In conclusion, I described for the first time the in vivo functions of PAK2 during cardiac development and its requirement for heart contractility
AIM1 – Characterization of Pak2a and Pak2b functions during cardiovascular system development: description of the phenotype triggered by the loss of expression of pak2b in the pak2a mutant Firstly, in addition to the confirmation of the published data regarding the pak2a mutant and morphant phenotype, I showed that pak2bbns159 mutant does not exhibit morphological defects, neither in the ISV formation nor in the brain vascular patterning. More importantly, I analyzed in more details the phenotypic consequences of pak2a and pak2b loss of expression in the trunk and brain vasculatures. Indeed, the lack of blood flow in the embryos, was associated with central arteries migration defects and reduced lumen in these central arteries and the ISVs. Moreover, pak2a and pak2b loss of expression resulted in cardiac failure.
AIM2 – Role of Pak2 on cardiac contractility From 40 -46 hpf, I found a weaker heart contractility in the pak2ami149/mi149;pak2bbns159/bns159. Although, the PAK proteins have been shown to impact the actin cytoskeleton organization, the heart morphological defects associated with the altered contractility, were not associated with acto-myosin filament reorganization. However, by analyzing in more details the structure of the sarcomeres, I was able to demonstrate that the proteins constituting the sarcomeres were strongly affected and showed an altered spatial organization. Then, I also described the effects of the loss of expression of both paralogs on the junctional protein localization. I demonstrated the loss of Pak2 function resulted in junction protein rearrangement in the cardiomyocytes in the pak2ami149/mi149;pak2bbns159/bns159 mutants at 40 and 46 hpf.
Thus, I was able for the first time to demonstrate in vivo PAK2 functions during cardiac development and its requirement for proper cardiac contractility activity.
AIM3 – Decipher mechanism of Pak2 signaling cascade involved during cardiac development Both pak2a and pak2b WT mRNAs were able to rescue the pak2ami149/mi149;pak2bbns159/bns159 mutant heart defects and the results indicated that these paralogs share overlapping function during cardiac development. Moreover, although I was not able to examine the control transgenic lines, myocardial and endothelial specific pak2a overexpression did not ameliorate the mutant cardiac deficiency. Thus,the absence of rescue by reactivating pak2a in cardiomyocytes indicates a non-cell autonomous function of Pak2a on cardiomyocytes.
For the first time, this study allowed to follow PAK2 in vivo functions during cardiovascular development. More importantly, its role on heart contractility regulation would enable further investigations to generate new tools for the treatment of cardiomyopathies.
Macrophages exert the primary cellular immune response. Pathogen components like bacterial lipopolysaccharides (LPS) stimulate macrophage migration, phagocytotic activity and cytokine expression. Previously, we identified the poly(A)+ RNA interactome of RAW 264.7 macrophages. Of the 402 RNA-binding proteins (RBPs), 32 were classified as unique in macrophages, including nineteen not reported to interact with nucleic acids before. Remarkably, P23 a HSP90 co-chaperone, also known as cytosolic prostaglandin E2 synthase (PTGES3), exhibited differential poly(A)+ RNA binding in untreated and LPS-induced macrophages. To identify mRNAs bound by P23 and to elucidate potential regulatory RBP functions in macrophages, we immunoprecipitated P23 from cytoplasmic extracts of cross-linked untreated and LPS-induced cells. RNAseq revealed that enrichment of 44 mRNAs was reduced in response to LPS. Kif15 mRNA, which encodes kinesin family member 15 (KIF15), a motor protein implicated in cytoskeletal reorganization and cell mobility was selected for further analysis. Noteworthy, phagocytic activity of LPS-induced macrophages was enhanced by P23 depletion. Specifically, in untreated RAW 264.7 macrophages, decreased P23 results in Kif15 mRNA destabilization, diminished KIF15 expression and accelerated macrophage migration. We show that the unexpected RBP function of P23 contributes to the regulation of macrophage phagocytotic activity and migration.
In large models of neuronal cell death, there is a tight correlation between Cdk5 deregulation and cell-cycle dysfunction. However, pathways that link Cdk5 to the cell cycle during neuronal death are still unclear. We have investigated the molecular events that precede p25/Cdk5-triggered neuronal death using a neuronal cell line that allows inducible p25 expression. In this system, no sign of apoptosis was seen before 24 hours of p25 induction. Thus, at that time, cell-cycle-regulatory proteins were analysed by immunoblotting and some of them showed a significant deregulation. Interestingly, after time-course experiments, the earliest feature correlated with p25 expression was the phosphorylation of the retinoblastoma protein (Rb). Indeed, this phosphorylation was observed 6 hours after p25 induction and was abolished in the presence of a Cdk5 inhibitor, roscovitine, which does not inhibit the usual Rb cyclin-D kinases Cdk4 and Cdk6. Furthermore, analyses of levels and subcellular localization of Cdk-related cyclins did not reveal any change following Cdk5 activation, arguing for a direct effect of Cdk5 activity on Rb protein. This latter result was clearly demonstrated by in vitro kinase assays showing that the p25-Cdk5 complex in our cell system phosphorylates Rb directly without the need for any intermediary kinase activity. Hence, Rb might be an appropriate candidate that connects Cdk5 to cell-cycle deregulation during neuronal cell death.
