Background & Aims: Spontaneous portosystemic shunts (SPSS) frequently develop in liver cirrhosis. Recent data suggested that the presence of a single large SPSS is associated with complications, especially overt hepatic encephalopathy (oHE). However, the presence of >1 SPSS is common. This study evaluates the impact of total cross-sectional SPSS area (TSA) on outcomes in patients with liver cirrhosis.
Methods: In this retrospective international multicentric study, CT scans of 908 cirrhotic patients with SPSS were evaluated for TSA. Clinical and laboratory data were recorded. Each detected SPSS radius was measured and TSA calculated. One-year survival was the primary endpoint and acute decompensation (oHE, variceal bleeding, ascites) was the secondary endpoint.
Results: A total of 301 patients (169 male) were included in the training cohort. Thirty percent of all patients presented with >1 SPSS. A TSA cut-off of 83 mm2 was used to classify patients with small or large TSA (S-/L-TSA). Patients with L-TSA presented with higher model for end-stage liver disease score (11 vs. 14) and more commonly had a history of oHE (12% vs. 21%, p <0.05). During follow-up, patients with L-TSA experienced more oHE episodes (33% vs. 47%, p <0.05) and had lower 1-year survival than those with S-TSA (84% vs. 69%, p <0.001). Multivariate analysis identified L-TSA (hazard ratio 1.66; 95% CI 1.02–2.70, p <0.05) as an independent predictor of mortality. An independent multicentric validation cohort of 607 patients confirmed that patients with L-TSA had lower 1-year survival (77% vs. 64%, p <0.001) and more oHE development (35% vs. 49%, p <0.001) than those with S-TSA.
Conclusion: This study suggests that TSA >83 mm2 increases the risk for oHE and mortality in patients with cirrhosis. Our results support the clinical use of TSA/SPSS for risk stratification and decision-making in the management of patients with cirrhosis.
Lay summary: The prevalence of spontaneous portosystemic shunts (SPSS) is higher in patients with more advanced chronic liver disease. The presence of more than 1 SPSS is common in advanced chronic liver disease and is associated with the development of hepatic encephalopathy. This study shows that total cross-sectional SPSS area (rather than diameter of the single largest SPSS) predicts survival in patients with advanced chronic liver disease. Our results support the clinical use of total cross-sectional SPSS area for risk stratification and decision-making in the management of SPSS.
Long non-coding RNAs (lncRNAs) contribute to cardiac (patho)physiology. Aging is the major risk factor for cardiovascular disease with cardiomyocyte apoptosis as one underlying cause. Here, we report the identification of the aging-regulated lncRNA Sarrah (ENSMUST00000140003) that is anti-apoptotic in cardiomyocytes. Importantly, loss of SARRAH (OXCT1-AS1) in human engineered heart tissue results in impaired contractile force development. SARRAH directly binds to the promoters of genes downregulated after SARRAH silencing via RNA-DNA triple helix formation and cardiomyocytes lacking the triple helix forming domain of Sarrah show an increase in apoptosis. One of the direct SARRAH targets is NRF2, and restoration of NRF2 levels after SARRAH silencing partially rescues the reduction in cell viability. Overexpression of Sarrah in mice shows better recovery of cardiac contractile function after AMI compared to control mice. In summary, we identified the anti-apoptotic evolutionary conserved lncRNA Sarrah, which is downregulated by aging, as a regulator of cardiomyocyte survival.
Fruiting body-forming members of the Basidiomycota maintain their ecological fitness against various antagonists like ascomycetous mycoparasites. To achieve that, they produce myriads of bioactive compounds, some of which are now being used as agrochemicals or pharmaceutical lead structures. Here, we screened ethyl acetate crude extracts from cultures of thirty-five mushroom species for antifungal bioactivity, for their effect on the ascomycete Saccharomyces cerevisiae and the basidiomycete Ustilago maydis. One extract that inhibited the growth of S. cerevisiae much stronger than that of U. maydis was further analyzed. For bioactive compound identification, we performed bioactivity-guided HPLC/MS fractionation. Fractions showing inhibition against S. cerevisiae but reduced activity against U. maydis were further analyzed. NMR-based structure elucidation from one such fraction revealed the polyyne we named feldin, which displays prominent antifungal bioactivity. Future studies with additional mushroom-derived eukaryotic toxic compounds or antifungals will show whether U. maydis could be used as a suitable host to shortcut an otherwise laborious production of such mushroom compounds, as could recently be shown for heterologous sesquiterpene production in U. maydis.
