Floodplains and other wetlands depend on seasonal river flooding and play an important role in the terrestrial water cycle. They influence evapotranspiration, water storage and river discharge dynamics, and they are the habitat of a large number of animals and plants. Thus, to assess the Earth’s system and its changes, a robust understanding of the dynamics of floodplain wetlands including inundated areas, water storages, and water flows is required.
This PhD thesis aims at improving the modeling of large floodplains and wetlands within the global-scale hydrological model WaterGAP, in order to better estimate water flows and water storage variations in different storage compartments. Within the scope of this thesis, I have developed a new approach to simulate dynamic floodplain inundation on a global-scale. This approach introduces an algorithm into WaterGAP, which has a spatial resolution of 0.5 degree (longitude and latitude) globally. The new approach uses subgrid-scale topography, based on high-resolution digital elevation models, to describe the floodplain elevation profile within each grid cell by applying a hypsographic curve. The approach comprises the modeling of a two-way river-floodplain interaction, the separate downstream water transport within the river and the floodplain – both with temporally and spatially different variable flow velocities – and the floodplain-groundwater interactions. The WaterGAP version that includes the floodplain algorithm, WaterGAP 2.2b_fpl, estimates floodplain and river water storage, inundated area and water table elevation, and also simulates backwater effects.
WaterGAP 2.2b_fpl was applied to model river discharge, river flow velocity, water storages, water heights and surface water extent on a global-scale. Model results were comprehensively validated against ground observations and remote sensing data. Overall, the modeled and observed data are in agreement. In comparison to the former version WaterGAP 2.2b, the model performance has improved significantly. The improvements are most remarkable in the Amazon River basin. However, the seasonal variation of surface water extent and total water storage anomalies are still too low in many regions on the globe when compared to observations. A detailed analysis of the simulated results suggests that in the Amazon River basin the introduction of backwater effects is important for realistically simulating water storages and surface water extent. Future efforts should focus on the simulation of water levels in order to better model the flow routing according to water slope. To further improve the model performance in specific regions, I recommend that the globally constant model parameters that affect inundation initiation, river-floodplain interaction, DEM correction for vegetation, and backwater amount at basin or subbasin-scale be adjusted.
Die paravertebralen Grenzstränge entwickeln sich aus Neuralleistenzellen des Rumpf- und Lendenbereichs. Diese sammeln sich im Hühnerembryo an Embryonaltag 2,5-3 an der dorsalen Aorta und formen die primären sympathischen Ganglien. Die dorsale Aorta sezerniert Morphogene, welche einen Teil der Vorläuferzellen zur Differenzierung zu Neuroblasten anregt. Die sympathischen Neuroblasten sind, obgleich sie bereits neurale und noradrenerge Marker exprimieren, zur Zellteilung fähig. Sie unterscheiden sich darin von anderen Neuralleistenderivaten wie beispielsweise den Neuronen der parasympathischen Ziliarganglien und der sensorischen Hinterwurzelganglien. Schließlich wandern die primären sympathischen Ganglien weiter und bilden lateral zum Notochord die paravertebralen Grenzstränge (Rohrer, 2011).
Der Homöodomänen-Transkriptionsfaktor PROX1 wird im Laufe der Entwicklung höherer Vertebraten in vielen Geweben exprimiert. Welche Wirkung PROX1 dabei auf Überleben, Migration, Proliferation und Differenzierung hat, hängt davon ab, in welchem Zelltyp er aktiv ist (Dyer et al., 2003; Lavado et al., 2010). Im peripheren Nervensystem konnte PROX1 embryonal in den Hinterwurzelganglien und den sympathischen Ganglien nachgewiesen werden (Becker et al., 2009; Diplomarbeit Julia Holzmann, 2010). Zielsetzung dieser Dissertation war es, die Expression und die Funktion von PROX1 in sympathischen Ganglien von Hühnerembryonen zu analysieren.
Die Expressionsanalyse von PROX1 zeigte, dass der Anteil der PROX1-positiven Neurone an Embryonaltag 5 (E5) ein Maximum erreicht und danach im Laufe der Entwicklung stetig abnimmt. Dies gilt ebenso für die Population der proliferierenden Neuroblasten, welche ebenfalls im Laufe der Hühnerentwicklung erstmals detailliert untersucht wurde. Diese Korrelation führte zu der Vermutung, dass PROX1 hauptsächlich in proliferierenden Zellen exprimiert wird, welche anschließend experimentell bestätigt werden konnte. Die Population der PROX1-positiven und die der p27-negativen Neuroblasten haben in allen untersuchten Hamburger Hamilton-Stadien (HH-St 21-37) eine vergleichbare Größe. Dennoch ist PROX1 durchgehend in einem kleinen Teil der p27-positiven Neurone enthalten. Diese Population verändert sich im Laufe der Entwicklung kaum und das Fluoreszenzsignal eines oder beider Proteine ist bei doppelpositiven Zellen deutlich schwächer. Diese und andere Daten dieser Arbeit weisen darauf hin, dass es sich um Neuroblasten handelt, welche gerade aus dem Zellzyklus austreten. In postmitotischen Neuronen geht PROX1 verloren. Obwohl PROX1 in allen untersuchten HH-Stadien stark in der Population proliferierender Neurone exprimiert wird, zeichnet sich ab E7 eine kleinere Population von Neuroblasten in S-Phase ab, welche kein PROX1 enthalten.
