Wir werden uns in dieser Arbeit vorwiegend mit einem Modell befassen, das Y. Peres, C. Kenyon, W. Evans und L.J. Schulman 1998 in ihrem Artikel \Broadcasting on trees and the Ising-Modell" eingeführt haben.
In diesem Modell wird ein Signal, das die Werte +1 oder -1 annehmen kann, von der Wurzel eines Baumes aus entlang der Äste eines unendlichgroßen Baumes übertragen. Die Kanten des Baumes agieren dabei als Übertragungskanäle zwischen den Knoten. Jede Kante kann das Signal korrekt übertragen oder es flippen, das heißt, das Vorzeichen des Signals umkehren.
Das Übertragungsverhalten der Kanten ist zufällig. Mit einer festen Wahrscheinlichkeit ϵ, mit 0 < ϵ <= 1/2 , verfälscht eine Kante das Signal. Dies geschieht an allen Kanten unabhängig mit der gleichen Wahrscheinlichkeit. Es stellt sich nun die Frage, wie groß diese Fehlerwahrscheinlichkeit höchstens sein darf, damit das, was in der Krone des Baumes ankommt, noch etwas zu tun hat mit dem, was in der Wurzel eingespeist wird. Mit anderen Worte: Sind die Signale auf Knoten, die einen Abstand >= n von der Wurzel haben, für n -> ∞ asymptotisch unabhängig vom Signal in der Wurzel? Eine Möglichkeit, den Grad der Abhängigkeit zu messen, ist die sogenannte Information, der Kullback-Leibler-Abstand von gemeinsamer Verteilung zur Produkt-Verteilung, die in Definition 16 eingeführt wird.
Wir werden sehen, daß es eine kritische Schwelle ϵc;I für Informationsübertragung gibt. Ist die Fehlerwahrscheinlichkeit größer als ϵc;I , so ist die Information, die zwischen Wurzel und Krone übertragen wird, 0. Ist die Fehlerwahrscheinlichkeit kleiner als ϵc;I , so wird Information übertragen. Dieser kritische Wert ϵc;I hängt nur von der Branching-Number, einer Art mittleren Verzweigungszahl, des Baumes (vgl. Definition 1) ab.
Wir werden sehen, daß das Broadcasting-Modell eine elegante Formulierung eines wohlbekannten Modells, des Ising-Modells, mit freien Randbedingungen, ist.
Im Ising-Modell hat jeder Knoten des Baumes einen "magnetischen" Spin, der entweder +1 oder -1 sein kann. Spins direkt benachbarter Knoten beeinflussen sich, in dem sie versuchen, den gleichen Wert anzunehmen. Diesem Effekt wirkt ein thermischer Einfluß entgegen, der mittels eines als Temperatur bezeichneten Parameters modelliert wird.
Die klassische Frage im Ising-Modell ist, ob Phasenübergang stattfindet. Wir wollen Phasenübergang als das Phänomen verstehen, daß die Wurzel des Baumes die Vorgabe von Randbedingungen auf der Krone des Baumes spürt. Ist dies der Fall, so sagen wir, daß Phasenübergang stattfindet. Auch dies ist eine
Form der gegenseitigen Beeinflussung zwischen Wurzel und Krone des Baumes. Russel Lyons hat 1989 in seinem Artikel \The Ising-Model on trees and treelike Graphs" das Ising-Modell auf Bäumen untersucht und gezeigt, daß es eine kritische Temperatur tc für Phasenübergang gibt. Ist die Temperatur höher als tc, so spürt die Wurzel nichts von den Randbedingungen der Krone; ist die Temperatur geringer als tc, so haben die Randbedingungen Einfluß auf die Wurzel. Auch hier hängt die kritische Temperatur nur von der Branching-Number des Baumes ab.
In der Broadcasting-Formulierung des Modells ist der Fluß von Information ein naheliegendes Werkzeug, um die Beeinflussung von Wurzel und Krone zu messen, in der Ising-Formulierung ist die Existenz von Phasenübergang ein ebenso naheliegendes Werkzeug, ebendiesen Einfluß zu messen.
Wir werden die beiden Arten der Beeinflussung miteinander vergleichen und können zeigen, daß für die Übertragung von Information stets eine stärkere Interaktion zwischen den Knoten notwendig ist, als für den Einfluß der Randbedingungen aus der Krone.
Als letztes Phänomen werden wir untersuchen, ob es einen Pfad im Baum gibt, der in der Wurzel startend nur Knoten gleichen Spins besucht und die unendlich weit entfernte Krone erreicht. Wir bezeichnen dieses Phänomen als Spinperkolation.