Space optimizations in deterministic and concurrent call-by-need functional programming languages

In this thesis the space consumption and runtime of lazy-evaluating functional programming languages are analyzed.
The typed and extended lambda-calculi LRP and CHF* as core languages for Haskell and Concurrent Haskell are used. For each LRP and CHF* compatible abstract machines are introduced.
Too lower the distortion of space measurement a classical implementable garbage collector is applied after each LRP reduction step. Die size of expressions and the space measure spmax as maximal size of all garbage-free expressions during an LRP-evaluation, are defined.
Program-Transformations are considered as code-to-code transformations. The notions Space Improvement and Space Equivalence as properties of transformations are defined. A Space Improvement does neither change the semantics nor it increases the needed space consumption, for a space equivalence the space consumption is required to remain the same. Several transformations are shown as Space Improvements and Equivalences.
An abstract machine for space measurements is introduced. An implementation of this machine is used for more complex space- and runtime-analyses.
Total Garbage Collection replaces subexpressions by a non-terminating constant with size zero, if the overall termination is not affected. Thereby the notion of improvement is more independent from the used garbage collector.
Analogous to Space Improvements and Equivalences the notions Total Space Improvement and Total Space Equivalence are defined, which use Total Garbage Collection during the space measurement. Several Total Space Improvements and Equivalences are shown.
Space measures for CHF* are defined, that are compatible to the space measure of LRP. An algorithm with sort-complexity is developed, that calculates the required space of independent processes that all start and end together. If a constant amount of synchronization restrictions is added and a constant number of processors is used, the runtime is polynomial, if arbitrary synchronizations are used, then the problem is NP-complete.
Abstract machines for space- and time-analyses in CHF* are developed and implementations of these are used for space and runtime analyses.
We study whether and how time preferences change over the life cycle, exploiting representative long-term panel data. We estimate the age patterns of discount rates from age 25 to 80. In order to identify age effects, we have to disentangle them from cohort and period factors. We address this identification problem by estimating individual fixed effects models, where we substitute period effects with determinants of time preferences that depend on calendar years. We find that discount rates decrease with age and the decline is remarkably linear over the life cycle.
Hintergrund und Ziel der Studie: Steigende Erwartungen der Kataraktpatienten führen zu stetigen Weiterentwicklungen in der Linsenchirurgie. Der Wunsch nach einem perfekten Sehvermögen und nach Brillenfreiheit ist ein häufiges Anliegen insbesondere jüngerer Patienten. Die Einführung von Multifokallinsen hat die Therapie der Katarakt revolutioniert. Mit ihnen kann der Patient sowohl in der Nähe und Ferne als auch im Intermediärbereich scharf sehen. Für die präoperative Berechnung der Linsenstärke sind verschiedene Formeln verfügbar. Bis jetzt wurde keine Studie publiziert, die die Präzision dieser Formeln für die Kalkulation der Stärke von Tri- oder Quadrifokallinsen untersucht hat. Das Ziel dieser Studie war die Evaluation neun moderner Formeln für die Berechnung der Linsenstärke der quadrifokalen AcrySof IQ PanOptix TFNT00.
Methoden:
Die vorliegende Studie ist eine retrospektive konsekutive Fallserie, die an der Augenklinik der Johann Wolfgang Goethe-Universität Frankfurt am Main durchgeführt wurde. Es wurden alle Patienten eingeschlossen, die sich einer Femtosekundenlaser-assistierten Operation mit Implantation der quadrifokalen Intraokularlinse unterzogen. Mit dem IOLMaster 500 wurden die präoperativen Biometrie- und Keratometriemessungen durchgeführt. Das Scheimpflug-System Pentacam wurde zur Bestimmung der zentralen Hornhautdicke genutzt. Drei Monate nach der Operation fand eine subjektive Refraktionsbestimmung statt.
