In addition to infectious viral particles, hepatitis B virus-replicating cells secrete high amounts of SVPs, which are ssembled by HBsAg in the shape of spheres and filaments but lack any capsid and genome. Filaments are characterized by a much higher amount of the surface protein LHBs as compared to spheres. Spheres are
released via the constitutive secretory pathway, while viral particles are ESCRT-dependently released via MVBs. The interaction of virions with the ESCRT machinery is mediated by α-taxilin that connects the PreS1 domain of LHBs with the ESCRT-component tsg101. Since viral particles and filaments contain a significant amount of LHBs, it is unclear whether filaments are secreted as spheres or released like viral particles. To study the release pathways of HBV filaments in the absence of viral particles, A core-deficient
HBV mutant (1.2×HBVΔCore) was generated by site-directed mutagenesis based on wt1.2x HBV. The start codon of core protein was mutated into stop codon, which was confirmed by DNA sequencing. Data from HBsAg ELISA, Western blot, immunofluorescence microscopy and immunoelectron microscopy showed that the lack of core protein did neither affect the production nor the secretion of HBV SVPs. The intracellular distribution of
LHBs and SHBs showed no difference between wtHBV and the core-deficient mutant expressing cells. Therefore, this system is suitable to investigate the release pathway of HBV filaments in the absence of viral particles. Confocal microscopy analysis of cells cotransfected core-deficient mutants with peYFPRab7 as marker for the endosomal/MVB pathway or with pGalT-eGFP as marker for the trans Golgi apparatus showed that YFP-Rab7, but not GalT-GFP, partially colocalized with LHBs. Furthermore, LHBs could be found in dilated MVBs by immune electron microscopy of ultrathin sections. This was confirmed by isolation of MVBs by cell fractionation using discontinuous sucrose gradient ultracentrifugation and percoll-based linear gradient ultracentrifugation, indicating that filaments enter MVBs in the absence of virion formation. Moreover, inhibition of MVB biogenesis by the small molecular inhibitor U18666A significantly abolished the release of filaments in a dose-dependent manner, but no inhibition could be observed in the production. In contrast, no inhibition on the secretion and production of spheres could be
detected. Inhibition of ESCRT-functionality by coexpression of transdominant negative mutants (Vps4A, Vps4B, CHMP3) abolished the release of filaments while secretion of spheres was not affected. These data indicate that in contrast Abstract 73 to spheres while are secreted via the secretory pathway, filaments are released via ESCRT/MVB pathway like infectious viral particles.
Three types of post-translation modifications (PTMs) containing N-glycosylation, phosphorylation, ubiquitylation were characterized in diffuse large B-cell lymphoma (DLBCL) on a global scale using quantitative mass spectrometry based proteomics technology in this study.
DLBCL is the most common type of malignant lymphomas and has a heterogeneous gene expression profiling, phenotype and clinical response to chemotherapy. DLBCL is a good model for the correct classification of cancers into molecularly different subtypes, which benefits for the selection of rational therapeutic strategies. It resulted in two histologically indistinguishable subtypes-activated B-cell-like (ABC) subgroup and germinal center B-cell-like (GCB) subgroup according to gene expression profiling. Signals originating from the B-cell receptor (BCR), the key protein on the surface of B cells, promote growth and survival of DLBCL. Antigen-dependent/independent BCR signaling is found in DLBCL subtypes.
Recent researches reveal that glycosylation plays role in human cells via site-specific regulation. Aberrant N-glycosylation in BCR-related effectors, such as, CD79a, immunoglobulin M or G (IgM or IgG), has been found to be associated with lymphoid malignancies. However, accurate quantification of intact glycopeptides and their individual glycan moieties in a cell-wide manner is still challenging. Here we established a site-specific quantitative N-glycoproteomics platform termed SugarQuant. It included a fast sample preparation workflow using Protein Aggregation Capture (PAC), an optimized multi-notch MS3 acquisition workflow (Glyco-SPS-MS3), a self-developed R-based tool (GlycoBinder). The robustness and accuracy of quantitation in SugarQuant were proved in a study using the different amounts of TMT-labelled IgM N-glycopeptides spiked into a background of TMT-labelled yeast peptides. Next, we used SugarQuant to identify and quantify more than 5000 unique glycoforms in Burkitt’s lymphoma cells treated with a series of doses of 2-deoxy-2-fluoro-L-fucose (2FF) and determine the more accurate site-specific glycosylation changes that occurred upon inhibition of fucosylation compared to using MS2 analysis. It revealed that 2FF-sensitive N-glycosylation on key players in BCR-mediated signaling in DG75. Furthermore, 2FF treatment also affects phosphorylation of the key players involving in B cell receptor signaling.