Wir werden die Berechnung der kritischen Interaktion für Spinperkolation in einem Bernoulli-Feld auf den Kanten rekapitulieren und dann zeigen, daß die Existenz eines Perkolationspfades nur von der Interaktionsstärke des Modells und nicht von etwaigen Randbedingungen abhängt. Dabei kombinieren wir Ergebnisse aus zwei Arbeiten von Lyons und die Erkenntnis, daß Broadcasting- Modell und freies Ising-Modell identisch sind. Wir erhalten so einen neuen, einfachen Beweis über die kritische Interaktion für Spinperkolation in der Plus-Phase des Ising-Modells, die Lyons bereits in [7] berechnet hat.
Im Rahmen dieser Arbeit möchte ich nun aufzeigen, dass ein Projekt zu Glücksspielen eine „reichhaltige Lernsituation“ darstellen kann, in der die Schüler Raum, Gelegenheit und Anlass haben, Grunderfahrungen mit zufälligen Vorgängen zu machen, darauf aufbauend wichtige Begriffe zu bilden und schließlich wesentliche stochastische Zusammenhänge zu erkennen. Der Projektmethode entsprechend lag ein Großteil meiner Tätigkeiten im Vorfeld in vorbereitenden und planenden Tätigkeiten. Während der Projektdurchführung trat ich als beratender „Hintergrundlehrer“ auf. Die Schüler arbeiteten weitgehend selbstständig. Der Schwerpunkt dieser Arbeit liegt daher auf meinen didaktischen und methodischen Überlegungen zur Vorbereitung des Projekts.
Die Weißtanne (Abies alba) – Symbol für naturnahe Waldwirtschaft – ist im Bayerischen Wald beheimatet. Die Tanne ist dort neben Fichte und Buche das prägende Element der Bergmischwälder auf Höhen von 600 bis 1250 m. Einstmals war sie mit etwa 20 % (ROTHE & BORCHERT 2003) am Waldaufbau beteiligt. Seit rund 150 Jahren ist jedoch ein drastischer Rückgang der Tanne - nicht nur im Bayerischen Wald – zu verzeichnen. Die Ursachen für den Rückgang der Tanne sind überwiegend anthropogen bedingt. In erster Linie ist hier die Forstwirtschaft zu erwähnen, die bestrebt ist durch waldbauliche Maßnahmen den Fichtenanteil anzuheben. An zweiter Stelle stehen die neuartigen Waldschäden, welche, bedingt durch Schadstoffimmissionen in den 70er und 80er Jahren des letzten Jahrhunderts, zu verstärktem Absterben der Tanne führten. Noch heute ist die Tanne die am stärksten geschädigte Baumart in Bayern (BAYSTMLF 2002). Ein weiterer nicht zu vergessender Faktor für den Rückgang der Tanne sind die z.T. hohen Wildbestände, durch deren Verbiss und Schälen junger Tannen das Heranwachsen einer neuen Generation verhindert wird. Mit dem Rückgang der Tanne verändert sich jedoch nicht nur die Baumartenzusammensetzung, sondern auch Tier- und Pflanzenarten sind direkt und indirekt betroffen. Die Arthropodenfauna der bisher kaum untersuchten Tanne wird als relativ artenarm beschrieben (BRÄNDLE & BRANDL 2001), jedoch gibt es wenige spezielle Untersuchungen an dieser Baumart (MÜLLER & GOßNER 2004).
Natürlicherweise war die Tanne (Abies alba) in den bayerischen Wäldern mit Anteilen von bis zu 20% vertreten (nach ROTHE & BORCHERT 2003). Seit 150 Jahren aber gehen die Tannenbestände in drastischem Ausmaß zurück, was nicht nur auf die wachsende Schadstoffbelastung der Luft, sondern auch auf hohe Wildbestände und die einseitige Waldbewirtschaftung zugunsten der Fichte (Picea abies) zurückzuführen ist. In jüngster Zeit wird seitens der Staatsforstverwaltung angestrebt, der Tanne ihren angestammten Platz in unseren Wäldern wieder einzuräumen (BAYERISCHES STAATSMINISTERIUM FÜR LANDWIRTSCHAFT UND FORSTEN 1993). Es stellt sich die Frage, welchen Beitrag die Tanne zur Biodiversität in Wäldern und speziell in Mischwäldern leistet. Frühere Untersuchungen beschrieben die Tannenfauna als artenarm im Vergleich mit anderen Baumarten (BÖHME 2001, BUCKING 1998), allerdings wurde die Kronenfauna dabei stets vernachlässigt. Da aber 90% eines Hochwaldes über der Reichhöhe eines Menschen liegt (BUßLER et al. 2004), sind Untersuchungen im Kronenraum höheren Straten für eine umfassende Aussage über die Fauna auf Bäumen von großer Bedeutung. Bis jetzt gibt es trotz des generell wachsenden Interesses an der Kronenfauna keine umfassenden Untersuchungen an Tanne (MÜLLER & GOSSNER 2004). Mit dem hier vorgestellten Projekt sollte begonnen werden, die Wissenslücke um die Insektenfauna in Tannenkronen zu schließen. Den xylobionten Käfern galt besondere Aufmerksamkeit, da sie als eine baumartengebundene Insektengruppe für vergleichendeUntersuchungen sehr gut geeignet und sowohl taxonomisch als auch ökologisch gut untersucht sind. Daneben wurden Heteroptera, Neuropterida und Hymenoptera bearbeitet .
