Präsenzdiagnostik in der Laboratoriumsmedizin ist die zuverlässige Erbringung von Laborbefunden in einer dem Krankheitszustand des Patienten angemessenen Zeit, unter wirtschaftlichen Bedingungen.
Ergänzend zu den konventionellen Analysenverfahren, die auf der Anwendung von Reagenzien in gelöster Form und photometrischer Messung beruhen, wurden seit Anfang der 80er Jahre alternative Systeme entwickelt.
Die alternativen Systeme bestehen aus Reagenzträgern sowie Meß- und Auswertegeräten. Die Reagenzträger enthalten die zur Analysenreaktion erforderlichen Reagenzien, entweder in trockener Form oder in Kammern von Reaktionskassetten. Die Meß- und Auswertegeräte sind vollmechanisiert und haben den Vorteil der einfachen Bedienbarkeit.
Aufgrund apparativer Einfachheit kehrt möglicherweise der Teil der Labordiagnostik, der aufgrund der Einführung der Qualitätskontrolle und steigender Kosten in Laborgemeinschaften verlagert wurde, zurück zum niedergelassenen Arzt. Dadurch wäre in diesem ärztlichen Bereich ähnlich den Krankenhäusern eine Präsenzdiagnostik möglich.
Zur Zeit ist die Qualität der alternativen Analysensysteme noch unzureichend gesichert, da die seit über einem Jahrzehnt bewährten Qualitätssicherungsmaßnahmen der konventionellen Techniken nicht anwendbar sind. Auch liegen teilweise nur unzureichende Kenntnisse über Störmöglichkeiten und Referenzbereiche für 25°C-Methoden vor.
Voraussetzung zur Diagnostik und Verlaufsbeurteilung des Krebses durch die Bestimmung von Tumormarkern ist eine gute Abstimmung zwischen dem klinisch bzw. praktisch tätigen Arzt und dem Labor. Für die Auswahl der Testkits und die Festlegung des Grenzwertes normal/pathologisch muß das Labor vom anfordernden Arzt wissen, ob die Tumormarkerbestimmung eingesetzt wird:
a) zur Verlaufsbeurteilung eines bekannten Krebses, also im Sinne der Longitudinalbeurteilung
b) im Rahmen der Tumordiagnostik, d. h. zur Transversalbeurteilung.
Zum effektiven Einsatz der Tumormarkerbestimmung für die Verlaufsbeurteilung sollte das Labor sicherstellen:
a) Verwendung eines Testkits hoher Nachweisempfindlichkeit
b) Keinen Wechsel des Testkits bei nicht standardisierten Markern vorzunehmen, ohne den anforderndenArzt in Kenntnis zu setzen
c) Patientenbezogenen Kumulativreport liefern, der die relativen Veränderungen zum Vorwert erkennen läßt
d) Aufbewahrung der jeweilig letzten Analysenprobe. Nochmalige Bestimmung mit der neuen Probe in dergleichen Analysenserie, falls der Analysenwert der neuen Probe ein „Ausreißer"zu sein scheint.
Im Rahmen der Tumordiagnostik kann bei symptomatischen Patienten die Bestimmung von AFP beim Leberzellkarzinom und von AFP und HCG bei Keimzelltumoren empfohlen werden. Andere Tumormarker sollten nur zur Diagnostik eingesetzt werden, wenn im Patientenkollektiv des anfordernden Arztes eine hohe Krankheitsprävalenz für den entsprechenden Krebs vorliegt In Kenntnis der Krankheitsprävalenz sollten Labor und anfordernder Arzt unter Auswahl einer optimalen Spezifität (noch vertretbare Zahl falsch positiver Ergebnisse), den Grenzwert normal/pathologisch für die Transversalbeurteilung festlegen.
A role of the Qв binding protein in the mechanism of cyanobacterial adaptation to light intensity?

Growth of the unicellular blue-green alga Anacystis nidulans in media containing sublethal concentrations of DCMU-type inhibitors of photosynthetic electron transport in strong white light gave rise to shade type appearance in this organism, as characterized by an increased ratio of phycocyanin to chlorophyll and reduced ratios, both, of carotenoids to chlorophyll and of total chlorophyll to P700. Shade type in Anacystis was caused neither by phenolic inhibitors tested nor by those known to bind to the cytochrome b6/f-complex. Surprisingly enough, the molar ratio of phycocyanin to chlorophyll in artificially shade adapted Anacystis1 grown in strong white light in the presence of 10-6 м atrazine, was found to increase with temperature for a given light intensity and with light intensity for a given temperature.
Mutants of Anaeystis with a reduced binding capacity for DCMU-type herbicides due to an amino acid exchange in the 32 kDa Qв-binding polypeptide, also called D-1 protein, were ob- served to show shade type appearance in strong light, to respond very little to changes in light intensity and to show a reduced capability to further change their appearance to shade type by binding of competitors of Ob to the 32 kDa polypeptide.
In Anaeystis a concentration of atrazine (10-7 м), ten times lower than the one causing the highest rate of shade adaptation (10-6 м), was shown to induce an optimum in cell density, which in turn resulted in an optimum in light-dependent O2 evolution. Both factors together might be responsible for the so-called greening effect observed in higher plants treated with sublethal concentrations of DCMU-type inhibitors of photosynthetic electron transport.