Die Vorläuferzellen von Ziliarganglien werden, ähnlich wie die der sympathischen Ganglien, durch BMP-Proteine zur Differenzierung angeregt (Müller und Rohrer, 2002). Aufgrund der Ähnlichkeiten in der Entwicklung beider Neuralleistenderivate wurde die Expression von PROX1 in dieser Dissertation auch in Ziliarganglien untersucht: Der Transkriptionsfaktor wird dort nur an E4 und E5 vereinzelt in Neuronen exprimiert und nahezu gar nicht in Vorläuferzellen. In späteren HH-Stadien ist PROX1 in Ziliarganglien nicht mehr nachweisbar.
Ebenfalls konnte hier gezeigt werden, dass PROX1 in primären sympathischen Ganglien an E3 (HH-St 21) in Vorläuferzellen exprimiert wird, welche bereits begonnen haben, sich zu Neuroblasten zu differenzieren. Noch bevor die Differenzierung dieser Zellen jedoch abgeschlossen ist, wird PROX1 transient herunterreguliert. Die entstehenden Neuroblasten treten in dieser Phase kurzzeitig aus dem Zellzyklus aus. Da sich die Größe der p27-negativen und der PROX1-positiven Population auch an E3 stark ähnelt, kann man schließen, dass die Zellteilung in den Neuroblasten erst bei erneuter PROX1-Expression wieder aufgenommen wird. Ab E5 ist PROX1 fast ausschließlich in Neuroblasten nachweisbar.
Eine Funktionsanalyse von PROX1 unter Kulturbedingungen und im Hühnerembryo sollte durch Knockdown und Überexpression zeigen, welchen Einfluss der Transkriptionsfaktor auf die Proliferation der Neuroblasten nimmt. Die Manipulation der PROX1-Expression hatte in vitro einen proproliferativen Effekt. In vivo unterschieden sich Knockdown und Überexpression aber nicht von der Kontrolle.
Zusammenfassend wurde in dieser Doktorarbeit die Expression von PROX1 in sympathischen Ganglien von Hühnerembryonen im Detail analysiert. Der Transkriptionsfaktor ist sowohl in Vorläuferzellen als auch in Neuroblasten nur transient vorhanden. Zwar konnte eine klare Korrelation zwischen der Expression von PROX1 und der Proliferation der sympathischen Neuroblasten festgestellt werden, allerdings konnte eine Wirkung von PROX1 auf die Proliferation durch Funktionsanalysen nur teilweise bestätigt werden. Zusammen weisen die Daten darauf hin, dass PROX1 eine Rolle in der Feinregulation der Proliferation spielt.
Testamente fungieren als Mittler zwischen Vergangenheit und Zukunft. Sie organisieren Memoria und Nachleben, beeinflussen Verwandtschaftsordnungen und bestimmen deren Reproduktion, sie verursachen oder lösen rechtliche, familiale, ökonomische Konflikte. Insofern zählt das Testament zu den wichtigsten kulturellen Formen des geregelten Übergangs von Leben zu Leben, durch den Tod getrennt.
Ulrike Vedders Studie analysiert das Testament im Spannungsfeld von Recht, Ökonomie und Kultur sowie im Zusammenhang mit Konzepten von Erbschaft, Vererbung und Nachleben. Sie entwickelt eine Poetologie des Testaments anhand der Literatur des 19. Jahrhunderts (von Jean Paul, Kleist, E.T.A. Hoffmann, Balzac, Heine, Droste-Hülshoff, Stifter, Melville, Keller, Storm, Fontane, Zola, James u.a.) und erforscht zudem das Testament als eine Form der Übertragung von Eigentum, Dingen, Rechten, Identität, Schuld, Leidenschaften. Als ein Medium für weitreichende - literarische und außerliterarische - Erbe- und Transferprozesse bildet das Testament eine zentrale Figur der kulturellen Tradierung sowie des Austausches zwischen verschiedenen Wissensfeldern, zwischen Literatur, alltäglicher Praxis und Wissenschaften.