Then we investigated the site-specific quantitative N-glycoproteome in the cell lines of DLBCL subtypes using SugarQuant. More than 7000 unique intact glycopeptides (glycoforms) were quantified in five ABC DLBCL and four GCB DLBCL cell lines. The glycoproteome mapping (intact glycopeptide expressions) in each cell line allows to segregate DLBCL subtypes. The majority of these glycoforms were from the key cell-surface BCR effectors, such as IgM, CD79 and PTPRC. Lastly, we investigated the change of fucosylated glycopeptides in TMD8 cell line upon knockout of the fucosyltransferase FUT8, which is responsible for core-fucose synthesis, and by the treatment with 2FF. The results revealed that FUT8 might also regulate the synthesis of sub/terminal fucose on glycan chain and the inhibition of fucosylation increased the sialyated glycopeptide expression.
Phosphorylation is involved in regulating multiple processes as an important mediator in BCR signaling. Likewise, ubiquitylation plays vital roles in the activation of the nuclear factor-kappaB (NF-κB) pathway in BCR signaling. There are two vital upstream BCR-proximal tyrosine kinases, Bruton’s tyrosine kinase (BTK) and spleen tyrosine kinase (SYK), which regulate the auto-phosphorylation and phosphorylation of other proteins in BCR signaling pathway. Here we investigated the dynamics of downstream phosphorylation and ubiquitylation signaling in ABC DLBCL and GCB DLBCL cell lines upon the inhibitions of BTK and SYK using quantitative proteomics strategy. In the phosphoproteome analysis, a large dataset of quantified phosphorylation sites was obtained in the three ABC and four GCB DLBCL cell lines. BCR signaling in the subtypes of DLBCL cell lines was found to be highly individual in distinct cell lines. These significantly regulated phosphorylation events in each cell line with individual treatment were involved in multiple Reactome pathways, such as, M phase, signaling by Rho GTPases and diseases of signal transduction. Moreover, the gene regulation-related biological processes including chromosome organization and medication, DNA metabolic process, nuclear export, were involved in the DLBCL cell lines. In the ubiquitinome analysis, we identified more than 15,000 ubiquitylation sites in two ABC and one GCB cell lines upon the inhibition of BTK and SYK. The different ubiquitylation events observed in ABC and GCB subtypes revealed distinct BCR signaling pathways in two subtypes. The similar signaling perturbations across each cell line upon BTK and SYK inhibition, which were obtained from the significantly regulated ubiquitylated peptides expression, revealed the cell-type-specific concordance in ubiquitylation regulation upon BTK and SYK inhibition. These ubiquitylation modified proteins who bore the significantly regulated ubi-peptides in the samples were also found to be highly involved in gene regulatory processes.

Das Verständnis von Tumorerkrankungen wurde durch neue technologische und ökonomische Verbesserungen für die Next-Generation-Sequencing Analyse (NGS) gefördert. Die Komplexität der Interpretation genomischer Daten erschwert jedoch die Anwendung von NGS-Analysen im klinischen Kontext. Die Herausforderung besteht darin bei stetig wachsendem medizinischem Wissen dieses im klinischen Kontext zu interpretieren und eine personalisierte Therapieempfehlung abzugeben. Einen ressourcensparenden Ansatz können KI-unterstützende Software-Programme bieten, welche die genomischen Varianten mit der aktuellen Literatur vergleichen, eine Bewertung der Therapieoptionen geben und klinische Studien empfehlen können. In dieser retrospektiven Arbeit wurden Patient:innen mit metastasiertem Brustkrebs (n=77) mittels gezielter NGS-Analyse anhand von sogenannten Genpanels mit 126 bzw. 540 krebsrelevanten Genen im Zeitraum von 01/2019-02/2022 untersucht. Mit Hilfe von bioinformatischen Methoden wurden patientenspezifische genomische Veränderungen mit Behandlungsoptionen abgeglichen. Diese Methoden stützen sich vollständig auf öffentliche Datenbanken über somatische Varianten mit prädiktiver Evidenz für das Ansprechen auf bestimmte Medikamente. Diese Versorgungsforschung einer repräsentativen Kohorte des Universitätsklinikums Frankfurt in Kooperation mit Regionalverbund OncoNet Rhein-Main wurden systematisch ausgewertet inklusive der Bedeutung genomischer Varianten. Das OncoNet Rhein-Main ist eine Kooperation aus führenden onkologischen Zentren und Praxen im Rhein-Main-Gebiet, welche sich als Netzwerk der Aufgabe angenommen haben Patient:innen optimal therapeutisch zu versorgen. Für 51% (39/77) der Patient:innen konnte mindestens eine gezielte Therapieoption mit einem effektivem Biomarker im gleichen Tumortyp gemäß Zulassung der Europäischen Arzneimittelbehörde (EMA) gefunden werden.