Die folgenden Formeln wurden untersucht: Holladay 1, Sanders-Retzlaff-Kraff/ theoretical (SRK/T), Hoffer Q, T2, Holladay 2, Haigis, Barrett Universal II, Olsen und Hill-radiale Basisfunktion (RBF). Aus den präoperativ gemessenen Parametern und der implantierten Linsenstärke wurde für jedes Auge die postoperative Refraktion berechnet. Der Vorhersagefehler (PE) ist definiert als die Differenz zwischen dem tatsächlich erreichten postoperativen sphärischen Äquivalent und dem vorhergesagten postoperativen sphärischen Äquivalent. Zur Reduktion systematischer Fehler und somit des mittleren PE wurden die Linsenkonstanten optimiert. Primäre Endpunkte waren die Unterschiede der mittleren absoluten Vorhersagefehler (MAE) zwischen den Formeln. Die Standardabweichung sowie mediane und maximale absolute PE wurden ebenso untersucht wie die Prozent-zahlen der Augen, deren PE innerhalb von ± 0,25 Dioptrien (dpt), ± 0,5 dpt, ± 1,0 dpt und ± 2,0 dpt lagen.
Ergebnisse: 75 Augen von 38 Patienten wurden in die vorliegende Studie eingeschlossen. Es gab signifikante Unterschiede zwischen den Formeln bezüglich des MAE (p Wert < 0,001). Die Rangfolge der Formeln entsprechend ihrer MAE lautet wie folgt: Barrett Universal II (0,294 dpt), Hill-RBF (0,332 dpt), Olsen (0,339 dpt), T2 (0,351 dpt), Holladay 1 (0,381 dpt), Haigis (0,382 dpt), SRK/T (0,393 dpt), Holladay 2 (0,399 dpt) und Hoffer Q (0,410 dpt). Den niedrigsten maximalen absoluten PE erreichte die Barrett Universal II. Bei der Verwendung der Haigis, Barrett Universal II, Olsen und Hill-RBF hatten 80% der Augen einen PE innerhalb von ± 0,5 dpt. In der Subgruppe der kurzen Augen wurde der niedrigste PE mit der Hill-RBF erreicht, in der Subgruppe der langen Augen mit der Barrett Universal II und mit der Olsen. Die Unterschiede zwischen den Formeln in den Subgruppenanalysen waren jedoch nicht signifikant.
Schlussfolgerung:Die exaktesten Vorhersagen der postoperativen Refraktion konnten mit der Barrett Universal II erzielt werden. Diese Formel sollte daher künftig für die Berechnung der Linsenstärke der quadrifokalen PanOptix genutzt werden. Die T2, Olsen und Hill-RBF führten ebenfalls zu niedrigen PE. Weitere Studien sollten durchgeführt werden, um insbesondere die neuen Formeln im Zusammenhang mit verschiedenen Multifokallinsen zu untersuchen. Um die Vorhersagegenauigkeit der Formeln in Augen mit extremen Achsenlängen zu bewerten, sind Studien mit größeren Fallzahlen nötig.
The branchial parasitic isopod Pleurocryptella altalis sp. nov. (Bopyridae: Pseudioninae) is described from the squat lobster host Munidopsis petalorhyncha Baba, 2005. The new species is morphologically similar to Pleurocryptella formosa Bonnier, 1900 and P. wolffi Bourdon, 1972b but can be distinguished based on male characters (differences in head, pleon and uropods) and female characters (differences in barbula, pleopods and pleotelson). The parasite specimens (a female and male pair) were collected with the squat lobster host at a depth of 5060–5130 m from the Kuril-Kamchatka Trench, representing the deepest record for any of the 850+ described bopyrid isopod species and for any record of an infested host. Dichotomous identification keys to females and males of Pleurocryptella species and subspecies are provided.