In diversen Wissenschaftsdisziplinen ist derzeit eine Konjunktur der Begriffe Modell und Modellierung zu beobachten. Darunter sind auch solche Disziplinen, die Begriff wie Praxis des Modells bislang nicht zu ihrem methodischen Kernbereich zählten. Die Aktualität des Begriffs wie die Notwendigkeit einer Arbeit mit Modellen zeigt sich beispielsweise im Kontext sogenannter 'global challenges' wie klimatischer oder ökonomischer Krisen, die immer schon die Zuständigkeit einzelner Wissenschaften überschreiten und eine interdisziplinäre Zusammenarbeit verlangen. Eine zunehmende Resonanz modellhaften Denkens lässt sich indessen auch dort erkennen, wo kanonische Themen aus dem Geltungsbereich der Geistes- und Kulturwissenschaften zur Diskussion stehen. Es liegt daher die Vermutung nahe, der Modellbegriff erweise sein Potenzial nicht nur hinsichtlich der Analyse und Prognose komplexer dynamischer Systeme, sondern auch im Hinblick auf die interdisziplinäre Untersuchung tradierter Gegenstände wie literarischer Formen und Epochen, deren geschichtlicher Wandel beleuchtet werden soll. Die neue Anschlussfähigkeit für literatur- und kulturwissenschaftliche Ansätze ist u. a. der jüngeren Modelltheorie zu verdanken, welche das Modell bzw. den Modellierungsvorgang selbst im Modell erfasst und zugleich auf die 'aktiven Potenziale' sowie auf die Grenzen von Modellen hinsichtlich ihres epistemischen Status oder ihrer Materialität hingewiesen hat.
Serine/arginine-protein kinase 1 (SRPK1) regulates alternative splicing of VEGF-A to pro-angiogenic isoforms and SRPK1 inhibition can restore the balance of pro/antiangiogenic isoforms to normal physiological levels. The lack of potency and selectivity of available compounds has limited development of SRPK1 inhibitors, with the control of alternative splicing by splicing factor-specific kinases yet to be translated. We present here compounds that occupy a binding pocket created by the unique helical insert of SRPK1, and trigger a backbone flip in the hinge region, that results in potent (<10 nM) and selective inhibition of SRPK1 kinase activity. Treatment with these inhibitors inhibited SRPK1 activity and phosphorylation of serine/arginine splicing factor 1 (SRSF1), resulting in alternative splicing of VEGF-A from pro-angiogenic to antiangiogenic isoforms. This property resulted in potent inhibition of blood vessel growth in models of choroidal angiogenesis in vivo. This work identifies tool compounds for splice isoform selective targeting of pro-angiogenic VEGF, which may lead to new therapeutic strategies for a diversity of diseases where dysfunctional splicing drives disease development.
Philadelphia-like B-cell precursor acute lymphoblastic leukemia (Ph-like ALL) is characterized by distinct genetic alterations and inferior prognosis in children and younger adults. The purpose of this study was a genetic and clinical characterization of Ph-like ALL in adults. Twenty-six (13%) of 207 adult patients (median age: 42 years) with B-cell precursor ALL (BCP-ALL) were classified as having Ph-like ALL using gene expression profiling. The frequency of Ph-like ALL was 27% among 95 BCP-ALL patients negative for BCR-ABL1 and KMT2A-rearrangements. IGH-CRLF2 rearrangements (6/16; P=0.002) and mutations in JAK2 (7/16; P<0.001) were found exclusively in the Ph-like ALL subgroup. Clinical and outcome analyses were restricted to patients treated in German Multicenter Study Group for Adult ALL (GMALL) trials 06/99 and 07/03 (n=107). The complete remission rate was 100% among both Ph-like ALL patients (n=19) and the “remaining BCP-ALL” cases (n=40), i.e. patients negative for BCR-ABL1 and KMT2A-rearrangements and the Ph-like subtype. Significantly fewer Ph-like ALL patients reached molecular complete remission (33% versus 79%; P=0.02) and had a lower probability of continuous complete remission (26% versus 60%; P=0.03) and overall survival (22% versus 64%; P=0.006) at 5 years compared to the remaining BCP-ALL patients. The profile of genetic lesions in adults with Ph-like ALL, including older adults, resembles that of pediatric Ph-like ALL and differs from the profile in the remaining BCP-ALL. Our study is the first to demonstrate that Ph-like ALL is associated with inferior outcomes in intensively treated older adult patients. Ph-like adult ALL should be recognized as a distinct, high-risk entity and further research on improved diagnostic and therapeutic approaches is needed.
Wie bereits die Beiträge in der letzten Ausgabe des FIB beschäftigen sich auch diese mit Begriffen auf der Grenze von Natur und Kultur, von Evolution und Geschichte: 'Environment', 'Ecosystem', 'Noosphäre' und 'Anthropozän' gehören zu einem sich überschneidenden Wortfeld, das im Deutschen der Ökologie- und Umweltproblematik zugeordnet ist. Diese Begriffe haben ihre gegenwärtige Brisanz nicht zuletzt durch die Klimaveränderung gewonnen, bei der die Frage, ob oder inwieweit der mit ihnen erfasste Sachverhalt der Natur oder der menschlichen Kultur/Gesellschaft zugehört, umstritten und damit höchst politisch ist.