We develop a simple theoretical model to motivate testable hypotheses about how peer-to-peer (P2P) platforms compete with banks for loans. The model predicts that (i) P2P lending grows when some banks are faced with exogenously higher regulatory costs; (ii) P2P loans are riskier than bank loans; and (iii) the risk-adjusted interest rates on P2P loans are lower than those on bank loans. We confront these predictions with data on P2P lending and the consumer bank credit market in Germany and find empirical support. Overall, our analysis indicates the P2P lenders are bottom fishing when regulatory shocks create a competitive disadvantage for some banks.
Hypertension is a primary risk factor for cardiovascular diseases including myocardial infarction and stroke. Major determinants of blood pressure are vasodilatory factors such as nitric oxide (NO) released from the endothelium under the influence of fluid shear stress exerted by the flowing blood. Defects in flow-induced NO formation go along with endothelial dysfunction, initiation and progression of atherosclerosis as well as with arterial hypertension. Previous work has identified several mechanotransducing signaling processes involved in fluid shear stress-induced endothelial effects. But how fluid shear stress initiates the response is poorly understood. Here, I show in human and bovine endothelial cells that the G-protein Gq/G11 and the purinergic receptor P2Y2 mediate fluid shear stress-induced endothelial responses such as Ca2+ release, nitric oxide (NO) formation and the phosphorylation of platelet-endothelial-cell-adhesion-molecule-1 (PECAM-1), vascular endothelial growth factor-2 (VEGFR-2) and Akt kinase as well as activation of the endothelial NO synthase (eNOS). P2Y2 receptor is activated by adenosine triphosphate (ATP) which is released from endothelial cells under the influence of fluid shear stress. Arteries with P2Y2 or Gαq/Gα11 deficiency have impaired flow-induced dilatation. Mice with induced endothelium-specific deficiency of P2Y2 or Gαq/Gα11 develop hypertension which is accompanied by reduced eNOS activation. My data identify P2Y2 and Gq/G11 as a critical endothelial mechano-signaling pathway located upstream of mechanotransducing processes described so far. Moreover, I demonstrate that P2Y2 and Gq/G11 are required for basal endothelial NO formation, vascular tone and blood pressure.
Temporal predictability is thought to affect stimulus processing by facilitating the allocation of attentional resources. Recent studies have shown that periodicity of a tonal sequence results in a decreased peak latency and a larger amplitude of the P3b compared with temporally random, i.e., aperiodic sequences. We investigated whether this applies also to sequences of linguistic stimuli (syllables), although speech is usually aperiodic. We compared aperiodic syllable sequences with two temporally regular conditions. In one condition, the interval between syllable onset was fixed, whereas in a second condition the interval between the syllables’ perceptual center (p-center) was kept constant. Event-related potentials were assessed in 30 adults who were instructed to detect irregularities in the stimulus sequences. We found larger P3b amplitudes for both temporally predictable conditions as compared to the aperiodic condition and a shorter P3b latency in the p-center condition than in both other conditions. These findings demonstrate that even in acoustically more complex sequences such as syllable streams, temporal predictability facilitates the processing of deviant stimuli. Furthermore, we provide first electrophysiological evidence for the relevance of the p-center concept in linguistic stimulus processing.