CD4+ T cell lymphopenia predicts mortality from Pneumocystis pneumonia in kidney transplant patients

Background: Pneumocystis jirovecii pneumonia (PcP) remains a life-threatening opportunistic infection after solid organ transplantation, even in the era of Pneumocystis prophylaxis. The association between risk of developing PcP and low CD4+ T cell counts has been well established. However, it is unknown whether lymphopenia in the context of post-renal transplant PcP increases the risk of mortality. Methods: We carried out a retrospective analysis of a cohort of kidney transplant patients with PcP (n = 49) to determine the risk factors for mortality associated with PcP. We correlated clinical and demographic data with the outcome of the disease. For CD4+ T cell counts, we used the Wilcoxon rank sum test for in-hospital mortality and a Cox proportional-hazards regression model for 60-day mortality. Results: In univariate analyses, high CRP, high neutrophils, CD4+ T cell lymphopenia, mechanical ventilation, and high acute kidney injury network stage were associated with in-hospital mortality following presentation with PcP. In a receiver-operator characteristic (ROC) analysis, an optimum cutoff of ≤200 CD4+ T cells/µL predicted in-hospital mortality, CD4+ T cell lymphopenia remained a risk factor in a Cox regression model. Conclusions: Low CD4+ T cell count in kidney transplant recipients is a biomarker for disease severity and a risk factor for in-hospital mortality following presentation with PcP.
The miRNA biogenesis is tightly regulated to avoid dysfunction and consequent disease development. Here, we describe modulation of miRNA processing as a novel noncanonical function of the 5-lipoxygenase (5-LO) enzyme in monocytic cells. In differentiated Mono Mac 6 (MM6) cells, we found an in situ interaction of 5-LO with Dicer, a key enzyme in miRNA biogenesis. RNA sequencing of small noncoding RNAs revealed a functional impact, knockout of 5-LO altered the expression profile of several miRNAs. Effects of 5-LO could be observed at two levels. qPCR analyses thus indicated that (a) 5-LO promotes the transcription of the evolutionarily conserved miR-99b/let-7e/miR-125a cluster and (b) the 5-LO-Dicer interaction downregulates the processing of pre-let-7e, resulting in an increase in miR-125a and miR-99b levels by 5-LO without concomitant changes in let-7e levels in differentiated MM6 cells. Our observations suggest that 5-LO regulates the miRNA profile by modulating the Dicer-mediated processing of distinct pre-miRNAs. 5-LO inhibits the formation of let-7e which is a well-known inducer of cell differentiation, but promotes the generation of miR-99b and miR-125a known to induce cell proliferation and the maintenance of leukemic stem cell functions.
An integrative redescription of Hypsibius pallidoides Pilato, Kiosya, Lisi, Inshina & Biserov, 2011 was undertaken following a reexamination of the type material and new material using highquality light microscopy, scanning electron microscopy and methods of molecular taxonomy. Detailed morphological investigations revealed a unique complex of characters that precluded the attribution of this species to the genus Hypsibius Ehrenberg, 1848. Furthermore, phylogenetic analyses indicated the affi nity of this species within the subfamily Pilatobiinae (Hypsibiidae). Notahypsibius gen. nov. is erected for H. pallidoides and two putatively related species: H. scaber Maucci, 1987 and Ramazzottius arcticus (Murray, 1907). An emended diagnosis for the genus Pilatobius is given, while the subfamily Pilatobiinae lacks a cohesive morphological diagnosis despite representing, at the same time, a wellsupported molecular clade. Obvious controversy between the results of the morphological and molecular analyses of the phylogeny of Hypsibioidea is discussed. The distribution of morphological characters such as the claw type, organization of the bucco-pharyngeal apparatus, and egg shell sculpture type within Eutardigrada is analyzed and their phylogenetic signifi cance discussed.