Bei 12/77 (16%) wurde mindestens eine Alteration mit einem effektivem Biomarker und einer OFF-Label Therapieoption gefunden. Bei 30% der Patient:innen wurden Veränderungen in optionalen Biomarkern gefunden, welche Resistenzmechanismen erklären. Die umfassende molekulare Analyse von Patient:innen mit fortgeschrittenem Brustkrebs erlaubt die Behandlungsoption zu verbessern und ermöglicht durch die Analyse von bekannten Resistenzmarkern auch den klinischen Verlauf besser zu verstehen. Die interdisziplinäre Besprechung der Befunde im molekularem Tumorboard ist im Hinblick auf kontinuierliches Lernen aller Beteiligten sowie zur Qualitätssicherung eine entscheidende obligate Maßnahme.
Global analysis of halogenated trace gases in the UTLS: from long-lived to short-lived substances

In this dissertation, the distribution of chlorinated and brominated substances in the upper troposphere and lower stratosphere is investigated. These substances contribute significantly to the catalytic decomposition of ozone and are involved in the recurrent formation of the polar ozone hole in the Antarctic winter and spring. The Montreal Protocol, a multilateral environmental treaty to protect the ozone layer, has successfully reduced emissions of long-lived chlorine- and bromine-containing substances. Short-lived chlorinated and brominated substances, some of which are natural and anthropogenic in origin, are not regulated by the Montreal Protocol and it can be assumed that their relative contribution to the stratospheric halogen budget will increase, while the contribution of long-lived compounds will steadily decrease. The distribution of long- and short-lived halogenated substances are part of current research. For the upper troposphere and lower stratosphere, the very short-lived substances are particularly important. The lower stratosphere needs special investigation in this respect, since its composition is influenced by different transport processes. The influences on ozone trends in the lower stratosphere are subject to great uncertainties. Especially in the Southern Hemisphere, the number of observations is very limited.
In this work, the GhOST (Gas chromatograph for Observational Studies using Tracers) instrument was used during the SouthTRAC measurement campaign on the German HALO (High Altitude and LOng range) research aircraft, providing observations of halogenated hydrocarbons in Antarctic late winter to early spring 2019, a generally poorly sampled region. The polar vortex was, compared to previous years, significantly weaker and shifted towards the eastern South Pacific and South America. From the airborne measurements of chlorinated source gases, inorganic chlorine (the sum of active chlorine and reservoir gases; Cly) could be inferred with the result that Cly within the vortex increased up to 1687 ± 19 ppt at 385 K potential temperature, accounting for about 50 % of the total chlorine within the vortex and only 15 % of the total chlorine in the southern mid-latitudes. A comparison with the Northern Hemisphere could be made using the PGS measurement campaign in the Arctic winter 2015/2016. Under comparable conditions (season and distance from the tropopause), only 40 % of the total chlorine was in the inorganic form within the Arctic polar vortex and about 20 % was found in the mid-latitudes of the Northern Hemisphere. In addition, about 540 ppt more Cly was present in the Antarctic vortex than in the Arctic vortex, exceeding the annual variations previously reported for Antarctica.