The cyanobacteria Anabaena cylindrica and Synechococcus leopoliensis (= Anacystis nidulans) were grown at different levels of UV-B radiation (439. 717, 1230 and 1405 J m -2d-1 weighted according Caldwell, 1971) for 2 days. Dry weight was hardly affected but phycocyanin content of both species decreased linearly to the level of UV-B radiation. Contents of protein, carotenoids and chlorophyll a were reduced only after exposure to high doses (1230 J m-2d-1) of UV-B radiation. Photosynthetic 14CO2 fixation of Anabaena cells was reduced linearly with increasing UV-B dose whereas no effect could be observed in Synechococcus. A depression of photosynthetic 15N-nitrate uptake was found after UV-B stress in both species. UV-B irradiance caused an increase of 15N-incorporation into glutamine, but no effect was noted for incorporation into alanine or aspartic acid. An increase of 15N-excess in glutamic acid linear with the UV-B dose was observed in Synechococcus, only. Patterns of 14C-labelled photosynthetic products were either less affected by UV-B radiation (Anabaena) or an enhancement of 14C-label in total amino acids was detected (Synechococcus). The amount of total free amino acids increased parallel to the level of UV-B radiation. Only, the high dose of UV-B (1405 J m-2d-1, weighted) results in a decrease of the glutamine pool. Our results indicate an inhibition of glutamate synthase by UV-B irradiation in Anabaena, only. Results were discussed with reference to the damage of the photosynthetic apparatus.
During photooxidation of polycyclic aromatic hydrocarbons (PAH) products can be formed which develop chemiluminescence on treatment with bases. Flash photolysis experiments show that this is the case only after previous formation of cation radicals, e.g. in the presence of CCl4 as solvent or of e-acceptors in aprotic solvents. These radicals react with oxygen to peroxy-radicals which can combine to several kinds of peroxides. Primary and secondary peroxides are the sources of chemiluminescent activity.
Chemiluminescent peroxides can also be obtained by irradiation of PA H carbonyl com pounds in protic solvents under nitrogen. It is assumed that two excited CO groups combine exceptionally with their O-atom s thus creating a peroxide bond. 24 aromatic aldehydes, ketones, dicarboxylic acid anhydrides and coumarines develop chemiluminescence after illumination with wavelengths ≥ 320 nm with intensities varying 4 magnitudes of order.
The sensitivity of the photochemiluminescent method is sufficient to detect amounts of PA H and their CO derivatives in the ppb to ppm range.
A phase equilibrium study of the system aluminiumchloride and pyridiniumchloride has been carried out. The phase diagram of the system indicates the existence of four congruently melting compounds of the molar ratio AlCl3/PyHCl 1:1, 1:2, 1:3, 2:3.
Potential energy, dipole moment, and electronic transition moment functions for the A 3Πand X3Σ- states of PH have been calculated from highly correlated electronic wavefunctions. The electric dipole moments in the vibrational ground state of PH are calculated to be 0.637 Debye (A 3Π) and 0.403 Debye (X3Σ-). The predicted rates of spontaneous emission between low lying vibrational states of the X state lie in the range of 46 to 109 sec-1 (PH) and 12 to 30 sec-1 (PD). The calculated radiative lifetime of the v' = 0 level in the A 3Π state of 400 ns is lower by about 10 percent than the most recent experimental value. The classical intersection of the 5Σ- and the A 3Πstate has been calculated to lie between v' = 2 and 3 with an expected uncertainty of about 500 cm−1, whereas the onset of the rotationally dependent predissociation lies at v' = 0, J' = 11.
Die vielfache Überlagerung epochaler Bewußtseinsschichten, die Rettung erotischer Sprachgewalt bei gleichzeitiger Präsentation christlicher Sublimation der Erotik, die Spannung zwischen Sensualismus und Spiritualismus disponieren das Volkslied zur Herausforderung neuer ästhetischer Lösungen; Heines Tannhäuser sucht sie im Widerspiel von Deutschland und Frankreich und im Zusammenspiel von alter und neuer Poesie.
Die Polemik lebt aus der Spannung extremer Gegensätze: von Affekt und Besonnenheit, von Argument und Bluff, von Logik und Paralogismen, von Dokument und Metapher, von akribischer Kriminalistik und wilder Spekulation, von rigorosem ethischem Prinzip und brutalem Vernichtungswillen, von kalkulierter Wirkungsstrategie und dem Spiel mit dem Zufall. Diese Spannungen sind in der Polemik so organisiert, daß sie die Sprache bis an die Grenze ihrer Verbalität, ja über sie hinaus zu treiben versuchen. Das Wort soll, was es nicht kann, nicht mittelbar, sondern unmittelbar Tat werden. Diese polemische Grenzüberschreitungssucht ihres Mediums der rational argumentierenden Sprache macht die Brisanz, das Interesse, aber auch die fortdauernde Distanzierung von der Polemik, zumal in der Aufklärung, verständlich. Im folgenden sollen einige Stationen der Verdächtigung und Legitimierung der Gattung Polemik von der Aufklärung bis zum Vormärz nachgezeichnet werden, wobei - unumgänglich - das Verhältnis der Polemik zur entstehenden Kritik in Aufklärung und Romantik ins Zentrum der Aufmerksamkeit rückt.