In this paper we describe Macrobiotus engbergi sp. nov. and Tenuibiotus zandrae sp. nov. from Greenland. Our study has involved both classical taxonomic methods, which include morphological and morphometric analyses conducted with the use of light and scanning electron microscopy, and genetic analysis based on four molecular markers (three nuclear: 18S rRNA, 28S rRNA, ITS-2, and one mitochondrial: COI). Moreover, we re-examined the type series of Tenuibiotus voronkovi (Tumanov, 2007) as well as the original sample where the species was found and we provide new morphological data from light and scanning electron microscopy which enabled us to amend its description. Finally, we also analysed slides with animals and egg of two populations from Nordaustlandet and Edgeøya (archipelago of Svalbard, Norway) designated as T. voronkovi within its recent redescription. The results and comparisons presented in our study question the validity of this designation.
The widespread Ophioderma hendleri sp. nov., from the Eastern Tropical Pacific (Mexico to Colombia) is distinguished from its congeners by having radial shields covered by granules, naked adoral shields, up to 11 arm spines, and by its brown and beige coloration. Ophioderma hendleri sp. nov. belongs to the group of species with naked adoral shields (i.e., O. pentacanthum H.L. Clark, 1917, O. variegatum Lütken, 1856), and it has frequently been misidentified as O. panamense Lütken, 1859 or O. variegatum. Therefore, the main aim of the present work was to describe Ophioderma hendleri sp. nov. and differentiate it from its congeners. The original description of O. panamense was incomplete; thus, we provide a redescription. Due to the confusion in previous designations of its type material, we designate a lectotype and paralectotype of O. variegatum. Finally, we expand the distribution range of O. pentacanthum to Cocos Island, Costa Rica. With this work, the total number of valid species of Ophioderma Müller & Troschel, 1840 in the world increases to 33 and in the Eastern Pacific to nine species.
A new species of the genus Mesobiotus is described from the Republic of South Africa using a traditional morphological approach (light and scanning electron microscopy) combined with molecular analysis (18S rRNA, 28S rRNA, ITS-2 and COI markers). Mesobiotus anastasiae sp. nov. differs from all known Mesobiotus species by having a unique combination of characters of the adult animals and the eggs. Adults of the new species have an oral cavity armature without elongate teeth in the second band, while the processes of the egg chorion have a basal collar and distinct rows of large pores. An updated key to the species of the genus Mesobiotus, including 66 of 70 currently described species, is given. An aquatic mite species from the Lobohalacarus weberi complex (freshwater Halacaridae) co-occurs with M. anastasiae sp. nov., suggesting that the newly described tardigrade inhabits constantly wet moss cushion habitats.
Pathogenic variants in PRRT2, encoding the proline-rich transmembrane protein 2, have been associated with an evolving spectrum of paroxysmal neurologic disorders. Based on a cohort of children with PRRT2-related infantile epilepsy, this study aimed at delineating the broad clinical spectrum of PRRT2-associated phenotypes in these children and their relatives. Only a few recent larger cohort studies are on record and findings from single reports were not confirmed so far. We collected detailed genetic and phenotypic data of 40 previously unreported patients from 36 families. All patients had benign infantile epilepsy and harbored pathogenic variants in PRRT2 (core cohort). Clinical data of 62 family members were included, comprising a cohort of 102 individuals (extended cohort) with PRRT2-associated neurological disease. Additional phenotypes in the cohort of patients with benign sporadic and familial infantile epilepsy consist of movement disorders with paroxysmal kinesigenic dyskinesia in six patients, infantile-onset movement disorders in 2 of 40 individuals, and episodic ataxia after mild head trauma in one girl with bi-allelic variants in PRRT2. The same girl displayed a focal cortical dysplasia upon brain imaging. Familial hemiplegic migraine and migraine with aura were reported in nine families. A single individual developed epilepsy with continuous spikes and waves during sleep. In addition to known variants, we report the novel variant c.843G>T, p.(Trp281Cys) that co-segregated with benign infantile epilepsy and migraine in one family. Our study highlights the variability of clinical presentations of patients harboring pathogenic PRRT2 variants and expands the associated phenotypic spectrum.