The mean age of air plays an essential role in the derivation of Cly via the organic source gases, as was done in this work. A new method for determining the mean age of air from observational data has been introduced that accounts for extra-tropical input to the stratosphere in addition to tropical input. This new method was compared with the previously used method, which considered only the tropical input. The new method shows more realistic values especially near the tropopause. On average, the air of the lower stratosphere in the Northern Hemisphere was older than in the Southern Hemisphere by about 0.5 ± 0.3 years. About 65 K above the tropopause, the pattern changed with older air in the mid-latitudes of the Northern Hemisphere, but older air in high latitudes of the Southern Hemisphere, which implies differences in the strength and isolation of the respective polar vortex as well as the wave forcing in the shallow branch of the Brewer-Dobson circulation of the respective hemisphere. This is in good agreement with the distribution of Cly. The difference in the lower stratosphere was not clearly evident with the old method and it can be assumed that investigations of the differences in Cly of Northern and Southern Hemisphere will benefit from the new method.
Finally, the global and seasonal distribution of the two most important representatives of the short-lived brominated substances, CH2Br2 and CHBr3, was investigated. For this purpose, two additional HALO measurement campaigns have been used, the 2012 TACTS measurement campaign and the 2017 WISE measurement campaign, as well as the HIAPER Pole-to-Pole Observations (HIPPO) and Atmospheric Tomography (ATom) measurement campaigns. Observations of CH2Br2 show a pronounced seasonality in the free and upper troposphere of both hemispheres with slightly larger values in the Northern Hemisphere. CHBr3, on the other hand, shows a generally higher variability and lower seasonality with larger mixing ratios at mid and high latitudes in the northern hemispheric winter and autumn. A comparison of the lower stratosphere is limited to autumn and spring of both hemispheres due to the limited data basis of the observations. The distributions in each spring are similar (less than 0.1 ppt differences for e.g., CH2Br2). In hemispheric autumn, larger differences are evident with substantially smaller mixing ratios in the southern hemispheric lower stratosphere. This suggests that the transport processes of the two hemispheres may be different and implies that the input of tropospheric air (flushing) to the Northern Hemisphere lowest stratosphere is more efficient than in the Southern Hemisphere. Vertical profiles of CH2Br2 and CHBr3 in the mid-latitudes of both hemispheres and resulting vertical gradients support this conjecture. However, the Southern Hemisphere data set is insufficient to quantify this difference and further measurements are needed.
Unter den weltweit in ständigem Gebrauch befindlichen Chemikalien befinden sich nicht nur Verbindungen mit akuter toxischer Wirkung, sondern auch solche mit Wirkung auf das endokrine System. Eine große Rolle spielt hier vor allem die Störung der Geschlechtsdifferenzierung und der Reproduktion, ausgelöst durch natürliche oder synthetische Chemikalien mit endokrinem Potential, sogenannte endokrine Disruptoren (ED). Diese Chemikalien können über unterschiedliche Eintragspfade in die Umwelt gelangen. Seit Mitte des 20. Jahrhunderts werden mehr und mehr Fälle bekannt, in denen anthropogene Chemikalien die Pflanzen- und Tierwelt belasten, darunter zahlreiche Befunde zu Störungen des Hormonsystems von Mensch und Tier.
Im Rahmen der Gefahren- und Risikobewertung steht bereits eine Vielzahl harmonisierter Prüfrichtlinien für die Identifizierung und Evaluierung der Effekte von (potentiellen) ED zur Verfügung. Um die Gesamtheit aller potentiellen Interaktionen von ED mit dem Hormonsystem detektieren zu können, ist die In-vivo-Untersuchung an Vertebraten in der Chemikalienregistrierung bisher unabdingbar. Bei der Untersuchung endokriner Potentiale in höheren Vertebraten spielen vor allem nager- und vogelbasierte Testsysteme eine wichtige Rolle. Diese bergen jedoch einen hohen zeitlichen, personellen und finanziellen Aufwand und erfordern eine massive Zahl an Versuchstieren, die für diese Tests benötigt werden. Darüber hinaus beinhalten Tierversuche eine Vielzahl von Problemen einschließlich ethischer Bedenken, die sich als Konsequenz der Tierhaltung unter Versuchsbedingungen ergeben. Ein sehr interessanter und vielversprechender Ansatz zur Reduktion von Tierversuchen ist die Entwicklung eines standardisierten Verfahrens für die Untersuchung potentieller ED in Vogelembryonen. Auf Vogelembryonen basierende In-ovo-Modelle stellen einen Mittelweg zwischen In-vitro- und In-vivo-Testsystemen dar. Mit dem Vogeleitest wird der sich entwickelnde Embryo, das für ED sensitivste Entwicklungsstadium im Leben eines Organismus, berücksichtigt.