Faces are thought to be processed primarily according to their configurations which is in-ferred from comparisons with non-facial stimuli. While the whole (face) seems to be more than the sum of its parts, the same does not apply to objects which are processed analytically according to their featural information. A recent recognition model stresses the importance of certain visual information within facial stimuli. By applying a specific filtering technique, stimuli can be generated that are restricted to contain information of only a certain orienta-tion. Dakin and Watt (2009) reported greatest recognition performance with faces that only contained horizontally aligned information with accuracy continuously declining at vertical. Furthermore, they showed that, compared with images of natural scenes, horizontal contours within faces have an unusual tendency to fall into vertically co-aligned clusters which were labelled biological ‘bar code’ referring to a highly constrained one-dimensional code. Con-secutive research tested for face-specific processing by comparing faces and objects that displayed information of different orientations. Results suggested configural processing only for faces that contained horizontal information (Goffaux & Dakin, 2010). The findings con-tribute important insight on a still unanswered question in face processing research: what information is extracted from faces for recognizing them. Despite the importance of remembering human faces on a daily basis, this ability seems to develop disadvantageously over lifetime. Decreased accuracy cannot be attributed to de-creased general cognitive ability (Hildebrandt, Wilhelm, Schmiedek, Herzmann, & Sommer, 2011) and slower reactions times are assumed to be a product of decision making rather than sensory speed (Habak, Wilkinson, & Wilson, 2008). Considering the amount of published work on face recognition, there is a lack of studies available assessing this important ability at a higher age. New theoretical concepts are rarely examined with older participants, appar-ently assuming their general validity. The current dissertation tries to help fill this gap by assessing the importance of horizontal information from a developmental perspective com-paring younger and older adults under different experimental variations. The first study showed, that presenting older participants with horizontally filtered faces has a dispropor-tional negative impact on recognizing younger unfamiliar faces suggesting differential pro-cessing mechanisms, since recognizing stimuli that only contained vertical information did not differ between age groups. On this basis, the following study manipulated the presented stimulus material, since some evidence suggests that own-age faces are more easily recog-nized compared to faces of other ages, which is referred to as “own-age bias”. Therefore, the second study systematically assessed the impact of stimulus age on recognition sensitivity. Moreover, encoding modalities were varied by providing increased exposure duration to the stimuli. The results of the first study were replicated, as older participants’ performance was still poor at recognizing younger faces, independent from encoding modalities. However, similar face recognition sensitivity compared to younger adults was observable when filtered faces of the older adults’ own age had to be recognized. Interestingly, correlations between recognizing filtered and unfiltered faces were obtained for younger adults but not for older adults suggesting age variant processing of horizontal information. The last study assessed the importance of horizontal information with stimulus material familiar to the observer. Although research highlights differences between recognizing unfamiliar and familiar stimu-lus material, this factor is often not considered by contemporary research. By presenting par-ticipants with their own faces, a stimulus of greatest individual familiarity was chosen. The superiority of own face recognition over other familiar material is referred to as “self-face advantage” and has been shown in comparison with personally familiar faces (Keyes & Brady, 2010) and famous faces (Caharel et al., 2002). While younger adults indeed recog-nized their self-faces better compared to famous faces independent from stimuli being fil-tered or unfiltered, older participants displayed a completely different pattern including the inability to recognize their filtered self-faces. Again, significant associations were obtained between filtered and unfiltered recognition conditions suggesting convergent processing mechanisms for younger adults but not for the older age group. This dissertation provides a first insight in the divergence of response behavior in older adults with a recent face processing model. While the obtained data undermine the im-portance of horizontal information in younger adults by replicating and extending previously published work, a profoundly different type of processing is suggested at a higher age which largely relies on low-level pictorial information due to the inability to process horizontally filtered faces configurally. Specifically, it is suggested that with age, focusing on aging-salient features with configural processing disrupted may function as a critical source of di-agnostic information which can ultimately result in performance similar to younger adults.
Wir haben für diese Ausgabe einmal Spezialisten gebeten, in kurzen Expertisen die Rezeption Kosellecks und der Begriffsgeschichte wenigstens für einige ausgewählte Ländern zu untersuchen. Dabei haben wir exemplarisch sowohl Länder ausgewählt, in denen es, wie in Spanien, Frankreich oder Italien, bereits eine längere Rezeption gibt als auch solche, in denen die Rezeption 'in status nascendi' beobachtet werden kann, wie in Polen oder der Türkei.