The specific temporal evolution of bacterial and phage population sizes, in particular bacterial depletion and the emergence of a resistant bacterial population, can be seen as a kinetic fingerprint that depends on the manifold interactions of the specific phage–host pair during the course of infection. We have elaborated such a kinetic fingerprint for a human urinary tract Klebsiella pneumoniae isolate and its phage vB_KpnP_Lessing by a modeling approach based on data from in vitro co-culture. We found a faster depletion of the initially sensitive bacterial population than expected from simple mass action kinetics. A possible explanation for the rapid decline of the bacterial population is a synergistic interaction of phages which can be a favorable feature for phage therapies. In addition to this interaction characteristic, analysis of the kinetic fingerprint of this bacteria and phage combination revealed several relevant aspects of their population dynamics: A reduction of the bacterial concentration can be achieved only at high multiplicity of infection whereas bacterial extinction is hardly accomplished. Furthermore the binding affinity of the phage to bacteria is identified as one of the most crucial parameters for the reduction of the bacterial population size. Thus, kinetic fingerprinting can be used to infer phage–host interactions and to explore emergent dynamics which facilitates a rational design of phage therapies.
This Ph.D. thesis demonstrates i) the highly precise performance of refined and new analytical setups for clumped isotope analysis (Δ47 and Δ48) and ii) the applicability of clumped isotope analyses to biogenic and abiogenic carbonated apatite (Δ47) and abiogenic carbonates (Δ47 and Δ48) for research related to paleothermophysiology and paleoclimatology, whereas the overall analytical precision has been increased.
A comprehensive Δ47 dataset with 122 replicate analyses is provided from which the temperature dependence of Δ47 for (bio)apatite (Δ47-1/T2) is calculated between 1 °C and 80 °C. The temperature dependence of oxygen isotope equilibrium fractionation between carbonated synthetic apatite and water (1,000ln(αCHAP-H2O)) is experimentally determined. When applied to tooth enameloid from a modern Greenland shark (Somniosus microcephalus), a Late Miocene megatooth shark (Carcharodon megalodon), and an Upper Cretaceous Tyrannosaurus rex, reconstructed Δ47-based temperatures and δ18OH2O are in line with previously published data.
An analytical setup for highly precise clumped isotope analysis is described that allows for the simultaneous measurement of ∆47 and ∆48 in CO2 with external reproducibilities close to the respective shot-noise limits. The analyte gases originate from pure carbonates that were digested in hypersaturated orthophosphoric acid and purified using a fully automated device. Δ47 data sets with 117 replicate analyses in total on 22 pedogenic carbonate nodules from two Spanish Middle Miocene sections reveal the continental Southern European thermal structure during the end of the Middle Miocene Climatic Optimum (MCO) and the complete Middle Miocene Climatic Transition (MMCT; from 15.33 to 12.98 Ma).
Astrozyten erfüllen verschiedene Funktionen im Zentralnervensystem, welche sich in die Bereiche Entwicklung, Durchblutung, Metabolismus, Strukturerhalt und Gliotransmission unterteilen lassen. Astrozyten sind an der synaptischen Informationsverarbeitung beteiligt und wirken an zahlreichen höheren Hirnfunktionen mit. Durch Regulation der synaptischen Transmission und Plastizität sind Astrozyten am Lernverhalten und Erinnerungsvermögen, sowie an der Verhaltensmodulation und Verarbeitung emotionaler Reize involviert. Im Zuge dieser zahlreichen Funktionen können Astrozyten auf externe Stimuli mit der gezielten Freisetzung von Gliotransmittern reagieren.
In kultivierten Astrozyten konnte Keil143 das TGN, bestehend aus Zisternen und Vesikeln, darstellen und mit anti-Rab6 identifizieren. Rab6 mit seinen Subtypen A und B gehört der Superfamilie der monomeren Ras-GTPasen an, die den intrazellulären Membran- und Vesikelverkehr regulieren. Rab6 spielt in HeLa-Zellen beim Transport vesikulärer Organellen vom TGN zur Zellmembran eine wichtige Rolle. Assoziationsanalysen von Rab6A mit vesikulären Glutamattransportern, Serinracemase und Markern der regulierten Exozytose in kultivierten Astrozyten143 deuten darauf hin, dass dieses Rab6A-Organellsystem die ultrastrukturelle Grundlage für die Freisetzung von Gliotransmittern wie D-Serin und Glutamat bildet.