Das Ziel der vorliegenden Arbeit war die Entwicklung und Eignungsuntersuchung eines auf dem Embryo des Haushuhns (Gallus gallus domesticus) basierenden Testsystems für den Nachweis von ED. Das resultierende Testsystem soll als Alternativmethode zu bisher etablierten nager- und vogelbasierten Testsystemen für die Untersuchung der Effekte hormonell aktiver Substanzen auf die Geschlechtsdifferenzierung in höheren Wirbeltieren eingesetzt werden.
Die im Rahmen der vorliegenden Dissertation durchgeführten Arbeiten umfassten sowohl die Charakterisierung der Normalentwicklung des Hühnerembryos, unbeeinflusst durch ED, als auch die morphologisch-histologischen Veränderungen der Gonaden von substanzexponierten Embryonen. Für die Untersuchung substanzbedingter Effekte, welche den Schwerpunkt der vorliegenden Arbeit darstellen, wurden die Embryonen gegenüber verschiedenen (anti)estrogenen und (anti)androgenen Substanzen exponiert. Unter Einfluss der Estrogene Bisphenol A (BPA) und 17α-Ethinylestradiol (EE2) entwickelten sich die Keimdrüsen der Männchen zu Ovotestes, während Weibchen ein Ovar mit deutlich schmalerem Cortex ausbildeten. Unter Einfluss der Antiestrogene Fulvestrant und Tamoxifen blieben Effekte auf die Gonaden männlicher Embryonen aus, eine durch das potente Estrogen EE2 hervorgerufene Feminisierung männlicher Gonaden konnte durch beide Substanzen jedoch effektiv antagonisiert werden. Weibchen bilden unter Einfluss von Tamoxifen deutlich schmalere linke Gonaden mit einem missgebildeten Cortex aus. Unter Einfluss der Androgene Tributylzinn (TBT) und 17α-Methyltestosteron (MT) blieben die Effekte auf männliche Embryonen aus, während die Weibchen anatomisch virilisierte Gonaden und eine Reduktion des linken gonadalen Cortex aufwiesen. Allein die untersuchten antiandrogenen Versuchssubstanzen Cyproteronacetat (CPA), Flutamid und p,p´-Dichlorodiphenyldichloroethen (p,p´-DDE) hatten keinen Effekt auf die gonadale Geschlechtsdifferenzierung männlicher und weiblicher Hühnerembryonen.
Es konnte gezeigt werden, dass der Embryo von G. gallus domesticus einen sensitiven Organismus innerhalb des Tierreichs darstellt und hinreichend sensitiv auf eine Reihe von endokrin wirksamen und reproduktionstoxischen Chemikalien reagiert. Anatomische und histologische Änderungen der Gonaden können daher als Biomarker für die Wirkung von ED bei Vögeln nützlich sein. Die untersuchten Endpunkte beziehen sich jedoch auf apikale Effekte und liefern keine mechanistischen Informationen zu den untersuchten Substanzen. Der
Hühnereitest ist eine sinnvolle Ergänzung zur bestehenden OECD-Testbatterie und zeichnet sich besonders durch seine kostengünstige und einfache Handhabung im Labor sowie einfach durchzuführende Tests aus. Durch die vergleichsweise kurze Versuchsdauer von nur 19 Tagen ist ein schnelles Substanzscreening möglich, welches zeitlich deutliche Vorteile gegenüber den etablierten nager- und vogelbasierten Testsystemen hat. Als Alternative zu bisherigen Assays könnte der vorgeschlagene Hühnereitest dazu beitragen, im Rahmen der (öko)toxikologischen Gefährdungs- und Risikobewertung von Chemikalien künftig weniger Versuchstiere zu verwenden.