Es steht ausser Zweifel, das der Schutz der Privatsphäre von Internet-Nutzern gegenwärtig unzureichend ist. Die Chance sich im Netz relativ weiträumig und frei zu bewegen, steht die Möglichkeit gegenüber allerlei Informationen über Internet-Nutzer zu sammeln und auszuwerten. Dies ist natürlich auch im Intranet möglich. Im Rahmen dieser Diplomarbeit wurden die verschiedenen Möglichkeiten überprüft, die zur Veröffentlichung von Datenschutzmassnahmen angeboten werden. Zunächst ist der OECD Privacy Statement Generator, dessen Principles Grundlage bei der Formulierung von Lufthansa Principles waren, auf Lufthansatauglichkeit untersucht worden. Dabei hat sich ergeben, dass trotz der theoretischen Übereinstimmung der Principles der Lufthansa AG mit denen der OECD, der Gebrauch des Generators bei Lufthansa in dieser Form nicht möglich ist. Da anfänglich eine Anpassung des Generators an Lufthansabedürfnisse geplant war, sind im Rahmen dieser Diplomarbeit Änderungsvorschläge gemacht worden. Die Anpassung des Codes erfolgte nicht, da dieser nur für öffentliche Stellen und nicht für Privatunternehmen zugänglich ist. Mit P3P entwickelte das W3C eine Datenschutztechnik, die für Nutzer die Kontrolle über persönliche Daten automatisiert und damit den Schutz der Privatsphäre und die Akzeptanz der User verbessert. Nach der Einführung des P3P- und APPEL-Vokabulars, mit dem man einerseits Datenschutzmassnahmen und andererseits Datenschutzpräferenzen ausdrücken kann, sollte daher geprüft werden, ob dieses Vokabular ausreicht, um Lufthansa-spezifische Aussagen zu machen und in wieweit diese erweiterbar bzw. anpassbar sind. Die Untersuchung hat ergeben, dass das Vokabular bis zu einem gewissen Masse ausreicht und es ein Element gibt, das EXTENSION Element, mit dem eine Erweiterung des P3P Standardvokabulars möglich ist. Im Rahmen dieser Arbeit wurden solche auf Lufthansa abgestimmte Erweiterungen sowohl für eine P3P Policy als auch für eine entsprechende APPEL Präferenz formuliert. Die Lufthansa AG hat somit mit P3P die Möglichkeit, Ihre Datenschutzpraktiken für den Mitarbeiter transparenter zu gestalten, da sie auch über das Standardvokabular hinausgehende Aussagen formulieren kann. In der Diplomarbeit sind ausserdem die sich zur Zeit auf dem Markt befindlichen Tools, die bei der Erstellung einer maschinenlesbaren Datenschutzmassnahme, der sog. Privacy Policy benutzt werden können, untersucht worden. Der IBM P3P Policy Editor scheint für den Gebrauch bei Lufthansa denkbar, da die Handhabung des Generators einfach ist. Der Mitarbeiter, der die Policy für seine Abteilung erstellen soll, braucht sich nicht mit den Einzelheiten des P3P Vokabulars auseinander setzten. Mit diesem Editor kann zunächst ein Basisgerüst einer Policy erstellt werden. Die mit dem P3P Element EXTENSION formulierten Erweiterungen müssen jedoch selbsterstellt werden und können nachträglich in das Basisgerüst der Policy miteingebunden werden. Zusätzlich zu der maschinenlesbaren Form einer Policy erstellt der IBM Editor auch eine menschenlesbare HTML-Version der Policy. Dies ist sehr von Vorteil, da in einem Arbeitsgang zwei Policy-Versionen erstellt werden. Im Vergleich zu dem Formulierungsentwurf des OECD Generators ist die menschenlesbare Version des IBM Editors ausserdem wesentlich kürzer und dadurch auch übersichtlicher. Im Ganzen ist es daher sinnvoller, den IBM Editor zu benutzen, als den OECD Generator neu zu programmieren und dann mit Hilfe eines anderen Tools die P3P Policy zu erstellen. Zur Erstellung einer APPEL Präferenz ist zur Zeit nur ein Hilfsmittel auf dem Markt erhältlich. Der Grund hierfür ist sicherlich, dass sich APPEL noch zu keinem Standard entwickelt hat, sondern sich noch in einem Entwurfsstadium befindet. Der APPEL Editor von JRC ist ähnlich wie der P3P Policy Editor von IBM aufgebaut. Auch hier müssen Erweiterungen selbst formuliert und in dem vom Editor erstellten Basisgerüst einer Präferenz eingebunden werden. Nachdem die grundsätzliche Erweiterbarkeit von P3P in dieser Diplomarbeit festgestellt wurde, sind die sog. User Agents behandelt worden. Von Bedeutung war neben der Funktionsweise der einzelnen User Agents ihre Handhabung des EXTENSION Elementes. Da zu erwarten ist, dass nur wenige Nutzer die Voreinstellungen ihrer Software selbst verändern und sich mit der APPEL Spezifikation auseinander setzten, wird der Standardkonfiguration eines P3P User Agents eine große Bedeutung beigemessen. Bei allen vorgestellten User Agents gab es verschiedene Sicherheitsniveaux bzgl. Datenschutz aus denen der Nutzer auswählen konnte. Entsprechend des Niveaux wurde die Präferenz des Nutzers automatisch konfiguriert. Bei allen war es ausserdem möglich, selbsterstellte