Zur Untersuchung, ob Rab6A tatsächlich ein System der Glia-Neuron-Kommunikation im Gehirn darstellt, war es zunächst unabdingbar das Vorkommen von Rab6A in situ zu untersuchen. Die durchgeführten immunzytochemischen Färbungen an Hirnschnitten der Maus zeigen das gleichmäßige und ubiquitäre Vorkommen von Rab6A in allen untersuchten Hirnregionen. Durch verblindet durchgeführte Kolokalisationsanalysen von Rab6A mit den etablierten astrozytären Markern Glutaminsynthetase (GS), Glial fibrillary acidic protein (GFAP), Aldh1L1 und Sox9 konnte eine Lokalisation von Rab6A in allen Astrozyten gezeigt werden. Weitere Analysen schließen die Lokalisation von Rab6A in Mikroglia (Iba1), NG2-Zellen (NG2) und Oligodendrozyten (CNPase) aus. Die Astrozyten unterscheiden sich in Größe und subzellulärem Verteilungsmuster der Rab6A+ Strukturen, wonach eine Kategorisierung in vier Typen vorgenommen wurde. Anhand der Einteilung kann vermutet werden, dass größere Rab6A+ TGN-Zisternen bis weit in die Zellperipherie transportiert werden und kleine Rab6A+ Vesikel erst dort ausknospen und der Exozytose zugeführt werden. Zur Frage der möglichen astrozytären Subpopulationen konnte gezeigt werden, dass alle untersuchten Astrozyten GS+, Aldh1L1+, Sox9+ und Rab6A+ sind, jedoch nicht GFAP+.
Um die prinzipielle Übertragbarkeit der gewonnenen Befunde auf den Menschen zu überprüfen, wurde reseziertes Cortex-Gewebe von drei Patienten mit unterschiedlicher pathologischer Genese untersucht. Rab6A ist im massiven Ausmaß in humanen Astrozyten lokalisiert, was nahelegt, dass die zuvor an der Maus gewonnenen Ergebnisse auf den Menschen übertragbar sind.
Die mögliche funktionelle Bedeutung von astrozytärem Rab6A im Gehirn wurde an HFS-Schnitten untersucht. Die Untersuchung zeigt einen signifikanten Anstieg der Rab6A+ Intensität in der gesamten Molekularschicht der Fascia dentata der stimulierten im Vergleich zur unstimulierten Seite. Da die HFS ein etabliertes LTP-Modell darstellt, könnte es infolge dieser zu einer strukturellen, intrazellulären Veränderung der Astrozyten mit erhöhter Freisetzung von D-Serin oder Glutamat aus Rab6A+ Vesikeln kommen, was das Lernverhalten beeinflussen könnte. Die dargestellten Ergebnisse legen eine Auswirkung der HFS auf Rab6A nahe.
Zur Bestätigung der immunzytochemischen Untersuchungen wurde die mRNA-Expression von Rab6A in Astrozyten bereits publizierter Transkriptomanalysen untersucht. Die in den Publikationen verwendeten Genom-Chips treffen allenfalls indirekt eine Aussage zu Rab6A, da Rab6 allgemein und nur Rab6B spezifisch untersucht wurde, jedoch keine spezifische Rab6A Sonde erwähnt wird.
Zusammenfassend kann Rab6A als spezifisches und selektiv in Astrozyten vorkommendes Protein dargestellt und als neuer astrozytärer Marker etabliert werden, der auch Astrozyten des humanen Gewebes markiert. Durch die gewonnenen Befunde kann in nachfolgenden Studien die mögliche Bedeutung von Rab6A in neuropathologischen und neurophysiologischen Prozessen untersucht werden.