In der vorliegenden Arbeit konzentrierte ich mich auf mediterrane wirbellose Tierarten, welche sich als Konsequenz ihrer Lebensweise nur schlecht ausbreiten können. Nichtsdestotrotz haben es Süßwasserkrabben der Gattung Potamon und Landschnecken der Gattung Tudorella geschafft, große Gebiete zu besiedeln, die heute durch das Mittelmeer getrennt sind. Für beide Gruppen wurde spekuliert, dass Menschen an ihrer Ausbreitung beteiligt waren. Es war mein Ziel die biogeographischen Muster dieser beiden Gattungen zu analysieren und abzuschätzen, ob Menschen tatsächlich Vektoren ihrer Ausbreitung waren. Meine Analysen fanden auf drei Ebenen statt: Taxonomie, Gattung und Art.
Untersuchungen zur Interaktion zytoplasmatischer Proteine mit dem Serotonintransporter Der plasmamembranständige Serotonintransporter SERT terminiert die Neurotransmission an serotonergen Synapsen durch den schnellen Rücktransport des Neurotransmitters Serotonin (5HT) in die Präsynapse. Neben seinem präsynaptischen Vorkommen wird der SERT auch axonal in Varikositäten immundetektiert. Der Substrattransport selbst unterliegt der Kontrolle des Ca 2 /CaM, des Stickoxid (NO) und des Proteinkinase C (PKC) Signaltransduktionsweges. Letzterer bestimmt die Oberflächenverfügbarkeit des SERT durch dessen Sequestrierung. Die Regulation der Lokalisation und des Substrattransports des SERT sollte durch assoziierte Proteine erzielt werden. Da bislang jedoch keine interagierenden Proteine des SERT bekannt waren, sollten in dieser Arbeit mit Hilfe des HefeZweiHybridSystems zytoplasmatische Interaktionspartner des SERT identifiziert werden. Erste Untersuchungen an heterolog exprimierten Deletionsmutanten der Termini des SERT ergaben, daß diese zytoplasmatischen Domänen für die Funktion des Proteins unabdingbar sind. Im folgenden wurde daher mit dem Carboxyterminus des SERT eine hirnspezifische cDNABibliothek der neugeborenen Ratte durchsucht. Es wurden vier cDNAKlone isoliert, die für putative Interaktionspartner des SERT kodierten. Die Relevanz dieser genetisch nachgewiesenen Interaktionen wurde biochemisch mittels affinitätschromatographischer Studien untersucht, während immunzytochemische Analysen die Interaktionen durch die Umverteilung oder durch die Colokalisation der coexprimierten Proteine in vivo validierte. Pharmakologische Untersuchungen sollten darüberhinaus Aufschluß über den regulatorischen Einfluß der Interaktion auf die Transportfunktion des SERT geben. Nach diesen Aspekten wurden zwei der putativen Interaktionspartner näher untersucht. Der erste untersuchte cDNAKlon kodierte für die ßUntereinheit der intrazellulären Acetylhydrolase des "plateletactivating factor" (ßPAFAH). Affinitätschromatographisch wurde die Interaktion durch die Präzipitation der eukaryotisch überexprimierten ß Untereinheit an einem bakteriell exprimierten Fusionsprotein des SERTCT bestätigt. Die immunzytochemische Analyse zeigte nach der Coexpression der ßPAFAH und dem SERT eine Umlokalisierung des zytoplasmatischen Proteins an Bereiche plasmamembranständiger SERTImmunreaktivität. Durch Wiederaufnahmestudien wurde kein Effekt der ßPAFAH auf die Transportfunktion des SERT ermittelt, weshalb die funktionelle Bedeutung dieser Interaktion vorerst unklar bleibt. Der zweite Klon kodierte für das Rattenspezifische Homolog des myristoylierten Alanin reichen Proteinkinase CSubstrats (MacMARCKS). Der biochemische Nachweis einer Interaktion konnte hier nicht erbracht werden. Dagegen zeigte die immunzytochemische Analyse eine Colokalisation von MacMARCKS und SERT in diskreten Bereichen der Plasmamembran. Hinweise auf die physiologische Bedeutung konnten durch die pharmakologische Analyse gewonnen werden. So induzierte die Coexpression von MacMARCKS und SERT eine Abnahme der maximalen Transportrate (Vmax) um 29,6 ± 18,6 % (n = 3) im Vergleich zu GFPexprimierenden Kontrollzellen. Die Stimulierung der PKC erzielte dagegen kaum eine weitere Reduktion der Vmax im Gegensatz zu den Kontrollzellen, während die Behandlung der Zellen mit einem PKCInhibitor zum Angleich der Vmax der Konrollzellen und der MacMARCKSexprimierenden Zellen führte. Daraus wurde gefolgert, daß MacMARCKS an der Regulation der Transportfunktion des SERT durch dessen Sequestrierung beteiligt ist. Zusammenfassend wurden in dieser Arbeit zwei Interaktionspartner des SERTCT identifiziert, deren mögliche Funktionen in der Regulation der intrazellulären 5HT Konzentration einerseits bzw. der Regulation der Transportfunktion des SERT andererseits liegen könnten.