APPEL Formulierungen zu importieren. Bei dem Proxy von JRC ist es möglich, Settings mit einzubinden, die das EXTENSION Element beinhalten. AT&T erlaubt dies nicht und von Microsoft fehlt hierzu jegliche Angabe. Bzgl. der Lufthansa AG erscheint es sinnvoll, den Mitarbeitern einen eigenen User Agent anzubieten, der alle zusätzlich formulierten Aspekte aufgreift und mit dem der Mitarbeiter seine Präferenzen bzgl. des Intranets leicht ausdrücken kann. Der Aufwand, den Mitarbeitern das erweiterte Vokabular für die Formulierung einer Präferenz zur Verfügung zu stellen, setzt voraus, dass jeder Mitarbeiter sich mit der Syntax und Semantik von P3P und APPEL auskennt. Dies ist im Vergleich zur Programmierung eines eigenen User Agents, der den Mitarbeitern zur Verfügung gestellt wird, aufwendiger und wahrscheinlich auch nicht realisierbar. Grundsätzlich kann durch diese Massnahmen das Vertrauen der Mitarbeiter ins Intranet und damit die Nutzung dieses Mediums gesteigert werden. Die vermehrte Nutzung des Intranets auch für private Zwecke, wie zum Beispiel der Buchung von Reisen oder die Abfrage nach Flügen etc., würde für beide Seiten auch wirtschaftlichen Nutzen bringen. Für die Mitarbeiter selbst käme es z.B. zu einer Zeitersparnis, da sie jetzt zur Buchung ihrer Reisen nicht mehr in die Reisestelle müssen. Dies hätte natürlich auch wirtschaftliche Auswirkungen, da der Aufwand, um zur Reisestelle zu kommen, wegfällt. Aber auch die Lufthansa AG hätte durch die transparentere Gestaltung ihrer Datenschutzpraktiken wirtschaftlichen Nutzen. Die Mitarbeiter z.B. in der Reisestelle würden durch das neue Vertrauen ihrer Kollegen ins Intranet und damit einhergehend die selbstständige Online-Buchungsmöglichkeit entlastet werden. Es könnten mehr Kapazitäten für andere Aufgaben frei werden. Das Ausmass der Vorteile für Lufthansa und Ihrer Mitarbeiter, die sich aus der Veröffentlichung von Datenschutzmaßnahmen ergeben und damit ist auch das vermehrte Vertrauen der Mitarbeiter ins Intranet gemeint, ist zur Zeit noch nicht in vollem Umfang erfassbar, da sich viele Intranetprojekte der Lufthansa AG noch im Entwicklungsstadium befinden. Für die Zukunft ist jedoch auch festzustellen, dass datenschutzfreundliche Technologien allein nicht die Lösung zur Sicherstellung des Datenschutzes im Inter- als auch im Intranet sein können. Vielmehr müssen die aufkommenden technischen Maßnahmen durch nationale und internationale Regelungen unterstützt und ergänzt werden. Erst durch Festlegung internationaler Konventionen, die den Datenschutz in Zusammenhang mit grenzüberschreitenden Computernetzwerken und Diensten regeln, kann ein effektiver und unabhängiger Kontrollmechanismus sowie die Möglichkeit zu Sanktionen gewährleistet werden. Die Veröffentlichung von Datenschutzmassnahmen gewährleistet leider nicht ihre Einhaltung.
Nitric oxide (NO) represents a short lived mediator that pivotally drives keratinocyte movements during cutaneous wound healing. In this study, we have identified p68 DEAD box RNA helicase (p68) from an NO-induced differential keratinocyte cDNA library. Subsequently, we have analyzed regulation of p68 by wound-associated mediators in human and murine keratinocytes. NO, serum, growth factors, and pro-inflammatory cytokines were potent inducers of p68 expression in the cells. p68 was constitutively expressed in the epithelial compartment of murine skin. Upon injury, we found a transient down-regulation of overall p68 protein in wound tissue. However, p68 did not completely disappear during early wound repair, as we found an expression of p68 protein in isolated wound margin tissue 24 h after wounding. Moreover, immunohistochemistry and cell fractionation analysis revealed a restricted localization of p68 in keratinocyte nuclei of the developing epithelium. Accordingly, cultured keratinocytes also showed a nuclear localization of the helicase. Moreover, confocal microscopy revealed a strong localization of p68 protein within the nucleoli of the cells. Functional analyses demonstrated that p68 strongly participated in keratinocyte proliferation and gene expression. Keratinocytes that constitutively overexpressed p68 protein were characterized by a marked increase in serum-induced proliferation and vascular endothelial growth factor expression, whereas down-regulation of endogenous p68 using small interfering RNA markedly attenuated serum-induced proliferation and vascular endothelial growth factor expression. Altogether, our results suggest a tightly controlled expression and nucleolar localization of p68 in keratinocytes in vitro and during skin repair in vivo that functionally contributes to keratinocyte proliferation and gene expression.
