der Rolle, die sein Erstlingswerk dabei gespielt hat. In der Tat beruft er sich in dem zu Beginn des neuen Jahrhunderts erschienen Band "Die Mystik, die Künstler und das Leben", für jeden Kundigen deutlich, in der allgemeinen geistigen Haltung vielfach auf Hofmannsthal und zitiert, offen oder verdeckt, an mehreren Stellen dessen Werk.
Bericht zur Interdisziplinären und Internationalen Tagung, 7.-10. Oktober 2002, Westfälische Wilhelms-Universität Münster
Die Geisteswissenschaften waren gewissermaßen immer schon eine gespenstische Angelegenheit. Versuchte man im Zeitalter der Hermeneutik, den reinen Geist zu beschwören, ohne ungebetene Gäste aus dem Reich der Toten einzuladen, so begrüßt man jene Gespenster heute herzlicher. Schließlich scheinen die Gespenster die Figuren des Zwischenraums, des Anderen, der Wiederholung sprachlich verfügbar zu machen und auf diese Weise zu Metaphern der Postmoderne zu werden. Um über die Diskursfigur dieser 'Un-Wesen' nachzudenken, luden Dr. Moritz Baßler (Rostock), PD Dr. Bettina Gruber (Bochum) und Prof. Dr. Martina Wagner-Egelhaaf (Münster) nach Münster ins Alexander-von-Humboldt-Haus ein zu einer unheimlichen Begegnung mit den 'Gespenstern' in ihren vielfältigen 'Erscheinungen - Medien - Theorien'.