The peroxisome proliferator activated receptor gamma (PPARgamma) plays an eminent role during alternative activation of macrophages and resolution of inflammation. As an antiinflammatory signaling molecule, it seems likely that it is tightly regulated dependent on the state of the immune response. There is growing evidence that PPARgamma expression is reduced during inflammation, whereas molecular mechanisms are illdefined. Even though, its role in immunosuppression is getting more definite. Apoptotic cells (AC) provoke an active repression of pro-inflammatory responses inter alia by the inhibition of pro-inflammatory cytokine expression or attenuated generation of reactive oxygen species (ROS). The reduced formation of ROS was attributed to PPARgamma activation, while mechanisms behind the reduced cytokine expression remained unclear. Therefore, my Ph.D. thesis addressed the role of PPARgamma during inhibited cytokine synthesis in response to AC and the regulation of PPARgamma expression during an inflammatory response, which was initiated by lipopolysaccharide (LPS) exposure. In the first part of the thesis, I investigated the role of PPARgamma in coordinating the attenuation of pro-inflammatory cytokine expression in response to AC. Exposing murine RAW264.7 macrophages to AC prior to LPS-stimulation, reduced NFKB transactivation and lowered target gene expression of e.g. TNFalpha and IL-6 compared to controls. In macrophages over-expressing a dominant negative (d/n) mutant of PPARgamma, NFKB transactivation in response to LPS was restored, while using macrophages from myeloid lineage-specific conditional PPARgamma knock-out mice proved that PPARgamma transmitted the anti-inflammatory response delivered by AC. Domain analysis revealed that amino acids 32-250 are essential for inhibition of NFKB. Mutation of a SUMOylation (SUMO: small-ubiquitin related modifier) site in this region (K77R) and interfering SUMOylation by silencing the SUMO E3 ligase PIAS1 (protein inhibitor of activated Stat1) eliminated AC-provoked NFKB inhibition and concomitant TNFalpha expression. Chromatin-immunoprecipitation assays demonstrated that AC prevented the LPS-induced removal of nuclear receptor co-repressor (NCoR) from the KB response element within the TNFalpha promoter. I concluded that AC induce PPARgamma SUMOylation to attenuate the removal of NCoR, thereby blocking transactivation of NFKB. This contributes to an anti-inflammatory phenotype shift in macrophages in response to AC, by lowering pro-inflammatory cytokine production. The second part addressed molecular mechanisms responsible for reduced PPARgamma expression upon LPS exposure. PPARgamma gained considerable interest as a therapeutic target during chronic inflammatory diseases. Remarkably, the pathogenesis of diseases such as multiple sclerosis or Alzheimer’s disease is associated with impaired PPARgamma expression. Initiation of an inflammatory response by exposing primary human macrophages to LPS revealed a rapid decline of PPARgamma1 expression. PPARgamma1 mRNA decrease was prevented by inhibition of NFKB and also after pre-treatment with the PPARgamma agonist rosiglitazone, suggesting a NFKB-dependent pathway, because activated PPARgamma is known to inhibit NFKB transactivation. Since promoter activities were not affected by LPS, I focused on mRNA stability and noticed a decreased PPARgamma1 mRNA half-life. RNA stability is often regulated via 3’ untranslated regions (UTRs). Therefore, I analyzed the impact of the PPARgamma-3’UTR by luciferase assays. LPS significantly reduced luciferase activity of pGL3-PPARgamma-3’UTR, suggesting that PPARgamma1 mRNA is destabilized. Deletion of a potential miR-27a/b binding site within the 3’UTR completely restored luciferase activity. Moreover, inhibition of miR-27b, which was induced upon LPS-exposure, partially reversed PPARgamma1 mRNA decay, whereas the mature miR-27 mimicked the effect of LPS. MiR-27b was at least partially induced by NFKB, thus correlating with NFKB-dependent PPARgamma1 mRNA decrease. Since deletion of the miR-27 site also containing an AU-rich element (ARE) completely abrogated LPS-induced reduction but inhibition of miR-27b only partially restored PPARgamma1 mRNA expression, I suggested an additional implication of an ARE-binding protein. I provide evidence that LPS induces miR-27b, which in turn destabilizes PPARgamma1 mRNA. Understanding the molecular mechanism of PPARgamma mRNA destabilization, might help to rationalize inflammatory diseases associated with impaired PPARgamma expression. Even though, further experiments are needed to clarify the potential involvement of ARE-binding proteins.