Background: The integration of the non-cross-resistant chemotherapeutic agents capecitabine and vinorelbine into an intensified dose-dense sequential anthracycline- and taxane-containing regimen in high-risk early breast cancer (EBC) could improve efficacy, but this combination was not examined in this context so far. Methods: Patients with stage II/IIIA EBC (four or more positive lymph nodes) received post-operative intensified dose-dense sequential epirubicin (150mg/m2 every 2 weeks) and paclitaxel (225mg/m2 every 2 weeks) with filgrastim and darbepoetin alfa, followed by capecitabine alone (dose levels 1 and 3) or with vinorelbine (dose levels 2 and 4). Capecitabine was given on days 1-14 every 21 days at 1000 or 1250 mg/m2 twice daily (dose levels 1/2 and 3/4, respectively). Vinorelbine 25 mg/m2 was given on days 1 and 8 of each 21-day course (dose levels 2 and 4). Results: Fifty-one patients were treated. There was one dose-limiting toxicity (DLT) at dose level 1. At dose level 2 (capecitabine and vinorelbine), five of 10 patients experienced DLTs. Therefore evaluation of vinorelbine was abandoned and dose level 3 (capecitabine monotherapy) was expanded. Hand-foot syndrome and diarrhoea were dose limiting with capecitabine 1250 mg/m2 twice daily. At 35.2 months' median follow-up, the estimated 3-year relapse-free and overall survival rates were 82% and 91%, respectively. Administration of capecitabine monotherapy after sequential dose-dense epirubicin and paclitaxel is feasible in node-positive EBC, while the combination of capecitabine and vinorelbine as used here caused more DLTs. Background: Simultaneous pancreas kidney transplantation (SPK), pancreas transplantation alone (PTA) or pancreas transplantation after kidney (PAK) are the only curative treatment options for patients with type 1 (juvenile) diabetes mellitus with or without impaired renal function. Unfortunately, transplant waiting lists for this indication are increasing because the current organ acceptability criteria are restrictive; morbidity and mortality significantly increase with time on the waitlist. Currently, only pancreas organs from donors younger than 50 years of age and with a body mass index (BMI) less than 30 are allocated for transplantation in the Eurotransplant (ET) area. To address this issue we designed a study to increase the available donor pool for these patients.
Methods/Design: This study is a prospective, multicenter (20 German centers), single blinded, non-randomized, two armed trial comparing outcome after SPK, PTA or PAK between organs with the currently allowed donor criteria versus selected organs from donors with extended criteria. Extended donor criteria are defined as organs procured from donors with a BMI of 30 to 34 or a donor age between 50 and 60 years. Immunosuppression is generally standardized using induction therapy with Myfortic, tacrolimus and low dose steroids. In principle, all patients on the waitlist for primary SPK, PTA or PAK are eligible for the clinical trial when they consent to possibly receiving an extended donor criteria organ. Patients receiving an organ meeting the current standard criteria for pancreas allocation (control arm) are compared to those receiving extended criteria organ (study arm); patients are blinded for a follow-up period of one year. The combined primary endpoint is survival of the pancreas allograft and pancreas allograft function after three months, as an early relevant outcome parameter for pancreas transplantation.
Discussion: The EXPAND Study has been initiated to investigate the hypothesis that locally allocated extended criteria organs can be transplanted with similar results compared to the currently allowed standard ET organ allocation. If our study shows a favorable comparison to standard organ allocation criteria, the morbidity and mortality for patients waiting for transplantation could be reduced in the future.

