Der mythische-sakrale Bericht scheint dazu bestimmt zu sein, eine Verkörperung zu finden, die der zu Grunde liegenden Vorstellung eine größere Anziehungskraft verleiht, wobei beides, Verkörperung und Vorstellung, nur eng miteinander verbunden ihre religiöse Wirkung erlangen können. Das sehen wir am Beispiel einer Pilgerfahrt, bei welcher die Pilger danach streben, ihrem ›Heiligen Objekt‹ körperlich wie geistig näher zu sein. [...] Unter dem ›Rätsel der Mythologie‹, sofern davon die Rede sein kann, würden wir eine Art Bewegung der Mythologie verstehen, die darin besteht, die sakralen Werte für bestimmte Institutionen wie Religion oder Politik festzulegen und ebenso in einer zweiten Phase ihrer Selbstbewegung die eigene ›Identität‹ als Mythologie aufzugeben und unter anderen ›Gestalten‹ in Erscheinung zu treten: Sie heißt ab jetzt nicht mehr ›Mythologie‹, sondern ›heilige Geschichte‹ oder ›nationale Identität‹.
MLL-r Leukemia

Chromosomal translocations of the human mixed-lineage leukemia (MLL) gene have been analyzed for more than 20 yr at the molecular level. So far, we have collected about 80 direct MLL fusions (MLL-X alleles) and about 120 reciprocal MLL fusions (X-MLL alleles). The reason for the higher amount of reciprocal MLL fusions is that the excess is caused by 3-way translocations with known direct fusion partners. This review is aiming to propose a solution for an obvious problem, namely why so many and completely different MLL fusion alleles are always leading to the same leukemia phenotypes (ALL, AML, or MLL). This review is aiming to explain the molecular consequences of MLL translocations, and secondly, the contribution of the different fusion partners. A new hypothesis will be posed that can be used for future research, aiming to find new avenues for the treatment of this particular leukemia entity.
Over the last 15 years the Diagnostic Center of Acute Leukemia (DCAL) at the Frankfurt University has diagnosed and elucidated the Mixed Lineage Leukemia (MLL) recombinome with >100 MLL fusion partners. When analyzing all these different events, balanced chromosomal translocations were found to comprise the majority of these cases (~70%), while other types of genetic rearrangements (3-way-translocations, spliced fusions, 11q inversions, interstitial deletions or insertion of chromosomal fragments into other chromosomes) account for about 30%. In nearly all those complex cases, functional fusion proteins can be produced by transcription, splicing and translation. With a few exceptions (10 out of 102 fusion genes which were per se out-of-frame), all these genetic rearrangements produced a direct MLL fusion gene, and in 94% of cases an additional reciprocal fusion gene. So far, 114 patients (out of 2454 = ~5%) have been diagnosed only with the reciprocal fusion allele, displaying no MLL-X allele. The fact that so many MLL rearrangements bear at least two fusion alleles, but also our findings that several direct MLL fusions were either out-of-frame fusions or missing, raises the question about the function and importance of reciprocal MLL fusions. Recent findings also demonstrate the presence of reciprocal MLL fusions in sarcoma patients. Here, we want to discuss the role of reciprocal MLL fusion proteins for leukemogenesis and beyond.

This chapter reviews the boron isotopic composition of the ocean floor, including pristine igneous oceanic crust such as mid-ocean ridge basalts and ocean island basalts and their implications for the B isotopic composition of the mantle. The chapter further discusses the B isotopic effects of assimilation of altered crustal materials in mantle-derived magmas. The systematics of seawater alteration on oceanic rocks are discussed, including sediments, igneous crust and serpentinization of ultramafic rocks and the respective marine hydrothermal vent fluids. The chapter concludes with a discussion of the secular evolution of the B isotopic composition of seawater.
Lifespan Extension of Podospora anserina Mic60-Subcomplex Mutants Depends on Cardiolipin Remodeling

Function of mitochondria largely depends on a characteristic ultrastructure with typical invaginations, namely the cristae of the inner mitochondrial membrane. The mitochondrial signature phospholipid cardiolipin (CL), the F1Fo-ATP-synthase, and the ‘mitochondrial contact site and cristae organizing system’ (MICOS) complex are involved in this process. Previous studies with Podospora anserina demonstrated that manipulation of MICOS leads to altered cristae structure and prolongs lifespan. While longevity of Mic10-subcomplex mutants is induced by mitohormesis, the underlying mechanism in the Mic60-subcomplex deletion mutants was unclear. Since several studies indicated a connection between MICOS and phospholipid composition, we now analyzed the impact of MICOS on mitochondrial phospholipid metabolism. Data from lipidomic analysis identified alterations in phospholipid profile and acyl composition of CL in Mic60-subcomplex mutants. These changes appear to have beneficial effects on membrane properties and promote longevity. Impairments of CL remodeling in a PaMIC60 ablated mutant lead to a complete abrogation of longevity. This effect is reversed by supplementation of the growth medium with linoleic acid, a fatty acid which allows the formation of tetra-octadecanoyl CL. In the PaMic60 deletion mutant, this CL species appears to lead to longevity. Overall, our data demonstrate a tight connection between MICOS, the regulation of mitochondrial phospholipid homeostasis, and aging of P. anserina.