Beim Übergang vom Wachstum zur Entwicklung entsteht in Dictyostelium discoideum aus einzelnen vegetativ wachsenden Zellen ein multizellulärer Organismus, der in der Lage ist ausdauernde Sporen zu bilden. Während der im multizellulären Verband stattfindenden morphologischen Veränderungen und Zelldifferenzierung nimmt die Proteinkinase A (PKA) eine Schlüsselfunktion ein. Der C-Modul bindende Faktor A (CbfA), ursprünglich aufgrund seiner spezifischen Bindung an eine regulatorische DNA-Sequenz des Non-LTR Retrotransposons TRE5-A.1 in D. discoideum entdeckt, ist für den Übergang vom Wachstum zur Differenzierungsphase essentiell. Zellen der CbfA-Mangelmutante JH.D2 weisen im Vergleich zu denen des Wildtyp-Stamms AX2 ein verlangsamtes Wachstum und eine verzögerte Einleitung der Aggregation und der Entwicklung auf. Charakteristisch für diese CbfA-Mutante ist die Unterexpression der mRNA für die katalytische Untereinheit der Proteinkinase A (PkaC). Da die Expression der PkaC sowohl auf der Transkriptions- als auch auf der Translationsebene reguliert ist, wurde in dieser Arbeit ein monoklonaler Antikörper gegen die katalytische Untereinheit hergestellt, um den Proteingehalt an PkaC in Zellen der CbfA-Mutante zu untersuchen. Dabei konnte eine starke Unterexpression der PkaC in den CbfA-depletierten Zellen nachgewiesen werden. Demnach scheint es einen mittelbaren Zusammenhang zwischen der Menge an CbfA und dem sich selbst verstärkenden cAMP-abhängigen Signaltransduktionssystem der Zellen zu geben, da ein Mangel an cAMP eine fehlende Aktivierung der sich selbst induzierenden PKA nach sich zieht. Für D. discoideum konnte erstmals eine reproduzierbare 2D-gelelektrophoretische Trennmethode etabliert werden, mit der es gelang, differentiell exprimierte Proteine im Stadium der frühen Entwicklung zwischen den untersuchten D. discoideum-Stämmen AX2 und JH.D2 aufzufinden. Die Anwendung der DIGE-Technologie ermöglichte dabei die Detektion von ca. 30-40 differentiell exprimierten Proteinen. Durch die massenspektrometrische Untersuchung dieser Proteine unter Verwendung der MALDI-TOF Methode in Kombination mit dem Peptid-Massen-Fingerabdruck konnten insgesamt ca. 30% der differentiell exprimierten Proteine identifiziert werden. Dabei stellte sich heraus, daß vor allem Proteine mit essentiellen Funktionen im zellulären Stoffwechsel in der CbfA-Mangelmutante JH.D2 auf geringerem Niveau exprimiert wurden als im Wildtyp-Stamm AX2. Dies könnte eine Erklärung für die letalen Auswirkungen eines vollständigen knockouts von CbfA in D. discoideum liefern. Zusätzlich wurde mit Hilfe der DIGE-Technologie die Funktion der C-terminalen Domäne des CbfA-Proteins untersucht. Für diese Experimente wurde eine CbfA-Mutante verwendet, die den C-Terminus konstitutiv überexprimiert. Die daraus resultierenden Proteinmuster haben gezeigt, daß die C-terminale Domäne unabhängig vom Rest des Proteins sowohl induzierenden als auch reprimierenden Einfluß auf die Proteinexpression nimmt. Diese Ergebnisse zeigen eine unerwartet komplexe Funktion des Proteins CbfA in der Regulation von Genen während der untersuchten Lebensphasen von D. discoideum.