Chronic granulomatous disease (CGD) is a primary immunodeficiency characterized by impaired antimicrobial activity in phagocytic cells. As a monogenic disease affecting the hematopoietic system, CGD is amenable to gene therapy. Indeed in a phase I/II clinical trial, we demonstrated a transient resolution of bacterial and fungal infections. However, the therapeutic benefit was compromised by the occurrence of clonal dominance and malignant transformation demanding alternative vectors with equal efficacy but safety-improved features. In this work we have developed and tested a self-inactivating (SIN) gammaretroviral vector (SINfes.gp91s) containing a codon-optimized transgene (gp91(phox)) under the transcriptional control of a myeloid promoter for the gene therapy of the X-linked form of CGD (X-CGD). Gene-corrected cells protected X-CGD mice from Aspergillus fumigatus challenge at low vector copy numbers. Moreover, the SINfes.gp91s vector generates substantial amounts of superoxide in human cells transplanted into immunodeficient mice. In vitro genotoxicity assays and longitudinal high-throughput integration site analysis in transplanted mice comprising primary and secondary animals for 11 months revealed a safe integration site profile with no signs of clonal dominance.
Epigenetic silencing of transgene expression represents a major obstacle for the efficient genetic modification of multipotent and pluripotent stem cells. We and others have demonstrated that a 1.5 kb methylation-free CpG island from the human HNRPA2B1-CBX3 housekeeping genes (A2UCOE) effectively prevents transgene silencing and variegation in cell lines, multipotent and pluripotent stem cells, and their differentiated progeny. However, the bidirectional promoter activity of this element may disturb expression of neighboring genes. Furthermore, the epigenetic basis underlying the anti-silencing effect of the UCOE on juxtaposed promoters has been only partially explored. In this study we removed the HNRPA2B1 moiety from the A2UCOE and demonstrate efficient anti-silencing properties also for a minimal 0.7 kb element containing merely the CBX3 promoter. This DNA element largely prevents silencing of viral and tissue-specific promoters in multipotent and pluripotent stem cells. The protective activity of CBX3 was associated with reduced promoter CpG-methylation, decreased levels of repressive and increased levels of active histone marks. Moreover, the anti-silencing effect of CBX3 was locally restricted and when linked to tissue-specific promoters did not activate transcription in off target cells. Thus, CBX3 is a highly attractive element for sustained, tissue-specific and copy-number dependent transgene expression in vitro and in vivo.
A wide variety of enzymatic pathways that produce specialized metabolites in bacteria, fungi and plants are known to be encoded in biosynthetic gene clusters. Information about these clusters, pathways and metabolites is currently dispersed throughout the literature, making it difficult to exploit. To facilitate consistent and systematic deposition and retrieval of data on biosynthetic gene clusters, we propose the Minimum Information about a Biosynthetic Gene cluster (MIBiG) data standard.
Impact of Docetaxel on blood-brain barrier function and formation of breast cancer brain metastases

Background: Breast cancer (BC) is the most frequent malignant tumor in females and the 2nd most common cause of brain metastasis (BM), that are associated with a fatal prognosis. The increasing incidence from 10% up to 40% is due to more effective treatments of extracerebral sites with improved prognosis and increasing use of MRI in diagnostics. A frequently administered, potent chemotherapeutic group of drugs for BC treatment are taxanes usually used in the adjuvant and metastatic setting, which, however, have been suspected to be associated with a higher incidence of BM. The aim of our study was to experimentally analyze the impact of the taxane docetaxel (DTX) on brain metastasis formation, and to elucidate the underlying molecular mechanism.
Methods: A monocentric patient cohort was analyzed to determine the association of taxane treatment and BM formation. To identify the specific impact of DTX, a murine brain metastatic model upon intracardial injection of breast cancer cells was conducted. To approach the functional mechanism, dynamic contrast-enhanced MRI and electron microscopy of mice as well as in-vitro transendothelial electrical resistance (TEER) and tracer permeability assays using brain endothelial cells (EC) were carried out. PCR-based, immunohistochemical and immunoblotting analyses with additional RNA sequencing of murine and human ECs were performed to explore the molecular mechanisms by DTX treatment.
Results: Taxane treatment was associated with an increased rate of BM formation in the patient cohort and the murine metastatic model. Functional studies did not show unequivocal alterations of blood-brain barrier properties upon DTX treatment in-vivo, but in-vitro assays revealed a temporary DTX-related barrier disruption. We found disturbance of tubulin structure and upregulation of tight junction marker claudin-5 in ECs. Furthermore, upregulation of several members of the tubulin family and downregulation of tetraspanin-2 in both, murine and human ECs, was induced.
Conclusion: In summary, a higher incidence of BM was associated with prior taxane treatment in both a patient cohort and a murine mouse model. We could identify tubulin family members and tetraspanin-2 as potential contributors for the destabilization of the blood-brain barrier. Further analyses are needed to decipher the exact role of those alterations on tumor metastatic processes in the brain.
